Thylakoid and cytoplasmic membranes of the cyanobacterium Syncchocystis sp. PCC 6803 were purified by sucrose gradient centrifugation. Both membranes oxidize NADH in a rotenone-sensitive reaction. Antibodies prepared against psbG/ndhKand ndhJ fusion proteins detect the corresponding polypeptides in both membrane preparations. This demonstrates that a NADH-dehydrogenase, homologous to the mitochondrial NADHubiquinone-oxidoreductase (complex I of the respiratory chain) is present in cyanobacteria, The NADH-dehydrogenase can be solubilized with the detergent /-D-dodecylmaltoside. Sedimentation analysis of the solubilized enzyme on a sucrose gradient indicates that it is a multisubunit protein complex.
An antibody against the NDH-K subunit of the NAD(P)H-dehydrogenase from the cyanobacterium Synechocystis sp. PCC6803 was used to isolate a subcomplex ofthe enzyme from Triton X-lOO solubilized total membranes by immunoaffinity chromatography. The isolated subcomplex consisted of seven major polypeptides with molecular masses of 43, 27, 24, 21, 18, 14 and 7 kDa. The amino-terminal amino acid sequences of the polypeptides were determined. By comparing the sequences with the amino acid sequences deduced from DNA. three proteins were identified as NDH-H (43 kDa). NDH-K (27 kDa) and NDH-J (24 kDa). A fourth subunit (NDH-J, 21 kDa) was identified by Western blot analysis with an NDH-J antibody.
The plastid genomes of higher plants contain eleven reading frames (ndhA-K) that are homologous to genes encoding subunits of the mitochondrial NADH-ubiquinone-oxidoreductase (complex I). The carboxyterminal end of the NDH-H subunit from rice (Oryza sativa L.) was expressed as a fusion protein in Escherichia coli and antibodies against the fusion protein were generated in rabbits. The antibody was used to study the expression of NDH-H, and the following results were obtained: (i) NDH-H is expressed in mono- and dicotyledonous plants, (ii) NDH-H is localized on the stroma lamellae of the thylakoid membrane and (iii) NDH-H is expressed in etioplasts. Together with the finding that two other ndh genes (ndhI and ndhK) are expressed in plastids, these results point to the existence of an NAD(P)H-plastoquinone-oxidoreductase on the thylakoid membrane. The possible function of the enzyme in plastids is discussed and it is suggested that it works in balancing the ATP/ADP and the NADPH/NADP ratios during changing external (i.e. light) or internal (i.e. ATP and NADPH demands of biosynthetic pathways of the plastid) conditions.
In der inkompatiblen Interaktion von Lycopersicon esculentum und Cuscuta reflexa wird die Ausbildung von Haustorien, spezieller Organe, die der Nahrungsaufnahme durch den Parasiten dienen,bereits in einem sehr frühen Stadium der Infektion gehemmt. Um einen Einblick in die Regulationsmechanismen der Tomatenreaktion zu gewinnen, wurde eine Subtraktive Hybridisierung durchgeführt und es konnten 20 Gene identifiziert werden, deren Transkripte nach Cuscuta-Befall im Wirtsgewebe akkumulieren. Entsprechend ihrer möglichen Proteinfunktion lassen sich die mRNAs verschiedenen Bereichen der Tomatenreaktion im Infektionsprozess zuordnen: (a) Abwehr-assoziierte Proteine, (b) Signaltransduktionsassoziierte Proteine, (c) Zellstreckungsassoziierte Proteine und (d) Proteine mit bislang vollständig unbekannter Funktion. Einige der identifizierten mRNAs wurden durch Northern Analysen näher charakterisiert. Da eine der mRNAs eine mögliche Xyloglucanendotransglycosylase (XET) kodiert, wurde die XET-Aktivität im Tomatengewebe nach Infektion bestimmt. Außerdem wurde der Einfluß des Phytohormons Auxin auf die Akkumulation der Xyloglucanendotransglycosylase LeEXT1 sowie des Aquaporins LeAqp2 untersucht. Trotz auxinregulierter Transkription nach Cuscuta-Befall zeigte die auxininsensitive Tomatenmutante diageotropica im Vergleich zum Wildtyp keine veränderte Kompatibilität.
Um einen Beitrag zum besseren Verständnis der Rolle der Bienenwachse in der Kommunikation der Honigbienen leisten zu können, wurden Wabenwachse unterschiedlichen Alters und Kutikulawachse unterschiedlicher Kasten,Geschlechter und Berufsgruppen mit Hilfe von Gaschromatographie, Massenspektroskopie und FTIR-Spektroskopie untersucht. Die chemischen Analysen zeigten mittels Diskriminantenfunktionsanalysen hochsignifikante Unterschiede in den aliphatischen Kohlenwasserstoffen zwischen Wabenwachsen unterschiedlichen Alters und Kutikulawachsen unterschiedlicher Kasten und Geschlechter. Erstmals konnte für ein komplexes Substanzgemisch (Bienenwachs) eine lineare Abhängigkeit zwischen dem Schmelzverhalten und der chemischen Zusammensetzung der Wachse nachgewiesen werden.Mit Hilfe von Verhaltensversuchen wurde der Frage nachgegangen, ob die chemischen Unterschiede für die Bienen überhaupt relevant sind. Mit Hilfe der differentielle Konditionierung des Rüsselreflexes wurde getestet, inwieweit Bienen die verschiedenen Wachse unterscheiden können. Eine Diskriminierung der Wachse aufgrund der aliphatischen Kohlenwasserstoffe war den Honigbienen nicht möglich. Dies ergab einen neuen und interessanten Einblick in die Kommunikation der Honigbienen
In der vorliegenden Arbeit wurden pflanzliche Aquaporine aus Nicotiana tabacum und Samanea saman mit molekularbiologischen Methoden analysiert und durch heterologe Expression in Oozyten von Xenopus laevis funktionell charakterisiert. Das aus Tabak isolierte Aquaporin NtAQP1 wird am höchsten in Wurzeln exprimiert, ist trotz vorhandener Cysteine nicht quecksilbersensitiv und permeabel für Glycerin und Harnstoff. Sowohl eine Ionenleitfähigkeit, als auch eine Regulation auf Proteinebene konnten im Oozytensystem nicht nachgewiesen werden. Die Leguminose Samanea saman bewegt im Tag-/Nachtrhythmus ihre Fiederblätter und -stengel. Dieses wird durch Variieren des Turgors im Flexorgewebe von Pulvini realisiert. Während SsAQP1 in seinen Charakteristika mit NtAQP1 übereinstimmt, ist SsAQP2 quecksilbersensitiv und hoch selektiv für Wasser. Der für SsAQP2 ermittelte Permeabilitätskoeffizient war um den Faktor 10 höher als der von SsAQP1. Northern Experimente zeigten, dass SsAQP2 ausschließlich im Flexorgewebe exprimiert wird. Die Expression ist zum Zeitpunkt der Streckungsbewegung des Pulvinus um 7 Uhr am stärksten. Dagegen wurde SsAQP1 zu allen untersuchten Zeitpunkten nur schwach in Pulvini exprimiert.
In der vorgestellten Arbeit wurden das Wachstum und das physiologische Verhalten von Zea mays auf Müllheizkraftwerk (MHKW) -Schlacke im Vergleich zu Gartenerde als Kulturmedium untersucht. Dabei stand das Wurzelsystem der Maispflanzen im Mittelpunkt des Interesses. Da feste Bodensubstrate verwendet wurden, mußten diese zu Beginn der Experimente chemisch, physikalisch und bodenbiologisch charakterisiert werden. Die Analyse der Schlacke zeigte, daß Schlacke ein multifaktorielles Streßsystem darstellt: Sie enthält einen hohen Gehalt an leicht löslichen Salzen, v.a. NaCl (bis zu 220 mM in der Bodenlösung). MHKW-Schlacke ist dagegen arm an Stickstoff und pflanzenverfügbarem Phosphat. Der pH-Wert der Bodenlösung von Schlacke ist stark alkalisch (pH 8.4 - 9.0). Darüber hinaus besitzt Schlacke einen hohen Gehalt an potentiell toxischen Schwermetallen und weist im Vergleich zum Kontrollsubstrat Gartenerde eine verdichtete Bodenstruktur mit erhöhtem mechanischen Widerstand auf. Im Vergleich zu der Kontroll-Anzucht auf Gartenerde reagierten die auf Schlacke kultivierten Mais-Pflanzen mit vermindertem Wachstum: Sproß und Wurzel erreichten nur die Hälfte der Länge der Kontrollpflanzen. Ein Vergleich der Biomassen von Sproß und Wurzel zeigte, daß das Sproßwachstum der Schlacke-Pflanzen stärker eingeschränkt ist als das Wurzelwachstum, woraus ein vergrößertes Wurzel / Sproß-Verhältnis resultiert. Das Wachstum von jungen Mais-Pflanzen auf Schlacke ist jedoch nicht in dem Maß eingeschränkt, wie es aufgrund der hohen Salzbelastung zu erwarten wäre. In einem Vergleichsexperiment mit Mais-Pflanzen, die in einer Nährlösung mit Zusatz von 100 mM NaCl kultiviert wurden, war das Wachstum erheblich schlechter und in den Blättern akkumulierte weitaus mehr Natrium als in Schlacke-Pflanzen. Hier wird der positive Einfluß des hohen Calciumgehaltes der Schlacke deutlich. Die Beeinträchtigung des Wachstums von Mais bei Kultur auf Schlacke wird hauptsächlich auf Phosphatmangel zurückgeführt, da durch Düngung eine beträchtliche Wachstumsverbesserung erzielt werden kann. Zudem wurden keine toxischen Konzentrationen an Schwermetallen im Blattgewebe von auf Schlacke kultivierten Pflanzen gefunden. Der Photosynthese-Apparat der Schlacke-kultivierten Pflanzen war sehr leistungsfähig: Es bestand keine Beeinträchtigung in der Energieverfügbarkeit (Quantenausbeute des Photosystems II) und die Lichtsättigung der photo-synthetischen Elektronentransportrate lag sogar höher als bei den Kontrollpflanzen. Die Bestimmung des „adenylate energy charge“ bestätigte diesen Sachverhalt. Das Wurzelsystem von Zea mays auf Schlacke wies strukturelle Veränderungen auf. Neben der verkürzten Wurzellänge und dem vergrößerten Wurzeldurchmesser der Schlacke-Pflanzen ergaben mikroskopische Untersuchungen, daß die Wurzeln durch Kultur auf Schlacke mit einer mechanischen Verstärkung reagieren: Stärker ausgeprägte tangentiale Zellwandverdickungen der Endodermis im tertiären Zustand und Zellwandmodifikationen in den radialen Zellwänden der Rhizodermis (Phi-Verdickungen). Für monokotyle Arten, insbesondere für Mais, gibt es bisher keine Beschreibung von Phi-Verdickungen in der Literatur. Gaschromatographische und massenspektrometrische Untersuchungen belegen, daß sich die Zellwände von auf Erde und Schlacke kultivierten Maiswurzeln im Hinblick auf den Gesamtgehalt an Lignin (endodermale Zellwandisolate) und in der Ligninzusammensetzung (hypodermale Zellwandisolate) unterscheiden: In Schlacke-kultivierten Maiswurzeln wurde ein höherer Anteil an dem Lignin-Monomer p Hydroxyphenyl gefunden, was zu einem höher verdichteten Lignin führt (Streßlignin). Die endodermalen Zellwände von auf Schlacke-kulivierten Pflanzen hatten dagegen einen höheren Gesamtlignin-Gehalt als die entsprechenden Kontrollen, was ebenfalls eine mechanische Verstärkung der Wurzel bewirkt. In Bezug auf Suberin konnten keine Unterschiede zwischen den verschiedenen Anzuchten gefunden werden, weder in den hypodermalen noch in den endodermalen Zellwandisolaten. Die verschiedenen Streßfaktoren führen demnach nicht zu einer verstärkten Imprägnierung der Zellwände mit lipophilem Material. Die Zellwände von Mais spielen eine wichtige Rolle bei der Immobilisierung von Schwermetallen. Die Zellwandisolate von auf Erde und Schlacke kultivierten Mais-Pflanzen wiesen je nach Schwermetall-Element 43 - 100 % des Gesamtgehaltes auf. Die absoluten Gehalte in den Zellwandisolaten von auf Schlacke angezogenen Pflanzen waren dabei höher als die entsprechenden Werte der Kontrolle. Eine Anreicherung in den Zellwänden wurde hauptsächlich für die Schwermetalle Zink, Blei, Nickel und Chrom beobachtet. Als unspezifische Streßantwort reagierten Maispflanzen auf die Kultur in Schlacke mit einer erhöhten Peroxidaseaktivität in der interzellulären Waschflüssigkeit. Die Peroxidaseaktivität des Symplastens der Wurzel unterscheidet sich zwischen den beiden Anzuchten dagegen nicht. Die Konzentration des Phytohormons Abscisinsäure (ABA) war in Blättern von auf Schlacke kultivierten Pflanzen von Zea mays und Vicia faba im Vergleich zu den Kontrollpflanzen erhöht. Dieser Anstieg ist eine Folge der erhöhten Salzbelastung der Schlacke, da die ABA-Gehalte entsprechender Blätter von auf gewaschener Schlacke kultivierten Pflanzen annähernd den Kontrollwerten entsprachen. Bei der Verteilung von ABA zwischen der Wurzel und der Bodenlösung der umliegenden Rhizosphäre konnte das als Anionenfalle bekannte Prinzip bestätigt werden. Nach diesem Modell reichert sich ABA im alkalischten Kompartiment an (hier: Schlacke-Bodenlösung). In den Wurzeln konnte nur in der Maiskultur auf Schlacke ein erhöhter Gehalt gefunden werden, nicht dagegen in der Vicia faba-Kultur. Dieser Unterschied liegt daran, daß Mais im Gegensatz zu Vicia faba eine exodermale Spezies ist und die Exodermis für ABA eine Barriere darstellt, was den ABA-Efflux in die Rhizosphäre verhindert. Im Wurzelgewebe von auf Schlacke kultivierten Maispflanzen wurde ein im Vergleich zur Kontrolle 15-facher Gehalt an wasserlöslichen, nicht proteingebundenen Sulfhydrylgruppen nachgewiesen. Diese auf Schwermetallstreß zurückzuführende Reaktion impliziert, daß die in der Schlacke-Bodenlösung vorhandenen Schwermetalle nicht ausreichend im Apoplasten zurückgehalten werden und bis in den Symplasten vordringen können.
A central objective of many ecophysiological investigations is the establishment of mechanistic explanations for plant distributions in time and space. The important, albeit mostly ignored, question arises as to the nature of the organisms that should be used as representative in pertinent experiments. I suggest that it is essential to use a “demographic approach” in physiological ecology, because physiological parameters such as photosynthetic capacity (PC, determined under non-limiting conditions with the oxygen electrode) may change considerably with plant size. Moreover, as shown for nine epiphyte species covering the most important taxonomic groups, the intraspecific variability in PC was almost always higher than the interspecific variability when comparing only large individuals. In situ studies with the epiphytic bromeliad V. sanguinolenta revealed that besides physiological parameters (such as PC) almost all morphological, anatomical and other physiological leaf parameters studied changed with plant size as well. Likewise, important processes proved to be size-dependent on whole-plant level. For example, long-term water availability was clearly improved in large specimens compared to smaller conspecifics due to the increased efficiency of the tanks to bridge rainless periods. As model calculations on whole-plant level for V. sanguinolenta under natural conditions have shown photosynthetic leaf carbon gain as well as respiratory losses of heterotrophic plant parts scaled with plant size. The resulting area related annual carbon balances were similar for plants of varying size, which corresponded to observations of size-independent (and low) relative growth rates in situ. Under favorable conditions in the greenhouse, however, small V. sanguinolenta exhibited surprisingly high relative growth rates, similar to annuals, which clearly contradicts the prevalent, but barely tested notion of epiphytes as inherently slow growing plants and simultaneously illustrates the profound resource limitations that epiphytes are subjected to in the canopy of a seasonal rain forest. From habitat conditions it seems that size-related differences in water availability are the driving force behind the observed size-dependent ecophysiological changes: the larger an epiphyte grows the more independent it is with regard to precipitation patterns. In conclusion, the results strongly emphasize the need to treat plant size as an important source of intraspecific variability and thus urge researchers to consider plant size in the design of ecophysiological experiments with vascular epiphytes.
Mit den Experimenten dieser Arbeit konnte erstmals gezeigt werden, dass ein Phytohormon wie Abscisinsäure mit dem "Solvent-drag" des Wasserflusses apoplastisch durch den Wurzelzellwandbereich in die Xylemgefäße transportiert werden kann. Es konnte ein Bypass-Fluss für ABA durch den gesamten Zellwandapoplasten, auch durch lipophile Barrieren wie Exo- und Endodermis nachgewiesen werden. Dies ist durch die speziellen Moleküleigenschaften von Abscisinsäure möglich: (i) der geringe Durchmesser des Moleküls (8 - 11 nm) und (ii) die hohe Lipophilie von ABA bei schwach sauren pH-Werte. Mit einer Penetration apoplastischer Barrieren ist demnach zu rechnen. Weiterhin wurde gezeigt, dass die Ausbildung solcher lipophilen Zellwandnetze einen signifikanten Einfluss auf den apoplastischen ABA-Transport besitzt. Die Ausbildung einer Exodermis in Mais, wie sie unter natürlichen Bedingungen zu beobachten ist, konnte den ABA-Fluss in das Xylem um die Faktoren 2 bis 4 reduzieren. Da gleichzeitig eine Verminderung der hydraulischen Wurzelleitfähigkeit um denselben Betrag auftrat, blieb das Wurzel-Spross-ABA-Signal, die Phytohormonkonzentration, im Xylem gleich. Die zu den Stomata geleitete Information über den Wasserzustand der Wurzel änderte sich also nicht. Im natürlichen System ist sogar eine Verstärkung des Signals zu erwarten, da eine Exodermis nicht als Aufnahme-Barriere für gewebeproduzierte ABA wirkt. Gleichzeitig verringert sie den Verlust von apoplastischer ABA an die Rhizosphäre. Außerdem wird der Wasserverlust aus dem Gewebe durch eine Exodermis signifikant reduziert wird. Somit sind solche Wurzeln gut an die Bedingungen eines eintrocknenden Bodens angepasst. Apoplastische Barrieren sind demnach, neben membran-lokalisierten Tranportern, wichtige Parameter für die Beurteilung von Wurzeltransporteigenschaften für Wasser und darin gelöste Substanzen. Der Beitrag der apoplastischen Komponente zum Gesamt-ABA-Transport ist abhängig von der untersuchten Pflanzenart, der aktuellen Transpirations- oder Wasserflussrate und von Umwelteinflüssen wie erhöhter ABA-Konzentration im Wurzelgewebe (z.B. durch Trockenstress), pH-Wert der Rhizosphäre und den Ernährungsbedingungen der Pflanze. Erhöhter radialer Wasserfluss, erhöhte ABA-Wurzelgewebegehalte und niedriger pH-Wert der Rhizosphäre verstärken den apoplastischen Bypass-Fluss unter physiologischen Bedingungen. Geringe Wassertransportraten, niedrige ABA-Konzentrationen im Gewebe, alkalische pH-Werte der Rhizosphäre und Ammoniumernährung verstärken dagegen den symplastischen Beitrag zum ABA-Transport. In der vorliegenden Arbeit konnten die sich widersprechenden Theorien bezüglich des ABA-Effektes auf die hydraulische Leitfähigkeit von Wurzeln erklärt werden. ABA erhöht über einen Zeitraum von 2 Stunden die Zellleitfähigkeit (Lp) mit einem Maximum 1 Stunde nach ABA-Inkubation. Dies wirkt sich in einem verstärktem Lpr von intakten Wurzelsystemen aus, das einem ähnlichen Zeitmuster folgt. Pflanzen sind demnach in der Lage, mittels ABA den zellulären Wassertransportweg reversibel zu optimieren, um so unter mildem Trockenstress, wie er in einem gerade eintrocknenden Boden auftritt, die Pflanze mit ausreichend Wasser zu versorgen. Tritt ein länger andauernder Wassermangel ein, versperrt die Pflanze diesen Weg wieder. Dieser transiente Effekt erklärt auch die aus der Literatur bekannten stimulierenden und inhibierenden ABA-Wirkungen. Durch den verstärkten Wasserfluss zu Beginn der Stresssituation erzeugt ABA auf diese Weise ein sich selbst verstärkendes, wurzelbürtiges Hormonsignal in den Spross. Das Blatt erreicht in effektiver Weise eine ABA-Menge, die ausreichend ist, um die Stomata zu schließen. Es folgt eine Reduktion der Transpiration. Eine weiter andauernde Erhöhung des symplastischen Wassertransportweges wäre ohne physiologische Bedeutung. Regulierende Membranstrukturen für diesen Vorgang könnten ABA-sensitive Wasserkanäle (Aquaporine) der Plasmamembran sein. Es wurde gezeigt, dass der Rezeptor für diesen Vorgang innerhalb von corticalen Maiswurzelzellen lokalisiert und hochspezifisch für (+)-cis-trans-ABA ist. Die Signaltransduktion für diesen Kurzzeiteffekt erfolgt nicht mittels verstärkter Aquaporintranskription, könnte aber über ABA-induzierte Aktivierung (Phosphorylierung), oder Einbau von Aquaporinen in die Zellmembran ablaufen. Der Abscisinsäure-Transport ist ein komplexer Vorgang. Er wird beeinflusst durch Umwelteinflüsse, Wurzelanatomie, ist gekoppelt mit dem Wasserfluss und durch sich selbst variierbar. Herkömmliche Vorstellungen einer simplen Hormondiffusion können diesen regulierbaren Vorgang nicht mehr beschreiben. Pflanzen besitzen ein ABA-Transportsystem, das schnell, effektiv und an sich verändernde Umweltbedingungen adaptierbar ist.
Die Methodik und Technik der Gaswechselmessung von Pflanzen wurde für den Modellorganismus Arabidopsis thaliana optimiert und für Untersuchungen zur Beteiligung des Aquaporins PIP1b an Wassertransportvorgängen während der Stomaöffnung verwendet. Die Messungen der Transpirationsraten von PIP1b-Antisense-Pflanzen ergaben keine Hinweise auf Veränderungen des zeitlichen Verlaufs der Stomaöffnung. Die Wasserpermeabilitäten von Schließzell-Plasmamembranen scheinen somit nicht von der Expression des Aquaporins PIP1b beeinflußt zu sein. - Gaswechselmessungen an Nicotiana tabacum NtAQP1-Antisense-Pflanzen zeigten eine Verringerung der Transpirationsraten im Licht und eine geringere Grundtranspiration im Dunkeln. Dies deutet auf eine Beteiligung von NtAQP1 am Wassertransport hin. - Ausgewählte Arabidopsis thaliana-Mutanten wurden hinsichtlich ihrer stomatären Antwort auf Rot- und Rot-/Blaulicht-Bestrahlung analysiert. Hierfür wurde ein Doppelbestrahlungs-Protokoll entwickelt. Vergleiche mit den Wildtypen ergaben signifikante Unterschiede bei der Phytochrom-Mutante phyA-103, der Abscisinsäure-Mutante aba3-2 und der Auxin-resistenten Mutante axr1-3. Ferner zeigte die Mutante npq1-2 nicht die beschriebene Abweichung der stomatären Antwort auf Blaulicht. - Die Expressionsmuster eines PIP1b-GFP-Reportergens in transgenen Arabidopsis thaliana-Pflanzen wurden analysiert. Hohe Promotor-Aktivitäten konnten in meristematischen Bereichen von Wurzel und Sproß, in Elementen der Leitbündel, in jungen Kotyledonen und in Staubblättern beobachtet werden. Es zeigte sich eine enge Korrelation zwischen PIP1b-Promotoraktivität und Streckungswachstum. - Eine Sequenzanalyse des NtAQP1-Promotors ergab Übereinstimmungen mit spezifischen Bindungsmotiven von MYB-ähnlichen Transkriptionsfaktoren. Mit Promotor-Reportergenen konnte die Beteiligung eines dieser Sequenzmotive an der GA- und ABA-induzierten Aktivierung des NtAQP1-Promotors gezeigt werden. Zur Analyse der Phytohormon-Wirkungen auf deletierte Promotorbereiche wurde ein duales Vektorsystem entwickelt und bei der transienten Transformation von BY2-Protoplasten eingesetzt. - Die Expression eines GFP::NtAQP1-Fusionsgens in BY2-Zellen zeigte die subzelluläre Lokalisation des Aquaporins in der Zytoplasmamembran. Ferner wurde Fusionsprotein in Vesikel-ähnlichen Strukturen beobachtet.
