Vom Monomer zum Polymer: Iterative Synthese und optische Spektroskopie von Squarain-Oligomeren

Mittels einer Schutzgruppenstrategie wurden Squarain-basierte monodisperse Oligomere synthetisiert. Die lösungsmittelabhängigen Konformationen (Random Coil vs. Helix) wie auch der Faltungsprozess der Homooligomere wurden mittels optischer Spektroskopie, verschiedener NMR-Experimenten, Kleinwinkelneutronenstreuungsexperimenten sowie quantenchemischen Berechnungen näher beleuchtet. Die optisch-spektroskopischen Beobachtungen wurden mithilfe der Exzitonenkopplungstheorie und einer Orientierungs- und Winkelabhängigkeit der Übergangsdipolmomente der Oligomere erklärt. Der hohe Windungsabstand der helikalen Konformation führt zu einer Interkalation von Lösungsmittel, wodurch eine Art Klathrat gebildet wird. Zusätzlich wurden mittels eines Frenkel-Exzitonenmodells die Absorptions- und Fluoreszenzspektren modelliert. Es konnten die Exzitonendelokalisationslängen abgeschätzt und die Auswirkung der energetischen und strukturellen Unordnungen auf die Absorptions- und Fluoreszenzspektren bestimmt werden. Die Absorptionsspektren werden vorwiegend durch strukturelle Unordnungen verbreitert, die Fluoreszenzspektren dagegen von energetischen Übergangsenergieabweichungen.
Weiterhin wurden auch alternierende Squarain-Cooligomere synthetisiert und mittels optischer Spektroskopie untersucht. Es wurde, abhängig von dem gewählten Lösungsmittel, eine Verschiebung der Hauptbande beobachtet, was durch einen Random Coil vs. helikale-/schlaufenartige Konformation erklärt wird. Gestützt wurde dies mittels quantenchemischen Berechnungen der jeweiligen Konformationen.
Abschließend wurden alternierende Squarain-Copolymere synthetisiert, in verschiedenen Größen aufgetrennt und mittels optischer Spektroskopie untersucht. Mittels EEI2D-Experimenten wurde die Exzitonendynamik in Abhängigkeit von der Kettenlänge eingehender untersucht. Hierbei wird eine steigende, aber relativ abnehmende Kohärenzlänge bestimmt, die Auswirkungen auf die Exzitonendynamik hat. Der Exzitonentransport weist erst wellenförmiges und dann subdiffuses Verhalten auf.
In this work the synthesis, the spectroscopic and electrochemical investigation as well as some applications of a broad diversity of indolenine squaraine dyes were presented. This diversity was based on two parent squaraine dyes, one standard trans-configured compound (M1) and one in which one central oxygen atom was replaced by a dicyanomethylene moiety (M2), which increased the acceptor strength and induced a cis-configuration. The variety of synthesised dyes included functionalised squaraine monomers, donor- and acceptor-substituted monomeric model squaraines, donor- and acceptor-squaraine copolymers, pure squaraine homopolymers, a squaraine-squaraine copolymer, as well as some conjugated cyclic oligomers.
In order to be able to synthesise all these different kinds of dyes, several bromine and boronic ester derivatives were synthesised, which enabled the use of the Suzuki cross coupling reaction, to generate model dyes and copolymers. In addition, the bromine derivatives were used to carry out the Yamamoto homocoupling reaction to the respective homopolymers and macrocycles.
The absorption maximum of unsubstituted reference dye M1 was found at ~ 15500 cm–1, while that of M2 was red-shifted to ~ 14300 cm–1 due to the increased acceptor strength of the central unit. The extinction coefficients were in the order of ~ 300000 M–1 cm–1 and ~ 200000 M–1 cm–1, respectively. It was found that the implementation of functional groups (M3–M9), additional electron donors (M10–M19) or acceptors (M20–M22) at the periphery lead to bathochromic shifts of the absorption depending on the strength of either - and/or -donating properties of the substituents.
For the bis- and triarylamine substituted dyes M10–M13 and the dibrominated dyes M5 and M7 the electronic structure of the mono- and diradical (di)cations was explored using the interplay of cyclic voltammetry, spectroelectrochemistry, and DFT calculations. It was demonstrated that the monoradical cations still show a cyanine-like character and are delocalised Robin-Day class III species due to the low redox potential of the squaraine bridge between the additional amine redox centres. To the best of my knowledge, this made M13+∙, with an N-N-distance of 26 bonds between the additional redox centres to the longest bis(triarylamine) radical cation that is completely delocalised. For the diradical dications, the situation was of larger complexity. The computed most stable energetic state of the dianisylamine-substituted dyes turned out to be a broken-symmetry state with almost equal contributions of an open-shell singlet and triplet state. In addition, it was shown that the HOMO–1→HOMO transition dominated the absorption spectra of the diradical dications where the trans-/cis-configuration of the squaraines had a direct impact due to symmetry reasons.
Based on the donor–squaraine model compounds M10–M19, a series of donor–squaraine copolymers was synthesised (P7–P12) in order to further red shift and broaden the low energy absorption band. However, these effects were only of marginal extent. Both the optical and the electrochemical derived band gaps were barely lowered compared to the respective monomeric model dyes. This was assigned to an increased squaraine-squaraine distance and resulting lower exciton coupling between the squaraine chromophores due to the bridging units. In addition, according to semiempirical calculations the bridges were twisted out of the squaraine plane what reduced conjugational effects between the chromophores. To sum up, the idea to insert additional electron rich bridging units in order to create copolymers with broad and red-shifted absorption did not fully work out for the presented systems.
The addition of strong electron accepting NDI units at the periphery resulted in M21, the most unique monomeric model squaraine in this work. The common picture of a sharp low energy squaraine absorption completely altered due to the addition of the NDIs and a rather broad and solvent dependent low energy absorption was found. Spectroelectrochemical experiments and semiempirical calculations showed that this band is a superposition of the common squaraine HOMO→LUMO transition and a partial squaraine→NDI charge transfer transition. The latter was lost upon oxidation of the squaraine and the absorption spectrum of the monocation of M21 was found to be nearly a 1:1 image of a pure squaraine monocation. Both the monomeric model M21 and the respective copolymer P13 showed low electrochemically obtained band gaps of 1.05–1.20 eV, which were the lowest of all squaraines in this work. For both dyes, transient absorption measurements in the fs-time regime revealed the ultrafast formation of a CS state via an intermediate CT state within a few ps. Besides, charge recombination to the ground state also occured within a few ps. In the polymer, there was barely any further energy or charge transfer within the excited state lifetime and therefore the CS state was confined on adjacent squaraine-NDI pairs and did not further travel along the polymer strand.
The Ni-mediated Yamamoto homocoupling reaction was applied for the synthesis of the homopolymers (P1–P5). In contrast to the donor–squaraine copolymers, those polymers revealed strongly red-shifted and broad absorption in the red to NIR region in addition to a sharp fluorescence. These features could be explained to originate mainly from the exciton coupling of localised excited states and the presence of different superstructures in solution. For the polymers P1 and P2, an elongated J-type polymer chain caused the strong lowest energy absorption band whereas a zig-zag type arrangement of the single chromophores lead to transitions into both low and high energy excited states of the excitonic manifold. For the polymers P3 and P4, several polymer fractions of different size were investigated. Here, also an elongated chain with J-type character induced the lowest energy absorption band whereas a helical H-type arrangement caused transitions to higher energies of the excitonic manifold. The fractions to which these structures were formed depended on the chain length and the solvent. In thin film measurements, it was shown that the initially in solution formed superstructures were partly retained in the thin film but could be altered by annealing procedures. A control of the superstructures should enable the controlled tuning of the optical properties. Despite the strong interaction of the chromophores in the excited state, the redox potentials of the homopolymers barely differed to those of the respective reference dyes, indicating negligible electronic interaction in the ground state.
In addition squaraine-squaraine copolymer P6, consisting of alternating parent dyes M1 and M2, was synthesised. Likewise to the homopolymers, a broad and red-shifted absorption was observed. This was explained by exciton coupling theory, which was extended to also suit alternating copolymers. In toluene, an extraordinary narrow and intense lowest energy absorption band was observed. This exchange narrowing might be a result of a highly ordered J-type structure of the polymer especially in this solvent because it was not found in others. The features of the polymer may be compared to typical J-aggregates formed from monomeric cyanine molecules for example and the polymer used as model for excitonic interactions in an alternating copolymer. Transient absorption measurements revealed a strong energy dependence of the decay traces of the copolymer, most strikingly at early decay times. This was assigned to the occurrence of multiple excitations of one polymer strand (due to the large extinction coefficients of the polymer) and resulting exciton-exciton annihilation. Due to the large exciton diffusion constants that were estimated, the static exciton-exciton annihilation was the rate limiting process of the decay, in contrast to other conjugated polymers, where in thin film measurements the decay was diffusion controlled.
To sum up, for the polymers consisting of exclusively squaraine chromophores, it was shown that the exciton coupling of single chromophores with strong transition dipole moments was a fruitful way to tune the absorption spectra.
As a side product of some of the polycondensation reactions, unprecedented cyclic conjugated oligomers such as the triarylamine-bridged dimer Dim1, the cyclic homotrimers Tri1–Tri3, and the tetramer Tet1 were obtained by recycling GPC in low yields. Especially the cyclic trimers showed unusual absorption and even more extraordinary fluorescence properties. They showed multiple fluorescence bands in the NIR that covered a range from ~ 8000–12500 cm–1 (800–1250 nm). First hints from theoretical calculations indicated that the trimer was not fully planar but comprised a mixture of both planar and bent single squaraine chromophores. However, final results of the calculations were still missing at the time of writing.
In the last part of this work, the application of some monomeric and polymeric squaraines in binary and ternary bulk heterojunction solar cells was demonstrated. Also the utilisation as a dopant in a polymer matrix in an OLED device was shown. The homopolymers P1–P4 were tested in the binary BHJ solar cells revealing poor performances and especially very low short circuit currents. The utilisation of the polymers P3 and P4 that carried the dicyanomethylene group resulted in higher open circuit voltages due to the lower LUMO energy levels but still an overall poor performance. Neither for the different alkyl chains nor for the size of the polymers was a trend observed. In the ternary BHJ solar cells, small amounts of either monomer M14 or polymers P1A, P4–1 or P13 were added to a P3HT/PCBM system in order to generate an additional pathway for charge or energy transfer that should result in a better device performance. However, for none of the tested squaraines, improved solar cells could be built. In similarity to the binary solar cells, the short circuit currents were lower compared to a P3HT/PCBM reference device. These low short circuit currents indicated that the morphology of the squaraine dyes was the major limitation in those devices. It is possible that the dimethyl groups at the indolenine hindered a favoured alignment of the compounds that would allow decent charge transport. In the squaraine doped OLED the squaraine M6 worked rather well as an NIR emitter. Already at low dye loads the fluorescence of the host polymer SY-PPV was completely quenchend and emission from the squaraine was observed. For electroluminescence measurements, a lower dye load (0.5 wt.%) compared to the photoluminescence measurements was sufficient, indicating that apart from FRET additional quenching mechanisms were at work in the electrically driven devices such as charge carrier dynamics.
Umsetzungen N-heterocyclischer Carbene mit Boranen führen zur Bildung von „Lewis-Säure-Base-Addukten“. In Abhängigkeit des Substitutionsmusters der eingesetzten Borane bzw. Carbene eignen sich die erhaltenen Addukte als Ausgangsverbindungen zur Realisierung verschiedener Strukturmotive. Mit geeigneten Übergangsmetallfragmenten gelingt die Darstellung von sigma-Boran-Komplexen bzw. Basen-stabilisierter Boryl-Komplexe, welche mittels spektroskopischer Methoden sowohl im Festkörper, als auch in Lösung untersucht wurden. Ebenfalls gelingt die Synthese Basen-stabilisierter Borirane und einer tetraedrischen Borid-Spezies. Zudem wird ein selektiver Zugang zu Basen-stabilisierten Diborenen entwickelt, wobei deren Bindungssituation und Reaktivität im Detail diskutiert wird. So kann das B=B-Fragment in polymere Spezies eingebunden werden oder als Ligand an Übergangsmetalle koordinieren.
Silbernanodrähte (AgNW) wurden in verschiedene Hybridpolymere und in eine als Referenz dienende Silikonzusammensetzung eingebaut. Durch Spincoating konnten transparente leitfähige Filme erhalten werden. Deren jeweilige Nanodrahtverteilung, thermische Aktivierung und visuelle Transparenz wurden charakterisiert. Die Perkolationsschwelle der Filme hängt dabei von der individuellen durchschnittlichen AgNW-Länge ab. Eine beträchtliche Leitfähigkeit wurde während des mechanischen Streckens bis zu 30 % aufrechterhalten. Mikrostrukturierte Hybridpolymer-Verbundfilme wurden durch UV-Lithographie erhalten. ...
Structure-property relationships in poly(2-oxazoline)/poly(2-oxazine) based drug formulations

According to estimates, more than 40% of all new chemical entities developed in pharmaceutical industry are practically insoluble in water. Naturally, the demand for excipients which increase the water solubility and thus, the bioavailability of such hydrophobic drugs is enormous. Poly(2-oxazoline)s (POx) are currently intensively discussed as highly versatile class of biomaterials. Although selected POx based micellar drug formulations exhibit extraordinarily high drug loadings > 50 wt.% enabling high anti-tumor efficacies in vivo, the formulation of other hydrophobic compounds has failed. This casts doubt on the general understanding in which a hydrophobic active pharmaceutical ingredient is dissolved rather unspecifically in the hydrophobic core of the micelles following the fundamental concept of “like dissolves like”. Therefore, a closer look at the interactions between all components within a formulation becomes increasingly important. To do so, a large vehicle platform was synthesized, loaded with various hydrophobic drugs of different structure, and the formulations subsequently characterized with conventional and less conventional techniques. The obtained in-depth insights helped to develop a more thorough understanding about the interaction of polymer and incorporated API finally revealing morphologies deviating from a classical core/shell structure. During these studies, the scarcely investigated polymer class of poly(2-oxazine)s (POzi) was found as promising drug-delivery vehicle for hydrophobic drugs. Apart from this fundamental research, the anti-tumor efficacy of the two APIs curcumin and atorvastatin has been studied in more detail. To increase the scope of POx and POzi based formulations designed for intravenous administration, a curcumin loaded hydrogel was developed as injectable drug-depot.
Ziel dieser Arbeit war die Stabilisierung von Cadmiumsulfid CdS mit Pluronic P123, einem Polymer.
CdS ist ein Halbleiter, der zum Beispiel in der Photonik und bei optischen Anwendungen eingesetzt wird und ist deshalb äußerst interessant, da seine Bandlücke als Nanopartikel verschiebbar ist. Für die Photovoltaik ist es ein attraktives Material, da es im sichtbaren Licht absorbiert und durch die Bandlückenverschiebung effektiver absorbieren kann. Dies ist unter dem Namen Quantum Size Effekt bekannt. Als Feststoff ist CdS für einen solchen Anwendungsbereich weniger geeignet, zumal der Effekt der Bandlückenverschiebung dort nicht auftritt. Wissenschaftler bemühen sich deshalb CdS als Nanopartikeln zu stabilisieren, weil CdS in wässrigen Lösungen ein stark aggregierendes System, also stark hydrophob ist. Es wurden zwei Kriterien für die erfolgreiche Stabilisierung von CdS festgelegt. Zum einen muss das Cds homogen im Medium verteilt sein und darf nicht agglomerieren. Zum anderen, müssen die CdS Nanopartikel kleiner als 100 A sein.
In meiner Arbeit habe ich solche Partikel hergestellt und stabilisiert, d.h. verhindert, dass die Partikel weiterwachsen und gleichzeitig ihre Bandlücke verschoben wird. Die Herausforderung liegt nicht in der Herstellung, aber in der Lösung von CdS im Trägerstoff, da CdS in den meisten Flüssigkeiten nicht löslich ist und ausfällt. Die Stabilisierung in wässrigen Lösungen wurde das erste Mal durch Herrn Prof. Dr. Rempel mit Ethylendiamintetraessigsäure EDTA erfolgreich durchgeführt. Mit EDTA können jedoch nur sehr kleine Konzentrationen stabilisiert werden. Zudem können Parameter wie Größe und Geschwindigkeit der Reaktion beim Stabilisieren der CdS-Nanopartikel nicht angepasst oder beeinflusst werden. Dieses Problem ist dem, vieler medizinischer Wirkstoffe sehr ähnlich, die in hohen Konzentrationen verabreicht werden sollen, aber nicht oder nur schwer in Wasser löslich sind (Bsp. Kurkumin). Ein vielversprechender Lösungsweg ist dort, die Wirkstoffe in große Trägerpartikel (sog. Mizellen) einzuschleusen, die ihrerseits gut löslich sind. In meiner Arbeit habe ich genau diesen Ansatz für CdS verfolgt. Als Trägerpartikel/Mizelle wurde das bekannte Copolymer Pluronic P123 verwendet. Aus dieser Pluronic Produktreihe wird P123 gewählt, da es die größte Masse bei gleichzeitig höchstem Anteil von Polypropylenoxid PPO im Vergleich zur Gesamtkettenlänge hat. P123 ist ein ternäres Polyether oder Dreiblockkopolymer und wird von BASAF industriell produziert. Es besteht aus drei Böcken, dem mittlere Block Polypropylenoxid PPO und den beiden äußeren Blöcken Polyethylenoxid PEO. Der Buchstabe P steht für pastös, die ersten beiden Ziffern in P123 mit 300 multipliziert ergeben das molare Gewicht und die letzte Ziffer mit 10 multipliziert entspricht dem prozentualen Gewichtsanteil PEO. Die Bildung von Mizellen aus den P123 Molekülen kann bewusst über geringe Temperaturänderungen gesteuert werden. Bei ungefähr Raumtemperatur liegen Mizellen vor, die sich bei höheren Temperaturen von sphärischen in wurmartige Mizellen umwandeln. Oberhalb einer Konzentration von 30 Gewichtsprozent wtp bilden die Mizellen außerdem einen Flüssigkristall. Ich habe in meiner Arbeit zunächst P123 mit Hilfe von Röntgenstreuung untersucht. Anders als andere Methoden gibt Röntgenstreuung direkten Aufschluss über die Morphologie der Stoffe. Röntgenstreuung kann die Mischung von P123 mit CdS abbilden und lässt darauf schließen, ob das Ziel erreicht werden konnte, stabile CdS Nanopartikel in P123 zu binden.
Für die Stabilisierung der Nanopartikel ist es zunächst notwendig die richtigen Temperaturen für die Ausgangslösungen und gemischten Lösungen zu finden. Dazu muss P123 viel genauer untersucht werden, als der momentane Kenntnisstand in der Literatur. Zu diesem Zweck als auch für die Analyse des stabilisierten CdS habe ich ein neues Instrument am LRM entwickelt, sowie eine temperierbare Probenumgebung für Flüssigkeiten fürs Vakuum, um morphologische Eigenschaften aus Streuamplituden und -winkeln zu entschlüsseln. Diese Röntgenstreuanlage wurde konzipiert und gebaut, um auch im Labor P123 in kleinen Konzentrationen messen zu können. Röntgenkleinwinkelstreuung eignet sich besonders als Messmethode, da die Probe mit einer hohen statistischen Relevanz in Flüssigkeit und in verschiedenen Konzentrationen analysiert werden kann.
Für die Konzentrationen 5, 10 und 30 wtp konnte das temperaturabhängige Verhalten von P123 präzise mit Röntgenkleinwinkelstreuung SAXS gemessen und dargestellt werden. Für 5 wtp konnten die Größen der Unimere und Mizellen bestimmt werden. Trotz der nicht vorhandenen Absolutkalibration für diese Konzentration konnten dank des neu eingeführten Parameters kappa eine Dehydrierung der Mizellen mit steigender Temperatur abgeschätzt, sowie eine Hysterese zwischen dem Heizen und Abkühlen festgestellt werden. Für die Konzentration von 10 wtp wurden kleinere Temperaturschritte gewählt und die Messungen zusätzlich absolut kalibriert. Es wurden die Größen und Streulängendichten SLD der Unimere und Mizellen präzise bestimmt und ein vollständiges Form-Phasendiagramm erstellt. Auch für diese Konzentration konnte eine Hysterese eindeutig an der Größe, SLD und am Parameter kappa gezeigt werden, sowie eine Dehydrierung des Mizellenkerns. Dies beweist, dass der Parameter kappa geeignet ist, um bei nicht absolut kalibrierten Messungen, Aussagen über die Hydrierung und Hysterese komplexer Kern-Hülle Modelle zu machen. Für die Konzentration von 30 wtp konnte zwischen 23°C und 35°C eine FCC Struktur nachgewiesen werden. Dabei vergrößert sich die Gitterkonstante der FCC Struktur von 260 A auf 289 A in Abhängigkeit der Temperatur.
Durch das Mischen zweier Lösungen, zum einen CdCl2 und 30 wtp P123 und zum anderen Na2S und 30 wtp P123, konnte CdS erfolgreich stabilisiert werden. Mit einer Kamera wurde die Gelbfärbung der Lösung, und somit die Bildung des CdS, in Abhängigkeit der Zeit untersucht. Es konnte festgestellt werden, dass das Bilden der CdS Nanopartikel je nach Konzentration und Temperierprogramm zwischen 30 und 300 Sekunden dauert und einer logistischen Wachstumsfunktion folgt. Höhere Konzentrationen CdS bewirken einen schnelleren Anstieg der Wachstumsfunktion. Mittels UV-Vis Spektroskopie konnte gezeigt werden, dass die Bandlücke von CdS mit steigender Konzentration konstant bei 2,52 eV bleibt. Eine solche Verschiebung der Bandlücke von ungefähr 0,05 eV im Vergleich zum Festkörper, deutet auf einen CdS Partikeldurchmesser von 80A hin. Mit SAXS konnte gezeigt werden, dass sich die flüssigkristalline Struktur des P123 bei zwei verschiedenen Konzentrationen CdS, von 0,005 und 0,1 M, nicht ändert. Das CdS wird zwischen den Mizellen, also durch die Bildung des Flüssigkristalls, und im Kern der Mizelle aufgrund seiner Hydrophobizität stabilisiert. Die Anfangs definierten Kriterien für eine erfolgreiche Stabilisierung wurden erfüllt.
P123 ist ein hervorragend geeignetes Polymer, um hydrophobes CdS, sowohl durch die Bildung eines Flüssigkristalls, als auch im Kern der Mizelle zu stabilisieren.
Serum half-life elongation as well as the immobilization of small proteins like cytokines is still one of the key challenges for biologics. This accounts also for cytokines, which often have a molecular weight between 5 and 40 kDa and are therefore prone to elimination by renal filtration and sinusoidal lining cells. To solve this problem biologics are often conjugated to poly(ethylene glycol) (PEG), which is the gold standard for the so called PEGylation. PEG is a synthetic, non-biodegradable polymer for increasing the hydrodynamic radius of the conjugated protein to modulate their pharmacokinetic performance and prolong their therapeutic outcome. Though the benefits of PEGylation are significant, they also come with a prize, which is a loss in bioactivity due to steric hindrance and most often the usage of heterogeneous bioconjugation chemistries. While PEG is a safe excipient in most cases, an increasing number of PEG related side-effects, such as immunological responses like hypersensitivity and accelerated blood clearance upon repetitive exposure occur, which highlights the need for PEG alternative polymers, that can replace PEG in such cases.
Another promising method to significantly prolong the residence time of biologics is to immobilize them at a desired location. To achieve this, the transglutaminase (TG) Factor XIIIa (FXIIIa), which is an important human enzyme during blood coagulation can be used. FXIIIa can recognize specific peptide sequences that contain a lysine as substrates and link them covalently to another peptide sequence, that contains a glutamine, forming an isopeptide bond. This mechanism can be used to link modified proteins, which have a N- or C-terminal incorporated signal peptide by mutation, to the extracellular matrix (ECM) of tissues.
Additionally, both above-described methods can be combined. By artificially introducing a TG recognition sequence, it is possible to attach an azide group containing peptide site-specifically to the TG, recognition sequence. This allows the creation of a site-selective reactive site at the proteins N- or C-terminus, which can then be targeted by cyclooctyne functionalized polymers, just like amber codon functionalized proteins.
This thesis has focused on the two cytokines human Interferon-α2a (IFN-α2a) and human, as well as murine Interleukin-4 (IL-4) as model proteins to investigate the above-described challenges. IFN-α2a has been chosen as a model protein because it is an approved drug since 1986 in systemic applications against some viral infections, as well as several types of cancer. Furthermore, IFN-α2 is also approved in three PEGylated forms, which have different molecular weights and use different conjugation techniques for polymer attachment. This turns it into an ideal candidate to compare new polymers against the gold standard PEG. Interleukin-4 (IL-4) has been chosen as the second model protein due to its similar size and biopotency. This allows to compare found trends from IFN-α2a with another bioconjugate platform and distinguish between IFN-α2a specific, or general trends. Furthermore, IL-4 is a promising candidate for clinical applications as it is a potent anti-inflammatory protein, which polarizes macrophages from the pro-inflammatory M1 state into the anti-inflammatory M2 state.
Polymer-Biokonjugationen, vornehmlich mit dem Goldstandard PEG, führen zu einer verbesserten Pharmakokinetik, beeinflussen aber auch die konformative Stabilität von Proteinen. Bisherige Mutationsstudien, in denen überwiegend (Asn)PEG4 -Konjugate der Beta-faltblattstrukturreichen, humanen Pin 1 WW-Domäne untersucht wurden, postulieren auf einer Proteindesolvatation beruhende Stabilisierungsmechanismen: eine Stärkung intramolekularer Salzbrücken und NH-pi-Bindungen, sowie entropisch günstige Wasserverdrängungen um apolare Aminosäuren und Hydroxylgruppen. Ziel dieser Arbeit ist es, die Protein-Polymer-Dynamik auf molekularer Ebene zu charakterisieren, um damit rationale Ansätze zum Design neuer Biokonjugate voranzutreiben und mögliche PEG-Alternativen zu etablieren. Hierzu wurde eine Vielzahl an Deskriptoren mittels Molekulardynamik-Simulationen der WW-Konjugate gewonnen und mit publizierten Stabilitätsdaten in multivariaten Regressions- und logistischen Klassifikationsmodellen korreliert. Die gewonnenen QSPR-Modelle decken im Vergleich zu einer bereits publizierten, kristallstrukturbasierten Richtlinie einen größeren und strukturell vielfältigeren Datensatz an Konjugaten ab und zeigen gleichzeitig, auch für ein Konjugat der Src SH3-Domäne, eine deutlich verbesserte Leistung. Die Modelldeskriptoren beschreiben sowohl eine Modulation der Solvatation als auch Protein-Polymer-Interaktionen. Metadynamik-Simulationen zeigten zudem die Polymerdynamik während einer partiellen Proteinentfaltung auf. Mithilfe weiterer Simulationen von Konjugaten des alpha-helikalen Her2-Affibodys wurde die Dynamik von PEG und verschiedener Alternativen (LPG, PEtOx, PMeOx) systematisch studiert. PEG interagierte mit positiv geladenen Lysinen und Argininen in der Nähe hydrophober Aminosäuren. LPG zeigte zusätzliche Wechselwirkungen der Hydroxylgruppen mit Aspartaten und Glutamaten. POx-Polymere interagierten mit Phenylalaninen, Tyrosinen und über Carbonylgruppen mit HB-Donatoren. Größere Konjugate (10 - 50 kDa PEG/LPG/PEtOx) des antiviralen Biologikums Interferon-alpha2a wurden mittels gaußbeschleunigter MDs und einer CG-Simulation analysiert. Charakteristische Wechselwirkungspartner stimmten mit den Beobachtungen zu Oligomer-Konjugaten überein. In Einklang mit experimentellen Daten der Kooperationspartner zu den 10-kDa-Varianten deuteten zusätzliche Constrained-Network-Analysen, welche die Proteinflexibilität evaluieren, auf eine thermische Destabilisierung hin. Die Bioaktivität der untersuchten Konjugate wurde weiterhin erfolgreich mit den Gyrationsdurchmessern der modellierten Strukturen korreliert.
The aim of the thesis was to develop water soluble poly(2-oxazoline) (POx) copolymers with new side group functionalities, which can be used for the formation of hydrogels in biomedical applications and for the development of peptide-polymer conjugates.
First, random copolymers of the monomer MeOx or EtOx with ButEnOx and EtOx with DecEnOx were synthesized and characterized. The vinyl functionality brought into the copolymer by the monomers ButEnOx and DecEnOx would later serve for post-polymerization functionalization. The synthesized copolymers were further functionalized with thiols via post-polymerization functionalization using a newly developed synthesis protocol or with a protected catechol molecule for hydrogel formation. For the formation of peptide-polymer conjugates, a cyclic thioester, namely thiolactone acrylamide and an azlactone precursor, whose synthesis was newly developed, were attached to the side chain of P(EtOx-co-ButEnOx) copolymers.
The application of the functionalized thiol copolymers as hydrogels using thiol-ene chemistry for cross-linking was demonstrated. The swelling behavior and mechanical properties were characterized. The hydrophilicity of the network as well as the cross-linking density strongly influenced the swelling behavior and the mechanical strength of the hydrogels. All hydrogels showed good cell viability results.
The hydrogel networks based on MeOx and EtOx were loaded with two dyes, fluorescein and methylene blue. It was observed that the uptake of the more hydrophilic dye fluorescein depended more on the ability of the hydrogel to swell. In contrast, the uptake of the more hydrophobic dye methylene blue was less dependent on the swelling degree, but much more on the hydrophilicity of the network.
For the potential application as cartilage glue, (biohybrid) hydrogels were synthesized based on the catechol-functionalized copolymers, with and without additional fibrinogen, using sodium periodate as the oxidizing agent. The system allowed for degradation due to the incorporated ester linkages at the cross-linking points. The swelling behavior as well as the mechanical properties were characterized. As expected, hydrogels with higher degrees of cross-linking showed less swelling and higher elastic modulus. The addition of fibrinogen however increased the elasticity of the network, which can be favorable for the intended application as a cartilage glue. Biological evaluation clearly demonstrated the advantage of degradable ester links in the hydrogel network, where chondrocytes were able to bridge the artificial gap in contrast to hydrogels without any ester motifs.
Lastly, different ways to form peptide-polymer conjugates were presented. Peptides were attached with the thiol of the terminal cysteine group to the vinyl side chain of P(EtOx-co-ButEnOx) copolymers by radical thiol-ene chemistry. Another approach was to use a cyclic thioester, thiolactone, or an azlactone functionality to bind a model peptide via native chemical ligation. The two latter named strategies to bind peptides to POx side chains are especially interesting as one and in the case of thiolactone two free thiols are still present at the binding site after the reaction, which can, for example, be used for further thiol-ene cross-linking to form POx hydrogels.
In summary, side functional poly(oxazoline) copolymers show great potential for numerous biomedical applications. The various side chain functionalities can be introduced by an appropriate monomer or by post-polymerization functionalization, as demonstrated. By their multi-functionality, hydrogel characteristics, such as cross-linking degree and mechanical strength, can be fine-tuned and adjusted depending on the application in the human body. In addition, the presented chemoselective and orthogonal reaction strategies can be used in the future to synthesize polymer conjugates, which can, for example, be used in drug delivery or in tissue regeneration.
The subject of this thesis is the synthesis and characterization of PBI-based fluorescent metallosupramolecular polymers and cyclic arrays. Terpyridine receptor functionalized PBIs of predesigned geometry have been used as building blocks to construct desired macromolecular structures through metal-ion-directed self-assembly. These metallosupramolecular architectures have been investigated by NMR, UV/Vis and fluorescence spectroscopy, mass spectrometry, and atomic force microscopy.
