Prostaglandin E2 (PGE2) increased the blood pressure, heart rate and body temperature, when administered at the doses ofO.OOI-IO,ug into the lateral cerebral ventricle (i.c.v.) of the urethane-anesthetised rat. The highest dose of 10 ,ug/rat induced a strong initial hypotensive effect. lntravenously (i.v.), PGE2 at the doses of 0.01-10 ,ug/rat caused a biphasic blood pressure response with dose-related initial decreases followed by slight increases in blood pressure. The heart rate and body temperature were slightly increased by i.v. administrations of PGE2 . The highest i.v. dose of 10 ,ug/rat initially decreased also the heart rate. Central pretreatment with indomethacin ( I mg/rat i.c.v.) partly antagonised all of the recorded central effects of PGE2 , while sodium meclofenamate (I mg/rat i.c. v.) abolished the hypertensive response to i.c. v. administered PGE2 but failed to significantly affect the PGE2-induced rises of heart rate and body temperature. The results support the previous suggestions that PGE2 may participate in the central cardiovascular and thermoregulatory contro!. The results also suggest that indomethacin and sodium meclofenamate antagonize the effects of exogenous prostaglandins. Since sodium meclofenamate, unlike indomethacin, affected preferentially the hypertensive response to centrally administered PGE2 , there may be differences in the sites and/or modes of action between these drugs.
Unique functions of DNA topoisomerase IIalpha and IIbeta have been suggested. A human cell line which carries a homozygeous mutation of the nuclear localization sequence of the topoisomerase IIalpha gene expresses the isoform outside the nucleus at the onset of mitosis. At mitosis topoisomerase IIbeta diffused away from the chromatin despite the nuclear lack of the IIalpha-form. Chromosome condensation and disjunction was performed with the aid of cytosolic topoisomerase IIalpha which bound to the mitotic chromatin with low affinity. Consequently an increased rate of nondisjunction is observed in these cells. It is concluded that high affinity chromatin binding of topoisomerase IIalpha is essential for chromosome condensation/disjunction and that topoisomerase IIbeta does not adopt these functions. A centrosomal protein was recognized by topoisomerase IIalpha. This topoisomerase IIalpha-like protein resembles a modified form of topoisomerase IIalpha with an apparent size of 205 kDa compared to 170 kDa. The expression of the protein is constant in all stages of the cell cycle and it appears in proliferating as well as in resting cells. If there is not sufficient topoisomerase IIalpha present at mitosis the centrosomal proteins might adopt the function and a mitotic catastrophe in the cells could therefore be prevented.
In der vorgestellten Arbeit wurden das Wachstum und das physiologische Verhalten von Zea mays auf Müllheizkraftwerk (MHKW) -Schlacke im Vergleich zu Gartenerde als Kulturmedium untersucht. Dabei stand das Wurzelsystem der Maispflanzen im Mittelpunkt des Interesses. Da feste Bodensubstrate verwendet wurden, mußten diese zu Beginn der Experimente chemisch, physikalisch und bodenbiologisch charakterisiert werden. Die Analyse der Schlacke zeigte, daß Schlacke ein multifaktorielles Streßsystem darstellt: Sie enthält einen hohen Gehalt an leicht löslichen Salzen, v.a. NaCl (bis zu 220 mM in der Bodenlösung). MHKW-Schlacke ist dagegen arm an Stickstoff und pflanzenverfügbarem Phosphat. Der pH-Wert der Bodenlösung von Schlacke ist stark alkalisch (pH 8.4 - 9.0). Darüber hinaus besitzt Schlacke einen hohen Gehalt an potentiell toxischen Schwermetallen und weist im Vergleich zum Kontrollsubstrat Gartenerde eine verdichtete Bodenstruktur mit erhöhtem mechanischen Widerstand auf. Im Vergleich zu der Kontroll-Anzucht auf Gartenerde reagierten die auf Schlacke kultivierten Mais-Pflanzen mit vermindertem Wachstum: Sproß und Wurzel erreichten nur die Hälfte der Länge der Kontrollpflanzen. Ein Vergleich der Biomassen von Sproß und Wurzel zeigte, daß das Sproßwachstum der Schlacke-Pflanzen stärker eingeschränkt ist als das Wurzelwachstum, woraus ein vergrößertes Wurzel / Sproß-Verhältnis resultiert. Das Wachstum von jungen Mais-Pflanzen auf Schlacke ist jedoch nicht in dem Maß eingeschränkt, wie es aufgrund der hohen Salzbelastung zu erwarten wäre. In einem Vergleichsexperiment mit Mais-Pflanzen, die in einer Nährlösung mit Zusatz von 100 mM NaCl kultiviert wurden, war das Wachstum erheblich schlechter und in den Blättern akkumulierte weitaus mehr Natrium als in Schlacke-Pflanzen. Hier wird der positive Einfluß des hohen Calciumgehaltes der Schlacke deutlich. Die Beeinträchtigung des Wachstums von Mais bei Kultur auf Schlacke wird hauptsächlich auf Phosphatmangel zurückgeführt, da durch Düngung eine beträchtliche Wachstumsverbesserung erzielt werden kann. Zudem wurden keine toxischen Konzentrationen an Schwermetallen im Blattgewebe von auf Schlacke kultivierten Pflanzen gefunden. Der Photosynthese-Apparat der Schlacke-kultivierten Pflanzen war sehr leistungsfähig: Es bestand keine Beeinträchtigung in der Energieverfügbarkeit (Quantenausbeute des Photosystems II) und die Lichtsättigung der photo-synthetischen Elektronentransportrate lag sogar höher als bei den Kontrollpflanzen. Die Bestimmung des „adenylate energy charge“ bestätigte diesen Sachverhalt. Das Wurzelsystem von Zea mays auf Schlacke wies strukturelle Veränderungen auf. Neben der verkürzten Wurzellänge und dem vergrößerten Wurzeldurchmesser der Schlacke-Pflanzen ergaben mikroskopische Untersuchungen, daß die Wurzeln durch Kultur auf Schlacke mit einer mechanischen Verstärkung reagieren: Stärker ausgeprägte tangentiale Zellwandverdickungen der Endodermis im tertiären Zustand und Zellwandmodifikationen in den radialen Zellwänden der Rhizodermis (Phi-Verdickungen). Für monokotyle Arten, insbesondere für Mais, gibt es bisher keine Beschreibung von Phi-Verdickungen in der Literatur. Gaschromatographische und massenspektrometrische Untersuchungen belegen, daß sich die Zellwände von auf Erde und Schlacke kultivierten Maiswurzeln im Hinblick auf den Gesamtgehalt an Lignin (endodermale Zellwandisolate) und in der Ligninzusammensetzung (hypodermale Zellwandisolate) unterscheiden: In Schlacke-kultivierten Maiswurzeln wurde ein höherer Anteil an dem Lignin-Monomer p Hydroxyphenyl gefunden, was zu einem höher verdichteten Lignin führt (Streßlignin). Die endodermalen Zellwände von auf Schlacke-kulivierten Pflanzen hatten dagegen einen höheren Gesamtlignin-Gehalt als die entsprechenden Kontrollen, was ebenfalls eine mechanische Verstärkung der Wurzel bewirkt. In Bezug auf Suberin konnten keine Unterschiede zwischen den verschiedenen Anzuchten gefunden werden, weder in den hypodermalen noch in den endodermalen Zellwandisolaten. Die verschiedenen Streßfaktoren führen demnach nicht zu einer verstärkten Imprägnierung der Zellwände mit lipophilem Material. Die Zellwände von Mais spielen eine wichtige Rolle bei der Immobilisierung von Schwermetallen. Die Zellwandisolate von auf Erde und Schlacke kultivierten Mais-Pflanzen wiesen je nach Schwermetall-Element 43 - 100 % des Gesamtgehaltes auf. Die absoluten Gehalte in den Zellwandisolaten von auf Schlacke angezogenen Pflanzen waren dabei höher als die entsprechenden Werte der Kontrolle. Eine Anreicherung in den Zellwänden wurde hauptsächlich für die Schwermetalle Zink, Blei, Nickel und Chrom beobachtet. Als unspezifische Streßantwort reagierten Maispflanzen auf die Kultur in Schlacke mit einer erhöhten Peroxidaseaktivität in der interzellulären Waschflüssigkeit. Die Peroxidaseaktivität des Symplastens der Wurzel unterscheidet sich zwischen den beiden Anzuchten dagegen nicht. Die Konzentration des Phytohormons Abscisinsäure (ABA) war in Blättern von auf Schlacke kultivierten Pflanzen von Zea mays und Vicia faba im Vergleich zu den Kontrollpflanzen erhöht. Dieser Anstieg ist eine Folge der erhöhten Salzbelastung der Schlacke, da die ABA-Gehalte entsprechender Blätter von auf gewaschener Schlacke kultivierten Pflanzen annähernd den Kontrollwerten entsprachen. Bei der Verteilung von ABA zwischen der Wurzel und der Bodenlösung der umliegenden Rhizosphäre konnte das als Anionenfalle bekannte Prinzip bestätigt werden. Nach diesem Modell reichert sich ABA im alkalischten Kompartiment an (hier: Schlacke-Bodenlösung). In den Wurzeln konnte nur in der Maiskultur auf Schlacke ein erhöhter Gehalt gefunden werden, nicht dagegen in der Vicia faba-Kultur. Dieser Unterschied liegt daran, daß Mais im Gegensatz zu Vicia faba eine exodermale Spezies ist und die Exodermis für ABA eine Barriere darstellt, was den ABA-Efflux in die Rhizosphäre verhindert. Im Wurzelgewebe von auf Schlacke kultivierten Maispflanzen wurde ein im Vergleich zur Kontrolle 15-facher Gehalt an wasserlöslichen, nicht proteingebundenen Sulfhydrylgruppen nachgewiesen. Diese auf Schwermetallstreß zurückzuführende Reaktion impliziert, daß die in der Schlacke-Bodenlösung vorhandenen Schwermetalle nicht ausreichend im Apoplasten zurückgehalten werden und bis in den Symplasten vordringen können.
Das Ziel der vorliegenden Arbeit war es, einen Beitrag zu leisten zur Klärung der Beziehung zwischen Alexithymie und den subjektiven und physiologischen Reaktionen auf emotionale Situationen. Kern des Persönlichkeitsmerkmals ‘Alexithymie’ ist die Schwierigkeit, eigene Gefühle zu identifizieren und anderen mitzuteilen (Bagby & Taylor, 1999a). Ähnlich wie bei anderen Formen emotionaler Hemmung wurde auch bei Alexithymie eine erhöhte physiologische Reaktivität angenommen, die auch mit einem erhöhten Erkrankungsrisiko verbunden sein soll (Stress-Alexithymie Hypothese, Martin & Pihl, 1985). Demnach führt eine in Stresssituationen durch mangelnde Emotionsregulation erhöhte und verlängerte physiologische Aktivität bei alexithymen Personen zu körperlichen Erkrankungen. In der Entkopplungshypothese (Papciak, Feuerstein & Spiegel, 1985) geht man bei Alexithymie unspezifischer als bei der Stress-Alexithymie Hypothese von einer Dissoziation der physiologischen Aktivität und den subjektiven Angaben zu Gefühlen oder emotionaler Erregung aus. Zu diesen Hypothesen liegen jedoch nur wenige und zudem widersprüchliche empirische Befunde vor. Die zentrale Frage der vorliegenden Arbeit lautete daher, ob sich hoch und niedrig alexithyme Personen in ihren subjektiven und physiologischen Reaktionen auf emotionale und belastende Situationen unterscheiden. Dazu wurde je eine experimentelle Untersuchung mit gesunden Probanden (n=43) und mit Patienten einer psychosomatischen Klinik (n=82) durchgeführt. Alle Probanden wurden nach der 20-Item Toronto-Alexithymieskala (Bagby, Parker & Taylor, 1994) in hoch und niedrig alexithyme Personen eingeteilt. Nach der Induktion von Emotionen und Belastungen (durch Filmausschnitte, Hyperventilation und einen modifizierten Stroop-Test) wurden die Reaktionen der Versuchspersonen hinsichtlich ihrer Gefühle, Körperempfindungen und physiologischen Parameter erfasst. Wie erwartet berichteten hoch alexithyme Gesunde und besonders Patienten im Vergleich zu niedrig Alexithymen stärkere negative Emotionen (v.a. Angst) und in einigen Bereichen stärkere körperliche Empfindungen im tonischen Niveau (vor der Emotionsinduktion). Jedoch ergaben sich entgegen den Erwartungen keine Gruppenunterschiede in den physiologischen Variablen. Durch Darbietung von Filmausschnitten wurden die Zielemotionen Traurigkeit und Ärger in ausreichender Stärke induziert. Während der Filme zeigten hoch Alexithyme stärkere Angst als niedrig Alexithyme. Signifikante Unterschiede zwischen hoch und niedrig alexithymen Personen in den Zielemotionen der Filmausschnitte oder anderen Emotionen fanden sich jedoch nicht. Allerdings beurteilten in beiden Untersuchungen weniger hoch als niedrig alexithyme Personen die Zielemotion Traurigkeit als stärkste Emotion während der traurigkeitsinduzierenden Filme. Hoch alexithyme Gesunde und stärker noch Patienten berichteten stärkere körperliche Empfindungen sowie größere Schwierigkeiten, ihre Gefühle während der Filmausschnitte in Worte zu fassen. Signifikante Unterschiede in der physiologischen Reaktivität auf die Filmausschnitte waren jedoch nicht nachweisbar. Vergleichbare Ergebnisse wie bei der Emotionsinduktion zeigten sich ebenfalls bei körperlicher und kognitiver Belastung. Die Befunde der vorliegenden Untersuchungen gelten damit für emotionale Situationen sowie auch für körperliche und kognitive Belastungen. Weder die Vorhersagen der Stress-Alexithymie Hypothese noch die der Entkopplungshypothese konnten in den vorliegenden Untersuchungen bestätigt werden. Ingesamt sprechen die Befunde daher dafür, dass eine mögliche höhere Vulnerabilität alexithymer Personen für körperliche Krankheiten nicht auf eine verstärkte physiologische Reaktivität auf spezifische emotionale Situationen zurückzuführen ist. Die Ergebnisse weisen allerdings auf eine in der Entkopplungshypothese nicht postulierte Dissoziation zwischen der objektiv messbaren und der wahrgenommenen physiologischen Reaktivität bei hoch alexithymen Patienten hin. Die stärkere Fokussierung auf körperliche Empfindungen lässt einen verstärkten Bericht körperlicher Symptome sowie ein verstärktes Krankheitsverhalten dieser Patienten erwarten.
Mediators of Social Anxiety - External Social Threat-Cues vs. Self-Related Negative Cognitions

Based on a review of models and empirical findings a working model is proposed, suggesting that self-related negative cognitions and biased processing of external social threat-cues are mediators of social anxiety. Hypotheses derived from this model were tested in three experiments. The first experiment examined whether levels of trait social anxiousness predicted fearful responding to external social threat-cues (angry vs. neutral and happy facial expressions) during social evaluation. Higher trait social anxiousness predisposes to an inward focus on one’s fear reaction to social threat. Using this strategy was expected to enhance fearful responding to angry facial expressions. A strategy of identifying with angry faces was expected to counteract fearful responding, but was expected to fail more often with increasing levels of trait social anxiousness. To examine these hypotheses, affective modulation of the startle eye-blink was assessed in forty-four undergraduate students. This measure served as a probe into the activation of brain structures involved in the automatic evaluation of environmental threat-cues. Trait and state anxiety as well as explicit emotional responding to the stimuli were assessed with questionnaires and ratings. Processing angry faces potentiated startle amplitudes as expected. Low arousal induced by the stimuli was a probable reason, why startle potentiation to happy faces emerged instead of attenuation. Trait social anxiousness and the cognitive strategies did not influence these effects. Yet, increased trait social anxiousness predicted decreased startle latency, indicating motor hyper-responsivity, which is part of the clinical representation of social anxiety disorder (SAD). Processing facial expressions and identifying with them disrupted this association. Previous studies support that similar strategies may enhance treatment of SAD. Individuals with SAD were expected to respond with increased arousal to external social threat-cues. Therefore, the second experiment examined whether nine individuals with SAD showed attentional (prepulse inhibition, PPI) or affective startle modulation to angry as compared to neutral and happy facial expressions. Corrugator supercilii activity was assessed as a behavioral indicator for effects of facial expressions. The remaining setup resembled the first experiment. Facial expressions did not modulate the startle reflex, but corrugator supercilii activity was sensitive to facial valence. However, the effects were not related to trait social anxiousness. Apparently, angry facial expressions do not act as phobic stimuli for individuals with SAD. The third experiment examined whether focusing on self-related negative cognitions or biased processing of external social threat-cues mediates relationships between trait social anxiety and anxious responding in a socially challenging situation. Inducing self-related negative cognitions vs. relaxation was expected to reveal a functional dependency on the supposed mediation in a multivariate assessment of criteria of the working model. Within this design, the impact of external social threat-cues (facial expressions and emotional words) was compared to control stimuli and context effects, using the startle paradigm. The findings provide first evidence for full statistical mediation of the associations between trait social anxiety and self-reported anxiety as well as parasympathetic withdrawal by self-related negative cognitions, when thirty-six undergraduate students anticipated public speaking. Apprehensive arousal, as indicated by increased skin conductance levels and heart rate, was present in all participants. Observer ratings of behavior during public speaking matched the self-rated quality of the performance. None of these measures were correlated with trait social anxiousness. Startle amplitude correlated with state and trait social anxiety, but was no mediator. Finally, there was no affective modulation of the startle amplitude by external social threat-cues. These studies advance both our current understanding of the factors that mediate social anxiety responses to situations and our knowledge of the physiological and anatomical mechanisms involved in social anxiety. Based on these findings a revised version of the working model on mediators of social anxiety is proposed in the hope it may aid further research for the ultimate goal of developing an empirically validated functional anatomical model of social anxiety.
Das Ziel der vorliegenden Arbeit war die Untersuchung der Reaktionen von Migränepatientinnen mit episodischer (EM) und häufiger Migräne (HM) auf verschiedene Aspekte des Triggerfaktors „Negativer Affekt“ wie Stress und negative Emotionen. Die Ergebnisse der beiden Gruppen wurden mit denen gesunder Kontrollpersonen verglichen (KG). Zur Ermittlung des Aufmerksamkeitsverhaltens gegenüber emotionalen Reizen wurden zwei Emotionale Stroop Tests (EST) durchgeführt. Erwartet wurde ein Aufmerksamkeitsbias der Patientinnen hinsichtlich negativer emotionaler Reize. Im EST 1 wurden allgemeine affektive Wörter der Valenzen positiv, neutral und negativ verwendet. Die Probandinnen sollten auf die Wortfarbe mit Tastendruck reagieren und den Wortinhalt ignorieren. Im EST 2 wurden emotionale Gesichtsausdrücke (ärgerlich, freundlich, neutral) als Reize verwendet. Dabei sollte die Rahmenfarbe der Bilder per Tastendruck bestimmt werden und der Inhalt ignoriert werden. Zur Auswertung wurden Emotionale Stroop Interferenzen (ESI) zum Vergleich Reaktionszeitdifferenzen negativ-neutral und negativ-positiv berechnet. Der erwartete Aufmerksamkeitsbias der HM für negative emotionale Reize wurde dabei nicht gefunden. Dafür zeigten im EST 2 die KG einen Aufmerksamkeitsbias für ärgerliche Gesichter. Ein signifikanter Gruppenunterschied in EST 2 mit sehr niedrigen, im Vergleich negativ-positiv sogar negativen ESI der HM ließ auf ein Vermeidungsverhalten dieser Gruppe ärgerlichen Gesichtern gegenüber schließen. Dieses wurde als Vermeidung negativer sozialer Reize interpretiert und zum gelernten, möglicherweise dysfunktionalen Vermeidungsverhalten von Migränepatienten potentiellen Triggersituationen gegenüber in Bezug gesetzt. Weiterhin wurden die Probandinnen mit dem „Paradigma der Öffentlichen Rede“ psychosozialem Stress ausgesetzt, indem sie vor einer Videokamera unter Beobachtung eine Rede halten sowie eine Kopfrechenaufgabe lösen sollten. Vorher und nachher wurden insgesamt vier Speichelproben zur Bestimmung des Stresshormons Kortisol genommen. Zudem wurden die Druckschmerzschwellen vor und nach dem Experimentalteil gemessen. Die erwartete Kortisolreaktion als Antwort auf die psychosoziale Stressaufgabe blieb aus. Ursache dafür kann die Stichprobenzusammensetzung mit 98% Frauen sein, deren Kortisolreaktion auf Stress durch hormonelle Schwankungen im Experiment nur unzuverlässig stimulierbar ist. Bei der Berechnung der Gesamtkortisolausschüttung über die Zeit zeigte sich im Gegensatz zu dem erwarteten erhöhten Kortisolspiegel der Migränepatientinnen ein linearer Abfall des Spiegels von KG, über EM zu HM, mit den niedrigsten Werten der HM. Diese Ergebnisse könnten auf Veränderungen der Hypophysen-Nebennieren (HHN)-Achse im Sinne eines Hypokortisolismus bei Migränepatientinnen widerspiegeln, der weiterer Klärung bedarf, z.B. durch die Bestimmung eines Kortisoltagesprofils bei Patientinnen. Eine veränderte Funktion der HHN-Achse könnte außerdem zu einer inadäquaten Reaktion auf Stresssituationen beitragen. Die bei Patientinnen ausbleibende Veränderung der Druckschmerzschwelle in Reaktion auf Stress lässt ebenfalls auf eine ungenügende Stressreaktion der Patientinnen schließen. Am Ende der Untersuchung, nach einer Entspannungsphase von 50 Minuten, wurde den Probandinnen Blut abgenommen, in dem die mRNA- und Proteinkonzentrationen ausgewählter pro- und antiinflammatorischer Zytokine bestimmt wurden. Die Analyse der Zytokinkonzentrationen mit Luminex ergab für die Proteindaten aufgrund zu geringer verwertbarer Daten kein interpretierbares Bild. Die mittels Real Time Quantitativer PCR erhaltenen mRNA-Konzentrationen spiegelten die Schmerzfreiheit der Patienten wieder, mit im Vergleich zu KG verringerten proinflammatorischen Zytokinen (TNF-alpha, IL-1beta, IL-2, IL-6) und dem ebenfalls verringerten antiinflammatorischen Zytokin IL-10, sowie dem deutlich erhöhten antiinflammatorischen IL-4. Die im Vergleich zur KG überregulierten Zytokine im schmerzfreien Intervall weisen auf veränderte Regulierungsmechanismen des Immunsystems für die Schmerzmediatoren Zytokine hin. Weitere Schmerzmediatoren könnten ebenfalls verändert sein, was weiterer Klärung in nachfolgenden Studien bedarf. Alles in allem konnten verschiedene Veränderungen in den psychologischen und endokrinen Reaktionen der Migränepatientinnen auf Bestandteile des Triggers „Negativer Affekt“ sowie in der Schmerzregulierung gefunden werden, wobei die Veränderungen bei Patientinnen mit Häufiger Migräne stärker auftraten. Dies weist auf eine mögliche Rolle der einzelnen untersuchten Komponenten bei der Migränechronifizierung hin, was in weiteren Studien vertiefend untersucht werden sollte.
Recent studies show that combinations of defined key developmental transcription factors (TFs) can reprogram somatic cells to pluripotency or induce cell conversion of one somatic cell type to another. However, it is not clear if single genes can define a cells identity and if the cell fate defining potential of TFs is also operative in pluripotent stem cells in vitro. Here, we show that ectopic expression of the neural TF Neurogenin2 (Ngn2) is sufficient to induce rapid and efficient differentiation of embryonic stem cells (ESCs) into mature glutamatergic neurons. Ngn2-induced neuronal differentiation did not require any additional external or internal factors and occurred even under pluripotency-promoting conditions. Differentiated cells displayed neuron-specific morphology, protein expression, and functional features, most importantly the generation of action potentials and contacts with hippocampal neurons. Gene expression analyses revealed that Ngn2-induced in vitro differentiation partially resembled neurogenesis in vivo, as it included specific activation of Ngn2 target genes and interaction partners. These findings demonstrate that a single gene is sufficient to determine cell fate decisions of uncommitted stem cells thus giving insights into the role of key developmental genes during lineage commitment. Furthermore, we present a promising tool to improve directed differentiation strategies for applications in both stem cell research and regenerative medicine.
In their natural environment animals face complex and highly dynamic olfactory input. This requires fast and reliable processing of olfactory information, in vertebrates as well as invertebrates. Parallel processing has been shown to improve processing speed and power in other sensory systems like auditory or visual. In the olfactory system less is known about olfactory coding in general and parallel processing in particular. With its elaborated olfactory system and due to their specialized neuroanatomy, honeybees are well-suited model organism to study parallel olfactory processing. The honeybee possesses a unique neuronal architecture - a dual olfactory pathway. Two mirror-imaged output projection neuron (PN) pathways connect the first olfactory processing stage, the antennal lobe (analog to the vertebrates olfactory bulb, OB), with the second, the mushroom body (MB) known to be involved in orientation and learning and memory, and the lateral horn (LH). The medial antennal lobe-protocerebral tract (m-APT) first innervates the MB and thereafter the LH, while the other, the lateral-APT (l-APT) projects in opposite direction. The neuroanatomy and evolution of these pathways has been analyzed, yet little is known about its physiology. To analyze the function of the dual olfactory pathway a new established recording method was designed and is described in the first chapter of this thesis (multi-unit-recordings). This is now the first time where odor response from several PNs of both tracts is recorded simultaneously and with high temporal precision. In the second chapter the PN odor responses are analyzed. The major findings are: both tracts responded to all tested odors but with differing characteristics. Since recent studies describe the input to the two tracts being rather similar, the results now indicate differential odor processing along the tracts, therefore this is a good indicator for parallel processing. PNs of the m-APT process odors in a sparse manner with delayed response latencies, but with high odor-specificity. PNs of the l-APT in contrast respond to several odor stimuli and respond in general faster. In some PN originating from both tracts, characteristics of odor-identity coding via response latencies were found. Analyzing the over-all dynamic range of the PNs both l- and m-APT PNs were tested over a large odor concentration range (10-6 to 10-2) (3. chapter). The PNs responded with linear and non-linear correlation of the response strength to the odor concentration. In most cases the l-APT is comparatively more sensitive to low odor concentrations. Response latency decreases with increasing odor concentration in both tracts. Alternative coding principles and elaboration on the hypothesis whether the dual olfactory pathway may contribute coincidental innervation to the next higher-order neurons, the Kenyon cells (KC), is subject of the 4. chapter. Cross-correlations and synchronous responses of both tracts show that in principle odors may be coded via temporal coding. Results suggest that odor processing is enhanced if both tracts contribute to olfactory coding together. In another project the distribution of the inhibitory neurotransmitter GABA (gamma-aminobutyric acid) was measured in the bee’s MB during adult maturation (5. chapter). GABAergic inhibition is of high importance in odor coding. An almost threefold decrease in the total amount of GABAergic innervation was found during adult maturation in the l- and m-APT target region, in particular at the change in division of labor during the transition from a young nurse bee to an older forager bee. The results fit well into the current understanding of brain development in the honeybee and other social insects during adult maturation, which was described as presynaptic pruning and KC dendritic outgrowth. Combining anatomical and functional properties of the bee’s dual olfactory pathway suggests that both rate and temporal coding are implemented along two parallel streams. Comparison with recent work on analog output pathways of the vertebrate’s OB indicates that parallel processing of olfactory information may be a common principle across distant taxa.
Mammalian haloacid dehalogenase (HAD)-type phosphatases are a large and ubiquitous family of at least 40 human members. Many of them have important physiological functions, such as the regulation of intermediary metabolism and the modulation of enzyme activities, yet they are also linked to diseases such as cardiovascular or metabolic disorders and cancer.
Still, most of the mammalian HAD phosphatases remain functionally uncharacterized.
This thesis reveals novel cell biological and physiological functions of the phosphoglycolate phosphatase PGP, also referred to as AUM. To this end, PGP was functionally characterized by performing analyses using purified recombinant proteins to investigate potential protein substrates of PGP, cell biological studies using the spermatogonial cell line GC1, primary mouse lung endothelial cells and lymphocytes, and a range of biochemical techniques to characterize Pgp-deficient mouse embryos.
To characterize the cell biological functions of PGP, its role downstream of RTK- and integrin signaling in the regulation of cell migration was investigated. It was shown that PGP inactivation elevates integrin- and RTK-induced circular dorsal ruffle (CDR) formation, cell spreading and cell migration. Furthermore, PGP was identified as a negative regulator of directed lymphocyte migration upon integrin- and GPCR activation.
The underlying mechanisms were analyzed further. It was demonstrated that PGP regulates CDR formation and cell migration in a PLC- and PKC-dependent manner, and that Src family kinase activities are required for the observed cellular effects. Upon integrin- and RTK activation, phosphorylation levels of tyrosine residues 1068 and 1173 of the EGF receptor were elevated and PLCγ1 was hyper-activated in PGP-deficient cells. Additionally, PGP-inactivated lymphocytes displayed elevated PKC activity, and PKC-mediated cytoskeletal remodeling was accelerated upon loss of PGP activity. Untargeted lipidomic analyses revealed that the membrane lipid phosphatidylserine (PS) was highly upregulated in PGP-depleted cells.
These data are consistent with the hypothesis that the accumulation of PS in the plasma membrane leads to a pre-assembly of signaling molecules such as PLCγ1 or PKCs that couple the activation of integrins, EGF receptors and GPCRs to accelerated cytoskeletal remodeling.
Thus, this thesis shows that PGP can affect cell spreading and cell migration by acting as a PG-directed phosphatase.
To understand the physiological functions of PGP, conditionally PGP-inactivated mice were analyzed. Whole-body PGP inactivation led to an intrauterine growth defect with developmental delay after E8.5, resulting in a gradual deterioration and death of PgpDN/DN embryos between E9.5 and E11.5. However, embryonic lethality upon whole-body PGP inactivation was not caused by a primary defect of the (cardio-) vascular system. Rather, PGP inactivated embryos died during the intrauterine transition from hypoxic to normoxic conditions.
Therefore, the potential impact of oxygen on PGP-dependent cell proliferation was investigated. Analyses of mouse embryonic fibroblasts (MEFs) generated from E8.5 embryos and GC1 cells cultured under normoxic and hypoxic conditions revealed that normoxia (~20% O2) causes a proliferation defect in PGP-inactivated cells, which can be rescued under
hypoxic (~1% O2) conditions. Mechanistically, it was found that the activity of triosephosphate isomerase (TPI), an enzyme previously described to be inhibited by phosphoglycolate (PG) in vitro, was attenuated in PGP-inactivated cells and embryos. TPI constitutes a critical branch point between carbohydrate- and lipid metabolism because it catalyzes the isomerization of the glycolytic intermediates dihydroxyacetone phosphate (DHAP, a precursor of the glycerol backbone required for triglyceride biosynthesis) and glyceraldehyde 3’-phosphate (GADP).
Attenuation of TPI activity, likely explains the observed elevation of glycerol 3-phosphate levels and the increased TG biosynthesis (lipogenesis). Analyses of ATP levels and oxygen consumption rates (OCR) showed that mitochondrial respiration rates and ATP production were elevated in PGP-deficient cells in a lipolysis-dependent manner. However under hypoxic conditions (which corrected the impaired proliferation of PGP-inactivated cells), OCR and ATP production was indistinguishable between PGP-deficient and PGP-proficient cells. We therefore propose that the inhibition of TPI activity by PG accumulation due to loss of PGP activity shifts cellular bioenergetics from a pro-proliferative, glycolytic metabolism to a lipogenetic/lipolytic metabolism.
Taken together, PGP acts as a metabolic phosphatase involved in the regulation of cell migration, cell proliferation and cellular bioenergetics. This thesis constitutes the basis for further studies of the interfaces between these processes, and also suggests functions of PGP for glucose and lipid metabolism in the adult organism.
