Altersbedingte Makuladegeneration (AMD) ist weltweit die häufigste Ursache von irreversibler Erblindung des alternden Menschen. Mit der anti-VEGF-Behandlung steht für die deutlich seltenere feuchte AMD eine zugelassene Therapie bereit, die deutlich häufigere trockene AMD entzieht sich aktuell jedoch jeglicher Therapie. Ein zentraler Pathomechanismus der AMD ist der progrediente Untergang des retinalen Pigmentepithels (RPE). Die Rarifizierung und letztendlich Atrophie des RPEs führt zum Untergang der funktionellen Einheit aus RPE, Photorezeptoren und Bruch’scher Membran und somit zum irreversiblen Funktionsverlust. Ein möglicher therapeutischer Ansatz, der progredienten Atrophie des RPEs entgegenzuwirken, ist, das prinzipiell post- mitotischen RPE zur Proliferation anzuregen.
Grundlage unserer in vitro Untersuchungen ist das ARPE-19 Zellmodell. Um die Proliferation anzuregen wurden die RPE-Zellen mit E2F2, einem Zellzyklus- regulierendem Transkriptionsfaktor, transfiziert.
Zunächst wurde ein nicht-proliferatives RPE-Zellmodell mit spontanem Wachstumsarrest etabliert. Innerhalb von zwei Wochen konnte die Ausbildung von Zonulae occludentes als Zeichen der Integrität des adhärenten Zellmonolayers beobachtet werden. Die chemische Transfektion von E2F2 unter einem CMV-Promoter führte zur Überexpression von E2F2-Protein. Der proliferationssteigernde Effekt von E2F2 konnte durch die Proliferationsmarker Cyclin D1 sowie Ki67, dem Anstieg der BrdU-Aufnahme und der nach Transfektion mit E2F2 zunehmenden Gesamtzellzahl nachgewiesen werden.
Der Zellzahlerhöhung standen jedoch potentiell qualitative und funktionelle Einbußen entgegen. So zeigten sich nach Behandlung mit E2F2 die Zellviabilität reduziert und die Apoptoserate sowie die Permeabilität des Epithels erhöht. Diese Einschränkungen waren jedoch nur passager bis 7 Tage nach Transfektion sichtbar und reversibel. Unsere Ergebnisse weisen darauf hin, dass diese Defizite nicht durch E2F2 selbst, sondern durch das Transfektionsreagenz PEI bedingt waren. Weitere funktionelle Defizite könnten durch epithelial-mesenchymale Transition (EMT) verursacht werden. Hier zeigte sich durch E2F2 keine De-Differenzierung im Sinne einer typischen EMT-Marker- Expression.
Die vorliegende Arbeit zeigt in einem in vitro Zellmodell die Grundlagen eines vielversprechenden Ansatzes zur Therapie der trockenen AMD: Durch Überexpression eines den Zellzyklus regulierenden Gens (hier E2F2) wurde die RPE-Regeneration angeregt. Analog zur schon zugelassenen Gentherapie des RPEs bei RPE65-assoziierten Netzhautdystrophien durch den Transfer von funktionstüchtigem RPE65-Gen mittels Adeno-assoziierten Viren könnte mittels E2F2, übertragen mit einem lentiviralen Verktor, eine Stimulation des RPEs zur Proliferation möglich sein. Entscheidend ist der möglichst gute Struktur- und Funktionserhalt des Photorezeptor-Bruch-Membran-RPE Komplexes. Eine Therapie sollte daher in frühen Krankheitsstadien erfolgen, um die Progression zu fortgeschrittenen Erkrankungsstadien mit irreversiblem Funktionsverlust zu verzögern oder zu verhindern.
Pflanzen müssen sich während der Samenkeimung und Keimlingsentwicklung über eingelagerte Speicherstoffe heterotroph versorgen, bis sie, nach Etablierung ihres Photosyntheseapparats, einen autotrophen Lebensstil führen können.
Diese Arbeit geht von der Hypothese aus, dass der evolutionär konservierten zentral-metabolischen Kinase Snf1-RELATED PROTEIN KINASE 1 (SnRK1) eine besondere Rolle bei der Mobilisierung von Speicherstoffen während der Keimlingsentwicklung zukommt. Während die Bedeutung von SnRK1 als zentraler Regulator katabolischer Prozesse unter Energiemangel- und Stresssituationen bereits gezeigt wurde, war die Funktion von SnRK1 im Zusammenhang mit der Samenkeimung weitgehend ungeklärt. In dieser Arbeit konnte erstmals gezeigt werden, dass SnRK1 in Arabidopsis die Mobilisierung und Degradation von Speicherstoffen, insbesondere von Triacylglyceride (TAGs), Samenspeicherproteinen und Aminosäuren, steuert. Sowohl Studien zur Lokalisation von SnRK1:GFP-Fusionsproteinen als auch Kinaseaktivitätsassays unterstützen eine mögliche Funktion von SnRK1 während der Keimlingsentwicklung. Eine induzierbare snrk1-knockdown Mutante zeigt neben einem eingeschränkten Wurzel- und Hypokotylwachstum auch keine Ausbildung eines Photosyntheseapparats, was die zentrale Rolle der SnRK1 in diesem frühen Entwicklungsstadium untermauert. Durch Fütterungsexperimente mit Glukose konnte der Phänotyp einer snrk1 -Mutante in Keimlingen gerettet werden. Dies zeigt, dass der metabolische Block durch externe Gabe von Kohlenhydraten umgangen werden kann. Die zentrale Funktion von SnRK1 ist folgich der Abbau von Speicherstoffen und keine allgemeine Deregulation des pflanzlichen Stoffwechsels. Durch massenspektrometrische Untersuchungen von Keimlingen des Wildtyps und der snrk1-Mutante konnte gezeigt werden, dass TAGs in der Mutante in der spä- ten Keimlingsentwicklung ab Tag 4 langsamer abgebaut werden als im Wildtyp. Ebenso werden Samenspeicherproteine in der Mutante langsamer degradiert, wodurch die Verfügbarkeit von freien Aminosäuren in geringer ist. Entgegen der allgemeinen Annahme konnte gezeigt werden, dass während der Keimlingsentwicklung zumindest in Arabidopsis, einer ölhaltigen Pflanze, zunächst Kohlenhydrate in Form von Saccharose abgebaut werden, bevor die Degradation von TAGs und Aminosäuren beginnt. Diese Abbauprodukte können dann der Glukoneogenese zugeführt werden um daraus Glukose herzustellen. Mittels Transkriptom-Analysen konnten zentrale SnRK1-abhängige Gene in der Speicherstoffmobilisierung von TAG, beispielsweise PEROXISOMAL NAD-MALATE DEHYDROGENASE 2 (PMDH2) und ACYL-CoA-OXIDASE 4 (ACX4), und Aminosäuren identifiziert werden. Somit wurde ein Mechanismus der SnRK1-abhängigen Genregulation während der Samenkeimung in Arabidopsis gefunden. Bei der Degradation von Aminosäuren wird die cytosolische PYRUVATE ORTHOPHOSPHATE DIKINASE (cyPPDK), ein Schlüsselenzym beim Abbau bestimmter Aminosäuren und bei der Glukoneogenese, SnRK1-abhängig transkriptionell reguliert. Durch Koregulation konnte der Transkriptionsfaktor bZIP63 (BASIC LEUCINE ZIPPER 63) gefunden werden, dessen Transkription ebenfalls SnRK1-abhängig reguliert wird. Außerdem konnte die Transkription von cyPPDK in bzip63-Mutanten nur noch sehr schwach induziert werden. In Protoplasten konnte der cyPPDK-Promotor durch Aktivierungsexperimente mit bZIP63 und SnRK1α1 induziert werden. Durch Mutationskartierung und Chromatin-Immunopräzipitation (ChIP)PCR konnte mehrfach eine direkte Bindung von bZIP63 an den cyPPDK-Promotor nachgewiesen werden. Zusammenfassend ergibt sich ein mechanistisches Arbeitsmodell, in dem bZIP63 durch SnRK1 phosphoryliert wird und durch Bindung an regulatorische G-Box cis-Elemente im cyPPDK- Promotor dessen Transkription anschaltet. Infolgedessen werden Aminosäuren abgebaut und wird über die Glukoneogenese Glukose aufgebaut. Dieser Mechanismus ist essentiell für die Übergangsphase zwischen heterotropher und autotropher Lebensweise, und trägt dazu bei, die im Samen vorhandenen Ressourcen dem Keimling zum idealen Zeitpunkt zugänglich zu machen. Darüber hinaus werden Gene im Abbau von verzweigtkettigen Aminosäuren ebenfalls durch bZIP63 reguliert. Dabei wird dem Keimling Energie in Form von Adenosin-Triphosphat (ATP) zur Verfügung gestellt.
Zusammengefasst zeigen die Ergebnisse dieser Arbeit, dass die Mobilisierung von Speicherstoffen auch während der Keimlingsentwicklung direkt von SnRK1 abhängig ist. Die umfangreichen Datensätze der RNA-Seq-Analysen bieten zudem die Möglichkeit, weitere SnRK1-abhängige Gene der Speichermobilisierung zu identifizieren und somit einem besseren Verständnis der Keimlingsentwicklung beizutragen. Aufgrund der zentralen Bedeutung der SnRK1-Kinase in diesem entscheidenden Entwicklungsschritt ist davon auszugehen, dass diese Erkenntnisse mittelfristig auch für bessere Keimungsraten und somit bessere Erträge in der Landwirtschaft genutzt werden können.
Microorganisms that colonize the human body face large fluctuations in their surroundings. Therefore, those microbes developed sophisticated mechanisms that allow them to adapt their cell biology and maintain cellular homeostasis. One organelle vital to preserve cell physiology is the vacuole. The vacuole exhibits a wide range of functions and is able to adjust itself in response to both external and internal stimuli. Moreover, it plays an important role in host interaction and virulence in fungi such as Candida albicans. Despite this connection, only a few regulatory proteins have been described to modulate vacuolar biology in fungal pathogens. Furthermore, whether such regulation alters fungus-host interplay remains largely unknown.
This thesis focuses on the characterization of ZCF8, a fungus-specific transcription regulator in the human-associated yeast C. albicans. To this end, I combined genome-wide protein-DNA interaction assays and gene expression analysis that identified genes regulated by Zcf8p. Fluorescence microscopy uncovered that several top targets of Zcf8p localize to the fungal vacuole. Moreover, deletion and overexpression of ZCF8 resulted in alterations in vacuolar morphology and in luminal pH and rendered the fungus resistant or susceptible to a vacuole-disturbing drug. Finally, in vitro adherence assays showed that Zcf8p modulates the attachment of C. albicans to human epithelial cells in a vacuole-dependent manner.
Given those findings, I posit that the previously uncharacterized transcription regulator Zcf8p modulates fungal attachment to epithelial cells in a manner that depends on the status of the fungal vacuole. Furthermore, the results highlight that vacuolar physiology is a substantial factor influencing the physical interaction between Candida cells and mammalian mucosal surfaces.
Ubiquitination is a posttranslational modification with immense impact on a wide range of cellular processes, including proteasomal degradation, membrane dynamics, transcription, translation, cell cycle, apoptosis, DNA repair and immunity. These diverse functions stem from the various ubiquitin chain types, topologies, and attachment sites on substrate proteins. Substrate recruitment and modification on lysine, serine or threonine residues is catalyzed by ubiquitin ligases (E3s). An important E3 that decides about the fate of numerous substrates is the HECT-type ubiquitin ligase HUWE1. Depending on the substrate, HUWE1 is involved in different processes, such as cell proliferation and differentiation, DNA repair, and transcription. One of the transcription factors that is ubiquitinated by HUWE1 is the MYC interacting zinc finger protein 1 (MIZ1). MIZ1 is a BTB/POZ (Bric-à-brac, Tramtrack and Broad-Complex/Pox virus and zinc finger) zinc finger (ZF) protein that binds to DNA through its 13 C2H2-type zinc fingers and either activates or represses the transcription of target genes, including genes involved in cell cycle arrest, such as P21CIP1 (CDKN1A). The precise functions of MIZ1 depend on its interactions with the MYC-MAX heterodimer, but also its heterodimerization with other BTB-ZF proteins, such as BCL6 or NAC1. How MIZ1 interacts with HUWE1 has not been studied and, as a consequence, it has not been possible to rationally develop tools to manipulate this interaction with specificity in order to better understand the effects of the interaction on the transcriptional function of MIZ1 on target genes or processes downstream. One aspect of my research, therefore, aimed at characterizing the MIZ1-HUWE1 interaction at a structural level. I determined a crystal structure of the MIZ1-BTB-domain in complex with a peptide, referred to as ASC, derived from a C terminal region of HUWE1, previously named ‘activation segment’. The binding mode observed in this crystal structure could be validated by binding and activity assays in vitro and by cell-based co-IP experiments in the context of N-terminally truncated HUWE1 constructs. I was not able to provide unambiguous evidence for the identified binding mode in the context of full-length HUWE1, indicating that MIZ1 recognition by HUWE1 requires yet unknown regions in the cell. While the structural details of the MIZ1-HUWE1 interaction remains to be elucidated in the context of the full-length proteins, the binding mode between MIZ1BTB and ASC revealed an interesting, atypical structural feature of the BTB domain of MIZ1 that, to my knowledge, has not been described for other BTB-ZF proteins: The B3 region in MIZ1BTB is conformationally malleable, which allows for a HUWE1-ASC-peptide-mediated β-sheet extension of the upper B1/B2-strands, resulting in a mixed, 3 stranded β-sheet. Such β-sheet extension does not appear to occur in other homo- or heterodimeric BTB-ZF proteins, including MIZ1-heterodimers, since these proteins typically possess a pre-formed B3-strand in at least one subunit. Instead, BCL6 co repressor-derived peptides (SMRT and BCOR) were found to extend the lower β-sheet in BCL6BTB by binding to an adjacent ‘lateral groove’. This interaction follows a 1:1 stoichiometry, whereas the MIZ1BTB-ASC-complex shows a 2:1 stoichiometry. The crystal structure of the MIZ1BTB-ASC-complex I determined, along with comparative binding studies of ASC with monomeric, homodimeric, and heterodimeric MIZ1BTB variants, respectively, suggests that ASC selects for MIZ1BTB homodimers. The structural data I generated may serve as an entry point for the prediction of additional interaction partners of MIZ1 that also have the ability to extend the upper β-sheet of MIZ1BTB. If successful, such interaction partners and structures thereof might aid the design of peptidomimetics or small-molecule inhibitors of MIZ1 signaling. Proof-of-principle for such a structure-guided approach targeting BTB domains has been provided by small-molecule inhibitors of BCL6BTB co-repressors interactions. If a similar approach led to molecules that interfere with specific interactions of MIZ1, they would provide intriguing probes to study MIZ1 biology and may eventually allow for the development of MIZ1-directed cancer therapeutics.
Die Regulation der Genexpression steht am Anfang vieler zellbiologischer Prozesse wie beispielsweise dem Zellwachstum oder der Differenzierung. Gene werden an Promotoren transkribiert, wobei ein Promotor selbst aus vielen logischen Einheiten aufgebaut ist, den Transkriptionsfaktorbindestellen (TFBSs). Diese können sehr nah beieinander liegen, aber auch weit entfernt voneinander sein. Sie werden spezifisch von Transkriptionsfaktoren (TFs) gebunden, die die Transkritptionsrate z.B. verstärken (Enhancer) oder schwächen (Silencer) können. Zwei oder mehr dieser TFBSs mit bestimmtem Abstand werden als "Module" zusammengefasst, die über Spezies hinweg konserviert sein können. Typischerweise findet man Module in Zellen mit einem Zellkern. Spezies mit gemeinsamen Modulen können ein Hinweis auf die gemeinsame phylogenetische Abstammung darstellen, aber auch gemeinsame Funktionsmechanismen von TFs über Gene hinweg aufdecken. Heutzutage sind verschiedene Anwendungen verfügbar, mit denen nach TFBSs in DNA gesucht werden kann. Zum Zeitpunkt des Verfassens dieser Arbeit sind aber nur zwei kommerzielle Produkte bekannt, die nicht nur TFBSs, sondern auch Module erkennen. Deshalb stellen wir hier die freie und quelloffene Lösung "AIModules" vor, die diese Lücke füllt und einen Webservice zur Verfügung stellt, der es erlaubt nach TFBSs sowie nach Modulen auf DNA- und auf RNA-Abschnitten zu suchen. Für die Motivesuche werden entweder Matrizen aus der Jaspar Datenbank oder Matrizen vom Anwender verwendet. Darüberhinaus zeigen wir, dass unser Tool für die TF Suche nur Sekunden benötigt, wohingegen conTraV3 mindestens eine Stunde für dieselbe Analyse braucht. Zusätzlich kann der Anwender bei unserem Tool den Grad der Konserviertheit für TFs mit angeben und wir zeigen, dass wir mit unserer Lösung, die die Jaspar Datenbank heranzieht, mehr Module finden, als ein kommerziell verfügbares Produkt. Weiterhin kann mit unserer Lösung auch auf RNA-Sequenzen nach regulatorischen Motiven gesucht werden, wenn der Anwender die dafür nötigen Matrizen liefert. Wir zeigen dies am Beispiel von Polyadenylierungsstellen. Zusammenfassend stellen wir ein Werkzeug vor, das erstens frei und quelloffen ist und zweitens entweder auf Servern veröffentlicht werden kann oder On-Site auf einem Notebook läuft. Unser Tool erlaubt es Promotoren zu analysieren und nach konservierten Modulen sowie TFBSs in Genfamilien sowie nach regulatorischen Elementen in mRNA wie z.B. Polyadenylierungsstellen oder andere regulatorische Elemente wie beispielsweise Enhancern oder Silencern in genomischer DNA zu suchen.
In der vorliegenden Arbeit wurde die Rolle der Transkriptionsfaktoren NFATc1/αA bzw. NFATc1/ßC und NFATc2 in NK-Zellen und die Rolle der Faktoren NFATc1-4 und NFAT5 in Keratinozyten analysiert. Die Familie der Nuclear Factor of Activated T-cell (NFAT) Transkriptionsfaktoren besteht aus fünf Mitgliedern, welche entscheidend die Gentranskription bei Immunantworten beeinflussen. Nach Antigenaktivierung wird in Lymphozyten die Expression des Nfatc1-Gens stark induziert. Die Akkumulation der dabei gebildeten kurzen Isoform NFATc1/αA ist für die Zytokinproduktion als Effektorfunktion sowie für die Proliferation und das Überleben aktivierter Zellen verantwortlich. Das kurze NFATc1-Protein unterscheidet sich nicht nur strukturell, sondern auch funktionell von fast allen anderen NFAT-Proteinen. Um neue Erkenntnisse über die Funktion der Isoform NFATc1/αA in Lymphozyten zu gewinnen, wurden KT12 NK-Zellen mit NFATc1/αA bzw. NFATc1/ßC und NFATc2- exprimierenden Vektoren transfiziert, geklont, stimuliert und anschließend auf ihre jeweilige Apoptoserate und die Zytokinsynthese bzw. -expression hin untersucht. Die gewählten KT12-Hybridoma-Zellen erwiesen sich allerdings in ihrer intendierten Funktion als Testzellen für die Über-Expression von NFATc-Proteinen in NK-Zellen als ungeeignet.
Fehlregulationen von NFAT-Signalwegen werden mit einer fehlerhaften Entwicklung des Immunsystems, mit der Entstehung von Autoimmunerkrankungen und mit Krebs in Verbindung gebracht. Um die Rolle von NFAT-Faktoren in Keratinozyten besser zu verstehen, wurden HaCaT-Zellen und primäre humane Keratinozyten mit Differenzierungssignalen bzw. UVB-Licht stimuliert. Änderungen der Transkription von NFAT-Faktoren, Keratinozyten-spezifischen Proteinen und Chemokinen wurden mittels qRT-PCR-Assays detektiert und analysiert. Insgesamt konnte die Beteiligung von NFAT-Faktoren am Differenzierungsprozess und an der UV-Antwort von Keratinozyten gezeigt werden. Es zeigte sich tendenziell eine stärkere Induktion der kurzen NFATc1-Isoform im Vergleich zu langen NFATc1-Isoformen, was die Frage nach einer besonderen Funktion der kurzen NFATc1-Isoform in Keratinozyten aufwirft. Generell ließen sich besonders hohe Expressionslevel des Transkriptionsfaktors NFAT5 - verglichen mit anderen NFAT-Faktoren - messen. Für die Entwicklung von Therapien, welche die Ursachen und Folgen einer dysregulierten Hautbarrierenbildung behandeln, könnten sich weitere Studien zu einzelnen NFAT-Faktoren bzw. NFATc1-Isoformen als zielführend erweisen.
Es wurden mögliche frühzeitige Immunoseneszenzparameter bei Kindern mit Autismus und ADHS im Vergleich zu gesunden Kontrollen mittels real-time quantitativer PCR ermittelt. Zusätzlich wurden T-Zell-spezifische Transkriptionsfaktoren mittels real-time quantitativer PCR gemessen. Es zeigte sich bei Autismus, im Vergleich zu gesunden Kontrollen, eine signifikant erniedrigte Anzahl von TRECs und eine signifikant erhöhte Expression des Th2-Zell-Transkriptionsfaktors GATA3. Schlußendlich scheint es eine schnellere Alterung des Thymus bei Kindern mit Autismus zu geben. Weitere Untersuchungen zur genaueren Differenzierung sind notwendig.
Gene expression and transfer of the genetic information to the next generation forms the basis of cellular life. These processes crucially rely on DNA, thus the preservation, transcription and translation of DNA is of fundamental importance for any living being. The general transcription factor TFIIH is a ten subunit protein complex, which consists of two subcomplexes: XPB, p62, p52, p44, p34, and p8 constitute the TFIIH core, CDK7, CyclinH, and MAT1 constitute the CAK. These two subcomplexes are connected via XPD. TFIIH is a crucial factor involved in both, DNA repair and transcription. The central role of TFIIH is underlined by three severe disorders linked to failure of TFIIH in these processes: xeroderma pigmentosum, Cockayne syndrome, and trichothiodystrophy. Only limited structural and functional data of TFIIH are available so far. Here, the model organism Chaetomium thermophilum was utilized with the aim to structurally and functionally characterize TFIIH. By combining the expression and purification of single TFIIH subunits with the co-expression and co-purification of dual complexes, a unique and powerful modular system of the TFIIH core subunits could be established, encompassing all proteins in high quality and fully functional. This system permits the step-wise assembly of TFIIH core, thereby making it possible to assess the influence of the intricate interaction network within TFIIH core on the overall enzymatic activities of TFIIH, which has not been possible so far. Utilizing the single subunits and dual complexes, a detailed interaction network of TFIIH core was established, revealing the crucial role of the p34 subunit as a central scaffold of TFIIH by linking the two proteins p44 and p52. Our studies also suggest that p62 constitutes the central interface of TFIIH to the environment rather than acting as a scaffold. TFIIH core complexes were assembled and investigated via electron microscopy. Preliminary data indicate that TFIIH adopts different conformational states, which are important to fulfill its functions in transcription and DNA repair. Additionally, a shortened construct of p62 was used to develop an easy-to-use, low cost strategy to overcome the crystallographic phase problem via cesium derivatization.
Oxylipine werden in der Pflanze unter Stressbedingungen gebildet. Die dafür notwendige Oxidation von Fettsäuren wird entweder nicht-enzymatisch über Radikale wie reaktive Sauerstoffspezies (ROS) oder enzymatisch über Lipoxygenasen katalysiert. Abhängig von der Position der Oxidation in der Fettsäure entstehen dabei C13- oder C9-Oxylipine. Sehr gut erforscht sind C13-Oxylipine wie Jasmonsäure (JA), die bei biotischem Stress und Verwundung gebildet werden und bei exogener Gabe das Wurzelwachstum von Arabidopsis thaliana hemmen. Die C9-Oxylipine wie 9-Hydroxyoktadekatriensäure (9-HOT) sind erst wenig erforscht. Ziel dieser Arbeit war die Charakterisierung von Transkriptionsfaktoren, mit dem Fokus auf 9-HOT-vermittelte Signalwegen in Arabidopsis thaliana. Da bekannt ist, dass auch sie zu einer Hemmung des Wurzelwachstums führen, wurde dazu die Untersuchung des Wurzelwachstums von 10 Tage alten Keimlingen etabliert. Funktionsgewinn-Mutanten des Transkriptionsfaktors TGA5 sowie des TGA5-Zielgens CYTOCHROM P450 MONOOXYGENASE CYP81D11 zeigten auf 9-HOT ein verglichen mit Col-0 deutlich besseres Wurzelwachstum. Die AtTORF-Ex-Kollektion, eine große Sammlung an Überexpressions-Linien verschiedener Transkriptionsfaktoren, wurde hinsichtlich Wurzelwachstums auf dem Oxylipin 9-HOT analysiert. Die Gesamtheit der untersuchten Pflanzen enthielt 263 unabhängige TF-Expressions-Konstrukte. Von 6087 untersuchten Pflanzen zeigten 201 Pflanzen keine Hemmung des Wurzelwachstums auf 9-HOT. Dabei konnten 80 verschiedene Transkriptionsfaktoren identifiziert werden, deren Überexpression die Wurzelwachstums-hemmende Wirkung von 9-HOT kompensiert. Es zeigte sich eine Häufung von Transkriptionsfaktoren der ERF- (ethylene responsive factor) Familie. Die verstärkte Expression der nahe verwandten Transkriptionsfaktoren ERF106 und ERF107 ermöglichte sowohl auf 9-HOT als auch auf 9-KOT ein längeres Wurzelwachstum im Vergleich zum Wildtyp. Die Genexpression von ERF106 und ERF107 wird durch Überflutung aktiviert. Durch Überflutung wird im Wildtyp die Expression von Hypoxia-Antwort-Genen wie HRE1, SUS4 oder PDC1 induziert. In den Funktionsverlust-Mutanten sind diese Gene in der Expression aber nicht beeinflusst. Auch ist nach Überflutung im normalen Tag / Nacht-Rhythmus kein signifikanter Unterschied im Überleben zwischen Col-0 und den Mutanten erf106, erf107 und erf106xerf107 nachweisbar. Zur Identifikation möglicher Ziel-Gene von ERF106 und ERF107 wurden Transkriptom-Analysen durchgeführt. Die Funktionsverlust-Mutanten erf106, erf107 und erf106xerf107 zeigten weder im Grundzustand noch nach 4 Stunden Überflutung Veränderungen in den bekannten Hypoxia-Antwort-Genen. Die Funktionsgewinn-Mutanten von ERF106 und ERF107 zeigten in der Transkriptom-Analyse eine deutliche Aktivierung von Genen, die wichtig für Entgiftung und Stressabwehr sind. Ebenso wurden wichtige Biosynthese-Gene aus der Camalexin- und Glukosinolat-Synthese in den Funktionsgewinn-Mutanten verstärkt exprimiert. Des Weiteren konnte eine verringerte Expression von Genen beobachtet werden, die wichtig für die Regulation der Eisen-Aufnahme sind, darunter bHLH-Transkriptionsfaktoren, der Eisen-Transporter IRON REGULATED TRANSPORTER 1 (IRT1) und die Eisen-Reduktase FERRIC REDUCTION OXIDASE 2 (FRO2). Zusammenfassend wurden in dieser Arbeit durch die Untersuchung der AtTORF-Ex-Kollektion mehrere TF identifiziert, die wichtige Abwehr-Gene gegen Stress- und Vergiftung sowie bedeutende Gene im Bereich der Biosynthese und Eisenaufnahme regulieren können, um so die Antwort auf C9-Oxylipine zu beeinflussen.
N-MYC is a member of the human MYC proto-oncogene family, which comprises three transcription factors (C-, N- and L-MYC) that function in multiple biological processes. Deregulated expression of MYC proteins is linked to tumour initiation, maintenance and progression. For example, a large fraction of neuroblastoma displays high N-MYC levels due to an amplification of the N-MYC encoding gene. MYCN-amplified neuroblastoma depend on high N-MYC protein levels, which are maintained by Aurora-A kinase. Aurora-A interaction with N-MYC interferes with degradation of N-MYC via the E3 ubiquitin ligase SCFFBXW7. However, the underlying mechanism of Aurora-A-mediated stabilisation of N-MYC remains to be elucidated.
To identify novel N-MYC interacting proteins, which could be involved in N-MYC stabilisation by Aurora-A, a proteomic analysis of purified N-MYC protein complexes was conducted. Since two alanine mutations in MBI of N-MYC, T58A and S62A (N-MYC mut), disable Aurora-A-mediated stabilisation of N-MYC, N-MYC protein complexes from cells expressing either N-MYC wt or mut were analysed. Proteomic analysis revealed that N-MYC interacts with two deubiquitinating enzymes, USP7 and USP11, which catalyse the removal of ubiquitin chains from target proteins, preventing recognition by the proteasome and subsequent degradation. Although N-MYC interaction with USP7 and USP11 was confirmed in subsequent immunoprecipitation experiments, neither USP7, nor USP11 was shown to be involved in the regulation of N-MYC stability. Besides USP7/11, proteomic analyses identified numerous additional N-MYC interacting proteins that were not described to interact with MYC transcription factors previously. Interestingly, many of the identified N-MYC interaction partners displayed a preference for the interaction with N-MYC wt, suggesting a MBI-dependent interaction. Among these were several proteins, which are involved in three-dimensional organisation of chromatin domains and transcriptional elongation by POL II. Not only the interaction of N-MYC with proteins functioning in elongation, such as the DSIF component SPT5 and the PAF1C components CDC73 and CTR9, was validated in immunoprecipitation experiments, but also with the POL III transcription factor TFIIIC and topoisomerases TOP2A/B. ChIP-sequencing analysis of N-MYC and TFIIIC subunit 5 (TFIIIC5) revealed a large number of joint binding sites in POL II promoters and intergenic regions, which are characterised by the presence of a specific motif that is highly similar to the CTCF motif. Additionally, N-MYC was shown to interact with the ring-shaped cohesin complex that is known to bind to CTCF motifs and to assist the insulator protein CTCF. Importantly, individual ChIP experiments demonstrated that N-MYC, TFIIIC5 and cohesin subunit RAD21 occupy joint binding sites comprising a CTCF motif.
Collectively, the results indicate that N-MYC functions in two biological processes that have not been linked to MYC biology previously. Furthermore, the identification of joint binding sites of N-MYC, TFIIIC and cohesin and the confirmation of their interaction with each other suggests a novel function of MYC transcription factors in three-dimensional organisation of chromatin.
In contrast to normal vessels, tumor vasculature is structurally and functionally abnormal. Tumor vessels are highly disorganized, tortuous and dilated, with uneven diameter and excessive branching. Consequently, tumor blood flow is chaotic, which leads to hypoxic and acidic regions in tumors. These conditions lower the therapeutic effectiveness and select for cancer cells that are more malignant and metastatic. The therapeutic outcome could be improved by increasing the functionality and density of the tumor vasculature. Tumor angiogenesis also shows parallels to epithelial to mesenchymal transition (EMT), a process enabling metastasis. Metastasis is a multi-step process, during which tumor cells have to invade the surrounding host tissue to reach the circulation and to be transported to distant sites.
We hypothesize that the variability in the phenotype of the tumor vasculature is controlled by the differential expression of key transcription factors. Inhibiting these transcription factors might be a promising way for angiogenic intervention and vascular re-engineering. Therefore, we investigated the interdependence of tumor-, stroma- and immune cell-derived angiogenic factors, transcription factors and resulting vessel phenotypes. Additionally, we evaluated whether transcription factors that regulate EMT are promising targets for vascular remodeling.
We used formalin fixed paraffin embedded samples from breast cancer patients, classified according to estrogen-, progesterone- and human epidermal growth factor receptor (HER) 2 status. Establishing various techniques (CD34 staining, laser microdissection, RNA isolation and expression profiling) we systematically analyzed tumor and stroma-derived growths factors. In addition, vascular parameters such as microvessel size, area, circularity and density were assessed. Finally the established expression profiles were correlated with the observed vessel phenotype. As the SNAI1 transcriptional repressor is a key regulator of EMT, we examined the effect of vascular knockdown of Snai1 in murine cancer models (E0771, B16-F10 and lewis lung carcinoma).
Among individual mammary carcinomas, but not among subtypes, strong differences of vascular parameters were observed. Also, little difference between lobular carcinomas and ductal carcinomas was found. Vessel phenotype of Her2 enriched carcinomas was similar to that of lobular carcinomas. Vessel morphology of luminal A and B and basal-like tumors resembled each other. Expression of angiogenic factors was variable across subtypes. We discovered an inverse correlation of PDGF-B and VEGF-A with vessel area in luminal A tumors. In these tumors expression of IL12A, an inhibitor of angiogenesis, was also correlated with vessel size. Treatment of endothelial cells with growth factors revealed an increased expression of transcription factors involved in the regulation of EMT. Knockdown of Snai1 in endothelial cells of mice increased tumor growth and decreased hypoxia in the E0771 and the B16-F10 models. In the lewis lung carcinomas, tumor vascularity and biodistribution of doxorubicin were improved. Here, doxorubicin treatment in combination with the endothelial cell-specific knockdown did slow tumor growth. This shows that SNAI1 is important for a tumor's vascularization, with the significance of its role depending on the tumor model.
The methods established in this work open the way for the analysis of the expression of key transcription factors in vessels of formalin fixed paraffin embedded tumors. This research enables us to find novel targets for vascular intervention and to eventually design novel targeted drugs to inhibit these targets.
Several important cellular processes, including transcription, nucleotide excision repair and cell cycle control are mediated by the multifaceted interplay of subunits within the general transcription factor II H (TFIIH).
A better understanding of the molecular structure of TFIIH is the key to unravel the mechanism of action of this versatile protein complex within these pathways. This becomes especially important in the context of severe diseases like xeroderma pigmentosum, Cockayne syndrome and trichothiodystrophy, that arise from single point mutations in some of the TFIIH subunits.
In an attempt to structurally characterize the TFIIH complex, we harnessed the qualities of the eukaryotic thermophile Chaetomium thermophilum, a remarkable fungus, which has only recently been recognized as a novel model organism. Homologues of TFIIH from C. thermophilum were expressed in E. coli, purified to homogeneity and subsequently utilized for crystallization trials and biochemical studies.
The results of the present work include the first crystal structure of the p34 subunit of TFIIH, comprising the N-terminal domain of the protein. The structure revealed a von Willebrand Factor A (vWA) like fold, which is generally known to be involved in a multitude of protein-protein interactions. Structural comparison allowed to delineate similarities as well as differences to already known vWA domains, providing insight into the role of p34 within TFIIH. These results indicate that p34 assumes the role of a structural scaffold for other TFIIH subunits via its vWA domain, while likely serving additional functions, which are mediated through its
C-terminal zinc binding domain and are so far unknown.
Within TFIIH p34 interacts strongly with the p44 subunit, a positive regulator of the XPD helicase, which is required for regulation of RNA Polymerase II mediated transcription and essential for eukaryotic nucleotide excision repair. Based on the p34 vWA structure putative protein-protein interfaces were analyzed and binding sites for the p34 p44 interaction suggested. Continuous crystallization efforts then led to the first structure of a p34 p44 minimal complex, comprising the N-terminal vWA domain of p34 and the C-terminal C4C4 RING domain of p44. The structure of the p34 p44 minimal complex verified the previous hypothesis regarding the involved binding sites. In addition, careful analysis of the complex interface allowed to identify critical residues, which were subsequently mutated and analyzed with respect to their significance in mediating the p34 p44 interaction, by analytical size exclusion chromatography, electrophoretic mobility shift assays and isothermal titration calorimetry. The structure of the p34 p44 complex also revealed a binding mode of the p44 C4C4 RING domain, which differed from that of other known RING domains in several aspects, supporting the hypothesis that p44 contains a novel variation of this domain.
Hintergrund: Die Überexpression von Hypoxia Inducible Factor 1 (HIF-1) wird mit Tumorprogression und schlechter Prognose in Zusammenhang gebracht. Wir untersuchten, ob die pharmakologische Hemmung des Transkriptionsfaktors HIF-1 mittels Chetomin, einem Inhibitor der Interaktion von HIF-1 mit dem Koaktivator Protein p300, die Hypoxie-induzierte Strahlenresistenz menschlicher Fibrosarkomzellen vom Typ HT 1080 beeinflusst.
Methoden: Die optimale Dosis von Chetomin wurde durch Versuchsreihen mit Hypoxie-sensiblem Promotor in mit destabilisiertem EGFP-Vektor transfizierten HT 1080 HRE-Zellen bestimmt. HT 1080 Zellen wurden mittels RT-PCR sowie Western Blot auf die Transkription der HIF-1-regulierten Gene Carboanhydrase IX (CA9) und Vascular Endothelial Growth Factor (VEGF) untersucht. Außerdem wurden sie zur Erstellung klonogener Assays unter normoxischen sowie hypoxischen (0,1% O2, 12 Stunden) Bedingungen in vitro mit 0, 2, 5 oder 10 Gy bestrahlt mit oder ohne Chetominbehandlung (150 nM, 12 Stunden, Vorbehandlung 4 Stunden).
Ergebnisse: In der RT-PCR zeigte sich eine signifikante Reduktion (Signifikanzniveau p<0,05) der mRNS-Expression von CA9 und VEGF unter Chetomin und Hypoxie auf 44,4 +/- 7,2% beziehungsweise 39,6 +/- 16,0%, im Western Blot supprimierte Chetomin auch die Downstream-Genprodukte von CA9 und VEGF. In den Überlebenskurven erhöhte Chetomin die Wirksamkeit der Bestrahlung wesentlich, der modifizierte Sauerstoffeffekt (modified Oxygen Enhancement Ratio, OER') war mit Ausnahme der 50% SF in Bezug auf die Kontrollen bei 50%, 37% und 10% Relativem Überleben (SF) von 1,57 auf 1,58, von 1,56 auf 1,42 und von 1,38 auf 1,22 reduziert.
Schlussfolgerung: Die HIF-1-Hemmung durch Chetomin reduziert effektiv die Hypoxie-abhängige Transkription und verstärkt die Strahlensensibilität von hypoxischen HT 1080 Fibrosarkomzellen in vitro.
Identifying novel driver genes in cancer remains a crucial step towards development of new therapeutic approaches and the basic understanding of the disease.
This work describes the impact of the AP1 transcription activator component FOSL1 on melanoma maintenance. FOSL1 is strongly upregulated during the progression of melanoma and the protein abundance is highest in metastases. I found that the regulation of FOSL1 is strongly dependent on ERK1/2- and PI3K- signaling, two pathways frequently activated in melanoma. Moreover, the involvement of p53 in FOSL1 regulation in melanoma was investigated. Elevated levels of the tumor suppressor led to decreased FOSL1 protein levels in a miR34a/miR34c- dependent manner.
The benefit of elevated FOSL1 amounts in human melanoma cell lines was analyzed by overexpression of FOSL1 in cell lines with low endogenous FOSL1 levels. Enhanced levels of FOSL1 had several pro-tumorigenic effects in human melanoma cell lines. Besides increased proliferation and migration rates, FOSL1 overexpression induced the colony forming ability of the cells. Additionally, FOSL1 was necessary for anchorage independent growth in 3D cell cultures. Microarray analyses revealed novel downstream effectors of FOSL1. On the one hand, FOSL1 was able to induce the transcription of different neuron-related genes, such as NEFL, NRP1 and TUBB3. On the other hand, FOSL1 influenced the transcription of DCT, a melanocyte specific gene, in dependence of the differentiation of the melanoma cell line, indicating dedifferentiation.
Furthermore, FOSL1 induced the transcription of HMGA1, a chromatin remodeling protein with reprogramming ability, which is characteristic for stem cells. Consequently, the influence of HMGA1 on melanoma maintenance was investigated. In addition to decreased proliferation and reduced anoikis resistance, HMGA1 knockdown reduced melanoma cell survival. Interestingly, the FOSL1 induced pro-tumorigenic effects were demonstrated to be dependent on the HMGA1 level. HMGA1 manipulation reversed FOSL1 induced proliferation and colony forming ability, as well as the anchorage independent growth effect.
In conclusion, I could show that additional FOSL1 confers a clear growth benefit to melanoma cells. This benefit is attributed to the induction of stem cell determinants, but can be blocked by the inhibition of the ERK1/2 or PI3K signaling pathways.
Die zunehmende Versalzung des Bodens führt weltweit zu starken Ernteeinbußen. Ob- wohl die Wurzeln der Pflanzen als erstes mit dem Salzstress in Berührung kommen, ist noch nicht viel über Signaltransduktionswege in Wurzeln zur Anpassung der Pflanze an Salzstress bekannt. Die bZIP-Transkriptionsfaktoren der Gruppe S1, bZIP1 und bZIP53, werden gewebespezifisch in der Wurzel nach Salzstress aktiviert. In dieser Arbeit werden diese bZIPs in ein Netzwerk eingeordnet, von der Aktivierung der Tran- skriptionsfaktoren bis zur Funktion in der Regulation des Stoffwechsels in der salzgest- ressten Pflanze.
Die Aktivierung von bZIP1 kann über verschiedene sowohl ionische als auch osmotische Stimuli erfolgen und ist abhängig von Calcium, der HEXOKINASE 1 und SnRK1- Kinasen (Snf1 RELATED PROTEIN KINASE 1). Die dunkelinduzierte Expression von bZIP1 wird HXK1-abhängig durch Glucose inhibiert, bei Energiemangelbedingungen ist die Aktivierung von bZIP1 SnRK1-abhängig. Beide Enzyme spielen auch in der salzinduzierten Expression von bZIP1 eine Rolle. Über Transkriptom- und Me- tabolomanalysen kann gezeigt werden, dass bZIP1 und bZIP53 an der Umprogram- mierung des Kohlenhydrat- und Aminosäuremetabolismus teilhaben. Besonders Gene der Glukoneogenese (PYRUVAT ORTHOPHOSPHAT DIKINASE und FRUCTOSE- 1,6-BISPHOS- PHATASE) bzw. des Aminosäurekatabolismus (BRANCHED- CHAIN AMINO ACID TRANSAMINASE 2, METHYLCROTONYL- COA-CARBOXYLASE A und HOMOGENTISATE 1,2-DIOXYGENASE ) werden von den Transkriptionsfaktoren reguliert. Das spricht für eine Umprogrammierung des Metabolismus und der Mobilisierung von Energie aus Aminosäuren zur Anpassung an die Stressbedingungen. Die Transkriptionsfaktoren der Gruppe S1 bilden vorzugsweise Heterodimere mit der Gruppe C. Mit Mutantenanalysen, die zum einen die Transkriptionsfaktoren des C/S1-Netzwerks und zum anderen Komponenten der Abscisinsäure (ABA) abhängigen Signaltransduktion beinhalten, konnte ein Signaltransduktionsnetzwerk aufgestellt werden, das die Antwort auf abiotischen Stress mittels des Signalwegs über ABA, SnRK2 und AREB (ABA RESPONSIVE ELEMENTS-BINDING PROTEIN) mit der SnRK1-vermittelten Antwort auf Energiemangelbedingungen in der Pflanze verknüpft. Die gefundenen stress- bzw. energieresponsiven Gene konnten nach den Mutantenana- lysen auf Grund ihrer unterschiedlichen Regulation in vier Klassen eingeteilt werden, wovon nur eine, die Klasse 4, von dem C/S1 Netzwerk reguliert wird. Die Klassen 1- 3 sind unabhängig von den bZIP-Transkriptionsfaktoren der Gruppe C. Die Klasse 1 bilden typische ABA-responsive Gene, die von den Gruppe A-bZIPs reguliert werden. Faktoren der Gruppe A sind auch an der Expression der Gene der Klasse 2 beteiligt, diese werden aber auch durch bZIP1 und bZIP53 induziert. Dieser Klasse konnten Gene zugeordnet werden, die im Abbau verzweigtkettiger Aminosäuren eine Rolle spielen. Am Aminosäureabbau sind außerdem die Gene der Klasse 2 beteiligt. Für diese Gene konnte eine Expressionsregulation durch bZIP1 und bZIP53 gezeigt werden. Für die Bestimmung möglicher Heterodimerisierungspartner bedarf es noch weiterer Analysen. Dieses Model, das den abitoschen Stress abhängigen ABA-Signalweg mit dem ener- gieabhängigen SnRK1-Signaltransduktionsweg verknüpft, zeigt die präzise Regulation von mindestens 4 Gen-Klassen, deren Expression durch die Kombination verschiedener bZIP-Transkriptionsfaktoren aktiviert wird.
While numerous experiments on NFAT were already performed with CD4+ T cells showing defective cytokine release and a reduced T helper cell development, no detailed studies existed for CD8+ T cells. From this point, we wanted to examine the impact of NFATc1 and c2 on the physiological functions of CD8+ T cells in vitro and in vivo. Therefore, we used a murine infection model with the bacteria Listeria monocytogenes and mice in which NFATc1 was specifically depleted in the T cell compartment.
Our first in vitro studies showed a typical NFATc1 and c2 nuclear translocation and changes on mRNA levels upon T cell activation similarly in CD4+ as well as in CD8+ T cells extracted from wild type mice. NFAT nuclear translocation is important for target gene activation and generation of effector functions. Stimulated T cell populations lacking NFATc1 and/or NFATc2 showed a markedly decreased expression of Th1/Tc1 cytokines, as e.g. IL 2 and IFNγ being important for the clearance of intracellular pathogens. From our in vitro model for the generation of allogenically reactive cytotoxic CD8+ T cells, we revealed a decreased killing and lytic granule-release capacity in Nfatc1 inactivated CD8+ T cells whereas NFATc2-/- cytotoxic T cells did not show an altered cytotoxic response compared to wild type cells.
Interestingly, we found lytic granules accumulated and mitochondria not getting translocated to the immunological synapse upon re-stimulation in NFATc1-deficient CD8+ T cells. Together with results showing the CsA insensitivity of the CTL killing/degranulation capacities, we assume that some major cellular processes are affected by NFATc1 which are not directly linked to the TCR-induced signal transduction cascade.
We also showed the importance of NFATc1 in T cells during intracellular infections with the bacteria Listeria monocytogenes in an in vivo mouse model. After five days, only few bacteria were detected in wt mice whereas high amounts of Listeria particles were extracted from livers of Nfatc1fl/fl x Cd4 cre mice. Although the reactivity towards the pathogen was similar in both groups, a decreased cytokine expression in NFATc1-/- CD8+ T cells was observed together with an altered memory cell generation.
Our results show the importance of NFATc1 in CD8+ T cells and give some clue for a possible connection to other basal cellular functions, as e.g. the formation of an immunological synapse.
Burkitt lymphoma (BL) is a highly aggressive B cell malignancy. Rituximab, a humanized antibody against CD20, in a combination with chemotherapy is a current treatment of choice for B-cell lymphomas including BL. However, certain group of BL patients are resistant to Rituximab therapy. Therefore, alternative treatments targeting survival pathways of BL are needed.
In BL deregulation of MYC expression, together with additional mutations, inhibits differentiation of germinal centre (GC) B cells and drives proliferation of tumor cells. Pro-apoptotic properties of MYC are counteracted through the B-cell receptor (BCR) and phosphoinositide-3-kinase (PI3K) pathway to ensure survival of BL cells. In normal B-cells BCR triggering activates both NF-κB and NFAT-dependent survival signals. Since BL cells do not exhibit constitutive NF-κB activity, we hypothesized that anti-apoptotic NFATc1A isoform might provide a major survival signal for BL cells.
We show that NFATc1 is constitutively expressed in nuclei of BL, in BL cell lines and in Eµ-Myc–induced B cell lymphoma (BCL) cells. Nuclear residence of NFATc1 in these entities depends on intracellular Ca2+ levels but is largely insensitive to cyclosporine A (CsA) treatment and therefore independent from calcineurine (CN) activity. The protein/protein interaction between the regulatory domain of NFATc1 and DNA binding domain of BCL6 likely contributes to sustained nuclear residence of NFATc1 and to the regulation of proposed NFATc1-MYC-BCL6-PRDM1 network in B-cell lymphomas.
Our data revealed lack of strict correlation between the expression of six NFATc1 isoforms in different BL-related entities suggesting that both NFATc1/alphaA and -betaA isoforms provide survival functions and that NFATc1alpha/betaB and -alpha/betaC isoforms either do not possess pro-apoptotic properties in BL cells or these properties are counterbalanced. In addition, we show that in BL entities expression of NFATc1 protein is largely regulated at post-transcriptional level, including MYC dependent increase of protein stability.
Functionally we show that conditional inactivation of Nfatc1 gene in Eµ-Myc mice prevents development of BCL tumors with mature B cell immunophenotype (IgD+). Loss of NFATc1 expression in BCL cells ex vivo results in apoptosis of tumor cells.
Together our results identify NFATc1 as an important survival factor in BL cells and, hence, as a promising target for alternative therapeutic strategies for BL.
The Notch signaling pathway is crucial for mammalian heart development. It controls cell-fate decisions, coordinates patterning processes and regulates proliferation and differentiation. Critical Notch effectors are Hey bHLH transcription factors (TF) that are expressed in atrial (Hey1) and ventricular (Hey2) cardiomyocytes (CM) and in the developing endocardium (Hey1/2/L). The importance of Hey proteins for cardiac development is demonstrated by knockout (KO) mice, which suffer from lethal cardiac defects, such as ventricular septum defects (VSD), valve defects and cardiomyopathy. Despite this clear functional relevance, little is known about Hey downstream targets in the heart and the molecular mechanism by which they are regulated.
Here, I use a cell culture system with inducible Hey1, Hey2 or HeyL expression to study Hey target gene regulation in HEK293 cells, in murine embryonic stem cells (ESC) and in ESC derived CM. In HEK293 cells, I could show that genome wide binding sites largely overlap between all three Hey proteins, but HeyL has many additional binding sites that are not bound by Hey1 or Hey2. Shared binding sites are located close to transcription start sites (TSS) where Hey proteins preferentially bind to canonical E boxes, although more loosely defined modes of binding exist. Additional sites only bound by HeyL are more scattered across the genome. The ability of HeyL to bind these sites depends on the C-terminal part of the protein. Although there are genes which are differently regulated by HeyL, it is unclear whether this regulation results from binding of additional sites by HeyL.
Additionally, Hey target gene regulation was studied in ESC and differentiated CM, which are more relevant for the observed cardiac phenotypes. ESC derived CM contract in culture and are positive for typical cardiac markers by qRT PCR and staining. According to these markers differentiation is unaffected by prolonged Hey1 or Hey2 overexpression. Regulated genes are largely redundant between Hey1 and Hey2. These are mainly other TF involved in e.g. developmental processes, apoptosis, cell migration and cell cycle. Many target genes are cell type specifically regulated causing a shift in Hey repression of genes involved in cell migration in ESC to repression of genes involved in cell cycle in CM.
The number of Hey binding sites is reduced in CM and HEK293 cells compared to ESC, most likely due to more regions of dense chromatin in differentiated cells. Binding sites are enriched at the proximal promoters of down-regulated genes, compared to up-or non-regulated genes. This indicates that up-regulation primarily results from indirect effects, while down-regulation is the direct results of Hey binding to target promoters. The extent of repression generally correlates with the amount of Hey binding and subsequent recruitment of histone deacetylases (Hdac) to target promoters resulting in histone H3 deacetylation.
However, in CM the repressive effect of Hey binding on a subset of genes can be annulled, likely due to binding of cardiac specific activators like Srf, Nkx2-5 and Gata4. These factors seem not to interfere with Hey binding in CM, but they recruit histone acetylases such as p300 that may counteract Hey mediated histone H3 deacetylation. Such a scenario explains differential regulation of Hey target genes between ESC and CM resulting in gene and cell-type specific regulation.
Der Transkriptionsfaktor C/EBPβ besitzt sehr vielgestaltige Funktionen und ist an Wachstums-und Differenzierungsvorgängen verschiedener Gewebe beteiligt. So fördert es in T-Lymphozyten über Transaktivierung des Il4-Promotors und Repression der TH1-Zytokine IL-2 und IFN-γ die Bildung eines TH2-Phänotyps [Berberich-Siebelt et al. 2000]. Durch Herabregulation von c-Myc bewirkt es einen Zellzyklusarrest in G1 und vermehrte Differenzierung der Zellen auch über eine reziproke Steigerung von Differenzierungsfaktoren wie Mad4 [Berberich-Siebelt et al. 2006]. In einer den G1-Arrest nachweisenden Zellzyklusanalyse von mit C/EBPβ transduzierten EL-4 Zellen zeigte sich daneben ein kleiner Sub-G1-Peak, der auf eine apoptotische Zellpopulation hinweist [Berberich-Siebelt et al. 2006]. Aufgabe dieser Arbeit war es, den möglichen Zusammenhang zwischen der Aktivierung von C/EBPβ und Auslösung von Apoptose in EL-4 Zellen hinsichtlich seiner Spezifität und dabei favorisierter Signalwege zu untersuchen.
Gegenstand der Untersuchungen waren mit dem C/EBPβ-ERTM-Konstrukt alleine und in Kombination mit dominant-negativen Mutanten der Caspase-3 und der Caspase-9 transduzierte EL-4 Kulturzellen. Durch Einbringen der Caspasemutanten sollte eine kompetitive Hemmung der entsprechenden endogenen Caspasen bewirkt werden. Methodisch erfolgten Apoptosenachweise mittels durchflusszytometrischer Analysen von mit Annexin V-PE und 7-Amino-Actinomycin (7-AAD) gefärbten EL-4 Zellen sowie die Detektion von gespaltener PARP (Poly-ADP-Ribose-Polymerase), einem Substrat der Caspase-3 im Western Blot. Des Weiteren erfolgten mittels Ribonuklease-Protektionsanalysen Untersuchungen der RNA-Expression von Zytokinen, Caspasen und von Mitgliedern der Myc- und der Bcl-2-Proteinfamilien, um das Verhalten der Zellen unter Hemmung von Apoptosewegen bzw. Caspasen bei Aktivierung von C/EBPβ näher betrachten zu können.
In Annexin V-PE- und 7-AAD-Färbungen sowie durch Nachweis der spezifischen Spaltung von PARP konnte gezeigt werden, dass C/EBPβ Zelluntergang und Apoptose fördert. Diese war durch den Pancaspaseinhibitor Z-VAD fmk hemmbar, was, wie auch die PARP-Spaltung, auf einen caspaseabhängigen Signalweg hinweist. Hemmung der Caspase-3 durch Transduktion der Zellen mit einer Caspase-3-Mutante besaß kaum Einfluss auf die durch C/EBPβ veränderte Zytokinexpression und die Repression von c-Myc, doch erschien eine vermehrte Hochregulation des Differenzierungsfaktors Mad4, der endogenen Caspase3- und der Caspase11-RNA. Die Steigerung von Caspase-3 unter Aktivierung von C/EBPβ fand sich auch auf Proteinebene. Allerdings konnte eine Hemmung der Caspase-3 bei den untersuchten EL-4 Zellen die durch C/EBPβ vermittelte Apoptose nicht verhindern, was auf andere Apoptosewege oder kompensatorischer Effekte verwies. Durch Beeinflussung des intrinsischen Signalweges mit Hemmung der Caspase-9 zeigten sich ebenfalls kaum Auswirkungen auf die Zytokinexpression der untersuchten Zellen. Hier fanden sich Hochregulationen sowohl der pro- als auch antiapoptotischen Mitglieder der Bcl-2-Familie. Funktionell konnte auch eine Hemmung von Caspase-9 die Zellen nicht vor der Apoptose durch Aktivierung von C/EBPβ bewahren.
So konnte hier gezeigt werden, dass Aktivierung von C/EBPβ in den untersuchten EL-4 Zellen Apoptose fördern kann, dies über eine Aktivierung der Caspasekaskade zu geschehen scheint und mit einer Steigerung endogener Caspase-3-Expression einhergeht. Aus den Untersuchungen dieser Arbeit konnte eine Favorisierung eines bestimmten Apoptosesignalweges nicht abgeleitet werden, da eine Hemmung des intrinsischen Weges die Zellen nicht vor dem Zelltod schützen konnte. Insgesamt lässt sich aber, obwohl nicht alle Details geklärt werden konnten, festhalten, dass C/EBPβLAP in T-Zellen neben Proliferationshemmung und Differenzierungsinduktion auch für Caspase vermittelten Zelltod verantwortlich ist.
In this work we wanted to investigate the role of NFATc1 in lymphocyte physiology and in pathological conditions (eg. psoriasis). NFATc1 is part of the signal transduction
pathways that regulates B cells activation and function. NFATc1 has different isoforms that are due to different promoters (P1 and P2), polyadenylation and alternative splicing. Moreover, we tried to elucidate the points of interactions between the NFAT and the NF-κB pathways in
activated B-cell fate. NFAT and NF-κB factors share several properties, such as a similar mode of induction and architecture in their DNA binding domain. We used mice which over-express a constitutive active version of NFATc1/α in their B cells with -or without- an ablated IRF4. IRF4 inhibits cell cycle progression of germinal center B cell-derived Burkitt’s lymphoma cells and
induces terminal differentiation toward plasma cells. Our experiments showed that a ‘double hit’ in factors affecting B cell activation (NFATc1 in this case) and late B cell Differentiation (IRF4 in this case) alter the development of the B cells, lead to increase in their numbers and increase in stimulation induced proliferation. Therefore, the overall picture indicates a link between these 2 genes and probable carcinogenic alterations that may occur in B cells.
We also show that in splenic B cells, c-Rel (of the NF-κB canonical pathway) Support the induction of NFATc1/αA through BCR signals. We also found evidence that the lack of NFATc1 affects the expression of Rel-B (of the NF-κB non-canonical pathway). These data suggest a tight interplay between NFATc1 and NF-κB in B cells, influencing the competence of B cells and their functions in peripheral tissues.
We also used IMQ-induced psoriasis-like inflammation on mice which either lack NFATc1 from B cell. Psoriasis is a systemic chronic immunological disease characterized
primarily by abnormal accelerated proliferation of the skin keratinocytes. In psoriasis, the precipitating event leads to immune cell activation. Our experiments showed that NFATc1 is needed for the development of psoriasis. It also showed that IL-10 is the link that enables NFAT
from altering the B cell compartment (eg Bregs) in order to affect inflammation. The important role of B cell in psoriasis is supported by the flared up psoriasis-like inflammation in mice that lack B cells. Bregs is a special type of B cells that regulate other B cells and T cells; tuning the immunological response through immunomodulatory cytokines.
