Background
Fabry disease is an inborn lysosomal storage disorder which is associated with small fiber neuropathy. We set out to investigate small fiber conduction in Fabry patients using pain-related evoked potentials (PREP).
Methods
In this case–control study we prospectively studied 76 consecutive Fabry patients for electrical small fiber conduction in correlation with small fiber function and morphology. Data were compared with healthy controls using non-parametric statistical tests. All patients underwent neurological examination and were investigated with pain and depression questionnaires. Small fiber function (quantitative sensory testing, QST), morphology (skin punch biopsy), and electrical conduction (PREP) were assessed and correlated. Patients were stratified for gender and disease severity as reflected by renal function.
Results
All Fabry patients (31 men, 45 women) had small fiber neuropathy. Men with Fabry disease showed impaired cold (p < 0.01) and warm perception (p < 0.05), while women did not differ from controls. Intraepidermal nerve fiber density (IENFD) was reduced at the lower leg (p < 0.001) and the back (p < 0.05) mainly of men with impaired renal function. When investigating A-delta fiber conduction with PREP, men but not women with Fabry disease had lower amplitudes upon stimulation at face (p < 0.01), hands (p < 0.05), and feet (p < 0.01) compared to controls. PREP amplitudes further decreased with advance in disease severity. PREP amplitudes and warm (p < 0.05) and cold detection thresholds (p < 0.01) at the feet correlated positively in male patients.
Conclusion
Small fiber conduction is impaired in men with Fabry disease and worsens with advanced disease severity. PREP are well-suited to measure A-delta fiber conduction.
Die Bcl-2-Familienmitglieder A1 und sein humanes Homolog Bfl-1 gewährleisten das Überleben der Zelle. Gleichzeitig trägt eine Dysregulation der Expression von A1/ Bfl-1 zur Krebsentstehung bei. Die Stabilität von A1/ Bfl-1 wird durch deren Ubiquitinylierung sowie die anschließende proteosomale Degradation gesteuert. Mit Hilfe eines Yeast-Two-Hybrid-Screens wurde die E3-Ubiquitinligase HectD1 als potentieller Interaktionspartner von A1/ Bfl-1 identifiziert. Die Interaktion von A1 und HectD1 des Yeast-Two-Hybrid-Screens konnte in Säugerzellen bestätigt werden. Desweiteren konnte gezeigt werden, dass lediglich 87 Aminosäuren für eine Interaktion von HectD1 und A1 nötig sind. Da membrangebundenes HectD1 zu einer Translokation von zytosolischem A1 an die Zellmembran führt, kann man davon ausgehen, dass beide Proteine auch in vivo miteinander interagieren. Eine dominant negative HectD1-Mutante schließlich beeinflusst die Ubiqutinylierung von A1 und führt somit zu dessen Stabilisierung. Diese Daten legen nahe, dass HectD1 ein wichtiger negativer Regulator von A1/ Bfl-1 ist und dass HectD1 für die Regulierung der A1/ Bfl-1-Proteinmenge in (Krebs)zellen sehr wichtig ist.
Profiling the Cross Reactivity of Ubiquitin with the Nedd8 Activating Enzyme by Phage Display

The C-terminal peptides of ubiquitin (UB) and UB-like proteins (UBLs) play a key role in their recognition by the specific activating enzymes (E1s) to launch their transfer through the respective enzymatic cascades thus modifying cellular proteins. UB and Nedd8, a UBL regulating the activity of cullin-RING UB ligases, only differ by one residue at their C-termini; yet each has its specific E1 for the activation reaction. It has been reported recently that UAE can cross react with Nedd8 to enable its passage through the UB transfer cascade for protein neddylation. To elucidate differences in UB recognition by UAE and NAE, we carried out phage selection of a UB library with randomized C-terminal sequences based on the catalytic formation of UB similar to NAE thioester conjugates. Our results confirmed the previous finding that residue 72 of UB plays a "gate-keeping" role in E1 selectivity. We also found that diverse sequences flanking residue 72 at the UB C-terminus can be accommodated by NAE for activation. Furthermore heptameric peptides derived from the C-terminal sequences of UB variants selected for NAE activation can function as mimics of Nedd8 to form thioester conjugates with NAE and the downstream E2 enzyme Ubc12 in the Nedd8 transfer cascade. Once the peptides are charged onto the cascade enzymes, the full-length Nedd8 protein is effectively blocked from passing through the cascade for the critical modification of cullin. We have thus identified a new class of inhibitors of protein neddylation based on the profiles of the UB C-terminal sequences recognized by NAE.
Functional magnetic resonance imaging (fMRI) has become a powerful and influential method to non-invasively study neuronal brain activity. For this purpose, the blood oxygenation level-dependent (BOLD) effect is most widely used. T2* weighted echo planar imaging (EPI) is BOLD sensitive and the prevailing fMRI acquisition technique. Here, we present an alternative to its standard Cartesian recordings, i.e. k-space density weighted EPI, which is expected to increase the signal-to-noise ratio in fMRI data. Based on in vitro and in vivo pilot measurements, we show that fMRI by k-space density weighted EPI is feasible and that this new acquisition technique in fact boosted spatial and temporal SNR as well as the detection of local fMRI activations. Spatial resolution, spatial response function and echo time were identical for density weighted and conventional Cartesian EPI. The signal-to-noise ratio gain of density weighting can improve activation detection and has the potential to further increase the sensitivity of fMRI investigations.
Das Signal-zu-Rausch-Verhältnis (SNR) stellt bei modernen Bildgebungstechniken in der Magnetresonanz-Tomographie heutzutage oftmals die entscheidende Limitation dar. Eine Verbesserung durch Modifikation der Hardware ist kostspielig und führt meistens zu einer Verstärkung anderer Probleme, wie zum Beispiel erhöhte Energiedeposition ins Gewebe. Im Gegensatz dazu ist Dichtegewichtung eine Methode, die eine SNR-Erhöhung durch Modifikation der Aufnahmetechnik ermöglicht. In der MR-Bildgebung erfolgt oftmals eine retrospektive Filterung des aufgenommenen Signalverlaufs, beispielsweise zur Artefaktreduktion. Damit einhergehend findet eine Veränderung der Modulationstransferfunktion (MTF) bzw. ihrer Fouriertransformierten, der räumlichen Antwortfunktion (SRF), statt. Optimales SNR wird nach dem Matched Filter-Theorem erzielt, wenn die nachträgliche Filterung dem aufgenommenen Signalverlauf proportional ist. Dies steht dem Ziel der Artefaktreduktion entgegen. Bei Dichtegewichtung steht durch nicht-kartesische Abtastung des k-Raums mit der k-Raum-Dichte ein zusätzlicher Freiheitsgrad zur Verfügung. Dieser ermöglicht es, im Falle eines konstanten Signalverlaufs eine gewünschte MTF ohne Filterung zu erreichen. Bei veränderlichem Signalverlauf kann ein SNR Matched Filter angewendet werden, dessen negative Einflüsse auf die MTF durch Dichtegewichtung kompensiert werden. Somit ermöglicht Dichtegewichtung eine vorgegebene MTF und gleichzeitig ein optimales SNR. In der vorliegenden Arbeit wurde Dichtegewichtung erstmals bei den schnellen Multi-Echo-Sequenzen Turbo-Spin-Echo und Echoplanar-Bildgebung (EPI) angewendet. Im Gegensatz zu bisherigen Implementierungen muss hier der Signalabfall durch T2- bzw. T2*-Relaxation berücksichtigt werden. Dies führt dazu, dass eine prospektiv berechnete dichtegewichtete Verteilung nur bei einer Relaxationszeit optimal ist. Bei Geweben mit abweichenden Relaxationszeiten können sich wie auch bei den kartesischen Varianten dieser Sequenzen Änderungen an SRF und SNR ergeben. Bei dichtegewichteter Turbo-Spin-Echo-Bildgebung des Gehirns konnte mit den gewählten Sequenzparametern ein SNR-Vorteil von 43 % gegenüber der kartesischen Variante erzielt werden. Die Akquisition wurde dabei auf die T2-Relaxationszeit von weißer Substanz optimiert. Da die meisten Gewebe im Gehirn eine ähnliche Relaxationszeit aufweisen, blieb der visuelle Gesamteindruck identisch zur kartesischen Bildgebung. Der SNR-Gewinn konnte in der dichtegewichteten Implementierung zur Messzeithalbierung genutzt werden. Dichtegewichtete EPI weist eine hohe Anfälligkeit für geometrische Verzerrungen, welche durch Inhomogenitäten des Hauptmagnetfeldes verursacht werden, auf. Die Verzerrungen konnten erfolgreich mit einer Conjugate Phase-Methode korrigiert werden. Dazu muss die räumliche Verteilung der Feldinhomogenitäten bekannt sein. Dazu ist zusätzlich zur eigentlichen EPI-Aufnahme die zeitaufwendige Aufnahme einer sogenannten Fieldmap erforderlich. Im Rahmen dieser Arbeit konnte eine Methode entwickelt werden, welche die zur Erlangung einer Fieldmap notwendige Aufnahmedauer auf wenige Sekunden reduziert. Bei dieser Art der Fieldmap-Aufnahme müssen jedoch durch Atmung hervorgerufene Effekte auf die Bildphase berücksichtigt werden. Die Fieldmap-Genauigkeit kann durch Aufnahme unter Atempause, Mittelung oder retrospektiver Phasenkorrektur erhöht werden. Für die gewählten EPI-Sequenzparameter wurde mit Dichtegewichtung gegenüber der kartesischen Variante ein SNR-Gewinn von 14 % erzielt. Anhand einer funktionellen MRT (fMRI)-Fingertapping-Studie konnte demonstriert werden, dass die SNR-Steigerung auch zu einer signifikant erhöhten Aktivierungsdetektion in Teilen der Hirnareale führt, die bei der Fingerbewegung involviert sind. Die Verwendung von zusätzlicher EPI-Phasenkorrektur und iterativer Optimierung der dichtegewichteten k-Raum-Abtastung führt zu weiteren Verbesserungen der dichtegewichteten Bildgebung mit Multi-Echo-Sequenzen.
In this thesis we study various aspects of chaos synchronization of time-delayed coupled chaotic maps. A network of identical nonlinear units interacting by time-delayed couplings can synchronize to a common chaotic trajectory. Even for large delay times the system can completely synchronize without any time shift. In the first part we study chaotic systems with multiple time delays that range over several orders of magnitude. We show that these time scales emerge in the Lyapunov spectrum: Different parts of the spectrum scale with the different delays. We define various types of chaos depending on the scaling of the maximum exponent. The type of chaos determines the synchronization ability of coupled networks. This is, in particular, relevant for the synchronization properties of networks of networks where time delays within a subnetwork are shorter than the corresponding time delays between the different subnetworks. If the maximum Lyapunov exponent scales with the short intra-network delay, only the elements within a subnetwork can synchronize. If, however, the maximum Lyapunov exponent scales with the long inter-network connection, complete synchronization of all elements is possible. The results are illustrated analytically for Bernoulli maps and numerically for tent maps. In the second part the attractor dimension at the transition to complete chaos synchronization is investigated. In particular, we determine the Kaplan-Yorke dimension from the spectrum of Lyapunov exponents for iterated maps. We argue that the Kaplan-Yorke dimension must be discontinuous at the transition and compare it to the correlation dimension. For a system of Bernoulli maps we indeed find a jump in the correlation dimension. The magnitude of the discontinuity in the Kaplan-Yorke dimension is calculated for networks of Bernoulli units as a function of the network size. Furthermore the scaling of the Kaplan-Yorke dimension as well as of the Kolmogorov entropy with system size and time delay is investigated. Finally, we study the change in the attractor dimension for systems with parameter mismatch. In the third and last part the linear response of synchronized chaotic systems to small external perturbations is studied. The distribution of the distances from the synchronization manifold, i.e., the deviations between two synchronized chaotic units due to external perturbations on the transmitted signal, is used as a measure of the linear response. It is calculated numerically and, for some special cases, analytically. Depending on the model parameters this distribution has power law tails in the region of synchronization leading to diverging moments. The linear response is also quantified by means of the bit error rate of a transmitted binary message which perturbs the synchronized system. The bit error rate is given by an integral over the distribution of distances and is studied numerically for Bernoulli, tent and logistic maps. It displays a complex nonmonotonic behavior in the region of synchronization. For special cases the distribution of distances has a fractal structure leading to a devil's staircase for the bit error rate as a function of coupling strength. The response to small harmonic perturbations shows resonances related to coupling and feedback delay times. A bi-directionally coupled chain of three units can completely filter out the perturbation. Thus the second moment and the bit error rate become zero.
Hintergrund: Die Schlafbezogene Atmungsstörung (SBAS) ist eine häufige Komorbidität der systolischen Herzinsuffizienz und mit einer kürzeren Lebenserwartung assoziiert. Es steht derzeit eine Vielzahl einfacher ambulanter SBAS-Screening-Geräte zur Verfügung. Keines davon wurde jedoch bisher für Patienten mit chronischer systolischer Herzinsuffizienz validiert.
Fragestellung: Die vorliegende Untersuchung diente der Prüfung eines einfachen ambulanten SBAS-Screenings hinsichtlich diagnostischer Trennschärfe und prognostischer Relevanz für Patienten mit systolischer Herzinsuffizienz.
Methoden: Bei Patienten mit symptomatischer systolischer Herzinsuffizienz (linksventrikuläre Ejektionsfraktion (LVEF) < 45%, NYHA ≥ II) wurden nächtlicher Atemfluss, Pulsfrequenz und Sauerstoffsättigung mit dem ApneaLinkTM (Fa. ResMed) ambulant aufgezeichnet. Hieraus werden der Apnoe-/Hypopnoe-Index (AHI) sowie der Entsättigungsindex (ODI) berechnet. Die Ergebnisse wurden der Diagnose des Schlafmediziners anhand einer PSG als gültigem Goldstandard der SBAS-Diagnostik gegenübergestellt. Der Überlebensstatus wurde bei allen Patienten durch ambulante bzw. telefonische Nachuntersuchungen erfasst. Mittels ROC-Analysen wurden diagnostische und durch Cox-Regressionen prognostische Schwellenwerte des SBAS-Screenings ermittelt.
Ergebnisse: Insgesamt wurden 131 Patienten eingeschlossen: das mittlere Alter lag bei 68±13 Jahren, 110 Patienten (84%) waren männlich, 53 Patienten (41%) hatten ein NYHA-Stadium ≥3, die mittlere LVEF lag bei 34%. Bei 69 Patienten (53%) konnte eine PSG durchgeführt werden, welche bei 55 Patienten (80%) eine SBAS diagnostizierte. Bei 38 Patienten (55%) lag eine mind. mittelgradige, therapiepflichtige SBAS vor. In ROC-Analysen für ein mind. mittelgradige SBAS ergaben sich AUCs von 0.77; 0.82; 0.81; 0,79 und 0,82 für AHI; ODI; (AHI+ODI)/2; (2xAHI+ODI)/3 und (AHI+2xODI)/3. Die größtmögliche Spezifität (0,90) und Sensitivität (0,66) für die Diagnose einer Therapiepflichtigen SBAS lab bei einem cut-off von (AHI+2xODI)/3 ≥ 21/h. Die mediane Follow-up Zeit der Studie lag bei 23 (18; 27) Monaten; es starben 21 Patienten (16%). Es fand sich eine positive Korrelation zwischen (AHI+2xODI)/3 und Mortalitätsrate. Bereits für den (AHI+2xODI)/3 ≥9/h war die Mortalität signifikant erhöht (3,55 (95%CI: 1.04-12.13) p=0,04).
Schlussfolgerung: Eine therapiepflichtige SBAS lässt durch ein ambulantes Screening zuverlässig diagnostizieren. Dabei ist der ODI dem AHI überlegen. Für die bestmögliche diagnostische Trennschärfe empfehlen wir einen (AHI+2xODI)/3 von 21/h heranzuziehen, wobei bereits ein Wert von 9/h prognostisch relevant ist, so dass bei Patienten mit einem (AHI+2xODI)/3 von 9-20/h zwar auf eine schlafmedizinische Untersuchung verzichtet werden kann, eine engmaschige kardiologische Kontrolle inklusive wiederholten Screenings jedoch unabdingbar bleibt.
Die vorliegende Dissertation behandelt zum einen die mechanistischen Details von Bindungsaktivierungs-Reaktionen an Disauerstoff und weißem Phosphor mit den Komplexfragmenten 1[Ni(iPr2Im)2] und 3[(η5-C5H5)Co(iPr2Im)] und zum anderen die Regioselektivität von oxidativen Insertionsreaktionen des 1[Ni(iPr2Im)2]-Komplexfragments in C–X-Bindungen substituierter Fluoraromaten (X = F, OCH3, CN, H).
1. Since the early nineteenth century describing (and understanding) patterns of distribution of biodiversity across the Earth has represented one of the most significant intellectual challenges to ecologists and biogeographers. Among the most striking patterns of species richness are: the latitudinal and elevational gradients, with peaks in number of species at low latitudes and somewhere at mid altitudes, although other patterns, e.g. declines with increasing elevation, are often observed. Even in highly diverse tropical regions, species richness is not evenly distributed but there are “hotspots” of biodiversity where an exceptional number of species, especially endemics, are concentrated. Unfortunately, such areas are also experiencing dramatic loss of habitat. Among vertebrate taxa, amphibians are facing the most alarming number of extinctions. Habitat destruction, pollution and emergence of infectious diseases such as chytridiomycosis, are causing worldwide population declines. Responses to these drivers can be multidirectional and subtle, i.e. they may not be captured at the species but at the genetic level. Moreover, present patterns of diversity can result from the influence of past geological, climatic and environmental changes.
In this study, I used a multidisciplinary and multilevel approach to understand how and to which extent the landscape influences amphibian diversity. Mount Kilimanjaro is an exceptional tropical region where the landscape is rapidly evolving due to land use changes; additionally, there is a broad lack of knowledge of its amphibian fauna. During two rainy seasons in 2011, I recorded anurans from the foothills to 3500 m altitude; in addition, I focused on two river frog species and collected tissue samples for genetic analysis and swabs for detection of chytridiomycosis, the deadly disease caused by Batrachochytrium dendrobatidis (Bd).
2. I analyzed how species richness and composition change with increasing elevation and anthropogenic disturbance. In order to disentangle the observed patterns of species diversity and distribution, I incorporated inferences from historical biogeography and compared the assemblage of Mt. Kilimanjaro and Mt. Meru (both recent volcanoes) with those of the older Eastern Arc Mountains. Species richness decreased with elevation and locally increased in presence of water bodies, but I did not detect effects of either anthropogenic disturbance or vegetation structure on species richness and composition. Moreover, I found a surprisingly low number of forest species. Historical events seem to underlie the current pattern of species distribution; the young age of Mt. Kilimanjaro and the complex biogeographic processes which occurred in East Africa during the last 20 million years prevented montane forest frogs from colonizing the volcano.
3. I focused on the genetic level of biodiversity and investigated how the landscape, i.e. elevation, topographic relief and land cover, influence genetic variation, population structure and gene flow of two ecologically similar and closely related river frog species, namely Amietia angolensis and Amietia wittei. I detected greater genetic differentiation among populations in the highland species (A. wittei) and higher genetic variation in the lowland species (A. angolensis), although genetic diversity was not significantly correlated with elevation. Importantly, human settlements seemed to restrict gene flow in A. angolensis, whereas steep slopes were positively correlated with gene flow in A. wittei. This results show that even ecologically similar species can respond differently to landscape processes and that the spatial configuration of topographic features combined with species-specific biological attributes can affect dispersal and gene flow in disparate ways.
4. River frogs of the genus Amietia seem to be particularly susceptible to chytridiomycosis, showing the highest pathogen load in Kenya and other African countries. In the last study, I collected swab samples from larvae of A. angolensis and A. wittei for Bd detection. Both species resulted Bd-positive. The presence of Bd on Mt. Kilimanjaro has serious implication. For instance, Bd can be transported by footwear of hikers from contaminated water and soil. Tourists visiting Mt. Kilimanjaro may translocate Bd zoospores to other areas such as the nearby Eastern Arc Mts. where endemic and vulnerable species may still be naïve to the fungus and thus suffer of population declines.
5. My study significantly contributed to the knowledge of the amphibian fauna of Mt. Kilimanjaro and of East Africa in general, and it represents a valuable tool for future conservation actions and measures. Finally, it highlights the importance of using a multidisciplinary (i.e. community ecology, historical biogeography, landscape genetics, disease ecology) and multilevel (i.e. community, species, population, gene) approach to disentangle patterns of biodiversity.
Human growth has an estimated heritability of about 80%-90%. Nevertheless, the underlying cause of shortness of stature remains unknown in the majority of individuals. Genome-wide association studies (GWAS) showed that both common single nucleotide polymorphisms and copy number variants (CNVs) contribute to height variation under a polygenic model, although explaining only a small fraction of overall genetic variability in the general population. Under the hypothesis that severe forms of growth retardation might also be caused by major gene effects, we searched for rare CNVs in 200 families, 92 sporadic and 108 familial, with idiopathic short stature compared to 820 control individuals. Although similar in number, patients had overall significantly larger CNVs \((p-value <1 x 10^{-7})\). In a gene-based analysis of all non-polymorphic CNVs >50 kb for gene function, tissue expression, and murine knock-out phenotypes, we identified 10 duplications and 10 deletions ranging in size from 109 kb to 14 Mb, of which 7 were de novo (p < 0.03) and 13 inherited from the likewise affected parent but absent in controls. Patients with these likely disease causing 20 CNVs were smaller than the remaining group (p < 0.01). Eleven (55%) of these CNVs either overlapped with known microaberration syndromes associated with short stature or contained GWAS loci for height. Haploinsufficiency (HI) score and further expression profiling suggested dosage sensitivity of major growth-related genes at these loci. Overall 10% of patients carried a disease-causing CNV indicating that, like in neurodevelopmental disorders, rare CNVs are a frequent cause of severe growth retardation.
