In order to survive, organisms avoid threats and seek rewards. Classical conditioning is a simple model to explain how animals and humans learn associations between events that allow them to predict threats and rewards efficiently. In the classical conditioning paradigm, a neutral stimulus is paired with a biologically significant event (the unconditioned stimulus – US). In virtue of this association, the neutral stimulus acquires affective motivational properties, and becomes a conditioned stimulus (CS+). Defensive responses emerge for pairings with an aversive US (e.g., pain), and appetitive responses emerge for pairing with an appetitive event (e.g., reward). It has been observed that animals avoid a CS+ when it precedes an aversive US during a training phase (CS+ US; forward conditioning); whereas they approach a CS+ when it follows an aversive US during the training phase (US CS+; backward conditioning). These findings indicate that the CS+ acquires aversive properties after a forward conditioning, whereas acquires appetitive properties after a backward conditioning. It is thus of interest whether event timing also modulates conditioned responses in such an opponent fashion in humans, who are capable of explicit cognition about the associations. For this purpose, four experiments were conducted in which a discriminative conditioning was applied in groups of participants that only differed in the temporal sequence between CS+ onset and US onset (i.e., the interstimulus interval – ISI). During the acquisition phase (conditioning), two simple geometrical shapes were presented as conditioned stimuli. One shape (CS+) was always associated with a mild painful electric shock (i.e., the aversive US) and the other one (CS-) was never associated with the shock. In a between-subjects design, participants underwent either forward or backward conditioning. During the test phase (extinction), emotional responses to CS+ and CS- were tested and the US was never presented. Additionally, a novel neutral shape (NEW) was presented as control stimulus. To assess cognitive components, participants had to rate both the valence (the degree of unpleasantness or pleasantness) and the arousal (the degree of calmness or excitation) associated with the shapes before and after conditioning. In the first study, startle responses, an ancestral defensive reflex consisting of a fast twitch of facial and body muscles evoked by sudden and intense stimuli, was measured as an index of stimulus implicit valence. Startle amplitude was potentiated in the presence of the forward CS+ whilst attenuated in the presence of the backward CS+. Respectively, the former response indicates an implicit negative valence of the CS+ and an activation of the defensive system; the latter indicated an implicit positive valence of the CS+ and an activation of the appetitive system. In the second study, the blood-oxygen level dependent (BOLD) response was measured by means of functional magnetic resonance imaging (fMRI) to investigate neural responses after event learning. Stronger amygdala activation in response to forward CS+ and stronger striatum activation in response to backward CS+ were found in comparison to CS-. These results support the notion that the defensive motivational system is activated after forward conditioning since the amygdala plays a crucial role in fear acquisition and expression. Whilst the appetitive motivational system is activated after backward conditioning since the striatum plays a crucial role in reward processing. In the third study, attentional processes underlying event learning were observed by means of steady-state visual evoked potentials (ssVEPs). This study showed that both forward and backward CS+ caught attentional resources. More specifically, ssVEP amplitude was higher during the last seconds of forward CS+ that is just before the US, but during the first seconds of backward CS+ that is just after the US. Supposedly, attentional processes were located at the most informative part of CS+ in respect to the US. Participants of all three studies rated both forward and backward CS+ more negative and arousing compared to the CS-. This indicated that event timing did not influence verbal reports similarly as the neural and behavioral responses indicating a dissociation between the explicit and implicit responses. Accordingly, dual process theories propose that human behavior is determined by the output of two systems: (1) an impulsive implicit system that works on associative principles, and (2) a reflective explicit system that functions on the basis of knowledge about facts and values. Most importantly, these two systems can operate in a synergic or antagonistic fashion. Hence, the three studies of this thesis congruently suggest that the impulsive and the reflective systems act after backward association in an antagonistic fashion. In sum, event timing may turn punishment into reward in humans even though they subjectively rate the stimulus associated with aversive events as being aversive. This dissociation might contribute to understand psychiatric disorders, like anxiety disorders or drug addiction.
Teil 1: Aus früheren Arbeiten geht hervor, dass Fibomyalgiepatienten über eine höhere Schmerzstärke in bezug auf ihren klinischen Schmerz nach negativem emotionalen Priming verglichen mit positivem Priming berichten als Patienten mit muskuloskelettalem Schmerz. Um die affektive Modulation von Druckschmerz bei Fibromyalgiepatienten (n = 30) unter kontrollierten Bedingungen im Vergleich mit Gesunden und Schmerzerkrankungen geklärter Genese (d.h. organisch oder psychisch) beschreiben zu können, wurde Schmerz experimentell induziert. Neben 30 Gesunden (schmerzfreien Personen) dienten 30 Rückenschmerzpatienten, bei denen eine organische Schmerzgenese vermutet wurde, und 30 somatoforme Schmerzpatienten als Vergleichsgruppen. Für die letzte Gruppe, bei der eine psychische Schmerzgenese angenommen wurde, wurde die gleiche Schmerzmodulation wie für die Fibromyalgiepatienten vermutet. Als Primes dienten positive, neutrale, negative und schmerzbezogene Bilder des International Affective Picture Systems. Schmerz wurde über einen konstanten tonischen Druckreiz ausgelöst; als abhängige Variable wurde die empfundene Schmerzstärke erfasst. Über alle Versuchspersonen hinweg modulierte die Bildervalenz die Schmerzstärke: nach schmerzbezogenen Bildern war die Schmerzstärke höher als nach negativen und nach negativen Bildern war sie höher als nach neutralen. Die Schmerzstärken nach neutralen im Vergleich zu positiven Bildern unterschieden sich jedoch nicht signifikant. Somatoforme Schmerzpatienten berichteten über höhere Schmerzstärken als Rückenschmerzpatienten und Gesunde. Die Fibromyalgiepatienten zeigten ähnlich hohe Schmerzstärken wie die somatoforme Schmerzgruppe, doch unterschieden sie sich weder von den Gesunden noch von den Rückenschmerzpatienten. Zwischen Priming und Gruppe gab es keine Interaktion: Die affektive Schmerzmodulation war bei den Fibromyalgie- und den somatoformen Schmerzpatienten nicht spezifisch verändert, doch die somatoformen Schmerzpatienten zeigten eine erhöhte Druckschmerzsensibilität als Gesunde und Rückenschmerzpatienten. Teil 2: Es wurde oft vermutet, dass sich Fibromyalgiepatienten hinsichtlich soziodemographischer Daten und psychologischer Merkmale von Schmerzpatienten unterscheiden, deren Schmerz organischer Genese ist wie bei Patienten mit (rheumatoider) Arthritis. Ob sie sich diesbezüglich von Patienten mit somatoformer Schmerzstörung unterscheiden, wurde bis jetzt noch nicht geklärt. Um das psychologische Profil von Fibromyalgiepatienten zu spezifizieren, wurden 25 Fibromyalgie- mit 29 somatoformen Schmerz-, 27 Rückenschmerzpatienten und 30 gesunden (schmerfreien) Kontrollpersonen (alle Versuchspersonen hatten am Teil 1 der Studie teil genommen) in bezug auf Unterschiede in soziodemographischen Merkmalen, klinischer Schmerzstärke, schmerzbezogene Selbstinstruktionen, Partnerreaktionen, Stressbewältigungsstrategien und Selbstwirksamkeits- und externale Kontrollüberzeugungen verglichen. Alle drei Schmerzgruppen berichteten über eine stärkere Depression, größere Trait-Angt und größere affektive Verstimmung als die Gesunden. Fibromyalgie- und somatoforme Schmerzpatienten zeigten außerdem eine niedrigere Lebenskontrolle, weniger Aktivitäten außer Haus und vermehrt negative Stressbewältigungsstrategien als die Gesunden. Die Fibromalgiepatienten berichteten zudem über eine stärkere Somatisierung und größere affektive und sensorische Schmerzstärken als die Rückenschmerzpatienten, aber unterschieden sich nicht von den somatoformen Schmerzpatienten. Des weiteren gaben die Fibromyalgiepatienten mehr Schmerzen an verschiedenen Körperstellen an als die Rückenschmerzpatienten. Ein unerwartetes Ergebnis war, dass die somatoformen Schmerzpatienten eine größere Ausprägung im Merkmal Bestrafung (i. s. einer Partnerreaktion) als die Fibromyalgiepatienten (und die Gesunden) zeigten.
Attention-Deficit/Hyperactivity Disorder (ADHD) is characterized by symptoms of inattentiveness and hyperactivity/impulsivity. Besides, increasing evidence points to ADHD patients showing emotional dysfunctions and concomitant problems in social life. However, systematic research on emotional dysfunctions in ADHD is still rare, and to date most studies lack conceptual differentiation between emotion processing and emotion regulation. The aim of this thesis was to systematically investigate emotion processing and emotion regulation in adult ADHD in a virtual reality paradigm implementing social interaction. Emotional reactions were assessed on experiential, physiological, and behavioral levels.
Experiment 1 was conducted to develop a virtual penalty kicking paradigm implying social feedback and to test it in a healthy sample. This paradigm should then be applied in ADHD patients later on. Pleasant and unpleasant trials in this paradigm consisted of hits respectively misses and subsequent feedback from a virtual coach. In neutral trials, participants were teleported to different spots of the virtual stadium. Results indicated increased positive affectivity (higher valence and arousal ratings, higher zygomaticus activations, and higher expression rates of positive emotional behavior) in response to pleasant compared to neutral trials. Reactions to unpleasant trials were contradictory, indicating increased levels of both positive and negative affectivity, compared to neutral trials. Unpleasant vs. neutral trials revealed lower valence ratings, higher arousal ratings, higher zygomaticus activations, slightly lower corrugator activations, and higher expression rates of both positive and negative emotional behavior. The intensity of emotional reactions correlated with experienced presence in the virtual reality.
To better understand the impact of hits or misses per se vs. hits or misses with coach feedback healthy participants’ emotional reactions, only 50% of all shots were followed by coach feedback in experiment 2. Neutral trials consisted of shots over the free soccer field which were followed by coach feedback in 50 % of all trials. Shots and feedback evoked more extreme valence and arousal ratings, higher zygomaticus activations, lower corrugator activations, and higher skin conductance responses than shots alone across emotional conditions. Again, results speak for the induction of positive emotions in pleasant trials whereas the induction of negative emotions in unpleasant trials seems ambiguous. Technical improvements of the virtual reality were reflected in higher presence ratings than in experiment 1.
Experiment 3 investigated emotional reactions of adult ADHD patients and healthy controls after emotion processing and response-focused emotion regulation. Participants successively
went through an ostensible online ball-tossing game (cyber ball) inducing negative emotions, and an adapted version of the virtual penalty kicking game. Throughout cyber ball, participants were included or ostracized by two other players in different experimental blocks. Participants were instructed to explicitly show, not regulate, or hide their emotions in different experimental blocks. Results provided some evidence for deficient processing of positive emotions in ADHD. Patients reported slightly lower positive affect than controls during cyber ball, gave lower valence ratings than controls in response to pleasant penalty kicking trials, and showed lower zygomaticus activations than controls especially during penalty kicking. Patients in comparison with controls showed slightly increased processing of unpleasant events during cyber ball (higher ratings of negative affect, especially in response to ostracism), but not during penalty kicking. Patients showed lower baseline skin conductance levels than controls, and impaired skin conductance modulations. Compared to controls, patients showed slight over-expression of positive as well as negative emotional behavior. Emotion regulation analyses revealed no major difficulties of ADHD vs. controls in altering their emotional reactions through deliberate response modulation. Moreover, patients reported to habitually apply adaptive emotion regulation strategies even more frequently than controls. The analyses of genetic high-risk vs. low-risk groups for ADHD across the whole sample revealed similar results as analyses for patients vs. controls for zygomaticus modulations during emotion processing, and for modulations of emotional reactions due to emotion regulation.
To sum up, the virtual penalty kicking paradigm proved to be successful for the induction of positive, but not negative emotions. The importance of presence in virtual reality for the intensity of induced emotions could be replicated. ADHD patients showed impaired processing of primarily positive emotions. Aberrations in negative emotional responding were less clear and need further investigation. Results point to adult ADHD in comparison to healthy controls suffering from baseline deficits in autonomic arousal and deficits in arousal modulation. Deficits of ADHD in the deliberate application of response-focused emotion regulation could not be found.
Die Aufmerksamkeitsdefizit-/Hyperaktivitätsstörung (ADHS) zeichnet sich durch eine starke klinische Heterogenität aus, deren Ursachen bislang noch nicht völlig geklärt sind. Als erfolgversprechendes Erklärungsmodell hat sich das Endophänotypenkonzept herausgestellt, das davon ausgeht, dass unterschiedliche Dysfunktionen den vielfältigen klinischen Phänotypen der ADHS zugrunde liegen. Emotional-motivationalen Defiziten wird hierbei eine große Bedeutung beigemessen, allerdings wurden diese bislang kaum untersucht. Die wenigen vorliegenden Studien bezogen sich auf subjektive Daten und differenzierten nicht nach klinischen Subgruppen, wodurch sich heterogene Ergebnisse ergaben. Die vorliegende Arbeit hatte somit zum Ziel, einen emotional-motivationalen Endophänotyp der ADHS bei unterschiedlichen klinischen Subgruppen von ADHS-Patienten mit subjektiven und objektiven psychophysiologischen Daten zu untersuchen. Dies wurde mithilfe eines emotionalen Bilderparadigmas untersucht, bei dem neben subjektiven Bildbewertungen die affektmodulierte Startlereaktion als Valenzindikator und die elektrodermale Aktivität als Arousalindikator emotional-motivationaler Reaktivität gemessen wurden. Studie 1 (N = 325) konzentrierte sich auf die klinischen Subtypen der ADHS bei erwachsenen Patienten. Diese Studie konnte aufzeigen, dass ADHS-Patienten in Abhängigkeit vom ADHS-Subtypus Defizite in der emotional-motivationalen Reaktivität aufwiesen. Der Mischtypus und der hyperaktiv-impulsive Typus zeichneten sich durch eine verminderte Reaktivität auf positive Stimuli aus, was sich in einer reduzierten Startleinhibition widerspiegelte. Der hyperaktiv-impulsive Typus reagierte zudem vermindert auf negative Stimuli, was sich in einer verringerten Startlepotenzierung zeigte. Im Gegensatz dazu reagierte der unaufmerksame Typus vergleichbar zu Kontrollpersonen mit einer leicht geringeren Startleinhibition bei positiven Stimuli. Die besonders beeinträchtigte emotionale Reaktivität des hyperaktiv-impulsiven Typus spiegelte sich auch in einem Bias zu positiveren Bewertungen aller Bilder und einer verminderten Arousaleinschätzung negativer Stimuli bei Männern dieses Typus wider. Die ADHS-Patienten zeigten keine elektrodermalen Arousaldysfunktionen, wobei auch hier der hyperaktiv-impulsive Typus deskriptiv auffallend abgeflachte Werte in der Reaktivität auf emotionale Stimuli aufwies. Die gefundenen Dysfunktionen könnten zu hyperaktiv-impulsivem Verhalten und Sensation Seeking durch die Suche nach Verstärkern führen. Gleichzeitig könnten die Ergebnisse die starken sozialen Dysfunktionen und antisoziales Verhalten von ADHS-Patienten mit hyperaktiv-impulsiven Symptomen erklären. Zur Berücksichtigung von Entwicklungsaspekten im Endophänotypenmodell und Untersuchung des emotional-motivationalen Endophänotyps bei Kindern mit ADHS konzentrierte sich Studie 2 (N = 102) auf Jungen mit ADHS, die mit und ohne Methylphenidat untersucht wurden. Durch die zusätzliche Methylphenidatgruppe sollten die klinische Relevanz emotional-motivationaler Dysfunktionen belegt und Erkenntnisse zur Wirkweise von Methylphenidat gewonnen werden. Diese Studie konnte aufzeigen, dass sich ADHS-Kinder ohne Methylphenidat durch Hypoarousal auszeichneten, was sich in verminderten Hautleitfähigkeitsreaktionen auf die Bilder und Startletöne sowie einem verminderten tonischen Hautleitfähigkeitsniveau widerspiegelte. Diese Dysfunktionen wurden durch Methylphenidat normalisiert. Die Startledaten konnten aus methodischen Gründen die affektive Modulation bei den Kindern nicht abbilden. Diese Daten lieferten jedoch Hinweise, dass Methylphenidat die emotional-motivationale Reaktivität steigerte, da die ADHS-Kinder mit Methylphenidat eine verstärkte Startlereaktivität während der Bildbetrachtung aufwiesen. Das gefundene Hypoarousal auf Stimuli könnte dazu führen, dass vermindert auf Umweltreize und auch auf Belohnung und Bestrafung reagiert wird. Dies könnte soziale Dysfunktionen und externalisierendes Verhalten nach sich ziehen. Hyperaktiv-impulsives Verhalten und Sensation Seeking könnten kompensatorisch zur Anhebung des Arousals resultieren. Unaufmerksamkeit könnte durch einen suboptimalen Aktiviertheitsgrad bedingt sein. Methylphenidat könnte durch eine Steigerung des Arousals und die Verstärkung der emotionalen Reaktivität diesen Symptomen entgegenwirken. Die vorliegende Arbeit konnte somit als erste einen emotional-motivationalen Endophänotyp der ADHS unter Berücksichtigung valenz- und arousalbezogener Maße bei unterschiedlichen klinischen Subgruppen mit objektiven psychophysiologischen Parametern aufzeigen. Die Normalisierung des Hypoarousals von der Kindheit zum Erwachsenenalter könnte mit der Veränderung der ADHS-Symptome über die Entwicklung zusammenhängen. Die weitere Erforschung des Endophänotypenmodells der ADHS ist eine wichtige Aufgabe für die Zukunft. Die vorliegende Arbeit versuchte, hierzu einen Beitrag zu leisten.

Geteilte Emotionen
In dieser Arbeit verteidige ich die Auffassung, dass geteilte Emotionen nicht nur metaphysisch möglich, sondern darüber hinaus auch ein integraler Bestandteil unseres sozialen Lebens sind. Dabei diskutiere ich in dieser Arbeit zunächst die begrifflichen und metaphysischen Vorannahmen, welche einer philosophischen Analyse von geteilten Emotionen zugrunde liegen. Ausgehend davon versuche ich eine Theorie der Emotionen zu entwickeln, welche Emotionen als wesentlich in soziale Kontexte eingebettet begreift. Daneben beleuchte ich die kognitiven und non-kognitiven Mechanismen, etwa die Gefühlsansteckung, Empathie oder geteilte Intentionalität, welche Konstitutiv für die Hervorbringung geteilter Emotionen sind. Das Ergebnis meiner Arbeit besteht darin, dass ich dafür argumentiere, dass die stark intellektualistische Ausrichtung in der gegenwärtigen Sozialphilosophie und in der Philosophie der Emotionen dazu führt, dass die entsprechenden Theorien die Phänomenologie geteilter emotionaler Erfahrungen nicht adäquat erfassen können. Als Konsequenz dessen versuche ich ein alternatives Modell zur Erklärung von geteilten Emotionen zu entwickeln, welches die Bedeutung von körperlicher Interaktion hervorhebt.

For formative evaluations of user experience (UX) a variety of methods have been developed over the years. However, most techniques require the users to interact with the study as a secondary task. This active involvement in the evaluation is not inclusive of all users and potentially biases the experience currently being studied. Yet there is a lack of methods for situations in which the user has no spare cognitive resources. This condition occurs when 1) users' cognitive abilities are impaired (e.g., people with dementia) or 2) users are confronted with very demanding tasks (e.g., air traffic controllers). In this work we focus on emotions as a key component of UX and propose the new structured observation method Proxemo for formative UX evaluations. Proxemo allows qualified observers to document users' emotions by proxy in real time and then directly link them to triggers. Technically this is achieved by synchronising the timestamps of emotions documented by observers with a video recording of the interaction.
In order to facilitate the documentation of observed emotions in highly diverse contexts we conceptualise and implement two separate versions of a documentation aid named Proxemo App. For formative UX evaluations of technology-supported reminiscence sessions with people with dementia, we create a smartwatch app to discreetly document emotions from the categories anger, general alertness, pleasure, wistfulness and pride. For formative UX evaluations of prototypical user interfaces with air traffic controllers we create a smartphone app to efficiently document emotions from the categories anger, boredom, surprise, stress and pride. Descriptive case studies in both application domains indicate the feasibility and utility of the method Proxemo and the appropriateness of the respectively adapted design of the Proxemo App.
The third part of this work is a series of meta-evaluation studies to determine quality criteria of Proxemo. We evaluate Proxemo regarding its reliability, validity, thoroughness and effectiveness, and compare Proxemo's efficiency and the observers' experience to documentation with pen and paper. Proxemo is reliable, as well as more efficient, thorough and effective than handwritten notes and provides a better UX to observers. Proxemo compares well with existing methods where benchmarks are available.
With Proxemo we contribute a validated structured observation method that has shown to meet requirements formative UX evaluations in the extreme contexts of users with cognitive impairments or high task demands. Proxemo is agnostic regarding researchers' theoretical approaches and unites reductionist and holistic perspectives within one method.
Future work should explore the applicability of Proxemo for further domains and extend the list of audited quality criteria to include, for instance, downstream utility. With respect to basic research we strive to better understand the sources leading observers to empathic judgments and propose reminisce and older adults as model environment for investigating mixed emotions.
Hintergrund: Ausgangspunkt für diese Arbeit sind korrelative Befunde, die das Bestehen eines Zusammenhangs zwischen einer negativen Emotionalität und der Verschlimmerung einer Schmerzproblematik nahe legen. Die motivationale Priming-Hypothese von Lang bietet einen theoretischen Rahmen zur Erklärung der Wirkung von Emotionen auf die Schmerzwahrnehmung. Allerdings wurden die Vorhersagen dieser Theorie bisher hauptsächlich für den Schreckreflex untersucht und müssen für die Schmerzreizverarbeitung noch validiert werden. Bis heute ist es außerdem eine offene Frage, welche Rolle Aufmerksamkeitsprozesse bei der affektiven Schmerzmodulation spielen. Experiment 1. Fragestellung war, ob die motivationale Priming-Hypothese auch für die Wahrnehmung und Verarbeitung von Schmerzreizen gültig ist. Methode: 30 Probanden erhielten schmerzhafte und nicht schmerzhafte elektrische Reize, während sie positive, neutrale und negative Bilder betrachteten. Zur Erfassung der Schmerzwahrnehmung wurden Schmerzintensitätsratings erhoben und zur Messung der kortikalen Schmerzreizverarbeitung wurden somatosensorisch evozierte Potentiale (SEPs) aufgezeichnet. Ergebnisse: Die Valenz der Bilder beeinflusst die Intensitätsratings und die N150 Amplituden mit höheren Ratings und N150 Amplituden bei negativen als bei positiven Bildern. Dagegen wurde die Amplitude der P260 durch das Arousal der Bilder moduliert mit höheren Amplituden bei neutralen als bei erregenden Hintergrundbildern. Interpretation: Die Vorhersagen der motivationalen Priming-Hypothese scheinen auch für die Verarbeitung und Wahrnehmung von Schmerzreizen valide zu sein. Während die Modulation der N150 Amplitude eine affektive Schmerzmodulation zu reflektieren scheint, ist die Arousalmodulation der P260 vermutlich auf schmerzunspezifische Aufmerksamkeitsprozesse zurückzuführen. Experiment 2. Fragestellung war, ob die affektive Schmerzmodulation von Aufmerksamkeitsprozessen unabhängig ist und ob unterschiedliche neuronale Mechanismen der aufmerksamkeitsbasierten und der affektiven Schmerzmodulation zugrunde liegen. Methoden: 30 Probanden sahen positive, neutrale und negative Bilder, während sie schmerzhafte und nicht schmerzhafte elektrische Reize erhielten. Zur Aufmerksamkeitsmanipulation wurden die Probanden vor jeder Bildpräsentation instruiert, sich auf die Bilder, auf die Intensität oder auf die Unangenehmheit des elektrischen Reizes zu konzentrieren. Zur Schmerzevaluation wurden affektive und sensorische Schmerzratings und SEPs erhoben. Die neuronalen Quellen der N150 und P260 Komponenten wurden mit Hilfe einer LORETA-Analyse bestimmt. Ergebnisse: Die Aufmerksamkeitsmanipulation beeinflusste die sensorischen Schmerzratings: Die Ratings waren am höchsten, wenn die Aufmerksamkeit auf die Reizintensität gerichtet war. Die Affektinduktion wirkte sich hauptsächlich auf die affektiven Schmerzratings aus mit höheren Ratings bei negativen als bei positiven Bildern. N150 Amplituden wurden durch die Valenz der affektiven Bilder moduliert mit höheren Amplituden bei negativen als bei positiven Bildern. Die Aufmerksamkeitsmanipulation hatte keinen Effekt auf die N150 Amplituden. P260 Amplituden wurden durch das Arousal der Bilder moduliert mit höheren Amplituden bei neutralen als bei erregenden Bildern. Außerdem waren die P260 Amplituden am höchsten bei einem Aufmerksamkeitsfokus auf die Reizintensität. Die LORETA-Analyse erbrachte für die N150 bei schmerzhaften im Vergleich zu nicht schmerzhaften Reizen eine maximale Aktivierung im ACC und im Präcuneus und für die P260 im superioren und medialen frontalen Gyrus und im ACC. Diskussion: Beide Experimente unterstützen die motivationale Priming-Hypothese für die Wahrnehmung und Verarbeitung von Schmerzreizen. Dies zeigt sich in einer affektiven Modulation der sensorischen und affektiven Schmerzratings und der N150 Amplituden. Die Befunde des zweiten Experiments deuten außerdem darauf hin, dass die Wirkungen von Emotionen und Aufmerksamkeit auf die Schmerzwahrnehmung weitestgehend unabhängig voneinander sind: Aufmerksamkeitsmanipulationen wirken sich nur auf die sensorische Schmerzkomponente aus und Affektmanipulationen modulieren hauptsächlich die affektive Schmerzkomponente. Der affektiven und der aufmerksamkeitsbasierten Schmerzmodulation scheinen unterschiedliche neuronale Mechanismen zugrunde zu liegen: Die LORETA-Analyse erbrachte verschiedene neuronale Generatoren für die N150 und die P260 und die Wirkung von Aufmerksamkeit und Emotion dissoziiert für diese beiden Komponenten: die Modulation der N150 reflektiert eine affektive Schmerzmodulation und die Modulation der P260 reflektiert Aufmerksamkeitsprozesse.

BD is a severe and highly prevalent psychiatric illness characterized by oscillating mood episodes, where patients express either depressed mood, anhedonia, decreased activation along with concentration difficulties and sleep disturbances, or elevated mood with hyperactivity and loss of inhibitions. Between mood episodes, patients return to a relatively normal state of functioning without mood symptoms. Previous research on underlying neuronal mechanisms has led to a model of neuronal dysfunction in BD which states that BD arises from disruption in early development within brain networks that modulate emotional behavior. These abnormalities in the structure and function of key emotional control networks then lead to decreased connectivity among ventral prefrontal networks and limbic brain regions. This in turn creates a loss of emotional homeostasis, putting bipolar patients at risk for developing extreme mood states and switching among mood states. Two core components for BD have been identified, a hyperactive emotion processing system and a hypoactive cognitive functions system. It is controversial whether these deficits are still detectable in euthymia, so it is unclear if hyper- and hypoactivations represent state or trait-like characteristics. The aim of this study was to research both core components of BD with a paradigm eliciting differential activations in both cognitive and emotion processing networks. For this, an emotional word working memory paradigm was constructed to test for differences between manic, depressive, and remitted patients as well as a healthy control group. Differences were assessed in behavior, brain activation (as a correlate for the hypoactive cognitive functions system), measured with near-infrared spectroscopy (fNIRS), and electrophysiological changes in the late positive potential (as a correlate for the hyperactive emotion processing system), an event-related potential (ERP) measured with electroencephalography. 47 patients in the acutely ill phase and 45 healthy controls were measured. Of the 47 patients, 18 returned to the clinic for a second testing while in remission for at least 3 months. Acutely ill patients were classified into 4 groups according to their disorder status: a mildly depressed group, a depressed group, a manic group, and a mixed group along DSM-IV criteria. Analyses were calculated for 3 load conditions (1-back, 2-back and 3-back) and 3 valence conditions (negative, neutral, positive) for behavioral measures reaction time and omission errors, for brain activation and event related potential changes.
Results indicate that ill patients differed from controls in their behavioral performance, but the difference in performance was modulated by the mood state they were in. Depressed patients showed the most severe differences in all behavioral measures, while manic and mixed patients differed from controls only upon different valence conditions. Brain activation changes were most pronounced in mildly depressed and manic patients, depressed patients and mixed patients did not differ as much from controls. ERP changes showed a significant difference only between mixed patients and controls, where mixed patients had an overall much higher ERP amplitude. When remitted patients were compared to controls, no differences in behavior, brain activation or ERP amplitude could be found. However, the same was true for differences in patients between acutely ill and remitted state. When looking at the overall data, the following conclusion can be drawn: assuming that the brain activation seen in the prefrontal cortex is part of the dorsal cognitive system, then this is the predominantly disturbed system in depressed patients who show only small changes in the ERP. In contrast, the predominantly disturbed system in manic and mixed patients is the ventral emotion processing system, which can be seen in a hyper-activation of ERP related neural correlates in mixed and hypo-activated neural correlates of the LPP in manic patients. When patients are remitted, the cognitive system regains temporary stability, and can be compared to that of healthy controls, while the emotion processing system remains dysfunctional and underlies still detectable performance deficits.
