Einfluß der Dialysetherapie auf den Genomschaden von Nierenpatienten in einer prospektiven Studie

Patienten mit terminaler Niereninsuffizienz haben im Vergleich zur Normalbevölkerung eine deutlich erhöhte Inzidenz maligner Erkrankungen. Frühere Untersuchungen zeigten, dass periphere Blutlymphozyten dieser Patienten einen höheren genetischen Schaden aufweisen, wodurch das Risiko einer malignen Entartung steigt. In dieser Arbeit wurde der genetische Schaden mithilfe zweier Testverfahren, Comet Assay und Mikrokerntest, untersucht. Es handelte sich um eine prospektive Studie mit zwei Patientenkollektiven. Die erste Gruppe bestand aus Patienten, die aufgrund einer terminalen Niereninsuffizienz innerhalb der nächsten Monate eine Dialysetherapie mittels konventioneller Hämodialyse beginnen mußten. Die zweite Gruppe bildeten Dialysepatienten, die im Verlauf von konventioneller Dialyse auf Hämodiafiltration umgestellt wurden. Bei allen Patienten wurde der genetische Schaden der peripheren Blutlymphozyten in den Monaten vor und nach Therapiebeginn bzw. Therapieumstellung regelmäßig untersucht. Unsere Ergebnisse zeigen, dass 4 der 10 Prädialysepatienten nach Beginn der Dialyse einen niedrigeren genetischen Schaden hatten, 2 Patienten hatten unterschiedliche Werte in Comet Assay und Mikrokerntest und bei 2 Patienten ergab sich im Verlauf eine höhere DNA-Schädigung. Die verbliebenen 2 Patienten mußten aufgrund einer konstant bleibenden Niereninsuffizienz nicht mit der Dialysetherapie beginnen. Bei Zusammenfassung aller Einzelwerte zeigte sich, dass das Kollektiv der Prädialysepatienten insgesamt vom Beginn der Behandlung profitiert hat. In der Gruppe der Dialysepatienten hatte 2 von 7 Patienten nach Umstellung auf Hämodiafiltration eine geringere DNA-Schädigung, 2 Patienten zeigten unterschiedliche Ergebnisse im Comet Assay und Mikrokerntest und 2 weitere Patienten wiesen eine höheren genetischen Schaden in den Lymphozyten auf. Ein Patient konnte bei fehlenden Vorwerten nicht berücksichtigt werden. Im Gruppenvergleich zeigte sich für alle Dialysepatienten ein gleichbleibender DNA-Schaden, gemessen mithilfe des Comet Assays bei leicht erhöhten Mikrokernraten. Jedoch hatte sich die Zellproliferation ebenfalls etwas verbessert. Zusammenfassend ergibt sich somit in beiden Gruppen kein eindeutiges Ergebnis, woraus neue Therapieempfehlungen für Patienten mit terminaler Niereninsuffizienz abzuleiten wären. Um weiter Einflußvariablen auf die Höhe des genetischen Schadens festzustellen, sind weiter Untersuchungen mit größeren Patientenkollektiven erforderlich.
The aim of the present study was to design different dosage forms as carrier systems to deliver sorafenib to the lung of BXB-23 transgenic mice using different routes of administration. Three dosage forms were used one of them was an oil-in-water emulsion and the oral route was chosen for this experiment. The other delivery system was a liposome preparation for intratracheal instillation. In this case the oral route was considered as a control experiment. The last dosage form was PLGA microspheres. Before sorafenib administration it was important to develop a HPLC method to assess sorafenib absorption after its administration and to determine its concentrations in mouse serum. The HPLC method allowed sorafenib quantification in small volumes (30 µl) of mouse serum and tissues. The developed HPLC method was validated resulting in satisfactory selectivity, good linearity, good accuracy and precision over the concentration range examined. Sorafenib was successfully incorporated in a fat emulsion (o/w) using a traditional method resulting in a white homogenous emulsion and no particle aggregation was observed. Sorafenib exhibited antitumor activity on the lung adenoma in BXB-23 transgenic mice when administered orally (2 mg sorafenib per mouse) in the emulsion preparation. The determined effect was an approximately 29 % reduction in the tumor area of the adenoma foci and a proliferation reduction. In order to improve the pharmacological effects of sorafenib on the lung adenoma in BXB-23 mice, the targeting of sorafenib directly to the site of action (the lung) was an attractive concept. For this purpose the intratracheal route was used. Since sorafenib administration by instillation required incorporation of sorafenib in a dosage form suitable for its lipophilic nature, a liposome suspension was the second dosage form used. A lyophilization method was employed for sorafenib liposome preparation utilizing dilauroylphosphatidylcholine (DLPC) which is safe and tolerable for the lung. Incorporation of sorafenib in the liposomes did not influence the particle size and its distribution. The sorafenib liposomes showed high encapsulation efficiency, good stability at 4 °C for one month and satisfactory in vitro release properties and inhibited Raf-1 mediated activation of ERK in cell culture assay. In a pharmacokinetic experiment sorafenib loaded liposomes were instilled directly into the lung. The results revealed that a significant level of sorafenib was achieved in the lung tissues after 2 hours and then reduced after 48 h and remained nearly constant for one week. On the other hand, only traces of sorafenib were found in the mice serum up to 48 h. Subsequently, the pharmacological activity of sorafenib (1 mg per mouse) was studied when delivered in a liposomal suspension intratracheally to treat the lung adenoma of BXB-23 mice. The data of this experiment demonstrated that sorafenib intratracheal instillation resulted in a reduction of tumor area of adenoma foci (67 %) and an elevation of the percent of apoptotic cells. In contrast, prolongation of the treatment period did not further enhance sorafenib activity on the lung adenoma. This previous finding suggested a development of multidrug resistance (MDR) by the adenoma foci cells against sorafenib instillation, which was examined by immunohistochemistry staining. The percent of MDR positive cells was higher after two and three weeks sorafenib liposome instillation treatment than that after one week treatment. The last dosage form used for sorafenib was microspheres, which were prepared by emulsion-diffusion-evaporation method using biodegradable PLGA 50:50 resulting in a white lyophilized powder. The system was characterized physicochemically and revealed a good microspheres yield, high encapsulation efficiency, a homogenous particle size distribution and slow in vitro release of sorafenib. The other strategy studied in the present research project was gene delivery to target the lung bearing tumor of BXB-23 mice using a non-viral vector (polyethylenimine). Polyethylenimine (PEI) was used to investigate its efficiency in transfecting lung bearing tumor of BXB-23 mice model and its ability to transfect the adenoma foci cells. LacZ, which encodes Beta-galactosidase was used in the present study as a reporter gene and was complexed with PEI before delivered intravenously. A high LacZ expression in the alveolar region with some expression in the adenoma foci was observed. On contrary, a low LacZ expression in the alveoli and in the adenoma foci was achieved after instillation of the same polyplex intratracheally.
Diese Dissertation enthält eine Edition und eine annotierte Übersetzung des dem Bhasa zugeschriebenen Sanskrit-Schauspiels "Pratijnayaugandharayana". Die Edition beruht auf zehn Manuskripten, von denen bisherige Ausgaben nur drei verwendet hatten. Um Zusammenhänge zwischen den Manuskripten sichtbar zu machen, und so deren jeweiligen Wert besser beurteilen zu können, werden quantitative Methoden verwendet. In diesem Zusammenhang wird die effektive Verwendung von EDV für die Veröffentlichung von Sanskrit-Schauspielen reflektiert. Die vorgestellte Herangehensweise ermöglichte auch die automatische Erstellung des als Anhang enthaltenen Index aller in dem Schauspiel vorkommenden Sanskrit- und Prakrit-Wortformen.
Diese Arbeit versucht, die wirtschaftliche Bedeutung der globalen Erwärmung zu erklären und die Lösung dieses Problems zu finden. Die globale Erwärmung als länderübergreifendes Problems kann durch das marktwirtschaftliche Preissystem gelöst werden, besonders durch den internationalen Handel der Schadstoffemission. Hierbei wird die dezentrale Lösungsprinzip betont. Die globale Erwärmung und Politik für die Lösung dieses Problem sind dauerhaft. Daher wird das intertemporale Wachstumsmodell zur Berechnung des Gewinns und der Kosten der Politik verlangt. Dabei wird ein Prinzip besagt, dass jede Generation verantwortlich auf ihre Generation ist. In dieser Arbeit wird versucht, die optimale Handelspolitik und die Kyoto-Politik zu vergleichen.
Die Ökonomik Konrads von Megenberg gehört zu einer Literaturgattung der Antike und des Mittelalters, die sich in Anlehnung an Aristoteles‘ Oeconomica mit dem Haushalt beschäftigt. Anhand zweier Traktate aus dem Kapitel über das nicht herrschaftliche Haus wird in dieser Arbeit Konrads Sicht auf die Ehe, die Familie und die Kindererziehung mentalitäts- und alltagsgeschichtlich untersucht.
Despite its precise agreement with the experiment, the validity of the standard model (SM) of elementary particle physics is ensured only up to a scale of several hundred GeV so far. Even more, the inclusion of gravity into an unifying theory poses a problem which cannot be solved by ordinary quantum field theory (QFT). String theory, which is the most popular ansatz for a unified theory, predicts QFT on noncommutative space-time as a low energy limit. Nevertheless, independently of the motivation given by string theory, the nonlocality inherent to noncommutative QFT opens up the possibility for the inclusion of gravity. There are no theoretical predictions for the energy scale Lambda_NC at which noncommutative effects arise and it can be assumed to lie in the TeV range, which is the energy range probed by the next generation of colliders. Within this work we study the phenomenological consequences of a possible realization of QFT on noncommutative space-time relying on this assumption. The motivation for this thesis was given by the gap in the range of phenomenological studies of noncommutative effects in collider experiments, due to the absence in the literature of Large Hadron Collider (LHC) studies regarding noncommutative QFTs. In the first part we thus performed a phenomenological analysis of the hadronic process pp -> Z gamma -> l^+l^- gamma at the LHC and of electron-positron pair annihilation into a Z boson and a photon at the International Linear Collider (ILC). The noncommutative extension of the SM considered within this work relies on two building blocks: the Moyal-Weyl star-product of functions on ordinary space-time and the Seiberg-Witten maps. The latter relate the ordinary fields and parameters to their noncommutative counterparts such that ordinary gauge transformations induce noncommutative gauge transformations. This requirement is expressed by a set of inhomogeneous differential equations (the gauge equivalence equations) which are solved by the Seiberg-Witten maps order by order in the noncommutative parameter Theta. Thus, by means of the Moyal-Weyl star-product and the Seiberg-Witten maps a noncommutative extension of the SM as an effective theory as expansion in powers of Theta can be achieved, providing the framework of our phenomenological studies. A consequence of the noncommutativity of space-time is the violation of rotational invariance with respect to the beam axis. This effect shows up in the azimuthal dependence of cross sections, which is absent in the SM as well as in other models beyond the SM. Thus, the azimuthal dependence of the cross section is a typical signature of noncommutativity and can be used in order to discriminate it against other new physics effects. We have found this dependence to be best suited for deriving the sensitivity bounds on the noncommutative scale Lambda_NC. By studying pp -> Z gamma -> l^+l^- gamma to first order in the noncommutative parameter Theta, we show in the first part of this work that measurements at the LHC are sensitive to noncommutative effects only in certain cases, giving bounds on the noncommutative scale of Lambda_NC > 1.2 TeV. Our result improved the bounds present in the literature coming from past and present collider experiments by one order of magnitude. In order to explore the whole parameter range of the noncommutativity, ILC studies are required. By means of e^+e^- -> Z gamma -> l^+l^- gamma to first order in Theta we have shown that ILC measurements are complementary to LHC measurements of the noncommutative parameters. In addition, the bounds on Lambda_NC derived from the ILC are significantly higher and reach Lambda_NC > 6 TeV. The second part of this work arose from the necessity to enlarge the range of validity of our model towards higher energies. Thus, we expand the neutral current sector of the noncommutative SM to second order in $\theta$. We found that, against the general expectation, the theory must be enlarged by additional parameters. The new parameters enter the theory as ambiguities of the Seiberg-Witten maps. The latter are not uniquely determined and differ by homogeneous solutions of the gauge equivalence equations. The expectation was that the ambiguities correspond to field redefinitions and therefore should vanish in scattering matrix elements. However, we proved that this is not the case, and the ambiguities do affect physical observables. Our conjecture is, that every order in Theta will introduce new parameters to the theory. However, only the experiment can decide to what extent efforts with still higher orders in Theta are reasonable and will also give directions for the development of theoretical models of noncommutative QFTs.
The continuously increase in resistance of human pathogenic microorganisms to the known antibiotics leads to the necessity for searching new sources for production of new active antimicrobial compounds from different natural sources especially plants, since many plants have been found to be able to produce antimicrobial compounds as a defense phenomenon against invading microorganisms. The aim of this work is to screen cultures for production of antimicrobial activity against representative of human pathogenic microorganisms and selection the most active cell culture producing antimicrobial protein(s) which are active against these pathogenic microorganisms and also isolation ,purification of the active protein(s) and cloning of its/their genes. Ten different plant suspension cultures have been screened in presence of nine elicitors for their antimicrobial activity against five selected human pathogenic microorganisms, and it has been found that the heterotrophic cultures are more active against the tester isolates than the autotrophic ones. The intracellular fraction of the mixotrophic Arabidopsis thaliana culture elicited with salicylic acid showed the highest antimicrobial activity against the tester isolates. The presence of proteinous antimicrobial activity has been elucidated by testing the activity of ammonium sulphate precipitate against Candida maltosa. High speed centrifugation technique has been used for partial purification of the active protein. The proteinous nature of the isolated compound has been confirmed by using bioautography technique and its molecular weight could be estimated to be around 26KDa. The active protein has been purified using gel filtration, and using mass spectrometry technique, for microsequencing of the active protein, it has been found that the function of the protein is unknown and we have termed it as AtPDP1 according to Arabidopsis thaliana Plat-Domain Protein1, since it contains a plant stress domain termed PLAT domain. It has been found that a second protein from the same plant with high homology level to AtPDP1 with the same domain, we termed it as AtPDP2. Genes for AtPDP1 and AtPDP2 have been cloned in E. coli using PGEM-T easy vector. The expression of both genes have been tested using Digital Northern program, and it has been observed that both genes are induced by different pathogens, chemicals known to induce defense in plant cells and also different hormones. We tried to clone the gene for AtPDP1 in PBI121 binary vector under the control of an elicitor inducible promoter of a proteinase inhibitor gene, to test its function in plant by overexpression, but we did not succeeded. Also the work aims to cloning the different known thaumatin genes from Arabidopsis thaliana for future work which represented by testing their expression under different stimuli, since most thaumatins have antimicrobial activity and some of them are active against Candida spp..Thirteen genes of known thaumatins from Arabidopsis thaliana have been cloned in PGEM-Teasy vector in DH5-alpha cells. coli cells. The expression of the thirteen genes has been done using Digital Northern program and it has been found that different genes show different expressions under different stimuli and the expression of At1g75800 gene was the maximum under all stimuli. The minimum expression of genes was for At1g75050. The rest of thaumatin genes showed moderate expressions under different stimuli.
In vorliegender Studie wurde an histologischen Präparaten mittels immunhistochemischen Färbungen der Übergangsbereich zwischen oraler Mukosa und transplantiertem Jejunumsegment untersucht. Mit der Fragestellung ob sich die Strukturen im Übergangsbereich zwischen freiem Jejunumtransplantat und oraler Mukosa funktionell verändern und ob die Dünndarmepithelien einer morphologischen Umwandlung unterliegen. Zu diesem Zweck wurden die Gewebeschnitte nach der LSABC Methode mit verschiedenen Antikörpern markiert die spezifisch das zu bestimmende Epithel anfärbten. Die Differentzierung der Epithelien erfolgte durch für jede Epithelart spezifische Zytokeratine. Nachgewiesen wurde eine Abflachung des Zottenreliefs sowie eine numerische Becherzellatrophie. In unseren histologisch nachuntersuchten Gewebeproben stellte sich die Grenze zwischen transplantiertem Jejunumsegment und mehrschichtigem Plattenepithel exakt und scharf begrenzt dar. Weder Zellcluster von mehrschichtigem Plattenepithel im Bereich des Dünndarms, noch eine Transformationszone, wie sie bei Zellmetaplasie vorkommt konnte nachgewiesen werden. Auch hielten die dünndarmeigenen Gefäße die Grenze zwischen den beiden Geweben strikt ein und verliefen ebenso scharf begrenzt. Subepithelial zeigte sich zwar zu Beginn eine entzündlich bedingte Gefäßproliferation mit schütterem leukozytären Infiltrat, welches aber im Laufe der Zeit durch kollagenfasereiches Bindegewebe narbig ersetzt wurde. Die nachweisbaren CD 4 positiven Entzündungszellen waren nicht über das gewöhnliche Ausmaß vermehrt, es kommt zur Ausbildung einer Gewebsgrenze die von beiden Epithelien respektiert wird unter der Vereinigung des subepithelialen Bindegewebes. Dies alles kann als Bestätigung dafür angesehen werden, dass die Rekonstruktion großer Defekte mittels freiem Jejunumtransplantat auch über einen Zeitraum von mehr als 21 Jahren erfolgreich ist und das Transplantat seinen Anforderungen in der neuen Umgebung standhält.
Mehrere Autoren haben schon die Intra- und Interobserver-Variabilität bei der Bestimmung des Schilddrüsenvolumens und knotiger Herdbefunde mit Hilfe des zweidimensionalen (2D) Ultraschalls evaluiert. Darüber hinaus wurde über Interobserver-Korrelationen für Schilddrüsenvolumenmessungen berichtet. Es gibt jedoch keine prospektive verblindete Studie, die die Intra- bzw. Interobserver-Variabilität bei der Volumenbestimmung der gesamten Schilddrüse an gesunden Probanden bzw. einzelner Knoten unterschiedlicher Echogenität an einem Phantom untersucht hat. Die Ergebnisse der Einzelstudien sollen hier vorgestellt und – soweit möglich – miteinander verglichen werden. Im Rahmen einer quantitativen Studie mit dem hier präsentierten Schilddrüsenphantom soll die Intra- und Interobserver-Variabilität bei der 2D-Ultraschallvolumetrie einzelner Knoten unterschiedlicher Größe und Echogenität und der Schilddrüsenlappen evaluiert werden. Da Schilddrüsenknoten wegen des geringeren Volumens und ihrer oft unscharfen Randkontur schwieriger zu entdecken und auszumessen sind als die Gesamtschilddrüse, soll untersucht werden, welche Größenordnungen des Messfehlers auftreten und in welcher Relation sie zueinander stehen. Außerdem soll der methodenimmanente Fehler quantifiziert und detektierbare Volumenänderungen erfassbar gemacht werden. Bisher war in der Schilddrüsensonographie kein geeignetes Phantom verfügbar, das kommerziell erhältlich ist und mit dem qualitativ unterschiedliche intrathyreoidale Herdbefunde untersucht werden können. Die vorliegende Studie an gesunden Probanden hatte das primäre Ziel, die Frage nach der Quantifizierbarkeit von Unsicherheitsfaktoren in der Schilddrüsenvolumetrie durch den konventionellen 2D-Ultraschall im Vergleich zu 3D-Referenzvolumina bei gesunden Erwachsenen möglichst exakt zu beantworten und die Untersucherabhängigkeit der Methode zu demonstrieren. Damit soll die Genauigkeit (Richtigkeit und Präzision) der sonographischen Schilddrüsendiagnostik mathematisch erfasst und eine bessere Bewertungsgrundlage für die Frage nach der Reproduzierbarkeit von Ultraschall-Volumenbestimmungen der Schilddrüse und ihrer pathologischen Veränderungen geschaffen werden. Hierfür wurden möglichst aussagekräftige statistische Parameter wie die Intra- und Interobserver-Variabilität, der systematische und zufällige Fehler, der reine Fehler der Messmethode, minimale, sicher detektierbare Volumenänderungen und im Rahmen einer multivariaten Reliabilitätsanalyse die Reliabilitätskoeffizienten untersucht. Ein weiteres Ziel dieser Studie bestand darin, die Reliabilität der in der klinischen Routine benutzten Ellipsoidformel zur Berechnung des Schilddrüsenvolumens zu überprüfen.
Infrared photodissociation spectroscopy of ionic hydrocarbons : microsolvation and protonation sites

This work has presented a spectroscopic analysis of three types of hydrocarbon cations: two ionized aromatic hydrocarbons, two protonated aromatic hydrocarbons and the cation of a fundamental radical hydrocarbon. The experiments were centered on the proton stretch vibrations of mass-selected complexes of these systems and polar (H2O) and non polar (Ar, N2, CO2) ligands. The experiments have been done in a tandem mass spectrometer coupled with an electron impact ionization ion source; an OPO laser system was used as tunable IR light source. All the proposed dimer structures have been also modeled using quantum chemical calculations (QCC). These calculations have consistently been matched with the experimental results and have enabled clear identification of the spectral features observed. This has enabled the evaluation of thermochemical properties which could not be extracted directly from experiment. The experiments done on complexes of 1-Np+ and Im+ have allowed for the acidity of their various groups to be probed: the shifts in the frequency as well as the enhancement in the intensity of the OH and NH stretch vibrations resulting from the complexation have yielded dependences on both the species (L) and the number (n) of the ligands. OH bound 1-Np+···Ar has been detected for the first time, showing that the REMPI-IRPD method is severely limited with respect to the production of the most stable isomer of a given cationic complex. The detection of c-1-Np+···(N2)n corresponds to the first observation of c-1-Np+ complexes and enables thus direct comparison of both 1-Np+ rotamers. The shift of the NH vibration of Im+···N2(H) yielded the first experimental estimate for the PA of the imidazyl radical. It was also found that the most stable 1-Np+···Ar and Im+···Ar structures differ qualitatively from that of the corresponding neutral dimers (H-bound vs pi-bound), emphasizing the large impact of ionization on the interaction potential and the preferred recognition motif between acidic aromatic molecules (A) and nonpolar ligands. The IRPD spectra of 1-Np+···Ln and Im+···Ln yielded spectroscopic information about the CH, NH and OH stretch vibrations of bare 1-Np+ and Im+. The dependence of the shifts in the frequency of the OH and NH stretch vibrations allows for creating microsolvation models. The spectroscopic results obtained on size-selected 1-NpH+···Ln show that, in the output of the presently used ion source, three classes of 1-NpH+ isomers can be identified: oxonium ions (1-Np protonated at the O atom); carbenium ions obtained by protonation in the para and ortho positions with respect to the OH functional group; carbenium ions obtained by the addition of a proton to well-defined sites on the second naphthalene ring. The spectral identification of these three classes of protonation sites is supported by their different photofragmentation patterns. It was demonstrated that the spectroscopic monitoring of the microsolvation of ImH+ in Ar and N2 together with the QCCs paint a very detailed picture of the microsolvation process, evidencing clear differences between the microsolvation models as function of the PA of the ligands. Important differences have also been identified between the various binding sites, enabling the creation of a clear scale of priorities for occupation of the binding sites during microsolvation. The application of IRPD to the study of microhydrated ImH+ provided for the first time direct spectroscopic information on the properties of the N-H bonds of this biomolecular building block under controlled microhydration. It was demonstrated that, as protonation enhances the acidity of the NH groups, the ability for proton conductivity of ImH+ increases. A very important result is derived from the IRPD spectroscopy of C2H5+···L (L = Ar, N2, CO2, CH4) dimers. The equilibrium geometry of the C2H5+ has long been debated. Now, IRPD spectra were recorded over the range of the CH stretch fundamentals (covering possible sp3 and sp2 hybridization of C). Depending on the ligand species, the spectra are found to be dominated by the fingerprint of two largely different dimer geometries. Using the experimental C2H5+···Ar spectrum and the corresponding QCCs, the structure of the (weakly perturbed) C2H5+ was found to be the nonclassical one, with one proton straddling across the C=C bond of the H2C=CH2. On the other hand, ligands like N2 and CH4 are strongly influencing the geometry, as seen in the spectral signatures of the C2H5+···N2 and C2H5+···CH4, which correspond to the classical [H2CCH3]+. It was thus demonstrated that while the nonclassical C2H5+ is the global minimum on the PES of the free [C2,H5]+, the structure of the C2H5+ can be strongly influenced by the chemical properties of the environment.
Teil 1: Aus früheren Arbeiten geht hervor, dass Fibomyalgiepatienten über eine höhere Schmerzstärke in bezug auf ihren klinischen Schmerz nach negativem emotionalen Priming verglichen mit positivem Priming berichten als Patienten mit muskuloskelettalem Schmerz. Um die affektive Modulation von Druckschmerz bei Fibromyalgiepatienten (n = 30) unter kontrollierten Bedingungen im Vergleich mit Gesunden und Schmerzerkrankungen geklärter Genese (d.h. organisch oder psychisch) beschreiben zu können, wurde Schmerz experimentell induziert. Neben 30 Gesunden (schmerzfreien Personen) dienten 30 Rückenschmerzpatienten, bei denen eine organische Schmerzgenese vermutet wurde, und 30 somatoforme Schmerzpatienten als Vergleichsgruppen. Für die letzte Gruppe, bei der eine psychische Schmerzgenese angenommen wurde, wurde die gleiche Schmerzmodulation wie für die Fibromyalgiepatienten vermutet. Als Primes dienten positive, neutrale, negative und schmerzbezogene Bilder des International Affective Picture Systems. Schmerz wurde über einen konstanten tonischen Druckreiz ausgelöst; als abhängige Variable wurde die empfundene Schmerzstärke erfasst. Über alle Versuchspersonen hinweg modulierte die Bildervalenz die Schmerzstärke: nach schmerzbezogenen Bildern war die Schmerzstärke höher als nach negativen und nach negativen Bildern war sie höher als nach neutralen. Die Schmerzstärken nach neutralen im Vergleich zu positiven Bildern unterschieden sich jedoch nicht signifikant. Somatoforme Schmerzpatienten berichteten über höhere Schmerzstärken als Rückenschmerzpatienten und Gesunde. Die Fibromyalgiepatienten zeigten ähnlich hohe Schmerzstärken wie die somatoforme Schmerzgruppe, doch unterschieden sie sich weder von den Gesunden noch von den Rückenschmerzpatienten. Zwischen Priming und Gruppe gab es keine Interaktion: Die affektive Schmerzmodulation war bei den Fibromyalgie- und den somatoformen Schmerzpatienten nicht spezifisch verändert, doch die somatoformen Schmerzpatienten zeigten eine erhöhte Druckschmerzsensibilität als Gesunde und Rückenschmerzpatienten. Teil 2: Es wurde oft vermutet, dass sich Fibromyalgiepatienten hinsichtlich soziodemographischer Daten und psychologischer Merkmale von Schmerzpatienten unterscheiden, deren Schmerz organischer Genese ist wie bei Patienten mit (rheumatoider) Arthritis. Ob sie sich diesbezüglich von Patienten mit somatoformer Schmerzstörung unterscheiden, wurde bis jetzt noch nicht geklärt. Um das psychologische Profil von Fibromyalgiepatienten zu spezifizieren, wurden 25 Fibromyalgie- mit 29 somatoformen Schmerz-, 27 Rückenschmerzpatienten und 30 gesunden (schmerfreien) Kontrollpersonen (alle Versuchspersonen hatten am Teil 1 der Studie teil genommen) in bezug auf Unterschiede in soziodemographischen Merkmalen, klinischer Schmerzstärke, schmerzbezogene Selbstinstruktionen, Partnerreaktionen, Stressbewältigungsstrategien und Selbstwirksamkeits- und externale Kontrollüberzeugungen verglichen. Alle drei Schmerzgruppen berichteten über eine stärkere Depression, größere Trait-Angt und größere affektive Verstimmung als die Gesunden. Fibromyalgie- und somatoforme Schmerzpatienten zeigten außerdem eine niedrigere Lebenskontrolle, weniger Aktivitäten außer Haus und vermehrt negative Stressbewältigungsstrategien als die Gesunden. Die Fibromalgiepatienten berichteten zudem über eine stärkere Somatisierung und größere affektive und sensorische Schmerzstärken als die Rückenschmerzpatienten, aber unterschieden sich nicht von den somatoformen Schmerzpatienten. Des weiteren gaben die Fibromyalgiepatienten mehr Schmerzen an verschiedenen Körperstellen an als die Rückenschmerzpatienten. Ein unerwartetes Ergebnis war, dass die somatoformen Schmerzpatienten eine größere Ausprägung im Merkmal Bestrafung (i. s. einer Partnerreaktion) als die Fibromyalgiepatienten (und die Gesunden) zeigten.
Oxygen-centered radicals are important intermediates in photobiological, mechanistic and synthetic studies. The majority of precursors of reactive oxyl radicals are labile and thus delicate to handle. Therefore N-(alkoxy)-pyridinethiones and N-(Alkoxy)-thiazolethiones have attracted attention as "mild'' photochemical source of alkoxyl radicals, in the last few years. A disadvantage of the pyridine compounds, is their sensibility to daylight. Despite of their similarities, both molecules behave surprisingly different, if photolyzed in the absence of trapping reagents. The pyridinethione compounds undergo highly efficient radical chain reactions under such conditions while the corresponding thiazolethiones react surprisingly sluggish and give rise to several unwanted side products. The properties of both compounds should be understood and optimized in the frame of this work. Additionally new compounds should be suggested that can also be applied in the photochemical alkoxyl radical generation. Some background information about the generation and application of alkoxyl radicals is provided in chapter 2. Electronic excitations and UV/vis spectroscopy together with a description of quantum chemical approaches that are able to calculate such phenomena are outlined in chapter 3. Chapter 4 deals with the description of the vertical excitation spectra. During the validation CASSCF, CASPT2, TD-DFT and RI-CC2 were tested with respect to their ability to describe the vertical excitations in both compounds. The CASPT2 approach gives accurate descriptions of the electronic excitation spectra of all compounds. The time-dependent DFT results are very sensitive on the choice of the functional and a validation of the results should be always done. On the basis of these computations the spectroscopic visible absorption bands of both compounds were assigned to a pi-->pi* transition in the thiohydroxamic acid functionality. In chapter 5 the mechanism of the thermally and the photochemically induced N,O homolysis in both compounds is unveiled. The near UV-induced N,O homolysis will start from the S2 state. The expected relaxation from the S2- to the S1-state and the dissociation process is expected to be very fast in the case of the thiazolethione compound. The potential surfaces of the pyridine compound in contrast point to a slower N,O bond dissociation. Due to the resulting faster dissociation process the excess energy which results from the photochemical activation is quenched only to small amounts. The maximal possible excess energy of the fragments is lower and a quenching is much more likely in the case of the pyridinethione compounds. This explaines the different reactivities of both compounds. For the also already successfully applied precursor system N-(alkoxy)-pyridineones the computed dissociation paths show courses that clearly predict a slow bond dissociation process. Chapter 6 deals with the tuning of the initial excitation wave length of the known pyridinethiones und thiazolethiones. In the first part the effects of substituents on the thiazolethione heterocycle was examined. The UV/vis spectra of 4 and 5 substituted thiazolethiones can be interpreted like the spectrum of the parent compound. The second part of chapter 6 deals with the identification of a substitution pattern on the pyridine heterocycle which induces a blue shift of the photo active band. The computations showed that electron rich and electron poor substituents result the same effects on the electronic excitation spectra. These substituent effects are additive, but the steric orientation of the substituents has to be taken into account. Chapter 7 describes a computer aided design of new alkoxyl radical precursors. Combining the advantages of both compounds the radical formation should be initiated by an irradiation with light at about 350 nm, and the amount of side products during the radical formation process should be small. To achieve this 18 test candidates were obtained by a systematic variation of the parent compound of the thiazolethione precursor. To identify the promising new precursor systems a screening of the lower electronic excitations of all resulting 18 systems was performed with TD-DFT. For promising systems the N,O or P,O dissociation paths, respectively, were analyzed according to the developed model. N-(methoxy)-azaphospholethione and N-(methoxy)-pyrrolethione seem to be the most promising candidates. The computations predict a strong absorption at about 350 nm respectively 320 nm. Due to the amounts of maximal excess energy and the shapes of the potential surfaces of the N,O bond dissociation paths their reactivity should resemble more the behavior of the pyridinethiones.
Transmissible spongiform encephalopathies (TSEs) or prion diseases are a group of infectious neurodegenerative diseases that are associated with misfolding of the cellular form of the cellular prion protein (PrPC) into a disease associated conformer (PrPSc). No therapy for prion diseases is available at present. So far, anti-PrPC vaccination is hampered by immunological tolerance of the mammalian immune system to endogenous PrPC. The aim of this thesis was to set up a new vaccination strategy based on virus-like particles (VLP) to induce anti-PrPC antibody responses in PrPC-competent mice. In a first step it was assessed whether VLP have the capacity to induce antibody responses that are protective against conventional pathogens. For this purpose, VLP displaying the vesicular stomatitis virus-gylcoprotein (VLP-VSV) were generated and tested for their immunogenicity. Similarly to live vesicular stomatitis virus (VSV), replication deficient VLP-VSV induced T help-independent VSV neutralizing IgM responses that switched to the IgG subclass in a T help-dependent manner. Furthermore, type I IFN receptor (IFNAR) triggering only marginally affected VLP-VSV induced neutralizing IgM responses, whereas it was critically required to promote the IgG switch. The analysis of conditional knockout mice with a lymphocyte-specific IFNAR deletion revealed that IFNAR triggering of lymphocytes did not play a crucial role, neither upon VLP-VSV nor VSV immunization. Collectively, these data verified the high immunogenicity of VLP. Therefore, in a next step VLP were generated displaying the C-terminal half of PrP (residues 121-231aa) fused to the platelet derived growth factor receptor (PDGFR) transmembrane region (VLP-PrPD111) for anti-PrPC immunization. On the surface of such retroparticles, PrPC was expressed at high levels as determined by electron microscopy. VLP-PrPD111 immunization of Prnp-deficient (Prnp0/0) mice resulted in antibody response specifically binding the cellular form of PrPC. Upon intravenous injection of wild-type mice, high PrPC-specific IgM responses were induced, whereas the T cell-dependent switch from the IgM to the IgG subclass was less pronounced. As a consequence, anti-PrPC titers were rather short-lived. The impaired subclass switch was probably related with host T cell tolerance to endogenous PrPC. Attempts to increase anti-PrPC IgG responses in wild-type mice via administration of VLP-PrPD111 emulsified in various different adjuvants failed. Nevertheless, in single individuals low IgG antibodies were induced after immunization of VLP-PrPD111 emulsified in CFA. To circumvent T cell tolerance in wild-type mice, a multitude of different immunization strategies was tested, including priming and boosting protocols with different types of VLP or VLP expressing PrPC together with foreign T helper epitopes. Overall, those efforts did not improve anti-PrPC IgG responses in wild-type mice. Interestingly, anti-PrPC antibodies induced in Prnp0/0 mice reduced PrPSc levels in prion infected cell cultures, whereas serum of vaccinated wild-type mice did not. To assess the protective capacity of VLP-PrPD111 induced immune responses, vaccinated wild-type mice were infected with scrapie (RML 5.0). Unfortunately, vaccinated mice did not show a significant delay in the onset of scrapie. In a last part of the thesis it was studied whether in the absence of T cell help activated “memory” B cells were able to produce anti-PrPC specific antibodies. To address this question, PrPC-specific memory B cells were sorted from vaccinated Prnp0/0 mice and adoptively transferred into wild-type recipient mice. Upon VLP-PrPD111 challenge, no PrPC-specific IgG titers were induced in the recipients. Nevertheless, several VLP-PrPD111 challenged recipient mice were protected against scrapie infection. In conclusion, VLP were characterized as highly immunogenic vaccines that were used to elucidate various questions concerning adaptive immune response and basic mechanisms of PrPC-specific tolerance vs. immunity. Remarkably, VLP-PrPD111 was able to induce native PrPC-specific antibodies in wild-type mice but major difficulties associated with PrPC-specific tolerance made efficacious scrapie vaccination impossible. New vaccination approaches are being tested to overcome these limitations.
Weltweit sind Trockengebiete in ständiger Veränderung, verursacht durch natürliche klimatische Schwankungen und oftmals durch Prozesse der Landdegradation. Auch weisen die meisten semi-ariden Naturräume eine große räumliche Heterogenität auf, hervorgerufen durch ein kleinräumiges Mosaik aus Gräsern, kleineren Sträuchern und Bereichen offenliegenden Bodens. Die Dichte der Vegetation wird primär vom pflanzenverfügbaren Wasser bestimmt, aber auch der Entwicklungs- und Degradationszustand der Böden sowie anthropogen bedingte Faktoren spielen hierbei eine Rolle. Zur Charakterisierung und Kartierung der Vegetation sowie zur Bewertung des Bodenerosionsrisikos und des Degradationszustands hat sich die Erhebung der Bedeckungsgrade von vitaler, photosynthetisch aktiver Vegetation (PV), von abgestorbener oder zeitweise vertrockneter und somit nicht photosynthetisch aktiver Vegetation (NPV) sowie von offenliegendem Boden als zweckmäßig herausgestellt. Die Nutzung der Fernerkundung für diese Aufgabe erfolgt zumeist nur für kleinmaßstäbige Kartierungen und – im Falle von Multispektralsensoren – unter Vernachlässigung nicht-photosynthetisch aktiver Vegetation. Die räumliche Variabilität der Vegetation-Boden-Mosaike liegt oftmals in der Größenordnung von wenigen Metern und somit unterhalb des räumlichen Auflösungsvermögens von Fernerkundungssystemen. Um dennoch die verschiedenen Anteile innerhalb eines Pixels identifizieren und quantifizieren zu können, sind Methoden der Subpixel-Klassifikation notwendig. In dieser Arbeit wird eine Methodik zur verbesserten und automatisierbaren Ableitung von Bodenbedeckungsgraden in semi-ariden Naturräumen vorgestellt. Hierzu wurde ein Verfahren zur linearen spektralen Entmischung in Form einer Multiple Endmember Spectral Mixture Analysis (MESMA) entwickelt und umgesetzt. Durch diese Methodik kann explizit die spektrale Variabilität von Vegetation und Boden in das Mischungsmodellmiteinbezogen werden, und quantitative Anteile für die funktionalen Klassen PV, NPV und Boden innerhalb eines Pixels erfasst werden. Durch die räumliche Kartierung der verwendeten EM wird weiterhin eine thematische Klassifikation erreicht. Die hierfür benötigten Informationen können – wie im Falle der Spektren reiner Materialien (EM-Spektren) – aus den Bilddaten selbst abgeleitet werden, oder können – wie ein Geländemodell und die Information über den Scanwinkel – im Zuge der Vorprozessierung aus weiteren Datenquellen erzeugt werden. Hinsichtlich der automatisierten EM-Ableitung wird eine zweistufige Methodik eingesetzt, welche auf einer angepassten Version des Sequential Maximum Angle Convex Cone (SMACC)-Verfahrens sowie der Analyse einer ersten Entmischungsiteration basiert. Die Klassifikation der gefundenen potentiellen EM erfolgt durch ein merkmalsbasiertes Verfahren. Weiterhin weisen nicht-photosynthetisch aktive Vegetation und Boden eine hohe spektrale Ähnlichkeit auf. Zur sicheren Trennung kann die Identifikation schmaler Absorptionsbanden dienen. Zu diesen zählen beispielsweise die Absorptionsbanden von Holozellulose und – je nach Bodentyp – Absorptionsbanden von Bodenmineralen. Auch die spektrale Variabilität der Klassen PV und NPV erfordert zur sicheren Unterscheidung die Verwendung biophysikalisch erklärbarer Merkmale im Spektrum. Hierzu zählen unter anderem die Stärke der Chlorophyll-Absorption, die Form und Lage der ’RedEdge’ und das Auftreten von Holozellulosebanden. Da diese spektrale Information bei herkömmlichen Entmischungsansätzen nicht berücksichtigt wird, erfolgt überwiegend eine Optimierung der Gesamtalbedo, was zu einer schlechten Trennung der Klassen führen kann. Aus diesem Grund wird in der vorliegenden Arbeit der MESMA-Ansatz dahingehend erweitert, dass spektrale Information in Form von identifizierten und parametrisierten Absorptionsbanden in den Entmischungsprozess mit einfließt und hierdurch das Potential hyperspektraler Datensätze besser genutzt werden kann. Auch wird in einer zusätzlichen Entmischungsiteration die räumliche Nachbarschaft betrachtet, um insbesondere die Verwendung des sinnvollsten Boden-EMs zu gewährleisten. Ein zusätzliches Problemfeld stellt die numerische Lösung des überbestimmten und oftmals schlecht konditionierten linearen Mischungsmodells dar. Hierzu kann durch die Verwendung des BVLS-Algorithmus und des Ausschlusses kritischer EM-Kombinationen eine numerisch stabile Lösung gefunden werden. Um die oftmals immense Rechenzeit von MESMA-Verfahren zu verkürzen, besteht die Möglichkeit einer iterativen EM-Auswahl und somit die Vermeidung einer Lösung des Mischungssystems durch Berechnung aller EM-Kombinationen (’Brute-Force’-Ansatz). Ein weiterer wichtiger Punkt ist die explizite pixelweise Angabe zur Zuverlässigkeit der Entmischungsergebnisse. Dies erfolgt auf Basis des Mischungsmodells selbst, durch den Vergleich zu empirischen Regressionsmodellen, durch die Berücksichtigung des lokalen Einfallswinkels sowie durch die Integration von Qualitätsangaben der Ausgangsdaten. Um das Verfahren systematisch und unter kontrollierten Bedingungen zu verifizieren und um den Einfluss verschiedener externer Parameter sowie die typischen Genauigkeiten auf einer breiten Datenbasis zu ermitteln, wird eine Simulationskette zur Erzeugung synthetischer Mischungen erstellt. In diese Simulationen fließen Feldspektren von Böden und Pflanzen verschiedener semi-arider Gebiete mit ein, um möglichst viele Fälle abdecken zu können. Die eigentliche Validierung erfolgt auf HyMap-Datensätzen des Naturparks ’Cabo de Gata’ in der andalusischen Provinz Almería sowie auf Messungen, die begleitend im Feld durchgeführt wurden. Hiermit konnte die Methodik auf ihre Genauigkeit unter den konkreten Anforderungen des Anwendungsbeispiels überprüft werden. Die erzielbare Genauigkeit dieser automatisierten Methodik liegt mit einem mittleren Fehler um rund 10% Abundanz absolut im selben Wertebereich oder nur geringfügig höher als die Ergebnisse publizierter manueller MESMA-Ansätze. Weiterhin konnten die typischen Genauigkeiten der Verifikation im Zuge der Validierung bestätigt werden. Den limitierenden Faktor des Ansatzes stellen in der Praxis fehlerhafte oder unvollständige EM-Modelle dar. Mit der vorgestellten Methodik ist somit die Möglichkeit gegeben, die Bedeckungsgrade quantitativ und automatisiert im Subpixelbereich zu erfassen.
Wie die vorliegenden und auch früheren Untersuchungen zeigen, können psychometrisch nur wenige, aber für unterschiedliche Fragebögen und unterschiedliche Stichproben sehr stabile Grunddimensionen des Lebensgeschehens erfasst werden, die auch nur einen Teil der Varianz zu erklären vermögen. Das bedeutet, dass psychometrisch nur ein Teil des Le-bensgeschehens erfasst werden kann. Jedoch kann die Möglichkeit, das Lebensgeschehen psychometrisch zu erfassen, durch zwei weitere psychometrische Ansätze ergänzt werden: Erstens durch die psychometrische Erfassung der Persönlichkeit mittels Persönlichkeitsfragebögen. Zweitens wird der Erklä-rungshorizont noch erweitert, wenn außer den Grunddimensionen des Lebensgeschehens und der Persönlichkeit noch bedeutsame Lebensereignisse berücksichtigt werden. Dies geschieht mittels Listen von bedeutsamen Lebensereignissen wie etwa Geburt von Ge-schwistern und später von eigenen Kindern, Tod der Eltern oder einer nahe stehenden Be-zugsperson, Hochzeit, Umzug in eine andere Umgebung, Erwerb von schweren chroni-schen Krankheiten oder beruflicher Erfolg bzw. Misserfolg. Biographische Fragebögen, Persönlichkeitsfragebögen und Listen bedeutsamer Lebens-ereignisse sind gemeinsam in der Lage, psychometrisch eine differenziertere Erklärung dessen zu geben, was sich im Leben ereignet.
Die Meiose ist eine besondere Art der Zellteilung, die während der Keimzellreifung stattfindet. Sie umfasst zwei aufeinander folgende Zellteilungen mit nur einer DNA-Repli-kationsrunde, wodurch aus einer diploiden Ausgangszelle vier haploide Gameten entstehen. In der ersten meiotischen Teilung werden die homologen Chromosomen miteinander rekombiniert und voneinander getrennt, in der Meiose II findet die Trennung der Schwesterchromatiden statt. Für den korrekten Ablauf dieser Prozesse musste sich eine spezielle molekulare Architektur des meiotischen Chromosoms entwickeln welche die Synapse der homologen Chromosomen durch den Synaptonemalkomplex (SC) beinhaltet. SCs sind evolutionär hochkonservierte, meiosespezifische Proteinkomplexe, die eine zentrale Bedeutung für Synapse, Rekombination und Segregation der homologen Chromosomen haben. Ein SC besteht aus zwei lateralen Elementen (LEs), die den Achsen der homologen Chromosomen aufgelagert sind, einer zentralen Region (CR) und einem zentralen Element (CE). Eine Hauptstrukturkomponente der LEs in Vertebraten ist das Synaptonemalkomplexprotein, SYCP3. Um die molekulare Architektur des SC besser zu verstehen und die Bedeutung von SYCP3 für die Zusammenlagerung der LE aufzudecken, wurden die Polymerisationseigenschaften von SYCP3, exprimiert in somatischen Zellen, erforscht. In diesem experimentellen Ansatz polymerisierte SYCP3 autonom zu stabilen, höher geordneten, filamentösen Strukturen. Die „Coiled-Coil“-Domäne und die flankierenden, evolutionär konservierten Motive sind dabei notwenig, und nach Deletion des weniger konservierten N-terminalen Bereichs auch ausreichend für die Bildung der höher geordneten Strukturen. Der N-Terminus hingegen spielt eine Rolle in der Stabilität der Polymärstrukturen, welche durch Phosphorylierung zweier Serinreste im N-terminalen Bereich beeinflusst werden könnte. Obwohl die Struktur des SC in der Evolution hochkonserviert ist, sind die Protein-komponenten auf Aminosäuresequenzebene sehr unterschiedlich und weisen wenn überhaupt eine strukturelle Homologie in ihrer Domänenorganisation auf. Um den SC-Aufbau und dessen Funktion besser verstehen zu können, wurden die orthologen SC-Proteine zwischen taxonomisch entfernten Spezies Ratte und Medaka verglichen. Es konnte gezeigt werden, dass trotz der Unterschiede in den Aminosäuresequenzen die sich in den letzen 450 Millionen Jahren zwischen Fisch- und Säugern-SYCP3 akkumuliert haben, die Eigenschaften der Proteine vergleichbar sind, und das sie unter experimentellen Bedingungen miteinander interagieren und zu höher geordneten Strukturen kopolymerisieren können.
Herzchirurgische Eingriffe bei Patienten mit terminaler, dialysepflichtiger Niereninsuffizienz

Patienten mit TDNI, bei denen herzchirurgische Eingriffe vorgenommen werden müssen, gelten als Hochrisikopatienten. Diese retrospektive Studie berichtet über 61 Patienten mit TDNI, die in unserer Klinik von 1984 bis 2001 unter Verwendung der Herz-Lungen-Maschine operiert wurden. Beschrieben wird deren präoperativer klinische Status und dessen Einfluss auf den peri- und postoperativen Verlauf, Morbidität und Letalität, sowie auf die Langzeitergebnisse. 1. Es wurden 46 Männer und 15 Frauen im Alter zwischen 34 und 76 Jahren (Durchschnittsalter 58.6 +10 Jahre) untersucht. 2. Bei 47 Patienten wurde eine ACB-Operation durchgeführt. 14 Patienten erhielten einen Herzklappenersatz, davon 10 einen Aortenklappenersatz (1 Patient mitzusätzlich ACB), 3 Patienten einen Mitralklappenersatz und 1 Patient eine AKE, MKE und zusätzlich ACB. 3. Alle 61 Patienten waren präoperativ seit 1 Monat bis 26 Jahre (im Durchschnitt 57.5 +37.5 Monate) dialysepflichtig. 4. Präoperativ hatten 20 Patienten einen Myokardinfarkt (33.8%), 13 Patienten einen Diabetes mellitus (21.3%), 42 Patienten eine Arterielle Hypertonie (68.8%), 23 Patienten eine Hyperlipoproteinämie (37.7%), 15 Patienten wiesen eine pAVK auf (24.6%) und 13 Patienten waren Raucher. Bei 13 Patienten (21:3%) konnten verschiedene neurologische Erkrankungen diagnostiziert werden. 5. 9 Patienten wurden notfallmäßig operiert, wovon 7 eine Herzbypassoperation und 2 eine Klappenoperation erhielten. 6. Die Patienten wurden mittels HLM und Blutkardioplegie operiert, lediglich ein Patient wurde in off-pump-Technik operiert. 7. Die Dauer des extrakorporalen Kreislaufes betrug im Durchschnitt 115,6 Minuten, die durchschnittliche postoperative Beatmungsdauer 18,35 +13,7 Stunden. 8. Perioperativ entwickelten 30 Patienten (49,2%) Komplikationen, betroffen waren 46% der Koronarpatienten und 50% Herzklappenpatienten. 9. Eine Mediastinitis trat bei 6,6% der Patienten auf. Wundheilungsstörungen an der Venenentnahmestelle mussten bei 8,8% aller Patienten behandelt werden. Punktionsbedürftige Pleuraergüsse wurden bei ebenfalls 8,8% behandelt. 10. 5 Patienten des Gesamtkollektivs verstarben perioperativ. Hieraus resultiert eine Operationsletalität von 8,2%. 3 Patienten verstarben im akuten Herzversagen, ein Patient an einer Sepsis aufgrund einer Peritonitis bei akuter Pankreatitis, ein Patient verstarb im Multiorganversagen nach langer HLM-Dauer und Gerinnungsstörung mit Massentransfusion. 11. Eine Reoperation war bei 2 Patienten nötig geworden; davon musste ein Patient 5 Monate nach der Erstoperation einer Aortenklappenendokarditis erneut wegen einer Prothesenendokarditis operiert werden. Der zweite Patient wurde 2 Jahre postoperativ wegen einer Klappenprotheseninsuffizinez und KHK reoperiert werden. Beide Patienten verstarben bei der durchgeführten Re-Operation. 12. Die postoperative Überlebensrate nach 1, 2, 3, 4 und 5 Jahren betrug jeweils 91,8%, 77,2%, 69,2%, 58,2% und 51,2%. 13. Eine klinisch spürbare Verbesserung der Symptomatik äußerten 71,8% aller untersuchten Patienten mit vollständigem Follow up (28 von 39). 14. Postoperativ erhielten 6 der Studienpatienten eine Nierentransplantation. 15. Eine fortgeschrittene Herzinsuffizienz mit Stadium NYHA III°-IV° insbesondere bei Herzklappenoperation und pAVK erhöhen die postoperative Letalität. 16. Limitierend für die Interpretation der Ergebnisse dieser Studie sind die retrospektive Analyse und das kleine Patientenkollektiv (61 Patienten), was jedoch durchaus vergleichbar ist mit anderen Studien zu diesem Thema (Tabelle 9). Andererseits sind in dem großen Untersuchungszeitraum von 1984 bis 2001 viele Weiterentwicklungen und Erfahrungen eingeflossen, die gerade diesem speziellen Kollektiv zu Gute kamen.
Die Dissertation India Book House und die Comic-Serie Amar Chitra Katha (1970-2002) ist eine interdisziplinär angelegte Studie. Sie vermittelt Erkenntnisse über einen vernachlässigten Bereich der Mediengeschichte Indiens. Im ersten Teil wird das Modell "Kreislauf der Kultur" auf das Medienprodukt Amar Chitra Katha angewandt. Dabei werden fünf Prozesse berücksichtigt: Repräsentation, Produktion, Identität, Konsum und Regulierung. Im zweiten Teil werden ausgewählte ACK-Ausgaben aus den Kategorien Mythologie und Geschichte formal-inhaltlich analysiert und verglichen. Die jeweiligen Unterschiede in der Bearbeitungsform der ACK-Ausgaben werden auf die Erkenntnisse des ersten Teils bezogen. Dadurch konnten die arbeitsbedingten und verlagspolitischen Motive der Transformationen der ACK-Serie belegt werden.
Bestimmung der adulten Neurogenese im Hippocampus von NOS-III-Knockout-Mäusen Ziel dieser Arbeit war es, einen möglichen Effekt von NOS-III auf die adulte Neurogenese (AN) zu erforschen. Einige Hinweise, wie z.B. die Expression dieses Enzyms im Endothel und die damit verbundene räumliche Nähe zu neuronalen Stammzellen, weisen darauf hin, dass dieses Enzym und das von ihm synthetisierte NO das Potential besitzen, die AN zu beeinflussen. Für die vorliegende Untersuchung wurden deshalb die Gehirne von NOS-III-Knockout-Mäusen im Vergleich zu den Wildtypen und den für dieses Gen heterozygoten Mäuse auf Proliferation und Survival neuronaler Stammzellen im Bereich des Gyrus dentatus untersucht. Die Auswertung der Ergebnisse zeigte eine signifikant (23%) verringerte Proliferationsrate bei NOS-III-Knockout-Mäusen. Bei den Werten für die Survivalrate gab es jedoch keine signifikanten Unterschiede zwischen den verschiedenen Genotypen. Die Doppelmarkierung der Gehirnschnitte mittels Konfokalmikroskopie und Antikörpern gegen BrdU, den neuronalen Marker NeuN bzw. den Astrozytenmarker GFAP zeigten, dass die neu gebildeten Zellen vorwiegend zu reifen Neuronen und kaum zu Astrozyten differenzierten. Geschlechtsspezifische Unterschiede wurden ebenso wenig beobachtet wie morphologische Unterschiede im Gyrus dentatus.
