Transforming-Growth-Factor-beta1 (TGF-b1) is a multifunctional cytokine that regulates cell growth and differentiation in many types of cells. TGF-b1 is especially known to exert a variety of regulatory functions in the immune system, such as T cell differentiation and T cell function. Signal transduction of TGF-b1 is mediated by phosphorylation of receptorassociated Smad proteins (R-Smads). R-Smads are phosphorylated by the activated type I receptor, which is itself phosphorylated by the high affinity type II receptor upon ligand binding. The phosphorylated R-Smads then associate with Co-Smads. Heterooligomers of R- and Co-Smads translocate into the nucleus where they regulate transcription of target genes in concert with other transcription factors such as CBP/p300 or AP-1. Recent findings suggest that the pleiotropic effects of TGF-b1 are conferred by crosstalks to other signal transduction pathways such as the MAP-kinases or the STAT-pathway. Here we describe the effect of long-term exposure to TGF-b1 on the effector function of differentially stimulated primary murine splenocytes and purified primary murine CD8+ cytotoxic T cells. Long-term exposure to TGF-b1 results in non-responsiveness to TGF-b1- induced Smad2 phosphorylation. This is seen either by no phosphorylation or sustained phosphorylation of Smad2. Furthermore, we observed a strong correlation between sustained Smad2 phosphorylation and resistance to TGF-b1 mediated growth inhibition. In contrast, splenocyte cultures strongly growth inhibited by TGF-b1 showed no Smad2 phosphorylation. Lytic activity of these cultures, however, was found to be suppressed regardless of proliferation properties and Smad2 phosphorylation pattern. We also describe that a functional MEK-1 pathway is a prerequisite for rendering murine splenocytes unresponsive to TGF-b1 mediated growth inhibition, and that inhibition of the MEK-1 cascade alters the Smad2 phosphorylation pattern. In addition, we show that resistance to TGF-b1 mediated growth inhibition correlates with the activation of the JNK pathway. However, the resistant phenotype was found unable to be reverted upon administration of exogeneous IFNg and/or aCD28 antibody. In human or mouse T cell lines, however, the described correlation between the type of stimulation and TGF-b growth resistance or growth sensitivity is not present. Thus, this correlation is specific for primary T cells. We also cloned a chimeric dominantnegative TGF-b receptor which is coupled to a suicide gene, in order to render T cells resistant to TGF-b mediated effects.These findings shed light on how TGF-b1 mediates its immunosuppressive role, and may help to gain knowledge of averting these TGF-b1 effects in the course of tumor therapy.
In der vorliegenden Untersuchung wurde der Einfluss des AT1-R -Antagonisten Valsartan auf die Nierenfunktion bei nierentransplantierten Ratten mit der Fragestellung analysiert, ob eine Langzeittherapie mit diesem Wirkstoff einen positiven Effekt auf die Nierenfunktion entfaltet und sich somit sein Einsatz gegen die Entwicklung einer chronischen Transplantatnephropathie empfiehlt. Die über den gesamten Versuchszeitraum gegenüber der allogenen Kontrollgruppe signifikant erhöhten Urinvolumina stellen allein kein Indiz für eine bessere Nierenfunktion unter Therapie mit Valsartan dar. Dieses Ergebnis ist am ehesten durch Veränderungen der glomerulären Hämodynamik post transplantationem zu erklären. Wie nunmehr in mehreren tierexperimentellen Untersuchungen und klinischen Patientenstudien nachgewiesen worden ist, zeigt sich auch in der Synopsis der eigenen Befunde ein signifikant günstigerer Verlauf des Serumkreatinins, des Serum-BUN, der Kreatinin-Clearance sowie der Proteinurie unter Blutdrucksenkung mit dem AT1-R-Antagonisten Valsartan. An einigen Zeitpunkten der Studie waren die Ergebnisse allerdings statistisch nicht signifikant. Eine positive Wirkung auf die Transplantatfunktion und auf das Langzeitüberleben der Versuchstiere ist anzunehmen, ist aber in dieser Studie nicht weiter verfolgt worden. Eine Untersuchung mit einer größeren Anzahl von Versuchstieren und über einem längeren Versuchzeitraum hin scheint sinnvoll, um signifikante Unterschiede zwischen den Kontrollgruppen und der Versuchgruppe unter Valsartan zu belegen. Die im Vergleich zu den Kontrollgruppen geringere Entwicklung des Körpergewichts hatte bei der o.g. Fragestellung keine Relevanz. Wie in zahlreichen klinischen Studien für die Progredienz des chronischen Nierenversagens seit längerem eindrucksvoll belegt ist, scheint eine pharmakologische Blockade des RAAS auch einen protektiven Effekt auf die Entstehung einer chronischen Transplantatnephropathie zu entfalten. Die eigene Untersuchung liefert hinreichend Belege für diese Vermutung. Auch wenn in einzelnen Studien über negative Auswirkungen einer Blockade des RAAS auf die Transplantatfunktion berichtet worden ist, gibt es genügend Anhaltspunkte für einen günstigeren Verlauf nach Transplantation sowohl in Tierversuchen als auch für den transplantierten Patienten. Das allmähliche Fortschreiten der chronischen Transplantatnephropathie kann damit allerdings nicht ganz aufgehalten werden. Somit bleibt trotz dieser erfolgversprechenden experimentellen Ergebnisse nach Organtransplantation durch diese neuen Therapieansätze (Immunsuppressiva, RAAS-Blockade, Plasmapherese u.a.) die chronische Transplantat-Abstoßung immer noch ein therapeutisch fortbestehendes Problem. Weitere Untersuchungen über die Zusammenhänge immunologischer sowie nicht-immunologischer Ursachen einer chronischen Transplantatnephropathie und eine Optimierung der Immunsuppression sind deshalb auch weiterhin dringend erforderlich.
Die Alveoläre Echinokokkose ist eine bedeutende, gefährliche Parasitose des Menschen. Über die molekularen Grundlagen und Mechanismen der Wirt-Parasit- Interaktion ist bislang nur wenig bekannt. In den letzten Jahren konnten Hinweise erlangt werden, dass Wirt und Parasit über evolutionsgeschichtlich konservierte Signalsysteme kommunizieren. Eines dieser Systeme ist das TGF-b/BMP-Signaltransduktionssystem. TGF-β-Signaltransduktionskomponenten steuern grundlegende Prozesse der Entwicklung und Differenzierung in allen Tieren. Über dieses Signalsystem wird ein weites Spektrum von zellulären Prozessen wie Proliferation, Apoptose und Differenzierung reguliert. Dieses System besteht aus strukturell verwandten Zytokinen der TGF-β (transforming growth factor β) bzw. BMP (bone morphogenetic protein)-Familie, membranständigen Rezeptoren der TGF-β-Rezeptorfamilie (Typ I und Typ II) sowie intrazellulären Signaltransduktoren der Smad-Familie. Bislang konnten verschiedene Echinokokken Smad-Faktoren (EmSmadA, EmSmadB, EmSmadC und EmSmadD) sowie drei Echinokokken Rezeptoren der Typ I Familie (EmRSK1, EmRSK2, EmRSK3) in E. multilocularis identifiziert werden. Ein Mitglied der TGF-β Typ II-Rezeptorfamilie war bislang noch nicht beschrieben. In dieser Arbeit wird ein solches Molekül vorgestellt, EmRSK4 (=TGF-b Typ IISerin/ Threonin Kinase Rezeptor aus Echinococcus multilocularis). Genexpressionsanalysen und immunhistochemische Untersuchungen zeigen an, dass EmRSK4 in der Germinalschicht des E. multilocularis Metacestoden zusammen mit EmRSK1 (=BMP Typ I-Serin/Threonin Kinase Rezeptor) exprimiert wird. Studien an heterolog exprimierten Rezeptoren zeigten, dass EmRSK4 funktionell aktiv ist und mit humanen Typ I-Rezeptoren einen Komplex bilden kann. Diese Studien zeigen auch, dass EmRSK4 mit EmRSK1 einen aktiven heterologen Typ I-/Typ II-Rezeptorkomplex in HEK293-T Zellen bildet, der durch Wirts-BMP2 stimuliert wird und EmSmadB aktiviert. In Untersuchungen mit EmRSK2 (= TGF-β Typ ISerin/ Threonin Kinase Rezeptor) konnte gezeigt werden, dass bei Anwesenheit beider Rezeptoren, EmRSK2 und EmRSK4, eine Phosphorylierung von EmSmadC nachweisbar ist, während eine Phosphorylierung von EmSmadA auch ohne die Anwesenheit von EmRSK4 stattfindet. Desweiteren konnte gezeigt werden, dass der Inhibitor SB-431452 die Kinaseaktivität von EmRSK2 hemmt. Nach Zugabe von exogenem BMP2 zu Metazestodenvesikel konnten Hinweise erhalten werden, dass ein bislang noch nicht charakterisiertes, zusätzliches EmSmad aktiviert wird. Zusammengenommen lässt die Co-Expression von EmRSK1 mit EmRSK4 in der Germinalschicht, die Bildung eines BMP-responsiven Komplexes aus beiden Rezeptoren und die Phosphorylierung mindestens eines zellulären Faktors nach exogener Zugabe von Wirts-BMP2 zu Metacestodenvesikeln darauf schließen, dass beide Rezeptoren während einer Infektion an der Sensierung von BMP Signalen des Wirts beteiligt sein könnten
Purpose:
The biologic relevance of human connective tissue growth factor (hCTGF) for primary human tenon fibroblasts (HTFs) was investigated by RNA expression profiling using affymetrix (TM) oligonucleotide array technology to identify genes that are regulated by hCTGF.
Methods:
Recombinant hCTGF was expressed in HEK293T cells and purified by affinity and gel chromatography. Specificity and biologic activity of hCTGF was confirmed by biosensor interaction analysis and proliferation assays. For RNA expression profiling HTFs were stimulated with hCTGF for 48h and analyzed using affymetrix (TM) oligonucleotide array technology. Results were validated by real time RT-PCR.
Results:
hCTGF induces various groups of genes responsible for a wound healing and inflammatory response in HTFs. A new subset of CTGF inducible inflammatory genes was discovered (e.g., chemokine [C-X-C motif] ligand 1 [CXCL1], chemokine [C-X-C motif] ligand 6 [CXCL6], interleukin 6 [IL6], and interleukin 8 [IL8]). We also identified genes that can transmit the known biologic functions initiated by CTGF such as proliferation and extracellular matrix remodelling. Of special interest is a group of genes, e.g., osteoglycin (OGN) and osteomodulin (OMD), which are known to play a key role in osteoblast biology.
Conclusions:
This study specifies the important role of hCTGF for primary tenon fibroblast function. The RNA expression profile yields new insights into the relevance of hCTGF in influencing biologic processes like wound healing, inflammation, proliferation, and extracellular matrix remodelling in vitro via transcriptional regulation of specific genes. The results suggest that CTGF potentially acts as a modulating factor in inflammatory and wound healing response in fibroblasts of the human eye.
Controversy surrounds neutrophil function in cancer because neutrophils were shown to provide both pro-and antitumor functions. We identified a heterogeneous subset of low-density neutrophils (LDNs) that appear transiently in self-resolving inflammation but accumulate continuously with cancer progression. LDNs display impaired neutrophil function and immunosuppressive properties, characteristics that are in stark contrast to those of mature, high-density neutrophils (HDNs). LDNs consist of both immature myeloid-derived suppressor cells (MDSCs) and mature cells that are derived from HDNs in a TGF-beta-dependent mechanism. Our findings identify three distinct populations of circulating neutrophils and challenge the concept that mature neutrophils have limited plasticity. Furthermore, our findings provide a mechanistic explanation to mitigate the controversy surrounding neutrophil function in cancer.
Gliomas have been classified according to their histological properties. However, their respective cells of origin are still unknown. Neural progenitor cells (NPC) from the subventricular zone (SVZ) can initiate tumors in murine models of glioma and are likely cells of origin in the human disease. In both, p53 signaling is often functionally impaired which may contribute to tumor formation. Also, TGF-beta, which under physiological conditions exerts a strong control on the proliferation of NPCs in the SVZ, is a potent mitogen on glioma cells. Here, we approach on the crosstalk between p53 and TGF-beta by loss of function experiments using NPCs derived from p53 mutant mice, as well as pharmacological inhibition of TGF-beta signaling using TGF-beta receptor inhibitors. NPC derived from p53 mutant mice showed increased clonogenicity and more rapid proliferation than their wildtype counterparts. Further, NPC derived from p53\(^{mut/mut}\) mice were insensitive to TGF-beta induced growth arrest. Still, the canonical TGF-beta signaling pathway remained functional in the absence of p53 signaling and expression of key proteins as well as phosphorylation and nuclear translocation of SMAD2 were unaltered. TGF-beta-induced p21 expression could, in contrast, only be detected in p53\(^{wt/wt}\) but not in p53\(^{mut/mut}\) NPC. Conversely, inhibition of TGF-beta signaling using SB431542 increased proliferation of p53\(^{wt/wt}\) but not of p53\(^{mut/mut}\) NPC. In conclusion, our data suggest that the TGF-beta induced growth arrest in NPC depends on functional p53. Mutational inactivation of p53 hence contributes to increased proliferation of NPC and likely to the formation of hyperplasia of the SVZ observed in p53 deficient mice in vivo.
