Dications 1 derived from 2,2-bipyridine are found to exist as fully reversible two step redox systems with persistent radical ions (SEM) of high thermodynamic stability, if the bridge X forces the two pyridine rings into co planar positions. The well known derivative 1a (Diquat R) is now complemented by the boronium ions 1c and 1e -1g., as shown by voltammetry and the uv spectra of the corresponding radicals.

A route to 2S,5S-and 2R,5S-hydroxypipecolic acid is presented, starting with the enantiopure 5S-5-hydroxy-piperidone 7. The key step of this reaction sequence is the chemoselsctive methylenation of the amide carbonyl group of 8 with dimethyltitanocene 9 to 10. The transformation of the exocyclic enecarbamate double bond to the carboxylic acid group is best accomplished via hydroboration/oxidation to the alcohol 11a,b. Separation and oxidation of the dlastereomers 11a,b, to 148. and 14b, and hydrolysis furnishes the diastereomeric pipecolic acids 15a and 15b in enantiopure form.
The 2-halo imidoyl chlorides 7 are obtained from the amide 5 and the 2-halo amides 6 by the action of phosphorus pentachloride and thionyl chloride, respectively. Non-racemic (S)-6a is converted into 7a which is racemic, however. The reaction of Lawesson's reagent with 6a furnishes the diastereomeric 1,3.2-thiazaphospholidine derivatives 15. Treatment of (S)-6a (98% eel with methyl triflate affords 2-chloro imidate 8 (95% eel which reacts with methanamine in the presence of methanammonium chloride to yield the 2-chloro amidine (S)-9a (90% eel. The 2-halo imidoyl halides 7a and b react with methanamine to produce the 2-halo amidines 9a and b. - Strong bases, e.g. potassium tert-butoxide or sodium hydride in the presence of catalytic amounts of tertbutyl alcohol, eliminate hydrogen chloride or bromide from the 2-halo amidines 9a and band (S)-9a to yield mixtures of Recently, we demonstrated that the formation of the chiral non-racemic aziridinone (R)-2 from the a-chloro amide (5)-1 by base-promoted dehydrochlorination[2) as well as the nucleophilic cleavage of the N-C(3) bond of (R)_2[3,4) occur with inversion of configuration, thus excluding the intervention of achiral (acyclic) intermediates. In the temperature range of lOO-170°C, however, slow racemization accompanies the thermolysis of (R)-2 and indicates the existence of an achiral or a racemic transient, e. g. (M)-3 + (P)-3. Indeed, high-level quantum-chemical calculations reveal that an activation energy of (170 ± 25) kJmol- 1 is required for the unimolecular ring opening of the parent aziridinone which affords a species of high diradical character[41. Subsequently, the unstable N-phenylaziridinone invoked in the decomposition of the (5)-2-bromopropananilide anion was shown to react with tert-butylamine or dimethylformamide with inversion of configuration at C(3)[51. Thus, the stereochemical evidence in the series of 3-alkylaziridinones excludes achiral (acyclic) aziridinone isomers as intermediates at low tempera tures [6J. Similar stereochemical studies are still missing in the related series of iminoaziridines. Therefore, we report on the synthesis and thermolysis of the diastereomeric chiral racemic (E)- and (Z)-(4)[71 and non-racemic iminoaziridines (E,R)- and (Z,R)-4. Racemic Iminoaziridines (E)- and (Z)-4 Though a photochemical route to the iminoaziridines (E)- and (Z)-4 has been devised more recently, i. e. the phothe 2-iminoaziridines (E)- and (Z)-4, and (E,R)- and (Z.R)-4 (83% eel, respectively. The 1.3-elimination of hydrogen bromide from 9b is diastereoselective at -30 to -40°C [(E)-4:(Z)-4 = <10:>90). The diastereomers equilibrate at 36°C with (kEZ + k ZE) = (5.92 ± 0.08) . 10-5 S-I (K = kEZlkzE = 0.428 ± 0.013). - The thermolysis of (E)- and (Z)-4 in [D61benzene solution yields the imine 16 and methyl isocyanide (17). The decomposition follows the first-order rate law. The following Arrhenius and Eyring parameters are calculated from five rate constants obtained in the temperature range of 70-110°C: Ea = (115.2 ± 0.4) kJmol-t, IgA = (12.06 ± 0.28), AH* = (112.1 ± 0.4) kJmol- l , AS'" = (-23.9 ± 0.7) JK-I mol-I, AGj73K = 121 kJmol-1 . The enantiomeric excess of the surviving fraction of (E,R)- and (Z.R)-4 is unchanged after two half-lives at 80°C.
Unsere Lebensmittel haben eine zweifache, auch vom Gesetz vorgegebene Zweckbestimmung, d.h. sie dienen einerseits dem Genuss und andererseits der Ernährung. Aktuelle For-schungsstrategien in der Lebensmittelchemie auf diesen Gebieten lassen sich dahingehend zusammenfassen, dass im ersten Bereich die Suche nach wirksamen Aromastoffen und deren Vorläufern sowie auch zunehmend Probleme der Herkunftsanalytik im Vordergrund stehen. Auf dem Gebiet der Ernährung spiegeln die Schlagworte „Funktionalität“ und „Bioaktivität“ die derzeitigen Entwicklungen wider. Für alle Bereiche bilden instrumentell-analytische Techniken die Grundlage für Bewertungen. Im Rahmen der vorliegenden Arbeit haben wir daher das uns zur Verfügung stehende analytische Potential genutzt, in den oben genannten Gebieten einschlägige Beiträge zu leisten, d.h. die Suche nach wertgebenden Aromastoffen fortzuführen, Zuckerkonjugate zu untersuchen, die sowohl als Aromavorläufer als auch im Hinblick auf Bioaktivitätsstudien von Bedeutung sein können, und schließlich Methoden zur Herkunftsanalytik zu forcieren. Zu diesen Studien sind als attraktive und wirtschaftlich be-deutsame Rohwaren Rucola (Eruca sativa Mill.) und Kaffee (Coffea arabica) eingesetzt worden. Mit Hilfe der Kopplung aus S-selektiver Detektion und strukturselektiver massen-spektro-metrischer Analytik sind in Extrakten von Rucolablättern 22 schwefelhaltige Verbindungen strukturell charakterisiert worden. Darunter befanden sich neben Minorkomponenten wie 3-Sulfanyl-1-hexanol, Glucosinolatabbauprodukte wie 4-Methylthiobutylnitril, 4-Methylthiobutylthiocyanat und 4-Methylthiobutylisothiocyanat. Mit [1,3]-Thiazepan-2-thion ist eine bislang nicht bekannte Komponente erstmals beschrieben wor-den. Das Vorkommen von 3-Sulfanyl-1-hexanol, welches im Übrigen anhand von MDGC-MS-Analytik in nahezu racemischer Form (44:56 Prozent, R:S) in Rucola gefunden wurde, forderte dazu heraus, in einer Modellstudie eine allgemeine Methode zur Bestimmung der Absolutkonfiguration azyklischer 2- und 3-Sulfanyl-1-alkanole mittels der ECCD-Methode zu entwickeln. Dabei wurden in einer Ein-Schritt-Synthese die funktionellen Grup-pen der 2- und 3-Sulfanyl-1-alkanole (in der Studie handelte es sich um enantiomerenreine 2- und 3-Sulfanyl-1-hexanole) jeweils mit dem Chromophor 9-Anthroylflourid derivatisiert. Die ent-sprechenden CD-Splitkurven erlaubten eine eindeutige Zuordnung der Konfiguration. Die entwickelte Mikro-Maßstab-Methode wurde ebenfalls er-folgreich zur Bestimmung der Absolutkonfiguration von in der Studie als zusätzliche Modellverbindungen eingesetzten 1,2- und 1,3-Diolen angewendet. 3-Sulfanyl-1-hexanol wurde abschließend in einer Zwei-Schritt-Synthese mit 9- und 2-Anthroylflourid derivatisiert, auch diese Variation erlaubte eine eindeutige Zuordnung der Absolutkonfiguration. Mittels präparativer HPLC-Techniken wurden drei neue Flavonoidglykoside aus Rucolablättern isoliert. Deren Charakterisierung erfolgte mittels Tandemmassenspektrometrie sowie ein- und zweidimensionalen NMR-Messungen. Die Verbindungen sind als Quercetin-3,3’,4’-tri-O-beta-D-glucopyranosid, Quercetin-3’-(6-sinapoyl-O-beta-D-gluco-py-ra-nosyl)-3,4’-di-O-beta-D-gluco-pyranosid und Quercetin-3-(2-sinapoyl-O-beta-D-gluco-pyranosyl)-3’-(6-sina-poyl-O-beta-D-glucopyranosyl)-4’-O-beta-D-gluco-py-rano-sid identifiziert worden. Diese Quercetin-Derivate zeigten sowohl hinsichtlich der Glucosilierung des Aglykons (an den Positionen 3, 3’ und 4’) als auch bezüglich der Acylierung ungewöhnliche Strukturen. Bei Rohkaffee hat man bislang keine Studien über das Vorkommen von Zuckerkonjugaten als Aromastoffvorläufer durchgeführt. Wir konnten diese Lücke anhand der Strukturaufklärung der nachfolgend aufgeführten Verbindungen schließen. Durch Kombination verschiedener präparativer Techniken (CCC, LC und HPLC) mit verschiedenen Methoden der Strukturaufklärung (HRGC-MS, HPLC-MS/MS, ein- und zweidimensionale NMR) sind aus Rohkaffee fünf Aromastoffprekursoren isoliert und identifiziert worden. Für die beiden aus Rohkaffee isolierten Linalyldisaccharide wurden als Strukturen 3(S)-Linalyl-3-O-beta-D-glucopyranosyl-(1-6)-alpha-L-arabinofuranosid (AK1) und 3(S)-Linalyl-3-O-beta-D-gluco-pyranosyl-(1-6)-beta-D-apiofuranosid ermittelt. Weiterhin sind erstmals drei neue Aromastoffprekursoren in Form von Zuckerestern, d.h. (3-Methylbutanoyl)-1-O-beta-D-glucopyranosyl-beta-D-apiofuranosid, (3-Methylbuta-noyl)-6-O-alpha-D-gluco-pyranosyl-(1-2)-beta-D-fructofuranosid und (3-Methyl-2-butenoyl)-O-beta-D-gluco-pyranosyl-beta-D-apio-furanosid in Rohkaffee charakterisiert worden. Die Kenntnis der Herkunft bestimmter Lebensmittel bzw. einzelner In-haltstoffe spielt für die Lebensmittelindustrie eine wichtige Rolle. Für die Zuordnung der geographischen Herkunft von Rohkaffee wurden in dieser Arbeit die 13C/12C-, 2H-/1H- und 18O/16O-Verhältnisse von extrahiertem Coffein mittels Elementaranalysator-Isotopen-massen-spektro-metrie (EA-IRMS) bestimmt. Zur Bewertung der Multielement-Messwerte wurden diese statistischen Berechnungen unterzogen. Zur Anwendung kamen die Lineare Diskrimi-nanzanalyse (LDA) und die Auswertung mittels sog. „Classification and Regression Trees“ (CART). In der Anpassung wurde dabei eine Probe aus der Klasse 1 (Provenienz Afrika) der Klasse 2 (Provenienz Amerika) zugeordnet und eine Probe der Klasse 2 der Klasse 1 zugeordnet (Fehlerrate von 5,1 Prozent). Bei Kreuzvalidierung wurden drei der 39 Proben falsch zugeordnet (jeweils zwei der Klasse 1 und eine der Klasse 2), die Fehlerrate betrug hier somit 7,7 Prozent. Die Proben der Klasse 3 (synthetisches Coffein) ließen sich zu 100 Prozent von denjenigen natürlichen Ursprungs unterscheiden.
In der vorliegenden Arbeit werden Studien zur selektiven bakteriellen Hydroxylierung von Fettsäuren vorgestellt. Unter Verwendung von Linolsäure als Substrat wurden aus Bodenproben verschiedene Mikroorganismen isoliert, die polare Metabolite bildeten. Die phänotypische und genotypische Charakterisierung eines Stammes führte zu dessen Identifizierung als Stenotrophomonas maltophilia. Die Strukturaufklärung der drei Hauptreaktionsprodukte erfolgte mittels Hochleistungsflüssigchromatographie-Massenspektrometrie (HPLC-MS), Gaschromatographie-Massenspektrometrie (GC-MS) sowie ein- und zweidimensionalen NMR-Experimenten (1H-NMR, 13C-NMR, 13C-DEPT, H/H-COSY, HMQC, HMBC). Linolsäure wurde von Stenotrophomonas maltophilia zu 3-Hydroxy-Z6-dodecensäure, 3-Hydroxy-Z5,Z8-tetradecadiensäure und 3-Hydroxy-Z7,Z10-hexadecadiensäure umgesetzt. In einem anschließenden Substratscreening wurden 32 Verbindungen als Edukte für die Biotransformation eingesetzt und so die strukturellen Voraussetzungen ermittelt, die für eine effiziente Umsetzung von Fettsäuren durch Stenotrophomonas maltophilia notwendig sind. Zum Einsatz kamen Substrate mit unterschiedlicher Anzahl an C-Atomen sowie mit Variationen bezüglich Anzahl, Position und Konformation von Doppelbindungen. Weiterhin wurden Substanzen verwendet, die bereits funktionelle Gruppen im Molekül aufwiesen (z. B. Ricinolsäure). Die Bestimmung der Enantiomerenverteilung der bakteriell gebildeten 3-Hydroxysäuren mittels multidimensionaler Gaschromatographie (MDGC) ergab einen deutlichen Enantiomerenüberschuss (ee 84 – 98 Prozent). Die Aufklärung der Absolutkonfiguration erfolgte über die Synthese von Dodecan-1,3-diolen und deren anschließende Analytik mittels MDGC. Zusätzlich wurde die Konfiguration mit Hilfe der CD Exciton Chirality-Methode bestimmt. Weiterhin wurde untersucht, ob die bakteriell gebildeten 3-Hydroxysäuren als Substrate oder Inhibitoren des Enzyms Lipoxygenase L-1 aus Sojabohnen fungieren. Die im Rahmen dieser Arbeit durchgeführten Studien zur Darstellung von optisch aktiven 3-Hydroxysäuren belegen das Potential des Bodenbakteriums Stenotrophomonas maltophilia, exogen zugeführte Fettsäuren im Rahmen der b-Oxidation zu kettenverkürzten, an Position 3 hydroxylierten Metaboliten abzubauen. Dabei liegen jedoch deutliche Abweichungen zur b-Oxidation in anderen Organismen vor, die auf Unterschieden in der Enzymausstattung bzw. deren Aktivität beruhen. Durch die gewonnenen Erkenntnisse zum b-Oxidationsmechanismus in Stenotrophomonas maltophilia kann diese Aktivität durch geeignete Substratauswahl gezielt zur Synthese von optisch aktiven 3-Hydroxysäuren eingesetzt werden, deren chemische Synthese gegenüber dieser Biotransformation deutlich schwieriger zu realisieren ist. Für solche Verbindungen besteht in der organischen Synthese von Naturstoffen wie Pheromonen, Vitaminen und Antibiotika Bedarf.
Die vorliegende Arbeit präsentiert neue Erkenntnisse zur Biosynthese von 2,5-Dimethyl-4-hydroxy-3(2H)-furanon (DMHF) und 2,5-Dimethyl-4-methoxy-3(2H)-furanon (DMMF), zwei wichtigen Aromakomponenten in Erdbeeren. Potentielle, mit stabilen Isotopen markierte Vorläufersubstanzen wurden an Erdbeeren appliziert und drei Tage bei Raumtemperatur inkubiert. Der Nachweis über den erfolgreichen Einbau erfolgte mittels Gaschromatographie-Massenspektrometrie. Anhand der Massenspektren von DMHF und DMMF, die aus den behandelten Erdbeeren mittels Festphasenextraktion isoliert wurden, konnte der Markierungsgrad der Verbindungen ermittelt werden und somit Rückschlüsse über die Effizienz der Metabolisierung der applizierten Zucker zu den beiden Furanonen DMHF und DMMF gezogen werden. Als mögliche Ausgangsstoffe dienten unterschiedlich markierte D-Glucose und D-Fructose, sowie Desoxyzucker, da diese als direkte Vorläufersubstanzen von DMHF und DMMF diskutiert werden. Isotopenmarkierte Desoxy-D-glucose oder Desoxy-D-fructose sind kommerziell nicht erhältlich, weshalb die Verbindungen [1-13C]-1-Desoxy-D-fructose, [6-2H1]-6-Desoxy-D-glucose und [5,6,6,6-2H4]-6-Desoxyhexulose-1-phosphat zuerst synthetisiert werden mussten. Nach Applikation der Desoxyzucker wurde keine Erhöhung des Markierungsgrades an stabilen Isotopen gegenüber dem natürlichen Isotopenverhältnis festgestellt. Somit können 6-Desoxy-D-glucose, 1-Desoxy-D-fructose und 6-Desoxy-D-fructose-1-phosphat als Prekursoren von DMHF und DMMF ausgeschlossen werden. Als gute Vorläufersubstanzen erwiesen sich D-Glucose und D-Fructose. Markierungen (13C und 2H) an Position C-1 oder C-6 der beiden Verbindungen wurden sowohl in DMHF als auch in dessen Methoxyderivat DMMF detektiert, wobei die Applikation von D-Fructose im Gegensatz zur D-Glucose einen höheren Markierungsgrad der Zielverbindungen zur Folge hatte. Durch den Einsatz positionsspezifisch markierter D-Glucose (2H-Markierung an Position C-1, C-2 oder C-4) sollten Aufschlüsse über den Metabolisierungsmechanismus gewonnen werden. Die Markierung der D-[2-2H]Glucose befand sich wie die der D-[1-2H]Glucose in den Methylgruppen der Furanone, was nur durch eine intramolekulare Verschiebung von C-2 nach C-1 erklärbar ist. Diese wurde bei der Glucosephosphatisomerase-katalysierten Umwandlung von D-Glucose-6-phosphat zu D-Fructose-6-phosphat beobachtet. Somit muss D-Glucose bei der Biosynthese von DMHF und DMMF zuerst in diese Intermediate überführt werden. Im Gegensatz zu an Position C-2 markierter D-Glucose ging das Proton an Position C-4 im Laufe der Metabolisierung verloren. Demzufolge findet der in der Natur verbreitete Desoxygenierungsmechanismus von Monosacchariden nicht statt und schließt die Beteiligung von Desoxyzuckern an der Biosynthese von DMHF und DMMF gänzlich aus. Nach Einsatz von uniform markierter D-Fructose konnte sechsfach markiertes DMHF und DMMF identifiziert werden, was durch den Einbau der intakten Kohlenstoffkette zu erklären ist. Dieser Befund und weitere Untersuchungen mit verschiedenen Glykolyse-regulierenden Substanzen deuteten darauf hin, dass die Furanone dem zentralen Kohlenhydratstoffwechsel, der Glykolyse, entspringen. Vor der Aldolase-katalysierten Spaltung in zwei C3-Einheiten muss jedoch eine Abzweigung erfolgen, da sonst die Kohlenstoffkette nicht unverändert vorliegen könnte. Im zweiten Teil der Arbeit ist erstmals mit Hilfe von molekularbiologischen Techniken die vollständige cDNA einer O-Methyltransferase (OMT) aus Erdbeeren isoliert worden. Hierfür wurde mRNA aus reifenden Erdbeeren extrahiert und eine cDNA Bibliothek hergestellt. Diese wurde mit einer OMT-spezifschen Sonde durchmustert, welche durch PCR mit degenerierten Primern synthetisiert worden war. Nach mehreren Vereinzelungs-Zyklen konnte die vollständige cDNA einer O-Methyltransferase (STOMT, Strawberry OMT) erhalten werden. Northern-Analysen ergaben, dass die entsprechende RNA ausschließlich in den verschiedenen Reifestadien der Frucht akkumuliert, mit den höchsten Transkriptmengen in der rot-werdenden und reifen Frucht. In anderen Gewebeteilen wie Wurzel, Blätter, Stängel und Blüte konnte keine STOMT-RNA nachgewiesen werden. Das korrespondierende Protein zeigte hohe Homologien zu Kaffeesäure-OMTs aus Weidengewächsen der Gattung Populus. Nach erfolgreicher, heterologer Expression von STOMT in E. coli wurde die Substratspezifität des Enzyms untersucht, dessen Temperaturoptimum bei 30°C lag. Alle eingesetzten Substrate mit phenolischem Grundgerüst, wie Brenzcatechin, Kaffeesäure, Kaffeeoyl-CoA und 3,4-Dihydroxybenzaldehyd, aber auch das Furanonderivat DMHF wurden von der rekombinanten O-Methyltransferase umgesetzt. Als bestes Substrat erwies sich 3,4-Dihydroxybenzaldehyd, das, im Gegensatz zu dessen Methylierungsprodukt Vanillin, bisher in Erdbeeren nicht nachgewiesen werden konnte. Kaffeesäure wurde ebenfalls effektiv methyliert, worin vermutlich die Hauptaufgabe von STOMT in der Pflanze liegt. Die Methylierung von Kaffeesäure oder 5-Hydroxyferulasäure ist ein wichtiger Prozess in der Entstehung von Lignin. Die Tatsache, dass Erdbeeren teilweise in den Leitbündeln und verstärkt in den Achenen lignifiziert sind, erklärt das Vorhandensein eines solchen Enzyms. DMHF, das als Dienol-Tautomer eine aromatische Struktur mit Hydroxylgruppen aufweist und somit strukturelle Ähnlichkeiten zu phenolischen Verbindungen zeigt, wurde ebenfalls von STOMT als Substrat akzeptiert. Die Bildung von DMMF, dem Methoxyderivat von DMHF erfolgte vergleichsweise langsam, war aber eindeutig auf die Methyltransferase-Aktivität zurückzuführen. STOMT ist aufgrund des Expressionsmusters als fruchtspezifisch und reifeinduziert einzustufen. Primäre Funktion ist vermutlich zu Beginn der Fruchtreifung die Lignifizierung der Leitbündel und später die der Achenen. Gleichzeitig scheint STOMT wesentlich an der Bildung der Aromastoffe DMMF und Vanillin in Erdbeeren beteiligt zu sein.
Ziel der vorliegenden Arbeit war die Entwicklung, Optimierung und Validierung von phyto- und bioanalytischen Analysemethoden. Dabei wurden exemplarische Fragestellungen aus dem Themenkreis Phytotherapie bzw. Bioverfügbarkeit und Metabolismus von einfachen und Polyphenolen bearbeitet. Quercetin und seine Derivate: Zur qualitativen Analyse der Flavonoide und -derivate in einem Trockenextraktgemisch aus Birkenblättern, Goldrutenkraut und Orthosiphonblättern wurde eine HPLC- UV/VIS Methode entwickelt. Nach oraler Applikation dieses Trockenextraktgemisches als orale Arzneiform wurde die systemische Verfügbarkeit von Quercetin bzw. Quercetinglykosiden untersucht, welche für Quercetin bzw. -glykosiden 0 Prozent betrug (LOD ~ 25 pg Quercetin on column), da Quercetin als Phase-I- bzw. -II- Metabolite vorlag. Nach Hydrolyse der Phase-II-Konjugate wurden die höchsten Quercetinspiegel (101±107 ng·mL-1) nach ca. 4 Stunden (tmax) gemessen. Der Zeitpunkt der höchsten renalen Quercetinausscheidung lag im Intervall von 4?8 Stunden nach Verum-Gabe, wobei die über 24 Stunden ausgeschiedene Quercetinmenge 604±1037 µg·mL-1 betrug. Im Rahmen der studienbegleitenden bioanalytischen Methodenentwicklung wurde eine Extraktionsmethode für Plasma und Urin entwickelt und validiert. Durch Anwendung eines neuartigen Analyseverfahrens, der coulometrischen Array-Detektion, konnte eine HPLC-Methode entwickelt werden, die erstmals Quercetin und seine Metabolite simultan, selektiv und ausreichend sensitiv in biologischen Matrices detektieren konnte. Unter Verwendung des analytischen Verfahrens der HPLC-Tandemmassenspektrometrie konnten erstmals fünf Quercetinglucuronide als die Hauptmetabolite des Quercetins identifiziert werden. Um weitere Erkenntnisse über den Metabolismus der systemisch verfügbaren Quercetinglucuronide am Venenendothelgewebe zu gewinnen, wurde ein In-vitro-Modell unter Verwendung von HUVEC (human umbilical endothelial cells)- Monolayerkulturen entwickelt und orientierend validiert. Die Experimente zeigten, dass das Venenendothel Glucuronidaseaktivität aufweist und somit Quercetin in-situ am oder im Zielgewebe aus systemisch vorliegenden Glucuroniden freisetzen kann. Die frei vorliegenden Aglykone können dann in das Venenendothelgewebe aufgenommen werden. Das In-vitro-Modell gibt damit erstmals einen plausiblen Erklärungsansatz für die Wirksamkeit von Quercetinderivat-haltigen Präparaten in-vivo, z.B. beim varikösen Symptomen-Komplex. Procyanidine in Weißdorn: Zur Gruppenbestimmung der Procyanide in Weißdornpräparaten wurde ein Analysenprotokoll optimiert und validiert, welches den derzeit üblichen Protokollen bezüglich des Grades an Selektivität überlegen ist. Weiterführend wurde zur selektiven Analyse der mono- bis trimeren Procyanidine in Weißdornpräparaten eine HPLC-Methode unter Adaption eines Nachsäulenderivatisierungsverfahrens (NSD) entwickelt und validiert. Unter Kombination beider Verfahren konnte erstmals das Procyanidinspektrum in handelsüblichen Weißdornpräparaten durch Einbeziehung der mono- bis trimeren Procyanidine, näher beschrieben, und marktführende Weißdornpräparate vergleichend analysiert werden. In Zusammenhang mit publizierten pharmakologischen Arbeiten mit teilweise identischen Extrakten, leisten die in dieser Arbeit erlangten Kenntnisse über Gehalt und Polymerisationsgrad der in den untersuchten Phytopharmaka enthaltenen Procyanidine einen Beitrag zum besseren Verständnis der Wirkungen von Weißdorn-Zubereitungen insgesamt. Hydrochinon und seine Derivate: Die renale Verfügbarkeit von Hydrochinon nach oraler Gabe einer flüssigen und einer festen Bärentraubenblätterextrakt-Zubereitung wurde anhand einer Probandenstudie gezeigt. Das mit dem Bärentraubenblätter-Trockenextrakt applizierte Arbutin (210 mg, ≈ 771,3 µmol) wurde zu ca. 0,18 Prozent als freies Hydrochinon (≈ 1,38 ± 0,79 µmol) ausgeschieden. Der Zeitpunkt der maximalen Hydrochinon-Ausscheidung (0,3±0,1 µg) lag bei beiden Arzneiformen im 4-8 Stunden-Intervall nach Applikation. Zur Analyse des Hydrochinons bzw. seiner Konjugate in humanem Urin wurde eine Extraktionsmethode zur simultanen Extraktion entwickelt und für den Zielanalyten Hydrochinon validiert. Durch Anwendung der coulometrischen Array-Detektion, konnten in Probandenurin erstmals Substanz-Zeit-Kinetiken freien Hydrochinons detektiert werden. Die in der vorliegenden Arbeit erhobenen In-vivo-Daten erlauben damit eine Korrelation von einerseits In-vitro-Daten zur bakteriziden Aktivität von Hydrochinon und andererseits epidemiologischen Daten zur Wirksamkeit von Bärentraubenblätter-Zubereitungen. Damit konnte erstmals unter therapiekonformen Bedingungen das für Bärentraubenblätter postulierte Wirkprinzip - das Hydrochinon - in-vivo bestätigt werden.
In der vorliegenden Arbeit wird die Massenbilanzierung einer Wirbelschicht-granulierung implementiert. Durch die umfangreiche Instrumentierung eines Laborgerätes zum Wirbelschichtgranulieren im Top-Spray-Verfahren (Glatt GPCG 1.1, Binzen, Deutschland) ist es möglich, die relative Luftfeuchte, die Temperatur und den Absolutdruck der Frischluft sowie der Abluft und ferner den Luftvolumenstrom zu messen. Die Berechnung der Dichten der feuchten Luft ermöglicht den Übergang zu Massenströmen von Luft und von Wasser. Aus der Differenz von in die Anlage eingebrachtem und herausgefördertem Wasser wird auf die aktuelle Masse an Wasser im Granulationsansatz geschlossen. Voraussetzung für eine derartige Massenbilanzierung ist dabei eine sehr präzise Bestimmung sämtlicher relevanter Parameter. Die relative Luftfeuchte wird mit kapazitiven Feuchtesensoren gemessen, die einem neu entwickelten Kalibrierverfahren unterzogen werden. Da das Ansprechverhalten der Feuchtesensoren einen kritischen Prozeßparameter darstellt, werden Vergleichsmessungen mit akustischen Feuchtesensoren durchgeführt. Die Messung des Volumenstroms erfolgt durch ein ungedämpftes Flügelradanemometer. Zur Kalibrierung dieses Sensors werden mit einem Hitzedrahtanemometer Strömungsprofile in einer Einlaufstrecke bei verschiedenen Volumenstrombedingungen aufgenommen. Aus der Integration der Strömungsgeschwindigkeit über den Rohrquerschnitt wird der aktuelle Volumenstrom ermittelt. In der gesamten Anlage herrschen stets turbulente Strömungsverhältnisse. Voraussetzung für die Massenbilanzierung ist, dass im Bereich der Feuchtemessstellen sämtliches Wasser gasförmig vorliegt, da Kondensat nicht von den Feuchtesensoren erfasst wird. Durch Messung der Rohrwandtemperatur und Berechnung der Taupunkttemperatur der Abluft kann eine Kondensation von Wasser an der Stelle der Abluftfeuchtemessung ausgeschlossen werden. Durch Anwendung der Massenbilanzierungsrechnung kann gezeigt werden, dass der Wassergehalt im Granulationsgefäß im Verlauf der Sprühphase kontinuierlich ansteigt, um während der Trocknungsphase in drei Trocknungsabschnitten wieder auf den Ausgangswert zurückzugehen. Mit dieser Arbeit werden die Voraussetzungen für die umfassende Steuerung von Wirbelschichtgranulationsanlagen sowie der darin stattfindenden Granulationsprozesse gelegt.
