In dieser Studie wurden Schlaganfallpatienten untersucht, die einen ausgedehnten Infarkt im Versorgungsgebiet der A.cerebri media erlitten und wegen Bewusstseinstrübung (sog. Maligner Mediainfarkt) auf der Neurologischen Intensivstation des Universitätsklinikums Würzburg im Zeitraum von 1991 bis 2005 behandelt wurde, um herauszufinden, welchen Einfluss eine operative Behandlung auf den kritisch erhöhten Hirnsdrucks zusätzlich zur konservativen Intensivtherapie auf Mortalität sowie langfristige Lebensqualität hatte. Insgesamt konnten die Daten von 292 Patienten ausgewertet werden, wovon 259 konservativ und 33 operativ behandelt worden waren. Es zeigte sich insgesamt, dass eine stillschweigende günstige Selektion für eine Trepanation sprach (jünger, eher keine Aphasie, weniger Komorbiditäten). Die Hemikraniektomie senkte die Mortalität in der Akutphase hochsignifikant (K: 22, 4%, T: 3,0%; p=0,009). Sie hatte erwartungsgemäß auch einen positiven Einfluss auf das Vigilanzniveau: die Quote von wachen Patienten war bei Entlassung der Trepanierten um 66% höher als bei Aufnahme, bei den konservativ Behandelten war sie nur um 33,3 % gestiegen. Das vorherrschende Symptom bei Aufnahme und Entlassung war eine motorische bzw. sensomotorische Hemiparese. Der Anteil der Aphasiker bei den 201 konservativ therapierten Patienten, die den stationären Aufenthalt überlebt haben, ist von bei Aufnahme 56,2% auf bei Entlassung 48,6% gesunken. Bei den 32 trepanierten Patienten ist er dagegen mit 50% gleich geblieben, obwohl 2/3 aller Patienten an der nicht-dominanten Hemisphäre operiert worden waren. Es war und ist auch nicht zu erwarten, dass eine Entlastung von Hirndruck qualitativ die hirninfarktbedingten Symptome beseitigt. Die Nachbefragung der Patienten fand im Schnitt 64,7 Monate nach erlittenem Mediainfarkt statt. Inzwischen waren von den 259 konservativ Behandelten 47,1% verstorben, von den 33 Hemikraniektomierten nur 24,2%. Die poststationäre Mortalität im weiteren Verlauf war anteilsmäßig gering (K: 24%, T: 21,2%). Die Überlebensdauer der Trepanierten war dreimal so lang wie die der nicht operierten (K: 11,6 Monate, T: 34,4 Monate). Diese Unterschiede im Langzeitüberleben sind wahrscheinlicher auf die geringeren Komorbiditäten der Trepanierten zurückzuführen, als auf die stattgehabte Operation an sich. Allerdings ist nicht auszuschließen, dass die durch Trepanation frühzeitiger verbesserte Wachheit sich auch günstig auf lebensverkürzende Folgekomplikationen ausgewirkt haben könnte. In der Nachbefragung zeigte sich, dass bezüglich der erworbenen körperlichen Funktionsdefizite, gemessen am Barthel Index, zwischen den beiden Kollektiven kein signifikanter Unterschied bestand. Die ehemals konservativ behandelten Patienten kamen auf durchschnittlich 75, die trepanierten Patienten auf 60 von 100 Punkten. Im Lebensalltag schlägt sich dieser Unterschied von 15 Punkten relevant nieder, aber insgesamt liegen beide Patientenkollektive im Bereich einer leichten bis nicht vorhandenen Abhängigkeit. Die vergleichbaren Langzeitdaten von Patienten mit Mediainfarkt liegen in einem ähnlichen Bereich. Erstmalig werden hier Langzeitdaten solcher Patienten über die Lebensqualität vorgelegt, gemessen mit dem SF-36. Nachvollziehbar zeigte sich ein deutlicher Unterschied zur Lebensqualität der Durchschnittsbevölkerung, insbesondere im Bereich der körperlichen Belastbarkeit. Für uns unerwartet günstig fielen die Antworten auf der eher psychologischen Ebene aus. Es zeigten sich bei allen Punkten des SF-36 keine signifikanten Unterschiede zwischen dem konservativ behandelten und den hemikraniektomierten Patienten, so dass die Operation als solche keinen eigenständigen Einfluss auf die langfristige Lebensqualität nahm. Zusammengefasst verbesserte die osteoklastische Trepanation des raumfordernden malignen Mediainfarkts die Überlebenschance in der Akutphase signifikant, was mit inzwischen publizierten kontrollierten Studienergebnissen übereinstimmt. Der Langzeitverlauf nach überlebter Akutkrankheit gestaltet sich unabhängig von der Trepanation. Es gibt aufgrund der erworbenen Behinderung eine weiterhin relativ hohe längerfristige Sterblichkeit. Bemerkenswert ist, dass die Selbsteinschätzung der Lebensqualität von Patienten mit einer erheblichen infarktbedingen körperlichen Behinderung psychologisch-emotional nur geringfügig von der Selbstwahrnehmung in der nicht- behinderten Durchschnittsbevölkerung. Dass bedeutet, dass Spekulationen über die zukünftige Lebensqualität keinen Einfluss auf die Operationsindikation nehmen sollten.
China’s monetary policy aims to reach two final targets: a paramount economical target (i.e. price stability) and a less important political target (i.e. economic growth). The main actor of monetary policy is the central bank, the People’s Bank of China (PBC). But the PBC is a non-independent central bank. The State Council approves the goals of monetary policy. Very limited instrument independence means that interest rates cannot be set at the PBC’s discretion, and in-sufficient personal independence fails to insulate central bank officials from political influence. Monetary policy in China applies to two sets of monetary policy instruments: (i) instruments of the PBC; and (ii) non-central bank policy instruments. The instruments of the PBC include price-based indirect and quantity-based direct instruments. Non-central bank policy instruments include price and wage controls. The simultaneous usage of all these instruments leads to various distortions that ultimately prevent the interest rate channel of monetary transmission from functioning. Moreover, the strong influences of quantity-based direct instruments and non-central bank policy instruments bring into question the approach of indirect monetary policy in general. The PBC officially follows the monetary targeting approach with monetary aggregates as intermediate targets. Domestic loan growth and the exchange rate are defined as additional intermediate targets. In an in-depth analysis of the intermediate targets two main issues are primarily explored: (i) Are the intermediate targets of the Chinese monetary policy controllable? (ii) Is a sufficient relationship between these targets and the inflation rate observable? It is then shown that monetary aggregates are very difficult to control, but they have a satisfactory relationship with the inflation rate. Similarly, domestic loan growth is difficult to control – a fact largely attributed to the interest rate elasticity of loans – while there is a particularly close relationship between credit growth and the inflation rate. The exchange rate as an intermediate target can be controlled through foreign exchange market interventions; at the same time the exchange rate appears to have a significant relationship to the domestic inflation rate. Discussing the special issue of sterilizing foreign exchange inflows, the study concludes that between 2002 and 2008 not only no costs were incurred by sterilization operations, but that the central bank was actually able to realize a profit through foreign exchange market interventions. Based on this, it is concluded that the exchange rate target has not adversely affected the domestic orientation of monetary policy on the whole. The final part of the study examines whether there are any alternative monetary policy approaches that may be able to describe the policy approach in China; special focus is placed on nominal GDP targeting, the Taylor rule, and inflation targeting. A literature review reveals that the concept of nominal GDP targeting may be able to detect inflationary tendencies in the economy and, in combination with other indicators, it could be a suitable concept to assess the overall economic situation. The author calculates a Taylor rule for China from 1994 to 2008 and concludes that there is no close relationship between the PBC lending and the Taylor rate. The author then designs an augmented Taylor rule expanded to include a credit component (credit-augmented Taylor rule). The study shows that the augmented Taylor rule does not perform much better than the original one, but that it maps high inflationary periods relatively well. This is attributed to direct interventions into the credit markets, which have played a major role in combating inflationary cycles over the past decades. The analysis ends with an introduction of the concept of inflation targeting and an examination of whether this could describe monetary policy in China. It is clear that the PBC does not currently follow the inflation targeting approach, although the Chinese authorities could actually be able to influence inflation expectations effectively, not least through direct instruments such as price controls. The author notes that the PBC indeed had a good track record of fighting inflation between 1994 and 2008, and that this may now indicate a good time to think about introducing inflation targeting in China. The central conclusion of the study is that the proven gradual approach to economic and monetary reforms in China is reaching its limit. To break the vicious cycle that relies on the continuous use of quantity-based instruments to compensate for the ineffective price-based instruments – which in turn arises from the simultaneous use of both types of instruments – a complete shift away from quantity-based instruments is needed. Only then the approach of indirect monetary policy, which was officially introduced in 1998, could come into full play.
Diese Dissertationsarbeit liefert einen Beitrag zur Erfassung und Bewertung von Degradationsprozessen im semi-humiden Süden Spaniens. Der erste Teil der Arbeit widmet sich der detaillierten physisch-geographischen Charakterisierung des Großraumes, um danach in dem kleinräumigen Einzugsgebiet des Arroyo del Alforzo, einem Tributär des Río Turón, zwei unterschiedliche Ansätze zur Erfassung von die degradationsbeeinflussenden Fatoren wie Landnutzungswechsel und Starkniederschlagsereignissen in diesem Raum zu untersuchen. Anhand von drei Satellitenbildern wurde der Landnutzungswechsel ermittelt und im Untersuchungsgebiet die Abhängigkeit zu den Hangneigungen untersucht. Vor dem Hintergrund, daß unterschiedlich starke Hangneigungen einen unterschiedlich starken Einfluss auf die Abtragsdynamik bei Niederschlägen hat, wurden anhand der Landnutzungsklassifizierungen in Kombination mit den Hangneigungnen sensible Räume ermittelt. Ein weiterer Ansatz ist die Untersuchung von Tagesniederschlagsdaten auf Starkniederschlagsereignisse, mit dem Ziel, diese zu separieren. Es galt die Annahme, daß diese Starkniederschlagsereignisse im Einzugsgebiet des Arroyo del Alforzo oberflächlichen Abfluss generieren und somit ein bedeutender Sedimenteintrag aus den sensiblen Bereichen des Untersuchungsgebiet in den Stausee Conde de Guadalhorce stattfindet. Mittels sedimentstratigraphischer Untersuchungen an den Sedimenten des 2006 gewonnenen Bohrkerns aus dem Mündungsbereich des Arroyo del Alforzo in den Stausee Conde de Guadalhorce sollte dieser Sedimenteintrag identifierziert werden und somit ein zeitlicher und räumlicher Rückschluss auf die die Abtragung beeinflussenden faktoren Landnutzungswechsel, Hangneigung und Niederschlag efolgen. Die Annahme, dass sich diese Rückschlüsse ziehen lassen können auf der Grundlage des Bindeglieds „Sedimentbohrung“ erwies sich jedoch als zu eng. In einer abschliessenden Bewertung wurde erläutert, daß durch eine gezielte methodische Ergänzung jedoch die Möglichkeit besteht, die Unsicherheiten, die durch eine räumlich wie zeitlich inkonsistente Datenlage der Niederschlagsdaten und die in einem Stausee herrschende spezielle Akkumulationsdynamik hervorgerufen wurde, beseitigt werden kann.
This thesis included the synthesis of conformationally stable chiral perylene bisimide (PBI) dyes, the study of their optical properties in solution and their chiral self-sorting behaviour in nonpolar solvents in which dimerization via pi-pi-stacking takes place. Furthermore, the influence of PBI core chirality on the properties of these dyes in the condensed state has been also studied. We have demonstrated and quantified the prevalence of chiral self-recognition over self-discrimination in pi-stacking dimerization of PBIs. It has been shown that this self-recognition event is compromised by the increasing flexibility of the structures related to the size of the OEG bridging units. Moreover, the inherent chirality of these PBIs has been proven to strongly influence their condensed state properties, for which large differences between the pure enantiomers and the racemates were revealed, as well as between the different bridged macrocyclic PBIs.
In the frame of this thesis vibronic and electronic states of organic molecules have been examined. A central question is the interaction within and between the molecules in thin films and at metal-organic interfaces. The main experimental tools were high resolution electron energy loss spectroscopy (HREELS) and high resolution near edge X-ray absortion fine structure (NEXFAS). The electronic and vibronic structure of thin NTCDA films was examined with low energy electrons as probe, i.e. HREELS. The spectra of the electronic excited molecular orbitals of submonolayer NTCDA on a Ag(111) shows a partially filled orbital. The interaction between this orbital and the total symetric molecular vibrations leads to the typical Fano peak profiles which are seen in the vibrational spectra. The sub-monolayer superstructure can be driven to a phase transition into an disordered phase upon cooling, which is also seen in the electronic and vibronic excitation spectra. Multilayers show flat lying or upright standing molecules as a function of the preparation conditions. The upright standing molecules show an island growth mode, where the islands are well ordered and exhibit a structure in diffraction experiments which can be attributed to the molecular crystal structure. In order to examine the order in more detail various thin films were examined using SPALEED as function of film thickness and preparation parameters. In case of a low temperature substrate no long range order leading to a diffraction pattern was found. In contrast growth on room temperature substrates leads to island growth of films in a structure of the molecular crystal, where two preferred orientations of the islands relative to the substrate were found. In case of thick films the reference to the substrate gets lost and the molecular crystals grow with a defined crystal direction with respect to the surface but with an arbitrary azimuthal orientation leading to circles in the diffraction pattern. NTCDA monolayers on a Ag(111) surface using HREELS as a tool were examined. The electronic excitation spectra reveal a partially filled molecular orbital which is strongly shifted compared to the multilayer. The existence of this state is responsible for the activation of normally forbidden Ag modes in the vibrational spectra. Due to the electron phonon coupling these modes exhibit a Fano like peak shape. Cooling a monolayer leads to a phase transition with strong changes in the spectroscopic features both in electronic and vibronic excitations. In case of the molecule ANQ the intramolecular interaction was examined. In the oxygen NEXAFS spectra a vibronic fine structure is found, which leads to the conclusion that asymmetric potentials are involved. It is an interesting question if the fundamental vibration is has C-H or C=O character. In order to address this question spectra of condensed and gas phase ANQ were compared to an ANQ derivate (ANQ- Br$_2$Cl$_2$), with the conclusion that the coupling is most likely to a C=O mode. High resolution C1s spectra of hydrogenated and fully deuterated naphthalene both in gas and condensed phase have been presented. Depending on the final state orbital distinct differences have been found between gas and condensed phase. A energetic shift of resonances (Res. B, C, D) is interpreted as effect of $\pi$-$\pi$ interaction in the condensed phase. This is especially notable for resonance B which is undoubtly assigned to an excitation into a $\pi^*$ orbital. The results lead to an interpretation, that for organic molecular crystals more than pure van-derWaals interaction has to be taken into account. In summary it is found that the intramolecular interaction in NEXAFS spectra is preferentially coupled to one or a few vibronic progressions. Due to the delocalized electronic system maybe even states which are not spatially near the core excited atom can be involved. It could be shown that a condensation of the molecules in thin films leads to changes within the spectra. The influence the intermolecular interaction can be clearly seen in this finding, where additional hints are found that more than mere van-der-Waals binding has to be taken into account.
1. Im Rahmen dieser Arbeit konnten neue Erkenntnisse hinsichtlich des angenomme-nen gerichteten Ionentransports zwischen Schließ- und Nebenzellen von Zea mays gewonnen werden: a. Mittels der Patch-Clamp-Technik wurden in beiden Zelltypen S-Typ-ähnliche Anionenkanäle identifiziert. In Nebenzellen konnten sie durch steigende zytosolische Ca2+-Konzentrationen gehemmt und durch ABA und zytosolische Alkalisierung stimuliert werden. Die S-Typ-Anionenkanäle der Schließzellen wurden hingegen durch eine Alkalisierung kaum beeinflusst und durch steigende zytosolische Ca2+-Konzentrationen stimuliert. b. Darüber hinaus konnte an intakten Mais-Pflanzen mit der Einstich-Elektroden-Technik gezeigt werden, dass Nebenzellen eine gegenläufige Polarisation des Membranpotentials während der Licht-/Dunkel-induzierten Stomabewegung aufweisen. Da das Membranpotential der Nebenzellen von Hordeum vulgare ein zu Mais ähnliches Verhalten während der Stomabewegung zeigte und gegenläu-fig zur Membranpolarisation der benachbarten Schließzellen war, ist ein ähnli-ches Verhalten bei Zea mays Schließzellen naheliegend. c. Zudem wurde in intakten Nebenzellen von Zea mays eine zytosolische Alkali-sierung während der Licht-induzierten Stomaöffnung beobachtet, die bei Stomaschluss wieder auf den Ursprungswert zurückkehrte. d. Mit Hilfe rekonstruktierter 3D-Modelle von intakten Mais-Stomakomplexen konnte ein Volumenverhältnis zwischen Schließ- und Nebenzellen von 1:6 bzw. 1:4 bei geöffneten und geschlossenen Stomata ermittelt werden. Unter Einbeziehung der Vorarbeiten unserer Arbeitsgruppe konnten die hier gewon-nenen Erkenntnisse schlüssig in ein Modell zur Beschreibung des Shuttle-Ionentransports zwischen Neben- und Schließzellen während der Licht-induzierten Stomabewegung eingebunden werden. 2. Des Weiteren wurden die S-Typ-Anionenstromantworten von A. thaliana Schließ-zellen in Patch-Clamp-Experimenten näher untersucht. Dabei waren die S-Typ-Anionenströme bei Ca2+- bzw. ABA-Stimulation in CPK23- und OST1-Verlustmutanten im Vergleich zum Wildtyp stark reduziert. Diese in vivo generierten Daten untermauern die in vitro Ergebnisse der Arbeitsgruppe von Prof. R. Hedrich (Universität Würzburg), dass OST1 und CPK23 Interaktionspartner des S-Typ-Anionenkanals SLAC1 in A. thaliana sind. Das SLAC1-homologe Gen SLAH3 ko-diert für einen Nitrat-permeablen S-Typ-Anionenkanal in Schließzellen, der zudem durch externes Nitrat aktiviert wird. Da in slac1-3 Verlustmutanten S-Typ-ähnliche Anionenströme generiert werden konnten, wenn Nitrat das dominierende Anion dar-stellte oder den Chlorid-basierten Lösungen externes Nitrat zugegeben wurde, scheint SLAH3 unter bestimmten Bedingungen einen alternativen Weg für die Ent-lassung von Anionen aus der Schließzelle darzustellen. 3. Die elektrophysiologische Charakterisierung der R-Typ-Anionenkanäle in A. thaliana Schließzellen belegt, dass dieser Kanal ähnliche Grundcharakteristika aufweist, die schon in Vicia faba beschrieben wurden: eine starke Spannungsab¬hängigkeit, sowie schnelle Aktivierungs- und Deaktivierungskinetiken. Im Gegensatz zu Vicia faba wurde die Spannungsabhängigkeit dieses Kanaltyps in A. thaliana nicht durch externes Malat beeinflusst. Jedoch war unter externen Malatbedingungen die Stromantwort einer almt12-Verlustmutante im Vergleich zu Wildtyp-Schließzellen erheblich reduziert, während unter externen Sulfatbe¬dingungen keine Unterschiede zwischen Wildtyp und almt12-Verlustmutante auszu¬machen waren. ALMT12 scheint demnach für den Malat-aktivierten Teil des R-Typ-Anionenkanals verantwortlich zu sein.
Bisherige Studien zur Prognose ehemaliger sehr kleiner Frühgeborener (Geburtsgewicht < 1500 g, Gestationsalter < 32 SSW) in Deutschland haben sich überwiegend mit der Entwicklung bis in das Pubertätsalter befasst. Bekannt ist die signifikante Häufung von kognitiven Defiziten, Zerebralparesen und Sinnesbehinderungen in Korrelation mit dem Gestationsalter im Vergleich zu ehemalig reifen Neugeborenen. Nach einem Pilotprojekt an der Univ.-Kinderklinik Würzburg wurden Daten von 291 Probanden, die zwischen 1979 und 1986 geboren und in zehn verschiedenen Kinderkliniken primär behandelt worden sind, gesammelt. Dabei konnte eine deutliche Zunahme der Zahl der Probanden in den Geburtsjahrgängen nach 1982 festgestellt werden. 50 (17 % ) des Kollektivs hatten ein Geburtsgewicht unter 1000 g. 27 % haben die Schule mit dem Abitur abgeschlossen, 37 % mit der mittleren Reife, 22 % mit einem Hauptschulabschluss, die meisten befanden sich aktuell noch in der Berufsausbildung. 22 % berichteten über Hinweise für eine chronische Bronchitis, 19 % hatten regelmäßig Physiotherapie. 54 % sind Brillenträger. Mit Hilfe des Fragebogens zur Lebensqualität KIDDO-KINDL konnten in den Bereichen körperliches und psychisches Wohlbefinden sowie Alltagsfunktionen keine signifikante Abweichung von Daten eines Normalkollektivs festgestellt werden. Auch ein Vergleich der gleich großen Gruppen aus den west- und ostdeutschen Bundesländern ergab in Bezug auf die Lebensqualität keinen signifikanten Unterschied. Weitere Auswertungen dieser Ergebnisse sind notwendig. Zusammenfassend ist trotz erhöhter Morbidität die subjektive Einschätzung der Lebensqualität der ehemalig sehr kleinen Frühgeborenen im Erwachsenenalter, die an unserer Erhebung teilgenommen haben, in Deutschland nicht signifikant schlechter als in einem Normalkollektiv und entspricht den Angaben der internationalen Literatur. Zur genaueren Analyse der vielfältigen Einflussfaktoren auf die Langzeitprognose sind konsequente multizentrische Langzeitstudien, besonders von den Frühgeborenen mit einem Geburtsgewicht unter 1000 g, unbedingt erforderlich.
Protein phosphatases can be classified into at least three major families based on amino acid sequences at their active sites. A newly emerging phosphatase family contains the active site sequence DXDX(T/V), and belongs to the haloacid dehalogenase (HAD) superfamily of hydrolases, a ubiquitous and evolutionarily conserved enzyme family. Although the existence of 58 human HAD enzymes has been predicted by database analysis, our understanding of their biological functions remains rudimentary.By database mining amd phylogenetic analysis of human HAD phosphatases, we have found a marked increase in cell area of spreading cells, as well as accelerated cell spreading onfibronectin. Taken together, we have identified and characterized AUM as a novel member of the emerging family of aspartate-dependent protein tyrosine phosphatases. Our findings implicate AUM as an important regulator of Src-dependent cytoskeletal dynamics during cell adhesion and migration. a previously unidentified enzyme with homology to Chronophin, a cytoskeletal regulatory HAD phosphatase. We have cloned and characterized this novel enzyme and named it AUM,for actin remodeling, ubiquitously expressed, magnesium-dependent HAD phosphatase. By Northern blot, real-time PCR and Western blot analysis, we show that AUM is broadly expressed in all major human and mouse tissues with highest levels found in testis. Using immunohistochemistry, we can show that AUM is specifically expressed in maturing germ cells and that its expression peaks during spermiogenesis. To characterize the substrate preference of AUM, we have conducted an in vitro phosphatase substrate screen with 720 phosphopeptides derived from human phosphorylation sites. AUM exclusively dephosphorylates phosphotyrosine (pTyr)-containing peptides. Furthermore, only 17 pTyr peptides (~2% of all pTyr peptides investigated) acted as AUM substrates, indicating a high degree of substrate specificity. Putative AUM substrates include proteins involved in cytoskeletal dynamics and tyrosine kinase signaling.In accordance with the phosphopeptide screen, phosphatase overlay assays employing whole-cell extracts of pervanadate-treated HeLa cells show that AUM dephosphorylates only a limited number of tyrosyl-phosphorylated proteins.The role of AUM for cellular signaling was investigated in response to epidermal growth factor (EGF) stimulation in a spermatogonial cell line (GC-1 spg). The overexpression of AUM reduces, whereas the RNAi-mediated depletion of endogenous AUM increases EGF inducedtyrosine phosphorylation, including changes in the phosphorylation of the EGF receptor itself. Interestingly, in vitro kinase/phosphatase assays with purified Src and AUM indicate that AUM can activate Src, which in turn phosphorylates and inactivates AUM. Although it is at present unclear how Src and AUM regulate each other, our initial findings suggests that AUM enhances Src kinase activity independently of its phosphatase activity, whereas Src diminishes AUM phosphatase activity in a kinase dependent manner. On a cellular level, AUM-depleted cells are characterized by altered actin cytoskeletal dynamics and adhesion, as indicated by stabilized actin filaments, enlarged focal adhesions,a marked increase in cell area of spreading cells, as well as accelerated cell spreading on fibronectin. Taken together, we have identified and characterized AUM as a novel member of the emerging family of aspartate-dependent protein tyrosine phosphatases. Our findings implicate AUM as an important regulator of Src-dependent cytoskeletal dynamics during cell adhesion and migration.
Ruptures of the anterior cruciate ligament (ACL) and defects of the rotator cuff represent the most common ligament and tendon injuries in knee and shoulder. Both injuries represent significant implications for the patients. After an injury, the ACL and the rotator cuff both exhibit poor intrinsic healing capacities. In order to prevent further defects such as arthritis of the knee and fatty infiltration of the rotator cuff, surgical interaction is essential. In both cases, the currently used surgical techniques are far from optimal because even after the therapy many patients report problems ranging from pain and reduced mobility to complete dysfunction of the involved joint and muscles. Tissue engineering may be a possible solution. It is a promising field of regenerative medicine and might be an advantageous alternative for the treatment of musculoskeletal injuries and diseases in the near future. In this thesis, different tissue engineering based approaches were investigated. For the reconstruction of damaged or diseased ligaments and tendons, the use of MSCs and gene therapy with growth factors is especially suitable and possesses a great therapeutic potential. Therefore, the first method studied and tested in this thesis was the development of a biomaterial based construct for the repair of a ruptured ACL. The second approach represents a cell based strategy for the treatment of the fatty infiltration in the rotator cuff. The third approach was a combined cell, biomaterial, and growth factor based strategy for ACL ruptures. Biomaterial based ACL construct The implant is currently tested in a preclinical in vivo study in mini pigs. This proof-of-principle study is performed to validate the functional capability of the collagen fiber based implant under load in vivo and its population with fibroblasts which produce a ligamentogenic matrix. Cell based treatment of the fatty infiltration in the rotator cuff Regarding the treatment of the fatty infiltration of the rotator cuff in a rabbit model, the in vivo results are also promising. The group treated with autologous MSCs (+MSC group) showed a lower fat content than the untreated group (–MSC group) 6 weeks after the treatment. Furthermore, the SSP muscle of the MSC-treated animals revealed macroscopically and microscopically only few differences compared to the healthy control group. The exact underlying mechanisms leading to the positive results of the treatment are not yet fully understood and have therefore to be further investigated in the future. Cell, biomaterial, and growth factor based treatment of ACL ruptures Studies described in current literature show that collagen hydrogel scaffolds are not ideal for a complete ligament or tendon reconstruction, because of their insufficient mechanical stability. Introduced as an alternative and superior therapy, the combined strategy used in this thesis proves that the cultivation of BMP-12, -13, and IGF-1 transduced MSCs and ACL fibroblasts in a collagen hydrogel is successful. The results of the performed in vitro study reveal that the cells exhibit a fibroblastic appearance and produce a ligamentogenic matrix after 3 weeks. Furthermore, the adenoviral transduction of MSCs and ACL fibroblasts showed no negative effects on proliferation or viability of the cells nor was apoptosis caused. Therefore, the application of these cells represents a possible future therapy for a partial ligament and tendon rupture where the mechanical stability of the remaining ligament or tendon is sufficient and the healing can be improved substantially by this therapy. In general, prospective randomized clinical trials still have to prove the postulated positive effect of MSCs for the treatment of various musculoskeletal diseases, but the results obtained here are already very promising. Ideally, the treatment with MSCs is superior compared to the standard surgical procedures. Because of current safety issues the use of genetically modified cells cannot be expected to be applied clinically in the near future. In summary, the different tissue engineering approaches for novel therapies for musculoskeletal injuries and diseases invested in this thesis showed very promising results and will be further developed and tested in preclinical and clinical trials.
Precise control of mitotic progression is vital for the maintenance of genomic integrity. Since the loss of genomic integrity is known to promote tumorigenesis, the identification of knew G2/M regulatory genes attracts great attention. LINC, a human multiprotein complex, is a transcriptional activator of a set of G2/M specific genes. By depleting LIN9 in MEFs, a core subunit of LINC, Gas2l3 was identified as a novel LINC target gene. The so far uncharacterized Gas2l3 gene encodes for a member of the family of growth arrest specific 2 (GAS2) proteins, which share a highly conserved putative actin binding CH and a putative microtubule binding GAS2 domain. In the present study GAS2L3 was identified as a LINC target gene also in human cells. Gene expression analysis revealed that GAS2L3 transcription, in contrast to all other GAS2 family members, is highly regulated during the cell cycle with highest expression in G2/M. The GAS2L3 protein showed a specific localization pattern during the M phase: In metaphase, GAS2L3 localized to the mitotic spindle, relocated to the spindle midzone microtubules in late anaphase and concentrated at the midbody in telophase where it persisted until the end of cytokinesis. Overexpression of a set of different GAS2L3 deletion mutants demonstrated that the localization to the mitotic microtubule network is dependent on the C-terminus, whereas the midbody localization is dependent on full length GAS2L3 protein. Additionally, exclusive overexpression of the CH domain induced the formation of actin stress fibers, suggesting that the CH domain is an actin binding domain. In contrast, the GAS2 domain was neither needed nor sufficient for microtubule binding, indicating that there must be an additional so far unknown microtubule binding domain in the C-terminus. Interestingly, immunoblot analysis also identified the C-terminus as the domain responsible for GAS2L3 protein instability, partially dependent on proteasomal degradation. Consistent with its specific localization pattern, GAS2L3 depletion by RNAi demonstrated its responsibility for proper mitosis and cytokinesis. GAS2L3 depletion in HeLa cells resulted in the accumulation of multinucleated cells, an indicator for chromosome mis-segregation during mitosis. Also the amount of cells in cytokinesis was enriched, indicating failures in completing the last step of cytokinesis, the abscission. Strikingly, treatment with microtubule poisons that lead to the activation of the spindle assembly checkpoint (SAC) indicated that the SAC was weakened in GAS2L3 depleted cells. Although the exact molecular mechanism is still unknown, fist experiments support the hypothesis that GAS2L3 might be a regulator of the SAC master kinase BUBR1. In conclusion, this study provides first evidence for GAS2L3 as a novel regulator of mitosis and cytokinesis and it might therefore be an important guardian against tumorigenesis.
