Die Zahnwerkstoffe HEMA (Hydroxyethylmethacrylat) und TEGDMA (Triethylenglycol-dimethacrylat) gehören zu den so genannten Restmonomeren. Sie liegen nach der Polymerisation noch ungebunden vor und werden anschließend freigesetzt. Sie gelangen in den Organismus über die Pulpa, die Gingiva oder über den Speichel und können biologisch wirksam werden. Bisherige Studien zeigen dosisabhängige mutagene Effekte in tierischen und menschlichen Zellen. HEMA und TEGDMA führen zu DNA-Strangbrüchen, Mikrokernbildung, Apoptosen und nehmen Einfluss auf den Zellzyklus (G1- und G2-Verzögerung). Ebenso wurden ein allergenes Potential und eine toxische Wirkung auf die Niere beschrieben. In dieser Arbeit wurden genotoxische Effekte von HEMA und TEGDMA in humanen Lymphozyten in Konzentrationsbereichen überprüft, wie sie auch im Körper auftreten können. Hierfür wurden die Lymphozyten 24 Stunden mit 10 µM, 100 µM und 1 mM HEMA und mit 1 µM, 10 µM und 100 µM TEGDMA behandelt. Mit dem Comet Assay werden DNA-Einzel- und Doppelstrangbrüche sowie die Reparatur zuvor induzierter DNA-Schäden erfasst. Durch die Modifikation des Comet Assay mit dem Fpg-Protein werden zusätzlich oxidativ geschädigte Basen mit hoher Sensitivität nachgewiesen. Der Mikrokerntest weist manifeste DNA-Schäden auf DNA-Ebene in Form von Mikrokernen nach. Daneben lassen sich auch andere zelluläre Reaktionen wie Mitosen und Apoptosen sowie die Proliferationsrate der Zellen bestimmen. Der Chromosomen-aberrationstest dient zum Nachweis von Veränderungen in der Struktur und/oder in der Anzahl von Chromosomen eines Genoms. Mit dem Schwesterchromatidaustauschtest werden ebenfalls Chromosomenmutationen nachgewiesen. Durchflusszytometrische Methoden werden zum Nachweis von Apoptosen und zur Zellzyklusanalyse eingesetzt. Im herkömmlichen Comet Assay zeigen HEMA und TEGDMA keine signifikante Wirkung auf die DNA (OTM < 2). Es kann aber gezeigt werden, dass die Behandlung mit Fpg zu einer Verdoppelung des OTM führt. Bei 1 mM HEMA und 100 µM TEGDMA wird dadurch das OTM auf > 2 angehoben. HEMA und TEGDMA wirken sich nicht auf die Mikrokernbildung aus, jedoch wird durch den Mikrokerntest ab 1 mM HEMA und 100 µM TEGDMA eine Einflussnahme auf die Proliferation gezeigt. Die Rate früher (< 10%) und später Apoptosen Apoptosen (< 4 %) bleibt im Durchschnitt weitgehend konstant. Eine Ausnahme sind 1 mM HEMA, die die frühen Apoptosen auf > 10 % anheben. Eine Einflussnahme auf den Zellzyklus, in Form einer Verzögerung, üben 1 mM HEMA in der S-Phase und 100 µM TEGDMA in der G1-Phase aus. In den Chromosomentests werden einerseits ein dosisabhängiger Anstieg der Aberrationen und andererseits vermehrte Chromatidaustausche beobachtet. In dieser Arbeit wird die Verbindung von HEMA und TEGDMA zu oxidativen Stress im Comet Assay mit Fpg gezeigt. Da die tatsächlich in vivo erreichbaren Konzentrationen unter 100 µM liegen, ist zu schließen, dass HEMA und TEGDMA in diesem niedrigen Konzentrationsbereich keine nachteiligen Effekte ausüben, denn nur die hohen Konzentrationen (1 mM HEMA, 100 µM TEGDMA) sind in der Lage eine genotoxische Wirkung zu entfalten. Jedoch kann das Auslösen von Mutationen mit dem Chromosomenaberrationstest und Schwesterchromatidaustauschtest bestätigt werden. Um das Schädigungsprofil dieser häufig eingesetzten Zahnwerkstoffe detaillierter beschreiben zu können, müssen Untersuchungen auf Chromatidebene intensiviert werden.
Die einvernehmliche Weiterbeschäftigung während des Kündigungsschutzprozesses kann ein grundsätzliches Problem des deutschen Kündigungsschutzrechts lösen: Dass dieses nur eine ex-post-Beurteilung zulässt, zwingt den gekündigten Arbeitnehmer regelmäßig dazu, seinen Arbeitsplatz zumindest zeitweise zu ver-lassen. Mit dem möglichen Nachteil, bestimmte Positionen oder Qualifikationen im Betrieb zu verlieren. Dagegen riskiert der Arbeitgeber bei Ausspruch einer Kündigung stets, in Annahmeverzug zu geraten und Lohn zahlen zu müssen, ohne im Gegenzug wenigstens die Arbeitsleistung erhalten zu haben. Damit ist die vorübergehende Weiterbeschäftigung „bis zum Abschluss des Kündigungs-schutzverfahrens“ für beide Seiten nur sinnvoll. Vorliegend wurde untersucht, wie ein solcher Prozessarbeitsvertrag geschlossen wird, welche Rechte und Pflichten er für Arbeitgeber und Arbeitnehmer begrün-det und unter welche Bedingung und Befristungen er (zwangsläufig) gestellt ist. Im Vordergrund steht dabei die Frage, ob und inwieweit das Schriftformerfor-dernis des § 14 Abs. 4 TzBfG zu beachten ist. Lehnt der Arbeitnehmer eine ihm angebotene Weiterbeschäftigung dagegen ab, so stellt sich die Frage, welche Auswirkungen dies auf seinen Verzugslohnan-spruch hat. Die Rechtsprechung des BAG, wonach der bei Ausspruch einer un-wirksamen Kündigung begründete Annahmeverzug des Arbeitgebers zwar nicht ende, der Arbeitnehmer sich aber über § 615 S. 2 BGB böswillig unterlassenen Zwischenverdienst anrechnen lassen müsse, wird hier einer kritischen Prüfung unterzogen.
In diesem Artikel wird ein betreutes Selbstlernprojekt für Sprachanfänger in Arabisch und Russisch beschrieben. Ziel war der eigenverantwortliche Spracherwerb mit Hilfe von Lernsoftware in Verbindung mit persönlicher tutorieller Betreuung von Muttersprachlern. Ein besonderes Augenmerk lag dabei auf der Bewusstmachung des Lernprozesses und der Aufrechterhaltung der Lernmotivation.
This thesis is devoted to Bernoulli Stochastics, which was initiated by Jakob Bernoulli more than 300 years ago by his master piece 'Ars conjectandi', which can be translated as 'Science of Prediction'. Thus, Jakob Bernoulli's Stochastics focus on prediction in contrast to the later emerging disciplines probability theory, statistics and mathematical statistics. Only recently Jakob Bernoulli's focus was taken up von Collani, who developed a unified theory of uncertainty aiming at making reliable and accurate predictions. In this thesis, teaching material as well as a virtual classroom are developed for fostering ideas and techniques initiated by Jakob Bernoulli and elaborated by Elart von Collani. The thesis is part of an extensively construed project called 'Stochastikon' aiming at introducing Bernoulli Stochastics as a unified science of prediction and measurement under uncertainty. This ambitious aim shall be reached by the development of an internet-based comprehensive system offering the science of Bernoulli Stochastics on any level of application. So far it is planned that the 'Stochastikon' system (http://www.stochastikon.com/) will consist of five subsystems. Two of them are developed and introduced in this thesis. The first one is the e-learning programme 'Stochastikon Magister' and the second one 'Stochastikon Graphics' that provides the entire Stochastikon system with graphical illustrations. E-learning is the outcome of merging education and internet techniques. E-learning is characterized by the facts that teaching and learning are independent of place and time and of the availability of specially trained teachers. Knowledge offering as well as knowledge transferring are realized by using modern information technologies. Nowadays more and more e-learning environments are based on the internet as the primary tool for communication and presentation. E-learning presentation tools are for instance text-files, pictures, graphics, audio and videos, which can be networked with each other. There could be no limit as to the access to teaching contents. Moreover, the students can adapt the speed of learning to their individual abilities. E-learning is particularly appropriate for newly arising scientific and technical disciplines, which generally cannot be presented by traditional learning methods sufficiently well, because neither trained teachers nor textbooks are available. The first part of this dissertation introduces the state of the art of e-learning in statistics, since statistics and Bernoulli Stochastics are both based on probability theory and exhibit many similar features. Since Stochastikon Magister is the first e-learning programme for Bernoulli Stochastics, the educational statistics systems is selected for the purpose of comparison and evaluation. This makes sense as both disciplines are an attempt to handle uncertainty and use methods that often can be directly compared. The second part of this dissertation is devoted to Bernoulli Stochastics. This part aims at outlining the content of two courses, which have been developed for the anticipated e-learning programme Stochastikon Magister in order to show the difficulties in teaching, understanding and applying Bernoulli Stochastics. The third part discusses the realization of the e-learning programme Stochastikon Magister, its design and implementation, which aims at offering a systematic learning of principles and techniques developed in Bernoulli Stochastics. The resulting e-learning programme differs from the commonly developed e-learning programmes as it is an attempt to provide a virtual classroom that simulates all the functions of real classroom teaching. This is in general not necessary, since most of the e-learning programmes aim at supporting existing classroom teaching. The forth part presents two empirical evaluations of Stochastikon Magister. The evaluations are performed by means of comparisons between traditional classroom learning in statistics and e-learning of Bernoulli Stochastics. The aim is to assess the usability and learnability of Stochastikon Magister. Finally, the fifth part of this dissertation is added as an appendix. It refers to Stochastikon Graphics, the fifth component of the entire Stochastikon system. Stochastikon Graphics provides the other components with graphical representations of concepts, procedures and results obtained or used in the framework of Bernoulli Stochastics. The primary aim of this thesis is the development of an appropriate software for the anticipated e-learning environment meant for Bernoulli Stochastics, while the preparation of the necessary teaching material constitutes only a secondary aim used for demonstrating the functionality of the e-learning platform and the scientific novelty of Bernoulli Stochastics. To this end, a first version of two teaching courses are developed, implemented and offered on-line in order to collect practical experiences. The two courses, which were developed as part of this projects are submitted as a supplement to this dissertation. For the time being the first experience with the e-learning programme Stochastikon Magister has been made. Students of different faculties of the University of Würzburg, as well as researchers and engineers, who are involved in the Stochastikon project have obtained access to Stochastikon Magister via internet. They have registered for Stochastikon Magister and participated in the course programme. This thesis reports on two assessments of these first experiences and the results will lead to further improvements with respect to content and organization of Stochastikon Magister.
This work is composed of three main parts: remote control of mobile systems via Internet, ad-hoc networks of mobile robots, and remote control of mobile robots via 3G telecommunication technologies. The first part gives a detailed state of the art and a discussion of the problems to be solved in order to teleoperate mobile robots via the Internet. The focus of the application to be realized is set on a distributed tele-laboratory with remote experiments on mobile robots which can be accessed world-wide via the Internet. Therefore, analyses of the communication link are used in order to realize a robust system. The developed and implemented architecture of this distributed tele-laboratory allows for a smooth access also with a variable or low link quality. The second part covers the application of ad-hoc networks for mobile robots. The networking of mobile robots via mobile ad-hoc networks is a very promising approach to realize integrated telematic systems without relying on preexisting communication infrastructure. Relevant civilian application scenarios are for example in the area of search and rescue operations where first responders are supported by multi-robot systems. Here, mobile robots, humans, and also existing stationary sensors can be connected very fast and efficient. Therefore, this work investigates and analyses the performance of different ad-hoc routing protocols for IEEE 802.11 based wireless networks in relevant scenarios. The analysis of the different protocols allows for an optimization of the parameter settings in order to use these ad-hoc routing protocols for mobile robot teleoperation. Also guidelines for the realization of such telematics systems are given. Also traffic shaping mechanisms of application layer are presented which allow for a more efficient use of the communication link. An additional application scenario, the integration of a small size helicopter into an IP based ad-hoc network, is presented. The teleoperation of mobile robots via 3G telecommunication technologies is addressed in the third part of this work. The high availability, high mobility, and the high bandwidth provide a very interesting opportunity to realize scenarios for the teleoperation of mobile robots or industrial remote maintenance. This work analyses important parameters of the UMTS communication link and investigates also the characteristics for different data streams. These analyses are used to give guidelines which are necessary for the realization of or industrial remote maintenance or mobile robot teleoperation scenarios. All the results and guidelines for the design of telematic systems in this work were derived from analyses and experiments with real hardware.
This article is about a measurement analysis based approach to help software practitioners in managing the additional level complexities and variabilities in software product line applications. The architecture of the proposed approach i.e. ZAC is designed and implemented to perform preprocessesed source code analysis, calculate traditional and product line metrics and visualize results in two and three dimensional diagrams. Experiments using real time data sets are performed which concluded with the results that the ZAC can be very helpful for the software practitioners in understanding the overall structure and complexity of product line applications. Moreover the obtained results prove strong positive correlation between calculated traditional and product line measures.
Bacteria lose or gain genetic material and through selection, new variants become fixed in the population. Here we provide the first, genome-wide example of a single bacterial strain’s evolution in different deliberately colonized patients and the surprising insight that hosts appear to personalize their microflora. By first obtaining the complete genome sequence of the prototype asymptomatic bacteriuria strain E. coli 83972 and then resequencing its descendants after therapeutic bladder colonization of different patients, we identified 34 mutations, which affected metabolic and virulence-related genes. Further transcriptome and proteome analysis proved that these genome changes altered bacterial gene expression resulting in unique adaptation patterns in each patient. Our results provide evidence that, in addition to stochastic events, adaptive bacterial evolution is driven by individual host environments. Ongoing loss of gene function supports the hypothesis that evolution towards commensalism rather than virulence is favored during asymptomatic bladder colonization.
Die zellulären Rho GTPasen kontrollieren und regulieren zentrale elementare Zellvorgänge wie Phagozytose, Migration und epitheliale Integrität. Aufgrund ihrer zentralen Stellung, interagiert eine Vielzahl von bakteriellen Cytotoxinen und Modulinen mit den Rho GTPasen und wirken so als Pathogenitätsfaktoren. Die zur W-xxx-E Familie gehörenden Effektoren IpgB1 und IpgB2 von Shigella und Map von E. coli (Pathotypen EHEC und EPEC) werden über ein Typ 3 Sekretionssystem (T3SS) in Wirtszellen injiziert und wirken als Rac1, RhoA bzw. Cdc42 GEF Mimetikum. In der vorliegenden Arbeit wurden die Effektor Funktionen von IpgB1 IpgB2 und Map mit Hilfe des Yersinia (Ysc)-T3SS untersucht, was zur Etablierung der „Yersinia-Toolbox“ führte. Damit können heterologe Effektoren isoliert im physiologischen Kontext der Erreger-Zell-Interaktion zellbiologisch untersucht werden unter Vermeidung von simultaner Injektion redundanter oder unbekannter Effektoren. Zur Etablierung der Yersinia-Toolbox wurden zunächst die Gene für die Rho GTPasen modulierenden Shigella Effektoren IpgB1 und IpgB2 sowie der E. coli (EHEC)-Effektor Map mit unterschiedlich langen Gensequenzen der N-terminalen Bereiche des Yersinia-Effektorproteins YopE fusioniert (Hybridproteine: YopEi-X:i = 18, 53 bzw. 138 Aminosäurereste, X = IpgB1, IpgB2 bzw. Map). In der vorliegenden Arbeit wird gezeigt, dass die Hybridproteine YopE53-X und YopE138-X (X=IpgB1, IpgB2, Map) in den Kulturüberstand sezerniert bzw. in Zielzellen injiziert wurden. In einem weiteren Schritt konnte die zellbiologische Aktivität der heterologen Proteine fluoreszenzmikroskopisch durch Aktinzytoskelettumlagerungen gezeigt werden. So wurden „Membrane Ruffles“ (Rac1-Aktivierung) durch YopE138-IpgB1, Stressfasern (RhoA-Aktivierung) durch E138-IpgB2 und „Mikrospikes“ (Cdc42-Aktivierung) durch YopE138-Map nachgewiesen. Invasionstudien zeigten, dass YopEi-IpgB1 (i = 53, 138) die Yersinia-Invasion induzierte, wohingegen YopEi-IpgB2 die Invasionsrate der Stämme WA (pT3SS, pEi-IpgB2) (i=53, 138) verglichen mit dem Stamm WA (pT3SS) reduziert war. Durch Kombination verschiedener Yersinia-Toolbox-Stämme konnte im Co-Infektionsmodell mit HeLa-Zellen gezeigt werden, dass (1) die YopE138-IpgB1 vermittelte Invasion durch YopE138-IpgB2 signifikant inhibiert werden kann, was auf eine antagonistische Wirkung zwischen IpgB1 und IpgB2 schließen lässt, dass (2) YopT ebenfalls die IpgB1 vermittelte Invasionsrate reduziert (inhibitorische Wirkung auf Rac1), und dass (3) YopE als GAP für RhoG/Rac1 (bevorzugt RhoG) praktisch nicht die IpgB1-vermittelte Invasion hemmt. Durch Klonierung der YopE138-IpgB1 und YopE138-IpgB2 kodierenden Fusionsgene in zwei kompatible Plasmidvektoren konnten die Hybridproteine simultan transloziert werden und die Co-Infektionsergebnisse bestätigt werden. In der Literatur ist beschrieben, dass die Ysc-Translokationspore YopB/YopD Rho-abhängig Membranporen-bedingte Zellschädigungen verursacht (LDH-Freisetzung, PI-Kernfärbung). Mit der Yersinia-Toolbox konnte mit dem Stamm WA (pT3SS) Zytoplasmamembranschädigung / Zytotoxizität nachgewiesen werden, nicht aber mit den Stämmen WA (pE138-X) X = IpgB1, IpgB2 oder Map. Co-Infektionen jedoch zeigen, dass vermehrt LDH bei der Infektion mit WA (pT3SS) + WA (pT3SS, pE138-IpgB1) detektiert wurde, wohingegen dieser Effekt von YopE138-IpgB2 in einer Co-Infektion von WA (pT3SS) + WA (pT3SS, pE138-IpgB2) inhibiert wurde. Auch hier wurde der Antagonismus zwischen IpgB1 und IpgB2 erneut sichtbar. Diese Befunde widersprechen publizierten Daten, die eine RhoA-Aktivierung/Aktinpolymerisierung mit verstärkter Porenbildung in einen Zusammenhang bringen. Rho GTPasen sind beteiligt an der Erhaltung der polarisierten Eipthelzellschichtintegrität über Adhäsionskomplexbildung. Mittels Infektion von polarisierten MDCK-Zellschichten mit verschiedenen Yersinia-Stämmen und Messung des transepithelialen elektrischen Widerstandes/Resistenz (TER) konnte gezeigt werden, dass die Ysc-T3SS vermittelte Injektion von YopE138-IpgB1 (Rac1-Aktivierung) oder YopE138-Map (Cdc42-Aktivierung) zur Abnahme der TER und damit Schädigung der Zellschichtintegrität führt, wogegen bei YopE138-IpgB2-Injektion der TER-Wert unverändert blieb. Um bakterielle Rho GTPasen-modulierende Effektorproteine detailliert untersuchen zu können und um die Rolle von Rho GTPasen im Mausinfektionsmodell mit Yersinia enterocolitica und Salmonellen zu bestimmen, wurden Mäuse mit deletierten Genen für RhoA, Rac1 bzw. Cdc42 in Makrophagen hergestellt.
Im Rahmen dieser retrospektiven Studie wurden 23 Patienten mit Erkrankungen aus dem rheumatischen Formenkreis, überwiegend rheumatoide Arthritis und 22 Kontrollpersonen mit degenerativ bedingter Omarthrose, welchen zwischen 1993 und 2005 eine Schulterendoprothese unterschiedlichen Typs implantiert worden war, durchschnittlich 28 Monate postoperativ klinisch und radiologisch nachuntersucht. Das Alter zum Zeitpunkt der Operation betrug im Mittel 65 Jahre. Die Ergebnisse wurden anhand des Constant Score erhoben. Durch die Operation verbesserten die 23 Patienten mit Erkrankungen aus dem rheumatischen Formenkreis ihren Constant Score von durchschnittlich 21,7% auf 79,9%. Die Kontrollgruppe wies sowohl präoperativ (28,7%) als auch postoperativ (96,3%) eine signifikant bessere Schulterfunktion auf. Innerhalb beider Kollektive waren ca. 95% der Patienten mit dem Operationsergebnis sehr zufrieden bzw. zufrieden.
