Implications of Advanced Glycation Endproducts in Oxidative Stress and Neurodegenerative Disorders

The reactions of reducing sugars with primary amino groups are the most common nonenzymatic modifications of proteins. Subsequent rearrangements, oxidations, and dehydrations yield a heterogeneous group of mostly colored and fluorescent compounds, termed "Maillard products" or advanced glycation end products (AGEs). AGE formation has been observed on long-lived proteins such as collagen, eye lens crystalline, and in pathological protein deposits in Alzheimer's (AD) and Parkinson's disease (PD) and dialysis-related amyloidosis. AGE-modified proteins are also involved in the complications of diabetes. AGEs accumulate in the the ß-amyloid plaques and neurofibrillary tangles (NFT) associated with AD and in the Lewy bodies characteristic of PD. Increasing evidence supports a role for oxidative stress in neurodegenerative disorders such as AD and PD. AGEs have been shown to contribute towards oxidative damage and chronic inflammation, whereby activated microglia secrete cytokines and free radicals, including nitric oxide (NO). Roles proposed for NO in the pathophysiology of the central nervous system are increasingly diverse and range from intercellular signaling, through necrosis of cells and invading pathogens, to the involvement of NO in apoptosis. Using in vitro experiments, it was shown that AGE-modified bovine serum albumin (BSA-AGE) and AGE-modified ß-amyloid, but not their unmodified proteins, induce NO production in N-11 murine microglia cells. This was mediated by the receptor for AGEs (RAGE) and upregulation of the inducible nitric oxide synthase (iNOS). AGE-induced enzyme activation and NO production could be blocked by intracellular-acting antioxidants: Ginkgo biloba special extract EGb 761, the estrogen derivative, 17ß-estradiol, R-(+)-thioctic acid, and a nitrone-based free radical trap, N-tert.-butyl-*-phenylnitrone (PBN). Methylglyoxal (MG) and 3-deoxyglucosone (3-DG), common precursors in the Maillard reaction, were also tested for their ability to induce the production of NO in N-11 microglia. However, no significant changes in nitrite levels were detected in the cell culture medium. The significance of these findings was supported by in vivo immunostaining of AD brains. Single and double immunostaining of cryostat sections of normal aged and AD brains was performed with polyclonal antibodies to AGEs and iNOS and monoclonal antibodies to Aß and PHF-1 (marker for NFT) and reactive microglia. In aged normal individuals as well as early stage AD brains (i.e. no pathological findings in isocortical areas), a few astrocytes showed co-localisation of AGE and iNOS in the upper neuronal layers of the temporal (Area 22) and entorhinal (Area 28, 34) cortices compared with no astrocytes detected in young controls. In late AD brains, there was a much denser accumulation of astrocytes co-localised with AGE and iNOS in the deeper and particularly upper neuronal layers. Also, numerous neurons with diffuse AGE but not iNOS reactivity and some AGE and iNOS-positive microglia were demonstrated, compared with only a few AGE-reactive neurons and no microglia in controls. Finally, astrocytes co-localised with AGE and iNOS as well as AGE and ß-amyloid were found surrounding mature but not diffuse ß-amyloid plaques in the AD brain. Parts of NFT were AGE-immunoreactive. Immunohistochemical staining of cryostat sections of normal aged and PD brains was performed with polyclonal antibodies to AGEs. The sections were counterstained with monoclonal antibodies to neurofilament components and a-synuclein. AGEs and a-synuclein were colocalized in very early Lewy bodies in the substantia nigra of cases with incidental Lewy body disease. These results support an AGE-induced oxidative damage due to the action of free radicals, such as NO, occurring in the AD and PD brains. Furthermore, the involvement of astrocytes and microglia in this pathological process was confirmed immunohistochemically in the AD brain. It is suggested that oxidative stress and AGEs participate in the very early steps of Lewy body formation and resulting cell death in PD. Since the iNOS gene can be regulated by redox-sensitive transcription factors, the use of membrane permeable antioxidants could be a promising strategy for the treatment and prevention of chronic inflammation in neurodegenerative disorders.
In der vorliegenden Arbeit wurde die Expression unterschiedlicher Marker für die Vaskularisation in verschiedenen Modifikationen zur Unterkieferaugmentation mit autologem kortikospongiösen Beckenkammtransplantaten in-vivo am Schafmodell untersucht. Wie schon aus Voruntersuchungen bekannt, zeigte vor allem die Modifikation des Transplantatlagers in der Kombination einer resorbierbaren Bio-Gide® Membran mit dem Knochenersatzmaterial (KEM) Bio-Oss® und einem kortikospongiösen Transplantat die geringsten Resorptionsraten. Dieser Konditionierung wurden verschiedene andere Modikifationen des Transplantatlagers gegenübergestellt und jeweils die Expression der Vaskularisationsmarker untersucht und verglichen. Sowohl die Untersuchungen zur Expression von Bone Morphogenetic Protein-2 (BMP-2), Vascular Endothelial Growth Factor (VEGF) und seinen Rezeptor-Isoformen VEGF-Rezeptor 1 und VEGF-Rezeptor 2, sowie den NOS-Isoformen eNOS und iNOS zeigen alle eine signifikant vermehrte Expression in der Konditionierung mit Bio-Oss® als KEM und einer resorbierbaren Bio-Gide® Membran. Lediglich die Expression des von-Willebrand-Faktors (vWF) als Marker des Endothels zeigte keine signifikant vermehrte Expression in der mit Bio-Oss® und Bio-Gide® modifizierten Gruppe. Hier wäre die Tatsache, dass der vWF auch als Marker der endothelialen Dysfunktion beschrieben wird und in den klinisch stärker atrophierten Modifikation vermehrt exprimiert wird, eine Erklärungsmöglichkeit. Die Untersuchungen zur Expression der Vaskularisationsmarker hinsichtlich der Regionen im Augmentat zeigen sowohl bei BMP-2 wie auch bei VEGF und seinen Rezeptorisoformen VEGF-R1 und VEGF-R2 eine signifikant vermehrte Expression im Bereich des Transplantates in der Bio-Oss® Modifikation. Diese Ergebnisse stützen den klinischen Aspekt der guten Vaskularisierung des Transplantates und unterstreichen die Bedeutung der Wachstumsfaktoren für die Vaskularisierung im Transplantat. Bei den Färbungen auf NOS findet sich eine signifikant vermehrte Expression im Transplantatlager. Diese Ergebnisse stellen die Bedeutung von NOS für die Regulation der Osteogenese und die direkten Wirkungen auf Osteoblasten und Osteoklasten. Die Tatsache, dass gerade in der Bio-Oss® Modifikation, in der klinisch die geringste Resorption zu beobachten war, signifikant mehr Vaskularisationsmarker exprimiert wurden, ist Nachweis der besseren Vaskularisation dieser Konditionierung und unterstreicht sowohl den Nutzen von KEM wie Bio-Oss® bei Transplantationen autologen Knochens, wie auch die Möglichkeiten der therapeutischen Verwendung einzelner Wachstumsfaktoren wie z. B. BMP-2.
Background
The mechanisms by which vaccinia virus (VACV) interacts with the innate immune components are complex and involve different mechanisms. iNOS-mediated NO production by myeloid cells is one of the central antiviral mechanisms and this study aims to investigate specifically whether iNOS-mediated NO production by myeloid cells, is involved in tumor eradication following the virus treatment.
Methods
Human colon adenocarcinoma (HCT-116) xenograft tumors were infected by VACV. Infiltration of iNOS\(^{+}\) myeloid cell population into the tumor, and virus titer was monitored following the treatment. Single-cell suspensions were stained for qualitative and quantitative flow analysis. The effect of different myeloid cell subsets on tumor growth and colonization were investigated by depletion studies. Finally, in vitro culture experiments were carried out to study NO production and tumor cell killing. Student’s t test was used for comparison between groups in all of the experiments.
Results
Infection of human colon adenocarcinoma (HCT-116) xenograft tumors by VACV has led to recruitment of many CD11b\(^{+}\) ly6G\(^{+}\) myeloid-derived suppressor cells (MDSCs), with enhanced iNOS expression in the tumors, and to an increased intratumoral virus titer between days 7 and 10 post-VACV therapy. In parallel, both single and multiple rounds of iNOS-producing cell depletions caused very rapid tumor growth within the same period after virus injection, indicating that VACV-induced iNOS\(^{+}\) MDSCs could be an important antitumor effector component. A continuous blockade of iNOS by its specific inhibitor, L-NIL, showed similar tumor growth enhancement 7–10 days post-infection. Finally, spleen-derived iNOS+ MDSCs isolated from virus-injected tumor bearing mice produced higher amounts of NO and effectively killed HCT-116 cells in in vitro transwell experiments.
Conclusions
We initially hypothesized that NO could be one of the factors that limits active spreading of the virus in the cancerous tissue. In contrast to our initial hypothesis, we observed that PMN-MDSCs were the main producer of NO through iNOS and NO provided a beneficial antitumor effect, The results strongly support an important novel role for VACV infection in the tumor microenvironment. VACV convert tumor-promoting MDSCs into tumor-killing cells by inducing higher NO production.
Diese Arbeit befasst sich mit dem NO-Stoffwechsel und der Wirkung von Rosiglitazon (RGZ) im ischämischen akuten Nierenversagen (iANV). Im Rattenmodell wurde mittels 60-minütigem Clamping beider Aa. renales ein iANV induziert. Die Unterteilung erfolgte in die Gruppen mit Gefäßclamping jeweils ohne bzw. mit Gabe von RGZ (Clamp+NaCl bzw. Clamp+RGZ) sowie in die entsprechenden Gruppen mit Scheinoperation (Sham+NaCl bzw. Sham+RGZ). 24 Stunden nach dem Eingriff wurde photometrisch die Inulin- und PAH-Clearance bestimmt. Die Expression der Enzyme, Proteine und Metabolite des NO-Stoffwechsels wurde mittels Western-Blot, real time-PCR aus Nierenhomogenisaten oder Flüssig¬chromato¬graphie mit Massenspektrometrie-Kopplung (LC-MS/MS) aus Serumproben quantitativ bestimmt. In der unbehandelten Clamp-Gruppe zeigte sich ein deutlicher Abfall (90%) der Inulin- und PAH-Clearance und PAH-Nettosekretion. Die Gabe von RGZ besserte die Inulin- und PAH-Clearance sowie die PAH-Nettosekretion. Die Applikation von RGZ im iANV bewirkte keine aktivitätssteigernde Phosphorylierung der endothelialen NO-Synthase (eNOS) an Serine 1177. An eNOS Serine 633 nahm durch RGZ die Phosphorylierung ab. Auch das, an vielen Signalkaskaden beteiligte, Akt zeigte keine vermehrte Aktivierung. Die Gesamtexpression der eNOS-mRNA wurde durch RGZ im iANV signifikant geringer (auf 60% des Ausgangswertes) vermindert als in unbehandelten Tieren (20% des Ausgangswertes). Im iANV stieg die Expression der induzierbaren NO-Synthase (iNOS) - mRNA um das vierfache an, dieser Anstieg wurde durch Gabe von RGZ halbiert. Der verminderte Anstieg von iNOS kann als Erklärung für den Anstieg von eNOS dienen. Der Anstieg von ED-1 als Marker der Inflammationsreaktion sowie der Anstieg der Cleaved caspase 3 als Marker der Apoptosereaktion im iANV konnte nach der Applikation von RGZ nicht mehr nachgewiesen werden. Insgesamt schienen Inflammationsreaktion und Apoptose keinen signifikanten Einfluss auf die funktionellen Parameter im iANV zu besitzen. Das L-Argininderivat „Asymmetrisches Dimethylarginin“ (ADMA), das eNOS kompetitiv hemmt, stieg im iANV in der Clamp+NaCl und in der Clamp+RGZ Gruppe um ungefähr 20% an. Das an der Synthese von ADMA beteiligte Enzym PRMT 1 (Proteinargininmethyltransferase) und das ADMA-abbauende Enzym DDAH II (Dimethylarginindiaminohydrolase) wurden im iANV nicht reguliert. DDAH I, ein funktionsgleiches Isomer von DDAH II, zeigte im iANV eine Herabregulation um 20%. Diese Herabregulation könnte den Anstieg von Serum-ADMA im iANV erklären. Die Applikation von RGZ hatte weder auf ADMA noch auf DDAH einen regulatorischen Effekt. Die Halbierung der Expression von PRMT 1 durch RGZ hatte keinen Einfluss auf den ADMA-Serumspiegel. L-Arginin (L-Arg) stieg mit 60% im iANV deutlich stärker an als ADMA und könnte den Anstieg von ADMA kompensieren. Der Anstieg von L-Arg war von RGZ unabhängig. Der Quotient aus L-Arg und ADMA stieg in unbehandelten Tieren im iANV signifikant an, unter der Gabe von RGZ jedoch nicht. Dieser fehlende Anstieg wirkte sich nicht wesentlich auf die Produktion von NO aus. Folglich stellen sowohl ADMA als auch der L-Arg/ADMA Quotient keine Erklärung für die unzureichende funktionelle Wirkung einer Expressionssteigerung von eNOS unter RGZ im iANV dar. „Symmetrisches Dimethylarginin“ (SDMA) inhibiert als Isomer von ADMA die Aufnahme von L-Arg in die Zelle kompetitiv. SDMA zeigte im iANV einen Anstieg um fast 400 % im Vergleich zu den Shamtieren. SDMA wurde durch die Gabe von RGZ nicht reguliert. Hieraus wurde die Hypothese abgeleitet, dass der erhöhte SDMA-Spiegel den transzellulären L-Arg-Transport blockiert. Dies kann den Serumanstieg von L-Arg im iANV erklären und würde zu einem intrazellulären Mangel an L-Arg führen. Die durch RGZ bewirkte Steigerung der Expression von eNOS bliebe ineffektiv, da durch den Substratmangel die Produktion von NO nicht adäquat ansteigen könnte. Das L-Arg-Paradox im iANV beschreibt die Tatsache, dass die Applikation von L-Arg im iANV zu einer Mehrproduktion von NO durch eNOS führt, obwohl der Serumspiegel von L-Arg bereits vor Applikation klar über dem Sättigungsbereich von eNOS liegt. Da der Anstieg von ADMA im iANV durch den deutlich höheren Anstieg von L-Arg überkompensiert wird, scheint ADMA als Erklärung des Paradoxes nicht hinreichend. Der deutliche Anstieg von SDMA im iANV hingegen könnte über eine Blockade des L-Arg-Transporters zu einem intrazellulären Mangel an L-Arg führen. Diese kompetitive Blockade könnte durch die Applikation von L-Arg aufgehoben werden. Somit wäre SDMA eine Erklärung für das L-Arg Paradox. Zusammenfassend wurde in dieser Arbeit gezeigt, dass der starke Anstieg von SDMA möglicherweise dem protektiven Effekt von RGZ im iANV entgegenwirkt. Außerdem konnte mit dem Anstieg von SDMA ein neuer Erklärungsansatz des L-Arg-Paradoxes im iANV aufgezeigt werden.
