"Eignet sich die kritische Flimmerfrequenz zur Diagnose einer minimal hepatischen Enzephalopathie?"

Korrelation und Kontingenzprüfung von Kritischer Flimmerfrequenz als diagnostischem Mittel bei minimal hepatischer Enzephalopathie mit anderen etablierten diagnostischen Mitteln und beschreibenden Parametern.
In den Ergebnissen lediglich Korrelation mit Alertness Testung in der Testbatterie.
Minimal hepatische Enzephalopathie braucht zur Diagnostik mindestens 2 verschiedene ergänzende diagnostische Verfahren (neuropsychologisch und -physiologisch), um sicher entdeckt werden zu können. Bei nur einem Testverfahren blieben zahlreiche Betroffene unentdeckt. Möglicherweise ist das verschiedenen pathophysiologischen Subgruppen geschuldet.
71 Studierende nahmen am Universitätsklinikum Würzburg in der Abteilung für Zahnärztliche Prothetik an einem freiwilligen Übungsseminar zum Aufpassen von Kronen mit Störstellen, die im 3D-Druckverfahren hergestellt wurden, teil. Das Übungsseminar fand an zwei Terminen statt. Zum Identifizieren der Störstellen standen Xantopren und Okklusionsspray zur Verfügung. Nach dem praktischen Teil der Übung wurde ein Fragebogen ausgefüllt. Zusätzlich wurden die aufgepassten Kronen mittels Laborscanner digitalisiert und mit einer Krone ohne Störstellen überlagert. Dadurch konnten positive und negative Oberflächenabweichungen für die Bereiche der Störstellen sowie der Gesamtinnenfläche der Kronen ermittelt werden.
Die flächenbezogenen Abweichungswerte zeigten einen signifikanten Lernerfolg – gemessen anhand der Passungsparameter - zwischen den beiden Terminen des Übungsseminars. Hierbei erreichten Kronen, die mit Okklusionsspray aufgepasst wurden, signifikant geringere flächenbezogene Abweichungswerte im Vergleich zu Kronen, die mit Xantopren aufgepasst wurden.
Die Auswertung der mit Schulnoten skalierten Fragen ergab signifikante Unterschiede bei der Bewertung der Härte, eines realitätsnahen Gefühls beim Einschleifen bzw. beim Aufpassen und Details wie Randschluss. Beim Vergleich der Aufpassmethoden im Fragebogen ergaben die Einfachheit beim Aufpassen, das Identifizieren der Störstellen und das präferierte Material signifikante Unterschiede. Der subjektive Lernerfolg mit den Materialien zeigte ebenfalls signifikante Unterschiede. Insbesondere die Materialeigenschaften und die Randgenauigkeit der Druckkronen wurden häufig kritisiert, die schnelle und einfache Möglichkeit zur Herstellung von Übungsmaterialien sowie deren Reproduzierbarkeit wurden von den Studierenden hingegen begrüßt.
In der vorliegenden Arbeit präparierten Studierende 3D-gedruckte Übungszähne, in denen die korrekte Präparation eines Veneers farblich abgesetzt war. Die neue Lehrmethode wurde durch die Teilnehmer in einem Fragebogen evaluiert und zusätzlich wurden die Präparationen digital mit einer Referenzpräparation verglichen. Die Teilnehmer des praktischen Kurses schätzten die Zweischichttechnik als gute Lehrmethode ein (Ø 2,0 ± 0,37) und gaben zahlreiche Vorteile der Zweischichttechnik an.
Die digitale Auswertung der präparierten Zähne konnte unter den Limitationen der vorliegenden Studie keine signifikant schlechtere Präparationsqualität nach zweimaligem Präparieren von einschichtigen Modellzähnen als nach zweimaligem Präparieren von zweischichtigen Übungszähnen nachweisen (p = 0,91).
Der Lernerfolg der Studierenden erwies sich durch in Zweischichttechnik gedruckte Zähne mit integriertem Veneer nicht besser als durch einschichtige Modellzähne (〖ΔL〗_A= -0,01; 〖ΔL〗_B= -0,03). Der Unterschied zwischen den Präparationsergebnissen des ersten und vierten Durchgangs war allerdings nicht signifikant (Gruppe A: Ø GMW+/- 0,17 ± 0,07 → Ø GMW+/- 0,18 ± 0,05, p = 0,317; Gruppe B: Ø GMW+/- 0,15 ± 0,07 → 0,18 ± 0,09, p = 0.066). Gründe hierfür könnten unter anderem Ermüdung und sinkende Motivation während des praktischen Kurses gewesen sein. Diesem Problem könnte Rechnung getragen werden, indem folgende Studien an mehreren Terminen durchgeführt werden. Auch eine mögliche Fokussierung der Studierenden auf das Ablösen der oberen Schicht sowie die unterschiedliche Härte der beiden Schichten könnten einen besseren Lernerfolg mit zweischichtigen Übungszähnen verhindert haben.
Die Teilnehmer, die ihre manuellen Fertigkeiten als besonders gut einschätzen, präparierten mit einer durchschnittlichen mittleren absoluten Abweichung von 0,17 ± 0,07 nicht signifikant besser als die Teilnehmer mit geringer Selbsteinschätzung, welche eine mittlere absolute Abweichung von 0,16 ± 0,05 (p = 0 ,967) erreichten.
Vitamin B6 deficiency has been linked to cognitive impairment in human brain disorders for decades. Still, the molecular mechanisms linking vitamin B6 to these pathologies remain poorly understood, and whether vitamin B6 supplementation improves cognition is unclear as well. Pyridoxal phosphatase (PDXP), an enzyme that controls levels of pyridoxal 5’-phosphate (PLP), the co-enzymatically active form of vitamin B6, may represent an alternative therapeutic entry point into vitamin B6-associated pathologies. However, pharmacological PDXP inhibitors to test this concept are lacking. We now identify a PDXP and age-dependent decline of PLP levels in the murine hippocampus that provides a rationale for the development of PDXP inhibitors. Using a combination of small molecule screening, protein crystallography and biolayer interferometry, we discover and analyze 7,8-dihydroxyflavone (7,8-DHF) as a direct and potent PDXP inhibitor. 7,8-DHF binds and reversibly inhibits PDXP with low micromolar affinity and sub-micromolar potency. In mouse hippocampal neurons, 7,8-DHF increases PLP in a PDXP-dependent manner. These findings validate PDXP as a druggable target. Of note, 7,8-DHF is a well-studied molecule in brain disorder models, although its mechanism of action is actively debated. Our discovery of 7,8-DHF as a PDXP inhibitor offers novel mechanistic insights into the controversy surrounding 7,8-DHF-mediated effects in the brain.

Abstract
Fabry disease (FD) is a life-limiting disorder characterized by intracellular globotriaosylceramide (Gb3) accumulations. The underlying α-galactosidase A (α-GAL A) deficiency is caused by variants in the gene GLA. Variants of unknown significance (VUS) are frequently found in GLA and challenge clinical management. Here, we investigated a 49-year old man with cryptogenic lacunar cerebral stroke and the chance finding of the VUS S126G, who was sent to our center for diagnosis and initiation of a costly and life-long FD-specific treatment. We combined clinical examination with in vitro investigations of dermal fibroblasts (HDF), induced pluripotent stem cells (iPSC), and iPSC-derived sensory neurons. We analyzed α-GAL A activity in iPSC, Gb3 accumulation in all three cell types, and action potential firing in sensory neurons. Neurological examination and small nerve fiber assessment was normal except for reduced distal skin innervation. S126G iPSC showed normal α-GAL A activity compared to controls and no Gb3 deposits were found in all three cell types. Baseline electrophysiological characteristics of S126G neurons showed no difference compared to healthy controls as investigated by patch-clamp recordings. We pioneer multi-level cellular characterization of the VUS S126G using three cell types derived from a patient and provide further evidence for the benign nature of S126G in GLA, which is of great importance in the management of such cases in clinical practice.
This thesis provides an edition and commentary of a manuscript discovered by Michael Stolberg in the archives of the central library in Zurich under the title “Mon aprendisage à l'Hôtel Dieu de Paris 1704.” (My apprenticeship at the Hôtel-Dieu de Paris 1704). The manuscript contains records of a midwifery student at the Hôtel-Dieu de Paris, an old hospital famous among others for its education in midwifery in the maternity ward. We read about managing different births, recipes for common remedies, direct questions answered by the maîtresse sage-femme, the leading midwife at the Hôtel-Dieu de Paris and more.
Although other accounts exist of the maternity ward at the Hôtel-Dieu de Paris, \(Mon\) \(Aprendisage\) is the first and only account from a midwife’s perspective that gives more than just instructions on obstetrical techniques. It takes us into the day-to-day experience of a woman as she progressed through her training at the Hôtel-Dieu.
A novel USP11-TCEAL1-mediated mechanism protects transcriptional elongation by RNA Polymerase II

Deregulated expression of MYC oncoproteins is a driving event in many human cancers. Therefore, understanding and targeting MYC protein-driven mechanisms in tumor biology remain a major challenge.
Oncogenic transcription in MYCN-amplified neuroblastoma leads to the formation of the MYCN-BRCA1-USP11 complex that terminates transcription by evicting stalling RNAPII from chromatin. This reduces cellular stress and allows reinitiation of new rounds of transcription. Basically, tumors with amplified MYC genes have a high demand on well orchestration of transcriptional processes-dependent and independent from MYC proteins functions in gene regulation. To date, the cooperation between promoter-proximal termination and transcriptional elongation in cancer cells remains still incomplete in its understanding.
In this study the putative role of the dubiquitinase Ubiquitin Specific Protease 11 (USP11) in transcription regulation was further investigated. First, several USP11 interaction partners involved in transcriptional regulation in neuroblastoma cancer cells were identified. In particular, the transcription elongation factor A like 1 (TCEAL1) protein, which assists USP11 to engage protein-protein interactions in a MYCN-dependent manner, was characterized. The data clearly show that TCEAL1 acts as a pro-transcriptional factor for RNA polymerase II (RNAPII)-medi- ated transcription. In detail, TCEAL1 controls the transcription factor S-II (TFIIS), a factor that assists RNAPII to escape from paused sites. The findings claim that TCEAL1 outcompetes the transcription elongation factor TFIIS in a non-catalytic manner on chromatin of highly expressed genes. This is reasoned by the need regulating TFIIS function in transcription. TCEAL1 equili- brates excessive backtracking and premature termination of transcription caused by TFIIS.
Collectively, the work shed light on the stoichiometric control of TFIIS demand in transcriptional regulation via the USP11-TCEAL1-USP7 complex. This complex protects RNAPII from TFIIS-mediated termination helping to regulate productive transcription of highly active genes in neuroblastoma.
Barth Syndrome (BTHS) is an inherited X-chromosomal linked disorder, characterized by early development of cardiomyopathy, immune system defects, skeletal muscle myopathy and growth retardation. The disease displays a wide variety of symptoms including heart failure, exercise intolerance and fatigue due to the muscle weakness. The cause of the disease are mutations in the gene encoding for the mitochondrial transacylase Tafazzin (TAZ), which is important for remodeling of the phospholipid cardiolipin (CL). All mutations result in a pronounced decrease of the functional enzyme leading to an increase of monolysocardiolipin (MLCL), the precursor of mature CL, and a decrease in mature CL itself. CL is a hallmark phospholipid of mitochondrial membranes, highly enriched in the inner mitochondrial membrane (IMM). It is not only important for the formation of the cristae structures, but also for the function of different protein complexes associated with the mitochondrial membrane. Reduced levels of mature CL cause remodeling of the respiratory chain supercomplexes, impaired respiration, defects in the Krebs cycle and a loss of mitochondrial calcium uniporter (MCU) protein. The defective Ca2+ handling causes impaired redox homeostasis and energy metabolism resulting in cellular arrhythmias and defective electrical conduction. In an uncompensated situation, blunting mitochondrial Ca2+ uptake provokes increased mitochondrial emission of H2O2 during workload transitions, related to oxidation of NADPH, which is required to regenerate anti-oxidative enzymes. However, in the hearts and cardiac myocytes of mice with a global knock-down of the Taz gene (Taz-KD), no increase in mitochondrial ROS was observed, suggesting that other metabolic pathways may have compensated for reduced Krebs cycle activation.
The healthy heart produces most of its energy by consuming fatty acids. In this study, the fatty acid uptake into mitochondria and their further degradation was investigated, which showed a switch of the metabolism in general in the Taz-KD mouse model. In vivo studies revealed an increase of glucose uptake into the heart and decreased fatty acid uptake and oxidation. Disturbed energy conversion resulted in activation of retrograde signaling pathways, implicating overall changes in the cell metabolism. Upregulated integrated stress response (ISR) was confirmed by increased levels of the downstream target, i.e., the activating transcription factor 4 (ATF4). A Tafazzin knockout mouse embryonal fibroblast cell model (TazKO) was used to inhibit the ISR using siRNA transfection or pharmaceutical inhibition. This verified the central role of

the ISR in regulating the metabolism in BTHS. Moreover, an increased metabolic flux into glutathione biosynthesis was observed, which supports redox homeostasis. In vivo PET-CT scans depicted elevated activity of the xCT system in the BTHS mouse heart, which transports essential amino acids for the biosynthesis of glutathione precursors. Furthermore, the stress induced signaling pathway also affected the glutamate metabolism, which fuels into the Krebs cycle via -ketoglutarate and therefore supports energy converting pathways. In summary, this thesis provides novel insights into the energy metabolism and redox homeostasis in Barth syndrome cardiomyopathy and its regulation by the integrated stress response, which plays a central role in the metabolic alterations. The aim of the thesis was to improve the understanding of these metabolic changes and to identify novel targets, which can provide new possibilities for therapeutic intervention in Barth syndrome.
The transition to school is a key juncture in an individual’s educational trajectory, with far-reaching effects on the development of children and their families. Successful transitions require flexibility in the design of the transition process, addressing the needs of the persons involved in an adaptive manner. Adaptivity is also considered crucial for the success of inclusive transitions. However, a systematic breakdown of the aspects that characterize the concept of adaptivity in the context of inclusive school entry is not available at this point. This article therefore provides a conceptualization of adaptivity in the inclusive transition to school as well as a review of the current literature focusing this topic. The goal is to develop a model that structures the various aspects of adaptivity at school entry and offers an overview of the way these aspects are important to design the transition successfully according to current findings of empirical research. Building on a concept of transitions informed by ecological systems theory, we are guided by the assumption that adaptivity at transition to school may occur in three forms: as a feature of the persons involved in the transition; as a feature of the processes that moderate the course of the transition; and as a feature of the structures that frame the transition. Based on this distinction, we develop a model that presents adaptivity in the inclusive transition to school.
Human prosociality, encompassing generosity, cooperation, and volunteering, holds a vital role in our daily lives. Over the last decades, the question of whether prosociality undergoes changes over the adult lifespan has gained increased research attention. Earlier studies suggested increased prosociality in older compared to younger individuals. However, recent meta-analyses revealed that this age effect might be heterogeneous and modest. Moreover, the contributing factors and mechanisms behind these age-related variations remain to be identified. To unravel age-related differences in prosociality, the first study of this dissertation employed a meta-analytical approach to summarize existing findings and provide insight into their heterogeneity by exploring linear and quadratic age effects on self-reported and behavioral prosociality. Additionally, two empirical research studies investigated whether these age-related differences in prosociality were observed in real life, assessed through ecological momentary assessment (Study 2), and in a controlled laboratory setting by applying a modified dictator game (Study 3). Throughout these three studies, potential underlying behavioral and computational mechanisms were explored. The outcome of the meta-analysis (Study 1) revealed small linear age effects on prosociality and significant age group differences between younger and older adults, with higher levels of prosociality in older adults. Explorative evidence emerged in favor of a quadratic age effect on behavioral prosociality, indicating the highest levels in midlife. Additionally, heightened prosocial behavior among middle-aged adults was observed compared to younger adults, whereas no significant differences in prosocial behavior were noted between middle-aged and older adults. Situational and contextual features, such as the setting of the study and specific paradigm characteristics, moderated the age-prosociality relationship, highlighting the importance of the (social) context when studying prosociality. For Study 2, no significant age effect on real-life prosocial behavior was observed. However, evidence for a significant linear and quadratic age effect on experiencing empathy in real life emerged, indicating a midlife peak. Additionally, across all age groups, the link between an opportunity to empathize and age significantly predicted real-life prosocial behavior. This effect, indicating higher levels of prosocial behavior when there was a situation possibly evoking empathy, was most pronounced in midlife. Study 3 presented age differences in how older and younger adults integrate values related to monetary gains for self and others to make a potential prosocial decision. Younger individuals effectively combined both values in a multiplicative fashion, enhancing decision-making efficiency. Older adults showed an additive effect of values for self and other and displayed increased decision-making efficiency when considering the values separately. However, among older adults, individuals with better inhibitory control were better able to integrate information about both values in their decisions. Taken together, the findings of this dissertation offer new insights into the multi-faceted nature of prosociality across adulthood and the mechanisms that help explain these age-related disparities. While this dissertation observed increasing prosociality across the adult lifespan, it also questions the assumption that older adults are inherently more prosocial. The studies highlight midlife as a potential peak period in social development but also emphasize the importance of the (social) context and that different operationalizations might capture distinct facets of prosociality. This underpins the need for a comprehensive framework to understand age effects of prosociality better and guide potential interventions.

Graphs provide a key means to model relationships between entities.
They consist of vertices representing the entities,
and edges representing relationships between pairs of entities.
To make people conceive the structure of a graph,
it is almost inevitable to visualize the graph.
We call such a visualization a graph drawing.
Moreover, we have a straight-line graph drawing
if each vertex is represented as a point
(or a small geometric object, e.g., a rectangle)
and each edge is represented as a line segment between its two vertices.
A polyline is a very simple straight-line graph drawing,
where the vertices form a sequence according to which the vertices are connected by edges.
An example of a polyline in practice is a GPS trajectory.
The underlying road network, in turn, can be modeled as a graph.
This book addresses problems that arise
when working with straight-line graph drawings and polylines.
In particular, we study algorithms
for recognizing certain graphs representable with line segments,
for generating straight-line graph drawings,
and for abstracting polylines.
In the first part, we first examine,
how and in which time we can decide
whether a given graph is a stick graph,
that is, whether its vertices can be represented as
vertical and horizontal line segments on a diagonal line,
which intersect if and only if there is an edge between them.
We then consider the visual complexity of graphs.
Specifically, we investigate, for certain classes of graphs,
how many line segments are necessary for any straight-line graph drawing,
and whether three (or more) different slopes of the line segments
are sufficient to draw all edges.
Last, we study the question,
how to assign (ordered) colors to the vertices of a graph
with both directed and undirected edges
such that no neighboring vertices get the same color
and colors are ascending along directed edges.
Here, the special property of the considered graph is
that the vertices can be represented as intervals
that overlap if and only if there is an edge between them.
The latter problem is motivated by an application
in automated drawing of cable plans with vertical and horizontal line segments,
which we cover in the second part.
We describe an algorithm that
gets the abstract description of a cable plan as input,
and generates a drawing that takes into account
the special properties of these cable plans,
like plugs and groups of wires.
We then experimentally evaluate the quality of the resulting drawings.
In the third part, we study the problem of abstracting (or simplifying)
a single polyline and a bundle of polylines.
In this problem, the objective is to remove as many vertices as possible from the given polyline(s)
while keeping each resulting polyline sufficiently similar to its original course
(according to a given similarity measure).
Aufgrund der Beeinträchtigung eines oder beider Beine sind viele Menschen zur selbstständigen Fortbewegung im Alltag auf ein Hilfsmittel angewiesen. Gehstützen sind das aufgrund hoher Nutzerfreundlichkeit am häufigsten eingesetzte Hilfsmittel bei kurz oder langandauernder Funktionseinschränkung.
Die im Vergleich zum normalen Gang erhöhte Belastung der oberen Extremitäten kann in schwerwiegenden Komplikationen wie Thrombosen, Gewebe- und Nervenschäden resultieren. Um diese möglichst gering zu halten, wurden im Laufe der Zeit verschiedene Gehstützenmodelle entwickelt.
Ziel der durchgeführten Studie war es, die Belastung der Hände bei verschiedenen hier zu Lande häufig genutzten Gehstützenmodellen und verschiedenen Entlastungen zu erfassen und diese miteinander zu vergleichen. Hierfür zogen wir 26 gesunde Probanden heran, die mit ergonomischen UAG, Arthritis-Gehstützen und anatomischen UAG eine definierte Strecke in festgesetzter, für alle gleicher Reihenfolge liefen. Die zwischen der Hand und dem Gehstützengriff angebrachten Sensormatten erfassten im ersten Studienteil die auf die komplette Hand einwirkende Last und im zweiten Studienteil die Belastung der radialen und ulnaren Hohlhandhälfte separat. Sensorsohlen erfassten zeitgleich die Bodenreaktionskraft.
Die Studie zeigte, dass die mit bis zu 70% des Körpergewichts (KG) beim Gehen mit UAG stark belastete Hand mit Arthritis-Gehstützen deutlich entlastet werden kann. Die Belastung konnte auf 16% verringert werden. Eine schwierigere Handhabe muss jedoch in Kauf genommen werden.
Bei Beschwerden der Hand, besonders im ulnaren Bereich, sollten anatomische UAG gegenüber ergonomischen UAG bevorzugt werden. Die Belastung der ulnaren Handfläche lag hiermit mit 17% des KG deutlich unter der Belastung mit ergonomischen UAG mit 23% des KG. Die Belastung der radialen Hälfte unterschied sich nicht.
Da die Belastung der Hand mit steigender Beinentlastung stieg, sollte die Entlastung möglichst gering und zeitlich kurzgehalten werden.
Die Maus mit progressiver motorischer Neuropathie (PMN) ist ein Modell für eine vererbte motorische Neuropathie mit progressiver Neurodegeneration. Die Degeneration der Axone geht mit homozygoten Mutationen des TBCE- Gens einher, das für das Tubulin-Chaperon-E- Protein kodiert. TBCE ist für die korrekte Dimerisierung von Alpha- und Beta-Tubulin verantwortlich. Wirkung von peg-IGF-1 auf das auditorische System durch Behandlung ab dem 15. postnatalen Tag (p15). Die histologische Analyse ergab positive Auswirkungen auf die OHC-Synapsen der medialen olivocochleären (MOC) neuronalen Fasern und eine kurzfristige Abschwächung des OHC-Verlustes. Peg-IGF-1 war in der Lage, die Desorganisation der OHC-Synapsen bedingt wiederherzustellen und die Bereitstellung von cholinerger Acetyltransferase in den Präsynapsen aufrechtzuerhalten. Zur Beurteilung der auditorischen Funktion wurden frequenzspezifische Hirnstammreaktionen und otoakustische Emissionen mit Verzerrungsprodukten bei Tieren mit p21 und p28 aufgezeichnet. Trotz der positiven Auswirkungen auf die MOC-Fasern und die OHC konnte jedoch keine Wiederherstellung des Hörvermögens erreicht werden. Die vorliegende Arbeit zeigt, dass die synaptische Pathologie der efferenten MOC-Fasern in PMN-Mäusen eine besondere Form der "efferenten auditorischen Neuropathie" darstellt. Peg-IGF-1 zeigte eine otoprotektive Wirkung, indem es die Degeneration von OHCs und efferenten Synapsen verhinderte. Es sind jedoch verstärkte Anstrengungen zur Optimierung der Behandlung erforderlich, um nachweisbare Verbesserungen der Hörleistung zu erzielen.
This paper examines the potential reinforcement of motivated beliefs when individuals with identical biases communicate. We propose a controlled online experiment that allows to manipulate belief biases and the communication environment. We find that communication, even among like-minded individuals, diminishes motivated beliefs if it takes place in an environment without previously declared external opinions. In the presence of external plural opinions, however, communication does not reduce but rather aggravates motivated beliefs. Our results indicate a potential drawback of the plurality of opinions - it may create communication environments wherein motivated beliefs not only persist but also become contagious within social networks.
N\(^6\)-methyladenosine (m\(^6\)A) is an important modified nucleoside in cellular RNA associated with multiple cellular processes and is implicated in diseases. The enzymes associated with the dynamic installation and removal of m\(^6\)A are heavily investigated targets for drug research, which requires detailed knowledge of the recognition modes of m\(^6\)A by proteins. Here, we use atomic mutagenesis of m\(^6\)A to systematically investigate the mechanisms of the two human m\(^6\)A demethylase enzymes FTO and ALKBH5 and the binding modes of YTH reader proteins YTHDF2/DC1/DC2. Atomic mutagenesis refers to atom-specific changes that are introduced by chemical synthesis, such as the replacement of nitrogen by carbon atoms. Synthetic RNA oligonucleotides containing site-specifically incorporated 1-deaza-, 3-deaza-, and 7-deaza-m\(^6\)A nucleosides were prepared by solid-phase synthesis and their RNA binding and demethylation by recombinant proteins were evaluated. We found distinct differences in substrate recognition and transformation and revealed structural preferences for the enzymatic activity. The deaza m\(^6\)A analogues introduced in this work will be useful probes for other proteins in m\(^6\)A research.
Auf der Suche nach Frieden. Zur Gestaltung von Nachkriegsordnungen – eine archäologische Spurensuche

Ein in der archäologischen Konfliktforschung bislang wenig beachtetes Thema, die Geschehnisse nach Kampfhandlungen und die Etablierung einer Nachkriegsordnung, verbunden mit der Entstehung symbolischer Landschaften als Element einer Erinnerungskultur, behandelt Stefan Burmeister, der unter anderem die Ereignisse nach dem sog. Karfreitagsgefecht der Bundeswehr am 2. April 2010 in Afghanistan und die Rolle der materiellen Kultur in diesem Kontext analysiert.
Die ESWT ist eine nichtoperative Option, kann aber auch ergänzend zur operativen Therapie zur Behandlung von verzögerter (Knochen-)Heilung (VH) und Pseudarthrosen (PA) eingesetzt werden. Ihre Wertigkeit sowie beeinflussende Faktoren wurden an der oberen Extremität bisher nicht ausreichend quantifiziert. Sechzig Patienten wurden retrospektiv nach Anwendung einer ESWT hinsichtlich Ausheilungsrate und Konsolidierungszeit untersucht.
Bei 70 % der Patienten kam es zur Ausheilung. Das Durchschnittsalter der Geheilten und Nichtgeheilten unterschied sich nicht signifikant. Das Rauchverhalten und die Zeit zwischen Trauma/OP und ESWT war bei Geheilten und Nichtgeheilten ohne signifikanten Unterschied. Die Ausheilungsrate war am höchsten an Mittelhandknochen/Finger/Daumen (91 %), gefolgt von Unterarmschaft (88 %), Epi-/Metaphyse des Unterarms (67 %) und zuletzt Handwurzelknochen (59 %). Nach konservativer Vorbehandlung heilten 55 %, bei > 2 Voroperationen 67 %, ohne Vorbehandlung 73 % und nach 1 Voroperation 75 %. Die weitere Analyse hinsichtlich der operativen Vorversorgung ergab nach alleiniger ORIF 85 %, ohne Voroperation 64 % und nach ORIF mit Knochenanfrischung/-transplantation 57 % Heilungsrate. Bei intraoperativer ESWT kombiniert mit Knochendebridement/-transplantation + ORIF heilten 67 %, kombiniert mit einer alleinigen ORIF 86 %. Bei alleiniger ESWT oder mit nur minimalen Maßnahmen konsolidierten 70 %.
Die ESWT ist in jedem Stadium der Knochenheilungsstörung gleich wirksam. Die Prinzipien von Stabilität und Auffüllung bei vorhandenen Knochendefekten müssen auch bei der Anwendung der ESWT berücksichtigt werden, dann wirkt die ESWT alleinig oder kombiniert mit einer Operation gleichermaßen. Der negative Einfluss von Knochendefekten/-resorption ist auch mit ESWT noch nachweisbar. Ebenso ist die Behandlung des Kahnbeins problematischer als übrige Lokalisationen. Eine vorangegangene Operation stellt keinen negativen Faktor dar, auch wenn Fremdmaterial einliegt.
Auswirkungen der Genpolymorphismen ASIC1, BDNF und NPSR1 auf die Antizipationsphase aversiver Reize

In dieser Arbeit wurden einerseits die Antizipationsphasen von aversiven gegenüber neutralen Reizen anhand von Messungen der Hautleitfähigkeit und der Startle-Reaktion untersucht. Andererseits wurde die Hautleitfähigkeit auch während der Präsentation aversiver und neutraler Reize mit dem Ziel gemessen, signifikante Unterschiede festzustellen. Insbesondere wurden die Auswirkungen der Allele der Gene ASIC1 und der Interaktion der Genallele BDNF und NPSR1 betrachtet.
Ziel der vorliegenden Arbeit war es, den Einfluss der Risikogene auf die physiologische Angstreaktion und die subjektive Angstwahrnehmung zu untersuchen. Hierzu wurden den genotypisierten Probanden aversive und neutrale Videos präsentiert. Vor jedem Video erfolgte die Ankündigung, ob es sich um ein neutrales oder aversives Video handelt, wodurch bei Letzterem im Allgemeinen antizipatorische Angst – Erwartungsangst – hervorgerufen wird.
Im Vergleich der Antizipationsphase vor Darbietung aversiver Videos mit der Antizipationsphase vor neutralen Videos konnte eine erhöhte Startle-Amplitude gemessen werden. Jedoch konnte weder anhand der Veränderung der Hautleitfähigkeit noch anhand der Startle-Amplitude ein signifikanter Unterschied bei Trägern und Nicht- Trägern der Risikogenallelen in der Antizipationsphase festgestellt werden. Während der Präsentation der Videos konnte für die aversiven Videos im Vergleich zu den neutralen eine erhöhte Hautleitfähigkeit gemessen werden. Ebenfalls konnte bei der Darbietung von aversiven Videos bei den Trägern der Genallel-Interaktion NPSR1 AT/TT * BDNF GG und den Trägern des Risikogenallels ASIC1TT eine erhöhte Hautleitfähigkeit gemessen werden. So konnte mit den Ergebnissen dieser Arbeit belegt werden, dass Antizipationsangst auslösbar und anhand der Startle-Amplitude messbar ist.
Um Antizipationsangst festzustellen oder diese bei Risikogenallel-Träger zu untersuchen, waren die Ergebnisse bezüglich der Hautleitfähigkeit jedoch weniger aussagekräftig als erwartet.
Allgemein konnte die Interaktion NPSR1 AT/TT * BDNF GG und ASIC1 TT als Risikogenallele bezüglich einer verstärken Reaktion auf aversive Reize bestätigt werden. Weitere Studien sind notwendig, um die genetische Komponente von Angst und damit auch von Angsterkrankungen näher zu beleuchten, damit zukünftige Diagnostik- und Therapieansätze präzise entwickelt werden können.
