Die häufigste Form der Herzinsuffizienz in Deutschland ist die dilatative Kardiomyopathie, wobei bei ca. 6/100.000 Einwohnern pro Jahr keine eindeutige Ursache erkennbar ist und somit eine idiopathische DCM diagnostiziert wird. Ein Faktor zur Entstehung einer idiopathischen DCM könnten Autoantikörper gegen den β1-adrenergen Rezeptor sein. Bei ca. 30% der Patienten, die an einer DCM (Äquivalent in der ETiCS-Studie: erste akute Myokarditis = AMitis) leiden, sowie bei ca. 13% der Patienten, die an einer ischämischen Kardiomyopathie (Äquivalent in der ETiCS-Studie: erster akuter Myokardinfarkt = FAMI) leiden, konnten in älteren Arbeiten β1-AAk nachgewiesen werden. Im Rahmen der ETiCS-Studie erfolgte erstmals eine prospektive Beobachtung entsprechender Patientenkollektive über 12 Monate mit Blutentnahme und klinischen Kontrollen zum Zeitpunkt 0 Monate (=Baseline), 2-3 Monate (Follow-Up 1), 6 Monate (FUP2) und 12 Monate (FUP3). Zu diesen Zeitpunkten wurden anhand der gewonnenen Blutproben der β1-AAk-Status sowie die immunologischen Marker der FAMI- und AMitis-Patienten bestimmt und mit der kardialen LV-Pumpfunktion korreliert.
Zentrales Thema dieser Arbeit war es, Zusammenhänge zwischen der β1-AAk-Ausbildung in Abhängigkeit von individuellen Zytokinprofilen und der Entwicklung der LV-Pumpfunktion nach dem jeweiligen kardialen Ereignis zu untersuchen, wobei FAMI- und AMitis-Patienten miteinander verglichen wurden. Darüber hinaus wurde auch der Einfluss der CTLA-4-Haplotypen, also die „genetische“ Suszeptibilität Autoantikörper zu entwickeln, untersucht.
Während bei FAMI-Patienten die Entwicklung von β1-AAk keinen Einfluss auf den Verlauf der LV-Pumpfunktion zu haben scheint, wird diese bei AMitis-Patienten durch hochaffine β1-AAk im Verlauf stark beeinträchtigt.
Bei FAMI-Patienten konnte nach einer größeren Herzschädigung (CK-Werte >1000 U/l) eine schlechtere Pumpfunktion im Vergleich zu kleineren Myokardinfarkten (CK-Werte <1000 U/l) nachgewiesen werden, unabhängig von β1-AAk-Status. Für die Prognose und die Erholung der LV-Pumpfunktion scheint bei FAMI-Patienten folglich die Infarktgröße, aber nicht die Entwicklung von β1-AAk wichtig zu sein.
Hinsichtlich der unterschiedlichen Zytokinprofile bei FAMI- und AMitis-Patienten, die hochaffine β1-AAk entwickeln, scheinen bestimmte Zytokine die Induktion einer kardialen Autoimmunität zu begünstigen, während andere Zytokine wohl eher protektive immunologische Reaktionen in Gang setzen: Die proinflammatorischen Zytokine IL-1β, IL-2, IL-7, IL-12, IL-17, GM-CSF, MIP-1α und IFN-γ waren bei β1-AAk-positiven AMitis-Patienten statistisch signifikant erhöht. Protektive Effekte könnten dagegen von den antiinflammatorischen Zytokinen IL-1RA, IL-10 und IL-13 ausgehen, deren Serumspiegel bei FAMI- gegenüber AMitis-Patienten im Vergleich erhöht waren.
Beim direkten Vergleich von AMitis-Patienten mit hochaffinen β1-AAk und solchen ohne β1-AAk, zeigten sich bei Patienten mit hochaffinen β1-AAk höhere Konzentrationen an IL-1β, IL-2, IL-6, IL-7, IL-12, IL-17, GM-CSF, MIP-1α und TNF-α. Bei Patienten ohne Autoantikörper waren demgegenüber die Spiegel von IL-1RA, IL-10 und IL-13 erhöht, was zu einer besseren Erholung der LV-Pumpfunktion führte.
Nach genetischer Typisierung der CTLA-4-Haplotypen (Polymorphismen SNP +49G/A und SNP CT60A/G) fand sich bei Patienten mit dem Allel G/G ein höheres Risiko β1-AAk zu entwickeln, während das Allel A/A jeweils mit einem geringeren Risiko kardiale Autoantikörper zu entwickeln assoziiert war und somit protektiv gegen Autoimmunphänomene wirken könnte.
Zielsetzung dieser Arbeit war die Untersuchung von Gehirngewebe in der asymptomatischen Phase der SIV-Infektion zur Erfassung immunologischer Vorgänge bei der Entstehung einer SIV-Enzephalitis am Beispiel der gewebeständigen Lymphozyten und Makrophagen. Wir benutzten das etablierte SIV-AIDS Modell mit Makakenaffen. Untersucht wurden 24 Versuchstiere im Zeitraum von bis zu 20 Wochen, die zuvor mit einem der beiden Virusstämme (SIVmac251/ 32H oder SIVmac251/ MPBMC) infiziert worden waren, vier der Versuchstiere entwickelten eine SIV-Enzephalitis, eines dieser Tiere war mit Selegilin behandelt worden. Fünf weitere infektfreie Makaken dienten als Kontrollen. Nach definierten Zeiträumen wurden die Versuchstiere getötet, die Gehirne präpariert und mittels Immunhistochemie Lymphozyten und Makrophagen angefärbt. Im weiteren führten wir eine lichtmikroskopische Morphometrie, getrennt nach einzelnen Kompartimenten des Gehirns, wie Cortex, subcorticalem weißem Mark und perivaskulärem Raum, durch. Die Ergebnisse unserer Arbeit — die infizierte, asymptomatische Tiere mit den Kontrolltieren vergleicht — zeigen eine ausgeprägte lymphozytäre Reaktion speziell im Cortex, zum Teil auch im subcorticalen Mark. Die Makrophagen zeigen eine Zunahme der Zelldichte perivaskulär und im Cortex, welche quantitativ deutlich unter der der Lymphoyzten bleibt. Die Tiere mit Enzephalitis zeigen allesamt eine corticale, wie auch subcorticale Zunahme der lymphozytären Zelldichte, bei den Makrophagen zeigt sich eine überwiegend generelle Stimulierung in allen drei Kompartimenten. Die Ergebnisse unserer Arbeit — die sich durch die Untersuchung asymptomatischer Versuchstiere sehr gut an die Verhältnisse bei der HIV-Infektion annähert — lassen vermuten, daß es dem Immunsystem mit Hilfe der gewebeständigen Lymphozyten im Cortex und subcorticalen Mark für eine ganze Weile gelingt, den Ausbruch einer Enzephalitis und damit der AIDS Erkrankung zu verzögern.
Visualization of type I immunity using bicistronic IFN-gamma reporter mice in vitro and in vivo

IFN-γ is the signature cytokine of Th1 and CD8+ effector cells generated in type I immune responses against pathogens, such as Influenza virus, Sendai virus and the intracellular protozoan parasite Toxoplasma gondii. Understanding the regulation of IFN-γ is critical for the manipulation of immune responses, prevention of immunopathology and for vaccine design. In the present thesis, IFN-γ expression by CD4+ and CD8+ T cells was characterized in detail and the requirement of IFN-γ receptor mediated functions for IFN-γ expression was assessed. Bicistronic IFN-γ-eYFP reporter mice, which allow direct identification and isolation of live IFN-γ expressing cells, were used to visualize IFN-γ expression in vitro and in vivo after infection with the afore mentioned pathogens. Expression of the IFN-γ-eYFP reporter by CD4+ and CD8+ T cells was broadly heterogeneous in vitro and in vivo after infection. Increased expression of the reporter correlated positively with the abundance of IFN-γ transcripts and IFN-γ protein production upon stimulation. eYFP reporter brightness reflected the potential for IFN-γ production, but actual secretion was largely dependent on antigenic stimulation. Increased expression of the reporter also correlated with enhanced secretion of additional proinflammatory cytokines and chemokines and cell surface expression of markers that indicate recent activation. Highly eYFP fluorescent cells were generally more differentiated and their anatomical distribution was restricted to certain tissues. The anatomical restriction depended on the pathogen. IFN-γ expressing CD4+ and CD8+ T cells were generated in IFN-γ receptor deficient reporter mice after infection with Sendai virus or Toxoplasma gondii. However, in the absence of IFN-γ receptor mediated functions, the frequency and brightness of the eYFP reporter expression was altered. Dual BM chimeric mice, reconstituted with wild-type and IFN-γ receptor deficient reporter BM, revealed a T cell-intrinsic requirement for the IFN-γ receptor for optimal IFN-γ expression. Reporter fluorescence intensities were regulated independently of IFN-γ receptor mediated functions. Finally, we propose a model for IFN-γ expression by CD4+ and CD8+ T cells. 2. SUMMARY 10 In summary, the expression of IFN-γ is differentially regulated in CD4+ and CD8+ T cells and after viral or protozoan infections. Additionally, the role of IFN-γ receptor mediated functions for the expression of IFN-γ was determined.
Das Kolonkarzinom besitzt aufgrund seiner hohen Inzidenz und der zahlreichen Möglichkeiten, mittels moderner Therapiestrategien auf den Krankheitsverlauf Einfluss zu nehmen, eine hohe Relevanz für den klinischen Alltag. Etwa die Hälfte der an einem kolorektalen Karzinom erkrankten Patienten weisen Lebermetastasen auf, welche entweder bereits bei der primären Diagnosestellung vorliegen oder sich im weiteren Krankheitsverlauf ausbilden. Bei der Mehrzahl der Patienten mit Lebermetastasen ist derzeit nur eine palliative Therapie möglich. Die genaue Analyse kolorektaler Lebermetastasen ist somit unerlässlich, um eine gezielte Entwicklung neuer, effizienter Therapiestrategien für dieses große Patientenkollektiv zu ermöglichen. Ziel dieser Arbeit war es, möglichst genau Zytokinprofil, lymphozytäres Infiltrationsmuster und Verhältnis von pro- zu antiangiogenetischen Faktoren im Lebermetastasengewebe über einen biologisch maßgeblichen Wachstumszeitraum zu charakterisieren, um zu einem besseren Verständnis von Tumorimmunologie und Wachstumsverhalten kolorektaler Lebermetastasen beizutragen. Zu diesem Zweck wurde in einem Mausmodell die Ausbildung kolorektaler Lebermetastasen nach intraportaler Injektion von Tumorzellen (CT26.WT) untersucht.
The work of the previous chapters describes the role of Nipah virus (NiV) V and W proteins regarding their role in interferon antagonism and regulation of viral replication. Previous publications have shown that NiV encodes IFN antagonist activity in its V, W and C protein (Park et al., 2003b; Rodriguez et al., 2002). In order to study the effect of both NiV proteins in the context of a virus infection, recombinant Newcastle disease viruses (rNDVs) expressing NiV V or NiV W were constructed. As a control virus served rNDV expressing NDV V proteins, which behaved like wildtype NDV. Growth kinetic experiments demonstrated that rNDVs expressing NiV V or W grew to higher titers than rNDV expressing NDV V in human A549 cells. This result suggested that both NiV V and W were able to render the avian virus, which normally does not replicate well in human cells, into a better growing virus. This hypothesis was supported by the fact that all rNDVs grew similarly in avian DF1 or Vero cells. When rNDV-infected A549 cells were specifically stained for NiV V or W protein it was observed that V is localized in the cytoplasm whereas W could be predominantly found in the nucleus. This observation was in agreement with previous studies reporting a nucleus export signal (NES) for NiV V and a nuclear localization signal (NLS) for NiV W (Rodriguez et al., 2004; Shaw et al., 2005). The specific localization of each NiV protein has also been shown to contribute to different functions in terms of IFN antagonism (Shaw et al., 2005). Here, NiV V and W proteins caused a severe attenuation of the immune response in rNDV-infected human A549 and dendritic cells. The transcription of type I interferons and ISGs was significantly downregulated in the presence of NiV V and W proteins. As a consequence of the transcriptional block, there was also an inhibition at the level of translation (as seen for A549 cells) and the secretion of IFNs and cytokines/chemokines (as seen for DCs). In contrast, NDV V protein induced a host immune response. Both NiV V and W also displayed a strong inhibitory effect on the function DCs. DCs represent a very important cell class because they link the innate immune response to the adaptive immune response (Banchereau & Steinman, 1998). By downregulating the production and secretion of important cytokines/chemokines that are important for the activation of B and T lymphocytes, NiV V and W were able to disrupt that link. Interestingly, NiV W seemed to be a stronger inhibitor than NiV V in both A549 cells and DCs. Overall, it was demonstrated that NiV V and W were able to prevent the induction of the innate and adaptive host immune response cascade by inhibiting the transcription of immune genes in DCs and A549 cells. The second part of this work addressed the question whether NiV V and W proteins have a regulatory role in viral replication. This has been previously reported for Nipah virus itself (Sleeman et al., 2008) and other viruses (Atreya et al., 1998; Horikami et al., 1996; Witko et al., 2006). In order to study the ability of the V and W proteins of NiV to regulate viral transcription and/or replication, an existing NiV minireplicon assay was used (Halpin et al., 2004). Here, it was shown that NiV V and W (but not C) proteins significantly downregulated NiV minireplicon activity. The common N terminal region was shown to harbor the inhibitory activity. Co-immunoprecipitation experiments showed that both NiV V and W (but not C) were able to interact with NiV N, one component of the NiV polymerase. This result was supported by immunofluorescence experiments that revealed co-localization of NiV N with V and W. The binding of NiV V or W to NiV N occurred via their N terminus and more specifically amino acids 1-50. This suggested that V and W might inhibit viral replication by interacting with the viral polymerase resulting in a loss of function. Exact mechanisms still have to be elucidated.
The cancer stem cell hypothesis is a cancer development model which elicited great interest in the last decades stating that cancer heterogeneity arises from a stem cell through asymmetrical division. The Cancer Stem Cell subset is described as the only population to be tumorigenic and having the potential to renew. Conventional therapy often fails to eradicate CSC resulting in tumor relapse. Consequently, it is of great inter-est to eliminate this subset of cells to provide the best patient outcome. In the last years several approaches to target CSC were developed, one of them being immunotherapeu-tic targeting with antibodies. Since markers associated with CSC are also expressed on normal stem cells or healthy adjacent tissue in colorectal cancer, dual targeting strate-gies are preferred over targeting only a single antigen. Subsequently, the idea of dual targeting two CSC markers in parallel by a newly developed split T cell-engaging anti-body format termed as Hemibodies emerged. In a preliminary single cell RNA sequenc-ing analysis of colorectal cancer cells CD133, CD24, CD166 and CEA were identified as suitable targets for the combinatorial targeting strategy. Therefore, this study focused on trispecific and trivalent Hemibodies comprising a split binding moiety against CD3 and a binding moiety against either CD133, CD24, CD166 or CEA to overcome the occurrence of resistance and to efficiently eradicate all tumor cells including the CSC compartment. The study showed that the Hemibody combinations CD133xCD24, CD133xCD166 and CD133xCEA are able to eliminate double positive CHO cells with high efficacy while having a high specificity indicated by no killing of single antigen positive cells. A thera-peutic window ranging between one to two log levels could be achieved for all combina-tions mentioned above. The combinations CD133xCD24 and CD133xCD166 further-more proved its efficacy and specificity on established colorectal cancer cell lines. Be-sides the evaluation of specificity and efficacy the already introduced 1st generation of Hemibodies could be improved into a 2nd generation Hemibody format with increased half-life, stability and production yield. In future experiments the applicability of above-mentioned Hemibodies will be proven on patient-derived micro tumors to also include variables like tumor microenvironment and infiltration.
Semaphorin receptors in the immunological synapse: regulation and measles virus-driven modulation

Measles virus (MV) infection causes approximately 164,000 deaths per year worldwide (WHO, 2008). The main cause of death is MV-induced immunosuppression but the underlying mechanisms are not fully understood. It has been suggested that MV renders T cells dysfunctional by disrupting the integrity of actin dynamics while MV infection of dendritic cells results in their inability to sustain T cell activation. During neuronal development, semaphorins (SEMAs), especially SEMA3A, induce a collapse of growing dendrites via the binding to plexin-A1 (plexA1) and its coreceptor neuropilin-1 (NP-1). The collapse results from a disruption of actin dynamics. In this study, the roles of these three molecules were investigated in human immune cells and their possible role in MV induced immunosuppression. The present data have shown that plexA1 is an important component of human immunological synapse (IS). It translocated transiently to the surface of T cells after CD3/28 ligation and accumulated at the stimulatory interface between T cells and DCs (or CD3/28 coated beads). When plexA1 expression was inhibited (RNAi) or its function was disrupted (exogenous blocking or dominant negative expression), T cell expansion was reduced. Upon MV exposure, translocation of plexA1 and NP-1, another important component of IS, towards the stimulatory interface in T cells was abrogated. Moreover, MV infection interfered with plexA1/NP-1 turnover in maturing DCs and promoted early and substantial release of SEMA3A from these cells, particularly in the presence of allogenic T cells. As revealed by scanning electron microscopy, the release of SEMA3A caused a transient loss of actin-based protrusions on T cells. SEMA3A affected chemotactic migration of T cells and DCs, and reduced formation of allogenic DC/T cell conjugates. In conclusion, MV targeted SEMA receptor function both by disrupting their recruitment to the IS and by promoting a premature release of their repulsive ligand, SEMA3A. Both of which could contribute to MV-induced immunosuppression.
Rolle der gammadelta T-Zellen in der Immunantwort bei Patienten mit gastrointestinalen Tumoren

Zusammenfassend konnte im Rahmen der vorliegenden Arbeit die Frage nach der Fähigkeit der selektiven Stimulierung mittels des Phosphorantigens HMBPP und den beiden BTN3 Antikörpern bestätigt werden. Es konnte zudem wie erwartet hierbei ein Unterschied zwischen den beiden Kohorten detektiert werden. Dabei zeigte die Kohorte der Normalspender erwartungsgemäß eine stärkere Aktivierungs- sowie Proliferations-fähigkeit. Normalspender ließen sich signifikant besser mit HMBPP aktivieren und bei bestimmter Konzentration signifikant besser proliferieren, bei BTN3A und sc20.1 konnten keine signifikanten Unterschiede ermittelt werden, allerdings anhand der Mittelwerte eine deutlich stärkere Aktivierung und Proliferation aufgezeigt werden. Außerdem konnten interessante interindividuelle Unterschiede detektiert werden, die neue Erkenntnisse brachten. Mit Hilfe der untersuchten Oberflächenmoleküle CD45RA und CD27 und der Einteilung der gammadelta T-Zellen in unterschiedliche Subgruppen konnten so mögliche Erklärungen für die Unterschiede zwischen den Kohorten aufgezeigt werden. Normalspender zeigten signifikant höhere Anteile an naiven gammadelta T-Zellen und nicht signifikant höhere Anteile an central memory T-Zellen, demnach eine deutliche Verschiebung in Richtung nicht differenzierter Subsets, wohingegen die Tumorkohorte signifikant höhere effector memory T-Zellen aufwiesen und somit eine deutliche Verschiebung in Richtung differenzierter Subsets. Dadurch kann erklärt werden, weshalb Normalspender besser aktiviert werden und besser proliferieren können. Auch die Einteilung in unterschiedliche Profile 1-6 anhand CD28, CD27 und CD16 lieferte Gründe für den Unterschied zwischen den Kohorten, wobei Normalspender der Gruppe 1 und 2, Tumorpatienten der Gruppe 3 und 4 angehörten. Durch Ermittlung weiterer signifikanter Änderungen einiger exprimierter Oberflächenmoleküle CD39, CD161 und PD1 wurde mit Hilfe der vorliegenden Arbeit bekräftigt, dass einige Faktoren betrachtet werden müssen, die die Proliferation und Aktivierung der gammadelta T-Zellen positiv und negativ beeinflussen können. Es konnte jedoch auch erneut verdeutlicht werden, wie komplex und weitgreifend der Aktivierungsmechanismus, die damit verbundene Expansion und die Auslösung der einzelnen Effektorfunktionen ist.
The sequencing of several ant genomes within the last six years open new research avenues for understanding not only the genetic basis of social species but also the complex systems such as immune responses in general. Similar to other social insects, ants live in cooperative colonies, often in high densities and with genetically identical or closely related individuals. The contact behaviours and crowd living conditions allow the disease to spread rapidly through colonies. Nevertheless, ants can efficiently combat infections by using diverse and effective immune mechanisms. However, the components of the immune system of carpenter ant Camponotus floridanus and also the factors in bacteria that facilitate infection are not well understood.
To form a better view of the immune repository and study the C. floridanus immune responses against the bacteria, experimental data from Illumina sequencing and mass-spectrometry (MS) data of haemolymph in normal and infectious conditions were analysed and integrated with the several bioinformatics approaches. Briefly, the tasks were accomplished in three levels. First, the C. floridanus genome was re-annotated for the improvement of the existing annotation using the computational methods and transcriptomics data. Using the homology based methods, the extensive survey of literature, and mRNA expression profiles, the immune repository of C. floridanus were established. Second, large-scale protein-protein interactions (PPIs) and signalling network of C. floridanus were reconstructed and analysed and further the infection induced functional modules in the networks were detected by mapping of the expression data over the networks. In addition, the interactions of the immune components with the bacteria were identified by reconstructing inter-species PPIs networks and the interactions were validated by literature. Third, the stage-specific MS data of larvae and worker ants were analysed and the differences in the immune response were reported.
Concisely, all the three omics levels resulted to multiple findings, for instance, re-annotation and transcriptome profiling resulted in the overall improvement of structural and functional annotation and detection of alternative splicing events, network analysis revealed the differentially expressed topologically important proteins and the active functional modules, MS data analysis revealed the stage specific differences in C. floridanus immune responses against bacterial pathogens.
Taken together, starting from re-annotation of C. floridanus genome, this thesis provides a transcriptome and proteome level characterization of ant C. floridanus, particularly focusing on the immune system responses to pathogenic bacteria from a biological and a bioinformatics point of view. This work can serve as a model for the integration of omics data focusing on the immuno-transcriptome of insects.
Listeria monocytogenes, ein Gram-positives, fakultativ intrazelluläres Bakterium, kann bei immunsupprimierten Menschen schwere Infektionen des Zentralnervensystems auslösen. In Folge seines ubiquitären Vorkommens, sowie seiner hohen Resistenz gegenüber Lebensmittel-Konservierungsmethoden besteht ein großes Interesse an seiner raschen Identifizierung und Differenzierung gegenüber den apathogenen Spezies seiner Gattung. Sein P60 als essentielles Housekeeping-Gen bietet sich auf Grund der zwischen den einzelnen Spezies konservierten und variablen Bereiche für die Etablierung gattungs- und speziesspezifischer Nachweissysteme an. Mit Hilfe des EPITOP-SPOT-Mappings wurde eine Immunogenitätskarte des P60 von L. innocua bzw. L. monocytogenes erstellt, P60-spezifische CD4-T-Zellinien charakterisiert und das Epitop eines dieser T-Zellklone exakt bestimmt. Transferexperimente mit diesen T-Zelllinien und Boosterinfektionen mit L. monocytogenes bzw. L. innocua zeigten, dass L. innocua alleine zwar nicht in der Lage ist, einen Immunschutz gegen L. monocytogenes zu etablieren, diesen jedoch in vitro und in vivo erhalten kann, indem es durch kreuzreaktive Epitope Gedächtnis-T-Zellen stimuliert. Die in vitro-Transkription von Phly, PplcA und PactA erfolgt strikt PrfA-abhängig, während Piap, PprfA1/2 und - unerwarteter Weise - auch Pmpl PrfA-unabhängig transkribiert werden. Sie erfolgt - ausgenommen bei PprfA - nur bei ausreichender Verfügbarkeit von ATP nicht jedoch GTP.
