Prostaglandin D2 (PGD2) is the most common prostaglandin type of tile rat brain. Recently a neurornodulator role for PGD2 has been suggested. In the present work the central cardiovascular and thermal effects of PGDz were studied in urethane-anaesthetised rats. Mlen adrndnistered at the doses of 0.001-10 ~g/rat into the lateral cerebral ventricle(i.c.v.), PGD2 slightly increased the blood pressure, heart rate and body ternpera~ ure. The highest dose caused also an initial hypotensive effect. Upon lntravenous injections PGD2 (0.1-10 ~g/rat) initially decreased and then weakly increased the blood pressure but had only negligible effects on heart rate and body temperature. Central pretreatment with sodium meclofenamate or indomethacin (1 mg/rat i.c.v.) antagonised effectively all the recorded central effects of PGD2. The central cardiovascular and thermal effects of PGD2 were much weaker than those obtained earlier with other prostaglandins, such as PGF2alpha and PGE2.. Therefore, in spite of its abundance in the brain PGD2 may not be very important for the central cardiovascular and thermal regulation in the rat.
Prostaglandin E2 (PGE2) increased the blood pressure, heart rate and body temperature, when administered at the doses ofO.OOI-IO,ug into the lateral cerebral ventricle (i.c.v.) of the urethane-anesthetised rat. The highest dose of 10 ,ug/rat induced a strong initial hypotensive effect. lntravenously (i.v.), PGE2 at the doses of 0.01-10 ,ug/rat caused a biphasic blood pressure response with dose-related initial decreases followed by slight increases in blood pressure. The heart rate and body temperature were slightly increased by i.v. administrations of PGE2 . The highest i.v. dose of 10 ,ug/rat initially decreased also the heart rate. Central pretreatment with indomethacin ( I mg/rat i.c.v.) partly antagonised all of the recorded central effects of PGE2 , while sodium meclofenamate (I mg/rat i.c. v.) abolished the hypertensive response to i.c. v. administered PGE2 but failed to significantly affect the PGE2-induced rises of heart rate and body temperature. The results support the previous suggestions that PGE2 may participate in the central cardiovascular and thermoregulatory contro!. The results also suggest that indomethacin and sodium meclofenamate antagonize the effects of exogenous prostaglandins. Since sodium meclofenamate, unlike indomethacin, affected preferentially the hypertensive response to centrally administered PGE2 , there may be differences in the sites and/or modes of action between these drugs.
Vorgestellt wird ein Versuch, die in der einschlägigen literatur postulierte multifaktorielle Bedingtheit von Rechtschreibleistungen in der Grundschule über ein angemessenes methodisches Design zu prüfen. Zentral ist dabei die Frage, ob sich identische Kausalstrukturen für beginnende und geübte Rechtschreiber (Schüler der zweiten w. vierten Klasse) nachweisen lassen. Konventioneffe Verfahren der Kausafanafysa hatten sich in der Primärstudie (Schneider 1980) als unökonomisch und wenig aussagekräftig erwiesen, 10 daß in der Sekundäranalvse auf eine flexiblere Prozedur zurückgegriffen wird. Mit diesem Verfahren zur AnaIVse von Strukturgleichungssysteman (LiSREL) ist es möglich, für die Gruppe der Zweitund Viertkläßler Modelle zu entwickeln und zu überprüfen, die mit den Ausgangsdaten kompatibel sind. Als wesentliches Ergebnis zeigt sich, daß die theoretisch postulierte Bedingungsstruktur nur für die Viertkläßler (eingeschränkt) bestätigt werden kann, während für die Schüler der zweiten Klassenstufe ein grundlegend verschiedenes Muster resultiert.

In an experimental analog of verbal examinations, the call-up situation, the effects of two dosages of a tranquillizing agent (lopirazepam) are compared to placebo treatment. 72 male and female, healthy, young volunteers have been randomly assigned to 12 groups of 6 subjects each. Pulse frequency and performance were registered. The results indicated differential drug effects which were interpreted according to the hypotheses of "differential effects of social stressors". If a situation was highly challenging for a subject, the application of a tranquillizer in an adequately high dosage enabled him to perform well in spite of or because of strong increases in pulse frequency.

A novel chromatin configuration is described in lampbrush chromosomes of Pleurodeles waltlii oocytes which is different from transcriptionally inactive chromatin as weil as from the various forms of transcribed chromatin hitherto described. This novel type of chromatin is not arranged in Christmas tree-Iike configurations of densely packed lateral ribonucleoprotein (RNP) fibriIs but is characterized by a periodic alternating pattern of thick and thin regions which occur in clusters 01 some 10,000 repeats. Each thickened unit with an average length of 45 nm contains two c10sely spaced particles, the putative RNA polymerases, and each thickened unit is separated from the next one by a beaded chromatin spacer with a length of about 80 nm. This chromatin spacer contains on average two particles of approximately 14 nm in diameter, assumed to be nucleosomes. The thickened regions are interpreted to represent short transcriptional units containing approximately 130 base pairs of DNA which are separated from each other by nontranscribed spacers of 240-400 base pairs of DNA. The possibility is discussed that these transcriptional units represent 5S rRNA or tRNA genes.
While clear evidence exists for the direct involvement of cytolysins in the pathogenesis of Gram-positive bacteria, the significance of Gram-negative haemolysins remains unclear. This paper presents briefly data indicating a role for haemolysin production in infections caused by Escherichia coli and also experiments which have allowed an analysis of the molecular basis of the haemolysis among pathogenic and non-pathogenic strains of this species.
We have cloned the chromosomal hemolysin determinants from Escherichia coli strains belonging to the four O-serotypes 04, 06, 018, and 075, The hemolysin-producing clones were isolated from gene banks of these strains which were constructed by inserting partial Sau3A fragments of chromosomal DNA into the cosmid pJC74. The hemolytic cosmid clones were relatively stable. The inserts were further sub cloned either as Sail fragments in pACYC184 or as BamHI-SaLI fragments in a recombinant plasmid (pANN202) containing cistron C (hlye) of the plasmid-encoded hemolysin determinant. Detailed restriction maps of each of these determinants were constructed, and it was found that, despite sharing overall homology, the determinants exhibited minor specific differences in their structure, These appeared to be restricted to cistron A (hlyA), which is the structural gene for hemolysin. In the gene banks of two of these hemolytic strains, we could also identify clones which carried the genetic determinants for the mannose-resistant hemagglutination antigens Vb and VIc. Both of these fimbrial antigens were expressed in the E. coli K-12 clones to an extent similar to that observed in the wild-type strains. These recombinant cosmids were rather unstable, and, in the absence of selection, segregated at a high frequency.
The role of macrophages in primary and secondary infection of mice with Salmonella typhimurium
(1982)
Elimination of macrophages with high-molecular dextran sulphate (OS) markedly impairs resistance of mice to primary infection with smooth, virulent strains of Salmonella typhimurium, whereas stimulation of this system by killed Bordetella pertussis organisms increases resistance. In infection with rough, avirulent strains of S. iyphimurium the elimination of macro phages was not followed by an essential loss of resistance, and it appears that other non-specific defence mechanisms, for example the complement system, may have compensated for the lack of macrophages. Macrophages, therefore, play an important role in defence during primary infection with virulent strains. In immunity to challenge infection with S. typhimurium, macrophages play an even more significant role. Treatment with OS completely removes immunity, and both humoral and cell-mediated immune mechanisms seem to require the participation of macrophages.
Die Kristallstruktur von 4.CH\(_3\)(C\(_6\)H\(_4\))-TI(S\(_2\)CN(C\(_2\)H\(_5\))\(_2\))\(_2\) (P2\(_1\)/c, a = 11,973(3) A, b = 10,692(3) A, c = 19,232(4) A, ß = 114,02(2)°, Z = 4) und C\(_6\)H\(_5\)-Bi(S\(_2\)COCH\(_3\))\(_2\) (P2\(_1\)/c, a = 6,395(2) A, b = 24,684(8) A, c = 9,732(3) A, ß = 101,38(3)°, Z = 4) konnte aus Röntgendiffraktometerdaten von Einkristallen ermittelt werden. Die interatomaren Abstände zeigen daß, die Koordination von Dithiocarbamat und Xanthogenat an die Metallatome wie schon vermutet stark asymmetrisch zweizähnig und ausschließlich über Schwefel erfolgt. Die Koordinationsunter· schiede zwischen Bismut und Thallium ergeben ein deutliches Indiz für die "stereochemische Aktivität" des freien Elektronenpaares am Bismutatom.
Visualizing nucleic acids (DNA, RNA), nucleoprotein complexes and chromatin requires the use of special electron microscopicspreading techniques. In part 4 (27 refs.), methods are outlined for spreading DNA and RNA molecules for electron microscopic observation, these methods using modifications of the basic protein film method developed by A. Kleinschmidt and R. K. Zahn (1959). Hybridization techniques that allow the observation of heteroduplexes formed between two DNA molecules or between DNA and RNA molecules are reviewed, with special emphasis being placed on the DNA-RNA hybrids as a tool for elucidating RNA splicing. Techniques for studying DNA-protein interactions without the use of a protein monolayer film are mentioned. Finally, the "Miller spreading technique" for visualizing the nucleosomal organization of eukaryotic chromatin as well as the transcription of genes is discribed and illustrated.

Repeat sequences are transcribed in the germinal vesicles of amphibian oocytes. In the hnRNA population both complements of the repeats are found and can be readily detected because they form intermolecular duplex structures. The structure and formation of duplex regions have been studied in the hnRNA of Xenopus laevis, Triturus cristatus, Amphiuma means and Necturus maculosus, a series of amphibians of increasing genome size (C-value). In T. cristatus, the duplex structures are mostly 600- 1200 bp in length, whereas in X. laevis they are shorter and in N. maculosus they tend to be longer. Although the proportion of RNA sequence capable of rapidly forming duplex structures is different in different organisms, this property bears no relationship to C-value. However the sequence complexity of complementary repeats, as estimated from the rate of duplex formation, does show an increasing trend with C-value. The complementary repeats found in oocyte hnRNA are transcribed from families of DNA sequence that are each represented in the genome by thousands of copies. The extent of cross-species hybridization is low, indicating that the repeat sequences transcribed in different amphibian genera are not the same. In situ hybridization experiments indicate that the repeat sequences are spread throughout the genome. The evolution and possible function of complementary repeats are considered.
