Four new tetromycin derivatives, tetromycins 1-4 and a previously known one, tetromycin B (5) were isolated from Streptomyces axinellae Pol001(T) cultivated from the Mediterranean sponge Axinella polypoides. Structures were assigned using extensive 1D and 2D NMR spectroscopy as well as HRESIMS analysis. The compounds were tested for antiparasitic activities against Leishmania major and Trypanosoma brucei, and for protease inhibition against several cysteine proteases such as falcipain, rhodesain, cathepsin L, cathepsin B, and viral proteases SARS-CoV M(pro), and PL(pro). The compounds showed antiparasitic activities against T. brucei and time-dependent inhibition of cathepsin L-like proteases with K(i) values in the low micromolar range.
The technology of organic photovoltaics offers the possibility of low-cost devices due to easy fabrication procedures and low material consumption and at the same time high flexibility concerning the applied substrates or design features such as the color palette. Owing to these benefits, this research field is highly active, being reflected by the continuously rising number of publications. Chapter 1 gives an extensive overview of a part of these reports, namely the field of solution-processed BHJ organic solar cells using small molecules as electron-donating materials. In the early years of this research area (2006-2008), well known hole transporting materials such as triphenylamine based chromophores, oligothiophenes and polyaromatic hydrocarbons were applied. However, many of these dyes lacked absorption at longer wavelengths and were therefore limited in their light harvesting qualities. Later, chromophores based on low band gap systems consisting of electron-donating and electron-accepting units showing internal charge transfer overcame this handicap. Today, donor-substituted diketopyrrolopyrroles (D-A-D chromophores), squaraines (D-A-D chromophores) and acceptor substituted oligothiophenes (A-D-A chromophores) are among the most promising dyes for small molecule based organic solar cells with PCEs of 4-5%. This work is based on the findings of the groups of Würthner and Meerholz, which tested merocyanine dyes for the first time in organic BHJ solar cells.4 According to the Bässler theory85, the high dipolarity of these dyes should hamper the charge transport, but the obtained first results with PCE of 1.7% proved the potenital of this class of dyes for this application. Merocyanine dyes offer the advantages of facile synthesis and purification, high tinctorial strength and monodispersity. Additionally, the electronic structure of the dyes, namely the absorption as well as the electrochemical properties, can be adjusted by using the right combination of donor and acceptor units. For these reasons, this class of dye is highly interesting for the application in organic solar cells. It was the aim of the thesis to build more knowledge about the potential and limitations of merocyanines in BHJ photovoltaic devices. By screening a variety of donor and acceptor groups a comprehensive data set both for the molecular materials as well as for the respective solar devices was generated and analyzed. As one focus, the arrangement of the chromophores in the solid state was investigated to gain insight about the packing in the solar cells and its relevance for the performance of the latter. To do so, X-ray single crystal analyses were performed for selected molecules. By means of correlations between molecular properties and the characteristics of the corresponding solar cells, several design rules to generate efficient chromophores for organic photovoltaics were developed. This thesis concerned the quantification of cell adhesion molecules (CAM) in and on thin hydrogel films as surface modification of biomaterials. The established and well characterized, per se inert NCO-sP(EO-stat-PO) hydrogel system which allows the easy and reproducible bioactivation with peptides was used as basis for this thesis. Two methods can be used to functionalize the coatings. Ligands can either be mixed into the prepolymer solution in prior to layer formation (mix-in method), or freshly prepared coatings can be incubated with ligand solution (incubation method). Divided into three major parts, the first part of the thesis dealt with the concentration of ligands in the bulk hydrogel, whereas the second part of the thesis focused on the surface sensitive quantification of CAMs at the biointerface. The results were correlated with cell adhesion kinetics. The third part of this thesis investigated the biochemical and the structural mimicry of the extracellular matrix (ECM). ECM proteins were presented via sugar-lectin mediated binding and cell behavior on these surfaces was analyzed. Cell behavior on three-dimensional fibers with identical surface chemistry as the coatings in the previous sections of the thesis was analyzed and correlated with the amount of peptide used for bioactivation. Overall, the main question of this work was ‘How much?’ regarding maximal as well as optimal ligand concentrations for controlled cell-hydrogel interactions. The focus in the first practical part of this thesis was to analyze the amount of ligands in NCO-sP(EO-stat-PO) hydrogels using classical quantification methods. Coatings in 96-well plates as well as on glass were functionalized with GRGDS and 125I-YRGDS for radioisotopic detection (Chapter 3). Using the incubation method for functionalization, a maximal ligand binding using peptide concentrations of 600 µg/mL could be determined. When functionalization was introduced via the mix-in method, a clear tendency for higher ligand concentrations with increasing ligand to prepolymer ratio was observed, but no maximal ligand binding could be detected with a ligand to prepolymer ratio of 2/1 being the highest ratio investigated. This ratio of 2/1 was not exceeded to ensure that complete crosslinking of the hydrogel was not affected. In Chapter 4, a fluorinated amino acid and an iodinated peptide were immobilized to the hydrogels using the mix-in method and were detected by X-ray photoelectron spectroscopy (XPS) and time-of-flight secondary ion mass spectrometry (TOF-SIMS). In these measurements, maximal ligand binding was detected for a ligand to prepolymer ratio of 1/1. Higher ligand to prepolymer ratios did not result in any significant increase in ligand concentrations in the surface near regions of the crosslinked hydrogels. To address the question of how many ligands were actually accessible for cell interaction at the interface, surface sensitive quantification methods were applied in the second part of this thesis. For the quantification with surface plasmon resonance (SPR) and surface acoustic wave technology (SAW) (Chapter 5), the hydrogel coating procedure needed to be transferred onto cystamine functionalized gold surfaces. Characterization with ellipsometry and atomic force microscopy (AFM) revealed inhomogeneous cystamine binding to the activated surfaces, which resulted in inhomogeneous coatings. Nevertheless, it could be shown that SPR as well as SAW were suitable methods for the surface sensitive quantification of the ligand concentration on NCO-sP(EO-stat-PO) hydrogels. Non-functionalized coatings resisted non-specific serum as well as streptavidin (SA) adsorption. Coatings functionalized with biocytin and GRGDSK-biotin introduced specific SA binding that was dependent on the biotin concentration at the surface. Additionally, enzyme linked immunosorbent assay (ELISA) and enzyme linked lectin assay (ELLA) (Chapter 6) were applied to coatings in 96-well plates and on glass. Coatings were functionalized with the model molecule biocytin, the biotinylated peptide GRGDSK-biotin, the ECM protein fibronectin (FN), as well as the carbohydrates N-acetylglucosamine (GlcNAc) and N-acetyllactosamine (LacNAc). All ligands could be successfully detected with antibodies or SA via ELISA or ELLA. Maximal GRGDSK-biotin binding to the hydrogel coatings on glass was achieved at a peptide to prepolymer ratio of 1/5, which was used as reference value in Chapter 8. Last but not least, cell adhesion (Chapter 7) was quantified depending on the GRGDS concentration on hydrogel coatings on glass. Maximal adhesion of primary human dermal fibroblast (HDF) was observed at GRGDS to prepolymer ratios of 1/5, when adherent cells were counted on life cell images. Quantification of adherent cells using the CASY® cell counter revealed maximal HDF adhesion at molar ligand to prepolymer ratios of 1/2. However, cell vitality detected by intracellular enzyme activities was not dependent on the GRGDS concentration. Cells which managed to adhere were vital regardless of the amount of ligands present. Additionally, adhesion of fibroblasts from the murine cell line NIH L929 was analyzed by counting on life cell images. These cells, being much smaller than the HDF cells, needed higher GRGDS to prepolymer ratios (2/1) for proper cell adhesion. All quantification methods applied to analyze hydrogels which were functionalized by the mix-in method in Chapter 3, 4, 6 and 7, were compared in Chapter 8. Radiodetection gave information about the ligand concentrations throughout the whole hydrogel and no maximal amount of ligands could be detected when increasing the peptide to prepolymer ratio. In contrast, XPS and TOF-SIMS which only penetrated the surface near regions of the coating, a maximal ligand binding to the hydrogel was detected for 1/1 ratios. SPR and SAW were not included in this comparison, as the coatings on gold need to be optimized first. The two surface sensitive quantification methods (ELISA and HDF adhesion) could give information about the quantity of peptide which was sterically available for SA or cell binding. With these methods, maximal SA and cell binding was detected at ratios of 1/5. These results underline the importance of carefully compare the different methods. Beside ligand quantification on hydrogels, the third part of this thesis was concerned with the biochemical and structural mimicry of the ECM by advanced ECM engineering to design biomimetic biomaterials that are better accepted by cells and tissue. The subject of Chapter 9 was the biomimetic and flexible presentation of the ECM protein FN. FN was attached via sugar-lectin mediated binding to NCO-sP(EO-stat-PO) hydrogels. The build-up of the covalently immobilized sugar poly-N-acetyllactosamine (polyLacNAc), the subsequent non-covalent binding of the fungal galectin His6CGL2, and FN could be elegantly proven by fluorescent staining on coatings which were functionalized with the sugar by micro contact printing (MCP). Further experiments were carried out on build-ups, where polyLacNAc was immobilized on the hydrogel by incubation. Optimal parameters for the layer build-up were determined by ELLA/ELISA. Only the complete build-up induced proper adhesion of HDFs. Compared to tissue culture polystyrene (TCPS), cells adhered and spread faster on the biomimetic surfaces. The flexible presentation of FN allowed HDFs to rearrange homogenously immobilized FN into fibrillar structures, which seemed not to be possible when FN was adsorbed on glass or covalently bound directly to the hydrogel coatings. This new approach of a flexible and biomimetic presentation of an ECM protein allows new ways to design biomaterials with best possible cell-material interactions. The work described in Chapter 10 focused on the structural mimicry of the fibrous ECM structures by electrospinning of synthetic, bioactive, and degradable fibers. Poly(D,L-lactide-co-glycolide) (PLGA) and NCO-sP(EO-stat-PO) were electrospun out of one solution in an easy one-step preparation resulting in fibers with an ultrathin inert hydrogel layer at the surface. By adding GRGDS to the solution prior to electrospinning, specifically interacting fibers could be obtained. In comparison to PLGA, the adsorption of bovine serum albumin (BSA) could be reduced by 99.2%. As a control, the non-active peptide GRGES was immobilized to the fiber. These fibers did not allow cell adhesion, showing that the integrity of the hydrogel coated fibers was not affected by the immobilization of peptides. HDF adhesion was obtained by functionalization with GRGDS, leading to the adhesion, spreading, and proliferation of HDFs. Also mesenchymal stem cells (MSC) could adhere to GRGDS functionalized fibers. Additionally, for ligand quantification, the ELISA technique was successfully transferred to fiber substrates. To highlight the potential of the approaches for the biochemical and structural mimicry of the ECM, the sugar polyLacNAc was immobilized on the PLGA/sP(EO-stat-PO) fibers followed by the subsequent layer build-up with His6CGL2 and FN. These fibers triggered HDF adhesion.
Background: Xenobiotics represent an environmental stress and as such are a source for antibiotics, including the isoquinoline (IQ) compound IQ-143. Here, we demonstrate the utility of complementary analysis of both host and pathogen datasets in assessing bacterial adaptation to IQ-143, a synthetic analog of the novel type N,C-coupled naphthyl-isoquinoline alkaloid ancisheynine. Results: Metabolite measurements, gene expression data and functional assays were combined with metabolic modeling to assess the effects of IQ-143 on Staphylococcus aureus, Staphylococcus epidermidis and human cell lines, as a potential paradigm for novel antibiotics. Genome annotation and PCR validation identified novel enzymes in the primary metabolism of staphylococci. Gene expression response analysis and metabolic modeling demonstrated the adaptation of enzymes to IQ-143, including those not affected by significant gene expression changes. At lower concentrations, IQ-143 was bacteriostatic, and at higher concentrations bactericidal, while the analysis suggested that the mode of action was a direct interference in nucleotide and energy metabolism. Experiments in human cell lines supported the conclusions from pathway modeling and found that IQ-143 had low cytotoxicity. Conclusions: The data suggest that IQ-143 is a promising lead compound for antibiotic therapy against staphylococci. The combination of gene expression and metabolite analyses with in silico modeling of metabolite pathways allowed us to study metabolic adaptations in detail and can be used for the evaluation of metabolic effects of other xenobiotics.
Im Laufe der Evolution entwickelten Pflanzen, Bakterien, Pilze und Tiere eine Vielzahl von Signal-, Boten- und Abwehrstoffen. Diese für die Organismen oft lebensnotwendigen Sekundärmetabolite werden von den jeweiligen Produzenten oft sehr effizient und teilweise auch hoch stereoselektiv produziert. Die Effizienz und Selektivität, mit denen diese Stoffe mit den in der Natur zur Verfügung stehenden biosynthetischen Mitteln hergestellt werden, ist für Chemiker im Labor, trotz eines breiten Methodenrepertoires, eine große Herausforderung und oft gelingt die Laborsynthese nur über viele Stufen sowie in geringen Ausbeuten und/oder mit schlechten Enantio- oder Diastereoselektivitäten. Daher wird in der chemischen Synthese immer wieder die Neuentwicklung und Erweiterung von Methoden vorangetrieben. Neben der Synthese im Labor ist auch die Untersuchung von Biosynthesewegen sowie die Charakterisierung der dafür verantwortlichen Enzyme ein interessantes Forschungsgebiet, um von der Natur zu lernen und diese Erkenntnisse für die Entwicklung neuer Synthesestrategien und Methoden zu nutzen. Viele der bislang isolierten Verbindungen zeigen neben den für die produzierenden Organismen wichtigen Wirkungen auch Aktivitäten gegen für Menschen gefährliche Krankheitserreger. Daher ist die Isolierung und Strukturaufklärung neuer bioaktiver Naturstoffe als Leitstrukturen für neue Medikamente ein lohnendes Ziel für Wissenschaftler. In den letzten Jahren wurden aber immer häufiger bereits bekannte Verbindungen isoliert. Zur Vermeidung solcher Mehrfachisolierung werden auch die analytischen Methoden zur Identifikation und Strukturaufklärung stetig verbessert Einen wichtigen Stellenwert hat hierbei die Kopplung von Messgeräten wie z.B. MS oder NMR mit chromatographischen Anlagen wie GC, HPLC oder UPLC eingenommen. Die Kombination von Flüssigchromatographie-Anlagen mit chiroptischen Detektoren wie CD-Spektrometern erlaubt hierbei manchmal sogar die Zuordnung von Absolutkonfigurationen direkt aus dem Rohextrakt. Ziel der vorliegenden Arbeit war einerseits die Anwendung neuer Methoden bei der Entwicklung von Syntheserouten zu axialchiralen Naturstoffen und andererseits die Aufklärung der Konstitution und Konfiguration von Naturstoffen und synthetischen Verbindungen sowie die Untersuchung von Biosyntheseintermediaten.
Die Chlorophylle stellen in der Natur die wichtigsten Pigmente dar, weil sie verantwortlich für die Photosynthese sind und hierbei vielfältige Funktionen wahrnehmen, die sich aus ihrer Selbstassemblierung sowie den vorteilhaften optischen und Redox-Eigenschaften ergeben. Die in dieser Arbeit untersuchten semisynthetischen Zinkchlorine stellen Modellverbindungen des natürlichen Bacteriochlorophylls c (BChl c) der Lichtsammelsysteme (light-harvesting: LH) in Chlorosomen von Bakterien, jedoch ohne Proteingerüst, dar. Die entscheidenden Vorteile dieser Zinkchlorine (ZnChl) gegenüber den natürlichen BChls bestehen im einfachen semisynthetischen Zugang ausgehend von Chlorophyll a (Chl a), ihrer gesteigerten chemischen Stabilität sowie der Möglichkeit ihre Selbstassemblierung durch gezielte chemische Modifizierung der Seitenketten in der Peripherie zu steuern. Während bereits mehrfach über die vielversprechenden Redox- und excitonischen Eigenschaften von Aggregaten von ZnChl und natürlichem BChl c und den damit verbundene Voraussetzungen für Excitontransport über große Distanzen berichtet wurde, sind die Ladungstransporteigenschaften von Aggregaten der biomimetischen ZnChl bis heute unerforscht. Die vorliegende Arbeit beschäftigt sich mit der Aufklärung der Struktur von Aggregaten einer Vielzahl von semisynthetischen Zinkchlorophyllderivaten im Feststoff, in Lösung und auf Oberflächen durch die Kombination verschiedenster spektroskopischer, kristallographischer und mikroskopischer Techniken an die sich Untersuchungen zum Ladungstransport in den Aggregaten anschließen. Schema 1 zeigt die verschiedenen, in dieser Arbeit synthetisierten ZnChls, die entweder mit einer Hydroxy- oder Methoxygruppe in der 31-Position funktionalisiert sind sowie Substituenten unterschiedlicher Art, Länge und Verzweigung an der Benzylestergruppe in 172-Position tragen.Die Packung dieser Farbstoffe hängt entscheidend von ihrer chemischen Struktur ab. Während die ZnChls 1a, 2a, 3 mit 31-Hydroxygruppe und Alkylseitenketten (Dodecyl bzw. Oligoethylenglykol) gut lösliche stabförmige Aggregate bilden, lagern sich die analogen Verbindungen mit 31-Methoxygruppe (1b, 2b) zu Stapeln in Lösung und auf Oberflächen zusammen. Diese supramolekularen Polymere wurden im Detail in Kapitel 3 mit Hilfe von UV/Vis- und CD-Spektroskopie (circular dichroism: CD) sowie dynamische Lichtstreuung (dynamic light scattering: DLS) untersucht. Darüber hinaus lieferten temperaturabhängige UV/Vis- in Kombination mit DLS-Messungen wertvolle Informationen über die Aggregationsprozess dieser beiden Sorten von Aggregaten. Während sich die ZnChl 1a mit 31 Hydroxygruppe entsprechend dem isodesmischen Modell zu röhrenförmigen Aggregaten zusammenlagern, bilden sich die stapelförmigen Aggregate von 1b nach einem kooperativen Keimbildungs-Wachstums-Mechanismus (nucleation-elongation mechanism). Detaillierte elektronenmikroskopische Studien lieferten erstmals überzeugende Beweise für röhrenförmige Nanostrukturen der Aggregate des wasserlöslichen 31-Hydroxy Zinkchlorin 3. Die gemessenen Durchmesser der Röhren von ~ 5-6 nm dieser Aggregate liegen in hervorragender Übereinstimmung mit den Elektronenmikroskopie-Daten von BChl c Stabaggregaten in Chlorosomen (Chloroflexus aurantiacus, Durchmesser ~ 5-6 nm) und entsprechen damit dem von Holzwarth und Schaffner postulierten röhrenförmigen Modell... Im Einklang mit ihren hoch geordneten, robusten Strukturen, die sich eindimensional in einer Größenordnung von Mikrometeren erstrecken, sowie ihrer Fähigkeit zum effizienten Ladungs-trägertransport stellen diese selbstassemblierten Nanoröhren von ZnChls vielversprechende Ausgangsmaterialien für die Fertigung supramolekularer elektronischer Bauteile dar. Wissenschaftliche Bemühungen einige dieser Moleküle und ihre entsprechenden supramolekularen Polymere für die Fertigung von (opto-)elektronischen Bauteilen wie organischen Feldeffekttransistoren zu benutzten, stellen lohnende Aufgaben für die Zukunft dar...
In dieser Arbeit wurde die Kupfer(I)-katalysierte Cycloaddition von Alkinen und Aziden ("Click-Chemie") als effiziente, vielseitige und milde Reaktion für die Funktionalisierung von Nanodiamantpartikeln etabliert. Es wurden verschiedene Alkin- oder Azid-funktionalisierte Nanodiamantsysteme hergestellt und deren Reaktivität in Click-Reaktionen anhand vielfältiger Beispiele demonstriert. Hierzu wurden neben einfachen Testverbindungen auch komplexere Substanzen (z. B. ein Mannose-Derivat, Polytriazole, Fluoreszenzfarbstoffe, ein Thiazolium-Organokatalysator) durch Triazolsynthese immobilisiert. Zusätzlich wurde eine Methode entwickelt, die die Herstellung funktionalisierter und vollständig dispergierter Nanodiamant-Primärteilchen ermöglicht, wobei gezeigt wurde, dass dieser Weg auch für die Funktionalisierung durch Click-Chemie geeignet ist. Die Analyse der Nanodiamantpartikel erfolgte u. a. durch FT-IR, TGA, Elementaranalyse, Partikelgrößen- und Zetapotentialbestimmung, NMR, HR-TEM, UV / Vis sowie Fluoreszenzspektroskopie und -mikroskopie.
n this work the synthesis and analysis of chromophore functionalized spherical gold nanoparticles is presented. The optical, electrochemical and spectroelectrochemical properties of these hybrid materials are furthermore studied. The work therefore is divided into two parts. The first part deals with triarylamine and PCTM-radical functionalized gold nanoparticles. The focus thereby was on the synthesis and on the investigations of chromophore-chromophore interactions and gold core-chromophore interactions. The chromopores, especially triarylamines, were attached to the gold core via different bridging units and were studied with optical and electrochemical methods. The purity and dimensions of the nanoparticles was determined by 1H-NMR spectroscopy, diffusion ordered NMR spectroscopy (DOSY), TGA, XPS and STEM. Furthermore a cyclic voltammetry technique was used to determine the composition of the particles via the Randles-Sevcik equation. An analysis of these parameters led to a model of a sea urchin-shaped nanoparticle. Optical measurements of the particles revealed an anisotropic absorption behavior of the triarylamine units due to gold core-chromophore interaction. However this behavior depends strongly on the relative orientation of the transition dipole moment of the chromophore to the gold surface and the distance of the chromophore to the surface. Hence, the anisotropic behavior was exclusively detected in the spectra of the Au-Tara1 particles. The short and rigid pi-conjugated bridging unit thereby facilitates this gold core-chromophore interaction. It was shown from electrochemical investigations that the triarylamine units can be chemically reversibly oxidized to the triarylamine monoradical cation. Furthermore, the measurements revealed a strong interligand triarylamine-triarylamine interaction which was only seen for the Au-Tara1 particles. The long pi-conjugated bridging units of the Au-Tara2 and Au-Tara3 particles as well as the aliphatic bridging unit of Au-Tara4 prevent any detectable interligand interactions. One may conclude that both the gold core-chromophore and the interligand triarylamine-triarylamine interaction depend on the length and the rigidity of the bridging unit. The electron transfer behavior of the triarylamine units adsorbed onto the gold core was additionally studied via spectroelectrochemical (SEC) measurements which are able to reveal weaker interactions. The investigations of Au-Tara1 and Au-Tara2 revealed a significant strong coupling between neighboring triarylamine units which is due to through-space intervalence interactions. This behavior was not detected for Au-Tara3 or for Au-Tara4. The SEC analysis also revealed that these observed interligand interactions depend on the length and the rigidity of the bridging unit. Thus, the systematic variation of the bridging unit gave a basic insight in the optical and electrochemical properties of triarylamines, located in the vicinity of a gold nanoparticle. The second part of this work aimed at the synthesis of new molecules, denoted as SERS-markers, for immuno SERS applications. For this purpose, the SERS-markers were designed to have a Raman-active unit and a thiol group for chemisorptions to Au/Ag nanoshells. In cooperation with the group of Schlücker (University of Osnabrück) the SERS-markers were absorbed onto Au/Ag nanoshells, denoted as SERS-labels, and characterized. The SERS spectra of the SERS-labels exhibited intense and characteristic SERS-signals for each marker. For immuno SERS investigations SEMA3 was functionalized with a hydrophilic end unit. This marker was adsorbed onto an Au/Ag nanoshell and encapsulated with silica. An anti-p63 antibody was bound to the silica surface in order to generate a SERS-labeled antibody for the detection of the tumor suppressor p63 in benign prostate. Immuno-SERS imaging of prostate tissue incubated with SERS-labeled anti-p63 antibodies demonstrated the selective detection of p63 in the basal epithelium. The results show the potential of the method for the detection of several biomolecules in a multiplexing SERS experiment.
Einleitung: Die α-Hydroxycarbonsäuren 2 sind nicht nur wichtige chirale Bausteine für die organische Synthese,[20–25] sondern ebenfalls für den Menschen von großer Bedeutung aufgrund ihrer vielfältigen Anwendungen in der Lebensmittelbranche, Kosmetikindustrie und Medizin.[13–15] Aus diesen Gründen ist ein effizienter Zugang auch zu enantiomenreinen Säuren 2 notwendig. Viele natürlich vorkommende Vertreter sind aus dem chiral pool isolierbar, doch gerade für wissenschaftliche Zwecke sind häufig spezielle Strukturen von Interesse, die nur synthetisch darstellbar sind. Deshalb wurden, basierend auf unterschiedlichen Ansätzen, zahlreiche stereoselektive Routen zu α-Hydroxycarbonsäuren 2 entwickelt.[26–79] Eine schnelle und zugleich breit anwendbare Methode, 2 durch Hydroxylierung unfunktionalisierter Carbonsäuren 20 zu synthetisieren, sucht man jedoch vergebens. Das Ziel des ersten Teils dieser Arbeit bestand in der Entwicklung einer stereoselektiven Eintopfsynthese zur α-Hydroxylierung α-unfunktionalisierter Carbonsäurederivate 20 ... Fazit: Mit der MHACU von chiralen O-acylierten Oxazolin-N-oxiden (S)-37 wurde eine schnelle und einfache Eintopf-Synthese für α-Acyloxyoxazoline (S,S)-39, α-Hydroxymethylester (S)-30 und -säuren (S)-2 ausgehend von unfunktionalisierten Orthoestern 29 erarbeitet, die exzellente Diastereomeren- und Enantiomerenüberschüsse erlaubt (94–98% de/ee). Einleitung zum zweiten Teil der Arbeit: L-Prolin [(S)-222] gilt als einer der bedeutendsten Organokatalysatoren überhaupt und besticht durch seine strukturelle Einfachheit und seine erstklassigen katalytischen Eigenschaften.[230–232] Zudem stehen bei keinem zweiten Katalysator die materielle Verfügbarkeit und die chemische Anwendbarkeit in einem so idealen Verhältnis. Für einen Grundlagenforscher ist es daher eine besonders interessante Aufgabe, strukturelle Merkmale von L-Prolin [(S)-222] mit Erkenntnissen aus mechanistischen Untersuchungen zu verschmelzen, um so neue, noch leistungsstärkere Katalysatoren zu entwickeln. Fazit: Die vorgelegten Konzepte zur Verbesserung der Reaktivität von Pyrrolidin-basierten Katalysatoren erschienen zwar aussichtsreich, jedoch lieferten die bisherigen Umsetzungen noch keine brauchbaren Ergebnisse. Die in diesem Rahmen synthetisierten Aminosäuren und Isoxazolidine besaßen nur sehr geringe katalytische Aktivitäten. Weitere Arbeiten, zum Beispiel Veränderungen an den Katalysatorstrukturen, sind nötig, um eine erfolgreiche Umsetzung dieser Konzepte noch zu realisieren.
Für Diamantpartikel basiert diese Funktionalisierungschemie auf den bereits vorhanden Oberflächengruppen, die durch die Synthese und Reinigung entstanden sind. Die stabilste Anbindung wäre eine C-C-Verknüpfung der Diamantatome auf der Oberfläche mit den organischen Reagenzien. Um diese Verknüpfung zu erreichen, wurde die Oberfläche mittels Diels-Alder-Reaktion funktionalisiert. Grundvoraussetzung hierfür ist eine Oberfläche, die über eine ausreichende Menge an C=C-Doppelbindungen verfügt. Dies wurde durch eine thermische Behandlung erreicht. Die gewonnenen Ergebnisse zeigen, dass ab einer Behandlungstemperatur von 750 °C keine funktionellen Gruppen durch die eingesetzten Analysemethoden mehr nachgewiesen werden können. Die Elementaranalyse der einzelnen Proben zeigt einen deutlichen Anstieg des Kohlenstoffgehaltes. Nach erfolgreicher thermischer Behandlung des Diamanten wurde durch die Verwendung von Cyclopentadien und Anthracen als Dien gezeigt, dass eine Funktionalisierung über eine Diels-Alder-Reaktion generell möglich ist. Daher wurde als alternatives Dien ein o-Chinodimethan eingesetzt. Hierzu wurde eine systematische Untersuchung der Umsetzung der unterschiedlichen thermisch behandelten Proben mit α,α’-Dibrom-o-xylol durchgeführt. Hierdurch wurde gezeigt, dass der Ausgangsdiamant bei mindestens 750 °C behandelt werden muss, um eine messbare Oberflächenbelegung zu erhalten. Im Anschluss wurde die Funktionalisierbarkeit der eingeführten Arylgruppen überprüft. Durch die Einführung unterschiedlicher funktioneller Gruppen ist es dann möglich, Moleküle für spezielle Anwendungen anzubinden. Durch die Anwendung der klassischen Aromatenchemie war es möglich, die aromatischen Oberflächen zu nitrieren und sulfonieren. Die Sulfonsäuregruppe konnte partiell zum Thiol reduziert werden. Durch Umsetzung dieser Thiolgruppen war es möglich, einen mannosemodifizierten sowie einen farbstoffmodifizierten Diamanten herzustellen. Da die nachträgliche Einführung von Carboxylgruppen mit hohem synthetischem Aufwand verbunden ist, wurde alternativ ein carboxyltragender Vorläufer mit dem thermisch behandelten Diamant umgesetzt. Hierbei wurde eine Oberflächenbelegung von ca. 0.8 mmol g-1 erreicht (Ausgangsdiamant thermisch behandelt bei 750 °C). Im Anschluss wurde die Säuregruppe mit einem einfach geschützten Diamin weiterfunktionalisiert. Hierbei wurden unterschiedliche Methoden zur Amidbildung getestet. Als effektivste Methode stellte sich die Synthese über ein Säurechlorid heraus. Infolgedessen war eine weitgehende Umsetzung (ca. 94 - 88 %) der Säuregruppe möglich. Eine weitere wichtige funktionelle Gruppe für die Anbindung größerer Einheiten stellen die Amine dar. Diese lassen sich einfach darstellen, z. B. durch Reduktion von Cyanogruppen. Daher wurde 2,3-Bis-(brommethyl)-5,6-dicyanopyrazin als Edukt für die Funktionalisierung eingesetzt. Nach anschließender Reduktion der Cyanogruppen wurden primäre Amine erhalten. Die so erzeugte Aminogruppe wurde mit Biotin weiter funktionalisiert. Die bisher eingesetzten o-Chinodimethane sind ausschließlich durch 1,4-Eliminierung zugänglich. Eine sinnvolle Alternative zu diesem Vorläufer ist die Verwendung eines Cyclobutenderivates. Der Vorteil liegt in einer einfacheren Reinigung des Produktdiamanten. Als Testverbindung wurde 2,4-Bis-(methylsulfonyl)-5,6-dihydro-cyclobuta-[d]-pyrimidin verwendet. Nach erfolgreicher Umsetzung mit dem Diamanten wurden die Thioethergruppen in Sulfone überführt und in 2-Position wurde das Sulfon gegen eine Aminogruppe ausgetauscht. Neben den durchgeführten Funktionalisierungsreaktionen wurde eine nasschemische Nachweismethode zur Quantifizierung von primären Aminen entwickelt. Ein Vorteil des Verfahrens ist, dass der Nachweis auf einfache Weise durchführbar ist und nur 1 – 2 mg Diamantprobe nötig sind. Die gewonnen Ergebnisse zeigen, dass durch den Nachweis verlässliche Angaben über die genaue Aminmenge erhalten werden.
Nanodiamant weist aufgrund seiner herausragenden mechanischen, optischen und biokompatiblen Eigenschaften ein enormes Potential auf. Für eine erfolgreiche Anwendung muss dieser jedoch zunächst desagglomerisiert und an seiner Oberfläche funktionalisiert werden. In dieser Arbeit werden zwei unterschiedlichen Methoden zur Deagglomerierung und Oberflächenfunktionalisierung vorgestellt. Im ersten Teil wird die thermochemische Methode beschrieben, um den ungewöhnlich stark agglomerierten Detonationsnanodiamant zu deagglomerieren. Dabei wird die Strategie verfolgt, den Nanodiamant erst durch eine thermische Behandlung im Vakuum die vorhandenen Oberflächengruppe zu entfernen, π-Bindungen zu etablieren und dann dessen Oberfläche durch kovalente Bindungen zu modifizieren. Im zweiten Teil wird eine mechanochemische Methode für die Deagglomerierung und Oberflächenfunktionalisierung vorgestellt. Dabei wird ein völlig neuer Ansatz (BASD-Verfahren) entwickelt. Die Ultraschallbehandlung in Kombination mit zusätzlichen Mikrokeramikpartikeln eignet sich hervorragend, um Nanodiamant-agglomerate aufzubrechen. Zwei unterschiedliche Reaktionen mit zwei unterschiedlichen Nanodiamantsorten werden getestet: Eine einfache Kondensationsreaktion durch Silanisierung und eine Radikalreaktion über das Diazoniumsalz. In weiteren Experimenten kann gezeigt werden, dass sich die zuerst vorgestellte thermochemische Methode auch eignet, um aus fluoreszierenden NV-Nanodiamant kolloidalen NV-Nanodiamant mit funktionellen Gruppen herzustellen. Der letzte Teil der vorliegenden Arbeit beschäftigt sich mit der Herstellung von Nanodiamanten aus Diamantfilmen mit Hilfe des Top-down Ansatzes, da der gezielte und kontrollierte Aufbau der Gitterstruktur des Diamanten sowie die kontrollierte Einführung bisher nur durch CVD oder Ionenimplantationstechnik möglich ist.
