Saposin-ähnliche Proteine (SAPLIPs) sind membraninteragierende Proteine, die sich durch die konservierte Position von drei Disulfidbrücken, einer typischen alpha-helikalen Proteinfaltung und der Fähigkeit mit Lipiden zu interagieren, auszeichnen. Ihre zellulären Funktionen sind äußerst vielfältig. Bis zum Beginn des Genomsequenzierungsprojektes waren die Amoebapores die einzigen bekannten und charakterisierten SAPLIPs von Entamoeba histolytica, dem Erreger der humanen Amöbenruhr. Aufgrund ihrer antimikrobiellen Aktivität stellen sie für diesen parasitischen Einzeller, der sich von phagozytierten Bakterien ernährt, wichtige Effektormoleküle dar. Sie können aber auch cytolytisch auf Wirtszellen wirken und werden deshalb als bedeutender Pathogenitätsfaktor angesehen. Die theoretische computergestützte Datenbankanalyse nach Abschluss der Genomsequenzierung ergab, dass es 16 weitere Gene kodierend für SAPLIPs zusätzlich zu den drei Amoebapore-Genen gibt. Die Sequenzen der neuen SAPLIPs sind abgesehen von dem Cysteinmotiv divers und auch die Größe der Proteine ist sehr unterschiedlich (77 - 1009 Aminosäuren). Alle besitzen sie jedoch eine einzige, C-terminal gelegene SAPLIP Domäne. Außer der SAPLIP-Domäne konnten keine weiteren bekannten funktionellen oder strukturellen Domänen in den relevanten Datenbanken identifiziert werden, die auf mögliche Funktionen hätten hinweisen können. Alle SAPLIP-Gene werden gleichzeitig in axenisch kultivierten Trophozoiten transkribiert wie durch reverse Transkriptions-PCR gezeigt wurde. Die vergleichende transkriptionelle Analyse im Mikroarray ergab, dass nach Kontakt mit menschlichen Kolonzellen keine Hochregulierung dieser Gene mit Ausnahme des Amoebapore A Gens stattfindet. Für die parallele Klonierung der verschiedenen SAPLIP-Domänen wurde ein ”Expressionsscreening” in E.coli mit dem grün fluoreszierenden Protein als Reporterprotein etabliert, das die erfolgreiche Klonierung und Expression eines Fragments aufgrund der Fluoreszenz der Bakterienkolonie bereits auf der Ebene der Transformation anzeigt. Die rekombinant exprimierte und bis zur Homogenität gereinigte SAPLIP-Domäne von SAPLIP 12 wies Amoebapore-ähnliche Aktivitäten auf. Unter Verwendung von Liposomen konnte porenbildende Aktivität nachgewiesen werden, wobei diese Aktivität stark an einen sauren pH-Wert gebunden ist. Die SAPLIP-Domäne 12 ist aber auch antibakteriell und dieses sogar mit vergleichbarer Selektivität wie Amoebapore A, nämlich Zelllyse von gram-positiven B. megaterium war nachweisbar, jedoch nicht von gram-negativen E. coli. Strukturell unterscheiden sich die SAPLIP-Domäne 12 und Amoebapore A bezüglich der Exposition positiver Ladungsansammlungen auf der Proteinoberfläche und des Fehlens des für den Mechanismus der Amoebapores essentiellen Histidinrestes an entsprechender Position in der Sequenz. Darüber hinaus übt die SAPLIP-Domäne 12 eine im Vergleich zum Amoebapore A geringere spezifische Aktivität aus. Diese Eigenschaften weisen darauf hin, dass es sich um einen anderen Wirkungsmechanismus handeln könnte. Für die SAPLIP-Domäne 12 wäre eine über die positiven Ladungen der Proteinoberfläche vermittelte Interaktion mit den negativ geladenen Phospholipidköpfen von Membranen denkbar, die bei Erreichen einer bestimmten Konzentration in einer Störung der Lipidordnung und letztendlich in der Auflösung der Membranstruktur resultieren könnte. SAPLIP 3 ähnelt den Amoebapores in der Größe und molekularen Architektur und kann somit als funktionelle Einheit angesehen werden, es unterscheidet sich aber durch eine hohe negative Nettoladung von den Amoebapores. Außerdem ist das rekombinante SAPLIP 3 nicht antibakteriell und die Membraninteraktionen dieses SAPLIPs unterscheiden sich grundlegend von denjenigen, die für die Amoebapores beschrieben sind. SAPLIP 3 zerstört nicht einfach die Liposomenstruktur wie von den porenbildenden Amoebapores bekannt, sondern es vermittelt die Fusion von multilamellaren Liposomen unter Freisetzung des Liposomeninhalts. Diese Aktivität ist abhängig von der Anwesenheit anionischer Lipide und von einem sauren pH-Wert. Die Fähigkeit zur Vesikelfusion sowie die Verteilung der negativen Ladungen von SAPLIP 3 auf der Proteinoberfläche ähneln Merkmalen des humanen Saposin C. Neben der Funktion als Cofaktor von Exohydrolasen, die im Sphingolipid Katabolismus involviert sind, wird angenommen, dass die Fähigkeit von Saposin C, Vesikel zu fusionieren, wichtig für die Reorganisation der humanen lysosomalen Kompartimente ist. Die Saposin C-ähnlichen Charakteristika von SAPLIP 3 geben Grund zu der Annahme, dass es bereits in einem so basalen Organismus wie der Amöbe ein Protein mit Saposin-ähnlichen membranfusionierenden Aktivitäten gibt und dass dieses SAPLIP entsprechende Funktionen während endo- und exozytotischer Transportprozesse in der Amöbe übernehmen könnte.
In der inkompatiblen Interaktion von Lycopersicon esculentum und Cuscuta reflexa wird die Ausbildung von Haustorien, spezieller Organe, die der Nahrungsaufnahme durch den Parasiten dienen,bereits in einem sehr frühen Stadium der Infektion gehemmt. Um einen Einblick in die Regulationsmechanismen der Tomatenreaktion zu gewinnen, wurde eine Subtraktive Hybridisierung durchgeführt und es konnten 20 Gene identifiziert werden, deren Transkripte nach Cuscuta-Befall im Wirtsgewebe akkumulieren. Entsprechend ihrer möglichen Proteinfunktion lassen sich die mRNAs verschiedenen Bereichen der Tomatenreaktion im Infektionsprozess zuordnen: (a) Abwehr-assoziierte Proteine, (b) Signaltransduktionsassoziierte Proteine, (c) Zellstreckungsassoziierte Proteine und (d) Proteine mit bislang vollständig unbekannter Funktion. Einige der identifizierten mRNAs wurden durch Northern Analysen näher charakterisiert. Da eine der mRNAs eine mögliche Xyloglucanendotransglycosylase (XET) kodiert, wurde die XET-Aktivität im Tomatengewebe nach Infektion bestimmt. Außerdem wurde der Einfluß des Phytohormons Auxin auf die Akkumulation der Xyloglucanendotransglycosylase LeEXT1 sowie des Aquaporins LeAqp2 untersucht. Trotz auxinregulierter Transkription nach Cuscuta-Befall zeigte die auxininsensitive Tomatenmutante diageotropica im Vergleich zum Wildtyp keine veränderte Kompatibilität.
Funktionelle Charakterisierung neuer Proteine der inneren Kernmembran von Drosophila melanogaster: Drosophila Lamin B Rezeptor (dLBR), ein integrales Membranprotein der inneren Kernmembran; Bocksbeutel alpha und Bocksbeutel beta, LEM-Domänen Proteine sowie deren potentiellen Interaktionspartner Drosophila Barrier-to-Autointegration Factor (dBAF).
The auditory system is an exquisitely complex sensory organ dependent upon the synchronization of numerous processes for proper function. The molecular characterization of hereditary hearing loss is complicated by extreme genetic heterogeneity, wherein hundreds of genes dispersed genome-wide play a central and irreplaceable role in normal hearing function. The present study explores this area on a genome-wide and single gene basis for the detection of genetic mutations playing critical roles in human hearing.
This work initiated with a high resolution SNP array study involving 109 individuals. A 6.9 Mb heterozygous deletion on chromosome 4q35.1q35.2 was identified in a syndromic patient that was in agreement with a chromosome 4q deletion syndrome diagnosis. A 99.9 kb heterozygous deletion of exons 58-64 in USH2A was identified in one patient. Two homozygous deletions and five heterozygous deletions in STRC (DFNB16) were also detected. The homozygous deletions alone were enough to resolve the hearing impairment in the two patients. A Sanger sequencing assay was developed to exclude a pseudogene with a high percentage sequence identity to STRC from the analysis, which further solved three of the six heterozygous deletion patients with the hemizygous, in silico predicted pathogenic mutations c.2726A>T (p.H909L), c.4918C>T (p.L1640F), and c.4402C>T (p.R1468X). A single patient who was copy neutral for STRC and without pathogenic copy number variations had compound heterozygous mutations [c. 2303_2313+1del12 (p.G768Vfs*77) and c.5125A>G (p.T1709A)] in STRC. It has been shown that STRC has been previously underestimated as a hearing loss gene. One additional patient is described who does not have pathogenic copy number variation but is the only affected member of his family having hearing loss with a paternally segregating translocation t(10;15)(q26.13;q21.1).
Twenty-four patients without chromosomal aberrations and the above described patient with an USH2A heterozygous deletion were subjected to a targeted hearing loss gene next generation sequencing panel consisting of either 80 or 129 hearing-relevant genes. The patient having the USH2A heterozygous deletion also disclosed a second mutation in this gene [c.2276G>T (p.C759F)]. This compound heterozygous mutation is the most likely cause of hearing loss in this patient. Nine mutations in genes conferring autosomal dominant hearing loss [ACTG1 (DFNA20/26); CCDC50 (DFNA44); EYA4 (DFNA10); GRHL2 (DFNA28); MYH14 (DFNA4A); MYO6 (DFNA22); TCF21 and twice in MYO1A (DFNA48)] and four genes causing autosomal recessive hearing loss were detected [GJB2 (DFNB1A); MYO7A (DFNB2); MYO15A (DFNB3), and USH2A]. Nine normal hearing controls were also included. Statistical significance was achieved comparing controls and patients that revealed an excess of mutations in the hearing loss patients compared to the control group. The family with the GRHL2 c.1258-1G>A mutation is only the second family published worldwide with a mutation described in this gene to date, supporting the initial claim of this gene causing DFNA28 hearing loss. Audiogram analysis of five affected family members uncovered the progressive nature of DFNA28 hearing impairment. Regression analysis predicted the annual threshold deterioration in each of the five family members with multiple audiograms available over a number of years.

Spinal muscular atrophy with respiratory distress type 1 (SMARD1) is a fatal monogenic motoneuron disease in children with unknown etiology caused by mutations in the immunoglobulin μ-binding protein 2 (IGHMBP2) gene coding for DNA/RNA ATPase/helicase. Despite detailed knowledge of the underlying genetic changes, the cellular mechanisms leading to this disease are not well understood. In the Nmd2J ("neuromuscular disorder") mouse, the mouse model for the juvenile form of SMARD1 patients, in which similar pathological features as diaphragmatic paralysis and skeletal muscle atrophy are observed. Ex vivo studies in Nmd2J mice showed that loss of the motor axon precedes atrophy of the gastrocnemius muscle and does not correlate with neurotransmission defects in the motor endplate. The already described independent myogenic anomalies in the diaphragm and heart of the Nmd2J mouse raised the question whether spinal motoneuron degeneration develops cell autonomously. Ighmbp2 is predominantly localized in the cytoplasm and seems to bind to ribosomes and polysomes, suggesting a role in mRNA metabolism.
In this Ph.D. thesis, morphological and functional analyses of isolated Ighmbp2-deficient (Ighmbp2-def.) motoneurons were performed to answer the question whether the SMARD1 phenotype results from dysregulation of protein biosynthesis. Ighmbp2-deficient motoneurons show only negligible morphological alterations with respect to a slight increase in axonal branches. This observation is consistent with only minor changes of transcriptome based on RNA sequencing data from Ighmbp2-deficient motoneurons. Only the mRNA of fibroblast growth factor receptor 1 (Fgfr1) showed significant up-regulation in Ighmbp2-deficient motoneurons. Furthermore, no global aberrations at the translational level could be detected using pulsed SILAC (Stable Isotope Labeling by Amino acids in cell culture), AHA (L-azidohomoalanine) labeling and SUnSET (SUrface SEnsing of Translation) methods. However, a reduced β-actin protein level was observed at the growth cones of Ighmbp2-deficient motoneurons, which was accompanied with a reduced level of Imp1 protein, a known β-actin mRNA interactor. Live-cell imaging studies using fluorescence recovery after photobleaching (FRAP) showed translational down-regulation of eGFPmyr-β-actin 3'UTR mRNA in the growth cones and the cell bodies, although the amount of β-actin mRNA and the total protein amount in Ighmbp2-deficient motoneurons showed no aberrations. This compartment-specific reduction of β-actin protein occurred independently of a non-existent direct IGHMBPF2 binding to β-actin mRNA. Fgfr1, which was upregulated on the RNA level, did not show an increased protein amount in Ighmbp2-deficient motoneurons, whereas a reduced amount could be detected. Interestingly, a correlation could be found between the reduced amount of the Imp1 protein and the increased Fgfr1 mRNA, since the IMP1 protein binds the FGFR1 mRNA and thus could influence the transport and translation of FGFR1 mRNA. In summary, all data suggest that Ighmbp2 deficiency leads to a local but modest disturbance of protein biosynthesis, which might contribute to the motoneuron defects of SMARD1.
Das Cytokin Interleukin-4 (IL-4) ist ein essentieller Faktor bei der Entstehung von Sofort-Typ Allergien. Die Bindung von IL-4 an seinen Rezeptor und die anschließende Phosphorylierung des IL-4 aktivierten Transkriptionsfaktors Stat6 ist ein Schlüsselereignis bei der allergischen Immunantwort. In der vorliegenden Arbeit werden Ergebnisse zur Hemmung der Stat6 vermittelten Signaltransduktion des IL-4 Rezeptors vorgestellt. Dazu wurde ein Vektorsystem etabliert, bei dem ein von dem Drosophila-Transkriptionsfaktor Antennapedia abgeleitetes 16 AS langes Peptid benutzt wird. Dieses Antennapediapeptid kann Plasmamembranen lebender Zellen energie- und rezeptorunabhängig durchqueren und dabei andere hydrophile Moleküle mittransportieren. Stat6 bindet über eine SH2 Domäne an phosphorylierte Reste von IL4Ra und bildet, nachdem es selbst phosphoryliert ist, mit anderen Stat6-Molekülen aktive Dimere. Ein aus der Stat6-Bindestelle des IL-4Ra abgeleitetes phosphoryliertes Peptid (Stat6BP) wurde mit Hilfe des Antennapediapeptids in verschiedene humane und murine Zellinien transportiert. Für Stat6BP konnte mit Hilfe von spezifischer Immunpräzipitation und Western-Blot gezeigt werden, dass es IL-4 induzierte Phosphorylierung und Aktivierung von Stat6 transient hemmen kann. Durch zusätzliche Applikation des Tyrosinphosphataseinhibitors Natriumpervanadat gelang es, die hemmende Wirkung von Stat6BP zu verlängern. Unter gleichen Bedingungen konnte auch gezeigt werden, dass Stat6BP spezifisch die Aktivierung von Stat6 hemmt, da die durch IL-4 oder IL-3 induzierte Phosphorylierung des eng verwandten Stat5 völlig unbeeinträchtigt bleibt. Ferner wurde durch das Peptid die Expression eines Stat6 kontrollierten Reportergens gehemmt. Im Rahmen dieser Arbeit wurde außerdem die Rolle der Src-Typ Kinasen p56lck und p59fyn in der IL-4 Signaltransduktion in unterschiedlichen T-Zellinien untersucht. Es zeigte sich, dass die Aktivierung der beide Kinasen stark von der getesteten Zellinie abhängt. In einigen T-Zellinien aktiviert IL-4 eher p56lck, in anderen eher p59fyn.
B-cells of the rheumatoid synovial tissue are a constant part of and, in some histopathological subtypes, the dominant population of the inflammatory infiltrate, located in the region of tissue destruction. The pattern of B-cell distribution and the relationship to the corresponding antigen-presenting cells (follicular dendritic reticulum cells: FDCs) show a great variety. B-cells may exhibit (i) a follicular organization forming secondary follicles; (ii) follicle-like patterns with irregularly formed FDC networks, and (iii) a diffuse pattern of isolated FDCs. Molecular analysis of immunoglobulin VH and VL genes from human synovial B-cell hybridomas and synovial tissue demonstrates somatic mutations due to antigen activation. The FDC formations in the synovial tissue may therefore serve as an environment for B-cell maturation, which is involved in the generation of autoantibodies. An autoantibody is defined as "pathogenic" if it fulfills the Witebsky-Rose-Koch criteria for classical autoimmune diseases: definition of the autoantibody; induction of the disease by transfer of the autoantibody; and isolation of the autoantibody from the disease-specific lesion. B-cells from rheumatoid synovial tissue show specificity for FcIgG, type II collagen, COMP, sDNA, tetanus toxoid, mitochondrial antigens (M2), filaggrin and bacterial HSPs. The contributions of these antigens to the pathogenesis of RA are still hypothetical. A possible contribution could derive from crossreactivity and epitope mimicry: due to crossreaction, an antibody directed originally against a foreign infectious agent could react with epitopes from articular tissues, perpetuating the local inflammatory process. The characteristic distribution pattern, the localisation within the area of tissue destruction, the hypermutated IgVH and IgVL genes, and their exclusive function to recognize conformation-dependent antigens suggest a central role for B-cells in the inflammatory process of rheumatoid arthritis. Therefore, the analysis of synovial B-cell hybridomas and experimental expression of synovial IgVH and IgVL genes will help to characterise the antigens responsible for the pathogenesis of rheumatoid arthritis. In the present study 55 IgVH genes amplified from 3 different anatomical regions of a RA patient were analysed adding further information on synovial B-cell maturation and recirculation in RA. This analysis demonstrated somatically mutated IgVh genes in all different regions with amino acid deletions and mixed IgVh molecules, suggesting the existence of a novel pathway to generate (auto)antibody specificities. The comparison of amino acid sequences of amplified genes belonging to the VH1 family (with predominantly the same germline counterpart) exhibited a strong homology, indicating an apparently conserved mutational pattern. This suggests that the number of antigens activating B-cells in the different locations is restricted. The most striking result was the finding of clonally related sequences in different anatomical regions indicating a recirculation of activated B-cells between the different affected joints. Also in the present study a synovial B-cell hybridoma was analyzed for its specific recognition of cartilage antigens. A heptameric peptide of cartilage oligomeric protein (COMP) could be defined as the target structure. The IgVH-gene (IgHV4-59*01) of the IgG2l hybridoma has somatically mutated genes with high R/S values in the CDR regions (9:2). Thus, indicating that this hybridoma originates from a synovial B-cell which has been antigen activated/selected for its affinity. To analyse the presence of the clonotypic IgHV4-59*01 sequences in other cases of RA and osteoarthritis (OA) synovitis, primers specific for the CDR3 rearrangement of this hybridoma were used. The clonotypic and clone related sequences (98 per cent ± 1 per cent homology) could only be detected in synovitis of RA cases but not in OA cases indicating that this B-cell is specific to RA synovitis. The identified heptameric peptide of COMP was used in a peptide ELISA to analyse whether there is a specific binding in RA serum samples. Serum samples (IgG) from RA patients (n=22) showed a significant higher efficiency to the COMP heptamer than the OA sera (n=24) and the age matched healthy controls (n=20) (for both p<1x10-4, Students t-test). The specificity of this B-cell hybridoma may therefore be defined as RA specific. Since COMP is restricted to cartilage and tendons which are organs specifically affected in RA this COMP specific autoantibody represents the first organ specific autoantibody in RA. The IgG2 COMP specific autoantibody with somatically mutated IgVH genes is different from germline encoded, antigen clearing IgM autoantibodies and may therefore be directly involved as an "arthritogenic autoantibody" in cartilage and tendons destruction by complement activation.
B-Zell-Lymphome vom mukosa-assoziierten lymphatischen Gewebe (MALT)-Typ sind die größte Gruppe der extranodalen Lymphome. Sie enstehen vor dem Hintergrund einer chronischen Entzündung, wie etwa einer Helicobacter pylori-assoziierten Gastritis im Magen. Die Mechanismen der Lymphomgenese sind weitgehend unverstanden. Der Befund, dass die malignen B-Zellen der MALT-Typ Lymphome autoreaktiv sind und durch Antigen oder T-Zell-vermittelte Signale stimuliert werden, weist auf ein mögliches Ver-sagen der T-Zell-Kontrolle hin. Zur Prüfung dieser Hypothese wurden in der vorliegenden Arbeit sowohl tumor-infiltrierende T-Zellen als auch maligne B-Zellen aus MALT-Typ Lymphomen untersucht. Mit Hilfe von Expressionsanalysen der Vb -Kette des T-Zell-Rezeptors (TCR) wurden antigen-induzierte klonale Expansionen in den tumor-infiltrierenden T-Zellen nachgewiesen. Weiterhin wurden ähnliche antigen-erkennende Regionen (CDR3) des TCR in tumor-infiltrierenden T-Zellen aus zwei verschiedenen MALT-Typ Lymphomen gefunden. Diese Ergebnisse charakterisieren die T-Zellen als funktionell und auch als potenziell tumor-reaktiv. Um mögliche Defekte bei malignen B-Zellen zu untersuchen, wurde eine in vitro T/B-Zell-Kokultur entwickelt. Damit war es möglich, die Interaktion des apoptose-induzierenden Oberflächenmoleküls FasL auf aktivierten T-Zellen mit dem entsprechenden Todesrezeptor Fas auf malignen B-Zellen zu untersuchen. Drei von sieben MALT-Typ Lymphomen und vier von fünf DLBL erwiesen sich als resistent gegen FasL-vermittelte Apoptose. Meine Untersuchungen deuten darauf hin, dass hierfür eine mutationsbedingte funktionelle Inaktivierung des Fas-Rezeptors verantwortlich ist. In Fas-Transkripten aller untersuchten malignen B-Zellen wurden insgesamt 14 verschiedene Punktmutationen gefunden, die zu Aminosäureaustauschen bei der Translation führen. Zehn dieser Mutationen waren mit der Apoptose-Resistenz maligner B-Zellen assoziiert. Durch ergänzende Untersuchungen konnten alternative Mechanismen der Apoptose-Resistenz wie etwa reduzierte Fas-Expression, Produktion von löslichem Fas (sFas) oder Störungen in der Fas-Signalkaskade weitgehend ausgeschlossen werden. Aus diesen Ergebnissen lässt sich folgender Schluss ziehen: Resistenz gegen FasL/Fas-vermittelte Apoptose ist ein Mechanismus der frühen MALT-Typ Lymphomgenese und möglicherweise auf bestimmte Fas-Mutationen zurückzuführen. Durch Apoptose-Resistenz entkommen die malignen B-Zellen der MALT-Typ Lymphome der vorhandenen T-Zell-Kontrolle. Gleichzeitig nehmen sie die parakrin angebotene T-Zell-Hilfe solange in Anspruch, bis sie völlige Autonomie erreicht haben. Durch das abnorm verlängerte Überleben der B-Zellen steigt dann die Wahrscheinlichkeit, weitere Aberrationen wie etwa die in 50 Prozent aller Fälle auftretende Chromosomentranslokation t(11;18)(q21;q21) zu erwerben.
