Die periodische Katatonie ist eine eigenständige Erkrankung im Rahmen der differenzierten Klassifikation nach Kleist und Leonhard. Obwohl die Trennung Leonhards nach monopolaren, bipolaren affektiven und zykloiden Psychosen in moderne Klassifikationssysteme nach ICD 10 und DSM IV Einzug erhalten hat, werden Katatonien nur als Subtyp der Schizophrenie mit geringer Langzeitstabilität betrachtet.
Ziel dieser retrospektiven Studie ist die klinische Eigenständigkeit in einer Verlaufsbeschreibung der Symptome der periodischen Katatonie zu überprüfen. Dabei wurden 262 Patienten, bei denen durch klinisch geübte Untersucher eine periodische Katatonie diagnostiziert wurde, auf Verlaufsparameter und Soziobiographie retrospektiv untersucht.
Das erfasste Durchschnittsalter bei Ersthospitalisation der Patienten korrelierte mit den zuvor beschriebenen Ergebnissen von Leonhard, Männer erkranken mit 24 Jahren früher als Frauen mit 28 Jahren. Eine Erstmanifestation ab 45 ist selten, ab dem 60. Lebensjahr kommt dies ausschließlich bei Frauen vor.
Die Anzahl der stationären Aufenthalte korrelierte mit vorhandenen Ergebnissen einer älteren Studie und belief sich auf sechs im Durchschnitt.
Der überwiegende Anteil der Patienten (73%) ist zum Zeitpunkt des letzten stationären Aufenthaltes ledig und geht keiner Arbeit nach (50%). Der Anteil der männlichen Patienten mit einer abgeschlossenen Berufsausbildung (73%) ist höher ist als der weiblichen Patienten (48%). Ledige Patienten ohne vorhandene Schul- oder Berufsbildung werden in einem früherem Alter das erste mal stationär behandelt, stabile familiäre Situation und schulische bzw. berufliche Bildung scheinen eine protektive Wirkung auf den Ausbruch der Erkrankung zu haben.
Ein Alkohol- und Drogenkonsum fand sich zu Beginn der Erkrankung bei fast doppelt so vielen männlichen als bei weiblichen Probanden, im Rahmen des Krankheitsverlaufs nahm jedoch der Anteil der Frauen mit Alkohol- und Drogenkonsum zu.

Der Verlauf der Symptome von Psychomotorik, Affekt und Antrieb zeigt einen plötzlichen, schubförmigen Beginn der Erkrankung. Neben einem Wechsel zwischen Symptomen des Plus- und Minuspols, kommt es auch zu einem parallelen Auftreten, so dass typische Symptome der Mischform, wie zum Beispiel Stereotypien, Grimassieren und Negativismus entstehen. In der vorliegenden Untersuchung fand sich ausserdem die Entwicklung des von Leonhard beschriebenen Residualzustandes mit vorwiegenden Symptomen des Minuspols, aber wiederkehrenden Impulsvermehrungen im Verlauf.
Zeitlich betrachtet ist eine Abnahme der Dauer der stationären Behandlungszeiten zu vermerken. Diese Tatsache ist auf eine zunehmend gut entwickelte Sozio- und Pharmakotherapie, wie auch suffiziente ambulante, bzw. tagesklinische Weiterbehandlung zurückzuführen.
Die Untersuchung der Familienanamnese ergab einen hohen Anteil an einem psychisch erkrankten Elternteil (78%). Außerdem die Tatsache, dass Patienten mit erkrankten Verwandten ersten Grades in jüngeren Jahren eine Manifestation von ersten Symptomen einer periodischen Katatonie entwickeln, verglichen mit denen ohne familiäre Vorbelastung.
Bei einem kleinen Prozentsatz (2–3 %) aller molekulargenetisch untersuchten Hä-mophilie-A-Fälle konnte bislang keine kausale Mutation innerhalb der F8-Gen-Region aufgedeckt werden. Die molekularen Ursachen der Hämophilie dieser Patienten sollten im ersten Teil der vorliegenden Doktorarbeit mittels zusätzlicher Methoden aufgeklärt werden. Bei zwei Patienten mit milder Hämophilie A konnte je ein Basenaustausch im Promotorbereich des F8-Gens identifiziert werden. Um die Kausalität dieser Austausche zu überprüfen, wurden für diese und zwei weitere bereits publizierte Promotor-Mutationen Luciferase-Assays durchgeführt. Diese Ergebnisse machten deutlich, dass die nachgewiesenen Promotor-Mutationen die Aktivität des Promotors deutlich herabsetzen und daher als ursächlich einzustufen sind. Weiterhin wurden die übrigen Patienten auf epigenetische Veränderungen in fünf CpG-Inseln im 5’UTR und Intron 1 des F8-Gens untersucht. Hierbei konnten bei drei Patienten auffällige Methylierungsmuster nachgewiesen werden, wobei diese auf ein Klinefelter-Syndrom und genomische Veränderungen im Intron 1 zurückzuführen sind, nicht jedoch auf einen aberranten Methylierungsstatus, der die FVIII-Expression beeinflussen könnte. Mit Hilfe von mRNA-Untersuchungen konnten bei vier Patienten mit mutmaßlichen F8-Spleißmutationen aberrante F8-Transkripte nachgewiesen werden und somit die Kausalität der Mutationen geklärt werden. Des Weiteren wurden aus der Literatur alle bisher als kausal identifizierten stillen Mutationen und Spleißmutationen zusammengestellt, um mit diesen Ergebnissen die Spleißvorhersage-Software Alamut zu validieren. Die große Mehrzahl (78 %) der Spleißvorhersagen stimmte mit den Resultaten der mRNA-Untersuchungen (zumindest im Trend) überein, während es bei 22 % der Vorhersagen und mRNA-Analysen zu unterschiedlichen Resultaten kam. Innerhalb einer vorangegangenen Diplomarbeit konnten zehn Duplikationsbruch-punkte im F8-Gen von nicht verwandten Hämophilie-A-Patienten aufgeklärt werden. Diese wurden nun mit verschiedenen in-silico-Programmen analysiert, um die Sequenzumgebung der Bruchpunkt genauer zu beschreiben. Die Untersuchung ergab, dass verschiedene Mechanismen zur Entstehung von Duplikationen führen können und vermutlich mehrere Sequenzmotive in direkter Nähe der Bruchpunkte hierzu beitragen. Im Rahmen der molekulargenetischen Hämophilie-A-Diagnostik zum Nachweis von Intron-1- bzw. Intron-22-Inversionen sind einige Patienten mit schwerer Hämophilie A aufgefallen, welche ungewöhnliche Bandenmuster in den analytischen PCRs bzw. Southern-Blots aufwiesen. Mittels MLPA-Analysen wurden bei diesen Patienten Deletionen oder Duplikationen (CNVs) aufgedeckt, die meist allein die auffälligen Bandenmuster nicht erklären konnten. Weitere Long-Range-PCR-Untersuchungen belegten dagegen, dass fünf der untersuchten Fälle auf ein kombiniertes Inversions- und Duplikations- bzw. Deletionsereignis zurückzuführen sind. Als zweiter Teil der Arbeit wurden Transkriptions- und Translationsuntersuchungen von Nonsense-Mutationen des F8-Gens in einem zellulären Expressionssystem durchgeführt. Es konnte nachgewiesen werden, dass trotz Nonsense-Mutation eine komplette F8-Tran¬skrip¬tion stattfindet. Antigenanalysen konnten die Expression von trunkierten Proteinen nachweisen, wenn die Nonsense-Mutationen in der leichten Kette, d.h. den distalen Domänen A3, C1 oder C2, lag. Bei Nonsense-Mutationen in der schweren Kette (den proximalen Domänen A1, A2 oder B) war keine Proteinexpression nachweisbar. Diese Daten konnte durch intrazelluläre Immunlokalisation der trunkierten Proteine bestätigt werden. Die Ergebnisse deuten darauf hin, dass die B-Domäne eine wichtige Rolle bei der Proteinprozessierung spielt, vermutlich indem sie die Bindung von Chaperonen ermöglicht und das FVIII-Protein vor Degradation schützt.
Systems biology looks for emergent system effects from large scale assemblies of molecules and data, for instance in the human platelets. However, the computational efforts in all steps before such insights are possible can hardly be under estimated. In practice this involves numerous programming tasks, the establishment of new database systems but as well their maintenance, curation and data validation. Furthermore, network insights are only possible if strong algorithms decipher the interactions, decoding the hidden system effects. This thesis and my work are all about these challenges. To answer this requirement, an integrated platelet network, PlateletWeb, was assembled from different sources and further analyzed for signaling in a systems biological manner including multilevel data integration and visualization. PlateletWeb is an integrated network database and was established by combining the data from recent platelet proteome and transcriptome (SAGE) studies. The information on protein-protein interactions and kinase-substrate relationships extracted from bioinformatical databases as well as published literature were added to this resource. Moreover, the mass spectrometry-based platelet phosphoproteome was combined with site-specific phosphorylation/ dephosphorylation information and then enhanced with data from Phosphosite and complemented by bioinformatical sequence analysis for site-specific kinase predictions. The number of catalogued platelet proteins was increased by over 80% as compared to the previous version. The integration of annotations on kinases, protein domains, transmembrane regions, Gene Ontology, disease associations and drug targets provides ample functional tools for platelet signaling analysis. The PlateletWeb resource provides a novel systems biological workbench for the analysis of platelet signaling in the functional context of protein networks. By comprehensive exploration, over 15000 phosphorylation sites were found, out of which 2500 have the corresponding kinase associations. The network motifs were also investigated in this anucleate cell and characterize signaling modules based on integrated information on phosphorylation and protein-protein interactions. Furthermore, many algorithmic approaches have been introduced, including an exact approach (heinz) based on integer linear programming. At the same time, the concept of semantic similarities between two genes using Gene Ontology (GO) annotations has become an important basis for many analytical approaches in bioinformatics. Assuming that a higher number of semantically similar gene functional annotations reflect biologically more relevant interactions, an edge score was devised for functional network analysis. Bringing these two approaches together, the edge score, based on the GO similarity, and the node score, based on the expression of the proteins in the analyzed cell type (e.g. data from proteomic studies), the functional module as a maximum-scoring sub network in large protein-protein interaction networks was identified. This method was applied to various proteome datasets (different types of blood cells, embryonic stem cells) to identify protein modules that functionally characterize the respective cell type. This scalable method allows a smooth integration of data from various sources and retrieves biologically relevant signaling modules.
Spatiotemporale Organisation der Interaktion von Gq Protein-Untereinheiten und der Phospholipase Cβ3

Die G-Protein vermittelte Aktivierung der Phospholipase Cβ (PLCβ) stellt einen primären Mechanismus dar, um eine Vielzahl von physiologischen Ereignissen zu regulieren, z.B. die Kontraktion glatter Muskelzellen, Sekretion oder die Modulation der synaptischen Transmission. Sowohl Gαq- als auch Gβγ-Untereinheiten sind dafür bekannt mit PLCβ Enzymen zu interagieren und diese zu aktivieren. Über die Dynamik dieser Interaktion und den relative Beitrag der G-Protein Untereinheiten ist jedoch nur wenig bekannt. Unter Verwendung Fluoreszenz Resonanz Energie Transfer (FRET)- basierter Methoden in lebenden Zellen, wurde die Kinetik der Rezeptor-induzierten Interaktion zwischen Gβγ und Gαq Untereinheiten, die Interaktion von sowohl der Gαq als auch der Gβγ-Untereinheit mit der PLCβ3 und die Interaktion des regulator of G-Protein signaling 2 (RGS2) mit Gαq-Untereinheiten untersucht. Um die Untersuchung der Protein-Protein-Interaktion auf die Zellmembran zu beschränken, wurde die Total-Internal Reflection Fluorescence (TIRF) Mikroskopie angewandt. Zeitlich hoch auflösendes, ratiometrisches FRET-Imaging offenbarte eine deutlich schnellere Dissoziation von Gαq und PLCβ3 nach Entzug purinerger Agonisten verglichen mit der Deaktivierung von Gq Proteinen in der Abwesenheit der PLCβ3. Dieser offensichtliche Unterschied in der Kinetik kann durch die GTPase-aktivierende Eigenschaft der PLCβ3 in lebenden Zellen erklärt werden. Weiterhin zeigte es sich, dass PLCβ3 die Gq Protein Kinetik in einem ähnlich Ausmaß beeinflusst wie RGS2, welches in vitro deutlich effizienter darin ist, die intrinsische GTPase Aktivität der Gαq-Untereinheit zu beschleunigen. Als Antwort auf die Rezeptorstimulation wurde sowohl eine Interaktion von Gαq-Untereinheiten als auch von Gq-abstammende Gβγ-Untereinheiten mit der PLCβ3 beobachtet. Darüber hinaus zeigte sich auch eine Agonist-abhängige Interaktion von Gαq und RGS2. In Abwesenheit einer Rezeptorstimulation konnte kein spezifisches FRET-Signal zwischen Gq Proteinen und der PLCβ3 oder RGS2 detektiert werden. Zusammengefasst ermöglichte das ratiometrische FRET-Imaging in der TIRF Mikroskopie neue Einsichten in die Dynamik und Interaktionsmuster des Gq-Signalwegs.
Interleukin-6 (IL-6), oncostatin M (OSM), leukaemia inhibitory factor (LIF) and cardiotrophin-1 (CT-1) are members of the IL-6-type cytokine family that is characterised by sharing the common receptor subunit gp130. While the involvement of these polypeptides in cell differentiation, cell survival, proliferation, apoptosis, inflammation, haematopoiesis, immune response and acute phase reaction has already been demonstrated, the description of their role in development and progression of cardiac hypertrophy is still rather limited. A model has been postulated that declares the transient expression of IL-6-type cytokines as protective, while a continuous cardiac secretion of these proteins seems to be rather harmful for the heart. Within the first part of the study (results 4.1, 4.2 and 4.3) it was shown that OSM induces hypertrophy of primary neonatal rat cardiomyocytes (NRCM), just as its related cytokines LIF, CT-1 and hIL-6/hsIL-6R (hsIL-6R, human soluble IL-6 receptor). Regarding the hypertrophic potentials the LIFR/gp130 utilising cytokines (hLIF, hOSM and hCT-1) are stronger inducers than the OSMR/gp130 utilising mOSM. Human IL-6/hsIL-6R which signals via a gp130 homodimer has the weakest hypertrophic effect. The thorough analysis of typical signalling pathways initiated by IL-6-type cytokines revealed that STAT3 phosphorylation at Y705 seems to be the most important hypertrophy promoting pathway. In addition and in contrast to published work, we clearly demonstrate that classical IL-6 signalling (upon pure IL-6 treatment) has no hypertrophic effect on cardiomyocytes, because they lack sufficient amounts of the membrane-bound IL-6R. This is also true for neonatal rat cardiac fibroblasts (NRCFB). Since these cells can also influence cardiac hypertrophy, signalling pathways and target genes were additionally examined in NRCFB in response to OSM, LIF and IL-6/sIL-6R. One of the key findings of this thesis is the selective change in expression of cytokines and receptors of the IL-6 family in both cell types upon IL-6-type cytokine stimulation. A striking difference between NRCM and NRCFB is the fact that the target gene induction in NRCM is of similar duration upon mOSM and hIL-6/hsIL-6R treatment, while hIL-6/hsIL-6R is capable of promoting the induction of OSMR and IL-6 significantly longer in NRCFB. By searching for transcription factors or intermediate cytokines which could be responsible for this difference, a strong correlation between increased Il6 transcription and amount of mRNA levels for C/EBPβ and C/EBPδ was observed in response to IL-6/sIL-6R stimulation. Interestingly, mOSM also mediates the induction of C/EBPβ and δ, but the initiation is significantly less efficient than in response to IL-6/sIL-6R. Therefore, we assume that mOSM stimulation fails to reach threshold values required for a prolonged IL-6 secretion. Since we additionally observe a slight IL-6R mRNA upregulation in NRCFB, we assume that the combination of IL-6, LIF, C/EBPβ, C/EBPδ and IL-6R expression might be responsible for the observed different kinetics with which IL-6 and OSM stimulate NRCFB. In addition to the aforementioned proteins, members of the renin-angiotensin system seem to support the IL-6-type cytokine mediated hypertrophy. Since it has already been shown that angiotensin II vice versa induces IL-6 expression in NRCM and NRCFB, this enhanced expression of AT1α and ACE could be of crucial interest for the hypertrophy supporting phenotype. The second part of the presented work dealt with the characterisation of the receptor complexes of rat OSM. The central question of this analysis was, whether rOSM, just like mOSM, only binds the type II (OSMR/gp130) receptor complex or is able to utilise the type II and type I (LIFR/gp130) receptor complex. Using different experimental approaches (knock-down of the OSMR expression by RNA interference, blocking of the LIFR by LIF-05, an antagonistic LIF variant, and generation of stably transfected Ba/F3 cells expressing the newly cloned rat OSMR/gp130 or LIFR/gp130 receptor complex) we can clearly show that rat OSM surprisingly utilises both, the type I and type II receptor complex. Therefore it closely mimics the human situation. Furthermore, rOSM displays cross-species activities and stimulates cells of human as well as murine origin. Its signaling capacities closely mimic those of human OSM in cell types of different origin in the way that strong activation of the JAK/STAT, the MAP kinase as well as the PI3K/Akt pathways can be observed. Therefore, the results obtained in the last section of this thesis clearly suggest that rat disease models would allow evaluation of the relevance of OSM for human biology much better than murine models.
Glioblastoma multiforme (GBM) represents the most aggressive form of malignant brain tumors and remains a therapeutically challenge. Intense research in the field has lead to the testing of oncolytic viruses to improve tumor control. Currently, a variety of different oncolytic viruses are being evaluated for their ability to be used in anti-cancer therapy and a few have entered clinical trials. Vaccinia virus, is one of the viruses being studied. GLV-1h68, an oncolytic vaccinia virus engineered by Genelux Corporation, was constructed by insertion of three gene cassettes, RUC-GFP fusion, β-galactosidase and β- glucuronidase into the genome of the LIVP strain. Since focal tumor radiotherapy is a mainstay for cancer treatment, including glioma therapy, it is of clinical relevance to assess how systemically administered oncolytic vaccinia virus could be combined with targeted ionizing radiation for therapeutic gain. In this work we show how focal ionizing radiation (IR) can be combined with multiple systemically delivered oncolytic vaccinia virus strains in murine models of human U-87 glioma. After initial experiments which confirmed that ionizing radiation does not damage viral DNA or alter viral tropism, animal studies were carried out to analyze the interaction of vaccinia virus and ionizing radiation in the in vivo setting. We found that irradiation of the tumor target, prior to systemic administration of oncolytic vaccinia virus GLV-1h68, increased viral replication within the U-87 xenografts as measured by viral reporter gene expression and viral titers. Importantly, while GLV-1h68 alone had minimal effect on U-87 tumor growth delay, IR enhanced GLV-1h68 replication, which translated to increased tumor growth delay and mouse survival in subcutaneous and orthotopic U-87 glioma murine models compared to monotherapy with IR or GLV-1h68. The ability of IR to enhance vaccinia replication was not restricted to the multi-mutated GLV-1h68, but was also seen with the less attenuated oncolytic vaccinia, LIVP 1.1.1. We have demonstrated that in animals treated with combination of ionizing radiation and LIVP 1.1.1 a strong pro-inflammatory tissue response was induced. When IR was given in a more clinically relevant fractionated scheme, we found oncolytic vaccinia virus replication also increased. This indicates that vaccinia virus could be incorporated into either larger hypo-fraction or more conventionally fractionated radiotherapy schemes. The ability of focal IR to mediate selective replication of systemically injected oncolytic vaccinia was demonstrated in a bilateral glioma model. In mice with bilateral U-87 tumors in both hindlimbs, systemically administered oncolytic vaccinia replicated preferentially in the focally irradiated tumor compared to the shielded non- irradiated tumor in the same mouse We demonstrated that tumor control could be further improved when fractionated focal ionizing radiation was combined with a vaccinia virus caring an anti-angiogenic payload targeting vascular endothelial growth factor (VEGF). Our studies showed that following ionizing radiation expression of VEGF is upregulated in U-87 glioma cells in culture. We further showed a concentration dependent increase in radioresistance of human endothelial cells in presence of VEGF. Interestingly, we found effects of vascular endothelial growth factor on endothelial cells were reversible by adding purified GLAF-1 to the cells. GLAF-1 is a single- chain antibody targeting human and murine VEGF and is expressed by oncolytic vaccinia virus GLV-109. In U-87 glioma xenograft murine models the combination of fractionated ionizing radiation with GLV-1h164, a vaccinia virus also targeting VEGF, resulted in the best volumetric tumor response and a drastic decrease in vascular endothelial growth factor. Histological analysis of embedded tumor sections 14 days after viral administration confirmed that blocking VEGF translated into a decrease in vessel number to 30% of vessel number found in control tumors in animals treated with GLV-164 and fractionated IR which was lower than for all other treatment groups. Our experiments with GLV-1h164 and fractionated radiotherapy have shown that in addition to ionizing radiation and viral induced tumor cell destruction we were able to effectively target the tumor vasculature. This was achieved by enhanced viral replication translating in increased levels of GLAF-2 disrupting tumor vessels as well as the radiosensitization of tumor vasculature to IR by blocking VEGF. Our preclinical results have important clinical implications of how focal radiotherapy can be combined with systemic oncolytic viral administration for highly aggressive, locally advanced tumors with the potential, by using a vaccinia virus targeting human vascular endothelial growth factor, to further increase tumor radiation sensitivity by engaging the vascular component in addition to cancer cells.
The mechanisms that enable cells to regulate their gene expression and thus their metabolism, proliferation or cellular behaviour are not only important to understand the basic biology of a living cell, but are also of crucial interest in cancerogenesis. Highly interwoven and tightly regulated pathways are the basis of a robust but also flexible regulatory network. Interference with these pathways can be either causative for tumorigenesis or can modify its outcome. The receptor tyrosine kinase (RTK) and RAS dependent pathways leading to AKT or ERK1/2 activation are of particular interest in melanoma. These signaling modules are commonly activated by different mutations that can be found in various pathway components like NRAS, BRAF or PTEN. The first part of this work deals with the diverse and versatile functions of the ERK1/2 pathway feedbackregulator MKP2 in different cellular, melanoma relevant settings. In addition, a functional role of the AP1-complex member FOSL1, an ERK1/2 transcriptional target being implicated in the regulation of proliferation, is demonstrated. Secondly, aspects of direct pharmacological inhibition of the ERK1/2 pathway with regard to the induction of apoptosis have been analysed. Due to the high frequency of melanoma related mutations occurring in the RAS/RAF/MEK/ERK pathway (e.g. NRASQ61K, BRAFV600E), inhibition of this signaling cascade is deemed to be a promising therapeutic strategy for the treatment of malignant melanoma. However, although in clinical trials mono-therapeutic treatment with MEK- or RAF inhibitors was successful in the short run, it failed to show satisfactory long-lasting effects. Hence, combination therapies using a MAPK pathway inhibitor and an additional therapy are currently under investigation. I was able to demonstrate that inhibition of MEK using the highly specific inhibitor PD184352 can have a protective effect on melanoma cells with regard to their susceptibility towards the apoptosis inducing agent cisplatin. Single application of cisplatin led to strong DNA damage and the induction of caspase-dependent apoptosis. Additional administration of the MEK inhibitor, however, strongly reduced the apoptosis inducing effect of cisplatin in several melanoma cell lines, These cells displayed an increased activation of the serine/threonine kinase AKT after MEK inhibition. This AKT activation concomitantly led to the phosphorylation of FOXO transcription factors, attenuating the cisplatin induced expression of the BH3-only protein PUMA. PUMA in turn was important to mediate the apoptosis machinery after cisplatin treatment. My results also indicate a participation of RTKs, in particular EGFR, in mediating MEK inhibitor induced activation of AKT. These results demonstrate that inhibition of the RAS/RAF/MEK/ERK signaling pathway in melanoma cell lines does not necessilary have favourable effects in a cytotoxic co-treatment situation. Instead, it can even enhance melanoma survival under pro-apoptotic conditions.
γ-Aminobutyric acid type A receptors (GABAARs) and glycine receptors (GlyRs) are the major mediators of fast synaptic inhibition in the central nervous system. For proper synaptic function their precise localization and exact concentration within the neuronal surface membrane is essential. These properties are mediated by scaffolding proteins which directly contact the large intracellular loops of the receptors and tether them to cytoskeletal elements of the neuronal cells. In my thesis I deciphered the molecular details of several underlying protein-protein interactions, namely the interaction of a subset of GABAAR and GlyR subunits with the scaffolding proteins gephyrin, radixin and collybistin. I determined short linear motifs within the large intracellular loops of the receptors that directly engage in subunit specific scaffold protein interactions. My quantitative binding studies revealed that gephyrins E domain primarily recognizes the GABAAR α1 (Kd = 17 M) and α3 (Kd = 5 M) subunits, in contrast, the SH3 domain of collybistin mainly interacts with the GABAAR α2 subunit (Kd = 1 µM), while the FERM domain of radixin tightly binds to the GABAAR α5 subunit (Kd = 8 µM). My work additionally demonstrated that this simple relationship is complicated by (i) missing or (ii) overlapping binding specificities between the scaffold proteins and the receptor subunits. Moreover, this thesis addressed the possibility of (iii) posttranslational negative regulation as well as amplification generated by (iv) avidity effects as summarized below. (i) First, using biochemical methods I mapped the radixin-GABAAR α5 interaction in detail. My structural analysis and competition assays suggest that radixin mediates the receptor subunit binding via a universal binding site within the F3 subdomain of its FERM domain. This binding site is formed by an α-helix that offers a large hydrophobic pocket, which accepts a variety of different hydrophobic residues adopting different conformations, and a β-strand that readily engages in peptide backbone interactions. Not surprisingly, this binding site has been implicated in a wide variety of different scaffold interactions, thus emphasizing the importance of the essential FERM activation mechanism described earlier and suggesting additional pathways to allow tight regulation of this interaction. (ii) Next, I analyzed in detail the process of gephyrin-mediated GABAAR clustering. My X-ray crystallographic studies and binding assays revealed that gephyrin mediates binding of the GABAAR α1, α2 and α3 subunit via a universal binding site that also mediates the interactions with the GlyR β subunit. Using structure-guided mutagenesis I identified key residues within gephyrin and the receptor subunits that act as major contributors to the overall binding strength. Namely, two conserved aromatic residues within the N-terminal half of the receptor binding region engage in crucial hydrophobic interactions with gephyrin. Accordingly, J. Mukherjee from the group of our collaborator Steven J. Moss verified a substantial decrease in GABAAR cluster number and size in primary hippocampal neurons upon exchange of these residues within the GABAAR α2 subunit. Extension of my studies to collybistin (CB) revealed an overlapping but reciprocal subunit preference for this protein in comparison to gephyrin. The GABAAR α3 subunit exclusively binds gephyrin, in contrast the GABAAR α1 subunit mainly targets gephyrin (Kd = 17 µM) but additionally displays a moderate affinity (Kd ≈ 400 µM) towards the SH3 domain of CB. The GABAAR α2 subunit binds tightly to the SH3 domain of CB (Kd = 1 µM) and additionally displays a weak gephyrin affinity (Kd ≈ 500 µM). Notably, I could exclude the possibility of synergistic effects between gephyrins E domain, the SH3 domain of CB and the GABAAR α2 subunit. Instead, I found that the GABAAR α2 subunit binds gephyrin and CB in a mutually exclusive manner. These results suggest that CBs role in receptor clustering is solely determined by competing binding events of its constituting domains. Namely, the intra-molecular association between the PH/DH domain and the SH3 domain within CB competes with different inter-molecular interactions of CB: GABAAR α2 binding to the SH3 domain, PIP2 binding to the PH domain and gephyrin presumably binding to the PH and DH domain of CB. (iii) Interestingly, the receptor motifs, which have been mapped in my thesis to directly interact with the scaffold proteins, were shown in earlier studies to be posttranslationally modified in vivo. In particular, the GABAAR α1 and GlyR β subunits have been implicated as targets of the ERK/MAPK and PKC phosphorylation-pathways, respectively, while the GABAAR α5 subunit motif was shown to be ubiquitinated. In this dissertation, I analyzed Thr348, a possible ERK phosphorylation site within GABAAR α1. My binding assays verified a severe reduction of the direct gephyrin binding strength upon introduction of the respective phosphomimetic residue. The relevance of this in vitro result was highlighted by J. Mukherjee who confirmed a significant reduction in GABAAR cluster number and size upon introduction of the same mutation. The ERK/MAPK pathway is therefore a promising candidate for regulation of GABAergic transmission. (iv) In vivo, gephyrin presumably forms a multivalent scaffold, which is based on the self-association of its G (GephG) and E domains (GephE). Given the multimeric nature of gephyrin and the pentameric receptor architecture, I tested the possibility of avidity in the clustering of inhibitory neurotransmitter receptors. Cocrystallization of selected minimum peptides with GephE and their crystal structure analyses enabled me to define a receptor-derived peptide that offers a maximized gephyrin affinity. The structure of the GephE-GlyR receptor complex reveals two receptor-binding sites in close spatial vicinity (15 Å). I therefore designed bivalent peptides that enable to target both GephE sites at the same time and, as expected, a variety of biophysical methods verified an avidity-potentiated and unmatched high gephyrin affinity for these bidentate compounds. Notably, I could extend the dimerization approach to low affinity gephyrin ligands, namely short GABAAR-derived peptides that could not be studied using conventional monomeric ligands. Additionally, I verified that this compound specifically targets GephEs receptor binding site, and that it thereby inhibits its receptor binding activity. Further development of this molecule may offer the possibility to specifically analyze the effect of uncoupling the gephyrin-receptor interaction in cell culture-based assays, without altering protein function or expression level that accompanies conventional methods such as protein knock-out, RNA interference or the usage of antibodies.
The objective of this prospective observational controlled study was to evaluate potential effects and dose-response relationship of LT4 administration on BMD, parameters of bone and muscle strength, and biochemical variables of calcium homoeostasis and bone turnover.Ninety-seven men and pre-menopausal women after near total thyroidectomy and ¹³¹I remnant ablation for well-differentiated thyroid carcinoma or after strumectomy for non-toxic goitre were stratified by degree of TSH suppression and by gender in three subgroups: 28 men and 46 women on LT4 suppressive treatment and 23 women on LT4 replacement therapy. Patients were matched for age, gender and BMI to 89 healthy controls with a negative history of thyroid disease. Patients and controls were followed and studied for a mean time of 1.1±0.2 years. Peripheral volumetric total and trabecular BMD as well as bone strength (pQCT) were determined at the ultra-distal radius. Central areal BMD (DXA) was measured at the lumbar spine, left and right femoral neck as well as left and right total hip. Maximum grip strength (dynamometer) of the non-dominant forearm and serum markers of calcium and bone metabolism were assessed. BMD at the axial skeleton and muscle strength were not impaired by LT4 medication irrespective of gender, underlying diagnosis or treatment regimen. By contrast, a general trend of inversely affected total and trabecular BMD and of decreased bone strength was detected at the ultra-distal radius. Only in women on LT4 suppressive treatment, loss of total BMD at the ultra-distal radius reached a level of high significance. In women on LT4 replacement therapy, a significant decline of maximum grip strength appeared in comparison with female controls, while appendicular total and trabecular BMD as well as bone strength remained unchanged and did not differ from respective controls. In men on LT4 suppressive treatment, greater reduction of bone strength as compared to female thyroid cancer patients was marginally significant. Calcium balance was stable and serum concentrations of bone metabolism markers levelled off or rather decreased contradicting (high turnover) bone loss. The study did not reveal any dose-related differential influence of LT4 administration either on primary or secondary study endpoints in female patients. A gender-related difference of bone strength in response to LT4 suppressive treatment might not be excluded, as male thyroid cancer patients showed greater decline of bone strength despite unaffected peripheral BMD and muscle strength. In conclusion, there was only little evidence of adverse LT4 effects. For the most part, LT4 administration irrespective of degree of TSH suppression was not associated with low or accelerated loss of BMD at the peripheral and central skeleton and loss of bone and muscle strength, a finding also confirmed biochemically. The ultra-distal radius as a non-weight bearing skeletal site might be at risk for BMD reduction. According to the results, pre-menopausal women on LT4 suppressive therapy might be at risk of bone loss. The more complex approach of this study also took into account biomechanical qualities of bone material as well as structural and geometrical characteristics of bone architecture implying a causal muscle-bone interrelationship.
Fanconi anemia (FA) is an autosomal recessive or X-chromosomal inherited disorder, which is not only phenotypically but also genotypically very heterogeneous. While its hallmark feature is progressive bone marrow failure, many yet not all patients suffer additionally from typical congenital malformations like radial ray defects and growth retardation. In young adulthood the cumulative risk for developing hematological or other malignancies is compared to the general population several hundred-fold increased. The underlying molecular defect is the deficiency of DNA interstrand crosslink (ICL) repair. ICLs are deleterious lesions, which interfere with crucial cellular processes like transcription and replication and thereby can lead to malignant transformation, premature senescence or cell death. To overcome this threat evolution developed a highly complex network of interacting DNA repair pathways, which is conserved completely only in vertebrates. The so called FA/BRCA DNA damage response pathway is able to recognize ICLs on stalled replication forks and promotes their repair through homologous recombination (HR). Today we know 15 FA genes (FANCA, -B, -C, -D1, -D2, -E, -F, -G, -I, -J, -L, -M, -N, -O and -P) whose products are involved in this pathway. Although more than 80% of FA patients carry biallelic mutations in either FANCA, FANCC or FANCG, there are still some who cannot be assigned to any of the known complementation groups. This work aimed to indentify the di¬sease causing mutations in a cohort of those unassigned patients. Initial screens of the candidate genes FAN1, MHF1 and MHF2 did not reveal any pathogenic alterations. Moreover, FAN1 could be excluded as FA candidate gene because patients carrying a homozygous microdeletion including the FAN1 locus did not show a phenotype comparable to FA patients. In the case of MHF1 and MHF2 the reason for the negative screening result is not clear. Mutation carriers might be rare or, regarding the diverse and also FA pathway independent protein functions, phenotypically not comparable to FA patients. Nevertheless, this study contri¬buted to the identification and characterization of the most recent members of the FA pathway - RAD51C (FANCO), SLX4 (FANCP) and XPF (FANCQ). FANCO is one of the RAD51 paralogs and is involved in crucial steps of HR. But since the only reported FA-O patient has so far not developed any hematological anomalies, FANCO is tentatively designated as gene underlying an FA-like disorder. In contrast, patients carrying biallelic mutations in FANCP do not only show hematological anomalies, but as well congenital malformations typical for FA. The distinct role of FANCP in the FA pathway could not be determined, but it is most likely the coordination of structure-specific nucleases during ICL excision. One of these nucleases is the heterodimer XPF/ERCC1. XPF is probably disease causing in the complementation group FA-Q and is the first FA gene, which was identified by Next Generation Sequencing (NGS). Extraordinarily is that mutations in this gene had previously been reported to cause two other disorders, xeroderma pigmentosum and segmental progeria. Despite some overlaps, it was shown that the divergent phenotypes could clearly be distinguished and are caused by distinct functional defects of XPF. Additionally, this work aimed to improve and accelerate the genotyping process of FA patients in general. Therefore, classical approaches should be complemented or fully replaced by approa¬ches using NGS. Massively parallel sequencing of the whole exome proved to be most appro¬priate and the establishment of an FA-specific analysis pipeline facilitated improved molecular diagnostics by combining complementation group assignment and mutation analysis in one step. Consequently two NGS studies revealed the pathogenic defect in several previously unassigned FA patients and thereby added another patient to one of the most recent subtypes, FA-P. In summary, this work contributed not only to further completion of the FA/BRCA DNA repair network by adding three novel genes, it also showed that classical molecular approaches for re¬search as well as for diagnostics could be replaced by NGS.
In der Nachsorge des Medulloblastoms wird standardmäßig auf die Bildgebung mittels Magnetresonanztomographie zurück gegriffen. Da die Erkrankung vor allem entlang der Liquorwege in Form einer kranialen oder spinalen Meningeose metastasiert, wurde anhand Daten der Therapieoptimierungsstudie "HIT 2000" verglichen, welche Lokalisation am häufigsten betroffen ist. Es zeigte sich, dass zu einem hohen Prozentanteil vor allem eine kombinierte Meningeose im Rezidiv oder Progress auftritt, gefolgt von einer kranialen Metastasierung. Dennoch gibt es eine Gruppe an Patienten, die eine isolierte spinale Meningeose entwickeln.
Das Nebennierenrindenkarzinom (NNR-Ca) gehört mit einer Inzidenz von 1-2/1000000 zu den seltenen malignen Neubildungen. Neben Sarkomen, Hirntumoren, Brustkrebs und Leukämie gehört das NNR-Ca zu den Kerntumoren, durch die das selten vorkommende autosomal dominante Tumor-Prädispositions Syndrom, das Li Fraumeni Syndrom (LFS) gekennzeichnet ist. Das LFS wird mit Keimbahnmutationen im Tumorsuppressor Gen TP53 in Verbindung gebracht. Die vorliegende Arbeit untersucht TP53 Keimbahnmutationen und -polymorphismen und ihre Auswirkung auf klinische Faktoren bei einem großen Kollektiv von erwachsenen NNR-Ca Patienten.
Es wurde DNS aus Blut und teilweise aus Tumorgewebe von Patienten aus dem Deutschen Nebennierenrindenkarzinom Register extrahiert und die Exons 2 bis 11 von TP53 sequenziert. Darüber hinaus wurde der Nachweis der Mutationen und eines Loss of Heterozgosity von TP53 im Tumorgewebe und die immunhistochemische Färbung von p53 vorgenommen. Die anschließende Auswertung der Daten erfolgte unter Einbeziehung des klinischen Verlaufs der Krankheit bei den Patienten.
In dieser Arbeit konnten vier NNR-Ca Patienten (3,9 %) mit mindestens einer Keimbahnmutation im TP53 identifiziert werden, bei den unter 40-jährigen entspricht dies einem Anteil von 13,0 %. Unter der Altersgrenze von 40 Jahren sollte daher ein TP53 Mutationsscreening erwogen werden.
Die Auswertung der Polymorphismen zeigte, dass diese einen Einfluss auf die Entstehung und den klinischen Verlauf des NNR-Cas zu haben scheinen, jedoch weitere Studien nötig sind.
Das Polyomavirus WU (WUPyV) wurde erstmalig im Jahr 2007 in respiratorischem Material bei Patienten mit respiratorischem Infekt beschrieben. Charakterisierung, Epidemiologie und Beurteilung des Krankheitswerts des neuen Virus sind seither Gegenstand vieler Studien weltweit. Retrospektiv wurde Probenmaterial aus dem Respirationstrakt auf WUPyV mittels PCR untersucht. Das Material war zur virologischen Routinediagnostik eingegangen und stammte von in der Universitätskinderklinik Würzburg stationär behandelten Kindern, deren klinische Diagnosen anonymisiert zur Verfügung standen. Es wurden 1277 Nasenrachensekrete (NRS) berücksichtigt aus dem Zeitraum zwischen Januar 2002 und September 2005 sowie zwischen Januar und Juli 2007. Von 1277 NRS waren 62 (4,9 %) positiv für WUPyV. Das Virus wurde in jedem Monat eines Jahres nachgewiesen, wobei Wintermonate insgesamt stärker vertreten waren. Das mediane Alter der betroffenen Patienten betrug 3,0 Jahre (4 Monate – 6,3 Jahre). Klinische Diagnosen bei WUPyV-Infektionen umfassten ein breites Spektrum an oberen und unteren Luftwegserkrankungen. Bei 33 NRS (53,2 %) waren neben WUPyV zuvor ein oder zwei weitere respiratorische Viren durch PCR oder Immunfluoreszenz-Antigentest nachgewiesen worden (Adenovirus: 10; Influenza A: 10; humanes Bocavirus: 9; RSV: 5; Parainfluenzavirus 1/2/3: 3). Die Sequenzanalyse eines 647 bp langen Abschnitts der nicht kodierenden Region bei 50 WUPyV-positiven NRS zeigte eine Übereinstimmung der Sequenzen von 98,5 %. Die Ergebnisse der vorliegenden Studie unterstützen bisherige Ergebnisse zur Epidemiologie und Verbreitung des WUPyV. Demnach konnte WUPyV-DNA bei akuter respiratorischer Infektion im menschlichen Respirationstrakt, bevorzugt bei Kleinkindern, detektiert werden. WUPyV wies eine hohe Koinfektionsrate mit anderen respiratorischen Viren auf. Es zeigte sich in der phylogenetischen Analyse zweier Genomabschnitte eine geringe Variabilität des Genoms. Bei bisheriger Datenlage bleibt unklar, ob der Nachweis von WUPyV-DNA in NRS mit einer akuten respiratorischen Erkrankung assoziiert werden kann.
Dieses Projekt soll zur Entwicklung eines Bluttests beitragen, um bei allogen stammzelltransplantierten (allo-SZT) Patienten eine drohende akute Graft-versus-Host
Disease (aGVHD) vorhersagen und von einer Infektionskomplikation unterscheiden zu
können. Die aGVHD und opportunistische Infektionen, wie etwa eine Zytomegalievirus-
Infektion, stellen die Hauptrisiken der allo-SZT dar. Eine durch einen prädiktiven Test verbesserte Vorhersage bzw. Differenzierung dieser beiden schweren omplikationen
könnte zu einem breiteren Einsatz der allo-SZT führen.
Eine Reihe von Erkenntnissen lassen die Vermutung zu, dass bestimmte Homing-
Rezeptoren auf T-Zellen im peripheren Blut als Marker zur Vorhersage und Differenzierung zwischen einer aGVHD und Infektionskomplikationen dienen könnten. So wurde in Mausmodellen gezeigt, dass die Pathogenese der aGVHD ein streng zeitlich regulierter und organspezifischer Immunprozess ist. Alloreaktive T-Zellen müssen bestimmte Homing-Rezeptoren exprimieren, um in die Zielorgane der aGVHD (Gastrointestinaltrakt, Leber und Haut) einwandern zu können. Die Immunreaktion auf eine Infektion setzt ebenfalls voraus, dass in der adaptiven Immunantwort T-Zellen organ- und entzündungsspezifische Homing-Rezeptoren exprimieren.
Eine zentrale Voraussetzung für einen prospektiven klinischen Test mit allo-SZT Patienten ist die Kenntnis der physiologischen Bandbreite der Oberflächenmarker auf
T-Lymphozyten in Gesunden, um diese von pathophysiologischen Veränderungen unterscheiden zu können. Ziel dieser Arbeit war es daher, eine effiziente und zuverlässige Methode zu entwickeln, um das Homing-Rezeptor-Profil von humanen T-Zellen im peripheren Blut analysieren zu können. Dazu wurde ein urchflusszytometrie-Test mit 25 Oberflächenmarkern etabliert, die eine Rolle in der Pathogenese der aGVHD und bei Infektionen spielen könnten. Mit diesem Test wurde die Expression der
Oberflächenmarker von 21 gesunden Probanden an 8 Zeitpunkten über einen Zeitraum von 3 Wochen analysiert.
Die untersuchten Oberflächenmarker lassen sich dabei entsprechend ihrer Expressionsmuster in drei Kategorien einteilen, nämlich (I) zeitlich stabile niedrige,
(II) zeitlich stabile hohe und (III) dynamisch schwankende Expression. Aufbauend auf diesen Ergebnissen für gesunde Probanden wurde am Universitätsklinikum Würzburg eine prospektive klinische Studie an aGVHD-Patienten begonnen.
Das Europarecht stellt eine gemeinsame Grundlage für die Förderung der erneuerbaren Energien in den Mitgliedstaaten dar. Sowohl die Slowakei als auch Deutschland mussten die Richtlinie 2009/28/EG in ihr nationales Recht umsetzen. Dabei ergeben sich Gemeinsamkeiten, aber auch Unterschiede. In der vorliegenden Arbeit werden die Förderungspolitiken beider Staaten erörtert und auch der Frage nachgegangen, inwiefern sie den Anforderungen der Richtlinie gerecht werden. Politische, kulturelle und historische Hintergründe spielen hierbei eine wichtige Rolle. Die Arbeit enthält auch Empfehlungen zum Handlungsbedarf.
Elektrooptische Transporteigenschaften und stochastisch aktivierte Prozesse Resonanter Tunneldioden

Im Rahmen der vorliegenden Arbeit wurden elektrooptische Transporteigenschaften und stochastisch aktivierte Prozesse Resonanter Tunneldioden (RTDs) bei Raumtemperatur untersucht. Die RTDs wurden auf dem III-V Halbleitermaterialsystem AlGaAs/GaAs durch Molekularstrahlepitaxie, Elektronenstrahllithographie und trockenchemischen Ätztechniken hergestellt. Im Bereich des negativen differentiellen Leitwerts konnte bistabi-les Schalten und hierbei stochastisch aktivierte Dynamik nichtlinearer Systeme untersucht werden. Die Flächenabhängigkeit der Ätzrate konnte ausgenutzt werden, um RTDs mit einem Stamm und zwei Transportästen zu realisieren, welche hinsichtlich ihrer optischen und elektrischen Eigenschaften untersucht wurden. Im ersten experimentellen Abschnitt 3.1 werden die elektrischen Transporteigenschaften Resonanter Tunneldioden bei Raum-temperatur und die Flächenabhängigkeit des kohärenten und nicht-kohärenten Elektronen-transports analysiert. Die Realisierung universeller logischer Gatter (NOR und NAND) und deren Rekonfigurierbarkeit durch einen externen Kontrollparameter wird in Abschnitt 3.2 gezeigt. In Abschnitt 3.3 wird die Lichtsensitivität Resonanter Tunneldioden als Photode-tektoren für den sichtbaren Wellenlängenbereich und in Abschnitt 3.4 für die Telekommu-nikationswellenlänge bei λ = 1,3 µm demonstriert.
Das Prostatakarzinom (PCa) ist weltweit das häufigste Malignom des Mannes mit weiter steigenden Inzidenz. Leider ist es bisher mit den gebräuchlichen Prognosefaktoren nicht möglich, diesen klinisch sehr heterogenen Tumor vor einer chirurgischen Therapie hinsichtlich des postoperativen Rezidivrisikos zu charakterisieren. Die Einführung neuer, hochsensitiver Biomarker, die diese Aufgabe zuverlässig erfüllen können, stellt vor allem für die Entwicklung individualisierter Therapieregimes von Patienten mit einem high-risk-PCa eine aktuelle Herausforderung dar. MicroRNAs rückten in den letzten Jahren als potentielle Biomarker zunehmend in den wissenschaftlichen Fokus. Sie stellen kleine RNA-Moleküle dar, welche posttranskriptionell die Genexpression in Organismen steuern und auch in der Regulation von Tumorsuppressoren und Onkogenen eine Rolle spielen. Differentielle Expressionslevels verschiedener RNAs wurden in sehr vielen Tumorentitäten, wie auch dem PCa beobachtet. Vorarbeiten aus unserem Labor zeigten, dass zwei microRNAs, miR-205 und miR-221, im PCa sehr stark unterexprimiert werden. Im Falle von mir-221 konnte zudem bereits eine Korrelation mit dem klinischen Verlauf und der Prognose des high-risk-PCa dargestellt werden. Aufbauend auf diesen Vorarbeiten sollte in einem Teilprojekt der vorliegenden Arbeit die tumorsuppressive Funktion von miR-221 im PCa untersucht werden. Mittels transienter Überexpression von miR-221 in PCa-Zelllinien konnte die Bedeutung von miR-221 als Regulator der Proliferation in PCa-Zellen beschrieben werden. Um die molekulare Wirkweise von miR-221 weiter darzustellen, wurde die posttranskriptionelle Inhibierung von p27kip1 und c-kit, zwei bekannten miR-221-Zielgenen, untersucht. In sogenannten Luciferase-Assays konnte eine direkte Interaktion von miR-221 mit definierten Sequenzen im 3‘-untranslatierten Bereich (=3‘UTR) beider Gene nachgewiesen werden. Erwartungsgemäß wurde aber in primären PCa-Tumoren, die anders wie viele andere Tumorentitäten eine reduzierte miR-221-Expression aufwiesen, keine Überexpression des potentiellen Tumorsuppressors p27kip1 beobachtet. Somit ist nachgewiesen, dass miR-221 zwar prinzipiell den Tumorsuppressor p27kip1 regulieren kann, dass allerdings dieser Mechanismus in primären PCa für die Regulation des Tumorsuppressors p27kip1 keine übergeordnete Rolle spielt. Anders verhält es sich bei dem potentiellen Onkogen c-kit. Die von mir druchgeführten Untersuchungen können erstmals eine direkte Interaktion von c-kit und miR-221 in PCa-Zellen nachweisen. Somit kann die beschriebene Proliferations-inhibierung und Apoptoseinduktion nach ektopischer miR-221-Überexpression im Zellmodell mit einer miR-221-vermittelten c-kit-Inhibierung in Zusammenhang gebracht werden. Dieses Ergebnis wird durch den Nachweise einer inversen Assoziation der mir-221- und ckit-Expression in primären PCa-Fällen untermauert (nicht gezeigte Ergebnisse). Betrachtet man diese Ergebnisse in Zusammenhang mit der bekannten tumor-progressiven Funktionen von c-kit, könnte durch die fehlende/reduzierte miR-221-Inhibierung der c-kit-Translation die Entstehung und Progression vieler PCas erklärt werden. In einem zweiten Projektansatz wurde die Bedeutung und Funktion von miR-205 als möglicher Tumorsuppressor im PCa untersucht. Dabei kann gezeigt werden, dass mir-205 ebenfalls in der Lage ist, nach transienter Überexpression die Proliferation von PCa-Zellen zu inhibieren. Um molekulare Mechanismen und Wirkweisen von mir-205 zu untersuchen, wurden im Zellmodell die Expression der Onkogene HER2/neu und HER3, beides vorausgesagte miR-205-Zielgene, analysiert. Es konnte gezeigt werden, dass in PCa-Zellen die Unterexpression von HER3 und HER/neu mit der ektopischen Überexpression von miR-205 assoziiert ist. Die HER2/neu-Expression konnte zusätzlich auch im primären Tumor mit der miR-205-Expression invers assoziiert werden, wodurch die Regulation dieses Zielgens auf nativer Ebene verifiziert wurde. Ein weiterer Fokus dieser Arbeit konzentrierte sich auf die Fragestellung, ob sich miR-205 als Prognosemarker im high-risk-PCa eignen könnte. Hierzu wurden in einem etablierten high-risk-PCa-Studienkollektiv die Expression von mir-205 analysiert und anschließend Korrelations- und Überlebensanalysen durchgeführt. Es konnte statistisch keine Assoziation zwischen der miR-205-Expression und verschiedenen Prognoseparametern, die in der Klinik präoperativ prognostische Vorraussagekraft besitzen, hergestellt werden. Allerdings fiel auf, dass Karzinome, die miR-205 relativ schwach herabregulierten, ein signifikant schlechteres prognostisches Outcome und Überlebensnachteile zeigten, im Vergleich zu Tumoren, die eine starke miR-205-Regulation aufweisen. Somit konnten im Hochrisikokollektiv mit Hilfe der miR-205-Expressionsanalyse Karzinome identifiziert werden, die ein erhöhtes Rezidivrisiko aufweisen. Die hier vorgelegten Untersuchungen stellen also erste Hinweise dar, dass miR-205 als unabhängiger prognostischer Marker im PCa Verwendung finden könnte. Zusammenfassend kann in der vorgelegten Arbeit gezeigt werden, dass die microRNAs miR-205 und miR-221 zwei tumorsuppressive RNAs im PCa darstellen. Eine mögliche zukünftige Implementation der Expressionsanalysen von miR-221 und/oder miR-205 als Progressionsmarker stellt eine vielversprechende Möglichkeit dar, in Zukunft die Prognose und vielleicht auch die Therapie des PCa zu verbessern.

The participating nations articulated a set of guiding principles in the form of rules, rights and responsibilities endorsed by all the early member-nations on June 26, 1945, but effective from October 24, same year. This is the birth of the United Nations Charter.
With the endorsement of the Charter, all member-nations assumed the responsibility of making the world a better place, peaceful and secure for humanity. They vowed never again to engage in unethical wars, they accepted to respect and foster human rights, to fight poverty, to spread democracy and to promote more healthy and robust international relations through a more vibrant cooperation and aggressive diplomacy. The Charter also reaffirmed the intrinsic right of self-defense of the victim of an armed attack, which sometimes has been utilized as well as exploited.
Die Magnetresonanztomographie stellt den Goldstandard zur kardialen Funktionsdiagnostik dar und ermöglicht die nicht-invasive Analyse der Herzfunktion mit valider Bestimmung von Volumina, Flüssen sowie der Ejektionsfraktion in vivo. In unserer Arbeitsgruppe erfolgt eine stetige Weiterentwicklung der Methode am Rattenmodell, wobei regelhaft eine Narkose des Versuchtstiers notwendig ist. Im Rahmen meiner Arbeit wurde der Effekt verschiedener Narkoseformen auf die Herzfunktion untersucht. Dabei wurde eine Isoflurannarkose einer Narkose mittels Propofol sowie Propofol in Kombination mit Pancuronium gegenübergestellt. Hierbei zeigen sich teilweise deutliche Unterschiede in den kardialen Funktionsparametern während der Untersuchung. Hieraus ist zu folgern, dass ein sinnvoller Vergleich der Herzfunktion von Versuchsreihen mit unterschiedlicher Narkosetechnik problematisch ist. Dies unterstreicht die Wichtigkeit einer Festlegung der Narkosetechnik vor Beginn einer Versuchsreihe in der kardiovaskulären Forschung und deren Konstanthaltung über die gesamte Versuchsdauer.
In der vorliegenden Arbeit sollten die unterschiedlichen Manifestationsformen der Borreliose anhand verschiedener Parameter verglichen und der Verlauf einer Borrelioseerkrankung analysiert werden.
Dabei konnte gezeigt werden, dass es sich um eine ganzjährige Erkrankung handelt, die jedoch - je nach Manifestationsform - verschiedene jahreszeitliche Gipfel aufweist. So kommen die Neuroborreliose und die Frühborreliose gehäuft im Frühling und im Sommer vor, während die Lyme Arthritis durchweg ganzjährig auftritt.
Da die Borrelien erst nach ca. 12 Stunden Latenz auf den Wirt übertragen werden, sind es vor allem die unbemerkten Zeckenstiche, die eine Infektion verursachen. Deshalb ist es nicht verwunderlich, dass sich nur rund 55 % der Patienten an einen zurück liegenden Zeckenstich erinnern konnten.
Bei der Untersuchung der verschiedenen Laborparameter ergaben sich keine wesentlichen Unterschiede zwischen den Manifestationsformen bezüglich Hämoglobingehalt, Blutsenkungsgeschwindigkeit, Thrombozyten und Leukozyten im Serum. Desweiteren ergaben sich keine Hinweise auf eine Beteiligung des Rheumafaktors oder antinukleärer Antikörper an den immunologischen Prozessen während einer Lyme Arthritis Erkrankung.
IL-17 konnte mittels ELISA weder in Serum/Plasma noch in Synovialflüssigkeit signifikant erhöht nachgewiesen werden. Es bleibt jedoch unklar, ob dies aufgrund einer langen Lagerung der Proben der Fall war oder ob IL-17 tatsächlich nicht signifikant erhöht vorlag. Aus der Literatur kann man entnehmen, dass IL-17 bei den Abläufen einer Infektion mit B. burgdorferi eine wichtige Rolle zu spielen scheint. Dies könnte einen Ansatz für neue Behandlungsmethoden der Lyme Arthritis darstellen, weshalb die Durchführung weitere Untersuchungen wichtig ist.
Durch die Analyse der initialen Serologie konnte die Antikörperreaktion während einer Infektion mit B. burgdorferi veranschaulicht werden, die mit der Bildung von IgM- Antikörpern beginnt und dann einen Shift zu IgG-Antikörpern vollzieht. Die Antigene p19, p31/34, p39 und p65 scheinen hierbei vor allem bei der Lyme Arthritis im Vordergrund zu stehen. Weiterhin konnte bei dieser Analyse beobachtet werden, dass teilweise zahlreiche serologische Untersuchungen bei ein und demselbem Patienten statt fanden, was darauf schließen lässt, dass diese Untersuchung zur Verlaufskontrolle benutzt wurde. Grundsätzlich ist die Serologie hierzu jedoch nicht geeignet. Vielmehr sollte sie lediglich zur Diagnosestellung dienen. In dieser Funktion ist sie bei korrekter Interpretation ein hervorragendenes diagnostisches Mittel.
Leider werden serologische Untersuchungen jedoch häufig falsch interpretiert und angewendet, was dazu führt, dass Patienten häufiger oder auch länger als nötig antibiotisch behandelt werden. So erhielten auch rund 8 % der Lyme Arthritis Patienten mehr als 2 Zyklen antibiotischer Therapie. Die Nebenwirkungen dieser langwierigen antibiotischen Behandlung sind nicht zu vernachlässigen. Die aktuellen Leitlinien sehen im Gegensatz dazu nämlich nach dem zweiten antibiotischen Zyklus den Beginn einer Therapie mit DMARD ́s vor, selten kommen auch intraartikuläre Steroide zum Einsatz. Diese beobachtete „Überdiagnostik und Übertherapie“ der Borrelioseerkrankung spiegelt vermutlich die Angst vor chronischen Verlaufsformen in der Bevölkerung und teilweise auch unter Ärzten wider.
Bereits in anderen Studien konnte die Existenz einer therapieresistenten Verlaufsform der Borreliose - insbesondere der Lyme Arthritis – nachgewiesen werden. In der vorliegenden Arbeit litten 24 % der Lyme Arthritis Patienten nach einer adäquaten antibiotischen Behandlung laut eigenen Angaben zum Zeitpunkt der Umfrage noch unter Gelenkbeschwerden. Ob es sich hierbei nun um eine schwerwiegendere Verlaufsform in Europa handelt, bleibt zunächst offen. Zur Objektivierung der Beschwerden, zum Ausschluss einer Zweitinfektion oder aber auch einer Fibromyalgie als mögliche Differentialdiagnose sind prospektive Studien mit klinischer Untersuchung der Patienten im Verlauf nötig.
Eindeutig ist jedoch, dass durch eine frühzeitige antibiotische Behandlung nach Infektion das Fortschreiten der Erkrankung effektiv verhindert werden kann. Chronische Beschwerden gaben hingegen auch die Patienten mit zurückliegender Neuroborreliose an. Rund 20% der Patienten litten nach eigener Angabe unter Allgemeinsymptomen, die ihre Lebensqualität subjektiv wesentlich einschränkten. Der spezifische Zusammenhang zwischen diesen Beschwerden und einer Neuroborreliose bzw. einer Borrelioseerkrankung im Allgemeinen ließ sich jedoch nicht endgültig klären. Es wäre hierfür der Vergleich mit einer gesunden Kohorte und die Anwendung spezieller neuropsychologischer Untersuchungen nötig.
In summary, structure-activity relationships in peptide and dendrimer carriers modified with different organometal complexes were studied on a human breast cancer cell line. Variation of the organometal cargo and carrier can significantly influence their biological properties and might open the way to new approaches in chemotherapy. Furthermore, the incorporation of complexes with different C≡O vibrational signatures in a model peptide was explored to examine information encoding in biomolecules in a barcoding strategy for potential imaging applications. In particular for the latter, additional stable metal-carbonyl markers need to be prepared in future work to expand the pool of vibrational labels available.
Recent development of proteomic approaches and generation of large-scale proteomic datasets calls for new methods for biological interpretation of the obtained results. Systems biological approaches such as integrated network analysis and functional module search have become an essential part of proteomic investigation. Proteomics is especially applied in anucleate cells such as platelets. The underlying molecular mechanisms of platelet activation and their pharmacological modulation are of immense importance for clinical research. Advances in platelet proteomics have provided a large amount of proteomic data, which has not yet been comprehensively investigated in a systems biological perspective. To this end, I assembled platelet specific data from proteomic and transcriptomic studies by detailed manual curation and worked on the generation of a comprehensive human platelet repository for systems biological analysis of platelets in the functional context of integrated networks (PlateletWeb) (http:/PlateletWeb.bioapps.biozentrum.uni-wuerzburg.de). I also added platelet-specific experimentally validated phosphorylation data and generated kinase predictions for 80% of the newly identified platelet phosphosites. The combination of drug, disease and pathway information with phosphorylation and interaction data makes this database the first integrative platelet platform available for platelet research. PlateletWeb contains more than 5000 platelet proteins, which can also be analyzed and visualized in a network context, allowing identification of all major signaling modules involved in platelet activation and inhibition. Using the wealth of integrated data I performed a series of platelet-specific analyses regarding the platelet proteome, pathways, drug targets and novel platelet phosphorylation events involved in crucial signaling events. I analyzed the statistical enrichment of known pathways for platelet proteins and identified endocytosis as a highly represented pathway in platelets. Further results revealed that highly connected platelet proteins are more often targeted by drugs. Using integrated network analysis offered by PlateletWeb, I analyzed the crucial activation signaling pathway of adenosine diphosphate (ADP), visualizing how the signal flow from receptors to effectors is maintained. My work on integrin inside-out signaling was also based on the integrated network approach and examined new platelet-specific phosphorylation sites and their regulation using kinase predictions. I generated hypothesis on integrin signaling, by investigating the regulation of Ser269 phosphorylation site on the docking protein 1 (DOK1). This phosphorylation site may influence the inhibiting effect of DOK1 on integrin a2bb3. Extending the integrated network approach to further cell lines, I used the assembled human interactome information for the analysis of functional modules in cellular networks. The investigation was performed with a previously developed module detection algorithm, which finds maximum-scoring subgraphs in transcriptomic datasets by using assigned values to the network nodes. We extended the algorithm to qualitative proteomic datasets and enhanced the module search by adding functional information to the network edges to concentrate the solution onto modules with high functional similarity. I performed a series of analyses to validate its performance in small-sized (virus-infected gastric cells) and medium-sized networks (human lymphocytes). In both cases the algorithm extracted characteristic modules of sample proteins with high functional similarity. The functional module search is especially useful in site-specific phosphoproteomic datasets, where kinase regulation of the detected sites is often sparse or lacking. Therefore, I used the module detection algorithm in quantitative phosphoproteomic datasets. In a platelet phosphorylation dataset, I presented a pipeline for network analysis of detected phosphorylation sites. In a second approach, the functional module detecting algorithm was used on a phosphoproteome network of human embryonic stem cells, in which nodes represented the maximally changing phosphorylation sites in the experiment. Additional kinases from the human phosphoproteome in PlateletWeb were included to the network to investigate the regulation of the signal flow. Results indicated important phosphorylation sites and their upstream kinases and explained changes observed in embryonic stem cells during differentiation. This work presents novel approaches for integrated network analysis in cells and introduces for the first time a systematic biological investigation of the human platelet proteome based on the platelet-specific knowledge base PlateletWeb. The extended methods for optimized functional module detection offer an invaluable tool for exploring proteomic datasets and covering gaps in complex large-scale data analysis. By combining exact module detection approaches with functional information data between interacting proteins, characteristic functional modules with high functional resemblance can be extracted from complex datasets, thereby focusing on important changes in the observed networks.
Die zur Gruppe der mikrobiellen Rhodopsine gehörenden lichtaktivierbaren Ionenkanäle Channelrhodopsin 1 (ChR1) und Channelrhodopsin 2 (ChR2) aus dem Augenfleck von C. rheinhardtii sind Bestandteile des visuellen Systems und an der Phototaxis beteiligt. Sie bestehen aus einem zytosolisch gelegenen C Terminus, dessen Funktion noch ungeklärt ist und einem, für die Kanalaktivität verantwortlichen, N terminalen Bereich aus sieben Transmembranhelices. Der lichtsensitive Kofaktor all trans Retinal ist kovalent an einen Lysinrest (K257) der siebten Transmembranhelix gebunden. Bei einer Belichtung mit Blaulicht isomerisiert das Chromophor zur 13 cis Form, was eine Konformationsänderung und das Öffnen des Kanals zur Folge hat. Im Zuge dessen strömen ein und zweiwertige Kationen in die Zelle und eine Depolarisation findet statt. Um einen tieferen Einblick in Struktur und funktionelle Mechanismen zu bekommen, wurden Wildtyp und Mutanten von Ch1 und ChR2 heterolog in Oozyten von X. laevis exprimiert. In Bakteriorhodopsin bilden die Seitenketten von T90 und D115 eine für Stabilität und Funktion wichtige Wasserstoffbrücke aus. Durch elektrophysiologische, fluoreszenzmikroskopische und biochemische Verfahren wurden Mutanten der entsprechenden Reste in ChR2 (C128, D156) untersucht. Diese zeigten eine deutlich verlangsamte Kinetik und eine 10 bis 100fache Erhöhung der Lichtempfindlichkeit. Die identischen Auswirkungen von Mutationen beider Reste deuten auf eine Bindung mit funktioneller Bedeutung zwischen C128 und D156 hin. Im Falle von ChR2 C128T, C128A und D156A konnte der Kanal nach Anregung mit Blaulicht, durch grünes und violettes Licht vorzeitig geschlossen werden. Diese Lichtqualitäten entsprechen den Absorptionswellenlängen zweier Intermediate des Photozyklus von ChR2 (P390 und P520). Durch Veränderung des externen pH-Wertes konnten Hinweise auf eine protonenabhängige Gleichgewichtsreaktion dieser Intermediate gefunden werden. Auch in dem für Protonen höher leitfähigen ChR1 konnten Hinweise auf eine Interaktion zwischen den Resten C167 und D195 gefunden werden. Elektrische Messungen von Mutanten zeigten eine deutliche Erhöhung des Photostroms bei verhältnismäßig geringem Anstieg der Schließzeit. Der Einfluss dieser Mutationen auf die Kinetik war somit weniger ausgeprägt als bei ChR2. Einen besonderen Stellenwert unter allen Channelrhodopsin Mutanten nehmen ChR2 D156C und ChR1 D195C ein. Mit einem Photostrom von 5 µA bei ChR1 D195C und bis zu 50 µA bei ChR2 D156C konnten für diese die höchsten Photoströme aller bisher charakterisierten ChR1 bzw. ChR2 Varianten nachgewiesen werden. Durch fluoreszenzmikroskopische Quantifizierung konnte für alle im Rahmen dieser Arbeit erstellten ChR1 und ChR2 Mutanten eine erhöhte Proteinmenge sowohl in Anwesenheit als auch Abwesenheit zusätzlichen all trans Retinals während der Inkubation nachgewiesen werden. Die Fluoreszenzintensitäten korrelierten hierbei mit der Höhe der Stromamplituden und erreichten ein Maximum bei ChR2 D156C. Biochemische Experimente mit der Gesamtmembranfraktion von ChR2 exprimierenden Oozyten lieferten Hinweise auf eine dimere Quartärstruktur von Channelrhodopsinen, was durch die Kristallstruktur einer Chimäre aus ChR1 und ChR2 von (Kato et al., 2012) bestätigt wurde. Unter der Annahme, dass die Poren in den Proteomeren gebildet werden, konnte eine gegenseitige Beeinflussung der Regionen in heterodimeren Kanälen aus ChR2 Wildtyp und Mutanten aufgrund kinetischer Unterschiede bei kurzer und langer Belichtung oder der Verwendung von unterschiedlichen Lichtintensitäten nachgewiesen werden. Eine Voraussetzung für diesen Effekt ist eine synchrone Anregung beider Untereinheiten. Die Interaktion von Channelrhodopsin Untereinheiten konnte in vivo mithilfe der bimolekularen Fluoreszenzkomplementation nachgewiesen werden. Dabei zeigte sich, dass die Wechselwirkung nicht nur auf identische Untereinheiten in Homodimeren beschränkt ist, sondern auch bei Heterodimeren aus verschiedenen ChR2 Untereinheiten und sogar zwischen ChR2 und ChR1 möglich ist.

Cancer remains the second leading cause of death in the industrialized. The data from many different studies investigating the nature of cancer-initiating cells coined the description ‘cancer stem cells’ and has major implications on conventional cancer therapy. Thus, to improve the outcome of cancer treatment and to lower negative side effects, the development of novel therapeutic regimens is indispensable. It has been demonstrated in many preclinical studies that oncolytic virotherapy using vaccinia virus may provide a powerful and well-tolerable new tool in cancer therapy which is currently investigated in several clinical trials (Phase I & II) as stand-alone treatment or in combination with conventional cancer therapy. Cancer-initiating cells and stem cells share a variety of characteristics like the ability to self-renew, differentiation potential, quiescence, drug and radiation resistance, activation and inhibition of similar signaling pathways as well as expression of cell surface markers and stem cell-related genes. In this work, two new recombinant vaccinia viruses expressing the transcription factors Nanog (GLV-1h205) and Oct4 (GLV-1h208) were engineered to provide deeper insight of these stem cell master regulators in their significance of cancer-initiation and their impact on oncolytic virotherapy. Both viruses were analyzed for their replication potential in A549 and PC-3 human cancer cells. Marker gene expression was assessed by RT-PCR, SDS-PAGE and Western blotting, ELISA or immunocytochemistry.Furthermore, the effect of GLV-1h205 infection on the cell cycle in A549 cells was analyzed. Next, the effects of virus-mediated expression of stem cell transcription factors on therapeutic efficacy and survival rates in A549 xenograft mouse models was analyzed. A non-functional Nanog mutant-expressing virus strain (GLV-1h321) was engineered to analyze whether the observed therapeutic benefits were promoter- or payload-driven. Furthermore, this study analyzed the potential of GLV-1h68 to infect, replicate in, and lyse colorectal cancer cell lines to study whether oncolytic vaccinia viruses can be potential new and less invasive treatment regimens for late stage colorectal cancer. Marker gene expression was assessed by fluorescence microscopy and FACS. The transcription factor Klf4 is highly expressed in quiescent, terminally differentiated cells in the colonic epithelium whereas it is dramatically downregulated in colon cancers. Klf4 expression leads to cell growth arrest and inhibits Wnt signaling by binding to beta-catenin. To further improve the treatment of colorectal cancers, new recombinant vaccinia viruses (GLV-1h290-292) mediating the expression of differing amounts of the tumor suppressor Klf4 by using different promoter strengths were engineered. Initial characterization of recombinant vaccinia viruses expressing Klf4 by replication assay, cell viability assay, SDS-PAGE and Western blotting, immuncytochemistry and analysis of protein functionality by qPCR and ELISA analysis for cellular beta-catenin expression, demonstrated promoter strength-dependent expression of and impact of Klf4. To further boost the effects of tumor suppressor Klf4, a vaccinia virus strain expressing Klf4 with a C-terminal fusion of the TAT transduction domain (GLV-1h391) was engineered. Treatment of HT-29 non-responder tumors in vivo with GLV-1h291 and GLV-1h391 led to significant tumor growth inhibition and improved overall survival compared to GLV-1h68. This makes the Klf4-TAT expressing GLV-1h391 a promising candidate for the treatment of colorectal cancer in man.
Spielregeln für das Überleben: Dietrich von Bern im "Nibelungenlied" und in der "Nibelungenklage"

Anknüpfend an Jan Dirk Müllers Studie ‘Spielregeln für den Untergang’ fragt der Beitrag nach jenen Verhaltensweisen im ‘Nibelungenlied’, mit denen die Katastrophe hätte verhindert werden können. Im Zentrum des Beitrags steht Dietrich von Bern, der wiederholt Handlungsalternativen aufzeigt, die eine andere Entwicklung des Geschehens ermöglicht hätten. Seine Spielregeln für ein Überleben funktionieren nur deshalb nicht, weil der Herr der Amelungen in der nibelungischen Welt keine Mitspieler findet, die sich dauerhaft an seine Regeln halten. Nach der Katastrophe werden seine Maximen jedoch bestätigt und anerkannt, wie die zeitgenössischen Reflexionen im Umfeld des Epos zeigen. In der ʻNibelungenklageʼ wird nicht nur rückblickend das Fehlverhalten einzelner Figuren getadelt, sondern ermöglichen die Anweisungen Dietrichs auch ein Weiterleben und die Gestaltung künftigen Geschehens. Um diese These zu stützen, werden zunächst die für Dietrich relevanten Episoden des ‘Nibelungenlieds’ in chronologischer Folge untersucht und die jeweiligen Handlungsmaximen bestimmt, anschließend die Bedeutung und Bewertung seiner Spielregeln in der ‘Klage’ untersucht. Flankierend werden auch die Werke der historischen Dietrichepik herangezogen.
2,3-Dipalmitoyl-S-Glycerylcystein (Pam2Cys), ein Toll-like-Rezeptor 2 (TLR2)-Ligand, aktiviert dendritische Zellen und erhöht die Expression von HLA-DR auf deren Zelloberfläche. Natürliche Killerzellen (NK-Zellen) können über die Ausschüttung von Zytokinen das erworbene Immunsystem stimulieren. Zudem weist die Entdeckung von HLA-DR auf NK-Zellen, besonders auf CD56bright-NK-Zellen, auf eine Verbindung zwischen angeborenem und erworbenem Immunsystem hin. Gekoppelt an ein immundominantes Peptidepitop aus dem Glykoprotein D des Herpes-Simplex-Virus (HSV) Typ 2, wurde Pam2Cys in dieser Arbeit als potentieller lipopeptidbasierter HSV-Impfstoff eingesetzt, um die Effekte dieser Vakzine auf NK-Zellen zu untersuchen. Die Einflüsse der Vakzine wurden anhand der CD69-, HLA-DR- und TLR2-Expression auf und in NK-Zellen nach Inkubation mit Pam2Cys untersucht. Die Analyse der Gesamt-NK-Zellpopulation ließ nur mit Hilfe der quantitativen Real-Time-PCR einen signifikanten Anstieg der TLR2-Expression erkennen, jedoch nicht durchflusszytometrisch. Inkubierte NK-Zellen zeigten deutlich mehr HLA-DR auf der Zelloberfläche, als unstimulierte NK-Zellen. Die Untersuchung der zwei großen NK-Zellsubpopulationen, CD56bright und CD56dim, ergab, dass stimulierte CD56bright-Zellen wesentlich mehr HLA-DR und TLR2 aufwiesen, als stimulierte CD56dim-Zellen. Die Expression des Aktivierungsmarkers CD69 stieg in der Gesamtpopulation der NK-Zellen nicht in signifikantem Maße an, zeigte jedoch eine Zunahme nach Inkubation mit den Pam2Cys-Molekülen. Die Aufnahme der Lipopeptide in NK-Zellen wurde mit der konfokalen Mikroskopie bestätigt. Diese Ergebnisse zeigen, dass die hier verwendete HSV-Vakzine von NK-Zellen aufgenommen wird und sie die Ausbildung von Oberflächenrezeptoren auf diesen Zellen moduliert. Insbesondere die Expressionssteigerung von HLA-DR auf CD56bright-NK-Zellen deutet auf die Fähigkeit dieser Zellen hin, Antigene zu präsentieren und stellt einen weiteren Mechanismus vor, der eine impfstoffbezogene Immunantwort unterstützen kann. Die vorliegende Arbeit bildet eine Basis für weiterführende Studien an NK-Zellen hinsichtlich der Antigenpräsentation im Allgemeinen und der Antigenpräsentation nach Stimulation durch eine Vakzine im Besonderen.
Background: Recently, contradictory results on foamy virus protease activity were published. While our own results indicated that protease activity is regulated by the viral RNA, others suggested that the integrase is involved in the regulation of the protease. Results: To solve this discrepancy we performed additional experiments showing that the protease-reverse transcriptase (PR-RT) exhibits protease activity in vitro and in vivo, which is independent of the integrase domain. In contrast, Pol incorporation, and therefore PR activity in the viral context, is dependent on the integrase domain. To further analyse the regulation of the protease, we incorporated Pol in viruses by expressing a GagPol fusion protein, which supported near wild-type like infectivity. A GagPR-RT fusion, lacking the integrase domain, also resulted in wild-type like Gag processing, indicating that the integrase is dispensable for viral Gag maturation. Furthermore, we demonstrate with a trans-complementation assays that the PR in the context of the PR-RT protein supports in trans both, viral maturation and infectivity. Conclusion: We provide evidence that the FV integrase is required for Pol encapsidation and that the FV PR activity is integrase independent. We show that an active PR can be encapsidated in trans as a GagPR-RT fusion protein.
One’s own name constitutes a unique part of conscious awareness – but does this also hold true for unconscious processing? The present study shows that the own name has the power to bias a person’s actions unconsciously even in conditions that render any other name ineffective. Participants judged whether a letter string on the screen was a name or a non-word while this target stimulus was preceded by a masked prime stimulus. Crucially, the participant’s own name was among these prime stimuli and facilitated reactions to following name targets whereas the name of another, yoked participant did not. Signal detection results confirmed that participants were not aware of any of the prime stimuli, including their own name. These results extend traditional findings on ‘‘breakthrough’’ phenomena of personally relevant stimuli to the domain of unconscious processing. Thus, the brain seems to possess adroit mechanisms to identify and process such stimuli even in the absence of conscious awareness.
The Influence of Anticipation and Warnings on Collision Avoidance Behavior of Attentive Drivers

This thesis deals with collision avoidance. Focus is on the question of under which conditions collision avoidance works well for humans and if drivers can be supported by a Forward Collision Warning (FCW) System when they do not react appropriately. Forward Collision Warning systems work in a way that tries to focus the driver's attention in the direction of the hazard and evoke an avoidance reaction by some sort of alert (e.g., tone or light). Research on these warning systems generally focuses on inattention and distraction as the cause for crashes. If the driver is inattentive, the results of a crash are thought to be worse as the driver‘s reaction is belated or might not mitigate the crash at all. To ensure effectiveness in the worst case, most of the experiments studying FCW systems have been conducted with visually distracted drivers. Research on the cause and possible countermeasures for crashes of attentive drivers are hardly available, although crash databases and field operational test data show that 40-60% of the drivers look at the forward scene shortly before they crash. Hence, only a few studies elaborated on ideas about the reasons for crashes with attentive drivers. On the basis of the literature, it is worked out that one reason for delayed avoidance behavior can be an incorrect allocation of attention. It is further elaborated that high level attention processes are strongly influenced by interpretation of the situation and the anticipation of future status. Therefore, it is hypothesized that alert drivers react later when they can not foresee a potential threat or even when they misinterpret the situation. If the lack of threat anticipation or incorrect anticipation is a reason for crashes, a FCW system could be a great help, when the FCW is easily comprehensible. It is hypothesized that a FCW can compensate for missing threat anticipation in the driver. The results of the experiments show that the level of threat anticipation has the largest influence on driver behavior in an imminent crash situation. The results further suggest that FCW systems - especially warnings of audible or haptic modality - can help attentive drivers who do not anticipate a threat or misinterpret a situation. The negative influence of missing or mislead threat anticipation on objective measures was small when the threat appeared suddenly. This is thought to be due to the visual appearance of the introduced threat. It is assumed that this type of stimulus triggers a lower level attentional process, as opposed to a top-down attention process controlled by an anticipatory process. In the other scenario types such a lower level process may not be triggered. An important result of the second study is that (Forward) Collision Warnings have to be learned. Participants with warnings reacted slower than participants without any FCW in the first critical event. Participants with a visual warning reacted particularly slow. Later in the experiment, the probands with warnings were constantly faster than their counterparts without them. Hence, the results of this study suggest that a haptic or audible modality should be used as a primary warning to the driver. The characteristic of visual warnings to draw the visual attention is both a blessing and a curse. It is suggested to use the visual warning component for only a short period of time to attract the driver's attention to the forward scene, but then end the display to not further distract him. Car manufacturers try to avoid as many unnecessary alarms as possible. If driver monitoring would be available, it is often planned to suppress warnings when the driver is looking through the windshield. The results suggest not to do so. If a driver reaches a critical situation represented by a low Time-to-collision (TTC) or a high need to decelerate, he should always get a warning, unless he is already braking or steering. The most important arguments for this are: - Looking at the street does not mean that the driver has the correct situational awareness. - The driver has to learn the meaning of the warning. - The driver will not be annoyed by a warning when the situation is considered critical.
Eisenproduktionswerkplätze der späten römischen Kaiserzeit (3.–5. Jh. AD) im inneren Barbaricum

Durch systematische Prospektionen in Südbrandenburg wurden auch bei den devastierten Ortschaften Klein Görigk und Kausche zahlreiche bisher unbekannte Fundplatze entdeckt (vgl. Abb. 1). Diese verdeutlichen den Fundreichtum dieser kargen Landschaft als „archäologisches Fenster“ einer fallbeispielhaft intensiv erforschten Region. Die sehr zahlreichen Werkplätze der späten römischen Kaiserzeit (3.–5. Jh. AD) belegen eine massenhafte Eisenproduktion, die über den Eigenbedarf weit hinausging und die Grundlage für Handel darstellte. Interessanterweise sind im Eisenverhüttungszentrum des Niederlausitzer Grenzwalls keine zeitgleichen Siedlungen und Gräberfelder entdeckt worden. Diese liegen etwas weiter entfernt in den fruchtbareren Niederungs- und Beckenlandschaften der Umgebung. Die Werkplätze sind also nur temporär zur Eisenverhüttung aufgesucht worden. Die stereotyp errichteten Eisenproduktionsstätten wurden in unmittelbarer Nähe zum lokal vorkommenden „Raseneisenerz“ im waldreichen Gebiet errichtet. Durch die massenhafte Eisenproduktion, die äußerst viel Holzkohle benötigte, ist auch von negativen Folgen auf die prähistorische Umwelt auszugehen. Indizien einer mutmaßlichen „ökologischen Krise“ zum Ende der spätgermanischen Kultur (Mitte 5. Jh. AD) konnten jedoch bisher nicht sicher belegt werden.
Die Dissertation beschäftigt sich mit spätbronzezeitlichen (ca. 1300 - 800 v. Chr.) Depotfunden aus Bayern, Baden-Württemberg und Westböhmen, die auf festem Grund niedergelegt wurden. Die aus diesen Depots stammenden Bronzen wurden gezielt hinsichtlich ihrer Verwendung und Fragmentierung untersucht. Zum einen, um neue Erkenntnisse zum Deponierungsverhalten in Süddeutschland zu gewinnen; zum anderen sollten sich dabei Argumente ergeben, die in die andauernde Diskussion um die Deutung der Deponierungen, zu der vor allem die Erschließung der Niederlegungsabsicht gehört, eingebracht werden können. Von besonderem Interesse war die Analyse und Dokumentation der aus den sogenannten Brucherzdepots stammenden Objekte, die innerhalb des spätbronzezeitlichen Deponierungsphänomens neben den Fertigwarendepots als eigene Gruppe herausgestellt werden können und deren Deutung am heftigsten diskutiert wird.
