We examined the regulation of NFATc1 in different lymphomas and observed an inversed correlation between the methylation status and expression of NFATc1. Our data demonstrate that aberrant DNA methylation associated with chromatin remodeling within nfatc1 locus is a major mechanism for the repression of NFATc1 expression, suggesting that the DNA methylation-mediated transcriptional silencing of NFATc1 may be a critical event in the tumorogenesis of ALCLs and cHLs. Furthermore, the DNA methylation of human nfatc1 promoter region could be used as a novel biomarker of tumor progression. Our results indicate a close link between the loss of immunoreceptor signaling and NFATc1 expression in human lymphomas. For both ALCLs and cHLs, defects in immunoreceptor signaling have been described which result in a loss of receptor-mediated gene expression programs (Schwering et al., 2003; Bonzheim et al., 2004; Marafioti et al., 2004). In T cells, one indicator gene of these programs appears to be the nfatc1 gene whose expression is controlled by TCR signals (Chuvpilo et al., 2002a). In contrast, in T cells NFATc1 expression is unaffected by TCR signals, and NFATc2 was found to be expressed at normal levels in ALCLs and cHLs (L.K., unpubl. data). Moreover, the activity of NF-kappaB factors which can bind to certain NFAT binding sites and share a distantly-related DNA binding domain with NFATs is strongly elevated in cHL cells (Bargou et al., 1997; Hinz et al., 2001; Hinz et al., 2002) suggesting that NFATs and NF-kappaBs exert very different effects on generation and maintenance of Hodgkin’s lymhomas. However, it should be mentioned that in Burkitt’s and further B cell lymphomas in which NFATc1 proteins are strongly expressed and controlled by receptor signals (Kondo et al., 2003), they could exert a promoting function in tumor development. The genes of p53 family members p63 and p73 are prominent examples for mammalian genes whose products can act both as oncoproteins and tumor suppressor genes (Hibi et al., 2000; Stiewe and Putzer, 2002), and it is likely that more genes exist which encode both tumor suppressors and oncoproteins. It remains to be shown whether the nfatc1 gene is one of them.
Infektionen durch C. albicans auf den Schleimhäuten sind eine häufige Erkrankung bei Patienten mit einer Schwächung der T-Zellimmunität. Blutstrominfektionen mit der Hefe C. albicans (Candidämie) stellen, vor allem bei Patienten auf Intensivstationen, eine nach wie vor bedrohliche Komplikation mit hoher Letalität dar.
Das pH-regulierte Antigen 1 (Pra1) ist ein Protein, das von C. albicans produziert wird, auf der Oberfläche des Pilzes gebunden vorkommt und auch vom Pilz in den Überstand sezerniert wird. Im humanen System bindet das Protein an T-Zellen an das Oberflächenprotein CD46. Es ist des Weiteren bekannt, dass das Pra1 an bestimmte Immunzellen der Maus (Monozyten und Phagozyten) binden kann. Eine Bindung an T-Zellen der Maus ist bisher nicht beschrieben. Eine genaue Charakterisierung der Interaktion von Pra1 mit Immunzellen der Maus ist interessant, da die Maus als biologischer Modellorganismus zur Erforschung der Infektion mit C. albicans dient. In dieser Arbeit konnte gezeigt werden, dass rekombinantes Pra1 (rPra1) auch an Maus-CD4+ T-Zellen binden kann.
Es wurden Einflussfaktoren auf die gefundene Bindung von Pra1 an CD4+ T- Zellen gesucht. Als ein Einflussfaktor wurde Zink identifiziert. Pra1 kann an freies Zink binden und durch Zugabe von ZnCl2 während der Inkubation von Pra1 mit T-Zellen kann das Signal von gebundenem Pra1 an CD4+ T-Zellen erhöht werden. Aspf2, ein Protein aus Aspergillus fumigatus mit großer Homologie zu Pra1, kann nicht an diese Zellen binden.
Im in-vivo-Experiment mit Tieren, die mit C. albicans infiziert wurden, konnte kein wildtypisches sezerniertes Pra1 gebunden an T-Zellen nachgewiesen werden. Zellkulturüberstände von C. albicans zeigten nach Inkubation in vitro mit T-Zellen ein Signal für gebundenes Pra1 an CD4+ T-Zellen.
Die Bindungskinetik von Pra1 an T-Zellen zeigte eine über die Zeit der Inkubation konstante Zunahme des Signals von zellgebundenem rPra1 an CD4+ T-Zellen. In der off-Kinetik fand sich eine Abnahme des Signals über die Zeit bis an die Grenze der Nachweisbarkeit.
Der Bindungspartner von Pra1 auf T-Zellen konnte nicht identifiziert werden. Die strukturell und funktionell verwandten Oberflächenproteine Crry, CD59a und CD55 wurden auf Bindungsfähigkeit an T-Zellen in entsprechenden Knockout- Mäusen getestet, konnten jedoch als Rezeptor für Pra1 ausgeschlossen werden. Durch die Bindung von sezerniertem Pra1 an neutrophile Granulozyten wird die Fähigkeit dieser Zellen zur Phagozytose eingeschränkt. Die Bindung von Pra1 an CD4+ T-Zellen führt zur Kostimulation der T-Zellen, also zur verstärkten Zellaktivierung und Proliferation. Durch die Zugabe von 10 μM Zinkchlorid wird die kostimulatorische Aktivität von Pra1 verstärkt.
Während der Zellaktivierung von Effektor-Memory-CD4+ T-Zellen reduziert rPra1 die Sekretion von IFN-γ. Diese Reduktion von IFN-γ-produzierenden Zellen entsteht nicht durch einen Einfluss von Pra1 während der Zellaktivierung von naiven CD4+ T-Zellen zu Th1-Zellen und auch nicht durch die Auslösung von Apoptose in IFN-γ-produzierenden Th1-Zellen. Die Bindung von Pra1 an CD4+- T-Zellen, die über den T-Zell-Rezeptor aktiviert werden, reduziert in vitro die Sekretion des Zytokins. Zusätzlich werden weitere Zytokine in ihrer sezernierten Menge reduziert wie IL-2 und TNF-α.
Mechanismen der TZR-abhängigen und -unabhängigen Aktivierung primärer T-Zellen der Ratte durch CD 28

Zur Aktivierung ruhender T-Zellen sind zwei Signale erforderlich: Eines wird antigen-abhängig über den T-Zellrezeptor (TZR) gegeben, ein zweites erfolgt über kostimulatorische Rezeptoren. Unter den bekannten kostimulatorischen Molekülen ist CD28 das potenteste. Die mitogene Aktivität einiger monoklonaler Antikörper (mAb) spezifisch für CD28 der Ratte, die alle ruhenden primären Ratten-T-Zellen ohne TZR-Ligation zu Proliferation und IL-2-Produktion anregen können, erlaubte, die Signalwege nach dieser direkten CD28-Stimulation und nach Kostimulation zu analysieren und zu vergleichen. Die erzielten Ergebnisse deuten auf unterschiedliche Signalwege nach Kostimulation mit TZR-Beteiligung und direkter CD28-Stimulation hin und zeigen, daß direkte CD28-Stimulation nicht die Signaltransduktion durch den TZR imitiert. Daher unterstützt CD28 als kostimulatorisches Molekül nicht nur TZR-vermittelte Signaltransduktion, sondern fungiert auch als eigenständiges Signalmolekül, das spezifische mitogene Signale generiert. Diese Eigenschaft von CD28 könnte für die Art der induzierten Immunreaktion von Bedeutung sein, da das Verhältnis der Stärke von TZR- und CD28-Signal die funktionelle Differenzierung von T-Zellen in Th1- oder Th2-Zellen bestimmt.
Die Kostimulation über den CD28 Oberflächenrezeptor spielt eine entscheidende Rolle sowohl in der Proliferation von T-Zellen, in deren Differenzierung zu Effektorzellen, als auch in der Entwicklung und Kontrolle von regulatorischen T-Zellen. Diese Schlüsselrolle macht CD28 zu einer interessanten Zielstruktur, um den Einfluss monoklonaler Antikörper auf die Entwicklung und Homöostase von T-Zellen zu analysieren. Die hier verwendeten Antikörper lassen sich funktionell in zwei verschiedene Kategorien einteilen: zum Einen in konventionelle Antikörper (E18 mAk), welche die physiologische Rezeptor-Liganden Interaktion blockieren, und zum Anderen in superagonistische Antikörper (D665 mAk), die in der Lage sind ruhende T-Zellen ohne eine Ligation des T-Zell Rezeptors voll zu aktivieren. In der vorliegenden Arbeit konnte in verschiedenen in-vitro und in-vivo Experimenten gezeigt werden, dass eine Behandlung mit dem CD28 Superagonisten keinen Einfluss auf die Differenzierung von T-Zellen im Thymus nimmt, und die Zusammensetzung der einzelnen Zellpopulationen in diesem Organ unbeeinflusst bleibt. Bei Verwendung blockierender anti-Maus CD28 mAk zeigte sich in sämtlichen Untersuchungen eine beeinträchtigte Entwicklung von regulatorischen T-Zellen innerhalb des Thymus, während die anderen Zellpopulationen unbeeinflusst blieben. Auch in Analysen peripherer Lymphknoten lag der Anteil regulatorischer T-Zellen nach Behandlung mit E18 mAk unter den Kontrollwerten. Durch eine chronische Blockade von CD28 mittels E18 mAk ließ sich die Gesamtanzahl an Tregs in peripheren Lymphknoten, Milz und Thymus drastisch senken, ohne dass es zu einem vollständigen Verschwinden dieser Zellpopulation kam. Auch die absolute Zahl an CD4 positiven T-Zellen innerhalb der peripheren Lymphknoten, Milz und Thymus der behandelten Tiere lag deutlich unter der Kontrolle. Diese Beobachtungen waren 13 Wochen nach Beendigung der Antikörper-Applikationen vollständig reversibel und zu keinem Zeitpunkt des Experimentes ergaben sich klinische oder serologische Hinweise auf die Entwicklung von Autoimmunität trotz niedriger Treg- Zahlen über mehrere Wochen.
Glukokortikoide induzieren Apoptose in vielen verschiedenen Zelltypen. Wenngleich die Glukokortikoid-induzierte Apoptose eine der zuerst entdeckten Formen des Programmierten Zelltodes war, ist sie dennoch kaum verstanden und daher heute noch Teil vieler Forschungen. Eine Bcl-2 Überexpression resultiert in einer Resistenz gegenüber GC- induzierter Apoptose in einer Reihe von Zellen wie einigen Lymphomzell-linien, aber auch normalen Thymozyten, sowie reifen T- und B-Zellen. Glukokortikoide können die Transkription der BH3-only Gene Bim (BCL-2-interacting mediator of cell death) und Puma (p53-upregulated modulator of apoptosis), einer proapoptotischen Untergruppe der Bcl-2 Familie, induzieren. Dies legt nahe, dass Bim und Puma am Weg der GC-induzierten Apoptose beteiligt sind. Ceramid ist in der Lage über die Aktivierung der proapoptotischen Bcl-2 Mitglieder Bax und Bak die äußere Mitochondrienmembran zu schädigen und damit zur Aktivierung von Caspasen zu führen. Verschiedene apoptotische Stimuli, wie z.B. Glukokortikoide, führen so über eine Erhöhung des endogenen Ceramid Levels zur Apoptose. Ceramid kann auf zwei verschiedenen Wegen gebildet werden: zum einen durch Hydrolyse von Sphingomyelin (katalysiert durch saure (a), oder neutrale (n) Sphingomyelinase (SMase)), zum anderen durch de novo Biosynthese. Studien an Thymozyten zeigen, dass von den beiden Möglichkeiten der Ceramidsynthese, lediglich die Inhibition der aSMase zu einer Resistenz gegenüber GC-induzierter Apoptose führt. Um nun die Rolle von Bim und der aSMase bei der Glukokortikoid-induzierten Apoptose zu untersuchen, wurde die murine T-Zelllymphomlinie WEHI7.15a zu Hilfe genommen. Während die Überexpression retroviral eingebrachter shRNAs gegen Bim und aSMase keine Wirkung auf die Glukokortikoid-induzierte Apoptose zeigten, führte der knockdown des Glukokortikoid-Rezeptors selbst zu einer Resistenz gegenüber Dexamethason. Auch der pharmakologische Inhibitor der Sphingomyelin-Hydrolyse, Imipramin, zeigte sowohl in vitro, als auch in vivo keine Wirkung auf die Glukokortikoid-induzierte Apoptose. Darüber hinaus waren sowohl Thymozyten, als auch periphere T-Zellen von aSMase knockout Mäusen genauso sensitiv auf die Glukokortikoid-induzierte Apoptose wie Wildtyp-Zellen. Die in vitro knockdown Ergebnisse von Bim und vom Glukokortikoid-Rezeptor selbst, konnten weiterhin ex vivo, durch das Einbringen der shRNAs in hämatopoetische Stammzellen, welche zur Rekonstitution bestrahlter Mäuse genutzt wurden, bestätigt werden. Während der Bim knockdown keinerlei Einfluss auf die Glukokortikoid-induzierte Apoptose ex vivo zeigte, konnte die verminderte Expression des Glukokortikoid-Rezeptors den Zelltod verhindern. Im Gegensatz hierzu zeigten sowohl der GR knockdown, als auch der Bim knockdown im hämatopoetischen System einen Einfluss auf die Thymozytenentwicklung in vivo.
Die Multiple Sklerose (MS) und ihr Tiermodell, die Experimentelle Autoimmune Enzephalomyelitis (EAE), sind Autoimmunerkrankungen des Zentralen Nervensystems (ZNS). Neben myelinspezifischen CD4+ T-Zellen tragen auch CD8+ T-Zellen zur Pathogenese dieser Erkrankungen bei. Allerdings ist die Rolle der CD8+ T-Zellen während der Induktionsphase der Erkrankung außerhalb des ZNS noch unklar. In dieser Arbeit wurde daher der Beitrag der CD8+ T-Zellen in der EAE der Lewis-Ratte näher untersucht.
Dazu wurde die Krankheitsaktivität der aktiven EAE in normalen Lewis-Ratten mit Tieren verglichen, in denen die CD8+ T-Zellen durch CD8-spezifische monoklonale Antikörper depletiert wurden. Die CD8-depletierten Tiere zeigten dabei eine verminderte Krankheitsaktivität im Vergleich zu den Kontrolltieren. Ebenso entwickelten CD8 knockout Ratten, die durch die Abwesenheit funktionsfähiger CD8+ T-Zellen gekennzeichnet sind, deutlich reduzierte Krankheitssymptome im Vergleich zu wildtypischen Tieren. Die reduzierte Krankheitsaktivität in den CD8-defizienten Tieren war von einer verminderten Infiltration von T-Zellen und Makrophagen in das ZNS begleitet. Zwar konnten aktivierte gpMBP-spezifische CD4+ T-Zellen in den drainierenden Lymphknoten von CD8-depletierten Ratten detektiert werden, diese produzierten jedoch in deutlich reduziertem Umfang pro-inflammatorische Zytokine wie beispielsweise Interferon-. Offensichtlich können in der aktiven EAE myelinspezifische CD4+ T-Zellen in Abwesenheit von CD8+ T-Zellen nicht vollständig zu Effektorzellen differenzieren und infolgedessen das ZNS nicht infiltrieren. Umgekehrt konnten nach adoptivem Transfer von voll ausdifferenzierten enzephalitogenen CD4+ Effektorzellen sowohl in normalen als auch CD8-defizienten Empfängertieren gleich starke Symptome einer AT-EAE beobachtet werden. Die Entfaltung des pathogenen Potentials voll ausgereifter CD4+ Effektorzellen scheint somit nicht von der Präsenz von CD8+ T-Zellen abzuhängen.
Mit Hilfe eines Ratten-IFN- ELISpots gelang erstmals die Detektion Interferon--produzierender gpMBP-spezifischer CD8+ T-Zellen in Tieren, die zuvor mit gpMBP immunisiert wurden. Zum direkten Nachweis von gpMBP-spezifischen CD8+ T-Zellen wurden RT1.Al-Ig Dimere generiert und mit verschiedenen gpMBP-Peptiden beladen. Tatsächlich konnten in den drainierenden Lymphknotenzellen von Ratten, die zuvor mit gpMBP in CFA immunisiert wurden, CD8+ T-Zellen detektiert werden, die gpMBP125-133-beladene RT1.Al-Ig Dimere erkennen.
Die Ergebnisse dieser Arbeit legen insgesamt den Schluss nahe, dass bei der EAE der Lewis-Ratte Interferon--produzierende CD8+ T-Zellen in der Peripherie mit myelinspezifischen CD4+ T-Zellen interagieren und damit deren Differenzierung zu ZNS-infiltrierenden Effektorzellen ermöglichen.
The immune system is responsible for the preservation of homeostasis whenever a given organism is exposed to distinct kinds of perturbations. Given the complexity of certain organisms like mammals, and the diverse types of challenges that they encounter (e.g. infection or disease), the immune system evolved to harbor a great variety of distinct immune cell populations with specialized functions. For instance, the family of T cells is sub-divided into conventional (Tconv) and unconventional T cells (UTCs). Tconv form part of the adaptive arm of the immune system and are comprised of αβ CD4+ or CD8+ cells that differentiate from naïve to effector and memory populations upon activation and are essential during infection and cancer. Furthermore, UTCs, which include γδ T cells, NKT and MAIT, are involved in innate and adaptive immune responses, due to their dual mode of activation, through cytokines (innate-like) or TCR (adaptive), and function. Despite our understanding of the basic functions of T cells in several contexts, a great number of open questions related to their basic biology remain. For instance, the mechanism behind the differentiation of naïve CD4+ and CD8+ T cells into effector and memory populations is not fully understood. Moreover, the exact function and relevance of distinct UTC subpopulations in a physiological context have not been fully clarified. Here, we investigated the factors mediating naïve CD8+ T cell differentiation into effector and memory cells. By using flow cytometry, mass spectrometry, enzymatic assays, and transgenic mouse models, we found that the membrane bound enzyme sphingomyelin-phosphodiesterase acid-like 3b (Smpdl3b) is crucial for the maintenance of memory CD8+ T cells. Our data show that the absence of Smpdl3b leads to diminished CD8+ T cell memory, and a loss of stem-like memory populations due to an aggravated contraction. Our scRNA-seq data suggest that Smpdl3b could be involved in clathrinmediated endocytosis through modulation of Huntingtin interacting protein 1 (Hip1) levels, likely regulating TCR-independent signaling events. Furthermore, in this study we explored the role of UTCs in lymph node-specific immune responses. By using transgenic mouse models for photolabeling, lymph node transplantation models, infection models and flow cytometry, we demonstrate that S1P regulates the migration of tissue-derived UTC from tissues to draining lymph nodes, resulting in heterogeneous immune responses mounted by lymph nodes draining different tissues. Moreover, our unbiased scRNAseq and single lineage-deficient mouse models analysis revealed that all UTC lineages (γδ T cells, NKT and MAIT) are organized in functional units, based on transcriptional homogeneity, shared microanatomical location and migratory behavior, and numerical and functional redundancy. Taken together, our studies describe additional cell intrinsic (Smpdl3b) and extrinsic (S1Pmediated migration) functions of sphingolipid metabolism modulating T cell biology. We propose the S1P/S1PR1/5 signaling axis as the potential survival pathway for Smpdl3b+ memory CD8+ T cells and UTCs, mainly in lymph nodes. Possibly, Smpdl3b regulates S1P/S1PR signaling by balancing ligandreceptor endocytosis, while UTCs migrate to lymph nodes during homeostasis to be exposed to specific levels of S1P that assure their maintenance. Our results are clinically relevant, since several drugs modulating the S1P/S1PR signaling axis or the levels of Smpdl3b are currently used to treat human diseases, such as multiple sclerosis and B cell-mediated diseases. We hope that our discoveries will inspire future studies focusing on sphingolipid metabolism in immune cell biology.
Modulation CD4+ humaner Treg- und Tconv-Zellen durch Inhibition der sauren Sphingomyelinase in vitro

Die saure Sphingomyelinase (ASM) stellt durch die Umwandlung von Sphingomyelin in Ceramid und Phosphorylcholin ein zentrales, fein reguliertes Enzym im Sphingolipidmetabolismus dar. Dadurch nimmt es Einfluss auf verschiedene zelluläre Mechanismen wie Signalvermittlung, Endo- und Exozytose und Zellaktivierung. Dementsprechend weitreichend ist auch die Bedeutung der ASM bei verschiedenen Krankheiten wie Arteriosklerose, Depression oder Neoplasien. Auch auf das Immunsystem, insbesondere auf die Signalvermittlung durch T-Zellen innerhalb des adaptiven Immunsystems, nimmt die saure Sphingomyelinase Einfluss. Aufbauend auf früheren Forschungsarbeiten zur pharmakologischen und genetischen Hemmung der ASM im Mausmodell untersuchten wir, welche Auswirkungen die Hemmung dieses Enzyms in humanen Zellkulturen auf die Population regulatorischer und konventioneller T-Zellen haben. Hierzu verwendeten wir die beiden selektiven Serotonin-Wiederaufnahmehemmer Sertralin und Citalopram; zwei antidepressiv wirksame Medikamente, die durch eine Verdrängung der ASM von der lysosomalen Membran eine hemmende Wirkung ausüben. Wir konnten zeigen, dass diese beiden Substanzen sowohl in Maus-T-Zellen, als auch in humanen T-Zellen, in der Lage sind, die Aktivität der sauren Sphingomyelinase zu inhibieren. Durch Kultivierung von Immunzellen der Maus zusammen mit den Inhibitoren konnte darüber hinaus eine Erhöhung der Treg-Zellfrequenz erreicht werden. Verschiedene Zellkulturexperimente mit humanen PBMCs zeigten weiterhin, dass unter gewissen Umständen so auch eine Vermehrung regulatorischer T-Zellen im Menschen möglich ist, und dass dies mutmaßlich durch Einbindung der ASM im CD3/CD28-Signalweg bedingt ist. In mit AntiCD3-Antikörper stimulierten experimentellen Ansätzen kam es jedoch nur bei einzelnen Individuen, die als Responder identifiziert werden konnten, zu einer Treg-Zellvermehrung. Umgekehrt kam es durch externe Zugabe von C6-Ceramid zu einer Verringerung des Anteils an regulatorischen T-Zellen. Des Weiteren wurden verschiedene Veränderungen im Expressionsverhalten von Treg- und Tconv-Zellen bezüglich CD25, CD69 und CTLA-4 in Anwesenheit der ASMInhibitoren beobachtet. Weiterhin bestätigte sich, dass die pharmakologische Hemmung der sauren Sphingomyelinase auch Auswirkungen auf die Effektorfunktion von T-Zellen hat. Während die Proliferation der Zellen weitgehend unbeeinträchtigt blieb, kam es zu einer verringerten Sekretion der Zytokine IFN-gamma, TNF, IL-5 und IL-10. In ihrer Gesamtheit sprechen diese Ergebnisse dafür, dass Inhibitoren der sauren Sphingomyelinase begünstigend auf Krankheitsgeschehen mit überschießender oder dysregulierter Aktivität des Immunsystems einwirken könnten. Immunmodulatorischen Wirkungen durch Inhibition der ASM erklären möglicherweise auch Einflüsse auf das Immunsystem, die für verschiedene Antidepressiva beschrieben wurden. Insgesamt ist die Bedeutung der sauren Sphingomyelinase innerhalb der Regulation des adaptiven Immunsystems jedoch noch ein weitgehend ungeklärtes Thema mit vielen offenen Fragen. Daher ist auch in Zukunft weitere klinische und experimentelle Forschung erforderlich, um zu klären, welchen Einfluss dieses Enzyms auf Immunzellen hat und wie sich dieser auch klinisch anwenden lässt.
The most commonly used assay to quantitate the response of peripheral T~cells upon stimulation with growth factors is determination of incorporated (JH]TdR. We compared thls test to three other methods: 1. direct countlog of cells with a Coulter type counter as reference assay, 2. a colorimetric assay using the tetrazolium dye 3-[ 4,S-dimethylthiazol-l-yl]-2,5diphenyl tetrazolium (MTT), which is a cheap and increasingly popular non-radioactive method and 3. incorporation of the thymidine analog 5-bromo-2'-deoxyuridine detection with a monoclonal antibody on cytospins. Primary human PHA-blasts from >30 healthy individuals were stimulated with IL-2, IL-4 aod IL-7 and assayed with up to four different methods. We discuss the advantages and disadvantages of the assays used and tbe effects of differences between cell preparations. We observed no significant variations between individuals for the dose dependence, but the relative emctency of IL4 compared to IL-2 and IL-7 was variable. This was probably due to the slower response observed upon stimulation with this factor.
In der vorliegenden Arbeit werden Studien zu verschiedenen Aspekten der Stimulation von humanen Vg9Vd2-T-Lymphozyten vorgestellt. Ein Schwerpunkt war die Charakterisierung der Erkennung von Bisphosphonaten durch Vg9Vd2-T-Zellen. Weder die Alkylmonophosphonate Ethyl- und Propylphosphonat noch 3-Aminopropylphosphonat bewirkten eine Stimulation von Vg9Vd2 T-Lymphozyten. Anscheinend ist also für die gd-T-Zell-stimulierende Aktivität von Aminobisphosphonaten das Vorhandensein sowohl der Methylenbisphosphonat-Gruppe als auch des Amino-Stickstoffs entscheidend. Es wurden verschiedene Pamidronat-Derivate untersucht, die sich durch Substituenten am Stickstoffatom unterscheiden. Die meisten dieser Verbindungen konnten Vg9Vd2-T-Zellen aktivieren, die Art der Substituenten hatte aber großen Einfluss darauf, welche Konzentration des jeweiligen Bisphosphonats für eine gd-T-Zell-Antwort nötig war. Besonders negativ auf die gd-T-Zell-stimulierende Aktivität wirkte sich aus, wenn das Stickstoffatom Teil einer Säureamidbindung war. Das lässt darauf schließen, dass die Gegenwart einer positiven Ladung (durch Protonierung des Stickstoffatoms) von Bedeutung für die Bioaktivität dieser Verbindungen ist. Beim Vergleich verschiedener Bisphosphonate mit stickstoffenthaltenden Heteroaromaten (Fünfringe mit ein bis drei Stickstoffatomen) zeigte sich, dass sowohl die Position des basischen Stickstoffatoms im Ring als auch Art und Position von Ringsubstituenten Einfluss auf deren gd-T-Zell-stimulierende Aktivität haben. Es ergaben sich Hinweise, dass sich eine gesteigerte Neigung eine positive Ladung in der Seitenkette zu tragen bei diesen Verbindungen genau wie bei Aminobisphosphonaten günstig auf das gd-T-Zell-aktivierende Potential auswirkt. Durch Behandlung mit Zoledronat wurde die monozytäre Zelllinie THP-1 stimulierend für Vg9Vd2-T-Zellen. Auch weitere Zelllinien und Lymphozyten des peripheren Bluts (PBL) konnten nach Vorinkubation mit Zoledronat Vg9Vd2-T-Zellen aktivieren. Dabei genügten bei den PBL deutlich geringere Zoledronat-Konzentrationen um einen Effekt zu erzielen als bei den untersuchten Zelllinien. Für die indirekte Stimulation von Vg9Vd2-T-Zellen durch Zoledronat mittels THP-1 Zellen oder PBL war Zell-Zell-Kontakt zwischen den präsentierenden Zellen und den gd-T-Zellen Voraussetzung. Die Anwesenheit von alkalischer Phosphatase hatte keine Auswirkungen auf die gd-T-Zell-Aktivierung durch Zoledronat. Dies spricht dafür, dass Vg9Vd2-T-Zellen Oberflächenstrukturen auf anderen Zellen erkennen und dass freie Phosphoantigene bei der gd-T-Zell-Stimulierung durch Stickstoff enthaltende Bisphosphonate keine Rolle spielen. Wurde die Vorinkubation von THP-1 Zellen mit Zoledronat in Gegenwart von Saponin, einem Detergenz das die Durchlässigkeit der Zellmembran reversibel erhöht, durchgeführt, reichten deutlich niedrigere Konzentrationen des Bisphosphonats aus, um die THP-1 Zellen stimulierend für gd-T-Lymphozyten zu machen. Das ist ein Hinweis darauf, dass für die Aktivierung von Vg9Vd2-T-Zellen durch Zoledronat intrazelluläre Vorgänge in den „antigenpräsentierenden“ Zellen verantwortlich sein könnten. Beim Vergleich verschiedener N-BPs hinsichtlich ihrer Aktivität gegenüber gd-T-Zellen und ihrer antiresorptiven Potenz ergab sich eine erstaunlich gute Korrelation. Dies könnte darauf hinweisen, dass beide Effekte durch den gleichen Mechanismus – die Hemmung der Farnesylpyrophosphat-Synthase – zustande kommen. Die Gegenwart von Farnesol oder Geranylgeraniol während der Vorinkubation von THP-1 Zellen mit Zoledronat verringerte deren gd-T-Zell-stimulierendes Potential nicht, so dass vielleicht nicht die Verarmung an längerkettigen Isoprenoiden sondern die Anreicherung von Vorläufern zu Veränderungen in den Zellen führt, die diese schließlich erkennbar für Vg9Vd2-T-Zellen machen. Zusätzlich wurden im Rahmen dieser Arbeit einige Versuche zu bakteriellen Phosphoantigenen durchgeführt. Mittels einer selektiven HPLC-MS/MS-Methode gelang uns in einer E. coli-Probe der Nachweis eines Vg9Vd2-T-Zell-stimulierenden Pyrophosphats mit der molekularen Masse von 3-Formyl-1-butyl-pyrophosphat, einem Phosphoantigen das in Mykobakterien gefunden worden war (Belmant und Mitarbeiter, 1999). Eine weitere Strukturaufklärung war aufgrund der äußerst geringen Konzentration des Phosphoantigens nicht möglich. 2-C-Methyl-D-erythritol-2,4-cyclodiphosphat (MEcPP) ist ein Zwischenprodukt des 2-C-Methylerythritol-4-phosphat- (MEP) Wegs der Isoprenoidbiosynthese. Es wird in manchen Bakterien – z. B. Corynebacterium ammoniagenes – bei oxidativem Stress verursacht durch Benzylviologen (BV) akkumuliert. Es konnte gezeigt werden, dass Extrakte aus C. ammoniagenes, die in Gegenwart von BV kultiviert wurden, in einem höherem Maße Vg9Vd2-T-Zellen stimulieren als Proben von Bakterien, die ohne BV-Zusatz wuchsen; MEcPP wirkte selbst nicht als Phosphoantigen. Dies war ein weiterer Hinweis darauf, dass bakterielle Phosphoantigene mit dem MEP-Weg assoziiert sind.
In der vorliegenden Arbeit wurden die in-vivo-Effekte eines „superagonistischen“ mAks mit Spezifität für den kostimulierenden Rezeptor CD28 der Ratte untersucht. Dieser Antikörper unterscheidet sich von konventionellen CD28-spezifischen mAk durch seine Fähigkeit, T Zellen auch ohne Ligation des TZR und damit polyklonal zu aktivieren. Die so ausgelöste Expansion der T Zellen verläuft so schnell und effizient wie eine durch Antigen gesteuerte Proliferation; die überproportionale Vermehrung anti-inflammatorischer regulatorischer T Zellen während der initialen Expansionsphase ist vermutlich für das Ausbleiben toxischer Effekte im Zuge der polyklonalen TZellvermehrung verantwortlich.
Transforming-Growth-Factor-beta1 (TGF-b1) is a multifunctional cytokine that regulates cell growth and differentiation in many types of cells. TGF-b1 is especially known to exert a variety of regulatory functions in the immune system, such as T cell differentiation and T cell function. Signal transduction of TGF-b1 is mediated by phosphorylation of receptorassociated Smad proteins (R-Smads). R-Smads are phosphorylated by the activated type I receptor, which is itself phosphorylated by the high affinity type II receptor upon ligand binding. The phosphorylated R-Smads then associate with Co-Smads. Heterooligomers of R- and Co-Smads translocate into the nucleus where they regulate transcription of target genes in concert with other transcription factors such as CBP/p300 or AP-1. Recent findings suggest that the pleiotropic effects of TGF-b1 are conferred by crosstalks to other signal transduction pathways such as the MAP-kinases or the STAT-pathway. Here we describe the effect of long-term exposure to TGF-b1 on the effector function of differentially stimulated primary murine splenocytes and purified primary murine CD8+ cytotoxic T cells. Long-term exposure to TGF-b1 results in non-responsiveness to TGF-b1- induced Smad2 phosphorylation. This is seen either by no phosphorylation or sustained phosphorylation of Smad2. Furthermore, we observed a strong correlation between sustained Smad2 phosphorylation and resistance to TGF-b1 mediated growth inhibition. In contrast, splenocyte cultures strongly growth inhibited by TGF-b1 showed no Smad2 phosphorylation. Lytic activity of these cultures, however, was found to be suppressed regardless of proliferation properties and Smad2 phosphorylation pattern. We also describe that a functional MEK-1 pathway is a prerequisite for rendering murine splenocytes unresponsive to TGF-b1 mediated growth inhibition, and that inhibition of the MEK-1 cascade alters the Smad2 phosphorylation pattern. In addition, we show that resistance to TGF-b1 mediated growth inhibition correlates with the activation of the JNK pathway. However, the resistant phenotype was found unable to be reverted upon administration of exogeneous IFNg and/or aCD28 antibody. In human or mouse T cell lines, however, the described correlation between the type of stimulation and TGF-b growth resistance or growth sensitivity is not present. Thus, this correlation is specific for primary T cells. We also cloned a chimeric dominantnegative TGF-b receptor which is coupled to a suicide gene, in order to render T cells resistant to TGF-b mediated effects.These findings shed light on how TGF-b1 mediates its immunosuppressive role, and may help to gain knowledge of averting these TGF-b1 effects in the course of tumor therapy.
In this thesis, three species were investigated for the conservation of two non-conventional T cell systems, the CD1d/ iNKT cell system and the BTN3/ Vγ9Vδ2 T cell system. Non-conventional T cells are αβ or γδ T cells that do not fit into the classical mode of antigen recognition and adaptive responses. These T cells recognize antigens different from classical peptide antigens and are not restricted to the polymorphic MHC molecules but rather to non-polymorphic antigen-presenting molecules. The iNKT cell subset is restricted by the lipid antigen-presenting molecule CD1d and carries out immunomodulatory functions by rapid cytokine secretion. The molecular basis of this system, the semi-invariant iNKT TCR chains and CD1d were proven to be expressed and compared to homologs in human and rodents. Cotton rats possess multiple members of the AV14 and BV8 family and only one isoform of CD1d which is comparable to findings in the rat.
Moreover, the reactivity of primary cells to glycolipid antigens could be shown, and an iNKT
cell-like population was detected in primary cells using newly developed cotton rat CD1d oligomers. These were also applied to test the capacity of CD1d to present typical glycolipid
antigens to iNKT TCR transductants. In addition, expression of cotton rat iNKT TCR α and β chains in TCR-negative cell lines was used to show successful pairing and detection of glycolipids in the context of CD1d. In summary, the conservation of a functional CD1d/iNKT cell system in the cotton rat could be shown, and tools were developed to study this cell subset in the course of infectious diseases. The Vγ9Vδ2 T cell subset is the major γδ T cell subset in human peripheral blood and has the unique ability to contribute to immune surveillance by detecting pyrophosphorylated metabolites of isoprenoid synthesis that indicate cell stress, transformation or infection. Up to this date, phosphoantigen-reactive γδ T cells have only been shown in primate species. However, evidence for the existence and functional conservation of the genes implied in the BTN3/Vγ9Vδ2 T cell system was found in several placental mammal species,
and two candidate species were chosen for further investigation. The nine-banded armadillo, a valuable model for leprosy research, was shown to possess homologous genes to TRGV9, TRDV2 and BTN3. In this study, the expression of productive rearrangements of TRDV2 gene segments could be shown in peripheral blood samples, but no evidence was found for the expression of a functional TRGV9 rearrangement or BTN3 molecules. Moreover, determinants of phosphoantigen-reactive Vγ9Vδ2 T cells and functional BTN3 molecules were found to still be prevalent in armadillo genes. This makes the armadillo an interesting model to study the structural determinants that allow phosphoantigen recognition by a functional Vγ9Vδ2 T cell subset although this species is merely a witness for a functional system in a placental mammal ancestor. In contrast, alpacas were shown to express functional Vγ9Vδ2 T cells which conserved many features of the human counterpart. Expression of Vγ9Vδ2 pairings could be shown by single-cell PCR and functional phosphoantigenreactive pairings were observed. This phosphoantigen reactivity was also shown in PBMC cultures with a newly developed antibody specific for alpaca Vδ2Jδ4 chains. Moreover, a more detailed study of the alpaca TCR repertoire showed similarities to “γδ high” species like
camelids and cattle which possess an extended family of TRDV genes. The γ and δ loci of alpaca
TCR genes were drafted based on genomic information and cDNA studies and provide an overview for more detailed studies. Conservation of phosphoantigen recognition by the single BTN3 molecule of alpacas was shown in 293T knock out cell lines, and BTN3 detection on PBMCs was investigated with a newly developed alpaca BTN3-specific antibody. These findings prove the existence of a functional BTN3-dependent phosphoantigen-reactive Vγ9Vδ2 T cell subset and provide a basis for the future study of this cell system in a non-primate species. Moreover, as the first non-primate candidate species with the BTN3/Vγ9Vδ2 T cell system the alpaca is an important outgroup for research in this field. The use of a single BTN3 variant in contrast to three human isoforms that work together renders the alpaca a unique and to this date indispensable model for Vγ9Vδ2 T cells.
In conclusion, this study provides an overview of the applicability of new animal models in the
study of the non-conventional T cell subsets iNKT cells and Vγ9Vδ2 T cells and leads the way for a better understanding of structural and functional relationships.
Innerhalb der Juvenilen Idiopathischen Arthritis (JIA) bilden Patienten mit Antinukleären Antikörpern (ANA) Subgruppen-übergreifend eine klinisch homogene Erkrankungsgruppe. Ob diesen klinischen Gemeinsamkeiten jedoch auch eine einheitliche Pathogenese zugrunde liegt, ist bisher unbekannt. Sogenannte periphere T-Helferzellen (TPH) spielen im Kontext zahlreicher Autoimmunerkrankungen eine entscheidende Rolle bei der Aktivierung autoreaktiver B-Zellen. Ziel dieser Arbeit war daher die phänotypische und funktionelle Analyse von PD-1hiCXCR5-CD4+ TPH-Zellen, sowie deren Verteilung in der Synovialflüssigkeit von Patienten unterschiedlicher Subgruppen der JIA.
Hierzu wurden Phänotyp und Zytokinprofil von PD-1hiCD4+ T-Zellen durchflusszytometrisch analysiert. Der funktionelle Einfluss von PD-1hiCD4+ T-Zellen auf die B-Zell-Differenzierung wurde mittels in vitro Kokulturen FACS-sortierter TPH-Zellen der Synovialflüssigkeit untersucht.
IL-21- und IL-17-produzierende T-Gedächtniszellen der Synovialflüssigkeit zeigten eine negative Korrelation zueinander. Die IL-21-Produktion ging besonders von PD-1hiCXCR5-HLA-DR+CD4+ T-Zellen aus, welche besonders in den Gelenken ANA-positiver JIA-Patienten akkumulierten. Diese Population zeigte phänotypische Ähnlichkeit mit TPH-Zellen und leistete in vitro effiziente B-Zell-Hilfe zu Plasmazelldifferenzierung und Immunglobulinsekretion, induzierte jedoch zudem einen CD21lo/-CD11c+T-bet+ Phänotyp in B-Zellen. Passend hierzu bestand auch ex vivo eine signifikante Korrelation zwischen TPH und CD21lo/-CD11c+T-bet+ doppelt-negativen B-Zellen (BDN).
Es konnte also die Expansion einer spezifischen T-Zellpopulation mit phänotypischen und funktionellen Charakteristika von TPH-Zellen beobachtet und deren funktioneller Zusammenhang mit CD21lo/-CD11c+T-bet+ BDN in der Synovialflüssigkeit von JIA-Patienten aufgezeigt werden. Dies könnte die Autoimmunantwort auf ubiquitäre Autoantigene innerhalb betroffener Gelenke ANA-positiver JIA-Patienten widerspiegeln.
T cell activation is supposed to require two signals via engagement of the TCR and a costimulatory molecule. However, the signaling cascade of costimulatory molecules has remained elusive. Here, I provide evidence that CD44 supports proliferation as well as apoptosis mainly, if not exclusively, by enhancing signal transduction via the TCR/CD3 complex. Blockade of CD44 interferes with mounting of an immune response. This has been demonstrated by the significantly decreased IL-2 production of a T helper line, when stimulated in the presence of a competing CD44 receptor globulin. To evaluate the underlying mechanism, CD44 was cross-linked by an immobilized antibody (IM7). Cross-linking of CD44 induces proliferation of peripheral T cells and apoptosis of thymocytes and a T helper line in the presence of subthreshold levels of anti-CD3. CD44-induced proliferation was accompanied by an upregulation of the activation markers CD25 and CD69 and an increased cytokine production. TCR-mediated apoptosis was accompanied by an upregulation of CD95 ligand and CD95 receptor, which could be greatly enhanced by costimulation via CD44. On the level of signal transduction, coligation of CD44 with CD3 resulted in a strong and sustained increase of early tyrosine phosphorylation events and upregulated downstream signal transduction pathways, such as the ras/ERK and the JNK signaling cascades. These pleiotropic effects of CD44 are due to its involvement in the most proximal events in TCR signaling, as demonstrated by a strong increase in the phosphorylation of the TCR z-chain and ZAP-70. Notably, cross-linking of CD44 was binding-site dependent and was only effective when supporting colocalization of the TCR/CD3 complex and CD44. Cross-linking of CD44 via immobilized IM7 also induced profound changes in cell morphology, characterized by strong adhesion, spreading and development of surface extensions, which were dependent on a functional tubulin and actin cytoskeleton. These cytoskeletal rearrangements were mediated by rac1, a small GTPase of the rho subfamily, and src-family kinases, two of which, fyn and lck, were found to be associated with CD44. By cross-linkage of CD44 these kinases were redistributed into so called lipid rafts. It is supposed that for T cell activation a relocation of the TCR/CD3 complex into the same membrane microdomains is required. The data are interpreted in the sense that the costimulatory function of CD44 relies on its cooperativity with the TCR. Most likely by recruitment of phosphokinases CD44 significantly lowers the threshold for the initiation of signaling via the TCR. The requirement for immobilized anti-CD44, the necessity for neighbouring anti-CD3 and the dependence on the binding site of CD44 strongly suggest that the costimulatory mechanism involves cytoskeletal rearrangements, which facilitate recruitment and redirection of src-family protein kinases in glycolipid enriched membrane microdomains.
Ceramide sind biologisch aktive Sphingolipide, die verschiedene zelluläre Signalwege regulieren, meist im Zusammenhang mit der Induktion von Apoptose oder der Regulation des Zellzyklus. Darüber hinaus wurde in der Literatur beschrieben, dass Ceramide die Zytoskelettdynamik unterschiedlicher Zelltypen beeinflussen, die Bedeutung von Ceramiden für die Funktion von T-Zellen wurde allerdings bisher wenig untersucht. In der vorliegenden Arbeit konnte gezeigt werden, dass die exogene Akkumulation von Ceramiden ebenso wie die Generierung von Ceramiden durch bSMase die Adhärenz von T-Zellen an FN bzw. ICAM-1 beeinträchtigt. Des Weiteren konnte eine verminderte T-Zell-Polarisierung auf FN sowie eine reduzierte Chemotaxis und Motilität ceramidmodifizierter T-Zellen in Antwort auf SDF-1 nachgewiesen werden. In Übereinstimmung mit der Unfähigkeit ceramidmodifizierter Zellen morphologisch zu polarisieren wird ferner die Relokalisation von Oberflächenmolekülen und intrazellulärer Proteine durch die Akkumulation von Ceramiden gestört. Überdies konnte in dieser Arbeit gezeigt werden, dass Ceramide mit dem Aktivierungsstatus von Akt und ERM-Proteinen interferieren, da eine verminderte stimulationsabhängige Phosphorylierung von Akt und ERM-Proteinen in ceramidmodifizierten Zellen nachgewiesen wurde. Ein wesentlicher Schritt im Verlauf der T-Zell-Aktivierung ist die Ausbildung einer immunologischen Synapse mit dendritischen Zellen. In dieser Arbeit konnte gezeigt werden, dass, obwohl ceramidreiche Membrandomänen von der Kontaktstelle ausgeschlossen werden, Konjugatfrequenz und Architektur der IS durch die Induktion von Ceramiden nicht beeinflusst werden, da eine normale Verteilung von CD3 und des MTOC beobachtet wurde. Allerdings wird die Funktionalität der Konjugate durch die Induktion von Ceramiden beeinträchtigt. Ceramidmodifizierte Zellen waren nur eingeschränkt in der Lage Orai1 und Stim1 zur Kontaktfläche mit DCs zu translozieren. In Übereinstimmung mit diesen Befunden wurde auch ein verminderter Calcium-Einstrom sowie eine verminderte Proliferation infolge der Akkumulation von Ceramiden detektiert. Zusammenfassend konnte in dieser Arbeit gezeigt werden, dass Ceramide wesentliche Prozesse im Verlauf der T-Zell-Aktivierung beeinflussen, so dass die pathogeninduzierte Generierung von Ceramiden einen möglichen Mechanismus darstellt, die Funktion von T-Zellen zu beeinträchtigen.
Leishmaniasis is a neglected tropical disease that can be manifested through different clinical forms, ranging from cutaneous to visceral. The host response against Leishmania spp. is greatly dependent on T cell-mediated immunity, in which T helper 1 responses are associated with macrophage activation and elimination of the parasite, while regulatory T cells and T helper 2 responses are correlated with parasite survival and persistence of infection. Leishmania uses different virulence factors as strategies for evading the immune response of the host. One of them are cathepsin-like cysteine proteases, which are currently under extensive investigation as targets for drug development. Previous studies with inhibitors of cathepsins B and L in vivo revealed an outstanding modulation of the host T helper cell response. However, the mechanisms behind these observations were not further investigated. Given the urgent need for better treatments against leishmaniasis, the aim of this study was to investigate the effects that the lack of cathepsin B and L activity have on the signals that dendritic cells use to instruct T helper cell polarization in response to infection with Leishmania major.
The cathepsin inhibitors tested showed low or no cytotoxicity in bone marrow-derived dendritic cells, and dendritic cells and macrophages could be generated from cathepsin B and cathepsin L-deficient mice without apparent alterations in their phenotype in comparison to wild-type controls. Furthermore, lack of cathepsin B and L activity showed no impact in the rate of promastigote processing by dendritic cells. Cathepsin B and cathepsin L-deficient macrophages showed no differences in parasite proliferation and capacity to produce nitric oxide in comparison to wild-type macrophages. In response to the parasite, dendritic cells treated with a cathepsin B inhibitor and dendritic cells from cathepsin B-deficient mice showed higher levels of expression of major histocompatibility complex (MHC) class II molecules than dimethyl sulfoxide (DMSO) or wild-type controls, but it was not accompanied by changes in the expression of costimulatory molecules. Wild-type dendritic cells and macrophages are not able to express the pro-inflammatory cytokine interleukin (IL)-12 in response to promastigotes. However, cells treated with a cathepsin B inhibitor or cells deficient for cathepsin B were able to express IL-12, whilethe expression of other cytokines -including IL-6 and tumor necrosis factor (TNF)-alpha-remained unchanged. These characteristics point towards a more “pro-Th1” profile of dendritic cells in the absence of cathepsin B.
This data is the first report on IL-12 regulation depending on cathepsin B. The IL-12 up-regulation observed was already present at the transcriptional level. Furthermore, it was also present in macrophages and dendritic cells in response to LPS, and the latter had a higher capacity to induce T cell helper 1 polarization in vitro than wild-type dendritic cells. The activation of different signaling pathways was analyzed, but the up-regulation of IL-12 could not be attributed to modulation of nuclear factor-kappaB (NFkappaB), p38 mitogen activated protein kinase (MAPK) and extra-cellular signal-regulated kinase (ERK)1/2 pathways. Thus, the mechanism behind IL-12 regulation by cathepsin B remains to be elucidated, and the impact of these effects is yet to be confirmed in vivo. Altogether it is tempting to speculate that cathepsin B, in addition to its role in processing endocytosed material, is involved in the modulation of the pro-inflammatory cytokine IL-12.
The cancer stem cell hypothesis is a cancer development model which elicited great interest in the last decades stating that cancer heterogeneity arises from a stem cell through asymmetrical division. The Cancer Stem Cell subset is described as the only population to be tumorigenic and having the potential to renew. Conventional therapy often fails to eradicate CSC resulting in tumor relapse. Consequently, it is of great inter-est to eliminate this subset of cells to provide the best patient outcome. In the last years several approaches to target CSC were developed, one of them being immunotherapeu-tic targeting with antibodies. Since markers associated with CSC are also expressed on normal stem cells or healthy adjacent tissue in colorectal cancer, dual targeting strate-gies are preferred over targeting only a single antigen. Subsequently, the idea of dual targeting two CSC markers in parallel by a newly developed split T cell-engaging anti-body format termed as Hemibodies emerged. In a preliminary single cell RNA sequenc-ing analysis of colorectal cancer cells CD133, CD24, CD166 and CEA were identified as suitable targets for the combinatorial targeting strategy. Therefore, this study focused on trispecific and trivalent Hemibodies comprising a split binding moiety against CD3 and a binding moiety against either CD133, CD24, CD166 or CEA to overcome the occurrence of resistance and to efficiently eradicate all tumor cells including the CSC compartment. The study showed that the Hemibody combinations CD133xCD24, CD133xCD166 and CD133xCEA are able to eliminate double positive CHO cells with high efficacy while having a high specificity indicated by no killing of single antigen positive cells. A thera-peutic window ranging between one to two log levels could be achieved for all combina-tions mentioned above. The combinations CD133xCD24 and CD133xCD166 further-more proved its efficacy and specificity on established colorectal cancer cell lines. Be-sides the evaluation of specificity and efficacy the already introduced 1st generation of Hemibodies could be improved into a 2nd generation Hemibody format with increased half-life, stability and production yield. In future experiments the applicability of above-mentioned Hemibodies will be proven on patient-derived micro tumors to also include variables like tumor microenvironment and infiltration.
The neuronal ceroid lipofuscinoses (NCLs) are fatal neurodegenerative disorders in which the visual system is affected in early stages of disease. A typical accompanying feature is neuroinflammation, the pathogenic impact of which is presently unknown. In this study, the role of inflammatory cells in the pathogenesis was investigated in Palmitoyl-protein thioesterase 1-deficient (Ppt1-/-) and Ceroidlipofuscinosis, neuronal 3-deficient (Cln3-/-) mice, models of the infantile and juvenile forms of NCL, respectively. Focusing predominantly on the visual system, an infiltration of CD8+ cytotoxic Tlymphocytes and an activation of microglia/macrophage-like cells was observed early in disease. To analyze the pathogenic impact of lymphocytes, Ppt1-/- mice were crossbred with mice lacking lymphocytes (Rag1-/-) and axonal transport, perturbation and neuronal survival were scored. Lack of lymphocytes led to a significant amelioration of neuronal disease and reconstitution experiments revealed a crucial role of CD8+ cytotoxic T-lymphocytes. Lack of lymphocytes also caused an improved clinical phenotype and extended longevity. To investigate the impact of microglia/macrophage-like cells, Ppt1-/- and Cln3-/- mice were crossbred with mice lacking sialoadhesin (Sn-/-), a monocyte lineage-restricted cell adhesion molecule important for interactions between macrophage-like cells and lymphocytes. Similar to the lack of lymphocytes, absence of sialoadhesin significantly ameliorated the disease in Ppt1-/- and Cln3-/- mice. Taken together, both T-lymphocytes and microglia/macrophage-like cells were identified as pathogenic mediators in two distinct forms of fatal inherited neurodegenerative storage disorders. These studies expand the concept of secondary inflammation as a common pathomechanistic feature in some neurological diseases and provide novel insights that may be crucial for developing treatment strategies for different forms of NCL.
In Ratten und Mäusen aktiviert der superagonistische anti-CD28 monoklonale Antikörper (CD28SA) vorzugsweise regulatorische T-Zellen. In niedriger Dosierung führt CD28SA zu einer fast ausschließlichen Aktivierung von regulatorischen T-Zellen (Tregs). Diese Beobachtung konnte inzwischen auch für menschliche Zellen in Zellkultur bestätigt werden.
In gesunden und freiwilligen Testpersonen deutet die Zytokin-Antwort nach Applikationen von niedrigen CD28SA-Dosen darauf hin, dass sich diese Beobachtung auch in-vivo bewahrheitet. Eine Gabe von CD28SA in niedriger Dosierung, die zu einer exklusiven Aktivierung von regulatorischen T-Zellen führt, könnte somit in der Behandlung von Autoimmunkrankheiten oder von entzündlichen Erkrankungen eingesetzt werden.
Eine mechanistische Erklärung für dieses Phänomen blieb lange Zeit unklar. Die CD28SA-vermittelte T-Zell-Aktivierung ist abhängig von der Verstärkung von basalen tonischen Signalen, die T-Zellen über ihren T-Zell-Rezeptor erhalten. Diese Tatsache führte zu der Hypothese, dass die schwachen, tonischen Signale, die konventionelle CD4+ T-Zellen in Abwesenheit ihrer spezifischen Antigene über den T-Zell-Rezeptor erhalten, ein stärkeres CD28 Signal für ihre Aktivierung benötigen als die selbstreaktiven regulatorischen T-Zellen, die ein stärkeres Selbstpeptid-TCR Signal erhalten.
In dieser Arbeit konnte gezeigt werden, dass die Blockade von MHC-Klasse-II-Molekülen in Mäusen, in-vitro und in-vivo, den Vorteil der regulatorischen T-Zellen gegenüber den konventionellen T-Zellen bezüglich der Antwort auf niedrige CD28SA Dosierungen, aufhebt.
