The extracellular matrix (ECM) of soft tissues in vivo has remarkable biological and structural properties. Thereby, the ECM provides mechanical stability while it still can be rearranged via cellular remodeling during tissue maturation or healing processes. However, modern synthetic alternatives fail to provide these key features among basic properties. Synthetic matrices are usually completely degraded or are inert regarding cellular remodeling. Based on a refined electrospinning process, a method is developed to generate synthetic scaffolds with highly porous fibrous structures and enhanced fiber‐to‐fiber distances. Since this approach allows for cell migration, matrix remodeling, and ECM synthesis, the scaffold provides an ideal platform for the generation of soft tissue equivalents. Using this matrix, an electrospun‐based multilayered skin equivalent composed of a stratified epidermis, a dermal compartment, and a subcutis is able to be generated without the use of animal matrix components. The extension of classical dense electrospun scaffolds with high porosities and motile fibers generates a fully synthetic and defined alternative to collagen‐gel‐based tissue models and is a promising system for the construction of tissue equivalents as in vitro models or in vivo implants.
In this study, well-defined, 3D arrays of air-suspended melt electrowritten fibers are made from medical grade poly(ɛ-caprolactone) (PCL). Low processing temperatures, lower voltages, lower ambient temperature, increased collector distance, and high collector speeds all aid to direct-write suspended fibers that can span gaps of several millimeters between support structures. Such processing parameters are quantitatively determined using a “wedge-design” melt electrowritten test frame to identify the conditions that increase the suspension probability of long-distance fibers. All the measured parameters impact the probability that a fiber is suspended over multimillimeter distances. The height of the suspended fibers can be controlled by a concurrently fabricated fiber wall and the 3D suspended PCL fiber arrays investigated with early post-natal mouse dorsal root ganglion explants. The resulting Schwann cell and neurite outgrowth extends substantial distances by 21 d, following the orientation of the suspended fibers and the supporting walls, often generating circular whorls of high density Schwann cells between the suspended fibers. This research provides a design perspective and the fundamental parametric basis for suspending individual melt electrowritten fibers into a form that facilitates cell culture.
The foreign body reaction to neuronal electrode implants limits potential applications as well as the therapeutic period. Developments in the basic electrode design might improve the tissue compatibility and thereby reduce the foreign body reaction. In this work, the approach of embedding 3D carbon nanofiber electrodes in extracellular matrix (ECM) synthesized by human fibroblasts for a compatible connection to neuronal cells was investigated. Porous electrode material was manufactured by solution coelectrospinning of polyacrylonitrile and polyamide as a fibrous porogen. Moreover, NaCl represented an additional particulate porogen. To achieve the required conductivity for an electrical interface, meshes were carbonized. Through the application of two different porogens, the electrodes' flexibility and porosity was improved. Human dermal fibroblasts were cultured on the electrode surface for ECM generation and removed afterwards. Scanning electron microscopy imaging revealed a nano fibrous ECM network covering the carbon fibers. The collagen amount of the ECM coating was quantified by hydroxyproline-assays. The modification with the natural protein coating on the electrode functionality resulted in a minor increase of the electrical capacity, which slightly improved the already outstanding electrical interface properties. Increased cell numbers of SH-SY5Y cell line on ECM-modified electrodes demonstrated an improved cell adhesion. During cell differentiation, the natural ECM enhanced the formation of neurites regarding length and branching. The conducted experiments indicated the prevention of direct cell-electrode contacts by the modification, which might help to shield temporary the electrode from immunological cells to reduce the foreign body reaction and improve the electrodes' tissue integration.
Kardiovaskuläre Erkrankungen, wie beispielsweise der Herzinfarkt, sind die häufigste Todesursache weltweit. Bei einem Herzinfarkt sterben Areale des Herzens aufgrund einer Unterversorgung mit Blut ab. Da das Herzmuskelgewebe ein sogenanntes terminal differenziertes Gewebe ist, kommt es zu keiner Regeneration des Gewebes, mit der Folge einer Herzinsuffizienz beziehungsweise dem Tod des Patienten. Eine alternative Behandlungsmöglichkeit zu einer Herztransplantation stellt das Tissue Engineering dar. Mit Hilfe des Tissue Engineerings können dreidimensionale Gewebe aufgebaut und kultiviert werden, um auf diese Weise ein funktionelles Gewebe zu erhalten, durch welches das abgestorbene Gewebeareal des Herzens zukünftig auch ersetzt werden könnte.
In der vorliegenden Arbeit wurden notwendige Technologien für den Aufbau von Geweben entwickelt sowie erste Versuche für die Erzeugung eines funktionellen Herzmuskelgewebes durchgeführt. Beim Aufbau von dreidimensionalen Geweben finden Trägerstrukturen Anwendung, die mit Zellen besiedelt werden. Solche Trägerstrukturen können aus biologischen oder synthetischen Polymeren hergestellt sein oder aus der extrazellulären Matrix eines dezellularisierten Gewebes bestehen. Für eine standardisierte Dezellularisierung von Geweben wurde eine computergesteuerte Pumpeneinheit, für die Herstellung von Nanofaserscaffolds eine Elektrospinninganlage entwickelt. Mit Hilfe der Dezellularisierungseinheit können komplexe Organe, wie ein Herz im Ganzen, reproduzierbar dezellularisiert werden. Untersuchungen der mittels Elektrospinning hergestellten Nanofaserscaffolds, welche als Alternative zu der dezellularisierten, natürlichen Matrix eingesetzt werden können, zeigten bei allen hergestellten Zusammensetzungen eine Orientierung der Zellen entlang der Fasern.
Die Kultivierung von Zellmatrixkonstrukten erfolgt im Tissue Engineering häufig unter dynamischen Bedingungen. Hierfür wurde ein mobiler Stand Alone Inkubator mit der erforderlichen Peripherie für eine Kultur unter Perfusion des Gewebes entwickelt. Als Weiterentwicklung des Stand Alone Inkubators ist eine modulare Bioreaktorplattform, bestehend aus Wärmetauscher, Beutelpumpe und Gasaustauscher, aufgebaut worden. In dieses System kann über Standard Anschlüsse jegliche Art von Bioreaktor in das System eingebunden werden. Durch die Kompaktheit des Systems ist es möglich mehrere Ansätze parallel auf engem Raum durchzuführen. Die Funktion der Plattform, wurde in der vorliegenden Arbeit durch die Gewebekultur einer nativen porzinen Karotis nachgewiesen.
Für den Aufbau des kardialen Gewebes dient die small intestinal submucosa ohne Serosa (SISser) als Trägerstruktur. Der Aufbau des Gewebekonstrukts erfolgte in verschiedenen Ansätzen unter Einsatz verschiedener Zellarten. Native, aus Herzbiopsien generierte Cardiosphere derived cells (CDCs) verteilten sich gleichmäßige über die Oberfläche der Matrix, jedoch konnten immunhistologisch keine spezifischen kardialen Marker bei den artifiziellen Geweben nachgewiesen werden. Zellmatrixkonstrukte aus einer Mono Kultur von Kardiomyozyten, differenziert aus induzierten pluripotenten Stammzellen (iPS Zellen) sowie einer Co Kultur dieser Kardiomyozyten mit mesenchymalen Stammzellen und Zellen aus einer Herzbiopsie zeigten nach wenigen Tagen in Kultur ein kontraktiles Verhalten. Immunhistologische Färbungen der beiden Gewebe bestätigten die Expression der spezifischen kardialen Marker, wie beispielsweise kardiales Troponin T, kardiales Troponin C und alpha Actinin. Die Kardiomyozyten der Mono Kultur sind jedoch nicht über die gesamte Matrixoberfläche verteilt, sondern bilden Aggregate. Bei der Co Kultur kann eine gleichmäßige Verteilung der Zellen auf der Matrix beobachtet werden. Der vielversprechendste Ansatz für den Aufbau eines Herzmuskelgewebes, welches als Implantat oder Testsystem eingesetzt werden kann, bildet nach den in dieser Arbeit erzielten Ergebnissen, ein Konstrukt aus der SISser und der Co Kultur der Zellen. Allerdings muss die Zusammensetzung der Co Kultur sowie das Verhältnis der Zellzahlen optimiert werden.
Elastic fibers are essential for the proper function of organs including cardiovascular tissues such as heart valves and blood vessels. Although (tropo)elastin production in a tissue-engineered construct has previously been described, the assembly to functional elastic fibers in vitro using human cells has been highly challenging. In the present study, we seeded primary isolated human vascular smooth muscle cells (VSMCs) onto 3D electrospun scaffolds and exposed them to defined laminar shear stress using a customized bioreactor system. Increased elastin expression followed by elastin deposition onto the electrospun scaffolds, as well as on newly formed fibers, was observed after six days. Most interestingly, we identified the successful deposition of elastogenesis-associated proteins, including fibrillin-1 and -2, fibulin-4 and -5, fibronectin, elastin microfibril interface located protein 1 (EMILIN-1) and lysyl oxidase (LOX) within our engineered constructs. Ultrastructural analyses revealed a developing extracellular matrix (ECM) similar to native human fetal tissue, which is composed of collagens, microfibrils and elastin. To conclude, the combination of a novel dynamic flow bioreactor and an electrospun hybrid polymer scaffold allowed the production and assembly of an elastic fiber-containing ECM.
