CD70-abhängige und spezifische Aktivierung von TRAILR1 oder TRAILR2 durch scFv:CD70-TRAIL-Mutanten

Das Ziel dieser Arbeit bestand darin, den T-Zell-inhibierenden Effekt eines CD70-blockierenden Antikörpers mit einer Fc-unabhängigen Zelltod-induzierenden Aktivität auf CD70-exprimierende Tumoren zu kombinieren. Dazu wurden Fusionsproteine hergestellt und untersucht, die aus einer CD70-bindenden scFv-Domäne sowie aus einer TRAIL-Domäne bestehen.
Der CD70-spezifische monoklonale Antikörper lαhCD70 sowie der beretis bekannte hCD70-spezifische Antikörper 1F6 blockieren mit hoher Effizienz die CD27/CD70-Interaktion von CD70-exprimierenden Zelllinien (Mino, OVCAR-3, U-266) und inhibieren dadurch die Induktion der IL8-Produktion durch diese Zellen in kokultivierten HT1080-CD27-Zellen. IL8 wird durch den klassischen NFκB-Signalweg reguliert und ist für den pro-angiogenetischen Effekt von entscheidender Bedeutung (Abb. 2, 3). Mit Hilfe zellulärer Gleichgewichtsbindungsstudien mit mono- und trimeren scFv:lαhCD70-GpL-Fusionsproteinen (Abb. 4) auf Mino- und OVCAR-3-Zellen konnte gezeigt werden, dass die Trimerisierung in beiden Zelllinien zu einer Steigerung der apparenten Affinität der scFv:lαhCD70-CD70 Interaktion führt und damit einen Effekt auf die CD70-Belegung hat (Abb. 5). Für die Konstruktion der Fusionsproteine wurde sowohl Wildtyp-TRAIL als auch TRAIL-Mutanten mit Präferenz für den TRAILR1 oder TRAILR2 verwendet. Die TRAILR-Präferenz der verwendeten TRAIL-Mutanten (wt, mutR1, mutR2) wurde nicht nur in zellulären GpL-Bindungsstudien (Abb. 7) sondern zusätzlich auch in TRAILR Immobilisierungsexperimenten (Abb. 8) bewiesen. Hier zeigte sich, dass bei TRAILR1 keine Interaktion mit TRAILmutR2, so wie bei TRAILR2 keine signifikante Bindung mit TRAILmutR1 erfolgte. Nur der TRAIL-Wildtyp band signifikant an beide TRAIL-Todesrezeptoren. Vitalitätsexperimente (Abb. 10) und Western-Blot Analysen der Caspase-Prozessierung (Abb. 11) bestätigten die starke TRAILR1- bzw. TRAILR2-Spezifität der TRAILmutR1- und TRAILmutR2-Varianten. Im Gegensatz zu den unvernetzten löslichen TRAIL-Trimeren waren nur die
quervernetzten TRAIL-TNC-Varianten in der Lage, eine signifikante Apoptose-Signalkaskade bei relativ geringen Konzentrationen zu induzieren. Die toxischen ED50-Konzentrationen der unoligomerisierten TRAIL-Formen lagen um einen Faktor 100 höher als die der oligomerisierten Varianten. Zusammenfassend zeigten die ED50-Werte der Zytotoxizitätsexperimente von M2-oligomerisierten zu -unoligomerisierten trimeren TRAIL-Varianten bei allen Fusionskonstrukten und Zelllinien eine eindeutige Verstärkung der Apoptoseinduktion durch die M2-Quervernetzung. Bei Jurkat- und Mino-Zellen konnte größtenteils erst nach Oligomerisierung überhaupt eine Bioaktivität bzw. eine Zelltodinduktion beobachtet werden. In OVCAR-3-Zellen zeigte sich eine 100-1000 fache apoptotische Verstärkung durch die Oligomerisierung (Abb. 10). Weiterhin zeigten Zytotoxizitätsexperimente, dass sich durch Bindung an hCD70 das Ausmaß der Toxizität der Fusionsproteine auf allen CD70-exprimierenden Zelllinien 10-100x verstärkte (Abb. 15, 17). In Übereinstimmung mit der verstärkten TRAIL-Todesrezeptor-Aktivierung durch die CD70-Bindung der scFv-TRAIL-Fusionsproteine, konnte durch eine CD70-Blockade die Caspase-8 Aktivierung und die Prozessierung von Caspase-3 signifikant unterbunden werden (Abb. 18). Die Trimerisierung des scFv:lαhCD70-Antikörpers führte zu keiner Apoptose und beeinflußte auch nicht die Aktivität von TRAIL (Abb. 19) was belegt, dass die beobachteten Effekte auf einer stärkeren TRAIL-induzierten Apoptose nach CD70-Bindung der Konstrukte beruhen muss.
Die Fusionsproteine beseitigen somit nachweislich einerseits das Problem der limitierenden Aktivität von löslichem TRAIL über ihre Verankerung an CD70 (Abb. 15-20) und anderseits die potentielle unerwünschte CD70-vermittelte protumorale CD27-Stimulation (Abb. 3). Darüber hinaus könnten die TRAILR-spezifischen TRAIL-Mutanten helfen, Nebeneffekte zu reduzieren, die primär durch den jeweils anderen TRAIL-Todesrezeptor vermittelt werden. Jedoch sind weiter Forschungen insbesondere in vivo Experimente notwendig, um Aussagen über Funktionalität, Halbwertszeiten, sowie Effektivität und Verträglichkeit treffen zu können.
To combine the CD27 stimulation inhibitory effect of blocking CD70 antibodies with an antibody-dependent cellular cytotoxicity (ADCC)-independent, cell death-inducing activity for targeting of CD70-expressing tumors, we evaluated here fusion proteins of the apoptosis-inducing TNF family member TRAIL and a single-chain variable fragment (scFv) derived from a high-affinity llama-derived anti-human CD70 antibody (lαhCD70). A fusion protein of scFv:lαhCD70 with TNC-TRAIL, a stabilized form of TRAIL, showed strongly enhanced apoptosis induction upon CD70 binding and furthermore efficiently interfered with CD70-CD27 interaction. Noteworthy, introduction of recently identified mutations that discriminate between TRAILR1 and TRAILR2 binding into the TRAIL part of scFv:lαhCD70-TNC-TRAIL resulted in TRAIL death receptor-specific fusion proteins with CD70-restricted activity.
Background
It is hypothesized that because of higher mast cell numbers and mediator release, mastocytosis predisposes patients for systemic immediate-type hypersensitivity reactions to certain drugs including non-steroidal anti-inflammatory drugs (NSAID).
Objective
To clarify whether patients with NSAID hypersensitivity show increased basal serum tryptase levels as sign for underlying mast cell disease.
Methods
As part of our allergy work-up, basal serum tryptase levels were determined in all patients with a diagnosis of NSAID hypersensitivity and the severity of the reaction was graded. Patients with confirmed IgE-mediated hymenoptera venom allergy served as a comparison group.
Results
Out of 284 patients with NSAID hypersensitivity, 26 were identified with basal serum tryptase > 10.0 ng/mL (9.2%). In contrast, significantly (P = .004) more hymenoptera venom allergic patients had elevated tryptase > 10.0 ng/mL (83 out of 484; 17.1%). Basal tryptase > 20.0 ng/mL was indicative for severe anaphylaxis only in venom allergic subjects (29 patients; 4x grade 2 and 25x grade 3 anaphylaxis), but not in NSAID hypersensitive patients (6 patients; 4x grade 1, 2x grade 2).
Conclusions
In contrast to hymenoptera venom allergy, NSAID hypersensitivity do not seem to be associated with elevated basal serum tryptase levels and levels > 20 ng/mL were not related to increased severity of the clinical reaction. This suggests that mastocytosis patients may be treated with NSAID without special precautions.
In this work, a model-based acceleration of parameter mapping (MAP) for the determination of the tissue parameter T1 using magnetic resonance imaging (MRI) is introduced. The iterative reconstruction uses prior knowledge about the relaxation behavior of the longitudinal magnetization after a suitable magnetization preparation to generate a series of fully sampled k-spaces from a strongly undersampled acquisition. A Fourier transform results in a spatially resolved time course of the longitudinal relaxation process, or equivalently, a spatially resolved map of the longitudinal relaxation time T1.
In its fastest implementation, the MAP algorithm enables the reconstruction of a T1 map from a radial gradient echo dataset acquired within only a few seconds after magnetization preparation, while the acquisition time of conventional T1 mapping techniques typically lies in the range of a few minutes. After validation of the MAP algorithm for two different types of magnetization preparation (saturation recovery & inversion recovery), the developed algorithm was applied in different areas of preclinical and clinical MRI and possible advantages and disadvantages were evaluated.
Aims: The aim of the current study was to establish a simple and yet as much as possible physiologic approach for a simulation of the pulmonary absorption process to compare different inhaled drugs or drug formulations.
Methodology: We designed a dialysis setting that allowed monitoring the drug release from human lung tissue into a continuous-flow plasma compartment. For proof-of-concept experiments we chose the glucocorticoid fluticasone propionate (FP) as model compound. For subsequent experiments we selected a commercially available metered dose inhaler delivering a fixed combination of the short-acting ß2-agonist fenoterol and the muscarinic antagonist ipratropium bromide.
Results: With the novel dynamic dialysis model we observed high drug transport rates from the lung tissue into plasma including an elimination phase. The concentration profile in the plasma compartment of our model system was similar to the plasma concentration courses after inhalation of FP. Compared to FP significantly higher drug fractions of fenoterol and ipratropium bromide were released into plasma and the transfer of ipratropium was more pronounced compared to fenoterol. Again, concentration profiles in plasma were alike to those described in clinical studies.
Conclusion: We suggest that this model is appropriate for rapid assessment of comparative diffusion behaviour of drugs or drug formulations from lung tissue into plasma.
Stress impacts negatively on plant growth and crop productivity, causing extensive losses to agricultural production worldwide. Throughout their life, plants are often confronted with multiple types of stress that affect overall cellular energy status and activate energy-saving responses. The resulting low energy syndrome (LES) includes transcriptional, translational, and metabolic reprogramming and is essential for stress adaptation. The conserved kinases sucrose-non-fermenting-1-related protein kinase-1 (SnRK1) and target of rapamycin (TOR) play central roles in the regulation of LES in response to stress conditions, affecting cellular processes and leading to growth arrest and metabolic reprogramming. We review the current understanding of how TOR and SnRK1 are involved in regulating the response of plants to low energy conditions. The central role in the regulation of cellular processes, the reprogramming of metabolism, and the phenotypic consequences of these two kinases will be discussed in light of current knowledge and potential future developments.
Background: Teleost fish present a high diversity of sex determination systems, with possible frequent evolutionary turnover of sex chromosomes and sex-determining genes. In order to identify genes involved in male sex determination and differentiation in the platyfish Xiphophorus maculatus, bacterial artificial chromosome contigs from the sex-determining region differentiating the Y from the X chromosome have been assembled and analyzed.
Results: A novel three-copy gene called teximY (for testis-expressed in Xiphophorus maculatus on the Y) was identified on the Y but not on the X chromosome. A highly related sequence called texim1, probably at the origin of the Y-linked genes, as well as three more divergent texim genes were detected in (pseudo) autosomal regions of the platyfish genome. Texim genes, for which no functional data are available so far in any organism, encode predicted esterases/lipases with a SGNH hydrolase domain. Texim proteins are related to proteins from very different origins, including proteins encoded by animal CR1 retrotransposons, animal platelet-activating factor acetylhydrolases (PAFah) and bacterial hydrolases. Texim gene distribution is patchy in animals. Texim sequences were detected in several fish species including killifish, medaka, pufferfish, sea bass, cod and gar, but not in zebrafish. Texim-like genes are also present in Oikopleura (urochordate), Amphioxus (cephalochordate) and sea urchin (echinoderm) but absent from mammals and other tetrapods. Interestingly, texim genes are associated with a Helitron transposon in different fish species but not in urochordates, cephalochordates and echinoderms, suggesting capture and mobilization of an ancestral texim gene in the bony fish lineage. RT-qPCR analyses showed that Y-linked teximY genes are preferentially expressed in testis, with expression at late stages of spermatogenesis (late spermatids and spermatozeugmata).
Conclusions: These observations suggest either that TeximY proteins play a role in Helitron transposition in the male germ line in fish, or that texim genes are spermatogenesis genes mobilized and spread by transposable elements in fish genomes.
Gemäß den Zielen der Intersektionalitätsforschung untersucht der Beitrag die gesteigerte Diskriminierung derjenigen in Literatur und Film, die mehreren minderprivilegierten Gruppen angehören: die Frauen von Bechelaren, deren narrative Existenz im ‚Nibelungenlied‘ nur gesichert ist, solange sie in einer Relation zu ihrem Ehemann und Vormund stehen. Der Vergleich des mittelhochdeutschen Epos mit den modernen Adaptationen von Thea von Harbou und Fritz Lang zeigt, wie sich die Bedeutung der Ungleichheitskategorien fundamental verschiebt. Am negativsten wirkt sich die Verstärkung der Machtdifferenzen in Thea von Harbous ‚Nibelungenbuch‘ und Fritz Langs Film ‚Die Nibelungen‘ auf Rüdigers Frau Gotelind aus. Für die Markgräfin bleibt in den modernen Adaptationen aufgrund der rassisierenden Überformung der hunnischen Welt kein Platz mehr. Während sie im Epos Repräsentationsaufgaben übernimmt, Herrschaftspflichten erfüllt und großzügige Geschenke verteilt, tritt sie im Buch als Figur nicht in Erscheinung und gerät im Film nicht einmal in der Utopie von Bechelaren in den Blick. Gotelind wird nicht nur marginalisiert, sondern vollständig eliminiert.
Background: The Ikkα kinase, a subunit of the NF-kappa B-activating IKK complex, has emerged as an important regulator of inflammatory gene expression. However, the role of Ikkα-mediated phosphorylation in haematopoiesis and atherogenesis remains unexplored. In this study, we investigated the effect of a bone marrow (BM)-specific activation-resistant Ikk alpha mutant knock-in on haematopoiesis and atherosclerosis in mice.
Methods and Results: Apolipoprotein E (Apoe)-deficient mice were transplanted with BM carrying an activation-resistant Ikkα gene (Ikkα(AA/AA) Apoe(-/-)) or with Ikkα(+/+) Apoe(-/-) BM as control and were fed a high-cholesterol diet for 8 or 13 weeks. Interestingly, haematopoietic profiling by flow cytometry revealed a significant decrease in B-cells, regulatory T-cells and effector memory T-cells in Ikkα(AA/AA) Apoe(-/-) BM-chimeras, whereas the naive T-cell population was increased. Surprisingly, no differences were observed in the size, stage or cellular composition of atherosclerotic lesions in the aorta and aortic root of Ikkα(AA/AA) Apoe(-/-) vs Ikkα(+/+) Apoe(-/-) BM-transplanted mice, as shown by histological and immunofluorescent stainings. Necrotic core sizes, apoptosis, and intracellular lipid deposits in aortic root lesions were unaltered. In vitro, BM-derived macrophages from Ikkα(AA/AA) Apoe(-/-) vs Ikkα(+/+) Apoe(-/-) mice did not show significant differences in the uptake of oxidized low-density lipoproteins (oxLDL), and, with the exception of Il-12, the secretion of inflammatory proteins in conditions of Tnf-α or oxLDL stimulation was not significantly altered. Furthermore, serum levels of inflammatory proteins as measured with a cytokine bead array were comparable.
Conclusion: Our data reveal an important and previously unrecognized role of haematopoietic Ikkα kinase activation in the homeostasis of B-cells and regulatory T-cells. However, transplantation of Ikkα AA mutant BM did not affect atherosclerosis in Apoe(-/-) mice. This suggests that the diverse functions of Ikkα in haematopoietic cells may counterbalance each other or may not be strong enough to influence atherogenesis, and reveals that targeting haematopoietic Ikkα kinase activity alone does not represent a therapeutic approach.
This thesis treats the thermopower and other thermal effects in single quantum dots (QD) and quantum dot systems. It contributes new experimental results to the broad and active field of research on thermoelectrics in low dimensional systems. The thermopower experiments discussed in this work focus on QDs which exhibit a net spin and on tunnel-coupled double QDs (DQD). Furthermore, experiments are presented which address the realization of a QD device which extracts thermal energy from a heat reservoir and converts it into a directed charge current in a novel way.
The samples used for these investigations have been fabricated from GaAs/AlGaAs heterostructures which contain a two dimensional electron gas. Using optical and electron beam lithography, the devices have been realized by means of the top-gate technology. All experiments have been performed at low temperature. In order to create a controllable temperature difference in the samples the current heating technique has been used. These experimental basics as well as fundamentals of electric and thermoelectric transport are introduced in Part I of this thesis.
The experiments on the thermopower of a single QD are described in Part II. Essentially, they deal with the problem of how a single spin situated on a QD influences the thermoelectric properties of the system. In this context, the Kondo-effect plays a crucial role. Generally, the Kondo effect is the result of a many-body state which arises from an antiferromagnetic coupling of a magnetic impurity with the surrounding conduction electrons. Here, the magnetic impurity is represented by a QD which is occupied with an odd number of electrons so that it exhibits a net spin. For the first time the thermopower of a Kondo-QD has been studied systematically as a function of two parameters, namely the QD coupling energy and the sample temperature. Both parameters are crucial quantities for Kondo-physics to be observed. Based on these data, it is shown that the thermopower line shape as a function of QD energy is mainly determined by two competing contributions: On the one hand by the enhanced density of states around the Fermi level due to Kondo-correlations and on the other hand by thermopower contributions from the Coulomb resonances. Furthermore, the experiments confirm theoretical predictions which claim that the spectral DOS arising from Kondo-correlations shifts away from the Fermi level for those QD level configurations which are not electron-hole symmetric. Comparison with model calculations by T. Costi and V. Zlatic [Phys. Rev. B 81, 235127 (2010)] shows qualitative and partly even quantitative agreement. A finite thermovoltage at the center of the Kondo-region, which occurred in previous investigations, is also observed in the experiments presented here. It is not covered by the current theory of the Kondo effect. The dependence of this signal on temperature, coupling energy and magnetic field, which differ from non-Kondo regions, is analyzed. In order to clarify the physics behind this phenomenon further studies are desirable.
Furthermore, it is shown by variation of the QD coupling energy over a wide range that Kondo-correlations can be detected in the thermopower even in the regime of very weak coupling. In contrast, no Kondo signatures are visible in the conductance in this energy range. It is found that in the limit of weak coupling the Kondo effect causes the thermopower to exhibit a diminished amplitude in close vicinity of a conductance resonance. Subsequent filling of spin-degenerate states then leads to a thermopower amplitude modulation (odd-even-effect). Although this effect had been observed in previous studies, no connection to Kondo physics had been established in order to explain the observations.
Hence, the experiments on a single QD presented in this thesis provide unique insight into the complex interplay of different transport mechanisms in a spin-correlated QD. Moreover, the results confirm the potential of thermopower measurements as a highly sensitive tool to probe Kondo-correlations.
In Part III thermal effects are investigated in systems which contain two coupled QDs.
Such QD-systems are particularly interesting with respect to thermoelectric applications: Many proposals utilize the extremely sharp energy filtering properties of such coupled QDs and also different kinds of inter dot coupling to construct novel and highly efficient thermoelectric devices. In the present work, thermopower characterizations are performed on a tunnel-coupled DQD for the first time. The key result of these investigations is the thermopower stability diagram. Here it is found, that in such a system maximal thermopower is generated in the vicinity of the so-called triple points (TP) at which three charge states of the DQD are degenerate. Along the axis of total energy, which connects two adjacent TP, a typical thermopower line shape is observed. It is explained and modeled within an intuitive picture that assumes two transport channels across the DQD, representing the TP. For those regions which are far away from the TP, the thermopower turns out to be very sensitive to the relative configuration of the QD energies. The conductance and thermopower data are well reproduced within a model that assumes transport via molecular states. Integration of both models into one then allows model calculations for a complete stability cell in conductance and thermopower to be done.
Furthermore, experiments on two capacitively coupled QDs are presented. In these studies the focus lies on testing the feasibility of such systems for the manipulation and generation of charge currents from thermal energy. In a series of experiments it is shown that such a system of QDs can be utilized to increase or decrease a current flowing between two electron reservoirs by varying the temperature in a third reservoir. This effect is based on the cross-correlation of occupation fluctuations of the individual QDs. These are positive for certain QD energy level configurations and negative for others, which increases or decreases the charge current in the experiments, respectively. In the stability diagram this is manifested in a characteristic clover leaf shaped structure of positive and negative current changes in vicinity of the TP. All main experimental results are reproduced qualitatively in simple model calculations. Due to the close analogy between electrical and thermal conductance of a QD, this effect of thermal switching can, in principle, also be used to built a thermal transistor.
Finally, it is shown that a system consisting of two Coulomb-coupled QDs, which couple a hot electron reservoir electrostatically to two cold electron reservoirs, can be utilized as a novel device which extracts heat from its environment and converts it into a directed charge current. The idea of this heat-to-current converter (HCC) was first proposed by R. Sánchez and M. Büttiker [Phys. Rev. B 83, 085428 (2011)]. It is not only characterized by the novelty of its working principle but also by the fact, that it decouples the directions of charge current and energy flow. In the experiments presented here, such HCC-currents are identified unambiguously: For certain QD-level configurations an electric current between the two cold reservoirs is observed if the temperature in the third reservoir is increased. The direction of this current is shown to be independent of an external voltage. In contrast, the direction of the current exhibits a characteristic dependence on the tunneling coefficients of the QDs, as predicted by theory: By adjusting the thickness and the shape of the respective tunnel junctions, a charge current can be generated between two cold reservoirs, and it can even be inverted. The experimental observations are quantitatively reproduced by model calculations by R. Sánchez and B. Sothmann. Thus, the results represent direct evidence for the existence of HCC-currents. Due to the novelty of the working principle of the HCC and its relevance from a fundamental scientific point of view, the results presented here are an important step towards energy harvesting devices at the nano scale.
Function and regulation of phospholipase D in blood platelets: in vitro and in vivo studies in mice

Summary
Platelet activation and aggregation are crucial for primary hemostasis but can also result in occlusive thrombus formation. Agonist induced platelet activation involves different signaling pathways leading to the activation of phospholipases (PL) which produce second messengers. While the role of PLCs in platelet activation is well established, less is known about the relevance of PLDs. In the current study, the function and regulation of PLD in platelets was investigated using genetic and pharmacological approaches.
In the first part of this thesis, adhesion, activation and aggregation of platelets from mice lacking PLD2 or both PLD1 and PLD2 were analyzed in vitro and in vivo. While the absence of PLD2 resulted in slightly reduced PLD activity in platelets, it had no detectable effect on the platelet function in vitro and in vivo. However, the combined deficiency of both PLD isoforms resulted in defective alpha-granule release and protection in a model of ferric chloride induced arteriolar thrombosis, effects that were not observed in mice lacking only one PLD isoform. These results revealed, for the first time, redundant roles of PLD1 and PLD2 in platelet alpha-granule secretion and indicate that this may be relevant for pathological thrombus formation. Thus, PLD might represent a promising target for antithrombotic therapy.
Thus, this hypothesis was tested more directly in the second part of this thesis. The effects of pharmacological inhibition of PLD activity on hemostasis, thrombosis and thrombo-inflammatory brain infarction in mice were assessed. Treatment of platelets with the reversible, small molecule PLD inhibitor 5-Fluoro-2-indolyl des-chlorohalopemide (FIPI) led to a specific blockade of PLD activity that was associated with reduced -granule release and integrin activation. Mice that received FIPI at a dose of 3 mg/kg displayed reduced occlusive thrombus formation upon chemical injury of carotid arteries or mesenterial arterioles. Similarly, FIPI-treated mice had smaller infarct sizes and significantly better motor and neurological function 24 hours after transient middle cerebral artery occlusion. This protective effect was not associated with major intracerebral hemorrhage or prolonged tail bleeding times. Thus, pharmacological PLD inhibition might represent a safe therapeutic strategy to prevent arterial thrombosis or ischemic stroke.
After revealing a central role for PLD in thrombo-inflammation, the regulation of PLD activity in platelets was analyzed in the last part of the thesis. Up to date, most studies made use of inhibitors potentially exerting off-target effects and consequently PLD regulation is discussed controversially. Therefore, PLD activity in mice genetically lacking potential modulators of PLD activity was determined to address these controversies. These studies revealed that PLD is tightly regulated during initial platelet activation. While integrin outside-in signaling and Gi signaling was dispensable for PLD activation, it was found that PLC dependent pathways were relevant for the regulation of PLD enzyme activity.
The skeleton is a preferred homing site for breast cancer metastasis. To date, treatment options for patients with bone metastases are mostly palliative and the disease is still incurable. Indeed, key mechanisms involved in breast cancer osteotropism are still only partially understood due to the lack of suitable animal models to mimic metastasis of human tumor cells to a human bone microenvironment. In the presented study, we investigate the use of a human tissue-engineered bone construct to develop a humanized xenograft model of breast cancer-induced bone metastasis in a murine host. Primary human osteoblastic cell-seeded melt electrospun scaffolds in combination with recombinant human bone morphogenetic protein 7 were implanted subcutaneously in non-obese diabetic/severe combined immunodeficient mice. The tissue-engineered constructs led to the formation of a morphologically intact 'organ' bone incorporating a high amount of mineralized tissue, live osteocytes and bone marrow spaces. The newly formed bone was largely humanized, as indicated by the incorporation of human bone cells and human-derived matrix proteins. After intracardiac injection, the dissemination of luciferase-expressing human breast cancer cell lines to the humanized bone ossicles was detected by bioluminescent imaging. Histological analysis revealed the presence of metastases with clear osteolysis in the newly formed bone. Thus, human tissue-engineered bone constructs can be applied efficiently as a target tissue for human breast cancer cells injected into the blood circulation and replicate the osteolytic phenotype associated with breast cancer-induced bone lesions. In conclusion, we have developed an appropriate model for investigation of species-specific mechanisms of human breast cancer-related bone metastasis in vivo.
Background: A growing number of non-coding regulatory mutations are being identified in congenital disease. Very recently also some exons of protein coding genes have been identified to act as tissue specific enhancer elements and were therefore termed exonic enhancers or "eExons".
Methods: We screened a cohort of 134 unrelated families with split-hand/split-foot malformation (SHFM) with high resolution array CGH for CNVs with regulatory potential.
Results: In three families with an autosomal dominant non-syndromic SHFM phenotype we detected microdeletions encompassing the exonic enhancer (eExons) 15 and 17 of DYNC1I1. In a fourth family, who had hearing loss in addition to SHFM, we found a larger deletion of 510 kb including the eExons of DYNC1I1 and, in addition, the human brain enhancer hs1642. Exons 15 and 17 of DYNC1I1 are known to act as tissue specific limb enhancers of DLX5/6, two genes that have been shown to be associated with SHFM in mice. In our cohort of 134 unrelated families with SHFM, deletions of the eExons of DYNC1I1 account for approximately 3% of the cases, while 17p13.3 duplications were identified in 13% of the families, 10q24 duplications in 12%, and TP63 mutations were detected in 4%.
Conclusions: We reduce the minimal critical region for SHFM1 to 78 kb. Hearing loss, however, appears to be associated with deletions of a more telomeric region encompassing the brain enhancer element hs1642. Thus, SHFM1 as well as hearing loss at the same locus are caused by deletion of regulatory elements. Deletions of the exons with regulatory potential of DYNC1I1 are an example of the emerging role of exonic enhancer elements and their implications in congenital malformation syndromes.
Herstellung und Charakterisierung spintronischer und caloritronischer (Ga,Mn)As-Nanostrukturen

Die elektronischen Bauteile, die aus unserer technischen Welt kaum wegzuddenken sind, werden immer kleiner. Aktuelle ICs bestehen zum Beispiel aus Milliarden von Transistoren, von denen jeder einzelne kleiner als 100nm (dem 100-stel des typischen Durchmessers eines Menschenhaars) ist. Dass die Entwicklung auch zukünftig weiter dem Trend des Mooreschen Gesetzes folgen wird, gilt hierbei als unbestritten.
Die interessanteste Fragestellung der Halbleiter- und Nanostrukturforschung in diesem Zusammenhang ist: Kann man die weitere Entwicklung der Informations- und Kommunikationstechnik dadurch erreichen, dass man die Miniaturisierung der Transistoren in Mikroprozessoren und Speicherbauelementen weiter vorantreibt oder ist man auf gänzlich neue Wege angewiesen?
Bei der weitergehenden Miniaturisierung ist die größte Hürde darin zu suchen, ob man in der
Lage sein wird die Verbrauchsleistung dieser Bauelemente weiter zu reduzieren, um die Überhitzung der Bauteile in den Griff zu bekommen und nicht zuletzt auch, um Energie zu sparen.
Die heutige Elektronik hat ihre Grundlagen in den 60er Jahren. Diese Art der Elektronik ist jedoch hinsichtlich der Effizienzsteigerungen und vor allem der Wärmeentwicklung an ihre Grenzen gestoßen. Hauptursache für diese problematische Wärmeentwicklung sind die elektrischen Verbindungen, die die Informationen zwischen der halbleiterbasierten Datenverarbeitung und den metallischen Speicherelementen hin und hertransportieren. Obwohl diese elektrischen Verbindungen zum aktuellen Zeitpunkt aus der Computerarchitektur nicht weg zu denken sind, ist es eines der Hauptziele diese Verbindungen nicht mehr verwenden zu müssen. Dies kann jedoch nur erreicht werden, wenn es gelingt, die Speicherelemente und Datenverarbeitung in einem einzigen Element (Halbleiter) zu vereinen.
Bisher wurde die Ladung eines Elektrons für die Verarbeitung von elektrischen Informationen
bzw. Zuständen benutzt. Was wäre jedoch, wenn man diese bisherige Basis völlig ändert? Der
Spin der Elektronen ist ein viel effektiverer Informationsträger als die Ladung der Elektronen
selbst, nicht zuletzt deshalb, weil die Veränderung des Spins eines Elektrons im Vergleich zu
dessen Bewegung einen weitaus geringeren Energiebetrag benötigt [1]. Die Technik, die zusätzlich zur Informationsverarbeitung durch makroskopische Elektronenströme den viel effektiveren Spin-Quantenzustand der Elektronen oder Löcher als Freiheitsgrad nutzt, ist die sogenannte Spintronik1. Die Spinfreiheitsgrade eröffnen, wegen der längeren Phasenkohärenzlänge, im Vergleich zu den orbitalen Freiheitsgraden, völlig neue Wege für zukünftige Entwicklungen wie z.B. den Quantencomputer. Damit wäre die Entwicklung niederenergetischer Bauelemente möglich, die fast keine Wärmeentwicklung aufweisen. Wegen dieser vielen Vorteile hat sich die Spintronik in Rekordzeit von einer interessanten wissenschaftlichen Beobachtung in Rekordzeit zu einer marktbewegenden Anwendung weiterentwickelt (Nobelpreis 2007). Seinen Anfang nahm diese Entwicklung 1988 mit der Entdeckung des GMR-Effekts. Nach nur 9 Jahren wurden 1997 erste Festplatten-Leseköpfe eingesetzt, die sich diesen Effekt zu Nutze machten. Leseköpfe, die den Riesenmagnetwiderstand nutzen, waren nunmehr um ein Vielfaches empfindlicher als es die konventionelle Technik zugelassen hätte. Die Speicherdichte und damit die Kapazitäten der Festplatten konnte somit erheblich gesteigert und Festplatten mit zuvor nie gekannter Speicherkapazität preiswert produziert werden. Seit dieser Zeit rückt der Elektronenspin immer weiter in den Brennpunkt von Forschung und Entwicklung.
Da sich der elektrische Widerstand von Halbleitern in einem weiten Bereich manipulieren lässt
(was für ferromagnetische Metalle nicht der Fall ist), werden logische Bauelemente aus halbleitenden Materialien hergestellt. Im Gegensatz dazu sind ferromagnetische Metalle sehr gute Kandidaten für die Speicherung von Informationen. Dies liegt vor allem daran, dass zufällige Magnetfelder viel schwächer sind, als zufällige elektrische Felder, was ferromagnetische Systeme wesentlich unanfälliger macht. Daher sind die magnetischen Speicher nicht flüchtig und zudem müssen deren Informationsgehalte nicht wie bei DRAM immer wieder aufgefrischt werden. Um die jeweiligen Vorteile der Materialklassen – die magnetisch energiesparende sowie dauerhafte Speicherfähigkeit der Metalle und die logischen Operationen der Halbleiter – miteinander kombinieren zu können und damit neuartige Bauelemente wie z.B. MRAMs (logische Operationen und dauerhafte Speicherung) zu bauen, sind ferromagnetischen Halbleiter unverzichtbar. Auf dieser Basis könnten Speicherelemente und Datenverarbeitung in einem einzigen Element (Halbleiter) dargestellt werden. Zugleich braucht man aber auch neue Wege, um diese Speicher zu magnetisieren und später auslesen zu können. Ein weiterer Vorteil liegt zudem darin, dass hierzu kein Einsatz beweglicher Teile notwendig ist. Die Magnetisierungskontrolle muss aber temperaturunabhängig sein!
Der am besten erforschte ferromagnetische Halbleiter ist (Ga,Mn)As, der deswegen die Modellrolle einnimmt und als Prototyp für alle ferromagnetischen Halbleiter dient. Die Kopplung seiner magnetischen und halbleitenden Eigenschaften durch Spin-Bahn-Wechselwirkung ist die Ursache vieler neuer Transportphänomene in diesem Materialsystem. Diese Phänomene sind vielfach die Grundlage für neuartige Anwendungen, Bauteildesigns und Wirkprinzipien.
Das Ziel dieser Arbeit ist es, die interessanten Anisotropien in (Ga,Mn)As, die von der sehr starken Spin-Bahn-Kopplung im Valenzband herrühren zu nutzen, sowie neue spinbezogene Effekte in verschiedenen magnetischen Bauelementen zu realisieren.
Die vorliegende Arbeit gliedert sich wie folgt: In Kapitel 1 wird auf die grundlegenden Eigenschaften des (Ga,Mn)As und einige neuartige Spineffekten, die dieses Material mit sich bringt, eingegangen. Das zur Erzeugung dieser Effekte notwendige fertigungstechnische Wissen, für die lithografische Erzeugung der spintronisch bzw. caloritronisch aktiven Nanostrukturen, wird im Kapitel 2 beschrieben.
Um mit dieser Welt der Spineffekte „kommunizieren“ und die Effekte kontrollieren zu können,
sind entsprechend angepasste und funktionsfähige Kontaktierungen notwendig. Mit der detaillierten Herstellung und Analyse dieser Kontakte beschäftigt sich das Kapitel 3. Es wurden
zwei Arten von Kontakten hergestellt und bei den Proben eingesetzt: in situ (innerhalb der
MBE-Wachstumskammer) und ex situ. Zusammenfassend lässt sich sagen, dass bei der ex situ-Probenpräparation, die Reproduzierbarkeit der Kontakte, besonders bei logisch magnetischen Elementen, nicht gewährleistet werden konnte. Bei funktionierender Kontaktierung war das magnetische Verhalten dann jedoch stets gleich. Bei den in situ-Kontakten war zwar einerseits das elektrische Verhalten reproduzierbar und sehr gut, aber das magnetische Verhalten war nicht zufriedenstellend, da die Relaxation nicht vollständig stattfand.
Im Rahmen dieser Arbeit konnten die ex situ-Kontakte optimiert werden. Dabei wurde auf die
Problematiken bereits existierender Proben eingegangen und es wurden verschiedene Lösungsan sätze dafür gefunden. So konnte z.B. gezeigt werden, dass die Haftungsprobleme hauptsächlich auf dem unsaubere Oberflächen zurückzuführen sind. Jede Schicht, die zwischen aufgedampfter Metallschicht und dem dotierten Halbleiter bestehen bleibt, unabhängig davon, ob es sich dabei um eine oxidierte Schicht, Lackreste oder eine, zum Teil verarmte Schicht handelt, beeinträchtigt die Funktionalität der Kontakte. Je kleiner die Dimension der Kontakte, desto stärker wirkt sich die unsaubere Oberfläche aus. So konnte gezeigt werden, dass ab einer Größe von ca. 500nm_500nm die Zuverlässigkeit der Kontakte elementar von der Reinheit der Oberflächen und deren Homogenität beeinflusst wird. Zur Abwendung dieser Komplikationen werden verschiedene Lösungsansätze vorgeschlagen. Wird die Oberfläche mit hochenergetischen Ionen versetzt, verarmt deren Dotierung, was zu einer massiven Änderung der Leitfähigkeit führt. Daher wurden entweder völlig andere Prozessparameter zur Reinigung eingesetzt, die den dotierten HL nicht verarmen oder einer der nasschemischen Schritte wurde so angepasst, dass die extrem verarmte Schicht der HL-Oberfläche entfernt wurde.
Die einfachsten spintronischen Bauelemente (Streifen) und magnetischen Logikelemente sowie
deren Ergebnisse werden im Kapitel 4 diskutiert.
Hier wurde eindeutig gezeigt, dass die Streifen bei niedrigen Stromdichten nicht völlig uniaxial
sind, während bei erhöhten Stromdichten die Uniaxialität immer dominanter wird. Dies war
jedoch zu erwarten, da bei erhöhten Stromdichten die Temperatur auch ansteigt und da, bei
erhöhter Temperatur, die biaxiale Anisotropie mit M4, die uniaxiale aber jedoch nur mit M2
abfällt – die dominante Anisotropie wechselt folglich von biaxial zu uniaxial [2]. Im Rahmen dieser Arbeit wurden die Grundlagen gelegt, um Speicherelemente und Datenverarbeitung in einem einzigen Halbleiter (Ga,Mn)As herzustellen. Auf Basis dieser Arbeit und den dabei gewonnenen litographischen Erkentnissen wurden, in nachfolgenden Arbeiten, solche Bauelemente realisiert [3].
Spin-Kaloritronik:
Wie schon Eingangs erwähnt, wird im Allgemeinen davon ausgegangen, dass die Miniaturisierung der zukünftigen Elektronik weitergeführt werden kann. Bei stetiger Verkleinerung der Strukturen kommt es in heutigen Anwendungen zu immer größeren Problemen bei der Wärmeabfuhr. Die Folgen der Temperaturdifferenzen innerhalb der Strukturen führen dabei zu sog. Hotspots oder sogar Materialschäden. Temperaturunterschiede müssen aber nicht nur negative Auswirkungen
haben. So wurde an einem ferromagnetischen System aus Nickel, Eisen und Platin der sogenannte Spin-Seebeck-Effekt gemessen, bei dem die Elektronen in den Regionen verschiedener Temperatur unterschiedliche Spinpolarisationen zeigen [4].
Eine Batterie, die diesen spinpolarisierten Strom nutzt, könnte einen entscheidenden Fortschritt
in der Spintronik bedeuten. Dieser Bereich der Forschung an thermoelektrischen Effekten, bei
denen ferromagnetische Materialien involviert sind, wird auch „spin-caloritronics“ genannt [5].
Die Kapitel 5 und 6 beschäftigen sich mit einer neuartigen Klasse spintronischer Bauteile. whärend das Kapitel 5 sich mit einer neuartigen Klasse spintronischer Bauteile, für die von uns
als Bezeichnung TAMT („tunnel anisotropic magneto thermopower“) eingeführt wurde, beschäftigt, wird in Kapitel 6 an einem veränderten Probenlayout der Nernst-Effekt nachgewiesen.
Die Geometrie wurde in beiden fällen so gewählt und hergestellt, dass durch die Anisotropien
des (Ga,Mn)As die beiden thermoelektrische Effekte (Seebeck- und Nernst-Effekt) auf einen
n+-p+-Übergang übertragen werden konnten. Durch einen Strom, in einem mit Silizium hoch
dotierten GaAs-Heizkanal, kann jeweils ein vertikaler Temperaturgradient erzeugt werden. Die
hierbei entstehenden Thermospannungen wurden durch eine vollständige elektrische Charaktri sierungsmessung mit Hilfe präziser Lock-in-Verstärker-Technik detektiert.
Das Kapitel 5 beschäftigt sich mit allen Bereichen, von der Idee bis hin zu Messungen und Analysen des Seebeck-Effektes an einem n-p-Übergang (TAMT). Außerdem ist ein sehr einfaches numerisches Modell dargestellt, dass den gefundenen Effekt theoretisch beschreibt.
Durch die bekannten thermoelektrischen Effekte ergibt sich ein Temperaturgradient der immer
zu einer Thermospannung und somit zu einem Thermostrom entlang des Gradienten führt. Für
zukünftige Entwicklungen ist es demnach wichtig, diese Effekte zu beachten und diese bei elektrischen Messungen an Nanostrukturen als mögliche, zusätzliche Ursache eines Messsignals in Betracht zu ziehen.
In den vorliegenden Proben ist der Seebeck-Effekt stark anisotrop, mit einem größeren Thermospannungswert für Magnetisierungen entlang der magnetisch harten Achsen des (Ga,Mn)As.
Es wurde ein einfaches Model entwickelt, welches das Tunneln von Elektronen zwischen zwei
unterschiedlich warmen Bereichen erklärt. Die Abhängigkeit des Effekts von der Temperatur des Heizkanals wurde anhand dieses Models sowohl qualitativ als auch größenordnungsmäßig korrekt beschrieben.
Die Nernst-Proben wurden von der Theorie bis zur Herstellung so entwickelt, dass in derselben
Anordnung eine im (Ga,Mn)As senkrecht zum Temperaturgradienten gerichtete Spannung
zusätzlich gemessen werden konnte. Diese wurde durch den Nernst-Effekt erklärt. Besonders interessant war, dass die Größe der Nernst-Spannung hierbei mit der Magnetisierung im (Ga,Mn)As verknüpft ist und somit ein aus der typischen Magnetisierungsumkehr hervorgehendes Verhalten zeigt.
Gegenüber den Magnetowiderstandseffekten entsteht beim Nernst-Effekt in sogenannten Fingerprints (vgl. Kapitel 1.3.3) ein dreistufiges Farbmuster anstelle eines zweistufigen hoch-tief-Systems. Die entstehende Temperatur im Heizkanal wird jeweils durch eine longitudinale Widerstandsmessung in einem senkrecht zum Kanal gerichteten äußeren Magnetfeld bestimmt. Die Magnetfeldabhängigkeit des Widerstands kommt hierbei durch den Effekt der schwachen Lokalisierung in dünnen Filmen zustande.
Zusammenfassend stellen die Magneto-Thermoelektrizitätseffekte eine wichtige weitere Transporteigenschaft in ferromagnetischen Halbleitern dar, die mit der Magnetisierung direkt zusammenhängen.
In dieser Arbeit wurden Thermospannungen an (Ga,Mn)As-Schichten mit vergleichsweise hoher
Mangankonzentration untersucht. Allerdings sind die Thermoelektrizitätseigenschaften zusammen mit Magneto-Widerstandsmessungen in Zukunft in der Lage, zusätzliche Informationen über die Bandstruktur sowie die Ladungsträgereigenschaften in Materialsystemen mit niedrigerem Mangangehalt, insbesondere in der Nähe des Metall-Isolator-Übergangs, zu liefern.
Inhalt des Anhangs ist eine ausführliche Anleitung zur Optimierung der Probenherstellung bzw.
der verschiedenen Bauelemente.
In May 2014, six patients presented in Germany with a Sarcocystis-associated febrile myositis syndrome after returning from Tioman Island, Malaysia. During two earlier waves of infections, in 2011 and 2012, about 100 travellers returning to various European countries from the island were affected. While the first two waves were associated with travel to Tioman Island mostly during the summer months, this current series of infections is associated with travel in early spring, possibly indicating an upcoming new epidemic.
Leptoquarks are hypothetical particles that attempt to explain the coincidental similarities between leptons and quarks included in SM. Their exact properties vary between different theoretical models, and there are no strong theoretical constraints on their possible mass values. They can possibly be produced from particle
collisions, and there have already been searching efforts at previous collider experiments. Their presence have yet been observed, and this fact has been translated into lower bound exclusions on their possible mass values. The Large Hadron Collider (LHC) being the most recently constructed particle collider with the highest collision energies ever achieved experimentally, provides a new platform to continue the search for Leptoquarks at even higher mass ranges.
This thesis describes a search for pair-produced second-generation Leptoquarks using 20.3 fb−1 of data recorded by the ATLAS detector of LHC at √s = 8 TeV. Events with two oppositely charged muons and two or more jets in the final state were used. Candidate leptoquark events were selected with the help of four observables: the di-muon invariant mass (Mμμ ), the sum of the pT of the two muons
(LT ), the sum of the pT of the two leading jets (HT ) and the average Leptoquark mass (MLQ ). Monte Carlo simulations of SM background processes have shown
to be in good agreement with data, both in the region constructed using selection requirements for candiate leptoquark events and in the designated control regions.
Since no significant excess of events was observed in data, a exclusion limit was set as a function of the Leptoquark mass.
Background: Gametogenesis and fertilization play crucial roles in malaria transmission. While male gametes are thought to be amongst the simplest eukaryotic cells and are proven targets of transmission blocking immunity, little is known about their molecular organization. For example, the pathway of energy metabolism that power motility, a feature that facilitates gamete encounter and fertilization, is unknown.
Methods: Plasmodium berghei microgametes were purified and analysed by whole-cell proteomic analysis for the first time. Data are available via ProteomeXchange with identifier PXD001163.
Results: 615 proteins were recovered, they included all male gamete proteins described thus far. Amongst them were the 11 enzymes of the glycolytic pathway. The hexose transporter was localized to the gamete plasma membrane and it was shown that microgamete motility can be suppressed effectively by inhibitors of this transporter and of the glycolytic pathway.
Conclusions: This study describes the first whole-cell proteomic analysis of the malaria male gamete. It identifies glycolysis as the likely exclusive source of energy for flagellar beat, and provides new insights in original features of Plasmodium flagellar organization.
Respiratory syncytial virus (RSV) is the leading cause of hospitalization especially in young children with respiratory tract infections (RTI). Patterns of circulating RSV genotypes can provide a better understanding of the molecular epidemiology of RSV infection. We retrospectively analyzed the genetic diversity of RSV infection in hospitalized children with acute RTI admitted to University Hospital Heidelberg/Germany between October 2012 and April 2013. Nasopharyngeal aspirates (NPA) were routinely obtained in 240 children younger than 2 years of age who presented with clinical symptoms of upper or lower RTI. We analyzed NPAs via PCR and sequence analysis of the second variable region of the RSV G gene coding for the attachment glycoprotein. We obtained medical records reviewing routine clinical data. RSV was detected in 134/240 children. In RSV-positive patients the most common diagnosis was bronchitis/bronchiolitis (75.4%). The mean duration of hospitalization was longer in RSV-positive compared to RSV-negative patients (3.5 vs. 5.1 days; p < 0.01). RSV-A was detected in 82.1%, RSV-B in 17.9% of all samples. Phylogenetic analysis of 112 isolates revealed that the majority of RSV-A strains (65%) belonged to the novel ON1 genotype containing a 72-nucleotide duplication. However, genotype ON1 was not associated with a more severe course of illness when taking basic clinical/laboratory parameters into account. Molecular characterization of RSV confirms the co-circulation of multiple genotypes of subtype RSV-A and RSV-B. The duplication in the G gene of genotype ON1 might have an effect on the rapid spread of this emerging RSV strain.
DNA binding properties of human Cdc45 suggest a function as molecular wedge for DNA unwinding

The cell division cycle protein 45 (Cdc45) represents an essential replication factor that, together with the Mcm2-7 complex and the four subunits of GINS, forms the replicative DNA helicase in eukaryotes. Recombinant human Cdc45 (hCdc45) was structurally characterized and its DNA-binding properties were determined. Synchrotron radiation circular dichroism spectroscopy, dynamic light scattering, small-angle X-ray scattering and atomic force microscopy revealed that hCdc45 exists as an alpha-helical monomer and possesses a structure similar to its bacterial homolog RecJ. hCdc45 bound long (113-mer or 80-mer) single-stranded DNA fragments with a higher affinity than shorter ones (34-mer). hCdc45 displayed a preference for 3' protruding strands and bound tightly to single-strand/double-strand DNA junctions, such as those presented by Y-shaped DNA, bubbles and displacement loops, all of which appear transiently during the initiation of DNA replication. Collectively, our findings suggest that hCdc45 not only binds to but also slides on DNA with a 3'-5' polarity and, thereby acts as a molecular 'wedge' to initiate DNA strand displacement.
Object six Degrees of Freedom (6DOF) pose estimation is a fundamental problem in many practical robotic applications, where the target or an obstacle with a simple or complex shape can move fast in cluttered environments. In this thesis, a 6DOF pose estimation algorithm is developed based on the fused data from a time-of-flight camera and a color camera. The algorithm is divided into two stages, an annealed particle filter based coarse pose estimation stage and a gradient decent based accurate pose optimization stage. In the first stage, each particle is evaluated with sparse representation. In this stage, the large inter-frame motion of the target can be well handled. In the second stage, the range data based conventional Iterative Closest Point is extended by incorporating the target appearance information and used for calculating the accurate pose by refining the coarse estimate from the first stage. For dealing with significant illumination variations during the tracking, spherical harmonic illumination modeling is investigated and integrated into both stages. The robustness and accuracy of the proposed algorithm are demonstrated through experiments on various objects in both indoor and outdoor environments. Moreover, real-time performance can be achieved with graphics processing unit acceleration.
Adenosinrezeptoren werden auf nahezu allen Körperzellen exprimiert und übernehmen dort vielfältige und wichtige Funktionen. Auch auf diversen Tumorzelllinien konnten bereits Adenosinrezeptoren nachgewiesen und – je nach Subtyp – mit Pro- oder Anti-tumor-Effekten in Zusammenhang gebracht werden.
In dieser Arbeit wurden Gebärmutterhalskrebszellen sowie endometriale und triple-negative Brustkrebszellen auf Expression und mögliche Funktionen von Adenosinrezep-toren untersucht. Da spezifische Antikörper bis heute nicht verfügbar sind, wurde ein pharmakologischer Ansatz mit subtypspezifischen Agonisten und Antagonisten gewählt.
In Radioliganden-Bindungsassays, konnte nachgewiesen werden, dass sich auf der Zer-vixkarzinom-Zelllinie SiHa und der Brustkrebs-Zelllinie HCC1806 Adenosinrezeptoren des Subtyps A1 befinden. Die endometrialen Krebszelllinien Ishikawa und HEC-1-A exprimieren Rezeptoren vom Subtyp A1 und A2A. A3-Adenosinrezeptoren wurden auf keiner der untersuchten Zelllinien gefunden.
Der Nachweis von A2B-Rezeptoren kann mit dem Radioliganden-Bindungsassay nicht erbracht werden, da bislang kein Radioligand bekannt ist, der eine ausreichende Affini-tät besitzt, um diesen Subtyp zweifelsfrei nachweisen zu können.
Obwohl die Mehrheit der untersuchten Zelllinien Adenosinrezeptoren exprimiert, konnte ein signifikanter Effekt auf die Adenylatcyclase bei Stimulation der auf den Zellen vorhandenen Adenosinrezeptoren nur bei den HEC-1-A-Zellen festgestellt werden. Auch auf funktionelle A2B-Rezeptoren fand sich im Adenylatcyclaseassy kein Hinweis.
Im durchgeführten Kristallviolettassay zeigte sich ein proapoptotischer Effekt auf Ishi-kawa- und HEC-1-A-Zellen bei hohen Adenosin-Konzentrationen (100 µM). Die im BrdU-Assay gemessene Proliferationsrate hingegen änderte sich nach Vorbehandlung mit Adenosin nicht. Das metabolisch stabilere NECA (in Kombination mit ADA) hatte im Kristallviolettassay einen stärkeren Einfluss auf die Apoptoserate der jeweiligen Zelllinie als Adenosin und auch im BrdU-Assay sank die Menge an inkorporiertem BrdU. Ein Synergismus zwischen Stimulation von Adenosinrezeptoren und diversen Todesliganden bzw. Chemotherapeutika konnte nicht nachgewiesen werden.
Freies extrazelluläres Adenosin kann auch aus dem Abbau von ATP generiert werden, wenn Zellen die Ektonukleotidasen CD39 und CD73 exprimieren. Aufgrund der im-munsuppressiven Wirkung von Adenosin können diese Enzyme T-Zell- und NK-Zellantworten im Mikromilieu von Tumoren hemmen. Die durchflusszytometrische Analyse von HEC-1-A- und Ishikawa-Zellen zeigte zwar, dass die Expression von CD39 und CD73 nach Stimulation der Adenosinrezeptoren unverändert blieb. Die Ex-pression von Enzymen, lässt aber vermuten, dass die Zellen in vivo von Adenosin profi-tieren könnten. Angesichts der in vitro Daten, die allenfalls einen wachstumshemmen-den Effekt von Adenosin zeigten, könnte die vorrangige Wirkung von Adenosin im Tumormikromilieu tatsächlich auf der Inhibition von Immunantworten beruhen. Mög-licherweise würden die Rezeptoren dann in erster Linie als Sensoren dienen.
Weitere Forschungsarbeit wird helfen, die Rolle der Adenosinrezeptoren im Tumorge-schehen vollständig zu verstehen und möglicherweise für die Krebstherapie nutzbar zu machen.
In this work, high-energy observables arising during different phases of SN explosions are studied with respect to their potential for allowing conclusions on suggested explosion scenarios and physical mechanisms that are thought to influence the evolution of SNe in a major way. The focus on selected observables at keV and MeV energies is motivated by the appearance of large degeneracies that can even be found for disparate scenarios in many wavelength regimes. Since the discussed emission in the high-energy regime is directly linked to nuclear processes being usually very distinct for different suggested physical models, the signatures at keV and MeV energies allow for meaningful comparisons of simulations with observations.
In Yaşar Kemal‘s The Pomegranate on the Knoll, the environment of the Çukurova peasants is depicted meticulously in their daily struggle for survival; however, Kemal transcends purely realist representations whenever the characters try either to make sense of or to gain control over their hostile environment. By using magical realist strategies in his text as well, he gives a voice to the suffering of the Çukurova peasants and engages in a form of political writing different from social realism or village literature. He helps to raise public awareness for the traumatic consequences of the changing social reality in rural Turkey not only by describing it, but also by using magical realist writing strategies which let readers feel it for themselves.
Endurance athletes integrate four conditioning concepts in their training programs: high-volume training (HVT), “threshold-training” (THR), high-intensity interval training (HIIT) and a combination of these aforementioned concepts known as polarized training (POL). The purpose of this study was to explore which of these four training concepts provides the greatest response on key components of endurance performance in well-trained endurance athletes.
Methods: Forty eight runners, cyclists, triathletes, and cross-country skiers (peak oxygen uptake: (VO2peak): 62.6 ± 7.1 mL·min−1·kg−1) were randomly assigned to one of four groups performing over 9 weeks. An incremental test, work economy and a VO2peak tests were performed. Training intensity was heart rate controlled.
Results: POL demonstrated the greatest increase in VO2peak (+6.8 ml·min·kg−1 or 11.7%, P < 0.001), time to exhaustion during the ramp protocol (+17.4%, P < 0.001) and peak velocity/power (+5.1%, P < 0.01). Velocity/power at 4 mmol·L−1 increased after POL (+8.1%, P < 0.01) and HIIT (+5.6%, P < 0.05). No differences in pre- to post-changes of work economy were found between the groups. Body mass was reduced by 3.7% (P < 0.001) following HIIT, with no changes in the other groups. With the exception of slight improvements in work economy in THR, both HVT and THR had no further effects on measured variables of endurance performance (P > 0.05).
Conclusion: POL resulted in the greatest improvements in most key variables of endurance performance in well-trained endurance athletes. THR or HVT did not lead to further improvements in performance related variables.
The dissertation at hand focuses on the enforcement of accounting standards in Germany. The legal basis of the external enforcement of accounting standards in Germany was created by the „Bilanzkontrollgesetz” (Financial Reporting Enforcement Act) at the end of 2004. An enforcement mechanism was installed to enforce accounting standard compliance by regular reviews of disclosed financial statements. The system was established as implementation of EU guidelines. Since 2005, International Financial Reporting Standards (IFRS) shall be applied for consolidated financial statement of firms listed on a regulated market segment within the European Union (EU) (Regulation EC No. 1606/2002). Simultaneously to the harmonization of accounting standards, the EU fostered the standardization of enforcement systems to ensure compliance with international accounting standards. Par. 16 of the so-called “IAS Regulation” mandates the “Committee of European Securities Regulators” (CESR) to “develop a common approach to enforcement". Germany’s unique two-tiered system operates since July 2005; it involves the “Deutsche Pruefstelle fuer Rechnungslegung” (Financial Reporting Enforcement Panel), a newly established private organization primarily assigned to conduct the reviews. As the second tier, the „Bundesanstalt fuer Finanzdienstleistungsaufsicht” (Federal Financial Supervisory Authority) has the sovereign authority to order the publication of errors („error announcements“) and if necessary, to force the cooperation of firms in the review process.
The dissertation is structured as follows. A general introduction focuses on the theoretical background and the reasoning for the need of external enforcement mechanisms. The common approach to enforcement in the European Union is described. Building on this, the thesis consists of three individual essays that analyze three specific questions in the context of the enforcement of financial reporting standards in Germany.
The first paper focuses on the systematical evaluation of the information contained in 100 selected error announcements (from a total population of 151 evaluable announcements). The study finds that error announcements on average contain 3.64 single errors and 77% affect the reported profit. Relatively small as well as big, highly levered and rather unprofitable firms are overrepresented in the sample of misstatement firms. In a second step, the essay investigates the development of censured firms over time; the pre- and post-misstatement development of the firms in terms of balance sheet data, financial ratios and (real) earnings management are tracked. The analysis detects increasing leverage ratios and a decline in profitability over time. In the year of misstatement firms report large total and discretionary accruals, indicating earnings management. Compared to matched control firms, significant differences in profitability, market valuation, earnings management and real activities manipulations are observable. A major contribution of this first study is the examination of trends in financial data and (real) earnings management over a number of years surrounding misstatements as well as the elaboration of the distinction to non-misstating firms. The results show the meaning of the enforcement of IFRS for the quality of financial reporting to standard setters, policy makers, and investors in Germany.
The second paper examines the interrelation of enforcement releases, firm characteristics and earnings quality. Prior literature documents the correlation between underperformance in financial ratios and the probability of erroneous disclosure of financial statements; this study provides evidence for differences in characteristics between firms with enforcement releases and control firms as well as a broad sample of German publicly traded firms (4,730 firm-year observations). Furthermore, research affirms the connection of financial ratios to earnings quality metrics. The accuracy of financial information is considered to be correlated with its quality and therefore the differences in earnings quality between various sub-samples is examined. Overall, the results document the underperformance in important financial ratios as well as indicate an inferior earnings quality of firms subject to enforcement releases vis-a-vis the control groups. These results hold with regard to both different earnings quality specifications and different periods observed. This study appends the earnings quality discussion and contributes to develop a comprehensive picture of accounting quality for the unique institutional settings of Germany. The paper shows that a conjoint two-tier public and private enforcement system is effective and might be an adequate model for other countries. Implications for the regulation of corporate governance, the enforcement panel and the auditor are identified.
The third essay additionally considers the role of the auditor. The firms subject to error announcements are used to evaluate the consequences of increasing earnings management over time on enforcement releases and their recognition in audit fees. Prior literature provides evidence on a phenomenon called „balance sheet bloat” that is due to income increasing earnings management and later influences the disclosure of misstated financial statements. The evidence of earnings management recognition in audit fees and findings on the content of future information in audit fees leads to the hypothesis that auditors recognize increasing audit risk in fees before the enforcement process starts. The study extends related earnings management and audit fee literature by modeling the development of earnings management within the misstatement firms and systematically link it to auditor reactions. Significant predictive power of different commonly used accrual measures for enforcement releases in the period prior and up to the misstatement period are found by the study. In the same period of time an increase in audit fees, e.g. the recognition of increased audit risk, can be observed. A possible audit fee effect after the misstatement period is investigated, but no significant relation is obtained.
The dissertation closes with a summary of the main findings, a conclusion to the connection of the three essays as well as subsumption of findings in the accounting literature.
The majority of patients with acute myeloid leukemia will relapse, and older patients often fail to achieve remission with induction chemotherapy. We explored the possibility that leukemic suppression of innate immunity might contribute to treatment failure. Natural killer cell phenotype and function was measured in 32 consecutive acute myeloid leukemia patients at presentation, including 12 achieving complete remission. Compared to 15 healthy age-matched controls, natural killer cells from acute myeloid leukemia patients were abnormal at presentation, with downregulation of the activating receptor NKp46 (P=0.007) and upregulation of the inhibitory receptor NKG2A (P=0.04). Natural killer cells from acute myeloid leukemia patients had impaired effector function against autologous blasts and K562 targets, with significantly reduced CD107a degranulation, TNF-alpha and IFN-gamma production. Failure to achieve remission was associated with NKG2A overexpression and reduced TNF-alpha production. These phenotypic and functional abnormalities were partially restored in the 12 patients achieving remission. In vitro co-incubation of acute myeloid leukemia blasts with natural killer cells from healthy donors induced significant impairment in natural killer cell TNF-alpha and IFN-gamma production (P=0.02 and P=0.01, respectively) against K562 targets and a trend to reduced CD107a degranulation (P=0.07). Under transwell conditions, the inhibitory effect of AML blasts on NK cytotoxicity and effector function was still present, and this inhibitory effect was primarily mediated by IL-10. These results suggest that acute myeloid leukemia blasts induce long-lasting changes in natural killer cells, impairing their effector function and reducing the competence of the innate immune system, favoring leukemia survival.
Serotonin receptor 1A gene (HTR1A) knockout mice show pronounced defensive behaviour and increased fear conditioning to ambiguous conditioned stimuli. Such behaviour is a hallmark of pathological human anxiety, as observed in panic disorder with agoraphobia (PD/AG). Thus, variations in HTR1A might contribute to neurophysiological differences within subgroups of PD/AG patients. Here, we tested this hypothesis by combining genetic with behavioural techniques and neuroimaging. In a clinical multicentre trial, patients with PD/AG received 12 sessions of manualized cognitive-behavioural therapy (CBT) and were genotyped for HTR1A rs6295. In four subsamples of this multicentre trial, exposure behaviour (n = 185), defensive reactivity measured using a behavioural avoidance test (BAT; before CBT: n = 245; after CBT: n = 171) and functional magnetic resonance imaging (fMRI) data during fear conditioning were acquired before and after CBT (n = 39). HTR1A risk genotype (GG) carriers more often escaped during the BAT before treatment. Exploratory fMRI results suggest increased activation of the amygdala in response to threat as well as safety cues before and after treatment in GG carriers. Furthermore, GG carriers demonstrated reduced effects of CBT on differential conditioning in regions including the bilateral insulae and the anterior cingulate cortex. Finally, risk genotype carriers demonstrated reduced self-initiated exposure behaviour to aversive situations. This study demonstrates the effect of HTR1A variation on defensive behaviour, amygdala activity, CBT-induced neural plasticity and normalization of defence behaviour in PD/AG. Our results, therefore, translate evidence from animal studies to humans and suggest a central role for HTR1A in differentiating subgroups of patients with anxiety disorders.
Ziel der vorliegenden Arbeit war es, die Materialeigenschaften der CPC hinsichtlich ihres Aushärteverhaltens mit unterschiedlichen Herstellungs- und Versuchsparametern systematisch zu analysieren. Die Zementkomponente Ca2KNa(PO4)2 wurde mit den Additiven SiO2 oder MgO in verschiedenen prozentualen Anteilen dotiert. Die Pulver wurden entweder bei 1050 °C gesintert oder bei > 1500 °C geschmolzen. Die Abbindereaktion erfolgte nach 24-stündiger Mahlung mit Wasser, Na2HPO4, 0,1 M Citronensäure oder mit MCPA und 0,5 M Citronensäure. Im Fokus stand die Beeinflussung der HA-Produktion in Abhängigkeit der Parameter.
Ursächlich für das Nicht-Abbinden zu HA bei höherer Dotierung war die abbindeverzögernde Wirkung der Additive, sowie die Transformation der Pulver zu β-TCP bei höherer Dotierung. Ein Einfluss der Sintertemperatur auf die HA-Produktion konnte nicht festgestellt werden.
Trotz der exzellenten Biokompatibilität von Silizium und Magnesium vermindert eine Metalloxid-Dotierung die Abbindefähigkeiten der Ca2KNa(PO4)2 –Zemente. Die resultierenden mechanischen Eigenschaften erweisen sich als klinisch unzureichend. Die undotierten Ca2KNa(PO4)2 –Zemente, die zu HA abbinden, eignen sich chirurgisch gesehen eventuell in gering kraftbelasteten Defektbereichen
In der vorliegenden Arbeit wurde das Verfahren der Zwei-Photonen-Polymerisation von anorganisch-organischen Hybridpolymeren (ORMOCER®e) untersucht. Untersuchungsschwerpunkte bildeten dabei die theoretischen Betrachtungen der Wechselwirkung zwischen Laser und Hybridpolymer, die experimentelle Charakterisierung unterschiedlicher ORMOCER®e sowie die Aufskalierung der Technologie im Hinblick auf die Herstellung von Scaffold-Strukturen für die regenerative Medizin. Hierbei wurde u. a. ein innovativer Belichtungsaufbau entworfen und aufgebaut, der es erlaubt makroskopische, poröse Scaffold-Strukturen mit minimalen Strukturgrößen im Bereich von wenigen Mikrometern herzustellen.
ORMOCER®e sind typischerweise für optische Anwendungen konzipiert, weisen allerdings z. T. biokompatible Eigenschaften auf. Das Material ORMOCER® MB-47 wurde von M. Beyer eigens für biologische Anwendungen synthetisiert. Es zeichnet sich durch Biokompatibilität, teilweiser Biodegradierbarkeit und hervorragende Strukturierbarkeit durch die Zwei-Photonen-Polymerisation aus.
Das in dieser Arbeit verwendete Mikrostrukturierungssystem beinhaltet im Wesentlichen einen Ultrakurzpulslaser, der 325 fs Pulse bei 1030 nm emittiert (verwendet wird die zweite Harmonische bei 515 nm), ein hochpräzises Positionierungssystem, bestehend aus drei luftgelagerten Lineartischen mit einer Reichweite von 10 cm (y-, z-Richtung) bzw. 15 cm (x-Richtung) sowie diversen Objektiven zur Fokussierung. Mit diesen Komponenten lassen sich komplexe dreidimensionale Strukturen mit minimalen Strukturgrößen von bis unter 100 nm erzeugen.
In Kapitel 5.1 wurden theoretische Untersuchungen im Hinblick auf das Wechselwirkungsverhalten zwischen der fokalen Intensitätsverteilung und dem Materialsystem zur Bildung eines Voxels durchgeführt, wobei das technische Wechselwirkungsvolumen und das chemische Wechselwirkungsvolumen samt den reaktionskinetischen Abläufen separat betrachtet wurde.
Das technische Wechselwirkungsvolumen beschreibt die Wechselwirkung zwischen der fokalen Intensitätsverteilung und dem Materialsystem im Rahmen eines Schwellwertprozesses, der es erlaubt Strukturdimensionen unterhalb des Beugungslimits zu realisieren. Die theoretischen Untersuchungen diesbezüglich ergaben, dass sphärische Aberrationen die fokale Intensitätsverteilung (Intensity-Point Spread Function (IPSF)) in Abhängigkeit der Belichtungskonfiguration z. T. sehr stark beeinflussen. Darüber hinaus wurde durch Betrachtung des Schwellwertverhaltens ein mathematischer Zusammenhang zwischen der IPSF und der Leistungsabhängigkeit der Charakteristik des technischen Wechselwirkungsvolumens geschaffen.
Das chemische Wechselwirkungsvolumen beschreibt das tatsächliche Volumen der stattfindenden Polymerisationsreaktion. Dieses geht über das des technischen hinaus, was eine Folge von raumeinnehmendem Kettenwachstum im Rahmen von reaktionskinetischen Teilprozessen ist. Durch die Simulationen dieser reaktionskinetischen Abläufe wurde das leistungsabhängige, zeitliche Verhalten der Reaktionsteilnehmer (Radikale, Monomer, Photoinitiator) und des Vernetzungsgrades ermittelt. Die Simulation wurden für sehr kurze Belichtungszeiten (< 10 ms) auf der Basis von gekoppelten Differentialgleichungen nach Uppal & Shiakolas durchgeführt. Dabei wurde der Einfluss der Teilchendiffusion sowie der Temperaturentwicklung als gering erachtet und in den Berechnungen vernachlässigt.
Die Simulationsergebnisse zeigen, dass eine geringe Belichtungszeit nicht unbedingt durch größere Laserleistungen ausgeglichen werden kann, um einen bestimmten Vernetzungsgrad zu erzielen. Vielmehr führt eine höhere Leistung zu einem raschen Verbrauch des Photoinitiators im Reaktionsvolumen und damit einem schnelleren Erliegen der Polymerisationsreaktion. Um dennoch hohe Vernetzungsgrade erzielen zu können, sind die Reaktionsgeschwindigkeitskoeffizienten der Propagation und der Terminierung k_P und k_T sowie eine ausreichende Photoinitiatorkonzentration von entscheidender Bedeutung. Je größer das Verhältnis k_P/k_T, desto höhere Vernetzungsgrade können auch bei kurzen Belichtungszeiten realisiert werden, wobei ein wesentlicher Teil der Polymerisation als Dunkelreaktion stattfindet. Diese Erkenntnis ist für die Aufskalierung der Technologie der Zwei-Photonen-Polymerisation von großer Bedeutung, welche mit einer Verkürzung der Belichtungszeiten einhergehen muss.
Des Weiteren zeigen die Simulationen, dass das spatiale Konversionsprofil eines Voxels ein lokales Minimum im Zentrum aufweisen kann. Dieses Phänomen tritt dann auf, wenn aufgrund der applizierten Leistung, welche gemäß des Profils der IPSF im Zentrum am höchsten ist, der Photoinitiator im Zentrum rasch verbraucht wird.
In Kapitel 5.2 wurde die Voxelbildung, das Vernetzungsverhalten sowie die mechanischen Eigenschaften belichteter ORMOCER®e bei unterschiedlichen Parametern und Materialsystemen experimentell untersucht.
An Hand von Voxelfeldern wurden die Voxelgröße, das Aspektverhältnis und das Voxelvolumen bei unterschiedlichen Laserleistungen ermittelt. Die Ergebnisse wurden mit den berechneten technischen Wechselwirkungsvolumina verglichen, wobei die Differenz von tatsächlicher Voxelgröße und technischem Wechselwirkungsvolumen als eine weitere charakteristische Größe eingeführt wurde. Dabei zeigte sich, dass besonders die Voxellänge von der Länge des technischen Wechselwirkungsvolumens derart abweicht, dass dies nicht durch raumeinnehmendes Kettenwachstum im Rahmen der Reaktionskinetik erklärt werden kann. Mögliche Erklärungsansätze basieren hierbei auf Wechselwirkungseffekte zwischen Lichtfeld und Material. Beispielsweise könnten durch den nichtlinearen optischen Kerr-Effekt oder die Polymerisation selbst Brechzahlinhomogenitäten induziert werden, welche die Voxelbildung durch Selbstfokussierung beeinflussen. Der Unterschied der Voxelbreite, also das laterale chemische Voxelwachstum, zur Breite des technischen Wechselwirkungsvolumens wurde hingegen mit Hilfe der Reaktionskinetik erklärt. Dabei zeigte sich, dass dieser Unterschied sowohl vom Material selbst als auch von der Fokussieroptik abhängt.
Des Weiteren wurde die Polymerisationsrate der unterschiedlichen Materialien aus der Auftragung des Voxelvolumens gegenüber der Laserleistung durch lineare Approximation bestimmt. Hierbei wurde festgestellt, dass die Materialsysteme z. T. erhebliche Unterschiede aufweisen. Als das Materialsystem mit der höchsten Polymerisationsrate hat sich das auf Acrylaten als vernetzbare Gruppen basierende OC-V in Kombination mit dem Irgacure® Oxe02 Photoinitiator herausgestellt. Aus diesem Grund wurde es für die Herstellung von makroskopischen Strukturen durch die Zwei-Photonen-Polymerisation bevorzugt verwendet.
Die unterschiedlichen Materialien wurden ferner mit Hilfe der µ-Raman-Spektroskopie auf ihr Vernetzungsverhalten untersucht. Konkret wurden hierbei Linienfelder unter Variation der Scan-Geschwindigkeit und der Laserleistung mit Hilfe der 2PP hergestellt und vermessen. Die Vernetzungsgrade wurden semi-quantitativ aus den Spektren ermittelt. Insgesamt wurden Vernetzungsgrade im Bereich zwischen 40 % und 60 % gemessen, wobei mit OC-V und 2 Gew.-% Irgacure® Ox02 die höchsten Vernetzungsgrade erzielt wurden. Des Weiteren hat sich gezeigt, dass die Konversionsgrade für die jeweiligen Materialsysteme bei allen Scan-Geschwindigkeiten sich auf einem im Rahmen der Fehlergrenzen gleichem Niveau befinden. Damit kann der durch Simulationen theoretisch prognostizierte Abfall des Sättigungskonversionsgrades mit zunehmender Scan-Geschwindigkeit mit entsprechend variierenden Belichtungszeiten nicht als verifiziert angesehen werden.
Die verschiedenen Materialsysteme wurden außerdem bezüglich ihrer mechanischen Eigenschaften charakterisiert. Zu diesem Zweck wurden zylindrische Formkörper unter verschiedenen Bedingungen (1PP, 2PP, verschiedene Photoinitiatorkonzentrationen) hergestellt und Druckfestigkeitsmessungen durchgeführt, sowie die Dichten und die Vernetzungsgrade aus den Formkörpern bestimmt.
Insgesamt wurden Elastizitätsmodule im Bereich zwischen 0,40 und 1,37 GPa und Bruchfestigkeitswerte zwischen 117 bis 310 MPa ermittelt. Es konnte festgestellt werden, dass die Konzentration des Photoiniators das Vernetzungsverhalten und damit die mechanischen Eigenschaften der Formkörper stark beeinflusst. Während geringe Konzentrationen zu geringeren Vernetzungsgraden und niedrigen Elastizitätsmodulen führten, zeigten die Formkörper höherer Konzentration ein deutlich spröderes Verhalten mit höheren Vernetzungsgraden und Elastizitätsmodulen. Das höchste Elastizitätsmodul wurde an Hand von Formkörpern vermessen, welche aus OC-V mit 2 Gew.-% Irgacure® Ox02 hergestellt wurden.
Darüber hinaus wurde festgestellt, dass sich die mechanischen Eigenschaften von durch 2PP hergestellten Formkörpern durch die applizierte Laserleistung beeinflussen lassen. Die Ursache hierfür ist, dass durch die Laserleistung die Voxelgröße und damit der Überlapp zwischen den Voxeln eingestellt werden kann. Im Bereich des Überlapps findet dann eine Doppelbelichtung des Materials statt, was zu höheren Vernetzungsgraden führen kann. Außerdem wurden durch die 2PP bei hinreichend großen Belichtungsleistungen auch Formkörper realisiert, welche höhere Elastizitätsmodule und Bruchfestigkeitswerte aufwiesen als Körper, welche durch UV-Belichtung hergestellt wurden.
Die Aufskalierung der Zwei-Photonen-Technologie wurde in Kapitel 5.3 behandelt. Neben einer ausführlichen Diskussion zu den Herausforderungen diesbezüglich, wurden zwei Belichtungsstrategien zur Herstellung von makroskopischen Scaffold-Strukturen eingesetzt und optimiert. Hierbei ist insbesondere der Badaufbau hervorzuheben, der es erlaubte Strukturen von prinzipiell unbegrenzter Höhe mit Hilfe der Zwei-Photonen-Polymerisation herzustellen.
Eine wesentliche Herausforderung der Aufskalierung der 2PP ist die Beschleunigung des Prozesses. Aus den Betrachtungen geht hervor, dass für eine gravierende Beschleunigung der 2PP-Strukturierung neben der Scan-Geschwindigkeit auch das Beschleunigungsvermögen des Positionierungssystems entscheidend ist. Des Weiteren sind auch Parallelisierungsmethoden mit z. B. diffraktiven optischen Elementen nötig, um ausreichende Prozessgeschwindigkeiten zu erzielen.
Der Standardaufbau mit Luftobjektiven wurde dazu verwendet millimetergroße Strukturen mit hoher Qualität aus ORMOCER®en herzustellen. Auch wenn die maximale Strukturhöhe durch den Arbeitsabstand des Objektivs beschränkt ist, hat sich gezeigt, dass dieser Aufbau sich für die einfache Herstellung von millimetergroßen Test-Scaffold-Strukturen eignet, welche z. B. für Zellwachstumsversuche oder mechanische Belastungstest eingesetzt werden können. Das biodegradierbare MB-47 wurde hierbei ebenfalls erfolgreich eingesetzt und u. a. für die Herstellung von Drug-Delivery-Strukturen verwendet.
Der Badaufbau, basierend auf einem Materialbad mit durchsichtigem Boden, einem darin befindlichen und in der Vertikalen (z-Richtung) beweglichen Substrathalter sowie einer Belichtung von unten durch eine sich in der Ebene bewegende Fokussieroptik, wurde verwendet um eine Freiheitsstatue mit 2 cm Höhe sowie millimetergroße Scaffold-Strukturen mit Porengrößen im Bereich von 40 bis 500 µm in ORMOCER-V zu realisieren. Weitere Strukturierungsresultate mit z. T. anwendungsbezogenem Hintergrund sind die Gehörknöchelchen des menschlichen Ohrs in Lebensgröße, ein Scaffold in Form eines Steigbügels des menschlichen Ohrs, Test-Scaffold-Strukturen für mechanische oder biologische Untersuchungen sowie Drug-Delivery Strukturen. Es wurden Bauraten von bis zu 10 mm^3/h erzielt.
Bezüglich der Prozessgeschwindigkeit und Strukturhöhe wurde bei Weitem noch nicht das Potential des luftgelagerten Positioniersystems ausgeschöpft. Dafür bedarf es einer Gewichtsoptimierung des bestehenden Belichtungsaufbau, um höhere Beschleunigungswerte und Scan-Geschwindigkeiten realisieren zu können. Unter Annahme einer effektiven Gewichtsoptimierung und der damit verbundenen Erhöhung der Beschleunigung auf 10 m/s^2 könnte eine Baurate bei einer Scan-Geschwindigkeit von 225 mm/s und einem Slice- und Hatch-Abstand von 15 und 10 µm von etwa 60 mm^3/h erzielt werden.
Im Rahmen der Aufskalierung wurde ebenfalls der experimentelle Einsatz von diffraktiven optischen Elementen zur Fokus-Multiplikation untersucht. Die Experimente wurden mit Hilfe eines Elements durchgeführt, welches eine 2 x 2 Punkte-Matrix neben der ungebeugten 0. Ordnung bereitstellt und Bestandteil eines experimentellen Setups war, welches aus Linsen, Blenden und einem Objektiv zur Fokussierung bestand. Mit Hilfe der erzeugten Spot-Matrix wurden zum einen simultan vier Drug-Delivery-Strukturen hergestellt und zum anderen einzelne Scaffold-Strukturen realisiert. In jedem Fall wurde eine Beschleunigung des Prozess bzw. eine Erhöhung der Polymerisationsrate um den Faktor 4 für die verwendeten Parameter erreicht. Bei der Herstellung der Scaffolds wurden zwei unterschiedliche Strategien verfolgt. Während zum einen die Periodizität der inneren Scaffold-Struktur auf die Fokusabstände angepasst und damit simultan vier aneinandergereihte Einheitszellen hergestellt wurden, konnte zum anderen auch demonstriert werden, dass durch die geschickte Bewegung der Fokusse eine ineinander verschobene Struktur möglich ist. Der Vorteil der letzteren Strategie ist, dass auf diese Weise eine komplette Schicht gescannt werden kann und damit hohe Scan-Geschwindigkeiten realisiert werden können. Die erzielten Bauraten waren dennoch nicht größer als die Bauraten, die mit einem einzelnen Spot im Rahmen des Standardaufbaus oder des Badaufbaus erreicht wurden. Hierfür bedarf es weiterer Optimierung der Parameter und des Setups.
Transmittiert fokussiertes Licht eine Grenzfläche zweier Medien mit unterschiedlichen Brechungsindizes, dann tritt sphärische Aberration auf, welche sich durch die Verbreiterung des Fokus besonders in axiale Richtung bemerkbar macht. Da diese im Rahmen der verwendeten Belichtungsstrategien die Strukturierungsergebnisse nachweislich beeinträchtigen, wurden experimentelle Untersuchungen sowie Optimierungsroutinen diesbezüglich durchgeführt.
Im Zusammenhang mit dem Standardaufbau wurde eine Leistungsanpassung während der Strukturierung vorgenommen. Auf diese Weise wurde erreicht, dass bei variabler Fokustiefe im Material die maximale Intensität trotz sphärischer Aberration konstant gehalten wurde, wodurch sich die strukturelle Homogenität der Scaffolds entlang der axialen Richtung (optische Achse) deutlich verbesserte.
Des Weiteren wurde der Badaufbau dazu verwendet, die axiale Intensitätsverteilung in-situ für diskrete Fokustiefen unter der Verwendung eines Objektivs mit der NA von 0,60 abzubilden. Zu diesem Zweck wurde aus hergestellten Voxelfeldern eine Voxelfeldfunktion ermittelt und mit der axialen IPSF korreliert. Dabei wurde angenommen, dass sich das chemische Wechselwirkungsvolumen vernachlässigbar gering vom technischen Wechselwirkungsvolumen unterscheidet. Die experimentellen Ergebnisse zeigten deutlich die für sphärische Aberrationen typischen Nebenmaxima auf. Die Lage bzw. Abstände dieser entsprachen in guter Übereinstimmung den jeweiligen Simulationen. Schließlich wurde noch die sphärische Aberration durch den Korrekturring der Objektive für verschiedene Deckglasdicken korrigiert. Die resultierende IPSF wurde ebenfalls mit Hilfe des Badaufbaus abgebildet, wobei keinerlei Nebenmaxima gefunden werden konnten. Die Breite des Hauptmaximums konnte deutlich verringert werden.
Zusammengefasst lässt sich sagen, dass im Rahmen dieser Arbeit erhebliche Fortschritte bei der Aufskalierung der 2PP zur Erzeugung von Scaffold-Strukturen für die regenerative Medizin erzielt wurden. Die erreichten Strukturdimensionen und die Bauraten übertreffen alle bis dato bekannten Ergebnisse. Dabei wurden auch durch theoretische Betrachtungen und experimentellen Methoden grundlegende Erkenntnisse über die Reaktionsdynamik der durch die Zwei-Photonen-Absorption initiierten Polymerisationsreaktion gewonnen. Nichtsdestotrotz sind einige Fragestellungen offen sowie Problematiken des Prozesses vorhanden, die für eine Realisierung von makroskopischen Scaffold-Strukturen gelöst werden müssen.
So sind die realisierten Bauraten noch zu gering, um in angemessener Zeit makroskopische Scaffolds-Strukturen herzustellen, welche deutlich größer als 1 cm^3 sind. Aus diesem Grund müssen weitere Verbesserungen bezüglich der Scan-Geschwindigkeit sowie des Einsatzes von diffraktiven optischen Elementen zur Erhöhung der Polymerisationsrate erzielt werden. Da bei der Verwendung von Multi-Spot-Arrays, welche mit Hilfe gewöhnlicher diffraktiver optischer Elemente erzeugt wurden, die Realisierung von beliebigen und detaillierten äußeren Scaffold-Formen eingeschränkt ist, empfiehlt es sich den Einsatz von Spatial Light-Modulatoren zu verfolgen. Diese fungieren als dynamisch modulierbares DOE, mit dem einzelne Spots gezielt ein- und ausgeblendet und Spotabstände dynamisch variiert werden können. Schließlich ist es vorstellbar, den Spatial Light-Modulator mit dem Badaufbau zu kombinieren, um uneingeschränkte, große Strukturen in annehmbarer Zeit mit hochaufgelösten Details herstellen zu können. Dieses Vorgehen bedarf allerdings noch der tiefgreifenden Untersuchung der Potentiale des Spatial Light-Modulators.
Darüber hinaus weisen die theoretischen und experimentellen Untersuchungen zur Reaktionskinetik darauf hin, dass die Voxelentstehung ein komplexer Prozess ist, der möglicherweise auch durch nichtlineare optische Wechselwirkungseffekte abseits der Zwei-Photonen-Absorption beeinflusst wird. Daher sind hier weitere Untersuchungen und Berechnungen zu empfehlen, um z. B. den Einfluss einer intensitätsabhängigen Brechzahl auf die Voxelbildung quantifizieren zu können. Entsprechende Ergebnisse könnten schließlich dazu dienen, dass im Rahmen dieser Arbeit entwickelte Modell zur Voxelbildung, welches auf der getrennten Betrachtung von technischen und chemischen Wechselwirkungsvolumen basiert, zu verbessern. Ein leistungsfähiges Modell, welches die Voxelbildung in Abhängigkeit des Materials und der Fokussieroptik präzise vorhersagen kann, wäre für das Erzielen optimaler Strukturierungsergebnissen ein Gewinn.
Unicellular parasites have developed sophisticated swimming mechanisms to survive in a wide range of environments. Cell motility of African trypanosomes, parasites responsible for fatal illness in humans and animals, is crucial both in the insect vector and the mammalian host. Using millisecond-scale imaging in a microfluidics platform along with a custom made optical trap, we are able to confine single cells to study trypanosome motility. From the trapping characteristics of the cells, we determine the propulsion force generated by cells with a single flagellum as well as of dividing trypanosomes with two fully developed flagella. Estimates of the dissipative energy and the power generation of single cells obtained from the motility patterns of the trypanosomes within the optical trap indicate that specific motility characteristics, in addition to locomotion, may be required for antibody clearance. Introducing a steerable second optical trap we could further measure the force, which is generated at the flagellar tip. Differences in the cellular structure of the trypanosomes are correlated with the trapping and motility characteristics and in consequence with their propulsion force, dissipative energy and power generation.
Premature implant loosening following total knee arthroplasty (TKA) can have several causes. In this article we report on a rare case of a 74 year old male patient suffering tibial component loosening 14 month after primary TKA. The patient did neither have any malignancies nor joint arthroplasty before. Upon clinical examination the range of motion in the diseased knee was painfully restricted to 80° of knee flexion, with the patient increasingly suffering sleeping and resting pain, and also at weight bearing. In standard radiographs, loosening of the TKA due to a large osteolysis at the tibial component was evident. Local computed tomography (CT) of the right knee revealed loosening of the tibial component due to a presumably malign bone tumor. For determination of the final diagnosis a representative biopsy of the tumor was taken by open surgery prior to the tumor resection. Histopathologic evaluation of the biopsy revealed a periprosthetic myxoid chondrosarcoma of the proximal tibia. Pre-operative staging examination included CT scans of lung and abdomen, as well as a bone scintigraphy which revealed no signs of tumor metastasis in the body. Surgical management comprised wide tumor resection and implantation of a hinged tumor knee arthroplasty with replacements of the distal femur and proximal tibia, as well as a patella tendon replacement using a synthetic ligament. Revision surgery was necessary twice due to impaired wound healing and critical soft tissue coverage, and treatment included a gastrocnemius muscle flap with skin mesh graft covering. Unfortunately long-term follow-up examinations could not be obtained, as the patient deceased due to an alveolitis during rehabilitation. In summary, the specifics of this rare case of aseptic TKA loosening, and the unusual circumstances of chondrosarcoma diagnosis and treatment are informative for those providing surgical treatment of similar cases.
Efficient Transient Transfection of Human Multiple Myeloma Cells by Electroporation - An Appraisal

Cell lines represent the everyday workhorses for in vitro research on multiple myeloma (MM) and are regularly employed in all aspects of molecular and pharmacological investigations. Although loss-of-function studies using RNA interference in MM cell lines depend on successful knockdown, no well-established and widely applied protocol for efficient transient transfection has so far emerged. Here, we provide an appraisal of electroporation as a means to introduce either short-hairpin RNA expression vectors or synthesised siRNAs into MM cells. We found that electroporation using siRNAs was much more efficient than previously anticipated on the basis of transfection efficiencies deduced from EGFP-expression off protein expression vectors. Such knowledge can even confidently be exploited in "hard-to-transfect" MM cell lines to generate large numbers of transient knockdown phenotype MM cells. In addition, special attention was given to developing a protocol that provides easy implementation, good reproducibility and manageable experimental costs.
The embryonic vertebrate heart tube develops an atrioventricular canal that divides the atrial and ventricular chambers, forms atrioventricular conduction tissue and organizes valve development. Here we assess the transcriptional mechanism underlying this localized differentiation process. We show that atrioventricular canal-specific enhancers are GATA-binding site-dependent and act as switches that repress gene activity in the chambers. We find that atrioventricular canal-specific gene loci are enriched in H3K27ac, a marker of active enhancers, in atrioventricular canal tissue and depleted in H3K27ac in chamber tissue. In the atrioventricular canal, Gata4 activates the enhancers in synergy with Bmp2/Smad signalling, leading to H3K27 acetylation. In contrast, in chambers, Gata4 cooperates with pan-cardiac Hdac1 and Hdac2 and chamber-specific Hey1 and Hey2, leading to H3K27 deacetylation and repression. We conclude that atrioventricular canal-specific enhancers are platforms integrating cardiac transcription factors, broadly active histone modification enzymes and localized co-factors to drive atrioventricular canal-specific gene activity.
Das Visus-VEP wird angewendet bei Patienten mit eingeschränkter Kooperation (Kinder, retardierte Personen), unklarer Visusminderung, funktionellen Sehstörungen, Verdacht auf Simulation sowie zur Verlaufskontrolle von Erkrankungen der Sehbahn. Ziel dieser Arbeit war es, die bisherigen Methoden zu optimieren.
An 20 augengesunden Probanden wurden monokular Messungen mit Sinusgitter-Reizen bei 12 Ortsfrequenzen zwischen 1 und 20 cpd durchgeführt. Die Präsentation erfolgte jeweils im Pattern-reversal- und Pattern-onset-Modus. Die VEP-Signale wurden über eine auf der Position Oz liegende Elektrode sowie über zusätzlich zwei laterale (L, R) und eine kraniale Elektrode (O) abgeleitet. Durch ein mittels Igor Pro© geschriebenes Programm wurden zur Auswertung für jede Versuchsperson insgesamt 12 Graphen erstellt, in denen die Fourier-Amplitude als Funktion der Ortsfrequenzen aufgetragen wurde, sog. Tuning-Kurven: Für beide Reizmuster wurde neben der Oz -Ableitung eine 3-Punkt- (VEP-Signale von Oz, L, R) und eine 4-Punkt-Laplacian-Ableitung (VEP-Signale von Oz, L, R, O) durchgeführt. Zusätzlich erfolgte jeweils eine alleinige Auswertung der Fourier-Amplitude der Reizfrequenz sowie eine Auswertung unter Einbeziehen der ersten Oberwelle der Reizfrequenz im Fourier-Spektrum.
Der Einfluss dieser Methoden auf die Grenzortsfrequenz, die Amplitude, das Signal-Rausch-Verhältnis (SNR) und die Anzahl der signifikanten Antworten wurde untersucht. Die Elektrodenanordnung hatte keinen signifikanten Einfluss auf die Grenzortsfrequenz. Beim Pattern-reversal-Reiz waren durch die Laplacian-Ableitungen die Amplituden signifikant niedriger, beim Pattern-onset-Reiz bestand diesbezüglich kein Unterschied. Für den Pattern-reversal-Reiz erbrachte 4-Punkt-Laplacian das beste SNR, für den Pattern-onset-Reiz gab es hierbei keine signifikanten Unterschiede. Bei der Zahl der signifikanten Datenpunkte erwiesen sich beide Laplacian-Anordnungen bei beiden Reizmustern als vorteilhaft. Auch Mackay et al. (2003a,b) erreichten durch eine 3-Punkt-Laplacian-Anordnung ein verbessertes SNR und mehr signifikante Ereignisse. Trotz des Mehraufwandes empfehlen wir aufgrund dieses Ergebnisses in der Klinik die Verwendung einer Laplacian-Ableitung. Wegen einer Verminderung der Amplitudenhöhe kann jedoch die Laplacian-Ableitung in wenigen Fällen bei Personen mit sehr niedrigen VEP-Amplituden von Nachteil sein.
Eine Auswertung mit Oberwelle lieferte keine signifikanten Unterschiede für die Grenzortsfrequenzen. Die Amplituden lagen hierbei signifikant höher. Für den Pattern-onset-Reiz wurde das SNR sogar niedriger, für den Pattern-reversal-Reiz änderte es sich nicht. Bezüglich der Anzahl der signifikanten Datenpunkte bestand kein Einfluss der Oberwelle. Wegen der letzten beiden Punkte bewerten wir eine Auswertung mit erster Oberwelle nicht als vorteilhaft in der klinischen Routine. Bei einzelnen Patienten mit besonders niedrigen Amplituden im VEP kann diese Auswertungsmethode jedoch eine Möglichkeit sein.
Für den Pattern-onset-Reiz lagen die Grenzortsfrequenzen signifikant höher, ihr Spektrum war vergleichbar mit denen der Studien von Strasburger et al. (1996) und Bach et al. (2008). Im Hinblick auf die Amplitude bestand zwischen den beiden Reizmuster entgegen Strasburger et al. (1996) und Parry et al. (1999) in unserer Studie kein signifikanter Unterschied.
Bei 12 Versuchspersonen konnten beim Pattern-onset-Reiz aus den Messdaten keine Tuning-Kurven erstellt werden, da es bei den niedrigen Ortsfrequenzen wieder zu einem Anstieg der VEP-Amplitude gekommen war. In einem Nachexperiment erscheinen Helligkeitsartefakte des Monitors als wahrscheinlichste Ursache, für die der Pattern-onset-Reiz anfällig ist (Fahle & Bach, 2006). Wegen eines zusätzlichen Zeitaufwandes durch nicht auswertbare Messungen oder sogar verfälschter objektiver Visus-Werte durch falsch hohe VEP-Amplituden beim Pattern-onset-Reiz, empfehlen wir für die Praxis den Einsatz von Pattern-reversal-Mustern.
In weiterführende Studien sollte der Einfluss der in unserer Arbeit untersuchten Parameter (Reizart, Oberwelle, Ableitung) bei unterschiedlichen Augenerkrankungen sowie bei Personen mit besonders niedrigem Visus erforscht werden.
Einen Satz syntaktisch zu analysieren heißt, ein Satzverstehen nach bestimmten Kriterien offenzulegen. Jeder Satzanalyse geht ein grundsätzliches, voranalytisch verborgenes Verstehen ebenso voraus, wie der Prozess der Analyse ein solches Verstehen konkretisiert und ausdrücklich festzurrt: Ohne Verstehen keine Analyse. Für Analysen, die ein Satzverstehen syntaktisch explizieren sollen, benötigt man einen theoretischen Hintergrund, der die Analysewerkzeuge bereitstellt: Ohne Beschreibungsmittel keine Analyse. Schließlich braucht jede Analyse auch noch eine Darstellungsmethode: Ohne Festhalten der Ergebnisse kein Zugriff auf die Analyse.
Der vorliegende Band versteht sich als eine Explikation von Satzverstehen anhand eines konkreten, valenzorientierten Beschreibungsinventars. Im Vordergrund stehen dabei die Visualisierungen der Analyse-Ergebnisse durch Baumgraphen. Über 100 Bäumchen sind hier zu einem Wald versammelt. Damit ist insbesondere (aber nicht nur) für Studierende, die nach dieser Methode Sätze analysieren, eine Möglichkeit gegeben, einen valenz- und dependenzgrammatischen Zugriff auf verschiedene syntaktisch beschreibbare Phänomene authentischen Sprachvorkommens zu erproben. Darüber hinaus stellt der „Würzburger Wald“ unter Beweis, dass die in diesem Band intensiv angewandte Analysemethode ein stabiles Instrument für adäquate syntaktische Analysen sprachlicher Einheiten aus der freien Wildbahn journalistischer Textproduktion ist.
[Purpose] A wide variety of accelerometer tools are used to estimate human movement, but there are no adequate data relating to gait symmetry parameters in the context of knee osteoarthritis. This study's purpose was to evaluate a 3D-kinematic system using body-mounted sensors (gyroscopes and accelerometers) on the trunk and limbs. This is the first study to use spectral analysis for data post processing. [Conclusions] The 3D-kinematic system is objective, inexpensive, accurate and portable, and allows long-term recordings in clinical, sport as well as ergonomic or functional capacity evaluation (FCE) settings. For fast post-processing, spectral analysis of the recorded data is recommended.
[Purpose] A wide variety of accelerometer tools are used to estimate human movement, but there are no adequate data relating to gait symmetry parameters in the context of knee osteoarthritis. The aim of this work was to synthesize and functionalize different bio-relevant nanomaterials like silica-coated superparamagnetic iron oxide nanoparticles (SPIONs) as contrast agents for T2 magnetic resonance imaging (MRI) and detonation nanodiamond (DND) with the neurohormone peptide allatostatin 1 (ALST1) and a fluorescent dye. Analytical techniques for the determination and quantification of surface functional groups like amines, azides, and peptides were also developed and established.
Thus, in the first part of the work, a TGF-1 binding peptide and allatostatin 1 (ALST1), both supposed to act as active tumour targeting vectors, were synthesized by solid-phase peptide synthesis (SPPS) and characterized by high pressure liquid chromatography (HPLC) and mass spectrometry. Then, azide-functionalized silica nanoparticles were synthesized by the Stöber process and characterized by transmission electron microscopy (TEM) and infrared spectroscopy (IR). The surface loading of amine and azide groups was determined by a new protocol. The azide groups were reduced with sodium boronhydride to amine and then functionalized with Fmoc-Rink Amide linker according to a standard SPPS protocol. Upon cleavage of Fmoc by piperidine, the resulting dibenzofulvene and its piperidine adduct were quantified by UV/Vis spectroscopy and used to determine the amount of amine groups on the nanoparticle surface. Then, ALST1 and related tyrosine- and phenylalanine substituted model peptides were conjugated to the azide-functionalized silica nanoparticles by copper(I)-catalyzed azide-alkyne dipolar cycloaddition (CuAAC). The successful peptide conjugation was demonstrated by the Pauly reaction, which however is only sensitive to histidine- and tyrosine-containing peptides. As a more general alternative, the acid hydrolysis of the peptides to their individual amino acid building blocks followed by derivatization with phenyl isothiocyanate (PITC) allowed the separation, determination, and quantification of the constituent amino acids by HPLC.
In the second part of the work, amine- and azide-functionalized silica-coated superparamagnetic iron oxide nanoparticles (SPIONs) were synthesized by co-precipitation and subsequent silica-coated based on the Stöber process and characterized by TEM and IR. The amine surface loading was determined by the method already established for the pure silica systems. The azide surface loading could also be quantified by reduction with sodium boronhydride to amine groups and then conjugation to Fmoc-Rink amide linker. Upon cleavage of Fmoc with piperidine, the total amine surface loading was obtained. The amount of azide surface groups was then determined from the difference of the total amine surface loading and the amine surface loading. Thus, it was possible to quantify both amine and azide surface groups on a single nanoparticle system. Superparamagnetic iron oxide nanoparticles (SPIONs) are potent T2 contrast agents for magnetic resonance imaging (MRI). Due to their natural metabolism after injection into the blood stream, SPIONs mostly end up inside macrophages, liver, spleen or kidneys. To generate a potential target-specific SPION-based T2 contrast agent for MRI, the neurohormone peptide ALST1 was conjugated by CuAAC to the azide- and amine functionalized superparamagnetic iron oxide nanoparticles, since ALST1 is supposed to target difficult-to-treat neuroendocrinic tumours due to its analogy to galanin and somastatin receptor ligands. The organic fluorescent dye cyanine 5 (Cy5) was also conjugated to the silica-coated superparamagnetic iron oxide nanoparticles (SPIONs) via a NHS-ester to the amines to enable cell uptake studies by fluorescence microscopy. These constructs were characterized by TEM, dynamic light scattering (DLS), and IR. The amino acids of the conjugated ALST1 were determined by the HPLC method as described before for peptide-modified silica nanoparticle surfaces. Then, the relaxivity r2 was measured at 7 T. However, a r2 value of 27 L/mmolFe·s for the dual ALST1-/Cy5-functionalized silica-coated SPIONs was not comparable to T2 contrast agents in clinical use, since their relaxivity is commonly determined at 1.5 T, and no such instrument was available. However, it can be assumed that the synthesized dual
ALST1-/Cy5-functionalized silica-coated SPION would show a lower r2 at 1.5 T than at 7T. Commercial T2 MRI contrast agents like VSOP-C184 from Ferropharm show at r2 values of about 30 L/mmolFe·s at 1.5 T. Still, the relaxivity of the new material has some potential for application as a T2 contrast agent. Then, the material was used in cell uptake studies by fluorescence microscopy with the conjugated Cy5 dye as a probe. The dual
ALST1-/Cy5-functionalized silica-coated SPION showed a high degree of agglomeration with no cellular uptake unlike described for ALST1-functionalized nanoparticles in literature. It is assumed that upon agglomeration of the particles, constructs form which are unable to be internalized by the cellular endocytotic pathways anymore. As a future perspective, the tendency of the particle to agglomerate should be reduced by changing the coating material to polyethylene glycol (PEG) or chitosan, which are known to be bio-compatible, bio-degradable and prevent agglomeration.
In the third part of the work, the rhenium compound [ReBr(CO)3(L)] with L = 2-phenyl-1H-imidazo[4,5-f][1,10]phenanthroline and its manganese analogue were synthesized by heating the ligand and rhenium pentacarbonyl bromide or and manganese pentacarbonyl bromide respectively, in toluene. However, [MnBr(CO)3(L)] was unstable upon illumination by UV light at 365 nm. Thus, it was dismissed for further application. The photophysical properties of [ReBr(CO)3(L)] were explored, by determination of the excited-state life time by the time-correlated single-photon counting (TCSPC) method and the quantum yield by a fluorescence spectrometer equipped with an integration sphere. A value of = 455 ns, a Stokes shift of 197 nm and a rather low quantum yield =were found. Metal complexes are supposed to have superior properties compared to organic dyes due to their large Stokes shifts, long excited-state life times, and high quantum yields. Thus, amine- and azide-functionalized detonation nanodiamond (DND) as an alternative biological inert carrier system was functionalized with ALST1 to enhance its cell uptake properties. A luminescent probe for cell uptake studies using fluorescence microscopy was also attached, either based on the new rhenium complex or the commercially available organic dye Cy5, respectively. The aldehyde-functionalized rhenium complex was conjugated to the DND via oxime ligation, which is known to be a mild and catalyst-free conjugation method. The amount of peptide ALST1 on the DND was analyzed and quantified after acid hydrolysis and PITC derivatization by HPLC as described before. Then, the ALST1-/luminescent probe-functionalized DND was investigated for its photophysical properties by fluorescence spectroscopy. The Cy5-functionalized material showed a slightly lower fluorescence performance in aqueous solution than reported in literature and commercial suppliers with a life time < 0.4 ns and quantum yields not determinable by integration sphere due to the week signal intensity. The rhenium complex-functionalized material had a very low signal intensity in only aqueous medium, and thus determination of life times and quantum yield by fluorescence spectroscopy was not possible. After incubation with MDA-MB 231 cells, the Cy5-functionalized DND could easily be detected due to its red fluorescence. However, it was not possible to visualize the rhenium complex-functionalized DND with fluorescence microscopy due to the low fluorescence intensity of the complex in aqueous medium and the lack of proper filters for the fluorescence microscope. Cy5-functionalized DND did not show any cellular uptake in fluorescence microscopy after conjugation with ALST1. Since the nanodiamond surface is known to strongly adsorb peptides and proteins, it is assumed that the peptide chain is oriented perpendicular to the nanoparticle surface and thus not able to interact with cell membrane receptors to promote cell uptake of the particles. As a future perspective, the ALST1-promoted cellular uptake of the DND should be improved by using different linker systems for peptide conjugation to prevent adsorption of the peptide chain on the particle surface.
The new analytical methods for amino-, azide-, and peptide-functionalized nanoparticles have great potential to assist in the quantification of nanoparticle surface modifications by UV/Vis spectroscopy and HPLC. The determination of surface amine and azide groups based on the cleavage of conjugated Fmoc-Rink amide linker and detected by UV/Vis spectroscopy is applicable to all amine-/azide-functionalized nanomaterials. However, particles which form very stable suspension with the cleavage mixture can cause quantification problems due to scattering, making an accurate quantification of dibenzofulvene and its piperidine adduct impossible. The detection of tyrosine- and histidine-containing peptides based on the Pauly reaction is well-suited as a fast and easy-to-perform qualitative demonstration of successful peptide surface conjugation. However, its major drawback as a colourimetric approach is that coloured particles cannot be evaluated by this method. The amino acid analysis based on HPLC after acid hydrolysis of peptides conjugated to nanoparticle surfaces to its individual building blocks and subsequent derivatization with PITC, can be used on all nanomaterials with peptide or protein surface modification. It allows detection of amino acids down to picomolar concentrations and even enables analysis of very small peptide surface loadings. However, the resulting HPLC traces are difficult to analyze.
Three new analytical methods based on UV/Vis and HPLC techniques have been developed and established. They assisted in the characterization of the synthesized DND and SPIONs with dual functionalization by ALST1 and Cy5 or [ReBr(CO)3(L)], respectively. However, the nanomaterials showed no cellular uptake due to a high tendency to agglomerate. The cellular uptake should be improved and the tendency to agglomerate of the SPIONs should be reduced by changing the surface coating from silica to either PEG or chitosan. Furthermore, different linker systems for connecting peptides to DND surfaces should be synthesized and evaluated to reduce potential peptide chain adsorption.
Background
Preterm birth, low birth weight, and infant catch-up growth seem associated with an increased risk of respiratory diseases in later life, but individual studies showed conflicting results.
Objectives
We performed an individual participant data meta-analysis for 147,252 children of 31 birth cohort studies to determine the associations of birth and infant growth characteristics with the risks of preschool wheezing (1-4 years) and school-age asthma (5-10 years).
Methods
First, we performed an adjusted 1-stage random-effect meta-analysis to assess the combined associations of gestational age, birth weight, and infant weight gain with childhood asthma. Second, we performed an adjusted 2-stage random-effect meta-analysis to assess the associations of preterm birth (gestational age <37 weeks) and low birth weight (<2500 g) with childhood asthma outcomes.
Results
Younger gestational age at birth and higher infant weight gain were independently associated with higher risks of preschool wheezing and school-age asthma (P < .05). The inverse associations of birth weight with childhood asthma were explained by gestational age at birth. Compared with term-born children with normal infant weight gain, we observed the highest risks of school-age asthma in children born preterm with high infant weight gain (odds ratio [OR], 4.47; 95% CI, 2.58-7.76). Preterm birth was positively associated with an increased risk of preschool wheezing (pooled odds ratio [pOR], 1.34; 95% CI, 1.25-1.43) and school-age asthma (pOR, 1.40; 95% CI, 1.18-1.67) independent of birth weight. Weaker effect estimates were observed for the associations of low birth weight adjusted for gestational age at birth with preschool wheezing (pOR, 1.10; 95% CI, 1.00-1.21) and school-age asthma (pOR, 1.13; 95% CI, 1.01-1.27).
Conclusion
Younger gestational age at birth and higher infant weight gain were associated with childhood asthma outcomes. The associations of lower birth weight with childhood asthma were largely explained by gestational age at birth."
For social insects, colony performance is largely dependent on the quantity and quality of food intake and thus on the efficiency of its foragers. In addition to innate preferences and previous experience, foragers can use social information to decide when and where to forage. In some stingless bee (Meliponini) species, individual foraging decisions are shown to be influenced by the presence of social information at resource sites. In dual choice tests, we studied whether visual and/or olfactory cues affect individual decision-making in rigona corvina Cockerell and if this information is species-specific. We found that T. corvina foragers possess local enhancement: they are attracted by olfactory and visual cues released by conspecifics but avoid feeders associated with heterospecific individuals of the species Tetragona ziegleri (Friese). Overall, olfactory cues seem to be more important than visual cues, but information by visual cues alone is sufficient for discrimination.
Weltweit leben ca. 2,5 Mrd. Menschen im Dengue Virus Verbreitungsgebiet. Dengue Virus Infektionen führen zum Dengue Fieber und können bei Re-Infektionen
mit anderen Serotypen das sog. Dengue Schocksyndrom mit einer Letalität von 10% verursachen. Momentan stehen jedoch weder Impfstoffe noch antivirale Substanzen zur Verfügung.
In der vorliegenden Arbeit sollten DENV2-Proteaseinhibitoren entwickelt werden.
Dazu wurde ein in vitro DENV Proteasetest etabliert, für den die DENV Protease in Bakterien exprimiert und anschließend gereinigt wurde. Mit diesem System wurden 144 Verbindungen getestet und Diaryl-Thioether, Thiazole und Zimtsäurederivate als Dengue PIs charakterisiert. Ein Diarythioether (FM 47) wurde an die Proteasestruktur modelliert und nach den Strukturdaten zielgerichtet
derivatisiert. Diese Derivate ihibierten die Protease im mikromolaren Bereich und wurden anschließend in einer Zellkultur getestet. Drei Substanzen -
HWu 11, HWu 51, HWu 62 - zeigten gute bis sehr gute Hemmung in vivo bei 2,5 μM. Die Charakterisierung der Inhibitoren zeigte eine nicht-kompetitive Hemmung.
Die gefundenen Substanzen bilden eine gute Grundlage für die weitere Inhibitorforschung.
Die vorliegende Arbeit setzt sich mit den europa- und verfassungsrechtlichen Rechtsfragen der Einführung sogenannter Eurobonds auseinander.
Einleitend ordnet der Autor das Konzept der Eurobonds in den Kontext der Finanzkrise ein, um anschließend eine Begriffsklärung vorzunehmen. Dabei erläutert er auch die veränderbaren „Bausteine“ der Eurobonds und skizziert die Vielzahl bereits diskutierter Modelle.
Im zweiten Abschnitt des Hauptteils widmet sich der Autor sodann den europarechtlichen Rechtsfragen der Eurobonds. Im Fokus stehen hier insbesondere die Erörterung der denkbaren Kompetenzgrundlagen und die Frage der Vereinbarkeit mit der sogenannten No-Bail-Out-Klausel des Art. 125 Abs. 1 AEUV. Dabei gelangt er zu dem Befund der Unvereinbarkeit von Eurobonds jeglicher Art mit dem Telos des Art. 125 Abs. 1 AEUV. Als Konsequenz wird auf die Notwendigkeit einer Vertragsänderung hingewiesen, wobei der Autor für die Durchführung des ordentlichen Vertragsänderungsverfahrens gem. Art. 48 Abs. 2-5 EUV plädiert.
Im dritten Abschnitt wendet sich der Autor den verfassungsrechtlichen Rechtsfragen zu. Dabei wirft er ausgehend von der Integrationsverantwortung der deutschen Staatsorgane die Frage der Vereinbarkeit von Eurobonds mit der Budgethoheit des Bundestages auf. So gelangt er schließlich zu dem Befund, dass die Einführung von Eurobonds mit Blick auf das Budgetrecht im Grundsatz möglich ist. Im Anschluss hieran werden die verfassungsrechtlichen Grenzen der Übernahme von Gewährleistungen und die sich daraus ergebenden Folgen für die Einführung von Eurobonds diskutiert.
In einem Resümee werden Ergebnisse der juristischen Analyse mit einem kurzen Seitenblick auf die US-amerikanische Geschichte zusammengefasst.
Obligate intracellular bacteria depend entirely on nutrients from the host cell for their reproduction. Here, we show that obligate intracellular Chlamydia downregulate the central tumor suppressor p53 in human cells. This reduction of p53 levels is mediated by the PI3K-Akt signaling pathway, activation of HDM2, and subsequent proteasomal degradation of p53. The stabilization of p53 in human cells severely impaired chlamydial development and caused the loss of infectious particle formation. DNA-damage-induced p53 interfered with chlamydial development through downregulation of the pentose phosphate pathway (PPP). Increased expression of the PPP key enzyme glucose-6-phosphate dehydrogenase rescued the inhibition of chlamydial growth induced by DNA damage or stabilized p53. Thus, downregulation of p53 is a key event in the chlamydial life cycle that reprograms the host cell to create a metabolic environment supportive of chlamydial growth.
The intracellular pathogen Chlamydia is the causative agent of millions of new infections per year transmitting diseases like trachoma, pelvic inflammatory disease or lymphogranuloma venereum. Undetected or recurrent infections caused by chlamydial persistence are especially likely to provoke severe pathologies. To ensure host cell survival and to facilitate long term infections Chlamydia induces anti-apoptotic pathways, mainly at the level of mitochondria, and restrains activity of pro-apoptotic proteins. Additionally, the pathogen seizes host energy, carbohydrates, amino acids, lipids and nucleotides to facilitate propagation of bacterial progeny and growth of the chlamydial inclusion.
At the beginning of this study, Chlamydia-mediated apoptosis resistance to DNA damage induced by the topoisomerase inhibitor etoposide was investigated. In the course of this, a central cellular protein crucial for etoposide-mediated apoptosis, the tumour suppressor p53, was found to be downregulated during Chlamydia infections. Subsequently, different chlamydial strains and serovars were examined and p53 downregulation was ascertained to be a general feature during Chlamydia infections of human cells. Reduction of p53 protein level was established to be mediated by the PI3K-Akt signalling pathway, activation of the E3-ubiquitin ligase HDM2 and final degradation by the proteasome. Additionally, an intriguing discrepancy between infections of human and mouse cells was detected. Both activation of the PI3K-Akt pathway as well as degradation of p53 could not be observed in Chlamydia-infected mouse cells. Recently, production of reactive oxygen species (ROS) and damage to host cell DNA was reported to occur during Chlamydia infection. Thus, degradation of p53 strongly contributes to the anti-apoptotic environment crucial for chlamydial infection.
To verify the importance of p53 degradation for chlamydial growth and development, p53 was stabilised and activated by the HDM2-inhibiting drug nutlin-3 and the DNA damage-inducing compound etoposide. Unexpectedly, chlamydial development was severely impaired and inclusion formation was defective. Completion of the chlamydial developmental cycle was prevented resulting in loss of infectivity. Intriguingly, removal of the p53 activating stimulus allowed formation of the bacterial inclusion and recovery of infectivity. A similar observation of growth recovery was made in infected cell lines deficient for p53.
As bacterial growth and inclusion formation was strongly delayed in the presence of activated p53, p53-mediated inhibitory regulation of cellular metabolism was suspected to contribute to chlamydial growth defects. To verify this, glycolytic and pentose phosphate pathways were analysed revealing the importance of a functioning PPP for chlamydial growth. In addition, increased expression of glucose-6-phosphate dehydrogenase rescued chlamydial growth inhibition induced by activated p53. The rescuing effect was even more pronounced in p53-deficient cells treated with etoposide or nutlin-3 revealing additional p53-independent aspects of Chlamydia inhibition. Removal of ROS by anti-oxidant compounds was not sufficient to rescue chlamydial infectivity. Apparently, not only the anti-oxidant capacities of the PPP but also provision of precursors for nucleotide synthesis as well as contribution to DNA repair are important for successful chlamydial growth.
Modulation of host cell signalling was previously reported for a number of pathogens. As formation of ROS and DNA damage are likely to occur during infections of intracellular bacteria, several strategies to manipulate the host and to inhibit induction of apoptosis were invented. Downregulation of the tumour suppressor p53 is a crucial point during development of Chlamydia, ensuring both host cell survival and metabolic support conducive to chlamydial growth.
While gene expression is a fundamental and tightly controlled cellular process that is regulated at multiple steps, the exact contribution of each step remains unknown in any organism. The absence of transcription initiation regulation for RNA polymerase II in the protozoan parasite Trypanosoma brucei greatly simplifies the task of elucidating the contribution of translation to global gene expression. Therefore, we have sequenced ribosome-protected mRNA fragments in T. brucei, permitting the genome-wide analysis of RNA translation and translational efficiency. We find that the latter varies greatly between life cycle stages of the parasite and ∼100-fold between genes, thus contributing to gene expression to a similar extent as RNA stability. The ability to map ribosome positions at sub-codon resolution revealed extensive translation from upstream open reading frames located within 5' UTRs and enabled the identification of hundreds of previously un-annotated putative coding sequences (CDSs). Evaluation of existing proteomics and genome-wide RNAi data confirmed the translation of previously un-annotated CDSs and suggested an important role for >200 of those CDSs in parasite survival, especially in the form that is infective to mammals. Overall our data show that translational control plays a prevalent and important role in different parasite life cycle stages of T. brucei.
Background
Advances in high-throughput sequencing have led to the discovery of widespread transcription of natural antisense transcripts (NATs) in a large number of organisms, where these transcripts have been shown to play important roles in the regulation of gene expression. Likewise, the existence of NATs has been observed in Plasmodium but our understanding towards their genome-wide distribution remains incomplete due to the limited depth and uncertainties in the level of strand specificity of previous datasets.
Results
To gain insights into the genome-wide distribution of NATs in P. falciparum, we performed RNA-ligation based strand-specific RNA sequencing at unprecedented depth. Our data indicate that 78.3% of the genome is transcribed during blood-stage development. Moreover, our analysis reveals significant levels of antisense transcription from at least 24% of protein-coding genes and that while expression levels of NATs change during the intraerythrocytic developmental cycle (IDC), they do not correlate with the corresponding mRNA levels. Interestingly, antisense transcription is not evenly distributed across coding regions (CDSs) but strongly clustered towards the 3′-end of CDSs. Furthermore, for a significant subset of NATs, transcript levels correlate with mRNA levels of neighboring genes.
Finally, we were able to identify the polyadenylation sites (PASs) for a subset of NATs, demonstrating that at least some NATs are polyadenylated. We also mapped the PASs of 3443 coding genes, yielding an average 3′ untranslated region length of 523 bp.
Conclusions
Our strand-specific analysis of the P. falciparum transcriptome expands and strengthens the existing body of evidence that antisense transcription is a substantial phenomenon in P. falciparum. For a subset of neighboring genes we find that sense and antisense transcript levels are intricately linked while other NATs appear to be regulated independently of mRNA transcription. Our deep strand-specific dataset will provide a valuable resource for the precise determination of expression levels as it separates sense from antisense transcript levels, which we find to often significantly differ. In addition, the extensive novel data on 3′ UTR length will allow others to perform searches for regulatory motifs in the UTRs and help understand post-translational regulation in P. falciparum.
Im Rahmen der Dissertation wurde die Reaktivitat des terminalen Mangan-Borylenkomplexes gegenüber verschiedenen polaren Doppelbindungssystemen, koordinativ ungesattigten Übergangsmetallfragmenten sowie Lewis-Basen untersucht. Mithilfe der spektroskopischen und strukturellen Daten der dabei synthetisierten Verbindungen konnten neue Erkenntnisse uber die Natur der Metall-Bor-Bindung erlangt werden.
In mammals, KSR1 functions as an essential scaffold that coordinates the assembly of RAF/MEK/ERK complexes and regulates intracellular signal transduction upon extracellular stimulation. Aberrant activation of the equivalent MAPK signaling pathway has been implicated in multiple human cancers and some developmental disorders. The mechanism of KSR1 regulation is highly complex and involves several phosphorylation/dephosphorylation steps. In the present study, a number of novel in vivo phosphorylation sites were detected in mKSR1 by use of mass spectrometry analysis. Among others, Tyr728 was identified as a unique regulatory residue phosphorylated by LCK, a Src kinase family member. To understand how phosphorylation of Tyr728 may regulate the function of KSR1 in signal transduction and cellular processes, structural modeling and biochemical studies were integrated in this work.
Computational modeling of the mKSR1(KD) protein structure revealed strong hydrogen bonding between phospho-Tyr728 and the residues surrounding Arg649. Remarkably, this pattern was altered when Tyr728 was non-phosphorylated or substituted. As confirmed by biochemical analysis, Arg649 may serve as a major anchor point for phospho-Tyr728 in order to stabilize internal structures of KSR1. In line with the protein modeling results, mutational studies revealed that substitution of Tyr728 by phenylalanine leads to a less compact interaction between KSR1 and MEK, a facilitated KSR1/B-RAF binding and an increased phosphorylation of MEK in complex with KSR1. From these findings it can be concluded that phospho-Tyr728 is involved in tightening the KSR1/MEK interaction interface and in regulating the phosphorylation of KSR1-bound MEK by either RAF or KSR1 kinases.
Beside the Tyr728, Ser722 was identified as a novel regulatory phosphorylation site. Amino acid exchanges at the relevant position demonstrated that Ser722 regulates KSR1-bound MEK phosphorylation without affecting KSR1/MEK binding per se. Due to its localization, Ser722 might consequently control the catalytic activity of KSR1 by interfering with the access of substrate (possibly MEK) to the active site of KSR1 kinase. Together with Ser722, phosphorylated Tyr728 may further positively affect the kinase activity of KSR1 as a consequence of its vicinity to the activation and catalytic loop in the KSR1(KD). As revealed by structural modeling, phospho-Tyr728 builds a hydrogen bond with the highly conserved Lys685. Consequently, phospho-Tyr728 has a stabilizing effect on internal structures involved in the catalytic reaction and possibly enhances the phosphate transfer within the catalytic cleft in KSR1. Considering these facts, it seems very likely that the LCK-dependent phosphorylation of Tyr728 plays a crucial role in the regulation of KSR1 catalytic activity.
Results of fractionation and morphology analyses revealed that KSR1 recruits LCK to cytoskeleton for its phosphorylation at Tyr728 suggesting that this residue may regulate cytoskeleton dynamics and, consequently, cell motility. Beside that, phosphorylation of Tyr728 is involved in the regulation of cell proliferation, as shown by a significantly reduced population doubling time of KSR1-Y728F cells compared to cells expressing wild type KSR1.
Taken together, tyrosine phosphorylation in KSR1 uncovers a new link between Src family kinases and MAPK signaling. Tyr728, the novel regulatory phosphorylation site in murine KSR1, may coordinate the transition between the scaffolding and the catalytic function of KSR1 serving as a control point used to fine-tune cellular responses.
We investigated the ionization profiles for open-ring (OR) and closed-ring (CR) forms of midazolam and drug-binding modes with heptakis-(2,3,6-tri-O-methyl)-β-cyclodextrin (trimethyl-β-cyclodextrin; TRIMEB) using molecular modeling techniques and quantum mechanics methods. The results indicated that the total net charges for different molecular forms of midazolam tend to be cationic for OR and neutral for CR at physiological pH levels. The thermodynamic calculations demonstrated that CR is less water-soluble than OR, mainly due to the maximal solvation energy (ΔG(CR)(solv = -9.98 kcal·mol ⁻¹), which has a minimal ΔG(OR)(solv) of -67.01 kcal·mol⁻¹. A cell viability assay did not detect any signs of TRIMEB and OR/CR-TRIMEB complex toxicity on the cEND cells after 24 h of incubation in either Dulbecco's Modified Eagles Medium or in heat-inactivated human serum. The molecular docking studies identified the more flexible OR form of midazolam as being a better binder to TRIMEB with the fluorophenyl ring introduced inside the amphiphilic cavity of the host molecule. The OR binding affinity was confirmed by a minimal Gibbs free energy of binding (ΔG(bind)) value of -5.57 ± 0.02 kcal·mol⁻¹, an equilibrium binding constant (K(b)) of 79.89 ± 2.706 μM, and a ligand efficiency index (LE(lig)) of -0.21 ± 0.001. Our current data suggest that in order to improve the clinical applications of midazolam via its complexation with trimethyl-β-cyclodextrin to increase drug's overall aqueous solubility, it is important to concern the different forms and ionization states of this anesthetic. The cytochrome P450 (CYP)3A4 enzyme affects the metabolism of most drug-like substances, and its inhibition may influence drug safety. Modulation of CYP3A4 by flavonoids, such as anthocyanins, has been shown to inhibit the mutagenic activity of mammalian cells. Considering the previous investigations addressing CYP3A4 inhibition by these substances, we studied the three-dimensional quantitative structure-activity relationship (3D-QSAR) in a series of anthocyanin derivatives as CYP3A4 inhibitors. For the training dataset (n=12), comparative molecular field analysis (CoMFA) and comparative molecular similarity index analysis (CoMSIA) yielded crossvalidated and non-crossvalidated models with a q (2) of 0.795 (0.687) and r (2) of 0.962 (0.948), respectively. The models were also validated by an external test set of four compounds with r (2) of 0.821 (CoMFA) and r (2) of 0.812 (CoMSIA). The binding affinity modes associated with experimentally derived IC50 (half maximal inhibitory concentration) values were confirmed by molecular docking into the CYP3A4 active site with r (2) of 0.66. The results obtained from this study are useful for a better understanding of the effects of anthocyanin derivatives on inhibition of carcinogen activation and cellular DNA damage.
Retroviral vectors are potent tools for gene delivery and various biomedical applications. To accomplish a gene transfer task successfully, retroviral vectors must effectively transduce diverse cell cultures at different phases of a cell cycle. However, very promising retroviral vectors based on the foamy viral (FV) backbone lack the capacity to efficiently transduce quiescent cells. It is hypothesized that this phenomenon might be explained as the inability of foamy viruses to form a pre-integration complex (PIC) with nuclear import activity in growth-arrested cells, which is the characteristic for lentiviruses (HIV-1). In this process, the HIV-1 central polypurine tract (cPPT) serves as a primer for plus-strand synthesis to produce a “flap” element and is believed to be crucial for the subsequent double-stranded cDNA formation of all retroviral RNA genomes. In this study, the effects of the lentiviral cPPT element on the FV transduction potential in dividing and growth-arrested (G1/S phase) adenocarcinomic human alveolar basal epithelial (A549) cells are investigated by experimental and theoretical methods. The results indicated that the HIV-1 cPPT element in a foamy viral vector background will lead to a significant reduction of the FV transduction and viral titre in growth-arrested cells due to the absence of PICs with nuclear import activity.
The blood–brain barrier choline transporter (BBB-ChT) may have utility as a drug delivery vector to the central nervous system (CNS). We therefore initiated molecular docking studies with the AutoDock and AutoDock Vina (ADVina) algorithms to develop predictive models for compound screening and to identify structural features important for binding to this transporter. The binding energy predictions were highly correlated with r2=0.88, F=692.4, standard error of estimate =0.775, and P-value<0.0001 for selected BBB-ChT-active/inactive compounds (n=93). Both programs were able to cluster active (Gibbs free energy of binding <−6.0 kcal*mol-1) and inactive (Gibbs free energy of binding >−6.0 kcal*mol-1) molecules and dock them significantly better than at random with an area under the curve value of 0.86 and 0.84, respectively. In ranking smaller molecules with few torsional bonds, a size-related bias in scoring producing false-negative outcomes was detected. Finally, important blood–brain barrier parameters, such as the logBBpassive and logBBactive values, were assessed to predict compound transport to the CNS accurately. Knowledge gained from this study is useful to better understand the binding requirements in BBB-ChT, and until such time as its crystal structure becomes available, it may have significant utility in developing a highly predictive model for the rational design of drug-like compounds targeted to the brain.
P-glycoprotein (P-gp) is an ATP (adenosine triphosphate)-binding cassette transporter that causes multidrug resistance of various chemotherapeutic substances by active efflux from mammalian cells. P-gp plays a pivotal role in limiting drug absorption and distribution in different organs, including the intestines and brain. Thus, the prediction of P-gp–drug interactions is of vital importance in assessing drug pharmacokinetic and pharmacodynamic properties. To find the strongest P-gp blockers, we performed an in silico structure-based screening of P-gp inhibitor library (1,300 molecules) by the gradient optimization method, using polynomial empirical scoring (POLSCORE) functions. We report a strong correlation (r2=0.80, F=16.27, n=6, P<0.0157) of inhibition constants (Kiexp or pKiexp; experimental Ki or negative decimal logarithm of Kiexp) converted from experimental IC50 (half maximal inhibitory concentration) values with POLSCORE-predicted constants (KiPOLSCORE or pKiPOLSCORE), using a linear regression fitting technique. The hydrophobic interactions between P-gp and selected drug substances were detected as the main forces responsible for the inhibition effect. The results showed that this scoring technique might be useful in the virtual screening and filtering of databases of drug-like compounds at the early stage of drug development processes.
In lymphocytes, the three NFAT factors NFATc1 (also designated as NFAT2), NFATc2 (NFAT1), and NFATc3 (NFAT4 or NFATx) are expressed and are the targets of immune receptor signals, which lead to a rapid rise of intracellular Ca++, the activation of phosphatase calcineurin, and to the activation of cytosolic NFATc proteins. In addition to rapid activation of NFAT factors, immune receptor signals lead to accumulation of the short NFATc1/αA isoform in lymphocytes which controls their proliferation and survival. In this mini-review, we summarize our current knowledge on the structure and transcription of the Nfatc1 gene in lymphocytes, which is controlled by two promoters, two poly A addition sites and a remote downstream enhancer. The Nfatc1 gene resembles numerous primary response genes (PRGs) induced by LPS in macrophages. Similar to the PRG promoters, the Nfatc1 promoter region is organized in CpG islands, forms DNase I hypersensitive sites, and is marked by histone tail modifications before induction. By studying gene induction in lymphocytes in detail, it will be important to elucidate whether the properties of the Nfatc1 induction are not only typical for the Nfatc1 gene but also for other transcription factor genes expressed in lymphocytes.
Autoimmune myositis encompasses various myositis-overlap syndromes, each being identified by the presence of serum marker autoantibodies. We describe a novel myositis-overlap syndrome in 4 patients characterized by the presence of a unique immunologic marker, autoantibodies to nuclear pore complexes. The clinical phenotype was characterized by prominent myositis in association with erosive, anti-CCP, and rheumatoid factor-positive arthritis, trigeminal neuralgia, mild interstitial lung disease, Raynaud phenomenon, and weight loss. The myositis was typically chronic, relapsing, and refractory to corticosteroids alone, but remitted with the addition of a second immuno-modulating drug. There was no clinical or laboratory evidence for liver disease. The prognosis was good with 100% long-term survival (mean follow-up 19.5 yr). By indirect immunofluorescence on HEp-2 cells, sera from all 4 patients displayed a high titer of antinuclear autoantibodies (ANA) with a distinct punctate peripheral (rim) fluorescent pattern of the nuclear envelope characteristic of nuclear pore complexes. Reactivity with nuclear pore complexes was confirmed by immunoelectron microscopy. In a cohort of 100 French Canadian patients with autoimmune myositis, the nuclear pore complex fluorescent ANA pattern was restricted to these 4 patients (4%). It was not observed in sera from 393 adult patients with systemic sclerosis (n = 112), mixed connective tissue disease (n = 35), systemic lupus (n = 94), rheumatoid arthritis (n = 45), or other rheumatic diseases (n = 107), nor was it observed in 62 normal adults. Autoantibodies to nuclear pore complexes were predominantly of IgG isotype. No other IgG autoantibody markers for defined connective tissue diseases or overlap syndromes were present, indicating a selective and highly focused immune response. In 3 patients, anti-nuclear pore complex autoantibody titers varied in parallel with myositis activity, suggesting a pathogenic link to pathophysiology. The nuclear pore complex proteins, that is, nucleoporins (nup), recognized by these sera were heterogeneous and included Nup358/RanBP2 (n = 2 patients), Nup90 (n = 1), Nup62 (n = 1), and gp210 (n = 1). Taken together the data suggest that nup autoantigens themselves drive the anti-nup autoimmune response. Immunogenetically, the 4 patients shared the DQA1*0501 allele associated with an increased risk for autoimmune myositis. In conclusion, we report an apparent novel subset of autoimmune myositis in our population of French Canadian patients with connective tissue diseases. This syndrome is recognized by the presence of a unique immunologic marker, autoantibodies to nuclear pore complexes that react with nups, consistent with an "anti-nupsyndrome.''
Background: Myofibrillar myopathies (MFM) are a group of phenotypically and genetically heterogeneous neuromuscular disorders, which are characterized by protein aggregations in muscle fibres and can be associated with multisystemic involvement.
Methods: We screened a large cohort of 38 index patients with MFM for mutations in the nine thus far known causative genes using Sanger and next generation sequencing (NGS). We studied the clinical and histopathological characteristics in 38 index patients and five additional relatives (n = 43) and particularly focused on the associated multisystemic symptoms.
Results: We identified 14 heterozygous mutations (diagnostic yield of 37%), among them the novel p. Pro209Gln mutation in the BAG3 gene, which was associated with onset in adulthood, a mild phenotype and an axonal sensorimotor polyneuropathy, in the absence of giant axons at the nerve biopsy. We revealed several novel clinical phenotypes and unusual multisystemic presentations with previously described mutations: hearing impairment with a FLNC mutation, dysphonia with a mutation in DES and the first patient with a FLNC mutation presenting respiratory insufficiency as the initial symptom. Moreover, we described for the first time respiratory insufficiency occurring in a patient with the p. Gly154Ser mutation in CRYAB. Interestingly, we detected a polyneuropathy in 28% of the MFM patients, including a BAG3 and a MYOT case, and hearing impairment in 13%, including one patient with a FLNC mutation and two with mutations in the DES gene. In four index patients with a mutation in one of the MFM genes, typical histological findings were only identified at the ultrastructural level (29%).
Conclusions: We conclude that extraskeletal symptoms frequently occur in MFM, particularly cardiac and respiratory involvement, polyneuropathy and/or deafness. BAG3 mutations should be considered even in cases with a mild phenotype or an adult onset. We identified a genetic defect in one of the known genes in less than half of the MFM patients, indicating that more causative genes are still to be found. Next generation sequencing techniques should be helpful in achieving this aim.

Ein besonderer Fokus liegt dabei auf den zahlreichen Formen von Intertextualität.
Mammalian haloacid dehalogenase (HAD)-type phosphatases are an emerging family of enzymes with important functions in physiology and disease. HAD phosphatases can target diverse metabolites, lipids, DNA, and serine/threonine or tyrosine phosphorylated proteins with often high specificity (Seifried et al., 2013). These enzymes thus markedly enlarge the repertoire and substrate spectrum of mammalian phosphatases. However, the basis of HAD phosphatase substrate specificity is still elusive and a number of mammalian HAD phosphatases remain uncharacterized to date. This study characterizes the biochemical and structural properties of AUM (aspartate-based, ubiquitous, Mg2+-dependent phosphatase), a previously unexplored mammalian HAD phosphatase.
In vitro phosphatase assays of purified, recombinant AUM showed phosphatase activity towards para-nitrophenyl phosphate and adenine and guanine nucleotide di- and triphosphates. Inhibitor studies indicated that similar to other HAD superfamily members, the AUM-catalyzed dephosphorylation reaction proceeds via a pentacovalent phosphoaspartate intermediate. In line with an aspartate-based catalytic mechanism, AUM was insensitive to inhibitors of serine/threonine phosphatases. The characterization of the purified recombinant murine enzyme also revealed that AUM exists in equilibrium between dimers and tetramers.
AUM was identified as the closest, yet functionally distinct relative of chronophin, a pyridoxal 5’-phosphate and serine/threonine-directed phosphatase. Phylogenetic analyses showed that AUM and chronophin evolved via duplication of an ancestral gene at the origin of the vertebrates. In contrast to chronophin, AUM acts as a tyrosine-specific HAD-type phosphatase in vitro and in cells. To elucidate how AUM and chronophin achieve these distinct substrate preferences, comparative evolutionary analyses, biochemical approaches and structural analyses were combined. Swapping experiments of less homologous regions between AUM and chronophin were performed. The mutational analysis revealed residues important for AUM catalysis and specificity. A single differently conserved residue in the cap domain of AUM or chronophin is crucial for phosphatase specificity (AUML204, chronophinH182). The X-ray crystal structure of the AUM cap fused to the catalytic core of chronophin (CAC, PDB: 4BKM) was solved to 2.65 Å resolution. It presents the first crystal structure of the murine AUM capping domain. The detailed view of the catalytic clefts of AUM and chronophin reveals the structural basis of the divergent substrate specificities. These presented findings provide insights into the design principles of capped HAD phosphatases and show that their substrate specificity can be encoded by a small number of predictable residues. In addition, the catalytic properties of AUM were investigated, identifying a mechanism of reversible oxidation regulating the activity of AUM in vitro. AUM phosphatase activity is inhibited by oxidation and can be recovered by reduction. The underlying molecular mechanism was revealed by mutational analyses. The cysteines C35, C104 and C243, located in the AUM core domain, are responsible for the inhibition of AUM by oxidation. C293 mediates the redox-dependent tetramerization of AUM in vitro. Based on the chronophin and CAC structure, a direct impact of the oxidation of C35 on the nucleophile D34 is proposed. In addition, a redox-dependent disulfide bridge (C104, C243), connecting the core and cap domain of AUM may be important for an open/close-mechanism. This hypothesis is supported by CD spectroscopy experiments that demonstrate a structural change in AUM upon reduction. These data present the first evidence for the regulation of AUM catalysis by reversible oxidation. This finding is so far unique in the field of HAD phosphatases.
In this context, the first cell-based AUM activity assay was developed. For this, the artificial substrate pNPP was combined with the reducing agent DTT to create a specific AUM activity readout. This fractionation-based assay is the first tool to differentiate between cell lines or tissues with different AUM concentrations or activities.
Taken together, the presented biochemical characterization reveals the specificity determinants and catalytic properties of AUM. General insights into structural determinants of mammalian HAD phosphatase substrate recognition are provided and reversible oxidation as possible regulatory mechanism for AUM is proposed. These findings constitute a framework for further functional analyses to elucidate the biomedical importance of AUM.
Fbw7, the substrate recognition subunit of SCF(Fbw7) ubiquitin ligase, mediates the turnover of multiple proto-oncoproteins and promotes its own degradation. Fbw7-dependent substrate ubiquitination is antagonized by the Usp28 deubiquitinase. Here, we show that Usp28 preferentially antagonizes autocatalytic ubiquitination and stabilizes Fbw7, resulting in dose-dependent effects in Usp28 knockout mice. Monoallelic deletion of Usp28 maintains stable Fbw7 but drives Fbw7 substrate degradation. In contrast, complete knockout triggers Fbw7 degradation and leads to the accumulation of Fbw7 substrates in several tissues and embryonic fibroblasts. On the other hand, overexpression of Usp28 stabilizes both Fbw7 and its substrates. Consequently, both complete loss and ectopic expression of Usp28 promote Ras-driven oncogenic transformation. We propose that dual regulation of Fbw7 activity by Usp28 is a safeguard mechanism for maintaining physiological levels of proto-oncogenic Fbw7 substrates, which is equivalently disrupted by loss or overexpression of Usp28.
Diese Arbeit befasst sich mit der Verwendung einer solvensfreien Synthesese zur Darstellung koordinationspolymerer Verbindungen basierend auf den 3d-Übergangsmetallen Mn, Fe, Co und Zn und N-heterozyklischen, aminischen Ligandenschmelzen. Die eingesetzten Metalle wurden entweder in ihrer elementaren Form in einer bereits bekannten, redox-basierten Synthese oder als Metallchloride in einer adduktbasierten Synthese verwendet. Synthesen mit den Liganden Imidazol (ImH), Pyrazol (PzH), Benzimidazol (bzImH), 1,2,3-Triazol (1,2,3-TzH), 1,2,4,-Triazol (1,2,4-TzH) und Piperazin (pipz) führten zu verschiedenen neuen Verbindungen und Kristallstrukturen, speziell bei Verwendung des Liganden pipz. Die erhaltenen Produkte wurden für Untersuchungen von Struktureigenschafts-beziehungen hinsichtlich des Polarisationsverhaltens und im Falle der 1,2,3-Triazolate für eine Untersuchung von Photolumineszenzeigenschaften in Abhängigkeit von Physisorptionsprozessen verwendet.
Unter den Piperazin-Verbindungen findet sich eine starke Varianz im Bereich des polymeren Charakters und des Vernetzungsgrades, welcher von Komplexen (null-dimensional) wie [CoCl2(pipz)] (26) and [ZnCl2(pipz)] (27) über Stränge (eindimensional) wie 1∞[ÜMCl2(py)2(pipz)] (ÜM = Mn (29), Fe (30), Co (31)) bis hin zu Netzen (zweidimensional) wie 2∞[Fe2Cl4(pipz)3]∙(pipz) (25) und Gerüsten (dreidimensional) wie 3∞[FeCl2(pipz)] (32) reicht. Thermoanalyse und darauf basierende Veränderungen in den Reaktionsbedingungen ermöglichen es, die Zusammenhänge zwischen den einzelnen Phasen aufzuzeigen und ihre thermodynamischen Stabilitäten zu untersuchen, was nicht nur für die Piperazin-Verbindungen, sondern auch für andere Systeme gelang, von denen die größte Gruppe die Imidazolate des Typs 3∞[ÜM3(Im)6(ImH)2] (ÜM = Mn (1), Fe (2), Co (3)) waren.
Der erfolgreiche Zusatz von geringen Mengen Mn2+ in das MOF 3∞[Zn(1,2,3-Tz)2] (21) führt zur Bildung der Phase 3∞[Zn0,9Mn0,1(1,2,3-Tz)2] (22) und zur Aktivierung von Photolumineszenz unter Beibehaltung von Mikroporosität. Das erhaltene lumineszierende, mikroporöse Gerüst wurde als Modellsystem für die Analyse von Lumineszenzänderungen während der Physisorption verschiedener Analysegase und bei unterschiedlichen Temperaturen verwendet. Die erhaltenen Ergebnisse können eine wichtige Rolle bei der Charakterisierung potentieller Sensormaterialien spielen und sind weiterhin ein möglicher Ausgangspunkt für mechanistische Überlegungen bei diesen hochinteressanten Struktureigenschaftsbeziehungen.
Die erfolgreiche Redoxreaktion elementarer Metalle mit den Liganden ImH, PzH, bzImH und 1,2,3-TzH erlaubte eine Untersuchung dielektrischer Eigenschaften einer Gruppe von verwandten Verbindungen bezüglich verschiedener Probenformen (Pulverschüttungen gegenüber gepressten Pellets) und Strukturmerkmale wie Rigidität der Kristallstrukturen. Diese Eigenschaften wurden darüber hinaus auch temperaturabhängig in einem Bereich von -100 °C bis 150 °C untersucht. Weiterhin konnte mit Untersuchungen an den MOFs 3∞[ÜM(1,2,3-Tz)2] (ÜM = Mn (18), Fe (19), Co (20), Zn (21)) auch ein Beitrag zu einem Feld der MOF-Chemie geleistet werden, welches bereits stark diskutiert wurde, jedoch bislang nur sehr schwach erforscht ist.
Die Extraktion von Metadaten aus historischen Dokumenten ist eine zeitintensive, komplexe und höchst fehleranfällige Tätigkeit, die üblicherweise vom menschlichen Experten übernommen werden muss. Sie ist jedoch notwendig, um Bezüge zwischen Dokumenten herzustellen, Suchanfragen zu historischen Ereignissen korrekt zu beantworten oder semantische Verknüpfungen aufzubauen. Um den manuellen Aufwand dieser Aufgabe reduzieren zu können, sollen Verfahren der Named Entity Recognition angewendet werden. Die Klassifikation von Termen in historischen Handschriften stellt jedoch eine große Herausforderung dar, da die Domäne eine hohe Schreibweisenvarianz durch unter anderem nur konventionell vereinbarte Orthographie mit sich bringt. Diese Arbeit stellt Verfahren vor, die auch in komplexen syntaktischen Umgebungen arbeiten können, indem sie auf Informationen aus dem Kontext der zu klassifizierenden Terme zurückgreifen und diese mit domänenspezifischen Heuristiken kombinieren. Weiterhin wird evaluiert, wie die so gewonnenen Metadaten genutzt werden können, um in Workflow-Systemen zur Digitalisierung historischer Handschriften Mehrwerte durch Heuristiken zur Produktionsfehlererkennung zu erzielen.
Krebs ist trotz der fortgeschrittenen Therapiemöglichkeiten eine Diagnose, die eine große emotionale Belastung für die betroffenen Menschen darstellt. Im Rahmen der Psychoonkologie wurde in den vergangenen Jahren den psycho-sozialen Problemen Krebserkrankter immer mehr Beachtung geschenkt. Dennoch werden im klinischen Alltag manifeste psychische Störungen bei onkologischen Patienten zu selten erkannt und folglich nicht therapiert. Um dieser Unterversorgung entgegen zu wirken, wird nach Prädiktoren psychischer Störungen bei Krebserkrankten geforscht, mit deren Hilfe betroffene Personen leichter erfasst werden könnten.
Auch die vorliegende Arbeit diente dem Ziel, Prädiktoren einer psychischen Erkrankung bei Krebserkrankten zu ermitteln. Hierzu wurden mögliche Zusammenhänge zwischen psychischer Störung und soziodemographischen, medizinischen sowie psychosozialen Merkmale geprüft. Daneben sollte die Prävalenz psychischer Störungen in der Stichprobe erhoben werden.
Der Untersuchung liegen die Daten aus dem Studienzentrum Würzburg zugrunde, die im Rahmen einer von der Deutschen Krebshilfe unterstützten, multizentrischen Studie („Prävalenz psychosozialer Belastungen, psychischer Störungen und ihr Behandlungsbedarf bei Tumorpatienten“) erhoben wurden. An der Untersuchung nahmen in Würzburg insgesamt 742 Krebserkrankte mit unterschiedlichen Tumorlokalisationen, Tumorstadien und Behandlungssettings teil. Gemäß eines zweistufigen Studiendesigns sollte bei allen Screening-positiven (PHQ ≥ 9) und der Hälfte der Screening-negativen Patienten (PHQ < 9), folglich bei 437 Studienteilnehmern, ein diagnostisches Interview zur Erfassung psychischer Störungen (Composite International Diagnostic Interview) durchgeführt werden. Zusätzlich wurden soziodemographische, medizinische und psychosoziale Merkmale erhoben. Mögliche Zusammenhänge zwischen den erhobenen Variablen und einer psychischen Störung wurden bivariat sowie multivariat mit einer logistischen Regression untersucht.
Die ermittelte 1 Monatsprävalenz psychischer Störungen bei Krebserkrankten betrug 18,5%, die 12 Monatsprävalenz 24,4% und die Lebenszeitprävalenz 42,0%. Bei Betrachtung des 1 Monatsintervalls waren Angststörungen mit 11,2%, affektive Störungen mit 5,9% und somatoforme Störungen mit 3,1% am häufigsten vertreten.
Als unabhängige Prädiktoren einer psychischen Störung bei Krebserkrankten stellten sich ein weibliches Geschlecht, jüngeres Alter, eine längere Zeit seit aktueller Diagnosestellung, eine stärker negative soziale Interaktion (SSUK 8) sowie das Vorliegen eines praktischen Problems (NCCN-Distress-Problemliste) heraus. Desweiteren ergaben sich signifikante bivariate Zusammenhänge zwischen einer psychischen Störung und den folgenden Merkmalen: niedrigere subjektiv eingeschätzte körperliche Funktionsfähigkeit (EORTC QLQ-C30), Angabe von Schmerzen im Zusammenhang mit der Krebserkrankung, abgeschlossene Strahlentherapie, allgemeine psychische Belastung (PHQ-9, HADS, GAD 7, NCCN-Distress-Thermometer), Vorliegen eines familiären Problems (NCCN-Distress-Problemliste), Inanspruchnahme psychotherapeutischer / psychologischer / seel-sorgerischer / sozialrechtlicher Unterstützung sowie Nutzen eines Internetforums mit Betroffenen.
Die in der vorliegenden Arbeit ermittelte Punktprävalenz psychischer Störungen bei Krebserkrankten steht weitgehend in Einklang mit den bisherigen Ergebnissen aus der Forschung. Sie deckt sich damit auch in etwa mit der Häufigkeit in der Allgemeinbevölkerung. Allerdings scheinen Angststörungen im onkologischen Setting etwas häufiger aufzutreten.
Wie in der Allgemeinbevölkerung sind auch im onkologischen Bereich ein weibliches Geschlecht und jüngeres Alter Risikofaktoren einer psychischen Störung. Medizinische Faktoren scheinen bezüglich der Voraussage einer psychischen Erkrankung von untergeordneter Bedeutung zu sein. Psychosoziale Prädiktoren bieten zugleich mögliche Ansatzpunkte für eine bessere Versorgung von Krebserkrankten, die an einer komorbiden psychischen Störung leiden.
Über die Nervensonographie wurde bereits in den 1980er Jahren erstmals berichtet. Die rasche Weiterentwicklung der Technik hat dazu geführt, dass es inzwischen zahlreiche Fallberichte und einige Studien gibt, die sich mit der Darstellung peripherer Nerven durch Ultraschall als Mittel zur Diagnose verschiedener Nervpathologien beschäftigen. Besonders bei der Diagnostik des epidemiologisch häufigen Karpaltunnelsyndroms ist die sonographische Beurteilung des N. medianus in dieser Lokalisation vielerorts bereits etablierter Bestandteil der Diagnostik.
In der hier vorgelegten Studie sollte der Stellenwert der B-Bild-Sonographie peripherer Nerven am Unterschenkel für die Diagnose einer Vaskulitischen Neuropathie geprüft werden. Dazu musste zunächst die Ultraschalluntersuchung spezieller Nerven am Unterschenkel etabliert werden. Diese ist technisch deutlich anspruchsvoller als die Darstellung von Karpaltunnelsyndrom oder Armplexus.
Durch die fünfmalige Untersuchung zehn junger Personen wurden ultraschalltechnisch leicht reproduzierbar anatomisch auffindbare und Anisotropie-vermeidende Abschnitte von N. suralis, N. peroneus communis, profundus, superfcialis und N. tibialis definiert und als Messpunkte der Studie zu Grunde gelegt.
In die von der Ethikkommission der Medizinischen Fakultät positiv beschiedene Studie wurden 26 Patienten/-innen, die klinisch und elektrophysiologisch nachgewiesen eine Polyneuropathie hatten und bei denen zur Ursachendiagnostik eine Biopsie und histologische Aufarbeitung des N. suralis durchgeführt wurde (Abteilung für Neuropathologie des Pathologischen Instituts der Universität sowie Histologielabor der Neurologischen Universitätsklinik), sowie 26 Kontrollpersonen ohne klinischen Hinweis auf eine Polyneuropathie nach informiertem Einverständnis aufgenommen. Für jede/-n Patienten/-in wurde ein/-e Proband/-in gleichen Geschlechts mit einem Altersunterschied von höchstens fünf Jahren in die Kontrollgruppe aufgenommen. Alle 52 Untersuchten mussten erwachsen und 160 – 180 cm groß sein.
Bei allen Patienten/-innen und Kontrollpersonen wurden jeweils der GD, der KD, der LD und die QSF des N. suralis am unteren Drittel der Wade und distal im Bereich des Außenknöchels, des N. tibialis nahe des Innenknöchels, des N. peroneus communis im Bereich des Fibulaköpfchens, des N. peroneus profundus am Fußrücken und nahe der Großzehen und des N. peroneus superficialis im Bereich des distalen Schienbeins bestimmt.
Alle gesuchten Nerven waren bei allen Versuchspersonen eindeutig identifizierbar. Die Untersuchungen wurden durch eine Untersucherin mit demselben Gerät, geblindet für das Ergebnis der Histologie, durchgeführt. Das gewährleistete eine Konstanz in der schwierigen und mit Unsicherheiten behafteten Messung der Nervenstrukturen, was ausführlich diskutiert wird.
Ein statistisch signifikanter Unterschied zwischen den sonographisch erhobenen Messdaten der PNP-Gruppe und der Kontrollgruppe konnte bei 20 der 28 Parameter gezeigt werden. Bei 11 der 28 Parameter konnte zwischen Vaskulitis-Patienten/-innen und allen anderen, also PNP-Patienten/-innen und der Kontrollgruppe, ein statistisch signifikanter Unterschied festgestellt werden. Außerdem ergab die statistische Analyse bei drei der 28 Messgrößen einen statistisch signifikanten Unterschied zwischen Patienten/-innen mit und ohne Demyelinisierung des N. suralis in der feingeweblichen Untersuchung.
Die sonographischen Ergebnisse der Vakulitis-Patienten/-innen unterschieden sich nicht von denen der PNP-Patienten/-innen mit anderer Ätiologie. Es wurde auch kein statistisch signifikanter Unterschied zwischen den Werten der PNP-Patienten/-innen mit und ohne histologisch gesicherte entzündliche Komponente beobachtet.
Gemäß der histologischen Untersuchung der Biopsate wurde bei sechs Patienten/-innen eine Vaskulitis diagnostiziert. Bei fünf dieser Patienten/-innen fielen teilweise Kalibersprünge im Sinne einer Zunahme der QSF oder Abflachung im Verlauf des N. suralis, N. peroneus superficialis und N. peroneus communis auf. Aber auch bei Patienten/-innen mit einer anderen Form der Polyneuropathie und einigen Kontrollpersonen waren Besonderheiten im sonographischen Bild einzelner Nerven zu beobachten.
Mit der vorgelegten Untersuchung konnte zwar nicht gezeigt werden, dass die Nervensonographie einen Beitrag zur differentialdiagnostischen Abgrenzung Vaskulitischer Polyneuropathien leisten kann, der den Goldstandard invasiver Nervenbiopse entbehrlich machen könnte. Das war bei der histologischen Unterschiedlichkeit der beschädigten Nervenanatomie bei Vaskulitis aber auch nicht ernsthaft zu erwarten.
Die vorgelegte Arbeit zeigt aber auch, dass kranke periphere Nerven von gesunden Nerven im Ultraschall unterscheidbar sind, wenn man wie hier systematisch mit 28 Parametern an sieben Messpunkten untersucht. Dies allerdings dauert auch für einen Geübten 40 bis 60 Minuten, so dass die Polyneuropathiediagnostik oder gar Differentialdiagnostik mittels Ultraschall aktuell noch als Forschungsinstrument an großen Fallzahlen anzusehen ist.
Dabei wird es künftig für die Gruppenbildung der sonographisch Untersuchten neben ätiologischer und histologischer Gruppenbildung darauf ankommen, das Krankheitsbild besser zu definieren, d.h. Ausmaß von Demyelinisierung, Remyelinisierung und axonalem Untergang in geeignete Skalen zu fassen.
Auch die Magnetresonanztomographie stellt eine Option als diagnostischer Baustein bei Vaskulitischer Polyneuropathie dar. Dieses bildgebende Verfahren kann bereits zur Diagnostik von traumatischen Nervverletzungen, Kompressionensyndromen, Raumforderungen im Bereich der Nerven und Plexusneuritis eingesetzt werden.
Muskeldystrophie Duchenne (DMD) (Xp21.2) und gehört mit 1:3500 männlichen Geburten zu den häufigsten genetisch-determinierten Erkrankungen. DMD ist bis heute nicht heilbar. Die genetische Beratung ist Teil der Betreuung dieser Patienten und bezieht auch die heterozygotenwahrscheinlichkeit weiblicher Angehöriger
mit ein. Für die Risikoberechnung zum Überträgerstatus weiblicher Angehöriger von DMD-Patienten sind neben Stammbauminformationen und Enzymwerten Verhältnisse der Mutationsraten ( k -Werte) essentiell, welche die
unterschiedliche Entstehungswahrscheinlichkeit der einzelnen Mutationstypen (Deletion, Duplikation, Punktmutation) in Spermatogenese oder Oogenese
beschreiben.Die Bestimmung des Verhältnisses k der Mutationsraten zeigte, dass einerseits Deletionen im Dystrophin-Gen viel häufiger großmütterlichen
Ursprungs (k Deletion ≈ 0,26) und andererseits Punktmutationen im Dystrophin-Gen meist großväterlichen Ursprungs (k Punktmutation ≈ 2,8) sind.
The number of fungal infections is rising in Germany and worldwide. These infections are mainly caused by the opportunistic fungal pathogen C. albicans, which especially harms immunocompromised people. With increasing numbers of fungal infections, more frequent and longer lasting treatments are necessary and lead to an increase of drug resistances, for example against the clinically applied therapeutic fluconazole. Drug resistance in C. albicans can be mediated by the Multidrug resistance pump 1 (Mdr1), a membrane transporter belonging to the major facilitator family. However, Mdr1-mediated fluconazole drug resistance is caused by the pump’s regulator, the transcription factor Mrr1 (Multidrug resistance regulator 1). It was shown that Mrr1 is hyperactive without stimulation or further activation in resistant strains which is due to so called gain of function mutations in the MRR1 gene.
To understand the mechanism that lays behind this constitutive activity of Mrr1, the transcription factor should be structurally and functionally (in vitro) characterized which could provide a basis for successful drug development to target Mdr1-mediated drug resistance caused by Mrr1. Therefore, the entire 1108 amino acid protein was successfully expressed in Escherichia coli. However, further purification was compromised as the protein tended to form aggregates, unsuitable for crystallization trials or further characterization experiments. Expression trials in the eukaryote Pichia pastoris neither yielded full length nor truncated Mrr1 protein. In order to overcome the aggregation problem, a shortened variant, missing the N-terminal 249 amino acids named Mrr1 ‘250’, was successfully expressed in E. coli and could be purified without aggregation. Similar to the wild type Mrr1 ‘250’, selected gain of function variants were successfully cloned, expressed and purified with varying yields and with varying purity. The Mrr1 `250’ construct contains most of the described regulatory domains of Mrr1. It was used for crystallization and an initial comparative analysis between the wild type protein and the variants. The proposed dimeric form of the transcription factor, necessary for DNA binding, could be verified for both, the wild type and the mutant proteins. Secondary structure analysis by circular dichroism measurements revealed no significant differences in the overall fold of the wild type and variant proteins. In vitro, the gain of function variants seem to be less stable compared to the wild type protein, as they were more prone to degradation. Whether this observation holds true for the full length protein’s stability in vitro and in vivo remains to be determined. The crystallization experiments, performed with the Mrr1 ‘250’ constructs, led to few small needle shaped or cubic crystals, which did not diffract very well and were hardly reproducible. Therefore no structural information of the transcription factor could be gained so far.
Infections with M. tuberculosis, the causative agent of tuberculosis, are the leading cause of mortality among bacterial diseases. Especially long treatment times, an increasing number of resistant strains and the prevalence of for decades persisting bacteria create the necessity for new drugs against this disease. The cholesterol import and metabolism pathways were discovered as promising new targets and interestingly they seem to play an important role for the chronic stage of the tuberculosis infection and for persisting bacteria.
In this thesis, the 3-ketoacyl-CoA thiolase FadA5 from M. tuberculosis was characterized and the potential for specifically targeting this enzyme was investigated. FadA5 catalyzes the last step of the β-oxidation reaction in the side-chain degradation pathway of cholesterol. We solved the three dimensional structure of this enzyme by X-ray crystallography and obtained two different apo structures and three structures in complex with acetyl-CoA, CoA and a hydrolyzed steroid-CoA, which is the natural product of FadA5. Analysis of the FadA5 apo structures revealed a typical thiolase fold as it is common for biosynthetic and degradative enzymes of this class for one of the structures. The second apo structure showed deviations from the typical thiolase fold. All obtained structures show the enzyme as a dimer, which is consistent with the observed dimer formation in solution. Thus the dimer is likely to be the catalytically active form of the enzyme. Besides the characteristic structural fold, the catalytic triad, comprising two cysteines and one histidine, as well as the typical coenzyme A binding site of enzymes belonging to the thiolase class could be identified. The two obtained apo structures differed significantly from each other. One apo structure is in agreement with the characteristic thiolase fold and the well-known dimer interface could be identified in our structure. The same characteristics were observed in all complex structures. In contrast, the second apo structure followed the thiolase fold only partially. One subdomain, spanning 30 amino acids, was in a different orientation. This reorientation was caused by the formation of two disulfide bonds, including the active site cysteines, which rendered the enzyme inactive. The disulfide bonds together with the resulting domain swap still permitted dimer formation, yet with a significantly shifted dimer interface. The comparison of the apo structures together with the preliminary activity analysis performed by our collaborator suggest, that FadA5 can be inactivated by oxidation and reactivated by reduction. If this redox switch is of biological importance requires further evaluation, however, this would be the first reported example of a bacterial thiolase employing redox regulation.
Our obtained complex structures represent different stages of the thiolase reaction cycle. In some complex structures, FadA5 was found to be acetylated at the catalytic cysteine and it was in complex with acetyl-CoA or CoA. These structures, together with the FadA5 structure in complex with a hydrolyzed steroid-CoA, revealed important insights into enzyme dynamics upon ligand binding and release. The steroid-bound structure is as yet a unique example of a thiolase enzyme interacting with a complex ligand. The characterized enzyme was used as platform for modeling studies and for comparison with human thiolases. These studies permitted initial conclusions regarding the specific targetability of FadA5 as a drug target against M. tuberculosis infection, taking the closely related human enzymes into account. Additional analyses led to the proposal of a specific lead compound based on the steroid and ligand interactions within the active site of FadA5.
An attempt has been made to define the extent to which metabolic flux in central plant metabolism is reflected by changes in the transcriptome and metabolome, based on an analysis of in vitro cultured immature embryos of two oilseed rape (Brassica napus) accessions which contrast for seed lipid accumulation. Metabolic flux analysis (MFA) was used to constrain a flux balance metabolic model which included 671 biochemical and transport reactions within the central metabolism. This highly confident flux information was eventually used for comparative analysis of flux vs. transcript (metabolite). Metabolite profiling succeeded in identifying 79 intermediates within the central metabolism, some of which differed quantitatively between the two accessions and displayed a significant shift corresponding to flux. An RNA-Seq based transcriptome analysis revealed a large number of genes which were differentially transcribed in the two accessions, including some enzymes/proteins active in major metabolic pathways. With a few exceptions, differential activity in the major pathways (glycolysis, TCA cycle, amino acid, and fatty acid synthesis) was not reflected in contrasting abundances of the relevant transcripts. The conclusion was that transcript abundance on its own cannot be used to infer metabolic activity/fluxes in central plant metabolism. This limitation needs to be borne in mind in evaluating transcriptome data and designing metabolic engineering experiments.
Despite evidence that deregulated Notch signalling is a master regulator of multiple myeloma (MM) pathogenesis, its contribution to myeloma bone disease remains to be resolved. Notch promotes survival of human MM cells and triggers human osteoclast activity in vitro. Here, we show that inhibition of Notch through the γ-secretase inhibitor XII (GSI XII) induces apoptosis of murine MOPC315.BM myeloma cells with high Notch activity. GSI XII impairs murine osteoclast differentiation of receptor activator of NF-κB ligand (RANKL)-stimulated RAW264.7 cells in vitro. In the murine MOPC315.BM myeloma model GSI XII has potent anti-MM activity and reduces osteolytic lesions as evidenced by diminished myeloma-specific monoclonal immunoglobulin (Ig)-A serum levels and quantitative assessment of bone structure changes via high-resolution microcomputed tomography scans. Thus, we suggest that Notch inhibition through GSI XII controls myeloma bone disease mainly by targeting Notch in MM cells and possibly in osteoclasts in their microenvironment. We conclude that Notch inhibition is a valid therapeutic strategy in MM.
Proteine bestehen aus einer spezifischen Sequenz verschiedener Aminosäuren, die ihre charakteristische Funktion bestimmt. Die große Variabilität an Aminosäuresequenzen ermöglichte die Evolution einer nahezu unbegrenzten Anzahl an Proteinen. Meistens nehmen diese Schlüsselpositionen ein, von robusten Baustoffen bis hin zu molekularen Maschinen. Daher kann eine Fehlfunktion gravierende Auswirkungen auf das Leben haben, z.B. Krankheiten wie Alzheimer oder Epilepsi. Um die Funktionen und Fehlfunktionen zu verstehen, ist eine umfassende Kenntnis der Proteinfaltung, der Protein-Protein Assoziation, sowie den Dynamiken innerhalb von Proteinen erforderlich. Diese Vorgänge wurden in dieser Arbeit an drei isolierten Proteindomänen durch die Anwendung der Fluoreszenzlöschmechanismen der H-Dimerbildung und des photoinduzierten Elektronentransfers untersucht.
Der entfaltete Zustand der Bindungsdomäne BBL, das Teil des 2-oxo-acid Dehydrogenasekomplexes ist, wurde unter physiologischen Bedingungen mit Zirkulardichroismus (CD) und einer Kombination aus photoinduziertem Elektronentransfer und Fluoreszenzkorrelationsspektroskopie analysiert. Beide Methoden zeigten übereinstimmend anhand von 20 in BBL einzeln eingefügten konservativen Punktmutationen, dass Seitenketteninteraktionen keine Auswirkungen auf die Sekundärstruktur des denaturierten Zustandes, den Ausgangspunkt der Faltung, haben. Mit Hilfe der Dekonvolation der CD-Spektren wurde zudem gezeigt, dass die Reststruktur im denaturierten Zustand der helikalen Proteindomäne von β-Strängen und β-Kehren dominiert wird, die eine entscheidende Funktion bei der Faltung in den nativen Zustand haben könnten.
Die N-terminale Domäne (NTD), der für die Materialforschung hochinteressanten Spinnen-seidenfaser, ist für die Polymerisation des Spinnenseidenfadens auf den pH-Wechsel von pH 7 auf pH 6 hin verantwortlich. Dieser für die Proteinfunktion wichtige Prozess wurde durch die Einbringung eines extrinsischen Fluoreszenzschalters, basierend auf der H-Dimerbildung, mit der Stopped-Flow-Technik untersucht. Es wurde gezeigt, dass die NTDs

mit einer Rate von 3 x 10^8 M-1 s-1 assoziieren und somit nahezu das Geschwindigkeitslimit der Protein-Protein Assoziation erreicht wird. Zwei geladenen Seitenketten, der D39 und D40, kommt eine entscheidende Funktion in dem Prozess zu, da eine Mutation dieser die Assoziation verhindert. Des Weiteren wurde gezeigt, dass sich die NTD auf eine Erhöhung der Ionenstärke entgegengesetzt zu anderen Proteinen verhält: die Dissoziation wird beschleunigt, die Assoziation nicht beeinflusst. Gleiches Verhalten wurde auf den einzelnen Austausch der übrigen protonierbaren Aminosäureseitenketten hin beobachtet, ausgenommen die Mutation der E119, welche die Dissoziation verlangsamt. Daher scheint der makromolekulare Dipol, der auf Grund der Ladungsverteilung in der NTD entsteht, die Assoziation maßgeblich zu beeinflussen.
Glutamatrezeptoren sind an der schnellen synaptischen Signalweiterleitung im Nervensys-tem von Vertebraten beteiligt. Die Konformationen der Ligandenbindungsdomäne (LBD) haben dabei entscheidende Auswirkungen auf die Funktion des Gesamtrezeptors. Diese wurden mit einer Kombination aus photoinduziertem Elektronentransfer und Fluoreszenzkorrelationsspektroskopie untersucht. Mit dieser Methode wurde ein dynamisches Bild der gebundenen sowie ungebundenen Form der AMPA-spezifischen Glutamatrezeptor 2-LBD gezeigt. Es wurde zudem gezeigt, dass sich die Dynamiken in Abhängigkeit der Bindung von den Agonisten Glutamat und AMPA, dem partiellen Agonisten Kainate oder Cyclothiazid (CTZ), welches eine Dimerisierung der LBDs bewirkt, unterschiedlich verändern. Dies könnte eine Auswirkung auf die Funktion der Rezeptoren haben.
Die Anwendung der Fluoreszenzlöschmechanismen der H-Dimerbildung und des photoinduzierten Elektronentransfers in dieser Arbeit hat gezeigt, dass diese die Möglichkeit bieten, unterschiedlichste Fragestellungen zu beantworten und so Einblicke in dynamische Funktionsweisen von Proteinen eröffnen. Kombiniert mit etablierten Fluoreszenzmethoden ist es so möglich quantitativ Kinetiken auf unterschiedlichen Zeitskalen zu untersuchen.

Diese Studie vergleicht die Ergebnisse einer 10-Jahres Nachuntersuchung nach arthroskopisch unterstütztem Ersatz des vorderen Kreuzbandes mittels zweier verschiedener autologer Transplantate. Zwischen November 1998 und November 1999 wurden in der Orthopädischen Universitätsklinik König-Ludwig-Haus in Würzburg 62 Studienteilnehmer (45 Männer, 17 Frauen) mit Ruptur des vorderen Kreuzbandes operiert. Bei 31 Patienten erfolgte die ACL-Ersatzplastik mittels Lig. Patellae Transplantat (BTB-Transplantat), bei der anderen Hälfte mittels Semitendinosussehnentransplantat. Die Gruppenzusammensetzung zeigte vor dem Verletzungszeitpunkt keinen signifikanten Unterschied bezüglich Alter, Geschlecht und Aktivitätsniveau. Alle Patienten erhielten die gleiche postoperative Behandlung mit Don-Joy Schiene (Ext/Flex 0/0/90), Teilbelastung bis 20 kg während der ersten 6 Wochen und externe Rehabilitationsmaßnahmen. Nach über 10 Jahren konnten 47 (24 LP/23 ST) Patienten vollständig klinisch untersucht und bei 9 (5 LP/4 ST) weiteren zumindest standardisierte Fragebögen ausgewertet werden. 2 Studienteilnehmer (beide ST) schieden wegen einer Transplantatruptur, ein weiterer Patient (ebenfalls ST) aufgrund einer postoperativen Kniegelenksinfektion vorzeitig aus der Studie aus.
Ergebnisse:
Die klinische Untersuchung ergab bezüglich der verwendeten Parameter IKDC- und Lysholm-Score, Tegener-Aktivitätsskala, KT-1000-Arthrometer-Messung und radiologischer Kontrolle keine signifikanten Unterschiede zwischen den beiden Gruppen. Auch in den verwendeten Fragebögen mit Evaluation der subjektiven Kniefunktion, möglicher Schmerzen im Kniegelenk, Entnahmestellenmorbidität und der Zufriedenheit mit dem Operationsergebnis konnten keine signifikanten Gruppenunterschiede festgestellt werden.
In der vorliegenden Studie fanden sich somit auch über 10 Jahre nach vorderem Kreuzbandersatz keine relevanten Unterschiede oder Vorteile für eines der beiden untersuchten autologen Sehnentransplantate.
Background
While the impact of volatile anaesthetics to induce malignant hyperthermia (MH) is abundantly clear, the role of succinylcholine still remains controversial. To evaluate the influence of succinylcholine on porcine MH events, the authors investigated the hemodynamic and metabolic responses in MH susceptible (MHS) and non-susceptible (MHN) swine following either succinylcholine or halothane application alone or a combination of both substances.
Methods
With approval of the local animal care committee 27 MHS and 30 MHN pigs were anaesthetized and mechanically ventilated. Fiberoptic probes for continuous PCO2 measurement were inserted into the femoral vein and the triceps muscle. Group A received succinylcholine 4 mg/kg, group B incremental doses of halothane (0.5, 1.0 vol%) and group C succinylcholine and halothane simultaneously. Vital signs were recorded continuously.
Results
Prior to drug application measured values did not differ between MHS and MHN. While MHN pigs did not show relevant alterations, succinylcholine, halothane and the combination of both lead to significant hemodynamic and metabolic changes in MHS swine.
Conclusions
Hemodynamic and metabolic alterations following succinylcholine were similar to halothane in MHS pigs. The combination of both pharmacological agents potentiated the observed effects. According to these results succinylcholine acted as an independent and supportive factor during onset of an MH episode.
Ziel dieser Studie war es, mögliche Veränderungen des psychischen Befindens sowie Ausprägungen der Persönlichkeitsdimensionen Extraversion und Neurotizismus im Verlauf der Übungen der Progressiven Muskelrelaxation zu untersuchen. Weiterhin sollte der Einfluss der Persönlichkeitsmerkmale auf die Veränderung des Befindens überprüft werden.
Im theoretischen Teil der Arbeit wurde die Entstehung und Wirkungsweise der PR, Eysencks Theorien und die bisherige Literatur zum Thema dargestellt. Zum Einen konnte festgesellt werden, dass die PR als Teil unterschiedlicher Therapieintervention Einfluss auf das Befinden und die Persönlichkeitsmerkmale hat. Zum Anderen wurde deutlich, dass die Wirkung der PR auf das psychische Befinden und Persönlichkeitsausprägung durch verschiedene, sich gegenseitig bedingende Faktoren, beeinflusst wird. Folgende Hypothesen wurden aufgestellt:
Im Verlauf eines Kurses der PR verbessert sich das psychische Befinden. Zugleich nimmt der Neurotizismuswert ab und der Extraversionswert zu. Außerdem hängt das Ausmaß der Veränderung des psychischen Befindens mit der Persönlichkeits-ausprägung zu Beginn der Durchführung der PR zusammen. Introvertierte und emotional Labile sollen deutlicher von der Wirkung der PR profitieren als Extrovertierte und emotional Stabile.
93 Probanden füllten zu vier Messzeitpunkten vor, während und nach der PR Fragebögen (Eysenck-Persönlichkeits-Inventar, Form A und Eigenschaftswörterliste, Normalform) aus.
Die Ergebnisse zeigen keine Veränderung des Extraversionswertes, wohingegen es nach Beendigung der Übungen zu einer signifikanten Besserung der emotionalen Stabilität kam, was sich in einer Verringerung des Neurotizismuswertes äußerte. Es konnte während und nach Durchführung der PR eine signifikante Verringerung des negativen und Zunahme des positiven Befindens festgestellt werden. Eine Abhängigkeit der Veränderung des Befindens von den Persönlichkeitsdimensionen ließ sich nicht nachweisen. Die Annahme, dass vor allem emotional Labile und Introvertierte eine Verbesserung des Befindens zeigen, ließ sich nicht bestätigen.
Unter Beachtung, dass die tatsächlich involvierten Daten etwa der Hälfte der insgesamt an der Studie teilgenommenen Probanden entsprechen sowie unter Berücksichtigung des Studiendesigns (Ein-Gruppen-Prä-Post-Studie ohne Kontrollgruppe), kann man abschließend formulieren, dass die PR zur Verbesserung der Stimmungslage und Erlangung einer psychischen Stabilität beiträgt, unabhängig von der Ausprägung des Extraversions- und Neurotizismusgrades.
Abstract
Glioblastomas, primary brain tumors, represent a tumor entity with a dismal prognosis and a median survival of only about one year. Invasion into the healthy brain parenchyma contributes substantially to the malignancy of this type of brain tumor. Therefore, a better understanding of the mechanisms promoting the invasive behavior of these brain tumors is needed to identify new therapeutic targets.
Cofilin, an actin regulatory protein, has been shown to be an important regulator of the invasive behavior of tumor cells in other types of cancer and the actin cytoskeleton is involved in the formation of a variety of cellular structures important for cell migration and invasion. Cofilin is regulated by phosphorylation on a single residue, serine 3. The aim of this thesis was to examine the role of the cofilin regulatory phosphatase chronophin for glioma cell migration and invasion.
First, it was established that chronophin depletion in the cell line GBM6840 leads to an increase in the ratio of phosphorylated cofilin to total cofilin. Higher chronophin levels were correlated with a decrease in F-actin in the cell lines GBM6840 and U87 as measured in an actin spin down assay and in a flow cytometry based assay.
Furthermore, it was shown that knockdown of chronophin in two different cell lines, GBM6840 and DBTRG-05-MG, strongly increased their invasiveness in vitro. Expression of human chronophin in the cell line U87 decreased its invasiveness substantially. There was no difference in cell proliferation between GBM6840 and DBTRG-05-MG cells expressing a chronophin targeting shRNA or a control shRNA and U87 cells transfected with an empty vector or a human chronophin encoding plasmid. The increase in invasiveness after chronophin depletion could be correlated with an increase in directionality in cell migration under 2D culture conditions in the cell lines U87 and GBM6840. Moreover, treatment with the ROCK inhibitor Y-27632 decreased directionality in GBM6840 cells under 2D culture conditions and reduced the invasiveness of GBM6840 chronophin shRNA cells back to control levels.
Expression of a non-phosphorylatable cofilin mutant, the S3A mutant, was able to reduce invasiveness and to reduce directionality under 2D culture conditions back to control levels in GBM6840 chronophin shRNA cells.
This provides important evidence for the involvement of cofilin phosphoregulation in the phenotypes described above.
In vivo, when injected into NOD-SCID mice, chronophin depleted cells showed a dramatic growth reduction as compared to control and rescue cells.
Transciptomic characterization of GBM6840 cells by microarray analysis and subsequent comparison of the data with microarray profiles of normal brain tissues and different glioma entities identified two specifically chronophin regulated transcripts potentially involved in tumor progression and invasion, MXI1 and EDIL3. Moreover, c-myc was identified as a significantly altered transcription factor after chronophin deregulation based on the number of c-myc target molecules in the microarray dataset.
MXI1 is a potential negative regulator of c-myc dependent transcription, and was strongly downregulated after chronophin knockdown in GBM6840. In line with this, the activity of a c-myc reporter plasmid was increased after chronophin depletion in GBM6840 and reduced after chronophin expression in U87 cells.
However, the protein level of the c-myc protein was reduced after chronophin depletion in GBM6840.
Finally, anaylsis of the expression of proteases known to be important for glioblastoma pathogenesis revealed no major changes in protease expression between chronophin depleted and control cells.
Therefore, a comprehensive analysis of chronophin in the context of glioma pathogenesis has been performed in this thesis. It has been shown that chronophin depletion strongly enhanced invasiveness of glioma cells and that it induced transcriptomic changes potentially involved in tumor progression. The proteins regulating cofilin phosphorylation are therefore valuable therapeutic targets for anti-invasive therapy in glioblastomas. Inhibitors for kinases upstream of cofilin, e.g. LIMKs and ROCKs, are available, and might be promising agents for anti-invasive therapy.
Zentrales Ziel dieser Arbeit war es, Methoden der Mikroskopie, Bildverarbeitung und Bilderkennung für die Charakterisierungen verschiedener Phyotplankter zu nutzen, um deren Analyse zu verbessern und zu vereinfachen.
Der erste Schwerpunkt der Arbeit lag auf der Analyse von Phytoplanktongemeinschaften, die im Rahmen der Überprüfung der Süßwasserqualität als Marker dienen. Die konventionelle Analyse ist dabei sehr aufwendig, da diese noch immer vollständig von Hand durchgeführt wird und hierfür speziell ausgebildetes Personal eingesetzt werden muss. Ziel war es, ein System zur automatischen Erkennung aufzubauen, um die Analyse vereinfachen zu können. Mit Hilfe von automatischer Mikroskopie war es möglich Plankter unterschiedlicher Ausdehnung durch die Integration mehrerer Schärfeebenen besser in einem Bild aufzunehmen. Weiterhin wurden verschiedene Fluoreszenzeigenschaften in die Analyse integriert. Mit einem für ImageJ erstellten Plugin können Organismen vom Hintergrund der Aufnahmen abgetrennt und eine Vielzahl von Merkmalen berechnet werden. Über das Training von neuralen Netzen wird die Unterscheidung von verschieden Gruppen von Planktontaxa möglich. Zudem können weitere Taxa einfach in die Analyse integriert und die Erkennung erweitert werden. Die erste Analyse von Mischproben, bestehend aus 10 verschiedenen Taxa, zeigte dabei eine durchschnittliche Erkennungsrate von 94.7% und eine durchschnittliche Falsch-Positiv Rate von 5.5%. Im Vergleich mit bestehenden Systemen konnte die Erkennungsrate verbessert und die Falsch Positiv Rate deutlich gesenkt werde. Bei einer Erweiterung des Datensatzes auf 22 Taxa wurde darauf geachtet, Arten zu verwenden, die verschiedene Stadien in ihrem Wachstum durchlaufen oder höhere Ähnlichkeiten zu den bereits vorhandenen Arten aufweisen, um evtl. Schwachstellen des Systemes erkennen zu können. Hier ergab sich eine gute Erkennungsrate (86.8%), bei der der Ausschluss von nicht-planktonischen Partikeln (11.9%) weiterhin verbessert war. Der Vergleich mit weiteren Klassifikationsverfahren zeigte, dass neuronale Netze anderen Verfahren bei dieser Problemstellung überlegen sind. Ähnlich gute Klassifikationsraten konnten durch Support Vektor Maschinen erzielt werden. Allerdings waren diese bei der Unterscheidung von unbekannten Partikeln dem neuralen Netz deutlich unterlegen.
Der zweite Abschnitt stellt die Entwicklung einer einfachen Methode zur Viabilitätsanalyse von Cyanobakterien, bei der keine weitere Behandlung der Proben notwendig ist, dar. Dabei wird die rote Chlorophyll - Autofluoreszenz als Marker für lebende Zellen und eine grüne unspezifische Fluoreszenz als Marker für tote Zellen genutzt. Der Assay wurde mit dem Modellorganismus Synechocystis sp. PCC 6803 etabliert und validiert. Die Auswahl eines geeigeneten Filtersets ermöglicht es beide Signale gleichzeitig anzuregen und zu beobachten und somit direkt zwischen lebendenden und toten Zellen zu unterscheiden. Die Ergebnisse zur Etablierung des Assays konnten durch Ausplattieren, Chlorophyllbestimmung und Bestimmung des Absorbtionsspektrums bestätigt werden. Durch den Einsatz von automatisierter Mikroskopie und einem neu erstellten ImageJ Plugin wurde eine sehr genaue und schnelle Analyse der Proben möglich. Der Einsatz beim Monitoring einer mutagenisierten Kultur zur Erhöhung der Temperaturtoleranz ermöglichte genaue und zeitnahe Einblicke in den Zustand der Kultur. Weitere Ergebnisse weisen darauf hin, dass die Kombination mit Absorptionsspektren es ermöglichen können bessere Einblicke in die Vitalität der Kultur zu erhalten.
Defined aggregates of polymers such as polymeric micelles are of great importance in the development of pharmaceutical formulations. The amount of drug that can be formulated by a drug delivery system is an important issue, and most drug delivery systems suffer from their relatively low drug-loading capacity. However, as the loading capacities increase, i.e., promoted by good drug–polymer interactions, the drug may affect the morphology and stability of the micellar system. We investigated this effect in a prominent system with very high capacity for hydrophobic drugs and found extraordinary stability as well as a profound morphology change upon incorporation of paclitaxel into micelles of amphiphilic ABA poly(2-oxazoline) triblock copolymers. The hydrophilic blocks A comprised poly(2-methyl-2-oxazoline), while the middle blocks B were either just barely hydrophobic poly(2-n-butyl-2-oxazoline) or highly hydrophobic poly(2-n-nonyl-2-oxazoline). The aggregation behavior of both polymers and their formulations with varying paclitaxel contents were investigated by means of dynamic light scattering, atomic force microscopy, (cryogenic) transmission electron microscopy, and small-angle neutron scattering. While without drug, wormlike micelles were present, after incorporation of small amounts of drugs only spherical morphologies remained. Furthermore, the much more hydrophobic poly(2-n-nonyl-2-oxazoline)-containing triblock copolymer exhibited only half the capacity for paclitaxel than the poly(2-n-butyl-2-oxazoline)-containing copolymer along with a lower stability. In the latter, contents of paclitaxel of 8 wt % or higher resulted in a raspberry-like micellar core.
Background
Information about genes, transcripts and proteins is spread over a wide variety of databases. Different tools have been developed using these databases to identify biological signals in gene lists from large scale analysis. Mostly, they search for enrichments of specific features. But, these tools do not allow an explorative walk through different views and to change the gene lists according to newly upcoming stories.
Results
To fill this niche, we have developed ISAAC, the InterSpecies Analysing Application using Containers. The central idea of this web based tool is to enable the analysis of sets of genes, transcripts and proteins under different biological viewpoints and to interactively modify these sets at any point of the analysis. Detailed history and snapshot information allows tracing each action. Furthermore, one can easily switch back to previous states and perform new analyses. Currently, sets can be viewed in the context of genomes, protein functions, protein interactions, pathways, regulation, diseases and drugs. Additionally, users can switch between species with an automatic, orthology based translation of existing gene sets. As todays research usually is performed in larger teams and consortia, ISAAC provides group based functionalities. Here, sets as well as results of analyses can be exchanged between members of groups.
Conclusions
ISAAC fills the gap between primary databases and tools for the analysis of large gene lists. With its highly modular, JavaEE based design, the implementation of new modules is straight forward. Furthermore, ISAAC comes with an extensive web-based administration interface including tools for the integration of third party data. Thus, a local installation is easily feasible. In summary, ISAAC is tailor made for highly explorative interactive analyses of gene, transcript and protein sets in a collaborative environment.
Background
To examine whether lack of measurement invariance (MI) influences mean comparisons among different disease groups, this paper provides (1) a systematic review of MI in generic constructs across chronic conditions and (2) an empirical analysis of MI in the Health Education Impact Questionnaire (heiQ™).
Methods
(1) We searched for studies of MI among different chronic conditions in online databases. (2) Multigroup confirmatory factor analyses were used to study MI among five chronic conditions (orthopedic condition, rheumatism, asthma, COPD, cancer) in the heiQ™ with N = 1404 rehabilitation inpatients. Impact on latent and composite mean differences was examined.
Results
(1) A total of 30 relevant studies suggested that about one in three items lacked MI. However, only four studies examined impact on latent mean differences. Scale means were only affected in one of these three studies. (2) Across the eight heiQ™ scales, seven scales had items with lack of MI in at least one disease group. However, in only two heiQ™ scales were some latent or composite mean differences affected.
Conclusions
Lack of MI among disease groups is common and may have a relevant influence on mean comparisons when using generic instruments. Therefore, when comparing disease groups, tests of MI should be implemented. More studies of MI and according impact on mean differences in generic questionnaires are needed.
The interaction with brain endothelial cells is central to the pathogenicity of Neisseria meningitidis infections. Here, we show that N. meningitidis causes transient activation of acid sphingomyelinase (ASM) followed by ceramide release in brain endothelial cells. In response to N. meningitidis infection, ASM and ceramide are displayed at the outer leaflet of the cell membrane and condense into large membrane platforms which also concentrate the ErbB2 receptor. The outer membrane protein Opc and phosphatidylcholine-specific phospholipase C that is activated upon binding of the pathogen to heparan sulfate proteoglycans, are required for N. meningitidis-mediated ASM activation. Pharmacologic or genetic ablation of ASM abrogated meningococcal internalization without affecting bacterial adherence. In accordance, the restricted invasiveness of a defined set of pathogenic isolates of the ST-11/ST-8 clonal complex into brain endothelial cells directly correlated with their restricted ability to induce ASM and ceramide release. In conclusion, ASM activation and ceramide release are essential for internalization of Opc-expressing meningococci into brain endothelial cells, and this segregates with invasiveness of N. meningitidis strains.
Author Summary
Neisseria meningitidis, an obligate human pathogen, is a causative agent of septicemia and meningitis worldwide. Meningococcal infection manifests in a variety of forms, including meningitis, meningococcemia with meningitis or meningococcemia without obvious meningitis. The interaction of N. meningitidis with human cells lining the blood vessels of the blood-cerebrospinal fluid barrier is a prerequisite for the development of meningitis. As a major pathogenicity factor, the meningococcal outer membrane protein Opc enhances bacterial entry into brain endothelial cells, however, mechanisms underlying trapping of receptors and signaling molecules following this interaction remained elusive. We now show that Opc-expressing meningococci activate acid sphingomyelinase (ASM) in brain endothelial cells, which hydrolyses sphingomyelin to cause ceramide release and formation of extended ceramide-enriched membrane platforms wherein ErbB2, an important receptor involved in bacterial uptake, clusters. Mechanistically, ASM activation relied on binding of N. meningitidis to its attachment receptor, HSPG, followed by activation of PC-PLC. Meningococcal isolates of the ST-11 clonal complex, which are reported to be more likely to cause severe sepsis, but rarely meningitis, barely invaded brain endothelial cells and revealed a highly restricted ability to induce ASM and ceramide release. Thus, our results unravel a differential activation of the ASM/ceramide system by the species N. meningitidis determining its invasiveness into brain endothelial cells.
Targeting Echinococcus multilocularis Stem Cells by Inhibition of the Polo-Like Kinase EmPlk1

Background
Alveolar echinococcosis (AE) is a life-threatening disease caused by larvae of the fox-tapeworm Echinococcus multilocularis. Crucial to AE pathology is continuous infiltrative growth of the parasite's metacestode stage, which is driven by a population of somatic stem cells, called germinative cells. Current anti-AE chemotherapy using benzimidazoles is ineffective in eliminating the germinative cell population, thus leading to remission of parasite growth upon therapy discontinuation.
Methodology/Principal findings
We herein describe the characterization of EmPlk1, encoded by the gene emplk1, which displays significant homologies to members of the Plk1 sub-family of Polo-like kinases that regulate mitosis in eukaryotic cells. We demonstrate germinative cell-specific expression of emplk1 by RT-PCR, transcriptomics, and in situ hybridization. We also show that EmPlk1 can induce germinal vesicle breakdown when heterologously expressed in Xenopus oocytes, indicating that it is an active kinase. This activity was significantly suppressed in presence of BI 2536, a Plk1 inhibitor that has been tested in clinical trials against cancer. Addition of BI 2536 at concentrations as low as 20 nM significantly blocked the formation of metacestode vesicles from cultivated Echinococcus germinative cells. Furthermore, low concentrations of BI 2536 eliminated the germinative cell population from mature metacestode vesicles in vitro, yielding parasite tissue that was no longer capable of proliferation.
Conclusions/Significance
We conclude that BI 2536 effectively inactivates E. multilocularis germinative cells in parasite larvae in vitro by direct inhibition of EmPlk1, thus inducing mitotic arrest and germinative cell killing. Since germinative cells are decisive for parasite proliferation and metastasis formation within the host, BI 2536 and related compounds are very promising compounds to complement benzimidazoles in AE chemotherapy.
Author Summary
The lethal disease AE is characterized by continuous and infiltrative growth of the metacestode larva of the tapeworm E. multilocularis within host organs. This cancer-like progression is exclusively driven by a population of parasite stem cells (germinative cells) that have to be eliminated for an effective cure of the disease. Current treatment options, using benzimidazoles, are parasitostatic only, and thus obviously not effective in germinative cell killing. We herein describe a novel, druggable parasite enzyme, EmPlk1, that specifically regulates germinative cell proliferation. We show that a compound, BI 2536, originally designed to inhibit the human ortholog of EmPlk1, can also inhibit the parasite protein at low doses. Furthermore, low doses of BI 2536 eliminated germinative cells from Echinococcus larvae in vitro and prevented parasite growth and development. We propose that BI 2536 and related compounds are promising drugs to complement current benzimidazole treatment for achieving parasite killing.
Altering gene expression by aminocoumarins: the role of DNA supercoiling in Staphylococcus aureus

BACKGROUND:
It has been shown previously that aminocoumarin antibiotics such as novobiocin lead to immediate downregulation of recA expression and thereby inhibit the SOS response, mutation frequency and recombination capacity in Staphylococcus aureus. Aminocoumarins function by inhibiting the ATPase activity of DNA gyrase subunit B with a severe impact on DNA supercoiling.
RESULTS:
Here, we have analysed the global impact of the DNA relaxing agent novobiocin on gene expression in S. aureus. Using a novobiocin-resistant mutant, it became evident that the change in recA expression is due to gyrase inhibition. Microarray analysis and northern blot hybridisation revealed that the expression levels of a distinct set of genes were increased (e.g., recF-gyrB-gyrA, the rib operon and the ure operon) or decreased (e.g., arlRS, recA, lukA, hlgC and fnbA) by novobiocin. The two-component ArlRS system was previously found to decrease the level of supercoiling in S. aureus. Thus, downregulation of arlRS might partially compensate for the relaxing effect of novobiocin. Global analysis and gene mapping of supercoiling-sensitive genes did not provide any indication that they are clustered in the genome. Promoter fusion assays confirmed that the responsiveness of a given gene is intrinsic to the promoter region but independent of the chromosomal location.
CONCLUSIONS:
The results indicate that the molecular properties of a given promoter, rather than the chromosomal topology, dictate the responsiveness to changes in supercoiling in the pathogen Staphylococcus aureus.
Atherosklerose ist ein wichtiger Faktor bezüglich der Pathogenese kardiovaskulärer Erkrankungen, indem sie Einfluss auf die strukturellen und funktionellen Eigenschaften der Gefäße nimmt. Wegen dieser Bedeutung ist es wichtig, diese Erkrankung und deren Zusammenhänge genau zu erforschen.
Einige Studien konnten zeigen, dass die Pulswellengeschwindigkeit mit Gefäßwand-eigenschaften, insbesondere deren Steifigkeit, zusammenhängt. So konnte in Untersuchungen von Gotschy et al.27 eine Zunahme der lokalen Pulswellen-geschwindigkeit, als Marker für Gefäßwandsteifigkeit, in Aortenanteilen von ApoE(-/-)-Mäusen im Zeitverlauf mittels MRT gezeigt werden, ohne dass makroskopische Veränderungen in frühen Stufen atherosklerotischer Entwicklung sichtbar waren. Dies wird nun als Ausgangspunkt herangezogen, um zu erforschen, durch welche histologischen Veränderungen des Gefäßes innerhalb der Atherogenese eine frühe Zunahme der lokalen PWV erklärt werden kann.
Hierzu wird zum einen die mit der Gefäßwandsteifigkeit in Zusammenhang gebrachte Elastinfragmentierung innerhalb der elastischen Laminae der Aorta ascendens und abdominalis mittels histologischer Untersuchung quantitativ bestimmt. Die elastischen Schichten werden durch modifizierte Elastika-van-Gieson-Färbung dargestellt. Zur vergleichenden Bestimmung wird ein Quotient aus der Anzahl bei mikroskopischer Betrachtung erkennbarer Brüche der elastischen Laminae und bestimmter Gefäßwandquerschnittsfläche erstellt. Die Ausprägung der Elastinfragmentierung unterscheidet sich signifikant in den zwei höchsten Altersklassen der Test- und Kontrollmäuse in beiden untersuchten Aortenabschnitten, wobei die ApoE(-/-)-Tiere stets den höheren Wert aufweisen. Anhand der Ähnlichkeit hinsichtlich der Entwicklung der Messwerte von beschriebener lokalen PWV und der hier gezeigten Elastinfragmentierung kann vermutet werden, dass diese zu einer Erhöhung der Gefäßwandsteifigkeit beiträgt. Die gezeigten histologischen Parameter können so als Indikator für Veränderungen der Gefäßeigenschaften während der Atherogenese und als möglicher Grund einer Veränderung der Gefäßwandsteifigkeit mit konsekutiver Erhöhung der lokalen PWV angesehen werden.
Als weiterer Einflussfaktor auf die mechanischen Eigenschaften des Blutgefäßsystems wird die Entwicklung des Kollagengehaltes anteilig an der Aortenwand bestimmt. Es wurde in vielen Publikationen beschrieben, welchen großen Stellenwert Kollagenfasern für die Gefäßwandsteifigkeit haben. Auch wird das untersuchte Kollagen hinsichtlich seiner Faserdicken nochmals genauer untersucht. Die Kollagenfasern werden mittels Pikro-Sirius-Rot gefärbt und anschließend unter zirkulär polarisiertem Licht analysiert. Die erhaltenen Ergebnisse zeigen keinen eindeutigen Trend hinsichtlich ihrer Entwicklung. Sie lassen sich auch nicht mit der aufgestellten Hypothese in Übereinkunft bringen, da keine zeitliche Dynamik verglichen mit der Entwicklung der anderen Parameter nachgewiesen werden kann.
Um den beschriebenen entzündlichen Charakter der Atherogenese ebenso zu berücksichtigen, wird das Einwanderungsverhalten von Makrophagen in die Gefäßwand betrachtet. Es wird die relative Fläche der durch Makrophagen besiedelten Gefäßwandanteile bestimmt und als Indikator einer Entzündungsreaktion angesehen, welche direkt oder indirekt Einfluss auf Gefäßwandeigenschaften haben könnte. Mittels eines Antikörpers gegen das makrophagenspezifische Protein CD68 und Kombination mit anti-antikörpervermittelt bindenden fluoreszierenden Substanzen, werden Makrophagen in einem Fluoreszenzmikroskop sichtbar gemacht und die besiedelte Fläche softwaregestützt bestimmt. Dabei kommt die vorliegende Untersuchung zu dem Ergebnis, dass sich die Makrophagenbesiedelung in beiden betrachteten Aortenabschnitten deutlich im Zeitverlauf bei den ApoE(-/-)-Tieren erhöht. Der relative Gefäßwandanteil der von Makrophagen besiedelten Flächen erreicht seinen höchsten Wert innerhalb der Aorta ascendens bei 18 Wochen alten ApoE(-/-)-Tieren; in der Aorta abdominalis bei 30 Wochen alten. Die absolute CD68-Fläche in beiden Aortenabschnitten steigt bis zum Versuchsende hin an, wobei der stärkste Anstieg in der Zeit von sechs zu 18 Wochen geschieht. Dagegen zeigt die Kontrollgruppe eine sehr geringe Zunahme dieser beiden Messgrößen, wobei diese stets auf einem sehr niedrigen Niveau bleiben und keineswegs mit der Entwicklung der Testtiere zu vergleichen sind. Besonders zu beachten ist, das bereits in der jüngsten Altersstufe von sechs Wochen ein höheres Maß an Makrophagenbesiedelung bei Aortenanteilen der ApoE(-/-)-Tiere zu verzeichnen ist, als bei den entsprechenden Kontrollen. Auf Grund dieser Beobachtung, lässt sich ein möglicher Zusammenhang mit der schon früh ansteigenden Gefäßsteifigkeit vor sichtbarer Plaqueentwicklung herstellen.
Bei der Klärung der zentralen Fragestellung dieser Arbeit nach einem histomorphologischen Korrelat für die frühzeitige Erhöhung der lokalen PWV zeigt sich folgendes Ergebnis: Insgesamt können die gemessenen histologischen Charakteristika, vor allem die Entwicklung der Elastinfragmentierung innerhalb der Gefäßwand und der Grad der Makrophagenbesiedelung, frühe Indikatoren für Veränderungen morphologischer und funktioneller Gefäßwandspezifikationen sein. Sie kommen daher als denkbare Ursache für den Anstieg der lokalen Pulswellengeschwindigkeit in Betracht, besonders da die zuvor erwähnten histologischen Parameter zeitlich wie diese einen ähnlichen Verlauf hinsichtlich ihrer Entwicklung zeigen.
Die Atherogenese und deren zahlreiche Einflussfaktoren bleiben weiterhin Forschungs-gegenstand und es gilt mehr denn je mögliche Mechanismen aufzudecken, um eine Gefäßwandschädigung frühzeitig und effektiv verhindern zu können. Hiervon würde ein Großteil der Bevölkerung profitieren, da die Atherosklerose durch ihre Folgeerkrankungen nach wie vor, nicht nur in Deutschland, die Todesursachenstatistik anführt.
Introduction: Since 1996, the preferred approach for positioning the active middle-ear implant Vibrant Soundbridge© is a mastoidectomy and a posterior tympanotomy. With this device, placement of the floating mass transducer (FMT) on the long incus process is the standard method for treatment of mild-to-severe sensorineural hearing loss in the case of normal middle-ear anatomy. The aim of this study was to determine the vibrational effectiveness of FMT placement at the short incus process. Materials and Methods: An extended antrotomy and a posterior tympanotomy were performed in 5 fresh human temporal bones. As a control for normal middle-ear function, the tympanic membrane was stimulated acoustically and the vibration of the stapes footplate and the round-window (RW) membrane were (sequentially) measured by laser Doppler vibrometry. Vibration responses for coupling of an FMT to the long incus process (standard coupling) were compared to those for coupling to the short incus process. Results: Apart from narrow frequency bands near 3 and 9 kHz for the stapes footplate and RW membrane, respectively, the velocity responses presented no significant differences between standard coupling of the FMT and coupling to the short incus process. Conclusion: Coupling the FMT to the short incus process may be a viable alternative in cases where the surgical approach is limited to an extended antrotomy. A reliable technique for attachment to the short incus process has yet to be developed.
Prenatal stress-induced programming of genome-wide promoter DNA methylation in 5-HTT-deficient mice

The serotonin transporter gene (5-HTT/SLC6A4)-linked polymorphic region has been suggested to have a modulatory role in mediating effects of early-life stress exposure on psychopathology rendering carriers of the low-expression short (s)-variant more vulnerable to environmental adversity in later life. The underlying molecular mechanisms of this gene-by-environment interaction are not well understood, but epigenetic regulation including differential DNA methylation has been postulated to have a critical role. Recently, we used a maternal restraint stress paradigm of prenatal stress (PS) in 5-HTT-deficient mice and showed that the effects on behavior and gene expression were particularly marked in the hippocampus of female 5-Htt+/- offspring. Here, we examined to which extent these effects are mediated by differential methylation of DNA. For this purpose, we performed a genome-wide hippocampal DNA methylation screening using methylated-DNA immunoprecipitation (MeDIP) on Affymetrix GeneChip Mouse Promoter 1.0 R arrays. Using hippocampal DNA from the same mice as assessed before enabled us to correlate gene-specific DNA methylation, mRNA expression and behavior. We found that 5-Htt genotype, PS and their interaction differentially affected the DNA methylation signature of numerous genes, a subset of which showed overlap with the expression profiles of the corresponding transcripts. For example, a differentially methylated region in the gene encoding myelin basic protein (Mbp) was associated with its expression in a 5-Htt-, PS- and 5-Htt × PS-dependent manner. Subsequent fine-mapping of this Mbp locus linked the methylation status of two specific CpG sites to Mbp expression and anxiety-related behavior. In conclusion, hippocampal DNA methylation patterns and expression profiles of female prenatally stressed 5-Htt+/- mice suggest that distinct molecular mechanisms, some of which are promoter methylation-dependent, contribute to the behavioral effects of the 5-Htt genotype, PS exposure and their interaction.
Interferon alpha (IFNα) is approved for adjuvant treatment of stage III melanoma in Europe and the US. Its clinical efficacy, however, is restricted to a subpopulation of patients while side effects occur in most of treated patients. Thus, the identification of predictive biomarkers would be highly beneficial to improve the benefit to risk ratio. In this regard, STAT3 is important for signaling of the IFNα receptor. Moreover, the STAT3 single-nucleotide polymorphism (SNP) rs4796793 has recently been reported to be associated with IFNα sensitivity in metastatic renal cell carcinoma. To translate this notion to melanoma, we scrutinized the impact of rs4796793 functionally and clinically in this cancer. Interestingly, melanoma cells carrying the minor allele of rs4796793 were the most sensitive to IFNα in vitro. However, we did not detect a correlation between SNP genotype and STAT3 mRNA expression for either melanoma cells or for peripheral blood lymphocytes. Next, we analyzed the impact of rs4796793 on the clinical outcome of 259 stage III melanoma patients of which one-third had received adjuvant IFNα treatment. These analyses did not reveal a significant association between the STAT3 rs4796793 SNP and patients' progression free or overall survival when IFNα treated and untreated patients were compared. In conclusion, STAT3 rs4796793 SNP is no predictive marker for the efficacy of adjuvant IFNα treatment in melanoma patients.
It has been argued that the emergence of roughly periodic orientation preference maps (OPMs) in the primary visual cortex (V1) of carnivores and primates can be explained by a so-called statistical connectivity model. This model assumes that input to V1 neurons is dominated by feed-forward projections originating from a small set of retinal ganglion cells (RGCs). The typical spacing between adjacent cortical orientation columns preferring the same orientation then arises via Moire 'Interference between hexagonal ON/OFF RGC mosaics. While this Moire-Interference critically depends on long-range hexagonal order within the RGC mosaics, a recent statistical analysis of RGC receptive field positions found no evidence for such long-range positional order. Hexagonal order may be only one of several ways to obtain spatially repetitive OPMs in the statistical connectivity model. Here, we investigate a more general requirement on the spatial structure of RGC mosaics that can seed the emergence of spatially repetitive cortical OPMs, namely that angular correlations between so-called RGC dipoles exhibit a spatial structure similar to that of OPM autocorrelation functions. Both in cat beta cell mosaics as well as primate parasol receptive field mosaics we find that RGC dipole angles are spatially uncorrelated. To help assess the level of these correlations, we introduce a novel point process that generates mosaics with realistic nearest neighbor statistics and a tunable degree of spatial correlations of dipole angles. Using this process, we show that given the size of available data sets, the presence of even weak angular correlations in the data is very unlikely. We conclude that the layout of ON/OFF ganglion cell mosaics lacks the spatial structure necessary to seed iso-orientation domains in the primary visual cortex.
A longstanding question in infection biology addresses the genetic basis for invasive behavior in commensal pathogens. A prime example for such a pathogen is Neisseria meningitidis. On the one hand it is a harmless commensal bacterium exquisitely adapted to humans, and on the other hand it sometimes behaves like a ferocious pathogen causing potentially lethal disease such as sepsis and acute bacterial meningitis. Despite the lack of a classical repertoire of virulence genes in N. meningitidis separating commensal from invasive strains, molecular epidemiology suggests that carriage and invasive strains belong to genetically distinct populations. In recent years, it has become increasingly clear that metabolic adaptation enables meningococci to exploit host resources, supporting the concept of nutritional virulence as a crucial determinant of invasive capability. Here, we discuss the contribution of core metabolic pathways in the context of colonization and invasion with special emphasis on results from genome-wide surveys. The metabolism of lactate, the oxidative stress response, and, in particular, glutathione metabolism as well as the denitrification pathway provide examples of how meningococcal metabolism is intimately linked to pathogenesis. We further discuss evidence from genome-wide approaches regarding potential metabolic differences between strains from hyperinvasive and carriage lineages and present new data assessing in vitro growth differences of strains from these two populations. We hypothesize that strains from carriage and hyperinvasive lineages differ in the expression of regulatory genes involved particularly in stress responses and amino acid metabolism under infection conditions.
T cell paralysis is a main feature of measles virus (MV) induced immunosuppression. MV contact mediated activation of sphingomyelinases was found to contribute to MV interference with T cell actin reorganization. The role of these enzymes in MV-induced inhibition of T cell activation remained equally undefined as their general role in regulating immune synapse (IS) activity which relies on spatiotemporal membrane patterning. Our study for the first time reveals that transient activation of the neutral sphingomyelinase 2 (NSM2) occurs in physiological co-stimulation of primary T cells where ceramide accumulation is confined to the lamellum (where also NSM2 can be detected) and excluded from IS areas of high actin turnover. Genetic ablation of the enzyme is associated with T cell hyper-responsiveness as revealed by actin dynamics, tyrosine phosphorylation, Ca2+-mobilization and expansion indicating that NSM2 acts to suppress overshooting T cell responses. In line with its suppressive activity, exaggerated, prolonged NSM2 activation as occurring in co-stimulated T cells following MV exposure was associated with aberrant compartmentalization of ceramides, loss of spreading responses, interference with accumulation of tyrosine phosphorylated protein species and expansion. Altogether, this study for the first time reveals a role of NSM2 in physiological T cell stimulation which is dampening and can be abused by a virus, which promotes enhanced and prolonged NSM2 activation to cause pathological T cell suppression.

Nach einem Myokardinfarkt setzen Wundheilungsprozesse ein, um die Durchblutung wieder herzustellen und nekrotisches Muskelgewebe durch Narbengewebe zu ersetzen. Die Einsprossung neuer Kapillaren vom bestehenden Gefäßnetz aus wird als Angiogenese bezeichnet. Das dabei vermehrt exprimierte proteolytische Enzym Aminopeptidase N (APN) spielt eine entscheidende Rolle bei der Einsprossung von Endothelzellen. Beim kardialen Remodeling werden abgestorbene Myozyten mithilfe der Einwanderung von Fibroblasten durch Binde- oder Stützgewebe ersetzt, dabei übernimmt das Fibroblasten-Aktivierungs-Protein alpha (FAP) Aufgaben bei der Proliferation und Fortbewegung von Fibroblasten.
Durch ihre erhöhte Expression bei den Wundheilungs- und Remodelingprozessen nach einem Herzinfarkt stellen die Metalloprotease APN und die Serinprotease FAP molekulare Targets für die Diagnostik und Therapie dar. Als Diagnosemethode besonders geeignet ist die Positronen-Emissions-Tomographie (PET), die es ermöglicht, biochemische Prozesse in Echtzeit im zu untersuchenden Organismus zu visualisieren und zu quantifizieren. Eine als Radiopharmakon oder Tracer bezeichnete biochemische Sonde kann im Falle eines Enzyms dessen radioaktiv markiertes Substrat oder ein Inhibitor sein.
Ziel dieser Arbeit war es, spezifische APN- und FAP-affine Tracer für die nicht-invasive Untersuchung der APN- und FAP-Expression mittels PET zu entwickeln und dadurch die Rolle von APN und FAP bei Remodelingprozessen nach Myokardinfarkt besser verstehen bzw. klären zu können.
Um die Protease APN mittels PET zu untersuchen, wurden die für APN affine Verbindung NOTA-NGR (Komplexbildner + cyclisches Peptid inkl. Asparagin-Glycin-Arginin) mit dem Positronen-emittierenden Nuklid Gallium-68 (68Ga) markiert. Das Potential von 68Ga-NOTA-NGR als PET-Tracer wurde in vivo am Infarktmodell mittels Kleintier-PET untersucht und mit 68Ga-NOTA-RGD, einem zur Visualisierung des neo-angiogenetischen alphavbeta3-Integrins etablierten Tracer, verglichen. Untersuchungen ergaben, dass 68Ga-NOTA-NGR einen vielversprechenden neuen PET-Tracer für die Visualisierung und Quantifizierung der APN-Expression im Rahmen der Angiogenese nach einem Myokardinfarkt darstellt. 68Ga-NOTA-NGR zeigte eine erhöhte Aufnahme im Bereich des Myokardinfarkts im Sinne einer vermehrten Angiogenese. Die Aufnahme des Tracers in infarzierten Arealen war quantitativ höher als in der Untersuchung mit 68Ga-NOTA-RGD. In Autoradiographie-Experimenten wurde 68Ga-NOTA-NGR ex vivo untersucht. Die Akkumulation von 68Ga-NOTA-NGR im ischämischen Bereich war deutlich höher als im gesunden Myokard. Der Nachweis der unterschiedlichen Bereiche des Herzens erfolgte mit HE-Färbung.
Die Expression von APN wurde immunohistochemisch mittels spezifischer Antikörper bestätigt. Zum Vergleich wurden ebenso einige andere an der Angiogenese beteiligte Faktoren untersucht. APN stellte sich auch hier als geeignetes Target zum Nachweis der Angiogenese heraus.
Um die Protease FAP mittels PET zu untersuchen, wurden eine Reihe peptidomimetischer Inhibitoren, die die Erkennungssequenz Glycin-Prolin mit einer Carbonitril-Gruppe als elektrophiler Einheit zur kovalent-reversiblen Hemmung des Enzyms enthalten, entwickelt. Ausgehend vom N-Acetylglycin-pyrrolidin-(2S)-carbonitril als Leitstruktur wurden Inhibitoren und Vorstufen zur Radiomarkierung inkl. verschieden substituierter Benzoesäuren dargestellt. Zusätzlich wurden noch bereits bekannte Inhibitoren synthetisiert, die zum Vergleich in den Enzymassays dienten.
Drei Verbindungen zeigten gute inhibitorische Wirkung an FAP und außerdem Selektivität gegenüber DPP IV. Keine der entwickelten Verbindungen zeigte einen KI-Wert im nanomolaren Bereich, erforderlich für einen potentiellen Tracer zur in-vivo-Visualisierung einer Enzymexpression mittels PET.
Um die Inhibitoren mit der besten Hemmung an FAP zum PET-Tracer weiterzuentwickeln, mussten sie mit einem Positronenemitter markiert werden. Die Markierung erfolgte über Isotopenaustausch, bei dem nicht-radioaktives Iod am aromatischen Ring des Precursors durch das radioaktive Iod-124 (124I) substituiert wurde. Es konnten dadurch die radioiodierten Verbindungen 1-(2-[124I]Iodhippursäure)-pyrrolidin-(2S)-carbonitril und 1-(4-[124I]Iod-hippursäure)-pyrrolidin-(2S)-carbonitril synthetisiert werden.
Trotz der relativ niedrigen Affinität für FAP wurde das neue 1-(2-[124I]Iodhippursäure)-pyrrolidin-(2S)-carbonitril in Ratten am Infarktmodell mittels Kleintier-PET getestet. Die Lage der ischämischen Zone wurde im Anschluss durch HE-Färbung bestimmt. In vivo zeigte sich eine nur sehr geringe Aufnahme des Radiopharmakons in der ischämischen Zone des Myokards. Damit ist 1-(2-[124I]Iod-hippursäure)-pyrrolidin-(2S)-carbonitril kein für den gewünschten Zweck geeigneter PET-Tracer. Nichtsdestotrotz war der Ansatz vielversprechend und es wurde zum ersten Mal ein PET-Tracer dieser Art zur Untersuchung des FAP im Myokardinfarkt hergestellt.
Background: Dose requirements of erythropoietin-stimulating agents (ESAs) can vary considerably over time and may be associated with cardiovascular outcomes. We aimed to longitudinally assess ESA responsiveness over time and to investigate its association with specific clinical end points in a time-dependent approach. Methods: The German Diabetes and Dialysis study (4D study) included 1,255 diabetic dialysis patients, of whom 1,161 were receiving ESA treatment. In those patients, the erythropoietin resistance index (ERI) was assessed every 6 months during a median follow-up of 4 years. The association between the ERI and cardiovascular end points was analyzed by time-dependent Cox regression analyses with repeated ERI measures. Results: Patients had a mean age of 66 ± 8.2 years; 53% were male. During follow-up, a total of 495 patients died, of whom 136 died of sudden death and 102 of infectious death. The adjusted and time-dependent risk for sudden death was increased by 19% per 5-unit increase in the ERI (hazard ratio, HR = 1.19, 95% confidence interval, CI = 1.07-1.33). Similarly, mortality increased by 25% (HR = 1.25, 95% CI = 1.18-1.32) and infectious death increased by 27% (HR = 1.27, 95% CI = 1.13-1.42). Further analysis revealed that lower 25-hydroxyvitamin D levels were associated with lower ESA responsiveness (p = 0.046). Conclusions: In diabetic dialysis patients, we observed that time-varying erythropoietin resistance is associated with sudden death, infectious complications and all-cause mortality. Low 25-hydroxyvitamin D levels may contribute to a lower ESA responsiveness.
Assistierte Reproduktionstechniken (ARTs) zur Behandlung von Infertilität werden mit einer erhöhten Häufigkeit von epigenetischen Aberrationen während der Gametogenese und der frühen Embryonalentwicklung in Verbindung gebracht, speziell durch eine Beeinträchtigung von geprägten Genen. Die in vitro-Maturation (IVM) von Eizellen ist eine ART, die bereits routinemäßig zur Reproduktion von ökonomisch wertvollen Zuchttieren wie dem Hausrind (Bos taurus) eingesetzt wird. IVM-Oozyten weisen jedoch eine verringerte Entwicklungs-kompetenz zum Blastozystenstadium dar, welche möglicherweise auf eine beeinträchtigte epigenetische Regulation zurückzuführen ist.
Von allen bekannten epigenetischen Mechanismen ist die DNA-Methylierung die meist untersuchte DNA-Modifikation. In dieser Arbeit wurden zur Klärung der Frage nach den Auswirkungen der IVM auf die DNA-Methylierung geprägter als auch nicht geprägter Gene Oozyten des Hausrinds analysiert. Diese Tierart weist eine ähnliche Präimplantations-entwicklung und Tragezeit wie der Mensch auf und wird daher zunehmend als Modell zum Studium der humanen Keimzell- und Embryonalentwicklung herangezogen. Im Gegensatz zu Mensch und Maus gibt es bislang nur wenig Information über bovine geprägte Gene. Das erste Ziel der hier dargestellten Forschungsarbeiten war daher die Identifizierung und Charakterisierung der bovinen differenziell methylierten Regionen (DMRs) der drei geprägten Genorte von IGF2/H19, SNRPN und PEG3, welche mit Imprintingdefekten des Menschen und/oder im Mausmodell assoziiert werden. Die hier erstmalig erfolgte Beschreibung von mehreren intergenischen DMRs mittels Bisulfitsequenzierung und Pyrosequenzierung belegt die Existenz und evolutionäre Konservierung der IGF2/H19-Imprintingkontrollregion (ICR) beim Rind. Der geprägte Zustand der IGF2/H19-ICR sowie der bovinen Gene SNRPN und PEG3 wurde durch den Nachweis differenzieller Methylierung in plazentalen und somatischen Geweben sowie in Spermien und parthenogenetischen Embryonen bestätigt. Die beobachteten Methylierungsprofile waren typisch für genomische Prägung.
Die direkte Bisulfitsequenzierung nach vorangegangener Limiting Dilution (LD) erlaubt die Analyse von Methylierungsmustern einzelner Allele (DNA-Moleküle) von einigen wenigen oder auch nur einer einzigen Zelle (El Hajj et al., 2011). In einem ersten LD-Versuch an bovinen Oozyten wurden die drei vorab charakterisierten und geprägten Gene hinsichtlich möglicher epigenetischer Veränderungen untersucht, welche durch verschiedene IVM-Bedingungen und -Medien (TCM und mSOF) hervorgerufen werden könnten. Die Gesamtrate von Methylierungsfehlern einzelner CpG-Stellen sowie die von ganzen Allelen (Imprintingfehlern) unterschied sich nicht wesentlich zwischen den beiden IVM-Gruppen und der in vivo-Gruppe. Dieses Ergebnis weist darauf hin, dass die gängigen IVM-Protokolle keinen oder nur einen geringfügigen Einfluss auf diese entscheidenden epigenetischen Markierungen haben.
IVM-Oozyten präpuberaler Kälber weisen eine herabgesetzte Entwicklungskompetenz im Vergleich zu IVM-Oozyten aus adulten Tieren auf. Aus diesem Grund wurde in einem zweiten LD-Versuchsansatz die Promotormethylierung von drei entwicklungsrelevanten, nicht geprägten Genen (SLC2A1, PRDX1, ZAR1) nach ovarieller Stimulation mit FSH und/oder IGF1 untersucht. Sowohl ungereifte als auch in vitro-gereifte Oozyten präpuberaler und adulter Kühe zeigten eine deutliche, unbeeinträchtige Hypomethylierung der drei Genpromotoren ohne jegliche Unterschiede zwischen den verschiedenen Alterstypen der Spendertiere oder deren Behandlung. Weder das Alter, die hormonelle Stimulation noch die IVM scheinen somit einen Einfluss auf den Methylierungsstatus dieser drei Gene zu haben.
Zusammenfassend spiegelte sich die reduzierte Entwicklungsfähigkeit von IVM-Eizellen aus adulten und präpuberalen Kühen nicht in abnormalen Methylierungsmustern der untersuchten geprägten und ungeprägten Gene wider. Dies lässt auf eine generelle Stabilität der etablierten DNA-Methylierungsprofile in Oozyten schließen. Aus diesem Grund müssen andere epigenetische Mechanismen als die DNA-Methylierung wie beispielsweise ncRNAs oder Histonmodifikationen zur Reduktion der Entwicklungskompetenz von präpuberalen und IVM-Oozyten beitragen. Diese Veränderungen behindern mutmaßlich die zytoplasmatische Reifung der Eizelle, welche wiederum zu einer späteren Beeinträchtigung der Entwicklung der Zygote und des Embryos führt.

Bei niedriggradigen Gliomen WHO I° und II° ist das Kontrastmittelverhalten variabel und nicht mit der Prognose assoziiert. Andererseits wurde bei spontanen Regressionen von niedriggradigen Gliomen bei Patienten sowohl mit als auch ohne eine Neurofibromatose Typ I eine Abnahme der Kontrastmittelaufnahme berichtet. Vielleicht deswegen verleitet eine Zunahme des Enhancements oder ein neu aufgetretenes Enhancement nicht selten zur Diagnose einer Tumorprogression. Es stellt sich also die Frage, ob eine Kontrastmittelaufnahme bei LGGs mit einer Größenzunahme somit einem Tumorprogress assoziiert ist.
Es lässt sich eine überdurchschnittliche Assoziation einer Änderung der Kontrastmittelaufnahme mit dem Wachstumsverhalten bei LGGs nachweisen. Allerdings ist das Maß für eine Progression oder Regression weiterhin die Größenänderung des Tumors. Auch eine neue Kontrastmittelaufnahme innerhalb eines Tumors hat keine Bedeutung für das aktuelle Staging. Sie findet sich zwar häufiger bei Wachstum aber auch bei regredienten Tumoren.
