Zwei Arten helikal-chiraler Verbindungen mit einem oder zwei Boratomen wurden nach einem modularen Ansatz synthetisiert. Die Bildung der helikalen Strukturen erfolgte durch Einführung von Bor in flexible Biaryl- bzw. Triaryl-Vorstufen, hergestellt aus kleinen achiralen Bausteinen. Die durchgehend ortho-fusionierten Azabora[7]helicene zeichnen sich dabei durch außergewöhnliche Konfigurationsstabilität, blaue oder grüne Fluoreszenz in Lösung mit Quantenausbeuten (Φ\(_{fl}\)) von 18–24 %, grüne oder gelbe Emission im Festkörper (Φ\(_{fl}\) bis zu 23 %) und starke chiroptische Resonanz mit großen Anisotropiefaktoren von bis zu 1.12×10\(^{-2}\) aus. Azabora[9]helicene, aufgebaut aus winkelförmig sowie linear angeordneten Ringen, sind blaue Emitter mit Φ\(_{fl}\) von bis zu 47 % in CH\(_{2}\)Cl\(_{2}\) und 25 % im Festkörper. DFT-Rechnungen zeigen, dass ihre P-M-Interkonversion über einen komplexeren Weg verläuft als im Fall von H1. Röntgenstrukturanalyse von Einkristallen zeigt deutliche Unterschiede in der Packungsanordnung von Methyl- und Phenylderivaten auf. Die Moleküle werden als Primärstrukturen verlängerter Helices vorgeschlagen.
The scope of this work is to develop a novel single-molecule imaging technique by combining atomic force microscopy (AFM) and optical fluorescence microscopy. The technique is used for characterizing the structural properties of multi-protein complexes. The high-resolution fluorescence microscopy and AFM are combined (FIONA-AFM) to allow for the identification of individual proteins in such complexes. This is achieved by labeling single proteins with fluorescent dyes and determining the positions of these fluorophores with high precision in an optical image. The same area of the sample is subsequently scanned by AFM. Finally, the two images are aligned and the positions of the fluorophores are displayed on top of the topographical data. Using quantum dots as fiducial markers in addition to fluorescently labeled proteins, fluorescence and AFM information can be aligned with an accuracy better than 10 nm, which is sufficient to identify single fluorescently labeled proteins in most multi-protein complexes. The limitations of localization precision and accuracy in fluorescence and AFM images are investigated, including their effects on the overall registration accuracy of FIONA-AFM hybrid images. This combination of the two complementary techniques opens a wide spectrum of possible applications to the study of protein interactions, because AFM can yield high resolution (5–10 nm) information about the conformational properties of multi-protein complexes while the fluorescence can indicate spatial relationships of the proteins within the complexes. Additionally, computer simulations are performed in order to validate the accuracy of the registration algorithm.
In this thesis the syntheses and detailed investigations on two foldable PBI systems were presented. The reversible, solvent-dependet folding/unfolding-behavior was used to study the ground and excited states properties of folda-dimer and folda-trimer by means of different spectroscopic methods as well as theoretical studies. The switching between charge transfer or excimer formation pathways of photoexcited molecules influenced by the spatial arrangement of chromophores within defined dye systems illustrates the impact of conformational preferences on functional properties.
We introduce fluorescence-detected pump–probe microscopy by combining a wavelength-tunable ultrafast laser with a confocal scanning fluorescence microscope, enabling access to the femtosecond time scale on the micrometer spatial scale. In addition, we obtain spectral information from Fourier transformation over excitation pulse-pair time delays. We demonstrate this new approach on a model system of a terrylene bisimide (TBI) dye embedded in a PMMA matrix and acquire the linear excitation spectrum as well as time-dependent pump–probe spectra simultaneously. We then push the technique towards single TBI molecules and analyze the statistical distribution of their excitation spectra. Furthermore, we demonstrate the ultrafast transient evolution of several individual molecules, highlighting their different behavior in contrast to the ensemble due to their individual local environment. By correlating the linear and nonlinear spectra, we assess the effect of the molecular environment on the excited-state energy.
Ziel der Dissertation „Oktapeptide als neue Organokatalysatoren zur Hydrolyse von Phosphaten und Estern“ war es neue Oktapeptide zu finden, die die Fähigkeit besitzen Phosphate und Ester zu Hydrolysieren. In der Natur ist bei den Katalysereaktionen die Sekundärstruktur von entscheidender Bedeutung. Aus diesem Grunde wurde im Rahmen dieser Arbeit zunächst ein Modellsystem entwickelt, mit dem gezeigt werden konnte, dass es möglich ist mit einfachen Bausteinen wie Diaminobutan und dem in der Gruppe von Schmuck entwickelten Guanidiniocarbonylpyrrol ein Molekül zu entwickeln, welches eine stabile intramolekulare Schleife in polaren Lösungsmitteln ausbildet. Aufgrund der geringen Größe des Moleküls, konnte kein β-Faltblatt gebildet werden. Dennoch konnte mit Hilfe der NMR-Spektroskopie gezeigt werden, dass in dem polaren Lösungsmittel Methanol eine stabile Schleifenbildung mit einer intramolekularen Komplexierung stattfindet. Nach erfolgreicher Synthese des oben beschriebenen Testsystems wurde als nächster Schritt eine kombinatorische Organokatalysatorbibliothek mit 625 Mitgliedern aufgebaut. Die Struktur der Peptide kann man in drei Teile untergliedern. Der erste Teil ist die feste Phase, das Amino-TentaGel, an das nacheinander die einzelnen Aminosäuren gekuppelt wurden. An Stelle der Butylgruppe als Schleifenelement wurde Aib-D-Pro als β-Turn Element eingesetzt. Den dritten Teil bilden die bei der Synthese der Oktapeptide eingesetzten Aminosäuren AA1, AA3, AA6, AA8, die ein β-Faltblatt ausbilden sollten. Die kombinatorische Synthese der Bibliothek erfolgte nach der „Split and Mix“ Methode. Zum Unterscheiden der einzelnen Mitglieder untereinander, wurde die feste Phase zusammen mit einem Radiofrequenzchip in IRORI MikroKans gegeben. Durch die unterschiedlichen Aminosäuren ist die Bibliothek für die Katalyse in polaren Lösungsmitteln wie Wasser konzipiert worden. Als exemplarische Katalysereaktionen wurden dabei zwei Hydrolysereaktionen (Phosphatspaltung und Esterspaltung) ausgesucht. Zunächst wurde für beide Reaktionen ein Screening mit unterschiedlichen Bedingungen durchgeführt. Dabei hat sich gezeigt, dass bei der Phosphatspaltung nur dann die Reaktion katalysiert wurde, wenn das künstliche Argininanalogon, welches in unserer Arbeitsgruppe synthetisiert wurde, vorhanden war. Der beste Katalysator hat die Reaktion 175-mal schneller katalysiert als die unkatalysierte Reaktion. Bei dem Screening der Esterspaltung hat sich herausgestellt, dass die Aminosäure Histidin essentiell für die katalytische Aktivität ist. Der beste Katalysator bei der Esterspaltung hat die Hydrolyse des Esters 345-mal schneller katalysiert als die unkatalysierte Reaktion. Bei beiden Reaktionen hat sich gezeigt, dass die Sequenz der Katalysatoren sehr wichtig für die Katalyse ist. So verringert z.B. bei der Esterhydrolyse der Austausch zweier Aminosäuren eine Verringerung der Aktivität von dem Faktor 294 auf den Beschleunigungsfaktor 35. Auch konnten beide Katalysereaktionen in wässrigem gepufferten Lösung durchgeführt werden. Damit ist es möglich gewesen neue Oktapeptide für die Katalyse von Phosphat- und Esterspaltung zu finden und diese erfolgreich im Screening als auch in Lösung zu untersuchen.
Complexation properties of 2,2':6',2''-terpyridine (tpy) have been studied with a series of first row transition metal ions by UV-vis, 1H NMR and isothermal titration calorimetry and ƒ´H values for the tpy complexation processes have been determined. These studies reveal that Zn2+ is the best suited metal ion for the reversible coordination of the terpyridine ligand. Thus, supramolecular coordination polymerization of perylene bisimide fluorophores containing terpyridine functionalities have been investigated by using Zn2+ as metal ion. The formation of the dimeric complexes in the case of monotopic model comounds and coordination polymerization of ditopic functional building blocks have been confirmed by 1H NMR studies. The optical properties of dimeric and polymeric complexes have been investigated by UV-vis and fluorescence spectroscopy. The Zn2+ coordination to the terpyridine unit does not effect the advantageous fluorescence properties of perylene bisimide moieties. The reversibility of the formation of coordination polymers has been established by 1H NMR and additionally by DOSY NMR and fluorescence anisotropy measurements. Coordination polymer strands can be visualized by atomic force microscopy (AFM), which also reveals the formation of an ordered monolayer film at higher concentration. The average polymer length has been determined by AFM to 15 repeat units, which correlates well with the value estimated by 1H NMR to >10 repeat units.
Einführung: Die physikalischen Eigenschaften des intravenösen Anästhetikums Propofol (2,6-Diisopropylphenol) erlauben dessen fluoreszenzspektrometrische Detektion. Zur Entwicklung eines direkten Online-Monitorings im optisch dichten Medium Blut wird das Signalverhalten von Propofol im mit Blutprodukten gefüllten Kreislaufsystem untersucht. Material und Methoden: Der kontinuierliche Umsatz von 140,2 ml Probenvolumen im Kreislaufmodell mit integrierter Durchflussquarzküvette bezweckt ein stabiles Fluoreszenzniveau des Messmediums, da Blutprodukte in statischer Versuchsanordnung unter der verwendeten Anregungsstrahlung (UV-C) starken photochemischen Bleichungseffekten ausgesetzt sind. Als Messmedien untersucht werden Gefrorenes Frischplasma (GFP), eine Suspension aus Erythrozytenkonzentrat und GFP (EK + GFP) sowie am Versuchstag gespendetes heparinisiertes Vollblut. Es erfolgt die standardisierte Injektionen von vier Propofolboli, durch die im System Konzentrationen von 35,7 μg/ml bis 3,6 μg/ml entstehen und den klinisch relevanten Wirkspiegeln bei Narkoseeinleitung sowie Narkoseaufrechterhaltung entsprechen. Unter Anregung mit Licht der Wellenlänge 274 nm liefert Propofol ein maximales Signal bei 300 nm. Anhand der in engen zeitlichen Abständen aufgenommen Fluoreszenzspektren werden die Propofoleffekte bei 300 nm im Summationsspektrum des Blut-Propofol-Gemischs analysiert. Ergebnisse: Die Signalanstiege bei 300 nm nach Injektion in das mit GFP bzw. EK + GFP gefüllte Kreislaufsystem sind hochsignifikant für die erzeugten Propofolspiegel von 35,7 μg/ml bis 3,6 μg/ml und weisen eine sehr gute lineare Korrelation von R2 = 0,73 bis zu R2 = 0,99 zwischen Fluoreszenzsignal und Propofolkonzentration auf. Allein für diese Messmedien kann durch den Einsatz des Kreislaufmodells ein ausreichend stabiles Fluoreszenzsignal zum Propofolnachweis erreicht werden. Dem Fluoreszenzanstieg nach Propofolinjektion folgt in allen Messmedien ein über 30 Minuten andauernder Signalabfall, für den nach fluoreszenzspektrometrischer Untersuchung von Schlauchproben des Kreislaufmodells die Adsorption des lipophilen Anästhetikums an Silikon als ein ursächlicher Faktor bestimmt werden kann. Schlussfolgerung: Der direkte konzentrationsabhängige Fluoreszenznachweis von klinisch eingesetzten Propofol-Wirkspiegeln gelingt allein in transfusionsmedizinisch aufbreiteten Blutprodukten.
This thesis will outline studies performed on the fluorescence dynamics of phenyl-benzo-
[c]-tetrazolo-cinnolium chloride (PTC) in alcoholic solutions with varying viscosity using
time-resolved fluoro-spectroscopic methods. Furthermore, the properties of femtosecond
Laguerre-Gaussian (LG) laser pulses will be investigated with respect to their temporal
and spatial features and an approach will be developed to measure and control the spatial
intensity distribution on the time scale of the pulse.
Tetrazolium salts are widely used in biological assays for their low oxidation and reduction
thresholds and spectroscopic properties. However, a neglected feature in these applications
is the advantage that detection of emitted light has over the determination of the
absorbance. To corroborate this, PTC as one of the few known fluorescent tetrazolium
salts was investigated with regard to its luminescent features. Steady-state spectroscopy
revealed how PTC can be formed by a photoreaction from 2,3,5-triphenyl-tetrazolium
chloride (TTC) and how the fluorescence quantum yield behaved in alcoholic solvents
with different viscosity. In the same array of solvents time correlated single photon counting
(TCSPC) measurements were performed and the fluorescence decay was investigated.
Global analysis of the results revealed different dynamics in the different solvents, but
although the main emission constant did change with the solvent, taking the fluorescence
quantum yield into consideration resulted in an independence of the radiative rate from
the solvent. The non-radiative rate, however, was highly solvent dependent and responsible
for the observed solvent-related changes in the fluorescence dynamics. Further studies
with the increased time resolution of femtosecond fluorescence upconversion revealed an
independence of the main emission constant from the excitation energy, however the dynamics
of the cooling processes prior to emission were prolonged for higher excitation
energy. This led to a conceivable photoreaction scheme with one emissive state with a
competing non-radiative relaxation channel, that may involve an intermediate state.
LG laser beams and their properties have seen a lot of scientific attention over the past two
decades. Also in the context of new techniques pushing the limit of technology further to
explore new phenomena, it is essential to understand the features of this beam class and
check the consistency of the findings with theoretical knowledge. The mode conversion
of a Hermite-Gaussian (HG) mode into a LG mode with the help of a spiral phase plate
(SPP) was investigated with respect to its space-time characteristics. It was found that
femtosecond LG and HG pulses of a given temporal duration share the same spectrum
and can be characterized using the same well-established methods. The mode conversion
proved to only produce the desired LG mode with its characteristic orbital angular momentum
(OAM), that is conserved after frequency doubling the pulse. Furthermore, it
was demonstrated that temporal shaping of the HG pulse does not alter the result of its
mode-conversion, as three completely different temporal pulse shapes produced the same
LG mode. Further attention was given to the sum frequency generation of fs LG beams
and dynamics of the interference of a HG and a LG pulse. It was found that if both are
chirped with inverse signs the spatial intensity distribution does rotate around the beam
axis on the time scale of the pulse. A strategy was found that would enable a measurement
of these dynamics by upconversion of the interference with a third gate pulse. The results
of which are discussed theoretically and an approach of an experimental realization had
been made. The simulated findings had only been reproduced to a limited extend due to
experimental limitations, especially the interferometric stability of the setup.
The thesis enhances the strategy of non-destructive fluorescence read-out in rylene bisimide-diarylethene containing photochromic systems. The fluorescence of the emitter unit is quenched by a photoinduced electron transfer only to one of the isomeric forms of the photochrome. The driving force of the fuorescence-quenching electron transfer was calculated by the help of the Rehm-Weller equation. The novel photochromic systems satisfy the necessary requirements for non-destructive read-out in write/read/erase fluorescent memory devices.
