Die vorliegende Arbeit behandelt die Synthese sowie die Eigenschaften einer Serie von organofunktionellen α-, β-, γ- und ζ-Silanen, mit einem Fokus auf Alkoxy(aminoalkyl)silanen. Der Großteil dieser Modellstrukturen wurde anschließend hinsichtlich ihrer Hydrolysekinetik in Abhängigkeit der Art der funktionellen Gruppe X (NMe3+, N(H)COOMe, N(Me)COOMe, NH2, N(H)Me, NMe2, Pip, Me), des Abstandes des Substituenten X zu dem Silicium-Atom
(α-, β-, γ- und ζ-Position), der Alkoxy-Abgangsgruppe am Silicium-Atom (MeO, iPrO, tBuO) und des pD-Wertes der Reaktionslösung systematisch untersucht.
Eine große Herausforderung dieser Studie war die Synthese von β-Amino-funktionalisierten Alkoxysilanen, deren Chemie aufgrund ihrer Labilität bisher kaum erforscht ist. Die einzigen literaturbekannten Vertreter stellten bislang das Trialkoxysilan (EtO)3Si(CH2)2NH2 (1) und sein Dialkoxy-Derivat (EtO)2SiMe(CH2)2NH2 (2) dar, welche durch Reaktion des entsprechenden 2-(Chlorethyl)silans mit Ammoniak unter hohem Druck im Autoklaven zugänglich sind. Unter Verwendung dieser Synthesemethode konnte sowohl die Synthese der Silane 1 und 2 reproduziert, als auch das Trimethoxy-Analogon (MeO)3Si(CH2)2NH2 (3) erstmals dargestellt werden.
Darüber hinaus wurde eine Serie von organofunktionellen Monoalkoxysilanen des Typs RORSiMe(CH2)2X und ROSiMe2C(H)MeCH2X (4b–18b) im präparativen Maßstab analyserein dargestellt.
Des Weiteren wurden die entsprechenden α-Silane 8a, 11a, 14a und 15a, die γ-Silane 6c, 8c, 11c, 13c–15c und 18c sowie die ζ-Silane 19 und 20 erstmals dargestellt.
Weiterhin wurden die bereits literaturbekannten α-Silane 16a–18a und γ-Silane 7c, 16c und 17c für die Verwendung in den Hydrolyseexperimenten synthetisiert.
Die Charakterisierung aller im Rahmen dieser Arbeit synthetisierten Verbindungen erfolgte mittels NMR-Spektroskopie (1H-, 13C-, 15N- und 29Si-NMR) und Elementaranalysen (C, H, N) bzw. HRMS-Experimente.
Die hydrolytische Spaltung der Si–OC-Bindung in Alkoxy(aminoalkyl)silanen stellt einen technisch sehr wichtigen Schlüsselschritt in der Synthese von Amino-funktionalisierten Polysiloxanen dar. Um den Mechanismus dieser Si–OC-Bindungsspaltung besser zu verstehen, wurden die Alkoxysilane 4b, 4c, 5b, 6b, 6c, 7b, 7c, 8a–8c, 9b, 11a–11c, 12b, 14a–14c,
15a–15c, 16a–16c, 17a–17c, 18a–18c, 19 und 20 hinsichtlich ihrer Hydrolysekinetik in CD3CN/D2O unter sauren und basischen Bedingungen mittels 1H-NMR-Spektroskopie untersucht.
Die Ergebnisse dieser Struktur–Reaktivitäts-Studie zeigten, dass die beobachteten unterschiedlichen Hydrolysegeschwindigkeiten das Resultat mehrerer Faktoren sind, wie beispielsweise elektronische und sterische Effekte, der große Einflusses des pD-Wertes und auch intramolekulare N–H∙∙∙O-Wasserstoffbrückenbindungen zwischen der protonierten Amino-Gruppe und der Alkoxy-Abgangsgruppe. Da der Einfluss dieser Effekte auf die Reaktivität der untersuchten α-, β-, γ- und ζ-Silane sehr unterschiedlich ist, kann kein klarer Zusammenhang zwischen der Hydrolysereaktivität und der Positionierung der stickstoff-haltigen funktionellen Gruppe (α-, β-, γ- und ζ-Position) erkannt werden. Die jeweils beobachtete Reaktivität entspricht vielmehr einer Summe aller zuvor genannten Teileffekte.
Die Erkenntnisse, die im Rahmen dieser Arbeit erhalten wurden, ermöglichen ein verbessertes grundlegendes Verständnis der Reaktivität von funktionalisierten α-, β-, γ- und ζ-Silanen, und sind für die Silicon-Industrie von großem Interesse, da sie eine gezieltere Anwendung der α-, β- und γ-Aminosilane in der Synthese von technisch wichtigen Amino-funktionalisierten Polysiloxanen erlauben.
The Venus flytrap, \textit{Dionaea muscipula}, with its carnivorous life-style and its highly
specialized snap-traps has fascinated biologist since the days of Charles Darwin. The
goal of the \textit{D. muscipula} genome project is to gain comprehensive insights into the
genomic landscape of this remarkable plant.
The genome of the diploid Venus flytrap with an estimated size between 2.6 Gbp to
3.0 Gbp is comparatively large and comprises more than 70 % of repetitive regions.
Sequencing and assembly of genomes of this scale are even with state-of-the-art
technology and software challenging. Initial sequencing and assembly of the genome
was performed by the BGI (Beijing Genomics Institute) in 2011 resulting in a 3.7 Gbp
draft assembly. I started my work with thorough assessment of the delivered assembly
and data. My analysis showed that the BGI assembly is highly fragmented and
at the same time artificially inflated due to overassembly of repetitive sequences.
Furthermore, it only comprises about on third of the expected genes in full-length,
rendering it inadequate for downstream analysis.
In the following I sought to optimize the sequencing and assembly strategy to obtain
an assembly of higher completeness and contiguity by improving data quality and
assembly procedure and by developing tailored bioinformatics tools. Issues with
technical biases and high levels of heterogeneity in the original data set were solved
by sequencing additional short read libraries from high quality non-polymorphic DNA
samples. To address contiguity and heterozygosity I examined numerous alternative
assembly software packages and strategies and eventually identified ALLPATHS-LG
as the most suited program for assembling the data at hand. Moreover, by utilizing
digital normalization to reduce repetitive reads, I was able to substantially reduce
computational demands while at the same time significantly increasing contiguity of
the assembly.
To improve repeat resolution and scaffolding, I started to explore the novel PacBio
long read sequencing technology. Raw PacBio reads exhibit high error rates of 15 %
impeding their use for assembly. To overcome this issue, I developed the PacBio
hybrid correction pipeline proovread (Hackl et al., 2014). proovread uses high
coverage Illumina read data in an iterative mapping-based consensus procedure to
identify and remove errors present in raw PacBio reads. In terms of sensitivity and
accuracy, proovread outperforms existing software. In contrast to other correction
programs, which are incapable of handling data sets of the size of D. muscipula
project, proovread’s flexible design allows for the efficient distribution of work load on high-performance computing clusters, thus enabling the correction of the Venus
flytrap PacBio data set.
Next to the assembly process itself, also the assessment of the large de novo draft
assemblies, particularly with respect to coverage by available sequencing data, is
difficult. While typical evaluation procedures rely on computationally extensive
mapping approaches, I developed and implemented a set of tools that utilize k-mer
coverage and derived values to efficiently compute coverage landscapes of large-scale
assemblies and in addition allow for automated visualization of the of the obtained
information in comprehensive plots.
Using the developed tools to analyze preliminary assemblies and by combining my
findings regarding optimizations of the assembly process, I was ultimately able to
generate a high quality draft assembly for D. muscipula. I further refined the assembly
by removal of redundant contigs resulting from separate assembly of heterozygous
regions and additional scaffolding and gapclosing using corrected PacBio data. The
final draft assembly comprises 86 × 10 3 scaffolds and has a total size of 1.45 Gbp.
The difference to the estimated genomes size is well explained by collapsed repeats.
At the same time, the assembly exhibits high fractions full-length gene models,
corroborating the interpretation that the obtained draft assembly provides a complete
and comprehensive reference for further exploration of the fascinating biology of the
Venus flytrap.
„Perspective-taking“ is the ability to put yourself into the place of somebody else. Psychological research distinguishes three kinds of perspective-taking, namely, perceptual (visuo-spatial), affective (empathy), and cognitive (theory of mind) perspective-taking. The last two kinds of perspective-taking are often summarized as “psychological perspective-taking”. This dissertation tackles the question of whether these three kinds of perspective-taking should be conceptualized as independent constructs or as facets of one and the same construct.
Prior research findings concerning this are equivocal. While some authors consider correlations between the different kinds of perspective-taking as too low for a unitary construct, others interpret correlations of the same magnitude as evidence for this. A less arbitrary way of deciding this would be to identify common mechanisms that underlie all kinds of perspective-taking and to examine whether manipulating these mechanisms in psychological experiments affects measures of perceptual, affective, and cognitive perspective-taking in parallel.
In accordance with this reasoning, the present dissertation assumes that the mental self-rotation of the body schema into the physical location of another person, the main mechanism of perceptual perspective-taking, is a common mechanism of all kinds of perspective-taking. Thus, contrary to previous research a unitary construct is not only justified on the grounds of a common central functionality of all kinds of perspective-taking, that is, overcoming one’s egocentrism in favor of an alternative (perceptual, affective or cognitive) point of view, but additionally on the grounds of a common psychological mechanism. From this, the simple hypothesis that inducing visuo-spatial perspective-taking also leads to psychological consequences is derived. This hypothesis was tested in 6 experiments.
In these experiments, participants first had to adopt the visual perspective of another person. To this end, they saw a person sitting at a table with two objects. During every trial, participants had to decide which hand the person would have to use in order to grab one of the two objects. Furthermore, the angular disparity between the participant and the target was manipulated in such a way that during half of the trials the target person was within the same visuo-spatial reference frame as the participant and thus no perspective-taking was necessary to solve the task correctly. During the remaining trials, the target person was sitting in another visuo-spatial reference frame so that the participants had to engage in perspective-taking to solve the task correctly. After every such trial, the target person was imbued with a mental state. This was done using an adapted paradigm for the investigation of the anchoring heuristic. Specifically, participants were asked to answer a trivia question and also saw what the target person from the visuo-spatial perspective-taking task was guessing.
In line with the hypothesis that visuo-spatial perspective-taking leads to psychological outcomes, too, it was found that participants adopted the thoughts of the target person more strongly after visuo-spatial perspective-taking. This was evident in the absolute size of the anchoring effect, as well as the differences between participant and target estimations. Further experiments ruled out sample and stimulus characteristics and task difficulty as alternative explanations for these effects. The last two experiments furthermore established that the effects were specific to constellations where an embodied self-rotation into the target’s perspective was necessary and that the adoption of the target’s thoughts was associated with feelings of similarity.
Taken together, these findings support the theoretically elaborated unitary view of perspective-taking and furthermore distinguish this construct from other related phenomena. In the general discussion, the significance of these findings for research on empathy, theory of mind, and perspective-taking, as well as practical implications are discussed.
The most energetic versions of active galactic nuclei (AGNs) feature two highly-relativistic plasma outflows, so-called jets, that are created in the vicinity of the central supermassive black hole and evolve in opposite directions. In blazars, which dominate the extragalactic gamma-ray sky, the jets are aligned close to the observer's line of sight leading to strong relativistic beaming effects of the jet emission. Radio observations especially using very long baseline interferometry (VLBI) provide the best way to gain direct information on the intrinsic properties of jets down to sub-parsec scales, close to their formation region.
In this thesis, I focus on the properties of three AGNs, IC 310, PKS 2004-447, and 3C 111 that belong to the small non-blazar population of gamma-ray-loud AGNs. In these kinds of AGNs, the jets are less strongly aligned with respect to the observer than in blazars. I study them in detail with a variety of radio astronomical instruments with respect to their high-energy emission and in the context of the large samples in the monitoring programmes MOJAVE and TANAMI. My analysis of radio interferometric observations and flux density monitoring data reveal very different characteristics of the jet emission in these sources. The work presented in this thesis illustrates the diversity of the radio properties of gamma-ray-loud AGNs that do not belong to the dominating class of blazars.
Attention-Deficit/Hyperactivity Disorder (ADHD) is characterized by symptoms of inattentiveness and hyperactivity/impulsivity. Besides, increasing evidence points to ADHD patients showing emotional dysfunctions and concomitant problems in social life. However, systematic research on emotional dysfunctions in ADHD is still rare, and to date most studies lack conceptual differentiation between emotion processing and emotion regulation. The aim of this thesis was to systematically investigate emotion processing and emotion regulation in adult ADHD in a virtual reality paradigm implementing social interaction. Emotional reactions were assessed on experiential, physiological, and behavioral levels.
Experiment 1 was conducted to develop a virtual penalty kicking paradigm implying social feedback and to test it in a healthy sample. This paradigm should then be applied in ADHD patients later on. Pleasant and unpleasant trials in this paradigm consisted of hits respectively misses and subsequent feedback from a virtual coach. In neutral trials, participants were teleported to different spots of the virtual stadium. Results indicated increased positive affectivity (higher valence and arousal ratings, higher zygomaticus activations, and higher expression rates of positive emotional behavior) in response to pleasant compared to neutral trials. Reactions to unpleasant trials were contradictory, indicating increased levels of both positive and negative affectivity, compared to neutral trials. Unpleasant vs. neutral trials revealed lower valence ratings, higher arousal ratings, higher zygomaticus activations, slightly lower corrugator activations, and higher expression rates of both positive and negative emotional behavior. The intensity of emotional reactions correlated with experienced presence in the virtual reality.
To better understand the impact of hits or misses per se vs. hits or misses with coach feedback healthy participants’ emotional reactions, only 50% of all shots were followed by coach feedback in experiment 2. Neutral trials consisted of shots over the free soccer field which were followed by coach feedback in 50 % of all trials. Shots and feedback evoked more extreme valence and arousal ratings, higher zygomaticus activations, lower corrugator activations, and higher skin conductance responses than shots alone across emotional conditions. Again, results speak for the induction of positive emotions in pleasant trials whereas the induction of negative emotions in unpleasant trials seems ambiguous. Technical improvements of the virtual reality were reflected in higher presence ratings than in experiment 1.
Experiment 3 investigated emotional reactions of adult ADHD patients and healthy controls after emotion processing and response-focused emotion regulation. Participants successively
went through an ostensible online ball-tossing game (cyber ball) inducing negative emotions, and an adapted version of the virtual penalty kicking game. Throughout cyber ball, participants were included or ostracized by two other players in different experimental blocks. Participants were instructed to explicitly show, not regulate, or hide their emotions in different experimental blocks. Results provided some evidence for deficient processing of positive emotions in ADHD. Patients reported slightly lower positive affect than controls during cyber ball, gave lower valence ratings than controls in response to pleasant penalty kicking trials, and showed lower zygomaticus activations than controls especially during penalty kicking. Patients in comparison with controls showed slightly increased processing of unpleasant events during cyber ball (higher ratings of negative affect, especially in response to ostracism), but not during penalty kicking. Patients showed lower baseline skin conductance levels than controls, and impaired skin conductance modulations. Compared to controls, patients showed slight over-expression of positive as well as negative emotional behavior. Emotion regulation analyses revealed no major difficulties of ADHD vs. controls in altering their emotional reactions through deliberate response modulation. Moreover, patients reported to habitually apply adaptive emotion regulation strategies even more frequently than controls. The analyses of genetic high-risk vs. low-risk groups for ADHD across the whole sample revealed similar results as analyses for patients vs. controls for zygomaticus modulations during emotion processing, and for modulations of emotional reactions due to emotion regulation.
To sum up, the virtual penalty kicking paradigm proved to be successful for the induction of positive, but not negative emotions. The importance of presence in virtual reality for the intensity of induced emotions could be replicated. ADHD patients showed impaired processing of primarily positive emotions. Aberrations in negative emotional responding were less clear and need further investigation. Results point to adult ADHD in comparison to healthy controls suffering from baseline deficits in autonomic arousal and deficits in arousal modulation. Deficits of ADHD in the deliberate application of response-focused emotion regulation could not be found.
Die begrenzte Reichweite ist einer der Hauptgründe für das derzeitige mangelnde Kaufinteresse an Elektrofahrzeugen. Neben rein komponentenoptimierenden Maßnahmen, wie der Verbesserung der Batterie, ist die Förderung von Eco-Driving, also einer energieeffizienten Fahrweise, ein effektiver Ansatz zur Steigerung der Reichweite. Trainings und visuell dargebotene Eco-Assistenten können Eco-Driving wirksam steigern, schöpfen aber nicht dessen gesamtes Potential aus. Angepasste Pedal- und Motorkennlinien könnten Eco-Driving zusätzlich fördern. Für deren Bewertung sind die Wirksamkeit und Akzeptanz bisher nicht gemeinsam berücksichtig worden oder sie wurden nicht im Elektrofahrzeug evaluiert und validiert. Zu diesen Anpassungen zählen eine Veränderung des Beschleunigungspedals, sodass mit diesem gleichzeitig beschleunigt und rekuperiert werden kann, die Limitierung von Drehmoment und Leistung und der Einsatz eines aktiven Beschleunigungspedals, welches Widerstände abhängig von Fahrzeug- oder Situationsparametern aktiviert.
Für diese Arbeit habe ich daher die Pedal- und Motorkennlinien entsprechend angepasst und in ein validiertes Elektroautomodell implementiert. Ziel war es, verschiedene Fahrverhaltensbereiche im Elektrofahrzeug, die Eco-Driving kennzeichnen (energieoptimales Beschleunigen und Verzögern, Einhalten von Geschwindigkeitsbegrenzungen, vorausschauendes Fahren), benutzerfreundlich, akzeptabel und wirksam zu unterstützen. Zu diesem Zweck habe ich vier Probandenstudien im bewegten Fahrsimulator durchgeführt und geeignete Pedal- und Motorkennlinien empirisch bestimmt. In der ersten Studie habe ich untersucht, ob und warum eine Pedalkennlinie zu bevorzugen ist, bei der mit dem Beschleunigungspedal anstelle des Bremspedals rekuperiert wird. Das Ziel der zweiten Studie war es, eine geeignete Rekuperationsstärke für ein kombiniertes Beschleunigungspedal, bei dem mit dem Beschleunigungspedal rekuperiert wird, zu bestimmen. In der dritten Studie habe ich evaluiert, ob die Limitierung der Leistung oder die des Drehmoments zu bevorzugen ist, um das Beschleunigungsverhalten zu optimieren und wie stark die Limitierungen optimaler Weise sein sollten. Basierend auf den Ergebnissen der dritten Studie, habe ich schließlich einen optimierten Limitierungsansatz konzipiert, diesen mit einem aktiven Beschleunigungspedal verglichen und bestimmt, welcher Ansatz zu bevorzugen ist.
Aufgrund der Studienergebnisse werden folgende Ansätze für die jeweiligen Eco-Driving-Fahrverhaltensbereiche empfohlen und es werden folgende Gestaltungsempfehlungen abgeleitet: Zur Förderung eines energieeffizienten Beschleunigungsverhaltens ist die Limitierung von Drehmoment und Leistung geeignet. Die Limitierung des Drehmoments ist hierbei besonders wirksam in geringen, die Limitierung der maximalen Leistung in höheren Geschwindigkeitsbereichen. Zu empfehlen sind parallele mittelstarke Limitierungen von maximalem Drehmoment und maximaler Leistung, die Beschleunigungen mit 2.0 m/s² erlauben, bei gleichzeitiger Bereitstellung eines Kick-Downs. Ein aktives Beschleunigungspedal ist insbesondere aus Gründen der Benutzerfreundlichkeit zur Förderung eines energieeffizienten Beschleunigungsverhaltens nur eingeschränkt empfehlenswert.
Zur Förderung eines energieeffizienten Verzögerungsverhaltens wird die Implementierung der Rekuperationsfunktion auf dem Beschleunigungspedal anstelle des Bremspedals empfohlen, da dies einerseits ermöglicht, auf hydraulisches Bremsen zu verzichten und gleichzeitig mehr Energie rekuperiert werden kann. Ersteres trägt zu einer hohen Akzeptanz bei, letzteres zu einer günstigen Energiebilanz. Besonders effektiv und akzeptabel ist ein kombiniertes Fahrbremspedal, wenn es eine starke Rekuperation ermöglicht (zwischen -1.7 und -2.1 m/s²). Weiterhin ist ein aktives Beschleunigungspedal, das den geeigneten Zeitpunkt für eine maximal energieeffiziente Verzögerung mit einem kombinierten Fahrbremspedal anzeigt, wirksam, um die rekuperierte Energie zu steigern. Auf diese Weise kann zudem eine vorausschauende Fahrweise unterstützt werden. Hierbei muss jedoch die Funktionalität und Benutzerfreundlichkeit optimiert werden, um eine gesteigerte kognitive Fahrerbeanspruchung und Minderungen der Akzeptanz zu vermeiden. Zur Unterstützung der Einhaltung von Geschwindigkeitsbegrenzungen ist ebenfalls das aktive Beschleunigungspedal geeignet. Der Fahrer sollte hierbei aber die Möglichkeit haben, individuell Grenzwerte einzustellen.
Die Verknüpfung eines kombinierten Fahrbremspedals mit einer Limitierung von Drehmoment und Leistung sowie einem aktiven Beschleunigungspedal kann abschließend, unter Berücksichtigung der abgeleiteten Gestaltungsempfehlungen, als effektive und akzeptable Möglichkeit zur Förderung unterschiedlicher Verhaltensbereiche von Eco-Driving bewertet werden. Die erwarteten Synergieeffekte der evaluierten Ansätze in Verbindung mit Eco-Trainings und visuell dargebotenen Eco-Feedback-Assistenten sowie deren Langfristigkeit sollten Gegenstand weiterführender Forschung sein.
Die postovulatorische Alterung sowie die ovarielle Alterung konnten bei der Anwendung assistierter Reproduktionstechniken (ARTs) als entscheidende Faktoren identifiziert werden, die den Reproduktionserfolg nachhaltig beeinträchtigen. Die postovulatorische Alterung tritt ein, sobald die reife Eizelle nicht mehr innerhalb ihres physiologischen Zeitfensters befruchtet wird. Die ovarielle Alterung beschreibt hingegen die Abnahme des Follikel-Vorrats mit zunehmendem Alter des weiblichen Individuums bzw. des Ovars. Sowohl die postovulatorische Alterung als auch die ovarielle Alterung führen u.a. zu einer reduzierten Oozytenqualität und einer geringeren Blastozystenrate. Die Zielsetzung dieser Arbeit bestand darin, den Einfluss der postovulatorischen Alterung und der ovariellen Alterung im Holstein-Rind (Bos taurus) auf die DNA-Methylierung entwicklungsrelevanter Gene in Eizellen und Embryonen zu untersuchen. Aus Schlachthof-Ovarien wurden Antralfollikeln unterschiedlicher Größe (<2 mm, 3-5 mm und >6 mm) isoliert. Eizellen aus Follikeln der Größe 3-5 mm wurden für 24h (physiologisch) und 48h (gealtert) in vitro gereift (IVM). Die gereiften Oozyten wurden anschließend in vitro fertilisiert und Embryonen im 4-6 Zellstadium generiert. Sowohl in den unreifen Eizellen aus Antralfollikeln unterschiedlicher Größe als auch in den gereiften Oozyten und den Embryonen wurde die Promotormethylierung der Gene bH19, bSNRPN, bZAR1, bDNMT3A, bOCT4, bDNMT3Lo und bDNMT3Ls analysiert. Zur Untersuchung der ovariellen Alterung wurden mittelgroßen Antralfollikel aus Ovarien lebender Rinder (in vivo) unterschiedlichen Alters (9-12 Monate, 3-7 Jahre und 8-11 Jahre) gewonnen. In den daraus isolierten unreifen Eizellen wurde die DNA-Methylierung der Promotorregionen der Gene bTERF2, bREC8, bBCL-XL, bPISD, bBUB1, bDNMT3Lo, bH19 und bSNRPN bestimmt. Als Methode zur Analyse der Promotormethylierung wurde die Limiting Dilution Bisulfit-Sequenzierung angewendet.
In unreifen Eizellen aus Antralfollikeln unterschiedlicher Größe (<2 mm, 3-5 mm und >6 mm) konnte ein erhöhtes Auftreten abnormal methylierter Allele in den geprägten Genen bH19 und bSNRPN von Eizellen kleiner Follikel (<2 mm) identifiziert werden. Dieses Ergebnis könnte eine mögliche Ursache einer bereits bekannten und mehrfach beschriebenen geringeren Entwicklungskompetenz von Eizellen kleiner Follikel (<2 mm) auf epigenetischer Ebene darstellen.
Die verlängerte Reifungsdauer der IVM-Eizellen hatte eine signifikante Hypermethylierung in der Promotorregion des Gens DNMT3Lo von 48h-gereiften Eizellen zur Folge. Beim Übergang von 48h-gereiften Eizellen zum Embryo konnte eine signifikante Hypomethylierung von CpG7 des stammzellspezifischen Transkripts DNMT3Ls beobachtet werden. Diese CpG-Stelle wies ebenfalls einen signifikanten Anstieg von CpGs mit nicht-eindeutigem Methylierungszustand in unreifen Eizellen mit steigender Follikelgröße auf. Da sich die CpG-Position innerhalb eines Sequenz-Motivs einer Bindungsstelle des Transkriptionsfaktors CREB befindet, könnten die Methylierungsdaten auf eine Interaktion zwischen dem Transkriptionsfaktor CREB und der DNA-Methylierung während der Entwicklung und Reifung der Eizelle sowie der Transition von der Eizelle zum Embryo hindeuten.
Die DNA-Methylierungsprofile der untersuchten Gene in unreifen Eizellen aus Kühen unterschiedlichen Alters (9-12 Monate, 3-7 Jahre und 8-11 Jahre) wiesen keine signifikanten Unterschiede zwischen den Altersgruppen auf. Die ovarielle Alterung bei Rindern zwischen 9 Monaten und 11 Jahren zeigte damit keinen Effekt auf die DNA-Methylierung der untersuchten Promotorregionen der Gene bTERF2, bREC8, bBCL-XL, bPISD, bBUB1, bDNMT3Lo, bH19 und bSNRPN.
Nach einer simulierten postovulatorischen Alterung durch eine in vitro Reifung für 48h konnte eine Veränderung der DNA-Methylierung der Oozyten-spezifischen (DNMT3Lo) und Stammzell-spezifischen (DNMT3Ls) Promotoren des katalytisch inaktiven Cofaktors von DNMT3A, DNMT3L, beobachtet werden. Die veränderte DNA-Methylierung von DNMT3Ls tritt dabei erst im frühen Embryo in Erscheinung und interagiert vermutlich mit dem Transkriptionsfaktor CREB. Die Veränderungen von DNMT3Lo in Eizellen und DNMT3Ls in den daraus generierten Embryonen lässt vermuten, dass es sich hierbei um eine dynamische Anpassung des Embryos auf äußere Umweltbedingungen der Eizelle über die Methylierung der DNA handelt.
11 Conclusion
11.1 Glaze compositions
Glazes from tiles of imposing Islamic buildings and some tableware glazes of the medieval epoch in Central Asia, the Middle East, Asia Minor, and North Africa are analysed regarding their main composition and colouring agents. Three major production recipes can be distinguished, i.e. alkali glazes, alkali lead glazes, and lead glazes. In the work of Tite (2011), Islamic glazes from Egypt, Iran, Iraq, and Syria are subdivided into four groups of composition, being partly consistent with those of this work. The alkali lime glazes with <2 wt% PbO correspond to the alkali glazes, but with higher content of CaO. The second and third group of low lead alkali and lead alkali glazes (2-10 wt% PbO and 10-35 wt% PbO) can be subsumed to the alkali lead group described here. Tite´s high lead group has PbO contents >35 wt% and is comparable to the lead glazes (>30 wt% PbO) of this study. The lead and the alkali oxides serve as a flux for the lowering the melting point.
In the interaction of ceramic body and glaze, primarily an influence from Si, Al, and K is observed in the line scans from the cross section of ceramic and glaze. However, the input of ceramic material doesn’t seem to be critical for the classification of glazes according to their alkali and alkali lead compositions.
In every epoch and locality, except of the Ilkhanate dynasty in Iran, lead glaze samples can be verified. This is also observed in previous investigations e.g. from medieval Iraq, Jordan and Iran (McCarthy, 1996; Al-Saad, 2002; Holakooei et al., 2014). In the Moroccan and Bulgarian glazes, lead seems to be the only important flux. In part, the lead flux is supplemented by additional alkali contents. The lack of alkali and alkali lead glazes in Bulgarian and Moroccan glazes (assuming that the Ottoman alkali lead glazes are imported tableware) seems to affect the regions with Roman-influenced history and with geographical distance to the Near East alkali flux tradition.
For the alkali lead glazes and alkali glazes, the overall characteristic is sodium dominated, although the absolute soda values are in part surprisingly low. Samples from Bukhara, Takht-i-Suleiman and the Turkish localities have the highest, but still moderate Na2O values up to 15 wt%, compared to other analyses from e.g. India (Gill & Rehren, 2011).
The source of the alkali flux is either mineral natron or plant ash. The source can be determined regarding the MgO values, limited to 1.3 wt% in mineral natron and exceeding 2.0 wt% in the case of plant ashes. In the samples of the present study, the K2O component is not suitable for the indication of the flux-relevant alkali source due to its broad scattering. The P2O5 contents are also enhanced in the plant ash compositions but the data set is not sufficient for statistical evaluation. An influence of the ceramic body on the glaze composition is observed only for SiO2, Al2O3, and K2O in quartz frit ceramics with slight K-feldspar content.
The earliest Uzbek tableware glazes from the 10th-11th century (Seljuq period) were generally produced using a lead flux. The same applies to part of the Uzbek tile glazes which were produced between the 13th and 16th century. In Iran, glazes from the 12th century (Khwarezmid period) are lead glazes, but also alkali-fluxed glazes with mineral natron characteristics can be found. Although the production of lead-rich glazes was established from the 8th-9th century on in Iraq, Syria, and Egypt (Henshaw, 2010; Tite et al., 2011), alkali glazes are found in almost all regions except of Bulgaria and Morocco.
Plant ash-fluxed alkali glazes are found in 13th century glazes from Takht-i-Suleiman. The plant ash flux technology is assumed to be continuously used in Mesopotamia, Iran, and Central Asia (Sayre & Smith, 1974; Henderson, 2009), but it could be shown that a parallel use of mineral natron parallel existed in the alkali glaze production from the 12th-15th century from Uzbekistan to Afghanistan. Mineral natron characteristics are also reported by Mason (2004) for Syrian and Iranian alkali glazes on lustre ware of the 8th-14th century. Tile glazes with partly mineral natron compositions are found in the Mughal architectural glazes from the 14th- 17th century from India (Gill et al., 2014).
Alkali and alkali lead tile glazes from Samarkand from the 13th century (Mongolian period) have mineral natron flux characteristics, but samples from the 15th century (Timurid period) show plant ash signature. Alkali fluxed Uzbek glazes from Bukhara from the 16th century (Sheibanid dynasty) are also made by plant ash flux and are subdivided into two groups with high and low sodium oxide content. The Afghan alkali glazes have sodium oxide contents similar to the sodium-poor Uzbek subgroup, which points to a possible exchange of glaze makers or glaze making technology from Uzbekistan and Afghanistan in the 15th-17th century. Regarding the extensive exchange of Timurid craftsmen in Central Asia, this option seems to be even more likely (Golombek, 1996). One sample from the 15th century from Afghanistan with mineral natron reveals that this material was parallel used in these centuries.
Concerning the colouring of the glazes, it has to be distinguished between pigments and colouring ions which are incorporated in the glassy matrix. The colouring agents for translucent glazes are cations of various transition metals. As ions, Co2+ (blue), Cu2+ (green in a lead rich matrix), Fe3+ (brown/black), Mn4+ (brown/black) and Mn3+ (violet) are determined by EPMA. For opaque yellow, white, and turquoise glazes, different pigments were used. The crystalline pigments are investigated by a µ XRD2 device with the result of SnO2, SiO2, and PbSiO4 as whitening agents. PbSiO4 and Pb2Sn2O6 are found in the yellow glaze, from which only the lead tin oxide causes the yellow colour. In the black glazes, different Cr-rich pigments, Cu-Cr-Mn-oxides and iron containing clinopyroxenes are found, even in samples of the same period and region. Cr-rich particles are also detected in two turquoise Afghan glazes from the 15th and 16th century. The use of the ions of Fe, Cu, Co, Cr, and Mn seems to be widely common in the Islamic glazes and corresponds to the described colouring agents in e.g. the study of Tite (2011). The use of opacifying SnO2 particles is widespread as it is reported from different Islamic glazes from Iraq, Iran, Egypt, and Syria (Henshaw, 2010; O´Kane, 2011; Tite, 2011). The colouring agents are known already from former, e.g. Egyptian, Roman and pre-islamic periods, but especially SnO2 pigments became increasingly widespread in the Islamic glazing tradition. The use of yellow and black pigments instead varies already within the buildings from Bukhara from Cr crystals and clino-pyroxenes in the mosque Khoja Zainuddin to a Cu-Cr-Mn-oxide in the madrassa Mir-i Arab of the same epoch.
Regarding the matrix compositions connected with the colouring, a certain assignment within the different locations and epochs can be seen. It is noticeable that e.g. the content of lead in turquoise glazes in Uzbekistan is in the range of 0.0-9.2 wt% Pb, whereas blue glazes are mostly alkali ones with PbO contents <2.0 wt%. The turquoise glazes show, that this restriction is not influenced by any defaults of availability and processability. The assumption of common addition of lead and tin to the glaze, which is already described for Iranian glazes of the 13th century (Allan et al., 1973) cannot be confirmed by correlations of tin and lead oxide in the compositions.
11.2 Portable XRF measurement
With the p-XRF, semi-quantitative information about the major element compositions is generated. The depth of the detectable signals depends on the analysed sample setup. The p-XRF data are collected with the XL3 Hybrid device of the company Analyticon Instruments. In the comparison of p-XRF results of the “mining” program from Uzbek glazes with EPMA results, the same major composition groups can be distinguished. The Moroccan glazes, all lead rich, are measured with the “mining” as well as with the “soil” program, revealing a better performance in the “mining” measurements. The deviations are nevertheless high, because of the high lead contents, which make the calculation of matrix correction difficult.
The measurement of the colouring oxides MnO2, CoO, and CuO is satisfying with the internal calibration of the device and even improved with the “mining” program measurement, if compared to the results of the “soil” program. The measurements of glaze imitations lead to better results than that of bulk glass. This can be attributed to the smoother surface texture.
In spite of the accuracy limits in the measurements of particular elements in glazes, the classification of flux composition into three groups could be confirmed with the p XRF analysis. The measurement precision is therefore sufficient for the semi-quantitative analysis of the flux characteristic of glazes. Especially for the on-site measurement of large sample quantities on historical buildings, the device is a suitable tool.
11.3 Restoration material
The ORMOCER® fulfils the requirements of stability, reversibility, and transparency, which are imposed to a modern restoration material. As pigments, historically coloured glass, cobalt blue, Egyptian blue, lead tin yellow, manganese violet, iron oxide, copper oxide, and cassiterite were used. The metal compounds have higher colour intensities than the pigments of coloured glass. It has to be considered that the proportion of ORMOCER® in the batch must be high enough (70 vol%) to guarantee the ORMOCER® properties of weathering and mechanical stability. The adhesion properties of the ORMOCER® and the homogeneity of the mixture are the best in a fraction of max. 30 vol% particles per ORMOCER®.
With integrated particles, the ORMOCER® G materials show homogeneous coatings, whereas the particles in the ORMCOER® E show more agglomeration. In the sedimentation and weathering experiments, the use of an ultrasonic finger in combination with a roller mill is favourable compared to the treatment with bead grinding mill. The treatments with ultrasonic finger and roller mill result in less sedimentation and better adhesion of the dispersions. The treatment of the dispersions in the bead grinding mill does not result in sufficient adhesion, certainly due to the sedimentation behaviour and a congregation of particles on the bottom of the coating.
The modification of dispersed nano-particles by 3-methacryl-oxypropyltrimethoxysilan leads to a further homogenization in the sedimentation tests. It is therefore approved for the use in coloured glaze supplements. In weathered coatings of nano-particle compounds, the surface modification shows certainly no enhancement of stability.
The treatment of pigmented coatings with an additional layer of pure ORMOCER® results in a bright and transparent appearing, which is closer to the original optical appearance of the glaze. A long-time test application on a historical building will be the next step to validate the suitability of the restoration material.
Graphs are a frequently used tool to model relationships among entities. A graph is a binary relation between objects, that is, it consists of a set of objects (vertices) and a set of pairs of objects (edges).
Networks are common examples of modeling data as a graph. For example, relationships between persons in a social network, or network links between computers in a telecommunication network can be represented by a graph.
The clearest way to illustrate the modeled data is to visualize the graphs. The field of Graph Drawing deals with the problem of finding algorithms to automatically generate graph visualizations. The task is to find a "good" drawing, which can be measured by different criteria such as number of crossings between edges or the used area. In this thesis, we study Angular Schematization in Graph Drawing. By this, we mean drawings
with large angles (for example, between the edges at common vertices or at crossing points).
The thesis consists of three parts. First, we deal with the placement of boxes. Boxes are axis-parallel rectangles that can, for example, contain text.
They can be placed on a map to label important sites, or can be used to describe semantic relationships between words in a word network. In the second part of the thesis, we consider graph drawings visually guide the
viewer. These drawings generally induce large angles between edges that meet at a vertex. Furthermore, the edges are drawn crossing-free and in a way that
makes them easy to follow for the human eye. The third and final part is devoted to crossings with large angles. In drawings with crossings, it is important to have large angles between edges at their crossing point, preferably right angles.
The present thesis considers the development and analysis of arbitrary Lagrangian-Eulerian
discontinuous Galerkin (ALE-DG) methods with time-dependent approximation spaces for
conservation laws and the Hamilton-Jacobi equations.
Fundamentals about conservation laws, Hamilton-Jacobi equations and discontinuous Galerkin
methods are presented. In particular, issues in the development of discontinuous Galerkin (DG)
methods for the Hamilton-Jacobi equations are discussed.
The development of the ALE-DG methods based on the assumption that the distribution of
the grid points is explicitly given for an upcoming time level. This assumption allows to construct a time-dependent local affine linear mapping to a reference cell and a time-dependent
finite element test function space. In addition, a version of Reynolds’ transport theorem can be
proven.
For the fully-discrete ALE-DG method for nonlinear scalar conservation laws the geometric
conservation law and a local maximum principle are proven. Furthermore, conditions for slope
limiters are stated. These conditions ensure the total variation stability of the method. In addition, entropy stability is discussed. For the corresponding semi-discrete ALE-DG method,
error estimates are proven. If a piecewise $\mathcal{P}^{k}$ polynomial approximation space is used on the reference cell, the sub-optimal $\left(k+\frac{1}{2}\right)$ convergence for monotone fuxes and the optimal $(k+1)$ convergence for an upwind flux are proven in the $\mathrm{L}^{2}$-norm. The capability of the method is shown by numerical examples for nonlinear conservation laws.
Likewise, for the semi-discrete ALE-DG method for nonlinear Hamilton-Jacobi equations, error
estimates are proven. In the one dimensional case the optimal $\left(k+1\right)$ convergence and in the two dimensional case the sub-optimal $\left(k+\frac{1}{2}\right)$ convergence are proven in the $\mathrm{L}^{2}$-norm, if a piecewise $\mathcal{P}^{k}$ polynomial approximation space is used on the reference cell. For the fullydiscrete method, the geometric conservation is proven and for the piecewise constant forward Euler step the convergence of the method to the unique physical relevant solution is discussed.
Im Rahmen dieser Arbeit wurde ein Rastertunnelmikroskop (STM) für Messungen bei tiefen Temperaturen und hohen Magnetfeldern konzipiert und aufgebaut. Die Probentemperatur kann dabei auf bis zu 1.4\,Kelvin reduziert werden, was spektroskopische Messungen mit extrem hoher Energieauflösung ermöglicht. Die thermische Verbreiterung spektroskopischer Merkmale liegt somit im Bereich eines Milli-Elektronenvolts, wie durch den Fit der Bandlücke eines supraleitenden Materials demonstriert wird. Ein linearer Bewegungsmechanismus ermöglicht die Positionierung des STM-Körpers innerhalb einer supraleitenden Spule, in der Magnetfelder von bis zu 12.5\,Tesla senkrecht zur Probenoberfläche erzeugt werden können. Das System erlaubt des Weiteren den Wechsel von Spitzen und Proben innerhalb des Kryostaten sowie das Aufdampfen von Einzelatomen auf die kalte Probenoberfläche ohne die Probe aus dem STM zu entfernen. Um den Einfluss mechanischer Vibrationen zu minimieren wurde ein innovatives Feder-Dämpfungssystem entwickelt, dass eine Stabilität des Tunnelkontakts von bis zu einem Pikometer gewährleistet. \\
\noindent Der zweite Teil dieser Arbeit präsentiert die Ergebnisse von STM-Messungen auf Antimon-Tellurid (Sb_{2}Te_{3}). Sb_{2}Te_{3}\, gehört zur relativ neu entdeckten Materialklasse der Topologischen Isolatoren (TI). Diese Verbindungen besitzen auf ihren Oberflächen Zustände mit linearer Dispersion, die durch die Zeitumkehr-Invarianz geschützt werden. Fokus unserer Messungen ist dabei der Einfluss eines magnetischen Feldes auf die Eigenschaften eines derartigen unkonventionellen 2D-Elektronengases. Dazu wurde die Entstehung von Landau Level (LL) innerhalb eines Magnetfelds genau untersucht. Die zwei in dieser Arbeit untersuchten Hauptaspekte sind: \medskip
\noindent(i) Die energetische Verbreiterung, die Rückschlüsse auf die Lebensdauer zulässt\\
(ii) Die örtliche Fluktuation. \medskip
\noindent Erstaunlicherweise kann die gemessene Verbreiterung der Landau Resonanzen nicht mit gängigen Mechanismen der Lebenszeit-Verbreiterung erklärt werden. Aus diesem Grund wird eine alternative Interpretation basierend auf der Heissenbergschen Unschärferelation vorgestellt, die im guten Einklang mit den von uns gewonnenen Daten steht. Des Weiteren zeigen örtlich aufgelöste Messungen systematische Abweichungen in der Dirac-Geschwindigkeit positiver und negativer Landau Resonanzen. Diese Fluktuationen stehen dabei in direktem Zusammenhang mit Änderungen im lokalen chemischen Potential. Da die physikalischen Ursachen dieser Abweichung im Rahmen dieser Arbeit nicht zweifelsfrei geklärt werden konnten, werden im letzten Teil die zugrundeliegenden Messergebnisse vorgestellt und mögliche Erklärungen des Verhaltens präsentiert.
Aufklärung der molekularen Struktur und Funktion des R-Typ Anionenkanals QUAC1 in Schließzellen
(2016)
Zum Gasaustausch mit Ihrer Umgebung besitzen höhere Pflanzen stomatäre Komplexe. Die Turgor-getrieben Atmungsöffnungen in der Epidermis der Blätter werden von zwei Schließzellen umsäumt. Um bei Trockenheit einen exzessiven Verlust von Wasser zu verhindern, synthetisieren/importieren Schließzellen das Stresshormon ABA (Abszisinsäure), das über eine schnelle ABA-Signalkaskade plasmamembrangebundene Ionenkanäle steuert. Dabei wird der Stomaschluss durch die Aktivität von R-(rapid) und S-(slow)Typ Anionenkanälen initiiert. Obwohl die R- und S-Typ Anionenströme in Schließzellen seit Jahrzehnten bekannt waren, konnte erst kürzlich das Gen identifiziert werden, das für den S-Typ Anionenkanal (SLAC1, Slow activating Anion Channel 1) kodiert. Daraufhin wurde schnell der Zusammenhang zwischen dem Stresshormon ABA, der ABA-Signalkette und der Aktivität des SLAC1 Anionenkanals im heterologen Expressionssystem der X. laevis Oozyten als auch in Schließzellprotoplasten aufgeklärt. Es konnte gezeigt werden, dass ABA durch einen zytosolischen Rezeptor/Phosphatasekomplex (RCAR1/ABI1) erkannt wird und die Aktivität von kalziumabhängigen Kinasen (CPK-Familie) sowie kalziumunabhängigen Kinasen der SnRK2-Familie (OST1) steuert. In Anwesenheit von ABA phosphorylieren diese Kinasen SLAC1 und sorgen so für die Aktivierung von Anionenströmen und damit für die Initiierung des Stomaschlusses.
Die genetische Herkunft der ABA-induzierten R-Typ Ströme in Schließzellen war zu Beginn der vorliegenden Arbeit noch nicht bekannt. R-Typ Ströme zeichnen sich durch eine strikte Spannungsabhängigkeit und sehr schnellen Aktivierungs- sowie Deaktivierungskinetiken aus. Die Charakterisierung von Verlustmutanten des Schließzell-exprimierten Gens ALMT12 (Aluminium-aktivierter Malattransporter 12) konnte in Zusammenarbeit mit der Arbeitsgruppe Martinoia (Zürich) erste Hinweise auf die Beteiligung dieses Gens an der Stomabewegung demonstrieren. Anschließende Patch-Clamp Untersuchungen an Schließzellprotoplasten aus Wildtyppflanzen und ALMT12-Verlustmutanten zeigten, dass ALMT12 für die Malat-aktivierte R-Typ Anionenstromkomponente verantwortlich ist. Deshalb wurde der Anionenkanal QUAC1 (Quickly activating Anion Channel 1) benannt - in Anlehnung an die Benennung des Anionenkanals SLAC1. Mit der Identifizierung von QUAC1 in planta war es nun meine Aufgabe, die elektrischen Eigenschaften von ALMT12/QUAC1 und dessen Aktivitätskontrolle durch die ABA-Signalkaskade im heterologen Expressionssystem der Xenopus Oozyten zu untersuchen.
Protein-Protein Interaktionsstudien mit der Hilfe der Bimolekularen Fluoreszenz-Technik, sowie die Beobachtung von markant erhöhten QUAC1 Anionenströmen in Anwesenheit der SnRK2 Kinase OST1 und den Calcium-abhängigen Kinasen CPK2 und CPK20, ließen den Schluss zu, dass QUAC1, ebenso wie SLAC1, unter der Kontrolle des schnellen ABA-Signalwegs steht. Eine zusätzliche Expression des negativen Regulators ABI1 unterdrückte die aktivierenden Eigenschaften der QUAC1-aktivierenden Kinasen, was die Hypothese der Koregulation von S- und R-Typ Anionenkanälen durch die gleiche ABA-Signalkaskade weiter unterstützt.
Zur weiteren Aufklärung der elektrischen Eigenschaften von QUAC1 wurden tiefgreifende elektrophysiologische Untersuchungen mit der Zwei-Elektroden-Spannungsklemmen Technik durchgeführt. Durch die Wahl von geschickten Spannungsprotokollen konnte sowohl die schnelle Aktivierungskinetik als auch die schnelle Deaktivierungskinetik von QUAC1 bestimmt und quantifiziert werden. Diese Stromantworten waren sehr ähnlich zu den R-Typ Strömen, die man von Patch-Clamp Untersuchungen an Schließzellprotoplasten kannte, was ein weiteres Indiz dafür war, dass es sich bei QUAC1 tatsächlich um eine Komponente des R-Typ Kanals aus Schließzellen handelt. Weiterführende Untersuchungen bezüglich der Spannungsabhängigkeit und der Selektivität von QUAC1 charakterisierten das Protein als einen Depolarisations-aktivierten Anionenkanal mit einer starken Präferenz für Dicarbonsäuren wie Malat und Fumarat. Zudem konnte auch eine Leitfähigkeit für Sulfat und Chlorid nachgewiesen werden. Interessanterweise erwies sich Malat nicht nur als ein permeierendes Ion, sondern auch als ein regulierendes Ion, welches das spannungsabhängige Schalten von QUAC1 maßgeblich beeinflusst. Extrazelluläres Malat verschob die Offenwahrscheinlichkeit von QUAC1 sehr stark zu negativeren Membranspannungen, so dass der Anionenkanal bereits bei typischen Ruhespannungen von Schließzellen (ca. -150 mV) aktiviert werden konnte. Eine Beladung von QUAC1-exprimierender Oozyten mit Malat bewirkte zum einen höhere Anioneneffluxströme, aber auch eine Verschiebung der spannungsabhängigen Offenwahrscheinlichkeit zu negativeren Membranpotentialen.
Struktur-Funktionsanalysen sollten die umstrittene Topologie von ALMT-ähnlichen Proteinen beleuchten und die molekulare Herkunft der Phosphorylierungsaktivierung aufzeigen, sowie die Malatabhängigkeit und die starke Spannungsabhängigkeit von QUAC1 aufklären. Es zeigte sich jedoch schnell, dass Punktmutationen und Deletionen im C-Terminus von QUAC1 sehr häufig zu nicht-funktionellen Mutanten führten. Diese Tatsache weist darauf hin, dass es sich um einen hoch-strukturierten und funktionell sehr wichtigen Bereich des Anionenkanals handelt. Auch die Topologie des Anionenkanalproteins wird in der Literatur kontrovers diskutiert. Sowohl die Lage des N- und C-Terminus (extrazellulär oder intrazellulär), als auch die Anzahl der membrandurchspannenden Domänen war nicht abschließend geklärt. Deshalb wurde in einem Fluoreszenz-basiertem Ansatz die Lage der Termini bestimmt. Im Rahmen meiner Arbeit konnte somit eindeutig gezeigt werden, dass sich beide Termini im Zytosol der Zelle befinden. Auf Grundlage von Modellen aus der Literatur und meiner Topologiebestimmungen konnte schließlich ein erweitertes Modell zur Struktur von QUAC1 entwickelt werden. Dieses Modell kann in Zukunft als Ausgangspunkt für weiterführende Struktur-Funktionsanalysen dienen.
Diese Arbeit hat somit gezeigt, dass das Gen QUAC1 tatsächlich eine Komponente der R-Typ Ströme in Schließzellen kodiert. Ebenso wie SLAC1 steht der Malat-induzierte Anionenkanal QUAC1 unter der Kontrolle der schnellen ABA-Signalkaskade. In Zukunft bleibt zu klären, welche weiteren Gene für die R-Typ Kanalproteine in Schließzellen kodieren und welche strukturelle Grundlage für die besonderen Eigenschaften von QUAC1 hinsichtlich seiner schnellen Kinetiken, seiner Selektivität und Aktivierbarkeit durch Malat.
Das Cochlea-Implantat (CI) ermöglichte bereits >300 000 hochgradig hörgeschädigten Menschen
weltweit eine grundsätzlich wiederhergestellte Hörfunktion. Es wird angenommen, dass sich das
Sprachverständnis von CI-Trägern verbessert, wenn die funktionale Trennung der CI-Kanäle erhöht
wird. Neben verschiedenen auf die auditorische Peripherie beschränkten Ansätzen gibt es Überlegungen, eine verbesserte Kanaltrennung durch die Rehabilitation taubheitsinduzierter Degenerationen in der spektralen Verarbeitung im zentralen auditorischen System zu erreichen. Es konnte in ertaubten Tieren bislang allerdings kein adäquates CI-Stimulationsmuster beschrieben werden, dass es erlaubte, eine gezielte neuronale Plastizität in der spektralen Verarbeitung zu induzieren.
Die Arbeitsgruppe um M.P. Kilgard (UT Dallas, USA) zeigte in mehreren Studien in hörenden Tieren,
dass auditorische Stimulation gepaart mit elektrischer Vagusnerv-Stimulation (VNS) zu einer gezielten kortikalen Plastizität führt. Diese gepaarte Stimulation konnte die spektrale Verarbeitung von Signalen im auditorischen Kortex (AC) gezielt beeinflussen und so z.B. pathologisch verbreiterte Repräsentationen von Tönen wieder verfeinern. Dieses hochgradige Potential für gezielte Plastizität im AC durch die gepaarte VNS scheint eine vielversprechende Lösung darzustellen, um die durch verbreiterte Repräsentation im ertaubten AC verminderte CI-Kanaltrennung zu verbessern. Vor diesem Hintergrund sollte in der vorliegenden Promotion die Übertragbarkeit dieses hochgradigen Potentials auf das ertaubte und CI-stimulierte auditorische System evaluiert werden.
Um die CI-Kanaltrennung zu untersuchen, wurde ein Multikanal-CI für die Mongolische Wüstenrennmaus (Gerbil) entwickelt. Trotz der kleinen Ausmaße von Cochlea und AC im Gerbil und der generell breiten neuronalen Erregung durch intracochleäre elektrische Stimulation konnte eine tonotop organisierte und selektive Repräsentation der neuronalen Antworten für mehrere CI-Kanäle im AC nachgewiesen werden. Für die gepaarte CI/VN-Stimulation wurden die Tiere zusätzlich mit einer Manschettenelektrode um den linken zervikalen Nervus vagus (VN) implantiert. Die chronischen Implantate erlaubten über mehrere Wochen hinweg eine stabile und zuverlässige elektrische Stimulation im frei-beweglichen Gerbil. Damit kombiniert das in dieser Promotion entwickelte Multikanal-CI-VNS-Modell die Vorteile einer tonotop selektiven und stabilen neuronalen Aktivierung mit den ethischen, kostenrelevanten und entwicklungsbezogenen Vorteilen, die der Einsatz von Kleinnagern bietet.
Als nächster Schritt wurde das grundsätzliche Potential der gepaarten CI/VN-Stimulation für gezielte plastische Veränderungen im AC des Gerbils getestet. Engineer et al. (2011) hatten bereits in akustischen Studien in hörenden Ratten die kortikale Überrepräsentation eines einzelnen chronisch mit VNS gepaarten Tones gezeigt. In der vorliegenden Promotion wurde versucht, die Ergebnisse aus der akustischen Studie in hörenden Ratten in zwei verschiedenen Studien im Gerbil zu reproduzieren. Analog zur gepaarten Ton/VN-Stimulation in der Ratte untersuchten wir zuerst in ertaubten Gerbils die Auswirkungen einkanaliger CI-Stimulation gepaart mit VNS. Im AC des Gerbils konnten keine Veränderung der zentralen Repräsentation des VNS gepaarten CI-Kanals festgestellt werden. Um speziesspezifische (Ratte vs. Gerbil) und stimulusspezifische (akustisch vs. elektrisch) Unterschiede zwischen den Studien als mögliche Gründe für das Ausbleiben der VNS induzierten Plastizität auszuschließen, wurde nun die gepaarte Ton/VN-Stimulation (Engineer et al., 2011) im hörenden Gerbil wiederholt. Eine kortikale Überrepräsentation des VNS gepaarten Signals konnte aber auch im hörenden Gerbil nicht reproduziert werden.
Mögliche Gründe für die Diskrepanz zwischen unseren Ergebnissen im Gerbil und den publizierten
Ergebnissen in der Ratte werden diskutiert. Die generelle Funktionsfähigkeit der VNS in den chronisch stimulierten Tieren wurde durch die Ableitung VNS evozierter Potentiale (VNEP) kontrolliert. Ein speziesspezifischer Unterschied erscheint bei der biologischen Nähe von Ratte und mongolischer Wüstenrennmaus unwahrscheinlich, kann allerdings durch die vorliegenden Studien nicht vollständig ausgeschlossen werden. Eine Abhängigkeit des plastischen Potentials der gepaarten VNS von der Stimulationsintensität ist bekannt. Da Ratten und Gerbils ähnliche VNEP-Schwellen zeigten und mit identischen VNS-Amplituden stimuliert wurden, gehen wir davon aus, dass Unterschiede im plastischen Potential gepaarter VNS zwischen beiden Spezies nicht auf die verwendete Stimulationsintensität zurückzuführen sind.
Die beschriebene Diskrepanz im Potential für kortikale Plastizität durch gepaarte VNS weckt Zweifel an der Übertragbarkeit des für die Ratte publizierten Potentials auf andere Spezies, einschließlich des Menschen.
In order to select the appropriate behavior, it is important to choose the right behavior at the right time out of many options. It still remains unclear nowadays how exactly this is managed. To address this question, I expose flies (Drosophila melanogaster) to uncontrollable stress to study their behavior under restrictive circumstances by using the so-called shock box. Exposing animals to uncontrollable stress may have an impact on subsequent behavior and can last for some time. The animal learns that whatever it does, it cannot change the situation and therefore can develop something called learned helplessness. The term was first conceptualized by two American psychologists Maier and Seligman (1967), who discovered this phenomenon while doing experiments with dogs. They found out that dogs which are exposed to inescapable stress, later fail in a learning task (‘shuttle box’).
In this work the walking patterns of three different types of experimental flies, walking in a small dark chamber, were evaluated. Using the triadic design (Seligman and Maier, 1967), flies were either exposed to electric shock randomly (yoked), could turn it off by being active (master) or did not receive punishment at all (control). Master flies were shocked whenever they sat for more than 0.9 seconds. At the same time yoked flies received a shock as well independent of what they were doing, to ensure the same amount of shocks received and to create random punishment pattern for the yoked group. With this so-called no-idleness paradigm flies were conditioned either 10 minutes, which resulted in a short (3 minutes) after-effect, or 20 minutes that turned out to be more stable (10 minutes).
In a second part, the behavior during the 20 minute conditioning and a 10 minutes post-test was described in detail. Female flies of the yoked group developed lower activity levels, longer pauses and walked more slowly than master and control flies during conditioning. In the time after the shocks while still in the box, the yoked flies also reduced the frequency and duration of walking bouts as well as their walking speed. Additionally, they took more time to resume walking after the onset of an electric shock than master flies (escape latency) and turned out to make less pauses lasting between 1-1.5 seconds which supports the finding concerning the escape latency.
Male flies, tested under the same conditions, showed a slightly weaker after-effect regarding the difference between master and yoked during conditioning and post-test when compared to female flies.
When comparing the 20 minutes conditioning with subsequent 10 minutes test in the heat and the shock box in parallel, one finds the same effect: Flies which do not have control over the shocks, lower their activity, make less but longer pauses and walk more slowly than their respective master flies. Despite the similar effect of heat and shock on the flies, some differences between the devices occurred, which can partly be explained by different humidity conditions as well as by different surfaces within the chambers.
When the control over the shocks is given back to the yoked flies, it takes them about seven minutes to realize it. One could also show that dopamine levels in the brain were reduced in comparison to flies which did not receive shocks. Yoked flies also were impaired in a place learning task (place learning) and their reaction to light (exit from the box towards the light) directly after conditioning.
After characterizing the walking behavior in the chambers, the study deals with the question whether the effects observed in the chambers transfer to different environments.
In free walk they only differed from flies which did not receive electric shocks and no effect of uncontrollability was transferred to courtship behavior. Handling as the cause could be excluded. Since handling could be exclude to be the cause of losing the effect, I assumed that the behavior shown in the boxes are context depend.
Not only were the after-effects of inescapable shock subject of the current research also the impact of the rearing situation on the response to electric shock was investigated in the present study. Flies which grew up in a single-reared situation turned out to be less affected by inescapable stress in both sexes.
In the next part, the first steps to unravel the neuronal underpinning were taken. A mutant – fumin – which is defective in the dopamine re-uptake transporter showed less reaction to inescapable foot shocks, while a mutant for the gene which encodes an adenylate cyclase (rutabaga2080) resulted in a good score during conditioning, but showed no stable after-effect. Downregulating the expression of the adenylate cyclase gene (rutabaga) in different parts of the mushroom bodies showed, that rutabaga is necessary in the α’β’-lobes for expressing the differences between master and yoked flies in the no-idleness paradigm. The study further confirmed previous findings, that rutabaga is needed in operant but not in classical conditioning.
As a result, the study could show that not the stimulus itself causes the state of uncontrollability but the fact that the fly learned that it was not in control of the stimulus. This state turned out to be context and time dependent.
Die vorliegende Arbeit stellt einen Beitrag zur Chemie des höherkoordinierten Siliciums dar. Dabei standen die Synthese und Charakterisierung neuer neutraler tetra-, penta- und hexakoordinierter Silicium(IV)-Komplexe sowie die Synthese, Charakterisierung und Reaktivität eines neuartigen Donor-stabilisierten Silylens im Vordergrund.
Im Rahmen dieser Arbeit wurden das Silan 16, die neutralen tetrakoordinierten Silicium(IV)-Komplexe 36, 37', 38'·C6H5CH3, 40'∙C6H5CH3, 41 und 42, die neutralen pentakoordinierten Silicium(IV)-Komplexe 2‒8, 10·0.5C6H5CH3, 11, 12, 15, 17‒20 und 39', die neutralen hexakoordinierten Silicium(IV)-Komplexe 21‒23, das Donor-stabilisierte trikoordinierte Silylen 25, der neutrale tetrakoordinierte Silicium(II)-Komplex 35 sowie das Lithiumamidinat 1·2Et2O erstmalig dargestellt und charakterisiert. Die Charakterisierung dieser Verbindungen erfolgte durch NMR-Spektroskopie in Lösung und im Festkörper, durch Kristallstrukturanalyse sowie durch Elementaranalyse. Die Synthesen und Eigenschaften dieser Verbindungen können wie folgt zusammengefasst werden:
Synthese und Charakterisierung neutraler pentakoordinierter Silicium(IV)-Komplexe
Ausgehend von entsprechenden Silicium(IV)-haltigen Vorstufen wurden die neutralen pentakoordinierten Silicium(IV)-Komplexe 2‒8, 10·0.5C6H5CH3, 11, 12, 15 und 17‒20 dargestellt. So konnten die Verbindungen 2, 5, 7 und 8 durch Umsetzung der entsprechemden Trichlorsilane bzw. Tetrachlorsilan mit 1 in Diethylether erhalten werden. Diese Verbindungen besitzen an den beiden Stickstoff-Atomen des Amidinato-Liganden jeweils eine sterisch sehr anspruchsvolle Diisopropylphenyl-Einheit (Dipp), welche den Einbau eines zweiten Amidinato-Liganden nicht zulässt und sich so ausschließlich pentakoordinierte Silicium(IV)-Komplexe bilden. Durch Weiterreaktion von 2 und 5 mit Lithiumdimethyl- bzw. Kaliumbis(trimethylsilyl)amid entstanden die Komplexe 3, 4 und 6.
Die Si-Koordinationspolyeder von 2‒8 im Kristall entsprechen denen von stark verzerrten trigonalen Bipyramiden, wobei die Stickstoff-Atome des Amidinato-Liganden eine axiale bzw. äquatoriale Position besetzen. Die zweite axiale Position wird jeweils von einem Chloro-Liganden eingenommen. Die genannten Verbindungen besitzen alle einen stark gespannten viergliedrigen SiN2C-Ring mit mehr oder weniger stark ausgeprägter Elektronendelokalisation innerhalb de N‒C‒N-Fragmentes, welcher durch den Amidinato-Liganden mit dem Si-Koordinationszentrum gebildet wird und hauptverantwortlich für die starke Verzerrung der Si-Koordinationspolyeder ist.
Die Verbindungen 10·0.5C6H5CH3, 11 und 12 entstanden durch Umsetzung der entsprechenden Trichlorsilane mit 9 und zwei Moläquivalenten Triethylamin in Tetrahydrofuran. Durch Weiterreaktion von 11 mit Benzolthiol bzw. Benzolselenol und Triethylamin in Tetrahydrofuran wurden die Komplexe 18 und 19 gebildet. Die Verbindungen 15 und 20 entstanden mittels einer Transsilylierungsreaktion von 14 mit Azidotrimethylsilan bzw. 11 mit Trimethylsilyl(phenyl)tellurid in Tetrahydrofuran. Verbindung 17 wurde durch Umsetzung von Cl2Si(OPh)Me (16) mit 9 und zwei Moläquivalenten Triethylamin in Tetrahydrofuran erhalten.
Die Si-Koordinationspolyeder von 10·0.5C6H5CH3, 11, 12, 15 und 17‒20 im Kristall entsprechen denen von stark verzerrten trigonalen Bipyramiden, wobei der tridentate N,N',S- bzw. N,N',O-Ligand zwei Fünfringe mit dem Si-Koordinationszentrum ausbildet. Das Pyridin-Stickstoff- und das Schwefel-Atom des N,N',S-Liganden (bzw. Sauerstoff-Atom des N,N',O-Liganden) besetzen die axialen Bindungspositionen.
Synthese und Charakterisierung neutraler hexakoordinierter Silicium(IV)-Komplexe
Die neutralen hexakoordinierten Silicium(IV)-Komplexe 21 und 22 wurden durch Umsetzung von Trichlorsilan mit zwei Moläquivalenten des entsprechenden Lithiumamidinats in Diethylether dargestellt.
Die Si-Koordinationspolyeder von 21 und 22 im Kristall entsprechen denen von stark verzerrten Oktaedern, wobei jeweils eines der beiden Stickstoff-Atome der zwei Amidinato-Liganden trans zueinander angeordnet sind. Die beiden anderen Stickstoff-Atome der Amidinato-Liganden befinden sich in trans-Position zum Chloro- bzw. Hydrido-Liganden.
Der neutrale hexakoordinierte Silicium(IV)-Komplex 23 wurde durch Umsetzung des pentakoordinierten Silicium(IV)-Komplexes 11 mit 8-Hydroxychinolin und Triethylamin in Tetrahydrofuran dargestellt.
Das Si-Koordinationspolyeder von 23 im Kristall entspricht dem eines stark verzerrten Oktaeders, wobei der dreizähnige N,N',S-Ligand eine mer-Anordnung einnimmt und das Chinolin-Stickstoff-Atom und das Kohlenstoff-Atom des Methyl-Liganden trans zueinander stehen.
Mit den hier beschriebenen Synthesen konnte gezeigt werden, dass pentakoordinierte Chlorosilicium(IV)-Komplexe ‒ wie beispielsweise 2, 5 oder 11 ‒ sehr gut geeignete Ausgangsstoffe für die Darstellung neuartiger penta- und hexakoordinierter Silicium(IV)-Verbindungen darstellen.
Synthese und Charakterisierung eines neuartigen Donor-stabilisierten Silylens
Nachdem alle Versuche, ein entsprechendes Donor-stabilisiertes Silylen durch Basen-induzierte reduktive HCl-Eliminierung der penta- bzw. hexakoordinierten Chlorohydridosilicium(IV)-Komplexe 2‒4, 21 und 22 darzustellen, fehlschlugen, wurde daraufhin der pentakoordinierte Dichlorosilicium(IV)-Komplex 6 mit zwei Moläquivalenten elementarem Kalium in Tetrahydrofuran erfolgreich zum trikoordinierten Donor-stabilisierten Silylen 25 umgesetzt.
Das Si-Koordinationspolyeder von 25 entspricht dem eines stark verzerrten (Pseudo)tetraeders, wobei die drei Bindungspositionen von den Stickstoff-Atomen und eine vierte von dem freien Elektronenpaar eingenommen werden. Die starke Verzerrung ist auf den stark gespannten viergliedrigen SiN2C-Ring des Komplexes zurückzuführen.
Reaktivität des Donor-stabilisierten Silylens 25
Der trikoordinierte Silicium(II)-Komplex 25 reagierte mit Eisenpentacarbonyl in Toluol im Sinne einer nukleophilen Substitutionsreaktion unter Ausbildung einer Si–Fe-Bindung zum neutralen tetrakoordinierten Silicium(II)-Komplex 35.
Das Si-Koordinationspolyeder von 35 im Kristall entspricht dem eines stark verzerrten Tetraeders. Das Fe-Koordinationspolyeder entspricht dem einer stark verzerrten trigonalen Bipyramide, wobei der sterisch sehr anspruchsvolle Silylen-Ligand interessanterweise eine axiale Bindungsposition am Eisen-Koordinationszentrum einnimmt.
Desweiteren wurde 25 mit den Aziden Me3SiN3, PhSCH2N3 und (PhO)2P(O)N3 in Toluol im Sinne einer oxidativen Addition unter Abspaltung von elementarem Stickstoff zu 36, 37' bzw. 38'·C6H5CH3 umgesetzt. Bemerkenswert ist, dass bei der Reaktion mit PhSCH2N3 zu 37' eine Umlagerungsreaktion stattfindet, wobei eine Si–S-Bindung geknüpft und ein Si–N=CH2-Fragment gebildet wird. Bei der Reaktion von 25 mit (PhO)2P(O)N3 zu 38'·C6H5CH3 wird ein Sauerstoff-verbrücktes Dimer gebildet, wodurch ein achtgliedriger Ring mit zwei Silicium(IV)-Zentren aufgebaut wird.
Die Si-Koordinationspolyeder von 36, 37' und 38'·C6H5CH3 im Kristall entsprechen denen von stark verzerrten Tetraedern, wobei der Amidinato-Ligand nur in Verbindung 36 bidentat an das Silicium-Zentrum koordiniert ist, während für 37' und 38'·C6H5CH3 ein monodentater Koordinationsmodus beobachtet wird.
Durch Umsetzung von 25 mit N2O, S, Se bzw. Te in Toluol entstanden ebenfalls im Sinne einer oxidativen Addition die tetra- bzw. pentakoodinierten Silicium(IV)-Komplexe 39', 40'·C6H5CH3, 41 und 42. Die Verbindungen 39' und 40'·C6H5CH3 sind Dimere der eigentlichen Zielverbindungen 39 und 40, wobei 40' bei höheren Temperaturen zu dem Monomer 40 dissoziiert, welches dann nach Abkühlen auf Raumtemperatur auch in Lösung stabil ist. Die Verbindungen 41 sowie 42 bilden jedoch ausschließlich Monomere.
Die Si-Koordinationspolyeder von 39' im Kristall entsprechen dem einer stark verzerrten trigonalen Bipyramide, während die Si-Koordinationspolyeder von 40'·C6H5CH3, 41 und 42 denen eines stark verzerrten Tetraeders entsprechen. Dabei ist der Amidinato-Ligand in 39', 41 und 42 bidentat, in 40'·C6H5CH3 dagegen monodentat an das Silicium-Koordinationszentrum koordiniert ist.
Mit den hier beschriebenen Synthesen konnte gezeigt werden, dass das Donor-stabilisierte Silylen 25 ein außergewöhnliches Reaktivitätsspektrum aufweist und damit ein sehr interessantes Synthesepotential zur Darstellung neuartiger Silicium(II)- und Silicium(IV)-Komplexe besitzt.
Beyond the state of the art, towards intuitive and reliable non-visual Brain-Computer-Interfacing

For the present work three main goals were formulated:
goal 1 To design a tactile BCI used for mobility which is
intuitive (G1.1), reliable and fast while being usable
by participants aged 50 years and above.
goal 2 To design an auditory BCI used for communication
which is intuitive and reliable.
goal 3 To examine the effects of training on tactile and
auditory BCI performance.
Three studies were performed to achieve these goals.
In the first study nine participants aged above 50 years
performed a five-session training after which eight participants
were able to navigate a virtual wheelchair with
mean accuracy above 95% and an ITR above 20 bits / min.
In the second study 15 participants, four of them endusers
with motor-impairment, were able to communicate
meaningful with high accuracies using an auditory BCI.
In the third study nine healthy and nine visually impaired
participants (regarded as sensory experts for non-visual
perception) performed tactile, auditory and visual (for
healthy participants only) copy tasks. Participants with
trained perception significantly outperformed control
participants for tactile but not for auditory performance.
Tactile performance of sensory experts was on equal levels
as the visual performance of control participants.
We were able to demonstrate viability of intuitive gazeindependent
tactile and auditory BCI. Our tactile BCI performed
on levels similar to those of visual BCI, outperforming
current tactile BCI protocols. Furthermore, we were
able to demonstrate significant beneficial effect of training
on tactile BCI performance. Our results demonstrate previously
untapped potential for tactile BCI and avenues for
future research in the field of gaze-independent BCI.
Biochemical and molecular characterization of an original master sex determining gene in Salmonids
(2016)
Sexual development is a fundamental and versatile process that shapes animal morphology, physiology and behavior. The underlying developmental process is composed of the sex determination and the sex differentiation. Sex determination mechanisms are extremely labile among taxa. The initial triggers of the sex determination process are often genetics called sex determining genes. These genes are expressed in the bipotential gonad and tilt the balance to a developmental program allowing the differentiation of either a testis or an ovary. Fish represent a large and fascinating vertebrate group to study both sex determination and sex differentiation mechanisms. To date, among the known sex determining genes, three gene families namely sox, dmrt and TGF-β factors govern this developmental program. As exception to this rule, sdY “sexually dimorphic on the Y” does not belong to one of these families as it comes from the duplication / evolution of an ancestor gene related to immunity, i.e., the interferon related factor 9, irf9. sdY is the master sex determining gene in salmonids, a group of fishes that include species such as rainbow trout and Atlantic salmon. The present study was aimed to firstly characterize the features of SdY protein. Results indicate that SdY is predominantly localized in the cytoplasm tested in various fish and mammalian cell lines and confirmed by different methods. Predictive in silico analysis revealed that SdY is composed of a β-sandwich core surrounded by three α-helices as well specific characteristics conferring a putative protein-protein interaction site. Secondly, the study was aimed to understand how SdY could trigger testicular differentiation. SdY is a truncated divergent version of Irf9 that has a conserved protein-protein domain but lost the DNA interaction domain of its ancestor gene. It was then hypothesized that SdY could initiate testicular differentiation by protein-protein interactions. To evaluate this we first conducted a yeast-two-hybrid screen that revealed a high proportion of transcription factors including fox proteins. Using various biochemical and cellular methods we confirm an interaction between SdY and Foxl2, a major transcription factor involved in ovarian differentiation and identity maintenance. Interestingly, the interaction of SdY with Foxl2 leads to nuclear translocation of SdY from the cytoplasm. Furthermore, this SdY translocation mechanism was found to be specific to fish Foxl2 and to a lesser extend Foxl3 and not other Fox proteins or mammalian FoxL2. In addition, we found that this interaction allows the stabilization of SdY and prevents its degradation. Finally, to better decipher SdY action we used as a model a mutated version of SdY that was identified in XY females of Chinook salmon natural population. Results show that this mutation induces a local conformation defect obviously leading to a misfolded protein and a quick degradation. Moreover, the mutated version compromised the interaction with Foxl2 defining a minimal threshold to induce testicular differentiation. Altogether results from my thesis propose that SdY would trigger testicular differentiation in salmonids by preventing Foxl2 to promote ovarian differentiation. Further research should be now carried out on how this interaction of SdY and Foxl2 acts in-vivo.
Die Sequenzierungstechnologien entwickeln sich stetig weiter, dies ermöglicht eine zuvor nicht erreichte Ausbeute an experimentellen Daten und auch an Neuentwicklungen von zuvor nicht realisierbaren Experimenten. Zugleich werden spezifische Datenbanken, Algorithmen und Softwareprogramme entwickelt, um die neu entstandenen Daten zu analysieren. Während der Untersuchung bioinformatischer Methoden für die Identifizierung und Klassifizierung somatischer Mutationen in hämatologischen Erkrankungen, zeigte sich eine hohe Vielfalt an alternativen Softwaretools die für die jeweiligen Analyseschritte genutzt werden können. Derzeit existiert noch kein Standard zur effizienten Analyse von Mutationen aus Next-Generation-Sequencing (NGS)-Daten. Die unterschiedlichen Methoden und Pipelines generieren Kandidaten, die zum größten Anteil in allen Ansätzen identifiziert werden können, jedoch werden Software spezifische Kandidaten nicht einheitlich detektiert.
Um eine einheitliche und effiziente Analyse von NGS-Daten durchzuführen war im Rahmen dieser Arbeit die Entwicklung einer benutzerfreundlichen und einheitlichen Pipeline vorgesehen. Hierfür wurden zunächst die essentiellen Analysen wie die Identifizierung der Basen, die Alignierung und die Identifizierung der Mutationen untersucht. Des Weiteren wurden unter Berücksichtigung von Effizienz und Performance diverse verfügbare Softwaretools getestet, ausgewertet und sowohl mögliche Verbesserungen als auch Erleichterungen der bisherigen Analysen vorgestellt und diskutiert. Durch Mitwirken in Konsortien wie der klinischen Forschergruppe 216 (KFO 216) und International Cancer Genome Consortium (ICGC) oder auch bei Haus-internen Projekten wurden Datensätze zu den Entitäten Multiples Myelom (MM), Burkitt Lymphom (BL) und Follikuläres Lymphom (FL) erstellt und analysiert. Die Selektion geeigneter Softwaretools und die Generierung der Pipeline basieren auf komparativen Analysen dieser Daten, sowie auf geteilte Ergebnisse und Erfahrungen in der Literatur und auch in Foren. Durch die gezielte Entwicklung von Skripten konnten biologische und klinische Fragestellungen bearbeitet werden. Hierzu zählten eine einheitliche Annotation der Gennamen, sowie die Erstellung von Genmutations-Heatmaps mit nicht Variant-Calling-File (VCF)-Syntax konformen Dateien. Des Weiteren konnten nicht abgedeckte Regionen des Genoms in den NGS-Daten identifiziert und analysiert werden. Neue Projekte zur detaillierten Untersuchung der Verteilung von wiederkehrender Mutationen und Funktionsassays zu einzelnen Mutationskandidaten konnten basierend auf den Ergebnissen initiiert werden.
Durch eigens erstellte Python-Skripte konnte somit die Funktionalität der Pipeline erweitert werden und zu wichtigen Erkenntnissen bei der biologischen Interpretation der Sequenzierungsdaten führen, wie beispielsweise zu der Detektion von drei neuen molekularen Subgruppen im MM. Die Erweiterungen, der in dieser Arbeit entwickelten Pipeline verbesserte somit die Effizienz der Analyse und die Vergleichbarkeit unserer Daten. Des Weiteren konnte durch die Erstellung eines eigenen Skripts die Analyse von unbeachteten Regionen in den NGS-Daten erfolgen.
Aktueller Goldstandard bei der Rekonstruktion des ACL des Menschen sind au-tologe Transplantate. Diese sind allerdings je nach Entnahmeort mit einer mehr oder weniger hohen Entnahmemorbidität und dem Risiko für Folgeerkrankungen verbunden. Um dies zu umgehen, wurde ein xenogenes Kollagenimplantat aus
Kollagen-I-Fasern von Ratten entwickelt und das native Konstrukt bereits in einer Vorläuferstudie getestet.
Im Rahmen dieser Arbeit wurden diese Kreuzbandkonstrukte mit Hilfe diverser
Crosslinker modifiziert und hinsichtlich ihrer Biomechanik, Biokompatibilität und ihres in-vivo Verhaltens untersucht.
Bewusst wurde dabei auf die Zellbesiedlung dieser Konstrukte verzichtet, da un-ter Berücksichtigung wirtschaftlicher Gesichtspunkte eines späteren humanen
Einsatzes hierfür eine Arzneimittelzulassung notwendig gewesen wäre. Mit Hilfe der Crosslinker wurde versucht, die mechanische Stabilität sowie die Resistenz gegen kollagenabbauende Enzyme der Synovia zu erhöhen, um die Gefahr post-operativer Instabilitäten zu verringern. Dabei sollten Fragen bezüglich Immun-antwort, Biokompatibilität sowie Biodegradierbarkeit genau berücksichtigt wer-den. Als Crosslinker wurden für einen Vergleich in vitro neben 0,5 % Genipin auch 10 % HMDI sowie Glukose und EDC/NHS herangezogen.
Dabei zeigten die Genipin-gecrosslinkten Einzelfasern die größte Reißfestigkeits-zunahme, wohingegen auf Minikonstruktbasis 10 % HMDI zu den höchsten UTS-Werten führte. Ebenso ließen sich bezüglich der Biokompatibilutät in vitro bei den Crosslinkern 0,5 % Genipin und 10 % HMDI Vorteile gegenüber den beiden an-deren erkennen.
Schließlich erfolgte im Rahmen eines Tierversuchs an 16 Minipigs der Einbau von 0,5 % Genipin-gecrosslinkten Konstrukten als Kreuzbandersatz und an-schließend die biomechanische Testung sowie nach Paraffineinbettung auch eine durchlichtmikrokopische deskriptive Auswertung der Transplantate.
Während nach 6 Wochen eine deutliche Reißfestigkeitsabnahme zu verzeichnen war, erreichte diese nach 6 Monaten wieder fast 60 % ihrer ursprünglichen UTS.
Somit konnte ein Remodeling des eingesetzten Implantats angenommen wer-den. Dies bestätigte sich in der durchgeführten histologischen Untersuchung.
Hier war das Implantat deutlich vaskularisiert, von zahlreichen Fibroblasten durchsetzt und wies eine synoviale Deckschicht auf. Allerdings scheint vor allem wegen der Schwäche der Konstrukte nach 6 Wochen sowie den vermutlich auf-grund des Crosslinkers auftretenden Reaktionserscheinungen innerhalb des
Kniegelenks ein Einsatz im humanen Bereich zum gegenwärtigen Zeitpunkt noch nicht ausgereift.
Dennoch lässt sich gerade anhand des stattfindenden Remodelings das große Potential kollagenbasierter Materialien für den Kreuzbandersatz erkennen. Eine weitere Optimierung des bestehenden Konstrukts sollte deshalb forciert werden.
Project Borylene
A new borylene ligand ({BN(SiMe\(_3\))(t-Bu)}) has been successfully synthesized bound in a terminal manner to base metal scaffolds of the type [M(CO)\(_5\)] (M = Cr, Mo, and W), yielding complexes [(OC)\(_5\)Cr{BN(SiMe\(_3\))(t-Bu)}] (19), [(OC)\(_5\)Mo{BN(SiMe\(_3\))(t- Bu)}] (20), and [(OC)\(_5\)W{BN(SiMe\(_3\))(t-Bu)}] (21) (Figure 5-1). Synthesis of complexes 19, 20, and 21 was accomplished by double salt elimination reactions of Na\(_2\)[M(CO)\(_5\)] (M = Cr (11), Mo (1), and W (12)) with the dihaloborane Br\(_2\)BN(SiMe\(_3\))(t-Bu) (18). This new “first generation” unsymmetrical borylene ligand is closely akin to the bis(trimethylsilyl)aminoborylene ligand and has been shown to display similar structural characteristics and reactivity. The unsymmetrical borylene ligand {BN((SiMe\(_3\))(t-Bu)} does display some individual characteristics of note and has experimentally been shown to undergo photolytic transfer to transition metal scaffolds in a more rapid manner, and appears to be a more reactive borylene ligand, than the previously published symmetrical {BN(SiMe\(_3\))\(_2\)} ligand, based on NMR and IR spectroscopic evidence.
Photolytic transfer reactions with this new borylene ligand ({BN((SiMe\(_3\))(t-Bu)}) were conducted with other metal scaffolds, resulting in either complete borylene transfer or partial transfer to form bridging borylene ligand interactions between the two transition metals. The unsymmetrical ligand’s coordination to early transition metals (up to Group 6) indicates a preference for a terminal coordination motif while bound to these highly Lewis acidic species. The ligand appears to form more energetically stable bridging coordination modes when bound to transition metals with high Lewis basicity (beyond Group 9) and has been witnessed to transfer to transition metal scaffolds in a terminal manner and subsequently rearrange in order to achieve a more energetically stable bridging final state.
Figure 5-2 lists the four different transfer reactions conducted between the chromium borylene species [(OC)\(_5\)Cr{BN(SiMe\(_3\))(t-Bu)}] (19) and the transition metal complexes [(η\(^5\)-C\(_5\)H\(_5\))V(CO)\(_4\)] (51), [(η\(^5\)-C\(_5\)Me\(_5\))Ir(CO)\(_2\)] (56), [(η\(^5\)-C\(_5\)H\(_4\)Me)Co(CO)\(_2\)] (59), and [{(η\(^5\)-C\(_5\)H\(_5\))Ni}\(_2\){μ-(CO)\(_2\)}] (53). These reactions successfully yielded the new “second generation” borylene complexes [(η\(^5\)-C\(_5\)H\(_5\))(OC)\(_3\)V{BN(SiMe\(_3\))(t-Bu)}] (55), [(η\(^5\)-C\(_5\)Me\(_5\))Ir{BN(SiMe\(_3\))(t-Bu)}\(_2\)] (58), [{(η\(^5\)-C\(_5\)H\(_4\)Me)Co}\(_2\)(μ-CO)\(_2\){μ- BN(SiMe\(_3\))(t-Bu)}] (61), and [{(η\(^5\)-C\(_5\)H\(_5\))Ni}\(_2\)(μ-CO){μ-BN(SiMe\(_3\))(t-Bu)}] (62), respectively.
Analysis of the accumulated data for all of the terminal borylene species discussed in this section, particularly bond distances, infrared spectroscopy, and \(^{11}\)B{\(^1\)H} NMR spectroscopic data, has been performed, and a trend in the data has led to the following conclusions:
[1] NMR spectroscopic data for the \(^{11}\)B{\(^1\)H} boron and \(^{13}\)C{\(^1\)H} carbonyl environments of the first generation borylene species ([(OC)\(_5\)M{BN(SiMe\(_3\))(t-Bu)}] (M = Cr (19), Mo (20), and W (21))) all show progressive up-field shifting as the Group 6 metal becomes heavier (Cr (19) to Mo (20) to W (21)), indicating maximum deshielding for these nuclei in the [(OC)\(_5\)Cr{BN(SiMe\(_3\))(t-Bu)}] (19) complex.
[2] The boron-metal-trans-carbon (B-M-C\(_{trans}\)) axes of the first generation borylene complexes [(OC)\(_5\)M{BN(SiMe\(_3\))(t-Bu)}] (M = Mo (20), and W (21)) are not completely linear, preventing direct IR spectroscopic comparison. The chromium analog [(OC)\(_5\)Cr{BN(SiMe\(_3\))(t-Bu)}] (19), however, is essentially linear and displays the expected three carbonyl IR stretching frequencies, all at higher energy than those of the chromium bis(trimethylsilyl)aminoborylene complex [(OC)\(_5\)Cr{BN(SiMe\(_3\))\(_2\)}] (13), indicating that the ({BN(SiMe\(_3\))(t-Bu)}) ligand is either a stronger σ-donor or a poorer π-acceptor compared to the chromium metal center.
[3] In transfer reactions, the {BN(SiMe\(_3\))(t-Bu)} fragment appears to be more stable as a terminal ligand when bound to more Lewis acidic first row transition metals and appears to prefer coordination in a bridging motif when coordinated to more Lewis basic first row transition metals.
Project Borirene
The synthesis of the first platinum bis(borirene) complexes are presented along with findings from structural and electronic examination of the role of platinum in allowing increased coplanarity and conjugation of twin borirene systems. This series of trans-platinum-linked bis(borirene) complexes (119/120, 122/123, and 125/126) all show coplanarity in the twin ring systems and stand as the first verified structural representations of two coplanar borirene systems across a linking unit. The role of a platinum atom in mediating communication between chromophoric ligands can be generalized by an expected bathochromic (red) shift in the absorption spectrum due to an increase in the electronic delocalization between the formerly independent aromatic systems when compared to the platinum mono-σ-borirenyl systems. The trans-platinum bis(borirene) scaffold serves as a simplified monomeric system that allows not only study of the effects of transition metals in mitigating electronic conjugation, but also the tunability of the overall photophysical profile of the system by exocyclic augmentation of the three-membered aromatic ring.
A series of trans-platinum bis(alkynyl) complexes were prepared (Figure 5-3) to serve as stable platforms to transfer terminal borylene ligands {BN(SiMe\(_3\))\(_2\)} onto 95, 102, 106, and 63. Mixing of cis-[PtCl\(_2\)(PEt\(_3\))\(_2\)] (93) with two equivalents of corresponding alkynes in diethylamine solutions successfully yielded trans-[Pt(C≡C-Ph)\(_2\)(PEt\(_3\))\(_2\)] (95), trans-[Pt(C≡C-p-C\(_6\)H\(_4\)OMe)\(_2\)(PEt\(_3\))\(_2\)] (102), trans-[Pt(C≡C-p-C\(_6\)H\(_4\)CF\(_3\))\(_2\)(PEt\(_3\))\(_2\)](106), and trans-[Pt(C≡C-9-C\(_{14}\)H\(_9\))\(_2\)(PEt\(_3\))\(_2\)] (63) through salt elimination reactions.
Three of the trans-platinum bis(alkynyl) complexes (95, 102, and 106) successfully yielded trans-platinum bis(borirenyl) complexes 119/120, 122/123, and 125/126 through photolytic transfer of two equivalents of the terminal borylene ligand {BN(SiMe\(_3\))\(_2\)} from [(OC)\(_5\)Cr{BN(SiMe\(_3\))\(_2\)}] (13) (Figure 5-4). Attempted borylene transfer reactions to the trans-platinum bis(alkynyl) complex trans-[Pt(C≡C-9-C\(_{14}\)H\(_9\))\(_2\)(PEt\(_3\))\(_2\)] (63) failed due to the complex’s photoinstability. Although a host of other variants of platinum alkynyl species were prepared and attempted, these three were the only ones that successfully yielded trans-platinum bis(borirenyl) units. Attempts were also made to create a cis variant for direct UV-vis comparison to the trans-platinum bis(borirenyl) variants, however, these attempts were also not successful. Gladysz-type platinum end-capped alkynyl species were also synthesized to serve as transfer platforms for borirene synthesis in sequential order, however, these species were also shown to not be photolytically stable.
A host of new monoborirenes: Ph-(μ-{BN(SiMe\(_3\))(t-Bu)}C=C)-Ph (148), trans- [PtCl{(μ-{BN(SiMe\(_3\))(t-Bu)}C=C)-Ph}(PEt\(_3\))\(_2\)] (149), and [(η\(^5\)-C\(_5\)Me\(_5\))(OC)\(_2\)Fe(μ- {BN(SiMe\(_3\))(t-Bu)}C=C)Ph] (150) were synthesized by photo- and thermolytic transfer of the unsymmetrical {BN(SiMe\(_3\))(t-Bu)} ligand from the complexes [(OC)\(_5\)M{BN(SiMe\(_3\))(t-Bu)}] (M = Cr (19), Mo (20), and W (21)) to organic and organometallic alkynyl species to verify that the borylene complexes all display similar reactivity to the symmetrical terminal borylenes of the type [(OC)\(_5\)M{BN(SiMe\(_3\))\(_2\)}] (M = Cr (13), Mo (14), and W (15)). These monoborirenes are all found to be oils when in their pure states and X-ray structural determination was impossible for these species.
Project Boratabenzene
The bis(boratabenzene) complex [{(η\(^5\)-C\(_5\)H\(_5\))Co}\(_2\){μ:η\(^6\),η\(^6\)-(BC\(_5\)H\(_5\))\(_2\)}] (189) was successfully prepared by treatment of tetrabromodiborane (65) with six equivalents of cobaltocene (176) in a unique reaction that utilized cobaltocene as both a reagent and reductant (Figure 5-5). The bimetallic transition metal complex features a new bridging bis(boratabenzene) ligand linked through a boron-boron single bond that can manifest delocalization of electron density by providing an accessible LUMO orbital for π-communication between the cobalt centers and heteroaromatic rings.
This dianionic diboron ligand was shown to facilitate electronic coupling between the cobalt metal sites, as evidenced by the potential separations between successive single-electron redox events in the cyclic voltammogram. Four formal redox potentials for complex 189 were found: E\(_{1/2}\)(1) = −0.84 V, E\(_{1/2}\)(2) = −0.94 V, E\(_{1/2}\)(3) = −2.09 V, and E\(_{1/2}\)(4) = −2.36 V (relative to the Fc/Fc+ couple) (Figure 5-6). These potentials correlate to two closely-spaced oxidation waves and two well-resolved reduction waves ([(189)]\(^{0/+1}\), [(189)]\(^{+1/+2}\), [(189)]\(^{0/–1}\), and [(189)]\(^{–1/–2}\) redox couples, respectively). The extent of metal-metal communication was found to be relative to the charge of the metal atoms, with the negative charge being more efficiently delocalized across the bis(boratabenzene) unit (class II Robin-Day system). Magnetic studies indicate that the Co(II) ions are weakly antiferromagnetically coupled across the B-B bridge.
While reduction of the bis(boratabenzene) system resulted in decomposition of the complex, oxidation of the system by one- and two-electron steps resulted in isolable stable monocationic (194) and dicationic (195) forms of the bis(boratabenzene) complex (Figure 5-7). Study of these systems verified the results of the cyclic voltammetry studies performed on the neutral species. These species are unfortunately not stable in acetonitrile or nitromethane solutions, which until this point are the only solvents that have been observed to dissolve the cationic species. Unfortunately, this instability in solution complicates reactivity studies of these cationic complexes.
Finally, reactivity studies were performed on the neutral bis(boratabenzene) complex 189 in which the compound was tested for: (A) cleavage of the boratabenzene (cyclo-BC\(_5\)H\(_5\)) ring from the cobalt center, and (B) oxidative addition of the B-B bond to a transition metal scaffold to attempt synthesis of the first ever L\(_x\)M-η\(^1\)-(BC\(_5\)H\(_5\)) complex. Both of these reactivity studies, however, proved unsuccessful and typically witnessed decomposition of the bis(boratabenzene) complex or no reactivity. After repeated attempts of these reactions, no oxidative addition of the bis(boratabenzene) system could be confirmed.
