Interleukin-4 (IL-4) is an anti-inflammatory and analgesic cytokine that induces opioid receptor transcription. We investigated IL-4 knockout (ko) mice to characterize their pain behavior before and after chronic constriction injury (CCI) of the sciatic nerve as a model for neuropathic pain. We investigated opioid responsivity and measured cytokine and opioid receptor gene expression in the peripheral and central nervous system (PNS, CNS) of IL-4 ko mice in comparison with wildtype (wt) mice. Naïve IL-4 ko mice displayed tactile allodynia (wt: 0.45 g; ko: 0.18 g; p<0.001), while responses to heat and cold stimuli and to muscle pressure were not different. No compensatory changes in the gene expression of tumor necrosis factor-alpha (TNF), IL-1β, IL-10, and IL-13 were found in the PNS and CNS of naïve IL-4 ko mice. However, IL-1β gene expression was stronger in the sciatic nerve of IL-4 ko mice (p<0.001) 28 days after CCI and only IL-4 ko mice had elevated IL-10 gene expression (p = 0.014). Remarkably, CCI induced TNF (p<0.01), IL-1β (p<0.05), IL-10 (p<0.05), and IL-13 (p<0.001) gene expression exclusively in the ipsilateral spinal cord of IL-4 ko mice. The compensatory overexpression of the anti-inflammatory and analgesic cytokines IL-10 and IL-13 in the spinal cord of IL-4 ko mice may explain the lack of genotype differences for pain behavior after CCI. Additionally, CCI induced gene expression of μ, κ, and δ opioid receptors in the contralateral cortex and thalamus of IL-4 ko mice, paralleled by fast onset of morphine analgesia, but not in wt mice. We conclude that a lack of IL-4 leads to mechanical sensitivity; the compensatory hyperexpression of analgesic cytokines and opioid receptors after CCI, in turn, protects IL-4 ko mice from enhanced pain behavior after nerve lesion.
Background: To perform a systematic review and meta-analysis on cytokine levels in patients with fibromyalgia syndrome (FMS). Methods: Through December 2010 we systematically reviewed the databases PubMed, MEDLINE, and PsycINFO and screened the reference lists of 22 review articles for suitable original articles. Original articles investigating cytokines in patients with FMS were included. Data were extracted by two independent authors. Differences of the cytokine levels of FMS patients and controls were summarized by standardized mean differences (SMD) using a random effects model. Study quality was assessed applying methodological scores: modified Center of Evidence Based Medicine, Newcastle-Ottawa-Scale, and Würzburg Methodological Quality Score. Results: Twenty-five articles were included investigating 1255 FMS patients and 800 healthy controls. Data of 13/25 studies entered meta-analysis. The overall methodological quality of studies was low. The results of the majority of studies were not comparable because methods, investigated material, and investigated target cytokines differed. Systematic review of the selected 25 articles revealed that FMS patients had higher serum levels of interleukin (IL)-1 receptor antagonist, IL-6, and IL-8, and higher plasma levels of IL-8. Meta-analysis of eligible studies showed that FMS patients had higher plasma IL-6 levels compared to controls (SMD = -0.34 [-0.64, -0.03] 95% CI; p = 0.03). The majority of investigated cytokines were not different between patients and controls. Conclusions: The pathophysiological role of cytokines in FMS is still unclear. Studies of higher quality and with higher numbers of subjects are needed.
Background: There is extensive evidence that explicit memory, which involves conscious recall of encoded information, can be modulated by emotions; emotions may influence encoding, consolidation or retrieval of information. However, less is known about the modulatory effects of emotions on procedural processes like motor memory, which do not depend upon conscious recall and are instead demonstrated through changes in behaviour. Experiment 1: The goal of the first experiment was to examine the influence of emotions on motor learning. Four groups of subjects completed a motor learning task performing brisk isometric abductions with their thumb. While performing the motor task, the subjects heard emotional sounds varying in arousal and valence: (1) valence negative / arousal low (V-/A-), (2) valence negative / arousal high (V-/A+), (3) valence positive / arousal low (V+/A-), and (4) valence positive / arousal high (V+/A+). Descriptive analysis of the complete data set showed best performances for motor learning in the V-/A- condition, but the differences between the conditions did not reach significance. Results suggest that the interaction between valence and arousal may modulate motor encoding processes. Since limitations of the study cannot be ruled out, future studies with different emotional stimuli have to test the assumption that exposure to low arousing negative stimuli during encoding has a facilitating effect on short term motor memory. Experiment 2: The purpose of the second experiment was to investigate the effects of emotional interference on consolidation of sequential learning. In different sessions, 6 groups of subjects were initially trained on a serial reaction time task (SRTT). To modulate consolidation of the newly learned skill, subjects were exposed, after the training, to 1 of 3 (positive, negative or neutral) different classes of emotional stimuli which consisted of a set of emotional pictures combined with congruent emotional musical pieces or neutral sound. Emotional intervention for each subject group was done in 2 different time intervals (either directly after the training session, or 6 h later). After a 72 h post-training interval, each group was retested on the SRTT. Re-test performance was evaluated in terms of response times and accuracy during performance of the target sequence. Emotional intervention did not influence either response times or accuracy of re-testing SRTT task performance. However, explicit awareness of sequence knowledge was enhanced by arousing negative stimuli applied at 0 h after training. These findings suggest that consolidation of explicit aspects of procedural learning may be more responsive toward emotional interference than are implicit aspects. Consolidation of different domains of skill acquisition may be governed by different mechanisms. Since skill performance did not correlate with explicit awareness we suggest that implicit and explicit modes of SRTT performance are not complementary. Experiment 3: The aim of the third experiment was to analyze if the left hemisphere preferentially controls flexion responses towards positive stimuli, while the right hemisphere is specialized towards extensor responses to negative pictures. To this end, right-handed subjects had to pull or push a joystick subsequent to seeing a positive or a negative stimulus in their left or right hemifield. Flexion responses were faster for positive stimuli, while negative stimuli were associated with faster extensions responses. Overall, performance was fastest when emotional stimuli were presented to the left visual hemifield. This right hemisphere superiority was especially clear for negative stimuli, while reaction times towards positive pictures showed no hemispheric difference. We did not find any interaction between hemifield and response type. Neither was there a triple interaction between valence, hemifield and response type. In our experimental context the interaction between valence and hemifield seems to be stronger than the interaction between valence and motor behaviour. From these results we suppose that under certain conditions a hierarchy scaling of the asymmetry patterns prevails, which might mask any other existing asymmetries.
Im Rahmen dieser Arbeit wurden Gewebe von jeweils 15 kolorektalen MSI- und CSIPrimärtumoren, sowie 3 MSI- und 2 CSI-Tumorzelllinien mittels SNP-Arrayanalyse molekulargenetisch auf uniparentale Disomie (UPD) untersucht. Es konnte bestätigt werden, dass die uniparentale Disomie ein wichtiger Mechanismus zur Inaktivierung von Tumorsuppressorgenen zu sein scheint. Die Ergebnisse zeigen, dass nicht nur in MSI-Tumoren, sondern auch in CSI-Tumoren UPD nachgewiesen werden konnte, wobei der UPD-Anteil an identifizierten LOH-Regionen in MSI-Tumoren mit 94% gegenüber 41% in CSI-Tumoren deutlich höher lag. Interessanterweise wurde auch in 68% der korrespondierenden MSI- und in 27% der entsprechenden CSINormalgewebe UDP festgestellt, was daraufhin deutet, dass hier möglicherweise schon kanzerogene Vorstufen vorliegen. Die Verteilung der UPDs in den 15 Tumorproben der jeweiligen Tumortypen (MSI und CSI) war innerhalb der einzelnen Proben und innerhalb der einzelnen Chromosomen sehr heterogen, es wurden jedoch gemeinsam Regionen mit UPD gefunden. Bei 27% der MSI-Tumoren wurde eine neue Kandidatenregion auf Chromosom 6 (6pter-p22) identifiziert, die auch einige potentielle Tumorkandidatengene, wie das Tumorsuppressorgen NOL7 oder den Transkriptionsfaktor AP2a enthält. In CSI-Tumoren waren die monoallelischen Regionen mit 59% hauptsächlich durch Deletionen charakterisiert, während die MSITumore im Gegensatz nur 3% Deletionen aufwiesen. In CSI-Tumoren fielen insbesondere vier Bereiche auf den Chromosomen 18, 17, 8 und 22 durch allelische Verluste auf und bestätigen damit eine Reihe von früheren Ergebnissen. In bestimmten Regionen (z.B. 22q12.1) wurden sowohl Deletionen als auch UPD festgestellt. Die Region 5q22.2 war fast auschließlich von uniparentaler Disomie betroffen. Ob die UPD vor allem in frühen Stadien der Kolonkarzinogenese eine Rolle spielt und in späteren Stadien eher in chromosomaler, bzw. Mikrosatelliteninstabilität vorkommt, werden weitere Studien zeigen müssen.
Trotz beträchtlicher Anstrengung Malaria zu kontrollieren bzw. zu eradizieren, stellt die Krankheit weiterhin eines der gravierendsten Gesundheitsprobleme unseres Jahrtausends dar. Malaria fordert jährlich zwischen 0,7 und 2,7 Millionen Menschenleben, beeinträchtigt schulische und soziale Entwicklung und hemmt erheblich das Wirtschaftswachstum der betroffenen Länder. In Burkina Faso, einem der ärmsten Länder der Welt, ist Malaria eines der größten Gesundheitsprobleme und ca. ein Drittel aller Todesfälle werden hier Malaria angelastet. Die sich weiter ausbreiteten Resistenzen gegen die gängigen Malariamedikamente machen die Bekämpfung der Malaria zunehmend schwierig. Artemisinin basierende Kombinationstherapien sind aktuell, trotz relativ hoher Therapiekosten und erster Resistenzen, die Erstlinien Behandlung. Effektive und billige neue Kombinationstherapien werden dringend benötigt. In dieser Doktorarbeit wurde das Resistenzpotential von Artemisinin modelliert. Die Homologiemodellierungen unterstützen die These von Krishna und Kollegen von SERCA als einzige Zielstruktur von Artemisinin. Des Weiteren wurde Methylenblau als neues altes Malariamittel evaluiert. Methylenblau ist das erste gegen Malaria eingesetzte Medikament, agiert als ein prooxidatives Agens und inhibiert selektiv und nicht-kompetitiv die P. falciparum Glutathion Reduktase. Die additiven und multiplen Zielprotein Effekte von Methylenblau wurden experimentell untersucht und hier in einem bioinformatischem Modell getestet. Unter dem Einfluss von Methylenblau werden einige Schlüsselenzyme des Redoxstoffwechsels in ihrer Aktivität beeinträchtigt und der Parasit wird verstärkt oxidativem Stress ausgesetzt. Des Weiteren konnte in dieser Dr. Arbeit eine starke Kooperationsbereitschaft der urbanen und ländlichen Bevölkerung an zukünftigen Malaria Projekten gezeigt werden.
Bildung für nachhaltige Entwicklung setzt einen systematischen Ansatz voraus, der sozioökonomische Umweltaspekte in enge Beziehung zueinander setzt. Bildung für nachhaltige Entwicklung bedeutet also, die Komplexität von Phänomenen und deren Beziehung zueinander zu begreifen lernen und Schlüsselkompetenzen zu entwickeln, um aktiv, bewusst, verantwortungsvoll und kritisch an der Gestaltung der Gegenwart und der Zukunft teilzuhaben. Gegenstand vorliegender Forschungsarbeit ist die Bildung für nachhaltige Entwicklung (BNE) aus Sicht der Geographie und im internationalen Vergleich zwischen der italienischen Stadt Padua in der Region Venetien und Würzburg in Bayern. Im Mittelpunkt stehen Aktivitäten und Projekte, die im formalen Bereich, d.h. in Kindergärten und Grundschulen, und im non-formalen Bereich, wie in Vereinen, Einrichtungen, lokale Körperschaften, durchgeführt wurden. Damit war die Voraussetzung geschaffen, das gesamte Spektrum an Angeboten und Möglichkeiten der Zusammenarbeit im Bereich BNE in den für die Fallstudie gewählten Städten zu erfassen, also einschließlich der Angebote von und der Angebote an die Schulen. Dem Forschungsverfahren liegt ein Fragebogen über die Wertesysteme und Wertvorstellungen von Erziehern und Lehrern in Bezug auf geographische Bildung zugrunde. Dabei stellt sich unter anderem heraus, dass der Zusammenhang zwischen BNE und geographischer Bildung kaum wahrgenommen wird. Diese Feststellung bestätigt sich im weiteren Verlauf des Untersuchungsverfahrens, das sich auf die Auswertung von dreizehn, aus der Fachliteratur und aus internationalen Dokumenten ausgewählten und behandelten BNE-Themen konzentriert. Die Ergebnisse zeigen, dass es in beiden Städten und in beiden Bereichen, im formalen wie im non-formalen, “ BNE-Best Practice” gibt und dass Themen mit Bezug zu Bildung zu Nachhaltigkeit gegenüber Themen mit Bezug zu entwicklungsbezogener Bildung der Vorzug gegeben wird. Weiter geht daraus hervor, dass in erster Linie Umweltaspekte behandelt werden, gefolgt von sozialen, während ökonomische Aspekte das Schlusslicht bilden. In Bezug auf das, was nachhaltige Entwicklung und BNE bedeuten, herrscht bei den Interviewpartnern ziemliche Unklarheit und ein geringes Bewusstsein. Im non-formalen Bereich Tätige wissen über die Grundlagen nachhaltiger Entwicklung Bescheid, bevorzugen jedoch die Bezeichnung “Umwelterziehung”, weil dieser Begriff für die Allgemeinheit angeblich besser verständlich ist. Die Mehrheit der Erzieher und Lehrer hingegen erkennt keinen Unterschied zwischen BNE und Umwelterziehung. Auch die staatlichen Lehrpläne und Richtlinien geben kaum Aufschluss darüber, was BNE ist. Manchmal wird BNE mit Umwelterziehung gleichgesetzt, dann ist sie wieder Teil davon, andere Male wird sie als Orientierungshilfe für Umwelterziehung empfohlen. Jedenfalls wird darin nichts anderes als Umwelterziehung damit in Verbindung gebracht. Die befragten Personen sind wesentlich mehr auf die “ Praxis” als auf die “ Theorie” bedacht. Sie führen interessante Projekte und Aktivitäten durch, sind sich aber des theoretischen Ansatzes, der BNE zugrunde liegt und der mit dem der Geographie in gar einigen Punkten konvergent ist, nicht wirklich bewusst. In diesem Sinne erschließt vorliegende Forschungsarbeit das Potential an Synergien von BNE und geographischer Bildung. Denn einerseits könnte die Geographie aufschlussreiche theoretische Überlegungen und Impulse liefern, andererseits könnte BNE den Anstoß zur Erneuerung geographischer Bildung geben und sie damit aus der gesellschaftlichen Isolation führen. Solche Grundüberlegungen sollten selbstverständlich auch in den Curricula – in Form der zur Verfügung stehenden Stundenzahl – ihren Niederschlag finden.
Der Zusammenhang zwischen den Parametern der Mikrostruktur des Myokards und der Spindephasierung wird hergestellt. Zur Beschreibung der Mikrostruktur des Myokards wurde das Kroghsche Kapillarmodell genutzt. In diesem Modell wird das Myokard auf eine einzige Kapillare reduziert, die von einem konzentrischen Gewebszylinder umgeben ist. In dem Gewebszylinder findet die Dephasierung und Diffusion statt. Mathematisch wird die Dephasierung durch die Bloch-Torrey-Gleichung beschrieben. Experimentell wurde der Signal-Zeit-Verlauf mittels einer PRESS-Sequenz und einer Gradienten-Echo-Sequenz gemessen. Mit den in dieser Arbeit vorgestellten Methoden ist der Zusammenhang zwischen Kapillarradius und Freien Induktionszerfall bekannt.
Frühgeborene Kinder haben ein erhöhtes Risiko für impfpräventable Erkrankungen. Von der STIKO wird explizit empfohlen, sehr und extrem Frühgeborene frühzeitig nach ihrem chronologischen Alter, also nach dem gleichen Impfplan, der auch für Reifgeborene gilt, zu impfen. Studien aus den USA oder der Schweiz konnten zeigen, dass Impfungen bei dieser Risikopopulation häufig verspätet durchgeführt werden. Für Deutschland gab es hierfür bisher keine Daten. In der hier vorliegenden Pilotstudie wird das Impfverhalten bei 332 extrem und sehr Frühgeborenen deutschlandweit im Alter von mind. 2 Jahren analysiert. Die Grundimmunisierungen mit dem 5 - bzw. 6-fach Impfstoff sowie mit dem Pneumokokkenimpfstoff wurden konsequent durchgeführt, während die Auffrischimpfungen im 2. Lebensjahr deutlich seltener erfolgten. Ein ähnliches Impfverhalten zeigte sich auch bei den MMR(V)-Impfungen mit einer deutlich verminderten Compliance bei der Auffrischimpfung. Nur bei einem der 332 erfassten Kinder kam es zu einer schweren Nebenwirkung mit Bradykardie und Abfall der Sauerstoffsättigung. Insgesamt zeigte sich v.a. ein Defizit bei der Durchführung der Auffrischimpfungen und bei der rechtzeitigen Applikation der Impfungen bei diesem speziellen Risikokollektiv.
Single-molecule microscopy is one of the decisive methodologies that allows one to clarify cellular signaling in both spatial and temporal dimentions by tracking with nanometer precision the diffusion of individual microscopic particles coupled to relevant biological molecules. Trajectory analysis not only enables determination of the mechanisms that drive and constrain the particles motion but also to reveal crucial information about the molecule interaction, mobility, stoichiometry, all existing subpopulations and unique functions of particular molecules. Efficacy of this technique depends on two problematic issues the usage of the proper fluorophore and the type of biochemical attachment of the fluorophore to a biomolecule. The goal of this study was to evolve a highly specific labeling method suitable for single molecule tracking, internalization and trafficking studies that would attain a calculable 1:1 fluorophore-to-receptor stoichiometry. A covalent attachment of quantum dots to transmembrane receptors was successfully achieved with a techinque that amalgamates acyl carrier protein (ACP) system as a comparatively small linker and coenzyme A (CoA)-functionalized quantum dots. The necessity of optimization of the quantum dot usage for more precise calculation of the membrane protein stoichiometries in larger assemblies led to the further study in which methods maximizing the number of signals and the tracking times of diverse QD types were examined. Next, the optimized techniques were applied to analyze behavior of interleukin-5 β-common chain receptor (IL-5Rβc) receptors that are endogenously expressed at low level on living differentiated eosinophil-like HL-60 cells. Obtained data disclosed that perused receptors form stable and higher order oligomers. Additionally, the mobility analysis based on increased in number (>10%) uninterrupted 1000-step trajectories revealed two patterns of confined motion. Thereupon methods were developed that allow both, determination of stoichiometries of cell surface protein complexes and the acquisition of long trajectories for mobility analysis. Sequentially, the aforementioned methods were used to scrutinize on the mobility, internalization and recycling dynamics characterization of a G protein-coupled receptor (GPCRs), the parathyroid hormone receptor (PTHR1) and several bone morphogenetic proteins (BMPs), a member of the TGF-beta superfamily of receptors. These receptors are two important representatives of two varied membrane receptor classes. BMPs activate SMAD- and non-SMAD pathways and as members of the transforming growth factor β (TGF-β) superfamily are entailed in the regulation of proliferation, differentiation, chemotaxis, and apoptosis. For effective ligand induced and ligand independent signaling, two types of transmembrane serine/threonine kinases, BMP type I and type II receptors (BMPRI and BMPRII, respectively) are engaged. Apparently, the lateral mobility profiles of BMPRI and BMPRII receptors differ markedly, which determinate specificity of the signal. Non-SMAD signaling and subsequent osteoblastic differentiation of precursor cells particularly necessitate the confinement of the BMP type I receptor, resulting in the conclusion that receptor lateral mobility is a dominative mechanism to modulate SMAD versus non-SMAD signaling during differentiation. Confined motion was also predominantly observed in the studies devoted to, entailed in the regulation of calcium homeostasis and in bone remodeling, the parathyroid hormone receptor (PTHR1), in which stimulation with five peptide ligands, specific fragments of PTH: hPTH(1–34), hPTHrP(107–111)NH2; PTH(1–14); PTH(1–28) G1R19, bPTH(3–34), first four belonging to PTH agonist group and the last to the antagonist one, were tested in the wide concentration range on living COS-1 and AD293 cells. Next to the mobility, defining the internalization and recycling rates of the PTHR1 receptor maintained in this investigation one of the crucial questions. Internalization, in general, allows to diminish the magnitude of the receptor-mediated G protein signals (desensitization), receptor resensitization via recycling, degradation (down-regulation), and coupling to other signaling pathways (e.g. MAP kinases). Determinants of the internalization process are one of the most addressed in recent studies as key factors for clearer understanding of the process and linking it with biological responses evoked by the signal transduction. The internalization of the PTH-receptor complex occurs via the clathrin-coated pit pathway involving β-arrestin2 and is initiated through the agonist occupancy of the PTHR1 leading to activation of adenylyl cyclase (via Gs), and phosphatidylinositol-specific phospholipase Cβ (via Gq). Taken together, this work embodies complex study of the interleukin-5 β-common chain receptor (IL-5Rβc) receptors, bone morphogenetic proteins (BMPs) and the parathyroid hormone receptor with the application of single-molecule microscopy with the newly attained ACP-quantum dot labeling method and standard techniques.
Ziel der Arbeit war die Entwicklung von lateral gekoppelten DFB-Halbleiterlasern für Hochleistungsanwendungen. Besonderes Augenmerk war dabei auf hohe COD-Schwellen und schmale Fernfeldverteilungen gerichtet. Ausgehend von einem LOC-Design wurden Simulationsrechnungen durchgeführt und ein neues Epitaxiedesign mit einer 2.5 μm dicken LOC, in welcher die aktive Schicht asymmetrisch positioniert ist, entwickelt. Durch die asymmetrische Anordnung der aktiven Schicht kann die im Falle von lateral gekoppelten DFB-Lasern sehr kritische Kopplung der Lichtmode an das modenselektive Gitter gewährleistet werden. Zudem reichen die Ausläufer der Lichtmode in diesem Design weiter in den Wellenleiter hinab als dies bei herkömmlichen Wellenleitern der Fall ist, so dass sich die Fernfeldeigenschaften der Laser verbessern. Die Fernfeldverteilungen solcher Laser weisen Halbwertsbreiten von 14° in lateraler und nur 19° in transversaler Richtung auf. Im Vergleich mit Standardstrukturen konnte die Ausdehnung des transversalen Fernfeldes also um mehr als 50 % reduziert werden. Außerdem ergibt sich eine nahezu runde Abstrahlcharakteristik, was die Einkopplungseffizienz in optische Systeme wie Glasfasern oder Linsen signifikant verbessert. Unter Ausnutzung der entwickelten Epitaxiestruktur mit asymmetrischer LOC wurde ein neues Lateraldesign entwickelt. Es handelt sich hierbei um Wellenleiterstege welche im Bereich der Facetten eine Verjüngung aufweisen. Durch diese wird die optische Mode tief in die 2.5 μm dicke Wellenleiterschicht geführt, welche sie in transversaler Richtung komplett ausfüllt. Durch den größeren Abstand der Lasermode vom Wellenleitersteg ergibt sich zudem eine deutliche schwächere laterale Führung, so dass sich die Mode auch parallel zur aktiven Schicht weiter ausdehnt. Die Lichtmode breitet sich folglich über eine deutlich größere Fläche aus, als dies bei einem gleichbleibend breiten Wellenleitersteg der Fall ist. Die somit signifikant kleinere Leistungsdichte auf der Laserfacette ist gleichbedeutend mit einem Anstieg der COD-Schwelle der Laser der im Einzelnen von den jeweiligen Designparametern von Schicht- und Lateralstruktur abhängig ist. Außerdem bewirkt die in lateraler und transversaler Richtung deutlich schwächere Lokalisation der Mode eine weitere Abnahme der Halbwertsbreiten der Laserfernfelder. Durch die im Vergleich zu herkömmlichen Laserstrukturen schwächere Lokalisation der Lichtmode im Bereich der Facetten ergeben sich äußerst schmale Fernfelder. Ein 1800 μm langer Laser, dessen Stegbreite über 200 μm hinweg auf 0.4 μm verringert wurde, zeigt Halbwertsbreiten von 5.2° in lateraler und 13.0° in transversaler Richtung. Damit sind die Fernfelder dieser Laser bedeutend kleiner als die bislang vorgestellter Laserdioden mit LOC. Die Geometrie der Taperstrukturen bestimmt, wie vollständig sich die Mode in den unteren Wellenleiterbereich ausbreiten kann und nimmt damit Einfluss auf die Laserfernfelder. Im CW-Modus durchgeführte Messungen an Lasern mit Taperstrukturen zeigen maximale Ausgangsleistung von 200 mW bevor die Laser in thermisches Überrollen übergehen. Bei einer Ausgangsleistung von 185 mW beträgt das Seitenmodenunterdrückungsverhältnis 33 dB. Im gepulsten Modus (50 ns Pulsdauer, 1MHz Wiederholungsrate) betriebene Laser zeigen hohe COD-Schwellen von mehreren hundert bis hin zu 1600 mW, die eine deutliche Abhängigkeit von der Endbreite der Taperstrukturen zeigen: Mit abnehmender Taperbreite ergibt sich eine starke Zunahme der COD-Schwelle. An einem 1800 μm langen Laser mit 200 μm langen Taperstrukturen die eine Endbreite von 0.3 μm aufweisen konnte eine COD-Schwelle von 1.6 W nachgewiesen werden. Im Gegensatz zu anderen Ansätzen, die ebenfalls longitudinal und lateral mono-modige DFB-Laser mit hohen Ausgangsleistungen zum Ziel haben, kann jedoch bei dem hier präsentierten Konzept aufgrund des Einsatzes von lateralen DFB-Gittern auf eine Unterbrechung des epitaktischen Wachstums verzichtet werden. Dies vereinfacht die Herstellung der Schichtstrukturen deutlich. Die hier vorgestellten Konzepte sind mit weiteren üblichen Vorgehensweisen zur Herstellung von Hochleistungslaserdioden, wie z.B. speziellen Facettenreinigungs- und Passivierungsverfahren oder Materialdurchmischung im Facettenbereich, kombinierbar. Zudem kann das hier am Beispiel des InGaAs/GaAs Materialsystems entwickelte Konzept auf alle zur Herstellung von Halbleiterlaserdioden üblichen Materialsysteme übertragen werden und eröffnet so eine völlig neue, material- und wellenlängenunabhängige Möglichkeit Abstrahlcharakteristik und Ausgangsleistung von Laserdioden zu optimieren.
EINLEITUNG: Zur Behandlung der pAVK existieren verschiedene Behandlungsansätze. In dieser Studie soll die konservative intravenöse Therapieform, bestehend aus den Prostaglandinen Ilomedin® und Prostavasin® sowie dem Medikament Dusodril®, dargestellt werden. Die Schwerpunkte liegen auf der Betrachtung des Beinerhalts und des Überlebens sowie einem Vergleich zur Bypasstherapie und der Betrachtung des Kosten-Erlös-Verhältnisses. METHODEN: Anhand der SAP-Abrechnungsdatei, einer Datenbank und der Stationsbücher wurden alle Patienten der gefäßchirurgischen Station erfasst, die zwischen den Jahren 1993 und 2007 an der Universitätsklinik Würzburg auf Grund einer pAVK konservativ behandelt wurden. Zusätzlich wurden die entsprechenden Hausärzte anhand einer Follow-Up Studie zu dem weiteren Krankheitsverlauf der Patienten befragt. ERGEBNISSE: Von den insgesamt 94 Patienten litten 80 (85,1 %) an einer kritischen Ischämie. Je 40 (42,5 %) Patienten erhielten Ilomedin® und Prostavasin®, 14 (15 %) wurden mit dem Medikament Dusodril® therapiert. Die mittlere Therapiedauer lag bei 9,1 Tagen. Insgesamt mussten 18 Majoramputationen (19,1 %) durchgeführt werden, wobei besonders Patienten im Stadium 4 (88,8 %) betroffen waren (p=0,0024). Der 5-Jahres-Beinerhalt der Gesamtkollektivs lag bei 77,6 %. Schon im ersten Jahr nach Therapieende verstarben 22 % der Patienten, wobei sich sowohl die Herzinsuffizient (p=0,004), als auch eine KHK (p=0,008) als prognostisch ungünstig erwiesen. Ein Zusammenhang zwischen den pAVK-Stadien und den Überlebenschancen der jeweiligen Patienten konnte nicht festgestellt werden. Die mittlere Lebenserwartung des Kollektivs lag bei 3,5 Jahren. Sowohl in der Gruppe mit, als auch in der Gruppe ohne Revaskularisationsmöglichkeit war der Wert für den 5-Jahres- Beinerhalt für die Patienten ohne Bypass günstiger (94 % bzw. 75 %), als für die Patienten, die sich einer Bypassoperation unterzogen hatten (70 % bzw. 60 %). Der Kosten-Erlös-Vergleich zeigte eine Unterdeckung in allen Stadien der pAVK, welche im Stadium 2 besonders gravierend war (3751,38 € Kosten pro Patient vs. 1532 € Erlös [F65D] nach DRG-Prinzip). 80,7 % dieser Kosten sind auf den stationären Aufenthalt zurückzuführen. SCHLUSSFOLGERUNG: Die konservative intravenöse Therapieform der pAVK hat, auch bei einer deutlich kürzeren Therapiedauer als empfohlen, einen anhaltend amputationsvermeidenden und präventiven Effekt in Fällen. Bei Patienten mit einer Revaskularisationsmögichkeit scheint sie zudem eine Alternative zur Bypasstherapie darzustellen. Das Kosten-Erlös-Verhältnis ist allerdings, auf Grund der hohen anfallenden Kosten des stationären Aufenthalts, als ungünstig einzustufen.
Hintergrund: Die Alzheimer-Erkrankung ist die häufigste neurodegenerative Erkrankung. Da es zurzeit für sie noch keine Heilung gibt, richtet sich das Hauptaugenmerk auf eine möglichst frühe Diagnose und die Behandlung mit krankheitsverzögernden Medikamenten. Vor allem die funktionelle Bildgebung gilt im Bereich der Frühdiagnose als vielversprechend. Neben dem Gedächtnis werden die visuell-räumliche Informationsverarbeitung, exekutive Funktionen und Aufmerksamkeitsprozesse untersucht. Hierbei zeigen sich zentralnervöse Aktivierungsauffälligkeiten in kortikalen Zielregionen etwa im präfrontalen und im parietalen Kortex. Verlaufsuntersuchungen konzentrieren sich vor allem darauf aus der Gehirnaktivierung Vorhersagen über kognitive Veränderungen bei älteren Personen mit und ohne Gedächtnisstörung treffen zu können. Nur wenige Studien erfassen dabei jedoch die Gehirnaktivierung zu mehreren Messzeitpunkten. Gerade für große Stichproben und wiederholte Messungen könnte die funktionelle Nahinfrarotspektroskopie (fNIRS) sich als Alternative zur Magnetresonanztomographie anbieten. Ziel: Ziel der Studie war es, mit fNIRS und ereigniskorrelierten Potentialen funktionelle Unterschiede zwischen Alzheimer-Patienten und gleichaltrigen Kontrollen in mehreren Funktionsbereichen darzustellen und ihre Veränderung über den Zeitraum eines Jahres zu verfolgen. Zum ersten Mal sollte im Rahmen einer prospektiven Untersuchung mit fNIRS geprüft werden ob kortikale Aktivierungen zur Vorhersage von neuropsychologischen Testwerten genutzt werden können. Zusätzlich stellte sich die Frage, ob fNIRS für Verlaufsuntersuchungen an älteren Stichproben geeignet ist. Methoden: Im Rahmen der vorliegenden Arbeit wurden zum ersten Messzeitpunkt (T1) 73 Patienten und 71 Kontrollen mit vier Paradigmen in den drei Funktionsbereichen visuell-räumliche Informationsverarbeitung, exekutive Funktionen und zentralnervöse Filtermechanismen mit fNIRS und ereigniskorrelierten Potentialen gemessen. Die Probanden durchliefen eine Line Orientation Aufgabe, zwei Versionen einer Wortflüssigkeitsaufgabe (phonologisch und semantisch) und das P50-Doppelklickparadigma. Zielparameter waren dabei die aufgabenbezogene Aktivierung im parietalen Kortex, im dorsolateralen Präfrontalkortex (DLPFC) und das sensorische Gating, gemessen durch die P50-Supression nach wiederholter Reizdarbietung. Zusätzlich wurden zwei typische Tests zur Demenzdiagnostik (MMST und DemTect) erhoben. Die zweite Messung (T2) fand nach 12 Monaten statt und lief identisch zur ersten Untersuchung ab. Zu T2 konnten 14 Patienten und 51 Kontrollen erneut rekrutiert werden. Ergebnisse: Zu T1 konnte mit fNIRS ein Aktivierungsdefizit für Patienten im DLPFC während der phonologischen Wortflüssigkeitsaufgabe und im rechten Parietalkortex während der Line Orientation Aufgabe festgestellt werden. Für die semantische Wortflüssigkeitsaufgabe und das sensorische Gating zeigten sich keine zentralnervösen Unterschiede. Über das Jahr hinweg nahm die aufgabenbezogene Aktivierung der Patienten im linken DLPFC für beide Versionen der Wortflüssigkeitsaufgabe deutlich ab, während gleichaltrige Kontrollpersonen keine kortikalen Veränderungen zeigten. Zu T2 war das sensorische Gating der Patienten außerdem deutlich schlechter im Vergleich zu gesunden Kontrollen. Die Veränderungen der Oxygenierung während der Wortflüssigkeitsaufgabe konnten für gesunde Kontrollen Verschlechterungen im MMST und im DemTect vorhersagen. Vor allem ein Verlust der Lateralisierung ging mit einem Abfall in den kognitiven Tests einher. Schlussfolgerung: Spezifische Defizite in der kortikalen Aktivierung konnten bei Alzheimer-Patienten mit fNIRS beobachtet und genauer beschrieben werden. Auch die Veränderung im Verlauf eines Jahres ließ sich mit dieser Methode verfolgen. Für Längsschnittuntersuchungen, die sich mit der kortikalen Aktivierung als Prädiktor für dementielle Entwicklungen beschäftigen, bietet sich fNIRS somit als praktische Alternative zur fMRT an, zumal die gemessenen Veränderungen in der Oxygenierung auch prognostischen Wert für ältere Kontrollpersonen besaßen. Vor allem die funktionelle Lateralisierung in frontalen Kortexbereichen scheint als Prädiktor kognitiver Leistungen im Alter von Bedeutung zu sein.
Die vorliegende Arbeit befasst sich mit Untersuchungen von Element-Element- Bindungsaktivierungsreaktionen des dinuklearen Nickel(0)-NHC-Komplex [Ni2(iPr2Im)4(COD)] A mit verschiedenen reaktionsträgen Substraten, die ihrerseits wichtige Ausgangsstoffe für katalytische Anwendungen sind. Die Arbeit gliedert sich dabei in vier verschiedene Teile.
In der vorliegenden Studie wurde untersucht, ob sich bei Patienten mit Bronchialkarzinom durch eine Second-Line-Chemotherapie mit dem Tyrosinkinaseinhibitor Erlotinib ein Nutzen erzielen lässt. Dabei wurde besonderes Augenmerk auf Faktoren gelegt, die das progressionsfreie Überleben bei diesen Patienten möglicherweise beeinflussen können. Grundlage der Untersuchungen bildete ein Probanden- kollektiv von 42 Patienten, die sich mit der Diagnose nicht-kleinzelliges Bronchialkarzinom bis zum 31.12.2007 einer Second-Line-Chemotherapie mit Erlotinib unterzogen. Zu diesem Stichtag lagen für 23 Patienten Todesdaten vor. Das mediane progressionsfreie Überleben für alle untersuchten 42 Patienten betrug 3,88 Monate, wobei sich die Spanne von 0 bis 21 Monate erstreckte. Im Gesamten zeigten 59,52% der Patienten ein Ansprechen, das heißt, das progressionsfreie Überleben dauerte länger als einen Monat. Folgende allgemeine Patientenmerkmale wurden aus den Krankenblättern erhoben: Geschlecht, Alter, histologische Tumordiagnose, Tumorstadium nach Stadieneinteilung durch die TNM-Klassifikation, Metastasensorte soweit Metastasen vorlagen, Zeitpunkt der Erstdiagnose, Rauchverhalten, Allgemeinzustand nach Karnofsky-Index, Vorerkrankungen, Begleitmedikation, ausgewählte Laborwerte sowie Zeitraum und Art der First-Line-Therapie. Was die Second-Line-Therapie unter Erlotinib betrifft, so wurden hier gezielt im Verlauf die aktuelle Dosis, Änderungen der Begleittherapie, Allgemeinzustand nach Karnofsky-Index, Toxizität (besonderes Augenmerk verdienten Nebenwirkungen an der Haut und im Gastrointestinaltrakt), Auffälligkeiten von einigen Laborwerten und das Tumorverhalten untersucht. Zum Abschluss wurden das Ende der Erlotinibtherapie, Grund des Absetzens, aktueller Stand, eventuell im Anschluss an die Erlotinibtherapie durchgeführte Therapien sowie im Falle des Todes der Todeszeitpunkt erfasst. Im Gesamtüberblick konnten somit das progressionsfreie Überleben unter Erlotinib, die Einnahmedauer des Tyrosinkinaseinhibitors und das absolute Überleben seit der Erstdiagnose festgestellt werden. Hauptziel der Untersuchung war es, herauszufinden, inwiefern unsere Patienten unter Erlotinib im Sinne einer Verlängerung ihres progressionsfreien Überlebens profitieren und welche Faktoren die Ansprechrate beeinflussen. Der Einfluss ausgewählter Merkmale wurde statistisch untersucht. Im Vergleich mit Angaben in der Literatur konnte für das untersuchte Probandenkollektiv eine repräsentative Verteilung der Patientenmerkmale ermittelt und somit eine gewisse Relevanz der Ergebnisse dieser Studie angenommen werden. Statistisch signifikante Unterschiede ergaben sich für folgende Faktoren: Einnahmedauer, Krankheitsverhalten bei Therapiebeginn, Karnofsky-Index während der Behandlung, Nebenwirkungen an der Haut sowie Veränderungen in der Serumchemie. Nonresponder nahmen folglich Erlotinib wie erwartet statistisch signifikant kürzer ein, zeigten zu Beginn der Erlotinibtherapie statistisch signifikant häufiger ein fortschreitendes Krankheitsgeschehen, wiesen während der Therapie statistisch signifikant häufiger einen schlechteren Karnofsky-Index auf, erlitten statistisch signifikant weniger häufig schwere Nebenwirkungen an der Haut (Grad 3 und 4) und ihnen waren statistisch signifikant häufiger Serumwertveränderungen zuzuschreiben. Das prognostische Hauptmerkmal, das sowohl bei unserem Patientengut als auch in der Literatur signifikante Bedeutung hat, ist wohl, dass Responder statistisch signifikant häufiger schwere Nebenwirkungen an der Haut erleiden. Da Auftreten und Schweregrad von follikulären Ekzemen also mit einem positiven Ansprechen von Erlotinib korreliert sind, könnte es vielleicht nützlich sein, wenn man bereits vor Beginn der Erlotinibtherapie vorhersagen könnte, wie wahrscheinlich Patienten solche Nebenwirkungen entwickeln werden. Insgesamt kann man sagen, dass der Einsatz dieser molekular zielgerichteten Substanz zu einer Erweiterung der Therapiemöglichkeiten im palliativen Setting bei Bronchialkarzinompatienten geführt hat. Die Behandlung mit Erlotinib bedeutet ein Benefit für das Gesamtüberleben und das progressionsfreie Überleben bei Patienten mit fortgeschrittenem Bronchialkarzinom, die im Voraus eine Chemotherapie erhalten hatten. Dieses Benefit beschränkt sich nicht auf klinische Subgruppen wie weibliches Geschlecht, Adenokarzinom in der Histologie und Nichtraucher. Diese klinischen Parameter allein sind wohl folglich nicht ausreichend, um Patienten im Vornherein zu identifizieren, die wahrscheinlich von der Therapie profitieren. Vielmehr sind bessere Methoden notwendig, um möglicherweise voraussagen zu können, wer von dem Tyrosinkinaseinhibitor profitieren wird. Hoffnungen ruhen auf der Etablierung molekularer prognostischer und prädiktiver Marker in der klinischen Routine.
Growth factor induced signaling cascades are key regulatory elements in tissue development, maintenance and regeneration. Perturbations of these cascades have severe consequences, leading to developmental disorders and neoplastic diseases. As a major function in signal transduction, activating mutations in RAF family kinases are the cause of human tumorigenesis, where B-RAF V600E has been identified as the prevalent mutant. In order to address the oncogenic function of B-RAF V600E, we have generated transgenic mice expressing the activated oncogene specifically in lung alveolar epithelial type II cells. Constitutive expression of B-RAF V600E caused abnormalities in alveolar epithelium formation that led to airspace enlargements. These lung lesions showed signs of tissue remodeling and were often associated with chronic inflammation and low incidence of lung tumors. The inflammatory cell infiltration did not precede the formation of the lung lesions but was rather accompanied with late tumor development. These data support a model where the continuous regenerative process initiated by oncogenic B-RAF-driven alveolar disruption provides a tumor-promoting environment associated with chronic inflammation.

Inflammatory cell infiltration did not precede the formation of emphysema-like lesions but was rather accompanied with late tumor development. These data support a model where the continuous regenerative process initiated by oncogenic B-RAF-driven alveolar disruption provides a tumor-promoting environment associated with chronic inflammation. In the second project we focused on wild type B-RAF and its role in an oncogenic-C-RAF driven mouse lung tumor model. Toward this aim we have generated compound mice in which we could conditionally deplete B-RAF in oncogenic-C-RAF driven lung tumors. Conditional elimination of B-RAF did not block lung tumor formation however led to reduced tumor growth. The diminished tumor growth was not caused by increased cell death instead was a consequence of reduced cell proliferation. Moreover, B-RAF ablation caused a reduction in the amplitude of the mitogenic signalling cascade. These data indicate that in vivo B-RAF is dispensable for the oncogenic potential of active C-RAF; however it cooperates with oncogenic C-RAF in the activation of the mitogenic cascade.
The present study concerned mainly on the source, facies, and sedimentary environments of the Middle to Upper Jurassic strata in the Kerman and Tabas areas, east-central Iran. The composition of sandstones, and heavy mineral analysis point to pre-existing sedimentary, low, middle to upper rank metamorphic, and plutonic rocks of the Kalmard, Posht-e-Badam, Bayazeh, and Zarand-Kerman areas as the source rocks. According to the diagram of WELTJE et al. (1998), most samples from the Middle-Upper Jurassic rocks suggest a moderate to high elevation of the source area, and indicate a semi-arid and mediterranean to sub-humid climate. In the Qt-F-L ternary diagrams of DICKINSON et al. (1983), most point counting data from the Lower Siliciclastic Member and the top of the Hojedk Formation plot in the recycled orogen (Quartzose recycled) area of the diagram. The sandstones in this area can be interpreted as being derived from the Mid-Cimmerian Movements. Sixteen different types of siliciclastic-carbonate, and evaporatic sedimentary environments have been recognized. Thirty-nine macroinvertebrate taxa have been identified. Ten ichnotaxa have been taxonomically described from the Middle to Upper Jurassic rocks. Quite likely, before rotation of CEIM which were associated with counterclockwise block-rotation, equivalent rocks of the Bidou Formation occurred along the tectonic zone between the Yazd and the Tabas blocks (probably during the Middle Jurassic to Lower Cretaceous). However, from the Cretaceous onwards, most of the Bidou Formation has been removed by a combination of strike-slip and reverse movements of the Kashmar-Kerman tectonic zone. Roughly, these block-rotation movements occurred after the Cretaceous. During the Middle to Upper Jurassic, the tectonic activities were vertical movements producing the sedimentary pattern in the CEIM.
Background:
Retinitis pigmentosa (RP) is an inherited eye disease characterized by the progressive degeneration of rod photoreceptor cells. Mutations in pre-mRNA splicing factors including PRPF31 have been identified as cause for RP, raising the question how mutations in general factors lead to tissue specific defects.
Results:
We have recently shown that the zebrafish serves as an excellent model allowing the recapitulation of key events of RP. Here we use this model to investigate two pathogenic mutations in PRPF31, SP117 and AD5, causing the autosomal dominant form of RP. We show that SP117 leads to an unstable protein that is mislocalized to the rod cytoplasm. Importantly, its overexpression does not result in photoreceptor degeneration suggesting haploinsufficiency as the underlying cause in human RP patients carrying SP117. In contrast, overexpression of AD5 results in embryonic lethality, which can be rescued by wild-type Prpf31. Transgenic retina-specific expression of AD5 reveals that stable AD5 protein is initially localized in the nucleus but later found in the cytoplasm concurrent with progressing rod outer segment degeneration and apoptosis. Importantly, we show for the first time in vivo that retinal transcripts are wrongly spliced in adult transgenic retinas expressing AD5 and exhibiting increased apoptosis in rod photoreceptors.
Conclusion:
Our data suggest that distinct mutations in Prpf31 can lead to photoreceptor degeneration through different mechanisms, by haploinsufficiency or dominant-negative effects. Analyzing the AD5 effects in our animal model in vivo, our data imply that aberrant splicing of distinct retinal transcripts contributes to the observed retina defects.
Neurophysiologische Evidenz für eine Störung des impliziten Gedächtnis bei Alkoholabhängigkeit

Im Rahmen der vorliegenden Arbeit wurden 21 reine Alkoholpatienten, 6 polytoxikomane Alkoholpatienten und 18 gesunde Kontrollen neurophysiologisch untersucht. Basierend auf einem Paradigma zu Negativem Priming wurden Unterschiede zwischen den genannten Kollektiven bezüglich der Amplitude und Latenz der P300 im EEG untersucht. Kontrollpersonen zeigten in dieser Studie generell eine signifikant kürzere Latenz der P3a als beide Patientenkollektive, was als Hinweis auf eine kognitive Ineffizienz bei Alkoholpatienten gesehen werden kann. Darüber hinaus konnte gezeigt werden, dass reine Alkoholpatienten und polytoxikomane Alkoholpatienten bezüglich der Veränderungen der P300 getrennt betrachtet werden müssen, da sich signifikante Unterschiede bezüglich Latenz und Amplitude zwischen den beiden Patientenkollektiven zeigten. Ebenso gibt es Hinweise darauf, dass bei Studien zur P300 Geschlechterunterschiede berücksichtigt werden müssen. Mit vorliegender Studie konnte zudem die Theorie einer prominenten frontocentralen Verteilung der P3a unterstützt werden.
