This thesis is divided into three parts with the main goal allocating novel antimicrobial compounds that could be used as future antibiotics. The first part aimed to evaluate the potential of plant suspension cultures for the production of antimicrobial proteins. The extracellular, intracellular and cell wall bound fractions of seven heterotrophic and photomixotrophic plant cell suspension cultures treated with nine different elicitors were tested for the elicitor dependent production of antimicrobial proteins. Bioactivities were tested against a selected panel of human isolates including Gram-positive and Gram-negative bacteria as well as fungi using the disc diffusion assay. The intracellular fractions of elicited cell cultures were more active than extracellular fractions while the cell wall bound fractions showed lowest activities. Among the 21 fractions tested, the intracellular fraction of Lavendula angustifolia elicited with DC3000 was most active against Candida maltosa. The second most active fraction was the intracellular fraction of Arabidopsis thaliana elicited with salicylic acid which was moreover active against all test strains. The antimicrobial activity of elicited Arabidopsis thaliana cell cultures was tested by bioautography to locate the antimicrobial proteins in the crude extract. The intracellular fraction of photomixotrophic Arabidopsis thaliana cells elicited with salicylic acid was selected for further gel filtration chromatography on S-200 column leading to the purification of one 19 kDa antimicrobially active protein, designated, AtAMP. Our findings suggest that elicited plant cell cultures may present a new promising alternative source of antimicrobial proteins. The second part comprises the isolation of actinomycetes associated with marine sponges and testing the bioactivities of new species for further investigations. Actinobacterial communities of eleven taxonomically different sponges that had been collected from offshore Ras Mohamed (Egypt) and from Rovinj (Croatia) were investigated by a culture-based approach using different standard media for isolation of actinomycetes and media enriched with aqueous sponge extract to target rare and new actinomycete species. Phylogenetic characterization of 52 representative isolates out of 90 based on almost complete sequences of genes encoding 16S rRNA supported their assignment to 18 different actinomycete genera. Altogether 14 putatively new species were identified based on sequence similarity values below 98.2% to other strains in the NCBI database. The use of M1 agar amended with aqueous sponge extract yielded a putative new genus related to Rubrobacter which highlighting the need for innovative cultivation protocols. Biological activity testing showed that five isolates were active against Gram-positives only, one isolate was active against Candida albicans only and one isolate showed activity against both groups of pathogens. Moreover, the antiparasistic activity was documented for four isolates. These results showed a high diversity of actinomycetes associated with marine sponges as well as highlighted their potential to produce anti-infective agents. The third part of the thesis focused on the isolation and structure elucidation of new bioactive compounds. Streptomyces strain RV15 recovered from sponge Dysidea tupha, was selected for further chemical analysis by virtue of the fact that it exhibited the greatest antimicrobial potential against Staphylococcus aureus as well as Candida albicans among the all tested strains. Moreover, members of the genus Streptomyces are well known as prolific producers of interesting pharmacologically active metabolites. Chemical analysis of the methanolic crude extract using different chromatographic tools yielded four new compounds. The structures of the new compounds were spectroscopically elucidated to be four new cyclic peptides, namely, cyclodysidins A-D. Their bioactivity was tested against different proteases, bacteria and Candida as well as tumor cell lines. The compounds did not show any significant activities at this point.
Terrestrial actinomycetes are noteworthy producers of a multitude of antibiotics, however the marine representatives are much less studied in this regard. In this study, 90 actinomycetes were isolated from 11 different species of marine sponges that had been collected from offshore Ras Mohamed (Egypt) and from Rovinj (Croatia). Phylogenetic characterization of the isolates based on 16S rRNA gene sequencing supported their assignment to 18 different actinomycete genera representing seven different suborders. Fourteen putatively novel species were identified based on sequence similarity values below 98.2% to other strains in the NCBI database. A putative new genus related to Rubrobacter was isolated on M1 agar that had been amended with sponge extract, thus highlighting the need for innovative cultivation protocols. Testing for anti-infective activities was performed against clinically relevant, Gram-positive (Enterococcus faecalis, Staphylococcus aureus) and Gram-negative (Escherichia coli, Pseudomonas aeruginosa) bacteria, fungi (Candida albicans) and human parasites (Leishmania major, Trypanosoma brucei). Bioactivities against these pathogens were documented for 10 actinomycete isolates. These results show a high diversity of actinomycetes associated with marine sponges as well as highlight their potential to produce anti-infective agents.
Die Internationalisierung im Einzelhandel ist eine vergleichsweise junge Entwicklung, die in den vergangenen beiden Dekaden stark vorangeschritten ist. Dabei haben viele Unternehmen die Erfahrung machen müssen, die Grundlage ihres im Heimatland erlangten Wettbewerbsvorteils zwar in einige Märkte erfolgreich transferieren zu können, in andere
Märkte jedoch nicht. Worin liegen die Ursachen hierfür? Trotz einer zunehmenden Zahl von Studien zur Internationalisierung im Einzelhandel gibt es immer noch ein mangelndes Verständnis dafür, unter welchen Bedingungen Unternehmen im Ausland erfolgreich sind, respektive wann sie scheitern. Einen theoretischen Rahmen zur Analyse der relevanten Erfolgsdeterminanten liefert die Institutionentheorie, die das Verhältnis zwischen Unternehmen und Gesellschaft betrachtet und gerade auch für interkulturelle Fragestellungen einen interessanten Untersuchungsansatz darstellt.
Vor diesem Hintergrund wird in der vorliegenden Arbeit die US-amerikanische
Expansion des Discounters Aldi analysiert. Aldi bietet sich für eine solche Fallstudie an, da der Discounter sein an den deutschen Markt angepasstes Erfolgskonzept Ende der 1970er Jahre weitestgehend unverändert in die USA transferiert hat - in eine institutionelle Umwelt, die sich deutlich von der deutschen unterscheidet. Wie erfolgreich kann Aldi in dieser Umwelt agieren?
Identifizierung und Strukturaufklärung von Anthocyanen und ihrer Metabolite erfolgten mit Hilfe der mittels Hochleistungsflüssigchromatographie-Diodenarray-Detektion-Elektro-spray-Tan¬dem¬massen¬spektrometrie (HPLC-DAD-ESI-MS/MS). Quantitative Analysen wurden via HPLC-DAD durchgeführt. Die hierzu erforderlichen Referenzverbindungen wurden mittels präparativer HPLC aus Heidelbeeren isoliert (Reinheit zwischen 85,8% und 99,4%). Der Gehalt an Anthocyanen in den untersuchten Heidelbeerfrüchten lag bei 6 g/kg. Bezüglich der mengen¬mäßigen Verteilung dominierten Delphinidin- und Cyanidin¬glykoside vor den Glykosiden von Malvidin, Petunidin und Peonidin. Als konjugierte Zucker¬reste kamen vor allem Glukose und Galaktose vor, der Gehalt an Arabinosiden war weit geringer. Bei oraler Aufnahme erfolgt ein erster Kontakt der Anthocyane mit Speichel. Daher wurde dessen Wirkung auf die Heidelbeeranthocyane in ex vivo-Studien über einen (unphysio-logisch langen) Zeitraum von bis zu 30 Minuten untersucht. Dabei konnte wurde ins-besondere der Einfluß des pH-Wertes auf die Stabilität der Anthocyane aufgezeigt werden. Zur Simulation des Verhaltens von Anthocyanen im Magen wurden die einzelnen Heidelbeeranthocyane mit künstlichem Magensaft (pH 1,81) über vier Stunden inkubiert. Hier erwiesen sich alle untersuchten Verbindungen als stabil. Die anschließend von uns mit simuliertem Duodenalsekret (pH 7,2) über einen Zeitraum von 24 Stunden durchgeführten Studien zeigten, dass die Anthocyane unterschiedlich starken Modifizierungen unterlagen. Unter den schwach alkalischen Bedingungen wurden vor allem die Glykoside des Delphinidins schnell abgebaut, aber auch die übrigen Anthocyane erwiesen sich unter diesen Bedingungen als nicht stabil; nach 24 h war kein Anthocyan mehr nachweisbar. Um die Metabolisierungsvorgänge der Anthocyane im Dünn- und Dickdarm zu untersuchen, wurden ex vivo-Inkubationen jeweils mit frischem Ileo- bzw. Kolo¬stoma-beutel¬inhalt durchgeführt. Während die Abbaugeschwindigkeit in der ilealen Flüssigkeit vor allem von der pH-Stabilität des Aglykons abhänig war, konnten im Dickdarm einzig die Arabinoside nach einer Stunde noch alle in geringen Konzentrationen identifiziert werden. Die meisten Glukoside und Galaktoside waren zu diesem Zeitpunkt schon vollständig abgebaut. Da im Darm von einer hydrolytischen Spaltung der Anthocyane in Anthocyanidin und Zucker ausgegangen wird, wurde die Metabolisierung von Anthocyanidinen unter physio-logischen pH-Bedingungen untersucht. Neben der jeweiligen Spaltung in das Benzoe¬säure-derivat des B-Ringes sowie Phloroglucinessigsäure traten verschiedene Poly¬merisierungs¬-produkte auf, deren Strukturen nicht aufgeklärt werden konnten. In einer weiteren Versuchsreihe wurde die renale Ausscheidung von Anthocyanen bei Ileostomieprobanden nach oraler Applikation von 300 g Heidelbeeren über einen Zeitraum von acht Stunden untersucht. Es zeigte sich, dass ein Stoma des terminalen Ileums keinen Einfluss auf die Absorption und Metabolisierung der Anthocyane hatte. Die Bilanzierung der Anthocyane im Urin erfolgte als Äquvalente von Malvidin-3-O-glukosid, da nicht alle Anthocyanmetabolite zur Verfügung standen. Der Zeitpunkt der maximalen renalen Anthocyanausscheidung sowie die Menge der ausgeschiedenen Anthocyane waren starken interindividuellen Schwankungen unterworfen. Das Aus¬sscheidungs¬maximum (tmax) lag zwischen 0,5 und zwei Stunden. Bei der ausge¬schiedenen Menge wurden Werte zwischen 0,007% und 0.019% der auf¬ge¬nommenen Anthocyane ermittelt. Aufgrund der literaturbekannten Unterschiede zwischen den in Serum und Urin gefunden Anthocyanmengen ist davon auszugehen, dass es nach Anthocyanverzehr zu Inter-aktionen mit Proteinen in Blut oder Geweben kommt. Mittels Blutfraktionierung wurde das humane Serumalbumin (HSA) als wichtigster Bindungspartner der Anthocyane im Blut identifiziert. Anhand spektroskopischer Methoden war es möglich, die Bindungs¬parameter zu berechnen. Als Bindungsort wurde der hydrophile Eingang der lipophilen Warfarin-Bindungstasche in der Subdomäne IIA des HSA-Moleküls mittels "molecular modelling" identifiziert. Nasschemische Untersuchungen ergaben, dass die Bindung der Anthocyane an HSA diese vor ihrem pH-abhängigen Abbau schützt. Eine signifikante Herab¬setzung der chemischen Abbaugeschwindig¬keit konnte auch für bovines Serumalbumin beobachtet werden. Diese Erkenntnis ließ sich auf andere, mit dem HSA-Molekül nicht strukurverwandte lebensmittelrelevante Albumine übertragen. So zeigten Anthocyane große Stabilität in Milch und Eiklar, wobei die Stabilisierung auf eine Wechselwirkung mit den Proteinen Laktalbumin und Ovalbumin zurückgeführt werden konnte. Die in dieser Arbeit erlangten Erkenntnisse hinsichtlich Absorption, Metabolisierung und systemischer Verfügbarkeit im menschlichen Organismus leisten einen Beitrag zum besseren Verständnis der Wirkungen von Anthocyanen. Die neuen Erkenntnisse der Protein¬bindung sind vor allem für die Bewertung der Verfügbarkeit der Anthocyane in humanem Gewebe relevant.
Malaria still persists as one of the deadliest infectious disease in addition to AIDS and tuberculosis. lt is a leading cause of high mortality and morbidity rates in the developing world despite of groundbreaking research on global eradication of the disease initiated by WHO, about half a century ago. Lack of a commercially available vaccine and rapid spread of drug resistance have hampered the attempts of extinguishing malaria, which still leads to an annual death toll of about one million people. Resistance to anti-malarial compounds thus renders search for new target proteins imperative. The kinome of the human malaria parasite Plasmodium falciparum comprises representatives of most eukaryotic protein kinase groups, including kinases which regulate proliferation and differentiation processes. Several reports till date have suggested involvement of parasite kinases in the human host and as well as in the mosquito vector. Kinases essential for life cycle stages of the parasite represent promising targets for anti-malarial compounds thus, provoking characterization of additional malarial kinases. Despite extensive research on most plasmodial enzymes, very little information is available regarding the four identified members of the cyclin dependent kinase like kinase (CLK) family. Thus, the present thesis dealt with the functional characterization of four members of the PfCLK kinase family of the parasite denoted as PfCLK-1/Lammer, PfCLK-2, PfCLK-3 and PfCLK-4 with a special focus on the first two kinases. Additionally, one Ca2+/Calmodulin dependent putative kinase-related protein, PfPKRP, presumed to be involved in sexual stage development of the parasite, was investigated for its expression in the life cycle of the parasite. In other eukaryotes, CLK kinases regulate mRNA splicing through phosphorylation of Serine/Arginine-rich proteins. Transcription analysis revealed abundance of PfCLK kinase genes throughout the asexual blood stages and in gametocytes. By reverse genetics approach it was demonstrated that all four kinases are essential for completion of the asexual replication cycle of P. falciparum. PfCLK 1/Lammer possesses two nuclear localization signals and PfCLK-2 possesses one of these signals upstream of the C-terminal catalytic domains. Protein level expression and sub-cellular localization of the two kinases was determined by generation of antiserum directed against the kinase domains of the respective kinase. Indirect immunofluorescence, Western blot and electron microscopy data confirm that the kinases are primarily localized in the parasite nucleus, and in vitro assays show that both enzymes are associated with phosphorylation activity. Finally, mass spectrometric analysis of co immunoprecipitated proteins shows interactions of the two PfCLK kinases with proteins, which have putative nuclease, phosphatase or helicase functions. PfPKRP on the other hand is predominantly expressed during gametocyte differentiation as identified from transcriptional analysis. Antiserum directed against the catalytic domain of PfPKRP detected the protein expression profile in both asexual and gametocyte parasite lysates. Via immunofluorescence assay, the kinase was localized in the parasite cytoplasm in a punctuated manner, mostly in the gametocyte stages. Reverse genetics resulted in the generation of PfPKRP gene-disruptant parasites, thus demonstrating that unlike CLK kinases, PfPKRP is dispensable for asexual parasite survival and hence might have crucial role in sexual development of the parasite. On one hand, characterization of PfCLK kinases exemplified the kinases involved in parasite replication cycle. Successful gene-disruption and protein expression of PfPKRP kinase on the other hand, demonstrated a role of the kinase in sexual stage development of the parasite. Both kinase families therefore, represent potential candidates for anti-plasmodial compounds.
Der Atypische teratoid/rhabdoide Tumor (ATRT) und der primitive neuroektodermale Tumor (PNET) sind hochmaligne Tumorentitäten (WHO-Grad IV) des zentralen Nervensystems, die überwiegend im Kleinkindalter auftreten. Beide zeigen eine sehr heterogene morphologische Struktur und sind bisher nur mittels Histopathologie und Immunhistochemie voneinander zu differenzieren. Bisherige Untersuchungen ließen noch keine neuroradiologische Unterscheidbarkeit zwischen beiden Tumorentitäten erkennen. Die vorliegende Arbeit befasst sich anhand eines diesbezüglich einmalig großen Patientenkollektives (23 ATRT, 36 PNET) mit den spezifisichen morphologischen Kriterien des supratentoriellen (st) ATRT und PNET in der Magnetresonanztomographie (MRT). Die Patienten rekrutierten sich aus der multizentrischen Hirntumorstudie HIT 2000 (Teil des Kompetenznetzes der Hirntumorstudien der „Gesellschaft für pädiatrische Onkologie und Hämatologie“). Retrospektiv wurden MRT-Bilder aus einem Zeitraum von 5 Jahren ausgewertet. Untersucht wurden T1- und T2-Wichtung, nativ und unter Kontrastmittelapplikation. Zur Abgrenzung beider Entitäten voneinander wurden verschiedene Kriterien herausgearbeitet. Dazu zählten zunächst die Darstellung in der nativen T1-Wichtung, die Schärfe der Tumorbegrenzung, das zeitgleiche Vorliegen von Zysten, Ödemen und Blutungen sowie die Ausprägung des Kontrastmittel-Enhancements. Als zentrales Ergebnis der Arbeit konnte ein markantes strukturelles Muster des Kontrastmittel-Enhancements herausgearbeitet werden, welches sich als charakteristisch für den stATRT erwies, während es nur bei einem sehr geringen Prozentsatz der stPNETs anzutreffen war. Hierbei handelt es sich um ein girlandenförmiges Band, welches den Tumor randständig um eine zentrale Nekrose herum auskleidet. Dieses als „ATRT-typisch“ bezeichnete Muster wiesen zehn der stATRTs (43,5%) und drei der stPNETs (8,3%) auf. Darüber hinaus konnte man bei fünf stATRTs (21,7%) Areale mit wie in der Girlande anzutreffenden vesikulären Strukturen aber ohne begleitende zentrale Tumornekrose beobachten. Nur ein stPNET (2,8%) wies ebenfalls vesikuläre Anteile ohne zentrale Nekrose auf. Es konnten somit charakteristische Muster identifiziert werden, welche auffällig häufig in Kontrastmittel-verstärkten T1-gewichteten MRT-Bildern des stATRT in Erscheinung treten, während sie bei stPNETs nur ausgesprochen selten vorzufinden sind.
Traumatic brain injury, a leading cause of death and disability, is a result of an outside force causing mechanical disruption of brain tissue and delayed pathogenic events which collectively exacerbate the injury. These pathogenic injury processes are poorly understood and accordingly no effective neuroprotective treatment is available so far. Experimental models are essential for further clarification of the highly complex pathology of traumatic brain injury towards the development of novel treatments. Among the rodent models of traumatic brain injury the most commonly used are the weight-drop, the fluid percussion, and the cortical contusion injury models. As the entire spectrum of events that might occur in traumatic brain injury cannot be covered by one single rodent model, the design and choice of a specific model represents a major challenge for neuroscientists. This review summarizes and evaluates the strengths and weaknesses of the currently available rodent models for traumatic brain injury.
Chlamydia trachomatis is an obligate intracellular pathogenic bacterium that has been refractory to genetic manipulations. Although the genomes of several strains have been sequenced, very little information is available on the gene structure of these bacteria. We used deep sequencing to define the transcriptome of purified elementary bodies (EB) and reticulate bodies (RB) of C. trachomatis L2b, respectively. Using an RNAseq approach, we have mapped 363 transcriptional start sites (TSS) of annotated genes. Semiquantitative analysis of mapped cDNA reads revealed differences in the RNA levels of 84 genes isolated from EB and RB, respectively. We have identified and in part confirmed 42 genome- and 1 plasmid-derived novel non-coding RNAs. The genome encoded non-coding RNA, ctrR0332 was one of the most abundantly and differentially expressed RNA in EB and RB, implying an important role in the developmental cycle of C. trachomatis. The detailed map of TSS in a thus far unprecedented resolution as a complement to the genome sequence will help to understand the organization, control and function of genes of this important pathogen.
In der vorliegenden Arbeit werden die Foraminiferenfaunen von 125 Proben, die aus drei Profilen des marinen Mitteljura (Bajocium bis Unteroxfordium) von Kachchh, West-Indien stammen, analysiert: Das Badi Nala-Jhura Village-Profil (ca. 550 m Mächtigkeit) und das Kamaguna-Profil (ca. 365 m Mächtigkeit) wurden am Jhura Hill (23°26’8’’ N; 69°37’00’’ E) ca. 17 km NW von Bhuj aufgenommen, während das dritte Profil durch den Jumara Dome (ca. 365 m Mächtigkeit) (23°40’40’’ N; 69°04’00’’ E) ca. 50 km NW des Jhura Hill liegt. Die Proben wurden lithologisch sowie nach ihren Foraminiferen-Vergesellschaftungen ausgewertet, welche wichtige Daten zur Palökologie lieferten. Die Profile umfassen die mitteljurassische Sedimentabfolge, die der Jhurio-, Patcham- und Chari-Formation des Jhura Dome und Jumara Dome angehört. Die Schichtenfolge des Bajocium und Bathonium besteht aus Karbonaten und gemischt karbonatisch-siliziklastischen Sedimenten der Jhurio- und Patcham-Formation, während im Callovium die Chari-Formation siliziklastisch dominiert ist. Die sedimentäre Abfolge des Kachchh-Beckens zeigt an der Bathonium-Callovium-Grenze signifikante Veränderungen in der Lithologie, im faunistischen Inhalt und in der Faunendiversität. Die lithologischen Veränderungen von einer Karbonat-dominierten Abfolge im mittleren und oberen Bathonium zu siliziklastischen Sedimenten im Callovium gehen mit einer Abnahme der zuvor hohen Diversität einher. Der Grund für den Faunenumschwung liegt vermutlich in einer deutlichen Änderung der Umweltbedingungen von sauerstoffreich im unteren zu sauerstoffarm im oberen Profilabschnitt, was sich in einem Wechsel von einer Kalkschaler- zu einer Sandschaler-dominierten Foraminiferenfauna dokumentiert. Eine weitere Ursache für den Fazies- und Faunenumschwung liegt in einer deutlichen KlimaÄnderung, indem heiße, aride Bedingungen im Bathonium von kühleren, feuchteren Verhältnissen zu Beginn des Callovium abgelöst wurden. Taxonomisch konnten insgesamt 111 Foraminiferentaxa identifiziert werden, die 43 Gattungen angehören. Insgesamt wurden 24 Sandschaler- und 85 benthische Kalkschaler-Taxa sowie 2 Arten planktischer Foraminiferen (Globuligerina) in den untersuchten Profilen bestimmt. Die benthischen Formen dominieren somit bei weitem an Diversität und Häufigkeit, da die planktischen Protoglobigerinen im mittleren Jura global noch keine große Rolle spielten. Aufgrund der spärlich vorhandenen Literatur wurde jedes Taxon inklusive Synonymieliste beschrieben, durch rasterelektronen-mikroskopische Aufnahmen (REM) dokumentiert und auf 9 Tafeln illustriert. In der Foraminiferen-Gesamtfauna dominieren Vertreter der Familie Nodosariidae mit den Gattungen Lenticulina, Astacolus, Citharina, Lingulina, Marginulinopsis, Nodosaria und Vaginulina. Mit der zweitgrößten Häufigkeit folgen die Familien Epistominidae, Textulariidae und Spirillinidae mit den Gattungen Epistomina, Spirillina sowie Reophax, Ammobaculites und Textularia. Die Arten Reophax sterkii, Triplasia althoffi, Verneuilinoides subvitreus, Nubeculinella bigoti, Dentalina filiformis, Saracenaria oxfordiana, Lingulina longiscata, Citharina flabellata, Palmula deslongchampsi, Vaginulina proxima, Ammodiscus asper, Ammodiscus siliceus, Triplasia bartensteini, Spirillina orbicula, Ophthalmidium carinatum, Tubinella inornata, Nodosaria fusiformis, Pyramidulina rara und Ramulina ascissa wurden erstmals in Gesteinen des Kachchh-Beckens nachgewiesen. Die dominierenden Taxa in den untersuchten Sedimenten sind epifaunale Formen, die einen hohen Sauerstoffgehalt im Bodenwasser benötigen. Infaunale, im Sediment lebende Arten treten dagegen etwas seltener auf. Diese lebten bevorzugt in sub- bis dysoxischen Milieus mit geringen Gehalten an gelöstem Sauerstoff im Bodenwasser und konnten auch Sauerstoffminima tolerieren. Aus der benthischen Foraminiferenfauna können folgende Rückschlüsse gezogen werden: • Die wichtigsten Parameter für die Verbreitung der Foraminiferen sind Substrat, Energieniveau und Sauerstoffgehalt. • Die höchste Diversität weisen die epifaunalen Vergesellschaftungen auf. • Mit zunehmender Sedimenttiefe nehmen Häufigkeit und Diversität ab. • Die Diversität der Kalkschaler ist höher als die der Sandschaler. • Minimale Diversitäten liegen in Stressmilieus mit geringen Sauerstoffgehalten vor. • Die hohen Werte des Evenness-Index weisen auf eine annähernd gleichmäßige Verteilung der benthischen Foraminiferen in den Profilen hin. • Epifaunale Arten werden von den physikochemischen Eigenschaften des Bodenwassers gesteuert. Sie sind auf partikuläres organisches Material und hohe Sauerstoffgehalte des Bodenwassers angewiesen. In der vorliegenden Arbeit wurden Probencluster gebildet, um Faunenassoziationen zu erfassen und danach die Umwelt- und Ablagerungsbedingungen zu rekonstruieren. Aus drei Profilen wurden 125 Proben für eine quantitative palökologische Analyse der Foraminiferen ausgewählt. Die W-mode Clusteranalyse zeigt eine große Ähnlichkeit zwischen den Proben, die sich in zwei Hauptcluster mit insgesamt sechs Subclustern gruppieren lassen. Die dadurch gebildeten Probengruppen stellen verschiedene Foraminiferenassoziationen dar, die sich beschreiben und weitgehend interpretieren lassen. • Assoziation A zeichnet sich durch hohe Anteile der Gattung Epistomina mosquensis aus. Diese Assoziation repräsentiert vollmarine Ablagerungsbedingungen mit normalmariner Salinität und guter Durchlüftung des Bodenwassers. • Assoziation B wird durchweg von Reophax metensis dominiert. Die Assoziation charakterisiert einen niedrig-energischen und teilweise sauerstoffarmen Sedimentationsraum. • Assoziation C wird stark von Dorothia prekummi dominiert und kennzeichnet vollmarine Bedingungen. • Assoziation D ist von Lenticulina subalata beherrscht und charakterisiert gutdurchlüftete Flachwasserbereiche. • Assoziation E wird von Spirillina polygyrata dominiert. Sie ist typisch für einen flachen Subtidalbereich unterhalb der Wellenbasis mit vollmarinen Bedingungen, normal mariner Salinität und gut durchlüftetem Bodenwasser. • Assoziation F zeichnet sich durch hohe Gehalte an Lenticulina quenstedti aus. Die Assoziation charakterisiert Bereiche, die überwiegend unterhalb der Schönwetter-Wellenbasis liegen und ein niedriges bis mittleres Sauerstoff-Niveau aufweisen. Aus einem Vergleich zwischen den Gehäusegrößen und der Häufigkeit von Lenticulina subalata und Epistomina mosquensis in den karbonatischen und siliziklastischen Sedimenten ergab sich, dass die Gehäuse von L. subalata und E. mosquensis in den Karbonaten größer sind als in den Siliziklastika. Die mitteljurassischen Ablagerungsräume des Kachchh-Beckens werden anhand der lithologischen und faunistischen Parameter rekonstruiert. Ferner werden die Foraminiferen-Vergesellschaftungen mit den von FÜRSICH et al. (2004) beschriebenen Makrofauna-Assoziationen verglichen. Aus diesen Untersuchungen ergaben sich folgende Schlussfolgerungen: • Die Ablagerungen im Jhura-Profil sind in flacherem Wasser sedimentiert worden als im Kamaguna-Profil und Jumara-Profil. • Die Mikro- und Makrofauna in diesen Profilen führt zu mehr oder weniger identischen palökologischen Schlussfolgerungen.
Die vorliegende Arbeit zeigt eine Möglichkeit auf, die bisher meist erfolglose Chemotherapie des malignen Melanoms zu verbessern: Durch Inhibition des Transkriptionsfaktors NF-kB, der für die Regulation vieler tumorrelevanter Gene verantwortlich ist, konnten die Tumorzellen gegenüber der Wirkung von Zytostatika sensibilisiert werden. Zunächst wurden acht verschiedene Melanomzellen in Bezug auf ihre NF-kB-Aktivität und der Expression NF-kB-regulierter Proteine vergleichen. Es konnte gezeigt werden, dass die Mehrzahl der Melanomzellen über konstitutive Aktivität von NF-κB verfügt. Dabei bestand kein eindeutiger Zusammenhang zwischen der Expression NF-kB-regulierter Proteine und der Aktivität dieses Transkriptionsfaktors im Kern, was komplexe Regulationsmechanismen bei der Transkription und Translation vermuten lässt. Anhand einer ausgewählten Melanomzelllinie konnte gezeigt werden, dass zwei verschiedene NF-kB-Inhibitoren, der Proteasom-Inhibitor Bortezomib und der neue IKK-Inhibitor KINK-1 die Aktivität von NF-kB deutlich hemmen. Beim Vergleich beider NF-kB-Inhibitoren ließen sich unerwartet verschiedene molekulare Wirkungsmechanismen nachweisen: Während Bortezomib konzentrationsabhängig eine sehr starke Induktion von NOXA, eine Induktion von p53 sowie eine Abnahme von Cyclin D1 bewirkte, zeigte KINK-1 seine Effekte vor allem in der Reduktion von Chemokinen wie IL-8 und MCP-1. Passend zur Veränderung der Expression zellzyklus-relevanter Proteine hatte Bortezomib einen stärkeren Effekt auf den Zellzyklus als KINK-1. Beide Inhibitoren wurden mit verschiedenen Zytostatika kombiniert und konnten einerseits die Apoptoseinduktion durch Zytostatika verstärken und andererseits die durch Zytostatika reduzierte Invasion weiter reduzieren. Allerdings zeigte sich bei der Untersuchung tumorrelevanter Chemokine, dass KINK-1 im Gegensatz zu Bortezomib synergistische Effekte mit Camptothecin und Doxorubicin aufweist. Trotz molekularer Unterschiede bewirkten beide NF-kB-Inhibitoren vergleichbare funktionelle Effekte auf zellulärer Ebene. Dies galt auch für ein präklinisches in-vivo-Modell, in dem die experimentelle Lungenmetastasierung von B16F10-Melanomzellen in Mäusen ermittelt wurde: Hier wurden die Mäuse mit Camptothecin, KINK-1 und Bortezomib allein im Vergleich zu den jeweiligen Kombinationen aus Zytostatikum und NF-kB-Inhibitor behandelt. Beide Kombinationen zeigten eine signifikante Reduktion des Lungengewichts im Vergleich zu Camptothecin allein. Diese Arbeit konnte also den Nutzen aus NF-kB-Inhibition in Kombination mit Zytostatika für die hier verwendeten Substanzen bekräftigen und dabei einige molekulare Unterschiede aufdecken.
In order to survive, organisms avoid threats and seek rewards. Classical conditioning is a simple model to explain how animals and humans learn associations between events that allow them to predict threats and rewards efficiently. In the classical conditioning paradigm, a neutral stimulus is paired with a biologically significant event (the unconditioned stimulus – US). In virtue of this association, the neutral stimulus acquires affective motivational properties, and becomes a conditioned stimulus (CS+). Defensive responses emerge for pairings with an aversive US (e.g., pain), and appetitive responses emerge for pairing with an appetitive event (e.g., reward). It has been observed that animals avoid a CS+ when it precedes an aversive US during a training phase (CS+ US; forward conditioning); whereas they approach a CS+ when it follows an aversive US during the training phase (US CS+; backward conditioning). These findings indicate that the CS+ acquires aversive properties after a forward conditioning, whereas acquires appetitive properties after a backward conditioning. It is thus of interest whether event timing also modulates conditioned responses in such an opponent fashion in humans, who are capable of explicit cognition about the associations. For this purpose, four experiments were conducted in which a discriminative conditioning was applied in groups of participants that only differed in the temporal sequence between CS+ onset and US onset (i.e., the interstimulus interval – ISI). During the acquisition phase (conditioning), two simple geometrical shapes were presented as conditioned stimuli. One shape (CS+) was always associated with a mild painful electric shock (i.e., the aversive US) and the other one (CS-) was never associated with the shock. In a between-subjects design, participants underwent either forward or backward conditioning. During the test phase (extinction), emotional responses to CS+ and CS- were tested and the US was never presented. Additionally, a novel neutral shape (NEW) was presented as control stimulus. To assess cognitive components, participants had to rate both the valence (the degree of unpleasantness or pleasantness) and the arousal (the degree of calmness or excitation) associated with the shapes before and after conditioning. In the first study, startle responses, an ancestral defensive reflex consisting of a fast twitch of facial and body muscles evoked by sudden and intense stimuli, was measured as an index of stimulus implicit valence. Startle amplitude was potentiated in the presence of the forward CS+ whilst attenuated in the presence of the backward CS+. Respectively, the former response indicates an implicit negative valence of the CS+ and an activation of the defensive system; the latter indicated an implicit positive valence of the CS+ and an activation of the appetitive system. In the second study, the blood-oxygen level dependent (BOLD) response was measured by means of functional magnetic resonance imaging (fMRI) to investigate neural responses after event learning. Stronger amygdala activation in response to forward CS+ and stronger striatum activation in response to backward CS+ were found in comparison to CS-. These results support the notion that the defensive motivational system is activated after forward conditioning since the amygdala plays a crucial role in fear acquisition and expression. Whilst the appetitive motivational system is activated after backward conditioning since the striatum plays a crucial role in reward processing. In the third study, attentional processes underlying event learning were observed by means of steady-state visual evoked potentials (ssVEPs). This study showed that both forward and backward CS+ caught attentional resources. More specifically, ssVEP amplitude was higher during the last seconds of forward CS+ that is just before the US, but during the first seconds of backward CS+ that is just after the US. Supposedly, attentional processes were located at the most informative part of CS+ in respect to the US. Participants of all three studies rated both forward and backward CS+ more negative and arousing compared to the CS-. This indicated that event timing did not influence verbal reports similarly as the neural and behavioral responses indicating a dissociation between the explicit and implicit responses. Accordingly, dual process theories propose that human behavior is determined by the output of two systems: (1) an impulsive implicit system that works on associative principles, and (2) a reflective explicit system that functions on the basis of knowledge about facts and values. Most importantly, these two systems can operate in a synergic or antagonistic fashion. Hence, the three studies of this thesis congruently suggest that the impulsive and the reflective systems act after backward association in an antagonistic fashion. In sum, event timing may turn punishment into reward in humans even though they subjectively rate the stimulus associated with aversive events as being aversive. This dissociation might contribute to understand psychiatric disorders, like anxiety disorders or drug addiction.
Um sich vor dem Austrocknen zu schützen, haben Pflanzen eine Transpirationsbarriere entwickelt, die als Membran alle primären, oberirdischen Pflanzenteile überzieht. Diese so genannte Kutikula besteht hauptsächlich aus den lipophilen Komponenten Kutin und Wachs und reduziert so effektiv den Verlust von Wasser und wasserlöslichen Nährstoffen aus dem Blattinneren. Trotzdem ist sie nicht vollständig undurchlässig, und so können Wasser und gelöste Substanzen wie organische und anorganische Nährstoffe, Pestizide oder Umweltchemikalien die Kutikula in beiden Richtungen permeieren. Dabei ist offensichtlich, dass die zu Grunde liegenden Transportmechanismen den Ernährungszustand der Pflanzen, die Effizienz von Pestiziden und die Wirkung von Umweltchemikalien beeinflussen. Ein genaues Verständnis der Transportprozesse auf denen die kutikuläre Permeation basiert, kann helfen die Wirkweise von blattapplizierten Dünge- und Pflanzenschutzmitteln zu optimieren, indem gezielt Wirk- oder Zusatzstoffe modelliert werden können, welche die Aufnahme steigern. In der vorliegenden Arbeit sollte deshalb der Einfluss physiko-chemischer Eigenschaften von hydrophilen Verbindungen auf die kutikuläre Permeation untersucht werden. Nicht zuletzt wegen ihrer strukturellen Ähnlichkeit mit den blattapplizierten Herbiziden Glufosinat und Glyphosat wurden Aminosäuren als Modellsubstenzen ausgewählt. Die verwendeten Aminosäuren sind gut wasserlöslich, wobei alle Oktanol/Wasser Verteilungskoeffizienten kleiner als 1 sind. Zusätzlich liegen alle Aminosäuren in gelöster Form als Ionen vor, was zu einer Hydratisierung der Moleküle führt. Es wird spekuliert, dass hydratisierte Moleküle keinen Zugang zur lipophilen Phase der Kutikula haben. Welche Rolle die Hydrathülle bei der Permeation tatsächlich spielt, ist allerdings noch unklar. Viele Aktivwirkstoffe liegen nur unter ganz bestimmten Bedingungen in geladener Form vor, während die Richtung der kontinuierlichen Nettoladung der Aminosäuren durch den pH Wert modifiziert wird. Damit kann der Einfluss verschiedener Ladungszustände auf die kutikuläre Permeation unter Verwendung eines einheitlichen Sets von Modellsubstanzen untersucht werden. Unter natürlichen Bedingungen sind Aminosäuren unter anderem auf Blattoberflächen zu finden, wo sie blattassoziierten Mikroorganismen eine profitable Nahrungsquelle bieten. Ob äußere Faktoren für die Deposition dieser Recourcen verantwortlich sind, oder ob der Ursprung innerhalb des Blattgewebes liegt, wird kontrovers diskutiert. Die Sorption von Aminosäuren in isolierte Kutikularmembranen ist sehr gering, und korreliert - anders als bei lipophilen Substanzen - nicht mit dem Oktanol/Wasser Verteilungskoeffizienten. Das zeigt, dass der Verteilung von lipophilen und hydrophilen Substanzen innerhalb der Kutikula verschiedene Mechanismen zu Grunde liegen. Unter einer gegebenen Bedingung werden die kutikulären Leitwerte der Aminosäuren negativ vom Molvolumen beeinflusst. Zudem übersteigt die Länge des Permeationswegs die eigentliche Dicke der Membran um ein Vielfaches. Diese Zusammenhänge kennzeichnen eine gehinderte Diffusion innerhalb einer engporigen und weit verzweigten Umgebung. Eine Änderung des pH Wertes wirkt sich in unterschiedlicher Form auf die Leitwerte von Wasser und Aminosäuren aus. Mit steigendem pH Wert erhöht sich die Wasserpermeabilität isolierter Kutikularmembranen, was durch eine zunehmende, messbare Wassersorption in die Kutikula erklärt werden kann. Eine pH abhängige Dissoziation funktioneller Gruppen bewirkt eine Schwellung des polaren Weges, weshalb auch für die anionischen Aminosäuren bei pH 11 die höchsten Leitwerte gemessen wurden. Die zwitterionischen Aminosäuren bei pH 6 wiesen hingegen die geringsten Leitwerte auf, was im Widerspruch zu der Beobachtung steht, dass bei pH 1 die geringste Wassersorption in die Kutikula stattfindet. Eine Erklärung hierfür liefern die Hydrathüllen, die bei den zwitterionischen Aminosäuren am stärksten und bei den anionischen Species am geringsten ausgeprägt sind. Eine negative Korrelation aller gemessenen Aminosäureleitwerte mit den entsprechenden hydratisierten Molvolumen zeigt eindeutig, dass die Hydrathülle eine wichtige Größe für die Permeation durch die Kutikula darstellt. Dabei nimmt der Leitwert einer hydrophilen Substanz mit definiertem Molvolumen mit kleiner werdender Hydrathülle zu. Intakte Blätter wurden in flüssiges Wasser als Rezeptorlösung getaucht, um steady-state Bedingungen aufrecht zu erhalten. Dabei konnte gezeigt werden, dass die Permeabilitäten von intakten Kutikularmembranen, die anhand der natürlichen Aminosäurekonzentration innerhalb der Blätter bestimmt wurden, in derselben Größenordnung liegen, wie die für isolierte Membranen gemessenen. Außerdem konnte ein Vergleich der Flussraten auf der Ober- und Unterseite der Blätter zeigen, dass die stomatären Poren nicht direkt in den Leachingprozess involviert sind.
Vergleichende Genexpressions-Analyse unterschiedlicher Populationen mesenchymaler Stammzellen

Neben den omnipotenten embryonalen Stammzellen existieren im menschlichen Körper adulte mesenchymale Stammzellen (MSZ). Diese Zellen sind in mesenchymalen Geweben über den gesamten Organismus verteilt und sorgen für die Entwicklung und Erneuerung von mesenchymalen Geweben wie Knochen, Knorpel und Bändern. Daher gelten die MSZ im Gegensatz zu den omnipotenten embryonalen Stammzellen als multipotent. Diese verschiedenen MSZ stellen keine homogene Population dar, zeigen aber sowohl in vivo und auch in vitro ein ähnliches Differenzierungsverhalten. In der vorliegenden Arbeit wurde nun eine aus den Knochentrabekeln selbst isolierte MSZ-Population, so genannte bhMSZ, mit MSZ aus dem Knochenmark, mhMSZ genannt, mittels Array-Analyse miteinander verglichen. Die technische Evaluation des Array respektive der zugehörigen SAM-Analyse (significance analysis of microarrays) mittels konventioneller oder Real-Time PCR diente dazu, die Verlässlichkeit der Aussage der Hybridisierungsverfahren zu überprüfen. Dies wurde mit einem Set an ausgewählten Genen durchgeführt, die signifikant differentiell exprimiert waren, und die im Rahmen der Stammzellbiologie relevant erschienen. Die Analyse zeigte, dass die Übereinstimmung der Aussage im Array in über 80 % mit den Ergebnissen der RT-PCR kongruent war. Auf Grund starker interindividueller Schwankungen zeigte sich aber auch, dass die Anzahl der Spender 5 nicht unterschreiten sollte. Im Rahmen der Untersuchungen ergab sich, dass offenbar bei MSZ der Passage 0 eine Kontamination der MSZ mit Plasmazellen vorliegt. Weitere Versuche zeigten, dass erst das Passagieren der MSZ kontaminierende Plasmazellen weitgehend aus der Zellkultur entfernte. Aus diesem Grund wurde in einer weiteren Array Analyse das Transkriptom von MSZ aus Knochentrabekeln mit MSZ aus dem Knochenmark in Passage 1 verglichen. Es zeigten sich in einer stringenten SAM-Analyse keine Unterschiede im Transkriptom. Für klinische Anwendungen scheinen die bhMSZ daher auf Grund der aufwendigeren Isolierung und des dennoch eher geringen Zellgewinns nicht im gleichen Maß für klinische Anwendungen geeignet wie mhMSZ.
The human gut is home for thousands of microbes that are important for human life. As most of these cannot be cultivated, metagenomics is an important means to understand this important community. To perform comparative metagenomic analysis of the human gut microbiome, I have developed SMASH (Simple metagenomic analysis shell), a computational pipeline. SMASH can also be used to assemble and analyze single genomes, and has been successfully applied to the bacterium Mycoplasma pneumoniae and the fungus Chaetomium thermophilum. In the context of the MetaHIT (Metagenomics of the human intestinal tract) consortium our group is participating in, I used SMASH to validate the assembly and to estimate the assembly error rate of 576.7 Gb metagenome sequence obtained using Illumina Solexa technology from fecal DNA of 124 European individuals. I also estimated the completeness of the gene catalogue containing 3.3 million open reading frames obtained from these metagenomes. Finally, I used SMASH to analyze human gut metagenomes of 39 individuals from 6 countries encompassing a wide range of host properties such as age, body mass index and disease states. We find that the variation in the gut microbiome is not continuous but stratified into enterotypes. Enterotypes are complex host-microbial symbiotic states that are not explained by host properties, nutritional habits or possible technical biases. The concept of enterotypes might have far reaching implications, for example, to explain different responses to diet or drug intake. We also find several functional markers in the human gut microbiome that correlate with a number of host properties such as body mass index, highlighting the need for functional analysis and raising hopes for the application of microbial markers as diagnostic or even prognostic tools for microbiota-associated human disorders.
Der vorliegende Artikel befasst sich mit einer Aktionsforschung, die im Rahmen eines Japanischunterrichts an der Ruhr-Universität Bochum durchgeführt wurde und stellt konkrete Schritte einer Aktionsforschung dar. Die Unterrichtende versucht, anhand der empirischen Daten aus ihrem eigenen Unterricht heraus zu verstehen, inwieweit der von ihr konzipierte und im Kurs regelmäßig verwendete Reflexionsbogen das selbstreflektierende Lernen fördert. Außerdem wird der Frage nachgegangen, in welchem Zusammenhang autonomes Lernen mit bestimmten Grammatikvermittlungsmethoden steht. Ist eine induktive Grammatikvermittlung zur Förderung autonomen Lernens erforderlich? Was muss gefördert werden, wenn man autonomes Lernen fördern möchte? Die in diesem Artikel dargestellten Fragestellungen und Erkenntnisse beziehen sich zwar auf den Japanischunterricht, aber bieten im weiteren Unterrichtskontext ebenfalls einen Ansatzpunkt für die Weiterentwicklung des Fremdsprachenunterrichts. Darüber hinaus bietet Aktionsforschung die Möglichkeit, nicht nur Sprachlehrforschende im engen Sinne, sondern alle Unterrichtende, die an der forschungsgeleiteten Entwicklung des Unterrichtsgeschehens interessiert sind, in die Forschung mit einzubeziehen, was zur Weiterentwicklung der Fremdsprachenlehr/lernforschung sicherlich beiträgt.
This thesis consists of three major chapters, each of which has been separately published or under the process for publication. The first chapter is about anatomical characterization of the mushroom body of adult Drosophila melanogaster. The mushroom body is the center for olfactory learning and many other functions in the insect brains. The functions of the mushroom body have been studied by utilizing the GAL4/UAS gene expression system. The present study characterized the expression patterns of the commonly used GAL4 drivers for the mushroom body intrinsic neurons, Kenyon cells. Thereby, we revealed the numerical composition of the different types of Kenyon cells and found one subtype of the Kenyon cells that have not been described. The second and third chapters together demonstrate that the multiple types of dopaminergic neurons mediate the aversive reinforcement signals to the mushroom body. They induce the parallel memory traces that constitute the different temporal domains of the aversive odor memory. In prior to these chapters, “General introduction and discussion” section reviews and discuss about the current understanding of neuronal circuit for olfactory learning in Drosophila.
Bei N-Acyl-Ethanolaminphosphaten handelt es sich um eine bislang wenig untersuchte Klasse polarer Substanzen, deren Erforschung aufgrund ihrer strukturellen Analogie zu apolaren, physiologisch wirksamen N-Acyl-Ethanolaminen von Interesse ist. Zu bear-beiten waren analytische Fragestellungen, die auch synthetische Aufgaben beinhalteten, wie Methodenentwicklung und Versuche zur Erfassung von N-Acyl-Ethanolamin-phosphaten in ausgewählten Lebensmitteln sowie strukturelle Studien zur „Bioaktivität“ der Verbindungen. Erstes Ziel der vorliegenden Arbeit war es demzufolge, eine geeig-nete Methode für deren qualitative und quantitative Analytik zu entwickeln. Gleichzei-tig wurden ausgewählte N-Acyl-Ethanolaminphosphate synthetisiert. Aufgrund des literaturbekannten Vorkommens von N-Acyl-Ethanolaminen in Wein wurden für die Lebensmitteluntersuchungen fermentierte Produkte, d.h. drei verschie-dene Sake (Japanischer Reiswein) und ein fermentierter Rotkohl verwendet. Parallel zu diesen Untersuchungen erfolgten auch Studien zur Stabilität der N-Acyl-Ethanolamin-phosphate. Versuchsreihen zur Überprüfung potentieller „Bioaktivität“ umfassten Studien mit al-kalischer Phosphatase, PhospholipaseA2, Lipoxygenase, Xanthinoxidase, β-N-Acetyl-hexosaminidase und dem Cannabinoidrezeptor-1.
Der Kalium-Chloridkanal KCC3 ist für die elektroneutrale Bewegung von Ionen durch die Zellmembran zuständig. Weiterhin zeigte sich, dass dieser Kanal, dessen Isoform KCC3a hauptsächlich im ZNS exprimiert ist, in der Regulation der Zellproliferation eine wichtige Rolle zu spielen scheint. Mutationen im Bereich des KCC3-Gens führen zur Entwicklung des sogenannten Andermann-Syndroms, einer progressiven sensomotorischen Neuropathie, die assoziiert ist mit einer Agenesie des Corpus callosum und psychotischen Episoden. Das Gen ist lokalisiert auf Chromosom 15q14, einer Region von der angenommen wird, dass sie eine Suszeptibilitätsregion für schizophrene Psychosen und bipolare Erkrankungen darstellt. Im Rahmen von Mutationsanalysen des KCC3-Gens konnten zwei seltenen G-Varianten im Bereich des Promotors und der 5-UTR, sowie eine seltene Thymidininsertion in Intron 4 des Gens detektiert werden. Untersucht wurden hierbei zwei an katatoner Schizophrenie (periodischer Katatonie) erkrankte Individuen eines als Familie 11 bezeichneten Stammbaums und drei gesunde Individuen. Mit Hilfe dieser Assoziationsstudie soll nun geklärt werden, ob diese Polymorphismen in der Pathogenese schizophrener Psychosen und/oder bipolarer Erkrankungen involviert sind. Die zwei seltenen G-Varianten und die Thymidininsertion in Intron 4 des KCC3- Gens fanden sich bei allen an periodischer Katatonie erkrankten Mitglieder der beschriebenen Familie 11, die gemeinsame Markerallele zwischen den Markern D15S144 and D15S132 aufweist. Die drei seltenen Polymorphismen wiesen ein Kopplungsungleichgewicht auf. Es zeigte sich, dass der Haplotyp bestehend aus allen drei Varianten, den zwei seltenen G-Varianten und der Thymidininsertion, und der Haplotyp bestehend aus 2 Varianten mit bipolaren Erkrankungen assoziiert ist, für den Fall, dass die seltene G-Variante im Bereich des Promotors beinhaltet ist. Im Rahmen der Einzelmarkeranalyse zeigte sich eine Assoziation der G-Variante im Bereich des Promotors mit bipolaren Erkrankungen. Hinsichtlich der schizophrenen Psychosen zeichnete sich bei nicht-signifikanten Ergebnissen ein Trend in Richtung Assoziation zwischen den untersuchten Varianten des KCC3-Gens und der Patientengruppe, die aus Patienten mit bipolaren und schizophrenen Erkrankungen bestand, ab. Zusammenfassend kann schließlich behauptet werden, dass SLC12A6 ein mögliches Risiko-Gen darstellt, welches die Entwicklung bipolarer Erkrankungen begünstigt. Die Rolle des SLC12A6-Gens für die Entwicklung einer schizophrenen Psychose scheint hingegen nicht eindeutig geklärt zu sein. Es ist jedoch durchaus denkbar, dass das SLC12A6-Gen möglicherweise in der Pathogenese schizophrener Psychosen, insbesondere der katatonen Schizophrenie involviert ist.
Regulation of pathogen-inducible volatile compounds in Arabidopsis and their role in plant defense

Plants are constantly attacked by pathogenic microbes. As a result, they have evolved a plethora of constitutive and inducible defense responses to defend against attempted pathogen infection. Although volatile organic compounds have been implicated in plant defense, direct evidence of their function in plant resistance is still lacking. I have examined the role of VOCs in Arabidopsis defense against the hemibiotrophic bacterial pathogen Pseudomonas syringae pv. maculicola. The obtained results show that the vegetative parts of Arabidopsis produces and emits the volatile phenylpropanoid MeSA and three kinds of terpenoids, (E,E)-4,8,12-trimethyltrideca-1,3,7,11-tetraene (TMTT), alpha-ionon and beta-farnesen, upon avirulent and virulent P. syringae inoculation. Whereas the most abundant volatiles, MeSA and TMTT, are already produced at early stages of infection in the compatible and incompatible interaction, enhanced emission of alpha-ionon and beta-farnesen can only be detected in later stages of the compatible interaction. It was revealed that pathogen-induced synthesis of TMTT in Arabidopsis requires the JA signaling pathway but occurs independently of SA defense signaling. Similarly, the production of MeSA is dependent on JA signaling but not on the SA defense signaling pathway. Furthermore, production of MeSA is dependent on the function of ISOCHORISMATE SYNTHASE1, which produces its precursor SA. Upon inoculation with avirulent P. syringae, endogenously produced JA activates the JA signalling pathway to mediate MeSA and TMTT synthesis. By contrast, in the compatible Arabidopsis-Psm interaction, production of MeSA predominantly depends on the P. syringea the virulence factor coronatine, which activates JA downstream signaling. To learn more about the role of inducible VOCs in plant defense responses, I have identified an Arabidopsis T-DNA insertions line with a defect in the TERPENE SYNTHASE4 (TPS4) gene. Emission profiles from this mutant revealed that the induced production of TMTT but not of alpha-ionone, beta-farnesene or MeSA are abolished, demonstrating that TPS4 specifically regulates the P. syringae-induced synthesis of TMTT in Arabidopsis. The lack of TMTT in tps4 mutants, however, does not affect plant defense responses and resistance induction against P. syringae. This excludes a role of the terpenoid as an effective phytoalexin in Arabidopsis leaves against the bacterial pathogen. Moreover, tps4 mutant plants are still able to mount a SAR response, excluding a signaling function of TMTT during SAR. An important aim of our studies was to address the defensive role of MeSA, the major VOC emitted from P. syringae-inoculated Arabidopsis leaves. MeSA has been recently proposed as a critical long distance signal in the development of SAR. I found that two independent T-DNA insertions lines with defects in expression of the pathogen-inducible SA methyl transferase gene BSMT1 are completely devoid of pathogen-induced production of MeSA. However, bsmt1 mutant plants are capable to increase the level of SA in systemic, non-infected leaves of Arabodopsis and develop SAR like wild-type plants upon local P. syringae-inoculation. Thus, MeSA does not function as a critical SAR signal in Arabidopsis. Further experiments showed that SA accumulation in distant leaves occurs due to de novo synthesis through isochorismate synthase. In addition, we also ruled out a critical defensive role of MeSA at inoculation sites, because bsmt1 mutants are able to build up SA-dependent defense responses and local resistance in a wild-type-like manner. The conversion of SA to MeSA and subsequently emission of MeSA from the plant might help the plant to detoxify an excess of SA. This process is regulated by the JA pathway and might be one means to mediate negative crosstalk between JA and SA signaling. Moreover, the COR-triggered conversion of SA to MeSA and emission of the volatile methyl ester could be a way by which virulent P. syringae is able to attenuate the SA-defense pathway.
p8 ist ein erstmals im Zusammenhang mit akuter Pankreatitis beschriebenes Protein, das im exokrinen und endokrinen Pankreas mit vermehrtem Zellwachstum assoziiert ist. Bei der Analyse seiner Primärstruktur wurde ein speziesübergreifend hoch konservierter Abschnitt, eine sogenannte NLS, ausgemacht, der HMG-Y/I-Proteinen ähnelt. Da HMG-Proteine oft als Transkriptionsfaktoren wirken, wurde die Hypothese formuliert, auch p8 sei ein HMG-Y/I-Protein und wirke als Transkriptionsfaktor im Nukleus. Um die Bedeutung der rp8-NLS näher zu charakterisieren, wurde in INS-1 beta-Zellen ein rp8(NLS-)-EGFP Fusionsprotein ektopisch exprimiert, um dessen subzelluläre Lokalisation zu untersuchen. Es zeigte sich, ähnlich wie bei Kontrollzellen mit ektoper Expression von EGFP allein, eine gleichmäßige Verteilung von rp8(NLS-)-EGFP zwischen Zytoplasma und Nukleus. Da rp8(NLS-) trotz fehlender NLS dennoch in den Kern translozieren kann, scheint die NLS für diesen Vorgang nicht essentiell zu sein. Diese Annahme wird gestützt durch die Beobachtung, dass einzeln exprimiertes rp8(NLS-) seine Proliferation induzierende Wirkung nicht verliert. In Zellzählungsexperimenten zeigte sich, dass ein rp8- bzw. p8(NLS-)-EGFP Fusionsprotein keinen proliferationsfördernden Einfluss in INS-1 und hMSC-TERT Zellen hat. Bei ektoper Expression von rp8 bzw. rp8(NLS-) und hrGFP als Einzelproteine konnte jedoch eine zwischen beiden rp8-Varianten ähnliche und insgesamt signifikante Stimulation der Zellvermehrung beobachtet werden. Dies belegt, dass die Fusion von rp8 an EGFP dessen biologische Funktion inhibiert, während die Deletion der NLS keinen Einfluß darauf hat. Da der proliferative Stimulus von p8 in menschlichen hMSC-TERT Zellen unabhängig von der Herkunft von p8 aus Ratte oder Mensch ist, scheint p8 bei Säugern hoch konserviert zu sein und speziesübergreifend zu wirken. Aus der hier vorgestellten Arbeit geht hervor, dass der molekulare Mechanismus, über den p8 glukoseabhängig proliferationsinduzierend in INS-1 beta-Zellen wirkt, nicht über die NLS vermittelt wird.
