Baclofen (1) is a potent and selective agonist for bicuculline-insensitive GABAB receptors and is used clinically as an antispastic and muscle relaxant agent. In the search for new bioactive chemical entities that bind specifically to GABAB receptors, we report here the synthesis of certain baclofen homologues, namely (R,S)-5-amino-3-arylpentanoic acid hydrochlorides (R,S)-1a–h as well as (R,S)-5-amino-3-methylpentanoic acid [(RS)-1i] to be evaluated as GABABR agonists. Compound 1a is an agonist to GABAB receptors with an EC50 value of 46 μM on tsA201 cells transfected with GABAB1b/GABAB2/Gqz5, being the most active congener among all the synthesized compounds.
Propofol und Methohexital hemmen die Dünndarmperistaltik. Untersuchungen des Wirkmechanismus am Meerschweinchendünndarm in vitro Fragestellung: Die Hemmung der Darmmotilität durch Anästhetika und Pharmaka zur Analgosedierung von Patienten in der Intensivmedizin kann Ursache weiterer Komplikationen sein. Diese Arbeit untersucht, ob die Hypnotica Propofol und Methohexital einen Einfluss auf die intestinale Peristaltik haben. Methodik: Dünndarmsegmente des Meerschweinchens wurden in vitro in einer Vorrichtung perfundiert, die propulsive Peristaltik ermöglicht. Durch Registrierung des intraluminalen Drucks kann die Schwelle (peristaltic pressure threshold, PPT), ab der peristaltische Kontraktionen ausgelöst werden, bestimmt werden. Propofol, Methohexital, sowie mögliche Antagonisten, wurden den Dünndarmsegmenten extraserosal zupipettiert und die Änderungen der PPT registriert. Ergebnisse: Propofol und Methohexital beeinflussten die Dünndarmperistaltik auf unterschiedliche Art und Weise. Methohexital führte konzentrationsabhängig zu einem Anstieg der PPT, z.T. bis zur kompletten Hemmung der Peristaltik. Im Gegensatz dazu führte die Gabe von Propofol in keinem Fall zur kompletten Hemmung, es zeigte sich lediglich ein Anstieg der PPT. Die Hemmung durch Methohexital trat nach Naloxon und z.T. nach Bicucullin vermindert auf. Die Hemmung der Peristaltik durch Propofol war zumeist unbeeinflusst durch die verwendeten Antagonisten. Schlussfolgerung: Propofol und Methoxital hemmen konzentrationsabhängig die Dünndarmperistaltik des Meerschweinchens. Der hemmende Effekt von Methohexital scheint durch endogene Einflüsse und durch Bindung an vermittelt GABAA- Rezeptoren vermittelt zu werden. Die vorliegenden Daten weisen darauf hin, dass Hypnotika wie Propofol und Methohexital auch auf die menschliche Peristaltik einen hemmenden Einfluss haben und bei Intensivpatienten einen Ileus induzieren oder verschlimmern können. GABA- Rezeptoren scheinen hierbei eine untergeordnete Rolle zu spielen.

In der vorliegenden Arbeit wurde der Einfluss von Dopamin, Serotonin und GABA auf das Schlafverhalten von Drosophila melanogaster genauer untersucht. Mit Hilfe von Mutanten in Wiederaufnahmetransportern für Dopamin und Serotonin konnte gezeigt werden, dass Dopamin und Serotonin entgegengesetzte Wirkungen auf die Schlafmenge der Fliegen haben. Dopamin hat eine schlafhemmende, Serotonin eine schlaffördernde Wirkung. Die Nutzung eines neuronal dopamindefizienten Fliegenstammes erweitert diese Erkenntnisse. Die Nutzung von RNAi zur Hinunterregulierung der Rezeptoren für Dopamin brachte keine weiteren Erkenntnisse, da sie zu keinem messbaren Effekt führen. Jedoch ergab eine parallel dazu durchgeführte Hinunterregulierung des GABABR2 Rezeptors, dass dieser maßgeblich für die Aufrechterhaltung des Schlafes in der zweiten Hälfte der Nacht verantwortlich ist. Es konnte gezeigt werden, dass für diese Aufgabe vor allem ihre Expression in den l-LNv Neuronen relevant ist. Dabei ist für die GABABR2 Rezeptoren kein Effekt, für Dopamin und Serotonin nur in geringen Ausmaß ein Effekt auf die Innere Uhr in Form von gering veränderter Periode zu beobachten.
Durch eine Kombination der Transportermutanten für Dopamin und Serotonin mit dem intakten, als auch mutierten WHITE Transporter zeigte sich eine interessante Interaktion dieser drei Transporter bei der Regulation der Gesamtschlafmenge, wobei die white Mutation zu einer Reduzierung der Gesamtschlafmenge führt. UPLC Messungen der Stämme ergaben, dass der Effekt von white vermutlich auf dessen Einfluss auf den beta-Alanyldopamingehalt der Fliegen basiert. beta-Alanyldopamin wird bei dem Transport von Dopamin über die Gliazellen durch das Enzym EBONY gebildet, dessen Mutation in der Kombination mit intaktem WHITE und mutiertem Dopamintransporter zu einer drastischen Reduktion des Schlafes während der Nacht führt. Im Rahmen der Untersuchung konnte zudem gezeigt werden, dass entgegen des bisherigen Wissens aus Zellkulturstudien in Drosophila melanogaster kein beta-Alanylserotonin gebildet wird. Möglicherweise wird nur Dopamin, nicht jedoch Serotonin über die Gliazellen recycelt. Dies ist ein interessanter Unterschied, der sowohl eine zeitliche, als auch lokale Feinregulation der Gegenspieler Dopamin und Serotonin ermöglicht.
Die Untersuchung der Dimerpartner BROWN und SCARLET zeigte, dass lediglich BROWN zu einer Reduktion des Schlafes führt. Ein Effekt, der auch in einer Fliegenlinie mit spontaner white Mutation beobachtet werden konnte. Die genaue Funktion dieses Heterodimertransporters und seine neuronale Lokalisation wurden im Rahmen dieser Arbeit noch nicht geklärt. Dennoch liegt eine Funktion als Dopamin- oder beta-Alanyldopamintransporter in Gliazellen auf Grund der ermittelten Ergebnisse nahe.
Zusätzlich konnte zum ersten Mal in Drosophila melanogaster eine Funktion der Amintransporter bei der Anpassung der Inneren Uhr an extreme kurze bzw. lange Photoperioden gezeigt werden.
Eine anatomische Lokalisierung des WHITE Transporters im Gehirn von Drosophila melanogaster, die weitere Charakterisierung der Rolle des WHITE/BROWN Dimers und die Zuordnung bestimmter dopaminerger und serotonerger Neurone bei der Modulation der Aktivitätsmaxima stellen spannende Fragen für zukünftige Arbeiten dar.
γ-Aminobutyric acid type A receptors (GABAARs) mediate the majority of fast synaptic inhibition in the central nervous system (CNS). GABAARs belong to the Cys-loop superfamily of pentameric ligand-gated ion channels (pLGIC) and are assembled from 19 different subunits. As dysfunctional GABAergic neurotransmission manifests itself in neurodevelopmental disorders including epilepsy and anxiety, GABAARs are key drug targets. The majority of synaptic GABAARs are anchored at the inhibitory postsynaptic membrane by the principal scaffolding protein gephyrin, which acts as the central organizer in maintaining the architecture of the inhibitory postsynaptic density (iPSD). This interaction is mediated by the long intracellular loop located in between transmembrane helices 3 and 4 (M3–M4 loop) of the receptors and a universal receptor-binding pocket residing in the C-terminal domain of gephyrin. In 2014, the crystal structure of the β3-homopentameric GABAAR provided crucial information regarding the architecture of the receptor; however, an understanding of the structure and assembly of heteropentameric receptors at the atomic level was lacking. This review article will highlight recent advances in understanding the structure of heteropentameric synaptic GABAARs and how these structures have provided fundamental insights into the assembly of these multi-subunit receptors as well as their modulation by diverse ligands including the physiological agonist GABA. We will further discuss the role of gephyrin in the anchoring of synaptic GABAARs and glycine receptors (GlyRs), which are crucial for maintaining the architecture of the iPSD. Finally, we will also summarize how anti-malarial artemisinin drugs modulate gephyrin-mediated inhibitory neurotransmission.

The transport of glucose across the cell plasma membrane is vital to most mammalian cells. The glucose transporter (GLUT; also called SLC2A) family of transmembrane solute carriers is responsible for this function in vivo. GLUT proteins encompass 14 different isoforms in humans with different cell type-specific expression patterns and activities. Central to glucose utilization and delivery in the brain is the neuronally expressed GLUT3. Recent research has shown an involvement of GLUT3 genetic variation or altered expression in several different brain disorders, including Huntington’s and Alzheimer’s diseases. Furthermore, GLUT3 was identified as a potential risk gene for multiple psychiatric disorders. To study the role of GLUT3 in brain function and disease a more detailed knowledge of its expression in model organisms is needed. Zebrafish (Danio rerio) has in recent years gained popularity as a model organism for brain research and is now well-established for modeling psychiatric disorders. Here, we have analyzed the sequence of GLUT3 orthologs and identified two paralogous genes in the zebrafish, slc2a3a and slc2a3b. Interestingly, the Glut3b protein sequence contains a unique stretch of amino acids, which may be important for functional regulation. The slc2a3a transcript is detectable in the central nervous system including distinct cellular populations in telencephalon, diencephalon, mesencephalon and rhombencephalon at embryonic and larval stages. Conversely, the slc2a3b transcript shows a rather diffuse expression pattern at different embryonic stages and brain regions. Expression of slc2a3a is maintained in the adult brain and is found in the telencephalon, diencephalon, mesencephalon, cerebellum and medulla oblongata. The slc2a3b transcripts are present in overlapping as well as distinct regions compared to slc2a3a. Double in situ hybridizations were used to demonstrate that slc2a3a is expressed by some GABAergic neurons at embryonic stages. This detailed description of zebrafish slc2a3a and slc2a3b expression at developmental and adult stages paves the way for further investigations of normal GLUT3 function and its role in brain disorders.
Based on genetic association and functional imaging studies, reduced function of tryptophan hydroxylase-2 (TPH2) has been shown to be critically involved in the pathophysiology of anxiety-disorders and depression. In order to elucidate the impact of a complete neuronal 5-HT deficiency, mice with a targeted inactivation of the gene encoding Tph2 were generated. Interestingly, survival of Tph2-/- mice, the formation of serotonergic neurons and the pathfinding of their projections was not impaired. Within this thesis, I investigated the influence of 5-HT deficiency on the γ-amino butyric acid (GABA) system. The GABAergic system is implicated in the pathophysiology of anxiety disorders. Therefore, measurement of GABA concentrations in different limbic brain regions was carried out. These measurements were combined with immunohistochemical estimation of GABAergic cell subpopulations in the dorsal hippocampus and amygdala. In Tph2-/- mice GABA concentrations were increased exclusively in the dorsal hippocampus. In heterozygous Tph2+/- mice concentrations of GABA were increased in the amygdala compared to Tph2-/- and wt control mice, while the reverse was found in the prefrontal cortex. The changes in GABA concentrations were accompanied by altered cell density of GABAergic neurons within the basolateral complex of the amygdala and parvalbumin (PV) neurons of the dorsal hippocampus and by adaptational changes of 5-HT receptors. Thus, adaptive changes during the development on the GABA system may reflect altered anxiety-like and depressive-like behavior in adulthood. Moreover, chronic mild stress (CMS) rescues the depressive-like effects induced by 5-HT deficiency. In contrast, 5-HT is important in mediating an increased innate anxiety-like behavior under CMS conditions. This is in line with a proposed dual role of 5-HT acting through different mechanisms on anxiety and depressive-like behavior, which is influenced by gene-environment interaction effects. Further research is needed to disentangle these complex networks in the future.
