Typisch für die Alzheimer' schen Erkrankung ist die Bildung unlöslicher Ablagerungen im Gehirn, sogenannter "seniler Plaques". Diese Plaques bestehen im Wesentlichen aus fibrillärem beta-Amyloid, das durch Glykierungen verändert vorliegen kann. Außerdem beinhalten die Plaques, sogenannte AGEs "Advanced Glycation Endproducts", die aus nichtenzymatisch glykierten Proteinen entstehen. Diese AGE-modifizierten Proteine sowie das fibrilläre beta-Amyloid sind in der Lage Mikrogliazellen zu aktivieren. Die sessilen Gehirnmakrophagen wirken in aktiviertem Zustand neurotoxisch, wobei es verschiedene Hypothesen gibt, wie die Mikrogliazellen zu dem neuronalen Zelltod führen. Um dieses zu untersuchen wurden murine Mikrogliazellen herangezogen, die als Merkmal ihrer Aktivierung auf die Translokation des Transkriptionsfaktors NF-kappa-B in den Zellkern überprüft wurden. In der vorliegenden Arbeit wurden die Rahmenbedingungen näher untersucht, die zu der AGE vermittelten Mikrogliaaktivierung führen. Es wurde in vitro gezeigt, daß die Mikrogliaaktivierung zunächst durch eine hochmolekulare Hyaluronsäure, wie sie nativ in der extrazellulären Matrix vorliegt, verhindert wird. Im Gegensatz dazu konnte NF-kappa-B in Mikrogliazellen aktiviert werden, die in Gegenwart von Hyaluronsäurefragmenten mit AGE behandelt wurden. In der vorliegenden Arbeit wurde festgestellt, daß die Mikrogliaaktivierbarkeit umgekehrt proportional zu der durchschnittlichen Hyaluronsäuremolekülgröße ist. Andere Glykosaminoglykane aus der extrazellulären Matrix, wie D-Glukuronsäure, N-Azetylglukosamin oder Chondroitin-4-sulfat reduzierten die Aktivierbarkeit der Mikrogliazellen nur geringfügig. Sowohl beta-Amyloid, als auch AGEs setzen während ihres Entstehungsprozesses reaktive Sauerstoffspezies frei, die Hyaluronsäure in kleinere Bruchstücke zerschneiden können. Die Signaltransduktion der AGE-aktivierten Mikrogliazellen wurde mittels unterschiedlicher Inhibitoren gehemmt und die Auswirkung auf die NF-kappa-B Aktivierung untersucht. Hier zeigte sich ein komplexes Netzwerk an aktivierten Signalwegen, so daß kein Rückschluß auf einen bestimmten Rezeptor möglich war. Daher wurde ein "in vitro Modell" entwickelt, um die ausschlaggebende neurotoxischen Komponenten der Mikrogliareaktion aufzufinden. Darin wurden die Signalkaskaden der aktivierten Mikroglia erneut durch pharmakologische Inhibierung unterbrochen, das zellfreie Medium das von diesen Mikrogliazellen sezerniert wurde, wurde als "konditioniertes Medium" für die Kultur muriner Neuronen eingesetzt. Diese wurden bezüglich ihrer Überlebensrate in diesem konditionierten Medium untersucht. Die Hemmung der Transkription oder der Translation in den Mikrogliazellen zeigte keine Reduktion der neurotoxischen Wirkung des konditionierten Mediums. Ebensowenig wirkten Inhibitoren der mitochondrialen Atmungskette, der Radikalquellen Xanthin Oxidase, Lipoxygenase oder Cyclooxygenase. Die Hemmung der NADPH Oxidase reduzierte die Neurotoxizität des konditionierten Mediums auf etwa 30 Prozent. Die NADPH Oxidase ist ein Enzymkomplex, der im Rahmen des "oxidativen bursts" große Mengen Superoxidanionen freisetzt. Um die Bedeutung der NADPH Oxidase Aktivierung für die neurotoxische Wirkung nachzuweisen, wurde eine Untereinheit der NADPH Oxidase, das membranständige gp91phox in den Mikrogliazellen deaktiviert. Dies führte dazu, daß diese Zellen kein Superoxid auf die Stimulation mit beta-Amyloid oder AGE hin abgaben, im Gegensatz zu den Mikrogliazellen mit funktioneller NADPH Oxidase. Das konditionierte Medium der NADPH Oxidase defizienten Zellen war nicht mehr neurotoxisch. Die freien Sauerstoffradikale die aufgrund der NADPH Oxidase Aktivierung entstehen, können zu einer NF-kappa-B Aktivierung führen. NF-kappa-B wurde erfolgreich in den Mikroglia durch exogenes Wasserstoffperoxid stimuliert, wobei aber keine neurotoxische Wirkung im Modellsystem festgestellt wurde. NF-kappa-B scheint damit nicht für die mikrogliavermittelte Neurotoxizität verantwortlich zu sein, im Gegensatz zu der NADPH Oxidase, deren Aktivität unmittelbar mit der Neurotoxizität korreliert ist.
Die Pathomechanismen der Neuropathie bei Neuroborreliose sind noch immer unklar. In der vorliegenden Studie untersuchten wir 22 Patienten mit einer Neuropathie bei Neuroborreliose (BN) (3 Patienten in Stadium 2, 19 Patienten in Stadium 3) und verglichen diese mit 9 Patienten einer vaskulitischen Neuropathie (VN) und 14 Patienten einer idiopathisch axonalen Neuropathie (AN). Histologische und immunhistochemische Färbungen wurden mit Antikörpern gegen Leu4, CD68, 27E10 (frühzeitig aktivierte Makrophagen) und 25F9 (späte Makrophagen), Membrane-Attack-Komplex C5b-9, Adhäsionsmolekül ICAM sowie inflammatorische Zytokine Tumor Nekrose-Faktor-alpha (TNF-alpha), Interleukin-1ß (IL-1ß) und Interleukin-6 (IL-6), als auch Metalloproteinase MMP-9 durchgeführt, ferner mit Antikörpern gegen das membranassoziierte Glykoprotein HLA-DR3. Zusätzlich wurden Semi-Dünn-Schnitte angefertigt. Klinisch wiesen die meisten BN Patienten eine distal symmetrische sensomotorische Neuropathie auf, nur 6/22 Patienten waren schmerzfrei. Die Mehrzahl (18/22) der BN Neuropathien waren primär axonal mit perivaskulären (6/22), in 8 Fällen vaskulitischen Infiltraten. Das Perineurium war schwerpunktmäßig im Rahmen einer Borrelien-assoziierten Neuropathie betroffen. Das lies sich aus dem gegenüber den Kontrollgruppen signifikant verdickten Perineurium, der vermehrten Vaskularisation des Perineuriums und der starken IR für das Zytokin TNF-α schliessen, in geringerem Ausmaß für IL-1β, und für die terminale Komplementkomponente C5b9. Perivaskuläre und vaskuläre Infiltrate sowie die betont perineurale Expression bestimmter inflammatorischer Zytokine und Adhäsionsmoleküle erschienen charakteristisch für eine Neuropathie bei Neuroborreliose. Autoimmune Reaktionen mit Angriff am Perineurium können für die Pathogenese der Neuropathie bei Neuroborreliose bedeutsam sein.
The prevention of restenosis after percutaneous coronary intervention is a major task for researchers and clinicians in cardiovascular pharmacology. Nearly 1.5 million PTCA are performed every year worldwide and, due to the implantation of stents, most of the cases can be treated successfully. 60% of those patients develop restenosis within 6 months. SMC migration and ECM deposition are known to be responsible for neointima formation. Among many processes, integrin initiated signalling events play a central role in SMC migration. Many integrins recognize a specific RGD sequence which is present in several ECM proteins and cell surface immunoglobulin super family molecules. Until now, there are various integrin antagonists such as antibodies, cyclic peptides, peptidomimetics, and non-peptides have been shown to interfere with such pathological situations indicating the importance of integrin initiated signalling pathways in SMC migration. Therefore, in this study SMC migration induced by ECM proteins was inhibited either using pharmacological inhibitor or by overexpressing the endogenous inhibitor of FAK by AAV vector system. In the first part of the thesis, the effect of integrin-ligand stimulation on hCASMCs was studied. The tyrosine phosphorylation of many cellular proteins was observed from serum starved hCASMCs replated on VN but not on PL coated plates. The major tyrosine phosphorylated protein was identified as FAK by immunoprecipitation and also phosphorylation was found at Tyr 397, the autophosphorylation site of FAK. Further, VN induced the dose dependent migration of hCASMCs in haptotaxis assay. The integrin v inhibitor was used to block those ECM stimulated integrin signalling pathways and cell migration. It inhibited the ECM stimulated tyrosine phosphorylation in a dose dependent manner. Interestingly, specific potent antagonism of integrin v abrogated both ECM induced haptotaxis and growth factor induced chemotaxis. The inhibition of migration is consistent with the replating assay results that show interference with integrin induced signalling pathways particularly the FAK tyrosine phosphorylation. The integrin v inhibitor also is able to interfere with hCASMC invasion through matrigel by reducing MMP-2 secretion. Importantly, integrin v inhibitor did not induce the apoptosis in hCASMCs. FAK is a key player in many cellular events and its involvement in cell migration was extensively studied in various cell types. The present study explored the function of FAK in hCASMC migration by overexpression of FRNK, the C-terminal domain of FAK. Overexpression of FRNK inhibited the in vitro SMC migration as well as the neointima formation in a porcine restenosis model in vivo. The last part of this thesis focused on the identification of putative binding partners for the N-terminal domain of FAK by bacterial two-hybrid screen. One of the interesting binding partners was a putative protein of 17.9 kDa. Its human homolog is AGS4, which acts as a GTPase activator. The preliminary results revealed that it is able to interact with N-FAK domain and its expression is high in haematopoietic cells. Taken together the above results suggest that integrin v and FAK are promising targets for inhibition of SMC migration. Disruption of FAK-mediated signalling pathways by a pharmacological inhibitor or by overexpression of FRNK, which acts as dominant-negative regulator, resulted in decreased migration of SMCs and thus can lead to reduction of neointima formation.
Integrins are transmembrane receptors transmitting mechanical signals from the extracellular matrix (ECM) to the cytoskeleton (outside-in-signaling). Many molecular defects in the link between cytoskeleton and ECM are known to induce cardiomyopathies. alpha v integrin appears to play a major role in several processes relevant to remodeling, such as binding and activation of matrix metalloproteinases as well as regulation of cell proliferation, migration, and differentiation. We hypothesized that alpha v integrin-mediated signaling is required for the compensatory hypertrophy after aortic banding (AB) and associated with the modulation of ECM protein expression. Mice were treated in vivo with a specific integrin alpha v inhibitor or vehicle via osmotic minipumps starting 1 day prior to aortic banding (AB). At day 2 and day 7 following AB or sham-operation, the mice were examined by echocardiography and hemodynamic analyses were performed. Treatment of alpha v Integrin inhibitor led to a dilated cardiomyopathy and congestive heart failure in AB mice (dilated left ventricle, depressed LV function, and pulmonary congestion), but not to hypertrophy as observed in mice without inhibitor treatment. Investigation of downstream signaling revealed significant activation of the p38 Mitogen-Activated Protein Kinase (MAPK), the Extracellular signal-Regulated Kinases 1 and 2 (Erk 1/2), Focal Adhesion Kinase (FAK) and tyrosine-phosphorylation of c-Src in mice 7 days after AB. This response was blunted in mice treated with integrin alpha v inhibitor. Microarrays probing for a total of 96 cell adhesion and ECM genes identified various genomic targets of integrin alpha v mediated signalling. 7 days after AB 18 ECM genes were up-regulated more than 2-fold (n=6), e.g. collagen (8.11 ± 2.2), fibronectin (2.32 ± 0.94), secreted protein, acidic and rich in cysteine (SPARC, 3.78 ± 0.12), A disintegrin-like and metalloprotease (reprolysin type) with trombospondin type 1 (Adamts-1, 3.51 ± 0.81) and Tissue inhibitor of metalloproteinase 2 (TIMP2, 2.23 ± 0.98), whereas this up-regulation was abolished in mice that were treatd by integrin alpha v inhibitor via mini pumps. We conclude that signaling downstream of integrin alpha v is mediated by the MAPK, FAK and c-Src pathways leading to an up-regulation of extracelluar matrix components necessary for the compensatory response of the heart under a condition of pressure overload.
Solid tumors are complex organ-like structures that consist not only of tumor cells but also of vasculature, extracellular matrix (ECM), stromal, and immune cells. Often, this tumor microenvironment (TME) comprises the larger part of the overall tumor mass. Like the other components of the TME, the ECM in solid tumors differs significantly from that in normal organs. Intratumoral signaling, transport mechanisms, metabolisms, oxygenation, and immunogenicity are strongly affected if not controlled by the ECM. Exerting this regulatory control, the ECM does not only influence malignancy and growth of the tumor but also its response toward therapy. Understanding the particularities of the ECM in solid tumor is necessary to develop approaches to interfere with its negative effect. In this review, we will also highlight the current understanding of the physical, cellular, and molecular mechanisms by which the pathological tumor ECM affects the efficiency of radio-, chemo-, and immunotherapy. Finally, we will discuss the various strategies to target and modify the tumor ECM and how they could be utilized to improve response to therapy.
Identification of articular cartilage progenitor cells (ACPCs) has opened up new opportunities for cartilage repair. These cells may be used as alternatives for or in combination with mesenchymal stromal cells (MSCs) in cartilage engineering. However, their potential needs to be further investigated, since only a few studies have compared ACPCs and MSCs when cultured in hydrogels. Therefore, in this study, we compared chondrogenic differentiation of equine ACPCs and MSCs in agarose constructs as monocultures and as zonally layered co-cultures under both normoxic and hypoxic conditions. ACPCs and MSCs exhibited distinctly differential production of the cartilaginous extracellular matrix (ECM). For ACPC constructs, markedly higher glycosaminoglycan (GAG) contents were determined by histological and quantitative biochemical evaluation, both in normoxia and hypoxia. Differential GAG production was also reflected in layered co-culture constructs. For both cell types, similar staining for type II collagen was detected. However, distinctly weaker staining for undesired type I collagen was observed in the ACPC constructs. For ACPCs, only very low alkaline phosphatase (ALP) activity, a marker of terminal differentiation, was determined, in stark contrast to what was found for MSCs. This study underscores the potential of ACPCs as a promising cell source for cartilage engineering.
