Within the scope of this thesis two main topics have been investigated: the examination of micromagnetic sensors and transport of massive and massless Dirac fermions in HgTe quantum wells. For the investigation of localized, inhomogeneous magnetic fields, the fabrication and characterization of two different non-invasive and ultra sensitive sensors has been established at the chair ”Experimentelle Physik” of the University of Würzburg. The first sensor is based on the young technique named micro-Hall magnetometry. The necessary semiconductor devices (Hall cross structures) were fabricated by high-resolution electron beam lithography based on two different two dimensional electron gases (2DEGs), namely InAs/(Al,Ga)Sb- and HgTe/(Hg,Cd)Te- heterostructures. The characteristics have been examined in two different ways. Measurements in homogeneous magnetic fields served for characterization of the sensors, whereas the investigation of artificially produced sub-µm magnets substantiates the suitability of the devices for the study of novel nanoscale magnetic materials (e.g. nanowires). Systematic experiments with various magnets are in accordance with the theory of single-domain particles and anisotropic behavior due to shapes with high aspect ratio. The highest sensitivity for strongly localized fields was obtained at T = 4.2 K for a (200x200) nm^2 Hall cross - made from shallow, high mobility HgTe 2DEG. Although the field resolution was merely δB ≈ 100 µT, the nanoscale sensor size yields an outstanding flux resolution of δΦ = 2 10^(−3) Φ0, where Φ0 = h/2e is the flux quantum. Translating this result in terms of magnetic moment, the sensitivity allows for the detection of magnetization changes of a particle centered on top of the sensor as low as δM ≈ 10^2 µB, with the magnetic moment of a single electron µB, the Bohr magneton. The further examination of a permalloy nanomagnet with a cross-section of (100x20) nm^2 confirms the expected resolution ability, extracted from the noise of the sensor. The observed high signal-to-noise ratio validates the detection limit of this sensor in terms of geometry. This would be reached for a magnet (same material) with quadratic cross-section for an edge length of 3.3 nm. Moreover, the feasibility of this sensor for operation in a wide temperature range (T = mK... > 200 K) and high magnetic fields has been confirmed. The second micromagnetic sensor is the micro-SQUID (micro-Superconducting-QUantum-Interference-Device) based on niobium. The typical sensor area of the devices built in this work was (1.0x1.0) µm^2, with constrictions of about 20 nm. The characterization of this device demonstrates an amazing field sensitivity (regarding its size) of δB < 1 µT. Even though the sensor was 25 times larger than the best micro-Hall sensor, it provided an excellent flux resolution in the order of δΦ ≈ 5 10^(−4) Φ0 and a similar magnetic moment resolution of δM ≈ 10^2 µB. Furthermore, the introduction of an ellipsoidal permalloy magnet (axes: 200 nm and 400 nm, thickness 30 nm) substantiates the suitability for the detection of minuscule, localized magnetic fields. The second part of the thesis deals with the peculiar transport properties of HgTe quantum wells. These rely on the linear contribution to the band structure inherent to the heterostructure. Therefore the system can be described by an effective Dirac Hamiltonian, whose Dirac mass is tunable by the variation of the quantum well thickness. By fabrication and characterization of a systematical series of substrates, a system with vanishing Dirac mass (zero energy gap) has been confirmed. This heterostructure therefore resembles graphene (a monolayer of graphite), with the difference of exhibiting only one valley in the energy dispersion of the Brillouin zone. Thus parasitical intervalley scattering cannot occur. The existence of this system has been proven by the agreement of theoretical predictions, based on widely accepted band structure calculations with the experiment (Landau level dispersion, conductivity). Furthermore, another particularity of the band structure - the transition from linear to parabolic character - has been illustrated by the widths of the plateaus in the quantum Hall effect. Finally, the transport of ”massive” Dirac fermions (with finite Dirac mass) is investigated. In particular the describing Dirac Hamiltonian induces weak localization effects depending on the Dirac mass. This mechanism has not been observed to date, and survives in higher temperatures compared to typical localization mechanisms.
Oxide heterostructures attract a lot of attention as they display a vast range of physical phenomena like conductivity, magnetism, or even superconductivity. In most cases, these effects are caused by electron correlations and are therefore interesting for studying fundamental physics, but also in view of future applications. This thesis deals with the growth and characterization of several prototypical oxide heterostructures. Fe3O4 is highly ranked as a possible spin electrode in the field of spintronics. A suitable semiconductor for spin injection in combination with Fe3O4 is ZnO due to its oxide character and a sufficiently long spin coherence length. Fe3O4 has been grown successfully on ZnO using pulsed laser deposition and molecular beam epitaxy by choosing the oxygen partial pressure adequately. Here, a pressure variation during growth reduces an FeO-like interface layer. Fe3O4 films grow in an island-like growth mode and are structurally nearly fully relaxed, exhibiting the same lattice constants as the bulk materials. Despite the presence of a slight oxygen off-stoichiometry, indications of the Verwey transition hint at high-quality film properties. The overall magnetization of the films is reduced compared to bulk Fe3O4 and a slow magnetization behavior is observed, most probably due to defects like anti-phase boundaries originating from the initial island growth. LaAlO3/SrTiO3 heterostructures exhibit a conducting interface above a critical film thickness, which is most likely explained by an electronic reconstruction. In the corresponding model, the potential built-up owing to the polar LaAlO3 overlayer is compensated by a charge transfer from the film surface to the interface. The properties of these heterostructures strongly depend on the growth parameters. It is shown for the first time, that it is mainly the total pressure which determines the macroscopic sample properties, while it is the oxygen partial pressure which controls the amount of charge carriers near the interface. Oxygen-vacancy-mediated conductivity is found for too low oxygen pressures. A too high total pressure, however, destroys interface conductivity, most probably due to a change of the growth kinetics. Post-oxidation leads to a metastable state removing the arbitrariness in controlling the electronic interface properties by the oxygen pressure during growth. LaVO3/SrTiO3 heterostructures exhibit similar behavior compared to LaAlO3/SrTiO3 when it comes to a thickness-dependent metal-insulator transition. But in contrast to LaAlO3, LaVO3 is a Mott insulator exhibiting strong electron correlations. Films have been grown by pulsed laser deposition. Layer-by-layer growth and a phase-pure pervoskite lattice structure is observed, indicating good structural quality of the film and the interface. An electron-rich layer is found near the interface on the LaVO3 side for conducting LaVO3/SrTiO3. This could be explained by an electronic reconstruction within the film. The electrostatic doping results in a band-filling-controlled metal-insulator transition without suffering from chemical impurities, which is unavoidable in conventional doping experiments.
Follow-ups across discourse domains: A cross-cultural exploration of their forms and functions

The edited volume documents the proceedings of the ESF workshop "Follow-ups across discourse domains: a cross-cultural exploration of their forms and functions". It examines the forms and functions of the dialogue act of a follow-up, viz. accepting or challenging a prior communicative act, in political discourse across spoken and written dialogic genres. Specifically, it considers (1) the discourse domains of political interviews, editorials, op-eds and discussion forums, (2) their sequential organization as regards the status of initial (or 1st order) follow-up, a follow-up of a prior follow-up (2nd order follow-up), or nth-order follow-up, and (3) their discursive realization as regards degrees of indirectness and responsiveness which are conceptualized as a continuum along the lines of degrees of explicitness and degrees of responsiveness. The chapters come from the fields of linguistics, discourse analysis, socio-pragmatics, communication, political science and psychology, examining the heterogeneous field of political discourse and its manifestation in diverse discourse genres with respect to evasiveness, indirectness and redundancy in mediated political discourse, professional discourse, discourse identity and doing politics, to name but the most prominent questions.
The sexual phase of Plasmodium falciparum begins with the differentiation of intraerythrocytic sexual stages, termed gametocytes, in the human host. Mature gametocytes circulate in the peripheral blood and are taken up by the mosquito during the blood meal. These stages are essential for the spread of the malaria disease and form gametes in the mosquito midgut within minutes. A highly conserved family of six secreted proteins has been identified in Plasmodium falciparum. They comprise multiple adhesive domains and are termed PfCCp1 through PfCCp5, and PfFNPA. It was revealed in this work that PfCCp multi-domain adhesion proteins form protein complexes in gametocytes and on the surface of newly emerged macrogametes by adhesion domain-mediated binding. Co-Immunoprecipitation assays with activated gametocyte lysates show interactions between PfCCp proteins and indicate surface association via Pfs230 and Pfs25. Pfs230 is connected with the plasma membrane of the parasite by its interaction partner Pfs48/45. This protein is linked to the plasma membrane by a GPI anchor and presumably retains the multi-protein complex on the surface of newly emerged macrogametes in the mosquito midgut. A WD40 domain containing protein was identified to be part of this protein complex. It might serve as platform for the assembly of the multi protein complex or mediate the interplay among proteins, as suggested from known functions of the WD40 domain repeats. During egress from the host erythrocyte, the emerging gametes become vulnerable to factors of the human complement, which is taken up with the blood meal. In this thesis it was found that the complement system is active for about one hour post feeding. Macrogametes defend against complement-mediated lysis by co-opting the human complement regulators Factor H and FHL-1 from the blood-meal. These serum proteins bind via its SCR domains 5-7 to the surface of macrogametes. Once bound, they trigger complement inactivation of the alternative pathway, which prevents induction of complement lysis on the surface of the malaria parasite. Antibodies against Factor H are able to impair the sexual development in vitro and are able to block transmission to the mosquito. Interaction studies on endogenous proteins and immobilized recombinant proteins revealed the PfGAP50 protein as binding partner of Factor H and FHL-1. This protein was hitherto described as a glideosome-associated protein in invasive parasite stages, but has not yet been characterized in gametes. First localization studies indicate a relocation of PfGAP50 from the inner membrane complex to the surface of macrogametes. Malaria still persists as one of the deadliest infectious diseases worldwide. Investigations on the essential transmissive stages, gametocytes and gametes of Plasmodium falciparum, stood in the background of research for a long time. This work deciphered details on protein interactions on the surface of the malaria parasite and provides first information about coactions between the parasite and the human complement in the mosquito midgut.
In der vorliegenden Arbeit wurde der Einfluss eines Mangels des antiinflammatorischen Zytokins Interleukin(IL)-4 am Tiermodell einer experimentellen Mononeuropathie (engl. chronic constriction injury, CCI) untersucht. Zentrale Fragestellung der Studie war, ob IL-4 knockout(ko)-Mäuse im Vergleich zu Wildtyp(wt)-Mäusen mit einem gesteigerten Schmerzverhalten sowie einer veränderten Zytokinantwort und Opioidrezeptor-Expression nach Anwendung eines neuropathischen Schmerzmodells (CCI) reagieren. In mehreren Tierstudien war zuvor eine antiinflammatorische und analgetische Wirkung von IL-4 belegt worden (Vale et al. 2003; Hao et al. 2006) und in klinischen Studien war ein verminderter IL-4-Spiegel bei Patienten mit verschiedenen neuropathischen Schmerzsyndromen mit einer gesteigerten Schmerzempfindung verbunden (Üçeyler et al. 2006; Üçeyler et al. 2007b). Da IL-4 die Transkription von Opioidrezeptoren induziert (Kraus et al. 2001; Börner et al. 2004), wurde zudem das Ansprechen von IL-4 ko-Mäusen auf Morphin und die Genexpression zentraler Opioidrezeptoren untersucht. Vor sowie bis vier Wochen nach Durchführung einer CCI wurden IL-4 ko- sowie wt- Mäuse hinsichtlich ihrer Empfindlichkeit auf mechanische und thermische Stimuli analysiert. Zum Zeitpunkt des Schmerzmaximums nach CCI (Tag 7 bis 9) wurde zudem das Ansprechen beider Genotypen auf Morphin untersucht. Die Genexpression pro- (IL-1 beta, TNF) und antiinflammatorischer Zytokine (IL-10, IL- 13) im peripheren (N. ischiadicus) und zentralen Nervensystem (lumbales und zervikales Rückenmark, Pons, Thalamus, Hypothalamus, Striatum, Kortex) sowie die Genexpression zentraler Opioidrezeptoren (mü-OR, delta-OR, kappa-OR) wurde bei beiden Genotypen vor sowie vier Wochen nach CCI mittels Real-Time-PCR bestimmt. Unbehandelte IL-4 ko-Mäuse zeigten im Vergleich zu wt-Mäusen bereits vor Durchführung einer CCI eine mechanische Überempfindlichkeit (Hyperalgesie), was möglicherweise durch die bei IL-4-Mangel fehlenden zentralen inhibitorischen Mechanismen bedingt ist. Nach CCI entwickelten sowohl IL-4 ko- als auch wt-Mäuse eine gleich ausgeprägte mechanische und thermische Hyperalgesie. Die Tatsache, dass die mechanische Überempfindlichkeit bei IL-4 ko-Mäusen nach Nervenläsion nicht überproportional steigt, kann Ausdruck der nachgewiesenen kompensatorisch stärker ausgeprägten Genexpression proinflammatorischer, aber insbesondere auch antiinflammatorischer Zytokine in diesem Genotyp sein. Nur bei IL-4 ko-Mäusen war vier Wochen nach CCI die Genexpression der anti- inflammatorischen Zytokine im N. ischiadicus (IL-10) und ipsilateralen Rückenmark (IL-10, IL-13), jedoch auch die der proinflammatorischen Zytokine im ipsilateralen Rückenmark (TNF, IL-1 beta) erhöht. Nach CCI sprachen IL-4 ko-Mäuse schneller auf Morphingabe an als wt-Mäuse, was durch den bei diesem Genotyp stärker ausgeprägten Anstieg der Genexpression der Opioidrezeptortypen delta-OR und kappa-OR im kontralateralen Thalamus bedingt sein kann.
Internet applications are becoming more and more flexible to support diverge user demands and network conditions. This is reflected by technical concepts, which provide new adaptation mechanisms to allow fine grained adjustment of the application quality and the corresponding bandwidth requirements. For the case of video streaming, the scalable video codec H.264/SVC allows the flexible adaptation of frame rate, video resolution and image quality with respect to the available network resources. In order to guarantee a good user-perceived quality (Quality of Experience, QoE) it is necessary to adjust and optimize the video quality accurately. But not only have the applications of the current Internet changed. Within network and transport, new technologies evolved during the last years providing a more flexible and efficient usage of data transport and network resources. One of the most promising technologies is Network Virtualization (NV) which is seen as an enabler to overcome the ossification of the Internet stack. It provides means to simultaneously operate multiple logical networks which allow for example application-specific addressing, naming and routing, or their individual resource management. New transport mechanisms like multipath transmission on the network and transport layer aim at an efficient usage of available transport resources. However, the simultaneous transmission of data via heterogeneous transport paths and communication technologies inevitably introduces packet reordering. Additional mechanisms and buffers are required to restore the correct packet order and thus to prevent a disturbance of the data transport. A proper buffer dimensioning as well as the classification of the impact of varying path characteristics like bandwidth and delay require appropriate evaluation methods. Additionally, for a path selection mechanism real time evaluation mechanisms are needed. A better application-network interaction and the corresponding exchange of information enable an efficient adaptation of the application to the network conditions and vice versa. This PhD thesis analyzes a video streaming architecture utilizing multipath transmission and scalable video coding and develops the following optimization possibilities and results: Analysis and dimensioning methods for multipath transmission, quantification of the adaptation possibilities to the current network conditions with respect to the QoE for H.264/SVC, and evaluation and optimization of a future video streaming architecture, which allows a better interaction of application and network.
This thesis concerns (i) the synthesis and olfactory characterisation of silicon-containing analogues of the musk odourant phantolide, (ii) the synthesis and pharmacological investigation of silicon-containing analogues of retinoids of the EC23 and TTNN type and (iii) the attempted syntheses of silicon-containing analogues of the antipsychotic penfluridol and the antidiarrhoeal agent loperamide. All target compounds and intermediates were characterised by multinuclear NMR studies (1H, 13C, 15N, 19F, 29Si) and elemental analyses or high-resolution mass spectrometry. Additionally, some of these compounds were characterized by single crystal X-ray diffraction studies.
Derivate von Vinylsulfonen (VS), die zur Klasse der Michael-Akzeptoren gehören, haben sich in den letzten Jahren als potente irreversible Inhibitoren von Cystein-Proteasen etabliert. Durch einen nucleophilen Angriff des Cys-Restes im aktiven Zentrum der Protease auf das beta-Kohlenstoffatom der C-C-Doppelbindung wird die Protease irreversibel alkyliert. Ziel dieser Arbeit war es, einfache theoretische und experimentelle Methoden zu entwickeln, um erste Schlussfolgerungen hinsichtlich der Reaktivität unterschiedlicher Vinylsulfone ziehen zu können, die zur vollständigen Aufklärung der Struktur-Wirkungsbeziehung von Vinylsulfonen mit diversen Cystein-Proteasen dienen. Im ersten Teil der Arbeit wurden quantenmechanische Rechnungen an kleinen Vinylsulfon-Bausteinen angestellt, um den Einfluss unterschiedlicher Substitutionsmuster an der Sulfoneinheit auf die Reaktionskinetik von Vinylsulfonen zu untersuchen. Anhand der jeweiligen Potentialflächen ließen sich die charakteristischen Punkte der Reaktion, wie der Reaktionskomplex, der Übergangszustand (transition state, TS) sowie das Produkt mitsamt ihren Energien und Geometrien bestimmen. Die Höhe der Energiebarriere, die zum Erreichen des TS überwunden werden muss, die sogenannte Aktiverungsenergie, hängt über die Arrhenius-Gleichung mit den kinetischen Parametern der Reaktion zusammen. Es lässt sich also durch die Kenntnis der Aktivierungsenergien die Reaktivitätsreihenfolge unterschiedlich substituierter Vinylsulfone VS vorhersagen. Im zweiten Teil dieser Arbeit wurden Vinylsulfonbausteine synthetisiert und an separat hergestellte Peptide gekuppelt, sodass potentielle Inhibitoren erhalten wurden. So konnten u.a. die peptidischen Inhibitoren Mu-D-Phe-L-HomoPhe-VS-Me und MP-D-Phe-L-HomoPhe-VS-Me hergestellt werden. Ein zweites Syntheseprojekt beschäftigte sich mit der Kupplung von Peptiden an neue Derivate der trans-Aziridin-2,3-dicarbonsäure. Die synthetisierten Inhibitoren waren Z-Phe-Ala-Azi, Boc-Leu-Pro-Azi und Z-Pro-Leu-Azi. Hierfür wurden die Peptide des Vinylsulfonsprojekts in umgekehrter Aminosäure-Reihenfolge synthetisiert, um sie an die Aziridinbausteine kuppeln zu können. Der dritte Teil der Doktorarbeit befasste sich mit der experimentellen Untersuchung der synthetisierten Vinylsulfonbausteine sowie den erhaltenen peptidischen VS- und Aziridin-basierten Inhibitoren. Es wurden einerseits Enzym-Assays durchgeführt, um die prozentuale Hemmung verschiedener Cystein-Proteasen durch die synthetisierten Moleküle zu messen. Keine der Verbindungen wies jedoch eine signifikannte Hemmung der Proteasen Rhodesain, Falcipain 2 und Cathepsin B auf. Andererseits wurden Modellsysteme entwickelt, um die Kinetik der Reaktionen der Vinylsulfon- und Aziridinbausteine mit einem geeigneten Thiol als Enzym-Imitat zu verfolgen. Ein zielführendes Modell konnte mit Phenylethanthiol in deuteriertem Methanol realisiert werden. Durch Zusatz von NaOH, KOH oder KOtBu konnte zusätzlich die Reaktion mit dem Thiolat untersucht werden. Die Reaktionen wurden sowohl mit IR- als auch NMR-Spektroskopie verfolgt und es wurden die Geschwindigkeitskonstanten 2. Ordnung bestimmt. Auf den ersten Blick konnte mit dem theoretischen Modell der experimentell gefundene Trend nicht vorhergesagt werden. Die Reihenfolge der Sulfonderivate aber, die an der Sulfongruppe ein weiteres Heteroatom tragen, Sulfonester und Sulfonamid, wurde richtig abgeschätzt. Der Unterschied in der Aktivierungsenergie zwischen den Sulfonestern beläuft sich auf 0.7 kcal pro mol. Über die Arrheniusgleichung, ergibt sich bei Annahme desselben Arrhenius-Faktors bei einer Temperatur von 25°C, dass OPhVS um einen Faktor 3 schneller als OMeVS reagieren sollte. Tatsächlich wurde im Experiment ein Faktor von 2.6 gefunden. Aufgrund der unterschiedlichen Substituenten am Stickstoffatom, ist das Amid nicht vollständig mit seinem H-substituierten theoretischen Pendant vergleichbar. Dass das Sulfonamid langsamer als die Sulfonester reagieren, wurde vom theoretischen Modell ebenfalls richtig vorhergesagt.
The nothotaxa of the genus Sedum series Rupestria Berger (Crassulaceae) are reviewed, three new hybrids (S. ×affomarcoi, S. ×henkii and S. ×pascalianum) are described and Sedum brevierei Chass. is typified. The identity of the western (French) populations of S. rupestre L. subsp. erectum ‘t Hart is also clarified, and a table with all known combinations is given.
Der vorliegende Bericht gibt Auskunft über die Ergebnisse der wissenschaftlichen Begleitung des Eisregel-Trainings der DLRG-Jugend Bayern bei Kindern im Vorschulalter. Das Eisregel-Training ist Bestandteil des Programms "Cool & Sicher", bei dem Heranwachsende mit den Gefahren im und am Wasser bzw. Eisflächen vertraut gemacht werden. Das Ziel des Programms ist es, durch das Training die Wahrscheinlichkeit eines Tods durch Ertrinken bei Kindern deutlich zu reduzieren, in dem die Kinder selbst, aber auch die Eltern über Präventionsmaßnahmen aufgeklärt werden.
Jugend. Engagement. Politische Sozialisation : Skalendokumentation des Längsschnitts 2010 - 2011

Die dargestellten Ergebnisse stammen aus dem Projekt "Jugend. Engagement. Politische Sozialisation" ("Persönlichkeitsentwicklung und politische Beteiligungsbereitschaft durch gemeinnützige Tätigkeit im Jugendalter") der Universität Würzburg. Hauptaugenmerk dieser repräsentativen Umfrage liegt auf der Erfassung des ehrenamtlichen Engagements bei Jugendlichen und erfolgt in mehreren Erhebungswellen. Ziel ist die differenzierte Darstellung des aktuellen Engagements sowie die Begleitung potentieller Veränderungen über die Zeit hinweg. Die vorgestellten Ergebnisse beziehen sich auf die Daten der von Oktober 2010 bis Mitte Januar 2011 durchgeführten ersten Erhebungswelle (t1). Es werden sowohl Design, als auch die Form der Erfassung des ehrenamtlichen Engagements sowie die Stichprobe beschrieben und anschließend ausgewählte Ergebnisse dargestellt. Die Auswahl der Befunde bezieht sich auf folgende Punkte: zeitlicher Aufwand und Beurteilung des Engagements, Tätigkeitsbereiche, "Wer die ehrenamtlich Engagierten sind", Motive, die Jugendliche dazu bewegen, ein Ehrenamt auszuüben sowie mögliche Auswirkungen.

Ziel ist die differenzierte Darstellung des aktuellen Engagements sowie die Begleitung potentieller Veränderungen über die Zeit hinweg. Die vorgestellten Ergebnisse beziehen sich auf die Daten der von Oktober 2010 bis Mitte Januar 2011 durchgeführten ersten Erhebungswelle (t1). Es werden sowohl Design, als auch die Form der Erfassung des ehrenamtlichen Engagements sowie die Stichprobe beschrieben und anschließend ausgewählte Ergebnisse dargestellt. Die Auswahl der Befunde bezieht sich auf folgende Punkte: zeitlicher Aufwand und Beurteilung des Engagements, Tätigkeitsbereiche, "Wer die ehrenamtlich Engagierten sind", Motive, die Jugendliche dazu bewegen, ein Ehrenamt auszuüben sowie mögliche Auswirkungen.
In this thesis the Drosophila mutant loechrig (loe), that shows progressive degeneration of the nervous system, is further described. Loe is missing a neuronal isoform of the protein kinase AMPK γ subunit (AMP-activated protein kinase- also known as SNF4Aγ) The heterotrimeric AMPK controls the energy level of the cell, which requires constant monitoring of the ATP/AMP levels. It is activated by low energy levels and metabolic insults like oxygen starvation and regulates multiple important signal pathways that control cell metabolism. Still, its role in neuronal survival is unclear. One of AMPK’s downstream targets is HMGR (hydroxymethylglutaryl-CoA- reductase), a key enzyme in cholesterol and isoprenoid synthesis. It has been shown that manipulating the levels of HMGR affects the severity of the neurodegenerative phenotype in loe. Whereas the regulatory role of AMPK on HMGR is conserved in Drosophila, insects cannot synthesize cholesterol de novo. However, the synthesis of isoprenoids is a pathway that is evolutionarily conserved between vertebrates and insects. Isoprenylation of target proteins like small G-proteins provides a hydrophobic anchor that allows the association of these proteins with membranes and following activation. This thesis shows that the loe mutation interferes with the prenylation of Rho1 and the regulation of the LIM kinase pathway, which plays an important role in actin turnover and axonal outgrowth. The results suggest that the mutation in LOE, causes hyperactivity of the isoprenoid synthesis pathway, which leads to increased farnesylation of RHO1 and therefore higher levels of phospho-cofilin. A mutation in Rho1 improves the neurodegenerative phenotype and life span. The increased inactive cofilin amount in loe leads to an up regulation of filamentous actin. Actin is involved in neuronal outgrowth and experiments analyzing loe neurons gave valuable insights into a possible role of AMPK and accordingly actin on neurite growth and stability. It was demonstrated that neurons derived from loe mutants exhibit reduces axonal transport suggesting that changes in the cytoskeletal network caused by the effect of loe on the Rho1 pathway lead to disruptions in axonal transport and subsequent neuronal death. It also shows that actin is not only involved in neuronal outgrowth, its also important in maintenance of neurons, suggesting that interference with actin dynamics leads to progressive degeneration of neurons. Together, these results further support the importance of AMPK in neuronal function and survival and provide a novel functional mechanisms how alterations in AMPK can cause neuronal degeneration
Aldose Reduktase ALR2 katalysiert den ersten Schritt des Sorbitol-Stoffwechselweges. In diesem wird mit Hilfe des Kofaktors NADPH Glukose zu Sorbitol reduziert. Bei erhöhtem Blutzuckerspiegel, wie dies bei Diabetes-Patienten der Fall ist, ist dieser metabolische Weg von Bedeutung. Bis zu einem Drittel der Blutglukose wird zu Sorbitol reduziert. Die Folge der Sorbitolakkumulation in den Zellen und der Verminderung der NADPH-Konzentration sind „osmotischer“ sowie „oxidativer“ Stress. Diese stehen in Zusammenhang mit den vielfach diskutierten Spätschäden des Diabetes, wie diabetischer Katarakt, Neuro- und Nephropathie. Das Enzym ist experimentell sehr gut untersucht und eignet sich daher als Modellsystem zur Untersuchung der intrinsischen Proteinflexibilität und thermodynamischer Daten mit Hilfe von Computermethoden. Unter diesen Voraussetzungen steht der Gewinn eines besseren Verständnisses von molekularer Erkennung und Proteinbeweglichkeit der ALR2 unter Verwendung von Molekulardynamik-Simulationen MD als primäres Ziel im Zentrum dieser Arbeit. Dabei wurden MD-Studien zu zwei kristallographisch erhaltenen Protein-Ligand-Komplexen durchgeführt. Die Liganden unterscheiden sich nur geringfügig in der Länge einer Seitenkette, ihre Bindung führt allerdings zu gänzlich unterschiedlichen Bindemodi. Einer davon ist bislang einzigartig für die ALR2. Mit Hilfe von MD-Simulationen wurde versucht, eine Erklärung für diese neue Konformation der Bindetasche im Vergleich zu jener eines strukturell sehr ähnlichen Liganden zu finden. Außerdem waren über diese Studien Aussagen über besonders flexible Bereiche der ALR2-Bindetasche möglich, die mit bereits existierenden Erkenntnissen über die Bindetaschenflexibilität verglichen werden konnten. Darüber hinaus gelang es, durch die Methode der gesteuerten Molekulardynamik SMD einen Übergang zwischen einer röntgenkristallografisch ermittelten kofaktorgebundenen Holo-Konformation und kofaktorfreien Apo-Konformation zu simulieren. Computergestützte Methoden ermöglichen es also, weitläufige Bewegungen von einer Proteinkonformation in die andere nachzuvollziehen bzw. die experimentell erhaltenen Strukturen zu bestätigen. Eine mechanistische Deutung des Kofaktorassoziations- und Kofaktordissoziationsprozesses wurde ebenfalls versucht. Dafür war es notwendig, strukturelle Veränderungen im Protein zeitlich zu verfolgen und entscheidende Vorgänge zu identifizieren. Die Methode der SMD wurde in dieser Arbeit auch auf ein weiteres, pharmakologisch interessantes System übertragen. Dabei wurde versucht auch an zwei Vertretern der Klasse der Nukleären Rezeptoren NRs, dem Androgenrezeptor AR und dem Estrogenrezeptor ER, eine solche weitreichende Bewegung nachzuvollziehen. Auch bei diesen Rezeptoren sind zwei in der Position einer alpha-Helix unterschiedliche Formen bekannt. Auch hier wurden mit Hilfe der genannten Methode, relevante Ereignisse hinsichtlich der Helixmobilität identifiziert. Abschließend wurde auf den thermodynamischen Aspekt der Protein-Ligand-Komplexe eingegangen. Durch Berechnungen anhand der Methode der thermodynamischen Integration TI wurden relative Bindungsaffinitäten am Modellsystem ALR2 gewonnen. Durch den Vergleich mit experimentell vorhandenen Daten konnte die Methode validiert werden. Das Verfahren der TI sollte in Zukunft eine Voraussage von Affinitäten beliebiger, sich geringfügig unterscheidender Inhibitoren, die aber denselben Bindemodus aufweisen, ermöglichen und damit den Prozess des Wirkstoffdesigns erleichtern. Zusammenfassend ergab sich eine gute Übereinstimmung der experimentell ermittelten Strukturen bzw. Daten mit den durch Computersimulationen erhaltenen.
Einfluss von Monozyten auf Heilungsvorgänge und kardiale Thromboembolien nach einem Myokardinfarkt

Beim Myokardinfarkt führt die Unterbrechung der Blutversorgung zur Nekrose von Myozyten. Hierauf werden im Rahmen der Entzündungsreaktion verschiedene Reaktionen angestoßen, die dazu dienen sollen, die Integrität und Funktion des Herzens zu erhalten. Einen wichtigen Anteil an der Inflammationsreaktion haben die Monozyten, deren Funktion in diesem Rahmen bis jetzt nur wenig erforscht ist. In dieser Studie wurde die Rolle von Monozyten auf die Heilungsvorgänge nach einem Infarkt untersucht. Durch die Gabe von Clodronat-Liposom wurden in der Behandlungsgruppe die Monozyten ausgeschaltet. Es zeigte sich, dass die Mortalität in der Behandlungsgruppe aufgrund der Bildung eines linksventrikulären Thrombus deutlich erhöht ist. Dieser war bereits innerhalb der ersten 24h in den echokardiographischen Untersuchungen sichtbar. Veränderungen im Gerinnungssystem konnten als Ursache ausgeschlossen werden. Durch eine CD-31-Färbung wurde deutlich, dass in den infarzierten Herzen der Behandlungsgruppe ein Defekt im Endokard entstanden war, der als Ursache für die Entstehung der Thromben gewertet werden kann. Zudem wurde in der Behandlungsgruppe neben einem verschlechterten Abräumvorgang eine signifikant erniedrigte Kollagen-1-Produktion, ein erhöhtes MMP-9, ein leicht erniedrigtes TGF sowie erhöhtes VEGF gemessen. In dieser Studie wurde somit gezeigt, dass Monozyten ein essentieller Bestandteil der Heilungsvorgänge nach einem Infarkt sind. Durch eine eingeschränkte Monozytenfunktion wird zudem aufgrund von Defekten im Endothel die Bildung eines linksventrikulären Thrombus und folgender thromboembolischer Ereignisse begünstigt.
ATP dependent chromatin remodeling complexes are multifactorial complexes that utilize the energy of ATP to rearrange the chromatin structure. The changes in chromatin structure lead to either increased or decreased DNA accessibility. SWI/SNF is one of such complex. The SWI/SNF complex is involved in both transcription activation and transcription repression. The ATPase subunit of SWI/SNF is called SWI2/SNF2 in yeast and Brahma, Brm, in Drosophila melanogaster. In mammals there are two paralogs of the ATPase subunit, Brm and Brg1. Recent studies have shown that the human Brm is involved in the regulation of alternative splicing. The aim of this study was to investigate the role of Brm in pre-mRNA processing. The model systems used were Chironomus tentans, well suited for in situ studies and D. melanogaster, known for its full genome information. Immunofluorescent staining of the polytene chromosome indicated that Brm protein of C. tentans, ctBrm, is associated with several gene loci including the Balbiani ring (BR) puffs. Mapping the distribution of ctBrm along the BR genes by both immuno-electron microscopy and chromatin immunoprecipitation showed that ctBrm is widely distributed along the BR genes. The results also show that a fraction of ctBrm is associated with the nascent BR pre-mRNP. Biochemical fractionation experiments confirmed the association of Brm with the RNP fractions, not only in C. tentans but also in D. melanogaster and in HeLa cells. Microarray hybridization experiments performed on S2 cells depleted of either dBrm or other SWI/SNF subunits show that Brm affects alternative splicing and 3´ end formation. These results indicated that BRM affects pre-mRNA processing as a component of SWI/SNF complexes. 1
Time-resolved optical spectroscopy has become an important tool to investigate the dynamics of quantum mechanical processes in matter. In typical applications, a first “pump” pulse excites the system under investigation from the thermal equilibrium to an excited state, and a second variable time-delayed “probe” pulse then maps the dynamics of the excited system. Although advanced nonlinear techniques have been developed to investigate, e.g., coherent quantum effects, all of these techniques are limited in their spatial resolution. The laser focus diameter has a lower bound given by Abbe’s diffraction limit, which is roughly half the optical excitation wavelength—corresponding to about 400nm in the presented experiments. In the time-resolved experiments that have been suggested so far, averaging over the sample volume within this focus cannot be avoided. In this thesis, two approaches were developed to overcome the diffraction limit in optical spectroscopy and to enable the investigation of coherent processes on the nanoscale. In the first approach, analytic solutions were found to calculate optimal polarizationshaped laser pulses that provide optical near-field pump–probe pulse sequences in the vicinity of a nanostructure. These near-field pulse sequences were designed to allow excitation of a quantum system at one specific position at a certain time and probing at a different position at a later time. In the second approach, the concept of coherent two-dimensional (2D) spectroscopy, which has had great impact on the investigation of coherent quantum effects in recent years, was combined with photoemission electron microscopy, which yields a spatial resolution well below the optical diffraction limit. Using the analytic solutions, optical near fields were investigated in terms of spectroscopic applications. Near fields that are excited with polarization-shaped femtosecond laser pulses in the vicinity of appropriate nanostructures feature two properties that are especially interesting in the view of spectroscopic applications: On the one hand, control of the spatial distribution of the optical fields is achieved on the order of nanometers. On the other hand, the temporal evolution of these fields can be adjusted on the order of femtoseconds. In this thesis, solutions were found to calculate the optimal polarizationshaped laser pulses that control the near field in a general manner. The main idea to achieve this deterministic control was to disentangle the spatial and temporal near-field control. First, the spatial distribution of the optical near field was controlled by assigning the correct state of polarization for each frequency within the polarization-shaped laser pulse independently. The remaining total phase—not employed for spatial control—was then used for temporal near-field compression, which, in experimental applications, would lead to an enhancement of the nonlinear signal at the respective location. In contrast to the use of optical near fields, where pump–probe sequences themselves are localized below the diffraction limit and the detection does not have to provide the spatial resolution, a different approach was suggested in this thesis to gain spectroscopic information on the nanoscale. The new method was termed “Coherent two-dimensional (2D) nanoscopy” and transfers the concept of “conventional” coherent 2D spectroscopy to photoemission electron microscopy. The pulse sequences used for the investigation of quantum systems in this method are still limited by diffraction. However, the new key concept is to detect locally generated photoelectrons instead of optical signals. This yields a spatial resolution that is well below the optical diffraction limit. In “conventional” 2D spectroscopy a triple-pulse sequence initiates a four wave mixing process that creates a coherence. In a quantum mechanical process, this coherence is converted into a population by emission of an electric field, which is measured in the experiment. Contrarily, in the developed 2D nanoscopy, four-wave mixing is initiated by a quadruple-pulse sequence, which leaves the quantum system in an electronic population. This electronic population carries coherent information about the investigated quantum system and can be mapped with a spatial resolution down to a few nanometers given by the spatial resolution of the photoemission electron microscope. Hence, 2D nanoscopy can be considered a generalization of time-resolved photoemission experiments. In the future, it may be of similar beneficial value for the field of photoemission research as “conventional” 2D spectroscopy has proven to be for optical spectroscopy and nuclear magnetic resonance experiments. In a first experimental implementation of coherent 2D nanoscopy coherent processes on a corrugated silver surface were measured and unexpected long coherence lifetimes could be determined.
In the verification of positive Harris recurrence of multiclass queueing networks the stability analysis for the class of fluid networks is of vital interest. This thesis addresses stability of fluid networks from a Lyapunov point of view. In particular, the focus is on converse Lyapunov theorems. To gain an unified approach the considerations are based on generic properties that fluid networks under widely used disciplines have in common. It is shown that the class of closed generic fluid network models (closed GFNs) is too wide to provide a reasonable Lyapunov theory. To overcome this fact the class of strict generic fluid network models (strict GFNs) is introduced. In this class it is required that closed GFNs satisfy additionally a concatenation and a lower semicontinuity condition. We show that for strict GFNs a converse Lyapunov theorem is true which provides a continuous Lyapunov function. Moreover, it is shown that for strict GFNs satisfying a trajectory estimate a smooth converse Lyapunov theorem holds. To see that widely used queueing disciplines fulfill the additional conditions, fluid networks are considered from a differential inclusions perspective. Within this approach it turns out that fluid networks under general work-conserving, priority and proportional processor-sharing disciplines define strict GFNs. Furthermore, we provide an alternative proof for the fact that the Markov process underlying a multiclass queueing network is positive Harris recurrent if the associate fluid network defining a strict GFN is stable. The proof explicitely uses the Lyapunov function admitted by the stable strict GFN. Also, the differential inclusions approach shows that first-in-first-out disciplines play a special role.
Effekte eines standardisierten Kiefernrindenextraktes und dessen Metabolit auf NO und NO-Synthasen

Um die Grundlagen für die in klinischen Studien beim Einsatz des standardisierten Kiefernrindenextraktes (Pycnogenol®) gefundenen Effekte auf einer mechanistischen zellulären Ebene aufzuklären, wurde in der hier vorliegenden Arbeit der Einfluss der Komponenten des Extraktes und dessen Metabolit M1 (chemisch benannt δ-(3,4-Dihydroxyphenyl)-γ-valerolacton bzw. 5 (3,4 Dihydroxybenzyl)dihydrofuran 2(3H) on) hinsichtlich der Wirkung auf Stickstoffmonoxid(= NO)-produzierende Systeme untersucht. NO ist an einer Vielzahl von physiologischen und pathophysiologischen Prozessen in lebenden Organismen beteiligt. Im Menschen sind bislang drei NO-Synthasen bekannt: die induzierbare (iNOS), die hinsichtlich der Pathologie vor allem mit entzündlichen Vorgängen assoziiert wird, die endotheliale (eNOS), die bei Gefäß- und Herzkreislauferkrankungen eine Rolle spielt, und die neuronale (nNOS), die mit der Gedächtnisbildung, aber auch mit zytotoxischen Prozessen im Gehirn etwa bei Morbus Alzheimer oder der Parkinson-Krankheit in Verbindung gebracht wird. Der nach peroraler Einnahme des Extraktes im Darm durch metabolisierende Kolonbakterien entstehende und darauf im Plasma erscheinende Metabolit M1, dem bei allen durchgeführten Untersuchungen besonderes Augenmerk zuteil wurde, zeigte eine starke konzentrationsabhängige Inhibierung der NO-Freisetzung der iNOS aus einer durch einen Entzündungsreiz stimulierten murinen Makrophagenzellkultur (IC50= 1,28 µg/mL). Im Vergleich mit Fraktion I des Kiefernrindenextraktes, die vor allem monomere Extraktbestandteile enthält, und Hydrocortison zeigte M1 zusätzlich einen stärkeren Hemmeffekt auf die NO-Freisetzung nach dem Entzündungsreiz. Die Zytotoxizität von M1 im Testsystem war dabei als gering einzustufen. Interessanterweise wurde neben den NO-Radikalfängereigenschaften von M1 auch ein deutlich hemmender konzentrationsabhängiger Effekt auf die iNOS-Proteinexpression gefunden (IC50= 3,78 µg/mL). Da die bislang im Plasma bestimmten M1-Konzentrationen deutlich geringer als die in Zellkulturversuchen wirksamen waren, wurde eine mögliche Anreicherung von M1 in Gegenwart von Serumproteinen in humanen Endothelzellen, primären Monozyten und murinen Makrophagen untersucht. Dabei wurde eine starke Bindung von M1 an die Zellen gezeigt und Hinweise für eine potentiell erleichterte Aufnahme von M1 durch membranständige Transporter unter Einsatz eines Influx-Hemmers (Phloretin) gefunden. Zur Untersuchung der eNOS, die sehr geringe Mengen NO produziert, wurden neue methodische Ansätze entwickelt. In diesem Zusammenhang wurden zuvor unbekannte Fallstricke bei der Verwendung der Fluoreszenzsonde DAF-2 (4,5-Diaminofluorescein) zur NO-Detektion und dem Einsatz unterschiedlicher Detektionssysteme entdeckt. DAF-2 zeigte unter verschiedenen Bedingungen auch ohne extern zugegebene NO-Quelle und besonders beim Einfrieren/Auftauen unerwarteterweise eine Konversion zum korrespondierenden NO-Addukt (DAF-2T). Die eingesetzten monomeren Testsubstanzen ((+)-Catechin, (-)-Epicatechin, Resveratrol, M1) waren über die Testzeiträume deutlich instabil mit dynamischer Eigenfluoreszenz. Sowohl über kurze (≤ 45 min) als auch über längere Zeiträume (14-20 h) wurde entsprechend der Redoxaktivität der eingesetzten Polyphenole eine konzentrationsabhängige scheinbar hemmende Wirkung auf die extrazelluläre NO-Freisetzung der eNOS gezeigt. Die eNOS-Proteinexpression blieb durch die verwendeten Monomere weitestgehend unbeeinflusst. Durch eine hohe Konzentration der Fraktion I des Kiefernrindenextraktes wurde eine Steigerung der eNOS-Proteinkonzentration in Endothelzellen gefunden, wobei zytotoxische Artefakte dabei nicht auszuschließen waren. Als kompetitive endogene Inhibitoren der NOS wurden in vivo in jüngster Zeit methylierte Arginine (ADMA= asymmetrisches, SDMA= symmetrisches Dimethylarginin) entdeckt. In einer randomisierten, kontrollierten, doppelt-blinden klinischen Studie mit einem Cross-over Design am Universitätsklinikum Zürich mit 28 Patienten, die an einer koronaren Herzerkrankung litten, wurden die Plasmaspiegel methylierter Arginine vor und nach 8 wöchiger Einnahme des Kiefernrindenextraktes bestimmt. Es zeigte sich dabei trotz einer Verbesserung der flussinduzierten Gefäßerweiterungskapazität (Flow-mediated dilation) und Verringerung der 15-F2t-Isoprostan-Plasmaspiegel keine signifikante Veränderung der Plasmakonzentrationen von ADMA, SDMA und ET-1 (Endothelin-1) durch die Einnahme des Extraktes. Die nNOS kommt vor allem im Gehirn, aber auch in Muskelzellen vor. Der Einsatz des Metaboliten M1 führte zu keinen deutlichen Effekten auf die konstitutive nNOS-Expression in einem Rhabdomyosarkom(A-673)-Zellkulturmodell. Zur Beantwortung der Frage, wie wahrscheinlich es ist, dass zur möglichen Beeinflussung von (patho)-physiologischen zerebralen Prozessen Polyphenole in vivo das Gehirn erreichen, wurde erstmals ein in silico-Modell zur Vorhersage der Verteilung von ausgewählten polyphenolischen Substanzen zwischen Blut und Gehirn entwickelt. Damit wurde anschließend eine Reihenfolge mit logBB-Werten (logarithmierter Quotient aus Konzentration im Blut und im Gehirngewebe) geordnet nach einer entsprechend dem Modell wahrscheinlich höheren Verteilung ins Gehirn für die untersuchten Substanzen berechnet: Protocatechusäure < Quercetin < Cyanidin < (+) Catechin < (-)-Epicatechin < Phloretin < M1. Insgesamt schienen die untersuchten polyphenolischen Substanzen eher schwach bluthirnschrankengängig zu sein. Der Metabolit M1 zeigte den höchsten logBB-Wert und somit die höchste Wahrscheinlichkeit der untersuchten Polyphenole, die Blut-Hirnschranke in vivo zu überwinden. Im Kontext einer möglichen Anwendung bei chronisch-entzündlichen Erkrankungen wurde zusätzlich ein Extrakt aus der Frucht von Morinda citrifolia L. in einem primären Monozyten-Zellkulturmodell auf seine Eigenschaften hin die Sekretion der Matrix-Metalloprotease-9 (MMP-9) aus Immunzellen nach einem Entzündungsreiz zu beeinflussen untersucht. Dabei zeigten die Extraktverdünnungen deutliche konzentrationsabhängige Hemmeffekte um bis zu ~50 % der maximalen MMP-9 Sekretion, die mit dem Einsatz von Hydrocortison vergleichbar waren. Somit konnten in der vorliegenden Arbeit neue Beiträge zur Wirkungsweise der untersuchten Pflanzenextrakte und vor allem zum Verständnis der möglichen Effekte von Polyphenolen auf physiologisch relevante NO-Systeme sowie zur methodischen Wissenserweiterung der komplexen NO-Analytik geleistet werden.
