Ziel dieser Arbeit war es, prognoserelevante Faktoren für das kolorektale Karzinom anhand des eigenen Patientengutes zu untersuchen. Insbesondere sollte die prognostische Relevanz der Tumormarker CEA, CA 19-9 und p53 in Bezug auf tumorbedingtes Überleben und rezidivfreie Zeit analysiert werden, um anhand ihrer Ausprägung die Prognose eines Patienten nach primär chirurgischer Therapie besser definieren zu können. Dementsprechend sollte ein möglichst adäquates therapeutisches Vorgehen gewährleistet sein. Bezüglich des tumorbedingten Überlebens wurde anhand des Kaplan-Meier Verfahrens nachgewiesen, dass sowohl das UICC-Stadium als auch die Darmwandinfiltration (T-Status), Lymphknotenbefall (N-Status) und CEA im Serum als hochsignifikante Parameter dienen, wohingegen eine signifikante Einflussnahme für das Geschlecht, Alter, Tumorlokalisation, Grading, CA 19-9 und p53 (Serummessung) auf das Überleben nicht festgestellt wurde. Bei näherem Betrachten der einzelnen UICC-Stadien in Abhängigkeit von CEA war dieser der aussagekräfitgste prognostische Tumormarker (Serummessung, cut-off point >= 5 ng/ml) für Patienten in höherem UICC-Stadium (UICC III). Bei weiterer Betrachtung der einzelnen Subgruppen des UICC III-Stadiums war CEA insbesondere für Patienten in UICC IIIA am relevantesten. Bezüglich der beiden anderen Tumormarker CA 19-9 und p53 konnte mittels der Serummessungen keine Korrelation mit dem UICC-Stadium bzw. auch keine signifikante Einflussnahme auf das tumorbedingte Überleben festgestellt werden. Für die rezidivfreie Zeit zeigten UICC-Stadium, T-Status, N-Status, CEA (Serummessung) und Tumorlokalisation (Kolon/Rektum) eine signifikante Einflussnahme. Tumoren des Rektums hatten ein höheres Risikoprofil als die des Kolons. Die immunhistologische und molekulargenetische Analyse bezüglich aller drei Tumormarker bestätigte das UICC Stadium III als ein Risikostadium. Dabei lies sich entgegen der Serummessungen auch für CA 19-9 und p53 eine stadienabhängige Risiko-Korrelation darstellen. Des Weiteren korrelierte das Auftreten p53 spezifischer IgG Antikörper stark mit der p53 Proteinexpression im Gewebe, was die Annahme eines Zusammenhangs zwischen intrazellulärer Ansammlung von p53 in Tumorzellen und einer humoralen Antwort auf p53 stützt. Mit Ausnahme von CEA, zeigten CA 19-9 und p53 für sich alleine keine Korrelation zwischen erhöhten Serumwerten und/oder der Expression im Tumor und dem postoperativen Auftreten von Rezidiven. Dahingegen wurde erstmals in dieser Arbeit festgestellt, dass Patienten, die mindestens drei erhöhte Parameter (CEA, CA 19-9 und p53) im Serum und/oder im Tumor (Protein- oder Genexpression) hatten, eine höhere Rezidivrate (Lokal-, Metastasen) während der Tumornachsorge (36±6,6 Monate) zeigten; dies unabhängig von ihrem UICC Stadium (UICC I-III). Speziell diese Risikogruppe könnte von einer adjuvanten Therapie profitieren.
Macroautophagy (hereafter referred to as autophagy) is a homeostatic process that preserves cellular integrity. In mice, autophagy regulates pancreatic ductal adenocarcinoma (PDAC) development in a manner dependent on the status of the tumor suppressor gene Trp53. Studies published so far have investigated the impact of autophagy blockage in tumors arising from Trp53-hemizygous or -homozygous tissue. In contrast, in human PDACs the tumor suppressor gene TP53 is mutated rather than allelically lost, and TP53 mutants retain pathobiological functions that differ from complete allelic loss. In order to better represent the patient situation, we have investigated PDAC development in a well-characterized genetically engineered mouse model (GEMM) of PDAC with mutant Trp53 (Trp53\(^{R172H}\)) and deletion of the essential autophagy gene Atg7. Autophagy blockage reduced PDAC incidence but had no impact on survival time in the subset of animals that formed a tumor. In the absence of Atg7, non-tumor-bearing mice reached a similar age as animals with malignant disease. However, the architecture of autophagy-deficient, tumor-free pancreata was effaced, normal acinar tissue was largely replaced with low-grade pancreatic intraepithelial neoplasias (PanINs) and insulin expressing islet β-cells were reduced. Our data add further complexity to the interplay between Atg7 inhibition and Trp53 status in tumorigenesis.
Die DNA-Durchflusszytometrie ist eine anerkannte Methode zur Erfassung genetischer Abweichungen und Änderungen der Proliferationsfraktion von Tumorzellen. Die Aufarbeitung von Tumorzellen mit vorhandenen Begleitpopulationen bewirkt allerdings bei der Einfachfärbung der DNA eine Ungenauigkeit der Zellzyklusanalyse. In dieser Arbeit wurde der Einfluss der Begleitpopulationen auf die Analyse von Ploidie und Zellzyklus mit Hilfe der DNA-Zytokeratin-Doppelfärbung am Mammakarzinom untersucht. Zum anderen wird auf Zusammenhänge zwischen p53, Proliferation und Ploidie von Mammakarzinomen eingegangen. Von 28 untersuchten Fällen waren 26 für die DNA-Zytokeratin-Doppelfärbung komplett auswertbar. Bei 9 dieser 26 ausgewerteten Fälle konnte immunhistochemisch p53 nachgewiesen werden. Die Zellen wurden aus Gefriermaterial mechanisch vereinzelt und mit Propidium Jodid und FITC-konjugiertem anti-Zytokeratin-Antikörper bzw. anti-p53-Antikörper gefärbt. Die Messungen wurden mit einem FACScan durchgeführt. Die Auswertung erfolgte mit Hilfe der Multicycle Software AV. Die Auswertung der Zytokeratin-Doppelfärbung ergab im Vergleich zur Einfachfärbung einen Anstieg sowohl des Anteiles aneuploider Tumoren als auch des mittleren DNA-Index. Die Proliferationsrate, insbesondere die S-Phasen-Fraktion erhöhte sich ebenfalls signifikant. Die vergleichenden Zellzyklusanalysen der Zytokeratin-positiven und der p53-positiven Tumorzellen zeigte eine geringere Proliferationsfraktion der p53-positiven Zellen, verglichen mit den p53-negativen Tumorzellen. Der überwiegende Anteil der p53-positiven Zellen zeigte eine Aneuploidie. Weiterhin ergaben sich in dieser Untersuchung Hinweise auf eine Zellzyklusabhängige Expression des p53-Proteins. Durch die Nutzung der Doppelfärbung für DNA und Zytokeratin in der Durchflusszytometrie gelingt die exaktere Wiedergabe der Biologie von Tumoren. Es ist zu erwarten, dass dies die prognostische Aussagekraft der DNA-flowzytometrischen Parameter Ploidie und Proliferationsfraktion erhöht.
Background
Most tumor cells show aberrantly activated Akt which leads to increased cell survival and resistance to cancer radiotherapy. Therefore, targeting Akt can be a promising strategy for radiosensitization. Here, we explore the impact of the Akt inhibitor MK-2206 alone and in combination with the dual PI3K and mTOR inhibitor PI-103 on the radiation sensitivity of glioblastoma cells. In addition, we examine migration of drug-treated cells.
Methods
Using single-cell tracking and wound healing migration tests, colony-forming assay, Western blotting, flow cytometry and electrorotation we examined the effects of MK-2206 and PI-103 and/or irradiation on the migration, radiation sensitivity, expression of several marker proteins, DNA damage, cell cycle progression and the plasma membrane properties in two glioblastoma (DK-MG and SNB19) cell lines, previously shown to differ markedly in their migratory behavior and response to PI3K/mTOR inhibition.
Results
We found that MK-2206 strongly reduces the migration of DK-MG but only moderately reduces the migration of SNB19 cells. Surprisingly, MK-2206 did not cause radiosensitization, but even increased colony-forming ability after irradiation. Moreover, MK-2206 did not enhance the radiosensitizing effect of PI-103. The results appear to contradict the strong depletion of p-Akt in MK-2206-treated cells. Possible reasons for the radioresistance of MK-2206-treated cells could be unaltered or in case of SNB19 cells even increased levels of p-mTOR and p-S6, as compared to the reduced expression of these proteins in PI-103-treated samples. We also found that MK-2206 did not enhance IR-induced DNA damage, neither did it cause cell cycle distortion, nor apoptosis nor excessive autophagy.
Conclusions
Our study provides proof that MK-2206 can effectively inhibit the expression of Akt in two glioblastoma cell lines. However, due to an aberrant activation of mTOR in response to Akt inhibition in PTEN mutated cells, the therapeutic window needs to be carefully defined, or a combination of Akt and mTOR inhibitors should be considered.
Trotz der Fortschritte in der Therapie des Multiplen Myeloms und des stetig wachsenden Arsenals effektiver anti-MM-Medikamente muss ein Teil der Patienten mit bestimmten zytogenetischen Veränderungen der Tumorzellen nach wie vor der Hochrisiko-Gruppe zugeordnet werden und hat eine Lebens-erwartung von nur wenigen Jahren. Einer der ungünstigsten prognostischen Marker ist die Inaktivierung des Tumorsuppressorgens TP53 durch Mutationen des Gens oder Deletionen des kurzen Arms von Chromosom 17, del(17p). Diese wird häufig mit einer Chemoresistenz der entarteten Plasmazellen in Verbindung gebracht.
In der vorliegenden Arbeit gelang es mittels des CRISPR/Cas9-Systems TP53-Läsionen zu erzeugen und isogene Klone der TP53wt/wt Zelllinie AMO-1 zu generieren. Diese wurden anhand der Sequenzanalysen von beiden TP53-Allelen den Gruppen der biallelisch TP53-inaktivierten, der monoallelisch TP53-inaktivierten und der TP53wt/wt Klone zugeordnet. Das gruppenspezifische Verhalten der Klone aller drei Gruppen hinsichtlich deren Expression von p53, p21 und Mdm2 unterstrich die Validität des etablierten Zelllinienmodells zur Untersuchung der Bedeutung von TP53-Läsionen beim Multiplen Myelom. Neben einer kompletten Ausschaltung durch biallelische TP53-Inaktivierung zeigten die Ergebnisse der vorliegenden Arbeit auch eine Haploinsuffizienz des p53-Systems. Diese äußerte sich in einer Abschwächung der Nutlin-3A-abhängigen p53-, p21- und Mdm2-Induktion bereits nach Inaktivierung eines TP53-Allels durch Frameshift-Mutation. Korrelierend zu dem Proteinexpressions¬muster konnte eine zunehmende Resistenzentwicklung der Klone je nach Grad der TP53-Inaktivierung (mono- bzw. biallelisch) gegen Nutlin 3A sowie genotoxische Substanzen nachgewiesen werden, während die Sensibilität der MM-Zellen gegen Proteasominhibitoren unbeeinträchtigt blieb. Einschränkungen hinsichtlich der Übertragbarkeit der Ergebnisse der vorliegenden Arbeit auf das Multiple Myelom im Allgemeinen bestehen in dem Umstand, dass die beschriebenen Beobachtungen lediglich an einer einzigen MM-Zelllinie gemacht werden konnten. Dies ist durch die geringe Auswahl an TP53wt/wt MM-Zelllinien, die zudem noch oft eine schlechte Transfektabilität und niedrige Zellteilungsrate nach Einzelzellselektion aufweisen, bedingt.
Die an der Zelllinie AMO-1 gemachten Beobachtungen stehen in Einklang mit der in klinischen Studien festgestellten Verkürzung des progressionsfreien- (PFS) und Gesamt-Überlebens (OS) bei MM-Patienten mit TP53-Alterationen. Die zunehmende Chemoresistenz der malignen Plasmazellen nach mono- bzw. biallelischer TP53-Inaktivierung kann als Grund für die Akkumulation entsprechender Klone im Rezidiv und in fortgeschrittenen Krankheitsstadien des MM angesehen werden. Mittels möglichst umfassender Erfassung des genauen TP53-Läsions-Status in zukünftigen klinischen Studien zu multiplen Zeitpunkten des Krankheitsverlaufs könnte der Einfluss verschiedener, in der Therapie des MM zum Einsatz kommender Substanzen auf die Selektion bzw. die Unterdrückung besonders virulenter Subklone mit TP53-Läsionen untersucht werden.
Precise control of progression through mitosis is essential to maintain genomic stability and to prevent aneuploidy. The DREAM complex is an important regulator of mitotic gene expression. Depletion of Lin9, one core-subunit of DREAM, leads to reduced expression of G2/M genes and impaired proliferation. In conditional mouse knockout cells (MEFs) Lin9 deletion causes defects in mitosis and cytokinesis and cells undergo premature senescence in order to prevent further proliferation. In this work it could be shown that the senescence phenotype in Lin9 knockout MEFs is independently mediated by the two tumor suppressor pathways p53-p21 and p16-pRB. Studies using the conditional Lin9 knockout mouse model demonstrated an important function of Lin9 in the regulation of mitotic gene expression and proliferation in vivo. Deletion of Lin9 caused reduced proliferation in the intestinal crypts resulting in atrophy of the intestinal epithelium and in rapid death of the animals. In the second part of this work, the pathways leading to p53 mediated G1 arrest after failed cytokinesis were analyzed by using a chemical inhibitor of the mitotic kinase Aurora B. In a high throughput siRNA screen the MAP kinase MAP3K4 was identified as an upstream activator of p53. It could be shown that MAP3K4 activates the downstream stress kinase p38b to induce the p53 mediated cell cycle arrest of tetraploid cells. p38b was required for the transcriptional activation of the p53 target gene p21 in response to Aurora B inhibition. In contrast, phosphorylation, stabilization and recruitment of p53 to the p21 promoter occured independently of p38 signaling. Partial inhibition of Aurora B demonstrated that chromosome missegregation also activates the MAP3K4-p38-p53 pathway, suggesting that subtle defects in mitosis are sufficient for inducing this stress signaling pathway. Although p38 was required for the G1 cell cycle arrest after mitotic failures, long-term co-inhibition of p38 and Aurora B resulted in reduced proliferation probably due to increased apoptosis. Presumably, MAP3K4-p38-p53 signaling is a common pathway that is activated after errors in mitosis or cytokinesis to arrest cells in G1 and to prevent chromosomal instability.
Brustkrebs ist gegenwärtig die häufigste bösartige Erkrankung der Frau weltweit und verantwortlich für 15 % der Krebs¬todes-ursachen in der westlichen Welt. Maligne Erkrankungen in metastasierten Stadien gelten generell als unheilbar mit einem medianen Überleben von wenigen Jahren. Das LIM und SH3 Domänen Protein (LASP-1) ist ein spezielles fokales Ad¬hä¬sions-protein, das an den Vorgängen der Zellproliferation und -migration beteiligt ist. Der Knockdown von LASP-1 in metastatischen Brust- und Eier¬stock¬krebs-zelllinien führt zu einer starken Hemmung der Zellmigration und -proliferation. Um¬ge-kehrt kommt es nach Überexpression des Proteins in nicht neoplastischen Zellen zu einer erhöhten Migration. Bei den von uns untersuchten Patientinnen mit Brust- oder Eierstockkrebs korreliert die Überexpression des Proteins mit fortgeschrittener Tumor-größe und Lymphknoten-Metastasierung. Die genetische Analyse von 63 mikrodissektierten histologischen Brust-krebs-Schnittpräparaten mit anschließender qRT PCR auf LASP-1 ergab (mit nur einer positiven Probe; 1,6 %) allerdings keine Amplifikation des Gens. Es scheint, dass die LASP 1 Proteinüberexpression als aktiver Prozess in der Tumorgenese aufgefasst werden kann und in der Mehrheit der Brustkrebsfälle bevorzugt durch trans¬krip-tionelle Regulation als durch Gen¬amplifi¬ka-tion hervorgerufen wird. LASP-1 ist nicht ausschließlich ein zytosolisch lokalisiertes Protein, sondern in malignen Zellen außerdem im Zellkern nachweisbar. In einer Langzeitstudie (Januar 1985 – Dezember 2007) wurde anhand anti-LASP-1 gefärbter histologischer Schnittpräparate die LASP Expression bestimmt und mit dem Patienten-Überleben korreliert. Patientinnen mit nukleärer LASP-1-Lokalisation zeigen, im Vergleich zu nukleär-LASP-1 negativen Schnitten, ein signifikant (p = 0,0250) reduziertes Langzeitüberleben. Mit diesen Ergebnissen lassen sich zukünftig vielleicht prognostische Aussagen über die Auswirkungen der LASP-1-Expression für den einzelnen Patienten treffen.
Inefficient vascularisation of solid tumours leads to the formation of oxygen and nutrient gradients. In order to mimic this specific feature of the tumour microenvironment, a multicellular tumour spheroid (SPH) culture system was used. These experiments were implemented in p53 isogenic colon cancer cell lines (HCT116 p53 +/+ and HCT116 p53-/-) since Tp53 has important regulatory functions in tumour metabolism. First, the characteristics of the cells cultured as monolayers and as spheroids were investigated by using RNA sequencing and metabolomics to compare gene expression and metabolic features of cells grown in different conditions. This analysis showed that certain features of gene expression found in tumours are also present in spheroids but not in monolayer cultures, including reduced proliferation and induction of hypoxia related genes. Moreover, comparison between the different genotypes revealed that the expression of genes involved in cholesterol homeostasis is induced in p53 deficient cells compared to p53 wild type cells and this difference was only detected in spheroids and tumour samples but not in monolayer cultures. In addition, it was established that loss of p53 leads to the induction of enzymes of the mevalonate pathway via activation of the transcription factor SREBP2, resulting in a metabolic rewiring that supports the generation of ubiquinone (coenzyme Q10). An adequate supply of ubiquinone was essential to support mitochondrial electron transport and pyrimidine biosynthesis in p53 deficient cancer cells under conditions of metabolic stress. Moreover, inhibition of the mevalonate pathway using statins selectively induced oxidative stress and apoptosis in p53 deficient colon cancer cells exposed to oxygen and nutrient deprivation. This was caused by ubiquinone being required for electron transfer by dihydroorotate dehydrogenase, an essential enzyme of the pyrimidine nucleotide biosynthesis pathway. Supplementation with exogenous nucleosides relieved the demand for electron transfer and restored viability of p53 deficient cancer cells under metabolic stress. Moreover, the mevalonate pathway was also essential for the synthesis of ubiquinone for nucleotide biosynthesis to support growth of intestinal tumour organoids. Together, these findings highlight the importance of the mevalonate pathway in cancer cells and provide molecular evidence for an enhanced sensitivity towards the inhibition of mitochondrial electron transfer in tumour-like metabolic environments.
Background
Immune checkpoint inhibition and in particular anti-PD-1 immunotherapy have revolutionized the treatment of advanced melanoma. In this regard, higher tumoral PD-L1 protein (gene name: CD274) expression is associated with better clinical response and increased survival to anti-PD-1 therapy. Moreover, there is increasing evidence that tumor suppressor proteins are involved in immune regulation and are capable of modulating the expression of immune checkpoint proteins. Here, we determined the role of p53 protein (gene name: TP53) in the regulation of PD-L1 expression in melanoma.
Methods
We analyzed publicly available mRNA and protein expression data from the cancer genome/proteome atlas and performed immunohistochemistry on tumors with known TP53 status. Constitutive and IFN-ɣ-induced PD-L1 expression upon p53 knockdown in wildtype, TP53-mutated or JAK2-overexpressing melanoma cells or in cells, in which p53 was rendered transcriptionally inactive by CRISPR/Cas9, was determined by immunoblot or flow cytometry. Similarly, PD-L1 expression was investigated after overexpression of a transcriptionally-impaired p53 (L22Q, W23S) in TP53-wt or a TP53-knockout melanoma cell line. Immunoblot was applied to analyze the IFN-ɣ signaling pathway.
Results
For TP53-mutated tumors, an increased CD274 mRNA expression and a higher frequency of PD-L1 positivity was observed. Interestingly, positive correlations of IFNG mRNA and PD-L1 protein in both TP53-wt and -mutated samples and of p53 and PD-L1 protein suggest a non-transcriptional mode of action of p53. Indeed, cell line experiments revealed a diminished IFN-ɣ-induced PD-L1 expression upon p53 knockdown in both wildtype and TP53-mutated melanoma cells, which was not the case when p53 wildtype protein was rendered transcriptionally inactive or by ectopic expression of p53\(^{L22Q,W23S}\), a transcriptionally-impaired variant, in TP53-wt cells. Accordingly, expression of p53\(^{L22Q,W23S}\) in a TP53-knockout melanoma cell line boosted IFN-ɣ-induced PD-L1 expression. The impaired PD-L1-inducibility after p53 knockdown was associated with a reduced JAK2 expression in the cells and was almost abrogated by JAK2 overexpression.
Conclusions
While having only a small impact on basal PD-L1 expression, both wildtype and mutated p53 play an important positive role for IFN-ɣ-induced PD-L1 expression in melanoma cells by supporting JAK2 expression. Future studies should address, whether p53 expression levels might influence response to anti-PD-1 immunotherapy.
Gliomas have been classified according to their histological properties. However, their respective cells of origin are still unknown. Neural progenitor cells (NPC) from the subventricular zone (SVZ) can initiate tumors in murine models of glioma and are likely cells of origin in the human disease. In both, p53 signaling is often functionally impaired which may contribute to tumor formation. Also, TGF-beta, which under physiological conditions exerts a strong control on the proliferation of NPCs in the SVZ, is a potent mitogen on glioma cells. Here, we approach on the crosstalk between p53 and TGF-beta by loss of function experiments using NPCs derived from p53 mutant mice, as well as pharmacological inhibition of TGF-beta signaling using TGF-beta receptor inhibitors. NPC derived from p53 mutant mice showed increased clonogenicity and more rapid proliferation than their wildtype counterparts. Further, NPC derived from p53\(^{mut/mut}\) mice were insensitive to TGF-beta induced growth arrest. Still, the canonical TGF-beta signaling pathway remained functional in the absence of p53 signaling and expression of key proteins as well as phosphorylation and nuclear translocation of SMAD2 were unaltered. TGF-beta-induced p21 expression could, in contrast, only be detected in p53\(^{wt/wt}\) but not in p53\(^{mut/mut}\) NPC. Conversely, inhibition of TGF-beta signaling using SB431542 increased proliferation of p53\(^{wt/wt}\) but not of p53\(^{mut/mut}\) NPC. In conclusion, our data suggest that the TGF-beta induced growth arrest in NPC depends on functional p53. Mutational inactivation of p53 hence contributes to increased proliferation of NPC and likely to the formation of hyperplasia of the SVZ observed in p53 deficient mice in vivo.
