Bis zum Jahr 2100 prognostiziert der Weltklimarat (IPCC 2021) einen Anstieg des Meeresspiegels von bis zu 63-101 cm gegenüber heutigen Wasserständen. Im Rahmen des Generalplans Küstenschutz Schleswig-Holstein(GKSH) soll als Klimafolgeanpassung eine Erhöhung und Profiländerung der meisten Nordseedeiche und Elbedeiche erfolgen (zusammen 363,3 km mit einer Vegetationsfläche von 3.500 ha). Diese Maßnahmen werden mit einem vollständigen Verlust der alten Deichvegetation einhergehen und zur Freisetzung von großen Mengen an CO₂ aus dem Bodenkohlenstoff führen. Die Seedeiche der Nordseeküste (262 km) zählen zu den artenreichen, semi-natürlichen und von Schafen beweideten Grasländern (Fläche von 2600 ha) in Schleswig-Holstein mit bis zu 18 Gras- und 64 zweikeim-blättrigen Blütenpflanzen und an die Vegetation gebundene 800-1000 Arten von Invertebraten (darunter 200 Käferarten). Auf die Außenböschung dringen Pflanzen der Salzwiesengesellschaften vor. Die steileren, wärmeexponierten (überwiegend nach Osten und Süden ausgerichtet) und durch Vertritt lückigen Innenböschungen der Seedeiche sind wertvolle Refugien wärmeliebender, konkurrenzschwacher Arten von Magerstandorten und Trittgesellschaften wie die folgenden mediterran-subatlantischen Arten: Knotenklettenkerbel (Torilis nodosa), Zwergklee/Armblütiger Klee (Trifolium micranthum) und Vogelfußklee (Trifolium ornithopodioides). Für die Erhaltung beider Kleearten (die aktuelle Verbreitung wird dokumentiert) besitzt Schleswig-Holstein eine nationale und nordwest-europäisch-kontinentale Verantwortlichkeit. Folgende Maßnahmen zum Schutz der reichhaltigen Deichvegetation und Teilen seiner Invertebratenfauna bei der Deichverstärkung im Rahmen des GKSH werden vorgeschlagen: 1. Abheben der Grasnarbe mit Wurzelraum und zeitnahe Wiederverlegung der alten Grasnarbe (Soden) auf das neue Deichprofil; das ist auch wichtig zum Erhalt des Bodenkohlenstoffs (Klimaschutz). 2. Einsaat von neuen Deichprofilen mit Saatgut von artenreichen Deichabschnitten. 3. Aufnahme substanzieller Forschungsprogramme/Forschungsförderung zur Ökologie der Seedeiche. Weiterhin sollte auf den Einsatz von Herbiziden auf Deichen zur Bekämpfung von Disteln verzichtet werden.
Die psychosoziale Komponente spielt bei der ADHS v.a. in der Bewältigung von Erziehungsaufgaben eine erhebliche Rolle, da sowohl die Eltern als auch das Kind von der Störung betroffen sein können.
Ziel der vorliegenden Studie war die Untersuchung von Beziehungen und Konfliktpotential in Familien, die von ADHS betroffen sind. Es wurde der Frage nachgegangen, ob eine störungsspezifische Therapie von Müttern mit ADHS und deren Kind, das ebenfalls an ADHS litt, bessere familienspezifische Messwerte erreicht als eine übliche Standardbehandlung. Die Behandlungsgruppe erhielt eine intensive Gruppenpsychotherapie und begleitende Pharmakotherapie mit Methylphenidat, die Kontrollgruppe wiederholte psychiatrische Beratungen, beide Gruppen erhielten zusätzlich ein Mutter-Kind-Training. Die Stichprobe bestand aus 144 Mutter-Kind Paaren mit ADHS, die im Rahmen einer Mutter-Kind Treatment Studie rekrutiert wurden.
Es zeigten sich Verbesserungen in den untersuchten familienbezogenen Outcomes (soziales Leben, negative Gefühle gegenüber der Erziehung), nicht aber in allen erfassten Bereichen. Diese Verbesserungen zeigten jedoch keine signifikanten Gruppenunterschiede im Hinblick auf die beiden Studienbehandlungen zur Therapie der ADHS der Mütter (Pharmakotherapie plus Verhaltenstherapie vs. alleinige unspezifische Beratung). Bei Müttern, die ein Krankheitsverständnis für die ADHS, sowie eine Behandlungsmotivation hatten, verbesserte das Mutter-Kind-Training die Outcomes der Kinder, auch wenn die Mutter nur eine unterstützende Beratung erhielt. Die multimodale Therapie der Mütter mit Gruppenpsychotherapie und MPH-Medikation war bezüglich der Symptomreduktion der Mütter effektiv. Jedoch beeinflusste die multimodale Therapie im Vergleich zur unterstützenden psychiatrischen Beratung das externalisierende Verhalten des Kindes nach dem Elterntraining nicht zusätzlich. Deshalb scheint es vielversprechend, Müttern mit ADHS, welche nicht die Möglichkeit einer Medikation oder spezifischen Psychotherapie haben, auch zukünftig Elterntraining anzubieten.
Untersuchungen zur Informationsweitergabe in Familien mit erblichem Brust- und Eierstockkrebs

Für die hier beschriebene Studie wurde ein Fragebogen erstellt, welcher von 80 Trägerinnen und Trägern einer pathogenen Mutation in den Genen BRCA1 oder BRCA2 ausgefüllt wurde. Die Befragung sollte untersuchen, ob den Befragten das Risiko ihrer Verwandten, ebenso Mutationsträger zu sein, bewusst war. Weiterhin sollte ermittelt werden, ob sie die jeweiligen Risikopersonen darüber informierten. Es zeigte sich, dass den meisten Befragten dieses Risiko bekannt war. Einigen Personen schienen jedoch nicht genau zu wissen, welche Verwandten als „Risikopersonen“ zählen. Insbesondere war nicht allen Befragten die Möglichkeit bewusst, dass auch Männer die Mutation tragen und an ihre Kinder weitergeben sowie selbst an Brustkrebs erkranken können. Weiterhin gaben mehr als ein Viertel der Befragten an, dass sie mindestens ein Familienmitglied, obwohl es ihnen als Risikoperson bekannt war, nicht informierten. Als häufigste Grund hierfür wurde mangelnder Kontakt genannt. Vor dem Hintergrund der Angaben der Befragten sowie der aktuellen Forschungslage werden in der vorliegenden Arbeit Möglichkeiten diskutiert, wie die Anzahl der informierten Angehörigen verbessert werden könnte.
Zum 01.06.2017 wurde in der Universitätsklinik Würzburg im Rahmen des 'Antimicrobial Stewardship' in der Herzchirurgie die perioperative Antibiotikaprophylaxe von Cefuroxim auf Cefazolin umgestellt. Diese Studie untersucht insgesamt 1029 Patienten vor und nach der Umstellung hinsichtlich ihrer Raten an Wundinfektionen, nosokomialen Infektionen und Risikofaktoren zur Entwicklung einer postoperativen Wundinfektion.
Die Ergebnisse zeigen, dass eine Umstellung der perioperativen Antibiotikaprophylaxe von Cefuroxim, einem Cephalosporin der zweiten Generation, auf Cefazolin, ein Cephalosporin der ersten Generation, zu keinem Anstieg der Wundinfektionen in herzchirurgischen Eingriffen führt.
Insgesamt lag keine signifikante Überlegenheit eines der beiden Antibiotika vor, weder in Hinblick auf die gesamten Wundinfektionen, die tiefen sternalen Wundinfektionen und die Infektionen der Beinwunde, noch bei nosokomialen Infektionen wie der Pneumonie, dem Harnwegsinfekt oder der Sepsis. Im Patientenkollektiv konnten weiterhin einige unabhängige prä-, intra- und postoperative Risikofaktoren ermittelt werden, die zu einer signifikanten Steigerung der Infektionsraten führten. Auch bei der differenzierten Betrachtung der Risikopatienten zeigte sich kein signifikanter Wirkungsunterschied zwischen den beiden untersuchten Antibiotika.
Diese Studie weist mit 1029 Patienten ein zur existierenden Literatur vergleichsweise großes Patientenkollektiv auf. Auch die erhobenen Parameter sind umfangreich gewählt und boten die Möglichkeit tiefergehender Analysen. Limitiert wird die Studie jedoch durch ihr retrospektives Design mit dem Fehlen eines kontrollierten Follow-Ups. Um die Ergebnisse bestätigen und bekräftigen zu können, müsste eine prospektive, randomisierte Studie hieran angeschlossen werden.
For the quality assurance of substances for pharmaceutical use, a variety of analytical techniques are available to address specific analytical problems. In this field of application, liquid chromatography (LC) stands out as the gold standard in the pharmaceutical industry. Various detectors can be employed, which are e.g. based on UV/Vis spectroscopy for the examination of molecules with a chromophore, or mass spectrometry (MS) for structural elucidation of analytes. For the separation of enantiomers, the use of capillary electrophoresis (CE) may be more favorable due to the high separation efficiency and easy-to-use and comparatively inexpensive chiral selectors, in contrast to chiral columns for LC, which are usually very expensive and limited to a restricted number of analytes. For structure elucidation in impurity profiling, one- and multidimensional 1H NMR spectroscopy is a valuable tool as long as the analyte molecule has got nuclei that can be detected, which applies for the magnitude of organic pharmaceutical substances.
For the evaluation of the amount of mineral oil aromatic hydrocarbons (MOAH) in various paraffin samples from different suppliers, a straightforward method based on 1H NMR spectroscopy was elaborated. The MOAH/MOSH ratio was used to indicate the amount of MOAH of paraffins and to evaluate the extent of refining. In addition, a representative paraffin sample was measured without sample solvent at high temperatures (about 340 K) to avoid the interfering residual solvent signals in the spectral regions of interest. The results of both methods were in good accordance.
Moreover, the 1H NMR results were complemented with the UV measurements from the purity testing of paraffins according to the DAB 8. Correlations of the NMR and UV spectroscopic data indicated a linear relationship of both methods for the determination of MOAH in paraffins.
Finally, the 1H NMR data was evaluated by principal component analysis (PCA) to explore differences within the paraffin samples and the spectral regions in the 1H NMR spectrum which are responsible for the formation of groups. It could be found that most variation is due to the MOSH of the paraffins. The PCA model was capable of differentiating between soft, liquid and solid paraffins on the one hand and between natural and synthetic liquid paraffins on the other hand.
The impurity profiling of L-ascorbic acid 2-phosphate magnesium (A2PMg) was performed by means of one- and two-dimensional NMR spectroscopy. Several ethylated impurities could be detected, which were likely to be formed during synthesis of A2PMg. The structures of two of the ethylated impurities were identified as ascorbic acid 2-phosphate ethyl ester and ethanol, (residual solvent from synthesis). NMR spectroscopic studies of the fractions obtained from preparative HPLC of A2PMg revealed two additional impurities, which were identified as phosphorylated derivatives of ascorbic acid, ascorbic acid 3,5-phosphate and ascorbic acid 5-phosphate.
Solid state mechanochemistry as an alternative approach for stress testing was applied on the drug substances S-Ibuprofen (Ibu) and Clopidogrel (CLP) using a ball mill, in order to study their degradation profile:
First, the isomerization of S-Ibu was investigated, which was stressed in the solid state applying several milling frequencies and durations under basic, acidic and neutral conditions. For the separation of Ibu enantiomers, a chiral CE method was developed and validated according to ICH Q2(R1). It was found that S-Ibu is overall very stable to isomerization; it shows minor conversion into the R-enantiomer under basic environment applying long milling times and high frequencies.
Last, the degradation profile of clopidogrel hydrogen sulfate (CLP) was investigated, which was stressed in the solid state under various oxidative conditions. An already existing HPLC-UV method was adjusted to sufficiently separate the degradation products, which were characterized by means of UV and MS/(MS) detection. Most of the degradation products identified were already reported to result from conventional CLP stress tests. The degradation profile of CLP was mainly influenced by the material of the milling jar and the type of catalyst used.
Virale Reaktivierungen treten im Rahmen der Immundefizienz und Immunsuppression nach allogener Stammzelltransplantation häufig auf und können zu schwerwiegenden Komplikationen führen. Ziel dieser retrospektiven Studie war die Charakterisierung von viralen Reaktivierungen im ersten Jahr nach allogener Stammzelltransplantation, die Identifikation von Risikofaktoren sowie die Untersuchung des Einflusses viraler Reaktivierungen auf das Transplantationsoutcome. 107 pädiatrische allogene Stammzelltransplantationen im Zeitrahmen von Januar 2005 bis Dezember 2015 wurden in diesem Zusammenhang auf Infektionen mit dem Epstein-Barr Virus (EBV), Cytomegalovirus (CMV), Humanen Herpesvirus 6 (HHV 6), Herpes simplex Virus (HSV), Varicella zoster Virus (VZV) und Adenovirus (ADV) untersucht.
Bouldern ist eine noch relativ junge Trendsportart aus der Familie des Klettersports, die in den letzten Jahren starken Zuwachs gewonnen hat. Es gibt bisher wenig Literatur zu Verletzungen durch Bouldern generell und insbesondere zu Indoor-Bouldern. Das Ziel dieser Studie war Daten zu Verletzungshäufigkeit, -schwere und -lokalisation durch Indoor-Bouldern zu erheben sowie den Einfluss von potenziell das Verletzungsrisiko modulierenden Faktoren zu untersuchen.
Mittels eines Online-Fragebogens wurden Boulderer retrospektiv zu anthropometrischen Daten, potenziell risikomodulierendem Verhalten sowie Verletzungen in der Vergangenheit befragt. Anschließend wurde monatlich prospektiv über ein Jahr das Auftreten neuer Verletzungen erhoben. Zusätzlich wurden Patienten der Notaufnahme und Klettersprechstunde mit Verletzungen durch Indoor-Bouldern zu Diagnose, Unfallhergang sowie potenziell risikomodulierendem Verhalten befragt.
Knapp 60% aller Probanden hatten in der Vergangenheit bereits mindestens eine Verletzung erlitten, 44% eine Verletzung, die eine ärztliche Konsultation erforderte. Während der prospektiv beobachteten 12 Monate trat bei 44% der Probanden mindestens eine Verletzung durch Indoor-Bouldern auf, davon 78% im UIAA Schweregrad 1, 19% im Schweregrad 2 und 3% im Schweregrad 3. Die obere Extremität war von 63% aller Verletzungen betroffen, die untere Extremität von 23%. Verletzungen der unteren Extremität waren häufiger im UIAA Schweregrad ≥ 2 klassifiziert (p = 0,007).
Probanden, die Bouldern erst seit maximal einem Jahr betrieben, hatten ein erhöhtes Risiko für Verletzungen der unteren Extremität (p = 0,027). Keine der untersuchten protektiven Maßnahmen inklusive Spotten konnten das Verletzungsrisiko senken. Das Nutzen von Kletterschuhen mit starkem Downturn und Vorspann erhöhte das Risiko für Verletzungen im UIAA Schweregrad ≥ 2 (p = 0,003). Verletzungen der Patienten aus Notaufnahme und Klettersprechstunde waren in 5,1% im UIAA Schweregrad 1, 48,7% im Schweregrad 2 und 46,2% im Schweregrad 3. Verletzungen der unteren Extremität waren häufiger im Schweregrad 3 (p = 0,015).
Verletzungen durch Indoor Bouldern sind häufig, der Großteil erfordert jedoch keine medizinische Behandlung. Verletzungen der unteren Extremität sind gehäuft in einem höheren Schweregrad. Die untersuchten Präventivmaßnahmen senkten das Verletzungsrisiko nicht. Einsteigerkurse sollten insbesondere sicheres Abspringen und Stürzen trainieren. Das Nutzen eines weiteren Paares Kletterschuhe ohne starken Downturn und Vorspann für das Training scheint ratsam. Zukünftige Forschung sollte sich der Verletzungsprävention insbesondere durch Stürzen und Abspringen widmen.
Einleitung: Die sog. Direct-Drive-Simulation (DDS) bietet Patienten präoperativ eine Klangsimulation des "Vibrant Soundbridge®-Hörens". Ein Floating-Mass-Transducer (FMT) wird auf das Trommelfell oder die rekonstruierte Paukenabdeckung aufgesetzt, worüber den Patienten Testsignale dargeboten werden. Ziel der Arbeit war die Evaluation einer Weiterentwicklung des Tests hin zur DDS-Tonaudiometrie und DDS-Sprachaudiometrie. Bei Bestätigung der ersten Hypothese, soll darüber hinaus geprüft werden, ob eine Hirnstammaudiometrie (BERA) über den DDS durchgeführt werden kann. Ziel der vorliegenden Arbeit ist es Referenzdaten an einer Population Normalhörender zu generieren. Zusammenfassung: Sowohl die DDS-Ton- und Sprachaudiometrie als auch die Ableitung einer BERA über den DDS-FMT sind möglich. Mit der DDS-BERA besteht nun erstmals die Möglichkeit eines objektiven DDS-Tests präoperativ. Die notwendige Einschätzung der Ankopplungsqualität zur korrekten Interpretation der DDS-BERA lässt sich elegant mit Hilfe der DDS-Tonaudiometrie ermitteln.
The charged aerosol detector (CAD) is an aerosol-based detector employed in liquid chromatography which has become established in the field of pharmaceutical analysis due to its outstanding performance characteristics, e.g. the almost uniform response for nonvolatile analytes. Owing to its principle of detection, the response of the CAD depends on the volatility of a compound and is inherently nonlinear. However, the newly implemented instrumental settings evaporation temperature and power function value (PFV) are valuable tools to overcome some of these drawbacks and can even enhance the detector’s capabilities when adjusted properly.
This thesis aimed to evaluate the impact of the new instrumental settings on the CAD performance. Additionally, the influence of modern separation techniques for small polar compounds on the CAD was assessed and the applicability of hyphenated UV-CAD techniques explored. The optimization strategies derived from the evaluation procedures and the conjunction of the instrumental and chromatographic techniques investigated were utilized for the challenging impurity profiling of amino acids and amino acid-like drugs.
The results of the method validation procedures confirmed the broad applicability of the CAD in the pharmaceutical analysis of nonvolatile compounds, supported by satisfactory sensitivity and reproducibility for meeting the regulatory requirements with respect to the ICH guidelines Q2(R1) and Q3A(R2). The limits of applicability include the analysis of semivolatile compounds, and the method transfer between current and legacy CAD models. Further advances in the definition and standardization of allowed ranges for the instrumental settings and the establishment of general optimization procedures in the method development could lead to a more widespread use of the detection technique in compendial methods.
Im Zuge der Erstdiagnostik einer Krankenhausaufnahme bei akuter Herzinsuffizienz ist die Röntgen-Thorax-Untersuchung fester Bestandteil. Ziel dieser Arbeit war es, ihren klinischen Stellenwert und die Aufnahmequalität systematisch zu untersuchen. In der AHF-Registerstudie wurden alle am Universitätsklinikum Würzburg vorstelligen Patienten mit akuter Herzinsuffizienz konsekutiv registriert und umfassend phänotypisiert. Die Röntgen-Thorax-Befunde wurden systematisch informationsextrahiert, auf Konsistenz überprüft, katalogisiert und klassifiziert.
Body representations are readily expanded based on sensorimotor experience. A dynamic view of body representations, however, holds that these representations cannot only be expanded but that they can also be narrowed down by disembodying elements of the body representation that are no longer warranted. Here we induced illusory ownership in terms of a moving rubber hand illusion and studied the maintenance of this illusion across different conditions. We observed ownership experience to decrease gradually unless participants continued to receive confirmatory multisensory input. Moreover, a single instance of multisensory mismatch – a hammer striking the rubber hand but not the real hand – triggered substantial and immediate disembodiment. Together, these findings support and extend previous theoretical efforts to model body representations through basic mechanisms of multisensory integration. They further support an updating model suggesting that embodied entities fade from the body representation if they are not refreshed continuously.
Voluntary actions and causally linked sensory stimuli are perceived to be shifted towards each other in time. This so-called temporal binding is commonly assessed in paradigms using the Libet Clock. In such experiments, participants have to estimate the timing of actions performed or ensuing sensory stimuli (usually tones) by means of a rotating clock hand presented on a screen. The aforementioned task setup is however ill-suited for many conceivable setups, especially when they involve visual effects. To address this shortcoming, the line of research presented here establishes an alternative measure for temporal binding by using a sequence of timed sounds. This method uses an auditory timer, a sequence of letters presented during task execution, which serve as anchors for temporal judgments. In four experiments, we manipulated four design factors of this auditory timer, namely interval length, interval filling, sequence predictability, and sequence length, to determine the most effective and economic method for measuring temporal binding with an auditory timer.
Purpose
Inhomogeneities of the static magnetic B\(_{0}\) field are a major limiting factor in cardiac MRI at ultrahigh field (≥ 7T), as they result in signal loss and image distortions. Different magnetic susceptibilities of the myocardium and surrounding tissue in combination with cardiac motion lead to strong spatio‐temporal B\(_{0}\)‐field inhomogeneities, and their homogenization (B0 shimming) is a prerequisite. Limitations of state‐of‐the‐art shimming are described, regional B\(_{0}\) variations are measured, and a methodology for spherical harmonics shimming of the B\(_{0}\) field within the human myocardium is proposed.
Methods
The spatial B\(_{0}\)‐field distribution in the heart was analyzed as well as temporal B\(_{0}\)‐field variations in the myocardium over the cardiac cycle. Different shim region‐of‐interest selections were compared, and hardware limitations of spherical harmonics B\(_{0}\) shimming were evaluated by calibration‐based B0‐field modeling. The role of third‐order spherical harmonics terms was analyzed as well as potential benefits from cardiac phase–specific shimming.
Results
The strongest B\(_{0}\)‐field inhomogeneities were observed in localized spots within the left‐ventricular and right‐ventricular myocardium and varied between systolic and diastolic cardiac phases. An anatomy‐driven shim region‐of‐interest selection allowed for improved B\(_{0}\)‐field homogeneity compared with a standard shim region‐of‐interest cuboid. Third‐order spherical harmonics terms were demonstrated to be beneficial for shimming of these myocardial B\(_{0}\)‐field inhomogeneities. Initial results from the in vivo implementation of a potential shim strategy were obtained. Simulated cardiac phase–specific shimming was performed, and a shim term‐by‐term analysis revealed periodic variations of required currents.
Conclusion
Challenges in state‐of‐the‐art B\(_{0}\) shimming of the human heart at 7 T were described. Cardiac phase–specific shimming strategies were found to be superior to vendor‐supplied shimming.
Background: In recent years, health care has increasingly become the focus of public interest, politics, health insurance companies, and research. This includes the development of therapeutic concepts that can respond individually to patients' resources in order to improve coping with chronic diseases. Research into psychosocial and biological resilience factors is very important and the basic objective of the present work. I studied patients with fibromyalgia syndrome (FMS), who suffer among others from chronic pain, fatigue, sleep and gastrointestinal problems. This patient cohort is characterized by a pronounced heterogeneity in terms of clinical outcome, degree in disability and coping. FMS has a prevalence of 3 – 8 % in the Western population and has a significant socio-economic impact. Validated psychosocial resilience factors include optimism, humor, coherence, self-efficacy, awareness with one's own resources and the ability to apply them profitably (coping), and a healthy social environment with positive relationships. Studies in patients with cancer revealed religiosity as positive and negative factor on the health outcome, but there is little data on religious aspects of pain resilience. Various genetic polymorphisms and anti-inflammatory cytokines are known as biological resilience factors. Various microRNA (miRNA) were detected to contribute to resilience in the context of stress and psychiatric disorders. Objective: The underlying research question of this work is to understand the factors that make some FMS patients resilient and others not, even though they suffer from the same disease. The long-term aim was to understand mechanisms and influencing factors of resilience to design preventive and resource-oriented therapies for FMS patients. Material and Methods: Three studies examined religious, physiological, biological, and psychosocial factors which may contribute to resilience in FMS patients. Study one combined data of questionnaires, a psychosocial interview, and regression analyses to investigate the relevance of religiosity for coping and resilience. Study two examined variance explaining factors and defined clusters among FMS patients by their differences in coping, pain phenotype and disability. The factor analysis used variables derived from questionnaires and qPCR of cytokines in white blood samples (WBC) of patients and healthy controls. Study three assessed cluster-wise miRNA signatures which may underly differences in behaviour, emotional and physiological disability, and resilience among patient clusters. A cluster-specific speculative model of a miRNA-mediated regulatory cycle was proposed and its potential targets verified by an online tool. Results: The data from the first study revealed a not very religious patient cohort, which was rather ambivalent towards the institution church, but described itself as a believer. The degree of religiosity played a role in the choice of coping strategy but had no effect on psychological parameters or health outcomes. The coping strategy "reinterpretation", which is closely related iv to the religious coping "reappraisal", had the highest influence on FMS related disability. Cognitive active coping strategies such as reappraisal which belongs to religious coping had the highest effect on FMS related disability (resilience) and could be trained by a therapist. Results from the second study showed high variances of all measured cytokines within the patient group and no difference between patient and control group. The high dispersion indicated cluster among patients. Factor analysis extracted four variance-explaining factors named as affective load, coping, pain, and pro-inflammatory cytokines. Psychological factors such as depression were the most decisive factors of everyday stress in life and represented the greatest influence on the variance of the data. Study two identified four clusters with respective differences in the factors and characterized them as poorly adapted (maladaptive), well adapted (adaptive), vulnerable and resilient. Their naming was based on characteristics of both resilience concepts, indicated by patients who were less stress-sensitive and impaired as a personal characteristic and by patients who emerged as more resilient from a learning and adaptive process. The data from the variance analysis suggests that problem- and emotion-focused coping strategies and a more anti-inflammatory cytokine pattern are associated with low impairment and contribute to resilience. Additional favorable factors include low anxiety, acceptance, and persistence. Some cluster-specific intervention proposals were created that combine existing concepts of behavioral and mindfulness therapies with alternative therapies such as vitamin D supplementation and a healthy intestinal flora. The results of the third study revealed lower relative gene expression of miR103a-3p, miR107, and miR130a-3p in the FMS cohort compared to the healthy controls with a large effect size. The adaptive cluster had the highest gene expression of miR103a-3p and tendentially of miR107, which was correlated with the subscale score "physical abuse" of the trauma questionnaire. Further correlations were found in particular with pain catastrophizing and FMS-related disability. MiR103a-3p and miR107 form a miRNA-family. Based on this, we proposed a miR103a/107 regulated model of an adaptive process to stress, inflammation and pain by targeting genetic factors which are included in different anti-inflammatory and stress-regulating pathways. Conclusion: All three studies provide new insights into resilience in FMS patients. Cognitive coping (reappraisal/reinterpretation) plays a central role and thus offers therapeutic targets (reframing in the context of behavioral therapy). Religosity as a resilience factor was only partially valid for our patient cohort. Basically, the use of resource-oriented therapy in large institutions still requires research and interdisciplinary cooperation to create a consensus between the humanities, natural sciences and humanism.
Miniaturized satellites on a nanosatellite scale below 10kg of total mass contribute most to the number of launched satellites into Low Earth Orbit today. This results from the potential to design, integrate and launch these space missions within months at very low costs. In the past decade, the reliability in the fields of system design, communication, and attitude control have matured to allow for competitive applications in Earth observation, communication services, and science missions. The capability of orbit control is an important next step in this development, enabling operators to adjust orbits according to current mission needs and small satellite formation flight, which promotes new measurements in various fields of space science. Moreover, this ability makes missions with altitudes above the ISS comply with planned regulations regarding collision avoidance maneuvering.
This dissertation presents the successful implementation of orbit control capabilities on the pico-satellite class for the first time. This pioneering achievement is demonstrated on the 1U CubeSat UWE–4. A focus is on the integration and operation of an electric propulsion system on miniaturized satellites. Besides limitations in size, mass, and power of a pico-satellite, the choice of a suitable electric propulsion system was driven by electromagnetic cleanliness and the use as a combined attitude and orbit control system. Moreover, the integration of the propulsion system leaves the valuable space at the outer faces of the CubeSat structure unoccupied for future use by payloads. The used NanoFEEP propulsion system consists of four thruster heads, two neutralizers and two Power Processing Units (PPUs).
The thrusters can be used continuously for 50 minutes per orbit after the liquefaction of the propellant by dedicated heaters. The power consumption of a PPU with one activated thruster, its heater and a neutralizer at emitter current levels of 30-60μA or thrust levels of 2.6-5.5μN, respectively, is in the range of 430-1050mW. Two thruster heads were activated within the scope of in-orbit experiments. The thrust direction was determined using a novel algorithm within 15.7° and 13.2° of the mounting direction. Despite limited controllability of the remaining thrusters, thrust vector pointing was achieved using the magnetic actuators of the Attitude and Orbit Control System.
In mid 2020, several orbit control maneuvers changed the altitude of UWE–4, a first for pico-satellites. During the orbit lowering scenario with a duration of ten days, a single thruster head was activated in 78 orbits for 5:40 minutes per orbit. This resulted in a reduction of the orbit altitude by about 98.3m and applied a Delta v of 5.4cm/s to UWE–4. The same thruster was activated in another experiment during 44 orbits within five days for an average duration of 7:00 minutes per orbit. The altitude of UWE–4 was increased by about 81.2m and a Delta v of 4.4cm/s was applied. Additionally, a collision avoidance maneuver was executed in July 2020, which increased the distance of closest approach to the object by more than 5000m.
Die WHO-Klassifikation der Hirntumoren von 2016 ebnete den Weg für molekulare Marker und Therapie-Angriffspunkte. Der Transkriptionsfaktor ATF5 könnte ein solcher sein. Er unterdrückt die Differenzierung von neuronalen Vorläuferzellen und wird in Glioblastomen (GBM) überexprimiert. Daten zur ATF5-Expression in WHO Grad II Gliomen (LGG) und GBM-Rezidiven sind nur spärlich vorhanden. Daher untersuchten wir 79 GBM, 40 LGG und 10 Normalhirnproben auf ihre ATF5-mRNA- und Proteinexpression mit quantitativer Echtzeit-PCR bzw. Immunhistochemie und verglichen sie mit multiplen, retrospektiv erhobenen klinischen Charakteristika der Patienten. ATF5 war in LGG und GBM verglichen zum Normalhirn sowohl auf mRNA-, als auch Proteinebene überexprimiert. Obwohl die ATF5-mRNA-Expression im GBM eine erhebliche Fluktuationsrate zeigte, gab es keine signifikanten Expressionsunterschiede zwischen GBM-Gruppen unterschiedlicher biologischer Wachstumsmuster. ATF5-mRNA korrelierte mit dem Alter der Patienten und invers mit der Ki67-Färbung. Kaplan Meier- und Cox-Regressionsanalysen zeigten eine signifikante Korrelation der ATF5-mRNA-Expression mit dem Überleben nach 12 Monaten sowie dem progressionsfreien Überleben. Die Methylierung des Promotors der O6-Methylguanin-DNA-Methyltransferase (MGMT) ist ein etablierter Marker in der Therapie des GBMs. Sie ist mit dem therapeutischen Ansprechen auf Temozolomid und dem Überleben assoziiert. Uns fielen inzidentell Veränderungen der MGMT-Promotormethylierung auf, woraufhin wir den aktuellen Wissensstand mittels einer ausführlichen Literatur-Metaanalyse zusammenfassten. Dabei fanden wir Veränderungen der MGMT-Promotormethylierung bei 115 der 476 Patienten. Wir schlussfolgern, dass die ATF5-mRNA-Expression als prognostischer Faktor für das Überleben der Patienten dienen könnte. Da seine in vitro-Inhibition zu einem selektiven Zelltod von Gliomzellen führte und wir eine Überexpression in glialen Tumoren nachweisen konnten, zeigt ATF5 Potential als ubiquitäres Therapieziel in Gliomen. Zum aktuellen Zeitpunkt ergibt sich keine klare Indikation, den klinischen Standard der MGMT-Teststrategie zu verändern. Trotzdem könnte eine erneute Testung der MGMT-Promotormethylierung für zukünftige Therapieentscheidungen sinnvoll sein und wir regen an, dass dieses Thema in klinischen Studien weiter untersucht wird.
One of the major motivations for the analysis and modeling of time series data is the forecasting of future outcomes. The use of interval forecasts instead of point forecasts allows us to incorporate the apparent forecast uncertainty. When forecasting count time series, one also has to account for the discreteness of the range, which is done by using coherent prediction intervals (PIs) relying on a count model. We provide a comprehensive performance analysis of coherent PIs for diverse types of count processes. We also compare them to approximate PIs that are computed based on a Gaussian approximation. Our analyses rely on an extensive simulation study. It turns out that the Gaussian approximations do considerably worse than the coherent PIs. Furthermore, special characteristics such as overdispersion, zero inflation, or trend clearly affect the PIs' performance. We conclude by presenting two empirical applications of PIs for count time series: the demand for blood bags in a hospital and the number of company liquidations in Germany.

Die vorliegende Studie untersucht das Langzeitoutcome der filtrierenden Trabe- kulotomie (FTO) und vergleicht diese mit der konventionellen Trabekulektomie. Nach- dem in der Arbeit von Matlach et al. bereits gezeigt werden konnte, dass die filtrierende Trabekulotomie nach einem Jahr im Bezug auf die Augeninnendrucksenkung und das Auftreten von postoperativen Komplikationen mit der konventionellen Trabekulektomie vergleichbar ist (32), sollte in der vorliegenden Arbeit der Langzeiterfolg der beiden Ope- rationsmethoden miteinander verglichen werden. Untersucht wurde deshalb der intraoku- lare Druck, Langzeitkomplikationen, Folgeoperationen und der Visus nach eineinhalb und zwei Jahren.
Methode:
In einer Fall-Kontroll-Studie wurden 30 Patienten nach einer filtrierenden Tra- bekulotomie (prospektiv) mit 87 Patienten nach einer konventionellen Trabekulektomie (retrospektiv) verglichen. Alle Patienten waren zwischen 2007 und 2012 von demselben Operateur an der Augenklinik der Universität Würzburg operiert worden. Eingeschlossen wurden Patienten mit primärem Offenwinkelglaukom, Pseudoexfoliationsglaukom und Pigmentdispersionsglaukom. Die Patienten waren nach Alter und intraokularem Druck (IOD) gematcht worden. Die Daten der vorliegenden Studie wurden an der Augenklink der Universität Würzburg oder von den weiterbehandelnden niedergelassenen Augenärz- ten mit Hilfe eines für diese Studie entworfenen Fragebogens erhoben.
Endpunkt: Als primärer Endpunkt wurde ein Augeninnendruck von < 18 mmHg und eine Augeninnendrucksenkung um ≥ 30 % des Ausgangswertes festgelegt. Wurden diese Kriterien ohne Medikamente erreicht, sprach man von einem „absoluten Erfolg“ (com- plete-success), andernfalls von einem „relativen Erfolg“ (qualified-success).
Als sekundäre Endpunkte galten der intraokulare Druckverlauf, die Erfassung der Kom- plikationen, Folgeeingriffe, der Visusverlauf und die Medikation.
Ergebnisse: Von den anfangs 117 eingeschlossenen Patienten konnten nach 2 Jahren die Daten von 95 Patienten (27 aus der FTO-Gruppe und 68 aus der TE-Gruppe) erhoben werden. Präoperativ unterschieden sich die beiden Studiengruppen nicht signifikant im Bezug auf das Alter und den IOD, nach denen die Gruppen gematcht worden waren, den Visus (p= 0,60), das Geschlecht (p = 0,30) und die präoperativ verwendeten Medikamente (p≥ 0,05). In der FTO-Gruppe lag die Zahl der Patienten mit Pigmentdispersionsglaukom (p= 0,02) und Pseudophakie (p = 0,02) schon präoperativ deutlich über der der TE-
Gruppe. Der IOD lag präoperativ in beiden Operationsgruppen bei 23 mmHg (IQR 20- 27), wobei im Median 3,0 verschiedene augendrucksenkende Medikamente angewendet wurden.
Der absolute Erfolg (complete-success) unterschied sich im Zeitraum der vorliegenden Studie zu keinem Zeitpunkt signifikant zwischen den Studiengruppen (p 18Mon. = 0,50 ; p 24Mon. = 0,067). Auch der relative Erfolg (qualified-success) unterschied sich zwischen den Gruppen nicht signifikant (p 18Mon. = 0,23 ; p 24Mon. = 0,60). Nach 2 Jahren erreichten 33,3 % der FTO- und 56,7 % der TE-Patienten einen absoluten Erfolg und 70,4 % bzw. 77,6 % einen relativen Erfolg.
Der mediane postoperative IOD konnte auch 1,5 und 2 Jahre nach den Operationen sig- nifikant unter den präoperativen IOD gesenkt werden (p < 0,001) und betrug nach 2 Jah- ren in der FTO-Gruppe 12,80 ± 3,79 mmHg und in der TE-Gruppe 11 mmHg (IQR 9- 13). Er unterschied sich auch zwischen beiden Gruppen zu keinem Zeitpunkt signifikant (p 18Mon. = 0,18 ; p 24Mon. = 0,12).
Der mediane postoperative Visus unterschied sich 1,5 und 2 Jahre nach der OP zwischen den Gruppen nicht signifikant (p 1,5a = 0,11; p 2a = 0,77). In der FTO-Gruppe verschlech- terte sich der Visus der Patienten von 0,10 logMAR (0,79) nach 18 Monaten auf 0,20 logMAR (0,63) nach 24 Monaten. Der Visus in der TE-Gruppe blieb konstant (0,15 log- MAR).
Insgesamt hatte sich der Visus der Patienten nach 1,5 und 2 Jahren im Vergleich zum präoperativen Visus signifikant verschlechtert (p 1,5a = 0,02; p 2a = 0,00).
Bei der separaten Auswertung der Patienten mit PEX-Glaukom konnte der IOD postope- rativ zu allen statistisch beurteilbaren Zeitpunkten signifikant unter den präoperativen IOD gesenkt werden. Bei einem Vergleich zwischen den Gruppen lag der IOD der FTO- Gruppe einmalig über dem der TE-Gruppe. In den anderen Fällen traten keine signifikan- ten Druckunterschiede zwischen den Gruppen auf. Der postoperative Visus unterschied sich zu keinem Zeitpunkt, ob mit oder ohne Kunstlinse, im prä- postoperativen Vergleich und zwischen den Gruppen.
Die Anzahl der Wirkstoffklassen unterschied sich zwischen den Gruppen nicht signifi- kant. Reoperationen traten nach beiden Operationsmethoden so selten auf, sodass eine statistische Bewertung nicht möglich war.
Die Verteilung der Glaukomformen hatte sich auch nach 1,5 und 2 Jahren nicht verändert.
Die Zahl pseudophaker Patienten war im Beobachtungszeitraum auch in der TE-Gruppe stetig angestiegen, so dass sie sich nach 2 Jahren nicht mehr signifikant zwischen den Gruppen unterschied.
Schlussfolgerung: Die FTO ist im Bezug auf den IOD, den Visus und die Erfolgsrate der TE äquivalent.
Im Langzeitverlauf traten in der FTO Gruppe nicht mehr Komplikationen als nach TE auf. Nach 1 Jahr waren in der FTO-Gruppe signifikant mehr frühe Komplikationen auf- getreten als in der TE. Nach 2 Jahren war die Rate an Komplikationen in den Gruppen nicht mehr signifikant unterschiedlich.
Educational robotics is an innovative approach to teaching and learning a variety of different concepts and skills as well as motivating students in the field of Science, Technology, Engineering, and Mathematics (STEM) education. This especially applies to educational robotics competitions such as, for example, the FIRST LEGO League, the RoboCup Junior, or the World Robot Olympiad as out-of-school and goal-oriented approach to educational robotics. These competitions have gained greatly in popularity in recent years and thousands of students participate in these competitions worldwide each year. Moreover, the corresponding technology became more accessible for teachers and students to use it in their classrooms and has arguably a high potential to impact the nature of science education at all levels. One skill, which is said to be benefitting from educational robotics, is problem solving. This thesis understands problem solving skills as engineering design skills (in contrast to scientific inquiry). Problem solving skills count as important skills as demanded by industry leaders and policy makers in the context of 21st century skills, which are relevant for students to be well-prepared for their future working life in today’s world, shaped by an ongoing process of automation, globalization, and digitalization. The overall aim of this thesis is to try to answer the question if educational robotics competitions such as the World Robot Olympiad (WRO) have a positive impact on students’ learning in terms of their problem solving skills (as part of 21st century skills). In detail, this thesis focuses on a) if students can improve their problem solving skills through participation in educational robotics competitions, b) how this skill development is accomplished, and c) the teachers’ support of their students during their learning process in the competition. The corresponding empirical studies were conducted throughout the seasons of 2018 and 2019 of the WRO in Germany. The results show overall positive effects of the participation in the WRO on students’ learning of problem solving skills. They display an increase of students’ problem solving skills, which is not moderated by other variables such as the competition’s category or age group, the students’ gender or experience, or the success of the teams at the competition. Moreover, the results indicate that students develop their problem solving skills by using a systematic engineering design process and sophisticated problem solving strategies. Lastly, the teacher’s role in the educational robotics competitions as manager and guide (in terms of the constructionist learning theory) of the students’ learning process (especially regarding the affective level) is underlined by the results of this thesis. All in all, this thesis contributes to the research gap concerning the lack of systematic evaluation of educational robotics to promote students’ learning by providing more (methodologically) sophisticated research on this topic. Thereby, this thesis follows the call for more rigorous (quantitative) research by the educational robotics community, which is necessary to validate the impact of educational robotics.
The following study, The Integration of Female Refugees in Germany: Perspectives of Women and an Analysis of Federal and Selected State and City Integration Policies from 1998-2019, is focused on the qualitative analysis of integration policy in Germany regarding female refugees. The states of North Rhine-Westphalia, Bavaria, and Saxony-Anhalt have been selected for this dissertation as well as the cities of Cologne, Wuerzburg, and Magdeburg. Through an analysis and comparison of integration policies and programs on the federal and selected state and city levels the question will be answered how recognized female refugees are taken into account with the development and formulation of integration policy in Germany. The analysis is then complemented through interviews with recognized female refugees in each of the states and cities. Through analyzing the results of the interviews the question will be answered how the women view their situation and integration. Through a comparison of the findings from the policy analysis and the interviews it will then be able to decipher if integration policies and programs are truly reaching their target group, if they are effective, or what hurdles they may be producing. The goal of the study is to provide initial findings on the overall integration of recognized female refugees in Germany in connection to integration policies in order to discover potential deficits or ineffective programs and policies which can then be further researched in order to produce concrete policy suggestions.
Sterol O-Acyltransferasen (SOATs) spielen eine zentrale Rolle im Cholesterinstoffwechsel von Zellen, indem sie die Veresterung von freiem Cholesterin und Speicherung in Lipid droplets katalysieren. In Tumorzellen findet häufig eine Aktivierung alternativer Pfade des Energiestoffwechsels, unter anderem des Lipidstoffwechsels statt. Präklinische und klinische Daten unterstützen den Mechanismus der SOAT-Inhibierung als Therapiekonzept für bestimmte Tumore. Eine genaue Kenntnis sowohl dieser Inhibitoren als auch der Expression des Zielmoleküls ist Voraussetzung für eine klinische Anwendung.
Im ersten Teil dieser Arbeit wurde ein in-vitro SOAT-Aktivitätsassay etabliert und auf Grundlage dessen ein Vergleich der mittleren Hemmstärken ausgewählter SOAT-Inhibitoren gezogen. SOAT-transfizierte AD-293 Zellen sowie NCI-H295R Nebennieren-Zellen wurden mit dem fluoreszierenden 22-NBD-Cholesterin sowie den SOAT-Inhibitoren inkubiert und die Veresterung des Lipid-Analogons dann zunächst mikroskopisch und anschließend quantitativ mittels chromatographischer Auftrennung untersucht. Mitotane stellte sich mit einer IC50 von 1,3x10⁻⁶ M als schwächster SOAT-Inhibitor dar, gefolgt von Sandoz58-035 (IC50=1,4x10\(^{-8}\) M), ATR101 (IC50=3,1x10\(^{-9}\) M) und schließlich AZD3988 (IC50=8,8x10\(^{-10}\) M).
Im zweiten Teil dieser Arbeit wurde die SOAT-Expression in Prostatektomiepräparaten von Hochrisiko Prostatakarzinom-Patienten mittels Immunhistochemie bestimmt. Eine starke SOAT1 Expression (SOAT H-Score 3) war sowohl in der univariaten als auch in der multivariaten Analyse hoch signifikant mit einem kürzeren biochemisch progressfreien Überleben der Patienten assoziiert unabhängig von etablierten Prognoseparametern [HR für den biochemischen Progress 2,33 (95%KI 1,48-3,68), p<0,001)]. Für SOAT2 war dies erwartungsgemäß nicht der Fall. SOAT1 scheint bei diesem bestimmten Kollektiv einen vielversprechenden Stellenwert als prognostischer Marker zu haben und könnte darüber hinaus zukünftig als Zielmolekül im Rahmen einer individualisierten Therapie des Prostatakarzinoms in Frage kommen.
Owing to climate change, natural forest disturbances and consecutive salvage logging are drastically increasing worldwide, consequently increasing the importance of understanding how these disturbances would affect biodiversity conservation and provision of ecosystem services.
In chapter II, I used long-term water monitoring data and mid-term data on α-diversity of twelve species groups to quantify the effects of natural disturbances (windthrow and bark beetle) and salvage logging on concentrations of nitrate and dissolved organic carbon (DOC) in streamwater and α-diversity. I found that natural disturbances led to a temporal increase of nitrate concentrations in streamwater, but these concentrations remained within the health limits recommended by the World Health Organization for drinking water. Salvage logging did not exert any additional impact on nitrate and DOC concentrations, and hence did not affect streamwater quality. Thus, neither natural forest disturbances in watersheds nor associated salvage logging have a harmful effect on the quality of the streamwater used for drinking water. Natural disturbances increased the α-diversity in eight out of twelve species groups. Salvage logging additionally increased the α-diversity of five species groups related to open habitats, but decreased the biodiversity of three deadwood-dependent species groups.
In chapter III, I investigated whether salvage logging following natural disturbances (wildfire and windthrow) altered the natural successional trajectories of bird communities. I compiled data on breeding bird assemblages from nine study areas in North America, Europe and Asia, over a period of 17 years and tested whether bird community dissimilarities changed over time for taxonomic, functional and phylogenetic diversity when rare, common and dominant species were weighted differently. I found that salvage logging led to significantly larger dissimilarities than expected by chance and that these dissimilarities persisted over time for rare, common and dominant species, evolutionary lineages, and for rare functional groups. Dissimilarities were highest for rare, followed by common and dominant species.
In chapter IV, I investigated how β-diversity of 13 taxonomic groups would differ in intact, undisturbed forests, disturbed, unlogged forests and salvage-logged forests 11 years after a windthrow and salvage logging. The study suggests that both windthrow and salvage logging drive changes in between-treatment β-diversity, whereas windthrow alone seems to drive changes in within-treatment β-diversity. Over a decade after the windthrow at the studied site, the effect of subsequent salvage logging on within-treatment β-diversity was no longer detectable but the effect on between-treatment β-diversity persisted, with more prominent changes in saproxylic groups and rare species than in non-saproxylic groups or common and dominant species.
Based on these results, I suggest that salvage logging needs to be carefully weighed against its long-lasting impact on communities of rare species. Also, setting aside patches of naturally disturbed areas is a valuable management alternative as these patches would enable post-disturbance succession of bird communities in unmanaged patches and would promote the conservation of deadwood-dependent species, without posing health risks to drinking water sources.
Neuroblastoma is the most abundant, solid, extracranial tumor in early childhood and the leading cause of cancer-related childhood deaths worldwide. Patients with high-risk neuroblastoma often show MYCN-amplification and elevated levels of Aurora-A. They have a low overall survival and despite multimodal therapy options a poor therapeutic prognosis. MYCN-amplified neuroblastoma cells depend on Aurora-A functionality. Aurora-A stabilizes MYCN and prevents it from proteasomal degradation by competing with the E3 ligase SCFFBXW7. Interaction between Aurora-A and MYCN can be observed only in S phase of the cell cycle and activation of Aurora-A can be induced by MYCN in vitro. These findings suggest the existence of a profound interconnection between Aurora-A and MYCN in S phase. Nevertheless, the details remain elusive and were investigated in this study.
Fractionation experiments show that Aurora-A is recruited to chromatin in S phase in a MYCN-dependent manner. Albeit being unphosphorylated on the activating T288 residue, Aurora-A kinase activity was still present in S phase and several putative, novel targets were identified by phosphoproteomic analysis. Particularly, eight phosphosites dependent on MYCN-activated Aurora-A were identified. Additionally, phosphorylation of serine 10 on histone 3 was verified as a target of this complex in S phase. ChIP-sequencing experiments reveal that Aurora-A regulates transcription elongation as well as histone H3.3 variant incorporation in S phase. 4sU-sequencing as well as immunoblotting demonstrated that Aurora-A activity impacts splicing. PLA measurements between the transcription and replication machinery revealed that Aurora-A prevents the formation of transcription-replication conflicts, which activate of kinase ATR.
Aurora-A inhibitors are already used to treat neuroblastoma but display dose-limiting toxicity. To further improve Aurora-A based therapies, we investigated whether low doses of Aurora-A inhibitor combined with ATR inhibitor could increase the efficacy of the treatment albeit reducing toxicity. The study shows that the combination of both drugs leads to a reduction in cell growth as well as an increase in apoptosis in MYCN-amplified neuroblastoma cells, which is not observable in MYCN non-amplified neuroblastoma cells. This new approach was also tested by a collaboration partner in vivo resulting in a decrease in tumor burden, an increase in overall survival and a cure of 25% of TH-MYCN mice. These findings indicate indeed a therapeutic window for targeting MYCN-amplified neuroblastoma.

Diese retrospektive Auswertung von PatientInnendaten der kardiologischen Ambulanz des Universitätsklinikums Würzburg konnte zeigen, dass die Bestimmung der diastolischen Dysfunktion prognostisch relevante Informationen enthält. Das Studienkollektiv wurde anhand der gemessenen Ejektionsfraktion in die zwei Untersuchungsgruppen HFrEF und HFmrEF eingeteilt. Diese zwei Untersuchungsgruppen wurden anhand ihrer klinisch und echokardiographisch bestimmten Charakteristika verglichen. Anschließend wurden drei diastolische Parameter (E/e’, LAVi und TRVmax) auf ihre prognostische Relevanz untersucht. Die abschließende Untersuchung gruppierte die PatientInnen anhand der Schwere ihrer diastolischen Dysfunktion (mild / moderat / schwer) und untersuchte ebenfalls das Langzeitüberleben.
Die HFmrEF-Gruppe zeigte ähnliche klinische Charakteristika wie die HFrEF-Gruppe. Eine ischämische Genese der Herzinsuffizienz wurde in der HFmrEF-Gruppe im Vergleich zur HFrEF-Gruppe häufiger beobachtet.
Die Überlebenszeitanalysen konnten bei PatientInnen in der HFmrEF-Gruppe zeigen, dass ein dilatierter linker Vorhof (LAVi) oder eine große Regurgitation über der Trikuspidalklappe (TRVmax) mit einer schlechten Prognose einhergehen. Bei HFrEF-PatientInnen hingegen konnte dies nicht nachgewiesen werden. Hier zeigte sich, dass insbesondere der Parameter E/e’septal prognostisch relevante Informationen enthält.
Die Auswertung der Untersuchungsgruppen nach Einteilung anhand der Schwere der diastolischen Dysfunktion konnte die gefunden Effekte bestätigen. Eine moderate bis schwere diastolische Dysfunktion war mit einer signifikant schlechteren Prognose behaftet, und zwar sowohl in der HFrEF- wie auch in der HFmrEF-Gruppe.
Die gefunden Ergebnisse zeigen, dass die diastolische Dysfunktion auch bei PatientInnen mit einer systolischen Herzinsuffizienz wichtige prognostische Informationen enthalten. In der klinischen Routine sollte die echokardiographische Bestimmung der diastolischen Herzfunktion standardmäßig durchgeführt werden.
Die Ergebnisse könnten nicht nur in der Diagnostik zur Kategorisierung der PatientInnen und Bestimmung der Prognose, sondern auch hinsichtlich der Therapie von großem zukünftigem Nutzen sein. Hierzu sollten perspektivisch vor allem therapeutische Aspekte in prospektiven, idealerweise randomisierten Studien untersucht werden, welche sich auf die Erkenntnisse dieser Arbeit beziehen.
Ärztliche Kommunikation wird vielerorts bereits im Studium eingeübt. Das Aufklärungsgespräch vor einer Operation ist ein spezifischer Kommunikationsanlass, der einer differenzierten Rückmeldung an die Studierenden bedarf. Ziel war es, im Rahmen eines Kommunikationstrainings die Rückmeldung verschiedener Feedbackgeber (ärztlicher Experte, geschulte Tutorinnen und Tutoren, Studierende der Peer Group, Aufklärende selbst und Simulationspersonen) anhand von Bewertungschecklisten zu strukturieren und die Ergebnisse zu vergleichen. 171 Humanmedizinstudierende des 8. Semesters der Universität Würzburg nahmen in Kleingruppen an einem Training zur präoperativen Aufklärung teil. 50 Personen davon führten ein Aufklärungsgespräch und erhielten Feedback. Im Fokus der Gespräche standen „Kommunikation“ sowie „Komplikationen“. Die Studierenden bereiteten sich mittels Unterrichtsmaterialien auf der universitätseigenen E-Learning-Plattform vor. Gegenstand der statistischen Auswertungen waren die Testgüte der Bewertungschecklisten, die Bewertungspunkte in den Skalen und die Übereinstimmung der Bewertungen auf Basis des Intraklassenkorrelationskoeffizienten (ICC). Die Bewertungschecklisten wiesen zufriedenstellende Werte für interne Konsistenz, Itemschwierigkeit und Trennschärfe auf. Die Mittelwerte der Scores zur „Kommunikation“ unterschieden sich teilweise signifikant durch die 5 Bewertungsquellen, wobei hier die Selbsteinschätzung durch die studentischen Aufklärenden am strengsten ausfiel. Die studentischen Tutorinnen und Tutoren bewerteten identisch zum Experten. In Bezug auf die „Komplikationen“ gab es keine signifikanten Abweichungen zwischen den Bewertenden. Es konnte gezeigt werden, dass innerhalb des hochspezifischen Settings eines Simulationstrainings und nach geplanter Vorbereitung geschulte studentische Tutoren/innen eine vergleichbar gute Rückmeldung wie der ärztliche Experte geben können. Bei überwiegenden Übereinstimmungen im strukturierten Feedback darf somit der Rückmeldung durch Tutoren/innen oder Peers zukünftig ein höherer Stellenwert eingeräumt werden.
Genome Wide Association Studies (GWAS) have revolutionized the way on
how genotype-phenotype relations are assessed. In the 20 years long history
of GWAS, multiple challenges from a biological, computational, and statistical
point of view have been faced. The implementation of this technique using
the model plant species Arabidopsis thaliana, has enabled the detection of many
association for multiple traits. Despite a lot of studies implementing GWAS
have discovered new candidate genes for multiple traits, different samples are
used across studies. In many cases, either globally diverse samples or samples
composed of accessions from a geographically restricted area are used. With
the aim of comparing GWAS outcomes between populations from different
geographic areas, this thesis describes the performance of GWAS in different
European samples of A. thaliana. Here, association mapping results for flowering
time were compared. Chapter 2 describes the analyses of random resampling
from this original sample. The aim was to establish reduced subsamples to
later carry out GWAS and compare the outcomes between these subsamples.
In Chapter 3, the European sample was split into eight equally-sized local
samples representing different geographic regions. Next, GWAS was carried
out and an attempt was made to clarify the differences in GWAS outcomes.
Chapter 4 contains the results of a collaboration with Prof. Dr. Wolfgang Dröge-
Laser, in which my mainly task was the analysis of RNAseq data from A.
thaliana plants infected by pathogenic fungi. Finally, Appendix A presents a very
short description of my participation in the GHP Project on Access to Care for
Cardiometabolic Diseases (HPACC) at the university of Heidelberg.
The substitution of selected CC units by their isoelectronic and isosteric BN units in π−conjugated organic compounds (BN/CC isosterism), especially polycyclic aromatic hydrocarbons (PAHs), has emerged as a viable strategy to produce novel organic–inorganic hybrid materials with structural similarities to their all-carbon congeners, but in many cases with intriguing properties and functions.
In the first two chapters the synthesis and properties of novel BNB-doped phenalenyls, dithienoazadiborepins and dithienooxadiborepins are presented. The optoelectronic properties of these new building blocks can be effectively tuned by variation of the incorporated Ar (Mes, Tip, FMes) and R groups (H, Me, i-Pr, t-Bu, Ph). Theoretical investigations, including NICS (Nucleus Independent Chemical Shift) scans and AICD (Anisotropy of the Induced Current Density) calculations, have been performed which provide insight into their aromatic or antiaromatic character, respectively.
The incorporation of BP units, on the other hand, which are valence isoelectronic with BN and CC, into unsaturated organic compounds, has been scarcely studied, though the potential of the resulting BCP hybrid materials for electronic applications has been recognized quite recently. Main chain conjugated polymers featuring BP fragments in the backbone are unknown so far. The first molecular model compounds for a BP analogue of the conjugated polymer poly(p-phenylene vinylene) (PPV) are presented in chapter 3. Theoretical investigations revealed that the Mes* group to fully planarizes the phosphorus center, increasing the B=P double bond character and enabling conjugation over the BP unit. Different synthetic approaches to the molecular model compounds have been investigated and a viable synthetic strategy was found.
Bei ICCC (intrahepatischen cholangiozellulären Karzinomen) handelt es sich um eine seltene Erkrankung der Gallenwege. Die Erkrankung schreitet langsam voran und verläuft lange Zeit asymptomatisch oder verursacht nur unspezifische Symptome, sodass sich die meisten Patienten bei Diagnosestellung bereits in einem fortgeschrittenen, nicht-resezierbaren Tumorstadium befinden.
Im Rahmen dieser retrospektiven Arbeit wurden Einflussfaktoren auf das Überleben und die Entwicklung eines Tumorrezidivs von Patienten untersucht, die aufgrund eines ICCC kurativ leberreseziert wurden. Hierbei wurden 42 Patienten eingeschlossen, die zwischen 2003 und 2015 am Universitätsklinikum Würzburg operiert wurden.
Die Datenauswertung lieferte die Erkenntnis, dass sowohl das Vorhandensein multifokal lokalisierter ICCC als auch ein erhöhtes Lebensalter (60 Jahre und
älter) zum Zeitpunkt der Operation mit einem geringeren perioperativen und langfristigen Überleben der Patienten assoziiert sind. Außerdem sind diese beiden Faktoren mit der Entstehung eines Tumorrezidivs, was die Überlebenszeit verkürzt, assoziiert.
Hingegen konnte bei Auftreten von postoperativen Komplikationen und dem Vorhandensein einer höhergradigen ASA-Klassifikation (ASA III und ASA IV) eine Assoziation mit einem geringeren perioperativen nicht aber mit einem reduzierten langfristigen Überleben gezeigt werden.
Aufgrund der sich umkehrenden Alterspyramide in Deutschland leiden bereits jetzt immer mehr Menschen an Gelenkknorpelschäden. Doch nicht nur das Alter, sondern auch Unfälle und Sportverletzungen und Übergewicht können zu irreversiblen Knorpeldefekten führen. Obwohl es diverse Behandlungsmöglichkeiten gibt, können die bisherige Methoden nicht als dauerhafte Heilung betrachtet werden. Im Rahmen des internationalen Forschungsprojektes BIO-CHIP sollte eine vielsprechende Behandlungsmethode mit neuartigen Arzneimitteln untersucht werden.
Als Ausgangsmaterial des Arzneimittels, ein hergestelltes Knorpelimplantat, dienen patienteneigene Knorpelzellen aus der Nase. Diese werden isoliert, vermehrt und letztlich auf einer Matrix zu einem Knorpelimplantat kultiviert. Wesentliche Voraussetzung für die Implantatfreigabe stellt neben toxikologischen und biologischen Unbedenklichkeitstests die Beurteilung der Viabilität dar. Diese wurde bisher anhand von Histologieschnitten von der Pathologie durchgeführt.
Ziel der vorliegenden Arbeit war die Entwicklung und Validierung eines standardisierten und objektiven Viabilitätstests für die Chondrozyten innerhalb der Knorpelmatrix. Hierfür wurde die LDH als Marker für irreversibel geschädigte Zellen verwendet. Die LDH Konzentration konnte mit dem CyQuant LDH-Assay durch die Messung der Absorption gemessen werden. Es konnte nachgewiesen werden, dass LDH die erforderliche Stabilität und Nachweisbarkeit im Medium besitzt. Mithilfe der Lyse, analog zum Herstellungsprozess, gezüchteter Mini-Knorpelimplantate, konnten die maximal erreichbaren LDH Konzentrationen ermittelt werden. Mithilfe dieser Konzentrationen wurde eine Eichkurve generiert. Diese dient als Beurteilung der Viabilität zukünftig gemessener Absorptionen des Überstandmediums.
Das entwickelte Verfahren erfordert keine invasiven Eingriffe am Implantat und zeichnet sich durch eine einfache Durchführung aus, da nur der Überstand gemessen werden muss. Die durchgeführte Validierung der Methode bescheinigte eine hohe Robustheit, Linearität, Genauigkeit und Präzision.
Mittels Tissue Engineering hergestellte humane 3D in vitro-Testsysteme sind ein neuer Ansatz, um u.a. Erkrankungen der Atemwege zu simulieren und zu untersuchen. Obwohl gegen B. pertussis, den Erreger des Keuchhustens, Impfstoffe zur Verfügung stehen, nimmt die Erkrankungs-Inzidenz in den letzten Jahren deutlich zu. Da B. pertussis zu den obligat humanpathogenen Erregern zählt, sind die aus Tierversuchen stammenden Daten nur unzureichend auf den Menschen übertragbar. Die genauen Pathomechanismen der Infektion sind bisher nicht geklärt.
Auf einer biologischen Kollagenmatrix wurde eine Ko-Kultur aus humanen tracheobronchialen Fibroblasten und humanen tracheobronchialen Epithelzellen (hTEC) angesiedelt und 3 Wochen unter apikaler Belüftung kultiviert. Die ausdifferenzierten 3D Testsysteme wurden mit Überständen von Bordetella pertussis-Kulturen inkubiert und auf licht- und elektronenmikroskopischer Ebene analysiert. Weiterhin wurden 2D Kulturen der hTEC mit Hilfe der Ramanspektroskopie nicht-invasiv auf intrazelluläre Veränderungen nach der Inkubation mit den bakteriellen Überständen untersucht.
Das 3D Testsystem der humanen Atemwegschleimhaut zeigte auf lichtmikroskopischer und ultrastruktureller Ebene eine hohe in vitro – in vivo-Korrelation. Die elektronenmikroskopische Analyse zeigte morphologische Veränderungen nach der Inkubation mit den B. pertussis Überständen, die mit vorbeschrieben Effekten einer B. pertussis Infektion korrelieren. Mittels der Ramanspektroskopie ließen sich Gruppen von unbehandelten Zellen von Gruppen, die zuvor mit Bakterienüberständen inkubiert wurden, trennen. Somit zeigte sich die Ramanspektroskopie sensitiv für intrazelluläre Infektionsfolgen.
Zusammenfassend wurde belegt, dass das 3D-Modell der humanen Atemwegschleimhaut zur Untersuchung obligat humanpathogener Infektionserreger geeignet ist und dass die Ramanspektroskopie eine nicht-invasive Methode ist, um durch Infektionen hervorgerufene intrazellulären Pathologien zu analysieren.
Anlass für diesen Aufsatz ist die Tatsache, dass die Duden-Redaktion bei zahlreichen
Personenbezeichnungen das sog. ‚generische Maskulinum‘ wegdefiniert hat. Das Anfang 2021 erschienene online Wörterbuch gibt als Bedeutungserklärung des Substantiv Arzt folgende Beschreibung an: „männliche Person, die nach Medizinstudium und klinischer Ausbildung die staatliche Zulassung (Approbation) erhalten hat, Kranke zu behandeln (Berufsbezeichnung)“. Demgegenüber hieß es im Duden Universalwörterbuch von 2015: „jemand, der nach Medizinstudium und klinischer Ausbildung die staatliche Zulassung (Approbation) erhalten hat, Kranke zu behandeln (Berufsbezeichnung)“
Der Aufsatz versucht, wieder einmal zu überprüfen, ob es das generische Maskulinum als geschlechtsübergreifende oder besser: geschlechtsneutrale Bezeichnung gibt oder ob es, nicht zuletzt aufgrund des jahrhundertelangen Einflusses des Patriarchats auf die Sprachentwicklung, sich doch um den Ausdruck männlicher Vormacht handelt. Dabei werden sowohl das Problem ‚Genus‘ als auch der Begriff ‚generisch‘ untersucht und auf seine Bezeichnungsfunktion hin getestet. Schließlich soll auch überprüft werden, ob die Sprache tatsächlich die Wirklichkeit widerspiegelt und ob die Sprache wirklich alle Phänomene in der außersprachlichen Realität benennen muss. Dazu kommt natürlich auch die Frage, ob die Duden-Redaktion ihre Kompetenzen richtig interpretiert (hat).
Der Begriff „Regelungsgegenstand“ meint ein gemeinschaftliches Problem, das entweder durch die Zentralmacht reguliert oder durch einen lokalen Akteur geregelt wird. Das vorliegende Papier bietet eine knappe Einführung in eine besonders konfliktträchtige, relative Subkategorie, die sogenannten „kritischen“ Regelungsgegenstände. Das Attribut „kritisch“ verweist dabei auf die Qualität der Konsequenzen, welche die Dysfunktionalität des Regelungssystems des Regelungsgegenstands nach sich ziehen kann.
Nach einer kurzen Vorstellung des Konzepts werden unter Bezugnahme auf die eingangs formulierten Charakteristika am Beispiel der antiken Metropole Antiocheia am Orontes je zwei Beispiele aus den Regelungsbereichen „Materielle Grundlagen“ (Lebensmittelversorgung; Wasserversorgung) und „Soziokulturelle Grundlagen“ (Pferderennen; Episkopat) identifiziert.
Zur Identifizierung kritischer Regelungsgegenstände eignen sich insbesondere zwei Anhaltspunkte:
1) Lokale Konflikte, die sich in öffentlichen Unmutsbekundungen, Gewaltausbrüchen oder restaurativen Interventionen der Zentralmacht manifestieren können;
2) Euergetische Handlungen, die auf das Generieren von sozialem Kapital abzielen, konzentrieren sich auf besonders wichtige oder populäre Regelungsgenstände.
Der Ausblick enthält ein Resümee und verweist auf transepochale und transdisziplinäre Übertragungsmöglichkeiten.
The present work builds on a conjugated electrochromic polymer with a highly transmissive and colorless bright state and its application in electrochromic devices. The main body of this work focuses on the investigation of the influence of moisture on electrochromic devices and solutions to overcome possible degradation of these devices due to moisture ingress.
Firstly, a series of EDOT derivatives with a terminal double bond in the lateral sidechain to potentially achieve a highly transmissive and fully colorless bright state was investigated. All of the EDOT derivatives were electrochemically polymerized and characterized by means of (in-situ) spectroelectrochemistry. The results highlight the dramatic influence of the terminal double bond on the improved visible light transmittance and color neutrality in the bright state. After detailed evaluation and comparison, the best performing compound, which contains a hexenyl sidechain (PEDOT-EthC6), was scaled-up by changing the deposition technique from an electrochemical to a chemical in-situ polymerization process on a R2R-pilot line in an industrially relevant environment. The R2R-processed PEDOTEthC6 half-cells were characterized in detail and provide enhanced electrochromic properties in terms of visible light transmittance and color neutrality in the bright state as well as short response times, improved contrast ratio, coloration efficiency and cycling stability (10 000 cycles).[21]
In a second step, the novel PEDOT-EthC6 electrochromic polymer was combined with a Prussian Blue counter electrode and a solid polymer electrolyte to form an all-solid-sate ECDs based on complementary switching electrodes and PET-ITO as flexible substrates. The fabricated ECDs were optically and spectroelectrochemically characterized. Excellent functionality of the S2S-processed flexible ECDs was maintained throughout 10 000 switching cycles under laboratory conditions. The ECDs offer enhanced electrochromic properties in terms of visible light transmittance change and color neutrality in the bright state as well as contrast ratio, coloration efficiency, cycling stability and fast response times. Furthermore, the final device assembly was transferred from a S2S-process to a continuous R2R-lamination process.[238]
In a third step, the PEDOT-EthC6/PB-based ECDs were submitted to conscious environmental aging tests. The emphasis of the research presented in this work, was mainly put at the influence of moisture and possible failure mechanisms regarding the PEDOT-EthC6/PB based ECDs. An intense brown coloration of the electrodes was observed while cycling the ECDs in humid atmospheres (90% rH) as a major degradation phenomenon. The brown coloration and a thereby accompanied loss of conductivity of the PET-ITO substrates was related to significant degradation of the ITO layers, inserted as the conductive layers in the flexible ECDs. A dissolution of the ITO thin films and formation of metallic indium particles on the surface of the ITO layers was observed that harmed the cycling stability enormously. The conductive layers of the aged ECDs were investigated by XRD, UV-Vis, SEM and spectroelectrochemical measurements and validated the supposed irreversible reduction of the ITO layers.[279]
In the absence of reasonable alternatives to PET-ITO for flexible (R2R-processed) ECDs, it is also important to investigate measures to avoid the degradation of ECDs. This is primarily associated with the avoidance of appropriate electrode potentials necessary for ITO reduction in humid atmospheres. As an intrinsic action point, the electrode potentials were investigated via electrochemical measurements in a three-electrode setup of an all-solid-state ECD. Extensive knowledge on the electrode potentials allowed the voltage-induced degradation of the ITO in flexible ECDs to be avoided through the implementation of an unbalanced electrode configuration (charge density ratio of working and counter electrode). It was possible to narrow the overall operational voltage window to an extent in which irreversible ITO reduction no longer occurs. The unbalanced electrode configuration lead to an improved cycling stability without harming other characteristics such as response time and light transmittance change and allows ECD operation in the presence of humidity.[279]
The avoidance of the mentioned degradation phenomena is further associated with appropriate sealing methods and materials as well as appropriate electrode and device fabrication processes. Since a variety of sealing materials is commercially available, due to the commercial launch of organic photovoltaic (OPV) and light emitting diodes (OLEDs), the focus in the present work was put to water-free electrode fabrication. As an extrinsic action point, a novel preparation method of a nanoscale PEDOT-EthC6 dispersion based on organic solvents is presented here in a final step. The water-free processing method gives access to straightforward printing and coating processes on flexible PET-ITO substrates and thus represents a promising and simplified alternative to the established PEDOT:PSS. The resulting nano-PEDOT-EthC6 thin films exhibit enhanced color neutrality and transmissivity in the bright state and are comparable to the properties of the in-situ polymerized PEDOT-EthC6 thin films.[280]
Im Zuge der Bemühungen um neue, tumorspezifische Therapieansätze für die Myelomerkrankung hat sich der C-X-C-Chemokinrezeptor 4 (CXCR4) aufgrund seiner zentralen Rolle in der Tumorgenese als vielversprechender Angriffspunkt hervorgetan. Im Sinne eines theranostischen Konzepts wird der Rezeptor mithilfe eines radioaktiv markierten Liganden quantifiziert und anschließend von rezeptorspezifischen Radiotherapeutika als Zielstruktur genutzt. Die CXCR4-Expression ist allerdings ein höchst dynamischer Prozess mit großer inter- und intraindividueller Heterogenität, der u.a. durch eine begleitende Chemotherapie beeinflusst werden kann. Ob sich therapieinduzierte Veränderungen der Rezeptorexpression gezielt nutzen lassen, um die CXCR4-Expression zu optimieren und so die Effektivität der CXCR4-gerichteten Strategien zu steigern, wurde bislang nicht untersucht.
Vor diesem Hintergrund wurden in der vorliegenden Arbeit verschiedene, in der Myelomtherapie etablierte Substanzen sowohl einzeln als auch in Kombination hinsichtlich ihres Einflusses auf die CXCR4-Expression von MM-Zelllinien und primären MM-Zellen unter in vitro Bedingungen analysiert.
In den durchgeführten Experimenten zeigte sich eine hohe Variabilität der CXCR4-Expression der MM-Zellen nach Therapieinduktion, die sich als substanz-, dosis- und zeitabhängig herausstellte. Die Ergebnisse bestätigten das große Potenzial der therapieinduzierten Modulation der CXCR4-Expression. Im weiteren Verlauf sind translationale Forschungsansätze gerechtfertigt, die die Übertragbarkeit der in vitro gewonnenen Ergebnisse auf die komplexen Vorgänge im lebenden Organismus überprüfen. Langfristiges Ziel ist der Entwurf eines patientenzentrierten, multimodalen Therapiekonzepts, welches das CXCR4-gerichtete theranostische Konzept mit einer individuell angepassten, medikamentösen MM-Therapie kombiniert.
One of the fascinating features of meiotic prophase I, is the highly conserved
vigorous movements of homologous chromosomes. These movements are
critical for the success of essential events as homologs alignment, synapsis and
recombination. Several organisms studied so far, including mammals, worms,
yeast and plants achieve these movements by anchoring the chromosome ends
to specialized sites in the nuclear envelope (NE). This attachment requires
telomere adaptor proteins which have to date been identified in fission yeast
and mice.
The mouse meiosis-specific telomere adaptor proteins TERB1, TERB2, and
MAJIN are involved in the attachment of ubiquitous shelterin telomere to the
LINC complex, in an analogous mechanism as those described in fission yeast.
Despite the essential role of meiosis-specific telomere adaptor proteins, the
precise mechanism of anchorage of telomeres to the nuclear envelope, as well
as their evolutionary history, are still not well understood. Therefore, the main
aim of this thesis is to investigate the organization of the mouse meiosis-specific
telomere adaptor complex TERB1-TERB2-MAJIN and its evolutionary history.
In the first part of this thesis high-resolution Structured Illumination Microscopy
(SIM), indirect immunofluorescence and Telo-FISH on mouse spermatocytes
were used to determine precisely how the telomere complex proteins are
localized with relation to the shelterin telomeric TRF1 protein and telomeric
DNA. During zygotene and pachytene stages staining patterns revealed
extensively overlapping of meiotic telomere complex proteins distributions in
which TERB2 organization is more heterogeneous than TERB1 and MAJIN at
the chromosome ends. Further, TRF1 localization was shown at the side of
lateral elements (LEs) ends with grasp-like distribution surrounding the TERB1
and MAJIN signals in zygotene and pachytene stages. Interestingly, telomeric
DNA was shown to be laterally distributed and partially overlapping with the
more central distribution displayed by meiotic telomere complex proteins of LEs
ends. The combination of these results allowed to describe an alternative model
of the telomere attachment to the NE during meiotic prophase I. The second part of this thesis, analyses mouse TERB1, TERB2, and MAJIN
evolutionary history. The lack of similarity between mouse and fission yeast
meiotic-specific telomere adaptor proteins has raised the question about the
origin of this specific complex through evolution. To identify mouse TERB1,
TERB2, and MAJIN putative orthologues, computational approaches and
phylogenetic analyses were performed. Besides, to test their potential function
during meiosis, expression studies were conducted. From these analyses, it was
revealed that mouse meiosis-specific telomere complex is ancient, as it
originated as early as eumetazoans pointing to a single origin. The absence of
any homologs in Nematoda and only a few candidates detected in Arthropoda
for meiosis-specific telomere complex, seemed, that these proteins have been
lost/replaced or highly diversified in these lineages. Remarkably, TERB1, TERB2,
and MAJIN protein domains involved in the formation of the complex as well as
those required for the interaction with the telomere shelterin protein and the
LINC complexes revealed high sequence similarity across all clades. Finally,
gene expression in the cnidarian Hydra Vulgaris provided evidence that the
TERB1-TERB2-MAJIN complex is selectively expressed in the germline
suggesting conservation of meiotic functions across metazoan evolution.
In summary, this thesis provides significant insights into the meiosis-specific
telomere complex mechanism to engage telomeres to the nuclear envelope and
the elucidation of its origin in metazoans.
Neben Alter, Geschlecht, Rauchen und genetischen Polymorphismen der metabolischen Enzyme können vor allem Arzneimittelinteraktionen die Pharmakokinetik und dynamik von Medikamenten beeinflussen und zu starken Unterschieden der Serumspiegelkonzentrationen führen. Eine im klinischen Alltag sehr häufig zu findende Arzneimittelkombination ist die von Antipsychotika und Antikonvulsiva. Trotz der häufigen gemeinsamen Gabe gibt es noch immer keine eindeutigen Daten über Interaktionen zwischen den beiden Klassen von Psychopharmaka und daraus resultierenden Veränderungen der jeweiligen Serumwirkspiegel. In der Arbeit werden Einflüsse von Alter und Geschlecht sowie mögliche Effekte antikonvulsiver Komedikation auf die mittels Therapeutischen Drug Monitorings gemessenen Serumwirkspiegel der Antipsychotika aufgezeigt. Genauer untersucht werden dabei die Kombinationen Clozapin und Valproat sowie Olanzapin und Valproat. Die Arbeit betont zudem die Bedeutung des Therapeutischen Drug Monitorings im klinischen Alltag.
Pyrrolizidinalkaloide (PA) sind sekundäre Pflanzenstoffe, welche über Nahrungsmittel in den menschlichen Organismus gelangen können. Zahlreiche Studien belegen, dass PA in der Leber verstoffwechselt und dabei in aktive genotoxische Metabolite umgewandelt werden. Diese verursachen vor allem in der Leber zelluläre Schäden, was sich klinisch in Form einer hepatischen venösen okklusiven Leberkrankheit, aber auch in der Entstehung von Tumoren zeigt. Die vorliegende Arbeit testet das genotoxische Potential der drei PA Lasiocarpin, Senecionin und Seneciphyllin anhand der Leberzelllinie Huh6 mit Hilfe des Mikrokerntests. Darüber hinaus wird die Wirkung von Lasiocarpin auf den intrazellulären Glutathion-Gehalt, die Superoxidproduktion und das mitochondriale Membranpotential analysiert. Zudem werden sowohl der eventuell negative Einfluss einer Glutathion Depletion, als auch die möglicherweise schützenden Effekte des pflanzlichen Antioxidans Delphinidin in Bezug auf die Genotoxizität von Lasiocarpin untersucht. Es konnte gezeigt werden, dass alle drei ausgewählten PA einen signifikanten Anstieg der Mikrokernfrequenz bewirken.Unsere Messungen zeigten für Lasiocarpin eine dezente Reduktion des Glutathion Gehalts. Dagegen führte eine Glutathion-Depletion in den Huh6 Zellen zu keiner Steigerung der Genotoxizität von Lasiocarpin. In Kombination mit dem Antioxidans Delphinidin zeigte sich für Lasiocarpin eine signifikante Reduktion der Mikrokernfrequenz. Abschließend ist anzumerken, dass in Zukunft vor allem die Wechselwirkung der PA untereinander und mit anderen (Pflanzen-)bestandteilen für eine verbesserte Risikoabschätzung der PA-Exposition untersucht werden sollte.
CD84 ist ein Transmembran-Glykoprotein vom Typ 1, welches ein Mitglied der Familie der SLAM 5 ist. Es ist ein homophiles Adhäsionsmolekül, das von unterschiedlichen Immunzellpopulationen exprimiert wird, einschließlich Monozyten, Makrophagen, Granulozyten, T-Zellen, dendritischen Zellen und Mastzellen, die im Zusammenhang mit der Entstehung der Atherosklerose stehen.
Um die funktionelle Bedeutung von CD84 in der Pathogenese der Atherosklerose bestimmen zu können, wurden CD84-/- Mäuse mit ApoE-/- Mäusen gekreuzt, um Tiere zu erhalten, die CD84-defizient und anfällig für Atherosklerose waren.
Die Bedeutung dieses Moleküls für die Atherogenese sowie für die Adhäsion, Transmigration, Immunzellrekrutierung allgemein und Integrinexpression und -aktivierung wurde im Mausmodell in vivo und in vitro untersucht.
Nach acht und 16 Wochen pro-atherogener, fettreicher Western-Type Diät waren die Ausprägung der atherosklerotischen Läsionen in der Aortenwurzel, sowie deren Gehalt an Makrophagen in den ApoE-/-.CD84-/- Tieren im Vergleich zu den ApoE-/- Kontrolltieren signifikant vermindert.
Weiter zeigten die Ergebnisse, dass die Viabilität von Makrophagen, denen CD84 fehlte, in vitro nicht verändert war.
Der Einfluss von CD84 auf die akute Immunzellrekrutierung wurde mittels verschiedener in vivo und in vitro Experimente untersucht. Eine verminderte Rekrutierung von CD84-/- proinflammatorischer Ly6Chigh Monozyten konnte in vivo im Rahmen des Hintergliedmaßen-Ischämiemodells festgestellt werden, nicht hingegen im Air pouch-Modell.
Es konnte weiterhin weder eine Veränderung der Adhäsion und chemotaktischen Transmigration von Monozyten in vitro noch der Immunzellrekrutierung in atherosklerotische Läsionen in Ldlr-/- Mäusen in vivo bei Abwesenheit von CD84 festgestellt werden.
Darüber hinaus konnte gezeigt werden, dass CD84 zwar keine Bedeutung für die Integri-nexpression auf Monozyten hat, jedoch für die adäquate Aktivierung von LFA-1 auf T-Zellen.
Diese Arbeit trägt summa summarum zum verbesserten Verständnis des Prozesses der Atherogenese sowie der funktionellen Bedeutung von CD84 innerhalb dieses Prozesses sowie im Rahmen der Immunzellrekrutierung und Integrinaktivierung bei.
Diese Erkenntnisse könnten in Zukunft dabei helfen, durch die Entwicklung neuer phar-makologischer Therapieansätze, spezifischer in von CD84 mitregulierte inflammatorische Prozesse, wie die Atherosklerose, einzugreifen.
Dies könnte im Zusammenspiel mit Forschungsarbeiten gelingen, die weitere präzise Erkenntnisse zu spezifischen funktionellen Eigenschaften von CD84 hinsichtlich der Im-munzellrekrutierung und Integrinaktivierung liefern.
π-Conjugated oligomers and polymers with tricoordinate boron centers incorporated into the main chain have attracted considerable attention as the interaction of the vacant p orbital on boron with an adjacent π system of the chain leads to conjugated materials with intriguing optical and electronic properties. This enables applicability in organic electronics and optoelectronics (OLEDs, OFETs, photovoltaics) or as sensory materials.
The potential of our B–C coupling protocol using metal-free catalytic Si/B exchange condensation is demonstrated by the synthesis of a series of π-conjugated monodisperse (het)aryl oligoboranes. Variation of the (het)aryl moieties allowed for tunability of the optoelectronic properties of the materials. Additionally, catalytic C–C cross-coupling strategies were applied to synthesize oligofuryl-based mono- and bisboranes, as well as polymers. These studies led to very robust and highly emissive compounds (f up to 97 %), which allow for tuning of their emission color from blue to orange. Furthermore, this work includes investigations of reaction routes to a kinetically stabilized tetraoxaporphyrinogen.
Being a key aspect of this work, a full investigation of the mechanism of the catalytic Si/B exchange was carried out. Additionally, this work presents the use of borenium cations to perform B–C coupling via intermolecular electrophilic borylation. Similar to the Si/B exchange, this route is capable of giving access to diaryl(bromo)boranes.
Prävention ist der zentrale Hebel, um dem gesundheitspolitischen und sozialen Problem der Alzheimer-Krankheit (AD) zu begegnen. Ein Ansatz ist der Einsatz krankheitsmodifizierender Therapien in der präklinischen Krankheitsphase. Zwei mögliche Früherkennungsmethoden sind die somatosensibel evozierten Potenziale des Nervus vagus (VSEP) und das Aufmerksamkeitsblinzeln (Attentional Blink, ATB). Beide werden möglicherweise über den Locus coeruleus vermittelt, der sehr früh im Krankheitsverlauf strukturelle Veränderungen aufweist.
Ziel der Arbeit war es, Korrelationen zwischen den Parametern beider Methoden zu untersuchen. Hierfür mussten individuumspezifische ATB-Parameter entwickelt werden. Außerdem wurden Korrelationen mit psychometrischen Parametern der Demenzdiagnostik und Gruppenunterschiede zwischen Personen mit und ohne Mild Cognitive Impairment (MCI) analysiert.
Es wurden insgesamt 108 Teilnehmer der „Vogel-Studie“, einer prospektiven Längsschnittstudie zur Frühdiagnostik dementieller Erkrankungen, untersucht. Die VSEP wurden mittels der durch Fallgatter et al. (2003) entwickelten Technik bestimmt. Die ATB-Messung erfolgte in einem an Zylberberg et al. (2012) angelehnten Versuchsablauf.
Die gemessenen Parameter siedelten sich zwischen dem aus der Literatur bekannten Wertebereich gesunder und an Alzheimer-Demenz erkrankter Probanden an. Auffallend war das Auftreten von Attentional Masking Errors (AME), die bisher ausschließlich bei Patienten mit Alzheimer- und Lewy-Body-Demenz beschrieben wurden. Somit sprechen die Ergebnisse für eine beginnende Alzheimer-Pathologie im untersuchten Studienkollektiv. Es konnten keine signifikanten Korrelationen zwischen VSEP- und ATB-Parametern nachgewiesen werden. Die explorative Analyse weist auf zahlreiche Zusammenhänge zwischen ATB-Parametern und psychometrischen Tests hin. 16 % der Probanden erfüllten die Kriterien eines MCI (Portet et al., 2006). Wie in der vorbestehenden Literatur ergaben sich auch in dieser Arbeit keine signifikanten Gruppenunterschiede zwischen Probanden mit und ohne MCI.
Die Ergebnisse dieser Arbeit unterstützen die bestehende Evidenz dahingehend, dass beide Methoden frühe subklinische Alzheimer-Pathologien detektieren könnten. Insbesondere AME scheinen ein vielversprechender Parameter zu sein. Weiterführende Ergebnisse zum Vorhersagewert der einzelnen Parameter wird das Follow-Up der „Vogel-Studie“ erbringen.
Die Multiple Sklerose ist eine bisher nicht heilbare, chronisch-inflammatorische demyelinisierende Erkrankung des zentralen Nervensystems. Trotz intensiver Forschungsbemühungen ist der exakte Pathomechanismus nicht vollkommen verstanden. Klar ist jedoch, dass der Blut-Hirn-Schranke eine entscheidende Rolle bei der Pathogenese zukommt. Seit Februar 2014 ist mit Dimethylfumarat ein neues orales Medikament für die schubförmige Multiple Sklerose zugelassen. Die Wirkungen von Fumarsäureestern auf humane zerebrale Endothelzellen als Grundsteine der Blut-Hirn-Schranke sind allerdings nur unzureichend untersucht.
Mehrere Forschungsgruppen demonstrierten an humanem Nabelschnurvenenendothel einen hemmenden Effekt von Fumarsäureestern auf die Adhäsion von Leukozyten und beschrieben eine Inhibition der Aktivierung des proinflammatorischen Transkriptionsfaktors NFB in den Endothelzellen. Aufgrund der charakteristischen Eigenschaften zerebralen Endothels ist eine Übertragung dieser Beobachtungen auf die Blut-Hirn-Schranke allerdings nicht ohne weiteres möglich. Daher galt es potentielle Effekte von Fumarsäureestern auf primäre humane zerebrale Endothelzellen als in vitro Modell der Blut-Hirn-Schranke zu überprüfen. Dabei wurden die Zellen nicht nur unter ruhenden Bedingungen, sondern auch unter inflammatorischer Stimulation mit TNF-α, IL-1 und IFN untersucht, einem Milieu, wie es in inflammatorischen MS Läsionen zu finden ist. In Leukozyten-Adhäsionsassays konnte durch Inkubation mit Monomethylfumarat und Dimethylfumarat keine funktionale Beeinflussung der Adhäsion von T-Lymphozyten an den verwendeten zerebralen Endothelzellen verzeichnet werden. Kongruent dazu fand sich in durchflusszytometrischen Analysen keine Hemmung der inflammatorisch vermittelten Expression des Adhäsionsmoleküls ICAM-1, welches eine tragende Rolle bei der Leukozytenmigration spielt. Inflammatorische intrazelluläre Signalwege, wie die NFB-Kerntranslokation oder die Phosphorylierung von p38 wurden in HECE im Gegensatz zu HUVEC durch Fumarsäureester ebenso wenig beeinflusst.
Diese in sich konsistenten Ergebnisse führen zu der Schlussfolgerung, dass im Gegensatz zu anderen Gefäßbetten weder Dimethylfumarat noch Monomethylfumarat direkt am zerebralen Endothel anti-inflammatorisch wirken.
The present thesis deals with optimisation problems with sparsity terms, either in the constraints which lead to cardinality-constrained problems or in the objective function which in turn lead to sparse optimisation problems. One of the primary aims of this work is to extend the so-called sequential optimality conditions to these two classes of problems. In recent years sequential optimality conditions have become increasingly popular in the realm of standard nonlinear programming. In contrast to the more well-known Karush-Kuhn-Tucker condition, they are genuine optimality conditions in the sense that every local minimiser satisfies these conditions without any further assumption. Lately they have also been extended to mathematical programmes with complementarity constraints. At around the same time it was also shown that optimisation problems with sparsity terms can be reformulated into problems which possess similar structures to mathematical programmes with complementarity constraints. These recent developments have become the impetus of the present work. But rather than working with the aforementioned reformulations which involve an artifical variable we shall first directly look at the problems themselves and derive sequential optimality conditions which are independent of any artificial variable. Afterwards we shall derive the weakest constraint qualifications associated with these conditions which relate them to the Karush-Kuhn-Tucker-type conditions. Another equally important aim of this work is to then consider the practicability of the derived sequential optimality conditions. The previously mentioned reformulations open up the possibilities to adapt methods which have been proven successful to handle mathematical programmes with complementarity constraints. We will show that the safeguarded augmented Lagrangian method and some regularisation methods may generate a point satisfying the derived conditions.
Die Implantation einer Endoprothese des Kniegelenks ist eine der häufigsten Operationen in Deutschland. In vielen Kliniken wurde im Rahmen des Patient-Blood-Managements ein präoperatives Anämie Screening eingeführt, um mit Hilfe einer präoperativen Anämiebehandlung das Risiko für eine Bluttransfusion und andere Anämie assoziierte Risiken zu senken. In den bisherigen Publikationen variieren jedoch sowohl die Anämieprävalenz als auch das Bluttransfusionsrisiko erheblich, weshalb für eine gezielte Behandlung eine klinikspezifische Analyse von Risikofaktoren für eine Bluttransfusion erforderlich zu sein scheint. Ziel dieser retrospektiven, monozentrischen Studie war es, den Einfluss der präoperativen Anämie und weiterer möglicher Einflussfaktoren auf die Transfusions- und Komplikationsrate nach Implantation einer primären Endoprothese des Kniegelenks zu untersuchen. Hierfür wurden die Daten von 1811 Patienten, welche zwischen 2016 und 2018 eine primäre Endoprothese des Kniegelenks erhalten hatten, erfasst und statistisch ausgewertet. 7,4% der Patienten besaßen eine präoperative Anämie. Patienten mit einer präoperativen Anämie erhielten in 25,4% ein Erythrozytenkonzentrat, nicht anämische Patienten in 3,1% der Fälle. Die gesamte Transfusionsrate für allogene Transfusionen lag bei 4,7%. In der Gruppe der präoperativ anämischen Patienten zeigte sich dabei ein signifikant höhere Wahrscheinlichkeit für die Transfusion eines Erythrozytenkonzentrats, nicht jedoch für eine Komplikation während des stationären Aufenthalts. Unsere logistischen Regressionsmodelle ermittelten neben der präoperativen Hämoglobinkonzentration auch die systemische Tranexamsäuregabe als unabhängigen Einflussfaktor für Transfusionen. Das Geschlecht der Patienten hatte hingegen keinen Einfluss auf das Risiko für eine Bluttransfusion. Männer und Frauen mit der gleichen präoperativen Hämoglobinkonzentration besaßen in etwa das gleiche individuelle Risiko für eine Bluttransfusion. Entsprechend erscheint es sinnvoll, für Männer und Frauen die gleiche präoperative Mindesthämoglobinkonzentration anzustreben. Da jedoch das individuelle Risiko auch durch die systemische Gabe der Tranexamsäure beeinflusst wird, könnte dies bei der Festlegung einer Schwelle zur präoperativen Behandlung berücksichtigt werden.
The Stiff-person syndrome (SPS) is a rare autoimmune disease that is characterized by symptoms including stiffness in axial and limb muscles as well as painful spasms. Different variants of SPS are known ranging from moderate forms like the stiff-limb syndrome to the most severe form progressive encephalomyelitis with rigidity and myoclonus (PERM). SPS is elicited by autoantibodies that target different pre- or postsynaptic proteins. The focus of the present work is on autoantibodies against the glycine receptor (GlyR). At start of the present thesis, as main characteristic of the GlyR autoantibody pathology, receptor cross-linking followed by enhanced receptor internalization and degradation via the lysosomal pathway was described. If binding of autoantibodies modulates GlyR function and therefore contributes to the GlyR autoantibody pathology has not yet been investigated. Moreover, not all patients respond well to plasmapheresis or other treatments used in the clinic. Relapses with even higher autoantibody titers regularly occur.
In the present work, further insights into the disease pathology of GlyRα autoantibodies were achieved. We identified a common GlyRα1 autoantibody epitope located in the far N-terminus including amino acids A1-G34 which at least represent a part of the autoantibody epitope. This part of the receptor is easily accessible for autoantibodies due to its location at the outermost surface of the GlyRα1 extracellular domain. It was further investigated if the glycosylation status of the GlyR interferes with autoantibody binding. Using a GlyRα1 de-glycosylation mutant exhibited that patient autoantibodies are able to detect the de-glycosylated GlyRα1 variant as well. The direct modulation of the GlyR analyzed by electrophysiological recordings demonstrated functional alterations of the GlyR upon autoantibody binding. Whole cell patch clamp recordings revealed that autoantibodies decreased the glycine potency, shown by increased EC50 values. Furthermore, an influence on the desensitization behavior of the receptor was shown. The GlyR autoantibodies, however, had no impact on the binding affinity of glycine. These issues can be explained by the localization of the GlyR autoantibody epitope. The determined epitope has been exhibited to influence GlyR desensitization upon binding of allosteric modulators and differs from the orthosteric binding site for glycine, which is localized much deeper in the structure at the interface between two adjacent subunits. To neutralize GlyR autoantibodies, two different methods have been carried out. Transfected HEK293 cells expressing GlyRα1 and ELISA plates coated with the GlyRα1 extracellular domain were used to efficiently neutralize the autoantibodies. Finally, the successful passive transfer of GlyRα1 autoantibodies into zebrafish larvae and mice was shown. The autoantibodies detected their target in spinal cord and brain regions rich in GlyRs of zebrafish and mice. A passive transfer of human GlyRα autoantibodies to zebrafish larvae generated an impaired escape behavior in the animals compatible with the abnormal startle response in SPS or PERM patients.
Attention-deficit/hyperactivity disorder (ADHD) is a neurodevelopmental disorder with an estimated heritability of around 70%. In order to fully understand ADHD biology it is necessary to incorporate multiple different types of research. In this thesis, both human and animal model research is described as both lines of research are required to elucidate the aetiology of ADHD and development new treatments. The role of a single gene, Adhesion G protein-coupled receptor L3 (ADGRL3) was investigated using a knockout mouse model. ADGRL3 has putative roles in neuronal migration and synapse function. Various polymorphisms in ADGRL3 have been linked with an increased risk of attention deficit/hyperactivity disorder (ADHD) in human studies. Adgrl3-deficient mice were examined across multiple behavioural domains related to ADHD: locomotive activity, visuospatial and recognition memory, gait impulsivity, aggression, sociability and anxiety-like behaviour. The transcriptomic alterations caused by Adgrl3-depletion were analysed by RNA-sequencing of three ADHD-relevant brain regions: prefrontal cortex (PFC), hippocampus and striatum. Increased locomotive activity in Adgrl3-/- mice was observed across all tests with the specific gait analysis revealing subtle gait abnormalities. Spatial memory and learning domains were also impaired in these mice. Increased levels of impulsivity and sociability accompanying decreased aggression were also detected. None of these alterations were observed in Adgrl3+/- mice. The numbers of genes found to exhibit differential expression was relatively small in all brain regions sequenced. The absence of large scale gene expression dysregulation indicates a specific pathway of action, rather than a broad neurobiological perturbation. The PFC had the greatest number of differentially expressed genes and gene-set analysis of differential expression in this brain region detected a number of ADHD-relevant pathways including dopaminergic synapses as well as cocaine and amphetamine addiction. The most dysregulated gene in the PFC was Slc6a3 which codes for the dopamine transporter, a molecule vital to current pharmacological treatment of ADHD. The behavioural and transcriptomic results described in this thesis further validate Adgrl3 constitutive knockout mice as an experimental model of ADHD and provide neuroanatomical targets for future studies involving ADGRL3 modified animal models.
The study of ADHD risk genes such as ADGRL3 requires the gene to be first identified using human studies. These studies may be genome based such as genome wide association studies (GWAS) or transcriptome based using microarray or RNA sequencing technology. To explore ADHD biology in humans the research described in this thesis includes both GWAS and trancriptomic data. A two-step transcriptome profiling was performed in peripheral blood mononuclear cells (PBMCs) of 143 ADHD subjects and 169 healthy controls. We combined GWAS and expression data in an expression-based Polygenic Risk Score (PRS) analysis in a total sample of 879 ADHD cases and 1919 controls from three different datasets. Through this exploratory study we found eight differentially expressed genes in ADHD and no support for the genetic background of the disorder playing a role in the aberrant expression levels identified. These results highlight promising candidate genes and gene pathways for ADHD and support the use of peripheral tissues to assess gene expression signatures for ADHD.
This thesis illustrates how both human and animal model research is required to increase our understanding of ADHD. The animal models provide biological insight into the targets identified in human studies and may themselves provide further relevant gene targets. Only by combining research from disparate sources can we develop the thorough understanding on ADHD biology required for treatment development, which is the ultimate goal of translational science research.
Main objectives of the present dissertation can be divided in two parts. The first part deals with setting up a spectroscopic technique for reliable and accurate measurements of the two-photon absorption (2PA) cross section spectra. In the second part, this firmly established experimental technique together with conventional spectroscopic characterization, quantum-chemical computations and theoretical modelling calculations was combined and therefore used as a tool to gain information for the so-called structure-property relationship through several molecular compounds.
Using Photoionization to Investigate Reactive Boron Species and the Kinetics of Hydrocarbon Radicals

This thesis highlights the importance of isomer-selective approaches for the complete analysis of chemical processes. The method of choice is photoelectron/photoion coincidence spectroscopy, which allows simultaneous detection of electrons and ions coming from a single ionization event. Ionization techniques are sensitive and can record multiple species simultaneously, rendering them ideal tools to probe molecular transformations. Coupling these setups to synchrotron radiation allows one to analyze complex mixtures with isomer selectivity, based on ionization energies and vibrational structure in the cation, without any prior separation steps. Only few setups exist that can be used to gather these data, although their impact and applicability is growing steadily in various fields. For closed-shell species an easier and more widely used method is gas-chromatography, but most open shell species would not survive the separation process. Due to the reactivity of radicals they have to be created by selectively converting stable precursor molecules. Depending on the radical generation method different properties can be investigated ranging from thermodynamic data, over concentrations in high temperature environments, to chemical kinetics.
The first part of this thesis deals with the determination of bimolecular rate constants. Isomeric hydrocarbon radicals were generated by a high intense UV light pulses and their kinetics with oxygen was measured. The pressure dependence of different isomers in the falloff region was compared to theoretical models, and their reactivity could be explained.
The second part deals with boron containing compounds in various electronic situations. The corresponding precursors were successfully synthesized or could be bought. They were subjected to fluorine atoms in chemical reactors or destroyed pyrolytically at high temperatures. Most investigated species exhibited vibronic effects that could be elucidated using high level computations.
Die Schlaganfallnachsorge in Deutschland wird von verschiedenen Leistungserbringern geprägt, die teilweise komplementäre und komplexe Dienstleistungen erbringen und sektorenübergreifend arbeiten. In Bad Neustadt wurde in Kooperation mit der Universität Würzburg und dem Zentrum für Telemedizin Bad Kissingen das Stroke Manager Programm entwickelt und evaluiert. Das strukturierte Nachsorgeprogramm Stroke Manager basiert auf einer standardisierten Informations- und Software Unterstützung von der Akutversorgung bis drei Monate nach Entlassung aus der stationären Versorgung.
Anhand der Ergebnisse des Stroke Manager Programms konnte eine vergleichsweise hohe Persistenz bzgl. der stationär verordneten medikamentösen Sekundärprävention über einen Zeitraum von drei Monaten festgestellt werden, ebenso konnten wir nachweisen, dass sich das Programm positiv auf die Versorgungsqualität sowie die Patientenzufriedenheit nach Schlaganfall auswirken kann. Die im Stroke Manager-Programm betreuten Schlaganfallpatienten wiesen im Vergleich signifikante Unterschiede bei den Faktoren Rauchverhalten, Schlaganfallschweregrad und subjektive, globale Lebensqualität auf.
Sicherheit und Effektivität von Amisulprid zur Therapie von Übelkeit und Erbrechen nach Narkosen

PONV stellt mit einer Inzidenz von circa 30 % ein relevantes Problem in der postoperativen Phase dar. Amisulprid ist ein atypisches Neuroleptikum, das zur Behandlung der Schizophrenie verwendet wird und gehört zur Gruppe der D2-Rezeptor-Antagonisten. Hierüber wirkt Amisulprid antiemetisch.
Ziel der Studie war, die Sicherheit und Effektivität einer intravenösen Einmalgabe von Amisulprid in den Dosierungen 5 mg und 10 mg zur Therapie von PONV, nach Versagen einer PONV-Prophylaxe, untereinander und gegenüber einem Placebo zu vergleichen. Es handelte sich hierbei um eine multizentrische, randomisierte, doppelblinde Phase-III-Studie.
Amisulprid zeigte ein benignes Nebenwirkungsprofil. Amisulprid zeigte in der Dosierung 10 mg eine statistisch signifikante Überlegenheit gegenüber Placebo sowohl zur Therapie von PONV, als auch im Auftreten signifikanter Übelkeit und im Gebrauch einer Rescue-Medikation.
Eine weitere interessante Fragestellung wäre der Vergleich der Sicherheit und Effektivität von Amisulprid mit einem weiteren Antiemetikum.
GDF-15 wird seit wenigen Jahren als prognostischer und prädiktiver Marker in der Tumortherapie diskutiert. Diese Pilotstudie sollte erstmals GDF-15 bei Patienten mit HER2/neu positivem Mammakarzinom im frühen Stadium im klinischen Verlauf untersuchen. Dazu wurden 22 Patienten rekrutiert und die GDF-15-Spiegel mittels ELISA vor und während einer Antikörpertherapie bestimmt. Um GDF-15 als prädiktiven Marker zu testen, wurde nach neoadjuvanter Therapie und anschließender Operation der Regressionsgrad nach Sinn bewertet. In der untersuchten Kohorte wurde ein medianer GDF 15-Spiegel von 0,33 ng/ml ermittelt. Im Therapieverlauf kam es zu keiner signifikanten Veränderung des Spiegels. Höhere GDF-15-Spiegel konnten allerdings bei größeren Tumoren und bei einem höheren BMI analysiert werden. Ebenfalls konnten wir zeigen, dass der GDF-15-Spiegel signifikant mit dem Alter steigt. Nicht signifikant, aber von Bedeutung ist der Zusammenhang zwischen GDF-15 und dem Regressionsgrad nach Sinn. Die untersuchten Patienten wiesen tendenziell höhere GDF-15-Werte bei niedrigem Regressionsgrad auf. Ein schlechteres Ansprechen auf eine Antikörpertherapie bei höheren GDF 15-Spiegeln ist somit anzunehmen.
Bacterial small RNAs are key mediators of post-transcriptional gene regulation. An increasing number of sRNAs have been implicated in the regulation of virulence programs of pathogenic bacteria. Recently, in the enteric pathogen Salmonella Typhimurium, the PinT sRNA has gained increased importance as it is the most upregulated sRNA as Salmonella infects mammalian host cells (Westermann et al., 2016). PinT acts as a temporal regulator of Salmonella‘s two major pathogenicity islands, SPI-1 and SPI-2 (Kim et al., 2019; Westermann et al., 2016). However, the complete set of PinT targets, its role in Salmonella infection and host response is not yet fully understood. Building on the MS2 affinity purification and RNA- seq (MAPS) method (Lalaouna et al., 2015), we here set out to globally identify direct RNA ligands of PinT, relevant to Salmonella infection. We transferred the classical MAPS technique, based on sRNA-bait overexpression, to more physiological conditions, using endogenous levels of the sRNA. Making the henceforth identified targets, less likely to represent artefacts of the overexpression. More importantly, we progressed the MAPS technique to in vivo settings and by doing so, we were able pull-down bacterial RNA transcripts bound by PinT during macrophage infection. While we validate previously known PinT targets, our integrated data revealed novel virulence relevant target. These included mRNAs for the SPI-2 effector SteC, the PhoQ activator UgtL and the 30S ribosomal protein S22 RpsV. Next, we follow up on SteC, the best characterized virulence relevant PinT target. Using genetic and biochemical assays, we demonstrate that PinT represses steC mRNA by direct base-pairing and translational interference. PinT-mediated regulation of SteC leads to alterations in the host response to Salmonella infection. This regulation impacts the cytokine response of infected macrophages, by altering IL10 production, and possibly driving the macrophages to an anti-inflammatory state, more permise to infection. SteC is responsible for F-actin meshwork rearrangements around the SCV (Poh et al., 2008). Here we demonstrate that PinT-mediated regulation of SteC, impacts the formation of this actin meshwork in infected cells. Our results demonstrate that SteC expression is very tightly regulated by PinT in two layers; indirectly, by repressing ssrB and crp; and directly by binding to steC 5’UTR. PinT contributes to post-transcriptional cross-talk between invasion and intracellular replication programs of Salmonella, by controlling the expression of both SPI-1 and SPI-2 genes (directly and indirectly). Together, our collective data makes PinT the first sRNA in Gram-negatives with a pervasive role in virulence, at the center of Salmonella virulence programs and provide molecular input that could help explain the attenuation of pinT-deficient Salmonella strains in whole animal models of infection.
In light of the rapidly increasing global demand of energy and the negative effects of climate change, innovative solutions that allow an efficient transition to a carbon-neutral economy are urgently needed. In this context, artificial photosynthesis is emerging as a promising technology to enable the storage of the fluctuating energy of sunlight in chemical bonds of transportable “solar fuels”. Thus, in recent years much efforts have been devoted to the development of robust water oxidation catalysts (WOCs) leading to the discovery of the highly reactive Ru(bda) (bda: 2,2’-bipyridine-6,6’-dicarboxylic acid) catalyst family. The aim of this thesis was the study of chemical and photocatalytic water oxidation with functionalized Ruthenium macrocycles to explore the impact of substituents on molecular properties and catalytic activities of trinuclear macrocyclic Ru(bda) catalysts. A further objective of this thesis comprises the elucidation of factors that influence the light-driven water oxidation process with this novel class of supramolecular WOCs.
Die vorliegende Arbeit behandelt im ersten Abschnitt die Synthese und Reaktivität neuartiger Diborane(4). Ebenfalls wurde die Reaktivität von Dihalogendiboranen(4) gegenüber Phenylazid untersucht, wobei symmetrische Vertreter unter Beibehalt der B-B-Bindung die fünfgliedrigen B2N3 Heterocyclen 14 und 15 lieferten. Der zweite Abschnitt dieser Arbeit beschäftigt sich mit der unerwarteten Reaktivität der NHC-stabilisierten μ-Hydridodiborane(5) XXIII und XXIV. Der abschließende Teil dieser Arbeit befasst sich mit den ersten Versuchen zur Darstellung eines CAAC-stabilisierten, Diboranyl-substituierten Borylens.
Chapter 1 deals with the reaction of [Rh(acac)(PMe3)2] with para-substituted 1,4-diphenylbuta-1,3-diynes at room temperature, in which a complex containing a bidentate organic fulvene moiety, composed of two diynes, σ-bound to the rhodium center is formed in an all-carbon [3+2] type cyclization reaction. In addition, a complex containing an organic indene moiety, composed of three diynes, attached to the rhodium center in a bis-σ-manner is formed in a [3+2+3] cyclization process.
Reactions at 100 °C reveal that the third diyne inserts between the rhodium center and the bis-σ-bound organic fulvene moiety. Furthermore, the formation of a 2,5- and a 2,4-bis(arylethynyl)rhodacyclopentadiene is observed. The unique [3+2] cyclization product was used for the synthesis of a highly conjugated organic molecule, which is hard to access or even inaccessible by conventional methods. Thus, at elevated temperatures, reaction of the [3+2] product with para-tolyl isocyanate led to the formation of a purple organic compound containing the organic fulvene structure and one equivalent of para-tolyl isocyanate.
The blue and green [3+2+3] complexes show an unusually broad absorption from 500 – 1000 nm with extinction coefficients ε of up to 11000 M-1 cm-1. The purple organic molecule shows an absorption spectrum similar to those of known diketopyrrolopyrroles.
Additionally, the reaction of [Rh(acac)(PMe3)2] with para-tolyl isocyanate was investigated. A cis-phosphine complex of the form cis-[Rh(acac)(PMe3)2(isocyanate)2] with an isocyanate dimer bound to the rhodium center by one carbon and one oxygen atom was isolated.
Replacing the trimethylphosphine ligands in [Rh(acac)(PMe3)2] with the stronger σ-donating NHC ligand Me2Im (1,3-dimethylimidazolin-2-ylidene), again, drastically alters the reaction. Similar [3+2] and [3+2+3] products to those discussed above could not be unambiguously assigned, but cis- and trans-π-complexes, which are in an equilibrium with the two starting materials, were formed.
Chapters 2 is about the influence of the backbone of the α,ω-diynes on the formation and photophysical properties of 2,5-bis(aryl)rhodacyclopentadienes. Therefore, different α,ω-diynes were reacted with [Rh(acac)(PMe3)2] and [Rh(acac)(P(p-tolyl)3)2] in equimolar amounts. In general, a faster consumption of the rhodium(I) starting material is observed while using preorganized α,ω-diynes with electron withdrawing substituents in the backbone. The isolated PMe3-substituted rhodacyclopentadienes exhibit fluorescence, despite the presence of the heavy atom rhodium, with lifetimes τF of < 1 ns and photoluminescence quantum yields Φ of < 0.01 as in previously reported P(p-tolyl)-substituted 2,5-bis(arylethynyl)rhodacyclopentadienes. However, an isolated P(p-tolyl)-substituted 2,5-bis(aryl)rhodacyclopentadiene shows multiple lifetimes and different absorption and excitation spectra leading to the conclusion that different species may be present.
Reaction of [Rh(acac)(Me2Im)2] with dimethyl 4,4'-(naphthalene-1,8-diylbis(ethyne-2,1-diyl))dibenzoate, results in the formation of a mixture trans- and cis-NHC-substituted 2,5-bis(aryl)rhodacyclopentadienes.
In chapter 3 the reaction of various acac- and diethyldithiocarbamate-substituted rhodium(I) catalysts bearing (chelating)phosphines with α,ω-bis(arylethynyl)alkanes (α,ω-diynes), yielding luminescent dimers and trimers, is described. The photophysical properties of dimers and trimers of the α,ω-diynes were investigated and compared to para-terphenyl, showing a lower quantum yield and a larger apparent Stokes shift.
Furthermore, a bimetallic rhodium(I) complex of the form [Rh2(ox)(P(p-tolyl)3)4] (ox: oxalate) was reacted with a CO2Me-substituted α,ω-tetrayne forming a complex in which only one rhodium(I) center reacts with the α,ω-tetrayne. The photophysical properties of this mixed rhodium(I)/(III) species shows only negligible differences compared to the P(p-tolyl)- and CO2Me-substituted 2,5-bis(arylethynyl)rhodacyclopentadiene, previously synthesized by Marder and co-workers.
Hormones are essential components in the body and their imbalance leads to pathological consequences. T2DM, insulin resistance and obesity are the most commonly occurring lifestyle diseases in the past decade. Also, an increased cancer incidence has been strongly associated with obese and T2DM patients.
Therefore, our aim was to study the influence of high insulin levels in accumulating DNA damage in in vitro models and patients, through the induction of oxidative stress. The primary goal of this study was to analyze the genotoxicity induced by the combined action of two endogenous hormones (insulin and adrenaline) with in vitro models, through the induction of micronuclei and to see if they cause an additive increase in genomic damage. This is important for multifactorial diseases having high levels of more than one hormone, such as metabolic syndrome and conditions with multiple pathologies (e.g., T2DM along with high stress levels).
Furthermore, the combination of insulin and the pharmacological inhibition of the tumor suppressor gene: PTEN, was to be tested in in vitro models for their genotoxic effect and oxidative stress inducing potential. As the tumor suppressor gene: PTEN is downregulated in PTEN associated syndromes and when presented along with T2DM and insulin resistance, this may increase the potential to accumulate genomic damage.
The consequences of insulin action were to be further elucidated by following GFP-expressing cells in live cell-imaging to observe the ability of insulin, to induce micronuclei and replicative stress. Finally, the detrimental potential of high insulin levels in obese patients with hyperinsulinemia and pre-diabetes was to be studied by analyzing markers of oxidative stress and genomic damage. In summary, the intention of this work was to understand the effects of high insulin levels in in vitro and in patients to understand its relevance for the development of genomic instability and thus an elevated cancer risk.
Es sollten Checkpoint inhibierende anti-TNFRSF Rezeptor Antikörper-Fusionsproteine hergestellt und charakterisiert werden. Die agonistische Aktivität TNFR-spezifischer Antikörper wird maßgeblich durch eine Immobilisation über Fcγ-Rezeptoren beeinflusst. In dieser Arbeit erfolgte die Immobilisation der Antikörper-Fusionsproteine über den PD-L1. In funktionellen Assays konnte eine Aktivitätssteigerung der TNFR-spezifischen Domänen mittels PD-L1 vermittelter Immobilisation gezeigt werden.
L-type voltage-gated calcium channels (LTCC) are heteromultimeric membrane proteins that allow Ca2+ entry into the cell upon plasma membrane depolarization. The β subunit of voltage-dependent calcium channels (Cavβ) binds to the α-interaction domain in the pore-forming α1 subunit and regulates the trafficking and biophysical properties of these channels. Of the four Cavβ isoforms, Cavβ2 is predominantly expressed in cardiomyocytes. This subunit associates with diverse proteins besides LTCC, but the molecular composition of the Cavβ2 nanoenvironments in cardiomyocytes is yet unresolved. Here, we used a protein-labeling technique in living cells based on an engineered ascorbate peroxidase 2 (APEX2). In this strategy, Cavβ2b was fused to APEX2 and expressed in adult rat cardiomyocytes using an adenovirus system. Nearby proteins covalently labeled with biotin-phenol were purified using streptavidin-coated beads and identified by mass spectrometry (MS). Analysis of the in situ APEX2-based biotin labeling by MS revealed 61 proteins located in the nanoenvironments of Cavβ2b, with a high specificity and consistency in all the replicates. These proteins are involved in diverse cellular functions such as cellular trafficking, sarcomere organization and excitation-contraction coupling. Among these proteins, we demonstrated an interaction between the ryanodine receptor 2 (RyR2) and Cavβ2b, probably coupling LTCC and the RyR2 into a supramolecular complex at the dyads. This interaction is mediated by the Src homology 3 (SH3) domain of Cavβ2b and is necessary for an effective pacing frequency‐dependent increase in Ca2+-induced Ca2+ release in cardiomyocytes.
Das Bullöse Pemphigoid (BP) ist eine blasenbildende Autoimmunerkrankung der Haut, die durch subepidermale Blasenbildung und Antikörper (AK) gegen bestimmte hemidesmosomale Proteine der Basalmembran (BM) charakterisiert ist. Zielantigene sind BP180 und BP230. Im Fokus dieser Arbeit stand die retrospektive Identifikation und Datenerhebung von Patienten mit BP, die in der Dermatologie der Uniklinik Würzburg behandelt wurden. Zudem wurde eine Kontrollgruppe aus Patienten mit Basalzellkarzinom etabliert. Es konnten (hoch-)signifikante Assoziationen zwischen dem BP und verschiedenen Laborparametern (u.a. Leukozytose, Eosinophilie, Thrombozytose, Anämie, Kreatinin erhöht) sowie Erkrankungen (u.a. neurologische Erkrankungen (Schlaganfall, Demenz, MP, MS und Epilepsie) sowie psychiatrischen Erkrankungen (HOPS, Depression) und Diabetes mellitus) nachgewiesen werden.
Tree species diversity is important to maintain saproxylic beetle diversity in managed forests. Yet, knowledge about the conservational importance of single tree species and implications for forest management and conservation practices are lacking.
We exposed freshly cut branch‐bundles of 42 tree species, representing tree species native and non‐native to Europe, under sun‐exposed and shaded conditions for 1 year. Afterwards, communities of saproxylic beetles were reared ex situ for 2 years. We tested for the impact of tree species and sun exposure on alpha‐, beta‐, and gamma‐diversity as well as composition of saproxylic beetle communities. Furthermore, the number of colonised tree species by each saproxylic beetle species was determined.
Tree species had a lower impact on saproxylic beetle communities compared to sun exposure. The diversity of saproxylic beetles varied strongly among tree species, with highest alpha‐ and gamma‐diversity found in Quercus petraea. Red‐listed saproxylic beetle species occurred ubiquitously among tree species. We found distinct differences in the community composition of broadleaved and coniferous tree species, native and non‐native tree species as well as sun‐exposed and shaded deadwood.
Our study enhances the understanding of the importance of previously understudied and non‐native tree species for the diversity of saproxylic beetles. To improve conservation practices for saproxylic beetles and especially red‐listed species, we suggest a stronger incorporation of tree species diversity and sun exposure of into forest management strategies, including the enrichment of deadwood from native and with a specific focus on locally rare or silviculturally less important tree species.
Considerable effort has previously been invested in a light‐controlled inhibition of the enzyme acetylcholinesterase (AChE). We found that a novel azobenzene‐based bistacrine AChE inhibitor switched faster than the known dithienylethene based bistacrine and inverted the photo‐controlled interactions of the photoisomers compared to its dithienylethene congener. Furthermore, we have optimized a previously described light‐controlled tacrine‐based AChE inhibitor. Isomerization upon irradiation with UV light of the novel inhibitor was observed in aqueous medium and showed no fatigue over several cycles. The cis‐enriched form showed an 8.4‐fold higher inhibition of hAChE compared with its trans‐enriched form and was about 30‐fold more active than the reference compound tacrine with a single‐digit nanomolar inhibition. We went beyond proof‐of‐concept to discover photoswitchable AChE inhibitors with pharmacologically desirable nanomolar inhibition, “cis‐on” effect, and pronounces differences between the photoisomers.

Das Ziel dieser Arbeit war es den Effekt des spezifischen Inhibitors AMD3100 auf die CXCR4 vermittelte Proliferation und Migration der Vestibularisschwannomzellen in verschiedenen Zellkuturmodellen zu analysieren.
Die nachgewiesene Inhibition von CXCR4 deutet auf einen möglichen Einsatz von AMD3100 in der systemischen Therapie von NF-2 Patienten hin.
Kardiale Ischämien sind die häufigste Todesursache in Deutschland. Aufgrund bisheri-ger Fortschritte in der Therapie und Entwicklung neuer präventiver Prozesse, gewinnen die Erkenntnisse kardioprotektiver Strategien zunehmend an Bedeutung und gelangen in den Mittelpunkt gegenwärtiger Forschung. Der Ablauf molekularer Vorgänge und die subzellulären Veränderungen im Rahmen der APost könnten zu einem möglichen Erklä-rungsansatz beitragen. Die APost wurde im Tiermodell bisher in diversen Spezies gezeigt und konnte experimentell etabliert werden. Diese Arbeit beschäftigt sich mit der Frage, ob die Veränderungen der GRK2 auf molekularer Ebene einer der Gründe für den kardioprotektiven Mechanismus der APost sein könnte. Die GRK2 ist eine myokardspezfische Kinase, die bereits im Rahmen von Herzinsuffizenzmodellen mit kardioprotektiven Eigenschaften in Zusammenhang gebracht worden ist. Ob die GRK2 ebenfalls im Ischämie/Reperfusionsmodell den letalen Reperfusionsschaden und die Infarktgöße beeinflusst, wird in dieser Arbeit in-vivo untersucht.
Zusammenfassend lassen sich aus dieser Studie folgende Erkenntnisse gewinnen:

Diese Daten im Ischämie/Reperfusionsmodell zu reproduzieren und translational weiterzuentwickeln, sollte ein fokussiertes Ziel der Grundlagenforschung und Organprotektion werden, da unter Berücksichtigung des demographischen Wandels und der Lebenszeitprävalenzen zu KHK und Herzinfarkten ein gezieltes pharmakologisches Target im Fokus stehen sollte.
Die Arbeit greift die seit mehreren Jahrzehnten bestehenden Reformbestrebungen im Bereich des Systems der UN-Menschenrechtsüberwachung auf und setzt sich unter Einbeziehung vergangener und aktueller Entwicklungen mit den Perspektiven des Systems auseinander. Dabei wird nicht nur das System der vertraglichen UN-Menschenrechtsüberwachung einer kritischen Analyse unterzogen, sondern in einem größeren Kontext auch die Menschenrechtsüberwachung durch spezifische UN-Organe wie den Menschenrechtsrat sowie die Wechselwirkung mit bereits bestehenden justiziellen Mechanismen wie dem Internationalen Gerichtshof und dem Internationalen Strafgerichtshof. Nach einer ausführlichen Bestandsaufnahme des Systems, in welcher die Ursachen für die seit Langem bestehenden Defizite herausgearbeitet werden, folgt eine detaillierte Auseinandersetzung mit möglichen Reformansätzen. Diese reichen von strukturell-prozeduralen Anpassungen bis hin zu grundlegenden Umstrukturierungen und der Schaffung neuer (justizieller) Überwachungsorgane wie einem ständigen einheitlichen Vertragsorgan oder einem Internationalen Gerichtshof für Menschenrechte. Im Anschluss daran wird die Frage der Reformierung des bestehenden Systems aus dogmatischer Sicht beleuchtet und ein dogmatisches Konzept erarbeitet, welches zu einer Stärkung der Reformbestrebungen und des UN-Menschenrechtsschutzes als solches beitragen kann. Zugleich erhebt die Arbeit den Anspruch, auch unabhängig von einem dogmatischen Grundkonzept einen nachhaltigen Beitrag zur kritischen Bestandsaufnahme und umsichtigen Weiterentwicklung des Systems der UN-Menschenrechtsüberwachung zu leisten.
The cuticle is constituted of the biopolymer cutin and intra- and epicuticular waxes. In some cases, it has epicuticular wax crystals, protruding from the epicuticular wax film. One of the most important tasks is protection against desiccation. Many investigations were conducted to find the transport limiting component of the cuticle. It is evidentially confirmed that the waxes form this barrier. These waxes are multifactorial blends made of very-long-chain aliphatic (VLCA) compounds and triterpenoids (TRP). The VLCAs were proposed to constitute the transpiration barrier to water. However, experimental confirmation was lacking so far. The present study focuses on the development of a method to selectively extract TRPs from the cuticle and the impact of the removal on the transpiration barrier.
The plants deployed in this study exhibited several features. They had no epicuticular crystals on their surfaces, were astomatous, had a rather durable and possibly isolatable cuticle. A broad range of wax compositions was covered from plants with no TRP content and low wax load like Hedera helix and Zamioculcas zamiifolia to plants with high TRP content and high wax load like Nerium oleander. The selective extraction was conducted using a sequence of solvents. TRPs were extracted almost exhaustively from CMs with the first MeOH extract. Only a minor amount of shorter chained VLCAs was obtained. The remaining waxes, consisting mostly of VLCAs and some remnant TRPs, were removed with the following TCM extract.
After the extractions, the water permeance of native cuticular membranes (CM), MeOH extracted (M) and dewaxed cuticular discs (MX) was investigated gravimetrically. Compared to the water permeance of CMs, Ms showed no or only a small increase in water conductance. MXs, however, always showed strongly increased values.
The knowledge about the wax compounds constituting the transport-limiting properties is vital for different projects. For various issues, it would be favourable to have a standardized wax mixture as an initial point of research. It could be used to develop screening procedures to investigate the impact of adjuvants on cuticular waxes or the influence of wax constituents on the properties of cuticular waxes. This work concentrated on the development of an artificial wax mixture, which mimics the physical properties of a plant leaf wax sufficiently.
As target wax, the leaf wax of Schefflera elegantissima was chosen. The wax of this plant species consisted almost exclusively of VLCAs, had a rather simple composition regarding compound classes and chain length distribution and CMs could be isolated. Artificial binary, ternary and quaternary waxes corresponding to the conditions within the plant wax were investigated using differential scanning calorimetry (DSC), X-ray diffraction (XRD) techniques and Fourier-transform infrared (FTIR) spectroscopy. Phase diagrams were mapped out for a series of binary, ternary and quaternary wax mixtures. FTIR experiments were conducted using, ternary and a quaternary artificial wax blends. The blends were chosen to represent the conditions within the wax of the adaxial CM plant wax. The FTIR experiments exhibited an increasing resemblance of the artificial wax to the plant wax (adaxial CM wax) with an increasing number of compounds in the artificial wax. The same trend was found for DSC thermograms. Thermograms of ternary and quaternary blends exhibited more overlapping peaks and occurred in a temperature range more similar to the range of the whole leaf plant wax. The XRD spectrum at room temperature showed good conformity with the quaternary blend.
The current work illustrates a method for selective extraction of TRPs from isolated CMs. It gives direct experimental proof of the association of the water permeance barrier with the VLCA rather than to the TRPs. Furthermore, the possibility to mimic cuticular waxes using commercially available wax compounds is investigated. The results show promising feasibility for its viability, enabling it to perform as a standardized initial point for further research (e.g. to examine the influence of different constituents on waxes), revealing valuable knowledge about the structure and the chemistry-function relationship of cuticular waxes.
After almost two decades of extensive research, some controversy has remained regarding the self-renewal of resident macrophages of the central nervous system (CNS). Concurrently, the vessel wall has emerged as a potentially ubiquitous niche for stem and progenitor cells, including committed macrophage precursors. It is conceivable that their occurrence in the CNS might explain the brain-resident hematopoietic potential, which has repeatedly been observed but not yet characterized in detail. In this work, the presence of hematopoietic progenitors inside and outside the vessel wall was studied in the adult mouse brain, as well as their possible contribution to the resident macrophage pool. An immunohistological analysis did not corroborate CD45+ SCA-1+ macrophage progenitors, which have been characterized in peripheral arteries, in the circle of Willis. Accordingly, the ex vivo culture of CNS vessels did not provide evidence for de novo formation of macrophages, but for the extensive proliferative capacity of mature cells. However, when analyzing whole brain suspensions in colony-forming unit (CFU) assays, rare Iba1- Cx3cr1- (immature) clonogenic cells were detected, which were enriched at the cerebral surface/meninges and differentiated into macrophages in culture. Intravenous antibody injection and cell sorting confirmed their residence behind the blood-brain barrier. Intriguingly, brain-derived CFUs produced a unique pattern of colony types compared to cells from bone marrow (BM) or blood. Still they displayed the same immunophenotype as BM-resident myeloid progenitors (CD45lo, LIN-, SCA-1-, IL7Rα-, c-KIT+) and could be further stratified into a progenitor hierarchy giving rise to all erythro-myeloid cell types in vitro. This similarity was substantiated by labeling of their progeny in Flt3Cre x Rosa26mT/mG mice, which indicated a descendance from hematopoietic stem cells. While forced repopulation of brain macrophages using the CSF-1R inhibitor PLX5622 did not point to a role of progenitors in in vivo microglia/macrophage maintenance, recent advances in hematology imply that they might be involved in CNS immunosurveillance. In conclusion, though there was no evidence for adventitial macrophage precursors in the CNS, this study confirms the presence of myeloid progenitors in the adult brain and provides the anatomical and phenotypical details necessary to elucidate their relevance in neuroinflammation.
Ratten sind in urbanen Räumen allgegenwärtig. Sie leben hier zumeist im Untergrund oder doch gut verborgen vor menschlichem Blick. Von hier aus gestalten sie ohne bewusstes menschliches Zutun oder gar Akzeptanz bereits seit Jahrhunderten Stadträume eifrig mit. Dies Zusammenspiel von Ratten und Menschen wird in der vorliegenden Studie ausgehend von der unterfränkischen Stadt Würzburg untersucht. Die Autorin fragt nach Aushandlungsprozessen innerhalb alltäglicher Begegnungen von Menschen und Ratten, den damit verbundenen historischen Bezügen, Kontinuitäten und Brüchen sowie nach den hier wirksamen Narrationen. Mit den Multispecies Studies werden dabei Ratten als wirk- und handlungsmächtige Akteur*innen gesehen, die durch ihre Agency unter anderem Missstände in menschlichen Gesellschaften offenbaren.
According to the “canonical” paradigm of GPCR signaling, agonist-bound GPCRs only signal to the downstream adenylyl cyclase enzyme when they are seated at the plasma membrane. Upon prolonged binding of an agonist, receptor internalization usually takes place, leading to the termination of this downstream signaling pathway and activation of alternative ones. However, a set of recent studies have shown that at least some GPCRs (e.g. thyroid stimulating hormone receptor) continue signaling to adenylyl cyclase after internalization. In this study, I aimed to investigate canonical signaling by internalized μ opioid receptors (MORs), which are Gi-coupled receptors, using a fluorescence resonance energy transfer (FRET) sensor for cyclic AMP (cAMP) known as Epac1-camps. My results show that the cyclic AMP inhibition signal induced by the binding of DAMGO, a MOR agonist, persists after agonist washout. We hypothesized that this persistent signal might come from internalized DAMGO-bound receptors located in the endosomal compartment. To test this hypothesis, I used dynasore and Dyngo 4a, two dynamin inhibitors that are known to prevent clathrin-mediated endocytosis. Interestingly, dynasore but not Dyngo 4a pretreatment largely blunted the response to MOR activation as well as to adenylyl cyclase activation with Forskolin (FSK). In addition, DAMGO-induced cAMP signal remained persistent even in the presence of 30 M Dyngo 4a. These results might point to a complex interplay between clathrin-mediated internalization and MOR signaling. Further experiments are required to elucidate the mechanisms underlying the persistent MOR signaling and to fully clarify whether MORs are capable of Gi signaling in the endosomal compartment.
Das Tissue Engineering von Fettgewebe befasst sich mit der Herstellung von biologisch äquivalenten Gewebekonstrukten mit dem Ziel, diese in der Regenerativen Medizin zur Deckung von Weichteildefekten einzusetzen. Für die Ausreifung, Funktion und das Überleben von Adipozyten wurde die Bedeutung der Extrazellulärmatrix (EZM) zunehmend deutlich.18-20 Untersuchungen zur EZM und ihrer Einflussnahme auf die Adipogenese wurden bislang hauptsächlich an konventionellen zweidimensionalen Zellkulturen unter Verwendung von mesenchymalen Stammzellen aus dem Knochenmark (bone marrow-derived MSC), Präadipozyten der Mauszelllinie 3T3-L1 und intramuskulären Präadipozyten aus Rindern (bovine intramuscular preadipocytes, BIP) vorgenommen.23,56,69,76,115 Ziel dieser Arbeit war es Erkenntnisse über den Einfluss der EZM auf die adipogene Differenzierungsfähigkeit unter Verwendung von humanen mesenchymalen Stammzellen des Fettgewebes (human adipose-derived stem cells, hASC) zu gewinnen. Um in vitro eine natürlichere Mikroumgebung der Zellen zu generieren, wurde neben einer 2D Kultur vergleichend ein 3D Modell bestehend aus multizellulären Sphäroiden verwendet.84,85 Zudem war die Bestimmung eines stabilen Housekeeping-Gens notwendig, um valide Ergebnisse in qPCR-Analysen von Genexpressionsstudien zu gewährleisten.
Die Auswertung statistischer Parameter (Standardabweichung und Interquartilsbereich) sowie die Ergebnisse dreier zur Stabilitätsprüfung eingesetzten Softwares identifizierten EF1α als robustestes HKG.
Der Zusammenhang zwischen der EZM-Entwicklung und der Adipogenese wurde durch Hemmung der Kollagenentwicklung unter Verwendung von Ethyl-3,4-dihydroxybenzoat (EDHB) untersucht. Bei Betrachtung der Triglyceridsynthese mittels Histologie und quantitativer Analyse (Triglyceridassay) konnte in beiden Kultursystemen eine konzentrationsabhängige Hemmung der Adipogenese festgestellt werden. Im Unterschied zur 2D Kultur konnte der Triglyceridgehalt im 3D Modell annähernd auf das Niveau der nicht-induzierten Kontrolle gesenkt werden und damit ein tendenziell stärkerer negativer Effekt im 3D Modell demonstriert werden. In Untersuchungen zur Genexpression wurde die Expressionsrate der späten adipogenen Marker aP2 und C/EBPα maximal durch Zugabe von 0,05 mM EDHB gesenkt, wobei der Effekt in 3D erneut stärker ausgeprägt war.
Bei Betrachtung der Kollagenentwicklung zeigte sich immunhistochemisch zunächst eine Adipogenese-assoziierte Entwicklung der Kollagene I, IV und VI im 2D und 3D Modell. Durch die Zugabe von EDHB ließ sich die Kollagenbildung gleichermaßen in 2D und 3D konzentrationsabhängig inhibieren. Damit konnte ein Rückgang der Synthese von drei für die Adipozyten relevanten Kollagenen zusammen mit der Störung der adipogenen Differenzierung nachgewiesen werden. Auf mRNA-Ebene hingegen war eine unterschiedliche Expression von Kollagen I und IV nachweisbar. Für Kollagen I wurde eine Abnahme der Expression bei Differenzierung der Zellen beobachtet, während die Expressionsrate von Kollagen IV erst mit Beginn der Adipogenese gesteigert wurde. Die Genexpression der untersuchten Kollagene wurde durch EDHB nicht negativ beeinflusst.
Insgesamt weisen die Ergebnisse auf einen engen Zusammenhang der Kollagensynthese mit der Adipogenese hin. Inwieweit eine durch Zell-Matrix-Interaktionen ausgelöste Signaltransduktion und regulatorische Mechanismen in den Präadipozyten die Adipogenese beeinflussen, bleibt jedoch Gegenstand zukünftiger Forschung.
Das Targeting des MEK-Proteins in Krebszellen führt in der Regel zu einer erworbenen Resistenz gegen MEK-Inhibitoren und zur Aktivierung des überlebenswichtigen Proteins Akt. Da sowohl MEK als auch Akt Clienten des Hsp90-Chaperonsystems sind, untersucht die vorliegende Arbeit die Reaktionen von bestrahlten Lungenkarzinom- (A549) und Glioblastom- (SNB19) Zelllinien auf eine kombinierte MEK- und Hsp90-Hemmung. Unerwarteterweise verbesserte der 24 h vor der Bestrahlung verabreichte MEK-Inhibitor PD184352 das Zellüberleben durch Hochregulation von MEK und Erk1/2, aber auch von Akt. Im Gegensatz dazu reduzierte PD184352, das 1 h vor der Bestrahlung zugegeben wurde, die Expression von Erk stark und regulierte Akt in beiden Zelllinien nicht hoch. Als Ergebnis verstärkte der MEK-Inhibitor die radiosensibilisierende Wirkung des Hsp90-Inhibitors NVP-AUY922 in Glioblastomzellen (SNB19).
We compute genus-0 Belyi maps with prescribed monodromy and strictly verify the computed results. Among the computed examples are almost simple primitive groups that satisfy the rational rigidity criterion yielding polynomials with prescribed Galois groups over Q(t). We also give an explicit version of a theorem of Magaard, which lists all sporadic groups occurring as composition factors of monodromy groups of rational functions.
The liver plays a pivotal role in maintaining energy homeostasis. Hepatic carbohydrate and lipid metabolism are tightly regulated in order to adapt quickly to changes in nutrient availability. Postprandially, the liver lowers the blood glucose levels and stores nutrients in form of glycogen and triglycerides (TG). In contrast, upon fasting, the liver provides glucose, TG, and ketone bodies. However, obesity resulting from a discrepancy in food intake and energy expenditure leads to abnormal fat accumulation in the liver, which is associated with the development of hepatic insulin resistance, non-alcoholic fatty liver disease, and diabetes. In this context, hepatic insulin resistance is directly linked to the accumulation of diacylglycerol (DAG) in the liver. Besides being an intermediate product of TG synthesis, DAG serves as second messenger in response to G-protein coupled receptor signaling. Protein kinase D (PKD) family members are DAG effectors that integrate multiple metabolic inputs. However, the impact of PKD signaling on liver physiology has not been studied so far. In this thesis, PKD3 was identified as the predominantly expressed isoform in liver. Stimulation of primary hepatocytes with DAG as well as high-fat diet (HFD) feeding of mice led to an activation of PKD3, indicating its relevance during obesity. HFD-fed mice lacking PKD3 specifically in hepatocytes displayed significantly improved glucose tolerance and insulin sensitivity. However, at the same time, hepatic deletion of PKD3 in mice resulted in elevated liver weight as a consequence of increased hepatic lipid accumulation. Lack of PKD3 in hepatocytes promoted sterol regulatory element-binding protein (SREBP)-mediated de novo lipogenesis in vitro and in vivo, and thus increased hepatic triglyceride and cholesterol content. Furthermore, PKD3 suppressed the activation of SREBP by impairing the activity of the insulin effectors protein kinase B (AKT) and mechanistic target of rapamycin complexes (mTORC) 1 and 2. In contrast, liver-specific overexpression of constitutive active PKD3 promoted glucose intolerance and insulin resistance. Taken together, lack of PKD3 improves hepatic insulin sensitivity but promotes hepatic lipid accumulation. For this reason, manipulating PKD3 signaling might be a valid strategy to improve hepatic lipid content or insulin sensitivity. However, the exact molecular mechanism by which PKD3 regulates hepatocytes metabolism remains unclear.
Unbiased proteomic approaches were performed in order to identify PKD3 phosphorylation targets. In this process, numerous potential targets of PKD3 were detected, which are implicated in different aspects of cellular metabolism. Among other hits, phenylalanine hydroxylase (PAH) was identified as a target of PKD3 in hepatocytes. PAH is the enzyme that is responsible for the conversion of phenylalanine to tyrosine. In fact, manipulation of PKD3 activity using genetic tools confirmed that PKD3 promotes PAH-dependent conversion of phenylalanine to tyrosine. Therefore, the data in this thesis suggests that PKD3 coordinates lipid and amino acid metabolism in the liver and contributes to the development of hepatic dysfunction.
Am Universitätsklinikum Würzburg wurden zwischen 2006-2015 447 Fälle einer akuten erregerbedingten Meningoenzephalitis in den Kliniken der Neurologie, Kinderklinik, Neurochirurgie und Psychiatrie behandelt. Es konnten sowohl Fälle durch Bakterien als auch Fälle durch Viren, Parasiten und Pilze gesichert werden. Diese Arbeit beschreibt die lokale Epidemiologie akuter erregerbedingter Meningoenzephalitiden.
TNFR1 is a crucial regulator of NF‐ĸB‐mediated proinflammatory cell survival responses and programmed cell death (PCD). Deregulation of TNFα‐ and TNFR1‐controlled NF‐ĸB signaling underlies major diseases, like cancer, inflammation, and autoimmune diseases. Therefore, although being routinely used, antagonists of TNFα might also affect TNFR2‐mediated processes, so that alternative approaches to directly antagonize TNFR1 are beneficial. Here, we apply quantitative single‐molecule localization microscopy (SMLM) of TNFR1 in physiologic cellular settings to validate and characterize TNFR1 inhibitory substances, exemplified by the recently described TNFR1 antagonist zafirlukast. Treatment of TNFR1‐mEos2 reconstituted TNFR1/2 knockout mouse embryonic fibroblasts (MEFs) with zafirlukast inhibited both ligand‐independent preligand assembly domain (PLAD)‐mediated TNFR1 dimerization as well as TNFα‐induced TNFR1 oligomerization. In addition, zafirlukast‐mediated inhibition of TNFR1 clustering was accompanied by deregulation of acute and prolonged NF‐ĸB signaling in reconstituted TNFR1‐mEos2 MEFs and human cervical carcinoma cells. These findings reveal the necessity of PLAD‐mediated, ligand‐independent TNFR1 dimerization for NF‐ĸB activation, highlight the PLAD as central regulator of TNFα‐induced TNFR1 oligomerization, and demonstrate that TNFR1‐mEos2 MEFs can be used to investigate TNFR1‐antagonizing compounds employing single‐molecule quantification and functional NF‐ĸB assays at physiologic conditions.
Stronger reactivity to social gaze in virtual reality compared to a classical laboratory environment

People show a robust tendency to gaze at other human beings when viewing images or videos, but were also found to relatively avoid gaze at others in several real‐world situations. This discrepancy, along with theoretical considerations, spawned doubts about the appropriateness of classical laboratory‐based experimental paradigms in social attention research. Several researchers instead suggested the use of immersive virtual scenarios in eliciting and measuring naturalistic attentional patterns, but the field, struggling with methodological challenges, still needs to establish the advantages of this approach. Here, we show using eye‐tracking in a complex social scenario displayed in virtual reality that participants show enhanced attention towards the face of an avatar at near distance and demonstrate an increased reactivity towards her social gaze as compared to participants who viewed the same scene on a computer monitor. The present study suggests that reactive virtual agents observed in immersive virtual reality can elicit natural modes of information processing and can help to conduct ecologically more valid experiments while maintaining high experimental control.
In der vorliegenden Arbeit wurde untersucht, ob die farbige DSA-Darstellungsweise besser als die monochromatische dazu geeignet ist, eine Entscheidung über eine mögliche Stentimplantation bei pAVK zu treffen.
Hierfür wurden DSA-Daten des Universitätsklinikums Würzburg im Zeitraum 04/2014 - 10/2015 retrospektiv ausgewertet. Drei Ärzte bewerteten die Bilder in zwei getrennten Durchgängen bezüglich ihrer Entscheidung zur Stentimplantation. Diese Entscheidungen wurden mit einem Konsensus aus 2 Ärzten verglichen. Anhand von ROC-Analysen konnte so die Treffsicherheit der Entscheidungen evaluiert werden.
In der Studie stellte sich die farbkodierte Darstellung im Vergleich zur monochromatischen Darstellung nicht als überlegen heraus.
Patellaluxationen sind eine vor allem bei jungen, aktiven Patienten häufige Verletzung komplexer Ätiologie. Die Rekonstruktion des medialen patellofemoralen Ligaments (MPFL) ist die aktuell etablierte Operationstechnik bei strecknaher patellofemoraler Instabilität, zu der in der Literatur eine Vielzahl an patellaren Fixationstechniken des autologen Sehnentransplantates beschrieben werden.
In dieser Studie wurden 71 Patienten 5 Jahre nach Rekonstruktion des MPFLs mit patellarer Fixation in Weichteiltechnik nachuntersucht und die klinischen Ergebnisse der Operationsmethode und die Zufriedenheit der Patienten ermittelt.
Dafür wurde die Reluxationsrate ermittelt und die Funktion der Kniegelenke im Alltag mithilfe des Kujala- und des Lysholm-Fragebogens, das Aktivitätsniveau der Patienten mit der Tegner-Aktivitätsskala erfasst. Im Rahmen einer Nachuntersuchung wurden die Beweglichkeit des Kniegelenks und die Stabilität der Kniescheibe klinisch untersucht. Die Ergebnisse wurden unter Berücksichtigung klinischer und radiologischer Risikofaktoren ausgewertet.
Die Studie ergab eine Reluxationsrate von 5,6% und ist somit vergleichbar mit der Rate anderer in der Literatur beschriebener Techniken. Die Ergebnisse der klinischen Untersuchung ergaben eine stabile ligamentäre Führung der Kniescheibe bei insgesamt guter Beweglichkeit der Kniegelenke, die Auswertung der Fragebögen zeigten signifikante Verbesserungen der Funktion der operierten Kniegelenke im Alltag bei unverändertem Aktivitätsniveau.
Im Ergebnis kann durch die vorliegende Studie belegt werden, dass durch die Rekonstruktion des MPFL mit weichteiliger patellarer Fixation langfristig gute Ergebnisse bei einer niedrigen Komplikationsrate erzielt werden können. Allerdings erhöht das Zusammentreffen verschiedener Pathologien wie eine Patella alta mit einer ausgeprägten Dysplasie der Trochlea das Risiko für eine persistierende Instabilität und eine erneute Luxation.
Advanced Glycation Endproducts (AGEs) akkumulieren bei zunehmendem Alter. Die Haut ist das einzige Organ der durch ultraviolettes Licht ausgelösten Vitamin D Synthese. Die Akkumulation von AGEs in der Haut könnte die Synthese von Vitamin D stören, während Mikroinflammation und oxidativer Stress (beides mit Vitamin D-Mangel assoziiert), sowohl die toxischen Effekte der AGEs, als auch deren Bildung selbst verstärken könnten. Wir untersuchten zunächst potentielle Zusammenhänge zwischen zirkulierendem Vitamin D3, AGEs im Blut und AGEs in der Haut mit Markern für Inflammation und oxidativem Stress bei Nichtdiabetikern. In der vorliegenden Studie untersuchten wir 146 Probanden (119 gesunde Probanden und 27 Patienten mit arterieller Hypertonie; 73 Männer und 73 Frauen; durchschnittliches Alter 57.0 ± 15.5 Jahre). Mit Hilfe des AGE-Readers wurden die Advanced Glycation Endproducts in der Haut (SAF) gemessen. Außerdem wurde Vitamin D3, AGE-assoziierte Fluoreszenz (AGE-Fl) im Plasma, hoch-sensitives C-reaktives Protein (hs-CRP), Advanced Oxidation Protein Products (AOPPs) und die Nierenfunktion bestimmt. Außerdem wurden in einer Untergruppe von 61 Probanden N-Carboxymethyllysin (CML), der lösliche Rezeptor für AGEs (soluble RAGE) und das lösliche Vascular Adhesion Protein-1 (sVAP-1) bestimmt. Der durchschnittlich gemessene Vitamin D-Spiegel betrug 22.5 ± 8.9 ng/ml. Eine Vitamin D-Insuffizienz (20 – 29 ng/ml) lag bei 43% und ein manifester Vitamin D-Mangel bei 37% vor. Der altersabhängige Anstieg der Haut-AGEs war bei Rauchern und Patienten mit arterieller Hypertonie stärker ausgeprägt. Einen Zusammenhang zwischen der Hautfluoreszenz (SAF) und Vitamin D-Mangel fand sich nicht. Bei Rauchern konnte eine inverse Beziehung zwischen Vitamin D3 und Plasma AGE assoziierter Fluoreszenz sowie dem Soluble Vascular Adhesion Protein-1 nachgewiesen werden. Unsere Ergebnisse lassen vermuten, dass bei Probanden mit nichtdiabetischer Stoffwechsellage ein Vitamin D-Mangel nicht zu einer vermehrten Toxizität und Akkumulation der Advanced Glycation Endproducts führt. Nur bei Rauchern wäre solch eine Wechselwirkung denkbar.
Weil bei Diabetes mellitus die Akkumulation von Advanced Glycation Endproducts mit vermehrter kardiovaskulärer Morbidität und Mortalität in Zusammenhang steht, fragten wir uns außerdem ob ein Vitamin D-Mangel mit vermehrter AGE-Bildung und Toxizität bei Diabetikern einhergeht. Hierzu untersuchten wir 276 Diabetiker (160 Männer und 116 Frauen; Alter 65 ± 13.4 Jahre; 43 Typ 1-Diabetiker, 233 Typ 2-Diabetiker) und 121 Nichtdiabetiker (60 Männer und 61 Frauen; Alter 58.6 ± 15.5 Jahre). Die gleichen Parameter wie zuvor wurden bestimmt. Diabetiker zeigten höhere Werte an SAF und AGE-Fl als die Kontrollen. SAF und AGE-Fl korrelierte mit Alter, Diabetesdauer und Einschränkung der Nierenfunktion. Bei den Typ 2-Diabetikern korrelierte der altersabhängige AGE-Anstieg direkt mit hs-CRP und sVAP-1. Die Vitamin D-Spiegel der Diabetiker und Nichtdiabetiker waren beide gleich erniedrigt und lagen im Durchschnitt bei 22.5 ng/ml. Eine Beziehung zwischen Vitamin D und den erhobenen Parametern fand sich außer mit sVAP-1 (bei den Diabetikern) nicht. Zusammenfassend scheint ein Vitamin D-Mangel bei Diabetikern nicht mit vermehrter AGE-Akkumulation und einem Anstieg der Marker für Mikroinflammation und oxidativem Stress, mit Ausnahme von sVAP-1, einherzugehen.
Differenzierte β-Arrestin2 Rekrutierung am μ-Opioid Rezeptor durch klinisch eingesetzte Opioide

Opioide gehören zu den potentesten Analgetika für die Behandlung akuter und chronischer Schmerzen, werden jedoch in ihrer Anwendung durch analgetische Toleranz aber auch Nebenwirkungen wie Abhängigkeit, Atemdepression und Obstipation limitiert. Opioid-Analgetika vermitteln dabei nahezu alle klinisch relevanten Wirkungen durch Stimulation des μ-Opioidrezeptors, einem G- Protein-gekoppelten Rezeptor. Die „klassische“ Signaltransduktion durch Aktivierung inhibitorischer Gi/0-Proteine kann durch G-Protein gekoppelte Rezeptorkinasen (GRKs) und β-Arrestine negativ reguliert werden. Zusätzlich können durch β-Arrestin-Bindung an den Rezeptor G-Protein-unabhängige Signalwege aktiviert werden. Die genauen Mechanismen wie β-Arrestin- assoziierte Rezeptordesensibilisierung, -internalisierung und G-Protein- unabhängige Signalwege an der physiologischen Antwort und insbesondere an Toleranzentwicklung und Abhängigkeit von Opioid-Analgetika beteiligt sind, können bislang nicht ausreichend erklärt werden.
In dieser Arbeit konnte in HEK293-Zellen mit Lebendzell-Konfokalmikroskopie und Luciferase-Komplementierung für 17 Opioide eine differenzierte β-Arrestin2- Rekrutierung zum μ-Opioidrezeptor gezeigt werden. Von den untersuchten Opioiden sind 13 häufig eingesetzte Opioid-Analgetika. Durch die Erstellung detaillierter pharmakologischer Profile ließen sich die Opioide bezüglich ihres β- Arrestin2-Rekrutierungsvermögens in Voll-, Partial und Antagonisten eingruppieren. Bemerkenswert war die fehlende β-Arrestin2-Rekrutierung für Buprenorphin, Tramadol und Tilidin, sodass diese interessante Substanzen für weitere Untersuchungen in physiologischerem Kontext sind. Durch Überexpression von GRK2 konnte die β-Arrestin2-Rekrutierung insbesondere für Partialagonisten gesteigert werden, was die Abhängigkeit der β-Arrestin- Rekrutierung vom GRK-Expressionslevel, das in verschiedenen Assays und Gewebetypen variieren kann, zeigt. Außerdem konnte ein heterogenes Bild der Rezeptorregulierung demonstriert werden, welches indirekt durch Endozytosehemmung unter Verwendung von Dynamin-Inhibitoren erfasst wurde. Die erhobenen Daten dienen als Anknüpfungspunkt für weiteren Arbeiten auf dem Gebiet der μ-Opioidrezeptorregulation. Ein besseres Verständnis der molekularen Mechanismen ist nötig, um sichere und nebenwirkungsärmere Opioid-Analgetika entwickeln zu können.
Cell culture models are helpful tools to study inflammatory diseases, like rheumatoid arthritis (RA), osteoarthritis (OA), arteriosclerosis or asthma, which are linked to increased matrix metalloproteinase (MMP) activity. Such cell culture models often focus on the secretion of cytokines and growth factors or the direct effects of disease on tissue destruction. Even though the crucial role of MMPs in inflammatory diseases is known, the results of MMP studies are contradictious and the use of MMPs as biomarkers is inconsistent. MMPs play an important role in disease pathology, as they are involved in elastin degradation in the walls of alveoli in chronic obstructive pulmonary disease (COPD), tumor angiogenesis and metastasis and in cartilage and bone degradation in arthropathies. In RA and OA MMPs are secreted by osteocytes, synoviocytes, and by infiltrating immune cells in response to the increased concentration of inflammatory mediators, like growth factors and cytokines. MMPs are zinc and calcium-dependent proteinases and play an important role in physiological and pathological extracellular matrix (ECM) turn over. Their substrate specificity gives them the ability to degrade all major ECM components, like aggrecan, elastin, gelatin, fibronectin and all types of collagen even the triple helix of collagen monomers. The ECM consists of two large three-dimensional cross-linked macromolecule classes: one are fibrous proteins, like collagen and elastin fibers that are responsible for ECM’s structure, tensile strength, resiliency, reversible extensibility, and deformability and the second class is comprised of proteoglycans composed of glycosaminoglycan (GAG) chains covalently attached to protein cores that are multifunctionally involved in signaling pathways and cell interactions. ECM is present within all tissues and organs and changes in ECM structure contribute to pathogenesis, e.g. wounded and fibrotic tissue, COPD or tumours.
This thesis primarily focuses on the development of a diagnostic peptide system, that enables to gain information on MMP activity from ECM by deploying the isobaric mass encoding strategy. The core element of the developed system is an isotopically labelled peptide sequence (mass tag), that is released in response to elevated levels of MMPs and allows multiplexed detection in tandem mass spectrometry (LC-MS/MS). The mass reporters possess a modular structure with different functionalities. C-terminal either a transglutaminase (TG) recognition sequence or a high molecular weight polyethylene glycol (PEG) moiety was attached to immobilize the mass reporters covalently or physically at the injection site. The following matrix metalloproteinase substrate sequence (MSS) is incorporated in two different versions with different sensitivity to MMPs. The MSS were applied in pairs for relative quantification consisting of the cleavable version synthesized with natural L-amino acids and the non-cleavable D-amino acid variant. The mass tag was synthesized with isotopically labelled amino acids and is separated from the MSS by a UV light-sensitive molecule. N-terminal the mass tag is followed by a tobacco etch virus protease (TEV) sensitive sequence, that is responsible to separate the mass tag from the affinity tag, which was either the Strep-tag II sequence or biotin and were added for purification purposes.
Chapter 1 presents a step-by-step protocol on how to design a mass tag family allowing for multiplexed analysis by LC-MS/MS. The multiplexing is achieved by developing an isobar mass tag family with four family members, which are chromatographically indistinguishable, but due to the mass encoding principles they fragment in distinct y-type ions with a mass difference of 1 or 2 Da each in MS2. Furthermore, it is explained how to covalently attach the mass reporter peptides onto ECM by the activated calcium-catalyzed blood coagulation transglutaminase factor XIII (FXIIIa). The lysine of mass reporter’s TG sequence (D-domain of insulin-like growth factor-I (IGF-I)) and a glutamine in fibronectin are covalently crosslinked by FXIIIa and build an isopeptide bond. Elevated levels of MMP release the mass reporters from ECM by recognizing the inter-positioned MSS.
The designed mass reporters were able to monitor enzyme activity in an in vitro setting with cell-derived ECM, which was shown in Chapter 2. The modular structured mass reporters were investigated in a proof of concept study. First, the different modules were characterized in terms of their MMP responsiveness and their sensitivity to TEV protease and UV light. Then the FXIIIa-mediated coupling reaction was detailed and the successful coupling on ECM was visualized by an immunosorbent assay or confocal laser scanning microscopy. Finally, the immobilized mass reporters on ECM were incubated with MMP-9 to investigate their multiplexing ability of MMP activity. The cleaved mass reporter fragments were purified in three steps and mass tags were analyzed as mix of all four in LC-MS/MS.
Chapter 3 describes the change from an immobilizing system as seen in chapter 1 and 2 to a soluble enzyme activity monitoring system that was applied in an osteoarthritic mouse model. Instead of the immobilizing TG sequence the C-terminal MMS was extended with two amino acids where one holds an azide moiety to perform a strain-promoted azide-alkyne cycloaddition to a high molecular weight dibenzocyclooctyne-polyethylene glycol (DBCO-PEG), which was chosen to retain the mass reporters at the injection site. Furthermore, the N-terminal affinity tag was extended with a 2.5 kDa PEG chain to increase the half-life of the mass reporter peptides after MMP release. The systems biocompatibility was proved but its enzyme monitoring ability in an in vivo setting could not be analyzed as samples degraded during shipping resulting from the Chinese customs blocking transport to Germany.
In summary the diagnostic peptide system was developed in two variants. The immobilized version one from chapter 1 and 2 was designed to be covalently attached to ECM by the transglutaminase-mediated cross-linking reaction. In an in vitro setting the functionality of the mass reporter system for the detection of MMP activity was successfully verified. The second variant comprises of a soluble mass reporter system that was tested in an OA mouse model and showed biocompatibility. With these two designed systems this thesis provides a flexible platform based on multiplexed analysis with mass-encoded peptides to characterize cell culture models regarding their MMP activity, to deploy cell-derived ECM as endogenous depot scaffold and to develop a mass tag family that enables simultaneous detection of at least four mass tags.
This paper examines situations where two vertically integrated firms consider supplying an input to an independent downstream competitor via privately observed contracts. We identify equilibria where competition in the upstream market emerges—the downstream competitor gets supplied—as well as when the downstream firm does not receive the input and is excluded from the market. The likelihood of the outcome in which the downstream firm does not get supplied depends not only on demand parameters, but also on contractual flexibility and observability. We show that when contracts are unobservable, downstream entry will occur less often. Furthermore, our results suggest that permitting contracts that enable the contracting parties to coordinate their behavior in the downstream market may improve welfare by increasing the likelihood that the downstream firm is supplied.
Die Corona-Krise stellt eine der größten Herausforderungen in der Geschichte der EU dar. Aufgrund der geringen Kompetenzen der EU im Gesundheitsbereich liegt die Pandemiebekämpfung fast ausschließlich in den Händen der Mitgliedstaaten. Diese reagierten jedoch zunächst mit „nationalen Reflexen“ und unsolidarischem Verhalten. Erst nach Überwindung des ersten Schocks im Frühjahr 2020 konnte die EU sichtbarer bei der Krisenbewältigung werden. Den Höhepunkt stellte die Einigung auf das historische 750 Mrd. EUR schwere Corona-Hilfspaket „Next Generation EU“ (NGEU) dar, welches mit einer gemeinsamen Schuldenaufnahme einen Präzedenzfall geschaffen hat.
Diese Arbeit untersucht, wie die EU auf die Pandemie reagiert hat und ob diese Reaktion zu ihrer Stärkung führen kann. Sie soll einen Beitrag zum besseren Verständnis der Geschehnisse in der EU zwischen Januar 2020 und Mai 2021 leisten. Hierfür werden zunächst die Kompetenzen der EU im Gesundheitsbereich und beim Katastrophenschutz sowie deren Nutzung in der Pandemie aufgezeigt. Hauptteil der Arbeit ist die Untersuchung von Entstehung und Inhalt des NGEU-Hilfspaktes. Hier zeigt sich, dass die EU – mit Hilfe des deutsch-französischen Motors – zur Solidarität zurückgefunden hat. Die Schwerpunktsetzung von NGEU verdeutlicht, dass neben dem Wiederaufbau auch die aktuellen Kernthemen der EU – Digitalisierung und Klimaschutz – einen zentralen Stellenwert einnehmen. Damit kann NGEU zur wesentlichen Stärkung der EU beitragen. Eine Stärkung ist ebenfalls im Gesundheitsbereich festzustellen, wo erste Schritte zu einer Gesundheitsunion vollzogen wurden.
This thesis is concerned with applying the total variation (TV) regularizer to surfaces and different types of shape optimization problems. The resulting problems are challenging since they suffer from the non-differentiability of the TV-seminorm, but unlike most other priors it favors piecewise constant solutions, which results in piecewise flat geometries for shape optimization problems.The first part of this thesis deals with an analogue of the TV image reconstruction approach [Rudin, Osher, Fatemi (Physica D, 1992)] for images on smooth surfaces. A rigorous analytical framework is developed for this model and its Fenchel predual, which is a quadratic optimization problem with pointwise inequality constraints on the surface. A function space interior point method is proposed to solve it. Afterwards, a discrete variant (DTV) based on a nodal quadrature formula is defined for piecewise polynomial, globally discontinuous and continuous finite element functions on triangulated surface meshes. DTV has favorable properties, which include a convenient dual representation. Next, an analogue of the total variation prior for the normal vector field along the boundary of smooth shapes in 3D is introduced. Its analysis is based on a differential geometric setting in which the unit normal vector is viewed as an element of the two-dimensional sphere manifold. Shape calculus is used to characterize the relevant derivatives and an variant of the split Bregman method for manifold valued functions is proposed. This is followed by an extension of the total variation prior for the normal vector field for piecewise flat surfaces and the previous variant of split Bregman method is adapted. Numerical experiments confirm that the new prior favours polyhedral shapes.
The central complex (CX) in the insect brain is a higher order integration center that controls a number of behaviors, most prominently goal directed locomotion. The CX comprises the protocerebral bridge (PB), the upper division of the central body (CBU), the lower division of the central body (CBL), and the paired noduli (NO). Although spatial orientation has been extensively studied in honeybees at the behavioral level, most electrophysiological and anatomical analyses have been carried out in other insect species, leaving the morphology and physiology of neurons that constitute the CX in the honeybee mostly enigmatic. The goal of this study was to morphologically identify neuronal cell types of the CX in the honeybee Apis mellifera. By performing iontophoretic dye injections into the CX, we traced 16 subtypes of neuron that connect a subdivision of the CX with other regions in the bee's central brain, and eight subtypes that mainly interconnect different subdivisions of the CX. They establish extensive connections between the CX and the lateral complex, the superior protocerebrum and the posterior protocerebrum. Characterized neuron classes and subtypes are morphologically similar to those described in other insects, suggesting considerable conservation in the neural network relevant for orientation.
Magnetic random access memory (MRAM) technology aims to replace dynamic RAM (DRAM) due to its significantly lower power consumption and non-volatility [Dong08]. During the last couple of years the commercial focus was set on spin-transfer torque MRAM (STT-MRAM) systems, where a current is pushed through a ferromagnetic (FM) free layer and a reference layer which are separated by an insulator. The free layer can be set to parallel or anti-parallel depending on the current direction [Kim11]. Unfortunately these currents have to be quite high which could lead to damages of the tunnel barrier of the magnetic tunnel junction resulting in higher power consumption as well as reliability issues. At this point a new effect, where the current is passed below the ferromagnetic layer stack, can be exploited to change the direction of the free layer magnetization. The effect is known as spin-orbit torque (SOT) and describes the transfer of angular momentum onto an adjacent magnetization either by the spin Hall effect (SHE) or inverse spin galvanic effect (iSGE) [Manchon19]. The latter describes a spin accumulation due to a current. This is similar to the process of spin accumulation in TIs, where a current corresponds to an effective spin due to spin-momentum locking [Qi11]. Thus TIs exhibit a high current-to-spin conversion rate, which makes them a promising material system for SOT experiments. Among all TIs it is HgTe, which can be reliably grown as an insulator. This thesis covers the development of a working device for SOT measurements (SOT-device) in a CdTe/CdHgTe/HgTe/CdHgTe heterostructure. It involves the development of a tunnel barrier (ZrOx) as well as the investigation of the behavior of a ferromagnetic layer stack on top of etched HgTe. The main result of this work is the successful construction and evaluation of a working SOT-device, which exhibits the up to date most efficient switching of in-plane magnetized ferromagnetic layer stacks.
In order to avoid hybridization between HgTe and the adjacent ferromagnetic atoms, which would cause a breakdown of the topological surface state, it is necessary to implement a thin tunnel barrier in between the TI and free layer [Zhang16]. Aside from hybridization a tunnel barrier avoids shunting of the current, that is pushed on the surface of the HgTe/CdHgTe interface. Thus a bigger part of the current can be used for spin accumulation and, at the same time, the resistance measurement of the ferromagnetic layer stack is not perturbed. In chapter 3 the focus is set on investigating the tunneling characteristics of ZrOx on top of dry etched HgTe. Thin barriers are used as the interaction of the current generated spin and the adjacent magnetization decreases with distance. On the other hand too small insulator thicknesses lead to leakage currents which disturb heavily the measurement of the resistance of the ferromagnetic layer stack. Thus an optimum thickness of 10 ALD cycles (\(d\approx 1.6\rm\, nm\)) is determined which yields a resistance area product of \(R\cdot A \approx 3\rm\, k\Omega\mu m^{2}\). This corresponds to a tunneling resistance of \(R_{T}\approx 20\rm\, k\Omega\) over a structure surface of \(A_{T} = 0.12\rm\, \mu m^2\). Multiple samples with different thicknesses have been produced. All samples have been examined on their tunneling behavior. The resistance area product as a function of thickness shows a linear behavior on a logarithmic scale. Furthermore all working samples show non-linear I-V curves as well as parabolic dI/dV-curves. Additionally the tunneling resistance \(R_{T}\) increases with decreasing temperature. All above mentioned properties are typical for tunnel barriers which do not include pinholes [Jonsson00]. The last part of chapter 3 deals with thermal properties of HgTe. By measuring the second harmonic of a biasing AC current in the channel below the tunnel barrier it is attempted to extract the diffusion thermopower of the heated electrons. Unfortunately the measured signal showed a far superior contribution of the first harmonic. According to electric circuit simulations a small asymmetry in the barrier (penetration and leaving point of electrons) could be responsible for this behavior.
A ferromagnetic layer stack, consisting of PY/Cu/CoFe, serves as a sensor for magnetization changes due to external fields and current induced spin accumulations. The layer stack exhibits a giant magnetoresistance (GMR) which has been measured by a resistance bridge. The biggest peculiarity in depositing a GMR stack on top of HgTe is that its easy axis forms along only one of the crystal axes (\((110)\) or \((1\overline{1}0)\)). The reason for this anisotropy is still unclear. Sources such as an influence of the terminating material, miscut, furrows during IBE or sputter ripples have been ruled out. It can be speculated that the surface states due to HgTe might have an influence on the development of this easy axis but this would need further investigation. A consequence of this unexpected anisotropy is that every CdTe/CdHgTe/HgTe/CdHgTe wafer has first to be characterized in SQUID in order to find the easy axis. A ferromagnetic resonance (FMR) measurement confirmed this observation. The shape of the ferromagnetic layer stack is chosen to be an ellipse in order to support the easy axis direction by shape anisotropy. Over 8 million ellipses are used to generate a SQUID signal of \(m > 10^{-5}\rm\, emu\). This is sufficient to extract the main characteristics of an average nano pillar under the influence of an external magnetic field. As in the case of bigger structures the ellipse shaped structure shows a step-like behavior. A measured minor loop confirms the existence of the irreversible anti-parallel stable magnetic state. Furthermore this state persists for both directions at \(m=0\) resulting in an anti-ferromagnetic coupling between Py and CoFe.
The geometry of the SOT-device is chosen in such a way that the current induced spin aligns either parallel or anti-parallel to the effective magnetic field \(\vec{B}_{eff}=\vec{B}_{ext}+\vec{B}_{aniso}+\vec{B}_{shape}\), which acts on the pillar. Due to interaction of the spin with the adjacent magnetization of Py the magnetization direction gets changed by a torque \(\vec{T}\). In general this torque can be decomposed into two components a field-like torque \(\vec{\tau}_{FL}\) and a damping-like torque \(\vec{\tau}_{DL}\) [Manchon19]. In the case of TIs \(\vec{T}\) is additionally depending on the z-component of \(\vec{m}\) [Ndiaye17]. In our case the magnetization is lying in the sample plane (\(m_{z}=0\)) which results in \(\vec{\tau}_{DL}=0\). Thus, in the case of \(\vec{S}\parallel\left(\vec{\hat{z}}\times\vec{j}\right)\) and \(\vec{j}\parallel\vec{\hat{y}}\), the only spin dependent effective magnetic field is \(\vec{B}_{FL}=\tau_{FL}\cdot\vec{\hat{x}}\) which is lying parallel or anti-parallel to \(\vec{B}_{eff}\). The evaluation of \(\vec{B}_{FL}\) can therefore be done in the following manner. First a high \(B_{ext}\) has to be set along the easy axis of the pillar. Then \(B_{ext}\) has to be reduced just a few \(\rm\, Oe\) before the switching occurs at the magnetic field \(B_{ext,0}\). At the magnetic field \(\Delta B = B_{ext}-B_{ext,0}\approx 0.5\rm\, Oe\) the lower resistive state should be stable over a longer time range (\(10-30\rm\, min\)) in order to exclude switching due to fluctuations. Now a positive or negative current can be pushed through the channel below the pillar. For one of the two current directions the magnetization of Py switches. It is therefore not a thermal effect that drives the change of \(\vec{m}\). Current densities that are able to switch \(\vec{m}\) at small \(\Delta B\neq 0\) lie in the range of \(j\approx 10^{4}\rm\, A/cm^{2}\). In all experiments the switching efficiency \(\Delta B/j\) decreases with rising \(j\). Furthermore the efficiency as a function of \(j\) depends on the temperature as \(\Delta B/j\) values tend to be up to 20 times higher at \(T=1.8\rm\, K\) and \(j\approx 0\) than at \(T=4.2\rm\, K\). This temperature dependence suggests that switching occurs not due to Oersted fields. Furthermore the Biot-Savart fields had been calculated for four different models: an infinite long rectangular wire, two infinite planes, a full volume and two thin volume planes. Every model shows an efficiency, which is at least three times lower than the observation.
The highest efficiencies in our samples show up to 10 times higher values than in heavy-metal/ferromagnets heterostructures. In contrast to measurement procedures of most other groups our method leads to direct determination of SOT parameters like the effective magnetic field \(\vec{B}_{FL}\). Other groups make use of spin-transfer FMR (ST-FMR) where they AC bias their structure and extract SOT parameters (like \(\tau_{FL}\) and \(\tau_{DL}\)) from second harmonics by fitting theoretical models. Material systems consisting of TIs and magnetic insulators (MIs) on the other hand show 10 times higher efficiencies [Khang18,Li19]. In those cases the magnetization points out of the sample plane which is conceptually different from in-plane magnetic anisotropy geometries like in our case. The greatest benefit in-plane magnetic anisotropy systems is its easy realisation [Bhatti17]. Here only an elliptical shape has to be lithographically implemented instead of conducting research on the appropriate combination of material systems that result in perpendicular magnetic anisotropies [Apalkov16]. Despite the fact that in our case only \(\vec{\tau}_{FL}\) acts as the driving force for changing \(m\) our device still exhibits the up to date highest efficiencies in the class of in-plane magnetized anisotropies of all material classes ever recorded.
Das erste Ziel der vorliegenden Dissertation bestand darin, ein bereits bestehendes TOF-MS-Setup dahingehend zu erweitern, um damit Velocity Map Imaging-Experimente durchführen zu können. Dies erforderte zunächst die Konzipierung und Programmierung einiger für die Datenaufnahme, -verarbeitung und -analyse benötigter LabView-Anwendungen. Anschließend konnten erste Kalibrierexperimente an Methyliodid, in denen wichtige experimentelle Parameter identifiziert und optimiert wurden, durchgeführt werden. Außerdem gelang es dadurch, die Messgenauigkeit des Setups auf 0.7 % und dessen Auflösungsvermögen auf 4.4 % zu bestimmen, was im Bereich für VMI-Apparaturen typischer Werte liegt. Zur weiteren Überprüfung der Funktionstüchtigkeit des Setups wurde in ersten zeitaufgelösten Experimenten im Folgenden die Desaktivierung des S1-Zustands von Pyridin untersucht. Neben der Reproduktion einiger bereits literaturbekannter Resultate konnten dabei zusätzlich die im Multiphotonen-Ionisationsschritt populierten Rydberg-Zustände identifiziert werden. Anschließend wurde mit Experimenten an bisher weniger gut untersuchten organischen Aromaten und Heteroaromaten fortgefahren. Das Ziel dieser Studien lag in der Aufklärung der photoinduzierten Dynamiken der Verbindungen, wobei das zur Verfügung stehende ps-Lasersystem die Möglichkeit bot, die Desaktivierung elektronisch angeregter Zustände gezielt in Abhängigkeit von deren Schwingungsenergie zu untersuchen. Der darin bestehende Vorteil zeigte sich vor allem in Studien an Tolan und Phenanthridin, deren erste angeregte, optisch aktive Zustände am Origin Lebensdauern im ns-Bereich aufweisen, die sich mit zunehmender vibronischer Anregung jedoch auf bis zu 10 ps verringern. Als Grund dafür konnten nichtstrahlende Desaktivierungsprozesse, für deren Eintreten eine energetische Barriere überwunden werden muss, identifiziert werden. Während in Tolan nach Photoanregung ein Übergang in einen (πσ∗)-Zustand, der zur Ausbildung einer trans-bent-Struktur führt, erfolgt, ist im Falle von Phenanthridin vermutlich ein El-Sayed-erlaubter ISC-Übergang in einen 3(nπ∗)-Zustand für die drastische Verkürzung der S1-Lebensdauer verantwortlich. Ein solcher konnte weder im zu Phenanthridin isomerischen Benzo[h]quinolin, noch in dessen PAH-Muttermolekül Phenanthren beobachtet werden, was auf die höhere energetische Lage bzw. die Abwesenheit des mittels ISC populierten 3(nπ∗)-Zustands in diesen Molekülen zurückgeführt werden kann. In weiteren im Rahmen der vorliegenden Arbeit durchgeführten Experimente wurden zudem die aromatischen Moleküle Acenaphthylen und 4-(Dimethylamino)benzethin (DMABE) untersucht. Zeitaufgelöste Studien zeigten dabei, dass die Desaktivierung der S2-Zustände beider Moleküle auf der sub-ps-Zeitskala stattfindet und mit dem vorhandenen Lasersystem daher nicht aufgelöst werden kann. In Acenaphthylen erfolgt die S2-Relaxation größtenteils über einen sequentiellen IC-Mechanismus, innerhalb dem der S1-Zustand des Moleküls intermediär besetzt wird. Dessen Lebensdauer konnte am Origin auf 380 ps bestimmt werden, fällt mit steigender Schwingungsanregung jedoch auf bis zu 55 ps ab. Für die Desaktivierung des S2-Zustands von DMABE konnte hingegen ein paralleles Relaxationsmodell, in dem neben dem S1-Zustand ein weiterer elektronisch angeregter Zustand populiert wird, nachgewiesen werden. Bei diesem könnte es sich möglicherweise um einen (πσ∗)-Zustand, dessen Besetzung die Ausbildung einer trans-bent-Geometrie innerhalb der Acetylen-Einheit des Moleküls zur Folge hat, handeln. Einen weiteren großen Teil der vorliegenden Dissertation nahmen Experimente an van-der-Waals-gebundenen Clustersystemen ein. Im Fokus der Studien standen dabei Moleküle mit ausgedehnten aromatischen π-Systemen, da solche eine hohe Relevanz für verschiedene materialwissenschaftliche Forschungsgebiete besitzen. Ein Beispiel hierfür ist Tetracen, welches als Modellsystem für die Untersuchung von Singlet Fission-Prozessen angesehen wird. In Kombination mit nichtadiabatischen Surface-Hopping-Simulationen zeigten Experimente an Tetracen-Dimeren, dass nach deren S2-Anregung zunächst ein schneller S1←S2-Übergang (τ < 1 ps), gefolgt von der Ausbildung einer Excimerstruktur, stattfindet. Letztere erfolgt mit einer Zeitkonstante von 62 ps und führt zu einem Anstieg des transienten Ionensignals, wohingegen die Desaktivierung des Excimer-Zustands von einem abklingenden Signalbeitrag mit τ = 123 ps repräsentiert wird. Wenngleich über die weitere Relaxation der Excimerspezies zum gegenwärtigen Zeitpunkt keine Aussage getroffen werden kann, besteht damit die Möglichkeit, dass Excimer-Zustände als Zwischenstufe im SF-Mechanismus isolierter Tetracen-Dimere auftreten. In zeitaufgelösten Experimenten an Phenanthren-Dimeren konnte ebenfalls ein Anstieg des transienten Signals mit einer vergleichbaren Zeitkonstante von τ = 86 ps, der jedoch auf einem konstanten Signaloffset endet, gefunden werden. Dies deutet darauf hin, dass auch Phenanthren-Dimere in der Lage sind, Excimerstrukturen, die im Gegensatz zu denen des Tetracens jedoch deutlich langlebiger sind, auszubilden. Studien an den Dimerspezies der Azaphenanthrene Benzo[h]quinolin und Phenanthridin offenbarten hingegen etwas schnellere Relaxationen mit Zeitkonstanten von 15 bzw. 40 ps. Zudem zeigten beide Spezies eine stark ausgeprägte Fragmentation, sodass für deren Untersuchung auf die VMI-Detektionsmethode zurückgegriffen werden musste. Dadurch wurde deutlich, dass sich Photoionen-Imaging-Experimente hervorragend für Studien an schwach gebundenen Clustersystemen eignen, da diese die Separation verschiedener Signalbeiträge innerhalb eines betrachteten Massenkanals ermöglichen.
Digitization and artificial intelligence are radically changing virtually all areas across business and society. These developments are mainly driven by the technology of machine learning (ML), which is enabled by the coming together of large amounts of training data, statistical learning theory, and sufficient computational power. This technology forms the basis for the development of new approaches to solve classical planning problems of Operations Research (OR): prescriptive analytics approaches integrate ML prediction and OR optimization into a single prescription step, so they learn from historical observations of demand and a set of features (co-variates) and provide a model that directly prescribes future decisions. These novel approaches provide enormous potential to improve planning decisions, as first case reports showed, and, consequently, constitute a new field of research in Operations Management (OM).
First works in this new field of research have studied approaches to solving comparatively simple planning problems in the area of inventory management. However, common OM planning problems often have a more complex structure, and many of these complex planning problems are within the domain of capacity planning. Therefore, this dissertation focuses on developing new prescriptive analytics approaches for complex capacity management problems. This dissertation consists of three independent articles that develop new prescriptive approaches and use these to solve realistic capacity planning problems.
The first article, “Prescriptive Analytics for Flexible Capacity Management”, develops two prescriptive analytics approaches, weighted sample average approximation (wSAA) and kernelized empirical risk minimization (kERM), to solve a complex two-stage capacity planning problem that has been studied extensively in the literature: a logistics service provider sorts daily incoming mail items on three service lines that must be staffed on a weekly basis. This article is the first to develop a kERM approach to solve a complex two-stage stochastic capacity planning problem with matrix-valued observations of demand and vector-valued decisions. The article develops out-of-sample performance guarantees for kERM and various kernels, and shows the universal approximation property when using a universal kernel. The results of the numerical study suggest that prescriptive analytics approaches may lead to significant improvements in performance compared to traditional two-step approaches or SAA and that their performance is more robust to variations in the exogenous cost parameters.
The second article, “Prescriptive Analytics for a Multi-Shift Staffing Problem”, uses prescriptive analytics approaches to solve the (queuing-type) multi-shift staffing problem (MSSP) of an aviation maintenance provider that receives customer requests of uncertain number and at uncertain arrival times throughout each day and plans staff capacity for two shifts. This planning problem is particularly complex because the order inflow and processing are modelled as a queuing system, and the demand in each day is non-stationary. The article addresses this complexity by deriving an approximation of the MSSP that enables the planning problem to be solved using wSAA, kERM, and a novel Optimization Prediction approach. A numerical evaluation shows that wSAA leads to the best performance in this particular case. The solution method developed in this article builds a foundation for solving queuing-type planning problems using prescriptive analytics approaches, so it bridges the “worlds” of queuing theory and prescriptive analytics.
The third article, “Explainable Subgradient Tree Boosting for Prescriptive Analytics in Operations Management” proposes a novel prescriptive analytics approach to solve the two capacity planning problems studied in the first and second articles that allows decision-makers to derive explanations for prescribed decisions: Subgradient Tree Boosting (STB). STB combines the machine learning method Gradient Boosting with SAA and relies on subgradients because the cost function of OR planning problems often cannot be differentiated. A comprehensive numerical analysis suggests that STB can lead to a prescription performance that is comparable to that of wSAA and kERM. The explainability of STB prescriptions is demonstrated by breaking exemplary decisions down into the impacts of individual features. The novel STB approach is an attractive choice not only because of its prescription performance, but also because of the explainability that helps decision-makers understand the causality behind the prescriptions.
The results presented in these three articles demonstrate that using prescriptive analytics approaches, such as wSAA, kERM, and STB, to solve complex planning problems can lead to significantly better decisions compared to traditional approaches that neglect feature data or rely on a parametric distribution estimation.
An N-heterocyclic-carbene-stabilized diboryne undergoes rapid, high-yielding and catalyst-free hydroamina- tion reactions with primary amines, yielding 1-amino-2-hydro- diborenes, which can be considered boron analogues of enamines. The electronics of the organic substituent at nitrogen influence the structure and further reactivity of the diborene product. With electron-rich anilines, a second hydroamination can occur at the diborene to generate 1,1-diamino-2,2-dihy- drodiboranes. With isopropylamine, the electronic influence of the alkyl substituent upon the diborene leads to an unprece- dented boron-mediated intramolecular N-dearylation reaction of an N-heterocyclic carbene unit.
Forschung über Bürgerwehren in Burkina Faso: Herausforderungen und Strategien des Feldzugangs

Dieses Working Paper veranschaulicht, wie sich die beiden Forscher*innen Janneke Tiegna und Nestor Zanté des Teilprojekts F „Lokale Selbstregelungen für die Herstellung von Sicherheit: Bürgerwehren in Burkina Faso“, während ihres bisherigen Forschungsaufenthalts von September 2019 bis Februar 2020 Zugang in ihre Forschungsgemeinden im Westen und Süden Burkina Fasos verschafft haben. Zanté forschte zu der Selbstverteidigungsgruppe Koglwéogo und Tiegna zu den Dozo-Jäger*innen.
Dieses Working Paper erklärt zunächst die Zielgruppe und die Forschungsgebiete und geht dabei kurz auf das Forschungsinteresse ein. Es folgen Erläuterungen zur Vorbereitung auf den Feldaufenthalt. Ausführlich werden die Herausforderungen der Forschung über diese sensible Thematik erläutert sowie Abwehrreaktionen und Widerstände im Feld dargestellt. Anschließend erklären Tiegna und Zanté, mit wel-chen Strategien sie diesen Herausforderungen begegnet sind.
The introductory chapter reviews the current state of mechanistic understanding of the hexadehydro-Diels-Alder (HDDA) reaction. With the rapid development of the HDDA reaction from its first discovery in 1997, the question of whether a concerted or stepwise mechanism better describes the thermally activated formation of ortho-benzyne from a diyne and a diynophile has been debated. Mechanistic and kinetic investigations were able to show that this is not a black or white situation, as minor changes can tip the balance. In chapter 2 of this thesis, the catalytic process leading from 1,11-bis(p-tolyl)undeca-1,3,8,10-tetrayne to fully-substituted naphthalene and azulene derivatives, by two different platinum-catalyzed dimerization pathways, was investigated. In chapter 3, the cannibalistic self-trapping reaction of an ortho-benzyne derivative generated from 1,11-bis(p-tolyl)undeca-1,3,8,10-tetrayne in an HDDA reaction was investigated. Without adding any specific trapping agent, the highly reactive benzyne is trapped by another bisdiyne molecule in at least three different modes. In chapter 4 direct UV/VIS spectroscopic evidence for the existence of an o-benzyne in solution is reported, and the dynamics of its formation in a photo-induced reaction are established. For this purpose, 1,11-bis(p-tolyl)undeca-1,3,8,10-tetrayne was investigated, using femtosecond transient absorption spectroscopy in the ultraviolet/visible region. In chapter 5, following the isolation and characterization of the reaction products discussed in chapter 3, further species resulting from reactions of the highly reactive ortho-benzyne derivative were identified.
Die Entwicklung des Schädeldachs beginnt beim Menschen bereits in der frühen Embryogenese und ist erst im Erwachsenenalter abgeschlossen. Das Wachstum der Schädelknochen muss sich während der Entwicklung fortwährend dem Gehirnwachstum anpassen. An den Stellen, wo zwei Schädelknochen aufeinandertreffen, formen sich Schädelnähte, die aus mesenchymalem Bindegewebe bestehen und als Wachstumsfugen des Schädels dienen. Tritt eine frühzeitige Verknöcherung innerhalb einer oder mehrerer Schädelnähte auf, spricht man von einer Kraniosynostose. Als Konsequenz wird ein weiteres Knochenwachstum verhindert, sodass sich das Neurokranium in dieser Region nicht dem expansiven Wachstum des Gehirns anpassen kann. Dies geht in der Regel mit einem kompensatorischen Wachstum des Schädels und infolgedessen mit kraniofazialen Dysmorphien und einem erhöhten intrakraniellen Druck einher. Klinische Studien und Forschungen an Modellorganismen konnten bereits eine Vielzahl an Genen mit der Entstehung von Kraniosynostosen assoziieren, darunter die Transkriptionsfaktoren TCF12 und TWIST1. Beim Menschen sind heterozygote Mutationen in TCF12 und TWIST1 mit Kraniosynostosen der Koronarnaht assoziiert. Bei Mäusen hingegen führt eine heterozygote Tcf12 Mutation nur in Kombination mit einer heterozygoten Twist1 Mutation zu Fusionen der Koronarnaht.
Der Zebrabärbling (Danio rerio, überwiegend auch Zebrafisch genannt) weist eine bemerkenswerte Ähnlichkeit bezüglich der Anatomie und Morphologie des Schädeldachs zum Menschen auf. Um die genaue Funktion von TCF12 bei der Ausbildung der Schädelnähte zu untersuchen, wurde im Rahmen dieser Arbeit der Zebrafisch als in vivo Modell für die Entstehung tcf12-induzierter Kraniosynostosen etabliert. Zu Beginn der Arbeit wurde das Expressionsmuster von tcf12 über die Entwicklung hinweg analysiert. Ein besonderer Fokus lag dabei auf einem Expressionsnachweis während der Entwicklung der Schädelplatten und der Schädelnähte. Ein erster Expressionsnachweis von tcf12 mittels PCR-Analysen und Whole-mount RNA in-situ Hybridisierungen zeigte eine breite Expression von tcf12 ab dem 1-3 Somiten Stadium an. Für tiefergehende in vivo Analysen wurden im Zuge dieser Arbeit tcf12:EGFP Reportergenlinien generiert. Mit diesen gelang ein Nachweis der tcf12 Expression entlang der Wachstumsfronten der Schädelplatten, innerhalb der Schädelnähte sowie im Periost und der Dura mater.
Mit den tcf12:EGFP Fischen als Referenz wurde in weiterführenden Experimenten die Aktivität drei hochkonservierter CNEs (engl. conserved non-coding elements) in vivo im Zebrafisch untersucht. Zwei der CNEs konnten als tcf12 Enhancer verifiziert werden, die eine Genexpression während der Neurogenese des zentralen Nervensystems (ZNS) steuern. Die beiden Enhancer-Elemente zeichnen sich durch eine hohe Konservierung vom Menschen bis hin zum Zebrafisch aus.
Aufgrund der unterschiedlichen Sensitivität gegenüber einem Funktionsverlust von TCF12 und TWIST1 in Mensch und Maus sollte die Auswirkung eines Knockouts der orthologen Gene auf die Entwicklung der Schädelnähte des Zebrafisches untersucht werden. Mittels CRISPR/Cas9 wurden verschiedene Knockout-Linien für die Gene tcf12, twist1a und twist1b generiert. Analysen der Knockoutmutanten zeigten, dass ein heterozygoter Verlust von tcf12 und twist1b in seltenen Fällen zu partiellen Fusionen der Koronarnähte im Zebrafisch führt. Des Weiteren konnte bei tcf12 und twist1b Einzel- und Doppelmutanten ein abnormes Wachstum der Schädelplatten im Bereich der Suturen beobachtet werden. Die Expressionsstudien und die Analysen der Knockoutmutanten deuten auf eine Regulation von TCF12 bei der Differenzierung der Stammzellen sowie der Proliferation der Osteoblasten innerhalb der Schädelnähte hin.
Um die Auswirkung von TCF12 Mutationen auf funktioneller Ebene zu untersuchen wurden im Verlauf dieser Arbeit Luciferase-Reporter Assays durchgeführt. Anhand dieser konnte nachgewiesen werden, dass Mutationen, die die basic helix-loop-helix (bHLH)-Domäne beeinträchtigen, die Transaktivierungsfähigkeit von TCF12 aufheben. Co-Transfektions-Experimente mit TWIST1 offenbarten eine Regulation der Transaktivierung von TCF12 durch TWIST1, sowohl im Menschen, als auch im Zebrafisch. Im Rahmen dieser Arbeit konnten die genauen Expressionsorte von TCF12 während der Morphogenese des Schädeldachs nachgwiesen und die Funktion von TCF12 und seinem Interaktionspartner TWIST1 bei der Entstehung von Kraniosynostosen weiter aufgeklärt werden.
Ziel dieser Arbeit war es, den Einfluss von IL-12 auf die Rezeptoren DNAM-1, TIGIT und CD96 auf NK-Zellen näher zu untersuchen. Wir konnten nachweisen, dass IL-12 den Rezeptor DNAM-1 hochreguliert. CD96 wurde nach 48-stündiger Inkubation mit IL-12 bei frisch isolierten NK-Zellen zunächst ebenfalls hochreguliert, nach längerer in vitro Kultur mit IL-2/IL-15 und anschließender Intervention mit IL-12 für 48h fiel CD96 allerdings wieder unter das Ausgangsniveau ab. Die höchste Steigerung der Expression durch IL-12 konnte an den Rezeptoren CD62L (Adhäsion) und CD161 (Inhibition) beobachtet werden.
Die Ergebnisse wiesen darauf hin, dass IL-12 einen Einfluss auf das Verhältnis der NK-Subpopulationen besitzt, indem es durch Hochregulation von CD56 und Herabregulation von CD16 zu einer Umwandlung von CD56dimCD16+ NK-Zellen zu CD56brightCD16- NK-Zellen beitrug. Während bei beiden Populationen DNAM-1 hochreguliert wurde, stieg die Expression von CD96 in der CD56dim Population, fiel aber in der CD56bright Population. Die Expression von TIGIT verhielt sich in der IL-15 Gruppe gegensätzlich dazu.
IFN-γ konnte die Expression der Liganden für DNAM-1, TIGIT und CD96 auf einer der untersuchten Tumorzelllinien (SK-ES-1) steigern.
Die Zytotoxizität von NK-Zellen konnte nicht durch IL-12 gesteigert werden. Indessen konnten wir feststellen, dass DNAM-1 für die Aufrechterhaltung der zytotoxischen Funktion essentiell war und eine Blockierung von DNAM-1 zu einer drastischen Verringerung derselben geführt hat. Dagegen konnten die NK-Zellen ihre Funktion nach der Blockierung von CD96 weitestgehend aufrechterhalten, es kam allerdings auch nicht zu einer gesteigerten Lyse von Tumorzellen.
Die Ergebnisse verdeutlichten, dass IL-12 zwar die Expression von DNAM-1 auf NK-Zellen zu steigern vermochte, dies allerdings nicht zu einer gesteigerten Zytotoxizität der NK-Zellen gegenüber den getesteten drei Tumorzelllinien geführt hat.
Introduction
Proximal ulna fractures are common in orthopaedic surgery. Comminuted fractures require a high primary stability by the osteosynthesis, to allow an early functional rehabilitation as fast as possible, to reduce long-term limitations of range of motion. Classical dorsal plating is related to wound healing problems due to the prominence of the implant. New low-profile double plates are available addressing the soft tissue problems by positioning the plates at the medial and lateral side. This study analysed whether, under high loading conditions, these new double plates provide an equivalent stability as compared to the rigid olecranon locking compression plate (LCP).
Materials and methods
In Sawbones, Mayo Type IIB fractures were simulated and stabilized by plate osteosyntheses: In group one, two low-profile plates were placed. In group two, a single dorsal plate (LCP) was used. The bones was than cyclically loaded simulating flexion grades of 0°, 30°, 60° and 90° of the elbow joint with increasing tension forces (150 , 150 , 300 and 500 N). The displacement and fracture gap movement were recorded. In the end, in load-to-failure tests, load at failure and mode of failure were determined.
Results
No significant differences were found for the displacement and fracture gap widening during cyclic loading. Under maximum loading, the double plates revealed a comparable load at failure like the single dorsal plate (LCP). The double plates failed with a proximal screw pull-out of the plate, whereas in the LCP group, in 10 out of 12 specimens the mode of failure was a diaphyseal shaft fracture at the distal plate peak.
Conclusion
Biomechanically, the double plates are a good alternative to the dorsal LCP providing a high stability under high loading conditions and, at the same, time reducing the soft tissue irritation by a lateral plate position.
The capabilities of small satellites have improved significantly in recent years. Specifically multi-satellite systems become increasingly popular, since they allow the support of new applications. The development and testing of these multi-satellite systems is a new challenge for engineers and requires the implementation of appropriate development and testing environments. In this paper, a modular network simulation framework for space–terrestrial systems is presented. It enables discrete event simulations for the development and testing of communication protocols, as well as mission-based analysis of other satellite system aspects, such as power supply and attitude control. Olfactory circuits change structurally and physiologically during development and adult life. This allows insects to respond to olfactory cues in an appropriate and adaptive way according to their physiological and behavioral state, and to adapt to their specific abiotic and biotic natural environment. We highlight here findings on olfactory plasticity and modulation in various model and non-model insects with an emphasis on moths and social Hymenoptera. Different categories of plasticity occur in the olfactory systems of insects. One type relates to the reproductive or feeding state, as well as to adult age. Another type of plasticity is context-dependent and includes influences of the immediate sensory and abiotic environment, but also environmental conditions during postembryonic development, periods of adult behavioral maturation, and short- and long-term sensory experience. Finally, plasticity in olfactory circuits is linked to associative learning and memory formation. The vast majority of the available literature summarized here deals with plasticity in primary and secondary olfactory brain centers, but also peripheral modulation is treated. The described molecular, physiological, and structural neuronal changes occur under the influence of neuromodulators such as biogenic amines, neuropeptides, and hormones, but the mechanisms through which they act are only beginning to be analyzed.
Neural stem cells (NSCs) have been recently identified in the inferior colliculus (IC). These cells are of particular interest, as no casual therapeutic options for impaired neural structures exist. This research project aims to evaluate the neurogenic potential in the rat IC from early postnatal days until adulthood. The IC of rats from postnatal day 6 up to 48 was examined by neurosphere assays and histological sections. In free-floating IC cell cultures, neurospheres formed from animals from early postnatal to adulthood. The amount of generated neurospheres decreased in older ages and increased with the number of cell line passages. Cells in the neurospheres and the histological sections stained positively with NSC markers (Doublecortin, Sox-2, Musashi-1, Nestin, and Atoh1). Dissociated single cells from the neurospheres differentiated and were stained positively for the neural lineage markers β-III-tubulin, glial fibrillary acidic protein, and myelin basic protein. In addition, NSC markers (Doublecortin, Sox-2, CDK5R1, and Ascl-1) were investigated by qRT-PCR. In conclusion, a neurogenic potential in the rat IC was detected and evaluated from early postnatal days until adulthood. The identification of NSCs in the rat IC and their age-specific characteristics contribute to a better understanding of the development and the plasticity of the auditory pathway and might be activated for therapeutic use.
Summary
In adult hypophosphatasia (HPP) patients, elevated lumbar spine dual X-ray absorptiometry (DXA) values are associated with markers of disease severity and disease-specific fracture risk while femoral bone mineral density (BMD), being largely unaffected by the disease severity, may still be useful to monitor other causes of increased fracture risk due to low BMD.
Introduction
Hypophosphatasia (HPP) is a rare inherited metabolic disorder due to deficient activity of the tissue-nonspecific alkaline phosphatase (TNAP). Clinical manifestation in adult HPP patients is manifold including an increased risk for fractures, but data regarding clinical significance of DXA measurement and associations with fracture risk and disease severity is scarce.
Methods
Retrospective single-center analysis of DXA scans in patients with confirmed HPP (documented mutation, clinical symptoms, low alkaline phosphatase activity). Further data evaluation included disease-related fractures, laboratory results (alkaline phosphatase, pyridoxalphosphate, phosphoethanolamine), and medical history.
Results
Analysis included 110 patients (84 female, mean age of 46.2 years) of whom 37.3% (n = 41) were harboring two mutations. Average T-Score level at the lumbar spine was − 0.1 (SD 1.9), and mean total hip T-Score was − 1.07 (SD 0.15). Both lower ALP activity and higher substrate levels (pyridoxalphosphate and phosphoethanolamine) were significantly correlated with increased lumbar spine T-Score levels (p < 0.001) while BMD at the hip was not affected by indicators of disease severity. Increased lumbar spine BMD was significantly associated with an increased risk for HPP-related fractures, prevalent in 22 (20%) patients (p < 0.001) with 21 of them having biallelic mutations.
Conclusion
BMD in adult HPP patients is not systematically reduced. Conversely, increased lumbar spine BMD appears to be associated with severely compromised mineralization and increased risk for HPP-related fractures while BMD at the hip appears unaffected by indicators of disease severity, suggesting suitability of this anatomic location for assessing and discerning disorders with increased fracture risk owing to reduced BMD like osteoporosis.

Tetrahydroisoquinolines (TIQs) such as salsolinol (SAL), norsalsolinol (NSAL) and their methylated derivatives N-methyl-norsalsolinol (NMNSAL) and N-methyl-salsolinol (NMSAL), modulate dopaminergic neurotransmission and metabolism in the central nervous system. Dopaminergic neurotransmission is thought to play an important role in the pathophysiology of chronic tic disorders, such as Tourette syndrome (TS). Therefore, the urinary concentrations of these TIQ derivatives were measured in patients with TS and patients with comorbid attention-deficit/hyperactivity disorder (TS + ADHD) compared with controls. Seventeen patients with TS, 12 with TS and ADHD, and 19 age-matched healthy controls with no medication took part in this study. Free levels of NSAL, NMNSAL, SAL, and NMSAL in urine were measured by a two-phase chromatographic approach. Furthermore, individual TIQ concentrations in TS patients were used in receiver-operating characteristics (ROC) curve analysis to examine the diagnostic value. NSAL concentrations were elevated significantly in TS [434.67 ± 55.4 nmol/l (standard error of mean = S.E.M.), two-way ANOVA, p < 0.0001] and TS + ADHD patients [605.18 ± 170.21 nmol/l (S.E.M.), two-way ANOVA, p < 0.0001] compared with controls [107.02 ± 33.18 nmol/l (S.E.M.), two-way ANOVA, p < 0.0001] and NSAL levels in TS + ADHD patients were elevated significantly in comparison with TS patients (two-way ANOVA, p = 0.017). NSAL demonstrated an AUC of 0.93 ± 0.046 (S.E.M) the highest diagnostic value of all metabolites for the diagnosis of TS. Our results suggest a dopaminergic hyperactivity underlying the pathophysiology of TS and ADHD. In addition, NSAL concentrations in urine may be a potential diagnostic biomarker of TS.
