The aim of this work was to synthesize and functionalize different bio-relevant nanomaterials like silica-coated superparamagnetic iron oxide nanoparticles (SPIONs) as contrast agents for T2 magnetic resonance imaging (MRI) and detonation nanodiamond (DND) with the neurohormone peptide allatostatin 1 (ALST1) and a fluorescent dye. Analytical techniques for the determination and quantification of surface functional groups like amines, azides, and peptides were also developed and established.
Thus, in the first part of the work, a TGF-1 binding peptide and allatostatin 1 (ALST1), both supposed to act as active tumour targeting vectors, were synthesized by solid-phase peptide synthesis (SPPS) and characterized by high pressure liquid chromatography (HPLC) and mass spectrometry. Then, azide-functionalized silica nanoparticles were synthesized by the Stöber process and characterized by transmission electron microscopy (TEM) and infrared spectroscopy (IR). The surface loading of amine and azide groups was determined by a new protocol. The azide groups were reduced with sodium boronhydride to amine and then functionalized with Fmoc-Rink Amide linker according to a standard SPPS protocol. Upon cleavage of Fmoc by piperidine, the resulting dibenzofulvene and its piperidine adduct were quantified by UV/Vis spectroscopy and used to determine the amount of amine groups on the nanoparticle surface. Then, ALST1 and related tyrosine- and phenylalanine substituted model peptides were conjugated to the azide-functionalized silica nanoparticles by copper(I)-catalyzed azide-alkyne dipolar cycloaddition (CuAAC). The successful peptide conjugation was demonstrated by the Pauly reaction, which however is only sensitive to histidine- and tyrosine-containing peptides. As a more general alternative, the acid hydrolysis of the peptides to their individual amino acid building blocks followed by derivatization with phenyl isothiocyanate (PITC) allowed the separation, determination, and quantification of the constituent amino acids by HPLC.
In the second part of the work, amine- and azide-functionalized silica-coated superparamagnetic iron oxide nanoparticles (SPIONs) were synthesized by co-precipitation and subsequent silica-coated based on the Stöber process and characterized by TEM and IR. The amine surface loading was determined by the method already established for the pure silica systems. The azide surface loading could also be quantified by reduction with sodium boronhydride to amine groups and then conjugation to Fmoc-Rink amide linker. Upon cleavage of Fmoc with piperidine, the total amine surface loading was obtained. The amount of azide surface groups was then determined from the difference of the total amine surface loading and the amine surface loading. Thus, it was possible to quantify both amine and azide surface groups on a single nanoparticle system. Superparamagnetic iron oxide nanoparticles (SPIONs) are potent T2 contrast agents for magnetic resonance imaging (MRI). Due to their natural metabolism after injection into the blood stream, SPIONs mostly end up inside macrophages, liver, spleen or kidneys. To generate a potential target-specific SPION-based T2 contrast agent for MRI, the neurohormone peptide ALST1 was conjugated by CuAAC to the azide- and amine functionalized superparamagnetic iron oxide nanoparticles, since ALST1 is supposed to target difficult-to-treat neuroendocrinic tumours due to its analogy to galanin and somastatin receptor ligands. The organic fluorescent dye cyanine 5 (Cy5) was also conjugated to the silica-coated superparamagnetic iron oxide nanoparticles (SPIONs) via a NHS-ester to the amines to enable cell uptake studies by fluorescence microscopy. These constructs were characterized by TEM, dynamic light scattering (DLS), and IR. The amino acids of the conjugated ALST1 were determined by the HPLC method as described before for peptide-modified silica nanoparticle surfaces. Then, the relaxivity r2 was measured at 7 T. However, a r2 value of 27 L/mmolFe·s for the dual ALST1-/Cy5-functionalized silica-coated SPIONs was not comparable to T2 contrast agents in clinical use, since their relaxivity is commonly determined at 1.5 T, and no such instrument was available. However, it can be assumed that the synthesized dual
ALST1-/Cy5-functionalized silica-coated SPION would show a lower r2 at 1.5 T than at 7T. Commercial T2 MRI contrast agents like VSOP-C184 from Ferropharm show at r2 values of about 30 L/mmolFe·s at 1.5 T. Still, the relaxivity of the new material has some potential for application as a T2 contrast agent. Then, the material was used in cell uptake studies by fluorescence microscopy with the conjugated Cy5 dye as a probe. The dual
ALST1-/Cy5-functionalized silica-coated SPION showed a high degree of agglomeration with no cellular uptake unlike described for ALST1-functionalized nanoparticles in literature. It is assumed that upon agglomeration of the particles, constructs form which are unable to be internalized by the cellular endocytotic pathways anymore. As a future perspective, the tendency of the particle to agglomerate should be reduced by changing the coating material to polyethylene glycol (PEG) or chitosan, which are known to be bio-compatible, bio-degradable and prevent agglomeration.
In the third part of the work, the rhenium compound [ReBr(CO)3(L)] with L = 2-phenyl-1H-imidazo[4,5-f][1,10]phenanthroline and its manganese analogue were synthesized by heating the ligand and rhenium pentacarbonyl bromide or and manganese pentacarbonyl bromide respectively, in toluene. However, [MnBr(CO)3(L)] was unstable upon illumination by UV light at 365 nm. Thus, it was dismissed for further application. The photophysical properties of [ReBr(CO)3(L)] were explored, by determination of the excited-state life time by the time-correlated single-photon counting (TCSPC) method and the quantum yield by a fluorescence spectrometer equipped with an integration sphere. A value of = 455 ns, a Stokes shift of 197 nm and a rather low quantum yield =were found. Metal complexes are supposed to have superior properties compared to organic dyes due to their large Stokes shifts, long excited-state life times, and high quantum yields. Thus, amine- and azide-functionalized detonation nanodiamond (DND) as an alternative biological inert carrier system was functionalized with ALST1 to enhance its cell uptake properties. A luminescent probe for cell uptake studies using fluorescence microscopy was also attached, either based on the new rhenium complex or the commercially available organic dye Cy5, respectively. The aldehyde-functionalized rhenium complex was conjugated to the DND via oxime ligation, which is known to be a mild and catalyst-free conjugation method. The amount of peptide ALST1 on the DND was analyzed and quantified after acid hydrolysis and PITC derivatization by HPLC as described before. Then, the ALST1-/luminescent probe-functionalized DND was investigated for its photophysical properties by fluorescence spectroscopy. The Cy5-functionalized material showed a slightly lower fluorescence performance in aqueous solution than reported in literature and commercial suppliers with a life time < 0.4 ns and quantum yields not determinable by integration sphere due to the week signal intensity. The rhenium complex-functionalized material had a very low signal intensity in only aqueous medium, and thus determination of life times and quantum yield by fluorescence spectroscopy was not possible. After incubation with MDA-MB 231 cells, the Cy5-functionalized DND could easily be detected due to its red fluorescence. However, it was not possible to visualize the rhenium complex-functionalized DND with fluorescence microscopy due to the low fluorescence intensity of the complex in aqueous medium and the lack of proper filters for the fluorescence microscope. Cy5-functionalized DND did not show any cellular uptake in fluorescence microscopy after conjugation with ALST1. Since the nanodiamond surface is known to strongly adsorb peptides and proteins, it is assumed that the peptide chain is oriented perpendicular to the nanoparticle surface and thus not able to interact with cell membrane receptors to promote cell uptake of the particles. As a future perspective, the ALST1-promoted cellular uptake of the DND should be improved by using different linker systems for peptide conjugation to prevent adsorption of the peptide chain on the particle surface.
The new analytical methods for amino-, azide-, and peptide-functionalized nanoparticles have great potential to assist in the quantification of nanoparticle surface modifications by UV/Vis spectroscopy and HPLC. The determination of surface amine and azide groups based on the cleavage of conjugated Fmoc-Rink amide linker and detected by UV/Vis spectroscopy is applicable to all amine-/azide-functionalized nanomaterials. However, particles which form very stable suspension with the cleavage mixture can cause quantification problems due to scattering, making an accurate quantification of dibenzofulvene and its piperidine adduct impossible. The detection of tyrosine- and histidine-containing peptides based on the Pauly reaction is well-suited as a fast and easy-to-perform qualitative demonstration of successful peptide surface conjugation. However, its major drawback as a colourimetric approach is that coloured particles cannot be evaluated by this method. The amino acid analysis based on HPLC after acid hydrolysis of peptides conjugated to nanoparticle surfaces to its individual building blocks and subsequent derivatization with PITC, can be used on all nanomaterials with peptide or protein surface modification. It allows detection of amino acids down to picomolar concentrations and even enables analysis of very small peptide surface loadings. However, the resulting HPLC traces are difficult to analyze.
Three new analytical methods based on UV/Vis and HPLC techniques have been developed and established. They assisted in the characterization of the synthesized DND and SPIONs with dual functionalization by ALST1 and Cy5 or [ReBr(CO)3(L)], respectively. However, the nanomaterials showed no cellular uptake due to a high tendency to agglomerate. The cellular uptake should be improved and the tendency to agglomerate of the SPIONs should be reduced by changing the surface coating from silica to either PEG or chitosan. Furthermore, different linker systems for connecting peptides to DND surfaces should be synthesized and evaluated to reduce potential peptide chain adsorption.
Luminescent organotransition metal complexes are of much current interest. As the large spin-orbit coupling of 2nd and 3rd row transition metals usually leads to rapid intersystem crossing from S1 to T1, which enables phosphorescence, there is a special interest in using triplet-emitting materials in organic or organometallic light emitting diodes (OLEDs). Marder et al. have found that, reductive coupling of both para-R-substituted diarylbutadiynes and diaryldodecatetraynes on Rh(PMe3)4X leads to quantitative yields of bis(arylethynyl)-rhodacyclopentadienes with complete regiospecificity (R = BMes2, H, Me, OMe, SMe, CF3, CN, CO2Me, NMe2, NO2, C≡C-TMS and X = -C≡C-TMS, -C≡C-C6H4-4-NMe2, -C≡C-C≡C-C6H4-4-NPh2, Me, Cl).47,49 Unexpectedly, these compounds show intense fluorescence rather than phosphorescence (ɸf = 0.33-0.69, t = 1.2 3.0 ns). The substituent R has a significant influence on the photophysical properties, as absorption and emission are both bathochromically shifted compared to R = H, especially for R = π-acceptor.
To clarify the mechanism of the formation of the rhodacyclopentadienes, and to investigate further their unique photophysical properties, a series of novel, luminescent rhodacyclopentadienes with dithiocarbamate as a bidentate ligand at the rhodium centre has been synthesised and characterised (R = NO2, CO2Me, Me, NMe2, SMe, Ar = C6F4-4-OMe). The rhodacyclopentadienes have been formed via reductive coupling of diaryl undecatetraynes with [Rh(k2-S,S`-S2CNEt2)(PMe3)2]. The structures of a series of such compounds were solved by single crystal X-ray diffraction and are discussed in this work. The compounds were fully characterised via NMR, UV/Vis and photoluminescence spectroscopy as well as by elemental analysis, high-resolution mass spectrometry (HRMS) and X-ray diffraction.
When heating the reactions, another isomer is formed to a certain extent. The so-called dibenzorhodacyclopentadienes already appeared during earlier studies of Marder et al., when acetylacetonate (acac) was employed as the bidentate ligand at the Rh-centre. They are probably formed via a [4+2] cycloaddition reaction and C-H activation, followed by a β-H shift.
Use of the perfluorinated phenyl moiety Ar = C6F4-4-OMe provided a total new insight into the mechanism of formation of the rhodacyclopentadiene isomers and other reactions. Besides the formation of the expected rhodacyclopentadiene, a bimetallic compound was generated, isolated and characterised via X-ray crystallography and NMR spectroscopy, elemental analysis and high resolution mass spectrometry.
For further comparison, analogous reactions with [Rh(k2 S,S` S2CNEt2)(PPh3)2] and a variety of diaryl undecatetraynes (R = NO2 CO2Me, Me, NMe2, SMe, Ar = C6F4-4-OMe) were carried out. They also yield the expected rhodacyclopentadienes, but quickly react with a second or even third equivalent of the tetraynes to form, catalytically, alkyne cyclotrimerisation products, namely substituted benzene derivatives (dimers and trimers), which are highly luminescent. The rhodacyclopentadienes (R = NO2, CO2Me, Me, SMe, Ar = C6F4-4-OMe) are stable and were isolated. The structures of a series of these compounds were obtained via single crystal X-ray crystallography and the compounds were fully characterised via NMR, UV/Vis and photoluminescence spectroscopy as well as by elemental analysis and HRMS.
Another attempt to clarify the mechanism of formation of the rhodacyclopentadienes involved reacting a variety of diaryl 1,3-butadiynes (R = CO2Me, Me, NMe2, naphthyl) with [Rh(k2 S,S` S2CNEt2)(PMe3)2]. The reactions stop at an intermediate step, yielding a 1:1 trans π-complex, confirmed by single crystal X-ray diffraction and NMR spectroscopy. Only after several weeks, or under forcing conditions (µw / 80 °C, 75 h), the formation of another major product occurs, having bound a second diaryl 1,3-butadiyne. Based on earlier results of Murata, the product is identified as an unusual [3+2] cycloaddition product, ϭ-bound to the rhodium centre.
Stickstoffmonoxid (NO) ist ein gasförmiges, relativ stabiles Radikal, das in Pflanzen u. a. durch Reduktion aus Nitrit unter Katalyse des Enzyms Nitratreduktase gebildet wird. In tierischen Organismen wird NO dagegen über einen oxidativen Syntheseweg aus der Ami-nosäure L-Arginin katalysiert durch verschiedene Isoformen der NO-Synthasen (NOS) hergestellt. Es besitzt im tierischen System vielfältige Funktionen u. a. als Neurotransmitter sowie Blutfluss und -druck regulierendes Agens. Im Pflanzenreich werden NO u. a. Aufgaben bei der Regulierung von Spaltöffnungen, der Abwehr von Pathogenen sowie der Differenzierung der Xylemelemente zugeschrieben. Die vorliegende Arbeit verfolgte zwei Ziele: - Erforschung alternativer oxidativer Synthesewege von NO in Pflanzen und - Untersuchung der NO-Spezifität der DAF-Fluoreszenzfarbstoffe ausgehend Diskrepanzen zwischen Daten aus Floureszenzanalysen und der Gasphasen-Chemilumineszenz in früheren Arbeiten unserer Arbeitsgruppe und zahlreichen weiteren Publikationen. a) NO-Produktion aus Hydroxylaminen Hydroxylamin ist ein Zwischenprodukt bei der bakteriellen Denitrifizierung und wurde auch als Intermediat bei der Nitratreduktion in Pflanzen diskutiert. Hier wird gezeigt, dass Tabaksuspensionzellen in der Lage waren, exogenes Hydroxylamin schon in sehr niedrigen Konzentrationen (4 µM) zu NO zu oxidieren. Auch ein anderes HA-Derivat, nämlich der Hemmstoff der Alternativen Oxidase (AOX) in Mitochondrien, Salicylhydroxamsäure (SHAM), wurde zu NO oxidiert. Die Vermutung, reaktive Sauerstoffspezies (ROS) könn-ten bei diesem Oxidationsprozess eine Rolle spielen, wurde überprüft: Nach Einwirkung des ROS-abbauenden Enzyms Superoxid-Dismutase (SOD) konnte aber überraschender-weise keine Verminderung, sondern eher eine Steigerung der NO-Emission beobachtet werden. Die Rolle der SOD in diesem Reaktionsprozess ist daher noch nicht verstanden. b) NO-Detektion mittels Fluoreszenzindikatoren Zur Visualisierung und Lokalisierung von NO in tierischen und pflanzlichen Zellen und Geweben (in situ) mittels mikroskopischer (LSM) oder fluorimetrischer Methoden werden Fluoreszenzfarbstoffe, z. B. DAF-2 oder DAF-FM verwendet. Diese Farbstoffe reagieren mit NO zu stark fluoreszierenden Triazol-Derivaten. Eine Situation, in der Pflanzen u. a. mit NO-Freisetzung reagieren, ist der Pathogenbefall. Wir untersuchten die Reaktion von Tabaksuspensionszellen auf den pilzlichen Elicitor Cryptogein, ein Protein des Oomyceten Phytophthora cryptogea. Im Filtrat der Zellen, die mit Cryptogein behandelt wurden, zeigte sich nach Zugabe von DAF-Farbstoffen ein starker Fluoreszenzanstieg. Um die fluoreszenzerhöhenden Stoffe zu charakterisieren, wurde das Filtrat vor der DAF-Zugabe verschiedentlich behandelt. Bei Zugabe von KCN bzw. Katalase zum Überstand, verringerte sich der Fluoreszenzanstieg. Gleichzeitige Behandlung der Zellen mit Cryptogein sowie dem NADPH-Oxidase-Inhibitor DPI unterband den Fluoreszenzanstieg im Überstand nahezu komplett. Enzym-Assays mit Amplex Rot zeigten die Anhäufung von H2O2 im Filtrat der elicitierten Zellen. Neben ROS werden von Pflanzenzellen auch Peroxidasen in den Apoplasten sekretiert, die mit Hilfe von H2O2 für eine verstärkte Quervernetzung der Zellwand sorgen. Sowohl in unbehandelten Kontrollzellen als auch in elicitierten Zellen wurde Peroxidase-Aktivität nachgewiesen. Nach Zugabe von H2O2 und DAF-2 zum Filtrat von Kontrollzellen ergab sich ein Fluoreszenzanstieg ähnlich dem im Filtrat von behandelten Zellen. Mit Hilfe eines einfachen In-vitro-Systems aus Meerettich-Peroxidase (MR-PO), Wasser-stoffperoxid (H2O2) und DAF-2 konnten noch höhere Fluoreszenzwerte erzielt werden, was die Vermutung der Fluoreszenzerhöhung ohne Anwesenheit von NO erhärtete. Um diese nicht aus einer Reaktion mit NO resultierenden DAF-Produkte näher zu charak-terisieren, wurden Trennungen mittels Umkehrphasen-Hochdruck-Flüssigkeitschro-matographie mit Fluoreszenzdetektion (RP-HPLC-FL) und Massenspektrometrie (UPLC-MS) durchgeführt. Dabei wurden tatsächlich zwei neue Reaktionsprodukte festgestellt, die sich eindeutig von DAF-2T unterschieden. Letzteres konnte nur bei Hinzufügen des NO-Donors DEA-NO detektiert werden. Zur Erfassung von intrazellulären Reaktionsprodukten von DAF wurden die chromatogra-phischen Trennmethoden auch auf Extrakte von mit DAF-2 DA aufgeladenen und danach elicitierten Zellen angewandt. Bei dieser Auftrennung tauchten noch mehr DAF-Reaktionsprodukte auf. Die Hauptfluoreszenz, die auch bei nicht inkubierten Zellen auf-trat, konnte auf eine Reihe sehr früh eluierender Substanzen zurückgeführt werden. Die zwei DAF-Derivate aus dem Überstand inkubierter Zellen bzw. der In-vitro-Reaktion (MR-PO+H2O2+DAF-2) tauchten jedoch überhaupt nicht auf. Vorläufige massenspektrometrische Analysen legen nahe, dass es sich bei den in Abwe-senheit von NO gebildeten zwei Verbindungen um isomere Dimere von DAF-2 handelt.
In the first part of the work three polycarbazoles poly[N-((4-dimesitylboryl)-3,5-dimethylphenyl)-carbazole]-2,7-diyl P1, poly[N-((4-dimesitylboryl)-3,5-dimethylphenyl)-carbazole]-3,6-diyl P2 and poly[N-(4-(diphenylmethylene)-phenyl)- carbazole]-2,7-diyl P3 were synthesized by Yamamoto coupling reaction and their spectroscopic and electrochemical properties were investigated. Absorption and fluorescence characteristics of P1 and P3 were found to be similar to other 2,7-linked polycarbazoles, whereas P2 shows a CT absorption band arising from a shift of electron density from the nitrogen of the carbazole donor to the triarylborane acceptor. This causes a negative solvatochromic absorption and a positive solvatochromic fluorescence behaviour and is responsible for the significantly enlarged fluorescence quantum efficiency in solution and solid state compared to other 3,6-linked polycarbazoles. Thus the spectroscopic properties are governed by the connection pattern: the 2,7-linked polycarbazoles are not affected by the acceptor substituent due to the rigid poly-para-phenylene-like backbone structure, whereas the 3,6-linked polycarbazole P2 is dominated by the properties of the monomer unit due to its more flexible (less conjugated) structure. The oxidative processes of P1-P3 have been investigated in detail by cyclic voltammetry, which are similar to known 2,7- and 3,6-polycarbazoles. The reversible reduction found for P1 and P2, respectively, is attributed to the reduction of the triarylborane moiety. No reduction process referring to the carbazole moiety was observed. Due to its better solubility compared to P1 and P3 only P2 was used as active layer in an OLED device (ITO/P2/Al). The electroluminescence spectrum revealed CIE coordinates of (0.17, 0.21). In the second part of the work the low band gap polyradical poly{[((2,3,4,5,6-pentachlorophenyl)-bis(2,3,5,6-tetrachlorophenyl)methyl radical)-4,4’-diyl]-alt-4,4’-bis(vinylphenyl)-4-(2-ethylhexyloxy)phenylamin} P4 was synthesized by Horner-Emmons reaction. It shows an IV-CT band in the NIR, which arises from an ET from the triarylamine donor to the PCTM radical acceptor. This transition is confined to one monomer unit as deduced from comparison with the monomer spectra. HOMO and LUMO of P4 determined by cyclic voltammetry are at -5.5 and -4.5 eV, respectively. The smaller electrochemical band gap (1.0 eV) compared to the optical band gap (1.2 eV) is probably caused by ion pairing effects in the electrochemical experiments and indicates a low exciton binding energy. Femtosecond-pump-probe transient absorption spectroscopy revealed the spectral features of the oxidized triarylamine donor and the reduced PCTM acceptor similar to the spectra obtained separately for positive and negative potentials by spectroelectrochemistry. Thus the ET event causing the IV-CT absorption band could unambiguously be identified. The decay of the IV-CT state was found to be biexponential. The fast solvent dependent decay component is ascribed to the direct decay from the IV-CT state to the ground state, whereas the slow solvent independent decay component is tentatively attributed to an equilibrium formation of the IV-CT state and a completely charge separated state formed by charge migration along the polymer backbone. Well balanced ambipolar charge transport with hole and electron mobilities of ca. 3 × 10-5 cm2 V-1 s-1 was found in OFET devices (BG/TC structure) comprising an additional insulating organic PPcB layer. Polymer/polymer BHJ solar cell devices with the structure glass/ITO/PEDOT:PSS/(P3HT/P4)/Ca/Al yielded a power conversion efficiency of 3.1 × 10-3 %, VOC = 0.38 V, JSC = 2.8 × 10-2 mA cm-2 and FF = 0.29 for the 1:4 (P3HT/P4) blend ratio. The improper solid state morphology of P4 that causes the unsatisfying performance of OFET and solar cell devices renders P4 less suitable for these applications, whereas the hypothesis of charge migration in the excited state is worth to be investigated in more detail.
The main objective of this thesis was the design and synthesis of perylene bisimide dyes with sufficient water-solubility for the construction of self-assembled architectures in aqueous solutions. Beside these tasks another goal of this project was the control over the self-assembly process in terms of aggregate size and helicity, respectively. Within this thesis an appropriate synthesis for spermine-functionalized perylene bisimide dyes was developed and conducted successfully. The characterization of these building blocks and their course of self-assembly were investigated by NMR, UV/Vis and fluorescence spectroscopy as well as by atomic force and transmission electron microscopy. For the better understanding of the experimental results theoretical calculations were performed.
Die Möglichkeit, durch Beeinflussung der Interaktion der Gegenspieler Apobec3G und Vif, ein neuartiges Medikament gegen HIV zu entwickeln, ist in der Literatur bereits vielfach beschrieben (Argyris und Pomerantz 2004, Cullen 2006, Sheehy et al. 2003). Als Teil des angeborenen Immunsystems bietet die Aufrechterhaltung der antiviralen Eigenschaften von Apobec3G einen viel versprechenden Ansatzpunkt, die Infektiosität des HI-Virus einzudämmen.
RN18 ist als ein Vif-Antagonist in der Literatur beschrieben (Nathans et al. 2008). Um Substanzen ausfindig zu machen, die den Abbau von Apobec3G durch Vif
verhindern können, wurden in dieser Arbeit niedermolekulare Substanzen auf deren Tauglichkeit diesbezüglich getestet. In einem ersten Schritt (Screening) wurde die Wirksamkeit der Testsubstanzen bei einer Konzentration von 30 μM ermittelt. Bei Substanzen, die eine ähnliche Hemmung des Abbaus des Reporterproteins EYFP-A3G im Vergleich zu RN18 bewirkten, wurde eine quantitative Analyse zur genaueren Bestimmung der halbmaximalen Hemm-konzentration durchgeführt (Titration).
Einerseits wurden Derivate des bekannten Vif-Antagonisten RN18 getestet. Durch schrittweise Verbesserung der Wirksamkeit der RN18-Derivate gelang es schließlich Substanzen zu finden, für die ein besserer Effekt als für RN18 ermittelt werden konnte, den Abbau von Apobec3G durch Vif zu verhindern. Zur Beurteilung der Ergebnisse wurde der EC50-Wert berechnet, um die Wirksamkeit der Substanzen miteinander vergleichen zu können. Es wurde nach Substanzen gesucht, die bei möglichst geringen Konzentrationen wirken. Das RN18-Derivat mit dem besten Ergebnis war FM86 (EC50-Wert: 4.5 μM).
Andererseits wurden niedermolekulare Substanzen aus verschiedenen Arbeitsgruppen untersucht, um weitere Substanzen zu finden, die ebenso wie RN18 in der Lage sind, die Vif/Apobec3G-Interaktion zu hemmen. Auch hier
wurden mehrere Substanzen ermittelt, die eine bessere Wirksamkeit als RN18 erkennen ließen. Derivate der Substanz CBA77a konnten am effektivsten den Abbau von Apobec3G durch Vif verhindern. Das beste Ergebnis
wurde für die Testsubstanz CBA82 ermittelt (EC50-Wert: 2.8 μM). Ob die Ergebnisse der Testsubstanzen ausschließlich auf die Hemmung der Vif/A3G Interaktion zurückzuführen sind, kann letztendlich nicht abschließend beurteilt werden. Eine Erweiterung des Testsystems durch
unsere Arbeitsgruppe sieht daher vor, falsch positive Ergebnisse zu erkennen.
Die Möglichkeit, durch Beeinflussung der Interaktion der Gegenspieler Apobec3G und Vif, ein neuartiges Medikament gegen HIV zu entwickeln, ist in der Literatur bereits vielfach beschrieben (Argyris und Pomerantz 2004, Cullen 2006, Sheehy et al. 2003). Auch hier wurden mehrere Substanzen ermittelt, die eine bessere Wirksamkeit als RN18 erkennen ließen. Derivate der Substanz CBA77a konnten am effektivsten den Abbau von Apobec3G durch Vif verhindern. Das beste Ergebnis
wurde für die Testsubstanz CBA82 ermittelt (EC50-Wert: 2.8 μM).
Ob die Ergebnisse der Testsubstanzen ausschließlich auf die Hemmung der Vif/A3G Interaktion zurückzuführen sind, kann letztendlich nicht abschließend beurteilt werden. Eine Erweiterung des Testsystems durch unsere Arbeitsgruppe sieht daher vor, falsch positive Ergebnisse zu erkennen.
The cyclic nucleotides cAMP and cGMP are two ubiquitous important second messengers, which regulate diverse physiological responses from vision and memory to blood pressure and thrombus formation. They act in cells via cAMP- and cGMP-dependent protein kinases (PKA and GK), cyclic nucleotide-gated channels and Epac. Although the concept of cyclic nucleotide signalling is well developed based on classical biochemical studies, these techniques have not allowed to analyze cAMP and cGMP in live cells with high temporal and spatial resolution. In the present study fluorescence resonance energy transfer was used to develop a technique for visualization of cAMP and cGMP in live cells and in vitro by means of fluorescent biosensors. Ligand-induced conformational change in a single nucleotide-binding domain flanked with green fluorescent protein mutants was used for dynamic, highly sensitive measurements of cAMP and cGMP. Such biosensors retained binding properties and chemical specificity of unmodified domains, allowing to image cyclic nucleotides in a physiologically relevant range of concentrations. To develop cAMP-sensors, binding domains of PKA, Epac and cAMP-gated HCN-channel were used. cGMP-sensors were based on single domains of GK and phosphodiesterases (PDEs). Sensors based on Epac were used to analyze spatio-temporal dynamics of cAMP in neurons and macrophages, demonstrating that cAMP-gradients travel with a high speed (~ 40 μm/s) throughout the entire cytosol. To understand the mechanisms of cAMP-compartmentation, kinetics properties of phosphodi-esterase (PDE2) were, next, analyzed in aldosterone producing cells. PDE2 is able to rapidly hydrolyze extensive amounts of cAMP, so that the speed of cAMP-hydrolysis is much faster than that of its synthesis, which might serve as a basis of compartmentation. cAMP-sensors were also used to develop a clinically relevant diagnostic method for reliable detection of β1-adrenergic receptor autoantibodies in cardiac myopathy patients, which has allowed to significantly increase the sensitivity of previously developed diagnostic approaches. Conformational change in a single binding domain of GK and PDE was, next, used to create novel fluorescent biosensors for cGMP. These sensors demonstrated high spatio-temporal resolution and were applied to analyze rapid dynamics of cGMP production by soluble and particulate guanylyl cyclases as well as to image cGMP in mesangial cells. In summary, highly sensitive biosensors for cAMP and cGMP based on single cyclic nucleotide-binding domains have been developed and used in various biological and clinically relevant applications.
Die Fluoreszenzmikroskopie ist eine vielseitig einsetzbare Untersuchungsmethode für biologische Proben, bei der Biomoleküle selektiv mit Fluoreszenzfarbstoffen markiert werden, um sie dann mit sehr gutem Kontrast abzubilden. Dies ist auch mit mehreren verschiedenartigen Zielmolekülen gleichzeitig möglich, wobei üblicherweise verschiedene Farbstoffe eingesetzt werden, die über ihre Spektren unterschieden werden können.
Um die Anzahl gleichzeitig verwendbarer Färbungen zu maximieren, wird in dieser Arbeit zusätzlich zur spektralen Information auch das zeitliche Abklingverhalten der Fluoreszenzfarbstoffe mittels spektral aufgelöster Fluoreszenzlebensdauer-Mikroskopie (spectrally resolved fluorescence lifetime imaging microscopy, sFLIM) vermessen. Dazu wird die Probe in einem Konfokalmikroskop von drei abwechselnd gepulsten Lasern mit Wellenlängen von 485 nm, 532nm und 640nm angeregt. Die Detektion des Fluoreszenzlichtes erfolgt mit einer hohen spektralen Auflösung von 32 Kanälen und gleichzeitig mit sehr hoher zeitlicher Auflösung von einigen Picosekunden. Damit wird zu jedem detektierten Fluoreszenzphoton der Anregungslaser, der spektrale Kanal und die Ankunftszeit registriert. Diese detaillierte multidimensionale Information wird von einem Pattern-Matching-Algorithmus ausgewertet, der das Fluoreszenzsignal mit zuvor erstellten Referenzpattern der einzelnen Farbstoffe vergleicht. Der Algorithmus bestimmt so für jedes Pixel die Beiträge der einzelnen Farbstoffe.
Mit dieser Technik konnten pro Anregungslaser fünf verschiedene Färbungen gleichzeitig dargestellt werden, also theoretisch insgesamt 15 Färbungen. In der Praxis konnten mit allen drei Lasern zusammen insgesamt neun Färbungen abgebildet werden, wobei die Anzahl der Farben vor allem durch die anspruchsvolle Probenvorbereitung limitiert war. In anderen Versuchen konnte die sehr hohe Sensitivität des sFLIM-Systems genutzt werden, um verschiedene Zielmoleküle voneinander zu unterscheiden, obwohl sie alle mit demselben Farbstoff markiert waren. Dies war möglich, weil sich die Fluoreszenzeigenschaften eines Farbstoffmoleküls geringfügig in Abhängigkeit von seiner Umgebung ändern. Weiterhin konnte die sFLIM-Technik mit der hochauflösenden STED-Mikroskopie (STED: stimulated emission depletion) kombiniert werden, um so hochaufgelöste zweifarbige Bilder zu erzeugen, wobei nur ein einziger gemeinsamer STED-Laser benötigt wurde.
Die gleichzeitige Erfassung von mehreren photophysikalischen Messgrößen sowie deren Auswertung durch den Pattern-Matching-Algorithmus ermöglichten somit die Entwicklung von neuen Methoden der Fluoreszenzmikroskopie für Mehrfachfärbungen.
We demonstrate two-quantum (2Q) coherent two-dimensional (2D)electronic spectroscopy using a shot-to-shot-modulated pulse shaper and fluorescence detection. Broadband collinear excitation is realized with the supercontinuum output of an argon-filled hollow-core fiber, enabling us to excite multiple transitions simultaneously in the visible range. The 2Q contribution is extracted via a three-pulse sequence with 16-fold phase cycling and simulated employing cresyl violet as a model system. Furthermore, we report the first experimental realization of one-quantum−two-quantum (1Q-2Q) 2D spectroscopy, offering less congested spectra as compared with the 2Q implementation. We avoid scattering artifacts and nonresonant solvent contributions by using fluorescence as the observable. This allows us to extract quantitative information about doubly excited states that agree with literature expectations. The high sensitivity and background-free nature of fluorescence detection allow for a general applicability of this method to many other systems.
Pyrene is a polycyclic aromatic hydrocarbon (PAH) that has very interesting photophysical properties which make it suitable for a broad range of applications. The 2,7-positions of pyrene are situated on nodal planes in both the HOMO and LUMO. Hence, electrophilic reactions take place at the 1-, 3-, 6-, and 8-positions. The goal of this project was to develop novel pyrene derivatives substituted at the 2- and 2,7-positions, with very strong donors or/and acceptors, to achieve unprecedented properties and to provide a deeper understanding of how to control the excited states and redox properties. For that reason, a julolidine-type moiety was chosen as a very strong donor, giving D-π and D-π-D systems and, with Bmes2 as a very strong acceptor, D-π-A system. These compounds exhibit unusual photophysical properties such as emission in the green region of the electromagnetic spectrum in hexane, whereas all other previously reported pyrene derivatives substituted at the 2,7-positions show blue luminescence. Furthermore, spectroelectrochemical measurements suggest very strong coupling between the substituents at the 2,7-positions of pyrene in the D-π-D system. Theoretical studies show that these properties result from the very strong julolidine-type donor and Bmes2 acceptor coupling efficiently to the pyrene HOMO-1 and LUMO+1, respectively. Destabilization of the former and stabilization of the latter lead to an orbital shuffle between HOMO and HOMO 1, and LUMO and LUMO+1 of pyrene. Consequently, the S1 state changes its nature sufficiently enough to gain higher oscillator strength, and the photophysical and electrochemical properties are then greatly influenced by the substituents.
In another project, further derivatives were synthesized with additional acceptor moieties at the K-region of pyrene. These target derivatives exhibit strong bathochromically shifted absorption maxima (519-658 nm), which is a result of the outstanding charge transfer character introduced into the D-π-D pyrene system through the additional acceptor moiety at the K-region. Moreover, emission in the red to NIR region with an emission maximum at 700 nm in CH2Cl2 is detected. The excited state lives unusual long for K-region substituted pyrenes; however, such a lifetime is rather typical for 2,7-substituted pyrene derivatives.
The polycyclic aromatic hydrocarbon perylene, especially perylene diimide, has received considerable attention in recent years and has found use in numerous applications such as dyes, pigments and semiconductors. Nevertheless, it is of fundamental importance to understand how to modulate the electronic and photophysical properties of perylene depending on the specific desired application. Perylenes without carboxyimide groups at the peri positions are much less well studied due to the difficulties in functionalizing the perylene core directly. In particular, only ortho heteroatom substituted perylenes have not been reported thus far (exception: (Bpin)4-Per was already reported by Marder and co-workers). Thus, the effect of substituents on the ortho positions of the perylene core has not been investigated.
Two perylene derivatives were synthesized that bear four strong diphenylamine donor or strong Bmes2 acceptor moieties at the ortho positions. These compounds represent the first examples of perylenes substituted only at the ortho positions with donors or acceptors.
The investigations show that the photophysical and electronic properties of these derivatives are unique and different compared to the well-studied perylene diimides. Thus, up to four reversible reductions or oxidations are possible, which is unprecedented for monomeric perylenes. Furthermore, the photophysical properties of these two ortho-substituted derivatives are unusual compared to reported perylenes on many regards. Thus, large Stokes shifts are obtained, and the singlet excited state of these derivatives lives remarkably long with intrinsic lifetimes of up to 94 ns.
In a cooperation with Dr. Gerard P. McGlacken at University College Cork in Ireland, different quinolones were borylated using an iridium catalyst system to study the electronic and steric effect of the substrates. It was possible to demonstrate that the Ir-catalyzed borylation with the dtbpy ligand allows the direct borylation of various 4-quinolones at the 6- and 7-positions. Thus, later stage functionalization is possible with this method and more highly functionalized quinolones are also compatible with this mild reaction conditions.
Kohlenhydrat-Protein-Wechselwirkungen sind häufig entscheidend beteiligt an verschiedenen einer Infektion oder malignen Erkrankung zugrunde liegenden molekularen Erkennungs-prozessen, die zu Adhäsion, Zell-Zell-Interaktion sowie Immunreaktion und -toleranz führen. Trotz der hohen Relevanz für Diagnostik und Therapie dieser Erkrankungen sind die betreffenden Strukturen und Mechanismen bisher nur ungenügend untersucht und verstanden. Ziel dieser stark interdisziplinär angelegten Arbeit war es daher, Methoden der Fachbereiche Chemie und Pharmazie, Biologie und Medizin, aber auch Physik zu kombinieren, um Kohlenhydraterkennungsprozesse im Detail zu untersuchen und auf dieser Basis strukturell neuartige diagnostische und therapeutische Anwendungen zu entwerfen.
Die hochkomplexe Zusammensetzung einer Zelloberfläche wurde zunächst auf ihren Glycan-anteil reduziert und stark vereinfacht auf der Oberfläche sogenannter Glycochips imitiert. Die verwendeten Systeme auf Basis einer Gold- bzw. Glasoberfläche ergänzen sich optimal in ihrer Eignung für komplementäre analytische Methoden wie Massenspektrometrie sowie quantifizierbare Fluoreszenzspektroskopie.
Der Übergang auf die lebende Zelloberfläche gelang mit Hilfe des Metabolic Glyco-engineering, das die kovalente Präsentation definierter Motive durch eine Cycloaddition zwischen zwei bioorthogonalen Reaktionspartnern (z.B. Azid und Alkin) ermöglicht.
Auf diese Weise wurden in Zusammenarbeit mit der Arbeitsgruppe Sauer (Universität Würzburg) zunächst die Dichte und Verteilung verschiedener Oberflächenglycane auf humanen Zellen mittels hochauflösender Fluoreszenzmikroskopie (dSTORM) bestimmt. Diese Parameter zeigten im Modell des Glycochips einen entscheidenden Einfluss auf Bindungsereignisse und multivalente Erkennung und zählen auch auf natürlichen Zelloberflächen – in engem Zusammenhang mit der lateralen und temporalen Dynamik der Motive – zu den wichtigen Faktoren molekularer Erkennungsprozesse.
Die gezielte Modifikation zellulärer Oberflächenglycane eignet sich aber auch selbst als Methode zur Beeinflussung molekularer Wechselwirkungsprozesse. Dies wurde anhand des humanpathogenen Bakteriums S. aureus gezeigt, dessen Adhäsion auf Epithelzellen der Blasenwand durch Metabolic Glycoengineering partiell unterdrückt werden konnte.
In einem ergänzenden Projekt wurden zwei potentielle Metabolite eines konventionellen Antibiotikums – des Nitroxolins – mit bakteriostatischer sowie antiadhäsiver Wirksamkeit dargestellt. Diese dienten als Referenzsubstanzen zur Verifizierung der postulierten Struktur der Derivate, werden aber auch selbst auf ihr Wirkprofil hin untersucht. Gleichzeitig stehen sie zusammen mit der Grundverbindung zudem als Referenz für die Wirkstärke potentieller neu entwickelter Antiadhäsiva zur Verfügung.
The present thesis introduce different synthetic strategies towards a variety of polycyclic aromatic dicarboximides (PADIs) with highly interesting and diverse properties. This included tetrachlorinated, tetraaryloxy- and tetraaryl-substituted dicarboximides, fused acceptor‒donor(‒acceptor) structures as well as sterically shielded rylene and nanographene dicarboximides. The properties and thus the disclosure of structure‒property relationships of the resulting dyes were investigated in detail among others with UV‒vis absorption spectroscopy, fluorescence spectroscopy, cyclic voltammetry and single crystal X-ray analysis. For instance, some of the fused and substituted PADIs offer strong absorption of visible and near infrared (NIR) light, NIR emission and low-lying LUMO levels. On the contrary, intriguing optical features in the solid-state characterize the rylene dicarboximides with their bulky N-substituents, while the devised sterically enwrapped nanographene host offered remarkable complexation capabilities in solution.
Obwohl Protein-Mikroarrays ursprünglich aus dem gut entwickelten und fest etablierten DNA-Pendant entstanden sind, repräsentierte jedoch die Umstellung der Mikroarray-Technik von der DNA- auf die Proteinanalyse aufgrund der enormen physikalisch-chemischen Variabilität der Proteine, deren relativ niedrigen Stabilität und der komplexen Mikrospot-Kinetik eine große technologische Herausforderung. Deshalb setzt das Vorhaben, die Technik der Antikörper–Mikroarrays von ihrem konzeptuellen Zustand ausgehend zu einem robusten, real funktionierenden Werkzeug zu etablieren, nicht nur eine Vielzahl an technologischen Lösungen, sondern auch eine systematische und physikalisch begründete Herangehensweise in dieser technologischen Entwicklung voraus. Das waren im Wesentlichen die zwei wichtigsten, der eigentlichen Entwicklung der Antikörper-Mikroarrays untergeordneten Ziele der Arbeit. Mit dem Ziel, Antikörper-Mikroarrays prinzipiell zu etablieren und eine optimale Immobilisierungschemie für deren Herstellung zu finden, wurden im ersten Teil dieser Arbeit mehrere chemische Beschichtungen von Glasslides optimiert, unterschiedliche Spotting-Bedingungen von Antikörpern für verschiedene Oberflächen getestet und verschiedene Blockierungsverfahren und Strategien zur Aufbewahrung von Slides analysiert. Anschließend wurde eine Reihe von kommerziellen und selbst hergestellten chemisch beschichteten Slides unter den optimierten Bedingungen miteinander verglichen. Als Hauptergebnis dieser Untersuchung wurde die Herstellung der Antikörper-Microarrays etabliert. Unter anderem konnte im Zuge dieser systematischen Analyse gezeigt werden, dass Epoxysilan-modifizierte Oberflächen am besten geeignet sind. Diese Oberfläche ist heutzutage auf dem Gebiet der Protein-Microarrays am weitesten verbreitet und wurde für alle weiteren Studien innerhalb dieser Dissertation verwendet. Die Entwicklung der Antikörper-Mikroarrays in den letzten Jahren demonstrierte erhebliche Schwierigkeiten im Erreichen der nötigen Sensitivität und Reproduzierbarkeit. Um dieser Problematik auf den Grund zu gehen, und die Mikrospot-Kinetik experimentell untersuchen zu können, wurde im Rahmen dieser Arbeit eine modifizierte und für den Fall der Mikrorrays angepasste Variante des Two-Compartment Modells (TCM) entwickelt. TCM ermöglicht auf eine phänomenologische Weise, d.h., dass Diffusionskoeffizienten, Mischintensität oder Dichte der Bindungsstellen nicht bekannt sein müssen, eine quantitative experimentelle Analyse der Mikrospot-Kinetik unter Berücksichtigung von Effekten des Massentransports. Um die phänomenologischen TCM-Werte interpretieren zu können und um den Mechanismus der Mikrospot-Reaktion zu untersuchen, wurden auch andere, für die Mikrospot-Kinetik relevante, klassische Theorien an die Bedingungen der Mikrospot-Reaktion angepasst und mit dem modifizierten TCM mathematisch verbunden. Als das erste in der Mikroarray-Technologie mathematisch-physikalische Werkzeug dieser Art hat die hier entwickelte Theorie ein großes Potential, auch in den anderen verwandten Techniken wie DNA- oder Peptid-Mikroarrays Verwendung zu finden. Außerdem wurde innerhalb dieser Arbeit ein anderes einheitliches theoretisches Modell entwickelt, das eine kinetische Simulation von verschiedenen Reaktionsphasen sowohl für konventionelle als auch für Mikrospot-Immunoassays ermöglicht. Im Rahmen dieser Arbeit konnte für einen typischen Standard-Antikörper-Mikroarray theoretisch und experimentell eine lang andauernde, stark massentransportabhängige Mikrospot-Kinetik beschrieben werden. Es konnte gezeigt werden, dass das Erreichen eines thermodynamischen Gleichgewichts in Mikroarrays wegen eines relativ langsamen Ligandentransports zum Spot eine lange Zeit dauert, je nach Bindungskonstante, Diffusionsgeschwindigkeit und Ligandenkonzentration mehrere Stunden bis hin zu Wochen. In dieser Arbeit wurde ein neues physikalisches Konzept, das dem heutzutage dominierenden Blickwinkel, der sogenannten ambient analyte Theorie, opponierend gegenübersteht, formuliert. Auch konnten viele Konsequenzen fürs Design und die zukünftige Entwicklung dieser relativ neuen Technologie gezogen werden. Als eine logische Folge der massentransportlimitierten Reaktionen ist das Design eines Antikörper-Mikroarray ein kritischer Punkt für die Leistung des Verfahrens. Im Laufe der experimentellen und/oder theoretischen Betrachtungen konnte gezeigt werden, dass eine Reihe allgemeiner Parameter wie Größe eines Spots, Spotting-Muster, Inkubationsgeometrie, Volumen und Konzentration einer Probe, Viskosität des Inkubationspuffers und Mischintensität die Reaktionsraten auf den Spots insgesamt um mehrere Größenordnungen beeinflusst. Ist die maximale Rate des Massentransports in einem Mikroarray-Verfahren gewährleistet, kann dann auch die maximale Bindungsleistung der Spots, die durch die Dichte der Bindungsstellen, Bindungsaffinität, Inkubationszeit und andere relevante Parameter eingestellt wird, erreicht werden. Aber nicht nur in der Inkubationsphase, sondern auch bei den Wasch- und Detektionsschritten sollte die gleiche Liste der Parameter berücksichtigt werden. Durch die Optimierung all dieser Parametern konnte eine deutliche Verbesserung der Sensitivität von Antikörper-Mikroarrays in der Protein-Expressionsanalyse von klinischen Blutproben erzielt werden In einer weiteren Studie zur Analyse von unterschiedlichen Detektionsverfahren konnte die Sensitivität und Reproduzierbarkeit der etablierten Antikörper-Mikroarrays weiter verbessert werden. Eine Reihe unterschiedlicher Markierungssubstanzen mit NHS (N-hydroxysuccinimide) und ULS (universal linkage system) reaktiven Gruppen wurden innerhalb drei Detektionsverfahren untersucht: 1) eine direkte Probenmarkierung mit Fluoreszenzfarbstoffen, 2) Markierung der Probe mit Biotin-Substanzen und nachfolgender Detektion mittels fluoreszenzmarkierten Extravidin und 3) Markierung der Probe mit Fluorescein-Substanzen mit Anti-Fluorescein-Detektion. Aus den Erfahrungen der vorherigen kinetischen Untersuchungen wurde hier vorerst das kinetische Verhalten des Testsystems analysiert und optimale Inkubationsbedingungen festgelegt. Anschließend wurden optimale Konzentrationen all dieser Substanzen für die Markierung von Blutplasma bestimmt. Im Vergleich zur direkten Fluoreszenzmarkierung resultierten sich die indirekten Detektionsverfahren mit Biotin- und Fluorescein-Substanzen in wesentlich besseren Signal-zu-Hintergrund-Verhältnissen. In einer anschließenden Vergleichsanalyse zeigten sich einige Substanzen wie Biotin-ULS oder Fluoresceine-NHS als am besten geeignet für eine Protein-Expressionsanalyse von Blutplasma. Sensitivitäten im femtomolaren Bereich konnten mittels der etablierten Antikörper-Mikroarrays sowohl für eine markierte Antigenmischung als auch für komplexe klinische Proben innerhalb dieser Dissertation erzielt werden. Viele niedrig konzentrierte Proteine wie beispielsweise Zytokine, die normalerweise in einer piko-oder femtomolaren Konzentration im Blut vorliegen, wurden in dieser Arbeit mit sehr hohen Signal-zu-Hintergrund-Verhältnissen detektiert. Das hier beschriebene Verfahren öffnet zusätzliche Möglichkeiten für schnelle, kostengünstige und unbeschränkt erweiterungsfähige Mikrospot-Immunoassays.
Polyneuropathien (PNP) können zu einer Reorganisation der nodalen und paranodalen Membranproteine mit in der Folge fehlerhafter Axon-Schwann-Zell-Interaktionen führen. Im Rahmen der Basisdiagnostik einer Polyneuropathie haben sich Hautbiopsien als weniger invasive Ergänzung zur Suralisbiopsie mit einem geringen Nebenwirkungsrisiko entwickelt. Die Morphologie dermaler Nervenfasern lässt sich mittels Immunohistochemie in der Haut gezielt untersuchen. In der vorliegenden Studie wurde die Hypothese überprüft, ob pathologisch auffällige Ranvier-Schnürringe Hinweise auf Unterschiede bei PNP-Subgruppen und Schädigungsmuster liefern. Daneben wurden die Hypothesen überprüft, ob Entzündungszellen an myelinisierten Nervenfasern kolokalisiert nachweisbar sind und ob Hautbiopsien einen zusätzlichen Nutzen zur PNP-Basisdiagnostik liefern. Von 92 Patienten wurden Hautbiopsien von Finger, Ober-und Unterschenkel wurden entnommen, daraus gewonnene myelinisierte Nervenfasern der Haut wurden mittels immunohistochemischer Antikörper-Doppelfärbungen analysiert. Neuropathische Schädigungsformen vom axonalen und demyelinisierenden Typ zeigten keine signifikante Korrelation mit dem Auftreten von verlängerten Ranvier-Schnürringen und der Dispersion charakteristischer paranodaler und nodaler Membranproteine (Neurofascin, Caspr, Pan-Natrium-Kanäle). Kolokalisierte Entzündungszellen an myelinisierten Nervenfasern bei entzündlichen PNP ließen sich nicht nachweisen. PNP-Subgruppen zeigten keine signifikanten Unterschiede in Hinblick auf eine pathologische nodale oder paranodale Organisation. Der Zusatznutzen von Hautbiopsien in der PNP-Basisdiagnostik kann in Bezug auf die vorliegende Arbeit nur eingeschränkt bestätigt werden. Da Fingerbiopsien im Vergleich zu Proben aus Ober- und Unterschenkel eine signifikant höhere Dichte myelinisierter Nervenbündel pro Fläche Dermis aufweisen, wäre es durchaus denkbar, zukünftig primär Fingerbiopsien zu entnehmen um diese auf etwaige pathologische Veränderungen infolge neuropathischer Erkrankungen zu untersuchen. Anamnese, Basisdiagnostik und klinischer Befund erbringen nach wie vor den wichtigsten Beitrag zur PNP-Diagnostik.
Sharpening super-resolution by single molecule localization microscopy in front of a tuned mirror

The „Resolution Revolution" in fluorescence microscopy over the last decade has given rise to a variety of techniques that allow imaging beyond the diffraction limit with a resolution power down into the nanometer range. With this, the field of so-called super-resolution microscopy was born. It allows to visualize cellular architecture at a molecular level and thereby achieve a resolution level that had been previously only accessible by electron microscopy approaches.
One of these promising techniques is single molecule localization microscopy (SMLM) in its most varied forms such as direct stochastic optical reconstruction microscopy (dSTORM) which are based on the temporal separation of the emission of individual fluorophores. Localization analysis of the subsequently taken images of single emitters eventually allows to reconstruct an image containing super-resolution information down to typically 20 nm in a cellular setting. The key point here is the localization precision, which mainly depends on the image contrast generated the by the individual fluorophore’s emission. Thus, measures to enhance the signal intensity or reduce the signal background allow to increase the image resolution achieved by dSTORM. In my thesis, this is achieved by simply adding a reflective metal-dielectric nano-coating to the microscopy coverslip that serves as a tunable nano-mirror.
I have demonstrated that such metal-dielectric coatings provide higher photon yield at lower background and thus substantially improve SMLM performance by a significantly increased localization precision, and thus ultimately higher image resolution. The strength of this approach is that ─ except for the coated cover glass ─ no specialized setup is required. The biocompatible metal-dielectric nano-coatings are fabricated directly on microscopy coverslips and have a simple three-ply design permitting straightforward implementation into a conventional fluorescence microscope. The introduced improved lateral resolution with such mirror-enhanced STORM (meSTORM) not only allows to exceed Widefield and Total Internal Reflection Fluorescence (TIRF) dSTORM performance, but also offers the possibility to measure in a simplified setup as it does not require a special TIRF objective lens.
The resolution improvement achieved with meSTORM is both spectrally and spatially tunable and thus allows for dual-color approaches on the one hand, and selectively highlighting region above the cover glass on the other hand, as demonstrated here.
Beyond lateral resolution enhancement, the clear-cut profile of the highlighted region provides additional access to the axial dimension. As shown in my thesis, this allows for example to assess the three-dimensional architecture of the intracellular microtubule network by translating the local localization uncertainty to a relative axial position. Even beyond meSTORM, a wide range of membrane or surface imaging applications may benefit from the selective highlighting and fluorescence enhancing provided by the metal-dielectric nano-coatings. This includes for example, among others, live-cell Fluorescence Correlation Spectroscopy and Fluorescence Resonance Energy Transfer studies as recently demonstrated.
Das menschliche Genom verschlüsselt 30000 bis 40000 Proteine, von denen ein Großteil kovalent gebundene Karbohydrat-Gruppen an Asparagin-, Serin-, Threonin- oder Hydroxylysin-Resten trägt. Diese sogenannten Glykoproteine sind allgegenwärtige Bestandteile der extrazellulären Matrix von Zelloberflächen. Sie steuern Zell-Zell- und Zell-Matrix-Kommunikationen, können bei der roteinfaltung helfen bzw. die Proteinstabilität erhöhen oder Immunantworten regulieren. Die Auslösung von biologischen Prozesse erfordert aber Übersetzer der zuckerbasierten Informationen. Solche Effektoren sind die Lektine, unter ihnen auch die Galektine. Galektine binden spezifisch β-Galaktosen, weisen strukturelle Übereinstimmungen in der Aminosäuresequenz ihrer Zuckererkennungsdomänen (CRDs) auf und zeigen ein „jelly-roll“-Faltungsmuster, bestehend aus einem β-Sandwich mit zwei antiparallelen Faltblättern. Strukturell werden die CRDs in drei verschiedenen, topologischen Formen präsentiert. Proto-Typen existieren als nicht-kovalent verknüpfte Dimere der CRDs, Chimera-Typen besitzen neben der CRD eine Nicht-Lektin-Domäne und bei den Tandem-Repeat-Typen sind zwei verschiedene CRDs über ein kurzes Linker-Peptid kovalent verbunden. Galektine werden sowohl in normalem wie auch pathogenem Gewebe exprimiert und das zunehmende Wissen über die Beteiligung an verschiedenen Krankheiten und Tumorwachstum liefert die Motivation, strukturelle Aspekte und die Vernetzung von Lektinen detailliert, insbesondere im Hinblick auf ihre intrafamiliären Unterschiede, zu untersuchen. Durch die Kombination verschiedener Spektroskopie-Techniken mit hoher zeitlicher und räumlicher Auflösung, basierend auf der Verwendung von Fluorophoren (intrinsisch und extrinsisch), werden in dieser Arbeit die Eigenschaften von Galektinen näher untersucht. Mit Fluoreszenz-Korrelations-Spektroskopie (FCS) und Anisotropie-Messungen wird gezeigt, dass eine Liganden-Bindung bei Proto-Typ-Galektinen mit einer Verringerung des hydrodynamischen Radius einhergeht. Bei Tandem-Repeat- und Chimera-Typen bleibt der Radius konstant. Dafür skaliert die Diffusionskonstante von Tandem-Repeat-Typen anormal mit der molaren Masse. Die Anisotropie-Messungen werden parallel zu den FCS-Messungen durchgeführt, um einen Einfluss des Fluoreszenzmarkers auszuschließen. Mit Hilfe dieser Technik wird außerdem gezeigt, dass unterschiedliche Dissoziationskonstanten und Kinetiken für den Bindungsprozess innerhalb der Proto-Typ-Gruppe möglichweise auf unterschiedliche Konformationsdynamiken zurückgehen. Der Vergleich von hGal-1 und cG-1B verdeutlicht, dass strukturelle Ähnlichkeiten zwar ein identisches Bindungsverhalten hervorrufen können, der Oxidationsprozess der Proteine aber unterschiedlich ablaufen kann. Beide Methoden können so als sehr sensitive Techniken zur Untersuchung von Strukturmerkmalen bei Galektinen etabliert werden, wobei die Übertragbarkeit auf andere Glykoproteine gewährleistet ist. Weiterhin gilt Quervernetzung als eine der wichtigsten Eigenschaften von Galektinen, da durch die Vernetzung von Glykoproteinen auf der Zelloberfläche Signalwege aktiviert und Immunantworten reguliert werden. Um die räumliche organisation und Quervernetzung von hGal-1 auf den Oberflächen von Neuroblastomzellen nachzuweisen, eignet sich das hochauflösende Mikroskopieverfahren dSTORM sehr gut. Durch Verwendung des photoschaltbaren Fluorophors Alexa647 als spezifischem Marker für hGal-1, einem Standard-Weitfeld-Aufbau und verschiedenen Analyseverfahren, kann eine Clusterformation von hGal-1 auf der Zelloberfläche bestätigt werden. hGal-1 bildet Cluster mit einem mittleren Durchmesser von 81±7 nm aus. Der Durchmesser ist unabhängig von der Konzentration, während die Anzahl der Cluster davon abhängt. Für die Clusterausbildung ist ein Startpunkt, also eine minimale Dichte der Galektin-Moleküle, notwendig. Durch Blockierung der CRDs mit Laktose wird die Clusterbildung unterdrückt und die Spezifität der CRDs gegenüber β-Galaktosen erneut herausgestellt. Anders als dimeres hGal-1 binden Monomere deutlich schlechter an die Membranrezeptoren. Es werden keine Cluster ausgebildet, eine Quervernetzung von Membranrezeptoren ist nicht möglich. Außerdem kann es durch die Monomere zu einer vollständigen Markierung und damit Abkugellung der Zellen kommen. Möglicherweise wird der Zelltod induziert. Hochauflösende Mikroskopieverfahren sind durch den Markierungsprozess limitiert. Die bioorthogonale Click-Chemie eröffnet jedoch neue Möglichkeiten zur Markierung und Visualisierung von Biomolekülen, ohne die Notwenigkeit genetischer Manipulationen. Es werden modifizierte Zuckermoleküle in die Zellmembranen eingebaut, über eine 1,3-polare Cycloaddition mit einem Alkin markiert und ihre Verteilung mit Hilfe von dSTORM untersucht. Es wird nachgewiesen, dass die Zuckermoleküle in Clustern auftreten und Click-Chemie trotz dem Katalysator Kupfer an lebenden Zellen durchführbar ist. Die Bewegung der Gesamtcluster wird mittels Mean Square Displacement aufgeschlüsselt und eine Diffusionskonstante für Cluster im Bereich von 40 - 250 nm bestimmt. Zusammenfassend stellt die Kombination verschiedener Spektroskopie-Techniken ein gutes Werkzeug zur Untersuchung von Karbohydrat-bindendenden Proteinen mit hoher räumlicher und zeitlicher Auflösung dar und ermöglicht einen neuen Einblick in die Biologie der Galektine.
Efficient quadrupolar chromophores (A–pi–A) with triarylborane moieties as acceptors have been studied by the Marder group regarding their non‐linear optical properties and two‐photon absorption ability for many years. Within the present work, this class of dyes found applications in live‐cell imaging. Therefore, the dyes need to be water‐soluble and water‐stable in diluted aqueous solutions, which was examined in Chapter 2. Furthermore, the influence of the pi‐bridge on absorption and emission maxima, fluorescence quantum yields and especially the two-photon absorption properties of the chromophores was investigated in Chapter 3. In Chapter 4, a different strategy for the design of efficient two‐photon excited fluorescence imaging dyes was explored using dipoles (D–A) and octupoles (DA3). Finding the optimum balance between water‐stability and pi‐conjugation and, therefore, red‐shifted absorption and emission and high fluorescence quantum yields, was investigated in Chapter 5
Es wurde ein Leitpartikeltyp mit hoher Fluoreszenz sowie einem Absorptionsbereich oberhalb von 600 nm evaluiert. Zur Anbindung der hochspezifisch wirkenden Antikörper wurde die Teilchenoberfläche mit Carboxylgruppen funktionalisiert. Die Darstellung dieser sphärischen, komplex aufgebauten erfolgte über eine nasschemische Synthese. Die synthetisierten Partikel besitzen eine hohe Fluoreszenzintensität, gutes Chromatographierverhalten und spezifische Beladbarkeit mit monoklonalen Antikörpern (z.B. Troponin T) auf einer mit Carboxylgruppen modifizierten Partikeloberfläche. Auf die Partikel mit dem favorisierten Fluorophor musste eine zusätzliche Silicathülle aufkondensiert werden, damit diese im Anschluss erfolgreich mit Antikörpern beladen werden konnte. Die erhaltenen partikulären Systeme wurden sowohl qualitativ als auch quantitativ charakterisiert. Die Fluoreszenzintensität dieser dotierten Kern-Schale-Partikel konnte soweit optimiert werden, dass sich klinisch relevante und noch höhere Sensitivitäten in Prüfteststreifen detektieren ließen. Weiterhin wurden neuartige Fluoralkylsilan und Fluorophor codotierte Silicat-Nanopartikel synthetisiert, die auf Anhieb eine gute untere Nachweisgrenze von Troponin erzielten. Durch UV-VIS- und Fluoreszenz-Untersuchungen sowie Konjugations- und Prüfteststreifen-Versuche konnte gezeigt werden, dass die Cokondensation des Fluoralkylsilans in einer Erhöhung von Absorption und Fluoreszenz der Partikel resultiert. Weitere Untersuchungen von zeigten, dass eine zusätzliche Oberflächenmodifizierung mit Fluoralkylsilan zu einer signifikanten Verschlechterung der Konjugationseigenschaften mit Antikörpern führt. Alternative Detekorreagenzien und -methoden wurden ebenfalls untersucht. So konnte der kationische Komplex Tris-(1,10-phenantrolin)ruthenium(II)-dichlorid erfolgreich in monodisperse Silicat-Partikel eingebaut werden. Aufgrund ihrer geringen Sauerstoffpermeabilität sind sie als impermeabler Referenzstandard in O2-Sensoren geeignet. Eine andere untersuchte Detektionsmethode basiert auf zeitaufgelöster Fluoreszenz (TRF). Hierbei werden hauptsächlich Lanthanoid-Komplexe eingesetzt. Am besten untersucht sind Europium-Komplexe, welche meistens Diketone als Liganden besitzen. Bislang konnten diese neutralen Komplexe jedoch nicht in polare Silicatpartikel-Matrizes eingebaut werden. Durch Einsatz von 3,3,3-Trifluoropropyltrimethoxysilan gelang es erstmalig, einen Europium(III)-tris-4,4,4-trifluoro-1-(2-naphthoyl)-1,3-butandion-Komplex (Eu(TNB)3) in hydrophobierte Silicat-Nanopartikeln physikalisch einzubauen. TRF-Messungen zeigten Abklingzeiten von ca. 300 µs. In diesem bislang nicht verfügbaren Partikel-Typ konnten positive Eigenschaften von Latex- und Silicatpartikeln kombiniert werden. Auch einige Porphyrinkomplexe mit langen Fluoreszenzlebensdauern sind in Silicat-Nanopartikel eingebaut worden. Der neutrale Komplex 5,10,15,20-Tetrakis(4-carboxyphenyl)-porphyrin-Pd(II) konnte nur durch vorhergehende Silanisierung erfolgreich eingebunden werden. Die erhaltenen sphärischen Partikel weisen eine Größenverteilung von 200-300 nm auf. Ein weiteres, kationisches Porphyrin (5,10,15,20-Tetrakis(N-methyl-4-pyridyl)-21,23H-porphyrin-Zn(II)) konnte ebenfalls erfolgreich in etwa 140 nm große Silicat-Nanopartikel blutungsstabil eingebaut werden.
Koordinationspolymere auf der Basis von Terpyridin und Dipyridyltriazin: Synthese und Anwendung

Der erste Teil der Arbeit untersucht den Einsatzes von 4,6-Di-(pyrid-2´-yl)-1,3,5-triazin als Baustein für Metallo-supramolekulare Polyelektrolyte. Die dafür nötigen ditopen Liganden werden mittels Stille Kreuzkupplungen dargestellt. Die Absorptions- und Fluoreszenzeigenschaften können durch den Einbau von Oligothiophenen eingestellt werden.
Im zweiten Teil der Arbeit werden die elektrorheologischen Eigenschaften von Metallo-supramolekularen Polyelektrolyten untersucht. Zu diesem Zweck werden die Koordinationspolymere in das Schichtsilikat Montmorillonit interkaliert. Die Interkalation wird mittels verschiedener analytischer Methoden wie Pulverdiffraktometrie, Thermoanalyse oder Infrarotspektroskopie untersucht. Die entstehenden Nanokomposite zeigen einen elektrorheologischen Effekt bei einer geringen Stromdichte.
Zwei Arten helikal-chiraler Verbindungen mit einem oder zwei Boratomen wurden nach einem modularen Ansatz synthetisiert. Die Bildung der helikalen Strukturen erfolgte durch Einführung von Bor in flexible Biaryl- bzw. Triaryl-Vorstufen, hergestellt aus kleinen achiralen Bausteinen. Die durchgehend ortho-fusionierten Azabora[7]helicene zeichnen sich dabei durch außergewöhnliche Konfigurationsstabilität, blaue oder grüne Fluoreszenz in Lösung mit Quantenausbeuten (Φ\(_{fl}\)) von 18–24 %, grüne oder gelbe Emission im Festkörper (Φ\(_{fl}\) bis zu 23 %) und starke chiroptische Resonanz mit großen Anisotropiefaktoren von bis zu 1.12×10\(^{-2}\) aus. Azabora[9]helicene, aufgebaut aus winkelförmig sowie linear angeordneten Ringen, sind blaue Emitter mit Φ\(_{fl}\) von bis zu 47 % in CH\(_{2}\)Cl\(_{2}\) und 25 % im Festkörper. DFT-Rechnungen zeigen, dass ihre P-M-Interkonversion über einen komplexeren Weg verläuft als im Fall von H1. Röntgenstrukturanalyse von Einkristallen zeigt deutliche Unterschiede in der Packungsanordnung von Methyl- und Phenylderivaten auf. Die Moleküle werden als Primärstrukturen verlängerter Helices vorgeschlagen.
The scope of this work is to develop a novel single-molecule imaging technique by combining atomic force microscopy (AFM) and optical fluorescence microscopy. The technique is used for characterizing the structural properties of multi-protein complexes. The high-resolution fluorescence microscopy and AFM are combined (FIONA-AFM) to allow for the identification of individual proteins in such complexes. This is achieved by labeling single proteins with fluorescent dyes and determining the positions of these fluorophores with high precision in an optical image. The same area of the sample is subsequently scanned by AFM. Finally, the two images are aligned and the positions of the fluorophores are displayed on top of the topographical data. Using quantum dots as fiducial markers in addition to fluorescently labeled proteins, fluorescence and AFM information can be aligned with an accuracy better than 10 nm, which is sufficient to identify single fluorescently labeled proteins in most multi-protein complexes. The limitations of localization precision and accuracy in fluorescence and AFM images are investigated, including their effects on the overall registration accuracy of FIONA-AFM hybrid images. This combination of the two complementary techniques opens a wide spectrum of possible applications to the study of protein interactions, because AFM can yield high resolution (5–10 nm) information about the conformational properties of multi-protein complexes while the fluorescence can indicate spatial relationships of the proteins within the complexes. Additionally, computer simulations are performed in order to validate the accuracy of the registration algorithm.
In this thesis the syntheses and detailed investigations on two foldable PBI systems were presented. The reversible, solvent-dependet folding/unfolding-behavior was used to study the ground and excited states properties of folda-dimer and folda-trimer by means of different spectroscopic methods as well as theoretical studies. The switching between charge transfer or excimer formation pathways of photoexcited molecules influenced by the spatial arrangement of chromophores within defined dye systems illustrates the impact of conformational preferences on functional properties.
We introduce fluorescence-detected pump–probe microscopy by combining a wavelength-tunable ultrafast laser with a confocal scanning fluorescence microscope, enabling access to the femtosecond time scale on the micrometer spatial scale. In addition, we obtain spectral information from Fourier transformation over excitation pulse-pair time delays. We demonstrate this new approach on a model system of a terrylene bisimide (TBI) dye embedded in a PMMA matrix and acquire the linear excitation spectrum as well as time-dependent pump–probe spectra simultaneously. We then push the technique towards single TBI molecules and analyze the statistical distribution of their excitation spectra. Furthermore, we demonstrate the ultrafast transient evolution of several individual molecules, highlighting their different behavior in contrast to the ensemble due to their individual local environment. By correlating the linear and nonlinear spectra, we assess the effect of the molecular environment on the excited-state energy.
Ziel der Dissertation „Oktapeptide als neue Organokatalysatoren zur Hydrolyse von Phosphaten und Estern“ war es neue Oktapeptide zu finden, die die Fähigkeit besitzen Phosphate und Ester zu Hydrolysieren. In der Natur ist bei den Katalysereaktionen die Sekundärstruktur von entscheidender Bedeutung. Aus diesem Grunde wurde im Rahmen dieser Arbeit zunächst ein Modellsystem entwickelt, mit dem gezeigt werden konnte, dass es möglich ist mit einfachen Bausteinen wie Diaminobutan und dem in der Gruppe von Schmuck entwickelten Guanidiniocarbonylpyrrol ein Molekül zu entwickeln, welches eine stabile intramolekulare Schleife in polaren Lösungsmitteln ausbildet. Aufgrund der geringen Größe des Moleküls, konnte kein β-Faltblatt gebildet werden. Dennoch konnte mit Hilfe der NMR-Spektroskopie gezeigt werden, dass in dem polaren Lösungsmittel Methanol eine stabile Schleifenbildung mit einer intramolekularen Komplexierung stattfindet. Nach erfolgreicher Synthese des oben beschriebenen Testsystems wurde als nächster Schritt eine kombinatorische Organokatalysatorbibliothek mit 625 Mitgliedern aufgebaut. Die Struktur der Peptide kann man in drei Teile untergliedern. Der erste Teil ist die feste Phase, das Amino-TentaGel, an das nacheinander die einzelnen Aminosäuren gekuppelt wurden. An Stelle der Butylgruppe als Schleifenelement wurde Aib-D-Pro als β-Turn Element eingesetzt. Den dritten Teil bilden die bei der Synthese der Oktapeptide eingesetzten Aminosäuren AA1, AA3, AA6, AA8, die ein β-Faltblatt ausbilden sollten. Die kombinatorische Synthese der Bibliothek erfolgte nach der „Split and Mix“ Methode. Zum Unterscheiden der einzelnen Mitglieder untereinander, wurde die feste Phase zusammen mit einem Radiofrequenzchip in IRORI MikroKans gegeben. Durch die unterschiedlichen Aminosäuren ist die Bibliothek für die Katalyse in polaren Lösungsmitteln wie Wasser konzipiert worden. Als exemplarische Katalysereaktionen wurden dabei zwei Hydrolysereaktionen (Phosphatspaltung und Esterspaltung) ausgesucht. Zunächst wurde für beide Reaktionen ein Screening mit unterschiedlichen Bedingungen durchgeführt. Dabei hat sich gezeigt, dass bei der Phosphatspaltung nur dann die Reaktion katalysiert wurde, wenn das künstliche Argininanalogon, welches in unserer Arbeitsgruppe synthetisiert wurde, vorhanden war. Der beste Katalysator hat die Reaktion 175-mal schneller katalysiert als die unkatalysierte Reaktion. Bei dem Screening der Esterspaltung hat sich herausgestellt, dass die Aminosäure Histidin essentiell für die katalytische Aktivität ist. Der beste Katalysator bei der Esterspaltung hat die Hydrolyse des Esters 345-mal schneller katalysiert als die unkatalysierte Reaktion. Bei beiden Reaktionen hat sich gezeigt, dass die Sequenz der Katalysatoren sehr wichtig für die Katalyse ist. So verringert z.B. bei der Esterhydrolyse der Austausch zweier Aminosäuren eine Verringerung der Aktivität von dem Faktor 294 auf den Beschleunigungsfaktor 35. Auch konnten beide Katalysereaktionen in wässrigem gepufferten Lösung durchgeführt werden. Damit ist es möglich gewesen neue Oktapeptide für die Katalyse von Phosphat- und Esterspaltung zu finden und diese erfolgreich im Screening als auch in Lösung zu untersuchen.
Complexation properties of 2,2':6',2''-terpyridine (tpy) have been studied with a series of first row transition metal ions by UV-vis, 1H NMR and isothermal titration calorimetry and ƒ´H values for the tpy complexation processes have been determined. These studies reveal that Zn2+ is the best suited metal ion for the reversible coordination of the terpyridine ligand. Thus, supramolecular coordination polymerization of perylene bisimide fluorophores containing terpyridine functionalities have been investigated by using Zn2+ as metal ion. The formation of the dimeric complexes in the case of monotopic model comounds and coordination polymerization of ditopic functional building blocks have been confirmed by 1H NMR studies. The optical properties of dimeric and polymeric complexes have been investigated by UV-vis and fluorescence spectroscopy. The Zn2+ coordination to the terpyridine unit does not effect the advantageous fluorescence properties of perylene bisimide moieties. The reversibility of the formation of coordination polymers has been established by 1H NMR and additionally by DOSY NMR and fluorescence anisotropy measurements. Coordination polymer strands can be visualized by atomic force microscopy (AFM), which also reveals the formation of an ordered monolayer film at higher concentration. The average polymer length has been determined by AFM to 15 repeat units, which correlates well with the value estimated by 1H NMR to >10 repeat units.
Einführung: Die physikalischen Eigenschaften des intravenösen Anästhetikums Propofol (2,6-Diisopropylphenol) erlauben dessen fluoreszenzspektrometrische Detektion. Zur Entwicklung eines direkten Online-Monitorings im optisch dichten Medium Blut wird das Signalverhalten von Propofol im mit Blutprodukten gefüllten Kreislaufsystem untersucht. Material und Methoden: Der kontinuierliche Umsatz von 140,2 ml Probenvolumen im Kreislaufmodell mit integrierter Durchflussquarzküvette bezweckt ein stabiles Fluoreszenzniveau des Messmediums, da Blutprodukte in statischer Versuchsanordnung unter der verwendeten Anregungsstrahlung (UV-C) starken photochemischen Bleichungseffekten ausgesetzt sind. Als Messmedien untersucht werden Gefrorenes Frischplasma (GFP), eine Suspension aus Erythrozytenkonzentrat und GFP (EK + GFP) sowie am Versuchstag gespendetes heparinisiertes Vollblut. Es erfolgt die standardisierte Injektionen von vier Propofolboli, durch die im System Konzentrationen von 35,7 μg/ml bis 3,6 μg/ml entstehen und den klinisch relevanten Wirkspiegeln bei Narkoseeinleitung sowie Narkoseaufrechterhaltung entsprechen. Unter Anregung mit Licht der Wellenlänge 274 nm liefert Propofol ein maximales Signal bei 300 nm. Anhand der in engen zeitlichen Abständen aufgenommen Fluoreszenzspektren werden die Propofoleffekte bei 300 nm im Summationsspektrum des Blut-Propofol-Gemischs analysiert. Ergebnisse: Die Signalanstiege bei 300 nm nach Injektion in das mit GFP bzw. EK + GFP gefüllte Kreislaufsystem sind hochsignifikant für die erzeugten Propofolspiegel von 35,7 μg/ml bis 3,6 μg/ml und weisen eine sehr gute lineare Korrelation von R2 = 0,73 bis zu R2 = 0,99 zwischen Fluoreszenzsignal und Propofolkonzentration auf. Allein für diese Messmedien kann durch den Einsatz des Kreislaufmodells ein ausreichend stabiles Fluoreszenzsignal zum Propofolnachweis erreicht werden. Dem Fluoreszenzanstieg nach Propofolinjektion folgt in allen Messmedien ein über 30 Minuten andauernder Signalabfall, für den nach fluoreszenzspektrometrischer Untersuchung von Schlauchproben des Kreislaufmodells die Adsorption des lipophilen Anästhetikums an Silikon als ein ursächlicher Faktor bestimmt werden kann. Schlussfolgerung: Der direkte konzentrationsabhängige Fluoreszenznachweis von klinisch eingesetzten Propofol-Wirkspiegeln gelingt allein in transfusionsmedizinisch aufbreiteten Blutprodukten.
This thesis will outline studies performed on the fluorescence dynamics of phenyl-benzo-
[c]-tetrazolo-cinnolium chloride (PTC) in alcoholic solutions with varying viscosity using
time-resolved fluoro-spectroscopic methods. Furthermore, the properties of femtosecond
Laguerre-Gaussian (LG) laser pulses will be investigated with respect to their temporal
and spatial features and an approach will be developed to measure and control the spatial
intensity distribution on the time scale of the pulse.
Tetrazolium salts are widely used in biological assays for their low oxidation and reduction
thresholds and spectroscopic properties. However, a neglected feature in these applications
is the advantage that detection of emitted light has over the determination of the
absorbance. To corroborate this, PTC as one of the few known fluorescent tetrazolium
salts was investigated with regard to its luminescent features. Steady-state spectroscopy
revealed how PTC can be formed by a photoreaction from 2,3,5-triphenyl-tetrazolium
chloride (TTC) and how the fluorescence quantum yield behaved in alcoholic solvents
with different viscosity. In the same array of solvents time correlated single photon counting
(TCSPC) measurements were performed and the fluorescence decay was investigated.
Global analysis of the results revealed different dynamics in the different solvents, but
although the main emission constant did change with the solvent, taking the fluorescence
quantum yield into consideration resulted in an independence of the radiative rate from
the solvent. The non-radiative rate, however, was highly solvent dependent and responsible
for the observed solvent-related changes in the fluorescence dynamics. Further studies
with the increased time resolution of femtosecond fluorescence upconversion revealed an
independence of the main emission constant from the excitation energy, however the dynamics
of the cooling processes prior to emission were prolonged for higher excitation
energy. This led to a conceivable photoreaction scheme with one emissive state with a
competing non-radiative relaxation channel, that may involve an intermediate state.
LG laser beams and their properties have seen a lot of scientific attention over the past two
decades. Also in the context of new techniques pushing the limit of technology further to
explore new phenomena, it is essential to understand the features of this beam class and
check the consistency of the findings with theoretical knowledge. The mode conversion
of a Hermite-Gaussian (HG) mode into a LG mode with the help of a spiral phase plate
(SPP) was investigated with respect to its space-time characteristics. It was found that
femtosecond LG and HG pulses of a given temporal duration share the same spectrum
and can be characterized using the same well-established methods. The mode conversion
proved to only produce the desired LG mode with its characteristic orbital angular momentum
(OAM), that is conserved after frequency doubling the pulse. Furthermore, it
was demonstrated that temporal shaping of the HG pulse does not alter the result of its
mode-conversion, as three completely different temporal pulse shapes produced the same
LG mode. Further attention was given to the sum frequency generation of fs LG beams
and dynamics of the interference of a HG and a LG pulse. It was found that if both are
chirped with inverse signs the spatial intensity distribution does rotate around the beam
axis on the time scale of the pulse. A strategy was found that would enable a measurement
of these dynamics by upconversion of the interference with a third gate pulse. The results
of which are discussed theoretically and an approach of an experimental realization had
been made. The simulated findings had only been reproduced to a limited extend due to
experimental limitations, especially the interferometric stability of the setup.
The thesis enhances the strategy of non-destructive fluorescence read-out in rylene bisimide-diarylethene containing photochromic systems. The fluorescence of the emitter unit is quenched by a photoinduced electron transfer only to one of the isomeric forms of the photochrome. The driving force of the fuorescence-quenching electron transfer was calculated by the help of the Rehm-Weller equation. The novel photochromic systems satisfy the necessary requirements for non-destructive read-out in write/read/erase fluorescent memory devices.
