The goal of the project was to establish knock down of mRNA in human mesenchymal stem cells. Since these cells are difficult to transfect, a viral approach is needed to achieve sufficient expression of e. g. shRNA in a high percentage of cells to allow for an efficient silencing of corresponding mRNAs. For this purpose for every gene product of interest, a number of shRNA clones have to be tested to detect an individual shRNA with sufficient efficacy. Lentiviral systems for shRNA approaches have recently become available. The principal advantage of the lentiviral system is that it allows gene silencing in nondividing cells and therefore expands the usefulness of the RNAi-based gene silencing system. Lentivirus-delivered shRNAs are capable of specific, highly stable and functional silencing of gene expression in a variety of cell types. Since the viral transfection of MSCs is a time consuming process that involves transfection of 293 FT cells plus transduction of target cells, for this thesis the following approach was chosen: genes of interest were checked for expression in 293FT cells by RT-PCR. These gene products can be silenced in 293FT cells simply by transfection of shRNA clones and efficacy was subsequently tested by RT-PCR. Beyond this thesis then the project can proceed with effective clones to transduce primary MSCs with individual shRNA clones identified as effective silencing tool in this thesis.
Type 1 diabetes affects around 0.5% of the population in developed countries and the incidence rates have been rising over the years. The destruction of beta cells is irreversible and the current therapy available to patients only manages the symptoms and does not prevent the associated pathological manifestations. The patients need lifelong therapy and intensive research is being carried out to identify ways to eliminate autoimmune responses directed against pancreatic beta cells and to replace or regenerate beta cells. The work presented herein aimed at analyzing the role of the Th17 T cell subset, characterized by secretion of the pro- inflammatory cytokine IL-17A, in autoimmune diabetes and also at generating a beta cell reporter mouse line in the NOD background, the most widely- used mouse model for type 1 diabetes. We generated IL- 17A knockdown (KD) NOD mice, using RNAi in combination with lentiviral transgenesis. We analyzed diabetes frequency in IL-17A deficient mice and found that the loss of IL-17A did not protect the transgenic mice from diabetes. Based on these observations, we believe that Th17 cells do not play a critical role in type 1 diabetes through the IL-17A pathway, though they might still be involved in the disease process through alternate pathways. We also generated NOD and NOD-SCID mice with a transgene that drives the beta cell specific expression of a luciferase reporter gene. We used a lentiviral construct, which combined a luciferase sequence and a short- hairpin RNA (shRNA) expression cassette, allowing gene- knockdown under the beta cell specific rat insulin promoter (RIP). These mice will be of use in studying beta cell phenotypes resulting from the knockdown of target genes, using non- invasive bioimaging. We believe that the generation of these reporter mouse lines for diabetes studies will prove valuable in future investigations. Furthermore, the demonstration that the loss of IL-17A does not alter susceptibility to type 1 diabetes should help clarify the controversial involvement of Th17 cells in this disease.
ZUSAMMENFASSUNG
Obwohl diverse Mutationen des NF-κB Systems in Myelomzelllinien und primären Myelomzellen eine pathogenetische Beteiligung andeuten, ist die Relevanz des IKK Komplexes als molekularer Angriffspunkt für die Entwicklung medikamentöser Therapieoptionen noch nicht ausreichend geklärt. Zwar führte die Applikation des IKK-β Inhibitors MLN120b dosisabhängig und längerfristig zu einer Reduktion der Zellviabilität in einer Vielzahl von Myelomzelllinien, doch kann nicht ausgeschlossen werden, dass bei höheren Konzentrationen unspezifische Wirkungen für die beobachteten Effekte (mit-) verantwortlich sind.
Aus diesem Grund erfolgte in der vorliegenden Arbeit eine spezifische Suppression von
IKK-α, IKK-β oder IKK-γ mittels transienter Transfektion von shRNA Expressionsvektoren oder Stealth-siRNA. Es folgte die Charakterisierung der verminderten Zielproteinspiegel mittels Western-Blot und die Messung der Viabilität der Zellen mittels FACS Analysen. Darüber hinaus wurde in TNF-α Stimulationsexperimenten der Effekt der Suppression von IKK-β mittels Stealth-siRNA auf (Phospho-)IκB-α analysiert. Schließlich erfolgte die Applikation des IKK-β Inhibitors
TPCA, dessen Wirkung auf die Zellviabilität und auf die TNF-α-vermittelte Phosphorylierung und Degradation von IκB-α in MM.1S Zellen untersucht wurde.
Die Experimente mit Stealth-siRNA zeigten, dass weder die Suppression von IKK-β, noch die Suppression von IKK-α oder IKK-γ in AMO-1, L363 oder MM.1S eine Verminderung der Zellviabilität bewirken konnte. Auch eine kombinierte Suppression von IKK-α zusammen mit IKK-β in L363 und MM.1S Zellen bewirkte keinen vermehrten Zelltod. Dagegen zeigte die Behandlung von MM.1S Zellen mit hohen Konzentrationen von TPCA einen geringen Effekt auf das Überleben dieser Zellen. Die Suppression von IKK-β mittels Stealth-siRNA in MM.1S konnte nicht die TNF-α vermittelte IκB-α
Phosphorylierung und Degradation verhindern. Sowohl die hohe TNF-α Konzentration von 100ng/ml, als auch eine unvollständige Suppression von IKK-β könnte dazu beigetragen haben. In analogen Experimenten mit TPCA konnte die TNF-α vermittelte IκB-α Phosphorylierung und Degradation dagegen effektiv unterdrückt werden.
In der Zusammenschau der Ergebnisse konnte somit eine potenzielle therapeutische Relevanz des IKK-Komplexes als molekularer Angriffspunkt für eine Myelomtherapie nicht gefunden werden.
Eine noch detailliertere Analyse der Funktionalität des Signalwegs (insbesondere eine Messung der Aktivität der NF-κB Transkriptionsfaktoren im Zellkern) und die Etablierung stabiler und induzierbarer Expressionssysteme für längerfristige Untersuchungen der RNAi Wirkungen in Myelomzellen, stellen weiterführende Wege zu einer umfangreicheren Beurteilung der pathobiologischen und therapeutischen Bedeutung des NF-κB Systems dar. Darüber hinaus sind die das NF-κB System betreffenden Mutationen genauer hinsichtlich ihrer potenziellen Wirkung auf NF-κB unabhängige Signalwege zu untersuchen.
Identification of human host cell factors involved in \(Staphylococcus\) \(aureus\) 6850 infection

Staphylococcus aureus is both a human commensal and a pathogen. 20%-30% of all individuals are permanently or occasionally carriers of S. aureus without any symptoms. In contrast to this, S. aureus can cause life-threatening diseases e.g. endocarditis, osteomyelitis or sepsis. Here, the increase in antibiotic resistances makes it more and more difficult to treat these infections and hence the number of fatalities rises constantly. Since the pharmaceutical industry has no fundamentally new antibiotics in their pipeline, it is essential to better understand the interplay between S. aureus and the human host cell in order to find new, innovative treatment options.
In this study, a RNA interference based whole genome pool screen was performed to identify human proteins, which play a role during S. aureus infections. Since 1,600 invasion and 2,271 cell death linked factors were enriched at least 2 fold, the big challenge was to filter out the important ones. Here, a STRING pathway analysis proved to be the best option. Subsequently, the identified hits were validated with the help of inhibitors and a second, individualised small interfering RNA-based screen.
In the course of this work two important steps were identified, that are critical for host cell death: the first is bacterial invasion, the second phagosomal escape. The second step is obligatory for intracellular bacterial replication and subsequent host cell death. Invasion in turn is determining for all following events. Accordingly, the effect of the identified factors towards these two crucial steps was determined. Under screening conditions, escape was indirectly measured via intracellular replication. Three inhibitors (JNKII, Methyl-beta-cyclodeytrin, 9-Phenantrol) could be identified for the invasion process. In addition, siRNAs targeted against 16 different genes (including CAPN2, CAPN4 and PIK3CG), could significantly reduce bacterial invasion. Seven siRNAs (FPR2, CAPN4, JUN, LYN, HRAS, AKT1, ITGAM) were able to inhibit intracellular replication significantly. Further studies showed that the IP3 receptor inhibitor 2-APB, the calpain inhibitor calpeptin and the proteasome inhibitor MG-132 are able to prevent phagosomal escape and as a consequence intracellular replication and host cell death.
In this context the role of calpains, calcium, the proteasome and the mitochondrial membrane potential was further investigated in cell culture. Here, an antagonistic behaviour of calpain 1 and 2 during bacterial invasion was observed. Intracellular calcium signalling plays a major role, since its inhibition protects host cells from death. Beside this, the loss of mitochondrial membrane potential is characteristic for S. aureus infection but not responsible for host cell death. The reduction of membrane potential can be significantly diminished by the inhibition of the mitochondrial Na+/Ca2+ exchanger.
All together, this work shows that human host cells massively contribute to different steps in S. aureus infection rather than being simply killed by bacterial pore-forming toxins. Various individual host cell factors were identified, which contribute either to invasion or to phagosomal escape and therefore to S. aureus induced cytotoxicity. Finally, several inhibitors of S. aureus infection were identified. One of them, 2-APB, was already tested in a sepsis mouse model and reduced bacterial load of kidneys.
Thus, this study shows valuable evidence for novel treatment options against S. aureus infections, based on the manipulation of host cell signalling cascades.
Die subakut sklerosierende Panenzephalitis ist eine demyelinisierende Erkrankung des ZNS. Ätiologisch liegt eine persistierende Infektion von Masernviren der Neuronen bzw. Gliazellen zugrunde. Bis heute gibt es keine spezifische Therapie und lediglich symptomatische Therapiemaßnahmen werden in deren Behandlung angewandt. Obwohl sich während des Krankheitsverlaufes eine ausgeprägte Immunantwort des natürlichen als auch adaptiven Immunsystems des Wirtes abspielt führt die Erkrankung unweigerlich zum Tode der Betroffenen. Eine Therapiemöglichkeit diesbezüglich könnten die Effekte der RNAi darstellen. Um geeignete Sequenzen für einen möglichen Genknockdown einzelner MV-Gene herauszufiltern, wurde ein plasmid-basierendes Testsystem entwickelt, das die mRNA des Fluoreszenzproteins DsRed2 zusammen mit mRNA-Sequenzen einzelner MV-Gene exprimiert. Gegen diese MV-Gene wiederum wurden verschiedene shRNAs ausgewählt, welche mithilfe eines lentiviralen Vektorsystems exprimiert werden. Als Expressionsindikator findet in diesem lentiviralen Vektorsystem simultan die Expression von eGFP als statt. Die Auswertung dieses Dual-Color-Systems erfolgte im Folgenden mittels FACS-Analysen. Die wirksamsten siRNA-Sequenzen, ausgewählt durch eine hohe Abnahme der Rotfluoreszenz in den untersuchten Zellen, wurden für weitere Versuche selektiert. Die auf diese Weise ausgewählten shRNA-Sequenzen wurden in einem weiteren Schritt durch die Generierung lentiviraler Partikel in persistierend infizierten NT2-Zellen (piNT2), die das Fluoreszenzprotein HcRed als Infektionsindikator exprimieren, transduziert. Weitere durchflusszytometrische Messungen zeigten eine äußerst hohe Reduktion viraler Replikation innerhalb von 7 Tagen bis unterhalb der Detektionsgrenze sofern die shRNAs gegen die Expression der MV-Proteine N, P und L gerichtet waren. Die Anzahl der virus-negativen Zellen blieb im Folgenden bis zu 3 Wochen nach stattgehabter Transduktion konstant, sofern das fusion-inhibiting-peptide den Zellkulturen hinzugegeben wurde. Die transduzierten piNT2-Zellen, welche keine Expression von HcRed zeigten wurden auf Zellrasen bestehend aus Vero-Zellen aufgetragen und führten im zeitlichen Verlauf zu keiner Fusion bzw. Synzytienbildung. Dieser Zusammenhang legt nahe, dass die transduzierten piNT2-Zellen die Fähigkeit einer Expression viraler Proteine verloren haben und eine „Heilung“ stattfinden kann, wenn die virale Proteinbiosynthese durch eine permanente Expression von siRNAs, wie beispielsweise durch lentivirale Vektorsysteme, gehemmt wird.
Protein-protein interaction (PPI) studies are gaining momentum these days due to the plethora of various high-throughput experimental methods available for detecting PPIs. Proteins create complexes and networks by functioning in harmony with other proteins and here in silico network biology hold the promise to reveal new functionality of genes as it is very difficult and laborious to carry out experimental high-throughput genetic screens in living organisms. We demonstrate this approach by computationally screening C. elegans conserved homologs of already reported human tumor suppressor and aging associated genes. We select by this nhr-6, vab-3 and gst-23 as predicted longevity genes for RNAi screen. The RNAi results demonstrated the pro-longevity effect of these genes. Nuclear hormone receptor nhr-6 RNAi inhibition resulted in a C. elegans phenotype of 23.46% lifespan reduction. Moreover, we show that nhr-6 regulates oxidative stress resistance in worms and does not affect the feeding behavior of worms. These findings imply the potential of nhr-6 as a common therapeutic target for aging and cancer ailments, stressing the power of in silico PPI network analysis coupled with RNAi screens to describe gene function.
