In this thesis the Drosophila mutant loechrig (loe), that shows progressive degeneration of the nervous system, is further described. Loe is missing a neuronal isoform of the protein kinase AMPK γ subunit (AMP-activated protein kinase- also known as SNF4Aγ) The heterotrimeric AMPK controls the energy level of the cell, which requires constant monitoring of the ATP/AMP levels. It is activated by low energy levels and metabolic insults like oxygen starvation and regulates multiple important signal pathways that control cell metabolism. Still, its role in neuronal survival is unclear. One of AMPK’s downstream targets is HMGR (hydroxymethylglutaryl-CoA- reductase), a key enzyme in cholesterol and isoprenoid synthesis. It has been shown that manipulating the levels of HMGR affects the severity of the neurodegenerative phenotype in loe. Whereas the regulatory role of AMPK on HMGR is conserved in Drosophila, insects cannot synthesize cholesterol de novo. However, the synthesis of isoprenoids is a pathway that is evolutionarily conserved between vertebrates and insects. Isoprenylation of target proteins like small G-proteins provides a hydrophobic anchor that allows the association of these proteins with membranes and following activation. This thesis shows that the loe mutation interferes with the prenylation of Rho1 and the regulation of the LIM kinase pathway, which plays an important role in actin turnover and axonal outgrowth. The results suggest that the mutation in LOE, causes hyperactivity of the isoprenoid synthesis pathway, which leads to increased farnesylation of RHO1 and therefore higher levels of phospho-cofilin. A mutation in Rho1 improves the neurodegenerative phenotype and life span. The increased inactive cofilin amount in loe leads to an up regulation of filamentous actin. Actin is involved in neuronal outgrowth and experiments analyzing loe neurons gave valuable insights into a possible role of AMPK and accordingly actin on neurite growth and stability. It was demonstrated that neurons derived from loe mutants exhibit reduces axonal transport suggesting that changes in the cytoskeletal network caused by the effect of loe on the Rho1 pathway lead to disruptions in axonal transport and subsequent neuronal death. It also shows that actin is not only involved in neuronal outgrowth, its also important in maintenance of neurons, suggesting that interference with actin dynamics leads to progressive degeneration of neurons. Together, these results further support the importance of AMPK in neuronal function and survival and provide a novel functional mechanisms how alterations in AMPK can cause neuronal degeneration
Melanoma arises from the malignant transformation of melanocytes and is one of the most aggressive forms of human cancer. In fish of the genus Xiphophorus, melanoma development, although very rarely, happens spontaneously in nature and can be induced by interspecific crossing. The oncogenic receptor tyrosine kinase, Xmrk, is responsible for melanoma formation in these fishes. Since Xiphophorus are live-bearing fishes and therefore not compatible with embryonic manipulation and transgenesis, the Xmrk melanoma model was brought to the medaka (Oryzias latipes) system. Xmrk expression under the control of the pigment cell specific mitf promoter leads to melanoma formation with 100% penetrance in medaka. Xmrk is an orthologue of the human epidermal growth factor receptor (EGFR) and activates several downstream signaling pathways. Examples of these pathways are the direct phosphorylation of BRAF and Stat5, as well as the enhanced transcription of C-myc. BRAF is a serine-threonine kinase which is found mutated at high frequencies in malignant melanomas. Stat5 is a transcription factor known to be constitutively activated in fish melanoma. C-myc is a transcription factor that is thought to regulate the expression of approximately 15% of all human genes and is involved in cancer progression of a large number of different tumors. To gain new in vivo information on candidate factors known to be involved in melanoma progression, I identified and analysed BRAF, Stat5 and C-myc in the laboratory fish model system medaka. BRAF protein motifs are highly conserved among vertebrates and the results of this work indicate that its function in the MAPK signaling is maintained in medaka. Transgenic medaka lines carrying a constitutive active version of BRAF (V614E) showed more pigmented skin when compared to wild type. Also, some transiently expressing BRAF V614E fishes showed a disrupted eye phenotype. In addition, I was able to identify two Stat5 copies in medaka, named Stat5ab/a and Stat5ab/b. Sequence analysis revealed a higher similarity between both Stat5 sequences when compared to either human Stat5a or Stat5b. This suggests that the two Stat5 copies in medaka arose by an independent duplication processes. I cloned these two Stat5 present in medaka, produced constitutive active and dominant negative gene versions and successfully established transgenic lines carrying each version under the control of the MITF promoter. These lines will help to elucidate questions that are still remaining in Stat5 biology and its function in melanoma progression, like the role of Stat5 phosphorylation on tumor invasiveness. In a third project during my PhD work, I analysed medaka C-myc function and indentified two copies of this gene in medaka, named c-myc17 and c-myc20, according to the chromosome where they are located. I produced conditional transgenic medaka lines carrying the c-myc17 gene coupled to the hormone binding domain of the estrogen receptor to enable specific transgene activation at a given time point. Comparable to human C-myc, medaka C-myc17 is able to induce proliferation and apoptosis in vivo after induction. Besides that, C-myc17 long-term activation led to liver hyperplasia. In summary, the medaka models generated in this work will be important to bring new in vivo information on genes involved in cancer development. Also, the generated transgenic lines can be easily crossed to the melanoma developing Xmrk medaka lines, thereby opening up the possibility to investigate their function in melanoma progression. Besides that, the generated medaka fishes make it possible to follow the whole development of melanocytes, since the embryos are transparent and can be used for high throughput chemical screens.
Attention-deficit/hyperactivity disorder (ADHD) is a genetically complex childhood onset neurodevelopmental disorder which is highly persistent into adulthood. Several chromo-somal regions associated with this disorder were identified previously in genome-wide linkage scans, association (GWA) and copy number variation (CNV) studies. In this work the results of case-control and family-based association studies using a can-didate gene approach are presented. For this purpose, possible candidate genes for ADHD have been finemapped using mass array-based SNP genotyping. The genes KCNIP4, CDH13 and DIRAS2 have been found to be associated with ADHD and, in addition, with cluster B and cluster C personality disorders (PD) which are known to be related to ADHD. Most of the associations found in this work would not withstand correction for multiple testing. However, a replication in several independent populations has been achieved and in conjunction with previous evidence from linkage, GWA and CNV studies, it is assumed that there are true associations between those genes and ADHD. Further investigation of DIRAS2 by quantitative real-time PCR (qPCR) revealed expression in the hippocampus, cerebral cortex and cerebellum of the human brain and a significant increase in Diras2 expression in the mouse brain during early development. In situ hybrid-izations on murine brain slices confirmed the results gained by qPCR in the human brain. Moreover, Diras2 is expressed in the basolateral amygdala, structures of the olfactory system and several other brain regions which have been implicated in the psychopatholo-gy of ADHD. In conclusion, the results of this work provide further support to the existence of a strong genetic component in the pathophysiology of ADHD and related disorders. KCNIP4, CDH13 and DIRAS2 are promising candidates and need to be further examined to get more knowledge about the neurobiological basis of this common disease. This knowledge is essential for understanding the molecular mechanisms underlying the emergence of this disorder and for the development of new treatment strategies.
All animals learn in order to cope with challenges imposed on them by their environment. This is true also for both larval and adult fruit flies as exemplified in pavlovian conditioning. The focus of this Thesis is on various aspects of the fruit flies learning ability. My main project deals with two types of learning which we call punishment-learning and pain-relief learning. Punishment learning happens when fruit flies are exposed to an odour which is followed by electric shock. After such training, flies have learned that that odour signals pain and consequently will avoid it in the future. If the sequence of the two stimuli is reversed such that odour follows shock, flies learn the odour as a signal for relief and will later on approach it. I first report a series of experiments investigating qualitative and parametric features of relief-learning; I find that (i) relief learning does result from true associative conditioning, (ii) it requires a relatively high number of training trials, (iii) context-shock training is ineffective for subsequent shock-odour learning. A further question is whether punishment-learning and pain-relief learning share genetic determinants. In terms of genetics, I test a synapsin mutant strain, which lacks all Synapsin protein, in punishment and relief-learning. Punishment learning is significantly reduced, and relief-learning is abolished. Pan-neuronal RNAi-mediated knock-down of Synapsin results in mutant-like phenotypes, confirming the attribution of the phenotype to lack of Synapsin. Also, a rescue of Synapsin in the mushroom body of syn97 mutants restores both punishment- and relief-learning fully, suggesting the sufficiency of Synapsin in the mushroom body for both these kinds of learning. I also elucidate the relationship between perception and physiology in adult fruit flies. I use odour-shock conditioning experiments to identify degrees of similarity between odours; I find that those similarity measures are consistent across generalization and discrimination tasks of diverse difficulty. Then, as collaborator of T. Völler and A. Fiala, I investigate how such behavioural similarity/dissimilarity is reflected at the physiological level. I combine the behaviour data with calcium imaging data obtained by measuring the activity patterns of those odours in either the sensory neurons or the projection neurons at the antennal lobe. Our interpretation of the results is that the odours perceptual similarity is organized by antennal lobe interneurons. In another project I investigate the effect of gustatory stimuli on reflexive behaviour as well as their role as reinforcer in larval learning. Drosophila larvae greatly alter their behaviour in presence of sodium chloride. Increasing salt concentration modulates choice behaviour from weakly appetitive to strongly aversive. A similar concentration-behaviour function is also found for feeding: larval feeding is slightly enhanced in presence of low salt concentrations, and strongly decreased in the presence of high salt concentrations. Regarding learning, relatively weak salt concentrations function as appetitive reinforcer, whereas high salt concentrations function as aversive reinforcer. Interestingly, the behaviour-concentration curves are shifted towards higher concentrations from reflexive behaviour (choice behaviour, feeding) as compared to associative learning. This dissociation may reflect a different sensitivity in the respective sensory-motor circuitry.
Seit der Entdeckung des ersten Gens für den Vitamin K Epoxid-Reduktase-Komplex (VKORC1), dem Schlüssel-Enzym für die Regenerierung von Vitamin K, sind keine zusätzlichen Komponenten des Komplexes beschrieben worden. Die einzige bekannte Funktion von VKORC1 ist bislang die Reduktion von Vitamin K-2,3-Epoxid, welches bei der post-translationalen Carboxylierung von Proteinen als oxidierter Kofaktor anfällt, und im sogenannten Vitamin K-Zyklus regeneriert wird. VKORC1 ist zugleich das Zielprotein antikoagulativer Medikamente der Coumarin-Gruppe, wie Warfarin oder Marcumar. Mutationen im VKORC1-Gen führen zu einem signifikanten Effekt auf die benötigte Coumarin-Dosis und die Stabilität der Hämostase in der Thrombosebehandlung mit Antikoagulanzien. Gleichzeitig mit VKORC1 wurde ein stark sequenz-homologes Protein identifiziert, das „VKORC1-like1“ (VKORC1L1) genannt wurde und dessen physiologische Funktion zu Beginn dieser Arbeit völlig unbekannt war. Die vorliegende Arbeit beschäftigte sich mit zwei Aspekten des Vitamin K-Stoffwechsels: A. Den enzym-kinetischen Eigenschaften und der subzellulären Lokalisation des VKORC1L1-Proteins sowie B. der Identifizierung und Charakterisierung von Proteinen, die Interaktionspartner der beiden VKOR-Proteine sein können. Die Ergebnisse können wie folgt zusammengefasst werden: A.1. Die enzym-kinetischen Untersuchungen zeigen starke Ähnlichkeiten zwischen VKORC1 und VKORC1L1: Beide Enzyme haben eine Vitamin K-Epoxidase- und -Reduktase-Aktivität, wobei die Affinitäten zu Vitamin K2-Epoxid deutlich höher sind als die zu Vitamin K1-Epoxid. Beide Enzyme sind durch Warfarin hemmbar und der Austausch der vermutlich am Elektronentransfer beteiligten Cysteine an homologen Positionen führt in beiden Proteinen zu einem fast vollständigen Verlust der Aktivität. A.2. Mittels Ko-Lokalisation konnte VKORC1L1 – wie VKORC1 – in der ER-Membran lokalisiert werden. Folglich schließen wir, dass VKORC1L1 eine ähnliche Struktur, die gleiche zelluläre Lokalisation und zumindest in vitro auch eine VKOR-Aktivität hat und daher eventuell eine weitere Komponente des VKOR-Komplexes darstellen könnte. Allerdings sprechen gewichtige Argumente dagegen, dass beide Proteine funktionell austauschbar sind: Sowohl bei Patienten mit Mutationen in VKORC1 (VKCDF2-Erkrankung), als auch bei VKORC1-Knock-out Mäusen kann das intaktes VKORC1L1-Protein die inaktivierende Mutation im C1-Gen nicht kompensieren. B.1. Mit einem für Membranproteine adaptierten, modifizierten Yeast-Two-Hybrid Screen (Split-Ubiquitin-System) konnten mit VKORC1 und VKORC1L1 als Köder 114 Proteine aus einer Leber-cDNA-Bank als potentielle Interaktionspartner identifiziert werden. Davon wurden 6 Proteine aufgrund ihrer Trefferhäufigkeit und funktioneller Überlegungen mit Hilfe von Ko-Immunpräzipitationsexperimenten und Ko-Immunlokalisation näher untersucht. Interessanterweise zeigen die beiden Trefferlisten starke Überschneidungen. B.2. Es konnte eine in vitro- Interaktion von VKORC1 mit sich selbst und mit VKORC1L1 nachgewiesen werden, die bisher nicht bekannt war. Dies könnte auf der hohen Sequenz- und Struktur-Homologie der beiden Proteine beruhen, führt aber auch zu neuen Hypothesen bezüglich des Vitamin K-Stoffwechsels. B.3. Die Interaktion von VKORC1 und dem „stress-associated endoplasmic reticulum protein 1“ (SERP1) bringt Vitamin K in Zusammenhang mit oxidativem Stress. Dazu passen auch die neuesten Ergebnisse aus der Arbeitsgruppe von Johannes Oldenburg (vormals Würzburg, jetzt Bonn) zur Funktion von VKORC1L1, die eine protektive Rolle von Vitamin K beim Schutz der Zelle vor reaktiven Sauerstoffverbindungen nahe legen. Ob und wie Vitamin K und VKORC1L1 einen neuen Schutzmechanismus gegen Sauerstoffradikale bilden bedarf weiterer Untersuchungen. B.4. Ferner wurde eine Interaktion zwischen VKORC1 und dem „Emopamil-binding-protein“ (EBP) nachgewiesen. Mutationen in EBP führen zu der seltenen genetischen Krankheit Chondrodysplasia punctata. Die Ähnlichkeit der Symptomatik zwischen Chondrodysplasia punctata und der sogenannten Warfarin-Embryopathie, die durch überhöhte Dosierung von Coumarinen während der Schwangerschaft verursacht wird, legt einen Zusammenhang zwischen Vitamin K- und dem Kalziumstoffwechsel nahe. B5. In den Ko-Immunpräzipitationsexperimenten nicht bestätigt haben sich die initial positiven Proteine Protein-Disulfid-Isomerase (PDIA6), CD63, und Fibrinogen-Gamma (FGG). Die Ergebnisse geben Hinweise auf neue Funktionen der VKOR-Proteine beim Schutz vor oxidativem Stress und in der Verbindung zum Kalzium-Stoffwechsel. Beide Aspekte bedürfen weiterführender Untersuchungen. Im Hinblick auf diese neuen Funktionen wäre auch eine kritische Betrachtung der übrigen 85 primären Treffer des Split-Ubiquitin-Screens sinnvoll.
The Contribution of Common and Rare Variants to the Complex Genetics of Psychiatric Disorders

Attention deficit/hyperactivity disorder (ADHD), one of the most frequent childhood-onset, chronic and lifelong neurodevelopmental diseases, affects 5 - 10% of school – aged children and adolescents, and 4% of adults. The classified basic symptoms are - according to the diagnostic system DSM-VI - inattentiveness, impulsivity and hyperactivity. Also daily life of patients is impaired by learning problems, relationship crises, conflicts with authority and unemployment, but also comorbidities like sleep - and eating problems, mood - and anxiety disorders, depression and substance abuse disorders are frequently observed. Although several twin and family studies have suggested heritability of ADHD, the likely involvement of multiple genes and environmental factors has hampered the elucidation of its etiology and pathogenesis. Due to the successful medication of ADHD with dopaminergic drugs like methylphenidate, up to now, the search for candidate genes has mainly focused on the dopaminergic and - because of strong interactions - the serotonergic system, including the already analyzed candidate genes DAT1, DRD4 and 5, DBH or 5-HTTLPR. Recently, DNA copy number changes have been implicated in the development of a number of neurodevelopmental diseases and the analysis of chromosomal gains and losses by Array Comparative Genomic Hybridization (Array CGH) has turned out a successful strategy to identify disease associated genes. Here we present the first systematic screen for chromosomal imbalances in ADHD using sub-megabase resolution Array CGH. To detect micro-deletions and -duplications which may play a role in the pathogenesis of ADHD, we carried out a genome-wide screen for copy number variations (CNVs) in a cohort of 99 children and adolescents with severe ADHD. Using high-resolution aCGH, a total of 17 potentially syndrome-associated CNVs were identified. The aberrations comprise four deletions and 13 duplications with approximate sizes ranging from 110 kb to 3 Mb. Two CNVs occurred de novo and nine were inherited from a parent with ADHD, whereas five are transmitted by an unaffected parent. Candidates include genes expressing acetylcholine-metabolising butyrylcholinesterase (BCHE), contained in a de novo chromosome 3q26.1 deletion, and a brain-specific pleckstrin homology domain-containing protein (PLEKHB1), with an established function in primary sensory neurons, in two siblings carrying a 11q13.4 duplication inherited from their affected mother. Other genes potentially influencing ADHD-related psychopathology and involved in aberrations inherited from affected parents are the genes for the mitochondrial NADH dehydrogenase 1 alpha subcomplex assembly factor 2 (NDUFAF2), the brain-specific phosphodiesterase 4D isoform 6 (PDE4D6), and the neuronal glucose transporter 3 (SLC2A3). The gene encoding neuropeptide Y (NPY) was included in a ~3 Mb duplication on chromosome 7p15.2-15.3, and investigation of additional family members showed a nominally significant association of this 7p15 duplication with increased NPY plasma concentrations (empirical FBAT, p = 0.023). Lower activation of the left ventral striatum and left posterior insula during anticipation of large rewards or losses elicited by fMRI links gene dose-dependent increases in NPY to reward and emotion processing in duplication carriers. Additionally, further candidate genes were examined via Matrix assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF MS). This method enables the analysis of SNPs directly from human genomic DNA without the need for initial target amplification by PCR. All these findings implicate CNVs of behavior-related genes in the pathogenesis of ADHD and are consistent with the notion that both frequent and rare variants influence the development of this common multifactorial syndrome. The second part of this work concentrates on MLC1, a gene associated with Megalencephalic leukoencephalopathy with subcortical cysts, located on chromosome 22q13.33. To get more insight in the disease itself, a targeting vector for a conditional knockout mouse was constructed using homologous recombination. Furthermore, MLC1 has been suggested as a risk gene for schizophrenia, especially the periodic catatonia subtype. An initially identified missense mutation was found to be extremely rare in other patient cohorts; however, a recent report again argued for an association of two intronic MLC1 SNPs with schizophrenia and bipolar disorder. A case-control study of these polymorphisms as well as SNPs in the transcriptional control region of MLC1 was conducted in 212 chronic schizophrenic patients, 56 of which suffered from periodic catatonia, 106 bipolar patients, and 284 controls. Both intronic and promoter polymorphisms were specifically and significantly associated with periodic catatonia but not schizophrenia or bipolar disorder in general. A haplotype constructed from all polymorphisms was also associated with periodic catatonia. The MLC1 variation is associated with periodic catatonia; whether it constitutes a susceptibility or a modifier gene has to be determined.
Asymptomatische Bakteriurie (ABU) stellt eine bakterielle Infektion der Harnblase über einen langen Zeitraum dar, die häufig von Escherichia coli hervorgerufen wird, ohne dass typische Symptome einer Harnwegsinfektion auftreten. Um die Charakteristika von ABU E. coli Isolaten genauer zu untersuchen, wurden die Geno- und Phänotypen von 11 ABU-Isolaten verglichen. Außerdem wurden in mehreren aufeinanderfolgenden in vivo-Reisolaten des Modell-ABU Stammes 83972 die Veränderungen im Transkriptom, Proteom und Genom während einer langfristigen Persistenz in der menschlichen Blase charakterisiert. Schließlich wurde der Effekt des menschlichen Wirtes auf die bakterielle Adaptation durch einen Vergleich von in vitro- mit in vivo-kultivierten Stämmen abgeschätzt. ABU-Isolate stellt eine heterogene Gruppe von Organismen dar. Diese können den vier phylogenetischen Hauptgruppen von E. coli sowie unterschiedlichen klonalen Gruppen zugeordnet werden. Dementsprechend unterscheiden sie sich erheblich bezüglich der Zusammensetzung des Genomes, der Genomgröße und auch der Ausstattung mit UPEC-typischen Virulenz-assoziierten Genen. Multi-Lokus-Sequenz-Typisierung legt nahe, dass bestimmte ABU Stämme sich durch Genomreduktion aus UPEC Stämmen entwickelt haben, die eine Harnwegsinfektion mit charakteristischen Symptomen auslösen konnten. Folglich erlaubt die hohe Genomplastizität von E. coli keine generalisierte Betrachtung einzelner Isolate eines Klons. Genomreduktion über Punktmutationen, Genom-Reorganisation und Deletionen resultierte in der Inaktivierung einiger Gene, die für einige UPEC Virulenz-Faktoren kodieren. Dies stützt die Vorstellung, dass eine verminderte bakterielle Aktivierung der Entzündung der Wirtsschleimhaut den Lebensstil von ABU (bei diesen E. coli-)Isolaten fördert. Genregulation und genetische Diversität sind Strategien, die es Bakterien ermöglichen unter sich fortlaufend ändernden Bedingungen zu leben bzw. zu überleben. Um die anpassungsbedingten Veränderungen bei einem langfristigen Wachstum in der Blase zu untersuchen, wurden aufeinanderfolgende Reisolate, denen eine langfristige in vivo-Kolonisierung im menschlichen Wirt beziehungsweise eine in vitro-Kultivierung vorausgegangen ist, im Hinblick auf Veränderungen Genexpression und Genomorganisation analysiert. In diesem Zusammenhang konnte gezeigt werden, dass E. coli in der Lage ist, seine metabolischen Netzwerke verschiedenen Wachstumsbedingungen anzupassen und individuelle bakterielle Kolonisierungsstrategien entwickeln kann. Transkriptom- und Proteom-Analysen zeigten verschiedene metabolische Strategien zur Nährstoffbeschaffung und Energieproduktion bei untersuchten in vivo-Reisolaten vom Stamm 83972, die es ihnen ermöglichen, den Wirt zu kolonisieren. Das Zurückgreifen auf D-Serin, Deoxy- und Ribonucleoside sowie die bidirektionale Umwandlung zwischen Pentose und Glucuronat waren hoch-regulierte Stoffwechselwege, die die in vivo-Reisolate mit zusätzlicher Energie für ein effizientes Wachstum in der Blase versorgen. Zudem wurden in dieser Studie die Netzwerke für eine Reaktion auf Abwehrmechanismen des Wirtes erforscht: Erstmals wurde hier die Rolle der Klasse-III-Alkoholdehydrogenase AdhC, bekannt durch ihre Bedeutung bei der Entgiftung von Stickstoffmonoxid, bei der Wirtsantwort während einer asymptomatischen Bakteriurie gezeigt. Aufeinanderfolgende in vivo- und in vitro-Reisolate vom Stamm 83972 wurden ebenfalls bezüglich ihrer Genomstruktur analysiert. Einige Veränderungen in der Genomstruktur der aufeinanderfolgenden Reisolate, die von einer humanen Kolonisierungsstudie stammen, implizieren die Bedeutung einer Interaktion der Bakterien mit dem Wirt bei der Mikroevolution der Bakterien. Dagegen war die Genomstruktur von Reisolaten eines langfristigen in vitro-Kultivierungsexperiments, bei dem sich der Stamm 83972 ohne Wirtskontakt vermehrt hat, nicht von Veränderungen betroffen. Das legt nahe, dass die Immunantwort eine Genomplastizität fördert und somit eine treibende Kraft für den ABU Lebensstil und die Evolution im Harnwegstrakt ist.
Zusammenfassung: Hintergrund: Mit einer Lebenszeitprävalenz von 1 % und einem Ersterkrankungsalter in der frühen Adoleszenzperiode verursachen Schizophrenien (SCZ) und bipolare Störungen (BPD) großes individuelles Leid. Die genetische Komponente beider Erkrankungen liegt mit einer Heritabilität von bis zu 80 % im Vergleich zum Anteil von Umweltfaktoren sehr hoch. Aufgrund seiner Lage auf dem Locus 12q22-24, einem Hot spot für SCZ und BPD, stellt DYNLL1 ein interessantes positionales Kandidatengen dar. Das Protein, eine 8 kD schwere leichte Dyneinkette ist ein multifunktionales Protein. Durch seine Funktion als Inhibitor von NOS-I, seiner Beteiligung am postsynaptischen NMDA-Proteinkomplex, einer möglichen Interaktion mit NUDEL/DISC1, seiner Ähnlichkeit zu KIF2 und nicht zuletzt wegen der Interaktion mit KIBRA stellt DYNLL1 auch aufgrund seiner Funktion ein relevantes funktionelles Kandidatengen für beide Erkrankungen dar. Methoden: In einer Fall-Kontrollstudie wurden daher sechs Single nucleotid polymorphismen (SNPs) und deren entsprechenden Haplotypen bei 284 Kontrollen, 246 Patienten, die an einer SCZ und 90 Patienten, die an einer BPD litten analysiert, um eine Assoziation dieses Gens mit den entsprechenden Phänotypen zu untersuchen. Ergebnisse: Es zeigte sich eine Assoziation des Markers rs787828 mit SCZ, darüber hinaus eine signifikante Assoziation eines Haplotyps (TTATAG), letztere allerdings nur mit einer Frequenz von 1%. Bei der bipolaren Störung waren dagegen sowohl zwei Polymorphismen (rs1167705 und rs580016), als auch ein Haplotyp (TTGTAG) signifikant mit der Erkrankung assoziiert. Aufgrund der kleineren Stichprobengröße ist es jedoch wichtig, diesen Befund nochmals zu replizieren, um falsch positive Befunde auszuschließen. Zusammenfassung: Sowohl SNPs als auch Haplotypen im DYNLL1 Gen zeigten Assoziationen mit Schizophrenie und der bipolaren Störung, was die These unterstützt, dass DYNLL1 ein relevantes Kandidatengen für beide Erkrankungen ist. Um die Bedeutung von DYNLL1 in der Pathophysiologie der SCZ und der bipolaren Störung weiter aufzuklären, müssen die assoziierten Varianten bezüglich möglicher Auswirkungen auf Genexpression, Proteinfunktion und physiologische Parametern hin weiter untersucht werden.
Die primäre Gonarthrose ist ein komplexer, multifaktoriell bedingter Krankheitsprozess mit genetischer Komponente und gilt aufgrund ihrer epidemiologischen und sozioökonomischen Bedeutung als Volkskrankheit. Die Gene bzw. Proteine der Wnt- Familie spielen eine wichtige Rolle in der komplexen Regulation der Morphogenese, der Zelldifferenzierung , -proliferation und der Homöostase von Knochen und Knorpel. LRP 1 und LRP 5 fungieren als Co-rezeptoren des Wnt- Signalweges. In der vorliegenden Untersuchung wurden der A217V Polymorphismus des LRP 1 Gens und der A1330V Polymorphismus des LRP 5 Gens auf eine signifikante Assoziation mit dem Auftreten primärer Gonarthrose in einem arthroskopisch definierten Patientenkollektiv untersucht. Ziel der Studie war es, klar definierte Kollektive mit möglichst extremen Zuständen gegenüberzustellen und eine hochspezifische und hochsensitive Diagnostik zu verwenden. Junge, bereits in frühen Jahren an Gonarthrose erkrankte Patienten (<45 Jahre, Gruppe 1, n= 100) wurden älteren Patienten (>55 Jahre, Gruppe 3, n= 103), die nicht an Gonarthrose erkrankt waren, gegenübergestellt. Als weitere Gruppe dienten Patienten, welche eine endoprothetische Versorgung bei fortgeschrittener Gonarthrose erhielten (>55 Jahre, Gruppe 2, n= 86). Die Diagnose der primären Gonarthrose wurde anhand anamnestischer Daten sowie arthroskopischer bzw. direkt visuell erhobender Befunde im Rahmen der Arthrotomie bei Prothesenimplantation gestellt. Der Arthroseausschluss (Gruppe 3) erfolgte ebenfalls arthroskopisch. Methodisch wurden der WOMAC- Score, die Restriktions- Fragment- Längen- Polymorphismus (RFLP) Analyse sowie die Gensequenzierung angewendet. Die Allelverteilung der untersuchten Polymorphismen von LRP 1 und LRP 5 konnte zwischen den beiden gegenübergestellten Gruppen 1 und 3 keinen signifikanten Unterschied aufweisen. Für den LRP 5 A1330V Polymorphismus ergab sich ein signifikanter Unterschied (p< 0,05) zwischen den Gruppen 2 und 3. Dies kann als Hinweis auf eine mögliche arthrosefördernde oder auch protektive Funktion dieses Polymorphismus gewertet werden. Im Vergleich der Extremkollektive (Gruppe 1 vs. Gruppe 3) deutet sich lediglich ein Trend an (p= 0,067), so dass eine Assoziation des A1330V Polymorphismus des LRP 5 Gens mit primärer Gonarthrose erst durch weitere Untersuchungen mit höherer Fallzahl eindeutig bestätigt bzw. widerlegt werden kann.
In dieser Arbeit wurde die systematische Suche nach krankheitsassoziierten Genen bei periodischer Katatonie fortgeführt. Für diese Erkrankung war die klinische Abgrenzbarkeit und die familiäre Häufung signifikant und ließ aufgrund der vertikalen Transmission und dem Auftreten über mehrere Generationen und hinweg auf einen Hauptgeneffekt schließen. Nach der Durchführung von Kopplungs-Analysen kristallisierten sich zwei koppelnde Regionen auf den Chromosomen 15 und 22 heraus. Mittels Haplotypanalyse konnte der Genort auf Chromosom 22q13 auf einen knapp 5 Mbp großen Bereich eingeschränkt werden. Im kodierenden Bereich des MLC1-Genes segregierte im mit periodischer Katatonie assoziierten Haplotyp eine Variante (p.Leu309Met). Da Mutationen im MLC1-Gen bereits im Zusammenhang mit Megalenzephaler Leukoenzephalopathie beschrieben worden waren, wurden in dieser Arbeit zunächst fünf Patienten mit dieser Erkrankung auf Mutationen in kodierenden Bereichen von MLC1 systematisch untersucht. Daran schloss sich eine Analyse dieses Gens bei 140 Patienten mit periodischer Katatonie an. Ein Zusammenhang zwischen Mutationen in MLC1 und dem Auftreten von Megalenzephaler Leukoenzephalopathie wurde untermauert, wohingegen die Ergebnisse eindeutig gegen eine Assoziation mit periodischer Katatonie sprachen. Ein weiteres im Gehirn exprimiertes Kandidatengen (KIAA0767/DIP) wurde in dieser Arbeit untersucht. Dabei wurden sechs SNPs im exonnahen intronischen Bereich entdeckt sowie eine Variante im Exonbereich (p.Glu156Asn). Dies ist eine seltene Normvariante, eine Assoziation zur periodischen Katatonie wurde in einer Fall-Kontroll-Studie ausgeschlossen. Insgesamt wurde durch die systematische Mutationsanalyse die Kandidatenregion auf Chromosom 22q13.3 weiter eingeengt. Gegen einen Zusammenhang zwischen MLC1 und periodischer Katatonie sprechen die vorgestellten validen Ergebnisse.
