Die vorliegende Arbeit nimmt einmal mehr die >Kleine Prosa< Kafkas in den Fokus der Betrachtung. Hierbei rekurriert sie auf ein primär historisches Diskursfeld des 19. Jahrhunderts, in dessen philosophischen Gemarkungen der Leib, und dies nicht erst seit Schopenhauers einschneidender "Achsendrehung im Begriff des Menschen" (Georg Simmel), eine zentrale Bedeutung erlangt. Kafka inszeniert nun diesen historisch fundamentierten >Leib< in seiner >Kleinen Prosa< als einen gepeinigten, einen wunden. Die eingeschriebenen Körper-Wunden, die von den kafkaschen Protagonisten allesamt als >brandmarkend< erlebt werden, klaffen derart in drei ganz elementaren Erzählungen auf: In der Erzählung 'Ein Landarzt' spielt die zu heilende Körper-Wunde eines Patienten und, unmittelbar damit liiert, eine aus dem Viehischen hervor kriechende Tierfigur eine besonders augenfällige Rolle. Eine Körper-Schrift, die um den kranken Körper zirkuliert, ist mithin inauguriert. Auch Kafkas Affe Rotpeter, der gleichermaßen mit zwei machtvoll beigefügten Körper-Wunden versehen wird, zeigt in seinem 'Bericht für eine Akademie' aufs deutlichste an, wie fortan mit dem tierischen Körper zu verfahren ist; eingefangen, sodann gezähmt, domestiziert und in die Menschenwelt eingeformt, wird er sukzessive zur Unterdrückung der "Affennatur" angehalten. Eine buchstäblich ins Fleisch einschneidende Macht-Schrift gibt sich hier eindringlich zu erkennnen. Auf seinem eigenen Leib wiederum erfährt auch der Verurteilte in der 'Strafkolonie' seine "Schuld", und zwar als eine Einschreibung in den Körper selbst. Eine Körpermacht-Schrift, die gemäß der Eigenlogik des Offiziers eine Entzifferung der Schrift qua Wunde herbeiführen soll, entlarvt sich jedoch mehr und mehr als grausames Macht-Derivat, dessen markante Chiffre "Blutwasser" heißt. Kafkas Narrationen veranschaulichen immer wieder aufs Neue, dass die Diskreditierung des Körpers, und dessen immer schon machtvoll praktizierte Ausschließung, mit dem logozentrischen Supremat metaphysischer Erhöhungstechniken einhergeht. Die signifikant in Erscheinung tretenden Körper-Wunden sind denn auch deren anschaulichste Signaturen - >brandmarken<.
Rifting and breakup of Westgondwana in the Late Jurassic/ Early Cretaceous initiated the formation of the South Atlantic and its conjugated pair of passive continental margins. The Walvis Basin offshore NW-Namibia is an Early Cretaceous to recent depositional centre with a typically wedge-shaped postrift sedimentary succession covering an area of 105000km2. A 2D model transect across the central Walvis Basin and adjacent onshore areas is used as a case study to investigate quantitatively the denudational history of the evolving passive margin and the related contemporaneous depositional postrift evolution offshore. The database for both the onshore and offshore part of the model traverse is well constrained by own field work, published data as well as by seismic and well data supported by samples. The ultimate goal of this project is to present an integrated approach towards a quantitative link between surface processes and internal processes in terms of a mass and process balance.
The availability of coherent soft x-rays through the nonlinear optical process of high-harmonic generation allows for the monitoring of the fastest events ever observed in the laboratory. The attosecond pulses produced are the fundamental tool for the time-resolved study of electron motion in atoms, molecules, clusters, liquids and solids in the future. However, in order to exploit the full potential of this new tool it is necessary to control the coherent soft x-ray spectra and to enhance the efficiency of conversion from laser light to the soft x-ray region in the harmonic-generation process. This work developed a comprehensive approach towards the optimization of the harmonic generation process. As this process represents a fundamental example of \emph{light}--\emph{matter} interaction there are two ways of controlling it: Shaping the generating laser \emph{light} and designing ideal states of \emph{matter} for the conversion medium. Either of these approaches was closely examined. In addition, going far beyond simply enhancing the conversion process it could be shown that the qualitative spectral response of the process can be modified by shaping the driving laser pulse. This opens the door to a completely new field of research: Optimal quantum control in the attosecond soft x-ray region---the realm of electron dynamics. In the same way as it is possible to control molecular or lattice vibrational dynamics with adaptively shaped femtosecond laser pulses these days, it will now be feasible to perform real-time manipulation of tightly bound electron motion with adaptively shaped attosecond light fields. The last part of this work demonstrated the capability of the herein developed technique of coherent soft-x-ray spectral shaping, where a measured experimental feedback was used to perform a closed-loop optimization of the interaction of shaped soft x-ray light with a sulfur hexafluoride molecule to arrive at different control objectives. For the optimization of the high-harmonic-generation process by engineering the conversion medium, both the gas phase and the liquid phase were explored both in experiment and theory. Molecular media were demonstrated to behave more efficiently than commonly used atomic targets when elliptically polarized driving laser pulses are applied. Theory predicted enhancement of harmonic generation for linearly polarized driving fields when the internuclear distance is increased. Reasons for this are identified as the increased overlap of the returning electron wavefunction due to molecular geometry and the control over the delocalization of the initial electronic state leading to less quantum-mechanical spreading of the electron wavepacket during continuum propagation. A new experimental scheme has been worked out, using the method of molecular wavepacket generation as a tool to enhance the harmonic conversion efficiency in `pump--drive' schemes. The latter was then experimentally implemented in the study of high-harmonic generation from water microdroplets. A transition between the dominant laser--soft-x-ray conversion mechanisms could be observed, identifying plasma-breakdown as the fundamental limit of high-density high-harmonic generation. Harmonics up to the 27th order were observed for optimally laser-prepared water droplets. To control the high-harmonic generation process by the application of shaped laser light fields a laser-pulse shaper based on a deformable membrane mirror was built. Pulse-shape optimization resulted in increased high-harmonic generation efficiency --- but more importantly the qualitative shape of the spectral response could be significantly modified for high-harmonic generation in waveguides. By adaptive optimization employing closed-loop strategies it was possible to selectively generate narrow (single harmonics) and broad bands of harmonic emission. Tunability could be demonstrated both for single harmonic orders and larger regions of several harmonics. Whereas any previous experiment reported to date always produced a plateau of equally intense harmonics, it has been possible to demonstrate ``untypical'' harmonic soft x-ray spectra exhibiting ``switched-off'' harmonic orders. The high degree of controllability paves the way for quantum control experiments in the soft x-ray spectral region. It was also demonstrated that the degree of control over the soft x-ray shape depends on the high-harmonic generation geometry. Experiments performed in the gas jet could not change the relative emission strengths of neighboring harmonic orders. In the waveguide geometry, the relative harmonic yield of neighboring orders could be modified at high contrast ratios. A simulation based solely on the single atom response could not reproduce the experimentally observed contrast ratios, pointing to the importance of propagation (phase matching) effects as a reason for the high degree of controllability observed in capillaries, answering long-standing debates in the field. A prototype experiment was presented demonstrating the versatility of the developed soft x-ray shaping technique for quantum control in this hitherto unexplored wavelength region. Shaped high-harmonic spectra were again used in an adaptive feedback loop experiment to control the gas-phase photodissociation reaction of SF$_6$ molecules. A time-of-flight mass spectrometer was used for the detection of the ionic fragments. The branching ratios of particular fragmentation channels could be varied by optimally shaped soft x-ray light fields. Although in one case only slight changes of the branching ratio were possible, an optimal solution was found, proving the sufficient technical stability of this unique coherent soft-x-ray shaping method for future applications in optimal control. Active shaping of the spectral amplitude in coherent spectral regions of $\sim$10~eV bandwidth was shown to directly correspond to shaping the temporal features of the emerging soft x-ray pulses on sub-femtosecond time scales. This can be understood by the dualism of frequency and time with the Fourier transformation acting as translator. A quantum-mechanical simulation was used to clarify the magnitude of temporal control over the shape of the attosecond pulses produced in the high-harmonic-generation process. In conjunction with the experimental results, the first attosecond time-scale pulse shaper could thus be demonstrated in this work. The availability of femtosecond pulse shapers opened the field of adaptive femtosecond quantum control. The milestone idea of closed-loop feedback control to be implemented experimentally was expressed by Judson and Rabitz in their seminal work titled ``Teaching lasers to control molecules''. This present work extends and turns around this statement. Two fundamentally new achievements can now be added, which are ``Teaching molecules to control laser light conversion'' and ``Teaching lasers to control coherent soft x-ray light''. The original idea thus enabled the leap from femtosecond control of molecular dynamics into the new field of attosecond control of electron motion to be explored in the future. The \emph{closed}-loop approach could really \emph{open} the door towards fascinating new perspectives in science. Coming back to the introduction in order to close the loop, let us reconsider the analogy to the general chemical reaction. Photonic reaction control was presented by designing and engineering effective media (catalysts) and controlling the preparation of educt photons within the shaped laser pulses to selectively produce desired photonic target states in the soft x-ray spectral region. These newly synthesized target states in turn could be shown to be effective in the control of chemical reactions. The next step to be accomplished will be the control of sub-femtosecond time-scale electronic reactions with adaptively controlled coherent soft x-ray photon bunches. To that end a time-of-flight high-energy photoelectron spectrometer has recently been built, which will now allow to directly monitor electronic dynamics in atomic, molecular or solid state systems. Fundamentally new insights and applications of the nonlinear interaction of shaped attosecond soft x-ray pulses with matter can be expected from these experiments.
Ziel der vorliegenden Arbeit war es, die Methode der adaptiven Pulsformung von Femtosekunden Laserpulsen in der flüssigen Phase experimentell zu realisieren. Eine Erweiterung dieser Technik auf die kondensierte Phase stellt einen wichtigen Schritt in Richtung einer breiten Anwendbarkeit zur Steuerung von chemischen Reaktionen dar. Die größere Teilchendichte im Vergleich zur Gasphase ermöglicht zum einen eine Erhöhung der erzielbaren absoluten Produktausbeuten. Andererseits ergibt sich erst dadurch die Möglichkeit, reale chemische Reaktionen, wie bimolekulare Reaktionen, gezielt zu steuern, da Stöße zwischen verschiedenen Molekülen wahrscheinlicher werden. Die Methode der adaptiven Quantenkontrolle ist für die Anwendung in der flüssigen Phase bestens geeignet, da sie eine kohärente Kontrolle von photoinduzierten molekularen Prozessen selbst in komplexen Quantensystemen erlaubt. In dieser experimentellen Umsetzung einer ,,geschlossenen Kontrollschleife'' wird die spektrale Phasenstruktur von fs-Laserpulsen in einem computergesteuerten Pulsformer moduliert. Der resultierende geformte Laserpuls wechselwirkt anschließend mit dem zu untersuchenden molekularen System und steuert aktiv die Entwicklung des erzeugten Wellenpakets auf der Potentialenergiefläche. Eine quantitative Messung der erzeugten Photoprodukte dieser Licht-Materie Wechselwirkung dient als Rückkopplungssignal eines selbstlernenden Computeralgorithmus. Der auf dem Prinzip der Evolutionstheorie arbeitende Algorithmus verbessert nun iterativ die Pulsform bis ein Optimum des gewünschten Reaktionskanals erreicht wird. Das modulierte elektrische Feld des Laserpulses passt sich somit entsprechend der gestellten Kontrollaufgabe automatisch den molekularen Eigenschaften an. Um jedoch die Anwendung dieser Technik auch in der kondensierten Phase zu demonstrieren, mussten Methoden zur Gewinnung eines Rückkopplungssignals gefunden werden. Im Rahmen dieser Arbeit wurden daher Möglichkeiten eines quantitativen Rückkopplungssignals für die adaptive Kontrolle in der flüssigen Phase untersucht, wie die Emissionsspektroskopie und die transiente Absorption im UV/VIS oder infraroten Spektralbereich. In einem ersten Experiment wurde die Emissionsspektroskopie verwendet, um einen Ladungstransferprozess (MLCT) in einem Ru(II)-Komplex ([Ru(dpb)3]2+) mit geformten fs-Laserpulsen zu steuern. Um die dominierende Intensitätsabhängigkeit der Anregung zu eliminieren, wurde die Emissionsausbeute mit dem SHG-Signal eines nichtlinearen Kristalls „normiert“. Diese Auslöschung des intensitätsabhängigen Faktors in beiden Prozessen ermöglichte es, Pulsformen zu finden, die dieses Verhältnis sowohl maximieren als auch minimieren. Ein Ansatz zur Erklärung der experimentellen Ergebnisse konnte mit Hilfe eines störungstheoretischen Modells beschrieben werden. In einem zweiten Experiment wurde erstmals eine photochemische Selektivität zwischen zwei verschiedenen Substanzen in der kondensierten Phase demonstriert. Dabei sollte die jeweilige Zwei-Photonen Anregung des Komplexes [Ru(dpb)3]2+ gegenüber dem Molekül DCM selektiv kontrolliert werden. Wiederum diente die spontane Emission beider Substanzen als Rückkopplungssignal für die Effektivität des Anregungsschritts. Verschiedene Ein-Parameter Kontrollmethoden, wie der Variation der Anregungswellenlänge, der Intensität sowie des linearen Chirps, konnten diese Kontrollaufgabe nicht erfüllen. Jedoch konnte eine Optimierung des Verhältnisses der beiden Emissionsausbeuten mit Hilfe der adaptiven Pulsformung erzielt werden. Das Ergebnis dieses Experiments zeigt, dass photoinduzierte Prozesse in zwei unterschiedlichen molekularen Substanzen trotz der Wechselwirkungen der gelösten Moleküle mit ihrer Lösungsmittelumgebung selektiv und simultan kontrolliert werden können. Das Ziel des dritten Experiments war eine gezielte Steuerung einer komplexeren chemischen Reaktion. Mit Hilfe der adaptiven Pulsformung konnte eine optimale Kontrolle der Photoisomerisierungsreaktion des Moleküls NK88 demonstriert werden. Das dazu benötigte Rückkopplungssignal für den evolutionären Algorithmus wird durch transiente Absorptionsspektroskopie im UV/VIS Spektralbereich bereitgestellt. Eine Untersuchung der Dynamik der Isomerisierungsreaktion mit Hilfe der Pump-Probe Technik erlaubte eine Zuordnung zweier verschiedener Absorptionsbereiche zu den jeweiligen Isomeren. Die Ergebnisse der Optimierung des Verhältnisses der Quantenausbeuten der beiden Isomere zeigten, dass die geformten Laserpulse eine Kontrolle der Effizienz der Photoisomerisierung in der flüssigen Phase ermöglichen. Zusammenfassend kann man sagen, dass im Rahmen dieser Arbeit mit Hilfe der fs-Lasertechnologie und der Technik der adaptiven fs-Quantenkontrolle Experimente durchgeführt wurden, die einen wichtigen Beitrag zu dem neuen Forschungsbereich der Femtochemie darstellen. Die Erweiterung dieser Technik auf die flüssige Phase beschreibt einen ersten Erfolg in Richtung einer neuartigen Chemie.
In a three-year study the current aeolian transportation processes were examined in a linear dune area previously used for grazing near Nizzana at the Israeli-Egyptian border. The research area was subject to heavy grazing across the border, which led to the total destruction of the natural vegetation in the period of 1967 to 1982. As a consequence, intensified aeolian activity and significant changes of the morphology of the dunes were observed. After the end of the grazingg on the Israeli side, a rapid return of the vegetation in the interdune corridors and on the footslopes of the dunes took place. In addition also a reduction of obviously active areas on the dune crests was observed. The situation on Egyptian territory west the border remained unchanged until today. This study is aimed at understanding the changed aeolian morphodynamics east the border. The emphasis was placed on the investigation of the spatial and temporal distribution of aeolian sand transport as well as on the influencing factors morphology, surface condition and vegetation.
The success of diagnostic knowledge systems has been proved over the last decades. Nowadays, intelligent systems are embedded in machines within various domains or are used in interaction with a user for solving problems. However, although such systems have been applied very successfully the development of a knowledge system is still a critical issue. Similarly to projects dealing with customized software at a highly innovative level a precise specification often cannot be given in advance. Moreover, necessary requirements of the knowledge system can be defined not until the project has been started or are changing during the development phase. Many success factors depend on the feedback given by users, which can be provided if preliminary demonstrations of the system can be delivered as soon as possible, e.g., for interactive systems validation the duration of the system dialog. This thesis motivates that classical, document-centered approaches cannot be applied in such a setting. We cope with this problem by introducing an agile process model for developing diagnostic knowledge systems, mainly inspired by the ideas of the eXtreme Programming methodology known in software engineering. The main aim of the presented work is to simplify the engineering process for domain specialists formalizing the knowledge themselves. The engineering process is supported at a primary level by the introduction of knowledge containers, that define an organized view of knowledge contained in the system. Consequently, we provide structured procedures as a recommendation for filling these containers. The actual knowledge is acquired and formalized right from start, and the integration to runnable knowledge systems is done continuously in order to allow for an early and concrete feedback. In contrast to related prototyping approaches the validity and maintainability of the collected knowledge is ensured by appropriate test methods and restructuring techniques, respectively. Additionally, we propose learning methods to support the knowledge acquisition process sufficiently. The practical significance of the process model strongly depends on the available tools supporting the application of the process model. We present the system family d3web and especially the system d3web.KnowME as a highly integrated development environment for diagnostic knowledge systems. The process model and its activities, respectively, are evaluated in two real life applications: in a medical and in an environmental project the benefits of the agile development are clearly demonstrated.
Die Dissertation beschreibt, dass die menschliche Gleichgewichtsregulation zur Aufrechterhaltung der Stellung im Raume, zur Regulation der Blickmechanismen, aber auch zur inneren Erzielung eines gleichgewichtes auf komplizierten kybernetischen Regelkreisen beruht. Mit Hilfe eines umpfangreichen neurootometrischen Netzwerkes unter Einschaltung aequilibriometrischer Gleichgewichtsmessungen und audiometrischer Gehörfunktionsmessungen lässt sich nachweisen, dass die Regulationsmuster mindestens im quantitativen Bereich während des gesamten Lebens wechseln. sie sind netzwerkartig miteinander verknüft. Wenn aber dieses System im Alter nicht mehr passend einreguliert ist, empfinden die Patienten zahlreiche Symptome, die wir mit der Vorsilbe Alter (Presby) wie zum Beispiel Altersschwindel, Presbyvertigo, Alterstaumligkeit, Presbytaxie, Altershörstörungen, Presbyakusis, Altersohrgeräusche, Presbytnnitus usw., bezeichnen Aus medizinischer Sicht kommt es unter anderem darauf an, altersbedingte Fehlregulationen rechtzeitig messend zu erfassen, damit diesen durch entsprechende Behandlungen unter Einsatz von Physiotherapie, Psychtherapie, aber auch medikamentöser Therapie begegnet werden kann.
Die muscarinischen Rezeptoren sind ein wichtiger Bestandteil des parasympathischen Nervensystems. Sie gehören zur großen Gruppe der G-Protein-gekoppelten Rezeptoren, die nach ihrer Verwandtschaft in drei große Klassen eingeteilt werden können. Die muscarinischen Rezeptoren gehören zur Klasse A, den rhodopsinähnlichen Rezeptoren. Durch die im Jahr 2000 vorgenommene Aufklärung der hochauflösenden Röntgenkristallstruktur des Rinderrhodpsins und die hohe Aminosäuresequenzähnlichkeit der G-Protein-gekoppelten Rezeptoren hat man eine sehr gute Modellvorstellung über den Aufbau der G-Protein-gekoppelten Rezeptoren. Die Rezeptoren bestehen aus sieben transmembranalen Helices, die von drei intrazellulären und drei extrazellulären Loops stabilisiert werden. Bis heute konnten fünf Rezeptorsubtypen gentechnisch klassifiziert werden, die sich durch ihre Gewebeverteilung und Funktion unterscheiden. Allen Subtypen ist eine hohe Sequenzhomologie im Bereich der orthosteren Bindungsstelle gemeinsam, so dass die Entwicklung von subtyp-spezifischen orthosteren Liganden sehr schwierig ist. Außer der orthosteren Bindungsstelle konnte noch eine weitere Bindungsstelle am muscarinischen Rezeptor identifiziert werden. Diese befindet sich weiter außerhalb im Rezeptor in einem Bereich, der über die fünf Rezeptorsubtypen nicht sehr stark konserviert ist, so dass die Entwicklung von subtyp-spezifischen Liganden möglich ist. An dieser zweiten Bindungsstelle binden allostere Modulatoren. Hierbei handelt es sich um Substanzen, die ohne den orthosteren Liganden keinen Effekt am Rezeptor auslösen, dafür aber die Gleichgewichtsbindung des orthosteren Liganden beeinflussen können. Der Einfluss auf die Gleichgewichtsbindung geschieht wechselseitig und kann positiv, neutral oder negativ kooperativ sein. Zusätzlich üben allostere Modulatoren einen Effekt auf die Dissoziation des orthosteren Liganden aus. Die meisten bisher gefunden allosteren Modulatoren erniedrigen die Dissoziationsgeschwindikeit des orthosteren Liganden vom Rezeptor. Die Summe dieser Eigenschaften machen die allosteren Modulatoren sehr interessant für die Arzneimitteltherapie. Das Ziel dieser Arbeit war die Synthese strukturell neuer allosterer Modulatoren des muscarinischen Rezeptors unter Anwendung des postulierten Pharmakophormodells. Als Ausgangspunkt sollten geländerhelicale Moleküle dienen, die strukturell abgewandelt dieses Pharmakophormodell sehr gut erfüllen. Die geländerhelicalen Moleküle ähneln in ihrem dreidimensionalen Aufbau dem Geländer einer Wendeltreppe. Sie sind durch die Brücken zwischen den aromatischen Bereichen sehr rigide Moleküle, so dass es nur wenige genau definierte Konformationen gibt. Grundsätzlich können drei Atropisomere unterschieden werden, wobei zwei zueinander enantiomer sind. Geplant war die Synthese eine Reihe von tertiären Aminen oder quartären Ammoniumsalzen. Die Synthese der Ausgangsverbindung konnte nach der Vorschrift von Kiupel erfolgen, war aber nur mit geringer Ausbeute möglich. Deshalb wurde dieser Syntheseweg nicht weiterverfolgt. Als Alternative bot sich an, auf die Brücken zwischen den aromatischen Ringen zu verzichten. Die so entstandenen Verbindungen sind weniger rigide und können sich deshalb gegebenenfalls besser an den Rezeptor anpassen. Grundsätzlich können je nach Substitutionsmuster zwei Synthesewege verfolgt werden. Beide Varianten erfüllen das postulierte Pharmakophormodell. Der Aufbau des Grundgerüstes erfolgt mittels einer nickelkatalysierten Grignard-Kupplung. Danach erfolgen eine Wohl-Ziegler-Seitenkettenbromierung und eine Verlängerung der Seitenkette im Sinne einer Alkylierung mittels Malonsäurediethylester und einer Hilfsbase. Anschließend erfolgen die Decarboxylierung und die Umsetzung zum Amid, das zum Amin reduziert werden kann. Betrachtet man die Lage der Pharmakophorelemente so variiert der Abstand der positiv geladenen Stickstoffe je nach Konformation zwischen 5 Å und 15 Å, so dass ein weiter Bereich abgedeckt werden kann. Der Abstand der aromatischen Bereiche bleibt relativ stabil. Die pharmakologische Testung der Verbindungen auf ihre allostere Potenz und Affinität zum muscarinischen Rezeptor erfolgte in der Arbeitsgruppe von Prof. Mohr in Bonn. Hierzu werden Membranhomogenate vom Herzventrikelgewebe des Hausschweines verwendet. Diese enthalten mit großer Prävalenz muscarinische M2-Rezeptoren. Es wurden Gleichgewichtsbindungs- und Dissoziationsexperimente durchgeführt. Bis jetzt sind noch nicht alle Verbindungen getestet worden. Die bisher getesteten Verbindungen weisen alle eine Affinität zum mit [3H]-N-Methylscopolamin besetzten muscarinischen M2-Rezeptor im mikro-molaren Bereich auf. Sie liegen damit im oberen Bereich der bisher synthetisierten allosteren Modulatoren. Das postulierte Pharmakophormodell konnte also mit Hilfe der synthetisierten Substanzen bestätigt werden.
Diabetes mellitus ist die häufigste endokrine Störung des Glukosestoffwechsels und betrifft einen großen Teil der Bevölkerung. Der progressive Verlauf der Erkrankung führt zu schweren Sekundärschäden und schränkt die Lebensqualität der Betroffenen deutlich ein. Als einzige kausale Therapiemöglichkeiten stehen bislang nur die Pankreas- und Inseltransplantation zur Verfügung. Der Mangel an Spenderorganen und die erforderliche lebenslange Immunsuppression schränken die weite Verfügbarkeit dieser Behandlung für die überwiegende Anzahl der Diabetiker stark ein. Daher ist es sehr wichtig, neue Therapiestrategien des Diabetes mellitus zu entwickeln. Hierbei ist die Weiterentwicklung der Zelltherapie von zentraler Bedeutung, um durch Differenzierung und Expansion insulinproduzierender Zellen den Mangel an Zellen zur Transplantation zu überwinden. In dieser Arbeit wurde eine Analyse der Expression des proliferationsassoziierten Proteins P8 in Betazellen des endokrinen Pankreas durchgeführt. Es konnte eine spezifische Genexpression von P8 in Betazellen und duktalen Vorläuferzellen des endokrinen Pankreas nachgewiesen werden. Durch die Etablierung eines spezifischen Antiserums wurde P8 im Zellkern der Betazellen lokalisiert. Expressionsanalysen zeigten im Folgenden eine positive Regulation der P8-mRNA-Expression durch Glukose als bekannten Stimulus für die Insulinsekretion und Betazellreplikation. Gleiches wurde für das Inkretinhormon GLP-1, das die Genexpression von bedeutsamen Transkriptionsfaktoren für die Betazellproliferation und -differenzierung induziert, in Betazellen als auch deren Vorläuferzellen nachgewiesen. Anhand von Transfektionsexperimenten und Funktionsuntersuchungen mittels ELISA konnte eine Dedifferenzierung der Betazellen durch eine Überexpression des potentiell proliferationsinduzierenden Proteins P8 ausgeschlossen werden. Hierbei wurden betazellspezifische Differenzierungsmarker, PDX-1 und Proinsulin, sowie die Fähigkeit der Betazellen, Insulin zu produzieren, während einer Überexpression von P8 analysiert. Zusammenfassend wurde ein proliferationsassoziiertes Protein, das als möglicher Transkriptionsfaktor in Betazellen des endokrinen Pankreas fungieren könnte, näher charakterisiert, um damit neue Aspekte zur Expansion von Betazellen unter Erhalt ihrer Funktion im Rahmen der Zelltherapie beizutragen.
