A recombinant plasmid was constructed containing the gene for bovine growth hormone joinea with the regulatory region and the region coding the signal sequence of the Escherichia coli alkaline phosphatase gene. In conditions of phosphorus starvation, which c~s derepression of alkaline phosphatase, expression was shown of the gene for bovine growth hormone, in addition to partial processing and secretion of protein into periplasm.
Primary infection of HEp-2 cells with rubella virus resulted in non-cytophatic longterm persistent infection. During four years of persistence the virus was produced in sufficient quantities (up to 6 logs PFU/ml) and did not differ from the parental variant in its pathogenicity for BHK-21 or RK-13 cells, or hemagglutinating activity, but formed smaller plaques. Persistent virus preserved the original antigenicity as judged from reciprocal hemagglutination-inhibition or plaque reduction-neutralization tests with polyclonal antisera. Both original and persistent rubella viruses were thermoresistant (T 56° C) and sligthly temperature-sensitive. Clonal analysis revealed presence of ts-mutants among both original and persistent virus clones with different degrees of plating efficiency at 40°/34° C. RNA fingerprinting showed only minor changes in persistent rubella virus.
Kanadaptin ist ein Multidomänenprotein, das in der Ratte in nahezu allen Geweben vorkommt und über kernimport- und kernexport-aktive Sequenzen verfügt. Die kernimport-aktive Sequenz konnte der NLS1 (189RKRK) zugeordnet werden. Die Kernexportaktivität wird möglicherweise durch eine Leucin-reiche Domäne (414LLQEPELELEAAV) vermittelt. Zusätzlich ist Kanadaptin in Mitochondrien nachweisbar. In humanen Zellen wird eine Isoform gebildet, die über eine zusätzliche aminoterminale FHA-Domäne verfügt. Solche Proteine zeichnen sich u.a durch DNA-bindende Eigenschaften aus (Murakami and Okayama, 1995; Allen et al., 1994; Durocher and Jackson, 2002). Dies erklärt auch die nukleäre Retention von Kanadaptin nach Verlust Zellkernmembranintegrität (Hübner et al, 2002) Für eine nukleäre Funktion spricht auch, dass infolge der Expression eines Kanadaptin-cDNA-Fragments eine unter diesen Umständen (d.h. infolge der Expression DNA-bindender/modifizierender Proteine) spezifische SOS-Antwort in E. coli ausgelöst werden kann (Perkins et al., 1999). Die Ergebnisse deuten somit eindeutig daraufhin, dass Kanadaptin an nukleären und/oder nukleinsäureabhängigen Prozessen beteiligt ist. In der Niere wurde mit Hilfe der in-situ-Hybridisierung Kanadaptin hauptsächlich in proximalen Tubuluszellen detektiert. Insgesamt scheint eine Interaktion zwischen Kanadaptin und kAE1 inzwischen immer unwahrscheinlicher. Schließlich ließ sich eine Interaktion zwischen Kanadaptin und kAE1 im Hefe-2-Hybrid-System nicht mehr reproduzieren (Hübner, persönliche Mitteilung) (Wongthida P. et al., 2006). Auch in embryonalen Nierenzellen (HEK293-Zellen) konnte keine Interaktion zwischen humanem Kanadaptin und kAE1 nachgewiesen werden (Kittanakom et al., 2004). Welche Funktionen Kanadaptin intrazellulär ausübt bleibt weiterhin enigmatisch und muss in zukünftigen Untersuchungen herausgefunden werden.
Die Stabiltät und Integrität der Epidermis beruht zu einem großen Teil auf der intakten Funktion der Desmosomen. Diese fleckförmigen Zellkontakte vermitteln extrazellulär die Haftung zwischen den Keratinozyten durch Desmocadherine und sind intrazellulär über Adaptorproteine im Intermediärfilamentsystem des Zellskeletts verankert. Diese Funktion ist bei der Autoimmunerkrankung Pemphigus gestört, die zu intraepidermaler Blasenbildung durch Akantholyse der Keratinozyten führt. Pemphigus vulgaris (PV) und Pemphigus foliaceus (PF) stellen die beiden Hauptvarianten dar, wobei PV durch Autoantikörper gegen die Desmocadherine Desmoglein (Dsg) 3 und oftmals zusätzlich gegen Dsg 1, PF durch Autoantikörper nur gegen Dsg 1 gekennzeichnet ist. Rho-GTPasen sind zelluläre Regulatorproteine, die das Aktinzytoskelett und verschiedene Zellkontakte beeinflussen. Die vorliegende Arbeit beschäftigte sich mit dem Einfluss von Rho-GTPasen bei der Regulation von desmosomal vermittelter Adhäsion. In einem zweiten Teil wurde die Beteiligung von Rho-GTPasen bei den Pemphigusvarianten PV und PF näher charakterisiert. Für den ersten Abschnitt wurden bakterielle Toxine verwendet, die spezifisch Rho GTPasen aktivieren bzw. inhibieren, während für den zweiten Teil IgG-Fraktionen von PV- und PF-Patienten in Kombination mit aktivierenden Toxinen zur Anwendung kamen. Eine Inhibition der drei Hauptvertreter der Rho-GTPasen in kultivierten Keratinozyten und humaner Epidermis führte zu einer Rarefizierung des Aktinfilamentsystems, zu Verlust von membranständig lokalisiertem Dsg 1 und 3 und zu Zelldissoziation sowie zu verminderter Dsg 1 und 3-vermittelter Haftung von Mikroperlen auf der Oberfläche von Keratinozyten. Die Aktivierung der GTPasen resultierte in vermehrter linearisierter Darstellbarkeit von Aktin und Dsg 3 an den Zellgrenzen und einer verstärkten Dsg-vermittelten Haftung. Pemphigus-IgG führten ebenfalls zu Zelldissoziation und Verlust von Dsg-Immunreaktivität in Keratinozytenkulturen, zu Spaltbildung in humaner Epidermis und zum Verlust der durch Dsg 1 und Dsg 3 vermittelten Adhäsion. Dies ging einher mit einer vermehrten Menge an nicht am Zytoskelett verankerten Dsg 3 und wurde durch eine p38MAPK-abhängige Verminderung der Aktivität von Rho A moduliert. Die Aktivierung von Rho A verhinderte die Ausbildung der Pemphigus-induzierten Effekte nahezu vollständig. Zusammenfassend regulieren Rho-GTPasen die desmosomale Haftung in Keratinozyten. Die Daten zeigen weiterhin, dass Pemphigus-IgG durch eine Inhibition von Rho A diese Regulation beeinträchtigt, was zu Schwächung der Zytoskelettverankerung von Desmogleinen und zu Haftungsverlust und Spaltbildung führt. Somit ist Rho A ein wichtiger Faktor der Pemphigus-Pathogenese und stellt einen Erfolg versprechenden Ansatzpunkt zur Entwicklung neuer Therapieoptionen dar.
The RS1 protein (gene RSC1A1) participates in regulation of Na+-D-glucose cotransporter SGLT1 and some other solute carriers. In subconfluent LLC-PK1 cells, RS1 inhibits release of SGLT1 from the trans-Golgi network and transcription of SGLT1. In subconfluent cells, RS1 is localized in the nucleus and the cytoplasm whereas confluent cells contain predominantly cytoplasmic RS1. In the present study, the mechanism and regulation of confluence-dependent nuclear location of RS1 was investigated. Confluence dependent nuclear location of RS1 was shown to be regulated by the cell cycle. A nuclear shuttling signal (NS) in pRS1 was identified that ensures confluence-dependent distribution of pRS1 and comprises nuclear localization signal (NLS) and nuclear export signal (NES). The NLS and NES of RS1 mediate translocation into and out of the nucleus via importin ß1 and CRM1, respectively, and the nuclear/cytoplasmic distribution of the RS1 protein is determined by the nuclear export activity. The adjacent protein kinase C (PKC) phosphorylation site at serine 370 of pRS1 was shown to control nuclear localization driven by NS and is necessary for the differential localization of RS1 in quiescent versus proliferating cells. Basing on the data of site-directed mutagenesis, PKC activation experiments and mass spectrometry analysis of RS1 phosphorylation, the following model of the regulation of RS1 nuclear location in LLC-PK1 cells was proposed. In subconfluent cells, RS1 is actively imported into the nucleus whereas nuclear export of RS1 is not active leading to accumulation of RS1 in the nucleus. After confluence, phosphorylation of serine 370 of pRS1 by PKC takes place leading to enhancement of RS1 nuclear export and predominantly cytoplasmic distribution of the protein in the confluent cells. The confluence-dependent regulation of RS1 localization may control SGLT1 expression during regeneration of enterocytes in small intestine and during regeneration of renal tubular cells after hypoxemic stress. Moreover, the gene expression profiling of mouse embryonic fibroblasts with RS1-/- genotype suggests that transcriptional regulation by RS1 might be important for the cell cycle and cell division. Since RS1 localization depends on the cell cycle, RS1 might play a role in the regulation of the solute carriers during specific phases of the cell cycle.

Verschluss- und Adherenskontakte zwischen olfaktorischen Neuronen, olfaktorischen Gliazellen und den epithelialen Zellen des peripheren olfaktorischen Systems bestimmen Barriere- und Adhäsionseigenschaften im olfaktorischen Epithel, sowie die Kompartimentierung und Axonwachstumsprozesse in den Fila olfactoria. Zur Untersuchung der zellulären und subzellulären Lokalisation von Verschlusskontaktproteinen (Occludin, Claudin 1-5, Zonula occludens proteins (ZO) 1-3) und Adherenskontaktproteinen (N-Cadherin, E-Cadherin, alpha-, beta-, p120-Catenin) wurden immunhistochemische und immunelektronenmikroskipsche Verfahren an Gewebeschnitten des olfaktorischen Systems der Ratte durchgeführt. Mit Ausnahme von Claudin 2 waren alle untersuchten Verschlusskontaktproteine in Kontakten des olfaktorischen Epithels kolokalisiert. Unterschiedliche Immunfluoreszenzintensitäten konnten in den Kontakten zwischen olfaktorischen Neuronen und epithelialen Zellen beobachtet werden. Immunreaktivität von Claudin 5 wurde in Kontakten der olfaktorischen Gliazellen in den Fila olfactoria lokalisiert, Claudin 1- Immunreaktivität konnte in peripheren Bereichen der Fila olfactoria beobachtet werden. Hierbei zeigten sich Unterschiede in der Lokalistaion der Claudine mit verschiedenen ZOs. Stützzellen bildeten durch N-Cadherin vermittelte Adherenskontakte mit den olfaktorischen Neuronen, E-Cadherin-vermittelte Adherenskontakte mit Drüsen- und Microvilluszellen, sowie durch N- und E-Cadherin vermittelte Adherenskontakte mit benachbarten Stützzellen aus. Alpha-, beta- und p120-Catenin konnten in allen Adherenskontakten des olfaktorischen Epithels nachgewiesen werden. Olfaktorische Neurone bildeten in Abhängigkeit von deren Reifestadium unterschiedlich häufig Adherenskontakte aus. Adherenskontakte in den Fila olfactoria wurden zwischen Gliazellen, zwischen Gliazellen und Axonen, sowie zwischen Axonen untereinander lokalisiert. In den meisten Adherenskontakten der Fila olfactoria zeigte sich Kolokalisation zwischen N-Cadherin und den verschiedenen untersuchten Cateninen. In der Peripherie der Fila olfactoria konnten durch E-Cadherin vermittelte Adherenskontakte beobachtet werden. Die Funktion von Interzellularkontakten wird maßgeblich durch ihren molekularen Aufbau bestimmt. Charakteristische molekulare Zusammensetzungen der neuronalen, epithelialen und glialen Verschlusskontakte im olfaktorischen Epithel und den Fila olfactoria lässt auf unterschiedliche Eigenschaften dieser Kontakte schließen. Adherenskontakte sind für Neuro- und Axogenesevorgänge von entscheidender Bedeutung, und es liegt nahe, dass diese Kontakte auch bei diesen Prozessen im peripheren olfaktorischen Epithel eine wichtige Rolle spielen. Um Untersuchungen zur Ausbildung und zu möglichen Funktionen von Kontakten zwischen olfaktorischen Gliazellen und olfaktorischen Neuronen auch in vitro zu ermöglichen, wurden Primärkulturen von olfaktorischen Gliazellen etabliert und erste Experimente zur Charakterisierung der Kulturen bezüglich ihrer Homogenität sowie bezüglich verschiedener in vivo gefundener Eigenschaften, wie die Expression von Kontaktproteinen und die Produktion des ciliary neurotrophic factor (CNTF) in Einzelkulturen und in Kokulturen mit olfaktorischen Neuronen durchgeführt.
Hieracium fallax Willd. und weitere Hieracium echioides-Zwischenarten im nordwestlichen Bayern

Die Grenze des riesigen eurasiatisch-kontinentalen Areals von Hieracium echioides Lumn. verläuft durch Mitteldeutschland, Zwischenarten aus der Hieracium echioides-Verwandtschaft (sect. Echinina) dringen westlich bis in die Oberrheinebene vor, sind aber im übrigen Süden und Südosten Deutschlands sehr selten oder fehlen. In den letzten Jahren wurden im Nordwesten Bayerns neue Wuchsorte von Hieracium auriculoides Láng (MTB 5526.31, 5924.44, 6125.13, 6223.22), H. calodon Tausch ex Peter (6123.21, 6125.13) und H. fallax Willd. (6223.21) nachgewiesen. Dies stellt den zweiten aktuellen Nachweis von H. fallax in Bayern dar, bemerkenswert ist ein Nachweis von H. auriculoides in der Rhön in ca. 700 m Meereshöhe.
Ein pflanzensoziologisches Modell der Schattentoleranz von Baumarten in den Bayerischen Alpen

The ecological niche, as a summary of the environment in which a tree species can live, is a central concept in vegetation ecology and its application in silviculture. While the fundamental niche focusses on the physiological constraints of survival and growth, the realised niche takes competition in real communities into account. To understand realised niches in a causal fashion requires knowledge of the life cycle of plant species. The concept of regeneration niche is based on the notion that establishment and juvenile growth are particularly sensitive stages. Obviously, silviculturalists must be particularly interested in regeneration niches. The database BERGWALD contains 4,934 phytosociological plots from mountain forests and related vegetation types of the Bavarian Alps. The detailed information on plant species composition (trees, tree regeneration, shrubs, herbs and bryophytes) and cover has so far been used extensively for deriving vegetation units, site types and groups of indicator species. In the present study the database content was analysed with regard to the ecology of tree species in general and their regeneration niche in particular. The availability of light as a crucial resource that changes during forest succession was estimated by calculating average Ellenberg indicator values (mL) based on total field and bryophyte layer composition. The relative frequency of plots across the mL gradient in the total database was juxtaposed to the occurrence of the 16 most common tree species in the tree and in the regeneration layer, respectively. [...] As expected, the realised niches of tree species on the light gradient corresponded broadly to Ellenberg's L-value of tree regeneration. As the regional climax, Abies alba and Fagus sylvatica have coincident optima of tree layer and juvenile occurrences in closed, mature stands. Ulmus glabra and Fraxinus excelsior, as species of lower altitude, exhibit niches most similar to these climax species, followed by Acer pseudoplatanus and Picea abies, two of the most frequent species, that occur from low elevations to the timber line. The intermediate role of these four species is confirmed, as regeneration occurs mostly at light levels higher than those found under adult trees of the same species. Against expectations, Taxus baccata clearly prefers stands with moderate to high light, as do Sorbus aria, Sorbus aucuparia and Alnus incana. While Larix decidua and Pinus cembra occupy very similar overall niches, tree layer and regeneration niches of Larix differ markedly, whereas coincident layer niches in Pinus cembra underpin its status as a climax species at tree line. Pinus sylvestris and Salix eleagnos are typical shade-intolerant pioneers, of which regeneration is practically restricted to non-forest vegetation. Pinus rotundata and Juniperus communis are small trees that are entirely restricted to open stands subject to geomorphological activity. The results demonstrate the potential of phytosociological databases for studying the niches of tree species. To be sure, such analyses are no replacement for physiological and experimental studies. The research community is invited to use this source as a reference framework and an empirical validation for more specialised research.
Iconography of the Genus Hieracium in central Europe - Part 1: General Description and Morphotypes

The genus Hieracium comprises more than one thousand sexual and apomictic species in Europe, with numerous intermediates and microspecies. Only a small fraction of the members of the genus Hieracium has been illustrated or photo-documented in the literature. Since many of these publications are difficult to obtain, only a few specialists are familiar with most of the species and subspecies described in the literature. In order to overcome this problem and encourage geobotanical research on the genus Hieracium, we decided to edit an iconography of central and southern European Hieracia in an electronical journal (Forum geobotanicum) with free international access through the internet. Part I of this endeavour contains descriptions and photographs of the morphological spectrum of the genus Hieracium. Here, we categorize the genus into 15 basic morphotypes. These types conform partly to the sections and subsections of the genus Hieracium, but are in some cases informal and may even include members of different sections. Classification of morphotypes is considered helpful to obtain a first rough picture of an unknown species that then can be traced to the species and subspecies level by using keys or, after completion of this iconography, simply by screening the relevant images. One particularly novel aspect of the present endeavour will be the regular inclusion of magnified images and scanning electron micrographs.
Zur Kenntnis des Ranunculus auricomus-Komplexes in Deutschland: Ranunculus suborbicularis spec. nova

Die bei Schwarz (1949) als R. vertumnalis abgebildeten Pflanzen entsprechen nicht dem Typusmaterial, sondern weichen durch fast kreisförmige Blattspreiten der Schlussblätter und geringe Blattteilung ab. Sie werden hier als R. suborbicularis spec. nov. beschrieben und mit ihrem Blattzyklus und weiteren Belegen abgebildet. Die bekannte Verbreitung erstreckt sich auf Berlin (wohl erloschen), Thüringen und Bayern. Insgesamt ist aufgrund der kleinen Zahl der Populationen eine starke Gefährdung anzunehmen.
In August 2004 the authors of this report undertook a survey on the Genus Oenothera (evening-primroses) in the valley of the river Main between the cities of Schweinfurt and Bamberg (Bavaria, Lower and Upper Franconia). 22 different taxa were identified, most of which are new to Bavaria, one new for Europe (Oenothera hazelae Gates) and another one new for Middle Europe (Oenothera stucchii Soldano). In this report a list of all Oenothera species found in Franconia is given and Oenothera stucchii, a particular tall (up to > 2 m) species with extremely long Hypanthia of 50-70 mm and fruit capsules with truncated teeth, is described and depicted in more detail.
Das Naturschutzgebiet "Garchinger Heide" ist ein Relikt der ehemals ausgedehnten Kalkmagerrasen auf Pararendzinen über Niederterrassenschotter im Alpenvorland. Bei jahrhundertelanger extensiver Landnutzung hat sich auf trockenen nährstoffarmen Böden eine artenreiche Vegetation mit zahlreichen seltenen und gefährdeten Arten entwickelt. Die "Altheide", welche den größten Teil des Naturschutzgebiets einnimmt, ist geprägt durch flachgründige, nährstoffarme Böden, welche eine geschlossene Vegetation mit einem hohen Anteil an Arten der Halbtrockenrasen (hauptsächlich Mesobromion erecti und Cirsio-Brachypodion) tragen. Das 1945 durch Oberbodenabtrag begonnene, aber nicht mehr ausgebaute und verwendete „Rollfeld“ weist einen niedrigen Feinbodenanteil, welcher auf eine geringe Wasserverfügbarkeit schließen lässt, sowie niedrige Gehalte an Gesamtstickstoff und CAL-austauschbaren P2O5 auf. Die Vegetation ist lückig mit einem hohen Anteil an Trockenrasenarten (Xerobromion, Sedo-Scleranthetea, Sesleritalia albicantis). Das heutige Vorkommen zahlreicher Arten, welche in früheren Untersuchungen (1956 und 1986) nicht gefunden wurden, lässt auf eine fortschreitende Sukzession der Vegetation des Rollfeldes hin zu der der Altheide schließen. Der 1959 zum Naturschutzgebiet hinzu gekaufte ehemalige Acker unterscheidet sich immer noch deutlich von der Altheide. Eine allmähliche Annäherung der Standortbedingungen ist jedoch zu beobachten. So sanken die Gehalte an CAL-austauschbarem P_ 2 O_5 und K_2 O im Vergleich zu Werten aus dem Jahr 1993 deutlich ab. Trotz des immer noch hohen Anteils an Grünlandarten (Molinio-Arrhenatheretea) konnten sich im Vergleich zu 1986 mehr Magerrasenarten etablieren.
Prunus spinosa L. (Rosaceae) is one of the most widespread members of the genus Prunus in middle europe. Its morphological plasticity resulted in a number of described taxa at subspecific level. Since the early neolithic times, drupes of the plum family are recorded and exhibit already a remarkable diversity in size and form. Here we present a short historical account to the use of P. spinosa and an overview of the different taxonomic treatments. We examined distribution patterns in general and in particular in the central valley of the river Saale (Thuringia) with respect to ecological, edaphic and climatic factors. We assessed within 16 populations the variability of 22 metric and 10 qualitative morphological characters at 7 different locations. Population sites included forest-, way- and fieldsides, as well as lightish pine forests. Pollen fertility did not increase during the flowering period, all flowers were directly fully fertile from the beginning. In contrast, glucose content varied significantly depending of the status of fertilization. Epicuticular wax structure was without variation amongst the populations. P. spinosa leaves are covered with a smooth layer of slightly striated wax. Morphological characters were scored on 270 branches and 506 fruits. Most of the characters showed enormous variability among and within populations such as metrics of leaves, thorns and character states of flower morphology. The lowest variability among populations and therefore not dependend of modificatory factors was found in fruit characters. Since kernel morphology seems to be genetically rather than modificatory controlled, we applied the 3 taxonomical concepts of Werneck, Kühn and Scholz u. Scholz to identify evolutionary units at subspecific levels. However, population variability was still so high, that from our study here we can not support an infraspecific classification of Prunus spinosa L.
Based on queries of the phytosociological databank BERGWALD, a compilation of 3.504 forest vegetation plots from the Bavarian Alps, the ecological niche of Abies alba is re-assessed. The tree species occurs mostly admixed in mountain forests with Fagus and Picea rather than forming distinctive communities of its own. Climatically, Abies is widely distributed to the upper limit of the montane belt, but occurs only sparsely in subalpine forest. Analysis of Ellenberg indicator values based on total species composition yielded the following results: As a tolerant species Abies regeneration has a marked preference for shady forest, which in turn Abies tree layers themselves help create. It also has a clear preference for acidic topsoil conditions. Sites with low N-supply, such as early successional stages on raw carbonate soils, are rarely colonised by Abies. Also, dry and markedly wet forest sites in the region are avoided by Abies. Permutation-based indicator species analysis found a large number of common forest species as being significantly associated with Abies and its frequent companion Fagus sylvatica, whereas there is a negative relationship with more specialised Seslerion and Erico-Pinion species. As Abies alba has very few specific companion species not shared with either Fagus or Picea, the delimitation of Abietetum-syntaxa appears mostly motivated by ecological rather than phytosociological considerations. As a result of its susceptibility towards game browsing, Abies regeneration is an indicator of high woody species richness. The study broadly confirms Abies alba's status as a climax species intermediate between Fagus and Picea, and demonstrates the potential of large phytosociological databanks for niche modelling.
Polyspezifische organische Kationentransporter (OCTs) der SLC22 Familie transportieren organische Kationen entlang eines elektrochemischen Gradienten und spielen eine entscheidende Rolle bei der Ausscheidung und Gewebeverteilung von endogenen organischen Kationen und bei der Aufnahme, Ausscheidung und Verteilung von kationischen Medikamenten und Toxinen. Zu den endogenen transportierten Substanzen gehört auch der Neurotransmitter Acetylcholin (ACh), der unter anderem in der menschlichen Haut eine wichtige Rolle in der Zelldifferenzierung und –proliferation spielt. Die dermatologischen Antiinfektiva Gentianaviolett (GV) und Brillantgrün (BG) aus der Gruppe der Triphenylmethanfarbstoffe werden in der Behandlung lokaler Wundinfektionen verwendet und zeigen im klinischen Gebrauch Nebenwirkungen wie Wundheilungsstörungen. Es konnte gezeigt werden, dass die Farbstoffe die OCTs konzentrationsabhängig hemmen und es wurden die Werte der halbmaximalen Hemmkonzentration ermittelt. Dabei zeigte sich, dass GV und BG zu den Hemmstoffen mit der höchsten Affinität zu den OCTs gehören. Ein Transport der Farbstoffe durch die OCTs konnte nicht nachgewiesen werden, die toxische Wirkung auf Keratinozyten in in-vitro Versuchen mit menschlichen Zellreihen wurde bestätigt. Ein Zusammenhang zwischen den beobachteten Wundheilungsstörungen unter der Therapie mit Triphenylmethanfarbstoffen und der Hemmung des ACh-Transportes durch die OCTs konnte nicht bestätigt werden.
Die Blut-Hirn-Schranke reguliert den Transport von Molekülen aus dem Blut in das Gehirn und aus dem Hirngewebe in das Blut. Die Grundlage dieser für den Erhalt der Homöostase im Gehirn wichtigen Schranke bilden zwischen Endothelzellen der Gehirnkapillaren (BCECs) entwickelte, besonders dichte Zonulae Occludentes (Tight Junctions). Viele Krankheiten, zum Beispiel die Multiple Sklerose, gehen mit einer Dysfunktion der BBB einher, die molekularen Grundlagen verschiedener Störungen und damit die Therapiemöglichkeiten sind bisher jedoch oftmals noch unbekannt. Ein grundlegendes Problem der Forschung an der BBB war bislang das Fehlen eines geeigneten immortalisierten in vitro-Modelles zum Verständnis der Differenzierung und Regulierung der Schrankenfunktion. Es gelang nun erstmals, aus murinen BCECs ein solches in vitro-Modell der BBB zu entwickeln, welches wichtige Charakteristika der BBB in vivo aufweist. Zu den Eigenschaften der BBB in vivo zählen allgemein ein hoher transendothelialer elektrischer Widerstand (TER) von bis zu 2000  x cm², die Expression der TJ-Proteine Occludin, Claudin-1, Claudin-3 und Claudin-5 sowie eine geringe Rate transzellulärer Transportvorgänge. Die Entwicklung einer immortalisierten Zelllinie als in vitro-Modell der BBB beinhaltete das Bereitstellen einer möglichst natürlichen Umgebung für die Endothelzellen. Durch Zugabe von Wachstums- und Differenzierungsfaktoren sowie Serumreduktion im Differenzierungsmedium konnte eine dichte Schrankenfunktion induziert werden, welche sich anhand von TER-Messungen nachweisen ließ. Mittels immuncytochemischen und molekularbiologischen Methoden wurde außerdem die Expression verschiedener TJ-Proteine in den immortalisierten BCECs gezeigt. Die Permeabilität der BBB wird durch eine Reihe von Faktoren beeinflusst. So war zu erkennen, dass Glucocorticoide und Insulin die Barrierenfunktion der BBB induzieren und die Zugabe dieser Faktoren die in vitro-Kultivierung von BCECs ermöglicht, ohne dass diese dabei für die BBB in vivo wesentliche Charakteristika verlieren. Diese Ergebnisse stimmen überein mit anderen Studien, denenzufolge für die Induktion und Aufrechterhaltung komplexer Tight Junctions bei kultivierten Endothelzellen Glucocorticoide förderlich sind. Auch klinisch wird dieser Einfluss von Glucocorticoiden bereits genutzt: so konnten im Falle der Multiplen Sklerose Therapieerfolge durch die Gabe von Corticosteroiden erzielt werden.
