The RAF family of protein kinases consists of three members, A-RAF, B-RAF and C-RAF. Unlike the other isotypes, B-RAF has been found to have an important function for normal development of the central nervous system (CNS), because newly generated embryonic neurons lacking B-RAF cannot respond to survival factors and undergo cell death in vitro. A second cell lineage affected by the absence of B-RAF are endothelial cells and their death leads to internal bleedings and lethality of B-RAF-/- mice between embryonic day 10.5 (E10.5) and E12.5 precluding an opportunity to further analyze neural B-RAF function at a later stage. In contrast to B-RAF-/- mice, B-RAFKIN/KIN mice, which are B-RAF deficient but express a chimeric protein consisting of the unique N terminus of B-RAF and all the domains of A-RAF in the B-RAF gene locus, survive after midgestation because their endothelial cells are protected from apoptosis. More importantly, overall prevention of abnormal neural apoptosis in the forebrain allows us to study proliferation- or differentiation-oriented function of B-RAF other than its survival effects in CNS development. The detailed investigation of B-RAFKIN/KIN animals was concentrated on cortical development. There were apparent cortical defects in B-RAFKIN/KIN forebrain: Loss of B-RAF led to severe reduction of Brn-2 expressing pyramidal projection neurons accompanied by a disruption of dendrite formation in the upper layers. In further analysis, BrdU labelling experiments showed that from E14.5 to E16.5 cell proliferation in the ventricular zone of the mutant mice was reduced and that the late-born cortical neurons failed to migrate properly. While the proliferation defect of cortical progenitors was associated with reduced ERK activation, the mechanism causing impaired neuronal migration remains to be determined. Our hypothesis is that the subcellular localization of phospho-ERK may be altered in migrating cortical neurons in B-RAFKIN/KIN mice. To confirm in vivo function of B-RAF and further study unknown roles in embryonic neurogenesis as well as other morphogenesis, conditional B-RAF knockouts would be the ideal models, which can efficiently avoid embryonic lethality, prevent unwanted pleiotropic side effects and exclude accumulative compensatory developmental changes from the earliest developmental stage on, through the deletion of genetic material/gene function in selected cells at a specific time. The use of site-specific recombinases such as Cre and the successful development of the reversible tetracycline-based switch have provided powerful venues for creating conditional loss-of-function mouse models. Generation of tetracycline-regulated B-RAF and floxed B-RAF mouse embryonic stem (ES) cell lines was performed. Up to now, high-grade chimeric mice were obtained after blastocyst injection of the modified ES cell clones. The germline transmission from these chimeric mice is currently under investigation. When either of conditional mouse lines is ready, detailed examination in their CNS development would be done to reveal how B-RAF plays a real role for normal development of the nervous system.
Cellular proliferation, differentiation and survival in response to extracellular signals are controlled by the signal transduction pathway of Ras, Raf and MAP kinase. The Raf proteins are serine/threonine kinases with essential function in growth/differentiation/survival - related signal transduction events. In mammals, three functional (A-, B-, and C-Raf) genes were described. Biochemical studies suggest overlapping and differential utilization of Raf isozymes. However, the frequent co-expression of Raf isozymes and their multiple activators and effectors impedes the full understanding of their specific roles. The elucidation of these roles is important due to the involvement of the Ras/Raf/MEK/MAP kinase cascade in human disorders especially in tumor development and progression. B-Raf was shown to posses the strongest kinase activity among Raf kinases and display antiapoptotic properties. Mice deficient in B-Raf show overall growth retardation and die between E10.5 and E12.5 of vascular defects caused by excessive death of differentiated endothelial cells. To elucidate the redundancy of Raf isozymes during embryonic development and to rescue B-Raf-/- (KO) phenotype, B-Raf alleles were disrupted by introducing A-Raf cDNA under the control of endogenous B-Raf promoter. The resulting BRaf A-Raf/A-Raf (KIN) phenotype depends on genetic background. The living embryos displaying normal development but size reduction were found with low incidence at E12.5d-16.5d. All of them displayed the rescue of vascular system. One adult p20 mouse without any visible defects in development and behavior was obtained. On the other hand, the processes of neurogenesis and neural precursors migration in survived embryos were disturbed which led in some cases to underdevelopment of different brain compartments. TUNEL and cell proliferation (PCNA staining) assays revealed more apoptotic (E13.5d) and less proliferating(E12.5d cells within ventricular and sub-ventricular zones of brain ventricles and in striatum of KIN embryos. In addition, more apoptotic cells were detected in many other tissues of E13.5d and in lung of E16.5d KIN embryos but not in adult KIN mouse. p20 KIN mouse demonstrated reduced fraction of neural precursor cells in sub-granular zone of hippocampus and mature neurons in olfactory bulb. The other processes of neurogenesis were not disturbed in adult KIN animal. Fibroblasts obtained from KIN embryos demonstrated less proliferative ability and were more susceptible to apoptotic stimuli compared to WT. This was accompanied by the reduction of active ERK and Akt required for survival, and with decrease of inactive phosphorylated BAD. The kinetic of both ERK and Akt phosphorylation upon serum stimulation was delayed. All these data indicate that moderate A-Raf kinase activity can prevent the endothelial apoptosis but is not enough to completely rescue the other developmental consequences.
Spontaneous neural activity has been shown to regulate crucial events in neurite growth including axonal branching and path finding. In animal models of spinal muscular atrophy (SMA) cultured embryonic mouse motoneurons show distinct defect in axon elongation and neural activity. This defect is governed by abnormal clustering of Ca2+ channels in the axonal regions and the protruding growth cone area. The mechanisms that regulate the opening of calcium channels in developing motoneurons are not yet clear. The question was addressed by blocking neural activity in embryonic cultured motoneurons by pharmacological inhibition of voltage-gated sodium channels (VGSC) by saxitoxin (STX) and tetrodotoxin (TTX). Low dosages of STX resulted in significant reduction of axon growth and neural activity in cultured motoneurons. This pharmacological treatment did not affect survival of motoneurons in comparison to control motoneurons that was grown in the presence of survival neurotrophic factors BDNF and CNTF. It was also found that STX was 10 times more potent than TTX a common inhibitor of VGSC with a reduced activity on the TTX-insensitive sodium channels NaV1.5, NaV1.8 and NaV1.9. Reverse Transcriptase-PCR experiments revealed the presence of NaV1.9 as the likely candidate that begins to express from embryonic stage sixteen in the mouse spinal cord. Immunolabelling experiments showed that the channel is expressed in the axonal compartments and axonal growth cones in cultured motoneurons. Suppression of NaV1.9 in cultured motoneurons by lentivirus mediated short hairpin-RNA (shRNA) resulted in shorter axon length in comparison with uninfected and scrambled constructs. Further, embryonic motoneurons cultured from NaV1.9 knockout mice also showed a significant reduction in neural activity and axon growth. The findings of this work highlight the role of NaV1.9 as an important contender in regulating activity dependent axon growth in embryonic cultured motoneurons. NaV1.9 could therefore be considered as a prospective molecule that could play an important role in regulating axon growth in motoneuron disease models like spinal muscular atrophy (SMA).

Members of the T-box gene family encode transcription factors that play key roles during embryonic development and organogenesis of invertebrates and vertebrates. The defining feature of T-box proteins is an about 200 aa large, conserved DNA binding motif, the T domain. Their importance for proper development is highlighted by the dramatic phenotypes of T-box mutant animals. My thesis was mainly focused on two Drosophila T-box genes, optomotor-blind (omb) and optomotor-blind related 1 (org-1), and included (i) a genetic analysis of org-1 and (ii) the identification of molecular determinants within OMB and ORG-1 that confer functional specificity. (i) Genetic analysis of org-1 initially based on a behavioral Drosophila mutant, C31. C31 is a X-linked, recessive mutant and was mapped to 7E-F, the cytological region of org-1. This pleiotropic mutant is manifested in walking defects, structural aberrations in the central brain, and "held-out" wings. Molecular analysis revealed that C31 contains an insertion of a 5' truncated I retrotransposon within the 3' untranslated transcript of org-1, suggesting that C31 might represent the first org-1 mutant. Based on this hypothesis, we screened 44.500 F1 female offspring of EMS mutagenized males and C31 females for the "held-out" phenotype, but failed to isolate any C31 or org-1 mutant, although this mutagenesis was functional per se. Since we could not exclude the possibility that our failure is due to an idiosyncracy of C31, we intended not to rely on C31 in further genetic experiments and followed a reverse genetic strategy . All P element lines cytologically mapping to 7E-7F were characterized for their precise insertion sites. 13 of the 19 analyzed lines had P element insertions within a hot-spot 37 kb downstream of org-1. No P element insertions within org-1 could be identified, but several P element insertions were determined on either side of org-1. The org-1 nearest insertions were used for local-hop experiments, in which we associated 6 new genes with P insertions, but failed to target org-1. The closest P elements are still 10 kb away from org-1. Subsequently, we employed org-1 flanking P elements to induce precise deletions in 7E-F spanning org-1. Two org-1 flanking P elements were brought together on a recombinant chromosome. Remobilization of P elements in cis configuration frequently results in deletions with the P element insertion sites as deficiency endpoints. In a first attempt, we expected to identify deficiencies by screening for C31 alleles. 8 new C31 alleles could be isolated. The new C31 chromosomes, however, did not carry the desired deletion. Molecular analysis indicated that C31 is not caused by aberrations in org-1, but by mutations in a distal locus. We repeated the P element remobilization and screened for the absence of P element markers. 4 lethal chromosomes could be isolated with a deletion of the org-1 locus. (ii) The consequences of ectopic org-1 were analyzed using UAS-org-1 transgenic flies and a number of different Gal4 driver lines. Misexpression of org-1 during imaginal development interfered with the normal development of many organs and resulted in flies with a plethora of phenotypes. These include a homeotic transformation of distal antenna (flagellum) into distal leg structures, a strong size reduction of the legs along their proximo-distal axis, and stunted wings. Like ectopic org-1, ectopic omb leads to dramatic changes of normal developmental pathways in Drosophila as well. dpp-Gal4/ UAS-omb flies are late pupal lethal and show an ectopic pair of wings and largely reduced eyes. GMR-Gal4 driven ectopic omb expression in the developing eye causes a degeneration of the photoreceptor cells, while GMR-Gal4/ UAS-org-1 flies have intact eyes. Hence, ectopic org-1 and omb induce profound phenotypes that are qualitatively different for these homologous genes. To begin to address the question where within OMB and ORG-1 the specificity determinants reside, we conceptionally subdivided both proteins into three domains and tested the relevance ofthese domains for functional specificity in vivo. The single domains were cloned and used as modules to assemble all possible omb-org-1 chimeric trans- genes. A method was developed to determine the relative expression strength of different UAS-transgenes, allowing to compare the various transgenic constructs for qualitative differences only, excluding different transgene quantities. Analysis of chimeric omb-org-1 transgenes with the GMR-Gal4 driver revealed that all three OMB domains contribute to functional specificity.
Mutationen im humanen Emerin-Gen verursachen beim Menschen eine angeborene Muskelschwäche, die X-gebundene Emery-Dreifuss. Der Phänotyp dieser Störung manifestiert sich in der zweiten und dritten Lebensdekade durch Verkürzungen der Nacken , Ellenbogen- und Achillessehnen, progressiven Muskelschwund am Oberkörper sowie Störung der Reizweiterleitung und eine Kardiomyopathie. Zwar wurden die Funktionen dieses ubiquitären Kernmembranproteins bislang intensiv erforscht, allerdings blieben die krankheitsverursachenden Mechanismen, die für den späten Ausbruch der gewebespezifischen Erkrankung verantwortlich sind, noch weitestgehend unverstanden. Um Erkenntnisse über die pathologische(n) Funktion(en) des integralen Membranproteins Emerin zu gewinnen, wurde dessen spatio-temporäre Transkriptions- und Expressionsmuster während der frühen Embryonalentwicklung im Modellsystem Xenopus laevis charakterisiert. Durch EST-Datenbankanalysen konnten in der pseudotetraploiden Spezies zwei Emerin-Gene (Xemerin1 und -2) identifiziert werden. Im Unterschied zu dem längeren Säuger-Emerin (254 Reste bei Homo sapiens ) konnte allerdings kein Kernlokalisationssignal und auch kein serinreicher Sequenzbereich festgestellt werden. Durch Herstellung monoklonaler Antikörper wurde die subzelluläre und gewebespezifische Lokalisation der Xemerin-Proteine untersucht. Interessanterweise war Xemerin weder in der Immunfluoreszenz noch im Immunblot in Oozyten nachweisbar. Mit dem zweidimensionalen Gelektrophorese-Verfahren NEPHGE konnte gezeigt werden, dass der von uns hergestellte monoklonale Antikörper 59/7 beide Xemerin-Formen erkannte und die Proteine durch unterschiedliche molekulare Massen und isoelektrische Punkte voneinander zu trennen waren. Durch Immunoblotting embryonaler Proteine aus unterschiedlichen Entwicklungsstadien konnte gezeigt werden, dass Xemerin1 und -2 im Laufe der Embryogenese von Xenopus laevis erstmals im Entwicklungsstadium 43 exprimiert werden. Unerwarteterweise konnte durch RT-PCR-Analysen eine Aktivität der Xemerin-Gene während der gesamten Embryogenese belegt werden. Northernblot- und Sequenzanalysen der Xemerin-mRNA zeigten außerordentlich große untranslatierte Bereiche mit snRNP-Bindungsmotiven. Durch zwei voneinander unabhängige Analyseverfahren wurde festgestellt, dass die Xemerin-Genaktivität ab dem Stadium 30 deutlich zunahm. Äußerst interessant war in diesem Zusammenhang die Beobachtung, dass exakt zu diesem Zeitpunkt die Aktivität des XMAN1-Gens, einem weiteren Protein der inneren Kernmembran, signifikant herunterreguliert wurde. Whole-mount in situ Hybridisierungsversuche zeigten einen Xemerin-Expressionsschwerpunkt in neuro-ektodermalen Geweben von Tadpole-Embryonen, wie dies auch von XMAN1 (auch SANE genannt) berichtet wurde. Aufgrund dieser Erkenntnisse wurde angenommen, dass Xemerin und XMAN1 überlappende Funktionen aufweisen. Durch die Herstellung rekombinanter Fusionproteine konnte gezeigt werden, dass XMAN1 eine identische subzelluläre Verteilung wie Xemerin aufwies. In vitro Bindungsassays wiesen eine direkte Wechselwirkung von XMAN1 mit beiden Xemerin-Formen sowie mit Xenopus Lamin A nach. Diese Arbeit konnte durch die Charakterisierung von Xenopus Emerin die Grundlagen für weitere intensive Forschungen legen und zeigt eindeutig, dass das Modellsystem Xenopus laevis mit dem Säugermodell Maus konkurrenzfähig ist, um die krankheitsverursachende Mechanismen der Emery-Dreifuss Muskeldystrophie aufzuklären.
Das Ziel der vorliegenden Arbeit war die Charakterisierung der molekularen Grundlagen der Proepikardentwicklung im Huhn. Das Proepikard (PE) stellt die Vorläuferpopulation des Koronargefäßsystems dar. Es wurden zwei Schwerpunkte bearbeitet, zum einen die Rolle von einem Mitglied der TGFß-Superfamilie (BMP, bone morphogenetic protein) während der Induktionsphase des PE und zweitens die molekularen Mechanismen, welche der links/rechts Asymmetrie der PE Entwicklung zugrunde liegen. Die Expressionsmuster von TBX18, WT1, CFC weisen diese als proepikardiale Marker aus. BMP4 wird ebenfalls im PE exprimiert, wenngleich wesentlich schwächer als BMP2 im benachbarten Sinusmyokard. Durch in vitro Experimente mit proepikardialen Primärkulturen wurde festgestellt, dass die Zugabe von rekombinatem BMP2 einen Teil der Zellen zu Kardiomyozyten differenzieren lässt. Daher wird angenommen, dass ein Teil der proepikardialen Zellen konzentrationsabhängig auf Wachstumsfaktoren reagieren und in der Lage sind, die epikardiale Linie zu verlassen und ein myokardiales Schicksal anzunehmen. Im zweiten Teil der Arbeit wurde die Rolle des NODAL-PITX2-Signalweges in Bezug auf die Unilateralität des Proepikards im Hühnchen analysiert. Dazu wurden Manipulationen des Hensenschen Knotens im HH Stadium 4 durchgeführt, mit dem Ziel linksseitige Markergene auf der rechten Seite ektopisch zu induzieren. Keine dieser Manipulationen, als auch eine virale Überexpression von PITX2 in sinuatrialen Progenitoren in vivo, liess eine Veränderung der TBX18-Lateralität erkennen, obwohl die PITX2-Expression randomisiert war. Dagegen führte die Inhibition des asymmetrischen Ionenfluxes sowie der Apoptose im Primitivstreifen zu einer völligen Randomisierung der TBX18-Expression, sowie unter anderem der Entwicklung bilateraler Proepikardien. Die Induktion bilateraler TBX18-Expressionsdomänen war ebenfalls durch ektopisches FGF8 auf der linken Seite des Knotens zu beobachten. Der Verlust der rechtsseitigen FGF Signalgebung im Knoten resultierte in einem Verlust der rechtsseitigen TBX18-Expression im Sinus. Daraus kann geschlossen werden, dass proepikardiale Progenitoren nicht nur durch den asymmetrischen Ionenflux sowie Apoptose im Primitivstreifen in ihrer Lateralität festgelegt werden, sondern auch von rechtsseitigen FGF-Signalen im Knoten und unabhängig vom linkseitigen NODAL-PITX2-Signalweg lateralisiert werden.
The experimental work of this thesis addresses the questions of whether established cell lines injected into murine blastocysts find their way back home and seed preferentially at the site of their origin. Furthermore, can they change their fate and differentiate to unrelated cell types when exposed to the embryonic environment. This survey was based on the fact that different cell lines have different potentials in developing embryos, dependent on their cellular identity. The cell lines used in this survey were AGM region-deriving DAS 104-4, DAS 104-8 cells, yolk sac-deriving YSE cells and bone marrow-deriving FDCP mix cells. These cells were injected into mouse blastocysts. Donor cells were traced in developing embryos via specific markers. Analysis of the embryos revealed that DAS cells are promiscuous in their seeding pattern, since they were found in all analysed tissues with similar frequencies. YSE cells showed preferences in seeding yolk sac and liver. YSE donor cells in chimaeric tissues were not able to change their immuno-phenotype, indicating that they did not change their destiny. Analysis of adult mice did not reveal any of YSE-derived cells donor contribution. In contrast, FDCP mix cells mostly engrafted haematopoietic tissues, although the embryos analysed by in situ hybridization had donor signals frequently in cartilage primordia, heads, and livers. Analysis of whether FDCPmix-derived cells found in foetal livers were of haematopoietic or hepatocytes nature showed that progeny of injected FDCP mix cells do not differentiate into cells that express a hepatocyte-specific marker. Further analysis showed that FDCPmix-derived donor cells found in brain express neural or haematopoietic markers. In order to reveal if they transdifferentiate to neurons or fuse with neurons/glial cells, nuclear diameters of donor and recipient cells were determined. Comparison of the nuclear diameters of recipient and donor cells revealed no differences. Therefore this suggests that progeny of FDCP mix in brain are not fusion products. Analysis of adult mice tissues revealed that presence of FDCP mix-derived cells was the highest in brains. These results confirmed the assumption that the developmental potential of the analysed cells cannot be easily modified, even when exposed to early embryonic environment. Therefore one can conclude that the analysed cell types had different homing patterns depending on their origins.
Der Notch Signalweg spielt während der Embryonalentwicklung eine zentrale Rolle in der Spezifizierung des Zellschicksales, der Proliferation und der Kommunikation benachbarter Zellen. Die Hey bHLH Transkriptionsfaktoren sind Zielgene des Notch-Signalweges und besitzen wichtige Funktionen in der kardiovaskulären Entwicklung. Hey2 Knockout (KO) Mäuse und Hey1/HeyL Doppelknockout-Mäuse (DKO) sind gekennzeichnet durch eine fehlerhafte Ausbildung der Herzscheidewand und der Herzklappen und durch eine unzureichende Differenzierung während der Blutgefäßentwicklung. Ziel dieser Arbeit war es, neue Zielgene der Hey Proteine zu finden, um ihre Funktion in der Organentwicklung und die Ausprägung der Hey KO Maus-Phänotypen besser verstehen zu können. Dazu wurde als Methode eine Kombination aus Microarray-Analyse und Chromatinimmunpräzipitation (ChIP) gewählt, um gleichzeitig einen Überblick über die regulierten Zielgene und der direkt gebundenen Promotoren zu gewinnen. Als Zellkulturmodell wurden HEK293-Zellen genutzt, die doxyzyklin-induzierbar Flag-markiertes Hey1, bzw. Hey2 Protein überexprimieren. Eine Microarray-Analyse nach Überexpression von Hey1, bzw. Hey2 ergab insgesamt ca. 100 bis zu 5-fach herunterregulierte Zielgene und nur für Hey2 15 Gene, die stärker als 2-fach hochreguliert waren. Eine ChIP mit αFlag-Antikörper zeigte eine direkte DNA-Bindung von Hey1, bzw. Hey2, im proximalen Promotorbereich von 4 herunterregulierten Zielgenen (HEY1, BMP2, KLF10 und FOXC1). Ist jedoch die DNA-bindende basische Domäne des Hey1-Proteins deletiert, bzw. durch Aminosäureaustausche (3 Arginine zu 3 Lysine) vermutlich nicht mehr DNA-bindend, kann eine Herunterregulation der Zielgene nach Überexpression der Hey1-Mutanten nicht mehr festgestellt werden. Ebenso kann eine Bindung der Hey1-Mutanten an die ausgewählten Promotoren von HEY1, BMP2, KLF10 oder FOXC1 mit ChIP nicht mehr nachgewiesen werden. Dies deutet darauf hin, dass die basische Domäne essentiell für die DNA-Bindung und für die Funktion der Hey Proteine ist. Mit ChIP-PET und anschließender Hochdurchsatz-Sequenzierung wurde ein genomweiter Screen der Hey1- und der Hey2-Bindungsstellen in HEK293-Zellen durchgeführt. Für Hey1 wurden 1453 Zielgene, für Hey2 4288 Zielgene bestimmt, wobei 1147 Gene gemeinsame Zielgene von Hey1 und Hey2 waren. Obwohl die Bindungsstellen in 5'- und 3'-Richtung von kodierenden Sequenzen und auch in Exons und Introns lokalisiert waren, waren 55 %, bzw. 49 % aller Bindungsstellen für Hey1, bzw. Hey2 im proximalen Promotorbereich von -0,5 kb und im ersten Exon lokalisiert. Eine in silico Analyse des Bindemotivs deutete auf eine repetitive GC-haltige Sequenz hin, die vermutlich in CpG Inseln lokalisiert ist. Diese Ergebnisse weisen auf eine direkte Regulation der Transkriptionsmaschinerie durch die Hey Proteine hin. Ein Vergleich der Zielgene aus den Microarray-Analysen mit den ChIP-PET Daten zeigte einen hohen Anteil an herunterregulierten Genen mit Bindestellen, die direkt von Hey gebunden waren. Während 60 % der herunterregulierten Hey2 Zielgene in der ChIP-PET Analyse eine direkte DNA-Bindung zeigen, weisen nur 20 % der hochregulierten Gene Bindestellen für Hey2 auf. Dies spricht für eine überwiegende Repressorfunktion der Hey Proteine. Um zu überprüfen, inwieweit die Hey Proteine zelltypspezifisch verschiedene Zielgene regulieren, wurden embryonale Stammzellen (ES-Zellen) generiert, die ebenfalls doxyzyklin-induzierbar Hey1, bzw. Hey2 überexprimieren. Diese ES-Zellen konnten effektiv zu Kardiomyozyten differenziert werden, so dass auch in diesen Zellen eine Hey Überexpression induziert und somit eine Genexpressionsanalyse durchgeführt werden konnte. Microarray Analysen der ES-Zellen und Kardiomyozyten ergaben mehr hoch- als herunterregulierte Gene im Vergleich zu HEK293-Zellen. Die Überlappung an gemeinsam regulierten Zielgenen in HEK293, ES-Zellen und Kardiomyozyten war sehr gering. Nur zwei Hey2-Zielgene wurden gleichzeitig in HEK293 und ES-Zellen stärker als 2-fach reguliert (Hes1, Zic2). Diese geringe Überlappung deutet auf ein enges zelltypspezifische Regulationspotential hin. Eine Genontologie-Analyse aller Zielgene zeigte Interaktionen der Hey Proteine mit verschiedenen Signalwegen (z.B. TGFβ-, Id- oder Wnt-Signalweg), die alle unersetzlich in frühen Entwicklungsprozessen sind. Diese Ergebnisse deuten darauf hin, dass die Hey Proteine zelltypspezifisch die Expression von Genen aus verschiedenen Signalwegen beeinflussen und modulieren können. Weiterhin eröffnen diese Daten neue Möglichkeiten für zukünftige Forschung, um die Rolle der Hey Proteine in der frühen Organentwicklung genauer ergründen.
Hey-mutant mouse hearts at embryonic day E14.5 were shown to react to the knock out of Hey2 with several up-regualted genes. This up-regulation is due to the lack of Hey2 and cannot be explained by the structural changes in heart morphology as shown using control animals. Part of the gene regulation was further validated using in situ hybridization. Hey1 was located to the nucleus in immunofluorescence experiments. However, experiments on protein level showed also amount of Hey1 within the cytoplasm. The nuclear localization of Hey1 was unchanged during all cell cycle phases as well as when CaMKII was co-expressed or other cellular pathways were inhibited or stimulated. Hey1 does not seem to interact with the nuclear transport proteins importin-alpha and -beta, therefore it still needs to be elucidated how Hey1 is transported into the nucleus.
