Untersuchung zur NO/cGMP-Signaltransduktion in der glatten Muskulatur von NO-GC-defizienten Mäusen

Die Stickstoffmonoxid (NO)/cGMP-Signaltransduktion besitzt eine entscheidende Rolle bei der Tonusregulation der glatten Muskulatur. Dabei ist NO neben seiner herausragenden Bedeutung für das vaskuläre System einer der wichtigsten inhibitorischen Neurotransmitter im Gastrointestinaltrakt. Die Wirkung von NO beruht hauptsächlich auf der Aktivierung der NO-sensitiven Guanylyl-Cyclase (NO-GC), die aus zwei Untereinheiten aufgebaut ist (α und ß). Die Deletion der ß1-Untereinheit in Mäusen resultiert in einem vollständigen NO-GC-Knockout (GCKO). Im Gastrointestinaltrakt ist die Expression von NO-GC in glatten Muskelzellen (SMC), interstitiellen Zellen von Cajal (ICC) und Fibroblasten-ähnlichen Zellen (FLC) nachgewiesen. In dieser Arbeit wurde die Bedeutung des NO/cGMP-Signalweges für die Regulation von Kontraktion und Relaxation innerhalb dieser drei Zelltypen anhand von zellspezifischen GCKO-Tieren untersucht. SMC- und ICC-spezifische GCKO-Tiere waren bereits vorhanden. FLC-spezifische GCKO-Tiere wurden generiert und mit den vorhandenen ICC- und SMC-GCKO-Linien gekreuzt, um Doppel- und Tripel-Knockout-Tiere zu erhalten. FLC-GCKO-Tiere zeigen eine NO-induzierte Relaxation glattmuskulären Gewebes, die der von WT-Tieren gleicht. Auch Gewebe von FLC/ICC- und FLC/SM-GCKO-Tieren kann durch NO relaxiert werden. Erst die Deletion der NO-GC in allen drei Zelltypen (Tripel-GCKO) führt zu einer Unterbrechung der NO-Relaxation, wie sie aus GCKO-Tieren bekannt ist. Überraschenderweise zeigt sich bei FLC-GCKO-Tieren eine beschleunigte Darmpassagezeit. Die Ergebnisse dieser Arbeit lassen darauf schließen, dass die NO-GC in allen drei Zelltypen des Gastrointestinaltrakts an der nitrergen Signaltransduktion beteiligt ist, wenn auch auf unterschiedliche Weise. Es besteht demnach eine Interaktion zwischen den verschiedenen Zelltypen, die durch weiterführende Versuche mit den vorhandenen Doppel-Knockout-Tieren sowie der Tripel-GCKO-Linie nähergehend untersucht werden muss. Der zweite Teil der Arbeit beschäftigte sich mit der Rolle der NO-GC im unteren Harntrakt. Dort liegt die NO-GC in verschieden Zelltypen vor. In Urethra-Gewebe wird die NO-GC ausschließlich in SMC exprimiert, während sie in der Harnblase einzig in interstitiellen Zellen, nicht aber in SMC, befindet. Funktionell hat dies zur Folge, dass die NO-induzierte Urethra-Relaxation ausschließlich von glatten Muskelzellen vermittelt wird. Die Harnblasenmuskulatur hingegen zeigt keine Relaxation auf NO-Gabe hin. Die Identifizierung der NO-GC-exprimierenden interstitiellen Zellen sowie ihre Funktion sind bislang ungeklärt. In einem dritten Projekt wurden Untersuchungen zur Effektivität der NO-GC-Inhibitoren ODQ und NS2028 durchgeführt. Die Ergebnisse zeigen, dass bei einem Einsatz der Inhibitoren nicht von einer vollständigen Hemmung der NO-GC ausgegangen werden sollte. Drei Faktoren beeinflussen nachhaltig die Inhibitor-Effektivität: (1) die Klasse des NO-Donors, (2) die Inkubationszeit mit dem Inhibitor und dem NO-Donor sowie (3) die Stärke der Vorkontraktion bei Versuchen mit Glattmuskelgewebe. Die Wahl dieser Parameter bestimmt, in welchem Ausmaß ODQ und NS2028 die NO-stimulierte NO GC inhibieren können. Aus diesem Projektteil resultiert, dass man den Einsatz dieser Inhibitoren nicht, wie vielfach in der Literatur vorzufinden, als Beweis für cGMP unabhängige Effekte nutzen sollte.
In mammals, KSR1 functions as an essential scaffold that coordinates the assembly of RAF/MEK/ERK complexes and regulates intracellular signal transduction upon extracellular stimulation. Aberrant activation of the equivalent MAPK signaling pathway has been implicated in multiple human cancers and some developmental disorders. The mechanism of KSR1 regulation is highly complex and involves several phosphorylation/dephosphorylation steps. In the present study, a number of novel in vivo phosphorylation sites were detected in mKSR1 by use of mass spectrometry analysis. Among others, Tyr728 was identified as a unique regulatory residue phosphorylated by LCK, a Src kinase family member. To understand how phosphorylation of Tyr728 may regulate the function of KSR1 in signal transduction and cellular processes, structural modeling and biochemical studies were integrated in this work.
Computational modeling of the mKSR1(KD) protein structure revealed strong hydrogen bonding between phospho-Tyr728 and the residues surrounding Arg649. Remarkably, this pattern was altered when Tyr728 was non-phosphorylated or substituted. As confirmed by biochemical analysis, Arg649 may serve as a major anchor point for phospho-Tyr728 in order to stabilize internal structures of KSR1. In line with the protein modeling results, mutational studies revealed that substitution of Tyr728 by phenylalanine leads to a less compact interaction between KSR1 and MEK, a facilitated KSR1/B-RAF binding and an increased phosphorylation of MEK in complex with KSR1. From these findings it can be concluded that phospho-Tyr728 is involved in tightening the KSR1/MEK interaction interface and in regulating the phosphorylation of KSR1-bound MEK by either RAF or KSR1 kinases.
Beside the Tyr728, Ser722 was identified as a novel regulatory phosphorylation site. Amino acid exchanges at the relevant position demonstrated that Ser722 regulates KSR1-bound MEK phosphorylation without affecting KSR1/MEK binding per se. Due to its localization, Ser722 might consequently control the catalytic activity of KSR1 by interfering with the access of substrate (possibly MEK) to the active site of KSR1 kinase. Together with Ser722, phosphorylated Tyr728 may further positively affect the kinase activity of KSR1 as a consequence of its vicinity to the activation and catalytic loop in the KSR1(KD). As revealed by structural modeling, phospho-Tyr728 builds a hydrogen bond with the highly conserved Lys685. Consequently, phospho-Tyr728 has a stabilizing effect on internal structures involved in the catalytic reaction and possibly enhances the phosphate transfer within the catalytic cleft in KSR1. Considering these facts, it seems very likely that the LCK-dependent phosphorylation of Tyr728 plays a crucial role in the regulation of KSR1 catalytic activity.
Results of fractionation and morphology analyses revealed that KSR1 recruits LCK to cytoskeleton for its phosphorylation at Tyr728 suggesting that this residue may regulate cytoskeleton dynamics and, consequently, cell motility. Beside that, phosphorylation of Tyr728 is involved in the regulation of cell proliferation, as shown by a significantly reduced population doubling time of KSR1-Y728F cells compared to cells expressing wild type KSR1.
Taken together, tyrosine phosphorylation in KSR1 uncovers a new link between Src family kinases and MAPK signaling. Tyr728, the novel regulatory phosphorylation site in murine KSR1, may coordinate the transition between the scaffolding and the catalytic function of KSR1 serving as a control point used to fine-tune cellular responses.
Die vorliegende Arbeit behandelt TRAIL-induzierte Apoptose und Nekroptose in verschiedenen Zelllinien. Im Speziellen wurden die verschiedenen Funktionen des TNF receptor-associated factor 2 (TRAF2) untersucht. Hierzu wurde ein transienter Knockdown etabliert und dessen Wirkung auf die Suszeptibilität der Zellen gegenüber dem Zytokin TRAIL untersucht. Es konnte gezeigt werden, dass ein Knockdown von TRAF2 nicht nur zur Sensitivierung für Apoptose führt, sondern auch in Nekroptose-kompetenten Zellen zu einer Verstärkung der durch Caspaseinhibition mittels zVAD-fmk nach TRAIL-Stimulation induzierten Nekroptose führt. Mittels des Zytokins Fc-TWEAK wurde Fn14-vermittelt TRAF2 aus dem Zytosol in ein Triton X100-unlösliches Kompartiment rekrutiert und dadurch physiologisch depletiert. Dies führte zwar kaum zu gesteigerter TRAIL-abhängiger Apoptose, sensitivierte jedoch analog zum TRAF2-Knockdown RIP3-exprimierende Zellen für Nekroptose. Durch Vergleich RIP3-negativer (HeLa-Leervektor) mit RIP3-exprimierenden Zellen (HeLa RIP3, HT29, HaCaT) konnte die Essentialität von RIP3 für die Nekroptose herausgestellt werden und Einsatz des RIP1-Kinase-Inhibitors Necrostatin-1 sowie des MLKL-Inhibitors Necrosulfonamide belegte die Beteiligung der Nekroptosomkomponenten RIP1 und MLKL. Antagonismus putativen autokrinen TNFs bewies, dass es sich bei dem durch Fc-TWEAK verstärkten Zelltod um einen direkten TRAIL-Effekt handelte und Inhibition kanonischen NFkBs durch IKK2-Inhibitor TPCA-1, dass die TRAF2-Knockdown-vermittelte Sensitivierung gegenüber TRAIL nicht auf verändertes NFkB-Signalling zurückzuführen ist. Einsatz des SMAC-Mimetikums BV6 rekapitulierte zudem stark das im TRAF2-Knockdown Gesehene und unterstrich die Bedeutung der cIAPs. Immunpräzipitation von Caspase 8 unter nekroptotischen Bedingungen zeigte bei TRAF2-Knockdown eine Depletion von TRAF2 und cIAP1/2 sowie RIP1 und RIP3 aus dem Komplex mit Caspase 8. Insgesamt wird deutlich, dass TRAF2 einerseits antiapoptotisch wirkt als K48-Ubiquitinligase, die die Halbwertszeit aktiver Caspase 8-Komplexe determiniert und andererseits eine antinekroptotische Funktion hat, da es durch Rekrutierung von cIAP1/2 an RIP1 die TRAIL-induzierte Nekroptose verhindert, wenn die Caspasen inhibiert sind.
Das Multiple Myelom (MM) ist eine unheilbare Erkrankung, die aus einer klonalen Proliferation maligner Plasmazellen im Knochenmark hervorgeht. Dabei liegt ein komplexes Signalnetzwerk vor, das zum Überleben und Wachstum der MM-Zellen führt. Das MM ist durch eine enorme genetische und phänotypische Heterogenität gekennzeichnet. Die konstitutive Aktivierung des PI3K/Akt-Signalwegs spielt bei ungefähr der Hälfte der Patienten mit MM eine wichtige Rolle für das Überleben der MM-Zellen und ist daher ein potentieller therapeutischer Ansatzpunkt. Isoform-spezifische Untersuchungen der katalytischen Untereinheiten der Klasse I-PI3K (p110α, p110β, p110γ, p110δ) sollten zur Erkenntnis führen, welche dieser Isoformen für das MM Zellüberleben wichtig sind, um spezifischere Behandlungen mit möglichst geringen Nebenwirkungen zu erlauben. Dafür wurden zunächst Isoform-spezifische Knockdown-Experimente mit MM Zelllinien durchgeführt und sowohl deren Überleben als auch die Aktivierung der nachgeschalteten Komponenten im PI3K Signalweg untersucht. Zur Verifizierung der Ergebnisse wurden sowohl MM Zelllinien als auch Primärzellen mit Isoform-spezifischen PI3K-Inhibitoren behandelt (BYL 719 für p110α, TGX 221 für p110β, CAY10505 für p110γ und CAL 101 für p110δ) und in gleicher Weise untersucht. In beiden Versuchsansätzen stellte sich die katalytische Untereinheit p110α als wichtigste Isoform für das Überleben von MM Zellen mit konstitutiv phosphoryliertem Akt Signal heraus. Weder der Knockdown noch die pharmakologische Inhibition der anderen drei Isoformen (p110β, p110γ, p110δ) führten in MM-Zelllinien zur Beeinträchtigung des Zellüberlebens. Auch reagierten die Primärzellen von MM Patienten größtenteils nicht mit Apoptose auf eine Behandlung mit TGX 221, CAY10505 oder CAL 101. Aufbauend auf der postulierten Bedeutung von p110α, wurde der dafür spezifische Inhibitor BYL 719 mit bereits klinisch etablierten Therapeutika in Kombination verwendet, woraus eine im Vergleich zur Einzelbehandlung verstärkte Apoptose resultierte. Insgesamt deuten diese Daten darauf hin, dass PI3K/p110α eine therapeutisch nutzbare Zielstruktur zur Behandlung des Multiplen Myeloms darstellt. Daher scheinen weitergehende prä-klinische Studien mit p110α Inhibitoren erfolgversprechend.
A subtly regulated and controlled course of cellular processes is essential for the healthy functioning not only of single cells, but also of organs being constituted thereof. In return, this entails the proper functioning of the whole organism. This implies a complex intra- and inter-cellular communication and signal processing that require equally multi-faceted methods to describe and investigate the underlying processes. Parasitic helminths share a large degree of common genetic heritage with their various hosts. This includes cell-cell-communication mechanisms mediated by small peptide cytokines and lipophilic/steroid hormones. These cytokines are candidate molecules for host-parasite cross-communication in helminth diseases. In this work the function of two evolutionary conserved signaling pathways in the model cestode Echinococcus multilocularis has been studied. First, signaling mechanisms mediated through fibroblast growth factors (FGF) and their cognate receptors (FGFR) which influence a multitude of biological functions, like homeostasis and differentiation, were studied. I herein investigated the role of EmFR which is the only FGFR homolog in E. multilocularis. Functional analyses using the Xenopus oocyte expression system clearly indicate that EmFR can sense both acidic and basic FGF of human origin, resulting in an activation of the EmFR tyrosine kinase domain. In vitro experiments demonstrate that mammalian FGF significantly stimulates proliferation and development of E. multilocularis metacestode vesicles and primary cells. Furthermore, DNA synthesis and the parasite’s Erk-like MAPK cascade module was stimulated in the presence of exogenously added mammalian FGF. By using the FGFR inhibitor BIBF1120 the activity of EmFR in the Xenopus oocyte system was effectively blocked. Addition of BIBF1120 to in vitro cultivated Echinococcus larval material led to detrimental effects concerning the generation of metacestode vesicles from parasite stem cells, the proliferation and survival of metacestode vesicles, and the dedifferentiation of protoscoleces towards the metacestode. In conclusion, these data demonstrate the presence of a functional EmFR-mediated signaling pathway in E. multilocularis that is able to interact with host-derived cytokines and that plays an important role in larval parasite development. Secondly, the role of nuclear hormone receptor (NHR) signaling was addressed. Lipophilic and steroid hormone signaling contributes to the regulation of metazoan development. By means of in silico analyses I demonstrate that E. multilocularis expresses a set of 17 NHRs that broadly overlaps with that of the related flatworms Schistosoma mansoni and S. japonicum, but also contains several NHR encoding genes that are unique to this parasite. One of these, EmNHR1, is homolog to the DAF-12/HR-96 subfamily of NHRs which regulate cholesterol homeostasis in metazoans. Modified yeast-two hybrid analyses revealed that host serum contains a ligand which induces homodimerization of the EmNHR1 ligand-binding domain. Also, a HNF4-like homolog, EmHNF4, was characterized. Human HNF4 plays an important role in liver development. RT-PCR experiments showed that both isoforms of the EmHNF4 encoding gene are expressed stage-dependently suggesting distinct functions of the two isoforms in the parasite. Moreover, specific regulatory mechanisms on the convergence of NHR signaling and TGF-β/BMP signaling pathways in E. multilocularis have been identified. On the one hand, EmNHR1 directly interacted with the EmSmadC and on the other hand EmHNF4b interacted with EmSmadD, EmSmadE which are all downstream signaling components of the TGF-β/BMP signaling pathway. This suggests cross-communication in order to regulate target gene expression. With these results, further studies on the role of NHR signaling in the cestode will be facilitated. Also, the first serum-free in vitro cultivation system for E. multilocularis was established using PanserinTM401 as medium. Serum-free co-cultivation with RH-feeder cells and an axenic cultivation method have been established. With the help of this serum-free cultivation system investigations on the role of specific peptide hormones, like FGFs, or lipophilic/steroid hormones, like cholesterol, for the development of helminths will be much easier.
Durch die im Labor der AG Topp standardisierte Verfahren konnten im Rahmen dieser Arbeit stabile T Zell Linien aus primär humanen CMV spezifischen T Zellen und CMV spezifischen T Zellen mit chimärischen Rezeptoren hergestellt werden. Die Anzahl der chimärischen Rezeptoren auf den unterschiedlichen T Zellinien ist nicht signifikant unterschiedlich und beträgt ca. 17000 Rezeptoren pro Zelle. Bei der Überprüfung des Lyseverhaltens CMV spezifischer T Zellen gegenüber CMV spezifischer T Zellen transduziert mit einem chimärischen Rezeptor zeigt sich das gleiche Lyseverhalten. Um die zeitliche Dynamik dieses Verhaltens besser darzustellen, wurde ein FACS basierter Lysierungsversuch selbstständig entwickelt und etabliert. Somit konnte herausgestellt werden, dass das Lyseverhalten der T Zelllinien sowohl der CMV spezifischen T Zellen als auch das der Erst- und Zweitgeneration- T Zelllinien CAR CD19z und CAR CD19/28z vergleichbar war. Alle genannten Zelllinien lysieren ihre Targetzellen sowohl in der Quantität als auch in der zeitlichen Dynamik vergleichbar. Dies ist die einzige Gemeinsamkeit der verglichenen T Zell Aktivierungssysteme. In allen anderen funktionellen Versuchen ist die Aktivierung der T Zellen über einen chimärischen Rezeptor dem der physiologischen Aktivierung über den T Zell Rezeptor unterlegen. Der Proliferationsversuch mit CFSE zeigt eindeutig, dass nur die T Zellen, die über den TCR aktiviert werden, das Potential haben, sich zu teilen. Die T Zellen, die über den CAR sowohl der ersten als auch der zweiten Generation aktiviert werden, teilen sich nicht. Der Defekt der CAR T Zellen zeigt sich auch in der Hochregulation von CD25, einem Aktivierungsmarker für T Zellen. Die T Zellen, die durch die chimärischen Rezeptoren sowohl der ersten als auch der zweiten Generation aktiviert werden, regulieren CD25 nicht so stark nach oben wie die T Zellen, die physiologisch durch den TCR aktiviert werden. Diese Minderaktivierung spiegelt sich auch in der Zytokinproduktion wieder. Auch hier zeigt sich die unvollständige Aktivierung der CAR T Zellen. In der intrazellulären Zytokinmessung sieht man, dass bei CAR T Zellen der ersten und zweiten Generation zum einen die Einzelzellen weniger IFNy produzieren, zum anderen die Gesamtzahl der Zellen, die Zytokine produzieren, weniger ist als im Vergleich zur TCR Aktivierung. Dieses Phänomen der defizienten Aktivierung und der funktionellen Defekte von CAR T Zellen ist bekannt. Dies konnte noch einmal unter standardisierten und kontrollierten Bedingungen bestätigt werden. Durch die relativ neue Methode der durchflusszytometrischen Bead Technologie konnte dies nun zum ersten Mal realisiert werden. Es zeigte sich, dass ein signifikanter Unterschied zwischen den verschiedenen Aktivierungsmodi der T Zellen besteht. Eine physiologische Aktivierung von T Zellen führt zu einer höheren Maximum-Phosphorylierung als eine Aktivierung über den Erstgeneration CAR. Der signifikante Unterschied zeigt sich bei den MAP Kinasen ERK, JNK und p38. Somit ist dies ein Hinweis, dass die Signaltransduktion auf breiter Ebene aufgrund des alleinigen Vorhandenseins der Zeta Kette bei den CAR CD19z T Zellen, defizient ist und nicht ausreicht für eine volle Aktivierung, die eine volle Funktionalität der T Zelle nach sich ziehen würde. Diese unzureichende Aktivierung soll durch die kostimulatorische Komponente CD28 beim Zweitgenerationrezeptor CAR CD19/28z aufgehoben werden. In der Proliferation, Zytokineproduktion und den Aktivierungsmarkern zeigt sich keine Verbesserung im Vergleich zum Erstgenerationrezeptor bei CMV spezifischen T Zellen. Dieses Ergebnis wird bestätigt und korreliert damit, dass es keinen Unterschied in der maximalen Phosphorylierung von ERK, JNK und p38 zwischen Erstgeneration CAR CD19z und Zweitgeneration CAR CD19/28z gibt. Somit ist dieses System geeignet, um aus dem Phosphorylierungsstatus von MAP Kinasen von CAR T Zellen auf die Funktionalität dieser T Zellen zu schließen.
Bone Morphogenetic Proteins (BMPs) are key regulators for a lot of diverse cellular processes. During embryonic development these proteins act as morphogens and play a crucial role particularly in organogenesis. BMPs have a direct impact on distinct cellular fates by means of concentration-gradients in the developing embryos. Using the diverse signaling input information within the embryo due to the gradient, the cells transduce the varying extracellular information into distinct gene expression profiles and cell fate decisions. Furthermore, BMP proteins bear important functions in adult organisms like tissue homeostasis or regeneration. In contrast to TGF-ß signaling, currently only little is known about how cells decode and quantify incoming BMP signals. There is poor knowledge about the quantitative relationships between signal input, transducing molecules, their states and location, and finally their ability to incorporate graded systemic inputs and produce qualitative responses. A key requirement for efficient pathway modulation is the complete comprehension of this signaling network on a quantitative level as the BMP signaling pathway, just like many other signaling pathways, is a major target for medicative interference. I therefore at first studied the subcellular distribution of Smad1, which is the main signal transducing protein of the BMP signaling pathway, in a quantitative manner and in response to various types and levels of stimuli in murine c2c12 cells. Results indicate that the subcellular localization of Smad1 is not dependent on the initial BMP input. Surprisingly, only the phospho-Smad1 level is proportionally associated to ligand concentration. Furthermore, the activated transducer proteins were entirely located in the nucleus. Besides the subcellular localization of Smad1, I have analyzed the gene expression profile induced by BMP signaling. Therefore, I examined two endogenous immediate early BMP targets as well as the expression of the stably transgenic Gaussia Luciferase. Interestingly, the results of these independent experimental setups and read-outs suggest oscillating target gene expression. The amplitudes of the oscillations showed a precise concentration-dependence for continuous and transient stimulation. Additionally, even short-time stimulation of 15’ activates oscillating gene-expression pulses that are detectable for at least 30h post-stimulation. Only treatment with a BMP type I receptor kinase inhibitor leads to the complete abolishment of the target gene expression. This indicated that target gene expression oscillations depend directly on BMP type I receptor kinase activity.
Multiple sclerosis is an autoimmune disease of the central nervous system characterized by inflammatory, demyelinating lesions and neuronal death. Formerly regarded as a variant of MS, neuromyelitis optica (NMO)/Devic’s disease is now recognized as a distinct neurological disorder exhibiting characteristic inflammatory and demyelinated foci in the optic nerves and the spinal cord sparing the brain. With the introduction of the double-transgenic “Devic mouse” model featuring spontaneous, adjuvant-free incidence of autoimmune neuroinflammation due to the interaction of transgenic MOG-specific T and B cells, a promising tool was found for the analysis of factors triggering or preventing autoimmunity. The co-inhibitory molecule B7-H1 has been proposed to contribute to the maintenance of peripheral tolerance and to confine autoimmune inflammatory damage via the PD-1/B7-H1 pathway. Compared to Devic B7-H1+/+ mice, Devic B7-H1-/- mice developed clinical symptoms with a remarkably higher incidence rate and faster kinetics emphasized by deteriorated disease courses and a nearly quadrupled mortality rate. Remarkably enlarged immune-cell accumulation in the CNS of Devic B7-H1-/- mice, in particular of activated MOG-specific CD4+ T cells, correlated with the more severe clinical features. Our studies showed that the CNS not only was the major site of myelin-specific CD4+ T-cell activation but also that B7-H1 expression within the target organ significantly influenced T-cell activation and differentiation levels. Analysis at disease maximum revealed augmented accumulation of MOG-specific CD4+ T cells in the peripheral lymphoid organs of Devic B7-H1-/- mice partly due to increased T-cell proliferation rates. Transgenic MOG-specific B cells of Devic B7-H1-/- mice activated MOG-specific CD4+ T cells more efficiently than B cells of Devic B7-H1+/+ mice. This observation indicated a relevant immune-modulating role of B7-H1 on APCs (antigen-presenting cells) in this mouse model. We also assumed altered thymic selection processes to be involved in increased peripheral CD4+ T-cell numbers of Devic B7-H1-/- mice as we found more thymocytes expressing the transgenic MOG-specific T-cell receptor (TCR). Moreover, preliminary in vitro experiments hinted on an enhanced survival of TCRMOG-transgenic CD4+ T cells of Devic B7-H1-/- mice; a mechanism that might as well have led to higher peripheral T-cell accumulation. Elevated levels of MOG-specific CD4+ T cells in the periphery of Devic B7-H1-/- mice could have entailed the higher quantities in the CNS. However, mechanisms such as CNS-specific proliferation and/or apoptosis/survival could also have contributed. This should be addressed in future investigations. Judging from in vitro migration assays and adoptive transfer experiments on RAG-1-/- recipient mice, migratory behavior of MOG-specific CD4+ T cells of Devic B7-H1+/+ and Devic B7-H1-/- mice seemed not to differ. However, enhanced expression of the transmigration-relevant integrin LFA-1 on CD4+ T cells in young symptom-free Devic B7-H1-/- mice might hint on temporally differently pronounced transmigration capacities during the disease course. Moreover, we attributed the earlier conversion of CD4+ T cells into Th1 effector cells in Devic B7-H1-/- mice during the initiation phase to the lack of co-inhibitory signaling via PD-1/B7-H1 possibly leading to an accelerated disease onset. Full blown autoimmune inflammatory processes could have masked these slight effects of B7-H1 in the clinical phase. Accordingly, at peak of the disease, Th1 and Th17 effector functions of peripheral CD4+ T cells were comparable in both mouse groups. Moreover, judging from titers of MOG-specific IgG1 and IgM antibodies, alterations in humoral immunity were not detected. Therefore, clinical differences could not be explained by altered T-cell or B-cell effector functions at disease maximum. B7-H1 rather seemed to take inhibitory effect in the periphery during the initiation phase only and consistently within the target organ by parenchymal expression. Our observations indicate that B7-H1 plays a relevant role in the regulation of T-cell responses in this mouse model for spontaneous CNS autoimmunity. By exerting immune-modulating effects in the preclinical as well as the clinical phase of the disease, B7-H1 contributed to the confinement of the immunopathological tissue damage in Devic B7-H1+/+ mice mirrored by later disease onsets and lower disease scores. As a model for spontaneous autoimmunity featuring a close to 100 % incidence rate, the Devic B7-H1-/- mouse may prove instrumental in clarifying disease-triggering and -limiting factors and in validating novel therapeutic approaches in the field of autoimmune neuroinflammation, in particular the human Devic’s disease.
Investigation on Distinct Roles of Smad Proteins in Mediating Bone Morphogenetic Proteins Signals

Bone morphogenetic proteins (BMPs) belong to the transforming growth factor-β (TGF-β) superfamily and play important roles in numerous biological events in the development of almost all multi-cellular organisms. Dysregulated BMP signaling is the underlying causes of numerous heritable and non-heritable human diseases including cancer. The vast range of biological responses induced by BMPs converges on three closely related Smad proteins that convey intracellular signals from BMP receptors to the nucleus. The specificity of BMP signaling has been intensively investigated at the level of ligand-receptor interactions, but how the different Smad proteins contribute to differential signals elicited by BMPs remains unclear. In this work, we investigated the BMP/Smad signaling in different aspects. In search for an appropriate fluorescence reporter in zebrafish, we compared different photo-switchable proteins and found EosFP the best candidate this model system for its fast maturation and fluorescence intensity. We modified and created appropriate vectors enabling Tol2-transposon based trangenesis in zebrafish, with which transgenic zebrafish lines were generated. We combined fluorescence protein tagging with high resolution microscopy and investigate the dynamics of Smad proteins in model system zebrafish. We observed that Smad5 undergoes nucleo-translocation as BMP signal transmitter during zebrafish gastrulation. We explored the Smad involvement during myogenic-to-osteogenic conversion of C2C12 cell line induced by BMP4. We created transient loss-of-function of Smads by siRNA-mediated knockdowns and analyzed the effects on these coupled yet distinct procedures by quantitative real-time PCR and terminal marker staining. We found that different Smad-complex stoichiometry might be responsible for distinct cellular signals elicited by BMPs.
