SAP47 ist ein Synapsenassoziiertes Protein von 47 kDa aus Drosophila melanogaster, das zu einer neuen Proteinfamilie gehört. Um eine Sap47 Mutante zu erzeugen wurden drei Methoden eingesetzt: Gezielte Mutagenese durch homologe Rekombination, RNA interference (RNAi) und Transposon Remobilisierung. Um einen Interaktionspartner für das SAP47 Protein zu identifizieren wurden ein Yeast-Two-Hybrid System und das "CytoTrap" Verfahren eingesetzt.
The steroid hormones corticosterone/cortisol and aldosterone are synthesized and secreted by the adrenal gland in response to stress or an altered salt-water balance. This is controlled by a negative feedback mechanism referred to as the HPA axis and the RAAS. Actions of these steroid hormones are mediated by the glucocorticoid receptor (GR) and the mineralocorticoid receptor (MR), which reside in the cytoplasm in a complex with heat-shock proteins. Both, the GR and the MR belong to the nuclear receptor superfamily and share a common protein structure consisting of three separate domains. However, they have different affinities for various ligands, their actions depend on hormone concentration, they are modulated by pre-receptor mechanisms such as the 11β-HSD2 and they are differently distributed in several tissues. Aldosterone acts via the MR in epithelial and in non-epithelial cells and regulates sodium-water homeostasis, cardiovascular function, neuronal excitability and adipocyte differentiation. So far the analysis of gene inactivation in vivo was limited to mice, but disease models in rats sometimes more closely reflect the situation encountered in humans. Since embryonic stem cells and thus gene targeting in rats is not available, we generated MR knock-down transgenic rats by lentiviral delivery of a shRNA. The F1 progeny of the founder rats showed a wide range of reduced MR mRNA and protein levels in kidney and hippocampus, the two major sites of MR expression. In contrast, expression of the highly homologous GR was unaltered, indicating specificity of gene inactivation. The two MR target genes, Sgk1 and ENaC, were up-regulated while the mRNA levels of other genes such as IK1 and SCD2 was reduced. Similar to the knock-out mice and human patients, the knock-down rats displayed typical signs of pseudohypoaldosteronism type I such as increased serum levels of aldosterone and renin as well as growth retardation. Importantly, we found a linear relationship between MR mRNA expression in kidney, serum aldosterone levels and body weight. Thus, our MR knock-down rats are amongst the first examples of RNAi in vivo and confirm that this technique allows to accomplish graded levels of gene inactivation that mimick human genetic diseases. Secondly, we investigated the role of the GR and the MR for the immunomodulatory activities of glucocorticoids (GCs) in peritoneal macrophages. GCs are involved in the modulation of macrophage function and thereby control the host’s immune responses to pathogens. Therefore, GCs are widely used for the treatment of inflammation and autoimmune diseases. However, concerning these GC activities neither the role of hormone concentration nor the differential contribution of the GR and the MR are known. At first we confirmed that both receptors but not 11β-HSD2 are expressed in peritoneal macrophages. Next, we showed that low levels of corticosterone enhance NO production as well as mRNA expression of pro-inflammatory cytokines, chemokines and enzymes required for mediator synthesis. In contrast, at high corticosterone concentrations macrophage function was strongly repressed. Importantly, inactivation of the GR by lentiviral delivery of siRNAs abrogated both the immunostimulatory and the immunosuppressive GC actions whereas inactivation of the MR had no effect. Furthermore, removal of endogenous GCs by adrenalectomy in vivo induced a pre-activated state in macrophages that could be modulated by corticosterone. We conclude that GCs exert distinct effects on macrophage function dependent on their concentration, and that they act through the GR despite concomitant expression of the MR. In summary, our results confirm that lentiviral delivery of shRNAs is an efficient means to down-regulation gene expression in primary cells and transgenic rats and thereby allows to perform functional studies on gene function that were previously limited to mice.
Immunsupprimierte Patienten besitzen ein erhöhtes Risiko für opportunistische Infektionen, die hauptsächlich durch das humane Cytomegalievirus (HCMV) und den Schimmelpilz Aspergillus fumigatus verursacht werden. Aufgrund ihrer Lokalisation in den Geweben unterhalb von Lungenepithelien und des Gastrointestinaltraktes werden dendritische Zellen (DCs) als diejenigen Zellen betrachtet, die während der frühen Phase einer Infektion in Kontakt mit HCMV und A. fumigatus kommen und eine Aktivierung von angeborenen und adaptiven Abwehrmechanismen vermitteln. Im Rahmen der vorliegenden Dissertation wurde die Bedeutung von humanen DCs bei der Bekämpfung von HCMV und A. fumigatus näher untersucht. Um mit dem klinisch relevanten HCMV Stamm TB40E arbeiten zu können, musste zuerst ein geeignetes Zellkultursystem zur Anzucht von HCMV etabliert werden. Die aus Fibroblasten aufgereinigten Viren eigneten sich zur erfolgreichen Infektion von DCs, was durch verschiedene Färbemethoden nachgewiesen werden konnte. Aus diesem Grund war es möglich, in Abhängigkeit der Zeit ein Expressionsprofil von Klasse I Interferonen (IFN-alpha, IFN-beta), ausgesuchten Cytokinen (CXCL10, CXCL11, Rantes) und den wichtigen Immunrezeptoren Toll-like Rezeptor 3 (TLR3) und dendritic cell-specific ICAM3-grabbing nonintegrin (DC-SIGN) zu erstellen. Nachdem ein RNA Interferenz (RNAi) System zur erfolgreichen Transfektion von DCs mit small interfering RNA (siRNA) etabliert werden konnte, gelang es die Expression von TLR3 signifikant herunterzuregulieren. Stimulationsexperimente mit dem synthetisch hergestellten Polymer poly I:C identifizierten TLR3 als den Rezeptor, der die Expression von IFN-beta vermittelt. Ferner konnte nachgewiesen werden, dass TLR9 bei ex vivo generierten DCs keine Funktion besitzt. Eine direkte Aktivierung von TLR3 durch HCMV konnte mittels siRNA nicht nachgewiesen werden. Durch den Einsatz von genomweiten Microarray-Analysen konnten eine Vielzahl an Genen gefunden werden, die nach Co-Kultivierung von DCs und lebenden A. fumigatus Keimschläuchen (KS) differentiell exprimiert waren. Dabei wurde ein breites Spektrum an Cytokinen (TNF-alpha, IL-6, IL-10, IL-12), Chemokinen (IL-8, CCL20, CXCL10), Co-stimulatorischen Molekülen (CD40, CD80, CD83, CD86), Prostaglandin Synthese Genen (PTGS2) und Immunrezeptoren (PTX-3, TLR2, TLR4) gefunden, deren zeitabhängiges Expressionsprofil mittels qRT-PCR eindeutig bestätigt wurde. Als Wachen des Immunsystems müssen DCs Krankheitserreger zu einem frühen Zeitpunkt der Infektion erkennen. Die Erkennung von Pilzen wird durch die unterschiedlichen Rezeptoren vermittelt, die TLRs, C-Typ Lektine und Pentraxine umfassen, wobei ihre Bedeutung für humane DCs bisher nur unzureichend geklärt ist. Durch den Einsatz von siRNA konnte die Expression von TLR2, TLR4, myeloid differentiation primary response gene 88 (MyD88), DC-SIGN, Pentraxin-3 (PTX-3) und caspase recruitment domain family member 9 (Card-9) signifikant verringert werden. Für TLR2, TLR4, PTX-3 und DC-SIGN konnte durch den Einsatz der RNAi aufgezeigt werden, dass diese Rezeptoren nicht an der Induktion einer pro-inflammatorischen Immunantwort von DCs nach Infektion mit A. fumigatus beteiligt sind. Sowohl die Stimulierung mit den TLR Liganden Zymosan und LPS, als auch mit A. fumigatus, führte zu einer erhöhten Expression von TNF-alpha und IL-12 (Light Cycler), die sich in einer vermehrten Cytokinfreisetzung (ELISA) bemerkbar machte. Im Gegensatz zur TLR4 siRNA Transfektion und LPS-Stimulation war keine Reduktion der Expression von TNF-alpha und IL-12 nach TLR2 und TLR4 siRNA Transfektion und anschließender Pilzinfektion zu beobachten. Auch der Einsatz von gegen TLRs gerichteten Antikörpern konnte eine mögliche Signaltransduktion bei DCs nicht unterbinden. Anstelle von TLR2 und TLR4 wurde Dectin-1 als DC-Immunrezeptor für A. fumigatus KS identifiziert. Mit Hilfe eines spezifischen Antikörpers gegen Dectin-1 war es möglich, die Freisetzung von TNF-alpha und IL-12 nach Pilzinfektion zu blockieren. In einem unabhängigen Experiment mit siRNA wurde Dectin-1 als Rezeptor für A. fumigatus bestätigt. Wie fortführende Experimente mit Candida albicans KS und Zymosan gezeigt haben, handelt es sich bei Dectin-1 auf humanen DCs um einen generellen Rezeptor für Pilze. Die durchgeführten SNP-Analysen (single nucleotide polymorphism) zur Ermittlung eines Zusammenhanges mit einem erhöhten Virus- und Pilzinfektionsrisiko für Patienten nach Stammzelltransplantation erbrachten die Erkenntnis darüber, dass zwei Marker (rs735240, rs2287886) in DC-SIGN mit einer erhöhten Empfänglichkeit für HCMV, und drei Marker (rs1554013, rs3921, rs4257674) in CXCL10 mit einem vergrößerten Riskio für eine invasive Aspergillose assoziiert waren. Ein Screening von Patienten auf das Vorhandensein dieser definierten SNPs könnte helfen, die individuelle Gefahr für HCMV und A. fumigatus nach nach allogener Stammzelltransplantation abzuschätzen.
Im Rahmen dieser Arbeit wurde die Expression des putativen Onkogens HDGF (Hepatoma Derived Growth Factor) in der humanen Kolonkarzinomzelllinie HT29 mittels RNA Interferenz herunterreguliert. Zwei unterschiedliche, für das Zielgen HDGF spezifische siRNAs wurden hierzu jeweils in einen Plasmidvektor kloniert. Zellen der Kolonkarzinomzelllinie HT29 wurden zunächst transient und stabil mit den beiden resultierenden Plasmidvektoren transfiziert, die Expression von HDGF in den transfizierten Zellen mittels Realtime-PCR quantifiziert und mit der Expression in mit Lipofektamin behandelten Kontrollzellen verglichen. Durch die stabile Transfektion beider Plasmidvektoren konnte die HDGF-Expression fast komplett supprimiert werden. Im Rahmen eines cDNA-Array konnten außer einer Expressionsverminderung von HDGF noch multiple Expressionsveränderungen anderer Gene identifiziert werden. Dies ist zum einen durch unspezifische, durch den Plasmidvektor bedingte Effekte erklärbar. Zum anderen aber ist die Deregulation vieler dieser Gene ein Effekt der Inaktivierung von HDGF.
Die subakut sklerosierende Panenzephalitis ist eine demyelinisierende Erkrankung des ZNS. Ätiologisch liegt eine persistierende Infektion von Masernviren der Neuronen bzw. Gliazellen zugrunde. Bis heute gibt es keine spezifische Therapie und lediglich symptomatische Therapiemaßnahmen werden in deren Behandlung angewandt. Obwohl sich während des Krankheitsverlaufes eine ausgeprägte Immunantwort des natürlichen als auch adaptiven Immunsystems des Wirtes abspielt führt die Erkrankung unweigerlich zum Tode der Betroffenen. Eine Therapiemöglichkeit diesbezüglich könnten die Effekte der RNAi darstellen. Um geeignete Sequenzen für einen möglichen Genknockdown einzelner MV-Gene herauszufiltern, wurde ein plasmid-basierendes Testsystem entwickelt, das die mRNA des Fluoreszenzproteins DsRed2 zusammen mit mRNA-Sequenzen einzelner MV-Gene exprimiert. Gegen diese MV-Gene wiederum wurden verschiedene shRNAs ausgewählt, welche mithilfe eines lentiviralen Vektorsystems exprimiert werden. Als Expressionsindikator findet in diesem lentiviralen Vektorsystem simultan die Expression von eGFP als statt. Die Auswertung dieses Dual-Color-Systems erfolgte im Folgenden mittels FACS-Analysen. Die wirksamsten siRNA-Sequenzen, ausgewählt durch eine hohe Abnahme der Rotfluoreszenz in den untersuchten Zellen, wurden für weitere Versuche selektiert. Die auf diese Weise ausgewählten shRNA-Sequenzen wurden in einem weiteren Schritt durch die Generierung lentiviraler Partikel in persistierend infizierten NT2-Zellen (piNT2), die das Fluoreszenzprotein HcRed als Infektionsindikator exprimieren, transduziert. Weitere durchflusszytometrische Messungen zeigten eine äußerst hohe Reduktion viraler Replikation innerhalb von 7 Tagen bis unterhalb der Detektionsgrenze sofern die shRNAs gegen die Expression der MV-Proteine N, P und L gerichtet waren. Die Anzahl der virus-negativen Zellen blieb im Folgenden bis zu 3 Wochen nach stattgehabter Transduktion konstant, sofern das fusion-inhibiting-peptide den Zellkulturen hinzugegeben wurde. Die transduzierten piNT2-Zellen, welche keine Expression von HcRed zeigten wurden auf Zellrasen bestehend aus Vero-Zellen aufgetragen und führten im zeitlichen Verlauf zu keiner Fusion bzw. Synzytienbildung. Dieser Zusammenhang legt nahe, dass die transduzierten piNT2-Zellen die Fähigkeit einer Expression viraler Proteine verloren haben und eine „Heilung“ stattfinden kann, wenn die virale Proteinbiosynthese durch eine permanente Expression von siRNAs, wie beispielsweise durch lentivirale Vektorsysteme, gehemmt wird.
Visual Attention in Flies-Dopamine in the Mushroom Bodies Mediates the After-Effect of Cueing

Visual environments may simultaneously comprise stimuli of different significance. Often such stimuli require incompatible responses. Selective visual attention allows an animal to respond exclusively to the stimuli at a certain location in the visual field. In the process of establishing its focus of attention the animal can be influenced by external cues. Here we characterize the behavioral properties and neural mechanism of cueing in the fly Drosophila melanogaster. A cue can be attractive, repulsive or ineffective depending upon (e.g.) its visual properties and location in the visual field. Dopamine signaling in the brain is required to maintain the effect of cueing once the cue has disappeared. Raising or lowering dopamine at the synapse abolishes this after-effect. Specifically, dopamine is necessary and sufficient in the αβ-lobes of the mushroom bodies. Evidence is provided for an involvement of the αβ\(_{posterior}\) Kenyon cells.
Cryptochrome (CRY) is the primary photoreceptor of Drosophila’s circadian clock. It resets the circadian clock by promoting light-induced degradation of the clock protein Timeless (TIM) in the proteasome. Under constant light, the clock stops because TIM is absent, and the flies become arrhythmic. In addition to TIM degradation, light also induces CRY degradation. This depends on the interaction of CRY with several proteins such as the E3 ubiquitin ligases Jetlag (JET) and Ramshackle (BRWD3). However, CRY can seemingly also be stabilized by interaction with the kinase Shaggy (SGG), the GSK-3 beta fly orthologue. Consequently, flies with SGG overexpression in certain dorsal clock neurons are reported to remain rhythmic under constant light. We were interested in the interaction between CRY, Ramshackle and SGG and started to perform protein interaction studies in S2 cells. To our surprise, we were not able to replicate the results, that SGG overexpression does stabilize CRY, neither in S2 cells nor in the relevant clock neurons. SGG rather does the contrary. Furthermore, flies with SGG overexpression in the dorsal clock neurons became arrhythmic as did wild-type flies. Nevertheless, we could reproduce the published interaction of SGG with TIM, since flies with SGG overexpression in the lateral clock neurons shortened their free-running period. We conclude that SGG does not directly interact with CRY but rather with TIM. Furthermore we could demonstrate, that an unspecific antibody explains the observed stabilization effects on CRY.
5’-3’ decay is the major mRNA decay pathway in many eukaryotes, including trypanosomes. After deadenylation, mRNAs are decapped by the nudix hydrolase DCP2 of the decapping complex and finally degraded by the 5’-3’ exoribonuclease. Uniquely, trypanosomes lack homologues to all subunits of the decapping complex, while deadenylation and 5’-3’ degradation are conserved. Here, I show that the parasites use an ApaH-like phosphatase (ALPH1) as their major mRNA decapping enzyme. The protein was recently identified as a novel trypanosome stress granule protein and as involved in mRNA binding. A fraction of ALPH1 co-localises exclusively with the trypanosome 5’-3’ exoribonuclease XRNA to a special granule at the posterior pole of the cell, indicating a connection between the two enzymes. RNAi depletion of ALPH1 is lethal and causes a massive increase in total mRNAs that are deadenylated, but have not yet started 5’-3’ decay. These data suggest that ALPH1 acts downstream of deadenylation and upstream of mRNA degradation, consistent with a function in mRNA decapping. In vitro experiments show that recombinant, N-terminally truncated ALHP1 protein, but not a catalytically inactive mutant, sensitises the capped trypanosome spliced leader RNA to yeast Xrn1, but only if an RNA 5’ polyphosphatase is included. This indicates that the decapping mechanism of ALPH1 differs from the decapping mechanism of Dcp2 by leaving more than one phosphate group at the mRNA’s 5’ end. This is the first reported function of a eukaryotic ApaH-like phosphatase, a bacterial-derived class of enzymes present in all phylogenetic super-groups of the eukaryotic kingdom. The substrates of eukaryotic ApaH-like phosphatases are unknown. However, the substrate of the related bacterial enzyme ApaH, diadenosine tetraphosphate, is highly reminiscent of a eukaryotic mRNA cap.
Als Teil des angeborenen Immunsystems spielen Natürliche Killer(NK)- Zellen eine entscheidende Rolle in der Interaktion mit Pathogenen und Tumorzellen. Mithilfe der RNA-Interferenz bestimmter Gene wie beispielsweise CD56 könnten Hinweise auf die genaue Funktionsweise der NK-Zellen gewonnen werden. Ziel dieser Arbeit war es eine geeignete Transfektionsmethode zum Einführen von siRNA in NK-Zellen zu finden, welche eine hohe Transfektionseffizienz bei gleichzeitig hoher Zellviabilität aufweist. Hierfür wurden drei verschiedene Lipofektionsreagenzien und sechs verschiedene Elektroporationsprogramme verglichen. Zunächst wurde die Transfektionseffizienz mit an AF488 gekoppelter negativer siRNA per Fluoreszenzmikroskopie und Durchflusszytometrie evaluiert und jeweils das effizienteste Lipofektionsreagenz bzw. Elektroporationsprogramm ausgewählt. Mit diesen wurde anschließend eine Herunterregulation des CD56-Gens mit CD56-siRNA per RNA-Interferenz versucht. Die CD56-Gen-Expression wurde auf Proteinebene per Durchflusszytometrie und auf mRNA-Ebene per PCR (Polymerase-Ketten-Reaktion) evaluiert.
A cascade of histone acetylation events with subsequent incorporation of a histone H2A variant plays an essential part in transcription regulation in various model organisms. A key player in this cascade is the chromatin remodelling complex SWR1, which replaces the canonical histone H2A with its variant H2A.Z. Transcriptional regulation of polycistronic transcription units in the unicellular parasite Trypanosoma brucei has been shown to be highly dependent on acetylation of H2A.Z, which is mediated by the histone-acetyltransferase HAT2. The chromatin remodelling complex which mediates H2A.Z incorporation is not known and an SWR1 orthologue in trypanosomes has not yet been reported. In this study, we identified and characterised an SWR1-like remodeller complex in T. brucei that is responsible for Pol II-dependent transcriptional regulation. Bioinformatic analysis of potential SNF2 DEAD/Box helicases, the key component of SWR1 complexes, identified a 1211 amino acids-long protein that exhibits key structural characteristics of the SWR1 subfamily. Systematic protein-protein interaction analysis revealed the existence of a novel complex exhibiting key features of an SWR1-like chromatin remodeller. RNAi-mediated depletion of the ATPase subunit of this complex resulted in a significant reduction of H2A.Z incorporation at transcription start sites and a subsequent decrease of steady-state mRNA levels. Furthermore, depletion of SWR1 and RNA-polymerase II (Pol II) caused massive chromatin condensation. The potential function of several proteins associated with the SWR1-like complex and with HAT2, the key factor of H2A.Z incorporation, is discussed.
