Gold nanoparticles of diameter ca. 60 nm have been synthesized based on Turkevich and Frens protocols. We have demonstrated that the carboxyl-modified gold nanoparticles can be coupled covalently with antibodies (Ab) of interest using the EDC/NHS coupling procedure. Binding studies with Ab-grafted AuNPs and GpL fusion proteins proved that conjugation of AuNPs with antibodies enables immobilization of antibodies with preservation of a significant antigen binding capacity. More importantly, our findings showed that the conjugation of types of anti-TNF receptors antibodies such as anti-Fn14 antibodies (PDL192 and 5B6) (Aido et al., 2021), anti-CD40, anti-4-1BB and anti-TNFR2 with gold nanoparticles confers them with potent agonism. Thus, our results suggest that AuNPs can be utilized as a platform to immobilize anti-TNFR antibodies which, on the one hand, helps to enhance their agonistic activity in comparison to “free” inactive antibodies by mimicking the effect of cell-anchored antibodies or membrane-bound TNF ligands and, on the other hand, allows to develop new generations of drug delivery systems. These constructs are characterized with their biocompatibility and their tunable synthesis process.
In a further work part, we combined the benefits of the established system of Ab-AuNPs with materials used widely in the modern biofabrication approaches such as the photo-crosslinked hydrogels, methacrylate-modified gelatin (GelMA), combined with embedded variants of human cell lines. The acquired results demonstrated clearly that the attaching of proteins like antibodies to gold nanoparticles might reduce their release rate from the crosslinked hydrogels upon the very low diffusion of gold nanoparticles from the solid constructs to the surrounding medium yielding long-term local functioning proteins-attached particles. Moreover, our finding suggests that hydrogel-embedded AuNP-immobilized antibodies, e.g. anti-TNFα-AuNPs or anti-IL1-AuNPs enable local inhibitory functions,
To sum up, our results demonstrate that AuNPs can act as a platform to attach anti-TNFR antibodies to enhance their agonistic activity by resembling the output of cell-anchoring or membrane bounding. Gold nanoparticles are considered, thus, as promising tool to develop the next generation of drug delivery systems, which may contribute to cancer therapy. On top of that, the embedding of anti-inflammatory-AuNPs in the biofabricated hydrogel presents new innovative strategy of the treatment of autoinflammatory diseases.
Titanium Dioxide Nanoparticles: Synthesis, X-Ray Line Analysis and Chemical Composition Study

TiO2 nanoparticleshave been synthesized by the sol-gel method using titanium alkoxide and isopropanolas a precursor. The structural properties and chemical composition of the TiO2 nanoparticles were studied usingX-ray diffraction, scanning electron microscopy, and X-ray photoelectron spectroscopy.The X-ray powder diffraction pattern confirms that the particles are mainly composed of the anatase phase with the preferential orientation along [101] direction. The physical parameters such as strain, stress and energy density were investigated from the Williamson- Hall (W-H) plot assuming a uniform deformation model (UDM), and uniform deformation energy density model (UDEDM). The W-H analysis shows an anisotropic nature of the strain in nanopowders. The scanning electron microscopy image shows clear TiO2 nanoparticles with particle sizes varying from 60 to 80nm. The results of mean particle size of TiO2 nanoparticles show an inter correlation with the W-H analysis and SEM results. Our X-ray photoelectron spectroscopy spectra show that nearly a complete amount of titanium has reacted to TiO2
Use of polyhexanide and nanomedicine approach for effective treatments of cutaneous leishmaniasis

Despite huge suffering caused by cutaneous leishmaniasis (CL), there is no effective and affordable treatment strategy against CL and no licensed vaccines. The current treatments show limited efficacy and high toxicity. Improved therapies through discovery of novel drugs and/or an alternative treatment approaches are/is urgently needed. We aimed at identifying a novel antileishmanial agent and developing an innovative nanoparticle (NP) based platform for safe and effective treatments against CL. We discovered that polyhexanide (PHMB), a widely used antimicrobial polymer and wound antisepsis, shows an inherent antileishmanial activity at submicromolar concentrations. PHMB appears to kill L. major parasites via a dual mechanism involving disruption of membrane integrity and selective chromosome condensation. However, host chromosomes binding appear to be limited by exclusion from mammalian cell nuclei. Moreover, we attempted to establish effective drug delivery systems that overcome the various shortcomings in the present treatment of CL. In this scenario, we initially studied the cellular interactions of NPs and their uptake mechanisms into mammalian cells before applying them in drug delivery system. We obtained clear evidence for the involvement of multiple endocytic routes to internalize NPs. Physicochemical properties of NPs, cell type, temperature and pathogenesis of the target diseases were shown to be determinant factors. Thereafter, a mechanism based host- and pathogen-directed combination therapy comprising PHMB and CpG ODN immunomodulator was established for overall synergistic effect against CL. It simultaneously targets the pathogen and the host immunity with effective delivery system. The results show that PHMB binds to CpG ODN and form stable nanopolyplexes for efficient cell entry and therapy. The nanopolyplexes displayed enhanced cellular uptake and antileishmanial potency while drastically reducing the toxicity against mammalian cells. In conclusion, our findings clearly indicate that PHMB can be used as effective candidate drug against CL and as non-viral delivery of immunomodulatorynucleic acids. Moreover, our proof-of concept study showed nanomedicine approaches are effective strategy to challenge CL and other human diseases.
Die Arbeit hatte das Ziel, Beiträge zur Synthese photorefraktiver Nanokomposite durch die geschickte Kombination spezieller organischer Farbstoffe mit elektronenleitenden, anorganischen Nanopartikeln oder mit lochleitenden Carbazol-Derivaten in hybriden Polymeren zu leisten. The green synthesis of silver nanoparticles (SNPs) using plant extracts is an eco-friendly method. It is a single step and offers several advantages such as time reducing, cost-effective and environmental non-toxic. Silver nanoparticles are a type of Noble metal nanoparticles and it has tremendous applications in the field of diagnostics, therapeutics, antimicrobial activity, anticancer and neurodegenerative diseases. In the present work, the aqueous extracts of aerial parts of Lampranthus coccineus and Malephora lutea F. Aizoaceae were successfully used for the synthesis of silver nanoparticles. The formation of silver nanoparticles was early detected by a color change from pale yellow to reddish-brown color and was further confirmed by transmission electron microscope (TEM), UV–visible spectroscopy, Fourier transform infrared (FTIR) spectroscopy, dynamic light scattering (DLS), X-ray diffraction (XRD), and energy-dispersive X-ray diffraction (EDX). The TEM analysis of showed spherical nanoparticles with a mean size between 12.86 nm and 28.19 nm and the UV- visible spectroscopy showed λ\(_{max}\) of 417 nm, which confirms the presence of nanoparticles. The neuroprotective potential of SNPs was evaluated by assessing the antioxidant and cholinesterase inhibitory activity. Metabolomic profiling was performed on methanolic extracts of L. coccineus and M. lutea and resulted in the identification of 12 compounds, then docking was performed to investigate the possible interaction between the identified compounds and human acetylcholinesterase, butyrylcholinesterase, and glutathione transferase receptor, which are associated with the progress of Alzheimer’s disease. Overall our SNPs highlighted its promising potential in terms of anticholinesterase and antioxidant activity as plant-based anti-Alzheimer drug and against oxidative stress.
Ziel dieser Arbeit war die Herstellung fluoreszent markierter Präpolymere sowie deren Optimierung, die kontrollierte und reproduzierbare Synthese von redox-sensitiven und nicht redox-sensitiven NG mit und ohne Fluoreszenzmarkierung in einem durchschnittlichen Partikelgrößenbereich von 150 – 300 nm und mit einer Konzentration > 10*10 Partikel/ml, die Charakterisierung der NG, ihre Untersuchung bezüglich ihrer Stabilität und des Assoziationsverhaltens zu BSA sowie die Erlangung von Erkenntnissen bezüglich des Aufnahmemechanismus der NG in Abhängigkeit vom Transportpeptid Tat.
Abschließend kann zusammenfassend gesagt werden:
1. Das große Potential von PG-basierten NG für biologische bzw. medizinische Einsatzgebiete konnte weiter untermauert werden.
2. Das mit Cy5-Alkin markierte PG PG-SH-Cy5 erscheint aufgrund des relativ hohen erreichten Markierungsgrades bei der Herstellung als aussichtsreichster Kandidat für weitere Untersuchungen. Diese Umsetzung besitzt noch Optimierungspotentiale bezüglich einer Verringerung des Polymerverlusts bei der Aufarbeitung, des erreichbaren Markierungsgrades und der Markierungsausbeute. Möglichkeiten, dies zu erreichen, wurden diskutiert.
3. Klare Aussagen über den Einfluss des esterhaltigen bzw. esterfreien Ausgangspolymers PG-SH auf die Konzentration und die Partikelgröße konnten aufgrund einer nicht ausreichenden Datenlage nicht getroffen werden.
4. Die esterhaltigen PG-SH-Moleküle erscheinen aufgrund ihrer Labilität gegenüber Hydrolyse für die NP-Synthese weniger geeignet (geringere Stabilität).
5. Die Charakterisierung der aus den markierten und unmarkierten Ausgangspolymeren hergestellten NG, welche teilweise zusätzlich mit dem Transportpeptid Tat funktionalisiert wurden, erfolgte mittels NTA und zeigt für die meisten Spezies relativ schmale, gut definierte, monomodale Größenverteilungen mit einem Maximum um 100-200 nm im Bereich von ca. 40 – max. 400 nm mit Partikelkonzentrationen im Bereich von 1010 - 1011 Partikeln/ml.
6. Insgesamt konnte gezeigt werden, dass der untersuchte, von PG-SH abgeleitete NP-Typ (z. B. NG_3, redox-sensitiv unmarkiert) aufgrund seiner Einheitlichkeit, Partikelgröße und der Reproduzierbarkeit der Herstellung als gut geeignet für den geplanten Einsatz in biologischen Systemen erscheint. Von den weiter derivatisierten NG erscheinen die folgenden aufgrund der oben geschilderten Kriterien als besonders geeignet für den geplanten Einsatz in biologischen Systemen und weiterer Untersuchungen wert: NG680_(TAT)_1-4 (redox-sensitiv, markiert), NGCy5_(TAT)_1 (redox-sensitiv, markiert), NG_MA_2 (nicht redox-sensitiv, unmarkiert), NGCy7_MA_1 (nicht redox-sensitiv, markiert). Aufgrund des relativ hohen erreichbaren Markierungsgrades bei der Markierung der Ausgangspolymere erscheinen die mit Cy5-markierten Verbindungen als besonders vorteilhaft.
7. Die esterfreien, redox-sensitiven NP erwiesen sich bei 14-tägiger Lagerung unter physiologischen Bedingungen als stabil. Ihre Konzentration nahm über 14 Tage um ca. 60 % vom Ausgangswert ab. Gleichzeitig nahm der Teilchendurchmesser während des Beobachtungszeitraums um ca. 25 % zu. Die Abnahme der Teilchenzahl ist - zumindest teilweise - durch eine Vergrößerung des mittleren Teilchendurchmessers und mögliche Adsorptionseffekte an die Gefäßwände des Versuchsaufbaus zu erklären.
8. Die Konzentration der esterfreien, nicht redox-sensitiven NP verringert sich bei 14-tägiger Inkubation unter physiologischen Bedingungen deutlich auf ca. 10 % des Ausgangswerts. Der mittlere Durchmesser der Partikel bleibt innerhalb des Untersuchungszeitraums innerhalb der Fehlergrenzen konstant. Die starke Abnahme der Partikelkonzentration ist wahrscheinlich auf die Hydrolyse des verwendeten esterhaltigen Crosslinkers PEGDA zurückzuführen. Desweiteren sind Adsorptionsphänomene an Oberflächen des Versuchsaufbaus nicht auszuschließen. Insgesamt hervorzuheben ist die wesentlich höhere Stabiliät der redox-sensitiven NP unter den Versuchsbedingungen. Diese Substanzklasse sollte daher weiter verfolgt werden.
9. Es wurde gezeigt, dass sowohl die NG, die das Aufnahmeprotein Tat enthalten, als auch die NG ohne Tat mit Fluoreszenz-markiertem BSA (8,3 µg/ml) wechselwirken und zusammen mit diesem bei der Zentrifugation abgeschieden werden. Über die Art der Wechselwirkung kann keine Aussage getroffen werden.
10. Durch in vitro Zellaufnahmeuntersuchungen an Hela-Zellen konnte gezeigt werden, dass die mit Tat funktionalisierten, redox-sensitiven, Fluoreszenz-markierten NP von den Zellen aufgenommen werden. Die Aufnahme erfolgt über eine deutlich erkennbare Vesikelbildung, die an der Plasmamembran verstärkt beobachtet werden kann. Im Gegensatz hierzu konnte bei den nicht mit Tat funktionalisierten NP keine vergleichbare in vitro Zellaufnahme beobachtet werden.
Die Ergebnisse dieser Arbeit bestätigen insgesamt das große Potential der von Thiol-funktionalisierten PG abgeleiteten NG für die medizinische Forschung und zukünftige Anwendungen in der Diagnostik und Therapie. Es wird eine Reihe von Ansatzpunkten aufgezeigt, auf deren Basis weitere vertiefende Untersuchungen zur Charakterisierung und Optimierung sowie zu zukünftigen nutzbringenden Anwendungen vorgenommen werden sollten.
