IRAK2 besitzt eine Schlüsselrolle im Signalweg des TLR4. Fehlregulationen dieses Signalwegs führen zu fehlgeleiteten Immunreaktionen, die auch die Entstehung und Progression von Krebserkrankungen fördern. Bevor IRAK2 als therapeutisches Ziel in Frage kommen kann, muss erst noch weitere Klarheit über die grundsätzliche Funktionsweise dieses Proteins bestehen. So ist für IRAK2 aufgrund der Substitution einer Aminosäure in der Kinase-Domäne im Vergleich zu IRAK1 noch nicht abschließend geklärt, ob es sich um eine aktive Kinase oder eine Pseudokinase handelt und ob diese Veränderung eine Erhöhung oder eine Erniedrigung der Funktion im TLR4-Signalweg nach sich zieht.
Um diese Fragen anzugehen, wurde in dieser Arbeit Asparagin im vermeintlich aktiven Zentrum (Aminosäure 333) wieder zur Asparaginsäure [N333D] revertiert und damit versucht die Phosphorylierungsaktivität zu steigern bzw. vergleichbar zu IRAK1 wiederherzustellen. Das Einbringen der Mutation in IRAK2 erfolgte mittels ortsspezifischer Mutagenese. Mit dieser und anderen Mutanten und mit wildtypischem IRAK2 wurden durch die CRISPR/Cas9-Methode generierte IRAK2-defiziente 264.7 Makrophagen rekonstituiert und damit ein System etabliert, mit dem der Einfluss der Mutation auf den Signalweg des TLR4 nach Stimulation mit LPS quantitativ analysiert werden konnte. Sowohl die indirekte NF-κB-Messung über CD40-Expression als auch die direkte NF-κB-Messung über die NF-κB-getriebene Expression eines Reportergens (cyan fluorescent protein) ergab, dass IRAK2[N333D] die LPS-abhängige NF-κB-Aktivierung über den TLR4 Signalweg schlechter ermöglicht als IRAK2.
Insgesamt deuten die Ergebnisse darauf hin, dass die in der Entwicklungsgeschichte aufgetretene Veränderung des aktiven Zentrums von IRAK2 im Vergleich zu IRAK1 zu einer besseren Aktivierung der MyD88-abhängigen NF-κB-Aktivität führte und somit eine erhöhte und länger anhaltende Signalleitung ermöglichte. Diese Erkenntnis kann als weiterer Schritt hin zu einem besseren Verständnis der Funktion des IRAK2-Proteins und zu einer möglichen zukünftigen Verwendung von IRAK2 als Ziel therapeutischer Behandlungen gesehen werden.
Die Bcl-2-Familienmitglieder A1 und sein humanes Homolog Bfl-1 gewährleisten das Überleben der Zelle. Gleichzeitig trägt eine Dysregulation der Expression von A1/ Bfl-1 zur Krebsentstehung bei. Die Stabilität von A1/ Bfl-1 wird durch deren Ubiquitinylierung sowie die anschließende proteosomale Degradation gesteuert. Mit Hilfe eines Yeast-Two-Hybrid-Screens wurde die E3-Ubiquitinligase HectD1 als potentieller Interaktionspartner von A1/ Bfl-1 identifiziert. Die Interaktion von A1 und HectD1 des Yeast-Two-Hybrid-Screens konnte in Säugerzellen bestätigt werden. Desweiteren konnte gezeigt werden, dass lediglich 87 Aminosäuren für eine Interaktion von HectD1 und A1 nötig sind. Da membrangebundenes HectD1 zu einer Translokation von zytosolischem A1 an die Zellmembran führt, kann man davon ausgehen, dass beide Proteine auch in vivo miteinander interagieren. Eine dominant negative HectD1-Mutante schließlich beeinflusst die Ubiqutinylierung von A1 und führt somit zu dessen Stabilisierung. Diese Daten legen nahe, dass HectD1 ein wichtiger negativer Regulator von A1/ Bfl-1 ist und dass HectD1 für die Regulierung der A1/ Bfl-1-Proteinmenge in (Krebs)zellen sehr wichtig ist.
Herstellung monoklonaler Antikörper gegen das von Aspergillus fumigatus produzierte Gift Gliotoxin

Diese Arbeit befasst sich mit der Herstellung monoklonaler Antikörper gegen Gliotoxin und eine Charakterisierung der Eigenschaften dieser Antikörper sowie ihrer Fab-Fragmente im ELISA sowie in Zellkulturen. Insgesamt konnten fünf monoklonale Antikörper generiert werden, die spezifisch für das Mykotoxin Gliotoxin waren.
Die subakut sklerosierende Panenzephalitis ist eine demyelinisierende Erkrankung des ZNS. Ätiologisch liegt eine persistierende Infektion von Masernviren der Neuronen bzw. Gliazellen zugrunde. Bis heute gibt es keine spezifische Therapie und lediglich symptomatische Therapiemaßnahmen werden in deren Behandlung angewandt. Obwohl sich während des Krankheitsverlaufes eine ausgeprägte Immunantwort des natürlichen als auch adaptiven Immunsystems des Wirtes abspielt führt die Erkrankung unweigerlich zum Tode der Betroffenen. Eine Therapiemöglichkeit diesbezüglich könnten die Effekte der RNAi darstellen. Um geeignete Sequenzen für einen möglichen Genknockdown einzelner MV-Gene herauszufiltern, wurde ein plasmid-basierendes Testsystem entwickelt, das die mRNA des Fluoreszenzproteins DsRed2 zusammen mit mRNA-Sequenzen einzelner MV-Gene exprimiert. Gegen diese MV-Gene wiederum wurden verschiedene shRNAs ausgewählt, welche mithilfe eines lentiviralen Vektorsystems exprimiert werden. Als Expressionsindikator findet in diesem lentiviralen Vektorsystem simultan die Expression von eGFP als statt. Die Auswertung dieses Dual-Color-Systems erfolgte im Folgenden mittels FACS-Analysen. Die wirksamsten siRNA-Sequenzen, ausgewählt durch eine hohe Abnahme der Rotfluoreszenz in den untersuchten Zellen, wurden für weitere Versuche selektiert. Die auf diese Weise ausgewählten shRNA-Sequenzen wurden in einem weiteren Schritt durch die Generierung lentiviraler Partikel in persistierend infizierten NT2-Zellen (piNT2), die das Fluoreszenzprotein HcRed als Infektionsindikator exprimieren, transduziert. Weitere durchflusszytometrische Messungen zeigten eine äußerst hohe Reduktion viraler Replikation innerhalb von 7 Tagen bis unterhalb der Detektionsgrenze sofern die shRNAs gegen die Expression der MV-Proteine N, P und L gerichtet waren. Die Anzahl der virus-negativen Zellen blieb im Folgenden bis zu 3 Wochen nach stattgehabter Transduktion konstant, sofern das fusion-inhibiting-peptide den Zellkulturen hinzugegeben wurde. Die transduzierten piNT2-Zellen, welche keine Expression von HcRed zeigten wurden auf Zellrasen bestehend aus Vero-Zellen aufgetragen und führten im zeitlichen Verlauf zu keiner Fusion bzw. Synzytienbildung. Dieser Zusammenhang legt nahe, dass die transduzierten piNT2-Zellen die Fähigkeit einer Expression viraler Proteine verloren haben und eine „Heilung“ stattfinden kann, wenn die virale Proteinbiosynthese durch eine permanente Expression von siRNAs, wie beispielsweise durch lentivirale Vektorsysteme, gehemmt wird.
To circumvent time-consuming clinical trials, testing whether existing drugs are effective inhibitors of SARS-CoV-2, has led to the discovery of Remdesivir. We decided to follow this path and screened approved medications "off-label" against SARS-CoV-2. Fluoxetine inhibited SARS-CoV-2 at a concentration of 0.8 mu g/ml significantly in these screenings, and the EC50 was determined with 387 ng/ml. Furthermore, Fluoxetine reduced viral infectivity in precision-cut human lung slices showing its activity in relevant human tissue targeted in severe infections. Fluoxetine treatment resulted in a decrease in viral protein expression. Fluoxetine is a racemate consisting of both stereoisomers, while the S-form is the dominant serotonin reuptake inhibitor. We found that both isomers show similar activity on the virus, indicating that the R-form might specifically be used for SARS-CoV-2 treatment. Fluoxetine inhibited neither Rabies virus, human respiratory syncytial virus replication nor the Human Herpesvirus 8 or Herpes simplex virus type 1 gene expression, indicating that it acts virus-specific. Moreover, since it is known that Fluoxetine inhibits cytokine release, we see the role of Fluoxetine in the treatment of SARS-CoV-2 infected patients of risk groups.
Zytotoxische T-Zellen (CD8+) spielen eine wichtige Rolle bei retroviralen Infektionen des Menschen, wie HIV und HTLV. Ihre molekulare Wirkweise war bisher aber nicht bekannt. Die Infektion von Mäusen mit dem retroviralen Friend Virus Komplex (FV) wurde daher als Modell verwendet, um die antiviralen Mechanismen gegen Retroviren aufzuklären. Als erstens wurde die Beteiligung von CD8+ T-Zellen bei der Kontrolle der FV Infektion in Depletion-Experimenten gezeigt. In akut FV infizierten Tieren konnten außerdem aktivierte Virus-spezifische CD8+ T-Effektorzellen nachgewiesen werden, welche zytotoxische Moleküle produzierten. Die Infektion von „knockout“ Mäusen ergab, dass die CD8+ T-Zellen hauptsächlich den Exozytoseweg benutzten, um FV-infizierte Zellen zu eliminieren. Die Anwesenheit eines der drei am Exozytoseweg beteiligten zytotoxischen Moleküle Perforin, Granzym A oder Granzym B war in der akuten Phase der Infektion ausreichend, um die Virusreplikation zu kontrollieren. Diese Ergebnisse weisen auf neue molekulare Mechanismen von Granzymen bei der Virusabwehr hin. In Gegensatz zur Abwehr des pathogenen FV Komplex benutzten CD8+ T-Zellen nicht den Exozytoseweg zur Abwehr von apathogenen Retroviren. Eine Infektion von Mäusen mit dem nicht-pathogenen F-MuLV induzierte keine Produktion von zytotoxischen Molekülen in CD8+ T-Zellen. Die Infektion von „knockout“ Mäusen zeigte, dass F-MuLV von CD8+ T-Zellen über den Fas/FasL Weg kontrolliert wurde. Das pathogene Potential von Retroviren scheint also einen Einfluß auf den jeweils verwendeten Abwehrmechanismus von zytotoxischen T-Zellen zu haben. Trotz der wichtigen Funktion von CD8+ T-Zellen bei der Kontrolle der akuten FV Infektion, spielten diese Zellen bei der persistierenden Infektion keine Rolle mehr. Die Untersuchungen von aktivierten CD8+ T-Zellen aus persistierend infizierten Mäusen zeigte, dass die Zellen agranulär waren und keine zytotoxischen Moleküle produzierten. Folglich hatten sie auch keine zytotoxische Aktivität in vitro. Der Exozytoseweg von Virus-spezifischen CD8+ T-Zellen war also in der persistierenden FV Infektion gestört. So entwickelten viele persistierend infizierte Tiere nach einer Reinfektion eine Splenomegalie, was zusätzlich beweist, dass die Funktion von Virus-spezifischen T-Zellen auch in vivo gestört war. Mäuse, die chronisch mit FV infiziert waren, wiesen also einen kompletten Funktionsverlust von CD8+ T-Zellen auf, der offensichtlich eine wichtige Voraussetzung für die Persistenz des Virus darstellt. Da die CD8+ T-Zellen nicht mehr funktionell waren, mussten vermutlich CD4+ T-Zellen über den Fas/FasL-Weg die Kontrolle über die chronische Infektion übernehmen. Das Verständnis der molekularen Mechanismen der CD8+ T-Zell Abwehr gegen Retroviren ermöglicht es neue Strategien der Immuntherapie gegen retroviralen Infektionen zu entwickeln.
In multizellulären Organismen ist die Apoptose, eine Art des programmierten Zelltods, ein hoch spezifischer, natürlicher Prozess um unerwünschte, überschüssige oder beschädigte Zellen auf einem geordneten, nicht entzündlichen, Weg zu beseitigen. Auch im menschlichen Körper werden täglich Milliarden an Zellen durch Apoptose abgebaut. Dadurch kann der Organismus die Zellzahl und Gewebegröße regulieren. Eine Fehlregulation der Apoptose kann weitreichende Konsequenzen haben. Während es bei einer unzureichenden Apoptose zu Autoimmunität und Tumoren kommen kann, führt ein gesteigerter Zelltod zu Immunschwäche oder akuten und chronischen degenerativen Erkrankungen. Aus diesem Grund ist die Erforschung der genauen Regulation des programmierten Zelltods von großer Bedeutung. Die Mitochondrien spielen eine Schlüsselrolle bei der Regulation des programmierten Zelltods. Während des Ablaufs der Apoptose kommt es zur Freisetzung von Mediatoren der Apoptose aus diesen Organellen. Diese Moleküle sind dann an der Aktivierung von Caspasen beteiligt, welche die Degradation von Proteinen und als Folge davon der DNA bewerkstelligen. Die Proteine der Bcl-2 Familie, zu der pro- und anti-apoptotische Mitglieder gehören, kontrollieren die Integrität der Mitochondrien hauptsächlich durch Protein-Protein und Protein-Membran Interaktionen. Viele anti-apoptotische Mitglieder der Bcl-2 Familie sind mittels einer C-terminalen Transmembrandomäne in der Lage an die äußere Mitochondrienmembran zu binden. A1, ein anti-apoptotisches Mitglied dieser Proteinfamilie, besitzt allerdings keine typische Transmembrandomäne. Die Funktion und Lokalisation des Proteins werden sehr kontrovers diskutiert. Daher ist das Ziel dieser Arbeit, die Lokalisation und Funktion von A1 zu analysieren. Um ein umfassendes Bild zu bekommen, wandten wir gentechnische Methoden zur Modifizierung des A1 C-Terminus an. Die intrazelluläre Lokalisation des Proteins wurde mittels konfokaler Mikroskopie untersucht. Die Bedeutung des proteasomalen Abbaus von A1 wurde schließlich durch Inhibitionsexperimente charakterisiert. Durch eine Fusion des C-terminalen Endes von A1 mit dem „enhanced green fluorescent protein“ haben wir die Funktion und Lokalisierungseigenschaften des A1 C-terminus mittels Durchflusszytometrie und konfokaler Mikroskopie untersucht. Wir konnten zeigen, dass das C-terminale Ende das so entstandene chimäre Protein deutlich destabilisieren konnte. Eine Blockade des proteasomalen Abbaus führte zu einer Stabilisierung des Fusionsproteins. Des Weiteren konnte in der konfokalen Mikroskopie eine diffuse Verteilung des chimären Proteins in 293T Zellen nachgewiesen werden. Diese Ergebnisse weisen darauf hin, dass das C-terminale Ende von A1 keine Lokalisation an spezifische intrazelluläre Organellen vermitteln kann, jedoch für die Instabilität und den proteasomalen Abbau des Proteins verantwortlich ist. Eine Analyse der Lokalisation des Gesamtproteins A1 in 293T Zellen mittels konfokaler Mikroskopie konnte eine Verteilung von A1 zugunsten des Zytoplasmas mit Anreicherung an den Mitochondrien nachweisen. Obwohl das C-terminale Ende von A1 für sich keine spezifische intrazelluläre Lokalisation vermittelt, zeigte das Protein eine Kolokalisation an den Mitochondrien. Somit scheinen noch andere N-terminale Bereiche des Proteins an der Lokalisation von A1 beteiligt zu sein. Zusammenfassend konnte mit dieser Arbeit gezeigt werden, dass der C-Terminus von A1 eine wichtige Rolle für die Stabilität des anti-apoptotischen Bcl-2 Familienmitglieds spielt.
The transcriptional repressor-Blimp-1 terminates differentiation of B lymphocytes as well as myeloid cells. Our data show that Blimp-1 is highly expressed in freshly isolated murine primary T lymphocytes, particularly its minor splice variant. Ectopic expression of Blimp-1 by retroviral transduction neither dramatically altered secretion of IFN-ã or IL-4 nor did it induce the ability to suppress as regulatory T cells. However, induction of Blimp-1 resulted in not only a significant reduction in the production of IL-2 but also an inability to proliferate as well as in the reduced viability. These results demonstrate that Blimp-1 might mark end stages of lineage differentiation in T cells.
Background
Herpesviruses can infect a wide range of animal species. Herpes simplex virus 1 (HSV-1) is one of the eight herpesviruses that can infect humans and is prevalent worldwide. Herpesviruses have evolved multiple ways to adapt the infected cells to their needs, but knowledge about these transcriptional and post-transcriptional modifications is sparse.
Results
Here, we show that HSV-1 induces the expression of about 1000 antisense transcripts from the human host cell genome. A subset of these is also activated by the closely related varicella zoster virus. Antisense transcripts originate either at gene promoters or within the gene body, and they show different susceptibility to the inhibition of early and immediate early viral gene expression. Overexpression of the major viral transcription factor ICP4 is sufficient to turn on a subset of antisense transcripts. Histone marks around transcription start sites of HSV-1-induced and constitutively transcribed antisense transcripts are highly similar, indicating that the genetic loci are already poised to transcribe these novel RNAs. Furthermore, an antisense transcript overlapping with the BBC3 gene (also known as PUMA) transcriptionally silences this potent inducer of apoptosis in cis.
Conclusions
We show for the first time that a virus induces widespread antisense transcription of the host cell genome. We provide evidence that HSV-1 uses this to downregulate a strong inducer of apoptosis. Our findings open new perspectives on global and specific alterations of host cell transcription by viruses.
Im Vergleich zu anderen Retroviren, zeichnen sich Foamyviren durch eine Reihe von Eigenschaften aus, die sie besonders attraktiv für die Vektorentwicklung und somatische Gentherapie machen. Foamyviren exprimieren ihr Pol Prekursorprotein unabhängig von Gag, d.h. von ihrer eigenen gespleisten mRNA. Zwar ist der genaue Pol-Verpackungsmechanismus von Foamyviren noch nicht vollständig aufgeklärt, frühere Studien zeigten jedoch, dass die prägenomische RNA essentiell für die Pol-Enkapsidierung ist. Zwei Pol-Verpackungssequenzen (PES) wurden identifiziert, welche sich in den cis-aktiven Sequenzen (CAS) der prägenomischen RNA befinden (Heinkelein et al., 1998; Peters et al., 2005). In dieser Arbeit wurde untersucht, ob die PESI und PESII Sequenzen alleine ausreichend für die Pol-Verpackung sind. Zusätzich wurde der Einfluss von verschiedenen Teilen der ca. 2000 nt langen CASII Sequenz auf den Vektortransfer ohne Verlust der Pol-Enkapsidierung untersucht. Es konnte gezeigt werden, dass PESI und PESII alleine nicht ausreichend für die Pol-Verpackung ins foamyvirale Partikel sind. Die Verkürzung des CASII Elements zeigte keinen Effekt auf die Pol-Verpackung und den Vektortransfer. Das Einfügen eines zusätzlichen zentralen Polypurintraktes führte jedoch zur signifikanten Erhöhung der Transduktionseffizienz von FV Vektoren. Diese Ergebnisse führten zur Entwicklung eines neuen foamyviralen Vektors (pTW01), der ca. 850nt kürzer ist als die früher etablierten FV Vektoren, aber immer noch die gleiche Transduktionseffizienz auf Fibroblasten und humanen Stammzellen zeigt. Dieser Vektor mit einer höheren Verpackungskapazität und Sicherheit, eignet sich hervorragend für den Einsatz in gentherapeutischen Studien. Zusätzlich konnte gezeigt werden, dass eine heterologe Verpackung zwischen zwei unterschiedlichen Foamyviren (PFV und SFVmac) zu einem geringen Prozentsatz stattfindet. Als erster Schritt in der Entwickung eines neues Systems für eine einfache und kostengünstige Vektorvirusproduktion wurde gezeigt, dass die Expression der foamyviralen Gag, Pol und Env Proteine in Saccharomyces cerevisiae stattfinden kann.
