In physiological conditions platelets have a major role in maintaining haemostasis. Platelets prevent bleeding from wounds by distinguishing normal endothelial cells in vasculature from areas with lesions to which they adhere. Interaction of platelet agonists and their receptors is controlled by intracellular signaling molecules that regulate the activation state of platelets. Very important intracellular signaling molecules are cyclic nucleotides (cGMP and cAMP), both involved in inhibition of platelet activation. Formation of cGMP and cAMP in platelets is stimulated by endothelial-derived NO and prostacyclin (PGI2), which then mediate inhibition of platelets by activating protein kinase G (PKG) and protein kinase A (PKA). Recently, it has been suggested that reactive oxygen species (ROS) represent new modulators of cell signaling within different cell types. The work summarized here describes the involvement of platelet ROS production in platelet activation, the relation of NO/cGMP/PKG I pathway to ROS and to mitogen-activated protein kinases (MAP kinase) signaling, and the involvement of cyclic nucleotides in megakaryocyte and platelet development. Platelets activated with different agonists produce intracellular but not extracellular ROS by activation of NAD(P)H oxidase. In addition, ROS produced in platelets significantly affects αIIbβ3 integrin activation but not alpha/dense granule secretion and platelet shape change. Thrombin induced integrin αIIbβ3 activation is significantly decreased after pretreatment of platelets with NAD(P)H oxidase inhibitors and superoxide scavengers. These inhibitors also reduce platelet aggregation and thrombus formation on collagen under high shear and achieve their effects independently of the NO/cGMP pathway. ADP secreted from platelet dense granules with subsequent activation of P2Y12 receptors as well as thromboxane A2 release are found to be important upstream mediators of p38 MAP kinase activation by thrombin. However, p38 MAP kinase activation does not significantly contribute to calcium mobilization, P-selectin expression, αIIbβ3 integrin activation and aggregation of human platelets in response to thrombin. Finally, PKG activation does not stimulate, but rather inhibit, p38 and ERK MAP kinases in human platelets. Further study revealed that cyclic nucleotides not only inhibit platelet activation, but are also involved, albeit differentially, in megakaryocyte and platelet development. cAMP is engaged in haematopoietic stem cell differentiation to megakaryocytes, and cGMP has no impact on this process. While PKA is already present in stem cells, expression of proteins involved in cGMP signaling (soluble guanylyl cyclase, sGC; PKG) increases with maturation of megakaryocytes. In the final step of megakaryocyte maturation that includes release of platelets, cGMP and cAMP have mild but opposing effects: cGMP increases platelet production while cAMP decreases it indicating a finely regulated process that could depend on stimulus coming from adjacent endothelial cells of sinusoids in bone marrow. The results of this thesis contribute to a better understanding of platelet regulation and of the possible molecular mechanisms involved in megakaryocyte maturation in bone marrow vascular microenvironment.
Bereits 1942 beschrieben FITZGERALD und HALLPIKE im Zusammenhang mit der kalorischen Vestibularisprüfung den Begriff der "Directional Preponderance". Dabei handelt es sich um das Überwiegen einer Nystagmusrichtung bzw. eine Richtungsenthemmung des Nystagmus bei der kalorischen Stimulation des lateralen Bogenganges. Seit dieser Erstbeschreibung ist die klinische bzw. diagnostische Wertigkeit eines Nystagmusrichtungsüberwiegens umstritten. Zur Untersuchung des Zusammenhanges zwischen der kalorischen Nystagmusrichtungsenthemmung und klinischen Befunden wurden in einer retrospektiven Studie die Datensätze von 148 Patienten aus der Neurootologie der Universitäts-HNO-Klinik Würzburg ausgewertet, die mittels eines PICKER-SCHWARZER-Achtkanal-Polygraphen registriert wurden. Im Ergebnisteil werden 14 Unterkollektive deskriptiv statistisch aufgearbeitet und in Diagrammen und Tabellen anschaulich dargestellt. In den folgenden Kasuistiken werden fünf typische Fälle von verschiedenen Gleichgewichtsfunktionsstörungen genau erläutert. In der Diskussion werden im ersten Teil die Neuroanatomie und Physiologie des vestibulären Systems, sowie die Nystagmusgenerierung ausführlich dargestellt. Es folgen Ausführungen zu den sog. JONGKEES'schen Formeln sowie zu den verschiedenen Begriffsdefinitionen "Richtungsenthemmung" und "unilaterale Schwäche" aus der Literatur und es werden Beispiele für verschiedene Konstellationen beim kalorischen Test z.B. mit und ohne Spontagnystagmus aufgezeigt. Die Zusammenfassung fasst die wichtigsten statistischen Ergebnisse anschaulich zusammen.
Die Bedeutung der genomischen Instabilität in humanen melanocytischen Läsionen ist Gegenstand vieler dermatologischer Studien und bis heute ungeklärt. Ist die Mikrosatelliteninstabilität bloße Begleiterscheinung der unkontrolliert proliferierenden Tumorzellen oder wird ihr ein pathogenetischer Mechanismus zuteil? In Fischen der Gattung Xiphophorus können durch klassische Kreuzungsexperimente melanocytische Läsionen verschiedener Malignitätsgrade induziert werden. Diese Läsionen sind auf Grund ihres genetisch kontrollierten Hintergrundes eindeutig definiert und reproduzierbar - und damit im Vorteil gegenüber der unsicheren Klassifikation humaner Hautläsionen. In der vorliegenden Arbeit wurde hauptsächlich der Frage nachgegangen, ob MIN+ ein obligates molekulares Ereignis für die Progression von Melanomen bis zum terminalen Stadium darstellt. Dieser Fragestellung wurde unter Verwendung PCR-basierter Mikrosatellitenanalysen sowie Multilocus DNA-Fingerprint Analysen nachgegangen. 23 Tumoren unterschiedlicher Malignität wurden im Vergleich zum tumorfreien Normalgewebe desselben Fisches an 12 Genloci unbekannter chromosomaler Lokalisation auf Mikrosatelliteninstabilität untersucht. Es wurde insgesamt eine Zahl von 263 informativen Loci erzielt, von denen sich nur 20 ( = 7.6%) als instabil erwiesen. Mittels der Multilocus-Fingerprint Analysen wurden 8 Melanome und deren korrespondiere Normalgewebe mit drei Sonden hybridisiert. Nur einer von 12 analysierbaren MLFPs zeigte eine genetische Aberration in Form einer Deletion einer einzelnen Bande im Tumorgewebe. Mit diesem Ergebnis wurde das in der Mikrosatellitenanalyse erhaltene Resultat einer nicht signifikanten Mikrosatelliteninstabilität im Xiphophorus-Melanom-Modell System bestätigt. Zusammenfassend kann gesagt werden, daß die Abwesenheit eines MIN+-Phänotypen in den Melanomen von Xiphophorus darauf hinweist, daß der Erwerb von MIN weder ein notwendiges Kriterium für die Initiation eines Tumors noch für seine Progression darstellt. Ein anderer Pfad der Tumorinitiation ist die direkte Schädigung oder der Verlust von Tumorsuppressorgenen, von in Mismatch Repair involvierten Genen oder durch die Transformation von Proto-Onkogenen zu Onkogenen. In dieser Arbeit wurde die Expression von drei Genen, die in der Regulation des Zellzyklus Schlüsselrollen spielen (rb, p53, cdkn2a), sowie einer Komponente des MMR-Systems (MSH2) auf RNA-Ebene untersucht. Ihre Expressionsstärke wurde in folgenden Geweben miteinander verglichen: Maligne Melanome, benigne Läsionen, tumorfreie Kontrollgewebe und die Xiphophorus-Melanomzellinie (PSM). MSH2 und p53 wurden in allen untersuchten Geweben auf gleichem Niveau exprimiert. Die vergleichende Expressionsanalyse des vermeintlichen "melanoma susceptibility gene" cdkn2a im Melanom-Modell und der PSM-Zellinie bestätigte die bereits vorbeschriebene Überexpression des Zellzyklusinhibitors in malignen Melanomen von Xiphophorus-Hybriden. Die Untersuchung des Expressionsmusters von Rb in den oben genannten Geweben ergab eine verminderte Transkription dieses Gens in malignen Melanomen. Eine mögliche Interpretation dieses Ergebnisses wäre, einen Defekt in Rb oder pRB zu Grunde zu legen und die Hochregulation von cdkn2a entsprechend einer negativen Rückkopplungsschleife als physiologische Konsequenz anzusehen.
This volume brings together several authors from different areas of psychology and the neighbouring social sciences. Each one contributes their own perspective on the growing interest topic of subjective well-being. The aim of the volume is to present these divergent perspectives and to foster communication between the different areas. Split into three parts, this volume initially discusses the general perspectives of subjective well-being and addresses fundamental questions, secondly it discusses the dynamics of subjective well-being and more specific research issues to give a better understanding of the general phenomenon, and thirdly the book emphasizes the social context in which people experience and report their happiness and satisfaction. The book will be of great interest to social and clinical psychologists, students of psychology and sociology and health professionals.
Gegenstand des Buches ist ein psychologisches Modell der Beantwortung von Fragen in standardisierten Situationen, wie z.B. sozialwissenschaftlichen Umfragen. Dazu werden Denk- und Kommunikationsprozesse, die bei der Generierung von Antworten eine Rolle spielen, aufgezeigt und durch experimentelle Untersuchungen belegt. Dabei wird die Problematik der Unterscheidung zwischen "wahrem Wert" und "Fehler" bzw. "Antworteffekten" bei sozialwissenschaftlichen Befragungen deutlich.
Heute arbeiten ungefähr sechs Prozent der Erwerbstätigen als Telearbeiter. In zehn Jahren werden 30 Prozent prognostiziert. Eine Herausforderung, die bewältigt werden will. Insbesondere in Branchen, in denen eine starke internationale Vernetzung gegeben ist und die sich bereits in starkem Maße auf Datenverarbeitung stützen, wurde virtuelle Teamarbeit bereits früh eingeführt. Häufig herrscht aber eher Skepsis. Fehlende Kenntnisse der Arbeitsstrukturen werden kritisiert. Diese Lücke will dieses Buch schließen. Das Buch beantwortet Fragen wie: Welche Merkmale und Formen virtueller Arbeit gibt es? Wie können die Vorteile virtueller Kooperation effektiv genutzt werden? Welche Anwendungsbedingungen und Gestaltungsfelder gibt es? Wie baue ich entsprechende Teams auf und optimiere bzw. korrigiere die Zusammenarbeit? Prägnant und verständlich entwickeln die Autoren Elemente, Strukturen und Lösungen virtueller Zusammenarbeit. Im Ersten Teil des Buches veranschaulichen sie, welche Grundlagen und Strategien der virtuellen Arbeit es gibt. Der zweite Teil demonstriert, wie virtuelle Kooperation effektiv und erfolgreich gemanagt werden kann. Checklisten und Fragebögen runden das Buch ab und machen es zu einem handlichen Leitfaden für die Einrichtung und Optimierung virtueller Teams.
In vorliegender Studie wurde an histologischen Präparaten mittels immunhistochemischen Färbungen der Übergangsbereich zwischen oraler Mukosa und transplantiertem Jejunumsegment untersucht. Mit der Fragestellung ob sich die Strukturen im Übergangsbereich zwischen freiem Jejunumtransplantat und oraler Mukosa funktionell verändern und ob die Dünndarmepithelien einer morphologischen Umwandlung unterliegen. Zu diesem Zweck wurden die Gewebeschnitte nach der LSABC Methode mit verschiedenen Antikörpern markiert die spezifisch das zu bestimmende Epithel anfärbten. Die Differentzierung der Epithelien erfolgte durch für jede Epithelart spezifische Zytokeratine. Nachgewiesen wurde eine Abflachung des Zottenreliefs sowie eine numerische Becherzellatrophie. In unseren histologisch nachuntersuchten Gewebeproben stellte sich die Grenze zwischen transplantiertem Jejunumsegment und mehrschichtigem Plattenepithel exakt und scharf begrenzt dar. Weder Zellcluster von mehrschichtigem Plattenepithel im Bereich des Dünndarms, noch eine Transformationszone, wie sie bei Zellmetaplasie vorkommt konnte nachgewiesen werden. Auch hielten die dünndarmeigenen Gefäße die Grenze zwischen den beiden Geweben strikt ein und verliefen ebenso scharf begrenzt. Subepithelial zeigte sich zwar zu Beginn eine entzündlich bedingte Gefäßproliferation mit schütterem leukozytären Infiltrat, welches aber im Laufe der Zeit durch kollagenfasereiches Bindegewebe narbig ersetzt wurde. Die nachweisbaren CD 4 positiven Entzündungszellen waren nicht über das gewöhnliche Ausmaß vermehrt, es kommt zur Ausbildung einer Gewebsgrenze die von beiden Epithelien respektiert wird unter der Vereinigung des subepithelialen Bindegewebes. Dies alles kann als Bestätigung dafür angesehen werden, dass die Rekonstruktion großer Defekte mittels freiem Jejunumtransplantat auch über einen Zeitraum von mehr als 21 Jahren erfolgreich ist und das Transplantat seinen Anforderungen in der neuen Umgebung standhält.
Integrins are transmembrane receptors transmitting mechanical signals from the extracellular matrix (ECM) to the cytoskeleton (outside-in-signaling). Many molecular defects in the link between cytoskeleton and ECM are known to induce cardiomyopathies. alpha v integrin appears to play a major role in several processes relevant to remodeling, such as binding and activation of matrix metalloproteinases as well as regulation of cell proliferation, migration, and differentiation. We hypothesized that alpha v integrin-mediated signaling is required for the compensatory hypertrophy after aortic banding (AB) and associated with the modulation of ECM protein expression. Mice were treated in vivo with a specific integrin alpha v inhibitor or vehicle via osmotic minipumps starting 1 day prior to aortic banding (AB). At day 2 and day 7 following AB or sham-operation, the mice were examined by echocardiography and hemodynamic analyses were performed. Treatment of alpha v Integrin inhibitor led to a dilated cardiomyopathy and congestive heart failure in AB mice (dilated left ventricle, depressed LV function, and pulmonary congestion), but not to hypertrophy as observed in mice without inhibitor treatment. Investigation of downstream signaling revealed significant activation of the p38 Mitogen-Activated Protein Kinase (MAPK), the Extracellular signal-Regulated Kinases 1 and 2 (Erk 1/2), Focal Adhesion Kinase (FAK) and tyrosine-phosphorylation of c-Src in mice 7 days after AB. This response was blunted in mice treated with integrin alpha v inhibitor. Microarrays probing for a total of 96 cell adhesion and ECM genes identified various genomic targets of integrin alpha v mediated signalling. 7 days after AB 18 ECM genes were up-regulated more than 2-fold (n=6), e.g. collagen (8.11 ± 2.2), fibronectin (2.32 ± 0.94), secreted protein, acidic and rich in cysteine (SPARC, 3.78 ± 0.12), A disintegrin-like and metalloprotease (reprolysin type) with trombospondin type 1 (Adamts-1, 3.51 ± 0.81) and Tissue inhibitor of metalloproteinase 2 (TIMP2, 2.23 ± 0.98), whereas this up-regulation was abolished in mice that were treatd by integrin alpha v inhibitor via mini pumps. We conclude that signaling downstream of integrin alpha v is mediated by the MAPK, FAK and c-Src pathways leading to an up-regulation of extracelluar matrix components necessary for the compensatory response of the heart under a condition of pressure overload.
Die vorliegende Studie sollte klären in welchen Persönlichkeitsdimensionen sich depressive Patienten spezifisch von einer parallelisierten gesunden und klinischen Kontrollgruppe unterscheiden und welchen Einfluss die Persönlichkeit auf die Stimmung bei depressiven Patienten hat. Neben persönlichkeitsbezogenen Gegenwartsskalen wie sie in persönlichkeitspsychologischen Untersuchungen verwendet werden, kamen auch biographiebezogene Vergangenheitsvariablen zur Anwendung. Anhand der biographischen Variablen sollte untersucht werden, in welchem Zusammenhang Persönlichkeitsstrukturen und biographische Faktoren stehen und welchen Einfluss biographische Faktoren auf die Persönlichkeit und Stimmung von depressiven Patienten haben. Die klinische Studie umfasste 165 Versuchspersonen, aufgeteilt in 55 gesunde Probanden, 55 Patienten mit der Diagnose einer „Major Depression“, definiert nach DSM-IV und 55 psychisch kranke, jedoch nicht depressive Patienten (DSM-IV: Anpassungsstörungen, Schlafstörungen, Angststörungen). Die Patienten befanden sich im Zeitraum von 2000-2003 in ambulant psychiatrischer Behandlung. Das Aufnahmekriterium in die Studie war die Diagnose einer „Major depression“ nach DSM-IV. Die Kontrollgruppen wurden entsprechend der Hauptgruppe parallelisiert nach: 1. Geschlecht, 2. Alter (+/- 5 Jahre), 3. Schulbildung oder ausgeübter Beruf. Bei der Datenanalyse des Fragebogen für Psychovulnerabilität und Psychoprotektion (FPVP) mittels unterschiedlichen statistischen Verfahren fiel auf, dass sich die Patienten (depressive und sonstige psychisch kranke Patienten) deutlich von der gesunden Kontrollgruppe unterscheiden. Neurotizismus (NE) ist dabei der Persönlichkeitsfaktor, der bei den Patienten im Unterschied zu den Gesunden besonders ausgeprägt ist. Entgegen der häufig postulierten Unspezifität der Beziehung zwischen Neurotizismus (NE) und psychischer Störung, zeigen die Ergebnisse der vorliegenden Studie den Zusammenhang differenzierter, da eine statistisch signifikante Trennung der beiden klinischen Gruppen (depressive und sonstige psychisch kranke Patienten) anhand der Dimension Neurotizismus (NE) möglich war. Neben der Skala Neurotizismus (NE) zeigten sich auch in den Skalen Arbeitsbezogenheit (AB), Zielgerichtetheit (ZG), Desorganisation (DO) und Kindliches Kontaktverhalten (KI) spezifische Skalenwertunterschiede zwischen den depressiven und sonstigen psychisch kranken Patienten. Die Skalen Rigidität (RI) und Idealität (ID) im Sinne des Typus melancholicus, stellten keine spezifischen Persönlichkeitsmerkmale von unipolar depressiven Patienten dar. Wir gehen daher wie Kronmüller et al. (2002a, b) von einer störungstypischen, nicht jedoch störungsspezifischen Persönlichkeitsstruktur im Sinne des Typus melancholicus bei Patienten mit Major Depression aus. Die empirisch aufgefundenen Zusammenhänge zwischen FPVP- und EWL-Skalen bestätigten weitgehend die aufgrund von inhaltlichen Hinweisen entwickelte These von den psychoprotektiven bzw. psychovulnerablen Qualitäten der einzelnen FPVP-Skalen. Darüberhinaus zeigte sich ein Einfluß von Persönlichkeits- bzw. biographischen Variabeln auf Stimmung und Befindlichkeit. Zusammenfassend assoziieren sich negativ zu wertende Persönlichkeitsvariable mit negativ erlebten Befindlichkeitsvariablen und positiv zu wertende Persönlichkeitsvariable mit positiv erlebten Befindlichkeits-variablen, d.h. es besteht eine Verbindung von eher überdauernden Eigenschaften der Persönlichkeit mit eher vergänglichen Erlebensweisen. Weiterhin zeigen die Ergebnisse der vorliegenden Studie, dass neben Persönlichkeitsmerkmalen auch die Biographie ein wichtiger Vulnerabilitäts-faktor einer Depression ist. Die Biographie scheint dabei ihre pathogene Wirkung u.a. über die Persönlichkeit zu entfalten. Insbesondere die biographische Skala Primärsozialisation (PS) im Sinne einer ungünstigen Primärsozialisation (PS) zeigt bei den depressiven Patienten starke Zusammenhänge mit den Skalen Neurotizismus (NE) und Zielgerichtetheit (ZG). Auch anhand der Vorhersage der aktuellen Stimmung von Depressiven zeigt sich die Bedeutung der Skala Primärsozialisation (PS), die in der depressiven Gruppe v.a. eine negative Befindlichkeit mit den Aspekten Emotionale Gereiztheit und Angst vorhersagt. Aufgrund unterschiedlicher Meinungen in der Literatur sind weitere empirische Studien zur Objektivierung des Zusammenhangs zwischen Biographie, Persönlichkeit und Stimmung bei Depressiven nötig.
