Die akute Graft-versus-Host Erkrankung (GvHD), insbesondere die Darm GvHD, stellt weiterhin eine der Hauptursachen für Mortalität und Morbidität nach allogener SZT dar. Aktivierte, alloreaktive Spender T-Zellen infiltrieren dabei über die Blutbahn die intestinale Lamina Propria. Erst kürzlich konnten wir zeigen, dass neben der vaskulären Migration ein Teil der Spender T-Zellen auch direkt aus den PP in die angrenzende Lamina Propria migrieren. Um Faktoren, die diese direkte Migration fördern, zu untersuchen und die direkt migrierenden T-Zellen genauer zu charakterisieren, verwendeten wir ein MHC-inkompatibles Mausmodell zur Induktion einer akuten GvHD.
Durch RNA Sequenzierung und Massenspektrometrie lasermikrodissezierter Darmschleimhautproben konnte eine starke Expression der Chemokine CXCL9, CXCL10, CXCL11, CCL3, CCL4 und CCL5 während der akuten intestinalen GvHD aufgezeigt werden. Neben CCL4 und XCL1 wiesen verschiedene Faktoren der T-Zellaktivierung, wie CD3ζ, LAT, Lck und ZAP70, sowie Faktoren der zytoskelettalen Reorganisation, wie Dock2, Coro1α und Parvin-γ, eine vermehrte Expression insbesondere nahe der PP auf. Die Expression der migrationsfördernden Faktoren Coro1α und Parvin-γ in Spender T-Zellen nahe der PP konnte anschließend mittels histologischen Immunfluoreszenzfärbungen bestätigt werden. Durchflusszytometrische Analysen konnten weiterhin eine vermehrte Expression von CCR5, CCR9 und Intgerin α4β7 auf den vornehmlich Tbet+ Spender T-Zellen nahe der PP nachweisen. Funktionelle in vitro Migrationsversuche zeigten abschließend, dass in vivo aktivierte Spender T-Zellen eine gerichtete Migration in Richtung auf CXCL11 und zu späterem Zeitpunkt auch auf CCL4 vollziehen können.
Zusammenfassend zeigt diese Arbeit die Bedeutung zahlreicher Chemokine für das sequenzielle T-Zell-Homing während der akuten intestinalen GvHD. Neben der insbesondere durch Faktoren der zytosekeletalen Reorganisation vermittelten amoeboiden Migration kann auch eine mesenchymale Fortbewegung über Faktoren wie CCR5, CCR9 und Integrin α4β7 die direkte Migration der T-Zellen fördern. Den direkt migrierenden vornehmlich TH1 polarisierten Zellen folgen weitere, CD27 und Integrin αLβ2 exprimierende, zytotoxische T-Zellen aus der Blutbahn. Die direkt migrierenden Zellen könnten als Initiator und Potentiator der intestinalen T-Zell Infiltration wirken und müssen für zukünftige therapeutische Strategien nicht nur der Darm GvHD, sondern der intestinalen Inflammation im Allgemeinen mitberücksichtigt werden.
Das Masernvirus (MV) kann in Erkrankten eine schwere, langanhaltende Immunsuppression verursachen, wodurch Infektionen mit opportunistischen Pathogenen begünstigt werden. Diese basiert auf einer Paralyse der hämatopoetischen Zellen, welche das Virus durch Kontakt eines viralen Glykoproteinkomplexes zu einem unbekannten RezeptorX auf der Zell- Oberfläche induzieren kann. Kerncharakterisitika hiervon sind unter anderem die Herabregulation der Akt-Kinase-Phosphorylierung, die Inhibition der zellulären Proliferation und die Aktivierung der neutralen Sphingomyelinase 2 (NSM2).
In einem kinetischen Phosphoproteom konnten zwei potentielle Interaktionsrezeptoren des MV identifiziert werden: CD43 und P2X3. Das hochglykosylierte Oberflächenmolekül CD43 ist auf hämatopoetischen Zellen ubiquitär exprimiert und reguliert in T-Zellen deren Überleben, Proliferation, Aktivierung, Migration und Adhäsion. P2X3 wird in hämatopoetischen Zellen nur in geringem Maße exprimiert. Seine funktionelle Bedeutung ist in diesem Kompartiment nicht bekannt. Beide Kandidaten wurden mittels CRISPR/Cas9 Verfahren einzeln oder kombiniert aus Jurkat-T-Zellen ablatiert, welche nachfolgend nach MV-Kontakt hinsichtlich der oben erwähnten MV-modulierten Parameter getestet wurden. Zusätzlich wurden iso- und allosterische P2X3-Inhibitoren an primären und Jurkat-T-Zellen verwendet, um dessen Rolle in Ca2+-Mobilisierung und Proliferation nach T-Zell-Rezeptor Co-Stimulation zu analysieren.
Die genetische Depletion beider Rezeptor-Kandidaten verringerte die Effekte des MV auf alle getesteten Parameter signifikant, was darauf hindeutet, dass beide Proteine entscheidend an der T-Zell-Suppression beteiligt sind. Während die isosterische Inhibition von P2X3 keinen Effekt hatte, wurde die Proliferation primärer T-Zellen durch dessen allosterische Inhibition vor Co-Stimulation fast verdoppelt und die Effizienz der Ca2+-Mobilisierung in Jurkat- und primären T-Zellen signifikant erhöht. In P2X3-depletierten Jurkat-Zellen hingegen war die Ca2+-Mobilisierung nach Stimulation signifikant geringer als in WT-Zellen.
In dieser Arbeit konnten zwei wichtige Mediatoren der MV induzierten T-Zell-Suppression identifiziert werden. Vor allem P2X3, dessen Expression, Regulation und funktionelle Bedeutung im hämatopoetischen Kompartiment noch nicht erforscht wurde, könnte ein vielversprechender Kandidat für eine antivirale Therapie darstellen, da ein klinisch getesteter P2X3-Inhibitor bereits verfügbar ist.
Präeklampsie ist einer der Hauptgründe für maternale und fetale Mortalität und Morbidität. Obwohl die Aetiologie weitgehend unklar ist, wird angenommen, dass eine mangelnde Toleranz von Seiten des mütterlichen Organismus gegenüber dem Fetus und dadurch dessen sukzessive Abstossung ursächlich sein könnte. T regulatorischen Zellen wurde in der Vergangenheit eine mögliche Rolle in der Aufrechterhaltung einer gesunden Schwangerschaft mit erfolgreicher Integration des Fetus, eines durch die paternalen Antigene charakterisierten semi-haploiden Allografts, zugesprochen. In dieser Arbeit wurden T regulatorische Zellen am Ende des dritten Trimesters im peripheren Blut nicht-Schwangerer, gesunder Schwangerer sowie Schwangerer mit Präeklampsie untersucht. Zusätzlich wurden T-regulatorische Zellen in decidualem Gewebe bestimmt. Aufgrund der verschiedenen Ergebnisse vorangegangener Studien und der Uneinigkeit über die Identifizierung T regulatorischer Zellen wurden T regulatorische Zellen in dieser Arbeit mittels der drei gängigsten Markerkombinationen (foxp3+, CD4+CD25hi sowie CD4+CD127loCD25+) charakterisiert. Im peripheren Blut gesunder Schwangerer gab es signifikant mehr T regulatorische Zellen als im peripheren Blut nicht-Schwangerer gemessen mit den Markerkombinationen CD4+CD25hi sowie CD4+CD127loCD25+. In Schwangerschaften mit Präeklampsie zeigten sich eine verminderte Frequenz von T regulatorischer Zellen verglichen mit gesunden Schwangerschaften für die Markerkombinationen foxp3+ und CD4+CD127loCD25+. Auf lokaler Ebene, der Decidua, wurde bei gesunden Schwangerschaften sowie Schwangerschaften mit Präeklampsie eine erhöhte Frequenz gegenüber dem peripheren Blut gemessen. T-regulatorische Zellen scheinen also eine Rolle in der Aufrechterhaltung gesunder Schwangerschaften zu spielen, während sie in Schwangerschaften mit Präeklampsie vermindert sind.
Human Vγ9Vδ2 T cells are the main γδ T cell subset in the circulation, accounting for up to 5% of the total peripheral blood lymphocyte population. They have been suggested to be important in response to tumors and infections. Their immune mechanisms encompass cell killing via cytotoxicity and secretion of pro-inflammatory cytokines such as IFNγ and tumor necrosis factor (TNF). The main stimulators of Vγ9Vδ2 T cells are isopentenyl pyrophosphate (IPP) and (E)-4-hydroxy-3-methyl-but-2-enyl pyrophosphate (HMBPP), denominated phosphoantigens (PAg).
A major advance in the understanding of PAg detection and Vγ9Vδ2 T cell activation has been the identification of the butyrophlin 3A (BTN3A) proteins as key mediators in these processes. In humans, three isoforms constitute the BTN3A family: BTN3A1, BTN3A2, and BTN3A3; and their genes are localized on the short arm of chromosome 6. The role of BTN3A1 has been highlighted by BTN3A-specific monoclonal antibody 20.1 (mAb 20.1), which has an agonist effect and causes proliferation, expansion, and activation of primary human Vγ9Vδ2 T cells. On the other hand, BTN3A-specific monoclonal antibody 103.2 (mAb 103.2) is antagonistic, inhibiting the Vγ9Vδ2 T cell response. The actual mechanism underlying both PAg- and mAb 20.1-mediated activation is not completely elucidated, but the importance of BTN3A1 is clear.
The main objective of this dissertation was to characterize the role of BTN3A1 in the PAg-dependent and PAg-independent Vγ9Vδ2 T cell activation and to evaluate its contribution in the response to influeza A virus infected cells. This research work demonstrated, by using Vγ9Vδ2 TCR MOP-transduced murine cells (reporter cells), that human chromosome 6 (Chr6) is mandatory for PAg-induced stimulation, but not for stimulation with mAb 20.1. The reporter cells responded to mAb 20.1 in cultures with BTN3A1-transduced Chinese hamster ovary cells (CHO BTN3A1) as antigen presenting cells. Nevertheless, for PAg-dependent activation the presence of Chr6 in CHO BTN3A1 was mandatory.
Although reporter cells expressing clonotypically different Vγ9Vδ2 TCRs showed similar PAg response, they clearly differed in the mAb 20.1 response. The reporter cell line transduced with Vγ9Vδ2 TCR D1C55 demonstrated essentially no response to mAb 20.1 compared to Vγ9Vδ2 TCR MOP cells. These findings were further supported by experiments performed with human PBMCs-derived Vγ9Vδ2 T cell clones. The results indicate heterogeneity in the PAg- and 20.1-dependent responses, in terms of CD25 and CD69 expression, among three different Vγ9Vδ2 T cells clones.
Co-cultures of reporter cells with Raji RT1BI and PAg plus mAb 20.1 or single chain antibody 20.1 (sc 20.1) revealed no additive or synergistic activating effects. In contrast, mAb 20.1 or sc 20.1 inhibited the PAg-mediated activation of the reporter cells.
The comparison of the relative contribution of the isoforms BTN3A2 and BTN3A3, in the activation of Vγ9Vδ2 T cells, was undertaken by overexpression of these isoforms in CHO cells. The results showed that BTN3A2 contributes to both PAg- and mAb-induced Vγ9Vδ2 T cell activation. On the contrary, BTN3A3 does not support PAg-mediated γδ T cell response.
Additionally, mutations in the proposed PAg- and mAb 20.1-binding sites of the extracellular BTN3A1 domain were generated by means of site-directed mutagenesis. These mutations revoked the mAb 20.1-induced Vγ9Vδ2 T cell activation, but not that induced by PAg.
Finally, co-cultures of Vγ9Vδ2 TCR MOP-transduced murine reporter cells with influenza A/PR/8/34-infected cells, or infection of PBMCs with this virus strain indicated that BTN3A1 might be dispensable for the Vγ9Vδ2 T cell response against influenza A.
The data of this research work points out that: i) in addition to BTN3A1, other Chr6-encoded genes are necessary for Vγ9Vδ2 T cell activation with PAg; ii) clonotypical (CDR3) differences influence the PAg- and mAb 20.1-mediated Vγ9Vδ2 T cell activation; iii) the PAg- and mAb 20.1-induced responses are not synergistic and interfere with each other; iv) BTN3A2 and BTN3A3 isoforms differ in the ability to support PAg- or mAb 20.1-dependent Vγ9Vδ2 T cell activation; v) the importance of the intracellular B30.2 domain of BTN3A1, in the Vγ9Vδ2 T cell activation, might be higher than that of the extracellular domain; and vi) in spite of the importance of BTN3A1 in the activation of Vγ9Vδ2 T cells, it is possible that many molecules with redundant functions are involved in the elimination of influenza virus infection by these cells.
In summary, it is possible to hypothesize a model in which BTN3A1 detects prenyl pyrophosphates in the cytoplasm via its B30.2 domain and in association with another protein(s). The binding of PAg to this domain induces a multimerization of BTN3A1 or a conformational change of its extracellular domain (mimicked by mAb 20.1). These modifications might be recognized by the Vγ9Vδ2 TCR or by an associated T cell protein. In the case that the TCR directly recognizes BTN3A1, the intensity of the response will depend on the Vγ9Vδ2 TCR clonotype. Future research will allow to gain a better understanding of BTN3A1, its interaction with other proteins, its actual role in the activation of Vγ9Vδ2 T cells, and its importance in specific models of cancer or infection. This knowledge will be necessary to transform these cells into effective tools in the clinic.
Zu den gefährlichen Komplikationen der Masern gehört die selten vorkommende ZNS-Erkrankung subakute sklerosierende Panenzephalitis (SSPE), die erst mehrere Jahre nach einer akuten Masernvirusinfektion auftritt. Die SSPE verläuft immer tödlich und bis heute gibt es keine spezifische Therapie gegen diese Erkrankung. Unsere Arbeitsgruppe hat ein Modell für eine persistierende, virale ZNS-Infektion entwickelt, in dem 2-Wochen-alte, immunologisch normale C57BL/6-Mäuse mit einem rekombinanten, neurotropen Masernvirus (MV), das das Hämagglutinin eines an Nagern angepassten MV-Stammes enthält, intrazerebral (i.c.) infiziert werden. Im Rahmen dieser Doktorarbeit wurde die Rolle regulatorischer CD4+ CD25+ Foxp3+ T-Zellen (Treg) in diesem Mausmodell analysiert und untersucht, ob die persistierende ZNS-Infektion durch Manipulation peripherer Treg beeinflusst werden kann. Außerdem wurde ein IFN-y-ELISPOT-Assay etabliert, der CD8+ zytotoxische T-Zellen (CTL) identifiziert, die spezifisch für die MV-Hämagglutinin-Epitope MV-H22-30 (RIVINREHL) bzw. MVH446-454 (SNHNNVYWL) sind. In Bezug auf das erstgenannte Epitop wurde desweiteren eine Pentamer-Färbung etabliert, um CTLs mit Hilfe der Durchflusszytometrie zu identifizieren, die H-2Db-gekoppelte MV-H22-30-Epitope erkennen. Die Ergebnisse dieser Doktorarbeit zeigen, dass sich trotz eines hohen Anteils Masern-spezifischer CTLs und nur sehr wenigen Treg im Gehirn eine persistierende ZNSInfektion ausbildet. Periphere Treg wurden während der persistierenden Phase der ZNS-Infektion expandiert bzw. depletiert und die Konsequenzen für die Virus-spezifische Immunantwort sowie das Ausmaß der persistierenden Infektion wurden analysiert. Die Expansion von Treg mit Hilfe des superagonistischen anti-Maus CD28 Antikörpers D665 verursachte eine transiente Immunsuppression, die die Virus-Replikation sowie -Ausbreitung im Gehirn verstärkte. Im Gegensatz dazu führte die Depletion von Treg in DEREG-Mäusen mittels Diphtherietoxin zu einem erhöhten Anteil Virus-spezifischer CTLs im Gehirn sowie zu einer Reduktion der persistierenden ZNS-Infektion. Diese Daten zeigen, dass Treg die Fähigkeit besitzen, die Persistenz von MV im Gehirn zu kontrollieren und somit möglicherweise Teil eines Therapiekonzeptes gegen ZNS-Infektionen mit dem Masernvirus sein können. Frühere Studien unserer Arbeitsgruppe haben außerdem gezeigt, dass das durch IFN-y induzierbare Enzym Indolamin-2,3-Dioxygenase (IDO) antivirale Aktivitäten gegen MV aufweist. Dies wurde in dieser Doktorarbeit in vivo in unserem Mausmodell anhand von IDOk.o.-Tieren bestätigt, die nach i.c. Infektion nicht nur eine erhöhte Mortalitätsrate aufwiesen sondern auch in den überlebenden Tieren eine verstärkte persistierende ZNS-Infektion zeigten.
Die Interaktion des Masernvirus mit T-Zellen stört die Aktivierung der TCR-Signalkaskade durch die Hemmung der Phosphatidylinositol-3-Kinase (PI3K), die zur Einstellung der T-Zell-Funktionen führt und dadurch nachgeschaltete (downstream) Signalwege sowie den Eintritt in den Zellzyklus, aber auch die Genexpression reguliert. Infolgedessen können die Aktivität spleißregulatorischer Faktoren sowie die Spleißmuster von mRNAs verändert werden, wie zum Beispiel bei der alternativ gespleißten SHIP1-Isoform SIP110, die eine T-Zell-inhibitorische Aktivität zeigt. Um alternativ gespleißte (AS) und differentiell regulierte (RG) Transkripte in T-Zellen infolge von PI3K-Inhibition zu erfassen, wurde ein Human Exon 1.0 ST Array an RNA-Proben von humanen T-Zellen, 24 h stimuliert und stimuliert/ PI3K-inhibiert, durchgeführt. Durch die Anwendung geeigneter bioinformatischer Algorithmen konnten spezifisch in PI3K-inhibierten Zellen angereicherte Transkripte nachgewiesen und in die Kategorien AS (2192 Gene) und RG (619 Gene) eingeteilt werden. Ausgewählte Gene wurde mittels RT-PCR und qPCR validiert, gefolgt von der funktionellen Annotation beider Genlisten. AS Gene konnten verstärkt in ECM-Rezeptor Interaktionen, fokaler Adhäsion, Proliferation, Zytoskelettorganisation und Tumorsignalwegen gefunden werden, während RG Gene eher in der DNA-Replikation, DNA-Reparatur und Stressantwort vertreten waren. Gene beider Gruppen konnten auf Signalwege bezogen werden, die essentiell für den TCR-Signalweg, die Zytoskelettdynamik und den Zellzykluseintritt waren. Das stützt die Annahme, dass die Außerkraftsetzung der PI3K-Schüsselprozesse der T-Zell-Aktivierung sowohl auf der Ebene der RG als auch der AS Gene wirkt. Über die Ingenuity Pathway Analyse konnten wir unsere Genlisten mit Genen vergleichen, die bereits auf solche Schlüsselprozesse und die Viabilität bezogen werden können. In der Überschneidung wurden z.B. die AS GTP-Austauschfaktoren Vav1 und Vav3 gefunden, die für die Übersetzung extrazellulärer Signale in Zytoskelettdynamik, Proteinphosphatasen und Adapter von Bedeutung sind.
Ausgewählte Gene (AS - FBXO6 und LAT2, RG - SLFN5) wurden aus PI3K-arretierten T-Zellen kloniert und deren Effekt auf grundlegende zelluläre Funktionen durch Überexpression in HEK293T-Zellen überprüft. Die Fusionsproteine veränderten weder die Zellviabilität noch die Proliferation dieser Zellen. Über einen auf siRNA basierenden Knockdown wurde überprüft, ob das RG Gene SLFN5 als Suppressor auf die T-Zell-Aktivierung agiert. Der Knockdown in primären T-Zellen zeigte keinen Einfluss auf die Zellviabilität, Proliferation und Polarisation. Jedoch konnte ein signifikanter Effekt auf die T-Zell-Adhärenz auf Fibronektin gezeigt werden, was darauf schließen lässt, dass SLFN5 die T-Zell-Adhärenz negativ reguliert.
Des Weiteren wurde die MV-induzierte Regulation selektierter Gene betrachtet und Unterschiede in der Regulation im Vergleich zur direkten PI3K-Inhibition festgestellt. Ein Grund dafür könnte sein, dass das MV eine Inhibition auf vielen Ebenen induziert, anstelle der alleinigen PI3K-Inhibition.
Abschließend wurde untersucht, ob ausgewählte Gene an der Regulation in verschiedenen T-Zelllinien beteiligt sind und als Tumorsuppressoren agieren könnten. FBXO6 als Regulator der CHK1-Stabilität wurde in den meisten Zelllinien nicht exprimiert. Die Annahme, dass eine Stress-induzierte defekte Ubiquitinierungsmaschinerie an der Resistenz von Tumorzellen auf Chemotherapeutika beteiligt ist, macht FBXO6 zu einem interessanten Kandidaten als Biomarker für Tumorsensitivität gegenüber Krebsmedikamenten. Diese Annahme bedarf jedoch weiterer Untersuchungen.
Effective T cell immunity was believed to occur by mature DC, whereas tolerogenicity was attributed strictly to immature DC phenotypes. However, intermediate DC maturation stages were identified conditioned by inflammatory mediators like TNF. Furthermore, the T cell tolerance mechanisms are dependent on distinct modes and intensities of co-stimulation. Therefore, in this study it was addressed how distinct DC maturation signatures instruct CD4+ T cell tolerance mechanisms. DC acquire antigens from apoptotic cells for self-peptide-MHC presentation and functionally adapt presumed tolerogenic DC phenotypes. Here, immature murine bone-marrow derived DC representing both inflammatory and conventional DC subsets adapted a maturationresistant DC signature upon apoptotic cell recognition but no additional tolerogenic features. Immature DC instruct CD4+ FoxP3+ regulatory T cells in a TGF-β prone micro-environment or generate anergic CD4+ T cells hampered in the TCR-induced proliferation and IL-2 secretion. Secondary stimulation of such anergic CD4+ T cells by immature DC increased primarily IL-10 production and conferred regulatory function. These IL-10+ regulatory T cells expressed high levels of CTLA-4, which is potently induced by immature DC in particular. Data in this work showed that anergic T cells can be re-programmed to become IL-10+ regulatory T cells upon ligation of CTLA-4 and CD28 signalling cascades by B7 costimulatory ligands on immature DC. In contrast, semi-mature DC phenotypes conditioned by the inflammatory mediator TNF prevented autoimmune disorders by induction of IL-10+ Th2 responses as demonstrated previously. Here, it was shown that TNF as an endogenous maturation stimulus and pathogenic Trypanosoma brucei variant-specific surface glycoproteins (VSG) induced highly similar DC gene expression signatures which instructed default effector Th2 responses. Repetitive administration of the differentially conditioned semi-mature DC effectively skewed T cell immunity to IL-10+ Th2 cells, mediating immune deviation and suppression. Collectively, the data presented in this work provide novel insights how immature and partially mature DC phenotypes generate T cell tolerance mechanisms in vitro, which has important implications for the design of effective DC-targeted vaccines. Unravelling the DC maturation signatures is central to the long-standing quest to break tolerance mimicked by malignant tumours or re-establish immune homeostasis in allergic or autoimmune disorders.
After priming in Peyer's patches (PPs) and mesenteric lymph nodes (mLN) T- cells infiltrate the intestine through lymphatic draining and homing through the bloodstream. However, we found that in mouse models of acute graft-versus-host disease (GvHD), a subset of alloreactive T-cells directly migrates from PPs to the adjacent intestinal lamina propria (LP), bypassing the normal lymphatic drainage and vascular trafficking routes. Notably, this direct migration occurred in irradiated and unirradiated GvHD models, indicating that irradiation is not a prerequisite for this observed behavior.
Next, we established a method termed serial intravascular staining (SIVS) in mouse models to systematically investigate the trafficking and migration of donor T- cells in the early stages of acute GvHD initiation. We found that the direct migration of T-cells from PPs to LP resulted in faster recruitment of cells after allogeneic hematopoietic cell transplantation (allo-HCT). These directly migrating T-cells were found to be in an activated and proliferative state, exhibiting a TH1/TH17-like phenotype and producing cytokines such as IFN-γ and TNF-α. Furthermore, we observed that the directly migrating alloreactive T-cells expressed specific integrins (α4+, αE+) and chemokine receptors (CxCR3+, CCR5+, and CCR9+). Surprisingly, blocking these integrins and chemokine-coupled receptors did not hinder the direct migration of T- cells from PPs to LP, suggesting the involvement of alternative mechanisms. Previous experiments ruled out the involvement of S1PR1 and topographical features of macrophages, leading us to hypothesize that mediators of cytoskeleton reorganization, such as Coro1a, Dock2, or Cdc42, may play a role in this unique migration process.
Additionally, we observed that directly migrating T-cells created a local inflammatory microenvironment, which attracts circulating T-cells. Histological analysis confirmed that alloreactive PPs-derived T-cells and bloodborne T-cells colocalized. We employed two experimental approaches, including either photoconversion of T-cells in PPs or direct transfer of activated T-cells into the vasculature, to demonstrate this colocalization. We hypothesize that cytokines released by migrating T-cells, such as IFN-γ and TNF-α, may play a role in recruiting T-cells from the vasculature, as inhibiting chemokine-coupled receptors did not impair recruitment.
To analyze the role of protein kinase B(PKB)on developmental and functional aspects of T cells, we have generated transgenic mouse lines expressing a constitutively active form of PKB (myrPKB) in early stages of T cell development.Peripheral CD4+ T cells from PKB tg mice are hyperreactive, more efficient in producing th1 and th2 cytokines and show faster and CD28 co-stimulation independent cell cycle progression.Interestingly PKB tg T cells are resistant to CsA treatment in proliferation and cytokine production.Further analysis show PKB tg CD4+ T cells have a drastically reduced nuclear translocation of NFAT proteins and this is due to a direct interaction between PKB and NFAT. To study whether the negative regulatiopn of NFATs by PKB affects T cell development, we analyzed double tg mice expressing both, a constitutively active version of calcineurin (dCam) and myrPKB. dCam tg mice have a severe block in thymocyte development at the DN3 stage.But in the dCam/PKB double tg mice this developmental block is significantly rescued.This rescue of thymocyte development by PKB is due to the expression of RAG1 and subsequent TCRb chain expression. CsA treatment of neonatal thymic lobes from dCam mice restores normal thymocyte development, indicating involvement of NFATs in the severe block in dCam thymocyte development.Confocal studies clearly established that compared to dCam DN cells there is a significant reduction in the nuclear levels of NFATc1 and NFATc3 in dCam/PKB cells.Downregulation of nuclear NFAT levels by myrPKB thus seems to be an essential parameter in dCam cells to proceed with normal differentiation. In summary, the data from PKB tg peripheral CD4+ T cells and dCam/PKB double tg thymocytes clearly establish PKB as an important modulator of T cell development and function and PKB as a novel negative regulator of NFAT activation.
Typ 1 NKT Zellen oder iNKT Zellen (invariante Natürliche Killer T Zellen) stellen eine Subpopulation der abT Zellen dar, die sich durch mehrere charakteristische Eigenschaften aus- zeichnet. Ihr Hauptmerkmal ist die Expression eines semi-invarianten T Zellrezeptors (TCR), der die Bindung von CD1d:Glycolipid Komplexen ermöglicht, wohingegen ‚klassische‘ T Zellen an Komplexe aus MHC (Haupthistokompatibilitätskomplex) Molekülen und Peptiden binden. Die während der Reifung im Thymus durch Transkriptionsfaktoren festgelegte Voraktivierung der iNKT Zellen ermöglicht das unmittelbare Freisetzen von Cytokinen bei Antigenkontakt, wodurch iNKT Zellen die adaptive Immunantwort stark beeinflussen können: Sie tragen sowohl zur Regulation von Autoimmunerkrankungen als auch der Bekämpfung von Krebs und Infektionen bei.
Der iNKT TCR setzt sich aus einer invarianten a-Kette (AV14/AJ18 in der Maus bzw. AV24/AJ18 im Menschen) und einer charakteristischen Auswahl an b-Ketten (vorwiegend BV8S2, BV7 und BV2 in der Maus und BV11 im Menschen) zusammen. Das Cerebrosid a-Galactosylceramid (aGC, KRN7000) stellt eines der potentesten Antigene für iNKT Zellen dar. Die Präsentation dieser Antigenklasse erfolgt durch CD1d Moleküle, die, abgesehen von tiefen hydrophoben Bindungstaschen, strukturell MHC I Molekülen ähneln, jedoch nicht polymorph sind und außerhalb des MHC Locus codiert sind. Die, zwischen Maus und Mensch hochkon- servierte, Interaktion von iNKT TCR und CD1d:aGC Komplex zeichnet sich bei potenten Antigenen durch die eingeschränkte Nutzung der Antigenspezifität bestimmenden Regionen aus: CDR1a, CDR3a und CDR2b. Die den CDR3b definierende V-D-J Umlagerung der b-Kette stellt im iNKT TCR den Bereich der höchsten Variabilität dar, beeinflusst jedoch nur die Bindung schwächerer Antigene. Natürlich auftretende Variabilität innerhalb der a-Kette kann durch Abweichungen von der kanonischen V-J Umlagerung am Beginn des CDR3a entstehen und beeinflusst ebenfalls die Bindung des iNKT TCR.
Die iNKT Zellpopulation in F344 Ratten ähnelt in Frequenz und Korezepotorexpression derjenigen des Menschen. Ratten besitzen ein CD1D Gen, welches hoch homolog zu denen der Maus ist und zwei dem BV8S2 Gensegment der Maus homologe BV Segmente (BV8S2 und BV8S4), die in F344 Ratten beide funktionell sind. Eine Besonderheit der Ratte ist jedoch das Auftreten einer AV14 Multigenfamilie von bis zu zehn Gensegmenten. Diese unterscheiden sich neben dem HV4 vor allem in ihren CDR2 Sequenzen und werden anhand dieser Unterschiede in zwei Gruppen (Typ 1 und 2) eingeteilt. Zusätzlich wurde in der iNKT Zellpopulation eine hohe Frequenz an natürlich auftretenden A93G Substitutionen in der TCR↵ Kette beschrieben und es wurde gezeigt, dass, im Gegensatz zur Kreuzreaktivität zwischen iNKT TCR und CD1d von Maus und Mensch, iNKT Zellen der Ratte nicht an Maus CD1d binden. Die Besonderheiten des Ratten iNKT TCR und deren Auswirkungen auf die TCR Expression und Ligandenbindung der Ratten iNKT Zellpopulation wurden in der vorliegenden Arbeit untersucht.
Durch in dieser Arbeit durchgeführte in vitro Mutagenesestudien konnten Position 68 in der vierten Hypervariablen Schleife (HV4↵) und Position 93 zu Be- ginn des CDR3↵ als entscheidende Modulatoren der CD1d Bindung im iNKT TCR von Ratte und Maus identifiziert werden, wobei auch speziesspezifische Unterschiede aufgedeckt werden konnten. Die Spezieskreuzreaktivität des Ratten iNKT TCR selbst hing stark von einer A93G Substitution im TCRa ab. Bei Untersuchungen der b-Kette zeigte sich, dass sowohl BV Segmente als auch CDR3b Region die Ligandenbindung in differenziellem Zusammenspiel beeinflussen, was bei Paarung mit unterschiedlichen AV14 Segmenten verschieden ausgeprägt sein konnte. Weiterhin wurden humane CD1d Dimere generiert und zum ersten Mal die Bindung von Ratten CD1d an humane iNKT TCR gezeigt.
Weiterhin wurde in dieser Arbeit das TCR Repertoire von iNKT Zellen der F344 Ratte und deren CD1d Bindungseigenschaften charakterisiert. Hierzu wurde die bereits etablierte Methode der in vitro Expansion von iNKT Zellen aus der Rattenmilz weiterentwickelt, was die Langzeitkultur und -expansion der sortierten iNKT Zellpopulation ermöglichte. Bei Untersuchung der TCR Expression konnte gezeigt werden, dass die Auswahl der im Ratten iNKT TCR genutzten BV Gensegmente ähnlich limitiert ist wie in der Maus. Neben der dominanten Nutzung der BV8S4 und BV8S2 Gensegmente wurden hauptsächlich BV8S1, BV14 und BV7 gefunden. Bei Untersuchungen der CD1d Dimerbindung der iNKT Zellpopulation konnte der Einfluss der na- türlich auftretenden A93G Substitution in der iNKT TCRa Kette bestätigt werden. Außerdem zeigte sich hier ebenfalls der Einfluss des BV Gensegments auf die Ligandenbindung, wobei BV8S4 negative Zellen im Vergleich zu BV8S4 positiven Zellen eine stärkere Ratten CD1d Dimerbindung zeigten.
Kinasen der SRC-Familie (SFKs) sind sowohl in Wachstum und Metastasierung von Tumor- und Leukämiezellen als auch an prominenter Stelle in vielgestaltige Signalwege aller Immunzellen involviert. Eine Hemmung von SFKs ist damit ein vielversprechendes Mittel zur Therapie maligner Erkrankungen, kann aber darüber hinaus auch sehr effektiv zur Immunmodulation genutzt werden. Für den zur Therapie von CML und AML zugelassenen Tyrosinkinaseinhibitor (TKI) Dasatinib (Handelsname Sprycel®), für den unter anderem SFKs die Hauptziele darstellen, wurden, neben der antitumoralen Wirkung, sowohl immunsuppressive als auch immunstimulierende Effekte beschrieben. Aus diesem Grund könnte Dasatinib ein für die Modulation von Immunantworten sehr interessantes Hilfsmittel darstellen. In der vorliegenden Arbeit werden die hemmenden und fördernden Einflüsse von Dasatinib auf zwei Typen von Immunzellen genauer untersucht, um so die Auswirkungen einer Dasatinib-Behandlung auf Zellen des Immunsystems besser zu verstehen und sich das immunmodulatorische Potenzial von Dasatinib besser nutzbar machen zu können.
Der erste Teil der Arbeit beschäftigt sich mit der Untersuchung möglicher kombinatorischer Effekte zwischen Dasatinib und dem Glucocorticoid Dexamethason auf verschiedene Subsets von T-Zellen vor dem Hintergrund eines potentiellen Einsatzes der Kombination bei der allogenen Hämatopoetischen Stammzelltransplantation (HSCT) zur Separation von Graft-versus-Leukemia (GvL)-Effekten und der Graft-versus-host Disease (GvHD). Während keine kombinatorischen Effekte bei der Aktivierung von T-Zellen auftraten, ergaben sich bei der Untersuchung des Einflusses auf die Proliferation besonders in CD8+ T-Zellen additive Effekte durch die Kombination. Die Proliferation naiver T-Zell-Subsets wurde bereits durch die beiden Einzelsubstanzen alleine stark gehemmt. Dagegen waren Memory T-Zell-Subsets deutlich unempfindlicher, allerdings konnte durch eine Kombination von Dexamethason und Dasatinib auch die Proliferation dieser Memory Subsets effektiv gehemmt werden. Hierbei zeigten sich bei CD8+ Memory Subsets die deutlichsten synergistischen Effekte. Da eine Kombination in stärkerem Maße auch CD8+ gegenüber CD4+ Memory Subsets hemmt und diese Subsets unterschiedliche Rollen in der Induktion von GvL-Effekten und der Auslösung einer GvHD zu spielen scheinen, ist eine Steigerung der GvL-Effektivität durch die Medikamenten-Kombination bei gleichzeitiger Minimierung eines GvHD-Risikos in Zusammenhang mit anderen publizierten Ergebnissen durchaus denkbar. Weil eine starke Hemmung von virus-spezifischen T-Zellen nur bei sehr hohen Konzentrationen auftrat, ist zudem das Risiko einer Virus-Reaktivierung, die ein großes Problem bei einer HSCT darstellt, eher als gering einzuschätzen.
Der zweite Teil der Arbeit befasst sich mit dem Einfluss von Dasatinib auf aus Monozyten generierte Dendritische Zellen (moDCs) mit einem Fokus auf der Beeinflussung ihrer Migration. Während eine Behandlung mit Dasatinib nur sehr geringe Auswirkungen auf die Ausreifung der moDCs und die Expression von kostimulatorischen Molekülen hatte, führte eine Dasatinib-Behandlung zu einer Zeit- und Dosis-abhängigen Verringerung der Zytokinsekretion (IL-10 und IL-12). Im Gegensatz dazu hatte Dasatinib keinen Einfluss auf die phagozytotische Aktivität der moDCs und auf ihre Fähigkeit, Virus-spezifische T-Zell-Antworten auszulösen. Dasatinib zeigte dagegen einen deutlich steigernden Einfluss auf die Migration von moDCs gegen einen CCL19-Gradienten im Transwell-Assay, ohne die Expression des CCL19-Rezeptors CCR7 zu beeinflussen. Da ähnliche Migrations-steigernde Effekte auch bei einer Behandlung mit dem spezifischen SFK-Inhibitor SKI-1 auftraten, eine Behandlung mit Nilotinib, einem TKI der nicht auf SFKs wirkt, im Gegensatz dazu aber zu einer Hemmung der Migration führte, liegt es nahe dass die Migrations-steigernde Wirkung von Dasatinib über SFKs vermittelt wird. Dasatinib führte zu einer deutlichen Inhibierung der Phosphorylierung der inhibitorischen Immunrezeptoren Siglec-9 und Siglec-3 (CD33) ohne ihre Expressionslevel zu beeinflussen. Eine mit spezifischen Antikörpern durchgeführte Blockierung dieser Immunrezeptoren, deren ITIM-Domänen mutmaßlich von SFKs phosphoryliert werden, hatte eine deutliche Steigerung der Migration und eine verringerte Phosphorylierung von Siglec-9, Siglec-3 und SHP-2 zur Folge. Letztere ist eine Phosphatase, die nach Bindung an phosphorylierte ITIM-Domänen von Rezeptoren wie den Siglecs verschiedene Zielmoleküle dephosphoryliert. Die Ergebnisse dieser Arbeit legen nahe, dass die Migrations-steigernde Wirkung von Dasatinib über eine Hemmung von SFKs und daraus resultierend auf dem Wegfall eines inhibitorischen Signalwegs erfolgt. Diese Steigerung der Migration könnte in der Tumor-Therapie von großem Nutzen sein, da bei einer Vakzinierung mit autologen DCs, die mit Tumor-assoziierten Antigenen stimuliert wurden, die schlechte Einwanderung in die Lymphknoten eines der Hauptprobleme darstellt. Zur Überwindung dieses Problems könnte Dasatinib ein sehr effektives Hilfsmittel darstellen und die Therapie-Effizienz deutlich verbessern. Da Dasatinib aber auch eine ganze Reihe weiterer, sehr vielfältiger Einflüsse auf alle Arten von Immunzellen ausübt, scheint eine Verwendung spezifischer blockierender α-Siglec-Antikörper auf Grund geringerer Nebenwirkungen im Vergleich zu Dasatinib möglicherweise sogar noch deutlich besser geeignet zu sein, das Migrationsverhaltens Dendritischer Zellen positiv zu beeinflussen. Die Verwendung gegen Siglec-Rezeptoren gerichteter Antikörper als Adjuvantien könnte somit zu einem erfolgreicheren Einsatz der Vakzination mit Dendritischen Zellen in der Tumor-Therapie führen.
Der Kontakt humaner T-Zellen mit dem MV Glykoproteinkomplex interferiert mit der CD3/CD28 stimulierten Aktivierung von PI3/Akt-Kinase Signalwegen. Damit verbunden ist der ineffiziente Transport PH-Domänen-enthaltender Proteine in Membran-rafts, wie der Akt-Kinase und Vav, den Guaninnukleotid-Austauschfaktor von Rho GTPasen. Es konnte gezeigt werden, dass infolge des MV-Kontaktes die CD3/CD28 stimulierte Aktivität der Rho GTPasen Cdc42 und Rac1 inhibiert ist. Dagegen war in MV-behandelten Zellen eine leichte RhoA Aktivierung festzustellen. Rho GTPasen spielen eine kritische Rolle in der Regulation von Zytoskelettorganisation von T-Lymphozyten. Übereinstimmend damit wurde gezeigt, dass der Kontakt mit MV die CD3/CD28 costimulierte Aktivierung und Polymerisation des F-Aktins inhibiert. Damit verbunden ist die reduzierte Fähigkeit MV-behandelter T-Zellen auf Fibronektin- und mit CD3/CD28 Antikörpern-beschichteten Objektträgern zu polarisieren. Die Ausbildung F-Aktin-getriebener morphologischer Veränderungen, wie Filopodien, Lamellipodien und Uropodien, ist drastisch reduziert. Rasterelektronenmikroskopische Auf-nahmen zeigten in nicht-stimulierten und CD3/CD28 costimulierten MV-behandelten T-Zellen einen nahezu kompletten Verlust an Mikrovilli und Lamellipodien. Die Bindung von MV induziert die Dephosphorylierung des F-Aktin–bindenden Proteins Cofilin und der ERM-Proteine. Es konnte demonstriert werden, dass der MV-Kontakt die Ausbildung einer reifen immunologischen Synapse stört. Trotz der morphologischen Veränderungen konjugieren MV-behandelte T-Zellen mit DCs. Die Anzahl MV-behandelter T-Zellen, die mit DCs inter-agieren, ist vergleichbar mit der mock-behandelter T-Zellen. Allerdings zeigt die 3-dimensionale Rekonstruktion der DC/T-Zell-Kontaktzone, dass in MV-behandelten T-Zellen die zentrale Akkumulation und Clusterbildung des CD3-Moleküls gestört ist und keine monozentrische Synapse ausbildet wird. Desweiteren erfolgt die Relokalisation des MTOC in T-Zellen in Richtung der DC unvollständig. Zusammenfassend kann gesagt werden, dass der MV Glykoproteinkomplex mit essentiellen Schritten einer erfolgreichen T-Zell-Aktivierung während der APC/T-Zell-Interaktion interferiert.
The advances in genetic engineering have enabled us to confer T cells new desired functions or delete their specific undesired endogenous properties for improving their antitumor function. Due to their efficient gene delivery, viral vectors have been successfully used in T-cell engineering to provide gene transfer medicinal products for the treatment of human disease. One example is adoptive cell therapy with T cells that were genetically modified with gamma-retroviral and lentiviral (LV) delivery vectors to express a CD19-specific chimeric antigen receptor (CAR) for cancer treatment. This therapeutic approach has shown remarkable results against B-cell malignancies in pilot clinical trials. Consequently, there is a strong desire to make CAR T cell therapy scalable and globally available to patients. However, there are persistent concerns and limitations with the use of viral vectors for CAR T cell generation with regard to safety, cost and scale of vector production. In order to address these concerns, we aimed to improve non-viral gene transfer and genome editing tools as an effective, safe and broadly applicable alternative to viral delivery methods for T-cell engineering.
In the first part of the study, we engineered CAR T cells through non-viral Sleeping Beauty (SB) transposition of CAR genes from minimalistic DNA vectors called minicircles rather than conventional SB plasmids. This novel approach dramatically increased stable gene transfer rate and cell viability and resulted in higher yield of CAR+ T cells without the need of long ex vivo expansion to generate therapeutic doses of CAR+ T cells. Importantly, CD19-CAR T cells modified by MC-based SB transposition were equally effective as LV transduced CD19-CAR T cells in vitro and in a murine xenograft model (NSG/Raji-ffLuc), where a single administration of CD8+ and CD4+ CAR T cells led to complete eradication of lymphoma and memory formation of CAR T cells after lymphoma clearance.
To characterize the biosafety profile of the CAR T cell products, we did the most comprehensive genomic insertion site analysis performed so far in T cells modified with SB. The data showed a close-to-random integration profile of the SB transposon with a higher number of insertions in genomic safe harbors compared to LV integrants. We developed a droplet digital PCR assay that enables rapid determination of CAR copy numbers for clinical applications.
In the second part of the study, we ablated expression of PD-1, a checkpoint and negative regulator of T cell function to improve the therapeutic index of CAR T cells. This was accomplished using non-viral CRISPR/Cas9 via pre-assemble Cas9 protein and in vitro-transcribed sgRNA (Cas9 RNP). Finally, we combined our developed Cas9 RNP tool with CAR transposition from MC vectors into a single-step protocol and successfully generated PD-1 knockout CAR+ T cells. Based on the promising results achieved from antibody-mediated PD-1 blockade in the treatment of hematological and solid tumors, we are confident that PD-1 knockout CAR T cells enhance the potency of CAR T cell therapies for treatment of cancers without the side effects of antibody-based therapies.
In conclusion, we provide a novel platform for virus-free genetic engineering of CAR T cells that can be broadly applied in T-cell cancer therapy. The high level of gene transfer rate and efficient genome editing, superior safety profile as well as ease-of-handling and production of non-viral MC vectors and Cas9 RNP position our developed non-viral strategies to become preferred approaches in advanced cellular and gene-therapy.
Der MHC der Lewis.1F-Ratte (RT1f) stimuliert die präferentielle Expansion von CD8-T-Zellen, die das V-Segment 8.2 (AV8S2) der TCR-alpha-Kette verwenden. Sowohl als Folge der positiven thymischen Selektion in der Lewis.1F-Ratte (LEW.1F) wie auch bei der RT1f-Alloerkennung. Es war Ziel dieser Arbeit, das MHC Kl. I-Molekül der LEW.1F-Ratte, das diese präferentielle Expansion von AV8S2-Zellen induziert, zu klonieren sowie die Kontaktpunkte der Interaktion zwischen dem MHC-Molekül und AV8S2 zu kartieren. Über RT-PCR wurde ein MHC-I-Gen der LEW.1F-Ratte (RT1.Af) isoliert, sequenziert und zusammen mit einem anderen MHC-I-Gen aus der LEW.1F-Ratte (A2f) analysiert, das von einer Arbeitsgruppe aus Babraham (Cambridge, UK) gefunden worden war. Durch einen Nukleotidsequenz-Vergleich von Af und A2f mit bekannten Ratte-MHC-I-Genen konnte gezeigt werden, daß die Sequenzen beider charakteristisch für Ratte-MHC-I-Gene sind. Beide Moleküle wurden stabil in L-Zellen, rCD80-transfizierte P815-Zellen und T-Zellhybridomen (THO35/1) exprimiert. Die serologische Charakterisierung mit 19 RT1f-reaktiven Alloantikörpern ergab, daß sich die LEW.1F-Serologie alleine durch Af erklären läßt. In einem Zytotoxizitätstest mit LEW.1F-reaktiven zytotoxischen T-Lymphozyten, wurde Af spezifisch erkannt, A2f dagegen nicht. Serologie und Zytotoxizitätstest zeigen, daß die RT1f-spezifische Alloerkennung das Af-Molekül fokussiert, A2f unbedeutend scheint. Möglicherweise, da dieses Molekül in der LEW.1F-Ratte nur schwach exprimiert ist. Die Interaktion von Af und A2f mit AV8S2-Zellen wurde mittels einer MLR (gemischte Lymphozytenreaktion) in der Alloerkennung untersucht: Af, nicht aber A2f, stimuliert die präferentielle Expansion AV8S2-positiver CD8-T-Zellen in der Alloreaktion. Somit ist Af als das Molekül identifiziert, daß die Überselektion von AV8S2-Zellen vermutlich auch in der thymischen Repertoire-Selektion induziert. Zur Kartierung der Af/AV8S2-Interaktion wurden Hybridmoleküle aus Aa und Af gentechnisch hergestellt: Aa und Af unterscheiden sich extrazellulär nur in sieben Aminosäuren, die konzentriert auf zwei Regionen des MHC-Moleküls liegen: "Region I" (AS 62, 63, 65, 69, 70) und "Region II" (167, 169). Aa stimuliert keine präferentielle AV8S2-Zell-Expansion, daher muß die charakteristische Kombination dieser sieben Aminosäuren für die präferentielle Interaktion mit AV8S2-positiven CD8-T-Zellen ausschlaggebend sein. Durch MLRs mit Hybrid-exprimierenden Stimulatorzellen wurde gezeigt, daß beide Regionen zur präferentiellen Expansion AV8S2-exprimierender CD8-T-Zellen beitragen. Durch den Vergleich mit anderen Daten unseres Labors konnte dargelegt werden, daß die beta-Kette zur präferentiellen Interaktion von AV8S2-Zellen mit Af keinen spezifischen Beitrag leistet. Ebenso unbeteiligt ist die Komplementarität-bestimmende Region 2 (CDR2) der alpha-Kette. Die charakteristischen Sequenzen von CDR1alpha und CDR3alpha dagegen sind für die präferentielle Expansion von AV8S2-positiven CD8-T-Zellen durch Af essentiell.

Tolerogenic dendritic cells (DC) and regulatory T (Treg) cells are able to prevent destructive immune responses. There is reason to hope that it may soon be possible to use DC and Treg cells to suppress immune responses antigen-specific, not only after transplantation, but also in the case of autoimmunity and allergy. At the moment, the generation of such cell types is very time-consuming and not suitable for clinical routine. In addition, it is not yet fully understood how these cells elicit a desired protective immune response in vivo and how the risks of an excessive immune suppression can be managed. The rat is one of the most important animal models in biomedical research. It is therefore surprising that tolerogenic DC and Treg cells in particular have not been more thoroughly investigated in this model. Thus, the aim of the present study was to systematically characterize these immune cells and investigate their impact on the immune system. Tolerogenic DC were generated from bone marrow precursors cultured with GM-CSF and IL-4 (= IL-4 DC). The proportion of naturally occurring Treg cells with a CD4posCD25posFoxp3pos phenotype comprises approximately 5-8% of the peripheral CD4pos T cells. The characterization of IL-4 DC revealed an up to 26-fold reduced expression of surface molecules such as MHC class II molecules, CD80, CD86, ICAM-1 and CD25 in comparison to mature splenic DC (S-DC). This low expression did not change when the cells where stimulated with different maturation-inducing signals such as replating, LPS, TNF- α and CD40L. Thus, these cells possess a robust phenotype resistant to maturation-inducing stimuli. IL-4 DC take up antigen via endocytosis and are not able to activate naïve T cells or to restimulate antigen-specific T cells. Furthermore, they are able to inhibit and prolongate mature S-DC induced T cell proliferation as well as mature S-DC induced restimulation of antigen-specific T cells, respectively. Thereby, the T cell proliferation was reduced up to 95%. This strong inhibitory effect was mediated within 24 hours in association with a reduced cytokine production (IL-2 about 49% and IFN-γ about 92%). The inhibitory properties of IL-4 DC don´t seem to be caused exclusively by the reduced expression of co-stimulatory molecules. In this study, the detection of the inhibitory molecules PD-L1 and PD-L2 on IL-4 DC suggests they have an impact on mediating inhibitory signals to the T cells. In addition, a suppressive effect of soluble factors was shown. The supernatant of one million IL-4 DC, collected after a 24 hour culture, suppressed mature S-DC induced proliferation of naïve T cells by about 90%. TGF-β, which was detected in the supernatant (up to 300 pg/ml), appears to be the causing soluble factor for this immune inhibition. By contrast, the supernatants of mature S-DC, which did not inhibit the activation of T cells, showed a TGF-β concentration of only about 100 pg/ml. The cytotoxic nitric oxide does not contribute to the IL-4 DC-mediated inhibition of T cell proliferation. The NO synthase inhibitor NMMA reduced the amount of NO by about 50%, but the decreased NO levels did not influence T cell proliferation. Indeed, IL-4 DC are not able to induce T cell proliferation, but this doesn´t mean that there is no change on the molecular level. For instance, T cells co-cultured with IL-4 DC during a first culture are not able to proliferate in the presence of mature S-DC during a second culture. This anergic-like state, however, could be abolished by adding exogenous IL-2. In addition, T cells co-cultured with IL-4 DC are able to inhibit the activation of naïve T cells. Naïve and activated T cells were not able to inhibit the mature S-DC induced T cell proliferation. This observation suggests the induction of Treg cells and was investigated in more detail. Indeed, flow cytometric analysis showed a 1.6-fold expansion of CD4posCD25posFoxp3pos T cells from naturally occurring Treg cells in the presence of IL-4 DC. Thereby, the expansion of CD4posCD25posFoxp3pos T cells occurs independently of the maturation state of DC. Both immature IL-4 DC as well as mature S-DC were able to expand the percentage of naturally occurring Treg cells. However, Treg cells pre-incubated with mature S-DC demonstrated a diminished inhibitory effect compared to Treg cells pre-incubated with IL-4 DC. Treg cells pre-incubated with IL-4 DC were able to inhibit the activation of naïve T cells. In this study it was shown that the regulatory potential of DC cannot be deduced solely by their phenotype or maturation state. Other factors, such as functional properties, need to taken into consideration, too. The induction of Treg cells with suppressive properties induced by in vitro generated tolerogenic IL-4 DC might provide an important mechanism for the maintenance of peripheral tolerance. However, for clinical application further investigation is necessary, not only to understand the interactions between tolerogenic DC and Treg cells, but also to investigate the impact of the transfer of a larger quantity of regulatory cells on the immune system of the recipient.
Der Myokardinfarkt (MI) gehört nach wie vor zu den führenden Todesursachen weltweit. Eine Minimierung der Infarktgröße, die durch die Dauer der Ischämie bestimmt wird, ist wesentlich für das Überleben und die Lebensqualität des Myokardinfarkt-Patienten. Die Reperfusion stellt aktuell eine zentrale klinische Intervention dar, um den myokardialen Schaden einzugrenzen. Dennoch führt die Reperfusion per se zu zusätzlichem Schaden am Herzen. Somit ist die Erforschung neuer Strategien zur Minimierung des myokardialen Reperfusionsschadens international von Interesse. Die Pathophysiologie des myokardialen Reperfusionsschadens ist vielschichtig und einige Komponenten sind auch heute in ihrer Wirkweise noch nicht vollständig mechanistisch verstanden. Die vorliegende Arbeit untersucht die Rolle von CD4+ T-Zellen und insbesondere deren Subpopulation der regulatorischen T-Zellen im myokardialen Reperfusionsschaden und stellt neue, auf T-Zellen abzielende, Therapien in Ergänzung zur myokardialen Reperfusion vor.
Zunächst wurde eine Infiltration von T-Zellen in das Myokard nach Ischämie-Reperfusion (I/ R) untersucht. Nach der Ischämie-Reperfusion wurden infiltrierende CD4+ T-Zellen als quantitativ führend und aktiviert identifiziert und erwiesen sich in der Infarktgrößenbestimmung als relevante Mediatoren des Reperfusionsschadens. CD25+Foxp3+ regulatorische T-Zellen (Treg) stellen eine Subpopulation von CD4+ T-Zellen mit immunsuppressiven Eigenschaften dar, die schnell und niederschwellig aktiviert werden können und kommen somit als zum Reperfusionsschaden beitragend in Frage. Mit Hilfe des DEREG (DEpletion of REGulatory T cells) -Mausmodells wurde gezeigt, dass regulatorische T-Zellen zum myokardialen Reperfusionsschaden beitragen; Treg-depletierte DEREG-Mäuse waren vor dem Reperfusionsschaden geschützt und zeigten kleinere Infarktgrößen als die Kontrolltiere. Zudem wurde mittels Transferexperimenten gezeigt, dass für den Treg-vermittelten Reperfusionsschaden die Anwesenheit von CD25- konventionellen T-Zellen (Tconv) erforderlich ist. Regulatorische T-Zellen stellen also einen in der vorliegenden Arbeit identifizierten potentiellen Angriffspunkt zur Reduktion des myokardialen Reperfusionsschadens dar.
Anhand von T-Zell-Rezeptor transgenen OT-II Mäusen und MHC (Major Histocompatibility Complex) Klasse II Knockout (KO) Tieren wurde gezeigt, dass Autoantigenerkennung im myokardialen Reperfusionsschaden eine Rolle spielt. Zur vollen T-Zell-Aktivierung notwendig ist neben dem MHC Klasse II-Signalweg und Kostimulatoren auch das Moleküle CD154 (CD40L). Die Gabe eines inhibitorischen anti-CD154-Antikörpers reduzierte die Infarktgröße in Wildtyp-Tieren sigifikant. Der myokardiale Reperfusionsschaden kann neben Zellen der adaptiven Immunität auch durch Neutrophile Granulozyten, Plättchen oder Inflammation des Endothels verstärkt werden. Knockout Mäuse mit einer Defizienz an CD4+ T-Zellen verfügten über eine verbesserte Mikroperfusion. Mechanistisch war nach 24h Reperfusion die absolute Zellzahl an Neutrophilen Granulozyten im CD4 KO im Vergleich zu Wildtyp-Mäusen unverändert; in Endothelzellen war die Regulation bestimmter Gene (VEGFα, TIMP-1 und Eng) nach I/ R im CD4 KO jedoch verändert.
Zusammengefasst zeigt die vorliegende Arbeit eine zentrale Rolle der Antigen-Erkennung durch den T-Zell-Rezeptor zur Aktivierung von CD4+ T-Zellen im myokardialen Reperfusionsschaden. In Anwesenheit von CD4+Foxp3+ T-Zellen ist der Reperfusionsschaden erhöht. Somit können CD4+Foxp3+ T-Zellen potentiell als Ziel für neuartige Therapien des Myokardinfarkts genutzt werden.
Summary: In the present work, two important negative regulators of T cell responses in rats were examined. At the molecular level, rat CTLA-4, a receptor important for deactivating T cell responses, was examined for the expression pattern and in vitro functions. For this purpose, anti-rat CTLA-4 mAbs were generated. Consistent with the studies in mice and humans, rat CTLA-4 was detectable only in CD25+CD4+ regulatory T cells in unstimulated rats, and was upregulated in all activated T cells. Cross-linking rat CTLA-4 led to the deactivation of anti-TCR- and anti-CD28 stimulated (costimulation) T cell responses such as reduction in activation marker expression, proliferation, and cytokine IL-2 production. Although T cells stimulated with the superagonistic anti-CD28 antibody alone without TCR engagement also increased their CTLA-4 expression, a delayed kinetics of CTLA-4 upregulation was found in cells stimulated in this way. The physiological relevance of this finding needs further investigation. At the cellular level, rat CD25+CD4+ regulatory T cells were examined here in detail. Using rat anti-CTLA-4 mAbs, the phenotype of CD25+CD4+ regulatory T cells was investigated. Identical to the mouse and human Treg phenotype, rat CD25+CD4+ T cells constitutively expressed CTLA-4, were predominantly CD45RC low, and expressed high level of CD62L (L-selectin). CD25+CD4+ cells proliferated poorly and were unable to produce IL-2 upon engagement of the TCR and CD28. Furthermore, rat CD25+CD4+ cells produced high amounts of anti-inflammatory cytokine IL-10 upon stimulation. Importantly, freshly isolated CD25+CD4+ T cells from naïve rats exhibited suppressor activities in the in vitro suppressor assays. In vitro, CD25+CD4+ regulatory T cells proliferated vigorously upon superagonistic anti-CD28 stimulation and became very potent suppressor cells. In vivo, a single injection of CD28 superagonist into rats induced transient accumulation and activation of CD25+CD4+ regulatory T cells. These findings suggest firstly that efficient expansion of CD25+CD4+ cells without losing their suppressive effects (even enhance their suppressive activities) can be achieved with the superagonistic anti- CD28 antibody in vitro. Secondly, the induction of disproportional expansion of CD25+CD4+ cells by a single injection of superagonistic anti-CD28 antibody in vivo implies that superagonistic anti-CD28 antibody may be a promising candidate in treating autoimmune diseases by causing a transient increase of activated CD25+CD4+ T cells and thus tipping ongoing autoimmune responses toward selftolerance.
This study focuses on phosphoantigen specific Vg9Vd2 T cells which only exist in human and non-human primates. This population accounts for 1%-5% of peripheral blood T-lymphocytes but their frequency can rise to 50% of total blood T cells upon infection. Vg9Vd2 T cells can be activated by nonpeptide compounds with critical phosphate moieties which are termed as phosphoantigens. These include isopentenyl pyrophosphate (IPP), a key compound of isoprenoid synthesis in all organisms, and (E)-4-Hydroxy-3-methyl-but-2-enyl pyrophosphate (HMBPP), a direct precursor of IPP in DOXP pathway which only exist in eubacteria, plants, apicomplexaen parasites. Its activity as phosphoantigen is at least 1000 fold higher than that of IPP. However, direct structural evidence of phosphoantigen binding to the TCR is missing so far. Moreover, Vg9Vd2 T cells have potent anti-tumor activity e.g. against the B-cell lymphoma Daudi, whose Vg9Vd2 T cell activating properties have been suggested to result from sensing of abnormal intracellular IPP levels by the Vg9Vd2 TCR or Vg9Vd2 TCR binding to other postulated ligands such as an ectopically expressed F1-ATPase or UL-16 binding protein 4 (ULBP4). Aminobisphosphonates and alkymines were hypothesized to activate Vg9Vd2 T cells indirectly by inhibiting the IPP consuming enzyme farnysyl pyrophosphates synthesis (FPPS) although off target effects of these drugs or a direct interaction with the Vg9Vd2 TCR could not be excluded. This thesis presents new approaches for the mechanistic analysis of Vg9Vd2 T cell activation. By employing retroviral transduction of FPPS specific shRNA, it shows that specific shRNA reduces expression of FPPS and is sufficient to convert hematopoietic and non-hematopoietic tumor cell lines into Vg9Vd2 T cell activators. FPPS knockdown cells activated Vg9Vd2 T cells as measured by increased levels of CD69 and CD107a, kill of FPPS knockdown cells and induction of IFN-γ secretion. The IPP-synthesis-inhibiting drug mevastatin reduced Vg9Vd2 T cell activation by FPPS knockdown cells or aminobisphosphonate treated cells but not activation by the phosphoantigen bromohydrin pyrophosphate (BrHPP). A reduced growth of the FPPS knockdown cells has not been observed which is different to what has been reported for aminobisphosphonate treated cells. Finally, the human B-cell lymphoma RAJI has been transduced with Tetracyclin-inducible FPPS specific shRNA and proven to gain and loose the capacity to activate Vg9Vd2 TCR transductants upon doxycylin provision or removal. Another approach for the analysis of Vg9Vd2 T cell activation is Vg9Vd2 TCR transduced mouse cell lines with specificity for phosphoantigens. In contrast to the previously used Vg9Vd2 TCR transduced Jurkat cells, these cells do not present phosphoantigens, and are therefore specially suited for analysis of phosphoantigen presentation. The response of the new TCR transductants to presumed Vg9Vd2 TCR ligands/activators such as phosphoantigens, aminobisphosphonates or FPPS knockdown cells, depended strongly on the expression of a rat/mouse CD28 molecule by the transductants and its ligation by the (CD80) counter receptor on the ligand-presenting cell. The response is likely to reflect recognition of cognate Vg9Vd2 TCR antigens since mutations in the TCR-δ chain CDR2 and 3 abolished this response but activation by TCR or CD3 specific antibodies. A major difference between TCR transductants and primary gd T cells, was the lacking response of TCR transductants to Daudi or IPP. In addition their sensitivity to other soluble phosphoantigens was about 100 fold weaker than that of primary cells, stimulation of both cell type to CD80 expressing FPPS knock down or aminobisphosphonates was similar. Finally, the transductants have also been used to analyze effects of over-expression or knockdown of enzymes of isoprenoid synthesis such as 3-hydroxy-3-methyl-glutaryl-CoA reductase (HMG-CoA reductase or HMGR), mevalonate-5-pyrophosphate decarboxylase (MVD), isopentenyl pyrophosphate isomerase (IDI), geranyl-geranyl pyrophosphate synthase (GGPPS) but no clear effects have been found. In conclusion, this thesis supports the concept of Vg9Vd2 T cells being sensors of a dysregulated isoprenoid metabolism and established new tools to study ligand recognition and TCR mediated activation of this T cell population. These tools will be most useful to address following questions: 1) How does the dysregulation of isoprenoid metabolism affect tumor growth? 2) What is the correlation between the modulation of IPP levels and the Vg9Vd2 TCR binding or expression of other postulated ligands? 3) Are there any mevalonate pathway enzymes other than FPPS and HMGR, which play an important role in Vg9Vd2 T cells activation? 4) What is/are the putative phosphoantigen-presenting molecule(s)?
Ziel dieser Arbeit war die Entwicklung optimaler DCs für die Generierung von therapeutischen Tumorvakzinen. Neben den essenziellen Eigenschaften der DCs wie der Migrationsfähigkeit, der Hochregulation kostimulatorischer Moleküle sowie der Zytokinausschüttung, ist auch die optimale Aufnahme von Tumor-spezifischen Antigenen und deren Präsentation besonders wichtig, um eine effiziente Immunantwort zur ermöglichen. Wichtigstes Ziel war es über einen optimalen Ausreifungscocktail der DCS eine effektive Antwort der zytotoxischen CD8\(^+\)-Zellen zu erzielen. Dies geschieht im Rahmen der Präsentation von z.B. exogenen Antigenen wie in unserem Falle eines CMV-Proteins über den Weg der sogenannten Kreuzpräsentation.
Hierzu wurde im Rahmen dieser Arbeit der in unserer Klinik etablierte zytokinbasierte Ausreifungscocktail (IL-1β, TNFα) mit einem Ausreifungscocktail, welcher Toll-like-Rezeptor-Agonisten mit PGE\(_2\) kombiniert, verglichen.
Wir konnten erstmals zeigen, dass PGE\(_2\) nicht nur einen Einfluss auf die Ausreifung, Zytokinproduktion und somit Migrationsfähigkeit der DCs hat wie in der Literatur beschrieben, sondern auch auf die Kreuzpräsentationsfähigkeit exogener Antigene.
Das Weglassen von PGE\(_2\) im Cocktail 2 führte zu einer signifikant besseren Kreuzpräsentationsfähigkeit. Somit lässt sich durch unsere Experimente auf eine hemmende Wirkung von PGE\(_2\) auf die T-Zell-Aktivierung über die Kreuzpräsentation schließen.
Als mögliche Schlussfolgerung unserer Experimente sollte bei der Arbeit mit Antigenen, welche über Kreuzpräsentation eine T-Zell-Antwort auslösen (Proteine, Tumorlysat, long-peptides) auf die Zugabe von PGE\(_2\) im Ausreifungscocktail verzichtet werden.

This study has identified cofilin, an actin binding protein, as a control element in the reorganization of the actin cytoskeleton which is highly relevant for T lymphocyte activation. Cofilin is regulated in its activity by reversible phosphorylation which is inducible by stimulation through accessory receptors such as CD2 and CD28. First it could be demonstrated that accessory receptor triggering induces the transient association of cofilin with the actin cytoskeleton and that only the dephosphorylated form of cofilin possesses the capacity to bind cytoskeletal actin in vivo. PI3-kinase inhibitors block both the dephosphorylation of cofilin and its association with the actin cytoskeleton. Importantly, cofilin, actin, PI3-kinase and one of its substrates, namely phosphatidylinositol 4,5-bisphosphate (PtdIns(4,5)P2) which can bind to cofilin, co-localize within CD2-receptor caps. The cofilin/F-actin interaction has been identified as a crucial regulatory element for receptor cap formation and the strength of signal transduction. To this end, appropriately designed cell permeable non-toxic peptides that are homologous to actin binding motifs of the human cofilin sequence were introduced into untransformed human peripheral blood T lymphocytes. These peptides competitively and dose dependently inhibit the activation induced interaction of cofilin with the actin cytoskeleton in vivo. By this approach it was possible to study, for the first time, the functional consequences of this interaction in immunocompetent T cells. The present data demonstrate that inhibition of the actin/cofilin interaction in human T lymphocytes by means of these cofilin derived peptides abolishes receptor cap formation and strongly modulates functional T cell responses such as T cell proliferation, interleukin-2 production, cell surface expression of CD69, gIFN production, and CD95L expression. Importantly, receptor independent activation by PMA and calcium ionophore circumvents these peptide produced inhibitory effects on lymphocyte stimulation and places the cofilin/actin interaction to a proximal step in the cascade of signaling events following T cell activation via surface signals. The present results are novel since as yet no information existed regarding the molecular elements which link cell surface receptor stimulation directly to the resulting reorganization of the actin cytoskeleton.
