Spinal muscular atrophy with respiratory distress type 1 (SMARD1) is a fatal monogenic motoneuron disease in children with unknown etiology caused by mutations in the immunoglobulin μ-binding protein 2 (IGHMBP2) gene coding for DNA/RNA ATPase/helicase. Despite detailed knowledge of the underlying genetic changes, the cellular mechanisms leading to this disease are not well understood. In the Nmd2J ("neuromuscular disorder") mouse, the mouse model for the juvenile form of SMARD1 patients, in which similar pathological features as diaphragmatic paralysis and skeletal muscle atrophy are observed. Ex vivo studies in Nmd2J mice showed that loss of the motor axon precedes atrophy of the gastrocnemius muscle and does not correlate with neurotransmission defects in the motor endplate. The already described independent myogenic anomalies in the diaphragm and heart of the Nmd2J mouse raised the question whether spinal motoneuron degeneration develops cell autonomously. Ighmbp2 is predominantly localized in the cytoplasm and seems to bind to ribosomes and polysomes, suggesting a role in mRNA metabolism.
In this Ph.D. thesis, morphological and functional analyses of isolated Ighmbp2-deficient (Ighmbp2-def.) motoneurons were performed to answer the question whether the SMARD1 phenotype results from dysregulation of protein biosynthesis. Ighmbp2-deficient motoneurons show only negligible morphological alterations with respect to a slight increase in axonal branches. This observation is consistent with only minor changes of transcriptome based on RNA sequencing data from Ighmbp2-deficient motoneurons. Only the mRNA of fibroblast growth factor receptor 1 (Fgfr1) showed significant up-regulation in Ighmbp2-deficient motoneurons. Furthermore, no global aberrations at the translational level could be detected using pulsed SILAC (Stable Isotope Labeling by Amino acids in cell culture), AHA (L-azidohomoalanine) labeling and SUnSET (SUrface SEnsing of Translation) methods. However, a reduced β-actin protein level was observed at the growth cones of Ighmbp2-deficient motoneurons, which was accompanied with a reduced level of Imp1 protein, a known β-actin mRNA interactor. Live-cell imaging studies using fluorescence recovery after photobleaching (FRAP) showed translational down-regulation of eGFPmyr-β-actin 3'UTR mRNA in the growth cones and the cell bodies, although the amount of β-actin mRNA and the total protein amount in Ighmbp2-deficient motoneurons showed no aberrations. This compartment-specific reduction of β-actin protein occurred independently of a non-existent direct IGHMBPF2 binding to β-actin mRNA. Fgfr1, which was upregulated on the RNA level, did not show an increased protein amount in Ighmbp2-deficient motoneurons, whereas a reduced amount could be detected. Interestingly, a correlation could be found between the reduced amount of the Imp1 protein and the increased Fgfr1 mRNA, since the IMP1 protein binds the FGFR1 mRNA and thus could influence the transport and translation of FGFR1 mRNA. In summary, all data suggest that Ighmbp2 deficiency leads to a local but modest disturbance of protein biosynthesis, which might contribute to the motoneuron defects of SMARD1.
Das invasive Potential maligner Gliome beeinflusst maßgeblich die schlechte Prognose dieser Tumorentität. Migration und Invasion von Tumorzellen werden entscheidend durch die Cofilin-vermittelte Umstrukturierung des Aktin-Zytoskeletts geprägt, die durch die Aktivität antagonistischer Cofilin-Kinasen und -Phosphatasen reguliert wird.
Im Rahmen der vorliegenden Arbeit konnte ein progressiver Expressionsverlust der Cofilin-Phosphatase Chronophin mit ansteigendem Malignitätsgrad astrozytärer Gliome aufgezeigt werden, der mit einer Zunahme der Phosphorylierung von Cofilin einhergeht. In den entsprechenden Gewebeproben gelang gleichzeitig der Nachweis einer gesteigerten Expression der Cofilin-Kinase LIMK-2.
Genetische und epigenetische Analysen des Chronophin-Locus konnten eine Hypermethylierung im Bereich der Promotorregion der Phosphatase identifizieren, die möglicherweise dem Verlust von Chronophin in Glioblastom-Gewebeproben zugrunde liegt.
In Glioblastom-Zelllinien, die unterschiedliche Expressionsmuster von Chronophin aufwiesen, konnten hingegen keine molekularen Alterationen festgestellt werden.
Untersuchungen des Einflusses von ROCK- und LIMK-Inhibitoren auf Glioblastomzellen konnten ausgeprägte Veränderungen der Zellmorphologie dokumentieren, wobei erstmals die Induktion eines stellate cell-Phänotyps unter Einfluss des LIMK-Inhibitors BMS-5 beschrieben wird. Während ROCK- und LIMK-Inhibitoren keinen Einfluss auf die 2D-Motilität der Tumorzellen hatten, wiesen die Glioblastomzellen in Abhängigkeit ihrer basalen Cofilin-Aktivität eine verstärkte bzw. verminderte 3D-Invasivität auf.
Die Erkenntnisse dieser Arbeit unterstreichen die Bedeutung des Cofilin-Signalweges für die Migration und Invasion von Gliomzellen, zeigen neue Angriffspunkte in der Therapie maligner Gliome auf und warnen zugleich vor einem unkritischen Einsatz neuer Wirkstoffe.
Regulation Tumornekrosefaktor (TNF) Rezeptor assoziierter Signalwege durch das Adapterprotein TRAF1

TWEAK ist ein zu der TNF-Superfamilie (Tumor Necrosis Factor) zugehöriges Zytokin, welches in Form löslicher und membranständiger Moleküle vorkommt. Beide Formen des Liganden können an den Rezeptor (Fn14) binden. Viele verschiedene intrazelluläre Signalwege werden durch den Fn14 aktiviert, beispielweise Erk1/2, JNK, Jun und STAT3, vor allem jedoch das NFkB. Lösliches und membranständiges TWEAK zeigen eine ähnliche Aktivierungseffizienz bezüglich des alternativen NFkB-Signalwegs, wohingegen membranständiges TWEAK weit besser als lösliches TWEAK den klassischen NFkB-Signalweg aktiviert. In der vorliegenden Arbeit wurde zunächst die TWEAK-vermittelte Induzierbarkeit von verschiedenen Zielgenen des NFkB-Systems untersucht. Lösliches TWEAK zeigte einen weit schwächeren aktivierenden Effekt auf den klassischen NFkB-Signalweg als TNF, das ein sehr guter Aktivator des klassischen NFkB-Systems ist (Abb. 5, 6). Nichtsdestotrotz war TWEAK imstande eine stärkere TRAF1-Induktion als TNF herbeizuführen (Abbildung 7, 8). TRAF1 ist ein durch NFkB-System stark reguliertes Gen. Um posttranskriptionelle TRAF1-Modifikationen als Ursache für die unerwartet gute TRAF1-Induktion durch lösliches TWEAK auszuschließen, wurde die TRAF1-Expression nach Proteasom- und Caspasen-Inhibition untersucht (Abbildung 9). Dies ergab keinen Hinweis auf einen Einfluss dieser Prozessen auf der TRAF1-Expression.
Mittels des IKK2-spezifischen Inhibitor TPCA-1 wurde die TWEAK-vermittelte TRAF1-Induktion Zelltyp-abhängig gehemmt, wohingegen die TNF-vermittelte Induktion von TRAF1 in allen Zelllinien vollständig inhibiert wurde (Abbildung 13). Versuche mit dem NEDD8-aktivierenden Enzym (NAE) Inhibitor MLN4924, resultierten in einer totalen Inhibition der TRAF1-Expression in allen TWEAK- und TNF-stimulierten Zellen (Abbildung 14). Diese Befunde sprechen dafür, dass bei der TWEAK-vermittelten TRAF1-Expression beide Zweige des NFkB-Signalwegs Zelltyp-abhängig beteiligt sind.
Die Oligomerisierung der Liganden der TNF-Familie verstärkt oft ihre Aktivität. Oligomerisiertes TWEAK imitiert die biologische Aktivität von membranständigem TWEAK. Lösliches TWEAK wurde mit einem anti-Flag Antikörper oligomerisiert und die TRAF1-Induktion durch den alternativen NFkB-Signalweg wurde analysiert
Oligomerisiertes TWEAK aktivierte Zelltyp-unabhängig den klassischen NFkB-Signalweg stärker als lösliches TWEAK, wohingegen kein Effekt auf die TRAF1-Induktion oder auf die Aktivierung den alternativen NFkB-Signalweg festgestellt wurde (Abbildung 11, 12). TWEAK ist imstande die TRAF2-vermittelte CD40-Induktion des klassischen NFkB-Signalwegs zu hemmen (Abbildung 16, 17). Um den Beitrag von TWEAK-Induziertem TRAF1 zur CD40-Inhibition zu herauszufinden, wurden TRAF1-stabil transfizierte 786O- und U2OS-Zellen hergestellt (Abbildung 19). Die CD40-Induzierte IkBa-Degradation und IL8/6 Produktion war in den TRAF1-Transfektanten als auch in mit löslichem TWEAK vorbehandelte Zellen stark inhibiert (Abbildung 21, 22), wobei die CD40-Expression und CD40/CD40L-Interaktion unverändert blieb (Abbildung 20). Diese Ergebnisse sprechen für einen wichtigen Beitrag des Adaptorprotein TRAF1 in der TWEAK-vermittelte Inhibition der CD40-induzierte Aktivierung des klassischen NFkB-Signalwegs.
The mechanisms that enable cells to regulate their gene expression and thus their metabolism, proliferation or cellular behaviour are not only important to understand the basic biology of a living cell, but are also of crucial interest in cancerogenesis. Highly interwoven and tightly regulated pathways are the basis of a robust but also flexible regulatory network. Interference with these pathways can be either causative for tumorigenesis or can modify its outcome. The receptor tyrosine kinase (RTK) and RAS dependent pathways leading to AKT or ERK1/2 activation are of particular interest in melanoma. These signaling modules are commonly activated by different mutations that can be found in various pathway components like NRAS, BRAF or PTEN. The first part of this work deals with the diverse and versatile functions of the ERK1/2 pathway feedbackregulator MKP2 in different cellular, melanoma relevant settings. In addition, a functional role of the AP1-complex member FOSL1, an ERK1/2 transcriptional target being implicated in the regulation of proliferation, is demonstrated. Secondly, aspects of direct pharmacological inhibition of the ERK1/2 pathway with regard to the induction of apoptosis have been analysed. Due to the high frequency of melanoma related mutations occurring in the RAS/RAF/MEK/ERK pathway (e.g. NRASQ61K, BRAFV600E), inhibition of this signaling cascade is deemed to be a promising therapeutic strategy for the treatment of malignant melanoma. However, although in clinical trials mono-therapeutic treatment with MEK- or RAF inhibitors was successful in the short run, it failed to show satisfactory long-lasting effects. Hence, combination therapies using a MAPK pathway inhibitor and an additional therapy are currently under investigation. I was able to demonstrate that inhibition of MEK using the highly specific inhibitor PD184352 can have a protective effect on melanoma cells with regard to their susceptibility towards the apoptosis inducing agent cisplatin. Single application of cisplatin led to strong DNA damage and the induction of caspase-dependent apoptosis. Additional administration of the MEK inhibitor, however, strongly reduced the apoptosis inducing effect of cisplatin in several melanoma cell lines, These cells displayed an increased activation of the serine/threonine kinase AKT after MEK inhibition. This AKT activation concomitantly led to the phosphorylation of FOXO transcription factors, attenuating the cisplatin induced expression of the BH3-only protein PUMA. PUMA in turn was important to mediate the apoptosis machinery after cisplatin treatment. My results also indicate a participation of RTKs, in particular EGFR, in mediating MEK inhibitor induced activation of AKT. These results demonstrate that inhibition of the RAS/RAF/MEK/ERK signaling pathway in melanoma cell lines does not necessilary have favourable effects in a cytotoxic co-treatment situation. Instead, it can even enhance melanoma survival under pro-apoptotic conditions.
The prevention of restenosis after percutaneous coronary intervention is a major task for researchers and clinicians in cardiovascular pharmacology. Nearly 1.5 million PTCA are performed every year worldwide and, due to the implantation of stents, most of the cases can be treated successfully. 60% of those patients develop restenosis within 6 months. SMC migration and ECM deposition are known to be responsible for neointima formation. Among many processes, integrin initiated signalling events play a central role in SMC migration. Many integrins recognize a specific RGD sequence which is present in several ECM proteins and cell surface immunoglobulin super family molecules. Until now, there are various integrin antagonists such as antibodies, cyclic peptides, peptidomimetics, and non-peptides have been shown to interfere with such pathological situations indicating the importance of integrin initiated signalling pathways in SMC migration. Therefore, in this study SMC migration induced by ECM proteins was inhibited either using pharmacological inhibitor or by overexpressing the endogenous inhibitor of FAK by AAV vector system. In the first part of the thesis, the effect of integrin-ligand stimulation on hCASMCs was studied. The tyrosine phosphorylation of many cellular proteins was observed from serum starved hCASMCs replated on VN but not on PL coated plates. The major tyrosine phosphorylated protein was identified as FAK by immunoprecipitation and also phosphorylation was found at Tyr 397, the autophosphorylation site of FAK. Further, VN induced the dose dependent migration of hCASMCs in haptotaxis assay. The integrin v inhibitor was used to block those ECM stimulated integrin signalling pathways and cell migration. It inhibited the ECM stimulated tyrosine phosphorylation in a dose dependent manner. Interestingly, specific potent antagonism of integrin v abrogated both ECM induced haptotaxis and growth factor induced chemotaxis. The inhibition of migration is consistent with the replating assay results that show interference with integrin induced signalling pathways particularly the FAK tyrosine phosphorylation. The integrin v inhibitor also is able to interfere with hCASMC invasion through matrigel by reducing MMP-2 secretion. Importantly, integrin v inhibitor did not induce the apoptosis in hCASMCs. FAK is a key player in many cellular events and its involvement in cell migration was extensively studied in various cell types. The present study explored the function of FAK in hCASMC migration by overexpression of FRNK, the C-terminal domain of FAK. Overexpression of FRNK inhibited the in vitro SMC migration as well as the neointima formation in a porcine restenosis model in vivo. The last part of this thesis focused on the identification of putative binding partners for the N-terminal domain of FAK by bacterial two-hybrid screen. One of the interesting binding partners was a putative protein of 17.9 kDa. Its human homolog is AGS4, which acts as a GTPase activator. The preliminary results revealed that it is able to interact with N-FAK domain and its expression is high in haematopoietic cells. Taken together the above results suggest that integrin v and FAK are promising targets for inhibition of SMC migration. Disruption of FAK-mediated signalling pathways by a pharmacological inhibitor or by overexpression of FRNK, which acts as dominant-negative regulator, resulted in decreased migration of SMCs and thus can lead to reduction of neointima formation.
