Die aktuellen Veränderungen der Arbeitswelt erfordern eine stetige Anpassung an immer neue Herausforderungen seitens der Arbeitgeber und Arbeitnehmer. Lebenslanges Lernen und damit eine kontinuierliche Weiterbildung der Mitarbeiter ist essentiell für Unternehmen, um auf dem schnelllebigen Arbeitsmarkt wettbewerbsfähig zu sein. Die Bereitschaft und die Motivation von Menschen, dazuzulernen, ist jedoch sehr unterschiedlich. Eine mögliche Erklärung dafür liefert die dispositionelle Zielorientierung, welche der Achievement-Goal-Theorie entstammt. Das Konstrukt beschreibt, ob Menschen eine individuelle Präferenz für Lernziele (z.B. Kompetenzzuwachs) oder Leistungsziele (z.B. gute Beurteilungen bekommen oder schlechte Beurteilungen vermeiden) haben. Neben den Persönlichkeitsaspekten konzentriert sich die Forschung im Rahmen der Achievement-Goal-Theorie auch auf den Einfluss der Umgebung auf Lern- und Leistungsprozesse. Die sogenannte arbeitsplatzbezogene Zielorientierung beschreibt die wahrgenommene Zielstruktur der Arbeitsumgebung und stellt das situationsbedingte Gegenstück der Dispositionen dar. Zahlreiche Befunde aus dem Bereich der pädagogischen Psychologie zu Zielstrukturen der Umgebung zeigen einen Einfluss auf beispielsweise Lernerfolg, Motivation, Selbstregulationsprozesse oder Leistung. Zielstrukturen im Arbeitskontext stellen hingegen ein bisher wenig beachtetes Konstrukt dar. Ausgehend von den aktuellen Befunden zu Zielstrukturen, könnte die arbeitsplatzbezogene Zielorientierung jedoch einen wichtigen Beitrag leisten, wenn es um die Frage geht, wie Mitarbeiter unterstützt und zu Lernprozessen angeregt werden können. Die Identifizierung von lern- und leistungsförderlichen Zielstrukturen der Arbeitsumgebung würde wertvolle Ansatzpunkte für die Mitarbeiterentwicklung in der betrieblichen Praxis liefern.
Im Rahmen von drei empirischen Studien wird der Relevanz der Zielorientierung im Arbeits-kontext nachgegangen. Neben der Überprüfung eines ins Deutsche übertragenen und angepassten Mess-instruments zur Erhebung der arbeitsplatzbezogenen Zielorientierung (Studie 1) steht vor allem die Untersuchung möglicher Einflüsse des Konstrukts auf arbeitsrelevante Variablen im Vordergrund, um förderliche Zielstrukturen zu identifizieren (Studie 1 & 2). Darüber hinaus werden erstmalig mögliche Person-Situation-Interaktionen in diesem Zusammenhang untersucht (Studie 2). Abschließend erfolgt eine Untersuchung möglicher Antezedenten der arbeitsplatzbezogenen Zielorientierung, woraus sich erste wichtige Anhaltspunkte für Interventionsmaßnahmen am Arbeitsplatz ableiten lassen (Studie 3).
Die Ergebnisse der Studien zeigen, dass insbesondere ein lernzielorientierter Arbeitsplatz förderlich für die untersuchten Variablen wie Lernerfolg, Leistung oder auch berufliche Selbstwirksamkeit ist. In Bezug auf die Interaktion von Person und Situation ergaben sich gemischte Befunde, die kein eindeutiges Interaktionsmuster aufweisen. Bei der Frage, wie ein lernzielorientierter Arbeitsplatz gefördert werden kann, erwies sich vor allem die Art und Weise, wie Führungskräfte mit Fehlern umgehen, als relevant. Die Studien liefern demnach wichtige erste Ansätze für theoretische und praktische Implikationen, wie Mitarbeiter in Lern- und Leistungsprozessen unterstützt werden können.
Stomata are pores in the leaf surface, formed by pairs of guard cells. The guard cells modulate the aperture of stomata, to balance uptake of CO2 and loss of water vapor to the atmosphere. During drought, the phytohormone abscisic acid (ABA) provokes stomatal closure, via a signaling chain with both Ca2+-dependent and Ca2+-independent branches. Both branches are likely to activate SLAC1-type (Slow Anion Channel Associated 1) anion channels that are essential for initiating the closure of stomata. However, the importance of the Ca2+-dependent signaling branch is still debated, as the core ABA signaling pathway only possesses Ca2+-independent components. Therefore, the aim of this thesis was to address the role of the Ca2+-dependent branch in the ABA signaling pathway of guard cells.
In the first part of the thesis, the relation between ABA-induced Ca2+ signals and stomatal closure was studied, with guard cells that express the genetically encoded Ca2+-indicator R-GECO1-mTurquoise. Ejection of ABA into the guard cell wall rapidly induced stomatal closure, however, only in ¾ of the guard cells ABA evoked a cytosolic Ca2+ signal. A small subset of stomata (¼ of the experiments) closed without Ca2+ signals, showing that the Ca2+ signals are not essential for ABA-induced stomatal closure. However, stomata in which ABA evoked Ca2+ signals closed faster as those in which no Ca2+ signals were detected. Apparently, ABA-induced Ca2+ signals enhance the velocity of stomatal closure. In addition to ABA, hyperpolarizing voltage pulses could also trigger Ca2+ signals in wild type guard cells, which in turn activated S-type anion channels. However, these voltage pulses failed to elicit S-type anion currents in the slac1/slah3 guard cells, suggesting that SLAC1 and SLAH3 contribute to Ca2+-activated conductance. Taken together, our data indicate that ABA-induced Ca2+ signals enhance the activity of S-type anion channels, which accelerates stomatal closure.
The second part of the thesis deals with the signaling pathway downstream of the Ca2+ signals. Two types of Ca2+-dependent protein kinase modules (CPKs and CBL/CIPKs) have been implicated in guard cells. We focused on the protein kinase CIPK23 (CBL-Interacting Protein Kinase 23), which is activated by the Ca2+-dependent protein CBL1 or 9 (Calcineurin B-Like protein 1 or 9) via interacting with the NAF domain of CIPK23. The CBL1/9-CIPK23 complex has been shown to affect stomatal movements, but the underlying molecular mechanisms remain largely unknown. We addressed this topic by using an estrogen-induced expression system, which specifically enhances the expression of wild type CIPK23, a phosphomimic CIPK23T190D and a kinase dead CIPK23K60N in guard cells. Our data show that guard cells expressing CIPK23T190D promoted stomatal opening, while CIPK23K60N enhanced ABA-induced stomatal closure, suggesting that CIPK23 is a negative regulator of stomatal closure. Electrophysiological measurements revealed that the inward K+ channel currents were similar in guard cells that expressed CIPK23, CIPK23T190D or CIPK23K60N, indicating that CIPK23-mediated inward K+ channel AKT1 does not contribute to stomatal movements. Expression of CIPK23K60N, or loss of CIPK23 in guard cells enhanced S-type anion activity, while the active CIPK23T190D inhibited the activity of these anion channels. These results are in line with the detected changes in stomatal movements and thus indicate that CIPK23 regulates stomatal movements by inhibiting S-type anion channels. CIPK23 thus serves as a brake to control anion channel activity. Overall, our findings demonstrate that CIPK23-mediated stomatal movements do not depend on CIPK23-AKT1 module, instead, it is achieved by regulating S-type anion channels SLAC1 and SLAH3.
In sum, the data presented in this thesis give new insights into the Ca2+-dependent branch of ABA signaling, which may help to put forward new strategies to breed plants with enhanced drought stress tolerance, and in turn boost agricultural productivity in the future.
The present thesis introduce different synthetic strategies towards a variety of polycyclic aromatic dicarboximides (PADIs) with highly interesting and diverse properties. This included tetrachlorinated, tetraaryloxy- and tetraaryl-substituted dicarboximides, fused acceptor‒donor(‒acceptor) structures as well as sterically shielded rylene and nanographene dicarboximides. The properties and thus the disclosure of structure‒property relationships of the resulting dyes were investigated in detail among others with UV‒vis absorption spectroscopy, fluorescence spectroscopy, cyclic voltammetry and single crystal X-ray analysis. For instance, some of the fused and substituted PADIs offer strong absorption of visible and near infrared (NIR) light, NIR emission and low-lying LUMO levels. On the contrary, intriguing optical features in the solid-state characterize the rylene dicarboximides with their bulky N-substituents, while the devised sterically enwrapped nanographene host offered remarkable complexation capabilities in solution.
Einflüsse der Photophysik und Photochemie von Cyaninfarbstoffen auf die Lokalisationsmikroskopie

In den letzten Jahren haben sich hochauflösende Fluoreszenzmikroskopiemethoden, basierend auf der Lokalisation einzelner Fluorophore, zu einem leistungsstarken Werkzeug etabliert, um Fluoreszenzbilder weit unterhalb der Auflösungsgrenze zu generieren. Hiermit können räumliche Auflösungen von ~ 20 nm erzielt werden, was weit unterhalb der Beugungsgrenze liegt. Dabei haben zahlreiche Optimierungen und Entwicklungen neuer Methoden in der Einzelmolekül-Lokalisationsmikroskopie die Genauigkeit der orstspezifischen Bestimmung einzelner Fluorophore auf bis zu ~ 1 – 3 nm erhöht. Eine Auflösung im molekularen Bereich, weit unterhalb von ~ 10 nm bleibt allerdings herausfordernd, da die Lokalisationsgenauigkeit nur ein Kriterium hierfür ist. Allerdings wurde sich in den letzten Jahren überwiegend auf die Verbesserung dieses Parameters konzentriert. Weitere Kriterien für die fluoreszenzmikroskopische Auflösung sind dabei unter anderem die Markierungsdichte und die Kopplungseffizienz der Zielstruktur, sowie der Kopplungsfehler (Abstand zur Zielstruktur nach Farbstoffkopplung), die sich herausfordernd für eine molekulare Auflösung darstellen. Auch wenn die Kopplungseffizienz und -dichte hoch und der Kopplungsfehler gering ist, steigt bei Interfluorophordistanzen < 5nm, abhängig von den Farbstoffen, die Wahrscheinlichkeit von starken und schwachen Farbstoffwechselwirkungen und damit von Energieübertragungsprozessen zwischen den Farbstoffen, stark an. Daneben sollten Farbstoffe, abhänging von der Lokalisationsmikroskopiemethode, spezifische Kriterien, wie beispielsweise die Photoschaltbarkeit bei dSTORM, erfüllen, was dazu führt, dass diese Methoden häufig nur auf einzelne Farbstoffe beschränkt sind. In dieser Arbeit konnte mithilfe von definierten DNA-Origami Konstrukten gezeigt werden, dass das Blinkverhalten von Cyaninfarbstoffen unter dSTORM-Bedingungen einer Abstandsabhängigkeit aufgrund von spezifischen Energieübertragungsprozessen folgt, womit Farbstoffabstände im sub-10 nm Bereich charakterisiert werden konnten. Darüber hinaus konnte diese Abstandsabhängigkeit an biologischen Proben gezeigt werden. Hierbei konnten verschiedene zelluläre Rezeptoren effizient und mit geringem Abstandsfehler zur Zielstruktur mit Cyaninfarbstoffen gekoppelt werden. Diese abstandsabhänigen Prozesse und damit Charakterisierungen könnten dabei nicht nur spezifisch für die häufig unter dSTORM-Bedingungen verwendeten Cyaninfarbstoffen gültig sein, sondern auch auf andere Farbstoffklassen, die einen Auszustand zeigen, übertragbar sein. Darüber hinaus konnte gezeigt werden, dass hochauflösende dSTORM Aufnahmen unabhängig vom Farbstoffkopplungsgrad der Antikörpern sind, welche häufig für Standardfärbungen von zellulären Strukturen verwendet werden. Dabei konnte durch Photonenkoinzidenzmessungen dargelegt werden, dass aufgrund komplexer Farbstoffwechselwirkungen im Mittel nur ein Farbstoff aktiv ist, wobei höhere Kopplungsgrade ein komplexes Blinkverhalten zu Beginn der Messung zeigen. Durch die undefinierten Farbstoffabstände an Antikörpern konnte hier kein eindeutiger Energieübertragungsmechanismus entschlüsselt werden. Dennoch konnte gezeigt werden, dass Farbstoffaggregate bzw. H-Dimere unter dSTORM-Bedingungen destabilisiert werden. Durch die zuvor erwähnten DNA-Origami Konstrukte definierter Interfluorophordistanzen konnten Energieübertragungsmechanismen entschlüsselt werden, die auch für die Antikörper diverser Kopplungsgrade gültig sind. Des Weiteren konnten, ausgelöst durch komplexe Energieübertragungsprozesse höherer Kopplungsgrade am Antikörper, Mehrfarbenaufnahmen zellulärer Strukturen generiert werden, die über die spezifische Fluoreszenzlebenszeit separiert werden konnten. Dies stellt hier eine weitere Möglichkeit dar, unter einfachen Bedingungen, schnelle Mehrfarbenaufnahmen zellulärer Strukturen zu generieren. Durch die Verwendung des selben Farbstoffes unterschiedlicher Kopplungsgrade kann hier nur mit einer Anregungswellenlänge und frei von chromatischer Aberration gearbeitet werden. Neben den photophysikalischen Untersuchungen der Cyaninfarbstoffe Cy5 und Alexa Fluor 647 wurden diese ebenso photochemisch näher betrachtet. Dabei konnte ein neuartiger chemischer Mechanismus entschlüsselt werden. Dieser Mechanismus führt, ausgelöst durch Singulett-Sauerstoff (1O2), zu einer Photozerschneidung des konjugierten Doppelbindungssystems um zwei Kohlenstoffatome, was zu strukturellen und spektroskopischen Veränderungen dieser Farbstoffe führt. Auf Grundlage dieses Mechanismus konnte eine neue DNA-PAINT Methode entwickelt werden, die zu einer Beschleunigung der Aufnahmezeit führt.
Aging is known to be a risk factor for structural abnormalities and functional decline in the nervous system. Characterizing age-related changes is important to identify putative pathways to overcome deleterious effects and improve life quality for the elderly. In this study, the peripheral nervous system of 24-month-old aged C57BL/6 mice has been investigated and compared to 12-month-old adult mice. Aged mice showed pathological alterations in their peripheral nerves similar to nerve biopsies from elderly human individuals, with nerve fibers showing demyelination and axonal damage. Such changes were lacking in nerves of adult 12-month-old mice and adult, non-aged humans. Moreover, neuromuscular junctions of 24-month-old mice showed increased denervation compared to adult mice. These alterations were accompanied by elevated numbers of macrophages in the peripheral nerves of aged mice. The neuroinflammatory conditions were associated with impaired myelin integrity and with a decline of nerve conduction properties and muscle strength in aged mice.
To determine the pathological impact of macrophages in the aging mice, macrophage depletion was performed in mice by oral administration of CSF-1R specific kinase (c-FMS) inhibitor PLX5622 (300 mg/kg body weight), which reduced the number of macrophages in the peripheral nerves by 70%. The treated mice showed attenuated demyelination, less muscle denervation and preserved muscle strength. This indicates that macrophage-driven inflammation in the peripheral nerves is partially responsible for the age-related neuropathy in mice.
Based on previous observations that systemic inflammation can accelerate disease progression in mouse models of neurodegenerative diseases, it was hypothesized that systemic inflammation can exacerbate the peripheral neuropathy found in aged mice. To investigate this hypothesis, aged C57BL/6 mice were intraperitoneally injected with a single dose of lipopolysaccharide (LPS; 500 μg/kg body weight) to induce systemic inflammation by mimicking bacterial infection, mostly via activation of Toll-like receptors (TLRs). Altered endoneurial macrophage activation, highlighted by Trem2 downregulation, was found in LPS injected aged mice one month after injection. This was accompanied by a so far rarely observed form of axonal perturbation, i.e., the occurrence of “dark axons” characterized by a damaged cytoskeleton and an increased overall electron density of the axoplasm. At the same time, however, LPS injection reduced demyelination and muscle denervation in aged mice. Interestingly, TREM2 deficiency in aged mice led to similar changes to LPS injection. This suggests that LPS injection likely mitigates aging-related demyelination and muscle denervation via Trem2 downregulation.
Taken together, this study reveals the role of macrophage-driven inflammation as a pathogenic mediator in age-related peripheral neuropathy, and that targeting macrophages might be an option to mitigate peripheral neuropathies in aging individuals. Furthermore, this study shows that systemic inflammation may be an ambivalent modifier of age-related nerve damage, leading to a distinct type of axonal perturbation, but in addition to functionally counteracting, dampened demyelination and muscle denervation. Translationally, it is plausible to assume that tipping the balance of macrophage polarization to one direction or the other may determine the functional outcome in the aging peripheral nervous system of the elderly.
Biomechanische Eigenschaften eines biomaterialbasierten Kreuzbandkonstruktes in-vivo und in-vitro

Kreuzbandrupturen stellen nach wie vor eine Herausforderung in der klinischen Praxis hinsichtlich kurz- und langfristiger unerwünschter Nebenwirkungen dar (z.B. Reruptur und Arthrosebildung).
In der vorliegenden Arbeit wird der entwickelte Ansatz eines Kollagen-I-basierten künstlichen Kreuzbandkonstruktes hinsichtlich der Reißfestigkeit, Lagerung, Verstärkungsmöglichkeit mittels Fiber-tape und langfristigen Arthroseentstehung untersucht mittels in-vitro und in-vivo Untersuchungen unter zur Hilfe nahme des Minipig Tiermodels.
Die Ergebnisse zeigen keinen Einfluss der Lagerungstemperatur sowie des Lagerungszeitraums auf die Reißfestigkeit des Konstruktes, sowie eine mögliche initiale Verstärkung mittels Fibertape im Minipig. Darüber hinaus wurde mikroskopisch wie makroskopische Arthroseentstehung nachgewiesen. Das Ausmaß der Arthroseentstehung ist diesbezüglich mit einer Abweichung der Konstruktimplantation von der ursprünglichen Kreuzbandinsertion mittels MRT bestätigt worden.
Emotional-associative learning processes such as fear conditioning and extinction are highly relevant to not only the development and maintenance of anxiety disorders (ADs), but also to their treatment. Extinction, as the laboratory analogue to behavioral exposure, is assumed a core process underlying the treatment of ADs. Although exposure-based treatments are highly effective for the average patient suffering from an AD, there remains a gap in treatment efficacy with over one third of patients failing to achieve clinically significant symptom relief. There is ergo a pressing need for intensified research regarding the underlying neural mechanisms of aberrant emotional-associative learning processes and the neurobiological moderators of treatment (non-)response in ADs.
The current thesis focuses on different applications of the fundamental principles of fear conditioning and extinction by using two example cases of ADs from two different multicenter trials. First, we targeted alterations in fear acquisition, extinction, and its recall as a function of psychopathology in panic disorder (PD) patients compared to healthy subjects using fMRI. Second, exposure-based therapy and pre-treatment patient characteristics exerting a moderating influence on this essential learning process later on (i.e. treatment outcome) were examined using multimodal functional and structural neuroimaging in spider phobia.
We observed aberrations in emotional-associative learning processes in PD patients compared to healthy subjects indicated by an accelerated fear acquisition and an attenuated extinction recall. Furthermore, pre-treatment differences related to defensive, regulatory, attentional, and perceptual processes may exert a moderating influence on treatment outcome to behavioral exposure in spider phobia. Although the current results need further replication, on an integrative meta level, results point to a hyperactive defensive network system and deficient emotion regulation processes (including extinction processes) and top-down control in ADs. This speaks in favor of transdiagnostic deficits in important functional domains in ADs.
Deficits in transdiagnostic domains such as emotion regulation processes could be targeted by enhancing extinction learning or by means of promising tools like neurofeedback. The detection of pre-treatment clinical response moderators, for instance via machine learning frameworks, may help in supporting clinical decision making on individually tailored treatment approaches or, respectively, to avoid ineffective treatment and its related financial costs. In the long run, the identification of neurobiological markers which are capable of detecting non-responders a priori represents an ultimate goal.
Körperpsychotherapie etabliert sich zunehmend und ist keine neue Entdeckung. Bereits vor über 120 Jahren war bekannt, dass über den Körper die Psyche erreicht werden kann und damit die verbale Psychotherapie effektiver und gegebenenfalls erst möglich wurde. Wissenschaftliche Untersuchungen sprechen dafür, dass Körperpsychotherapie heute als fünfte Säule der allgemein anerkannten psychotherapeutischen Verfahren (PA, TP, VT, ST) angesehen werden kann. Sie hat sich aus der atemtherapeutischen und der Bewegung der Gymnastik sowie der Verwendung in der Psychoanalyse entwickelt. Sie ist weitestgehend in die tiefenpsychologische und verhaltenstherapeutische Psychotherapie integriert und kann zu den humanistischen Verfahren gezählt werden. Anwendung findet die Körperpsychotherapie beispielsweise in der Psychosomatischen Medizin sowie auf verschiedenen Gebieten der Psychotherapie. Laut den hier vorgelegten Befunden erreicht die Arbeit am Körper nonverbal Verarbeitetes, das sich tief in das implizite Körpergedächtnis eingegraben hat, lange bevor ein junger Mensch das Sprechen erlernte. Eine Möglichkeit, dies konzeptuell einzuordnen und therapeutisch nutzbar zu machen, ist das Modell der „verkörperten Selbstwahrnehmung“ nach Fogel, das Teile des Körperschemas beinhaltet. In der Bindungsbeziehung nicht adäquates Eingehen auf die kindlichen Bedürfnisse hat weitreichende Folgen auf das weitere Leben. In Untersuchungen konnte gezeigt werden, wie sich Störungen in der Entwicklung eines Kindes in Form von Körperschemastörungen und Körperdissoziationen, in Emotionsregulations- und als Entwicklungstraumastörung manifestieren können. Diese sind weit verbreitet und Teil einer Gesellschaft, die auf Leistung und Effizienz ausgerichtet ist und in Zusammenhang mit chronischem Stress stehen. Evolutionsgeschichtlich begründete Überlebensmuster werden durch chronischen Stress aktiviert und sind Ursache zahlreicher Erkrankungen. Hierfür liefert Porges mit seiner Polyvagal-Theorie einen neuen neurobiologischen Erklärungsansatz. Durch eine Imbalance stressauslösender und entspannender Faktoren zugunsten des Stresses werden körpereigene Selbstheilungskräfte der Selbstregulation verhindert und die Resilienzfähigkeit eingeschränkt. Selbstregulation und Resilienz sind vorhanden, wenn das Ruhe- und Bindungssystem dominiert im Gegensatz zur Kampf-, Flucht- und Erstarrungsreaktion. In seiner Hypothese zeigt Porges auf, wie das autonome Nervensystem Verhaltensweisen beeinflusst und wie diesen begegnet werden kann. Durch den sympathischen Zweig wird die An- und Verspannungsreaktion auf körperlicher Seite mit den auch auf der psychischen Seite verbundenen Reaktionen vermittelt. Diesem kann durch die parasympathisch vermittelte Oxytocin-Freisetzung begegnet werden. Durch eine Balance dieser beiden Waagschalen kann körperliche und seelische Gesundheit sowie Resilienzfähigkeit gefördert werden. Die Körperpsychotherapie bietet auch aus meiner Sicht eine noch unterschätzte Möglichkeit, die Balance wieder herzustellen. Eine Methode, die positive durch Oxytocin vermittelte heilsame Reaktionen in Gang zu setzt, stellt die berührende Körperarbeit dar wie sie beispielsweise nach der Rosen-Methode praktiziert wird. Körperpsychotherapie im Allgemeinen kann in der Behandlung von Depressionen, Angst- und psychosomatischen Störungen hilfreich sein. Sie ist empirisch in einer umfassenden Theorie begründet und fundiert auf neurobiologischen und neurowissenschaftlichen Erkenntnissen. Aus Sicht der Autorin handelt es sich bei der Körperpsychotherapie angesichts der vorliegenden Befunde und theoretischen Wirkkonzepte um einen therapeutischen Ansatz, der wesentlich dazu beitragen kann, die Behandlung psychischer Störungen kosteneffizienter und wirksamer zu gestalten. Um differenzierter zwischen theoretischem Potential und tatsächlich nachweisbaren Effekten körperpsychotherapeutischer Methoden unterscheiden zu können, ist es aus meiner Sicht dringend zu empfehlen, körperpsychotherapeutische Arbeitsansätze exakter zu erforschen. Beispielsweise wäre es lang- oder mittelfristig auch wünschenswert, Forschungsdaten für eine präzisere Indikationsstellung zur Verfügung zu haben. Dabei wäre beispielweise zu klären, welche Verfahren für welche Störungsbilder, in welchem Behandlungssetting und für welche Behandlungsdauer in Frage kommen. Auch fehlen hinsichtlich der Kontraindikationen belastbare Forschungsdaten zu den oben benannten Empfehlungen diverser Vertreter der Körperpsychotherapie. Aufgrund des hohen Erklärungspotentials für das individuelle Erleben psychisch beeinträchtigter Personen, das beispielsweise die Polyvagal-Theorie nach Porges oder die verkörperte Selbstwahrnehmung nach Fogel bieten, erscheint mir auch die Forderung nach einer Berücksichtigung körperpsychotherapeutischer Theorien und Methoden in der Ausbildung von Ärzten und Psychologen nachvollziehbar und sinnvoll. Aufgrund der in dieser Arbeit zusammengetragenen Ergebnisse halte ich es für dringend empfehlenswert, die Körperpsychotherapie als eigenständiges Behandlungselement in die fachgerechte Versorgung psychisch Erkrankter aufzunehmen, sofern keine der erwähnten Kontraindikationen dem widersprechen.
In dieser Arbeit geht es um die Phosphoglykolatphosphatase (PGP), die als Phosphatase vom Haloazid Dehalogenase-Typ (HAD-Phosphatase) zu der ubiquitär vorkommenden Superfamilie der HAD-Hydrolasen gehört. In der Literatur ist eine in vitro Phosphatase-Aktivität gegenüber 2-Phospho-L-Laktat (2PL), 4-Phospho-D-Erythronat (4PE), Phosphoglykolat (PG) und Glycerol-3-Phosphat (G3P) beschrieben. 2PL und 4PE entstehen in Nebenreaktionen während der Glykolyse und hemmen bei Akkumulation die Glykolyse bzw. den Pentosephosphatweg. PG kann auch in einer Nebenreaktion während der Glykolyse oder im Rahmen der Reparatur von oxidativen DNA-Schäden entstehen. G3P entsteht aus Dihydroxyacetonphosphat und bildet das Kohlenhydratgerüst der Triacylglyceride (TAG). Zelluläre Studien konnten Hinweise auf die Regulierung des epidermalen wachstumsfaktor-(EGF-)induzierten Zytoskelettumbaus durch die PGP liefern und die Untersuchung von Mäusen mit PGP-Inaktivierung zeigte einen Einfluss auf die Zellproliferation und embryonale Entwicklung. Die Regulation der PGP-Expression führte zu Veränderungen im Kohlenhydrat- und Fettstoffwechsel.
Die Untersuchung der PGP-Funktionen erfolgte bislang ausschließlich mit genetischen Ansätzen. Aufgrund von möglichen Kompensationsmechanismen und Off-Target-Effekten müssen genetische und pharmakologische Methoden als sich ergänzende Ansätze verstanden werden. Um die Funktionen der PGP besser zu verstehen, fokussiert sich die vorliegende Arbeit auf die gezielte pharmakologische PGP-Inhibition. In Vorarbeiten wurden 41.000 Moleküle gescreent und fünf potentielle Inhibitoren identifiziert. Ziele dieser Arbeit waren zum einen die Implementierung der Inhibitor # 1-Behandlung in der Zellkultur, zum anderen die Charakterisierung der PGP-Hemmung durch Inhibitor # 48 und die Durchführung erster Selektivitätstestungen mit Inhibitor # 48.
Zusammenfassend kann festgehalten werden, dass Inhibitor # 1 in der Lage ist, die endogene PGP in Zelllysaten der murinen spermatogonialen Zelllinie (GC1) zu hemmen. Unter bestimmten Bedingungen führte die Inhibitor # 1-Behandlung der GC1-Zellen zur Hemmung der PGP. Erste Analysen zellulärer Inhibitoreffekte konnten eine Steigerung der TAG-Konzentration in behandelten GC1-Zellen nachweisen. Die PGP-Hemmung durch Inhibitor # 48 wurde als unkompetitive Inhibition charakterisiert und es zeigten sich keine relevanten Inhibitoreffekte auf die HAD-Phosphatasen Magnesium-abhängige Phosphatase 1 (MDP1), Lysin-Histidin-Pyrophosphat-Phosphatase (LHPP) und Polynukleotidase 5'-Kinase/3'-Phosphatase (PnkP). Dagegen konnte eine Aktivitätssteigerung von Phospho 2 beobachtet werden. Die vorliegende Arbeit liefert somit erste Erkenntnisse über die Anwendung des PGP-Inhibitors # 1 in der Zellkultur und schafft die Grundlage für nachfolgende Untersuchungen mit Inhibitor # 48. Weitere Experimente sind notwendig, die die Inhibitorbehandlung in der Zellkultur optimieren und die Selektivität weiter charakterisieren, um mithilfe der Inhibitoren neue Erkenntnisse über die physiologische und pathophysiologische Rolle der PGP gewinnen zu können.
Neurodegeneration plays an essential role in Parkinson’s disease (PD). Several crucial neuronal pro-and antidegeneration markers were described to be altered in disease models accompanied by neurodegeneration. In the AAV1/2-A53T-aSyn PD rat model progressive time-dependent motor impairment and neurodegeneration in the nigrostriatal tract starting from 2 weeks after PD model induction could be found. Downregulation of Nrf2 in SN and nigrostriatal axon localization, a trend of Tau downregulation in SN and upregulation in axon localization in the AAV1/2-A53T-aSyn PD rat model were observed, indicating potential therapeutic value of these two molecular targets in PD. No alterations of SARM1 and NMNAT2 could be detected, indicating little relevance of these two molecules with our AAV1/2-A53T-aSyn rat model.
