Durch Untersuchungen zur Reaktivität von Boryl- und Boridverbindungen konnten deren Bindungssituationen aufgeklärt und neuartige Koordinationsmotive von Übergangsmetall-Bor-Verbindungen erhalten werden. Die erhaltenen Verbindungen wurden mittels NMR-Spektroskopie, IR-Spektroskopie, Elementaranalyse und Röntgendiffraktometrie untersucht und zusätzlich wurden DFT-Rechnungen angefertigt.
An verschieden substituierten Eisenborylkomplexen wurden Reaktivitätsuntersuchungen gegenüber Halogenidabstraktionsmitteln und Reduktionsmitteln durchgeführt und im Falle der Boridkomplexe wurden Verbindungen mit bis dato unbekanntem Strukturmotiv erhalten.

Au Niger, les milieux dunaires sont les plus affectés par la dégradation des paysages aux conséquences parfois irréversibles. Cette étude tente de montrer que, malgré les modifications climatiques et les pressions anthropiques, une régénération des sols dunaires serait possible, dans cette dynamique complexe. Cela a été démontré à travers l’analyse micromorphologique des matériaux des parties superficielles des sols (0-20 cm). L’étude des caractéristiques particulières des croûtes (organisations pelliculaires de surface des sols) offre des pistes de recherches pouvant proposer des moyens et méthodes de fixation des dunes dégradées. Elle propose également des alternatives de lutte contre l’érosion éolienne et hydrique dans les écosystèmes sahéliens. Ceci cadre parfaitement avec la situation au Niger, où les phénomènes de désertification et d’ensablement des cuvettes interdunaires constituent une préoccupation majeure en matière de protection de l’environnement.

Der Artikel fragt zunächst nach Konzepten, die in klassischer Zeit von Außenpolitik existierten. Überlegungen zu diesem Thema sind in den antiken philosophischen Schriften greifbar, wobei Vorstellungen von der Funktionsweise des Oikos und der Polis auf den griechischen Raum übertragen wurden. Ähnliche Kategorien, um polisübergreifende Beziehungen zu beschreiben, griff die politische Rhetorik auf. Bezogen sich antike Autoren jedoch in allgemeinen Abhandlungen auf die Außenpolitik, wurde der Krieg als bestimmendes Element identifiziert. Das theoretische Konzept von Außenpolitik konnte demnach für die Zeitgenossen in der Praxis wenig Wirkung entfalten. Ursachen hierfür waren sowohl die unsteten Verhältnisse des 4. Jahrhunderts als auch die konkrete Konzeption der Außenpolitik, da die Interaktion im griechischen Raum anders funktionierte als in kleineren sozialen Einheiten. Zudem wurden real existierende Mechanismen zur Befriedung von Hellas nur unzureichend in die Konzeption integriert.
Much research on first language (L1) acquisition carried out in the last decades has proven that language acquisition is based on a biological endowment, the language faculty, which is triggered by the exposure to linguistic data. The language acquisition process undergoes similar stages in the same time span, independently of the specific language. Non-native acquisition differs from L1 acquisition, as the speaker already has an internal grammar with all parameters set. Transfer should therefore take place, bringing the learner to analyse the new input according to the properties of the L1, but a reanalysis is possible because of the availability of UG (Schwartz/Sprouse 1996). This article explores a syntactic domain, namely the properties of the functional categories constraining the verb position in main and subordinate clauses, by means of empirical data from Italian L1-speakers acquiring German as a second language (L2). It will be shown that the interlanguage grammars reflect properties of L1 and that resetting can be achieved, although optionality still exists and full convergence to the target language cannot be guaranteed.
In der nuklearmedizinischen Therapie werden Radiopharmaka meist systemisch verabreicht. Primär werden dafür, wegen der kurzen Reichweite, beta-Strahler eingesetzt. Als Folge davon verteilt sich das Radiopharmakon im Körper, reichert sich in Organen und Zielstrukturen an und bestrahlt somit den Körper intern, im Gegensatz zur externen Bestrahlung bei der Strahlentherapie.
Das Verteilungsmuster der verabreichten Aktivität im Körper wird durch die chemischen und physikalischen Eigenschaften des Radiopharmakons bestimmt. Außerdem sind die Aktivität und die Art der Anreicherung ausschlaggebend für die durch ionisierende Strahlung deponierte Energie im Körper, der Energiedosis.
Gemeinsam haben externe und interne Bestrahlungsverfahren, dass der Patient ionisierender Strahlung ausgesetzt ist, die nicht nur die kranken Zellen zerstört, sondern auch gesunde Zellen schädigen kann. Dies geschieht durch direkte oder indirekte Wechselwirkung der Strahlung mit der DNA, die zur Schädigung der DNA-Struktur führt. Am häufigsten sind dabei Einzelstrangbrüche und Basenschäden. Die Doppelstrangbrüche sind im Vergleich zu Einzelstrangbrüchen und Basenschäden sehr selten aber sehr viel schädlicher für die Zelle, da die Reparatur komplizierter ist. Somit sind diese primär für den Zelltod oder für die Folgen nach fehlerhafter Reparatur verantwortlich.
Eine sehr schnelle Antwort auf strahleninduzierte oder durch andere Stoffe, wie z.B. zytotoxische Substanzen, induzierte Doppelstrangbrüche ist die Phosphorylierung der Histon H2 Variante H2AX, die gamma-H2AX genannt wird. Zusätzlich reichert sich das Protein 53BP1 nach dem Erkennen eines Doppelstrangbruches durch Sensorproteine sofort am Chromatin, das den Doppelstrang umgibt, an. Damit ist 53BP1 ein weiterer Biomarker, der strahleninduzierte Doppelstrangbrüche sehr effektiv nachweisen kann und der auf sehr verlässliche Weise mit gamma-H2AX kolokalisiert. Mittels Immunfluoreszenzfärbung lassen sich gamma-H2AX und 53BP1 als umschriebene „Foci“, im Zellkern mikroskopisch darstellen und zählen. Unter der Annahme, dass ein Focus einem Doppelstrangbruch entspricht, kann die Anzahl der Foci im Zellkern als quantitativer Biomarker für DNA Doppelstrangbrüche und damit für die Strahlenexposition und Strahlenwirkung verwendet werden.
Zudem zeigen Studien der Induktion von gamma-H2AX nach externer Bestrahlung von unterschiedlichen Gewebearten Linearität zwischen der Energiedosis und der Zahl der Foci im Zellkern. Weitere Studien beschäftigen sich mit den Auswirkungen externer Bestrahlung auf Patienten, aber nur wenige mit offenen radioaktiven Substanzen. Ziele dieser Arbeit waren daher:
1. Die Generierung einer bisher noch nicht beschriebenen in-vitro Kalibrierkurve nach interner Bestrahlung von Vollblut mit den in der Therapie eingesetzten beta-Strahlern.
2. Die gleichzeitige Bestimmung der physikalischen Dosis sowie der strahleninduzierten Anzahl der Foci in Lymphozyten, gewonnen aus Blutproben von Patienten nach Radiopeptidtherapie mit Lu-177 und Radioiodtherapie mit I-131.
3. Eine umfassende Beschreibung der Induktion und der Abnahme der Foci in den Lymphozyten aus den Blutproben der Patienten unter Einbeziehung der in-vitro Kalibrierung, um den dosis- und zeitabhängigen Verlauf der Anzahl der strahleninduzierten Foci zu bestimmen.
Für die in-vitro Kalibrierung mit I-131 und Lu-177 wurden bei Probanden Blutproben gewonnen und mit unterschiedlichen Aktivitätskonzentrationen ergänzt. Das Ziel war, eine Energiedosis bis 100mGy zu erhalten. Das Ergebnis war, dass sich die Zahl der strahleninduzierten Foci in Abhängigkeit von der Energiedosis gut durch eine lineare Funktion beschreiben lässt, so wie es auch für die externe Bestrahlung bereits gezeigt wurde.
Die Patientenstudien befassten sich mit dem Zusammenhang zwischen der im Blut deponierten Energiedosis und der Anzahl und dem zeitlichen Verlauf der induzierten Doppelstrangbrüche im peripheren Blut von Patienten unter Peptidrezeptor-Radionuklidtherapie mit Lu-177 DOTATATE/-TOC und Patienten unter Radioiodtherapie mit I-131 bei Ablationstherapien nach Operation eines differenzierten Schilddrüsenkarzinoms.
Die durchschnittliche Anzahl induzierter DSB-Foci zeigte in den frühen Zeitpunkten einen linearen dosisabhängigen Anstieg. In den ersten Stunden nach Therapie stimmten die in-vitro Kalibrierung und die Zahl der strahleninduzierten Foci sowohl für Lu-177 als auch für I-131 für die Patientendaten gut überein.
Die späteren Zeitpunkte werden durch eine Abnahme der Dosisrate und der Foci-Anzahl, bedingt durch Reparatur der DNA-Schäden, charakterisiert. Überstiegen die Blutdosiswerte in der ersten Stunde jedoch 20mGy (nur nach I-131-Gabe beobachtet), dann war die Induktion eines schnellen Reparaturprozesses festzustellen.
Diese experimentellen Ergebnissen und Modellierungen beschreiben erstmalig die Dosisabhängigkeit und den zeitlichen Verlauf der in-vitro und in-vivo DNA-Schadensantwort nach Inkorporation von beta-emittierenden Radionukliden.
Einwandige Kohlenstoffnanoröhren weisen aufgrund ihrer besonderen Struktur viele für ein rein kohlenstoffhaltiges Makromolekül ungewöhnliche Eigenschaften auf. Dies macht sie sowohl für die Erforschung grundlegender Phänomene in eindimensionalen Nanostrukturen als auch für potenzielle Anwendungen äußerst interessant. Da alle Atome einer SWNT Oberflächenatome sind, führt dies zu einer besonders ausgeprägten Empfindlichkeit ihrer elektronischen Eigenschaften auf Wechselwirkungen mit der Umgebung. Lokale zeitabhängige Änderungen in diesen Wechselwirkungen führen daher zu Phänomenen wie dem Photolumineszenz-Blinken und spektraler Diffusion. Die Erforschung und Kontrolle der Parameter, die für die Beeinflussung der elektronischen Eigenschaften von SWNTs durch Umgebungseinflüsse entscheidend sind, wird neben der spezifischen Synthese eine maßgebliche Rolle dabei spielen, ob und in welcher Form SWNTs in optoelektronischen Bauteilen zukünftig Anwendung finden. Die vorliegende Arbeit liefert einen Beitrag zum Verständnis dieser Wechselwirkungen, indem die Dynamik von Energietransferprozessen innerhalb von SWNTs und zwischen SWNTs untersucht wurde.
Im Rahmen dieser Arbeit wurden homogene und inhomogene Beiträge zur Linienverbreiterung von in einer Matrix eingebetteten SWNTs bestimmt. Dabei wurde erstmals beobachtet, dass die spektrale Diffusion sowohl bei Raumtemperatur als auch bei 17 K auf einer ultraschnellen Zeitskala, d. h. innerhalb von weniger als 1 ps abläuft. Mittels transienter Lochbrennspektroskopie konnte gezeigt werden, dass die homogene Linienbreite von (6,5)-SWNTs mit 3.6 meV nur den geringsten Beitrag zur Absorptionslinienbreite liefert, während die größte Verbreiterung mit mehr als 99 % inhomogen ist. Die inhomogene Linienbreite wurde aus inkohärenten 2D-Spektren, welche durch spektrale Lochbrennexperimente bei Variation der Anregungswellenlänge erhalten werden konnten, zu \(54\pm5\)meV bestimmt. Die Dynamik der spektralen Diffusion wird mit einer Exzitonendiffusion in einer durch lokale Umgebungswechselwirkungen verursachten inhomogenen Energielandschaft entlang der Nanorohrachse erklärt. Durch zeitaufgelöste Lochbrennexperimente unter nichtresonanter Anregung konnte gezeigt werden, dass die Populationsumverteilung innerhalb dieser Energielandschaft für eine energetisch abwärts gerichtete Relaxation ein spontaner Prozess ist. Im umgekehrten Fall ist sie dagegen thermisch aktiviert. Mögliche Einflüsse von Artefakten wurden anhand von Referenzmessungen diskutiert und die Bestimmung der homogenen Linienbreite durch komplementäre CW-Lochbrennexperimente ergänzt.
Durch Monte-Carlo-Simulationen konnten erstmals Informationen über die Form der Potenzialenergielandschaft entlang einer SWNT erhalten und die Größenordnung der Plateaubreite mit nahezu konstanter Energie innerhalb der Potenziallandschaft zu 5.8-18.2nm ermittelt werden. Dies gelang durch eine Kalibrierung der Simulationszeit anhand experimenteller transienter Absorptionsspektren. Im Rahmen dieses Modells wurde darüber hinaus die Zeit für einen Sprung zu einem benachbarten Gitterplatz der Energielandschaft zu 0.1 ps bestimmt.
Inter- und intraband-Relaxationsprozesse von SWNTs wurden mittels Photolumineszenzspektroskopie untersucht. Die Ergebnisse deuten auf eine temperaturunabhängige Effizienz der internen Konversion und die photostimulierte Generierung von Löschzentren hin. Anhand temperaturabhängiger PL-Messungen, die erstmals bei Anregung des \(S_1\)-Zustands durchgeführt wurden, konnte die Energiedifferenz zwischen dem hellen und dunklen Exziton für (6,5)-SWNTs im Rahmen des Modells eines Dreiniveausystems zu \(\delta = (3.7\pm0.1)\)meV bestimmt werden. Aus der guten Übereinstimmung des temperaturabhängigen Trends der PL-Intensität unter \(S_1\)-Anregung mit in früheren Studien erhaltenen Ergebnissen unter \(S_2\)-Anregung konnte geschlussfolgert werden, dass die Effizienz der internen Konversion nicht ausgeprägt temperaturabhängig ist. Für SWNT-Gelfilme wurde unter \(S_2\)-Anregung eine deutliche Abweichung zur \(S_1\)-Anregung in Form eines Bleichens der Photolumineszenz beobachtet. Dieses Phänomen ist in der Literatur wenig diskutiert und wurde daher in leistungsabhängigen PL-Experimenten weiter untersucht. Dabei wurde für die \(S_2\)- im Vergleich zur \(S_1\)-Anregung eine stärker ausgeprägte sublineare Leistungsabhängigkeit gefunden. Die Abweichung vom linearen Zusammenhang der PL-Intensität mit der Leistung trat hier schon bei um eine Größenordnung geringeren Leistungsdichten auf als in früheren Studien und kann mit einer Exziton-Exziton-Annihilation allein nicht erklärt werden. Möglicherweise ist die Öffnung zusätzlicher Zerfallskanäle durch metastabile Löschzentren für dieses Verhalten verantwortlich. Die PL-Experimente zeigten zudem ein zeitabhängiges irreversibles Bleichen unter \(S_2\)-Anregung, welches bei 30 K stärker ausgeprägt war als bei Raumtemperatur. Dessen Abhängigkeit von der eingestrahlten Photonenzahl lässt auf eine Akkumulation von Löschzentren schließen. Daher wird eine mögliche Redoxreaktion mit Wasser, ausgelöst durch die intrinsische p-Dotierung der SWNTs, als Quelle der Löschzentren diskutiert.
Das Verzweigungsverhältnis für die Relaxation nach \(S_2\)-Anregung von SWNTs wurde in Form der relativen Quantenausbeute bestimmt und eine nahezu quantitative interne Konversion des \(S_2\)-Exzitons gefunden. Dieses Ergebnis hat eine wichtige Bedeutung für potenzielle Anwendungen von SWNTs in der Photovoltaik, da die Verluste durch die interband-Relaxation bei einer Anregung des zweiten Subband-Exzitons <3% zu sein scheinen. Die Herausforderung des Experiments wird hier durch die geringe Stokes-Verschiebung von SWNTs verursacht, die eine quantitative Trennung von PL- und Streulicht unmöglich macht. Daher wurde ein Aufbau realisiert, in dem ein großer Teil des Streulichts bereits räumlich entfernt wird und die PL unter \(S_1\)- bzw. \(S_2\)-Anregung quantifizierbar und ohne eine Annahme über Streulicht-Anteile direkt vergleichbar ist. Sowohl für SDS- als auch für Polymer-stabilisierte SWNTs wurde eine relative Quantenausbeute von \(\xi \approxeq 1\) erhalten, was eine nahezu quantitative interne Konversion von \(S_2\)- zu \(S_1\)-Exzitonen innerhalb der PL-Lebensdauer nahelegt.
Anregungsenergietransferprozesse zwischen Kohlenstoffnanoröhren in mono- und bidispersen SWNT-Netzwerkfilmen definierter Zusammensetzung wurden mittels zeitaufgelöster Polarisationsanisotropie untersucht. Dabei wurden neben einem ultraschnellen Energietransfer in weniger als 1 ps auch Hinweise auf Beiträge des \(S_2\)-Exzitons an diesem Prozess gefunden. Die Ergebnisse der Experimente mit bidispersen SWNT-Netzwerkfilmen bestätigen den auch in PLE-Spektren beobachteten energetisch abwärts gerichteten Energietransfer von SWNTs mit großer zu solchen mit kleiner Bandlücke und liefern darüber hinaus eine Zeitskala von weniger als 1 ps für diesen Prozess. Die umgekehrte Transferrichtung konnte weder aus dem \(S_1\)- noch aus dem \(S_2\)-Exziton beobachtet werden. Eine Beschleunigung der Anisotropiedynamik bei \(S_2\)- im Vergleich zu S\uu1-Anregung deutet auf einen Beitrag des \(S_2\)-Exzitons am Energietransferprozess in Konkurrenz zur internen Konversion hin. Durch Referenzexperimente mit monodispersen Netzwerkfilmen konnte eine Beteiligung von Energietransferprozessen zwischen SWNTs der gleichen Chiralität auf einer Zeitskala von 1-2ps nachgewiesen werden. Dadurch konnten Beobachtungen von zeitabhängigen Anisotropieänderungen, die einen energetisch aufwärts gerichteten Energietransfer suggerieren, mit einem intra-Spezies-Transfer erklärt werden - Hinweise auf energetisch aufwärts gerichtete EET-Prozesse wurden nicht gefunden. Eine wichtige Erkenntnis aus diesen Experimenten ist die Tatsache, dass die Überlappung von Signalbeiträgen zu einer Verfälschung der Anisotropie und damit zu fehlerhaften Interpretationen führen kann. Darüber hinaus wurde auf den Einfluss der Probenheterogenität und der Alterung von SWNT-Netzwerkfilmen hingewiesen. Diese Untersuchungen legen nahe, dass ein effizienter Exzitonentransfer in SWNT-Netzwerkfilmen auch zwischen den einzelnen Röhrensträngen erfolgen kann und es somit möglich ist, die Effizienz entsprechender Solarzellen zu verbessern.
Im letzten Teil der Arbeit wurden erstmals transiente Absorptionsexperimente im Femtosekundenbereich mit SWNTs unter \(Gate-Doping\) durchgeführt. In ersten Experimenten konnte gezeigt werden, dass analog zur chemischen Dotierung von SWNTs die Dynamik des \(S_1\)-Bleichens eines (6,5)-SWNT-Netzwerkfilms nach \(S_2\)-Anregung unter \(Gate-Doping\) eine Beschleunigung durch zusätzliche Zerfallskanäle erfährt. Die elektrochemische Bandlücke wurde für (6,5)-Nanoröhren zu 1.5 eV bestimmt. Eine Verringerung der Photoabsorptionsamplitude mit zunehmendem Potenzial lässt Vermutungen über die Natur dieses in transienten Absorptionsexperimenten beobachteten PA-Merkmals in Form der Absorption einer dotierten SWNT-Spezies zu. Diese Untersuchungen liefern erste Einblicke in die Art und Weise, wie eine elektrochemische Modifizierung von SWNTs die elektronische Bandstruktur und Ladungsträgerdynamik verändert.
Die klinische Symptomatik verschiedener erblicher Muskelerkrankungen verläuft oft erstaunlich ähnlich mit Muskelschwäche und -schwund als den hervorstechenden Alltagsproblemen. Dem gegenüber sind die genetischen Grundlagen sehr vielfältig mit > 250 bisher identifizierten Genen (musclegenetable.org). Auch innerhalb eines definierten Krankheitsbildes werden verschiedene genetische Ursachen nebeneinander gefunden, was durch die Verknüpfung in einem gemeinsamen Pathomechanismus begründet sein kann. Die vorliegende Arbeit beschäftigt sich mit verschiedenen Aspekten dieser genetischen Heterogenität am Beispiel der beiden häufigen Muskelerkrankungen Myotone Dystrophie (DM) und Facioscapulohumerale Muskeldystrophie (FSHD), bei denen alternative genetische Ursachen, sowie anknüpfende Fragestellungen untersucht wurden.
Das erste Projekt dieser Arbeit beschäftigt sich mit Fragestellungen, welche die DM betreffen. Die DM Typ 1 und Typ 2 (DM1 und DM2) bilden zusammen die häufigste Muskelerkrankung im Erwachsenenalter. Sie ist durch die gemeinsamen Symptome Myotonie, Muskelschwäche und Katarakt sowie die Beteiligung weiterer Organsysteme gekennzeichnet, was sie zu einer multisystemischen Erkrankung macht. Die genetische Ursache liegt für beide Formen in einer Repeatexpansion eines Mikrosatelliten in der untranslatierten Region zweier Gene (DMPK in DM1, CNBP in DM2). Dem gemeinsamen Pathomechanismus liegt eine toxische Funktionsgewinn-Mutation des expandierten RNA-Transkripts zugrunde.
Die beiden bekannten Formen der DM sind phänotypisch häufig nicht unterscheidbar, weshalb in vielen Fällen beide Erkrankungen molekulargenetisch untersucht werden müssen. Dabei ist die Diagnostik der DM durch die Notwendigkeit des Nachweises von sehr großen Repeatexpansionen recht aufwändig und die Bestimmung der Repeatlänge im Fall der DM2 nur eingeschränkt möglich. Im Rahmen dieser Arbeit wurde ein Test zum Nachweis der Repeatexpansionen auf der Basis der Methode des Molecular Combing entwickelt, welche den gleichzeitigen Nachweis der beiden Loci von DM1 und DM2 erlaubt und zusätzlich eine direkte Messung der Repeatlänge ermöglicht. Das Molecular Combing ist eine fluoreszenz-mikroskopische Einzelmolekül-Analysemethode, durch die es erstmals möglich wurde, die vermutete somatische Instabilität bei DM2 darzustellen.
Das zweite DM-Teilprojekt beschäftigt sich mit der Identifikation möglicher alternativer genetischer Ursachen für die Erkrankung. Dies wurde anhand einer Kohorte von 138 DM1- und DM2-negativen Indexpatienten mit dem typischen DM-Phänotyp untersucht. Ausgehend von dem gemeinsamen Pathomechanismus wurden die primären Krankheitsgene DMPK und CNBP, sowie CELF1 und MBNL1, welche wichtige Rollen auf sekundärer Ebene des Pathomechanismus spielen, mittels Next Generation Sequencing untersucht. Dabei wurde eine auffällige Variante in DMPK gefunden, keine Varianten in CNBP oder CELF1 und drei Varianten in MBNL1, was auf MBNL1 als Kandidatengen einer alternativen Ursache für DM hinweist. MBNL1 ist ein gewebespezifischer Spleißregulator, welcher einen Wechsel von einem fetalen zu einem adulten Spleißmuster im Muskel steuert. Die Pathogenität einer der Varianten wurde in einem RNA-Spleißassay mit MBNL1-Targetgenen untersucht. Dabei konnten keine spezifischen Spleiß-Effekte festgestellt werden, aber eine Verminderung des Expressionsniveaus im Sinne einer Haploinsuffizienz. Die 3D-Modellierung dieser Variante deutet auf Änderungen der Oberflächenladungen in MBNL1 hin. Der Nachweis der Pathogenität der Varianten und somit die Ursächlichkeit von MBNL1-Mutationen für DM konnte hiermit nicht abschließend geklärt werden. Die gefundenen Ergebnisse regen jedoch hoffentlich zu nachfolgenden Studien an.
Das zweite Projekt dieser Arbeit beschäftigt sich mit Fragestellungen um die FSHD. Diese bildet die dritthäufigste Muskelerkrankung, charakterisiert durch eine oft asymmetrische Schwäche der Muskulatur von Gesicht, Schultergürtel und Oberarmen. Genetisch ist die FSHD Typ 1 (FSHD1) mit einer Kontraktion des Makrosatelliten D4Z4 verknüpft, was eine Relaxation der Chromatinstruktur der Region mit sich bringt und damit die ektopische Expression des apoptotisch wirkenden Proteins DUX4 ermöglicht. Die pathogene Ausprägung dieser Funktionsgewinn-Mutation findet dabei nur in Verbindung mit einem FSHD-permissiven Haplotyp statt.
Auf der Grundlage des gleichen Pathomechanismus wurde eine zweite Form der FSHD (FSHD2) vorgestellt, bei der die Chromatinrelaxation unabhängig von der Länge von D4Z4 durch einen Defekt in dem an der DNA-Methylierung beteiligten Gen SMCHD1 assoziiert sein soll. Die Vererbung von FSHD2 verläuft digenisch mit Mutationen in SMCHD1 und dem FSHD-permissiven Haplotyp auf zwei unabhängigen Loci. Im Rahmen dieser Arbeit wurde eine Kohorte von 55 FSHD1-negativen Patienten mit dem typischen FSHD-Phänotyp untersucht. Dabei wurden der Haplotyp, die Methylierung von D4Z4 sowie das SMCHD1-Gen analysiert. Es konnten neun Patienten mit einem Defekt in SMCHD1 identifiziert werden. In einer zweiten Kohorte von 45 FSHD1-positiven Patienten wurde untersucht, ob SMCHD1-Mutationen auch in Kombination mit einer Kontraktion von D4Z4 vorkommen. Dieser Fall von FSHD1+2 konnte für drei Patienten gezeigt werden, welche außerdem einen auffällig schweren Phänotyp zeigten. SMCHD1 kann also als Modifier-Gen für die Schwere der Erkrankung bei FSHD1 angesehen werden. Damit wurden insgesamt zwölf SMCHD1-Mutationsträger identifiziert, davon sind zehn der Varianten noch nicht beschrieben worden. Für alle erkrankten Mutationsträger konnte eine Methylierung von D4Z4 ≤ 20 % ermittelt werden, was als diagnostisches Kriterium verwendet werden kann. Mit einem Anteil von 16,3 % Mutationsträger in der FSHD1-negetiven Kohorte bildet FSHD2 einen bedeutenden Anteil an dem Krankheitsbild der FSHD, weshalb die entwickelten Analysen in die Routinediagnostik eingegliedert wurden.
Das zweite Teilprojekt der FSHD beschäftigt sich mit der Funktion des SMCHD1-Gens bei der X-Inaktivierung (XI). Es ist bekannt, dass SMCHD1 bei weiblichen Mäusen an der Aufrechterhaltung der XI mitwirkt. Die Untersuchung der XI bei FSHD2-Frauen ergab eine extreme Verschiebung der erwarteten XI von 50:50 auf 0:100 oder 100:0 bei sechs von 13 Patientinnen. Die übrigen sieben zeigten eine XI im Normalbereich von > 20:80 oder < 80:20. Der Befund der einseitigen Verschiebung könnte auf einen negativen Selektionsdruck gegenüber Zellen mit unvollständiger XI hindeuten. Es wäre interessant zu untersuchen, ob sich der gleiche Effekt auch in einer größeren Kohorte wiederfindet und ob er sich mit der Art der Mutation korrelieren lässt.

In the past, there have been contradictory statements about the acquisition of the Bremer Papyri, particularly in respect of their relationship to the “Deutsches Papyruskartell”. The article reconstructs in detail the history of the Bremen collection and the beginning of the Gießen collection by publishing the relevant documents held by Deutsches Archäologisches Insitut in Cairo and the state and university library Bremen.
