This article exemplifies Gunter Grimm’s concept of productive reception by analyzing César Fernández García’s young adult novel La última bruja de Trasmoz (2009) and the episode Tiempo de hechizos (2017) of the TV series El ministerio del tiempo as two modern works which artistically and creatively deal with Gustavo Adolfo Bécquer’s Cartas desde mi celda (1864). The study is based on the core assumption that the choice of genre greatly influences the treatment of motives such as the perception of nature and the idea of dreams, as well as the image of the witch. While Bécquer’s letters, which focus on the rural people’s superstition, present the witch of Aragonese folklore, the modern examples portray two of her fantastic counterparts: the novel depicts its magical protagonist as an evil supernatural being, whereas the TV episode of the science fiction series shows the witch as a time traveller and a victim.
Background
Atrial fibrillation (AF) is present in 15–20% of patients with acute ischemic stroke. Oral anticoagulation reduces the risk of AF-related recurrent stroke but clinical guideline recommendations are rather vague regarding its use in the acute phase of stroke. We aimed to assess the current clinical practice of medical stroke prevention in AF patients during the acute phase of ischemic stroke.
Methods
In April 2017, a standardized anonymous questionnaire was sent to clinical leads of all 298 certified stroke units in Germany.
Results
Overall, 154 stroke unit leads participated (response rate 52%). Anticoagulation in the acute phase of stroke is considered feasible in more than 90% of AF patients with ischemic stroke. Clinicians assume that about two thirds of all AF patients (range 20–100%) are discharged on oral anticoagulation. According to local preferences, acetylsalicylic acid is given orally in the majority of patients with delayed initiation of oral anticoagulation. A non-vitamin K-dependent oral anticoagulant (NOAC) is more often prescribed than a vitamin K-dependent oral anticoagulant (VKA). VKA is more often chosen in patients with previous VKA intake than in VKA naive patients. In the minority of patients, stroke unit leads discuss the prescription of a specific oral anticoagulant with the treating general practitioner. Adherence to medical stroke prevention after hospital discharge is not assessed on a regular basis in any patient by the majority of participating stroke centers.
Conclusions
Early secondary stroke prevention in AF patients in German stroke units is based on OAC use but prescription modalities vary in clinical practice.
Bei der Multiplen Sklerose (MS) handelt es sich um eine Autoimmunerkrankung des zentralen Nervensystems (ZNS). Abhängig von der betroffenen ZNS-Region kann es zu vielfältigen Symptomen kommen. Neben neurologischen Symptomen verursacht durch ZNS-Läsionen leidet ein Großteil der MS-Patienten auch unter gastrointestinalen Funktionsstörungen. Diese gastrointestinalen Symptome wurden bisher eher auf Läsionen im Rückenmark zurückgeführt und nicht direkt in Verbindung mit der autoimmunen Ätiologie der Erkrankung gebracht.
In dieser Studie wurde das enterische Nervensystem (ENS) in einem B-Zell- und Antikörper-abhängigen Mausmodell der MS untersucht. Dafür wurde der Autoimmunprozess durch Immunisierung mit MP4, einem Fusionsprotein aus dem Myelin-Basischen-Protein (MBP) und dem Proteolipid-Protein (PLP), ausgelöst. Das ZNS und ENS wurden in den unterschiedlichen Erkrankungsstadien immunhistochemisch und elektronenmikroskopisch analysiert. Neben der Immunpathologie des ZNS konnte dabei eine Degeneration des ENS schon vor dem Einsetzen der ersten neurologischen Defizite nachgewiesen werden. Die ENS-Pathologie war antikörper-mediiert und ging einher mit einer verringerten gastrointestinalen Motilität sowie mit einer Gliose und Neurodegeneration des ENS.
Mithilfe von Immunpräzipitation und Massenspektrometrie konnten im ENS vier mögliche Zielstrukturen des Autoimmunprozesses identifiziert werden, was auf sog. epitope spreading hindeutet. Auch im Plasma von MS-Patienten konnten Antikörper gegen drei dieser Antigene nachgewiesen werden. Des Weiteren zeigten sich in Kolon-Resektaten von MS-Patienten erste Ansätze einer Neurodegeneration und Gliose des ENS.
In dieser Studie wurde zum ersten Mal ein direkter Zusammenhang zwischen der Autoimmunreaktion gegen das ZNS und einer simultanen Reaktion gegen das ENS gezeigt. Dies kann einen Paradigmenwechsel im Verständnis der Immunpathogenese der MS anstoßen und neue therapeutische und diagnostische Ansätze initiieren.
Die Konzertlandschaft im Deutschen Kaiserreich wurde neben den zahlreichen Opern- und Sinfonieorchestern vor allem durch die vielseitigen Darbietungen der Militärkapellen bei verschiedensten Anlässen geprägt. Dabei zeigten sich die Musikkorps als vielseitig einsetzbare Formationen und konnten einen spezifischen Beitrag zur Musikkultur leisten. Die Militärorchester hatten durch ihr konzertantes Wirken die Möglichkeit, großen Einfluss auf das musikkulturelle Profil innerhalb einer Region zu nehmen. Diese und andere Aspekte können am Beispiel der „Turnhallenkonzerte“ zu Arolsen aufgezeigt werden. Militärkapellmeister Hugo Rothe konnte sich bei diesen Konzerten sowohl durch seine Programmauswahl als auch durch die Zusammenarbeit mit vielen nationalen und internationalen Gastsolisten auszeichnen. Diese Ergänzung zum militärmusikalischen Tagesgeschäft (Paraden, Zeremonielle) war eine notwendige Facette, die sich besonders aus einem eigenen künstlerischen Ehrgeiz heraus zur musikalischen Selbstverwirklichung – sowohl für den Dirigenten als auch für die Orchestermusiker – entwickelte. Auf diese Weise konnte musikalische Volksbildung durch Popularisierung verschiedenster Werke sowie durch ein speziell zusammengestelltes Repertoire erfolgen, was sich anhand des umfangreichen Nachlassmaterials von Militärkapellmeister Hugo Rothe veranschaulichen lässt.
The synthetic compound dendritic polyglycerol sulfate (dPGS) is a pleiotropic acting molecule but shows a high binding affinity to immunological active molecules as L‐/P‐selectin or complement proteins leading to well described anti‐inflammatory properties in various mouse models. In order to make a comprehensive evaluation of the direct effect on the innate immune system, macrophage polarization is analyzed in the presence of dPGS on a phenotypic but also metabolic level. dPGS administered macrophages show a significant increase of MCP1 production paralleled by a reduction of IL‐10 secretion. Metabolic analysis reveals that dPGS could potently enhance the glycolysis and mitochondrial respiration in M0 macrophages as well as decrease the mitochondrial respiration of M2 macrophages. In summary the data indicate that dPGS polarizes macrophages into a pro‐inflammatory phenotype in a metabolic pathway‐dependent manner.
Insulin-like growth factor-I (IGF-I) is a 70-amino acid polypeptide with a molecular weight of approximately 7.6 kDa acting as an anabolic effector. It is essential for tissue growth and remodeling. Clinically, it is used for the treatment of growth disorders and has been proposed for various other applications including musculoskeletal diseases. Unlike insulin, IGF-I is complexed to at least six high-affinity binding proteins (IGFBPs) exerting homeostatic effects by modulating IGF-I availability to its receptor (IGF-IR) on most cells in the body as well as changing the distribution of the growth factor within the organism.1-3 Short half-lived IGF-I have been the driving forces for the design of localized IGF-I depot systems or protein modification with enhanced pharmacokinetic properties. In this thesis, we endeavor to present a versatile biologic into which galenical properties were engineered through chemical synthesis, e.g., by site-specific coupling of biomaterials or complex composites to IGF-I. For that, we redesigned the therapeutic via genetic codon expansion resulting in an alkyne introduced IGF-I, thereby becoming a substrate for biorthogonal click chemistries yielding a site-specific decoration.
In this approach, an orthogonal pyrrolysine tRNA synthetase (PylRS)/tRNAPyl CUA pair was employed to direct the co-translational incorporation of an unnatural amino acid—¬propargyl-L-lysine (plk)—bearing a clickable alkyne functional handle into IGF-I in response to the amber stop codon (UAG) introduced into the defined position in the gene of interest. We summarized the systematic optimization of upstream and downstream process alike with the ultimate goal to increase the yield of plk modified IGF-I therapeutic, from the construction of gene fusions resulting in (i) Trx-plk-IGF-I fusion variants, (ii) naturally occurring pro-IGF-I protein (IGF-I + Ea peptide) (plk-IGF-I Ea), over the subsequent bacterial cultivation and protein extraction to the final chromatographic purification. The opportunities and hurdles of all of the above strategies were discussed. Evidence was provided that the wild-type IGF-I yields were pure by exploiting the advantages of the pHisTrx expression vector system in concert with a thrombin enzyme with its highly specific proteolytic digestion site and multiple-chromatography steps. The alkyne functionality was successfully introduced into IGF-I by amber codon suppression. The proper folding of plk-IGF-I Ea was assessed by WST-1 proliferation assay and the detection of phosphorylated AKT in MG-63 cell lysate. The purity of plk-IGF-I Ea was monitored with RP-HPLC and SDS-PAGE analysis. This work also showed site-specific coupling an alkyne in plk-IGF-I Ea by copper (I)-catalyzed azide-alkyne cycloaddition (CuAAC) with potent activities in vitro. The site-specific immobilization of plk-IGF-I Ea to the model carrier (i.e., agarose beads) resulted in enhanced cell proliferation and adhesion surrounding the IGF-I-presenting particles. Cell proliferation and differentiation were enhanced in the accessibility of IGF-I decorated beads, reflecting the multivalence on cellular performance.
Next, we aimed at effectively showing the disease environment by co-delivery of fibroblast growth factor 2 (FGF2) and IGF-I, deploying localized matrix metalloproteinases (MMPs) upregulation as a surrogate marker driving the response of the drug delivery system. For this purpose, we genetically engineered FGF2 variant containing an (S)-2-amino-6-(((2-azidoethoxy)carbonyl)amino)hexanoic acid incorporated at its N-terminus, followed by an MMPs-cleavable linker (PCL) and FGF2 sequence, thereby allowing site-directed, specific decoration of the resultant azide-PCL-FGF2 with the previously mentioned plk-IGF-I Ea to generate defined protein-protein conjugates with a PCL in between. The click reaction between plk-IGF-I Ea and azide-PCL-FGF2 was systematically optimized to increase the yield of IGF-FGF conjugates, including reaction temperature, incubation duration, the addition of anionic detergent, and different ratios of the participating biopharmaceutics. The challenge here was that CuAAC reaction components or conditions might oxidize free cysteines of azide-PCL-FGF2 and future work needs to present the extent of activity retention after conjugation. Furthermore, our study provides potential options for dual-labeling of IGF-I either by the introduction of unnatural amino acids within two distinct positions of the protein of interest for parallel “double-click” labeling of the resultant plk-IGF-I Ea-plk or by using a combination of enzymatic-catalyzed and CuAAC bioorthogonal coupling strategies for sequentially dual-labeling of plk-IGF-I Ea.
In conclusion, genetic code expansion in combination with click-chemistry provides the fundament for novel IGF-I analogs allowing unprecedented site specificity for decoration. Considerable progress towards IGF-I based therapies with enhanced pharmacological properties was made by demonstrating the feasibility of the expression of plk incorporated IGF-I using E. coli and retained activity of unconjugated and conjugated IGF-I variant. Dual-labeling of IGF-I provides further insights into the functional requirements of IGF-I. Still, further investigation warrants to develop precise IGF-I therapy through unmatched temporal and spatial regulation of the pleiotropic IGF-I.
Die essenzielle, Ubiquitin-selektive ATPase p97 reguliert eine Vielzahl unterschiedlicher Prozesse in Eukaryoten. Dazu zählen Proteinqualitätskontrolle, DNA-Reparatur, Signaltransduktion, Zellzykluskontrolle, Autophagie sowie das endolysosomale System. Diese unterschiedlichen Funktionen von p97 werden durch die Bindung von Kofaktoren engmaschig gesteuert und kontrolliert. Die größte und am besten untersuchte Gruppe von p97-Kofaktoren sind die Proteine der UBX Familie. Diese zeichnen sich durch den Besitz einer UBX-Domäne aus, welche die Bindung an p97 vermittelt. Das in höheren Eukaryoten konservierte Familienmitglied UBXD1 besitzt darüber hinaus mit einer PUB-Domäne und einem VIM-Motiv noch mindestens zwei weitere p97-Bindemodule. UBXD1 kann an Vesikel des endolysosomalen Degradationssytems lokalisieren, seine genauen zellulären Funktionen sind jedoch noch weitgehend unbekannt.
Ziel dieser Arbeit war die funktionelle Charakterisierung von humanem UBXD1. Dafür wurden Kandidaten eines zuvor durchgeführten Yeast-Two-Hybrid-Screens auf ihre Two Hybrid-Interaktion mit unterschiedlichen UBXD1-Varianten getestet. Darüber hinaus wurde durch Immunpräzipitationsexperimente untersucht, ob die Kandidatenproteine auch in Säugerzellen mit UBXD1 interagieren. Als vielversprechende neue Bindungspartner von UBXD1 wurden so die Ubiquitin-Ligase TRIAD3A und das Ubiquitin-editierende Protein A20 identifiziert. Desweiteren konnte gezeigt werden, dass die Interaktion zwischen UBXD1 und A20 von einer funktionellen PUB Domäne und dem siebten Zinkfinger Motiv von A20 abhängig ist.
Da sowohl TRIAD3A als auch A20 negative Regulatoren des NF B Signalweges sind, wurde daraufhin untersucht, ob auch UBXD1 eine Funktion in diesem Signalweg besitzt. Tatsächlich war in UBXD1-depletierten HeLa 57A-Zellen die NF B-abhängige Expression eines Reportgens nach Aktivierung des Signalweges durch TNF, IL-1, Doxorubicin und H2O2 stark reduziert. Dabei spricht die verringerte Aktivierung nach unterschiedlichen Stimuli für eine generelle Rolle von UBXD1 im NF B Signalweg. Durch quantitative Echtzeit-PCR konnte gezeigt werden, dass in HeLa- und HEK293T-Zellen nach UBXD1-Depletion auch die Expression endogener NF B Zielgene verringert ist. Da in UBXD1-depletierten Zellen nach Stimulation mit TNF oder IL-1 bereits die Kerntranslokation des NF B-Transkriptionsfaktor p65 reduziert ist, ist davon auszugehen, dass UBXD1 an einer früheren Phase der Aktivierung des Signalweges beteiligt ist. Möglicherweise ist dies darauf zurückzuführen, dass UBXD1 bekannte Funktionen von A20 reguliert und etwa die Bindung von A20 an Vesikel des endolysosomalen Systems oder an lineare Ubiquitinketten beeinflusst. Diese Arbeit beschreibt somit eine neue Funktion des p97-Kofaktors UBXD1 im NF B-Signalweg.
In Science-Fiction-Literatur und -Filmen begegnen sie uns ständig: Mond- und Marsbasen, menschliche Außenposten im All, die die Fortschrittlichkeit und den Erkenntnisgewinn der Menschheit signalisieren und Reisen in die Tiefen des Alls ermöglichen. Blockbuster wie Der Marsianer (2015) oder Kultserien wie Star Trek werden millionenfach gesehen. Die Menschen scheinen gerne von extraterrestrischen Abenteuern in anderen Zeiten und anderen Welten zu träumen. Aber wie sieht es aus, wenn die Ideen der Leinwände ins Hier und Jetzt geholt werden?
SpaceX, ein privates, amerikanisches Raumfahrtunternehmen und sein Gründer, Elon Musk, haben es sich zum Ziel gesetzt, dem Menschen ein (Über-)Leben auf anderen Planeten zu ermöglichen – nicht erst in Jahrzehnten oder gar Jahrhunderten, sondern in ein paar Jahren. Auf dem International Astronautical Congress 2016 stelle Elon Musk zum ersten Mal konkrete Pläne für eine Kolonisierung des Mars vor, von technischen Vorbereitungen bis zum ersten Eintreffen von Menschen auf dem Roten Planeten im Jahr 2025.
Angesichts des regen Interesses an der fiktionalen Seite des Themas, stellt sich die Frage, wie eine – zumindest angekündigte – Verschiebung in die Realität aufgenommen wird. Anhand einer Analyse der Diskurslage in zwei deutschen Leitmedien (Bild.de und Focus Online) untersucht diese Arbeit die Bewertung des Vorhabens, Einschätzungen zu dessen Machbarkeit und zur Glaubwürdigkeit von SpaceX und Elon Musk und kommt dabei zu gänzlich unerwarteten Ergebnissen.
Elucidating the mechanism of action of domatinostat (4SC-202) in cutaneous T cell lymphoma cells

Background
Targeting epigenetic modifiers is effective in cutaneous T cell lymphoma (CTCL). However, there is a need for further improvement of this therapeutic approach. Here, we compared the mode of action of romidepsin (FK228), an established class I histone deacetylase inhibitor, and domatinostat (4SC-202), a novel inhibitor of class I HDACs, which has been reported to also target the lysine-specific histone demethylase 1A (LSD1).
Methods
We performed MTS assays and flow cytometric analyses of propidium iodide or annexin V-stained cells to assess drug impact on cellular proliferation, cell cycle distribution, and survival. Histone acetylation and methylation as well as caspase activation was analyzed by immunoblot. Gene expression analysis was performed using NanosString technology. Knockdown and knockout of LSD1 was achieved with shRNA and CRISPR/Cas9, respectively, while the CRISPR/Cas9 synergistic activation mediator system was used to induce expression of endogenous HDACs and LSD1. Furthermore, time-lapse fluorescence microscopy and an in vitro tubulin polymerization assay were applied.
Results
While FK228 as well as 4SC-202 potently induced cell death in six different CTCL cell lines, only in the case of 4SC-202 death was preceded by an accumulation of cells in the G2/M phase of the cell cycle. Surprisingly, apoptosis and accumulation of cells with double DNA content occurred already at 4SC-202 concentrations hardly affecting histone acetylation and methylation, and provoking significantly less changes in gene expression compared to biologically equivalent doses of FK228. Indeed, we provide evidence that the 4SC-202-induced G2/M arrest in CTCL cells is independent of de novo transcription. Furthermore, neither enforced expression of HDAC1 nor knockdown or knockout of LSD1 affected the 4SC-202-induced effects. Since time-lapse microscopy revealed that 4SC-202 could affect mitotic spindle formation, we performed an in vitro tubulin polymerization assay revealing that 4SC-202 can directly inhibit microtubule formation.
Conclusions
We demonstrate that 4SC-202, a drug currently tested in clinical trials, effectively inhibits growth of CTCL cells. The anti-cancer cell activity of 4SC-202 is however not limited to LSD1-inhibition, modulation of histone modifications, and consecutive alteration of gene expression. Indeed, the compound is also a potent microtubule-destabilizing agent.
Kann ein gesunder, körperlich unversehrter Mensch einen längeren Sterbeprozess durchleben? Und wenn dies so sein sollte, wie sieht die Sterbebegleitung in einem solchen Fall aus? Anhand des vier Monate währenden Briefwechsels des Ehepaares von Moltke vor und nach der Verurteilung des Grafen durch den Volksgerichtshof im Januar 1945 werden das Sterben im psychologischen Sinne und die Begleitung des Sterbenden ausführlich dargestellt. Auch die Situation der Ehefrau als Sterbebegleiterin wird beleuchtet. Es ergeben sich vielfältige Gemeinsamkeiten zwischen dem Sterben infolge einer Krankheit einerseits und den gedanklichen und emotionalen Anpassungsvorgängen in Erwartung der eigenen Hinrichtung andererseits. Sterben im psychologischen Sinne erweist sich als äußerst bewegtes Leben.
The aim of this thesis was the application of the functional prepolymer NCO-sP(EO-stat-PO) for the development of new biomaterials. First, the influence of the star-shaped polymers on the mechanical properties of biocements and bone adhesives was investigated. 3-armed star-shaped macromers were used as an additive for a mineral bone cement, and the influence on the mechanical properties was studied. Additionally, a previously developed bone adhesive was examined regarding cytocompatibility. The second topic was the examination of novel functionalization steps which were performed on the surface of electrospun fibers modified with NCO-sP(EO-stat-PO). This established method of functionalizing electrospun meshes was advanced regarding the modification with proteins which was then demonstrated in a biological application. Two different kinds of antibodies were immobilized on the fiber surface in a consecutive manner and the influence of these proteins on the cell behavior was investigated. The final topic involved the quantification of surface-bound peptide sequences. By functionalization of the peptides with the UV-reactive molecule 2-mercaptopyridine it was possible to quantify this compound via UV measurements by cleavage of disulfide bridges and indirectly draw conclusions about the number of immobilized peptides.
In the field of mineral biocements and bone adhesives, NCO-sP(EO-stat-PO) was able to influence the setting behavior and mechanical performance of mineral bone cements based on calcium phosphate chemistry. The addition of NCO-sP(EO-stat-PO) resulted in a pseudo-ductile fracture behavior due to the formation of a hydrogel network in the cement, which was then mineralized by nanosized hydroxyapatite crystals following cement setting. Accordingly, a commercially available aluminum silicate cement from civil engineering could be modified.
In addition, it could be shown that the use of NCO-sP(EO-stat-PO) is beneficial for adjusting specific material properties of bone adhesives. Here, the crosslinking behavior of the prepolymer in an aqueous medium was exploited to form an interpenetrating network (IPN) together with a photochemically curing poly(ethylene glycol) dimethacrylate (PEGDMA) matrix. This could be used for the development of a bone adhesive with an improved adhesion to bone in a wet environment. The developed bone adhesive was further investigated in terms of possible influences of the initiator systems. In addition, the material system was tested for cytocompatibility by using different cell lines.
Moreover, the preparation of electrospun fiber meshes via solution electrospinning consisting of poly(lactide-co-glycolide) (PLGA) as a backbone polymer and NCO-sP(EO-stat-PO) as functional additive is an established method for the application of the meshes as a replacement of the native extracellular matrix (ECM). In general, these fibers reveal diameters in the nanometer range, are protein and cell repellent due to the hydrophilic properties of the prepolymer and show a specific biofunctionalization by immobilization of peptide sequences. Here, the isocyanate groups presented on the fiber surface after electrospinning were used to carry out various functionalization steps, while retaining the properties of protein and cell repellency. The modification of the electrospun fibers involved the immobilization of analogs or antagonists of tumor necrosis factor (TNF) and the indirect detection of these by interaction with a light-producing enzyme. Here, a multimodal modification of the fiber surface with RGD to mediate cell adhesion and two different antibodies could be achieved. After culturing the cell line HT1080, the pro- or anti-inflammatory response of cells could be detected by IL-8 specific ELISA measurements.
Furthermore, the quantification of molecules on the surface of electrospun fibers was investigated. It was tested whether the detection by means of super-resolution microscopy would be possible. Therefore, experiments were performed with short amino acid sequences such as RGD for quantification by fluorescence microscopy. Based on earlier results, in which a UV-spectrometrically active molecule was used to detect the quantification of RGD, it was shown that short peptides can also be quantified in a small scale on flat functional substrates (2D) such as NCO-sP(EO-stat-PO) hydrogel coatings, and modified electrospun fibers produced from PLGA and NCO-sP(EO-stat-PO) (3D). In addition, a collagen sequence was used to prove that a successful quantification can be carried out as well for longer peptide chains.
These studies have revealed that NCO-sP(EO-stat-PO) can serve as a functional additive for many applications and should be considered for further studies on the development of novel biomaterials. The rapid crosslinking reaction, the resulting hydrogel formation and the biocompatibility are to be mentioned as positive properties, which makes the prepolymer interesting for future applications.
LTD-artige zentralmotorische Plastizität im Schubereignis bei Patienten mit Multipler Sklerose

Die Multiple Sklerose ist eine chronisch entzündliche Erkrankung des zentralen Nervensystems. Durch ein komplexes Zusammenspiel von Genetik, Autoimmunvorgängen und proinflammatorischen Prozessen kommt es zur Demyelinisierung sowie zu axonalen Schäden und kortikalen Läsionen (Calabrese et al., 2010; Ciccarelli et al., 2014; International Multiple Sclerosis Genetics et al., 2011; Leray et al., 2015). In den Industrieländern ist diese Erkrankung eine der häufigsten Ursachen für langfristige Behinderung bereits im frühen Lebensalter (Flores-Alvarado, Gabriel-
46 Ortiz, Pacheco-Mois, & Bitzer-Quintero, 2015). Die Diskrepanz allerdings zwischen klinischer Symptomatik und den Befunden der Bildgebung (Barkhof, 2002) gibt Anlass dafür, Adaptionsmöglichkeiten detailliert zu erforschen. Vorgänge der Neuroplastizität mit LTP und LTD als Basismechanismen erscheinen dabei zunehmend Beachtung zu finden (Dayan & Cohen, 2011; Zeller et al., 2011). Welche Rolle diese Prozesse allerdings im akuten Schub, während der häufig stark ausgeprägten Symptomatik, insbesondere aber auch während ihrer Rückbildung spielen, bleibt bisher weitgehend ungeklärt. Eine Untersuchung zu stimulationsinduzierter LTP-artiger Plastizität im Schub deutete auf einen möglichen Zusammenhang zwischen Ausmaß der Symptomrückbildung und PAS25-induziertem LTP-Effekt hin (Mori et al., 2014).
In der vorliegenden Arbeit wurde komplementär hierzu die stimulationsinduzierte LTD-artige Plastizität bei 19 MS- bzw. CIS-Patienten während des steroidbehandelten akuten Schubes untersucht. Als Kontrollgruppe wurden alters- und geschlechtsgematchte gesunde Probanden untersucht. Die Messungen wurden mithilfe eines Protokolls der assoziativen Paarstimulation durchgeführt. Paarstimulation wird die Kombination aus der peripher elektrischen und transkraniell magnetischen Stimulation genannt. Das in unserer Studie verwendete Protokoll sieht ein Interstimulusintervall von 10ms vor (PAS10). Der Effekt der Paarstimulation wird durch Messungen der Exzitabilität des motorischen Kortex mittels motorisch evozierter Potenziale (MEP) jeweils vor und nach der Intervention gemessen. Bei den MS-Patienten wurden diese Daten zum Zeitpunkt des Schubes (t1) und 12 Wochen danach (t2) erhoben; die gesunden Kontrollen wurden nur einmal gemessen. Daneben wurde bei den Schubpatienten zur Quantifizierung der klinischen Symptomatik jeweils zum ersten und zum zweiten Zeitpunkt der MSFC erhoben.
Die MS-Patienten zeigten im akuten MS-Schub im Gegensatz zu der Kontrollgruppe aus Gesunden keinen LTD-artigen, sondern einen inversen, sprich einen signifikant LTP-artigen Effekt; dieser war zum Zeitpunkt t2 nicht mehr zu erkennen. Der Unterschied zwischen den PAS10-Effekten der MS- und der Kontrollgruppe war ebenfalls signifikant. Der Vergleich der MSFC-Werte der MS-Gruppe zwischen t1 und t2 erbrachte eine signifikante klinische Besserung. Eine signifikante Korrelation zwischen

den neurophysiologischen und klinischen Daten bzw. ihren Veränderungen zwischen t1 und t2 zeigte sich nicht.
Diese Ergebnisse untermauern und erweitern bereits bestehende Hinweise, dass während der akuten Inflammationsprozesse des MS-Schubes veränderte Voraussetzungen für die Induzierbarkeit von Plastizität gegeben sind. Nicht nur, wie bereits gezeigt, die LTP-artige, sondern offenbar auch die LTD-artige assoziative Plastizität zeigt sich stark von den humoralen Veränderungen im steroidbehandelten Schub beeinflusst. Weitere Studien in stärker vorselektierten Patientengruppen sollten der Frage nachgehen, inwieweit LTD-artige Plastizität sich in verschiedenen Subgruppen mit unterschiedlichen Schubsymptomen unterscheidet. Des Weiteren ist der Frage weiter nachzugehen, ob LTD-artige Plastizität funktional zur Adaption im Rahmen des Schubereignisses notwendig ist und inwieweit deren Unterdrückung bzw. Ersatz durch Langzeitpotenzierung potenziell einer Adaption im Wege steht. Sollten potenzielle Folgestudien bestätigen, dass LTD- und LTP-artige Plastizität im Schub möglicherweise häufig dysfunktional ausgeprägt ist und einer optimalen Regeneration entgegensteht, wären daraus praktische Implikationen zu ziehen. Die Entwicklung neuer Trainingsprogramme oder elektrophysiologischer Konzepte könnte ein nächstes Ziel dieses Forschungszweiges sein, um potenziell dysfunktionale Plastizität zu vermeiden und physiologische Prozesse bereits im Schub zu fördern.
The detection of toxic gases, such as NH\(_{3}\) and CO, in the environment is of high interest in chemical, electronic, and automotive industry as even small amounts can display a health risk for workers. Sensors for the real‐time monitoring of these gases should be simple, robust, reversible, highly sensitive, inexpensive and show a fast response. The indicator supraparticles presented herein can fulfill all of these requirements. They consist of silica nanoparticles, which are assembled to supraparticles upon spray‐drying. Sensing molecules such as Reichardt's dye and a binuclear rhodium complex are loaded onto the microparticles to target NH\(_{3}\) and CO detection, respectively. The spray‐drying technique affords high flexibility in primary nanoparticle size selection and thus, easy adjustment of the porosity and specific surface area of the obtained micrometer‐sized supraparticles. This ultimately enables the fine‐tuning of the sensor sensitivity and response. For the application of the indicator supraparticles in a gas detection device, they can be immobilized on a coating. Due to their microscale size, they are large enough to poke out of thin coating layers, thus guaranteeing their gas accessibility, while being small enough to be applicable to flexible substrates.
Purpose
4D flow cardiovascular magnetic resonance (CMR) and the assessment of wall shear stress (WSS) are non-invasive tools to study cardiovascular risks in vivo. Major limitations of conventional triggered methods are the long measurement times needed for high-resolution data sets and the necessity of stable electrocardiographic (ECG) triggering. In this work an ECG-free retrospectively synchronized method is presented that enables accelerated high-resolution measurements of 4D flow and WSS in the aortic arch of mice.
Methods
4D flow and WSS were measured in the aortic arch of 12-week-old wildtype C57BL/6 J mice (n = 7) with a radial 4D-phase-contrast (PC)-CMR sequence, which was validated in a flow phantom. Cardiac and respiratory motion signals were extracted from the radial CMR signal and were used for the reconstruction of 4D-flow data. Rigid motion correction and a first order B0 correction was used to improve the robustness of magnitude and velocity data.
The aortic lumen was segmented semi-automatically. Temporally averaged and time-resolved WSS and oscillatory shear index (OSI) were calculated from the spatial velocity gradients at the lumen surface at 14 locations along the aortic arch. Reproducibility was tested in 3 animals and the influence of subsampling was investigated.
Results
Volume flow, cross-sectional areas, WSS and the OSI were determined in a measurement time of only 32 min. Longitudinal and circumferential WSS and radial stress were assessed at 14 analysis planes along the aortic arch. The average longitudinal, circumferential and radial stress values were 1.52 ± 0.29 N/m2, 0.28 ± 0.24 N/m2 and − 0.21 ± 0.19 N/m2, respectively. Good reproducibility of WSS values was observed.
Conclusion
This work presents a robust measurement of 4D flow and WSS in mice without the need of ECG trigger signals. The retrospective approach provides fast flow quantification within 35 min and a flexible reconstruction framework.
Im Rahmen dieser Arbeit konnte der erste metallfreie, an Raumtemperatur ablaufende Wasserstofftransfer von Amminboran (NH3BH3) auf Iminoborane durchgeführt werden. Bei der Umsetzung von NH3BH3 mit Di tert butyliminoboran XXIIIa wurde die Bildung des hydrierten Iminoborans 1 beobachtet. DFT Rechnungen belegen einen konzertierten H+/H- Transfer von NH3BH3 mit einer Aktivierungsenergie weit unter der vergleichbarer Wasserstoffübertragungsreaktionen. Da trotz zahlreicher Versuche keine geeigneten Einkristalle von 1 für eine strukturelle Charakterisierung erhalten werden konnten, wurde 1 mit IPr bzw. etherischer HCl umgesetzt, wobei das Addukt 2 bzw. das 1,2 Additionsprodukt 3 isoliert und vollständig charakterisiert werden konnte.
Darüber hinaus konnten weitere sterisch anspruchsvolle disubstituierte Amminborane dargestellt werden, die ebenfalls die Fähigkeit besitzen, das Di tert butyliminoboran XXIIIa zu hydrieren. Dafür wurden die Arylamminborane (Aryl = Dur (4) & Tip (5)) dargestellt und mit XXIIIa umgesetzt. Hierbei bildeten sich neben 1 die entsprechenden Arylaminoborane 6 und 7.
Ein weiteres Ziel war die Synthese neuartiger Iminoborane. Dafür wurden zwei ferrocenylsubstituierte Aminoboranpräkursoren dargestellt (Schema 53). Über eine Chlorsilan Eliminierung sollte aus Verbindung 8 das entsprechende Iminoboran 9 synthetisiert werden. Jedoch zeigten sich in 11B NMR spektroskopischen Untersuchungen nur Zersetzungsprodukte. Des Weiteren wurde versucht, eine HCl Abstraktion mit Hilfe von verschiedenen Abstrahierungs-Reagenzien, ausgehend von dem Aminoboranpräkursor 10, zu initiieren. Diesbezüglich wird 10 mit diversen Substraten (Na[BAr4F], Na[BAr4Cl], NaH, LiTmp und Na[HMDS]) umgesetzt, wobei sich allerdings für keine der eingesetzten Reagenzien die gewünschte HCl Eliminierung zeigte.
Ferner wurde der Aminoboranpräkursor 1,4-Bis{[tert-butyl(trimethylsilyl)amino]-chlorboranyl}durylen 14 erfolgreich dargestellt. Allerdings verlief die Chlorsilan-Eliminierung zu dem Bis-(tert-butylimino)durylenboran 15 im Pyrolyseofen erfolglos.
Weitere Syntheseversuche der Aminoborane 16, 17 und 19 als Präkursoren für das (tert-Butylimino) tris bzw. bis (trifluormethyl)phenylboran führten nicht zum gewünschten Ziel (Abbildung 29). Die Bildung dieser Aminoborane läuft unselektiv ab und es werden erhebliche Mengen unerwünschter Nebenprodukte beobachtet. Daher können keine analysenreinen Endprodukte isoliert werden.
Im Gegensatz dazu gelang die Darstellung des CF3 meta substituierten Präkursors 21. Durch anschließende Chlorsilan-Eliminierung konnte das (tert Butylimino) 3,5 bis(trifluormethyl)phenylboran 22 isoliert werden. Als Oligomerisierungsprodukt von 22 wurde das Dewar Borazin-Derivat 23 nachgewiesen.
Weiterhin wurde der Aminoboranpräkursor Trietylphenyl[tert-butyl(trimethylsilyl)amino]chlorboran 24 erfolgreich dargestellt (Schema 56). Nach anschließender Chlorsilaneliminierung im Pyrolyseofen konnte die Bildung des (tert Butylimino)-2,4,6-triethylphenylborans 25 beobachtet werden. Durch Erwärmen von 24 für zwei Stunden auf 100 °C wurde das Dimerisierungsprodukt 26 detektiert.
Die Umsetzung des Iminoborans 25 mit cAAC lieferte bei tiefen Temperaturen das Addukt 28. Erwärmen des Reaktionsgemisches auf Raumtemperatur führte zur Bildung des Cyclopropan Intermediats 30. Ferner ergab das Erhitzen auf 50 °C unter intramolekularem Ringschluss das bicyclische 1,2-Azaborolidin 31.
Außerdem wurde das Azaborinin 27 über eine metallkatalysierte Syntheseroute, ausgehend von dem Iminoboran 25 und dem Rhodiumkatalysator [{RhCl(PiPr3)2}2] LXV, unter Acetylenatmosphäre dargestellt.
Angesichts weiterer Reaktivitätsstudien von Iminoboranen gegenüber Lewissäuren, wurde das Di tert butyliminoboran XXIIIa mit Tris(pentafluorphenyl)boran umgesetzt (Schema 59). Bei der Reaktion findet eine Carboborierung statt; demgemäß handelt es sich um eine 1,2 Addition des Borans an das Iminoboran unter Bor-Kohlenstoff Bindungspaltung. DFT Rechnungen zufolge beträgt die freie Energie der Reaktion -17.4 kcal mol-1 und läuft damit exergonisch ab.
Des Weiteren konnte mit dem NHC IDip (1,3-Bis(2,6-diisopropylphenyl)imidazol-2-yliden) in Reaktivitätsstudien mit (tert Butylimino)mesitylboran XXIIIb die Bildung des Aminoborans 34 beobachtet werden. Dieses entsteht durch die Migration eines Dipp Restes des NHC´s zum Stickstoffatom des vormaligen Iminoborans. Dieser Reaktionstyp stellt ein neues Beispiel für die Reaktivität zwischen Iminoboranen und NHCs im Gegensatz zu klassischen Adduktformationen dar.
Einen weiteren neuartigen Beitrag zur Iminoboranchemie lieferte die Reaktion von Di tert butyliminoboran XXXII und dem von Tacke et al. entwickelten Bis(amidinato)silylen XCVI. Bei dem Reaktionsprodukt handelt es sich um eine bicyclische Silaiminboranstruktur, welche aus der Insertion von XCVI in die BN Dreifachbindung des Iminoborans hervorgeht. DFT Rechnungen bestätigen eine exergonische Reaktion mit einer Gesamtenergie von -28.4 kcal mol-1. Eine Insertion in eine Bor-Stickstoff-Dreifachbindung ist unter den Reaktivitätsstudien mit Iminoboranen ein seltener Reaktionstyp.
Überdies wurde die Reaktion zwischen dem Pentafluorphenyl-(tert-butyl)iminoboran (XCVII) und PiPr3 beobachtet. Dabei fand formal eine CH Aktivierung der drei Isopropyleinheiten von PiPr3 mit den Boratomen von drei XCVII-Einheiten statt. Zusätzlich bindet ein Sauerstoff an das Phosphoratom. Die Ursache dieser Beobachtung und der Mechanismus dieser Reaktion ist bislang nicht geklärt.
Bei der Implantatversorgung von Patienten mit Osteoporose besteht weiterhin eine hohe Komplikationsrate vor allem durch aseptische Prothesenlockerungen. Eine vielversprechende Möglichkeit diese zu minimieren stellt eine Funktionalisierung der Implantate mit Strontium dar.
Ziel der vorliegenden Arbeit war es dabei die Wirkung lokal verfügbaren Strontiums auf osteoklastäre und osteoblastäre Zellen zu untersuchen.
Mittels elektrochemischer Abscheidung erfolgte die Beschichtung von Titanproben mit strontiumdotiertem Struvit, wobei sieben verschiedene Dotierkonzentrationen zwischen 6 µg und 487 µg Strontium pro Probe hergestellt wurden. Die Untersuchungen an osteoklastären RAW 264.7 Zellen erfolgten mittels Bestimmung von Zellzahl und -aktivität, verschiedener mikroskopischer Methoden sowie auf genetischer Ebene. Osteoblastäre MG63-Zellen wurden orientierend anhand von Zellzahl und Zellaktivität untersucht.
Zellbiologisch konnte ein hemmender Einfluss von Strontium auf Differenzierung sowie Proliferation und Aktivität osteoklastärer Zellen gezeigt werden. Die Dotierkonzentration mit den günstigsten Eigenschaften war unter vorliegenden Versuchsbedingungen 487 µg Strontium pro Probe, da sich hierbei zudem eine erhaltene ostoblastäre Proliferation und Aktivität zeigte.
Die Knieendoprothetik ist ein etabliertes Verfahren zur Behandlung der symptomatischen Gonarthose, mit welchem gute Ergebnisse erzielt werden können. Dennoch kommt es in 10-20% zu einer Beschwerdepersistenz und die aseptische Lockerung der Prothesen stellt weiterhin ein Problem dar.
Als mögliche Ursache hierfür wird die unzureichend abgebildete Anatomie und Biomechanik diskutiert.
Die konvex gekrümmten Femurkondylen sind nicht symmetrisch. Die Krümmungsradien nehmen von dorsal nach ventral zu. Zudem ist die mediale Gelenkfläche des Tibiaplateaus nach kranial konkav und die laterale nach kranial konvex zulaufend. Die Instabilität wird durch das vordere Kreuzband gesichert. Bei der Bewegung des Kniegelenkes kommt es zu einer Roll/Gleitbewegung zwischen Femur und Tibia. Bisher gibt es nur wenige Knieendoprothesen, die das natürliche Roll-Gleit-Verhalten des natürlichen Kniegelenkes nachempfinden. Die Aequos G1 Endoprothese berücksichtigt die Besonderheiten (mediolaterale Asymmetrie) und soll so die natürliche Kinematik besser nachempfinden.
Ziel dieser Arbeit war es, Daten vom klinischen und radiologischen Ergebnis nach Implantation der Prothese innerhalb eines Jahres zu erheben. Es wurde Wert gelegt auf die Erfassung von funktionellem Outcome, gesundheitsbezogener Lebensqualität und Probleme der Patella wie dem vorderen Knieschmerz.
Im Zeitraum von 2010 bis 2011 wurden 26 Patienten in einer prospektiven, klinischen Verlaufsstudie erfasst. Es erfolgte eine Untersuchung präoperativ, eine vor Entlassung aus dem Krankenhaus und nach 6 und 12 Monaten. Die Patienten wurden mittels standardisierter Fragebögen befragt und Daten erhoben zu Knee Society Score (KSS), Oxford Knee Score (OKS), Visuelle Analogskala Schmerz (VAS), Kujala- Score, Turba- Score und SF-36. Zudem erfolgten eine klinische Untersuchung und eine standardisierte radiologische Diagnostik.
Das mittlere Alter der Patienten zum Zeitpunkt der Versorgung lag bei 68,77 Jahren (Min. 55 – Max. 83). Der Frauenanteil lag bei 76,92%, der durchschnittliche BMI betrug 31,48kg/m². Insgesamt konnten 22 Patienten nach 12 Monaten inkludiert werden. Eine Patientin war zum Zeitpunkt der Nachuntersuchung verstorben, eine Patientin erschien nicht zur Nachuntersuchung nach 6 Monaten, zwei Patienten erhielten einen sekundären Retropatellarersatz und schieden aus der Studie aus. Bei einer Patientin zeigte sich 12 Monate postoperativ eine Lockerung des Tibiaplateaus, es erfolgte ein Wechsel der tibialen Komponente. Eine Patientin klagte nach 6 Monaten überstarke, wirbelsäulenbedingte Beinschmerzen.
Der mittlere Bewegungsumfang besserte sich nicht signifikant. Er betrug präoperativ 106,68° (±22,86°) und nach 12 Monaten 113,18° (±8,67°). Sowohl die VAS, als auch der KSS, der OKS und der Kujala-Score waren 12 Monate postoperativ hochsignifikant besser als präoperativ. Die berechnete körperliche Summenskala des SF-36 war ebenfalls hoch signifikant besser als präoperativ. Die Daten der psychischen Summenskala waren signifikant besser als präoperativ.
Der subjektive Teil des Turba-Scores war hoch signifikant besser als präoperativ. Im objektiven Teil zeigte sich keine signifikante Besserung.
Insgesamt zeigten sich gute postoperative Ergebnisse nach Implantation der Aequos G1 Endoprothese nach einem Jahr mit signifikanter Verbesserung der Funktions- und Schmerzscores sowie der individuellen Gesundheitswahrnehmung. Im Vergleich mit den Ergebnissen der verfügbaren Literatur, scheint die Prothese nicht unterlegen.
Abzuwarten bleibt, ob sich die guten Ergebnisse langfristig bestätigen.
Quantencomputer können manche Probleme deutlich effizienter lösen als klassische Rechner. Bisherige Umsetzungen leiden jedoch an einer zu geringen Dekohärenzzeit, weshalb die Lebenszeit der Quantenzustände einen limitierenden Faktor darstellt. Topologisch geschützte Anregungen, wie Majorana-Fermionen, könnten hingegen dieses Hindernis überwinden. Diese lassen sich beispielsweise in topologischen Supraleitern realisieren. Bis zum jetzigen Zeitpunkt existieren nur wenige Materialien, die dieses Phänomen aufweisen. Daher ist das Verständnis der elektronischen Eigenschaften für solche Verbindungen von großer Bedeutung.
In dieser Dissertation wird die Koexistenz von Supraleitung an der Probenoberfläche und topologischem Oberflächenzustand (engl. topological surface state, TSS) auf potentiellen topologischen Supraleitern überprüft. Diese beiden Bedingungen sind essentiell zur Ausbildung von topologischer Supraleitung in zeitumkehrgeschützten Systemen. Hierzu wird mittels Landaulevelspektroskopie und Quasiteilcheninterferenz das Vorhandensein des TSS am Ferminiveau auf Tl$_{x}$Bi$_{2}$Te$_{3}$ und Nb$_{x}$Bi$_{2}$Se$_{3}$ verifiziert, die mittels Transportmessungen als supraleitend identifiziert wurden. Anschließend folgen hochaufgelöste Spektroskopien an der Fermienergie, um die supraleitenden Eigenschaften zu analysieren.
Zur Interpretation der analysierten Eigenschaften wird zu Beginn der Ni-haltige Schwere-Fermion-Supraleiter TlNi$_{2}$Se$_{2}$ untersucht, der eine vergleichbare Übergangstemperatur besitzt. Anhand diesem werden die gängigen Messmethoden der Rastertunnelmikroskopie und -spektroskopie für supraleitende Proben vorgestellt und die Leistungsfähigkeit der Messapparatur demonstriert. Im Einklang mit der Literatur zeigt sich ein $s$-Wellencharakter des Paarungsmechanismus sowie die Formation eines für Typ~II-Supraleiter typischen Abrikosov-Gitters in schwachen externen Magnetfeldern.
Im folgenden Teil werden die potentiellen topologischen Supraleiter Tl$_{x}$Bi$_{2}$Te$_{3}$ und Nb$_{x}$Bi$_{2}$Se$_{3}$ begutachtet, für die eindeutig ein TSS bestätigt wird. Allerdings weisen beide Materialien keine Oberflächensupraleitung auf, was vermutlich durch eine Entkopplung der Oberfläche vom Volumen durch Bandverbiegung zu erklären ist. Unbeabsichtigte Kollisionen der Spitze mit der Probe führen jedoch zu supraleitenden Spitzen, die wesentlich erhöhte Werte für die kritische Temperatur und das kritische Feld zeigen.
Der letzte Abschnitt widmet sich dem supraleitenden Substrat Nb(110), für den der Reinigungsprozess erläutert wird. Hierbei sind kurze Heizschritte bis nahe des Schmelzpunktes nötig, um die bei Umgebungsbedingungen entstehende Sauerstoffrekonstruktion effektiv zu entfernen. Des Weiteren werden die elektronischen Eigenschaften untersucht, die eine Oberflächenresonanz zum Vorschein bringen. Hochaufgelöste Messungen lassen eine durch die BCS-Theorie gut repräsentierte Struktur der supraleitenden Energielücke erkennen. Magnetfeldabhängige Experimente offenbaren zudem eine mit der Kristallstruktur vereinbare Anisotropie des Paarungspotentials. Mit diesen Erkenntnissen kann Nb(110) zukünftig als Ausgang für das Wachstum von topologischen Supraleitern herangezogen werden.
Die verfügbaren in vitro Genotoxizitätstests weisen hinsichtlich ihrer Spezifität und ihres Informationsgehalts zum vorliegenden Wirkmechanismus (Mode of Action, MoA) Einschränkungen auf. Um diese Mängel zu überwinden, wurden in dieser Arbeit zwei Ziele verfolgt, die zu der Entwicklung und Etablierung neuer in vitro Methoden zur Prüfung auf Genotoxizität in der Arzneimittelentwicklung beitragen.
1. Etablierung und Bewertung einer neuen in vitro Genotoxizitätsmethode (MultiFlow Methode)
Die MultiFlow Methode basiert auf DNA-schadensassoziierten Proteinantworten von γH2AX (DNA-Doppelstrangbrüche), phosphorylierten H3 (S10) (mitotische Zellen), nukleären Protein p53 (Genotoxizität) und cleaved PARP1 (Apoptose) in TK6-Zellen. Insgesamt wurden 31 Modellsubstanzen mit dem MultiFlow Assay und ergänzend mit dem etablierten Mikrokerntest (MicroFlow MNT), auf ihre Fähigkeit verschiedene MoA-Gruppen (Aneugene/Klastogene/Nicht-Genotoxine) zu differenzieren, untersucht. Die Performance der „neuen“ gegenüber der „alten“ Methode führte zu einer verbesserten Sensitivität von 95% gegenüber 90%, Spezifität von 90% gegenüber 72% und einer MoA-Klassifizierungsrate von 85% gegenüber 45% (Aneugen vs. Klastogen).
2. Identifizierung mechanistischer Biomarker zur Klassifizierung genotoxischer Substanzen
Die Analyse 67 ausgewählter DNA-schadensassoziierter Gene in der QuantiGene Plex Methode zeigte, dass mehrere Gene gleichzeitig zur MoA-Klassifizierung beitragen können. Die Kombination der höchstrangierten Marker BIK, KIF20A, TP53I3, DDB2 und OGG1 ermöglichte die beste Identifizierungsrate der Modellsubstanzen. Das synergetische Modell kategorisierte 16 von 16 Substanzen korrekt in Aneugene, Klastogene und Nicht-Genotoxine. Unter Verwendung der Leave-One-Out-Kreuzvalidierung wurde das Modell evaluiert und erreichte eine Sensitivität, Spezifität und Prädiktivität von 86%, 83% und 85%. Ergebnisse der traditionellen qPCR Methode zeigten, dass Genotoxizität mit TP53I3, Klastogenität mit ATR und RAD17 und oxidativer Stress mit NFE2L2 detektiert werden kann.
Durch die Untersuchungen von posttranslationalen Modifikationen unter Verwendung der High-Content-Imaging-Technologie wurden mechanistische Assoziationen für BubR1 (S670) und pH3 (S28) mit Aneugenität, 53BP1 (S1778) und FANCD2 (S1404) mit Klastogenität, p53 (K373) mit Genotoxizität und Nrf2 (S40) mit oxidativem Stress identifiziert.
Diese Arbeit zeigt, dass (Geno)toxine unterschiedliche Gen- und Proteinveränderungen in TK6-Zellen induzieren, die zur Erfassung mechanistischer Aktivitäten und Einteilung (geno)toxischer MoA-Gruppen (Aneugen/Klastogen/ Reaktive Sauerstoffspezies) eingesetzt werden können und daher eine bessere Risikobewertung von Wirkstoffkandidaten ermöglichen.
In Tissue Engineering, scaffolds composed of natural polymers often show a distinct lack in stability. The natural polymer gelatin is highly fragile under physiological conditions, nevertheless displaying a broad variety of favorable properties. The aim of this study was to fabricate electrospun gelatin nanofibers, in situ functionalized and stabilized during the spinning process with highly reactive star polymer NCO-sP(EO-stat-PO) (“sPEG”). A spinning protocol for homogenous, non-beaded, 500 to 1000 nm thick nanofibers from different ratios of gelatin and sPEG was successfully established. Fibers were subsequently characterized and tested with SEM imaging, tensile tests, water incubation, FTIR, EDX, and cell culture. It was shown that adding sPEG during the spinning process leads to an increase in visible fiber crosslinking, mechanical stability, and stability in water. The nanofibers were further shown to be biocompatible in cell culture with RAW 264.7 macrophages.
