Via reduction of benzvalene (1) with diirnine tricyclo[3.1.0.02•6]hexane is obtained in good yield. The procedure renders 3, which has already been synthesized by Lemal and Shim, accessible much easier and in larger quantities. IH and 13C n.m.r. spectroscopic data are discussed. Both the thermal and the AgBF4-catalyzed rearrangernent of 3 yield 1,3-cyclohexadiene (8). - The ozonolysis of 1 with subsequent LiAIH4-reduction results in cis-I,3- bis(hydroxyrnethyl)cyclobutane (13a).

Diels-Alder reaction of dimethyl 1,2,4,5-tetrazine-3,6-dicarboxylate (5) with benzvalene (4), norbornene, and norbornadiene afforded the azo compounds 7 and 8. Theseare derivatives of 2,3-diazabicyclo[2.2.2]oct-2-ene as is azo compound 3, which had been obtained previously from 5 and 2 equiv of benzvalene (4). The photochemical extrusion of nitrogen from 3, 7, and 8 has been studied. Whereas 7 and 8 on direct irradiation in benzene gave rise exclusively to the bicyclo[2.2.0]hexane derivatives 9 and 10, respectively, from 3 in addition to the bicyclo[2.2.0]hexane 11, the diolefin 1l was formed. Diolefin 12 has cisdouble bonds in the nine-membered ring and is fixed in a boat conformation in a manner so that the two bicyclobutane systems approach each other very closely. This geometry suggests the unusual ring opening of the intermediate 1,4-cyclohexanediyl diradical from a boat conformation, which arises by inversion of the primarily generated boat conformation. Sensitized photolysis of 3 as weilasthat of ll produced the saturated isomer 13 of 11 and 12. The proximity of the bicyclobutane systems in 1l causes unprecedented reactions leading to cage compounds. When ll was heated at 90 °C, a rearrangement to the pentacyclic product 10 took place. Utilization of tetradeuteriated substrate ll-d4 supported a pathway with two diradical intermediates. Behaving in a convcntional manncr, bicyclobutane 9 and bis(bicyclobutane) 11 took up 1 and 2 equiv of thiophenol most probably in a radical-chain addition to give the thioethers 28 and 19, respectively. In contrast, bis(bicyclobutane) ll was converted by 1 equiv of thiophenol into cagc compound 30 in a process involving both the strained a systems. Heating at 80 °C subjected 30 to a reversible Copc rearrangement, resulting in a 6:1 mixture of 31 and 30. When it was treated with bromine, 11 was transformed to cage compound 38. This addition is believed to proceed via a cationic intermediate. The structure of cage compound 10 was established by a singlc-crystal X-ray analysis of dialcohol 11 prepared from 20 and methyllithium.

The γ-oxoketenes, which are formed from oxadiazinone Ja and strained cyclopentene der1vat1ves, are shown to undergo a pericyclic ring enlargement to give the title compounds 2a, 2b, and 5. In the case of 5, two configurations, one having a cis and the other a trans Iactone functionality, are in equilibrium.
Aus Benzvalen (J) und 3,6-Bis(2-pyridyl)-l,2,4,5-tetrazin wurde das Dihydropyridazin 2c erhalten, das ebenso wie die bekannten Dihydropyridazine 2a, b mit Cyclopropen in die entsprechende Azoverbindung 3 überging. Addition von Thiophenol an 3a, c und 5 lieferte unter Beteiligung des Bicyclobutan-Systems und der Azofunktion die Halbkäfigverbindungen 4a, c bzw. 6. Aus der Umsetzung von 7, in dem die Azobrücke durch zwei BicycJobutan- Systeme flankiert ist, mit Thiophenol sowie Bromtrichlormethan gingen die Halbkäfigverbindungen 8 bzw. 9 hervor. An diesen ~eaktionen wirken beide Bicyclobutan-Systeme und die Azofunktion mit.
Carbon-13 shieldings and one-bond \(^{13}\)C-H coupling constants of bicydo[2.1.1]hexane, bicydo[2.l.l]hex- 2-ene, tricydo[3.1.1.0\(^{2.4}\)]heptane and benzvalene are presented and compared. to the data of related. compounds. H a bicydo[3.1.0]hexane system is part of a rigid skeleton, the cydopropane ring exerts spedfk: 'Y substituent eflects of two ldnds. In the case of the bicyclobexane boat form an upfield shift of the C-3 signal is observed and in tbe esse of the chair form a downfield shift of 15-20 ppm. Compared to the corresponding cydopentanes the double bond in strained cydopentenes causes downfield shifts of the C-4 absorption. 1bis eftect increases witb increasing strain, reaching 8 45.9 ppm maximum in benzvalene. Hence it is tbe only known bicydo[l.l.O]butane baving 8 reversed order of carbon shieldings. The downfield shifts are e:xplained by means of simple orbital interaction schemes.

Phenyl(tricyclo[4.1.0.0\^(^{2,7}\)] hept-4-en-3-yl)sulfone 8 has been prepared in two steps from 4,S-dlbromohomobenzvalene (6) and deprotonated to give the title compound 9. The carbon-13 NMR spectrum of 9 reveals a considerable interaction between the allyl anion moiety and the bicyclobutane system.

In den Reaktionen von Tetracyanethylen (TCNE) und 5,6-Dichlor-2,3-dicyan-p-benzochinon mit Benzvalen haben wir kürzlich die ersten Beispiele für die lange gesuchte einstufige 1,4-Cycloaddition eines Alkens an ein Vinylcyclopropan gcfunden(I~J. Sie ist als [(.,2.+.2s)+ 112J-Prozeß der Dicls-Alder-Addition nahe verwandtllbl. Allerdings entsteht das betreffende TCNE-Addukt, ein Dihydrosemibullvalen-Derivat, nur in einer Ausbeute von wenigen Prozent. Die Hauptprodukte gehen aus einer Zwitterionischen Zwischenstufe hervor, die durch Anlagerung von TCNE an die Benzvalen-n-Bindung resultiert.

Das bekannte 6-0xo-2,5-diphenyl-6H-l,3,4-oxadiazin (1 a) und die neuen 2,5-Diarylderivate 1 b - g lagern Alkene an, und es entstehen stickstofIfreie Produkte. Im Falle von Cyclopropenen und Cyclobuten bilden sich die a,p-ungesättigten Siebenring-Enollactone 42 -45 bzw. das a,ß-ungesättigte Achtring-Enollacton 47. 42a und d erleiden an basischem Aluminiumoxid die H-Verschiebung zu den ß,y-ungesättigten Siebenring-Enollactonen 46a bzw. d. Bei weiteren Olefinen wurden die 'Y-Ketoketene des Typs 33 als Zwischenprodukte nachgewiesen oder sind aufgrund der Konstitution der Endprodukte wahrscheinlich. Aus 1 a werden mit Norbomen, Norbornadien, Cyc1openten, trans-Cycloocten und Styrol die Sechsring-Enollactone 10, 13, 15, 19 bzw. 21 und 22 erhalten. 10 isomerisiert mit Kalium-tert-butoxid zu 34, und die Methanolyse dieser Enollactone erbringt die diastereomeren Methy]ester 12 bzw. 35. Das y-Ketoketen 16 aus trans-Cycloocten nimmt schneller Wasser und Methanol zu 17 bzw. 18 auf als es zum Enollacton 19 cyclisiert. cis,trans-l,5-Cyc1ooctadien erbringt über die Stufe des nachgewiesenen y-Ketoketens 40 in einer intramolekularen [2 + 2J-Cyc1oaddition den Tricyclus 41. Tetracyclische Verbindungen des Typs 4 gehen aus der Umsetzung von Benzvalen (2) mit 1 hervor. Durch Röntgenstrukturanalysen wurden Konstitution und Konfiguration von 10, 19 und 41 aufgeklärt

By means of the BC NMR spectra of tricyclo{2.2.0~rfJ6Jhexane and thirteen of its derivatives the effects of substituents in endo-3- and endo-5-positions on the HC chemical shifts have been determined. The y-anti effects are at least as Jarge as in monosubstituted cyc1obutanes, where the shielding values of second-row hetero substituents exceed those in unstrained systems by far, and higher-row and carbon substituents still cause substantial upfield shifts. In the title system the y-anti effect of a substituent in the endo-3- and endo-5-position are operative additively, and thus shift the absorption of C-J upfieJd by a maximum of 27 ppm with respect to the unsubstituted hydrocarbon.

The 130 chemical shifts were determined of the carbons in 12 cycloheptanes, 21 cycloheptanols, and 8 cycloheptanones. In some cyc1oheptanols and cyc1oheptanones, the assignments have been obtained unambiguously by the synthesis of deuterated derivatives and the use of paramagnetic-shift reagents. Substituent effects for the different types of groups have been calculated. The most informative data about the cyc10heptane conformations were provided by the relatively well understood I' effects. The results are generally in,good agreement with predictions based on the twist-chair form, which has been predicted by Hendrickson to be the most stable conformation. Pairs of cis-trans isomers are found to have rather characteristic differences in their 130 spectra. This fact was used to assign the resonances found for cis-trans mixtures of methyl-substituted cyc1oheptanols to specific isomers.
Das exo/endo--lsomerenverhältnis Ja: Jb bei der bekannten [2 + 2]-Cycloaddition von Styrol an 1,2-Cyclohexadien (2) wurde als tempcraturabhingiaaefunden. Der Einsatz von (Z)-Dcutcriostyrollicfene den Beweis der Zweistufiakeit dieser Reaktion, und das Diradikal 4 wird als wahrscheinlichste Zwischenstufe anaesehen. Erhitzen von Jb auf 140-170°C führte zur Binstellung des thermodynamischen Gleichgewichts mit Ja (Ja:3b = 93:7), wobei wieder das Diradikal4 als Zwischenstufe fungieren dürfte. Mit Hilfe kinetischer Messungen ermittelte man die Aktivierungsparameter für das System Ja~ 3b. - Aus 2 und den Abfangreagenzien p-Methoxystyrol, 1,1-Diphenylethylen sowie 1-Phenylpropen gingen mit bescheidenen Ausbeuten die Titelverbindungen 6a, b, 7 bzw. 8 hervor. Analoa zu 2 wurde sein l-Methylderivat 13 aus 6,6-Dibrom-1-methylbicyclo[3.1.0]hexan (9) durch Methyllithium freigesetzt. In Gegenwart von Styrol entstand neben den Abfanaprodukten 14a, b auch das Dimere 12 von lJ. - Die 1H-NMR-Spektren der Titelverbindungen belegen eine starre Halbsesselkonformation des Cyclohexentcils mit äquatorial anellienem Cyclobutanring.

Im Rahmen der vorliegenden Arbeit wurden mit gespannten Ringen überbrückte p-Elektronensysteme untersucht. Im ersten Teil wurde die Synthese von 1,2,3,5-Tetrahydro-1,2,3-methenopentalen (I) verbessert und die Reaktivität dieses gespannten Cyclopentadiens untersucht. Die Diels-Alder-Addukte von I (Benzvalenderivate) waren bei Raumtemperatur nicht beständig, da sie einer Umlagerung entweder in das aromatische Valenzisomer oder in die formalen [2+2]-Addukte (bei PTAD, Tetracyanoethen) unterliegen. Die Addukte von TCNE und PTAD waren bei 233 K im NMR-Spektrum beobachtbar. Das aus der Reaktion von I mit Singulett-Sauerstoff (1O2) hervorgehende Endoperoxid nahm in für diese Substanzklasse präzedenzloser Weise ein weiteres Molekül 1O2 auf. Die dabei gebildete Verbindung konnte nicht beobachtet werden und fragmentiert auf zwei verschiedenen Wegen unter Bildung stabiler Folgeprodukte. Quantenchemische Rechnungen an den unsubstituierten Stammkörpern der Diels-Alder-Addukte ergaben eine starke Pyramidalisierung dieser Alkene (21.2° bzw. 20.3°). Die ungewöhnlichen 13C-NMR-chemischen Verschiebungen dieser Verbindungen sind vermutlich auf diesen Effekt zurückzuführen. Im zweiten Teil wurden die Bindungslängenalternanz in mit gespannten Ringen 1,2-überbrückten Benzolen und Pyridazinen untersucht. Alle durch Röntgenstrukturanalyse bestimmten Strukturen weisen gering, aber sigifikant alternierende Bindungslängen im Aromaten auf. In den Pyridazinen wurde eine Zunahme der Alternanz in der Reihenfolge Cyclopentan-<Bicyclo[1.1.0]butan-<Cyclobutan-Brücke gefunden, die durch eine quantenchemische Berechnung der Enthalpie einer Modellreaktion (überbrücktes Ethen + cis-Butadien ® 2,3-überbrücktes Butadien + Ethen) korrekt vorhergesagt wird.
Es wurde eine Vielzahl neuer, flüssigkristalliner Phthalocyanin-Sternmesogene synthetisiert. Die Struktur-Eigenschaftsbeziehungen und die thermotropen Eigenschaften neuer Phthalocyanin-Sternmesogene mit Freiraum sowie von sterisch überfrachteten Verbindungen wurden insbesondere hinsichtlich der Freiraumfüllung untersucht. Diesbezüglich wurde ein neuer supramolekularer, freiraumfüllender "Klick-Prozess" zwischen einem Molekül mit Freiraum und einem sterisch überfrachteten Molekül mit vier Fullerenen beobachtet. Die photophysikalischen Eigenschaften wurden zudem insbesondere im Hinblick auf die Anwendung für die Organische Photovoltaik untersucht.
In der vorliegenden Dissertation werden optisch aktive Hydroperoxide I als enantioselektive Oxidationsmittel in der Weitz-Scheffer-Epoxidierung von Enonen II eingesetzt. Dabei sollten zunächst die besten Reaktionsbedingungen für eine effektive asymmetrische Induktion gefunden werden, um anhand dieser den Mechanismus des enantioselektiven Sauerstofftransfers aufzuklären. In einer weiteren Studie werden Chinconin- und Chinconidin-abgeleitete optisch aktive Phasentransferkatalysatoren (PTK) IV zur asymmetrischen Epoxidierung von Enonen II mit racemischen Hydroperoxiden I genutzt, wobei vordergründig die kinetische Racematspaltung der verwendeten Hydroperoxide I untersucht werden sollte. Darauf aufbauend wurde eine höchst effektive Methode zur enantioselektiven Epoxidierung von Isoflavonen V mit kommerziell erhältlichen, achiralen Hydroperoxiden entwickelt. 1. Die Optimierung der Reaktionsbedingungen an Chalkon IIa zeigt, dass die höchste Enantioseitendifferenzierung mit (S)-(-)-1-Phenylethylhydroperoxid (Ia) und KOH in Schema A: Asymmetrische Weitz-Scheffer-Epoxidierung mit optisch aktiven Hydroperoxiden I und den Basen KOH oder DBU als Katalysatoren Acetonitril bei –40 °C möglich ist. Dabei bildet sich das (alphaS,betaR)-Epoxid IIIa in 51 Prozent ee. Im Gegensatz dazu wird in Toluol bei 20 °C mit der Base DBU das entgegengesetzt konfigurierte (alphaR,betaS)-Epoxid IIIa in einem Enantiomerenüberschuss von 40 Prozent gebildet. Die Art der Base beeinflusst demnach grundlegend den stereochemischen Verlauf der Reaktion. Um diesen Effekt mechanistisch zu ergründen wird der elektronische Charakter der Arylreste im Enon II systematisch variiert, was allerdings nur zu einer geringen Veränderung der Enantioselektivität führt. Einen größeren Einfluss auf das Ausmaß der Enantioseitendifferenzierung in dieser asymmetrischen Weitz-Scheffer-Epoxidierung hat, sowohl bei der Reaktionsführung mit DBU in Toluol als auch mit KOH in CH3CN, der sterische Anspruch des beta-Substituenten im Enon II. Aufgrund der maßgeblichen Signifikanz der Größe des beta-Substituenten wird eine Templatstruktur T+ (Abbildung A) vorgeschlagen, in der eine sterische Wechselwirkung zwischen dem beta-Substituenten des Enons II und dem Hydroperoxyanion I- den Abbildung A: Bevorzugte Anordnungen in der Templatstruktur für die KON-vermittelte und die DBU-vermittelte Epoxidierung stereochemischen Verlauf der Epoxidierung bestimmt. Das Aggregat aus Substrat, Hydroperoxid und Gegenion wird in Form eines Templats T+ durch das K+-Ion oder das protonierte Amin DBU-H+ zusammengehalten. Dadurch wird den entgegengesetzten Enantioselektivitäten Rechnung getragen, die für diese beiden Basen beobachtet werden. Aus Abbildung A wird ersichtlich, dass die unterschiedliche Größe der K+- oder DBU-H+-Kationen und des beta-Substituenten im Templat wichtig für eine effektive Diskriminierung der beiden möglichen Angriffe T+-(Si) und T+-(Re) ist. Für das relativ kleine Kaliumion dominiert die Wechselwirkung zwischen dem beta-Substituenten und dem Hydroperoxid I. Diese wird im T+-(Si)-Angriff minimiert, indem das Wasserstoffatom am stereogenen Zentrum des Hydroperoxids I auf der Seite des Enons II steht. In der Epoxidierung mit der sterisch anspruchsvolleren Base DBU tritt die Wechselwirkung zwischen DBU-H+ und dem Hydroperoxid im Templat in den Vordergrund, was den Angriff auf der Re-Seite bedingt. Demnach werden mit KOH die besten Enantioselektivitäten für große beta-Substituenten beobachtet, wohingegen für die Amin-vermittelte Epoxidierung eine große Base, wie DBU, vorteilhaft ist. Sowohl für KOH als auch für DBU als Basenkatalysatoren wird die Validität der Templatstruktur durch weitere Variation der Reaktionsbedingungen geprüft. Wenn K+ durch den Kronenether 18-Krone-6 komplexiert wird oder anstelle von DBU-H+ eine nicht-koordinierende Schwesinger Base verwendet wird, das Templat also nicht durch Koordination gebildet werden kann, werden deutlich niedrigere Enantioselektivitäten in der Epoxidierung beobachtet. Die Notwendigkeit der S-cis-Konformation des Enons II für die Bildung des Templats, wird durch Untersuchungen mit konformationell fixierten Enonen untermauert. So wird die Enantioselektivität bei der Epoxidierung eines S-cis-fixierten Enons (IIb) auf bis zu 90 Prozent ee erhöht, während sie bei einer S-trans-Fixierung des Enons deutlich auf < 5 Prozent ee abfiel. Fazit: Mit den optisch aktiven Hydroperoxiden I wird in der Weitz-Scheffer-Epoxidierung durch die Wahl geeigneter Basen, KOH oder DBU, sowohl das (alphaS,betaR)-Epoxid III (bis zu 90 Prozent ee) als auch das (alphaR,betaS)-Epoxid (bis zu 72 Prozent ee) erhalten. Welches Enantiomer überwiegt kann dabei allein durch die Wahl der Base gesteuert werden. Die Enantioseitendifferenzierung wird durch sterische Wechselwirkungen in einem Templat aus Enon II, Hydroperoxid I und den Kationen K+ oder DBU-H+ bestimmt. Die kinetische Racematspaltung chiraler Hydroperoxide I durch Weitz-Scheffer-Epoxidierung mit optisch aktiven Chinconin-basierten Phasentransferkatalysatoren (PTK) IV wird untersucht, bei der als willkommenes „Nebenprodukt" optisch aktive Isoflavonepoxide VI (Schema B) mit bis zu 92 Prozent ee entstehen. Die Racematspaltung ist Schema B: Kinetische Racematspaltung des chiralen Hydroperoxids Ia mittels Weitz-Scheffer-Epoxidierung und dem optisch aktiven PTK IV jedoch nicht effektiv, es werden ee-Werte von maximal 33 Prozent erzielt. Auf dieser Basis wird eine Methode zur asymmetrischen Epoxidierung der Isoflavonen (V) (Schema C) mit dem Schema C: Enantioselektivitäten für die Epoxidierung der Enone IIb,c und des Isoflavons Vb in Anwesenheit des PTK IV kommerziell verfügbaren Cumylhydroperoxid entwickelt, die für das Isoflavon Vb bis zu 98 Prozent ee zu Gunsten des (1aR,7aS)-Epoxids ergibt. Die hohe Enantioselektivität wird mit dem Templat A (Schema D) erklärt, in dem eine H-Brücke von der Hydroxy-Funktion des PTK IV Schema D: Wasserstoffbrückengebundene Templatstrukturen A und B zum endocyclischen Ethersauerstoffatom des Isoflavons V ausgeht. Die Relevanz einer solchen H-Brücke ist durch Methylierung der Hydroxy-Funktion des PTK IV demonstriert. Zudem ist die Wichtigkeit dieses Ethersauerstoffatoms durch die Tatsache untermauert, dass das konformationell ähnliche Enon IIc (Schema C) nahezu unselektiv epoxidiert wird (18 Prozent ee). Eine analoge H-Brücke nunmehr zum Carbonylsauerstoffatom des S-cis-fixierten Enons IIb wird als Erklärung für dessen hoch enantioselektive Epoxidierung (95 Prozent ee) postuliert (Templat B, Schema D). Fazit: Die asymmetrische Weitz-Scheffer-Epoxidierung mit dem optisch aktiven Phasentransferkatalysator IV wird zur Herstellung fast enantiomerenreiner Epoxide (bis zu 98 Prozent ee) genutzt. Für die Enantioseitendifferenzierung zeigt sich die Ausbildung einer H-Brücke zwischen PTK IV und Substrat II oder V als essentiell. In der kinetischen Racematspaltung chiraler Hydroperoxide I ist diese Epoxidierung nicht effektiv.

The present work is part of the currently only rudimentary understanding of the structure-property relationships in the self-assembly of pi-conjugated organic molecules. Such structures may reveal favorable photophysical and semiconducting properties due to the weak non-covalent pi-pi interactions between the monomer units. The specific mutual orientation of the dyes is known to evoke individual functional properties for the condensed matter, however, the related electronic processes are still not well-understood and further enhancements of functional properties are seldom triggered by rational design. The pi-pi self-assembly structures of perylene bisimide (PBI) dyes are promising, versatile materials for organic electronic devices and have been elected for this thesis as an archetype aggregate system to investigate the dye-dye interactions in more detail. In cooperation with experts in the field of spectroscopy and theory the development of reliable routines towards a better understanding of the origins of the functional properties may be feasible, and, on a longer time-line, such knowledge may enable optimization of functional organic materials. Having designed such structures entailed the challenge of developing feasible synthesis strategies, and to actually generate the targeted molecules by synthesis. Several synthesis approaches were conducted until finally a perylene bisimide foldamer was obtained based on a Sonogashira co-polymerization reaction. After purification and enrichment of the larger-sized species by means of semi-preparative gel permeation chromatography (GPC) the average size of an octamer (8500 Da) species was determined by analytical GPC. The low polydispersity index (PD) of 1.1 is indicative of a sharp size distribution of the oligomers. This average size was confirmed by performing diffusion ordered NMR spectroscopy (DOSY). Furthermore, MALDI-TOF mass analysis substantiated the structural integrity of the co-polymerization product. Solvent-dependent UV/vis spectroscopic investigations demonstrated that intramolecular PBI aggregates are reversibly formed, indicating that this oligomer is able to fold and unfold in the intended manner upon changing external conditions. In the unfolded states, the PBI moieties are closely arranged due to the short OPE bridges (< 2.4 nm), which is expressed by an exciton coupling interaction of the dyes and therefore the characteristic monomer absorption pattern of the PBI chromophore cannot be obtained in the unfolded states. More interestingly, the folded state revealed a pronounced aggregate spectrum of the PBIs, however, striking differences in the shape of the absorption spectrum compared to our previously investigated PBI self-assembly were obtained.
Within this thesis, synthetic strategies for self-assembled organic cage compounds have been developed that allow for both stimuli-responsive control over assembly/disassembly processes and spatial control over functionalization. To purposefully operate the reversible assembly of organic cages, boron-nitrogen dative bonds have been exploited for the formation of a well-defined, discrete bipyramidal organic assembly in solution. Thermodynamic association equilibria for cage formation have been investigated by Isothermal Titration Calorimetry (ITC). Temperature-dependent NMR studies revealed a reversible cage opening upon heating and quantitative reassembly upon cooling. For the spatial functionalization of organic cages, two divergent molecular building units have been designed and synthesized, namely tribenzotriquinacene derivatives possessing a terminal alkyne moiety at the apical position and a meta-diboronic acid having a pyridyl group at the 2-position. Facile access to a variety of apically functionalized tribenzotriquinacenes has been illustrated by post-synthetic modifications at the terminal alkyne group by Sonogashira cross-coupling and azide-alkyne click reactions. Finally, these apically functionalized tribenzotriquinacene building blocks have been implemented into boronate ester-based organic cage compounds showing modular exohedral functionalities.
Die photolytische Stickstoffabspaltung aus Azoalkanen vom DBH-Typ verläuft stereoselektiv unter bevorzugter Bildung des invertierten Hausans. Bei cyclopentenannelierten DBH-Derivaten kann die Selektivität in Abhängigkeit von den Brückenkopfsubstituenten auch umgekehrt sein. Bei der Photolyse von Azoalkan DBH-d2 zeigt sich, dass das Verhältnis von Inversions- zu von der Viskosität des Lösungsmittels abhängig ist. Die Viskosität wird sowohl durch Verwendung einer Serie von Alkoholen verschiedener Viskosität als auch durch Variation der Temperatur in n-Butanol geändert. Der Wert für die Photolyse in Acetonitril fügt sich in die Reihe der alkoholischen Solventien ein, womit eine Beteiligung von Wasserstoffbrücken ausgeschlossen ist. Der Viskositätseffekt ist mit einem schrittweisen Mechanismus der Stickstoffabspaltung vereinbar, der über ein unsymmetrisches Singulettdiazenyldiradikal verläuft. Die Abweichung der Viskositätprofile des kinv/kret-Verhältnisses für die Viskositätsänderung durch Lösungsmittel- und Temperaturvariation lässt einen kleinen aber messbaren Unterschied in der Aktivierungsenergie für den Inversions- und Retentionsprozess ableiten. Das kinv/kret-Verhältnis bei der Photolyse von DBH-d2 wird auch in Abhängigkeit vom Druck in superkritischem Ethan und Kohlendioxid untersucht, wofür zunächst eine spezielle Apparatur aufgebaut werden musste. Die Analyse der beobachteten Druckabhängigkeit im Hinblick auf Stoß- (Selbstdiffusionskoeffi-zient) und Reibungseffekte (Viskosität) lässt schließen, dass eine Behinderung des Inversionsprozesses durch Reibung mit Mediummolekülen die experimentellen Beobachtungen am besten erklärt. Dies entspricht den Beobachtungen in flüssiger Phase und bestätigt den Mechanismus. In einer vergleichenden Untersuchung der Photolyse (bei + 25 °C) von Azaolkan Ib und der thermischen (bei +25 °C) syn-zu-anti-Isomerisierung des entsprechenden Hausans IIb wird festgestellt, dass die kinv/kret-Verhältnisse bei der Photolyse des Azoalkans in einer Serie von Alkoholen und die Geschwindigkeitskonstanten kiso der Isomerisierung in der gleichen Reihe von Lösungsmitteln einer sehr ähnlichen Viskositätsabhängigkeit gehorchen. Daraus wird geschlossen, dass die Bewegung bei der Gerüstinversion in beiden Fällen durch Reibung mit Lösungsmittelmolekülen gehemmt und damit die Stereoselektivität bestimmt wird. Die Photolyse von DBH-d2 in Isooctan/Nujol-Gemischen zeigt die gleiche Viskositätsabhängigkeit des kinv/kret-Verhältnisses wie die in alkoholischen Medien. Aus dem Unterschied der absoluten kinv/kret-Werte der beiden Serien und durch die Verwendung weite-rer aprotischer Lösungsmittel wird eine Beeinflussung der Selektivität durch die "bulk" Polarität des Mediums festgestellt. Fazit: Durch die Untersuchung des Einflusses der Viskosität und Polarität des Lösungsmittels auf die Stereoselektivität bei der Photolyse von bicyclischen Azoalkanen und die thermi-sche Isomerisierung der entsprechenden Hausane wird das Auftreten eines Diazenyldiradi-kals als Schlüsselintermediat bestätigt und dynamische Effekte werden ausgeschlossen. Bei der Photolyse der cyclopentenannelierten Azoalkane Ic,d mit n-Propyl- und Acetoxy-methylsubstituenten an den Brückenkopfpositionen (Schema IV) entstehen unter Singulettbedingungen (direkte Photolyse bei höherer Temperatur) unter Retention hauptsächlich die anti-IIc,d Hausane. Unter Triplettbedingungen (direkte Photolyse bei tiefer Temperatur oder sensibilisierte Photolyse) wird das Inversionsprodukt syn-IIc,d bevorzugt. Die favorisierte Inversion beim Triplettweg wird mit der unsymmetrischen Natur der Brückenkopfsubstituenten nPropyl und Acetoxymethyl bei der Rotation um die Brückenkopfposition des planaren Cyclopentan-1,3-diyltriplettdiradikal erklärt. Die rotationsunsymmetrischen Brückenkopfsubstituenten stehen in ihrer Konformation niedrigster Energie (AM1-Rechnungen) auf der gegenüberliegenden Seite des Diylrings als der annelierte Cyclopentenring. Nach ISC führt der Ringschluss aufgrund sterischer Wechselwirkungen zwischen den Brückenkopfsubstituenten und der gem-dimethylsubstituierten Methylenbrücke bevorzugt zum syn-Hausan. Beim Vergleich des Verhältnisses von Inversion zu Retention bei der Photolyse des ungesät-tigten und gesättigten Azoalkans Ie und If (Schema IV) zeigt sich, dass bei beiden Derivaten unter Singulettbedingungen das syn-Hausan in etwa gleichem Ausmaß entsteht. Unter Triplettbedingungen führt die Photolyse zum Retentionsprodukt anti-IIe,f als Hauptdiastereomer, aber mit einem beträchtlichen Unterschied im syn/anti-Hausan-Verhältnis für Ie (38 : 62) und If (6 : 94). Dieser signifikante Unterschied der anti-Stereoselektivität im Triplettweg wird mechanistisch durch weitreichende sterische Wechselwirkungen zwischen dem annelierten Ring und der gem-dimethylsubstituierten Methylenbrücke während des Ringschlusses nach ISC des planaren Cyclopentan-1,3-diyltriplettdiradikals gedeutet. Im Gegensatz dazu ist die Denitrogenierung des intermediären Diazenyldiradikals Ie,f-1DZ (analog zu Ia-1DZ) im SH2-Prozess (Inversion) des Singulettwegs relativ unempfindlich gegenüber solchen sterischen Effekten zwischen den entfernten Substituenten. Fazit: Bei der Photolyse von fünfringannelierten Azoalkanen wirken sich kleinere strukturelle Variationen (rotationsunsymmetrische Brückenkopfsubstituenten oder die Hydrierung der Doppelbindung im annelierten Ring) vor allem im Triplettweg über sterische Wechselwirkungen im planaren Cyclopentan-1,3-diyltriplettdiradikal auf das Verhältnis der entstehenden syn/anti-Hausane aus. Der SH2-Prozess im Singulettweg ist relativ unempfindlich gegenüber solchen sterischen Effekten.

Nucleic acids are not only one of the most important classes of macromolecules in biochemistry but also a promising platform for the defined arrangement of chromophores. Thanks to their precise organization by directional polar and hydrophobic interactions, oligonucleotides can be exploited as suitable templates for multichromophore assemblies with predictable properties. To expand the toolbox of emissive, base pairing nucleobase analogs several barbituric acid merocyanine (BAM) chromophores with tunable spectroscopic properties were synthesized and incorporated into RNA, DNA and glycol nucleic acid (GNA) oligonucleotides. A multitude of duplexes containing up to ten BAM chromophores was obtained and analysis by spectroscopic methods revealed the presence of dipolarly coupled merocyanine aggregates with properties
strongly dependent on the chromophore orientation toward each other and the backbone conformation. These characteristics were exploited for various applications such as FRET pair formation and polymerase chain reaction (PCR) experiments. The observed formation of higher-order aggregates implies future applications of these new oligonucleotide-chromophore systems as light-harvesting DNA nanomaterials. Besides oligonucleotide templated covalent assembly of chromophores also non-covalent nucleic acid-chromophore complexes are a broad field of research. Among these, fluorogenic RNA aptamers are of special interest with the most versatile ones based on derivatives of the GFP chromophore hydroxybenzylidene imidazolone (HBI). Therefore, new HBI-derived chromophores with an expanded conjugated system and an additional exocyclic amino group for an enhanced binding affinity were synthesized and analyzed in complex with the Chili aptamer. Among these, structurally new fluorogenes with strong fluorescence activation upon binding to Chili were identified which are promising for further derivatization and application as color-switching sensor devices for example.
Exciton coupling between two or more chromophores in a specific environment is a key mechanism associated with color tuning and modulation of absorption energies. This concept is well exemplified by natural photosynthetic proteins, and can also be achieved in synthetic nucleic acid nanostructures. Here we report the coupling of barbituric acid merocyanine (BAM) nucleoside analogues and show that exciton coupling can be tuned by the double helix conformation. BAM is a nucleobase mimic that was incorporated in the phosphodiester backbone of RNA, DNA and GNA oligonucleotides. Duplexes with different backbone constitutions and geometries afforded different mutual dye arrangements, leading to distinct optical signatures due to competing modes of chromophore organization via electrostatic, dipolar, - stacking and hydrogen-bonding interactions. The realized supramolecular motifs include hydrogenbonded BAM–adenine base pairs and antiparallel as well as rotationally stacked BAM dimer aggregates with distinct absorption, CD and fluorescence properties.
We report the synthesis and spectroscopic analysis of RNA containing the barbituric acid merocyanine rBAM2 as a nucleobase surrogate. Incorporation into RNA strands by solid-phase synthesis leads to fluorescence enhancement compared to the free chromophore. In addition, linear absorption studies show the formation of an excitonically coupled H-type dimer in the hybridized duplex. Ultrafast third- and fifth-order transient absorption spectroscopy of this non-fluorescent dimer suggests immediate (sub-200 fs) exciton transfer and annihilation due to the proximity of the rBAM2 units.
Complexation properties of 2,2':6',2''-terpyridine (tpy) have been studied with a series of first row transition metal ions by UV-vis, 1H NMR and isothermal titration calorimetry and ƒ´H values for the tpy complexation processes have been determined. These studies reveal that Zn2+ is the best suited metal ion for the reversible coordination of the terpyridine ligand. Thus, supramolecular coordination polymerization of perylene bisimide fluorophores containing terpyridine functionalities have been investigated by using Zn2+ as metal ion. The formation of the dimeric complexes in the case of monotopic model comounds and coordination polymerization of ditopic functional building blocks have been confirmed by 1H NMR studies. The optical properties of dimeric and polymeric complexes have been investigated by UV-vis and fluorescence spectroscopy. The Zn2+ coordination to the terpyridine unit does not effect the advantageous fluorescence properties of perylene bisimide moieties. The reversibility of the formation of coordination polymers has been established by 1H NMR and additionally by DOSY NMR and fluorescence anisotropy measurements. Coordination polymer strands can be visualized by atomic force microscopy (AFM), which also reveals the formation of an ordered monolayer film at higher concentration. The average polymer length has been determined by AFM to 15 repeat units, which correlates well with the value estimated by 1H NMR to >10 repeat units.
Natural light harvesting as well as optoelectronic and photovoltaic devices depend on efficient transport of energy following photoexcitation. Using common spectroscopic methods, however, it is challenging to discriminate one-exciton dynamics from multi-exciton interactions that arise when more than one excitation is present in the system. Here we introduce a coherent two-dimensional spectroscopic method that provides a signal only in case that the presence of one exciton influences the behavior of another one. Exemplarily, we monitor exciton diffusion by annihilation in a perylene bisimide-based J-aggregate. We determine quantitatively the exciton diffusion constant from exciton–exciton-interaction 2D spectra and reconstruct the annihilation-free dynamics for large pump powers. The latter enables for ultrafast spectroscopy at much higher intensities than conventionally possible and thus improves signal-to-noise ratios for multichromophore systems; the former recovers spatio–temporal dynamics for a broad range of phenomena in which exciton interactions are present.
Substituierte Tetrahydrofurane sind Bausteine vieler pharmakologisch interessanter Natur- und Wirkstoffe. Deshalb ist die Entwicklung effizienter Methoden zur Darstellung dieser Verbindungsklasse von großer Bedeutung. Bei den Übergansmetall-katalysierten Verfahren erwiesen sich sowohl Vanadium(V)-, als auch Cobalt(II)-Komplexe als besonders vielversprechende Oxidationskatalysatoren. Bis dato ist allerdings kein universelles Verfahren bekannt, mit welchem nur durch Wahl der Reaktionsbedingungen ein Substrat unter vollständiger Regio- und Stereokontrolle selektiv in einen definierten cyclischen Ether umgewandelt werden kann. Ziel dieser Arbeit war daher die Ausarbeitung von Vanadium(V)- und Cobalt(II)-Komplex-katalysierten Oxidationen zur selektiven Synthese substituierter Tetrahydrofurane. Die Reaktionen wurden ausgehend von Bishomoallylalkoholen entwickelt, optimiert und mechanistisch untersucht. Für die Vanadium(V)-katalysierten Oxidationen wurden Reaktionen evaluiert, die nach dem Peroxo- (H2O2 als Primäroxidans) und solche, die nach dem Peroxy-Mechanismus (TBHP als Primäroxidans) verlaufen. Bei den Cobalt(II)-katalysierten Oxidationen wurde molekularer Sauerstoff als Primäroxidans eingesetzt. Im Zentrum des Interesses stand weiterhin die breit angelegte Synthese neuartiger Komplexe, welche auf ihre Eignung als Katalysatoren hin überprüft wurden. Die Metallionen wurden durch strukturell unterschiedliche Liganden stabilisiert, die auf Grund ihrer sterischen und elektronischen Natur Reaktivitäts- und Selektivitätseinflüsse bei den oxidativen Cyclisierungen ausüben sollten. Hierbei kamen Picolin- und Hydroxamsäure-, Diketonat-, sowie macrocyclische Diamin-Schiffbase-Liganden zum Einsatz. In Test-Oxidationen unterschiedlicher Bishomoallylalkohole zur Optimierung der Reaktionsbedingungen hinsichtlich Solvens, Primäroxidans und Reaktionstemperatur erwiesen sich für Vanadium-katalysierte Oxidationen TBHP in Dichlormethan und H2O2 in tert-Butanol bei jeweils 25°C am geeignetsten. Für Cobalt(II)-katalysierte Oxidationen konnten molekularer Sauerstoff als Primäroxidans, Isopropanol als Solvens und eine Reaktionstemperatur von 60°C als Reaktionsparameter der Wahl ermittelt werden. Im Folgenden wurden Vanadium(V)-katalysierte Oxidationen unterschiedlich substituierter bishomoallylischer Alkohole unter optimierten Reaktionsbedingungen durchgeführt. Sämtliche Oxidationen führten regioselektiv zu Tetrahydrofuranen als Hauptprodukte. Die gewonnenen Erkenntnisse zur Vanadium(V)-katalysierten Darstellung substituierter Tetrahydrofurane wurden im Rahmen der Synthese eines Naturstoff-Derivats angewandt. Ziel war es dabei in einer vierstufigen Totalsynthese Derivate der natürlich vorkommenden Carboline Cyclocapitellin und Isocyclocapitellin zu synthetisieren. Die auf diese Weise erhaltenen enantiomerenreinen Indolalkaloide werden zur Zeit pharmakologischen Testungen unterzogen. In einem weiteren Projekt wurden aerobe Cobalt(II)-katalysierte Oxidationen unterschiedlich substituierter Bishomoallylalkohole durchgeführt. Wie bei den Vanadium-katalysierten Reaktionen konnten auch hier regioselektiv Tetrahydrofurane als Hauptprodukte erhalten werden. Abschließend wurden durch Oxidation prochiraler Alkenole in Gegenwart chiraler Cobalt(II)-Diketonat-Komplexe enantiomerenangereicherte Tetrahydrofurane dargestellt. Diese Oxidationen lieferten nur geringe Enantioselektivitäten, welche durch Verwendung von Propionaldehyddiethylacetal als Solvens gesteigert werden konnten. Zusammenfassend kann festgestellt werden, dass sich für Vanadium(V)- und Cobalt(II)-katalysierte Oxidationen, im Hinblick auf die Substituenten in unmittelbarer Nähe der Alkenol-Doppelbindung, ein komplementäres Reaktivitäts- und Selektivitätsbild ergibt. So werden bei Cobalt(II)-katalysierten Oxidationen Alkenole ohne terminale Methylgruppen in höheren Ausbeuten und Stereoselektivitäten zu Tetrahydrofuranen oxidiert, bei Vanadium(V)-Katalysen hingegen 5,5-Dimethyl-substituierte Alkenole.
Naphthylisochinolin-Alkaloide stellen eine Klasse pharmakologisch, biogenetisch und chemotaxonomisch interessanter Sekundärmetabolite dar, die ausschließlich in den beiden tropischen Pflanzenfamilien Ancistrocladaceae und Dioncophyllaceae vorkommen. Diese Biarylnaturstoffe zeigen viel versprechende Wirksamkeiten gegen zahlreiche Pathogene und sind somit ein lohnendes Ziel zur Isolierung. In der vorliegenden Dissertation wurden zwei neue Ancistrocladus-Arten aus Ostafrika (A. tanzaniensis) und Malaysia (A. benomensis) sowie die bekannte indische Art Ancistrocladus heyneanus phytochemisch und chemotaxonomisch untersucht. Es wurden insgesamt zehn neue und fünf bekannte Naphthylisochinolin-Alkaloide isoliert, die einer breit angelegten biologischen Testung zugeführt wurden. Weitere Zielsetzungen bestanden in der stereochemischen Analyse von Naturstoffen unserer Kooperationspartner und in der methodischen Erweiterung der phytochemischen Analytik.
Ziel der Dissertation „Oktapeptide als neue Organokatalysatoren zur Hydrolyse von Phosphaten und Estern“ war es neue Oktapeptide zu finden, die die Fähigkeit besitzen Phosphate und Ester zu Hydrolysieren. In der Natur ist bei den Katalysereaktionen die Sekundärstruktur von entscheidender Bedeutung. Aus diesem Grunde wurde im Rahmen dieser Arbeit zunächst ein Modellsystem entwickelt, mit dem gezeigt werden konnte, dass es möglich ist mit einfachen Bausteinen wie Diaminobutan und dem in der Gruppe von Schmuck entwickelten Guanidiniocarbonylpyrrol ein Molekül zu entwickeln, welches eine stabile intramolekulare Schleife in polaren Lösungsmitteln ausbildet. Aufgrund der geringen Größe des Moleküls, konnte kein β-Faltblatt gebildet werden. Dennoch konnte mit Hilfe der NMR-Spektroskopie gezeigt werden, dass in dem polaren Lösungsmittel Methanol eine stabile Schleifenbildung mit einer intramolekularen Komplexierung stattfindet. Nach erfolgreicher Synthese des oben beschriebenen Testsystems wurde als nächster Schritt eine kombinatorische Organokatalysatorbibliothek mit 625 Mitgliedern aufgebaut. Die Struktur der Peptide kann man in drei Teile untergliedern. Der erste Teil ist die feste Phase, das Amino-TentaGel, an das nacheinander die einzelnen Aminosäuren gekuppelt wurden. An Stelle der Butylgruppe als Schleifenelement wurde Aib-D-Pro als β-Turn Element eingesetzt. Den dritten Teil bilden die bei der Synthese der Oktapeptide eingesetzten Aminosäuren AA1, AA3, AA6, AA8, die ein β-Faltblatt ausbilden sollten. Die kombinatorische Synthese der Bibliothek erfolgte nach der „Split and Mix“ Methode. Zum Unterscheiden der einzelnen Mitglieder untereinander, wurde die feste Phase zusammen mit einem Radiofrequenzchip in IRORI MikroKans gegeben. Durch die unterschiedlichen Aminosäuren ist die Bibliothek für die Katalyse in polaren Lösungsmitteln wie Wasser konzipiert worden. Als exemplarische Katalysereaktionen wurden dabei zwei Hydrolysereaktionen (Phosphatspaltung und Esterspaltung) ausgesucht. Zunächst wurde für beide Reaktionen ein Screening mit unterschiedlichen Bedingungen durchgeführt. Dabei hat sich gezeigt, dass bei der Phosphatspaltung nur dann die Reaktion katalysiert wurde, wenn das künstliche Argininanalogon, welches in unserer Arbeitsgruppe synthetisiert wurde, vorhanden war. Der beste Katalysator hat die Reaktion 175-mal schneller katalysiert als die unkatalysierte Reaktion. Bei dem Screening der Esterspaltung hat sich herausgestellt, dass die Aminosäure Histidin essentiell für die katalytische Aktivität ist. Der beste Katalysator bei der Esterspaltung hat die Hydrolyse des Esters 345-mal schneller katalysiert als die unkatalysierte Reaktion. Bei beiden Reaktionen hat sich gezeigt, dass die Sequenz der Katalysatoren sehr wichtig für die Katalyse ist. So verringert z.B. bei der Esterhydrolyse der Austausch zweier Aminosäuren eine Verringerung der Aktivität von dem Faktor 294 auf den Beschleunigungsfaktor 35. Auch konnten beide Katalysereaktionen in wässrigem gepufferten Lösung durchgeführt werden. Damit ist es möglich gewesen neue Oktapeptide für die Katalyse von Phosphat- und Esterspaltung zu finden und diese erfolgreich im Screening als auch in Lösung zu untersuchen.
In this work, a series of redox cascades was synthesised and investigated in view of their photophysical and electrochemical properties. The cascades are based on a perchlorinated triphenylmethyl radical acceptor and two triarylamine donors. Absorption spectra showed the presence of charge-transfer bands in the NIR range of the spectra, which pointed to the population of a charge-transfer state between a triarylamine donor and the radical acceptor. A weak to moderate emission in the NIR range of the spectra was observed for all compounds in cyclohexane. Spectroelectrochemical measurements were used to investigate the characteristic spectral features of the oxidised and reduced species of all compounds. Transient absorption spectra in the ns- and fs-time regime revealed an additional hole transfer in the cascades between the triarylamine donors, resulting in a charge-separated state. Charge-separation and -recombination processes were found to be located in the ps-time regime.
A fine balance of regulatory (T\(_{reg}\)) and conventional CD4\(^+\) T cells (T\(_{conv}\)) is required to prevent harmful immune responses, while at the same time ensuring the development of protective immunity against pathogens. As for many cellular processes, sphingolipid metabolism also crucially modulates the T\(_{reg}\)/T\(_{conv}\) balance. However, our understanding of how sphingolipid metabolism is involved in T cell biology is still evolving and a better characterization of the tools at hand is required to advance the field. Therefore, we established a reductionist liposomal membrane model system to imitate the plasma membrane of mouse T\(_{reg}\) and T\(_{conv}\) with regards to their ceramide content. We found that the capacity of membranes to incorporate externally added azide-functionalized ceramide positively correlated with the ceramide content of the liposomes. Moreover, we studied the impact of the different liposomal preparations on primary mouse splenocytes in vitro. The addition of liposomes to resting, but not activated, splenocytes maintained viability with liposomes containing high amounts of C\(_{16}\)-ceramide being most efficient. Our data thus suggest that differences in ceramide post-incorporation into T\(_{reg}\) and T\(_{conv}\) reflect differences in the ceramide content of cellular membranes.
Die vorliegende Arbeit behandelt die Synthese sowie die Eigenschaften einer Serie von organofunktionellen α-, β-, γ- und ζ-Silanen, mit einem Fokus auf Alkoxy(aminoalkyl)silanen. Der Großteil dieser Modellstrukturen wurde anschließend hinsichtlich ihrer Hydrolysekinetik in Abhängigkeit der Art der funktionellen Gruppe X (NMe3+, N(H)COOMe, N(Me)COOMe, NH2, N(H)Me, NMe2, Pip, Me), des Abstandes des Substituenten X zu dem Silicium-Atom
(α-, β-, γ- und ζ-Position), der Alkoxy-Abgangsgruppe am Silicium-Atom (MeO, iPrO, tBuO) und des pD-Wertes der Reaktionslösung systematisch untersucht.
Eine große Herausforderung dieser Studie war die Synthese von β-Amino-funktionalisierten Alkoxysilanen, deren Chemie aufgrund ihrer Labilität bisher kaum erforscht ist. Die einzigen literaturbekannten Vertreter stellten bislang das Trialkoxysilan (EtO)3Si(CH2)2NH2 (1) und sein Dialkoxy-Derivat (EtO)2SiMe(CH2)2NH2 (2) dar, welche durch Reaktion des entsprechenden 2-(Chlorethyl)silans mit Ammoniak unter hohem Druck im Autoklaven zugänglich sind. Unter Verwendung dieser Synthesemethode konnte sowohl die Synthese der Silane 1 und 2 reproduziert, als auch das Trimethoxy-Analogon (MeO)3Si(CH2)2NH2 (3) erstmals dargestellt werden.
Darüber hinaus wurde eine Serie von organofunktionellen Monoalkoxysilanen des Typs RORSiMe(CH2)2X und ROSiMe2C(H)MeCH2X (4b–18b) im präparativen Maßstab analyserein dargestellt.
Des Weiteren wurden die entsprechenden α-Silane 8a, 11a, 14a und 15a, die γ-Silane 6c, 8c, 11c, 13c–15c und 18c sowie die ζ-Silane 19 und 20 erstmals dargestellt.
Weiterhin wurden die bereits literaturbekannten α-Silane 16a–18a und γ-Silane 7c, 16c und 17c für die Verwendung in den Hydrolyseexperimenten synthetisiert.
Die Charakterisierung aller im Rahmen dieser Arbeit synthetisierten Verbindungen erfolgte mittels NMR-Spektroskopie (1H-, 13C-, 15N- und 29Si-NMR) und Elementaranalysen (C, H, N) bzw. HRMS-Experimente.
Die hydrolytische Spaltung der Si–OC-Bindung in Alkoxy(aminoalkyl)silanen stellt einen technisch sehr wichtigen Schlüsselschritt in der Synthese von Amino-funktionalisierten Polysiloxanen dar. Um den Mechanismus dieser Si–OC-Bindungsspaltung besser zu verstehen, wurden die Alkoxysilane 4b, 4c, 5b, 6b, 6c, 7b, 7c, 8a–8c, 9b, 11a–11c, 12b, 14a–14c,
15a–15c, 16a–16c, 17a–17c, 18a–18c, 19 und 20 hinsichtlich ihrer Hydrolysekinetik in CD3CN/D2O unter sauren und basischen Bedingungen mittels 1H-NMR-Spektroskopie untersucht.
Die Ergebnisse dieser Struktur–Reaktivitäts-Studie zeigten, dass die beobachteten unterschiedlichen Hydrolysegeschwindigkeiten das Resultat mehrerer Faktoren sind, wie beispielsweise elektronische und sterische Effekte, der große Einflusses des pD-Wertes und auch intramolekulare N–H∙∙∙O-Wasserstoffbrückenbindungen zwischen der protonierten Amino-Gruppe und der Alkoxy-Abgangsgruppe. Da der Einfluss dieser Effekte auf die Reaktivität der untersuchten α-, β-, γ- und ζ-Silane sehr unterschiedlich ist, kann kein klarer Zusammenhang zwischen der Hydrolysereaktivität und der Positionierung der stickstoff-haltigen funktionellen Gruppe (α-, β-, γ- und ζ-Position) erkannt werden. Die jeweils beobachtete Reaktivität entspricht vielmehr einer Summe aller zuvor genannten Teileffekte.
Die Erkenntnisse, die im Rahmen dieser Arbeit erhalten wurden, ermöglichen ein verbessertes grundlegendes Verständnis der Reaktivität von funktionalisierten α-, β-, γ- und ζ-Silanen, und sind für die Silicon-Industrie von großem Interesse, da sie eine gezieltere Anwendung der α-, β- und γ-Aminosilane in der Synthese von technisch wichtigen Amino-funktionalisierten Polysiloxanen erlauben.
Chemical investigation of the methanolic extract of the Red Sea cucumber Holothuria spinifera led to the isolation of a new cerebroside, holospiniferoside (1), together with thymidine (2), methyl-α-d-glucopyranoside (3), a new triacylglycerol (4), and cholesterol (5). Their chemical structures were established by NMR and mass spectrometric analysis, including gas chromatography–mass spectrometry (GC–MS) and high-resolution mass spectrometry (HRMS). All the isolated compounds are reported in this species for the first time. Moreover, compound 1 exhibited promising in vitro antiproliferative effect on the human breast cancer cell line (MCF-7) with IC\(_{50}\) of 20.6 µM compared to the IC50 of 15.3 µM for the drug cisplatin. To predict the possible mechanism underlying the cytotoxicity of compound 1, a docking study was performed to elucidate its binding interactions with the active site of the protein Mdm2–p53. Compound 1 displayed an apoptotic activity via strong interaction with the active site of the target protein. This study highlights the importance of marine natural products in the design of new anticancer agents.
Reliable prediction of the isotropic hyperfine coupling constant A\(_{iso}\) is still a difficult task for ab initio calculations. Strang dependence on the method employed for its ca1culation has been found. Within a CI ansatz A\(_{iso}\) is considerably affected by the excitation classes taken into account within the CI calculation. In the present work the influence of various excitation classes on A\(_{iso}\) is examined. Calculations including all single, double, triple and a large part of the quadruple excitations are performed and the individual effects of the excitation classes are studied. It is found that the surprisingly good agreement found for S-CI treatments is due to large error cancellations. The importance of higher than double excitations arises from their indirect influence on the single excitations.
A reliable prediction of the isotropic hyperfine coupling constant A\(_{iso}\) is still a difficult task for ab initio calculations. In previous studies, the configuration selected multireference configuration interaction method in combination with perturbation theory to correct the wave function (MRCI/ B\(_K\)) yielded accurate isotropic hyperfine coupling constants very economically. The present study gives a detailed analysis of the MRCI/ B\(_K\) method based on the X\(^2 \pi\) state of CH as a test case. Furthermore, a comparison to various other methods such as Maller-Ptesset perturbation theory and the coupled cluster approach is made. The success of the MRCI/ B\(_K\) method in predicting isotropic hyperfine coupling constants is explained in terms of the inßuence of higher than double excitations.
Reliable prediction of the isotropic hyperfine coupling constant, a\(_{iso}\), is still a difficult task for ab initio calculations. Strong dependence on the method used for its calculation is found. Within a truncated multi-referencc ansatz a\(_{iso}\) is strongly affected by the size ofthe reference space and the nurober of terms in the truncated Cl expansion. In the present paperdifferent effects of the neglected Cl space are discussed. Modified B\(_K\) and A\(_K\) methods are used to estimate the contributions ofthe neglected configurations. lt can be shown that a combination of both methods is able to recover about 90-9 S% of the total error in a\(_{iso}\)· Furthermore, it was found that to obtain about 90% of the B\(_K\) correction only about I 0-20% ofthe configurations within H0 have to be corrected.
The hyperfine coupling constants (hfcc) A\(_{iso}\) and A\(_{ij}\) are calculated for the atoms of NH\(_2\) in its, two lowest-lying electronk states at various molecular geometries by means of the ab initio multireference configuration interaction .method. The vibronically averaged values of the hfccs for the K = 0 and 1 levels in \(^{14}\)N \(^1\)H\(_2\) in the energy range up to 20 000 cm\(^{-1}\) are computed. Polarization elfects which determine A\(_{iso}\) as well as a simple model to describe the dipolar hfccs are discussed. All resrilts are in excellent agreement with experimental data.
Large-acale multi-reference configuration interaction (MRD-CI) calculations in a quite flexible AO basis are employed to study the energy hypersurface for the reaction intermediates XC\(_3\)H\(_4\) with X = Cl, Br and F. Particular emphasis is therby placed on determining the equilibrium conformations, the CH\(_2\) rotation barrier and the energy surface for a possible bridging (shuttling motion (1a] of X between the two carbon centers). The absolute minimum in the potential energy surface is found in all three cases for the asymmetric ß-halo radical in general agreement with ESR data at an XCC angle of ca. 110°, a c-c separation somewhat shorter than a single bond and an approximate sp3 type hybridization (\(\alpha _2 \approx \) 135-140°). In FC\(_2\)H\(_4\) the energy difference between the minimum in the symmetric conformation and the absolute minimum is found to be more than 30 kcal so that shuttling seems impossible in agreement with experimental findings. In BrC\(_2\)H\(_4\) the difference between these two potential minima is only between 1-2 kcal, i.e., smaller than the barrier to CH\(_2\), rotation, so that· shuttling is favored, while ClC\(_2\)H\(_4\) takes an intermediate position between these extremes. The use of correlated wavefunctions is found to be quite important for such a study; the results are related to various kinetic studies of these radicals.
Large-scale multireference configuration interaction (MRD-CI) calculations in a quite flexible AO basis are employed to study the energy hypersurface for the reaction intermediate FC\(_2\)H\(_4\) • The reaction F + C\(_2\)H\(_4\) -> FC\(_2\)H\(_4\) as weil as the 1,2 migration of the fluorine atom in FC\(_2\)H\(_4\) is investigated. In addition the rotation around the CC bond in the optimum conformation is studied. The absolute minimum in the potential energy is found for the asymmetric structure but the symmetric structure is also found to be stable with respect to the dissociation, so that a shuttling of the fluorine atom is in principle possible but highly unlikely because ( l) the activation energy is high ( II 5-130 kJ fmol) and the saddle point lies only 4(}-50 kJ jmol below the dissociation Iimit of F + C\(_2\)H\(_4\) and (2) the competitive motion, i.e., rotation around the CC axis, is nearly free (I 1-17 kJ/mol).
The hyperfine coupling constants for the \(^3\)Σ\(-\) ground state of the NH molecule are determined by configuration interaction calculations whereby the infl.uence of polarization functions as weil as of the configuration space on the spin polarization mechanism is analysed. The dipolar part Au(N) and Au(H) can be obtained very reliably without much computational effort (A .. (N) == -45·3 MHz and A"(H) = -62·3 MHz). The value for the isotropic contribution a1.., in the best AO basis and MRD-CI treatment is - 64·5 MHz for H and 16·6 MHz for nitrogen compared to the corresponding experimental quantities of -66 MHz and 19 MHz respectively. Their determination depends on a subtle balance of the lu, 2u and 3u shell correlation description, whereby the dominant contribution to a1..,(H) results from the 2u shell. It is shown that the often good agreement of a110 values with experiment in a small basis singledouble configuration interaction treatment results from a cancellation of two errors.
Multi-reference configuration interaction calculations employing various orbital transformations are undertaken to obtain the isotropic hyperfine coupling constant a\(_{iso\) in nitrogen and a\(_{iso\) (H) in the CH molecule. The natural orbital (NO) basis is found to be more effective than the simple RHF-MO basis; the most obvious is a basis of spin natural orbitals (SNO). It is found that a\(_{iso\) is approached from opposite sides in the NO and 2s shell SNO basis if the CI expansion is increased. Both results are within a few percent of the full CI Iimit for the nitrogen atorn (in the given AO basis) and the experimental value for Hin the CH radical. Various features ofthe SNO are discussed.
Study of the 1s and 2s shell contributions to the isotropic hyperfine coupling constant in nitrogen
(1988)
The istropic part of the hyperfine coupling constant is investigated by means of multireference configuration interaction calculations employing Gaussian basis sets. A detailed study of the 1s and 2s spin polarisation in the nitrogen atom and the NH molecule shows that the structure of the lower-energy space of the unoccupied orbitals is essential for the results. A contraction of the Gaussian basis is possible without loss of accuracy if enough flexibility is retained to describe the main features of the original space of unoccupied functions. Higher than double excitations are found to be non-negligible for the description of α\(_{iso}\).
The hyperfine coupling constant for the nitrogen atom is evaluated by large-scale MRD-CI calculations. A detailed analysis of the charge density at the nucleus and the spin polarization in the ls and 2s shell as a function of various technical parameters is undertaken. Various (s, p) AO basis sets and the inftuence of correlation orbitals is investigated as weil as selection threshold and other properlies in CI calculations. The best value, obtained for the isotropic hyperfine coupling constant in an s, p, d basis, based on theoretical judgment of' best' quantities, is 9·9 MHz compared to 10·4509 MHz.
Large-scale multireference configuration interaction calculations in a double·t·type AO basis including polarization functions are carried out for the potential surface of the ClC\(_2\)H\(_4\9 system. The charge distribution for various extreme points of the surface is discussed. The absolute minimum is found for an asymmetric ClC2H4 structure. The symmetrical bridged nuclear conformation is also found to be stable with respect to dissociation into Cl + C\(_2\)H\(_4\)• The activation energy for rotation about the C-C axis is calculated tobe around 18 kJ/mol, which is comparable tothat for the 1,2 migration {around 26 kJ/mol). The stereochemistry is governed by the fact that addition of CI to C\(_2\)H\(_4\) (or dissociation) is a two-step reaction proceeding through a symmetrica1 intermediate. The direct addition pathway possesses a small barrier of about 8 kJ jmol.
Wben irradiated at 360 nm, furocoumarins with a hydroperoxide group in a side chain effciently give rise to a type of DNA damage that can best be explained by a photoinduced generation of hydroxyl radicals from the excited pbotosensitizers. The observed DNA damage profiles, i.e. the ratios of single-strand breaks, sites of base loss (AP sites) and base modifications sensitive to fonnamidopyrimidine-DNA glycosylase (FPG protein) and endonuclease m, are similar to the DNA damage profile produced by hydroxyl radicals generated by lonizing radiation or by xanthine and xanthine oxidase in the presence of Fe(III)-EDTA. No such damage is observed with the corresponding furocoumarin alcohols or in the absence of near-UV radiation. The damage caused by the photo-excited hydroperoxides is not influenced by superoxide dismutase (SOD) or catalase or by D2O as solvent. The presence of t-butanol, however, reduces both the formation of single-strand breaks and of base odifications sensitive to FPG protein. The cytotoxicity caused by one of the hydroperoxides in L5178Y mome lymphoma cells is found to be dependent on the near-UV irradiation and to be much higher than that of the corresponding alcohol. Therefore the new type of photoinduced damage occurs inside cells. Intercalating photosensitizers with an attached hydroperoxide group might represent a novel and versatile class of DNA damaging agents, e.g. for phototherapy.
Enzym-Modifikationen finden in der Natur in Form von posttranslationalen Protein-Modifikationen statt und sind ein faszinierender Mechanismus, um die biologische Vielfalt und Funktion von Proteinen um ein Vielfaches zu erhöhen. Daher ist es für ein ganzheitliches Verständnis bestimmter biologischer Prozesse oder enzymatischer Struktur-Funktions-Beziehungen unerlässlich, chemische Methoden zu entwickeln, die in der Lage sind, diese natürliche Diversität nachzuahmen.[61] Die wohl größte Herausforderung der chemischen Protein-Konjugation ist die chemo- und regioselektive Modifikation einer gezielten Aminosäure bei gleichzeitig milden und physiologischen Reaktionsbedingungen. Trotz zahlreich beschriebener Ansätze zur selektiven Protein-Modifikation, bedarf es weiterhin neuer Methoden, da viele bestehende Herangehens¬weisen auf ein spezielles System zugeschnitten sind.[9, 63]
Aus diesem Grund sollte im Rahmen dieser Arbeit eine breit anwendbare Methode zur selektiven chemischen Tyrosin-Modifikation am Modell der Levansucrase aus Bacillus megaterium entwickelt werden. Durch eine zweistufige Protein-Modifikation, bestehend aus einer En-Reaktion im ersten Schritt und einer Click-Reaktion im zweiten Konjugationsschritt, gelang es die Produktspezifität der Bm Levansucrase rational zu beeinflussen. Zunächst wurde die Tyrosin-spezifische En-Reaktion mit der Luminol-Verbindung 1 an natürlich vorkommenden Tyrosin-Seitenketten der Levansucrase erprobt und analysiert. Hierbei zeigte sich durch massenspektrometrische Untersuchungen, dass hauptsächlich zwei der 25 vorhandenen Tyrosin-Reste mit dem Luminol-Tag 1 modifiziert wurden, zu denen die Seitenketten Y247 und Y196 gehörten. Um die Auswirkungen der Tyrosin-Modifikation leichter interpretieren zu können und eine gegenseitige Beeinflussung auszuschließen, wurde vorerst mit der Einzelmutante Y247F gearbeitet. Da nach der ersten Modifikation der Variante Y247F geringe Veränderungen im Produkt¬spektrum beobachtet wurden, insbesondere im hoch-molekularen Bereich, wurde die Click-Reaktion im zweiten Schritt mit der Intention durchgeführt, diesen Effekt zu verstärken. Schließlich bewirkte die Click-Reaktion mit Azidoglucose (AzGlc) bei Variante Y247F-1-AzGlc eine erhebliche Verschiebung der Produktverteilung von kleinen Fructooligosacchariden (ca. 1100 Da) hin zu hoch-molekularem Levan (ca. 2,1∙106 Da).
Drei weitere Positionen, die in der dritten Zone des Enzyms liegen, wurden für die gentechnische Substitution gegen nicht-native Tyrosin-Reste ausgewählt. Dadurch wurden die Varianten E314Y, D248Y sowie F445Y erhalten und anschließend wie zuvor in zwei Schritten chemisch modifiziert. Die Modifikation dieser Varianten führte hinsichtlich der Veränderung des Produktprofils zu ähnlichen Ergebnissen, wie sie mit dem Enzym Y247F erhalten wurden (Übersicht 1, A). Um den Einfluss verschiedener Seitenketten zu analysieren, wurden neben der Azidoglucose vier weitere Azido-Verbindungen in der Click-Reaktion getestet.
Die Resultate aus den genannten Untersuchungen und die Einbeziehung molekular¬-dynamischer Simulationen ließen erste Rückschlüsse auf die mechanistischen Prozesse der Bm Levansucrase und deren gezielte Manipulation zu: Die Größe der eingeführten Seitenkette sowie die Fähigkeit des Tags polare Wechselwirkungen auszubilden, spielen eine entscheidende Rolle zur rationalen Modulation der Produkt¬spezifität. Insbesondere die räumliche Orientierung und Bewegung der Seitenkette 1 AzGlc und die damit einhergehende sterische Hinderung trugen dazu bei, eine vorzeitige Dissoziation der wachsenden Fructane zu verhindern und ermöglichten dadurch die prozessive Polymersynthese.
Weitere Erkenntnisse über den Levan-Elongationsmechanismus wurden durch die Modifikation der Varianten N126Y und S125Y erhalten. Diese lagen im Gegensatz zu den zuvor untersuchten Tyrosin-Resten nicht im Wachstumsverlauf des Substrats und besaßen zudem eine kürzere Distanz zum aktiven Zentrum. In beiden Fällen führte bereits die erste Modifikation mit Luminol-Derivat 1 zu völlig unter¬schiedlichen Produktprofilen im Vergleich zu den zuvor untersuchten Enzym-Varianten. Während mit der Variante N126Y-1 eine signifikante Akkumulation (bis zu 800 % Zunahme) verschiedener Oligosaccharide erzielt wurde, synthetisierte die Variante S125Y-1 schon nach dem ersten Modifikationsschritt Levan-Polymer (Übersicht 1, B/C). Die zugrunde-liegenden Interaktionen und Trajektorien der eingeführten Seitenkette wurden ebenfalls mit Hilfe von MD Simulationen analysiert und bestätigten die zuvor getroffenen Annahmen. Durch die räumliche Nähe zur Substrat-Bindungstasche reichte bei Variante S125Y 1 bereits die Luminol-Verbindung aus, um die Substrat-Dissoziation zu verhindern und damit die Polymer¬synthese zu induzieren. Hingegen dazu ergaben die Simulationen eine sehr dynamische und fluktuierende Seitenkette für N126Y-1, was vermutlich zur Destabilisierung initialer Wechselwirkungen zwischen Substrat und der Protein¬oberfläche führte und dadurch die Freisetzung und Akkumulation kurzer Oligo-saccharide begünstigte.
Durch die bioorthogonale chemische Einführung einer artifiziellen Seitenkette war es schließlich möglich, das Produktspektrum der Bm Levansucrase sowohl in Richtung Polymersynthese als auch in Richtung kurzer Oligosaccharide zu lenken. Unter Verwendung der Tyrosin-spezifischen En-Reaktion wurden dafür gezielt native und nicht-native Tyrosin-Reste selektiv modifiziert und in einer Folge¬reaktion mittels Click-Chemie zusätzlich derivatisiert. Die Auswirkungen der Modifikations-Reaktionen auf den Elongationsmechanismus des Substrats konnten durch MD-Simulationen aufgeklärt werden. Das Ziel, die Produktspezifität der Levansucrase rational zu beeinflussen und in eine gezielte Richtung zu steuern, wurde damit erfolgreich umgesetzt.
Ein weiterer Fokus dieser Arbeit lag darin, eine effiziente und einfache Methode zur Reinigung eines Fructan-Gemisches zu entwickeln, um damit den Zugang zu Oligo-sacchariden definierter Größen zu vereinfachen. Die Verfügbarkeit bestimmter Oligosaccharide in ausreichender Menge und Reinheit würde die Untersuchung von Fructanen auf ihre präbiotischen Eigenschaften erleichtern und zum Verständnis der Korrelation zwischen dem Darmmikrobiom und verschiedenen Krankheits¬bildern beitragen.[125] Mit Hilfe der Levansucrase-Variante K373L wurde ein Fructan-Gemisch synthetisiert, das im Vergleich zum Produkt¬profil des Wildtyps einen höheren Anteil kurzkettiger Oligosaccharide aufwies. In einem dreistufigen Reinigungsprozess wurde das Produktgemisch im ersten Schritt von den Monosacchariden Glucose und Fructose sowohl fermentativ durch den Hefe¬stamm H. polymorpha als auch chromatographisch per Silicagel separiert. Anschließend erfolgte eine grobe Trennung der Oligosaccharide nach dem Größen¬ausschlussprinzip mit einer Bio-Gel®P2-Säule. Im letzten Schritt wurde die Oligosaccharidfraktion, die hauptsächlich Tri- und Tetrasaccharide enthielt, schließlich mittels Umkehrphasen-Säulenchromatographie (RP18-HPLC) in die gewünschten Produkte aufgetrennt. Auf diese Weise gelang es, die Oligosaccharide 1 Kestose (28 %), 6 Kestose (56 %) und 6 Nystose (20 %) in hoher Reinheit (> 95 %) und moderaten Ausbeuten zu isolieren (Übersicht 2).
Der letzte Teil dieser Arbeit sollte die verschiedenen Disziplinen der Biokatalyse, chemischen Protein-Modifikation und Click-Reaktion mit einer neuen Kompontente, der Photokatalyse, verbinden und in einem innovativen Konzept die Grundlage für die Kombination dieser Forschungsbereiche schaffen. In diesem Kontext wurde einerseits eine lineare photo-biokatalysierte Kaskaden-Reaktion entworfen und vorbereitet, während andererseits die Synthese eines clickbaren Photokatalysators durchgeführt wurde (Übersicht 3). Für den enzymatischen Teil der Kaskaden-Reaktion wurden die Halogenasen RebH und RadH mit den zugehörigen Regenerationssystemen Fre und GDH erfolgreich in E. coli exprimiert, gereinigt und deren Aktivität nachgewiesen. Darüber hinaus wurde ein aktiver Alkin-funktionalisierter Photokatalysator synthetisiert, dessen Aktivität auch nach der Click-Reaktion mit einer Aminosäure und einem Peptid erhalten blieb. Damit wurden die Grundlagen geschaffen, um z. B. photoaktive Bausteine in ein Enzym einzubringen und somit neue lichtabhängige Reaktionszentren oder sogenannte Designer-Enzyme zu erzeugen.
This thesis deals with the isolation and structural elucidation of bioactive naphthylisoquinoline alkaloids and related analogs. The mode of action of the antiplasmodial activity exhibited by the naphthylisoquinoline alkaloids was explored and compared to that of the antimalarial drug chloroquine. Furthermore, the phase 1 and 2 metabolism of dioncophyllines A and C and dioncopeltine A were investigated. In detail the following results have been obtained: • From the leaves of the recently discovered East African liana A. tanzaniensis six naphthylisoquinoline alkaloids were isolated. • The leaves of a botanical yet undescribed Ancistrocladus species, collected by Prof. Dr. V. Mudogo in the Democratic Republic of Congo in the habitat Yeteto near the town Ikela, were analyzed for naphthylisoquinoline alkaloids for the first time. The isolation work led to the first identification of an N,C-coupled naphthyldihydroisoquinoline alkaloid; ancistrocladinium B. Phytochemical investigation of the roots of the Congolese Ancistrocladus species (habitat Yeteto), , afforded five new derivatives of known naphthylisoquinoline alkaloids, namely 5'-O-demethylhamatine, 5'-O-demethylhamatinine, 6-O-demethylancistroealaine A, 6,5'-O,O-didemethylancistroealaine A, and 5-epi-6-O-methylancistrobertsonine A, along with six known naphthylisoquinoline alkaloids. • The antiplasmodial activity guided purification of 60Co irradiated samples containing commercially available naphthylisoquinoline related substances, afforded the isolation of the irradiation products 3,4-dihydro-1-isoquinolinone, 3,4-dihydro-1-isoquinolineamine, and 1,2,3,4-tetrahydro-1,2-diazirino-isoquinoline. The compounds were found to be more active than the starting material, although only exhibiting weak antiplasmodial activity against P. falciparum. • The effect on the absorption spectrum of FPIX due to complex formation with the naphthylisoquinoline alkaloids dioncophyllines A and C, dioncopeltine A korupensamine A, and ancistrocladine was examined by a titration study. Job's plot analyses by UV-spectroscopy determined the stoichiometry for the complex formation of FPIX and naphthylisoquinoline alkaloids to be 2:1. Furthermore, the dissociation constants for the complexation with FPIX were determined for each of the naphthylisoquinoline alkaloids investigated. Dioncophylline C and dioncopeltine A were found to possess dissociation constants, which are comparable to the one reported for the antimalarial drug chloroquine. The ability of ESI to transfer noncovalent solution-phase assemblies intact into the gas phase, was conducted on solution mixtures of naphthylisoquinoline alkaloid and FPIX, as well as on mixtures of chloroquine and FPIX. The mass spectrometry analyses revealed several peaks, which corresponded to the complex formation of FPIX to the respective ligands investigated. The most interesting results obtained were the detection of peaks corresponding to the complex formation between a chelated dimer of FPIX and dioncophylline Cand of peaks corresponding to a double protonated tetramer of FPIX – consisting of two chelated -oxo dimers of FPIX – in complex formation with two molecules of chloroquine. • Two phase 1 metabolism products of dioncophylline A were identified. Coelution in combination with HPLC-MS/MS, NMR, and CD investigations assigned the major metabolic product as 5'-O-demethyldioncophylline A. The minor metabolic product was only present in small amounts, which disabled an unambiguous structural characterization of the compound. However, as deduced from the mass spectrometry analyses and exclusion of a possible metabolic oxidation product by coelution with authentic reference material, the metabolite should possess a 4-hydroxylated isoquinoline portion and is assumed to be represented by structure. Dioncophylline C and dioncopeltine A were found to be stable to phase 1 metabolism reactions caused by rat liver microsomes.
A new strategy is demonstrated for the synthesis of warped, negatively curved, all‐sp\(^2\)‐carbon π‐scaffolds. Multifold C−C coupling reactions are used to transform a polyaromatic borinic acid into a saddle‐shaped polyaromatic hydrocarbon (2 ) bearing two heptagonal rings. Notably, this Schwarzite substructure is synthesized in only two steps from an unfunctionalized alkene. A highly warped structure of 2 was revealed by X‐ray crystallographic studies and pronounced flexibility of this π‐scaffold was ascertained by experimental and computational studies. Compound 2 exhibits excellent solubility, visible range absorption and fluorescence, and readily undergoes two reversible one‐electron oxidations at mild potentials.
The West African liana Ancistrocladus abbreviatus is a rich source of structurally most diverse naphthylisoquinoline alkaloids. From its roots, a series of four novel representatives, named ancistrobrevolines A–D (14–17) have now been isolated, displaying an unprecedented heterocyclic ring system, where the usual isoquinoline entity is replaced by a ring-contracted isoindolinone part. Their constitutions were elucidated by 1D and 2D NMR and HR-ESI-MS. The absolute configurations at the chiral axis and at the stereogenic center were assigned by using experimental and computational electronic circular dichroism (ECD) investigations and a ruthenium-mediated oxidative degradation, respectively. For the biosynthetic origin of the isoindolinones from ‘normal’ naphthyltetrahydroisoquinolines, a hypothetic pathway is presented. It involves oxidative decarboxylation steps leading to a ring contraction by a benzilic acid rearrangement. Ancistrobrevolines A (14) and B (15) were found to display moderate cytotoxic effects (up to 72%) against MCF-7 breast and A549 lung cancer cells and to reduce the formation of spheroids (mammospheres) in the breast cancer cell line.
A unique series of six biaryl natural products displaying four different coupling types (5,10 , 7,10 , 7,80 , and 5,80) were isolated from the roots of the West African liana Ancistrocladus abbreviatus (Ancistrocladaceae). Although at first sight structurally diverse, these secondary metabolites all have in common that they belong to the rare group of naphthylisoquinoline alkaloids with a fully dehydrogenated isoquinoline portion. Among the African Ancistrocladus species, A. abbreviatus is so far only the second one that was found to produce compounds with such a molecular entity. Here, we report on four new representatives, named ancistrobreveines A–D (12–14, and 6). They were identified along with the two known alkaloids 6-O-methylhamateine (4) and entdioncophylleine A (10). The two latter naphthylisoquinolines had so far only been detected in Ancistrocladus species from Southeast Asia. All of these fully dehydrogenated alkaloids have in common being optically active despite the absence of stereogenic centers, due to the presence of the rotationally hindered biaryl axis as the only element of chirality. Except for ent-dioncophylleine A (10), which lacks an oxygen function at C-6, the ancistrobreveines A–D (12–14, and 6) and 6-O-methylhamateine (4) are 6-oxygenated alkaloids, and are, thus, typical ‘Ancistrocladaceae-type’ compounds. Ancistrobreveine C (14), is the first – and so far only – example of a 7,80-linked fully dehydrogenated naphthylisoquinoline discovered in nature that is configurationally stable at the biaryl axis. The stereostructures of the new alkaloids were established by spectroscopic (in particular HRESIMS, 1D and 2D NMR) and chiroptical (electronic circular dichroism) methods. Ancistrobreveine C (14) and 6-O-methylhamateine (4) exhibited strong antiproliferative activities against drug-sensitive acute lymphoblastic CCRF-CEM leukemia cells and their multidrugresistant subline, CEM/ADR5000.
