Bei der vorliegenden Arbeit handelt es sich um eine klinische und radiologische Nachuntersuchung von insgesamt 114 Patientinnen und Patienten, die zwischen 2009 und 2012 in der Poliklinik für Zahnerhaltung und Parodontologie der Universität Würzburg von approbierten Zahnärztinnen und Zahnärzten endodontisch behandelt wurden. Dabei kamen drei verschiedene Obturationsmethoden zum Einsatz.
1. Single-Cone-Technik mit Guttapercha und AH Plus® (SCGP)
2. Single-Cone-Technik mit Guttapercha und GuttaFlow® (SCGF)
3. Adhäsive Obturation in Continuous-Wave-Technik mit Resilon® (CWR)
Die Erhebung der Ausgangsvariablen (zum Behandlungszeitpunkt) erfolgte retrospektiv unter Zuhilfenahme der klinischen und radiologischen Dokumentation. Die Reevaluation des periapikalen Zustands der Zähne und die Erhebung weiterer klinischer Parameter erfolgte im Rahmen eines aktiven Patientenrecalls nach durchschnittlich 6,3 Jahren. Dabei wurden mit möglichst hoher Standardisierung postoperative Einzelzahnaufnah-men angefertigt. Diese wurden anhand der PAI-Klassifikation ausgewertet, um den pe-riapikalen Zustand der Zähne vor und nach Therapie zu bestimmen. PAI-Werte von 1 und 2 galten als Behandlungserfolg, Grad 3 bis 5 als Misserfolg. Im Hinblick auf die de-finierten Arbeitshypothesen wurden die Erfolgsraten innerhalb der Kohorten miteinander verglichen. Das vorrangige Ziel der hier vorliegenden Arbeit war, zu untersuchen, ob der endodontische Behandlungserfolg abhängig von der jeweiligen Obturationsmethode ist und ob technikspezifische Unterschiede sich einerseits auf die Qualität der Obturation und andererseits auf das Auftreten möglicher Komplikationen, wie der periapikalen Extrusion von Wurzelfüllmaterial, auswirken. Ferner sollten diese Aspekte neben weite-ren zahn- und patientenbezogenen Variablen bezüglich ihres Einflusses auf die Erfolgs-rate der endodontischen Therapie analysiert werden.
Es konnten keine signifikanten Unterschiede der endodontischen Erfolgsraten zwischen den hier untersuchten Obturationsmethoden ermittelt werden (p = ,16). In der SCGP-Kohorte lag die Erfolgsrate bei 85 % (34/40) verglichen mit 68,8 % (44/64) für CWR und 80 % (8/10) für SCGF. Die Homogenität der Obturation (p = ,2) und die Extrusion von Wurzelfüllmaterial in das periapikale Gewebe (p = ,93) zeigten keine Abhängigkeit von der gewählten Obturationstechnik. Die Länge der Wurzelkanalfüllung hingegen unter-schied sich signifikant zwischen den Kohorten (p = ,04*). Die Obturation mittels SCGP-Technik erzielte den höchsten Anteil adäquater Wurzelkanalfüllungen (92,5 %, 37/40) gegenüber SCGF (80 %, 8/10) und CWR (71,88 %, 46/64). Die CWR-Methode zeigte mit 18,8 % (12/64) den höchsten Anteil an unterfüllten Obturationen (SCGP: 7,5 %, 3/40; SCGF: 0 %).
Unabhängig von der Obturationsmethodik zeigte sich der endodontische Behandlungs-erfolg im Allgemeinen unbeeinflusst von der Qualität der Wurzelkanalfüllungen. Die Va-riablen Obturationslänge (p = ,12) und -homogenität (p = ,11) sowie die Extrusion von Wurzelfüllmaterial in die periapikale Region (p = 1,00) zeigten keinen signifikanten Ein-fluss auf die Erfolgsrate.
Das Durchschnittsalter im Patientenkollektiv betrug 60 Jahre mit einer tendenziellen Überrepräsentation weiblicher Probandinnen (60,5 %, 69/114). 73 % (81/111, 3 Mis-sings) der Studienteilnehmer/-innen wurden ab einem PSI-Grad von 3 als parodontal erkrankt eingestuft und 23,7 % (27/114) zeigten eine positive Raucheranamnese. Der BMI betrug im Durchschnitt 26,3 kg/m2. 42,3 % (47/111, 3 Missings) der Studienteil-nehmer/-innen wurden anhand der Einnahme von Medikamenten zur Therapie bzw. Prävention von kardiovaskulären Erkrankungen und/oder oraler Antidiabetika als chro-nisch erkrankt klassifiziert (chronic disease medication, CDM). Das Recallintervall be-trug durchschnittlich 6,3 Jahre mit einem Minimum von 4,7 und einem Maximum von 8,7 Jahren. Die patientenbezogenen Variablen Alter (p = ,45), Geschlecht (p = ,67), Pa-rodontitis (p = ,08), BMI (p = ,58), CDM (p = ,19), Recallintervall (p = ,08) und Rauchen (p = ,34) zeigten keinen signifikanten Einfluss auf den endodontischen Behandlungser-folg.
Unter den zahnbezogenen Variablen beeinflusste lediglich der präoperative apikale Sta-tus den endodontischen Erfolg signifikant (p = ,007*). Zähne mit präoperativer apikaler Läsion zeigten eine Erfolgsrate von 66,2 % (47/71) gegenüber 90,7 % (n = 39/43) bei Fällen ohne apikale Läsion. Die Misserfolgswahrscheinlichkeit bei Vorliegen einer präoperativen Läsion war um den Faktor 4,98 erhöht (OR = 4,98, 95 % KI: 1.60, 15,57, p = ,006*). Zwischen Kompositfüllungen, Teilkronen, Vollkronen, Teleskopkronen und Brückenversorgungen konnten keine relevanten Unterschiede in den Erfolgsraten er-mittelt werden (p = ,29). Gleiches galt für adäquate (76,6 %, 82/107) und inadäquate (57,1 %, 4/7) Restaurationen (p = ,36). Ebenso zeigten die Erfolgsraten von Wurzelka-nalrevisionen (70,5 %, 31/44) und Primärbehandlungen (78,6 %, 55/70) keine signifikan-ten Abweichungen voneinander (p = ,45). Molaren waren im Studienkollektiv mit 56,1 % (64/114) gegenüber Prämolaren und Frontzähnen mit je 21,9 % (25/114) überrepräsen-tiert. Der Zahntyp (p = ,07) und die Ausgangsdiagnose (p = ,22) stellten keine relevanten Einflussfaktoren des endodontischen Erfolgs dar.
Die AML stellt mit einem Anteil von 80 % an den akuten Leukämien bei Erwachsenen eine bedeutende Erkrankung für die Gesellschaft dar. Aufgrund fehlender durchbrechender Erfolge in der Therapieentwicklung liegt die durchschnittliche Fünfjahresüberlebensrate dennoch nur bei etwa 25 %. Der Blick auf die Kraft des Graft-versus-Leukämie-Effekts nach allogener Stammzelltransplantation, eine Langzeitremission der AML erzielen zu können, weist jedoch auf die Immunogenität und Eignung der Erkrankung für neue immuntherapeutische Ansätze hin.
Anhand der Kartierung der in-vivo präsentierten MHC-Klasse-I-Peptidome auf
AML-Blasten sollten in dieser Arbeit potenziell geeignete Therapietargets identifiziert werden, um eine breitere Anwendung immuntherapeutischer Strategien bei AML-Patienten zu ermöglichen. Auf primären Patientenmaterialien, Zelllinien und benignen Zellen wurden hierzu über eine Immunoaffinitätschromatographie mit nachfolgenden Purifizierungsschritten
die MHC-präsentierten Peptide massenspekrometrisch-basiert
identifiziert. Zusätzlich erfolgte eine Quantifizierung der Oberflächen- und intrazellulären MHC-Klasse-I-Moleküle der verwendeten Proben durch einen indirekten Immunfluoreszenz-Assay.
Unter der Gesamtheit von 17.750 identifizierten nicht-redundanten MHC-Klasse-
I-präsentierten Peptiden konnte eine Vielzahl von 5.626 Peptiden mit Präsentationsfrequenzen bis zu 72 % als AML-exklusiv beschrieben werden. Hierunter wurden 240 kryptische Peptide vermeintlich nicht-codierenden Ursprungs identifiziert. Zudem wurden mehrere potenziell CMV-kreuzreaktive AML-Peptide erfasst, die zu der reduzierten Rezidivrate bei CMV-Infektion nach allogener Stammzelltransplantation führen könnten. Bei der MHC-Quantifizierung wiesen die AML-Blasten keine verminderte MHC-Expression auf und stellten sich somit als geeignete Target-Zellen für eine T-Zell-Immuntherapie dar.
Die nächtliche (24-stündige) Überwachung des intraokularen Drucks (IOD) bei stationären Glaukompatienten wird in Europa seit mehr als 100 Jahren eingesetzt, um Spitzenwerte zu erkennen, die während der regulären Sprechstundenzeiten übersehen werden. Daten, die diese Praxis unterstützen, fehlen, zum Teil weil es schwierig ist, manuell erstellte IOD-Kurven mit dem objektiven Verlauf des Glaukoms zu korrelieren. Um dieses Problem zu beheben, haben wir automatisierte IOD-Datenextraktionswerkzeuge eingesetzt und auf eine Korrelation mit einem fortschreitenden Verlust der retinalen Nervenfaserschicht auf der optischen Kohärenztomographie im Spektralbereich (SDOCT) getestet.
Candida glabrata ist die zweithäufigste Ursache von Candidämien und invasiven Hefepilzinfektionen in Europa. Im Gegensatz zu C. albicans zeigt C. glabrata eine reduzierte Empfindlichkeit gegen bestimmte Antimykotika und kann unter Therapie rasch Resistenzen entwickeln.
Diese Arbeit umfasst eine systematische geno- und phänotypische Resistenzanalyse einer der größten europäischen - durch das NRZMyk in 5 Jahren zusammengetragenen - C. glabrata Stammsammlungen bestehend aus 176 klinisch relevanter Isolate. 84 der Stämme wurden anhand Referenztestung nach EUCAST zunächst als Anidulafungin (AND) resistent eingestuft. 71 wiesen konkordante Mutationen in den für die Glucan-Synthetase kodierenden FKS-Genen auf (13 % in FKS1, 87 % in FKS2). Vor allem die Position Ser-663 (FKS2-HS1) imponierte mit signifikant erhöhten AND MHK-Werten. 11 FKS-Wildtyp-Isolate, die ursprünglich als AND resistent klassifiziert wurden, wiesen in multiplen Nachtestungen um den Breakpoint undulierende AND MHK-Werte auf. 2 FKS-Wildtyp Isolate zeigten durchgängig hohe AND MHK-Werte und mussten daher - trotz fehlender Zielgenmutationen - als resistent eingestuft werden. Diese extremen Phänotypen wurden durch einen verblindeten nationalen Ringversuch bestätigt. Über ein Drittel der Isolate war multiresistent. Stämme aus Blutstrominfektionen und Ser-663 Mutation waren mit einer erhöhten Mortalität assoziiert. Ein weiteres Kernelement war die Detektion von Azol-resistenten C. glabrata petite-Phänotypen in der Routinediagnostik. Hier wurden innerhalb von 8 Monaten 20 relevante Isolate identifiziert.
Die Ergebnisse belegen das regelmäßige Auftreten single- / multidrug-resistenter C. glabrata Isolate in Deutschland. Phänotypische Resistenztestungen können zu Fehlklassifizierung von sensiblen Isolaten führen. FKS-Genotypisierungen hingegen sind ein nützliches Tool zur Identifizierung relevanter Resistenzen. In seltenen Fällen scheint jedoch eine Echinocandin-Resistenz ohne genotypisches Korrelat möglich zu sein.
The interaction between circadian clocks and metabolism is of increasing interest, since clock dysfunction often correlates with metabolic pathologies. Many research articles have been published analysing the impact of factors such as circadian clock, light, feeding time and diet-type on energy homeostasis in various tissues/organs of organisms with most of the findings done in mammals. Little is known about the impact of circadian clock and the above-mentioned factors on circulating lipids, especially the transport form of lipids - diacylglycerol (DG) and membrane lipids such as phosphatidylethanolamine (PE) and phosphatidylcholine (PC) in the Drosophila hemolymph. The fruit fly Drosophila is a prime model organism in circadian, behaviour and metabolism research.
To study the role of circadian clock and behaviour in metabolism, we performed an extensive comparative hemolymph lipid (diacylglycerol: DG, phosphatidylethanolamine: PE, phosphatidylcholine: PC) analysis using ultra performance liquid chromatography coupled to time-of-flight mass spectrometry (UPLC-MS) between wild-type flies (WTCS) and clock disrupted mutants (per01). In addition, clock controlled food intake– feeding behaviour was investigated. Time-dependent variation of transport (DG) and membrane lipids (PE and PC) were not rhythmic in WTCS under constant darkness and in per01 under LD, suggesting an impact of light and clock genes on daily lipid oscillations. Day-time and night-time restriction of food led to comparable lipid profiles, suggesting that lipid oscillations are not exclusively entrained by feeding but rather are endogenously regulated. Ultradian oscillations in lipid levels in WTCS under LD were masked by digested fatty acids since lipid levels peaked more robustly at the beginning and end of light phase when flies were fed a lipid- and protein-free diet. These results suggest that metabolite (DG, PE and PC) oscillation is influenced by complex interactions between nutrient-type, photic conditions, circadian clock and feeding time.
In conclusion, the results of this thesis suggest that circadian clocks determine transport and membrane lipid oscillation in Drosophila hemolymph in complex interactions between nutrient-type, photic conditions and feeding behaviour.
Im Zellkern eukaryotischer Zellen werden Gene in mRNAs transkribiert, welche umfangreich prozessiert und aus dem Zellkern exportiert werden. Im Zytoplasma erfolgt die Translation der mRNAs in Proteine, ein Prozess, welcher viel Energie benötigt und daher mittels vielfältiger Mechanismen streng reguliert wird. Ein Beispiel hierfür stellt die Klasse der TOP-mRNAs dar, eine RNA-Spezies, welche hauptsächlich Transkripte von Genen umfasst, die selbst in die Translation involviert sind. Die prominentesten Vertreter dieser Klasse sind die Proteine der kleinen und großen ribosomalen Untereinheiten. TOP-mRNAs zeichnen sich durch ein gemeinsames Sequenz-Motiv am Anfang Ihrer 5’-UTR aus, welches aus einem Pyrimidinstrang besteht und unmittelbar nach dem Cap mit einem Cytosin beginnt. Dieses allen TOP-RNAs gemeinsame Motiv ermöglicht die zeitgleiche Translationskontrolle dieser RNA-Klasse. So kann die Translation der TOP-mRNAs unter Stressbedingungen wie z.B. Nährstoffmangel koordiniert inhibiert werden, wodurch Energie eingespart wird.
Bereits lange wird nach einem Regulator gesucht, der an dieses TOP-Motiv bindet und die koordinierte Regulation ermöglicht. Man kann sich hier einen Inhibitor oder auch einen Aktivator vorstellen. Verschiedene Proteine wurden bereits in Erwägung gezogen. In dieser Arbeit wurde das Protein TIAR mittels Massenspektrometrie als TOP-interagierender Faktor identifiziert und dessen Bindungseigenschaften mit dem TOP-Motiv durch Shift Assays untersucht. Hierbei konnten Minimalkonstrukte verschiedener Organismen sowie RNA-TOP – Sequenzen identifiziert werden, welche sich für Strukturanalysen eignen würden. Als weiterer TOP-interagierender Faktor wurde über verschiedene sequenzielle Reinigungsschritte das Protein 14-3-3ε identifiziert.
Weiterhin wurden die TOP-Motiv-bindenden Proteine LARP1 und LARP7 auf Ihre Bindungseigenschaften mit Ihren Zielsequenzen untersucht. Während gezeigt werden konnte, dass LARP1 einen inhibierenden Einfluss auf TOP-RNAs hat, wurde in weiteren Shift-Assays die Bindungseigenschaften von LARP7 mit 7SK untersucht, wobei ebenfalls ein minimales LARP7–Konstrukt sowie 7SK-Konstrukte für Strukturanalysen identifiziert werden konnten. Weiterhin konnte gezeigt werden, dass verschiedene Substanzen wie tRNA und Arginin einen starken Einfluss auf die LARP7-7SK – Interaktion ausüben, welcher in weiteren Studien berücksichtigt werden sollte.
In der vorliegenden prospektiven Pilotstudie wurden die Hypothesen überprüft, dass es durch die nicht-invasive aurikuläre Vagusnervstimulation, jedoch nicht durch eine Kontrollstimulation am Ohrläppchen, zu einer Steigerung der Befindlichkeit, einer Verbesserung der Kognition und einem positiven Effekt auf die Herzratenvariabilität kommt.
Zusammenfassend konnten dabei in dieser Studie geringe Effekte der t-VNS auf einen kognitiven Parameter (F%-Wert des d2-Tests) sowie einen einzelnen HRV-Parameter (pNN50) gezeigt werden, wobei es Hinweise auf eine Intensitätsabhängigkeit der einzelnen Effekte gab. Auf die übrigen erfassten kognitiven Parameter und die weiteren gemessenen HRV-Parameter sowie die Befindlichkeit konnte kein Einfluss der t-VNS nachgewiesen werden. Bestätigt werden konnte das gute Sicherheitsprofil und die gute Tolerabilität der t-VNS.
Understanding the causal relationship between genotype and phenotype is a major objective in biology. The main interest is in understanding trait architecture and identifying loci contributing to the respective traits. Genome-wide association mapping (GWAS) is one tool to elucidate these relationships and has been successfully used in many different species. However, most studies concentrate on marginal marker effects and ignore epistatic and gene-environment interactions. These interactions are problematic to account for, but are likely to make major contributions to many phenotypes that are not regulated by independent genetic effects, but by more sophisticated gene-regulatory networks. Further complication arises from the fact that these networks vary in different natural accessions. However, understanding the differences of gene regulatory networks and gene-gene interactions is crucial to conceive trait architecture and predict phenotypes.
The basic subject of this study – using data from the Arabidopsis 1001 Genomes Project – is the analysis of pre-mature stop codons. These have been incurred in nearly one-third of the ~ 30k genes. A gene-gene interaction network of the co-occurrence of stop codons has been built and the over and under representation of different pairs has been statistically analyzed. To further classify the significant over and under- represented gene-gene interactions in terms of molecular function of the encoded proteins, gene ontology terms (GO-SLIM) have been applied. Furthermore, co- expression analysis specifies gene clusters that co-occur over different genetic and phenotypic backgrounds. To link these patterns to evolutionary constrains, spatial location of the respective alleles have been analyzed as well. The latter shows clear patterns for certain gene pairs that indicate differential selection.
The Amduat is one of the most important Netherworld Books which was recorded in various kinds of Ancient Egyptian sources since the beginning of the 18th dynasty, especially the walls of the royal tombs. The main theme of the Amduat is the journey of the sun god through the underworld where the solar bark and its crew is the central scene of the journey. The study focuses on finding the reasons of choosing the crew’s members who manage the sun bark’s journey in the Amduat. It also aims at illustrating the functions and responsibilities of each crew member. Following a historical approach, the study analyzes the Pyramid Texts and Coffin Texts as the most important documents before the New Kingdom, and proceeding to the inscriptions and writings of the monuments which contain portrayals and inscriptions of the Amduat in the New Kingdom. Furthermore, it sheds some light on the solar cycle’s main features and primary aspects, and tries to scrutinize the date, meaning, and symbolisms of the Amduat and its indications in the earlier sources.
Im Zeitraum vom 30.01.2019 bis zum 26.05.2020 wurden aus dem Universitätsklinikum Würzburg alle Patienten, die sowohl eine Charmi-Erstellung als auch eine Anlage des ASD-Dokuments erhalten haben, in das zu untersuchende Patientenkollektiv aufgenommen. Von diesen, so ermittelten, 1458 Patienten werden die stattgefundenen Operationen der Patienten eruiert. Die Bewegungswerte werden mithilfe des hierarchisch angeordneten Charmi-Systems von 0 bis 10 von den Physiotherapeuten erhoben. Dabei stellt der Charité Mobilitätsindex (Charmi-Index) ein neuartiges Diagnostikum dar, welches als Bewertungssystem zu Beginn 2019 erstmalig von den Physiotherapeuten an der Universität Würzburg eingesetzt wurde.
Der Schmerzzustand und Schmerzverlauf wird im Rahmen der Schmerzvisiten durch den Akutschmerzdienst (ASD) mithilfe der NRS-Skala festgehalten. Auch die NRS-Skala stellt eine hierarchisch aufgebaute Skala von 0 bis 10 dar. Schmerzwerte werden täglich im Verlauf der postoperativen Visiten erhoben. Charmi-Werte werden zu Beginn der postoperativen Phase, bei Zwischenereignissen und am Ende des stationären Aufenthalts oder bei Verlegung in einen anderen Fachbereich erhoben. Bei zeitgleich erhobenen Charmi-Werten und Schmerzwerten können diese zueinander in Bezug gesetzt werden.
Dabei kommt heraus, dass das Schmerzniveau in Ruhe und bei Belastung in der frühen postoperativen Phase negativ mit der Mobilität assoziiert ist. Zusätzlich zeigt sich, dass bei zunehmender Medikamentendosis die Mobilität sinkt. Je größer die Schmerzen der Patienten sind, desto schlechter die Mobilität. Wegen fehlender Charmi-Zwischenbefunde und zeitlich versetzt erhobener Charmi-Endbefunde ist kaum ein Zusammenhang zur Mobilität ersichtlich. Bei zeitlich passenden Charmi-Endwerten zeigt sich dennoch, dass bei steigenden Belastungsschmerzen die Mobilität abnimmt.
Eine weitere Erkenntnis ist, dass der Belastungsschmerz bei niedrigen Schmerzwerten durchschnittlich um 1,5-1,6 höher ist als der Ruheschmerz, bei mittleren Schmerzwerten um 1,5-1,4 höher und bei hohen Schmerzwerten um 1,4-1,2 höher.
Bei steigendem Ruheschmerz wird von den Patienten mehr Medikament angefordert als bei steigendem Belastungsschmerz, was die Vermutung zulässt, dass Patienten eher auf die Bewegung verzichten. Ein Therapieregime vermehrt mit Blickrichtung auf den Belastungsschmerz scheint daher an Bedeutung zuzunehmen.
Synthese einer Bibliothek von Aminosäure-basierten Oligopeptid-Amphiphilen mittels Festphasensynthese, deren kovalente Knüpfung an einen nukleophilen Kern zu C3-symmetrischen Sternmesogenen und die Analyse der Einflüsse der verwendeten Aminosäuren auf die Sekundärstruktur des synthetisierten Moleküls.
Hintergrund - Die Multiple Sklerose (MS) ist bis heute eine nur teilweise verstandene Autoimmunerkrankung des zentralen Nervensystems (ZNS). Das Tiermodell der experimentellen autoimmunen Enzephalomyelitis (EAE) ermöglicht die Erforschung von Teilaspekten der Pathogenese der MS und kann zur Etablierung von Therapeutika herangezogen werden. Die MP4-abhängige EAE ermöglicht als Mausmodell die gezielte Erforschung der Rolle der B-Zelle als Akteur in der Pathogenese der MS. Diese Dissertation untersuchte den Effekt des S1P1-Rezeptor-Modulators FTY720 (Fingolimod) auf die Immunantwort der autoreaktiven B-Zellen in der Peripherie sowie im ZNS.
Methoden - MP4-immunisierte Mäuse erhielten 50 Tage nach dem EAE-Krankheitsbeginn oral appliziertes FTY720 über einen Zeitraum von 30 Tagen. Die Tiere wurden nach dem Auftreten der Krankheitssymptome täglich klinisch evaluiert. Die MP4-spezifische B-Zell-Immunantwort und die MP4-spezifische humorale Immunreaktion wurden mittels ELISPOT und ELISA ausgewertet. Die Verteilung der T- und B-Zell-Anteile im peripheren Blut der Mäuse sowie die Aufteilung der B-Zell-Subsets in der Milz wurden mittels Durchflusszytometrie quantifiziert. Mittels Immunhistochemie wurden die B- und T-Zell-Ansammlungen im ZNS der Mäuse hinsichtlich ihrer Entwicklung in tertiär lymphatische Organe (TLOs) untersucht.
Ergebnisse - In diesem Versuchsaufbau zeigte FTY720 keine signifikante Verbesserung des klinischen Krankheitsverlaufes der Tiere. Der Anteil von T-Zellen im peripheren Blut der Mäuse war unter der Therapie mit FTY720 signifikant reduziert, während die Anzahl an B-Zellen nicht-signifikant beeinflusst wurde. Bei der Untersuchung der B-Zell-Subtypen in der Milz fiel zunächst ein erhöhter Anteil an B220+-B-Zellen auf, während die Verteilung der weiteren Subsets nicht-signifikant verändert war. Unter der Therapie mit FTY720 zeigte sich keine Reduktion bereits etablierter B-Zell-Aggregate im ZNS, allerdings ist eine inhibierte Entwicklung in TLOs zu diskutieren.
Zusammenfassung - Diese Arbeit impliziert unterschiedliche Effekte von FTY720 auf die B-Zellen in einem B-Zell-abhängigen chronischen Mausmodell der MS.
Die Raumfahrt ist eine der konservativsten Industriebranchen. Neue Entwicklungen von Komponenten und Systemen beruhen auf existierenden Standards und eigene Erfahrungen der Entwickler. Die Systeme sollen in einem vorgegebenen engen Zeitrahmen projektiert, in sehr kleiner Stückzahl gefertigt und schließlich aufwendig qualifiziert werden. Erfahrungsgemäß reicht die Zeit für Entwicklungsiterationen und weitgehende Perfektionierung des Systems oft nicht aus. Fertige Sensoren, Subsysteme und Systeme sind Unikate, die nur für eine bestimme Funktion und in manchen Fällen sogar nur für bestimmte Missionen konzipiert sind. Eine Neuentwicklung solcher Komponenten ist extrem teuer und risikobehaftet. Deswegen werden flugerprobte Systeme ohne Änderungen und Optimierung mehrere Jahre eingesetzt, ohne Technologiefortschritte zu berücksichtigen.
Aufgrund des enormen finanziellen Aufwandes und der Trägheit ist die konventionelle Vorgehensweise in der Entwicklung nicht direkt auf Kleinsatelliten übertragbar. Eine dynamische Entwicklung im Low Cost Bereich benötigt eine universale und für unterschiedliche Anwendungsbereiche leicht modifizierbare Strategie. Diese Strategie soll nicht nur flexibel sein, sondern auch zu einer möglichst optimalen und effizienten Hardwarelösung führen.
Diese Arbeit stellt ein Software-Tool für eine zeit- und kosteneffiziente Entwicklung von Sternsensoren für Kleinsatelliten vor. Um eine maximale Leistung des Komplettsystems zu erreichen, soll der Sensor die Anforderungen und Randbedingungen vorgegebener Anwendungen erfüllen und darüber hinaus für diese Anwendungen optimiert sein. Wegen der komplexen Zusammenhänge zwischen den Parametern optischer Sensorsysteme ist keine
„straightforward" Lösung des Problems möglich. Nur durch den Einsatz computerbasierter Optimierungsverfahren kann schnell und effizient ein bestmögliches Systemkonzept für die gegebenen Randbedingungen ausgearbeitet werden.
Protein-DNA interactions are central to many biological processes and form the bedrock of gene transcription, DNA replication, and DNA repair processes. Many proteins recognize specific sequences in DNA- a restriction enzyme must only cut at the correct sequence and a transcription factor should bind at its consensus sequence. Some proteins are designed to bind to specific structural or chemical features in DNA, such as DNA repair proteins and some DNA modifying enzymes. Target-specific DNA binding proteins initially bind to non-specific DNA and then search for their target sites through different types of diffusion mechanisms. Atomic force microscopy (AFM) is a single-molecule technique that is specifically well-suited to resolve the distinct states of target-specific as well as nonspecific protein-DNA interactions that are vital for a deeper insight into the target site search mechanisms of these enzymes. In this thesis, protein systems involved in epigenetic regulation, base excision repair (BER), and transcription are investigated by single-molecule AFM analyses complemented by biochemical and biophysical experiments.
The first chapter of this thesis narrates the establishment of a novel, user-unbiased MatLab-based tool for automated DNA bend angle measurements on AFM data. This tool has then been employed to study the initial lesion detection step of several DNA glycosylases. These results promoted a model describing the altered plasticities of DNA at the target lesions of DNA glycosylases as the fundamental mechanism for their enhanced efficiency of lesion detection.
In the second chapter of this thesis, the novel automated tool has been further extended to provide protein binding positions on the DNA along with corresponding DNA bend angles and applied to the study of DNMT3A DNA methyltransferase. These AFM studies revealed preferential co-methylation at specific, defined distances between two CpG sites by the enzyme and when combined with biochemical analyses and structural modelling supported novel modes of CpG co-methylation by DNMT3A.
In the third chapter of this thesis, the role of 8-oxo-guanine glycosylase (hOGG1) in Myc-mediated transcription initiation has been investigated. AFM analyses revealed that in the presence of oxidative damage in DNA, Myc is recruited to its target site (E-box) by hOGG1 through direct protein-protein interactions, specifically under oxidizing conditions. Intriguingly, oxidation of hOGG1 was further observed to result in dimerization of hOGG1, which may also play a role in the mechanism of transcription regulation by hOGG1 under oxidative stress.
Impacts of climate variability and change on Maize (\(Zea\) \(mays\)) production in tropical Africa

Climate change is undeniable and constitutes one of the major threats of the 21st century. It impacts sectors of our society, usually negatively, and is likely to worsen towards the middle and end of the century. The agricultural sector is of particular concern, for it is the primary source of food and is strongly dependent on the weather. Considerable attention has been given to the impact of climate change on African agriculture because of the continent’s high vulnerability, which is mainly due to its low adaptation capac- ity. Several studies have been implemented to evaluate the impact of climate change on this continent. The results are sometimes controversial since the studies are based on different approaches, climate models and crop yield datasets. This study attempts to contribute substantially to this large topic by suggesting specific types of climate pre- dictors. The study focuses on tropical Africa and its maize yield. Maize is considered to be the most important crop in this region. To estimate the effect of climate change on maize yield, the study began by developing a robust cross-validated multiple linear regression model, which related climate predictors and maize yield. This statistical trans- fer function is reputed to be less prone to overfitting and multicollinearity problems. It is capable of selecting robust predictors, which have a physical meaning. Therefore, the study combined: large-scale predictors, which were derived from the principal component analysis of the monthly precipitation and temperature; traditional local-scale predictors, mainly, the mean precipitation, mean temperature, maximum temperature and minimum temperature; and the Water Requirement Satisfaction Index (WRSI), derived from the specific crop (maize) water balance model. The projected maize-yield change is forced by a regional climate model (RCM) REMO under two emission scenarios: high emission scenario (RCP8.5) and mid-range emission scenario (RCP4.5). The different effects of these groups of predictors in projecting the future maize-yield changes were also assessed. Furthermore, the study analysed the impact of climate change on the global WRSI. The results indicate that almost 27 % of the interannual variability of maize production of the entire region is explained by climate variables. The influence of climate predictors on maize-yield production is more pronounced in West Africa, reaching 55 % in some areas. The model projection indicates that the maize yield in the entire region is expected to decrease by the middle of the century under an RCP8.5 emission scenario, and from the middle of the century to the end of the century, the production will slightly recover but will remain negative (around -10 %). However, in some regions of East Africa, a slight increase in maize yield is expected. The maize-yield projection under RCP4.5 remains relatively unchanged compared to the baseline period (1982-2016). The results further indicate that large-scale predictors are the most critical drivers of the global year-to-year maize-yield variability, and ENSO – which is highly correlated with the most important predictor (PC2) – seems to be the physical process underlying this variability. The effects of local predictors are more pronounced in the eastern parts of the region. The impact of the future climate change on WRSI reveals that the availability of maize water is expected to decrease everywhere, except in some parts of eastern Africa.
We present a technique for computing multi-branch-point covers with prescribed ramification and demonstrate the applicability of our method in relatively large degrees by computing several families of polynomials with symplectic and linear Galois groups.
As a first application, we present polynomials over \(\mathbb{Q}(\alpha,t)\) for the primitive rank-3 groups \(PSp_4(3)\) and \(PSp_4(3).C_2\) of degree 27 and for the 2-transitive group \(PSp_6(2)\) in its actions on 28 and 36 points, respectively. Moreover, the degree-28 polynomial for \(PSp_6(2)\) admits infinitely many totally real specializations.
Next, we present the first (to the best of our knowledge) explicit polynomials for the 2-transitive linear groups \(PSL_4(3)\) and \(PGL_4(3)\) of degree 40, and the imprimitive group \(Aut(PGL_4(3))\) of degree 80.
Additionally, we negatively answer a question by König whether there exists a degree-63 rational function with rational coefficients and monodromy group \(PSL_6(2)\) ramified over at least four points. This is achieved due to the explicit computation of the corresponding hyperelliptic genus-3 Hurwitz curve parameterizing this family, followed by a search for rational points on it. As a byproduct of our calculations we obtain the first explicit \(Aut(PSL_6(2))\)-realizations over \(\mathbb{Q}(t)\).
At last, we present a technique by Elkies for bounding the transitivity degree of Galois groups. This provides an alternative way to verify the Galois groups from the previous chapters and also yields a proof that the monodromy group of a degree-276 cover computed by Monien is isomorphic to the sporadic 2-transitive Conway group \(Co_3\).
In the heart the β\(_1\)-adrenergic receptor (AR) and the β\(_2\)-AR, two prototypical G protein-coupled receptors (GPCRs), are both activated by the same hormones, namely adrenaline and noradrenaline. Both receptors couple to stimulatory G\(_s\) proteins, mediate an increase in cyclic adenosine monophosphate (cAMP) and influence the contractility and frequency of the heart upon stimulation. However, activation of the β\(_1\)-AR, not the β\(_2\)-AR, lead to other additional effects, such as changes in gene transcription resulting in cardiac hypertrophy, leading to speculations on how distinct effects can arise from receptors coupled to the same downstream signaling pathway.
In this thesis the question of whether this distinct behavior may originate from a differential localization of these two receptors in adult cardiomyocytes is addressed. Therefore, fluorescence spectroscopy tools are developed and implemented in order to elucidate the presence and dynamics of these endogenous receptors at the outer plasma membrane as well as on the T-tubular network of intact adult cardiomyocytes. This allows the visualization of confined localization and diffusion of the β\(_2\)-AR to the T-tubular network at endogenous expression. In contrast, the β\(_1\)-AR is found diffusing at both the outer plasma membrane and the T-tubules. Upon overexpression of the β\(_2\)-AR in adult transgenic cardiomyocytes, the receptors experience a loss of this compartmentalization and are also found at the cell surface. These data suggest that distinct signaling and functional effects can be controlled by specific cell surface targeting of the receptor subtypes.
The tools at the basis of this thesis work are a fluorescent adrenergic antagonist in combination of fluorescence fluctuation spectroscopy to monitor the localization and dynamics of the lowly expressed adrenergic receptors. Along the way to optimizing these approaches, I worked on combining widefield and confocal imaging in one setup, as well as implementing a stable autofocus mechanism using electrically tunable lenses.
Chronische Nierenerkrankungen gehen mit einer erhöhten kardiovaskulären Morbidität und Mortalität einher. Charakteristisch für chronische Nierenerkrankungen, insbesondere im Stadium der Dialysepflichtigkeit, ist eine ausgeprägte Voralterung der Gefäße. Die Vorgänge, die den beschleunigten vaskulären Alterungsprozessen zugrunde liegen, umfassen ein Zusammenspiel aus einem gestörten Mineralstoffwechsel, der Akkumulation urämischer Toxine und chronischer Inflammation. Das Renin-Angiotensin-Aldosteron-System (RAAS) nimmt dabei eine zentrale Rolle ein. Eine gesteigerte Aktivität des RAAS ist ein Merkmal von kardiorenalen Syndromen und moduliert jenseits seiner Effekte auf den Blutdruck vaskuläre Entzündungs- und Remodelingprozesse. Durch das vaskuläre Altern kommt es zur Abnahme arterieller Compliance und zur Erhöhung der Pulswellengeschwindigkeit (PWV). Dadurch erhöht sich das Risiko für Endorganschäden. Die arterielle Gefäßsteifigkeit ist ein unabhängiger Prädiktor für Mortalität bei chronisch-dialysepflichtiger Niereninsuffizienz und eine Reduktion arterieller Rigidität geht mit einem verbesserten Überleben einher. Randomisierte Studien bei Dialysepatienten konnten bislang keinen eindeutigen Nutzen etablierter pharmakologischer Interventionen zur Reduktion des kardiovaskulären Risikos und vaskulärer «Stiffeningprozesse» feststellen. Als ein potentiell wirksamer Therapieansatz werden Mineralokortikoidrezeptorantagonisten (MRA) angesehen. Die vorliegende Arbeit evaluierte im Rahmen der Placebo-kontrollierten, randomisierten «Mineralocorticoid-Receptor Antagonists in End-Stage Renal Disease» (MiREnDa) Studie, ob die tägliche Einnahme von 50 mg Spironolacton über neun Monate einen Effekt auf die vaskuläre Funktion bei Patienten mit dialysepflichtiger chronischer Nierenerkrankung hat. Neben aortaler PWV, Augmentationsindex, zentralem Puls- und Blutdruck wurden zur Evaluation der vaskulären Funktion die Compliance der thorakalen Aorta und der A. carotis communis sowie die Distensibilität der A. carotis communis und die fluss-vermittelte Dilatation der A. brachialis vor Studienbeginn als sekundäre Endpunkte festgelegt. Ein weiterer Aspekt, der evaluiert wurde, war die Frage nach Korrelationen zwischen PWV und Augmentationsindex einerseits und weiteren Parametern vaskulärer Funktion, klinischen Merkmalen und Biomarkern andererseits. Die vorliegende Arbeit versuchte darüber hinaus, klinische Merkmale (Komorbiditäten, Inflammation), die ein Therapieansprechen von MRA potentiell modulieren, zu identifizieren. Das zentrale Ergebnis der Arbeit war, dass eine MRA-Therapie mit 50 mg Spironolacton täglich über neun Monate im untersuchten Kollektiv keinen Effekt auf die vaskuläre Funktion zeigte.
Spectroscopic methods were established decades ago in a wide variety of fields. This also applies to the pharmaceutical field, although they initially were mostly used for identity testing or structure elucidation only. Technical developments, such as miniaturization (NMR benchtop devices), Fourier transformations (for NMR, MIR spectroscopy) or the combination with chemometric evaluation (e.g., in Process Analytical Technology, PAT), have further increased their importance and opened up new applications. The aim of this work was to investigate further new approaches and to find new applications for already established methods and to show their benefits.
By means of MIR, NIR and NMR data and their chemometric evaluation (principal component analysis, PCA; hierarchical cluster analysis, HCA; linear discriminant analysis, LDA), possibilities were presented to successfully determine the manufacturer or the pharmaceutical company of various paracetamol preparations. In the course of this, various similarities and correlations between the preparations of individual companies could also be identified. For this purpose, a suitable sample preparation was developed for each spectroscopic method, and suitable measurement parameters in order to obtain reproducible spectra for the chemometric evaluation were determined. Furthermore, the results of the two unsupervised methods (HCA, PCA) were compared with each other. The HCA was able to confirm those of the PCA for the very most part. Additionally, through these methods it was possible to characterize many of the preparations based on clusters formed by comparable tablet compositions.
In order to be able to measure unmortared, whole tablets using the NIR spectrometer, an attachment was developed and manufactured using 3D printing. Its functionality was demonstrated by measuring and analyzing the tablets of two different batches of nine paracetamol preparations. The batches were clearly distinguished on the basis of a PCA and a significant difference was also demonstrated by means of statistical tests.
For NMR spectroscopy, a method was developed to obtain optimized "fingerprint" spectra of drug formulations. For this purpose, a 1D DOSY measurement was elaborated, in which the signals of the active ingredient could be filtered out by the appropriate choice of measurement parameters. The chemometric evaluation can thus focus on the remaining signals of the excipients, on the basis of which the preparations of the same API can be distinguished. Especially in the case of formulations that consist largely of active ingredient, data pre processing of the spectra can thus be simplified and greater importance can be assigned to the originally very small excipient signals.
A quantitative 1H NMR method was developed for the comparison of a high field spectrometer (400 MHz) with a benchtop spectrometer (80 MHz) for two finished drugs. It was shown that it is possible to obtain comparable results with both instruments, but that the influence of the excipients on the signals and the lower resolution of the benchtop instrument must be taken into account. Therefore, it was not possible to obtain comparable results without further optimization of the method for one of the active ingredients.
In the investigation of various reactions between APIs and excipients using DOSY, its usefulness as a screening method in stability testing was demonstrated. For this purpose, three different APIs and excipients were stressed together and the reaction mixtures were subsequently measured using DOSY. Based on the translational diffusion coefficient, the reaction products could be identified and distinguished from the active ingredients and the excipients used. The importance of thoughtful processing could also be demonstrated. If all peak heights are selected when evaluating signals split by direct spin spin coupling, this allows the detection of hidden signals as long as not all signals have the same diffusion coefficient. The selective selection of individual peak heights in the case of split signals also enables the evaluation of signals that overlap slightly. However, the limitations of this method were also shown when two signals overlap too much and differ too little in their diffusion coefficients.
Hence, it has been successfully demonstrated in the various projects that the new chemometric approaches, as well as the new applications of already established methods, enable in depth findings and thus have a clear added value.
Das fortgeschrittene Nebennierenkarzinom stellt Mediziner:innen im klinischen Alltag mit seinem aggressiven Verhalten vor eine große therapeutische Herausforderung. Bisher nimmt Mitotane die zentrale Rolle der medikamentösen Tumortherapie ein. Obwohl es seit über 60 Jahren im Einsatz ist, wurde die Frage der Effektivität einer Monotherapie im fortgeschrittenen Stadium in der Literatur bisher nur wenig untersucht. Daher wurden in dieser retrospektiven Studie Daten von 127 Patient:innen aus dem deutschen Nebennierenkarzinomregister untersucht, welche eine Mitotanemonotherapie bei Erst/- Rezidivdiagnose eines fortgeschrittenen ACC erhielten. Anhand dieses Datensatzes wurde die Ansprechrate, das progressionsfreie und Gesamtüberleben mithilfe der Kaplan- Meier- Methode ermittelt. Darüber hinaus wurde der Versuch einer Definition potenzieller Prädiktoren für ein verbessertes Ansprechen mittels Cox- Regressionsanalyse unternommen. Insgesamt wurde bei 26 Patient:innen (20,5%) ein objektives Ansprechen erzielt, von denen 3 Patient:innen eine komplette und 23 Patient:innen eine partielle Remission erreichten. Bei 32 weiteren Patient:innen (25,2%) wurde eine Stabilisierung der Erkrankung verzeichnet. Das progressionsfreie Überleben lag im Median bei 4,1 Monaten, das mediane Gesamtüberleben betrug 18,5 Monate. In der multivariaten Cox- Regressionsanalyse konnten zwei wesentliche, unabhängige Faktoren identifiziert werden: Patient:innen mit einer niedrigen Tumorlast (definiert als <10 Läsionen) scheinen im Vergleich zu Patient:innen mit einer höheren Tumorlast von der Therapie hinsichtlich des progressionsfreien Überlebens (HR:0,51; 95%-CI:0,33-0,79; p=0,02) und Gesamtüberlebens (HR:0,59; 95%-CI: 0,30-0,91; p=0,017) besser zu profitieren. Interessanterweise zeigen Patient:innen, die Mitotane erst zum Zeitpunkt eines Spätrezidivs (>360 Tage nach Erstdiagnose) erhalten haben, ein deutlich besseres Ansprechen im Vergleich zu Patient:innen, welche Mitotane unmittelbar nach Erstdiagnose erhielten: Es zeigte sich ein verlängertes progressionsfreies Überleben (HR: 0,35; 95%-CI: 0,23-0,55; p=0,001) und Gesamtüberleben (HR: 0,34; 95%-CI: 0,22-0,52; p=0,001). In der weiteren Analyse zeigte sich, dass sich ein Mitotanespiegel von >14mg/l günstig auf ein verbessertes Gesamtüberleben auswirkt. Zusammenfassend zeigt unsere bisher weltweit größte zu diesem Thema durchgeführte Studie, dass eine Mitotanemonotherapie unter bestimmten Voraussetzungen zu sehr guten Ergebnissen führen kann: Dabei scheinen Patient:innen mit einer niedrigen Tumorlast und dem späten Beginn einer Mitotanetherapie besonders geeignet für diese Therapiemodalität zu sein, während man Patient:innen mit Metastasen bei Erstdiagnose bzw. einer hohen Tumorlast zum Zeitpunkts des Rezidivs besser direkt mit einer Kombination aus Mitotane plus zytotoxischer Chemotherapie behandelt.
