The present study focuses on Rulin waishi 儒林外史 (The Unofficial History of the Scholars), a well-known Qing novel, from the perspective of gender. It attempts to contribute to the discussion about Chinese masculinity by identifying the representation of the scholars' masculinity in Rulin waishi and offer a better understanding of the novel's position regarding femininity and women.
This project shows that the novel nevertheless reflects rather than challenges gender ideologies of its time. The ideal manhood showed in the novel comprises real virtues and authentic learning. It goes against the traditional, orthodox Confucian masculinity which advocates that officialdom is the glorious path to fulfill a learned man's masculinity. It is mainly due to Wu Jingzi's own failure in the civil service examinations and official careers. Regarding the relation of masculinity and sexuality, the novel reveals that a masculine man is not tempted by female charm but can enjoy a harmonious and companionate marriage. Besides, scholars show great anxiety about their masculinity since they are in a marginal position in society. Their manliness is challenged by officials, merchants, and even commoners, as well as their colleagues.
Through a careful examination of stories of Pinniang, Miss Lu, and Mrs. Wang, it reveals that the novel holds a conventional opinion on women although it criticizes widow suicide and shows an egalitarian husband-wife relationship. It praises Confucian womanly virtues, such as following and serving the husband, managing the household, and keeping chastity. Female sexuality is blamed as an evil temptation to lead men to go astray. Women’s learning gains legitimacy when serving to fulfill domestic responsibilities. It carries the Confucian message that men should take the lead and maintain order in the household and reinforces the rightful patriarchy.
In a word, rather than go ahead of its time, Rulin waishi holds a conservative attitude towards gender issues.
Der zweizeitige Endoprothesenwechsel gilt als Goldstandard der Therapie periprothetischer Gelenkinfektionen. Ein sicherer Ausschluss einer Infektpersistenz ist für den Therapieerfolg essentiel. Es wurde der diagnostische Nutzen synovialer Marker zum Ausschluss einer Infektpersistenz bei einliegendem Kniegelenksspacer untersucht. Weder mikrobiologische noch zytologische Synovia-Analysen sind zum Ausschluss einer Infektpersistenz im Rahmen des zweizeitigen Knieendoprothesenwechsels geeignet.
Recent progress in nanotechnology has attracted interest to a biomedical application of the carbon nanoparticle C60 fullerene (C60) due to its unique structure and versatile biological activity. In the current study the dual functionality of C60 as a photosensitizer and a drug nanocarrier was exploited to improve the efficiency of chemotherapeutic drugs towards human leukemic cells.
Pristine C60 demonstrated time-dependent accumulation with predominant mitochondrial localization in leukemic cells. C60’s effects on leukemic cells irradiated with high power single chip LEDs of different wavelengths were assessed to find out the most effective photoexcitation conditions. A C60-based noncovalent nanosized system as a carrier for an optimized drug delivery to the cells was evaluated in accordance to its physicochemical properties and toxic effects. Finally, nanomolar amounts of C60-drug nanocomplexes in 1:1 and 2:1 molar ratios were explored to improve the efficiency of cell treatment, complementing it with photodynamic approach.
A proposed treatment strategy was developed for C60 nanocomplexes with the common chemotherapeutic drug Doxorubicin, whose intracellular accumulation and localization, cytotoxicity and mechanism of action were investigated. The developed strategy was revealed to be transferable to an alternative potent anticancer drug – the herbal alkaloid Berberine.
Hereafter, a strong synergy of treatments arising from the combination of C60-mediated drug delivery and C60 photoexcitation was revealed. Presented data indicate that a combination of chemo- and photodynamic treatments with C60-drug nanoformulations could provide a promising synergetic approach for cancer treatment.
Peptide receptor radionuclide therapy (PRRT) is a molecular targeted radiation therapy involving the systemic administration of radiolabeled somatostatin receptor binding peptides designed to target with high affinity and specificity receptors overexpressed on tumors. Peptides are applied which either target as agonist (with internalization) or antagonist (little to no internalization). Recently, two novel antagonistic agents have been developed for clinical use: OPS202 and OPS201. 68Ga-labelled OPS202 is used for diagnostic purposes with positron emission tomography and 177Lu-labelled OPS201 is used for the therapy in patients with neuroendocrine tumors (NETs). Both agents are presently under clinical evaluation. Despite the very low internalization rate, the use of somatostatin receptor antagonists which target more binding sites on receptors are expected to result in higher specificity, more favorable pharmacokinetics and higher tumor retention and better visualization than the agonists. The main goal of this thesis was analyzing the biodistribution, biokinetics and internal dosimetry of the recently developed somatostatin receptor antagonists (OPS201 and OPS202) for therapeutic and diagnostic purposes in different species (mice, pigs and patients). In addition, an analysis of the influence of image quantification and the integration of time activity curves on kidney dosimetry in a pig model was carried out. Furthermore, extrapolation methods, which are used for predicting organ absorbed doses for humans based on preclinical animal models, were systematically compared for blood, liver, and kidneys of OPS201 injected species. Based on the OPS202 injected patients’ investigations, 68Ga-OPS202 shows promising biodistribution and imaging properties with tumor contrast which is optimal one hour after injection of the radiotracer. OPS202 is well tolerated and delivers absorbed doses to organs that are lower than those by 18F-FDG and similar to other 68Ga-labeled somatostatin receptor ligands. As a result of 68Ga OPS202 injection, the highest absorbed doses were observed in the urinary bladder (0.10 mGy/MBq) and kidneys (0.84 mGy/MBq). The calculated mean effective dose coefficient of 68Ga-OPS202 injected patients was 0.024 mSv/MBq (3.6 mSv for 150 MBq 68Ga-OPS202 injection) which is similar to other 68Ga-labeled compounds. Based on the OPS201 biokinetics and dosimetry investigations, after the injection of 177Lu-OPS201, a fast blood clearance of the compound is observed in the first phase (half-life: 1.83 h) for each species. 10 min after injection, less than 5% of the injected activity per milliliter of blood circulates in pigs and humans. The analysis of the mice, pig and preliminary patient data provides evidence that, patients enrolled in a phase 1 177Lu-OPS201 trial would not be at risk of overexposure. Based on our results, for 177Lu labelled studies, late time points after 72 h have a great impact on absorbed dose calculations. That is why follow-up times especially at late time points (more than 72 h) are required for the time-integrated activity coefficient (TIAC) calculations in order to represent the area under the curve appropriately and to analyze both biokinetics and dosimetry accurately. In addition, to find the most adequate extrapolation methods that minimize the interspecies differences of dosimetry data, several extrapolation methods from animal to human have been tested. For OPS201 time scaling or combination of relative mass and time scaling results in most similar TIAC values, if the organ mass ratios between the species are high. In time scaling, the scan/sampling time is scaled by using the ratio of the whole body masses of the respective species. In relative mass scaling, the TIACs are scaled based on the ratio of the whole body and organ mass of respective species. Other methods tested showed higher deviations. For the study on the influence of image quantification and the choice of the optimal scanning time points, a study in a pig model, which was performed in collaboration with Aalborg University and Octreopharm Sciences GmbH, was reanalyzed. As kidneys are organs-at-risk in PRRT with 177Lu labelled peptides, several quantification methods, based on 2D and 3D quantitative imaging were chosen. For this purpose, a 3D printed pig kidney phantom was prepared and measured with/without background activities representing the activities in the pig SPECT/CT scans. The phantom dosimetry data based on multiple SPECT/CT images and based on multiple planar images in combination with one SPECT/CT scan (MP1S Imaging) were compared to the pig dosimetry. The calculated TIACs of the phantom with background based on multiple SPECT/CT and MP1S imaging were quite similar to the multiple SPECT/CT based pig TIAC. In addition, in order to investigate the effect of late time points on dosimetry and absorbed dose values in 177Lu therapies, the difference, associated with eliminating the late two scan time points, on the TIACs was analyzed. When the TIACs (including all time points) of the pig based on multiple SPECT/CT and MP1S imaging were investigated, the use of MP1S imaging results in considerably lower TIAC values to the kidney (by a factor of 1.4). With eliminating late time points from the created time activity curve, the factor increases up to 2.4 times with a corresponding increase in TIAC uncertainties. As a consequence, further evaluation of 68Ga-OPS202 for PET/CT imaging and 177Lu-OPS201 for the treatments of NET patients is necessary. In particular, a head-to-head comparison of agonists and OPS peptides with respect to biokinetics, biodistribution and dosimetry would be helpful. In addition, the influence of the late scan time points on dosimetry needs further attention in particular for kidney dosimetry
In dieser Dissertation wird der MEK5/ERK5- Signalweg als möglicher Angriffspunkt in der zielgerichteten Melanomtherapie identifiziert. Die Adressierung von ERK5 bietet eine Alternative, um einer Resistenzentwicklung gegenüber Inhibitoren des MAPK- Signalwegs entgegenzuwirken. Das maligne Melanom ist ein hochaggressiver Tumor mit steigender Inzidenz. Zunehmende Sonnenstunden im Rahmen des Klimawandels mit erhöhter Belastung der Haut durch UV-Strahlung werden die Problematik des malignen Melanoms für den Menschen in den nächsten Jahren weiter zunehmen lassen.
Die Aktivierung des MEK5/ERK5- Signalwegs scheint eine Reaktion von Tumorzellen auf Therapiestress zu sein. Diese Aktivierung liefert den Melanomzellen einen Überlebensvorteil und verhindert ein langfristiges Therapieansprechen. ERK5 beeinflusst den Zellzyklus von Melanomzellen und ist somit möglicherweise von wichtiger Bedeutung in der Tumorgenese des malignen Melanoms.
Patienten mit NRAS- Mutation profitieren auffallend weniger von einer gezielten MEKi-Therapie als solche mit BRAF Mutation. Für ersteres Patientenkollektiv steht aktuell lediglich die Immuntherapie zur Verfügung, wodurch oft nur ein kurzes, progressionsfreies Intervall erreicht werden kann und die Patienten häufig unter schweren Nebenwirkungen leiden. Grund für die problematische Behandlung könnte das häufige Auftreten einer basalen ERK5- Aktivierung in NRAS- mutierten Melanomen sein. Diese Arbeit liefert eine positive Prognose über den Nutzen einer ERK5- Inhibition als Erweiterung des Therapieschemas. Diese These gilt auch für Melanompatienten mit einer BRAF- Mutation. Patienten, die an einem malignen Melanom erkrankt sind, weisen zu 80% eine Mutation in einem dieser beschriebenen Onkogene auf. Die Arbeit lässt darauf schließen, dass eine ERK5- Inhibition in der Therapie von beiden Gruppen erfolgreich sein könnte und somit das Leben nahezu aller Melanompatienten betrifft.
This work revealed spin states that are involved in the light generation of organic light-emitting diodes (OLEDs) that are based on thermally activated delayed fluorescence (TADF). First, several donor:acceptor-based TADF systems forming exciplex states were investigated. Afterwards, a TADF emitter that shows intramolecular charge transfer states but also forms exciplex states with a proper donor molecule was studied. The primary experimental technique was electron paramagnetic resonance (EPR), in particular the advanced methods electroluminescence detected magnetic resonance (ELDMR), photoluminescence detected magnetic resonance (PLDMR) and electrically detected magnetic resonance (EDMR). Additional information was gathered from time-resolved and continuous wave photoluminescence measurements.
The first Borono-Strecker reaction has been developed to synthesize α-aminoboronates via a multicomponent reaction of readily available carbonyl compounds (aldehydes and ketones), amines and B2pin2. The preparation of α-amino cyclic boronates can be achieved via multicomponent coupling of salicylaldehydes, amines, and B2(OH)4. In addition, the diazaborole-based PBP pincer palladium chloride and the diazaborole-based PBP pincer palladium trifluoromethanesulfonate complexes were synthesized and fully characterized for the first time, and used as catalysts for Suzuki-Miyaura cross-coupling reactions.
The quantum Hall (QH) effect, which can be induced in a two-dimensional (2D) electron gas by an external magnetic field, paved the way for topological concepts in condensed matter physics. While the QH effect can for that reason not exist without Landau levels, there is a plethora of topological phases of matter that can exist even in the absence of a magnetic field. For instance, the quantum spin Hall (QSH), the quantum anomalous Hall (QAH), and the three-dimensional (3D) topological insulator (TI) phase are insulating phases of matter that owe their nontrivial topology to an inverted band structure. The latter results from a strong spin-orbit interaction or, generally, from strong relativistic corrections. The main objective of this thesis is to explore the fate of these preexisting topological states of matter, when they are subjected to an external magnetic field, and analyze their connection to quantum anomalies. In particular, the realization of the parity anomaly in solid state systems is discussed. Furthermore, band structure engineering, i.e., changing the quantum well thickness, the strain, and the material composition, is employed to manipulate and investigate various topological properties of the prototype TI HgTe.
Like the QH phase, the QAH phase exhibits unidirectionally propagating metallic edge channels. But in contrast to the QH phase, it can exist without Landau levels. As such, the QAH phase is a condensed matter analog of the parity anomaly. We demonstrate that this connection facilitates a distinction between QH and QAH states in the presence of a magnetic field. We debunk therefore the widespread belief that these two topological phases of matter cannot be distinguished, since they are both described by a $\mathbb{Z}$ topological invariant. To be more precise, we demonstrate that the QAH topology remains encoded in a peculiar topological quantity, the spectral asymmetry, which quantifies the differences in the number of states between the conduction and valence band. Deriving the effective action of QAH insulators in magnetic fields, we show that the spectral asymmetry is thereby linked to a unique Chern-Simons term which contains the information about the QAH edge states. As a consequence, we reveal that counterpropagating QH and QAH edge states can emerge when a QAH insulator is subjected to an external magnetic field. These helical-like states exhibit exotic properties which make it possible to disentangle QH and QAH phases. Our findings are of particular importance for paramagnetic TIs in which an external magnetic field is required to induce the QAH phase.
A byproduct of the band inversion is the formation of additional extrema in the valence band dispersion at large momenta (the `camelback'). We develop a numerical implementation of the $8 \times 8$ Kane model to investigate signatures of the camelback in (Hg,Mn)Te quantum wells. Varying the quantum well thickness, as well as the Mn-concentration, we show that the class of topologically nontrivial quantum wells can be subdivided into direct gap and indirect gap TIs. In direct gap TIs, we show that, in the bulk $p$-regime, pinning of the chemical potential to the camelback can cause an onset to QH plateaus at exceptionally low magnetic fields (tens of mT). In contrast, in indirect gap TIs, the camelback prevents the observation of QH plateaus in the bulk $p$-regime up to large magnetic fields (a few tesla). These findings allowed us to attribute recent experimental observations in (Hg,Mn)Te quantum wells to the camelback. Although our discussion focuses on (Hg,Mn)Te, our model should likewise apply to other topological materials which exhibit a camelback feature in their valence band dispersion.
Furthermore, we employ the numerical implementation of the $8\times 8$ Kane model to explore the crossover from a 2D QSH to a 3D TI phase in strained HgTe quantum wells. The latter exhibit 2D topological surface states at their interfaces which, as we demonstrate, are very sensitive to the local symmetry of the crystal lattice and electrostatic gating. We determine the classical cyclotron frequency of surface electrons and compare our findings with experiments on strained HgTe.
DD is a cardiac disturbance, which has gained increasing importance in recent years due to its important role in different cardiac disease and cardiomyopathies including ischemic cardiomyopathy, arterial hypertension and diabetic cardiomyopathy.
ECG-gated 18F-FDG PET is an imaging technique, that can distinguish between districts of myocardial viability and myocardial scars and further provides information of great interest on the efficacy of experimental approaches designed to improve the cardiac function and/or myocardial metabolism in experimental small animal models. However, ECG-gated 18F-FDG PET is a technique whose feasibility in the assessment of the LV diastolic function in small animals has not been a subject of study.
In this thesis, the ability of the ECG-gated 18F-FDG PET for the assessment of both the systolic and diastolic function in eight control rats and in seven ZDF rats, which are an experimental animal model mimicking T2DM conditions and diabetic related complications in humans including DCM, has been investigated The ECG-gated 18F-FDG PET imaging was performed under hyperinsulinemic-euglycemic clamping and the data were stored in list mode files and retrospectively reconstructed. The systolic and diastolic parameters were achieved from the time/volume and the time/filling curve calculated from the software HFV. Additionally, the influence of the number of gates per cardiac cycle on the LV volumes and function parameters has been studied.
Hyperinsulinemic-euglycemic clamp procedure and blood glucose measurement did confirm the development of a manifest diabetes in the ZDF rats at the timepoint of the experiments.
Regarding the systolic parameters, no significant difference could be detected between the ZDF and ZL rats. The values for the CO were similar in both groups, which demonstrates a similar LV systolic function in the ZDF and the ZL rats at the age of 13 weeks. Values for the systolic parameters are in good line with previous PET, MRI and cardiac catheterization-based studies in diabetic rats.
The main finding of this study was that by using in vivo ECG-gated 18F-FDG PET and the software HFV, reliable diastolic parameters could be calculated. Moreover, it was possible to detect the presence of a mild impaired diastolic filling in the ZDF rats in absence of any systolic alteration. This impaired diastolic function in an early stage of diabetes could also be detected by other investigators, who used echocardiography or cardiac catheterization. Therefore, this is the first study showing, that the assessment of the diastolic function in rats can be carried out by ECG-gated 18F-FDG PET imaging.
In conclusion, additionally to calculating LV volumes and LV EF, ECG-gated 18F-FDG PET can evaluate the diastolic function of healthy and diabetic rats and is able to detect a DD in ZDF rats.
Remdesivir is the only FDA-approved drug for the treatment of COVID-19 patients. The active form of remdesivir acts as a nucleoside analog and inhibits the RNA-dependent RNA polymerase (RdRp) of coronaviruses including SARS-CoV-2. Remdesivir is incorporated by the RdRp into the growing RNA product and allows for addition of three more nucleotides before RNA synthesis stalls. Here we use synthetic RNA chemistry, biochemistry and cryoelectron microscopy to establish the molecular mechanism of remdesivir-induced RdRp stalling. We show that addition of the fourth nucleotide following remdesivir incorporation into the RNA product is impaired by a barrier to further RNA translocation. This translocation barrier causes retention of the RNA 3ʹ-nucleotide in the substrate-binding site of the RdRp and interferes with entry of the next nucleoside triphosphate, thereby stalling RdRp. In the structure of the remdesivir-stalled state, the 3ʹ-nucleotide of the RNA product is matched and located with the template base in the active center, and this may impair proofreading by the viral 3ʹ-exonuclease. These mechanistic insights should facilitate the quest for improved antivirals that target coronavirus replication.
Im Zeitraum von 2004 bis 2016 erhielten an der Neurologischen Universitätsklinik Würzburg Patienten mit einer chronisch progredienten Multiplen Sklerose insgesamt 595 Injektionen von intrathekalem Triamcinolonacetonid. Diese Arbeit befasst sich mit Sicherheit, Nebenwirkungen und Wirksamkeit der intrathekalen Therapieform.
Bei der Autoimmunerkrankung Pemphigus vulgaris führen Antikörper zur charakteristischen suprabasalen Akantholyse und Blasenbildung der Epidermis, indem sie an spezifische Antigene, Dsg3 (Desmoglein 3) und Dsg1 (Desmoglein 1), auf der Zelloberfläche der Keratinozyten binden. Die Art und Weise, wie die multiplen zellulären Pathomechanismen zusammenwirken und das potenziell tödliche Krankheitsbild hervorrufen, ist jedoch bislang noch weitgehend unklar. In der vorliegenden Arbeit wurden entscheidende, durch die Autoantikörper hervorgerufene, pathologische intrazelluläre Prozesse genauer untersucht und deren Stellenwert beleuchtet.
Constraining graph layouts - that is, restricting the placement of vertices and the routing of edges to obey certain constraints - is common practice in graph drawing.
In this book, we discuss algorithmic results on two different restriction types:
placing vertices on the outer face and on the integer grid.
For the first type, we look into the outer k-planar and outer k-quasi-planar graphs, as well as giving a linear-time algorithm to recognize full and closed outer k-planar graphs Monadic Second-order Logic.
For the second type, we consider the problem of transferring a given planar drawing onto the integer grid while perserving the original drawings topology;
we also generalize a variant of Cauchy's rigidity theorem for orthogonal polyhedra of genus 0 to those of arbitrary genus.
G-protein-coupled receptors (GPCRs) regulate diverse physiological processes in the human body and represent prime targets in modern drug discovery. Engagement of different ligands to these membrane-embedded proteins evokes distinct receptor conformational rearrangements that facilitate subsequent receptor-mediated signalling and, ultimately, enable cellular adaptation to altered environmental conditions. Since the early 2000s, the technology of resonance energy transfer (RET) has been exploited to assess these conformational receptor dynamics in living cells and real time. However, to date, these conformational GPCR studies are restricted to single-cell microscopic setups, slowing down the discovery of novel GPCR-directed therapeutics. In this work, we present the development of a novel generalizable high-throughput compatible assay for the direct measurement of GPCR activation and deactivation. By screening a variety of energy partners for fluorescence (FRET) and bioluminescence resonance energy transfer (BRET), we identified a highly sensitive design for an α2A-adrenergic receptor conformational biosensor. This biosensor reports the receptor’s conformational change upon ligand binding in a 96-well plate reader format with the highest signal amplitude obtained so far. We demonstrate the capacity of this sensor prototype to faithfully quantify efficacy and potency of GPCR ligands in intact cells and real time. Furthermore, we confirm its universal applicability by cloning and validating five further equivalent GPCR biosensors. To prove the suitability of this new GPCR assay for screening purposes, we measured the well-accepted Z-factor as a parameter for the assay quality. All tested biosensors show excellent Z-factors indicating outstanding assay quality. Furthermore, we demonstrate that this assay provides excellent throughput and presents low rates of erroneous hit identification (false positives and false negatives). Following this phase of assay development, we utilized these biosensors to understand the mechanism and consequences of the postulated modulation of parathyroid hormone receptor 1 (PTHR1) through receptor activity-modifying protein 2 (RAMP2). We found that RAMP2 desensitizes PTHR1, but not the β2-adrenergic receptor (β2AR), for agonist-induced structural changes. This generalizable sensor design offers the first possibility to upscale conformational GPCR studies, which represents the most direct and unbiased approach to monitor receptor activation and deactivation. Therefore, this novel technology provides substantial advantages over currently established methods for GPCR ligand screening. We feel confident that this technology will aid the discovery of novel types of GPCR ligands, help to identify the endogenous ligands of so-called orphan GPCRs and deepen our understanding of the physiological regulation of GPCR function.

Wenn diese mitochondriale Ca2+-Aufnahme durch Dantrolen oder Ru360 blockiert wird, dann steigt nachfolgend das zytosolische [Ca2+] an. Nach Blockierung des mRyR durch Dantrolen steigt zusätzlich durch die Beeinflussung der passiven Komponente des nukleären Ca2+-Transienten das nukleäre [Ca2+] an. Dieses erhöhte nukleäre [Ca2+] hat letztlich eine Hypertrophie zur Folge. Somit können Mitochondrien, die in ihrer Funktion gestört sind, zur Entwicklung der Hypertrophie beitragen.
These days, we are living in a digitalized world. Both our professional and private lives are pervaded by various IT services, which are typically operated using distributed computing systems (e.g., cloud environments). Due to the high level of digitalization, the operators of such systems are confronted with fast-paced and changing requirements. In particular, cloud environments have to cope with load fluctuations and respective rapid and unexpected changes in the computing resource demands. To face this challenge, so-called auto-scalers, such as the threshold-based mechanism in Amazon Web Services EC2, can be employed to enable elastic scaling of the computing resources. However, despite this opportunity, business-critical applications are still run with highly overprovisioned resources to guarantee a stable and reliable service operation. This strategy is pursued due to the lack of trust in auto-scalers and the concern that inaccurate or delayed adaptations may result in financial losses.
To adapt the resource capacity in time, the future resource demands must be "foreseen", as reacting to changes once they are observed introduces an inherent delay. In other words, accurate forecasting methods are required to adapt systems proactively. A powerful approach in this context is time series forecasting, which is also applied in many other domains. The core idea is to examine past values and predict how these values will evolve as time progresses. According to the "No-Free-Lunch Theorem", there is no algorithm that performs best for all scenarios. Therefore, selecting a suitable forecasting method for a given use case is a crucial task. Simply put, each method has its benefits and drawbacks, depending on the specific use case. The choice of the forecasting method is usually based on expert knowledge, which cannot be fully automated, or on trial-and-error. In both cases, this is expensive and prone to error.
Although auto-scaling and time series forecasting are established research fields, existing approaches cannot fully address the mentioned challenges: (i) In our survey on time series forecasting, we found that publications on time series forecasting typically consider only a small set of (mostly related) methods and evaluate their performance on a small number of time series with only a few error measures while providing no information on the execution time of the studied methods. Therefore, such articles cannot be used to guide the choice of an appropriate method for a particular use case; (ii) Existing open-source hybrid forecasting methods that take advantage of at least two methods to tackle the "No-Free-Lunch Theorem" are computationally intensive, poorly automated, designed for a particular data set, or they lack a predictable time-to-result. Methods exhibiting a high variance in the time-to-result cannot be applied for time-critical scenarios (e.g., auto-scaling), while methods tailored to a specific data set introduce restrictions on the possible use cases (e.g., forecasting only annual time series); (iii) Auto-scalers typically scale an application either proactively or reactively. Even though some hybrid auto-scalers exist, they lack sophisticated solutions to combine reactive and proactive scaling. For instance, resources are only released proactively while resource allocation is entirely done in a reactive manner (inherently delayed); (iv) The majority of existing mechanisms do not take the provider's pricing scheme into account while scaling an application in a public cloud environment, which often results in excessive charged costs. Even though some cost-aware auto-scalers have been proposed, they only consider the current resource demands, neglecting their development over time. For example, resources are often shut down prematurely, even though they might be required again soon.
To address the mentioned challenges and the shortcomings of existing work, this thesis presents three contributions: (i) The first contribution-a forecasting benchmark-addresses the problem of limited comparability between existing forecasting methods; (ii) The second contribution-Telescope-provides an automated hybrid time series forecasting method addressing the challenge posed by the "No-Free-Lunch Theorem"; (iii) The third contribution-Chamulteon-provides a novel hybrid auto-scaler for coordinated scaling of applications comprising multiple services, leveraging Telescope to forecast the workload intensity as a basis for proactive resource provisioning. In the following, the three contributions of the thesis are summarized:
Contribution I - Forecasting Benchmark
To establish a level playing field for evaluating the performance of forecasting methods in a broad setting, we propose a novel benchmark that automatically evaluates and ranks forecasting methods based on their performance in a diverse set of evaluation scenarios. The benchmark comprises four different use cases, each covering 100 heterogeneous time series taken from different domains. The data set was assembled from publicly available time series and was designed to exhibit much higher diversity than existing forecasting competitions. Besides proposing a new data set, we introduce two new measures that describe different aspects of a forecast. We applied the developed benchmark to evaluate Telescope.
Contribution II - Telescope
To provide a generic forecasting method, we introduce a novel machine learning-based forecasting approach that automatically retrieves relevant information from a given time series. More precisely, Telescope automatically extracts intrinsic time series features and then decomposes the time series into components, building a forecasting model for each of them. Each component is forecast by applying a different method and then the final forecast is assembled from the forecast components by employing a regression-based machine learning algorithm. In more than 1300 hours of experiments benchmarking 15 competing methods (including approaches from Uber and Facebook) on 400 time series, Telescope outperformed all methods, exhibiting the best forecast accuracy coupled with a low and reliable time-to-result. Compared to the competing methods that exhibited, on average, a forecast error (more precisely, the symmetric mean absolute forecast error) of 29%, Telescope exhibited an error of 20% while being 2556 times faster. In particular, the methods from Uber and Facebook exhibited an error of 48% and 36%, and were 7334 and 19 times slower than Telescope, respectively.
Contribution III - Chamulteon
To enable reliable auto-scaling, we present a hybrid auto-scaler that combines proactive and reactive techniques to scale distributed cloud applications comprising multiple services in a coordinated and cost-effective manner. More precisely, proactive adaptations are planned based on forecasts of Telescope, while reactive adaptations are triggered based on actual observations of the monitored load intensity. To solve occurring conflicts between reactive and proactive adaptations, a complex conflict resolution algorithm is implemented. Moreover, when deployed in public cloud environments, Chamulteon reviews adaptations with respect to the cloud provider's pricing scheme in order to minimize the charged costs. In more than 400 hours of experiments evaluating five competing auto-scaling mechanisms in scenarios covering five different workloads, four different applications, and three different cloud environments, Chamulteon exhibited the best auto-scaling performance and reliability while at the same time reducing the charged costs. The competing methods provided insufficient resources for (on average) 31% of the experimental time; in contrast, Chamulteon cut this time to 8% and the SLO (service level objective) violations from 18% to 6% while using up to 15% less resources and reducing the charged costs by up to 45%.
The contributions of this thesis can be seen as major milestones in the domain of time series forecasting and cloud resource management. (i) This thesis is the first to present a forecasting benchmark that covers a variety of different domains with a high diversity between the analyzed time series. Based on the provided data set and the automatic evaluation procedure, the proposed benchmark contributes to enhance the comparability of forecasting methods. The benchmarking results for different forecasting methods enable the selection of the most appropriate forecasting method for a given use case. (ii) Telescope provides the first generic and fully automated time series forecasting approach that delivers both accurate and reliable forecasts while making no assumptions about the analyzed time series. Hence, it eliminates the need for expensive, time-consuming, and error-prone procedures, such as trial-and-error searches or consulting an expert. This opens up new possibilities especially in time-critical scenarios, where Telescope can provide accurate forecasts with a short and reliable time-to-result.
Although Telescope was applied for this thesis in the field of cloud computing, there is absolutely no limitation regarding the applicability of Telescope in other domains, as demonstrated in the evaluation. Moreover, Telescope, which was made available on GitHub, is already used in a number of interdisciplinary data science projects, for instance, predictive maintenance in an Industry 4.0 context, heart failure prediction in medicine, or as a component of predictive models of beehive development. (iii) In the context of cloud resource management, Chamulteon is a major milestone for increasing the trust in cloud auto-scalers. The complex resolution algorithm enables reliable and accurate scaling behavior that reduces losses caused by excessive resource allocation or SLO violations. In other words, Chamulteon provides reliable online adaptations minimizing charged costs while at the same time maximizing user experience.
In der vorliegenden klinischen retrospektiven Studie wurden 147 Goldteilkronen und 149 Keramikteilkronen an 296 Patienten nachuntersucht. Der Beobachtungszeitraum umfasste 9-25 Jahre nach Insertion (M = 14,28, SD = 3,59).
Die systematische Bewertung der Restaurationsqualität beruhte auf modifizierten USPHS-Kriterien, klinischen Parametern sowie einer röntgenologischen Beurteilung anhand eines Zahnfilms.
Die Goldteilkronen waren zu 68% (n = 100), Keramikteilkronen zu 75,2% (n = 112) in einem exzellenten bis akzeptablen Zustand. Eine Reparatur unter Erhalt der Restauration war bei 25,9% (n = 38) der Gold- und 20,8% (n = 31) der Keramikrestaurationen notwendig. Goldrestaurationen konnten in 6,1% der Fälle (n = 9) nicht erhalten werden, Keramikrestaurationen in 4% der Fälle (n = 6).
Die Qualität der Keramikrestaurationen war hinsichtlich der Kriterien Randverfärbung, Oberflächendefekte/Retention, Randqualität und Karies niedriger als die der hochgold-haltigen Restaurationen. Die Gesamtqualität wies keine signifikanten Unterschiede zwischen den Restaurationsgruppen auf. Vorhandene Plaqueakkumulation, erhöhtes Kariesrisiko und steigendes Patientenalter wirkten sich qualitätsmindernd auf die indirekten Restaurationen aus.
Gold- und Keramikteilkronen wiesen zufriedenstellende klinische Ergebnisse auf und können gleichermaßen als langfristige, hochqualitative Versorgung für ausgedehnte Defekte im Seitenzahnbereich angesehen werden.
Ziel der prospektiven, klinischen und multizentrischen Kohortenstudie war die Untersuchung des Einflusses demographischer (Alter, SÖS), psychologischer (KIDSCREEN-10, RSES, SDQ) und kieferorthopädischer Parameter auf die Mundgesundheitsbezogene Lebensqualität (COHIP-G19) von Kindern und Jugendlichen. Hierzu wurden 250 kieferorthopädische Patienten akquiriert. Die Ergebnisse zeigten signifikante Zusammenhänge zwischen der Mundgesundheitsbezogenen Lebensqualität und: (1) dem Alter, (2) der gesundheitsbezogenen Lebensqualität, (3) dem Selbstwertgefühl, (4) Verhaltensauffälligkeiten, (5) dem objektiven kieferorthopädischen Behandlungsbedarf, (6) spezifischen kieferorthopädischen Befunden.
Rezeptor-vermittelte Chemotherapie von ovarialen Karzinomzellen mit Disorazol-GnRH-Konjugaten

Das Ovarialkarzinom stellt einen häufigen maligen Tumor der Frau dar, der meist spät diagnostiziert wird. Therapeutische Optionen sind nur eingeschränkt verfügbar und nebenwirkungsbehaftet. In der modernen Tumortherapie sind zielgerichtete medikamentöse Ansätze von immer größer Bedeutung und sind bei verschiedenen Entitäten bereits zugelassen. Da Ovarialkarzinome häufig GnRH-Rezeptoren exprimieren, stellt dies einen guten Angriffspunkt für mögliche Therapeutika dar.
In dieser Arbeit wurde die Wirkung von Disorazol, einem potenten Zytotoxin, in Kopplung an GnRH auf Ovarialkarzinom-Zellen untersucht. Unter anderem wurden hierbei RT-PCR, Kristallviolettversuche, WST-Versuche und FACS-Analysen durchgeführt.
Molekularbiologisch war eine deutliche Expression von GnRH-Rezeptoren auf ovarialen Karzinomzellen zu sehen. Es zeigte sich eine spezifische Toxizität von GnRH-Disorazol-Konjugaten auf Ovarialkarzinom-Zelllinien und andere GnRH-tragende Zellen. Lymphozyten aus dem peripheren Blut waren nicht im besonderen Maße anfällig für Disorazol. Verapamil konnte in einzelnen Zelllinien die Toxizität des Konjugats verstärken, eine Cisplatin-Resistenz hatte jedoch keinen Einfluss darauf. Apoptose-inhibierende Substanzen wie zVAD verminderten den Anteil an toten Zellen, Necrostatin war dazu nicht in der Lage.
Die spezifische Wirksamkeit von GnrH gekoppeltem Disorazol auf Ovarialkarzinomzellen bestätigt das ursprüngliche Therapiekonzept. Eine ausgeprägtere Hämatotoxizität konnte nicht nachgewiesen werden, was im Hinblick auf den klinischen Einsatz eine bedeutende Rolle spielt. Da einige weitere Entitäten wie das triple-negative Mamma-Karzinom GnRH-Rezeptor-exprimierende Zellen aufweisen, ist ein Einsatz auch in diesen Krankheitsbildern denkbar.
Fluorescence microscopy is a form of light microscopy that has developed during the 20th century and is nowadays a standard tool in Molecular and Cell biology for studying the structure and function of biological molecules. High-resolution fluorescence microscopy techniques, such as dSTORM (direct Stochastic Optical Reconstruction Microscopy) allow the visualization of cellular structures at the nanometre scale (10−9 m). This has already made it possible to decipher the composition and function of various biopolymers, such as proteins, lipids and nucleic acids, up to the three-dimensional (3D) structure of entire organelles. In practice, however, it has been shown that these imaging methods and their further developments still face great challenges in order to achieve an effective resolution below ∼ 10 nm. This is mainly due to the nature of labelling biomolecules. For the detection of molecular structures, immunostaining is often performed as a standard method. Antibodies to which fluorescent molecules are coupled, recognize and bind specifcally and with high affnity to the molecular section of the target structure, also called epitope or antigen. The fluorescent molecules serve as reporter molecules which are imaged with the use of a fluorescence microscope. However, the size of these labels with a length of about 10-15 nm in the case of immunoglobulin G (IgG) antibodies, cause a detection of the fluorescent molecules shifted to the real position of the studied antigen. In dense regions where epitopes are located close to each other, steric hindrance between antibodies can also occur and leads to an insuffcient label density. Together with the shifted detection of fluorescent molecules, these factors can limit the achievable resolution of a microscopy technique. Expansion microscopy (ExM) is a recently developed technique that achieves a resolution improvement by physical expansion of an investigated object. Therefore, biological samples such as cultured cells, tissue sections, whole organs or isolated organelles are chemically anchored into a swellable polymer. By absorbing water, this so-called superabsorber increases its own volume and pulls the covalently bound biomolecules isotropically apart. Routinely, this method achieves a magnifcation of the sample by about four times its volume. But protocol variants have already been developed that result in higher expansion factors of up to 50-fold. Since the ExM technique includes in the frst instance only the sample treatment for anchoring and magnifcation of the sample, it can be combined with various standard methods of fluorescence microscopy. In theory, the resolution of the used imaging technique improves linearly with the expansion factor of the ExM treated sample. However, an insuffcient label density and the size of the antibodies can here again impair the effective achievable resolution. The combination of ExM with high-resolution fluorescence microscopy methods represents a promising strategy to increase the resolution of light microscopy. In this thesis, I will present several ExM variants I developed which show the combination of ExM with confocal microscopy, SIM (Structured Illumination Microscopy), STED (STimulated Emission Depletion) and dSTORM. I optimized existing ExM protocols and developed different expansion strategies, which allow the combination with the respective imaging technique. Thereby, I gained new structural insights of isolated centrioles from the green algae Chlamydomonas reinhardtii by combining ExM with STED and confocal microscopy. In another project, I combined 3D-SIM imaging with ExM and investigated the molecular structure of the so-called synaptonemal complex. This structure is formed during meiosis in eukaryotic cells and contributes to the exchange of genetic material between homologous chromosomes. Especially in combination with dSTORM, the ExM method showed its high potential to overcome the limitations of modern fluorescence microscopy techniques. In this project, I expanded microtubules in mammalian cells, a polymer of the cytoskeleton as well as isolated centrioles from C. reinhardtii. By labelling after expansion of the samples, I was able to signifcantly reduce the linkage error of the label and achieve an improved label density. In future, these advantages together with the single molecule sensitivity and high resolution obtained by the dSTORM method could pave the way for achieving molecular resolution in fluorescence microscopy
Strahlentherapie des Zervixkarzinoms untergliedert sich in die Teletherapie von extrakorporal und die Brachytherapie von intrauterin und intravaginal. Während in Bezug auf die Teletherapie international standardisierte Therapieschemata bestehen, fehlen diese bei der Brachytherapie. In Würzburg wurde ein Therapieregime von 3x8,5 Gy gewählt, während in anderen Therapierzentren mit z.B. 4x7 Gy oder 5x6 Gy bestrahlt wurde. Somit wurden in Würzburg deutlich höheren Einzeldosen appliziert.
Ziel der Arbeit war es, das in Würzburg angewandte Fraktionierungsschema auf Verträglichkeit und Effektivität zu überprüfen. In die Studie eingeschlossen wurden 74 Patientinnen, die zwischen Februar 1999 und September 2014 bei gesichertem Zervixkarzinom einer primären Radiochemotherapie unterzogen wurden. In einem medianen Follow-Up von 48 Monaten wurden Nebenwirkungen an Blase und Rektum sowie lokale und regionäre Rezidive und Fernmetastasen erhoben.
Bei neun Patientinnen (12,2%) wurde am Rektum eine Spätnebenwirkung vom Grad III oder IV nach CTCAE festgestellt, an der Blase wurde dies bei drei Patientinnen (4,8%) gefunden. Bei zwei (2,7%) Patientinnen fand sich ein lokaler, bei drei Patientinnen (4%) ein regionärer Progress. 19 Patientinnen (25,66%) entwickelten Fernmetastasen.
Es zeigte sich zusammenfassend, dass es im Rahmen des angewandten Therapieschemas zu einer im internationalen Vergleich sehr guten lokalen Kontrolle kam bei vor allem am Rektum leicht erhöhten höhergradigen Nebenwirkungsraten. Mittels moderner bildgestützter IMRT- und Brachytherapie-Planung werden sich diese Toxizitäten in Zukunft weiter reduzieren lassen.
Die Stammzellforschung beschäftigt sich bereits seit Jahren mit der Frage, wie Gewebe oder sogar Organe im Labor hergestellt werden können. Als besonders vielversprechend erscheinen hierfür humane Mesenchymale Stammzellen (hMSC), da diese in vielen Fällen direkt vom Empfänger gewonnen werden können und so keine Organ- oder Gewebeabstoßung durch Abwehrreaktionen zu erwarten ist. Für die weitere Erforschung des Verhaltens von Stammzellen in vivo ist es notwendig, diese nicht-invasiv darstellen zu können. Dies ist zum Beispiel mittels Magnetischer Partikel Bildgebung (MPI) möglich. Hierfür müssen die Stammzellen mit einer geeigneten Substanz markiert werden. Eine solche sind beispielsweise superparamagnetische Eisenoxidnanopartikel (SPION). Derzeit gibt es keine von den medizinischen Zulassungsbehörden zugelassenen SPION die ohne TA in hMSC aufgenommen werden. In der hier vorliegenden Arbeit sollte also im Rahmen des
EU-weiten „IDEA-Projekts“ ein geeigneter SPION identifiziert werden, der eine optimale Zell-Partikel-Interaktion aufweist und sowohl mittels MPI als auch mit Raman-Spektroskopie nachweisbar ist. Zudem sollte die Nachweisbarkeit des SPION über einen längeren Zeitraum gegeben und kein Einfluss auf die hMSC feststellbar sein. Es wurden hMSC mit den Eisenoxidnanopartikeln M4E, M4F, M4F2 und M3A-PDL in unterschiedlichen Konzentrationen markiert. Für M3A-PDL und M4E erfolgten bei einer Konzentration von 0,5 mg/ml Untersuchungen in Zellkultur sowie auf SIS-ser als Matrix im 3D-Modell. Desweiteren wurde das Differenzierungsverhalten der mit M4E markierten hMSC bei chondrogener Differenzierung untersucht. Außerdem kamen Magnetische Partikel Spektroskopie (MPS) und Raman-Spektroskopie als nicht-invasive Nachweisverfahren zum Einsatz. Der SPION-Nachweis erfolgte histologisch mittels Berliner Blau Färbung. Untersuchungen zu Zellviabilität und Proliferation erfolgten durch Trypanblau sowie Ki67-Antikörper-Färbung. Um Nachzuweisen ob auch markierte Zellen proliferieren wurde eigens ein kombiniertes Färbeprotokoll zur Kombination von Berliner Blau und immunhistochemischer Färbung
etabliert. Der Erfolg der chrondrogenen Differenzierung wurde mittels Alcianblau, Aggrecan- und Kollagen-II-Antikörper Färbung überprüft. Es konnte gezeigt werden, dass M4E bei der Markierung von hMSC eine sehr gute Zell-Partikel-Interaktion aufweist und im Gegensatz zu M3A auch ohne TA in die Zellen aufgenommen wird. Durch beide Partikel werden Zellviabilität und Proliferation nicht beeinflusst. M4F sowie M4F2 ist zur Markierung nicht geeignet. Die Markierung ließ sich im 3D-Modell mit vier Wochen deutlich länger nachweisen als in 2D Zellkultur mit maximal zwei Wochen. Die chondrogene Differenzierung wird durch die Markierung mit 0,5 mg/ml M4E beeinflusst. M3A-PDL sind durch MPS nachweisbar. Die Raman-Spektroskopie eignet sich zur Differenzierung zwischen mit M3A-PDL markierten und unmarkierten hMSC. Es ist im Rahmen dieser Arbeit gelungen, einen
Eisenoxidnanopartikel mit hervorragender Zell-Partikel-Interaktion zu identifizieren, der ohne zusätzliches TA eine intensive Markierung der hMSC ermöglicht und mit MPS nachweisbar ist. Für M4E konnte in weiteren Arbeiten am Institut bereits gezeigt werden, dass auch eine Differenzierung zwischen markierten und unmarkierten Zellen mittels Raman-Spektroskopie möglich ist. Die chondrogene Differenzierung der hMSC wurde in der vorliegenden Arbeit allerdings beeinträchtigt. In der Literatur finden sich Hinweise auf eine dosisabhängige Inhibition der Differenzierung. Es sind daher weitere Versuche notwendig, um herauszufinden, ob die Inhibition der Differenzierung möglicherweise bei geringerer SPION-Konzentration weniger ausgeprägt ist. Zudem sollte untersucht werden, ob auch geringere Konzentrationen in den Zellen über mehrere Wochen mittels MPS nachweisbar bleiben. Desweiteren sollten Untersuchungen in, der in vivo Situation ähnlicheren,
Systemen, wie dem dynamischen Umfeld einer BioVaSc-TERM® durchgeführt werden um bessere Vorhersagen zum Verhalten markierter hMSC in vivo treffen zu können.
E-Mails, Online Banking und Videokonferenzen sind aus unserem heutigen Alltag nicht mehr wegzudenken. Bei all diesen Aktivitäten werden zahlreiche personenbezogene Informationen und vertrauenswürdige Daten digital übertragen und gespeichert. Zur Sicherstellung der digitalen Daten vor unbefugten Zugriffen und Manipulationen existieren verschiedenste Konzepte, Methoden und Verfahren, die sich unter dem Begriff IT-Sicherheit zusammenfassen lassen. Klassische Sicherheitslösungen aus dem Bereich IT-Sicherheit sind Firewalls und Virenscanner. Derartige Ansätze sind meist regelbasiert und prüfen Dateien beziehungsweise eingehenden Netzwerkverkehr anhand einer Liste bekannter Angriffssignaturen. Folglich können diese Systeme nur bereits bekannte Angriffsszenarien detektieren und bieten keinen Schutz vor neuartigen Angriffen. Somit entsteht im Bereich IT-Sicherheit ein Wettlauf zwischen Hackern und IT-Sicherheitsexperten, bei dem die Hacker stets nach neuen Mitteln und Wegen suchen, die existierenden Sicherheitslösungen zu überwinden, während IT-Sicherheitsexperten stetig ihre Schutzmechanismen verbessern.
Die vorliegende Arbeit widmet sich der Detektion von Angriffsszenarien in Unternehmensnetzwerken mithilfe von Data Mining-Methoden. Diese Methoden sind in der Lage anhand von repräsentativen Daten die darin enthaltenen Strukturen zu erlernen und zu generalisieren. Folglich können sich Data Mining-Methoden grundsätzlich zur Detektion neuer Angriffsszenarien eignen, wenn diese Angriffsszenarien Überschneidungen mit bekannten Angriffsszenarien aufweisen oder sich wesentlich vom bekannten Normalverhalten unterscheiden. In dieser Arbeit werden netzwerkbasierte Daten im NetFlow Format analysiert, da diese einen aggregierten Überblick über das Geschehen im Netzwerk bieten. Häufig können Netzwerkdaten aufgrund datenschutzrechtlicher Bedenken nicht veröffentlicht werden, was für die Erzeugung synthetischer, aber realistischer Netzwerkdaten spricht. Des Weiteren führt die Beschaffenheit der Netzwerkdaten dazu, dass eine Kombination von kontinuierlichen und kategorischen Attributen analysiert werden muss, was vor allem das Vergleichen der Daten bezüglich ihrer Ähnlichkeit erschwert.
Diese Arbeit liefert methodische Beiträge zu jeder der drei genannten Herausforderungen. Im Bereich der Abstandsberechnung kategorischer Werte werden mit ConDist und IP2Vec zwei unterschiedliche Ansätze entwickelt. ConDist ist ein universell einsetzbares Abstandsmaß zur Berechnung von Abständen zwischen Datenpunkten, die aus kontinuierlichen und kategorischen Attributen bestehen. IP2Vec ist auf Netzwerkdaten spezialisiert und transformiert kategorische Werte in kontinuierliche Vektoren.
Im Bereich der Generierung realistischer Netzwerkdaten werden neben einer ausführlichen Literaturrecherche zwei unterschiedliche Ansätze vorgestellt. Zunächst wird ein auf Simulation basierter Ansatz zur Generierung flowbasierter Datensätze entwickelt. Dieser Ansatz basiert auf einer Testumgebung und simuliert typische Benutzeraktivitäten durch automatisierte Python Skripte. Parallel hierzu wird ein zweiter Ansatz zur synthetischen Generierung flowbasierter Netzwerkdaten durch Modellierung mithilfe von Generative Adversarial Networks entwickelt. Dieser Ansatz erlernt die zugrundeliegenden Eigenschaften der Netzwerkdaten und ist anschließend in der Lage, neue Netzwerkdaten mit gleichen Eigenschaften zu generieren.Während sich der erste Ansatz zur Erstellung neuer Datensätze eignet, kann der zweite Ansatz zur Anreicherung existierender Datensätze genutzt werden.
Schließlich liefert diese Arbeit noch zwei Beiträge zur Detektion von Angriffsszenarien. Im ersten Beitrag wird ein Konzept zur Detektion von Angriffsszenarien entwickelt, welches sich an die typischen Phasen eines Angriffsszenarios orientiert. Im zweiten Beitrag werden eine überwachte und eine unüberwachte Methode zur Detektion von langsamen Port Scans vorgestellt.
High-resolution nuclear magnetic resonance (NMR) spectroscopy is used in structure elucidation and qualitative as well as quantitative examination of product components. Despite the worldwide development of numerous innovative NMR spectroscopic methods, several official methods that analyze specific substances and do not represent a holistic analysis, are still in use for the quality control of drugs, food and chemicals. Thus, counterfeit or contaminated products of inferior quality can be brought onto the market and distributed despite previous quality controls. To prevent this, three NMR spectroscopic methods have been developed within the scope of this work (1) to study the peroxide value in vegetable and animal oils, (2) for the qualitative and quantitative analysis of metal cations and (3) to determine the enantiomeric excess in chiral alcohols. In oil analysis, titration methods are used to determine the bulk quality parameters such as peroxide value, which represents the concentration of peroxides. Titrations show several drawbacks, such as the need of a large amount of sample and solvents, cross reactions and the low robustness. Thus, an alternative NMR spectroscopic method was developed to improve the peroxide analysis by using triphenylphosphine as a derivatization reagent, which reacts with peroxides in a stoichiometric ratio of 1:1 forming triphenylphosphine oxide. In the 1H-31P decoupled NMR spectrum, the signals of the unreacted triphenylphosphine and the reacted triphenylphosphine oxide are detected at 7.4 ppm and 7.8 ppm, respectively. The ratio of the two signals is used for the calculation of the peroxide concentration. 108 oil samples with a peroxide value between 1 meq/kg and 150 meq/kg were examined using the developed method. Oils with a very low peroxide value of less than 3 meq/kg showed a relative standard deviation of 4.9%, highly oxidized oils with a peroxide value of 150 meq/kg of 0.2%. The NMR method was demonstrated as a powerful technique for the analysis of vegetable and krill oils. Another 1H NMR spectroscopic method was developed for the qualitative determination of Be2+, Sr2+ and Cd2+, and for the qualitative and quantitative determination of Ca2+, Mg2+, Hg2+, Sn2+, Pb2+ and Zn2+ by using ethylenediamine tetraacetate (EDTA) as complexing agent. EDTA is a hexadentate ligand that forms stable chelate complexes with divalent cations. The known amount of added EDTA and the signal ratio of free and complexed EDTA are used to calculate the concentrations of the divalent cations, which makes the use of an internal standard obsolete. The use of EDTA with Be2+, Sr2+, Cd2+, Ca2+, Mg2+, Hg2+, Sn2+, Pb2+ and Zn2+ result in complexes whose signals are pH-independent, showing cation-specific chemical shifts and couplings in the 1H NMR spectrum that are used for identification and quantification. In the presented NMR method, the limit of quantification of the cations Ca2+, Mg2+, Hg2+, Sn2+, Pb2+, and Zn2+ was determined with 5-22 μg/mL. This method is applicable in the food and drug sectors. The third NMR spectroscopic method introduced an alternative determination of the enantiomer excess (ee) of the chiral alcohols menthol, borneol, 1-phenylethanol and linalool using phosgene as a derivatizing reagent. Phosgene reacts with a chiral alcohol to form carboxylic acid diesters, made of two identical (RR, SS) or two different enantiomers (RS, SR). These two different types of diastereomers can be examined by the difference of their chemical shifts. In the presented method, the integration values of the carbonyl signals in the 13C NMR spectrum are used for the determination of the enantiomer excess. The limit of quantification depends, among others, on the sample and on the non-labelled or 13C-labelled phosgene used for the analysis. In the case of menthol, a quantification limit of ee=99.1% was determined using non-labelled phosgene and ee=99.9% using 13C-labelled phosgene. The 13C NMR method was also applied for the quality control of the enantiomeric purity of borneol, 1-phenylethanol and linalool. The developed 13C NMR method represents a powerful alternative to Mosher’s reagent for investigating the enantiomeric excess in chiral alcohols. This work demonstrates the variety of possibilities of applications for the quantitative nuclear magnetic resonance spectroscopy in the chemical analysis of drugs, food and chemicals using tagging reactions such as derivatizations and complexations. The nuclear resonance spectroscopic methods developed in this research work represent powerful alternatives to the previously used quality control techniques.
Cognitive control is what makes goal-directed actions possible. Whenever the environment or our impulses strongly suggests a response that is incompatible with our goals, conflict arises. Such conflicts are believed to cause negative affect. Aversive consequences of conflict may be registered in a conflict monitoring module, which subsequently initiates attentional changes and action tendencies to reduce negative affect. This association suggests that behavioral adaptation might be a reflection of emotion regulation. The theoretical cornerstone of current research on emotion regulation is the process model of emotion regulation, which postulates the regulation strategies situation selection, situation modification, attentional deployment, cognitive change, and response modulation. Under the assumption that conflict adaptation and affect regulation share common mechanisms, I derived several predictions regarding cognitive control from the process model of emotion regulation and tested them in 11 experiments (N = 509). Participants engaged in situation selection towards conflict, but only when they were explicitly pointed to action-outcome contingencies (Experiments 1 to 3). I found support for a mechanism resembling situation modification, but no evidence for a role of affect (Experiments 4 to 10). Changing the evaluation of conflict had no impact on the extent of conflict adaptation (Experiment 11). Overall, there was evidence for an explicit aversiveness of cognitive conflict, but less evidence for implicit aversiveness, suggesting that conflict may trigger affect regulation processes, particularly when people explicitly have affect regulation goals in mind.
The family of trypanosomatid parasites, including the human pathogens Trypanosoma brucei and Leishmania, has evolved sophisticated strategies to survive in harmful host environments. While Leishmania generate a safe niche inside the host’s macrophages, Trypanosoma brucei lives extracellularly in the mammalian bloodstream, where it is constantly exposed to the attack of the immune system. Trypanosoma brucei ensures its survival by periodically changing its protective surface coat in a process known as antigenic variation. The surface coat is composed of one species of ‘variant surface glycoprotein’ (VSG). Even though the genome possesses a large repertoire of different VSG isoforms, only one is ever expressed at a time from one out of the 15 specialized subtelomeric ‘expression sites’ (ES). Switching the coat can be accomplished either by a recombination-based exchange of the actively-expressed VSG with a silent VSG, or by a transcriptional switch to a previously silent ES.
The conserved histone methyltransferase DOT1B methylates histone H3 on lysine 76 and is involved in ES regulation in T. brucei. DOT1B ensures accurate transcriptional silencing of the inactive ES VSGs and influences the kinetics of a transcriptional switch. The molecular machinery that enables DOT1B to execute these regulatory functions at the ES is still elusive, however. To learn more about DOT1B-mediated regulatory processes, I wanted to identify DOT1B-associated proteins.
Using two complementary approaches, specifically affinity purification and proximity-dependent biotin identification (BioID), I identified several novel DOT1B-interacting candidates. To validate these data, I carried out reciprocal co-immunoprecipitations with the most promising candidates. An interaction of DOT1B with the Ribonuclease H2 protein complex, which has never been described before in any other organism, was confirmed. Trypanosomal Ribonuclease H2 maintains genome integrity by resolving RNA-DNA hybrids, structures that if not properly processed might initiate antigenic variation. I then investigated DOT1B’s contribution to this novel route to antigenic variation. Remarkably, DOT1B depletion caused an increased RNA-DNA hybrid abundance, accumulation of DNA damage, and increased VSG switching. Deregulation of VSGs from throughout the silent repertoire was observed, indicating that recombination-based switching events occurred. Encouragingly, the pattern of deregulated VSGs was similar to that seen in Ribonuclease H2-depleted cells. Together these data support the hypothesis that both proteins act together in modulating RNA-DNA hybrids to contribute to the tightly-regulated process of antigenic variation.
The transmission of trypanosomatid parasites to mammalian hosts is facilitated by insect vectors. Parasites need to adapt to the extremely different environments encountered during transmission. To ensure their survival, they differentiate into various specialized forms adapted to each tissue microenvironment. Besides antigenic variation, DOT1B additionally affects the developmental differentiation from the mammalian-infective to the insect stage of Trypanosoma brucei. However, substantially less is known about the influence of chromatin-associated proteins such as DOT1B on survival and adaptation strategies of related Leishmania parasites. To elucidate whether DOT1B’s functions are conserved in Leishmania, phenotypes after gene deletion were analyzed. As in Trypanosoma brucei, generation of a gene deletion mutant demonstrated that DOT1B is not essential for the cell viability in vitro. DOT1B deletion was accompanied with a loss of histone H3 lysine 73 trimethylation (the lysine homologous to trypanosomal H3K76), indicating that Leishmania DOT1B is also solely responsible for catalyzing this post-translational modification. As in T. brucei, dimethylation could only be observed during mitosis/cytokinesis, while trimethylation was detectable throughout the cell cycle in wild-type cells. In contrast to the trypanosome DOT1B, LmxDOT1B was not essential for differentiation in vitro. However, preliminary data indicate that the enzyme is required for effective macrophage infection.
In conclusion, this study demonstrated that the identification of protein networks and the characterization of protein functions of orthologous proteins from related parasites are effective tools to improve our understanding of the parasite survival strategies. Such insights are a necessary step on the road to developing better treatments for the devastating diseases they cause.
Die Dissertation untersucht hospitalisierte Patienten in der Kinderklinik des Universitätsklinikum Innsbruck, die wegen einer rotaviren assoziierten Gastroenteritis hospitalisiert wurden. Der Untersuchungszeitraum war von Januar 2002 bis Dezmber 2012.Untersucht wurde die Hospitalisierungsrate, Hospitalisierungsdauer, Hospitalisierungsalter, die Schwere der Erkrankung sowie eine Änderung von nosokomialen und ambulanten Erwerb vor und nach Impfeinführung im Juli 2007.
Zielvorgabe dieser Dissertation stellte die Konstruktion eines Plasmides mit mehreren shRNA-Expressionskassetten für den simultanen Knockdown onkogener Zielstrukturen im Multiplen Myelom dar. Es sollte sich zudem um ein Konstrukt handeln, bei dem ein beliebig häufiges Einfügen oder Ersetzen von Expressionskassetten durch einfache Klonierungsschritte möglich ist.
Im Rahmen dieser Arbeit wurden Plasmide mit bis zu vier unterschiedlichen shRNA-Expressionskassetten konstruiert und getestet. Mittels Elektroporation und anschließender Analyse der Knock-down-Effizienz im Western Blot konnte beispielhaft an Proteinen des MEK-ERK-Signalweges gezeigt werden, dass mithilfe der klonierten Plasmide die Spiegel von mindestens vier Zielproteinen gleichzeitig herunterreguliert werden können, ohne dass es dabei zu Einschränkungen im Vergleich zur Verwendung von Einzel-shRNA-Expressionsvektoren kommt. Es konnte gezeigt werden, dass weder die zunehmende Menge an hintereinander klonierten Kassetten noch die Reihenfolge der einzelnen Expressionskassetten im Plasmid einen negativen Einfluss auf das Knock-down-Ergebnis haben. Dieses Ergebnis konnte in verschiedenen Zelllinien des Multiplen Myeloms bestätigt werden. Die Vorteile des Verfahrens liegen vor allem in der Kosteneffizienz, der einfachen Herstellung und Modifikation, sowie der niedrigen biologischen Sicherheitsstufe der Versuchsschritte. Bedeutend ist zudem die Vermeidung von toxischen Effekten, welche das Einbringen von Fremd-DNA in Zellen ab gewissen Mengen mit sich bringt, durch Klonierung aller shRNA-Expressionskassetten zu lediglich einem Plasmidkonstrukt. Einschränkungen erfährt die etablierte Methodik dadurch, dass die zu untersuchenden Zellen sich für die Transfizierung mit Plasmiden eigenen müssen und ein Verfahren der Selektion transfizierter von nativen Zellen verfügbar sein muss. Durch genannte Vorteile stellt das Konstrukt jedoch ein hervorragendes Mittel zur Erforschung zellulärer Signalwege und ihrer Interaktionen weit über die Erkrankung des Multiplen Myeloms hinaus dar. Es kann zudem als kostengünstige molekulare Methode zur Identifizierung neuer pharmakologischer Angriffspunkte bei Tumorerkrankungen dienen.
Die Ergebnisse dieser Arbeit wurden in Folgeversuchen genutzt um eine stabile Integration der shRNA-Expressionskassetten in die zelluläre DNA sowie eine pharmakologische Induzierbarkeit zu erreichen. Es konnte somit der Sprung von den transienten Knock-down-Versuchen dieser Arbeit hin zur langfristig verminderten Proteintranslation geschafft werden. Am Ende dieser Arbeit verblieb zu eruieren, inwieweit die Anzahl der shRNA-Expressionskassetten über die getestete Anzahl von vier erweiterbar wäre und ob die guten Erfahrungen mit dieser Methodik auch bei anderen Arten von Tumorzellen replizierbar sind. Das etablierte Vektorsystem könnte in künftigen Versuchen zur schnellen Erforschung bisher wenig untersuchter oder unbekannter Signalwege dienen.
Morbus Fabry betrifft als lysosomale Speicherkrankheit viele Organsysteme durch die Ablagerung von Gb3 in verschiedenen Geweben. Besonders durch die Beteiligung von Nieren und Herz, wird die Lebenszeit von den Patienten häufig verkürzt. Eine Beschreibung konkreter klinischer Symptome, welche auch durch Allgemeinmediziner oder Zahnärzte erkannt werden könnten, könnte eine frühzeitigere Diagnose und damit frühzeitige Therapie ermöglichen. Besonders extraorale gesichtsspezifische Merkmale können von verschiedensten Gruppen von Ärzten erkannt werden.
Die extraorale Auswertung zeigte, wie in der Literatur beschrieben, das Vorkommen von periorbitaler Fülle, prominente Arcus superciliaris, eine kürzere und bullösere Nase. Die Auffälligkeiten waren besonders bei den Männern zu beobachten.
Die intraorale Auswertung wurde in dentale Auffälligkeiten und Ereignisse des Hart- und Weichgewebes eingeteilt. Bei den dentalen Ereignissen zeigte sich eine Diskrepanz zwischen der Kiefergröße und dem Zahnmaterial. So neigte das Patientenkollektiv eher zu einem Breitkiefer, was eine Erklärung für die multiplen Lücken im Frontzahnbereich der Patienten darstellt. An der Mundschleimhaut und perioral konnten vermehrt Angiokeratome und Teleangiektasien festgestellt werden, sowie das vermehrte Vorkommen von Exostosen. Speziell die Zunge der Patienten zeigte auch Auffälligkeiten in Form von einer subjektiven Makroglossie, einer Furchenzunge und Veränderungen der Papillen.
Die Auffälligkeiten in der Mundhöhle und im Kopf-Hals Bereich der Morbus Fabry Patienten sind, wie der Literatur beschrieben, vorhanden, jedoch stellen sie keine Schlüsselrolle in der Diagnose dar, da sie in allen Bereichen nur leichte Abweichungen oder Auffälligkeiten zeigen, welche nicht immer Auftreten und daher schwer zu diagnostizieren sind.
Typ 1 Diabetes mellitus (T1DM) ist eine Autoimmunerkrankung, welche durch die progrediente Schä¬digung der βZellen der Pankreas mit dadurch resultierendem absolutem Insulin¬mangel gekennzeichnet ist. Obwohl sich eine Vielzahl an Studien in den ver¬gangenen Jahrzehnten den zellulären Grundlagen dieser Erkrankung ge¬widmet hat, fehlt bis dato ein geeigneter kausaler Therapieansatz, sodass nach wie vor lediglich die eher symptomatisch orientierte Insulin-Substitution im Vor-dergrund steht.
Die hier vorliegende Arbeit beschäftigte sich mit der genauen Charakterisierung von Immunzellen – insbesondere Lymphozyten – aus dem peripheren Blut er¬krankter Kinder mit dem Ziel, Verschiebungen im Phänotyp dieser Zellen aufzudecken, um potenzielle Ansatzpunkte zur weiteren Forschung und Therapiefindung zu finden. Hierfür wurde die Durchflusszytometrie genutzt, um den genauen Phänotyp der Lymphozyten zu untersuchen und Veränderungen im Oberflächenmarker-Expressionsmuster einzelner spezifischer Subpopulati¬onen zuweisen zu können.
In erster Linie wurden hierbei zwei Zellpopulationen identifiziert, welche bei erkrankten Kindern deutlich vermehrt im peripheren Blut vorkommen, als bei gesunden Vergleichspersonen.
Die Erste dieser Populationen sind CCR6-exprimierende CD4+ Zellen. Dies ist interessant, da CCR6 in verschiedenen früheren Studien mit Autoim¬munität in Verbindung gebracht wurde. In der weiteren Analyse zeigte sich, dass eine erhöhte IFNγ-Produktion in der vermutlich selben Zellpopulation vorlag, die auch CCR6 verstärkt exprimierte (Gedächtnis-T-Zellen vom Typ CD4+CD45RO+CCR7+CD28+), während kein Anhalt auf eine geänderte IL 17-Produktion gesehen wer¬den konnte.
Die zweite in unserer Studie bei T1DM deutlich vermehrte Zellpopulation sind CXCR3-exprimierende CD8+ Zellen. Hierbei handelte es sich in erster Linie um Zellen, welche auch CCR7 exprimierten.
In der hier vorliegenden Studie wurde die T1DM-Gruppe außerdem aufgeteilt nach der Krankheitsdauer (recent onset T1DM RO und long-standing T1DM LS), um im Krankheitsverlauf entstehende Veränderungen erkennen zu können. Es konnten jedoch insgesamt keine klaren derartigen Veränderungen festgestellt werden, was allerdings möglicherweise auch an einer zu geringen überblickten Zeitspanne liegen könnte (geringe durchschnittliche und maximale Krankheitsdauer).
Aufgrund unserer Ergebnisse kann spekuliert werden, dass es sich bei den prozentuell vermehrt auftre¬tenden CCR6+ Zellen um IFNγ-produzierende Zellen handelt. Dies würde neueren Erkenntnissen von anderen Studien entsprechen, welche nicht – wie zuvor vermu¬tet wurde – Th17 Zellen, sondern nicht-klassische Th1 Zellen als pathogene Population diskutieren. Ein quantitativer Mangel an Treg als Ursache der ge¬steigerten IFNγ-Produktion erscheint nach unseren Ergebnissen unwahrscheinlich, da wir erhöhte Mengen CD25+CD127 /dim Zellen bei T1DM nachweisen konnten.
Die beobachtete Koexpression von CXCR3 und CCR7 ist interessant vor dem Hintergrund, dass CXCR3-Überexpression im Pankreas in einer früheren Studie mit T1DM in Verbindung gebracht wurde, jedoch eine CXCR3-Blockade bisher keinen geeigneten therapeutischen Ansatz lieferte. Zunächst kann hier aber nur eine Assoziation beschrieben werden, wobei ein kausaler Zusammenhang in geeigneten Studiendesigns überprüft werden müsste.
Eine genauere Betrachtung der CD8+CCR7+CXCR3+ Zellen könnte nach unserer Ansicht interessante neue Aspekte und potenzielle Therapieansätze offenlegen. Insbesondere scheint die Untersuchung CCR7-vermittelter Migration dieser Zel¬len einen geeigneten Studienansatz darzustellen. Ebenso verweisen unsere Er-gebnisse erneut auf eine pathogene Rolle CCR6-exprimierender Zellen, welche in kommenden Studien näher beleuchtet werden sollte.
Zusammenfassung:
Unsere Arbeit bestätigt die aus kleineren Studien bekannte hohe Expression der Chemokinrezeptoren CXCR4 und CXCR7 in der normalen Nebenniere und in der Mehrheit der Nebennierenkarzinome. Das auf mRNA Ebene bestätigte Vorkommen beider Chemokinrezeptoren im gesunden Nebennierengewebe deutet auf eine überwiegend für die normale Nebennierenphysiologie wichtige Rolle dieser Chemokinrezeptoren hin. Eine eventuell dennoch bestehende pathophysiologische Relevanz der Rezeptoren wurde ergänzend überprüft und ergab keinen signifikanten Einfluss auf die Prognose des Nebennierenkarzinoms.
Hintergrund: Das Catechol-O-Methyltransferase-Gen (COMT) ist ein vielversprechendes Kandidatengen zur Untersuchung kognitiver und emotionaler Funktionen sowie deren pathologischer Veränderungen. Ein einzelner Basenaustausch in diesem Gen führt zu einer 3-4fach höheren COMT-Aktivität der Val Variante. Ein dadurch vermitteltes dopaminerges Defizit wird als relevanter Faktor für eine veränderte Hirnfunktion angenommen.
Mit dem kognitiven Stroop-Paradigma wurden kognitive Verarbeitungsprozesse bisher gut erforscht. Zur Erfassung emotionaler Verarbeitungsprozesse wurde eine emotionale Variante entwickelt, deren neurale Grundlagen bislang weniger gut bekannt sind. Ziel: Unsere imaging genetics-Arbeit untersucht den Einfluss genetischer Varianten auf die neurale Funktion. Ziel dieser experimentellen Arbeit war es, den Einfluss des COMT-Polymorphismus (COMT-PM) auf die Frontalkortex-Funktion in ausgewählten Regionen von Interesse (ROI) zu erfassen und der Frage nachzugehen, ob das Val-Allel als Risiko-Allel zur Pathogenese einer Angststörung (AS) beitragen könnte. Zudem sollte die Tauglichkeit des emotionalen Stroop- Paradigmas als angstsensibles Messinstrument zur Untersuchung dieser Fragestellung geprüft werden. Demgegenüber steht die Annahme, das emotionale Stroop-Paradigma könnte lediglich eine Arbeitsgedächtnis (AG)-Aufgabe darstellen. Methoden: Mittels funktioneller Nahinfrarotspektroskopie (fNIRS) und ereigniskorrelierter Potentiale untersuchten wir 121 gesunde nach dem COMT- Val158Met-PM stratifizierte Probanden während eines kombiniert emotional- kognitiven Stroop-Paradigmas. Als neurale Korrelate von Exekutivfunktionen und AG-Aufgaben waren die ROI dabei der laterale präfrontale und inferiore Kortex, die auch mit emotionaler Regulation in Verbindung gebracht werden. Als Parameter der Reaktion des autonomen Nervensystems (ANS) diente die Erfassung der elektrodermalen Aktivität sowie die kontinuierliche Messung von Blutdruck, Herzfrequenz und Herzratenvariabilität. Ergebnisse: Bei allen drei COMT Varianten zeigte sich ein kognitiver Stroop-Effekt mit verlängerter Reaktionszeit und erhöhter Fehleranzahl während der Präsentation inkongruenter Farbworte. Als Reaktion des ANS stellte sich eine erhöhte elektrodermale Aktivität bei inkongruenten Farbworten dar. Die funktionelle Bildgebung ließ in den analysierten Regionen eine erhöhte präfrontale Aktivierung während der Verarbeitung inkongruenter Farbworte nachweisen. Es fanden sich keine Gruppenunterschiede im kognitiven Stroop-Paradigma. Der einzige emotionale Stroop-Effekt zeigte sich in der P300. Der einzig nachweisbare Gruppeneffekt stellte sich im emotionalen Stroop-Paradigma als höhere Fehleranzahl bei Met-Homozygoten verglichen mit Heterozygoten dar. Schlussfolgerung: Genetische Information und funktionelle Bildgebung kombiniert sollten ermöglichen, neurale Mechanismen zu definieren, die mit genetischen Varianten verlinkt sind. Die Ergebnisse bezogen auf die analysierten Regionen liefern keinen Hinweis auf ein Val-Allel assoziiertes Risiko für die Entwicklung einer AS. Damit gelingt es nicht, bisher gewonnene Ergebnisse zum Einfluss des COMT-PM auf die präfrontale Funktion zu replizieren. Fraglich ist jedoch, ob sich das emotionale Stroop-Paradigma zur Untersuchung dieser Frage eignet, da weder in den fNIRS-, noch in den autonomen oder Verhaltensdaten ein emotionaler Stroop-Effekt nachgewiesen werden konnte.
Die Narbenhernie stellt eine häufige Komplikation nach Laparotomien dar. Die Therapie der Narbenhernie erfolgt mittels chirurgischer Netzimplantation. Dieses Verfahren erfordert detaillierte anatomische Kenntnisse. Dem ethischen Imperativ folgend, wurde ein kosten-effizientes Modell entwickelt, welches den humanen Situs imitiert und an dem sich eine retromuskuläre Netzimplantation durchführen lässt. Das High-Fidelity-Modell besteht zum Hauptteil aus 2-Komponenten-Silikon. Das Modell wurde entwickelt und im Rahmen dieser Studie validiert. Zur Ermittlung der Testpersonenanzahl wurde die Methodik des sequentiellen Dreieckstests genutzt. Nachdem 6 Anfänger (PJ-Studierende) und 6 Könner (Fachärzte für Viszeralchirurgie) jeweils ein Modell operiert hatten, wurde die Kontent-, die Konstrukt- und die Kriterienvalidität untersucht. Anschließend wurde das Modell und die Operationsdurchführung mit drei Methoden untersucht. Zum einen füllten die Teilnehmenden einen Fragebogen bezüglich der Realitätsnahe des Modells direkt nach der Operation aus. Außerdem bewerteten drei verblindete Bewerter die Operationen anhand des Competency assessment tool (CAT), welcher eine modifizierte Version des Fragebogens nach Miskovic darstellt, nach den folgenden Subskalen: „Instrumentengebrauch“, „Umgang mit dem Gewebe“, „Mängel und Fehler“, „Qualität des Endprodukts“. Zuletzt wurden die operierten Modelle bezüglicher der „Endergebnisse“ autopsiert und bewertet.
Die Ergebnisse zeigen, dass am SUBsON-Modell eine Narbenhernienoperation mit Netzimplantation authentisch durchgeführt werden kann. Die Testpersonen bewerteten das Modell als realitätsnah. Die Reliabilität war in allen Kategorien gut bis exzellent. Die Könner waren in allen Subskalen des CATs den Anfängern überlegen. Bei Betrachtung der Kriterienvalidität zeigte sich ein paradoxer Effekt: Bei der Präparation des Fatty Triangles erbrachten die Anfänger eine signifikant (p< 0,05) höhere Leistung als die Könner. Mögliche Erklärungen dafür sind mannigfaltig.
Die Leistungsunterschiede zwischen Anfängern und Könnern bestätigen die Konstruktvalidität von Modell und Fragebogen sowie die Realitätsnähe des Modells. In dieser Studie konnten Defizite vor allem unter Könnern bezüglich anatomischer Kenntnisse bei der Präparation des Fatty Triangles aufgezeigt werden. Das Modell kann zukünftig genutzt werden, um die Netzimplantation und die Präparation des Fatty Triangles zu üben als auch um die chirurgischen Leistungen zu evaluieren.
Einleitung:
Volumensubstitution ist ein elementarer Bestandteil bei der hämodynamischen Stabilisierung von Patienten bei großen operativen Eingriffen. Die Art und Menge der zu applizierenden Lösungen ist Gegenstand von kontrovers geführten Diskussionen. Ziel dieser Arbeit war es, die unterschiedlichen Auswirkungen der intraoperativ verabreichten kolloidalen Infusionslösungen 6% HES 130/0,4 (HES) und Humanalbumin 5% (HA) zu untersuchen.
Material & Methoden:
Retrospektive Datenanalyse (2000-2007) von neurochirurgisch versorgten Kindern <5 Jahren mit einem intraoperativen Blutverlust von > 10%, die entweder HES oder HA zur Volumensubstitution verabreicht bekommen hatten.
Ergebnisse:
913 Patienten wurden zwischen 2000-2007 operiert, wovon 30 mit HES (Alter 26,8±17,6 [Monate]; Gewicht 12,3±4,0 [kg]) und 56 mit HA behandelte Kinder (Alter 20,0±17,9; Gewicht 11,0±3,9) eingeschlossen werden konnten. Der Blutverlust in der HA-Gruppe (54,8±45,0 [% Blutverlust vom Gesamtblutvolumen]) war signifikant höher als in der HES-Gruppe (30,5±30,0). In der HA-Gruppe wurden signifikant mehr Kolloide (25,7±11,3 HA vs. 16,4±9,2 HES [ml/kgKG]) im Vergleich zu den mit HES behandelten Kindern verabreicht. Sowohl intraoperativ, als auch nach dem Intensivstation-Aufenthalt hatte die HA-Gruppe eine signifikant höhere positive Bilanz gegenüber der HES-Gruppe. Die Urinproduktion unterschied sich sowohl intra-, als auch postoperativ nicht zwischen den Gruppen. Bei den HA-Kindern war postoperativ die partielle Thromboplastinzeit (PTT) signifikant erhöht und das Fibrinogen erniedrigt. Interessanterweise zeigten aber die mit HA behandelten Kinder tendenziell einen kürzeren Intensivaufenthalt (40,2±25,1 HES vs. 31,6±24,3 HA [h]) und eine verkürzte postoperative Krankenhausverweildauer (HES 11,0±3,8; HA 9,8±4,6 [d]).
Schlussfolgerung:
Trotz größerem Blutverlust, höherem Bedarf an Kolloiden und erhöhter PTT zeigten die mit HA behandelten Kinder sowohl einen Trend zu einem kürzeren Intensivaufenthalt, als auch zu einer verkürzten postoperativen Krankenhausverweildauer. Für eine valide Aussage PRO HES oder HA zur Volumentherapie in der pädiatrischen Neurochirurgie, müsste eine doppelblinde randomisierte Studie durchgeführt werden.
Salt formation is a routinely used strategy for poorly water-soluble drugs and traditionally performed with small inorganic counterions. High energy crystal lattices as well as effects on the local pH within the aqueous boundary layer during dissolution drive the increased dissolution rate and apparent solubility. Ionic liquids however, by definition low melting ionic salts with often large organic counterions, combine an increased dissolution rate with solubilization of the drug by the counterion itself. Long lasting supersaturation profiles of increased kinetic solubility were reported for several drugs formulated as ionic liquids increasing their overall bioavailability. Furthermore, aggregation and micellization between highly lipophilic compounds and amphiphilic bile acids was described before, demonstrating the capabilities of the human body itself to utilize solubilization of poorly water-soluble compounds. Development of novel counterions not only tailoring the desired physicochemical properties e.g. dissolution rate of the parent drug but adding – in a best-case scenario synergistic – pharmacological activity has been driven forward in the last years. However, salt formation can only be applied for ionizable i.e. acidic or basic compounds. While co-crystals can be used as a nonionized alternative, their formation is not always successful leading to an urgent need for other formulation strategies. In these lines, development of 2D and 3D printing techniques has been ongoing for the last decades and their pharmaceutical application has been demonstrated. The versatile nature and commercial availability allow a decentralized production further elaborating this technique for a highly flexible and patient-oriented supply with medication.
This thesis focuses on the theoretical background and potential application of salt formation in the pharmaceutical development of a drug candidate. The first section presents the current knowledge and state of the art in preparation of low melting ionic liquids i.e. salts and is translated to the in vitro investigation of molecular interaction between the poorly water-soluble drug imatinib and components of the human intestinal fluid in the second section. Development of novel antibiotic counterions and assessment of their potential use in pharmaceutical formulations with fluoroquinolones is described in the last two sections.
Chapter I describes the application of low melting ionic liquids in pharmaceutical formulation and details their development in the last two decades from versatile organic solvents in chemical synthesis towards amorphous strategies for drug delivery. The chapter gives a general overview on molecular structure and physicochemical properties of several drug containing ionic liquids and details the mechanisms which attribute to a typically fast dissolution, increased aqueous solubility as well as enhanced permeation which was reported in several publications.
Chapter II translates the increased aqueous solubility of drugs by an organic counterion to the human gastrointestinal tract with taurocholate and lecithin as main drivers for the solubilization of highly lipophilic and poorly water-soluble drugs. Investigation of the interaction of imatinib – a poorly water-soluble weak base – with fasted- and fed state simulated intestinal fluids revealed a complex interplay between the components of the intestinal fluid and the drug. Mixed vesicles and micelles were observed in concentration dependent aggregation assays and revealed differences in their size, molecular arrangement as well as composition, depending on the tested drug concentration. Overall, the study outlines the effective interaction of weakly basic drugs with taurocholate and lecithin to minimize recrystallization during intestine passage finally leading to favorable supersaturation profiles.
Chapter III focuses on the development of novel antibiotic counterions which potentially move the evolution of ionic liquids from a pharmaceutical salt with tailored physicochemical properties to a synergistic combination of two active pharmaceutical ingredients. The natural occurring anacardic acid derived from the cashew nut shell inspired a series of antibacterial active acidic compounds with increasing alkyl chain length. Their physicochemical properties, antibacterial activity, bacterial biofilm inhibition and cytotoxicity were detailed and in vivo activity in a Galleria mellonella model was assessed. This group of anacardic acid derivatives is synthetically accessible, easily modifiable and yielded two compounds with favorable activity and physicochemical profile for further drug development.
Chapter IV outlines the potential application of anacardic acid derivatives in pharmaceutical formulations by salt formation with fluoroquinolone antibiotics as well as novel techniques such as 2D/3D printing for preparation of drug imprinted products. Despite anacardic acid derivatives demonstrated promising physicochemical properties, salt formation with fluoroquinolone antibiotics was not feasible. However, 2D/3D printed samples with anacardic acid derivative alone or in combination with ciprofloxacin demonstrated physical compatibility between drug and matrix as well as antibacterial activity against three S. aureus strains in an agar diffusion assay. Conclusively, drug printing can be applied for the herein tested compounds, but further process development is necessary.
In summary, preparation of low melting ionic liquids, salts or co-crystals is an appropriate strategy to increase the aqueous solubility of poorly water-soluble drugs and tailor physicochemical properties. The counterion itself solubilizes the drug and furthermore potentially interferes with the complex micellar environment in the human intestine. However, salt formation as routinely used formulation strategy is not feasible in every case and development of alternative techniques is crucial to hurdle challenges related to unfavorable physicochemical properties. The outlined techniques for 2D/3D drug printing provide versatile production of drug products while extending the design space for novel drug development.
Das Nebenschilddrüsenkarzinom ist eine sehr seltene Erkrankung mit einem hohen krankheitsspezifischen Überleben.
Die vorliegende Studie ist mit 83 Patienten die bisher größte multizentrische Studie zu dieser seltenen Tumorerkrankung in Deutschland.
Aufgrund der Seltenheit der Erkrankung finden sich bislang keine prospektiven Studien zum Nebenschilddrüsenkarzinom, sodass sich die bisherigen Handlungs- und Therapieempfehlungen auf retrospektive Studien, Reviews sowie auf Erfahrungen aus Einzelfallberichten beziehen.
Ziel dieser Arbeit war die Evaluation von diagnostischen Mitteln, Krankheitsverlauf und prognostischen Faktoren sowie die Gegenüberstellung der Erfolgsaussichten verschiedener Therapieverfahren des Nebenschilddrüsenkarzinoms.
Veränderung der Ranvier’schen Schnürringarchitektur bei Patienten mit diabetischer Neuropathie

In der Krankheitsentstehung der diabetischen Neuropathie wird die paranodale Demyelinisierung als ein möglicher Pathomechanismus diskutiert, wobei Studien mit Gewebeproben von Patienten aufgrund der Invasivität limitiert sind. In der vorliegenden Studie wurden periphere Nervenfasern in Hautbiopsien von Patienten mit diabetischer Neuropathie und in Patienten mit Diabetes mellitus ohne Neuropathie untersucht. Ziel war es, nodale und paranodale Veränderungen, wie eine Dispersion der paranodalen Proteine Caspr und Neurofascin oder der nodalen Na-Kanäle, zu detektieren und die Proben auf verlängerte Ranvier`sche Schnürringe zu untersuchen.
Es wurde die Hypothese überprüft, dass paranodale Demyelinisierungen bei Patienten mit diabetischer Neuropathie in Hautbiopsien, als minimal-invasive Methode, nachweisbar sind. Hautproben von Patienten mit Diabetes mellitus ohne Neuropathie sollten zudem in einem frühen Krankheitsstadium untersucht werden.
Für die Untersuchung konnten 35 Patienten mit einer diabetischen Neuropathie, 17 Patienten mit Diabetes mellitus und 31 Kontrollen eingeschlossen werden. Immunfluoreszenzfärbungen mit Antikörpern gegen Caspr, Neurofascin und Natrium-Kanälen wurden zur Analyse der Ranvier`schen Schnürringarchitektur durchgeführt und ausgewertet.
Eine erhöhte Anzahl an verlängerten Schnürringen, als Zeichen einer segmentalen Demyelinisierung, konnte in den Patienten mit diabetischer Neuropathie aber auch in Patienten mit Diabetes mellitus nachgewiesen werden. Weiterhin waren vermehrt Veränderungen der paranodalen Proteine, wie eine Dispersion von Caspr und Neurofascin in den Proben des Fingers der Patienten mit diabetischer Neuropathie sowie eine Dispersion von Neurofascin im Unterschenkel in beiden Patientengruppen nachweisbar. Interessanterweise waren einzelne Veränderungen auch in den gesunden Kontrollen auffindbar.
Veränderungen der Schnürringarchitektur lassen sich mithilfe der Hautbiopsie nachweisen und quantifizieren. Nodale und paranodale Veränderungen weisen auf demyelinisierende Prozesse in Patienten mit diabetischer Neuropathie hin und finden sich auch bereits in einem frühen Krankheitsstadium.
Fluids in Gravitational Fields – Well-Balanced Modifications for Astrophysical Finite-Volume Codes

Stellar structure can -- in good approximation -- be described as a hydrostatic state, which which arises due to a balance between gravitational force and pressure gradient. Hydrostatic states are static solutions of the full compressible Euler system with gravitational source term, which can be used to model the stellar interior. In order to carry out simulations of dynamical processes occurring in stars, it is vital for the numerical method to accurately maintain the hydrostatic state over a long time period. In this thesis we present different methods to modify astrophysical finite volume codes in order to make them \emph{well-balanced}, preventing them from introducing significant discretization errors close to hydrostatic states. Our well-balanced modifications are constructed so that they can meet the requirements for methods applied in the astrophysical context: They can well-balance arbitrary hydrostatic states with any equation of state that is applied to model thermodynamical relations and they are simple to implement in existing astrophysical finite volume codes. One of our well-balanced modifications follows given solutions exactly and can be applied on any grid geometry. The other methods we introduce, which do no require any a priori knowledge, balance local high order approximations of arbitrary hydrostatic states on a Cartesian grid. All of our modifications allow for high order accuracy of the method. The improved accuracy close to hydrostatic states is verified in various numerical experiments.
Stomata sind kleine Poren in der Blattoberfläche, die Pflanzen eine Anpassung ihres Wasserhaushalts an sich ändernde Umweltbedingungen ermöglichen. Die Öffnungsweite der Stomata wird durch den Turgordruck der Schließzellen bestimmt, der wiederum durch Ionenflüsse über die Membranen der Zelle reguliert wird. Ein Netzwerk von Signaltransduktionswegen sorgt dafür, dass Pflanzen die Stomabewegungen an die Umgebungsbedingungen anpassen können. Viele molekulare Komponenten dieser Signaltransduktionketten in Schließzellen von Angiospermen sind inzwischen bekannt und Calcium spielt darin als Signalmolekül eine wichtige Rolle. Weitgehend unbekannt sind dagegen die Mechanismen, die zur Erzeugung von transienten Erhöhungen der Calciumkonzentration führen. Auch die molekularen Grundlagen der Regulierung der Stomaweite in Nicht-Angiospermen-Arten sind bisher nur wenig verstanden. Um zur Aufklärung dieser Fragestellungen
beizutragen, wurden in dieser Arbeit Mechanismen zur Erhöhungen der cytosolischen Calciumkonzentration sowie elektrophysiologische Eigenschaften von Schließzellen untersucht. Der Fokus lag hierbei insbesondere auf der Visualisierung cytosolischer Calciumsignale in Schließzellen. Im ersten Teil der Arbeit wurde durch die Applikation hyperpolarisierender Spannungspulse mittels TEVC (Two Electrode Voltage Clamp) gezielt eine Erhöhung der cytosolischen Calciumkonzentration in einzelnen Schließzellen von Nicotiana tabacum ausgelöst. Um die Dynamik der cytosolischen Calciumkonzentration dabei zeitlich und räumlich hoch aufgelöst zu visualisieren, wurde simultan zu den elektrophysiologischen Messungen ein
Spinning-Disc-System für konfokale Aufnahmen eingesetzt. Während der Applikation
hyperpolarisierender Spannungspulse wurde eine transiente Vergrößerung des cytosolischen Volumens beobachtet. Diese lässt sich durch einen osmotisch getriebenen Wasserfluss erklären, der durch die Veränderung der Ionenkonzentration im Cytosol verursacht wird. Diese wiederum wird durch die spannungsabhängige Aktivierung einwärtsgleichrichtender Kaliumkanäle in der Plasmamembran der Schließzellen und durch den Kompensationsstrom der eingestochenen Mikroelektrode hervorgerufen. Mit Hilfe des calciumsensitiven Farbstoffs Fura-2 konnte gezeigt werden, dass die Erhöhung der freien cytosolischen Calciumkonzentration während der Applikation hyperpolarisierender Spannungspulse durch zwei Mechanismen verursacht wird. Der erste Mechanismus ist die Aktivierung hyperpolarisationsaktivierter, calciumpermeabler Kanäle (HACCs) in der Plasmamembran, die schon 1998 von Grabov & Blatt beschrieben wurde. Zusätzlich zu diesem Mechanismus der Calciumfreisetzung, konnte ein zweiter bislang unbekannter Mechanismus aufgedeckt werden, bei dem Calcium aus intrazellulären Speichern in das Cytosol freigesetzt wird. Dieser Mechanismus hängt mit der oben beschriebenen Vergrößerung des cytosolischen Volumens zusammen und ist wahrscheinlich durch die Änderungen der mechanischen Spannung der Membran bzw. der Osmolarität innerhalb der Zelle bedingt. Diese könnten zu einer Aktivierung mechanosensitiver, calciumpermeabler Kanäle führen.
Der zweite Teil der Arbeit beschäftigt sich mit den molekularen Grundlagen der Regulierung von Stomata in Nicht-Angiospermen. In Schließzellen von Polypodium vulgare konnten durch die Anwendung der TEVC-Technik ähnliche spannungsabhängige Ströme über die Plasmamembran gemessen werden wie in Angiospermen. Ebenso wurden durch die Applikation hyperpolarisierender Spannungspulse an Schließzellen von Polypodium und Asplenium Erhöhungen der cytosolischen Calciumkonzentration ausgelöst, die auf die Existenz spannungsabhängiger, calciumpermeabler Kanäle in der Plasmamembran
hinweisen. Die Diffusion von Fluoreszenzfarbstoffen in die Nachbarschließzellen nach der iontophoretischen Beladung in Polypodium, Asplenium, Ceratopteris und Selaginella zeigte, dass in diesen Arten eine symplastische Verbindung zwischen benachbarten Schließzellen besteht, die an Schließzellen von Angiospermen bisher nicht beobachtet werden konnte. Anhand elektronenmikroskopischer Aufnahmen von Polypodium glycyrrhiza Schließzellen konnte gezeigt werden, dass diese Verbindung wahrscheinlich durch Plasmodesmata zwischen benachbarten Schließzellen gebildet wird. Durch die Analyse der Calciumdynamik in benachbarten Schließzellen nach hyperpolarisierenden Spannungspulsen stellte sich heraus, dass die Calciumhomöostase trotz symplastischer Verbindung in beiden Schließzellen unabhängig voneinander reguliert zu werden scheint. Im Rahmen der Untersuchungen an Farnschließzellen wurde desweiteren eine Methode zur Applikation von ABA etabliert, die es erlaubt mithilfe von Mikroelektroden das Phytohormon iontophoretisch in den Apoplasten zu laden. Im Gegensatz zu den Schließzellen von Nicotiana tabacum, die auf eine so durchgeführte ABA-Applikation mit dem Stomaschluss reagierten, wurde in Polypodium vulgare auf diese Weise kein Stomaschluss ausgelöst. Da die ABA-Antwort der Farnstomata aber auch von anderen Faktoren wie Wachstumsbedingungen abhängig ist (Hõrak et al., 2017), kann eine ABA-Responsivität in dieser Farnart trotzdem nicht vollkommen ausgeschlossen werden.
Die Freisetzung von Calcium aus intrazellulären Speichern, wie sie in dieser Arbeit gezeigt wurde, könnte eine wichtige Rolle bei der Regulierung der Stomaweite spielen. Zur Aufklärung dieser Fragestellung wäre die Identifizierung der Kanäle, die an der osmotisch/mechanisch induzierten Calciumfreisetzung aus internen Speichern beteiligt sind, von großem Interesse. Weiterführende Studien an Schließzellen von Farnen könnten die physiologische Bedeutung der aus Angiospermen bekannten Ionenkanäle für die Stomabewegungen in evolutionär älteren Landpflanzen aufklären und so maßgeblich zum Verständnis der Evolution der Regulierunsgmechanismen von Stomata beitragen. Außerdem stellt sich die Frage, welche Rolle die hier gezeigte symplastische Verbindung der Nachbarschließzellen durch Plasmodesmata für die Funktion der Stomata spielt.
Bei den primär herzgesunden Tieren wurde durch Frequenz-Überstimulation mit Hilfe eines implantierten biventrikulären Herzschrittmachers eine chronische Herzinsuffizienz induziert. Im Rahmen der Verlaufsbeobachtungen wurde in-vivo die Druckanstiegsgeschwindigkeit dP/dtmax, der enddiastolische sowie endsystolische Druck durch einen implantierten Drucksensor gemessen. Anhand der gemessen dP/dtmax-, EDP- und ESP-Werte konnte der Bowditcheffekt dargestellt werden. Mit Ausprägung einer chronischen Herzinsuffizienz fiel dieser im Verlauf deutlich geringer aus, blieb aber stets nachweisbar.
Hintergrund:
Die zeitgerechte Integration des Entlassmanagements ist ein wesentlicher Bestandteil des umfassenden Therapiekonzepts auf Palliativstation. Speziell zur Entlassung in ein stationäres Hospiz sollte die verbleibende Überlebenszeit gegen den Benefit eines stressbehafteten Versorgungswechsels diskutiert werden.
Ziel der Studie:
Aus der Vielzahl der vorhandenen und international validierten palliativmedizinischen Prognosescores wurden für diese Studie die Palliative Performance Scale (PPS) und der Palliative Prognostic Index (PPI) ausgewählt. Ziel war erstens die Überprüfung ihrer Anwendbarkeit auf eine deutsche Palliativpopulation. Zweitens wurden sie neben Symptomen der Terminalphase auf ihre Fähigkeit zur Kurzzeitprognose getestet, um Patienten mit kurzer Überlebenszeit in der stationären Hospizversorgung nach Entlassung identifizieren zu können.
Methodik:
Am Zentrum für Palliativmedizin des Universitätsklinikums Würzburg wurden retrospektiv PPS, PPI, ausgewählte Symptome der Sterbephase sowie die Überlebensdauer bei 112 Patienten erhoben, die von 2012 bis 2016 in ein Hospiz entlassen worden waren. Mittels ANOVA und Kaplan-Meier-Statistik wurden Überlebensdauer und Höhe der Prognosescores in Beziehung gesetzt und Risikogruppen gebildet. Zur Identifizierung von Risikopatienten mit einer Hospizverweildauer ≤ 7 Tagen wurden diese mit der Gruppe der Langverweiler (> 7 Tage) hinsichtlich Höhe der PPS, des PPI und das Vorhandensein von Terminalsymptomen verglichen.
Ergebnisse:
Mittels ANOVA und Kaplan-Meier-Kurven konnte die signifikante Korrelation zwischen Höhe des Prognosescores und der Überlebenszeit für die untersuchte Kohorte belegt werden. Risikopatienten mit einer Hospizverweildauer ≤ 7 Tagen wiesen einen signifikant niedrigeren PPS (40 % vs. 50 %) respektive einen höheren PPI-Wert (6,5 vs. 4,5 P.) als die Langverweiler auf. Die Terminalsymptome Dysphagie und eine reduzierte orale Nahrungsaufnahme waren unter Risikopatienten häufiger vertreten.
Schlussfolgerung:
Die Prognosefähigkeit der palliativmedizinischen Prognosescores PPS und PPI konnte für die untersuchte Kohorte belegt werden. Eine Kurzzeitprognose erwies sich allerdings aufgrund der geringen Trennschärfe der Cut-Off-Werte als praxisuntauglich. Sie können dennoch in speziellen Fällen als Orientierungshilfe im Entlassmanagement dienen.
In recent years <mi>PI</mi>-conjugated organoboron polymers and BN-doped polycyclic aromatic hydrocarbons have attracted a lot of interest due to their great potential in organic electronics. However, there are only few known examples of conjugated polymers with BN units in their main chain. Within this work silazane cleavage with silicon-boron (Si/B) exchange for the synthesis of a novel class of inorganic-organic hybrid polymers is demonstrated. These polymers consist of alternating NBN and para-phenylene units in the main chain. Photophysical studies and TD-DFT calculations for the polymer and molecular model systems were carried out, revealing a low extent of <mi>PI</mi>-conjugation across the NBN units. The new polymers can be used as macromolecular polyligands by a cross-linking reaction with a ZrIV compound. In the next chapter the synthesis and characterization of the first poly(p-phenylene iminoborane) is presented. This novel inorganic–organic hybrid polymer can be described as a BN analogue of the well-known poly(p-phenylene vinylene) (PPV) and is also accessible using the previously described Si/B exchange as synthetic strategy. Photophysical investigations and TD-DFT calculations on the polymer and corresponding model oligomers provide clear evidence for <mi>PI</mi>-conjugation across the B=N units and extension of the conjugation path with increasing chain length. Furthermore, a possible application of Si/B exchange for the synthesis of polysulfoximines was explored. Herein, diaryl sulfoximines and a p-phenylene bisborane serve as building blocks for new BN- and BO-doped alternating inorganic–organic hybrid copolymers. While the BN-linked polymers were accessible by a facile silicon/boron exchange protocol, the synthesis of polymers with B–O linkages in the main chain is achieved by salt elimination. In the last chapter the concept of Si/B exchange was investigated for the synthesis of BP-linked oligomers. Herein oligomers with sterically less demanding substituents (substituents: 2,4,6-trimethylphenyl or 2,4,6-tri-iso-propylphenyl) at the phosphorus are accessible using Si/B exchange, but the oligomer with Mes* (2,4,6-tri-tert-butylphenyl) as substituent needed a salt elimination pathway to give the desired product. Experimental data and theoretical investigations indicate, that the P-substituent has a high influence on the geometry of the phosphorus center and therefore on the possible conjugation over the BP units.
Das Ziel der vorliegenden Arbeit war die retrospektive Analyse von Patienten mit einer mykobakteriellen Hautinfektion, die sich an der Klinik und Poliklinik für Dermatologie, Venerologie und Allergologie des Universitätsklinikums Würzburg oder in der Fachabteilung für Tropenmedizin der Missioklinik Würzburg in dem Zeitraum zwischen 1998 und 2018 vorgestellt haben. Es wurden verschiedene Aspekte wie die Anamnese, der mutmaßliche Infektionsweg, klinische und diagnostische Daten sowie die durchgeführte Therapie untersucht. Es konnten insgesamt 32 Fälle erfasst werden, die in 4 erregerspezifische Gruppen unterteilt wurden. Die Gruppe der Mycobacterium (M.) marinum-Infektionen war mit 18 Fällen am größten, gefolgt von 9 Infektionen mit weiteren nichttuberkulösen Mykobakterien (NTM; M. chelonae, M. avium, M. fortuitum, M. wolinskyi/ mageritense sowie M. haemophilum), 3 Fällen mit dem Erreger M. leprae sowie 2 kutanen M. tuberculosis-Infektionen. Der Anteil immunsupprimierter Patienten mit einer M. marinum-Infektion lag bei 22 %. In unserer Untersuchung stellte der Umgang mit Zierfischaquarien den größten Risikofaktor für eine Infektion mit M. marinum dar. Wie unsere Studie weiterhin zeigte, sollte auch bei knotigen Hautveränderungen in für kutane Mykobakteriosen untypischen Körperarealen, wie z.B. dem Gesicht, eine solche Infektion differentialdiagnostisch erwogen werden. Der kulturelle Nachweis misslang lediglich bei 2 Patienten mit einer M. marinum-Infektion. Die Diagnose konnte in diesen beiden Fällen anhand einer charakteristischen Anamnese, Klinik und Histopathologie gestellt werden. In der vorliegenden Untersuchung erfolgte die Therapie bei allen Patienten mittels einer Antibiose. In der M. marinum-Gruppe erwies sich insbesondere der Wirkstoff Clarithromycin als effektiv. Unter der Gabe von Doxycyclin kam es dagegen in 80 % der Fälle zu einer Therapieumstellung. In beiden Gruppen nichttuberkulöser Mykobakteriosen konnten die Monotherapien im Durchschnitt früher erfolgreich beendet werden als die Kombinationstherapien. Letztere kamen allerdings zum Einsatz bei einer zugrundeliegenden Immunsuppression bzw. bei einem ausgedehnten Hautbefund.
Bacterial small non-coding RNAs (sRNAs) play fundamental roles in controlling and finetuning gene expression in a wide variety of cellular processes, including stress responses, environmental signaling and virulence in pathogens. Despite the identification of hundreds of sRNA candidates in diverse bacteria by genomics approaches, the mechanisms and regulatory capabilities of these posttranscriptional regulators have most intensively been studied in Gram-negative Gammaproteobacteria such as Escherichia coli and Salmonella. So far, almost nothing is known about sRNA-mediated regulation (riboregulation) in Epsilonproteobacteria, including the major human pathogen Helicobacter pylori. H. pylori was even thought to be deficient for riboregulation as none of the sRNAs known from enterobacteria are conserved in Helicobacter and since it lacks the major RNA chaperone Hfq, which is crucial for sRNA function as well as stability in many bacteria. Nonetheless, more than 60 cis- and trans-acting sRNA candidates were recently identified in H. pylori by a global RNA sequencing approach, indicating that this pathogen, in principle, has the capability to use riboregulation for its gene expression control. However, the functions and underlying mechanisms of H. pylori sRNAs remained unclear.
This thesis focused on the first functional characterization and target gene identification of a trans-acting sRNA, RepG (Regulator of polymeric G-repeats), in H. pylori. Using in-vitro and in-vivo approaches, RepG was shown to directly base-pair with its C/Urich terminator loop to a variable homopolymeric G-repeat in the 5’ untranslated region (UTR) of the tlpB mRNA, thereby regulating expression of the chemotaxis receptor TlpB. While the RepG sRNA is highly conserved, the length of the G-repeat in the tlpB mRNA leader varies among different H. pylori isolates, resulting in a strain-specific tlpB regulation. The modification of the number of guanines within the G-stretch in H. pylori strain 26695 demonstrated that the length of the homopolymeric G-repeat determines the outcome of posttranscriptional control (repression or activation) of tlpB by RepG. This lengthdependent targeting of a simple sequence repeat by a trans-acting sRNA represents a new twist in sRNA-mediated regulation and a novel mechanism of gene expression control, since it uniquely links phase variation by simple sequence repeats to posttranscriptional regulation.
In almost all sequenced H. pylori strains, tlpB is encoded in a two gene operon upstream of HP0102, a gene of previously unknown function. This study provided evidence that HP0102 encodes a glycosyltransferase involved in LPS O-chain and Lewis x antigen production. Accordingly, this glycosyltransferase was shown to be essential for mice colonization by H. pylori. The coordinated posttranscriptional regulation of the tlpB-HP0102 operon by antisense base-pairing of RepG to the phase-variable G-repeat in the 5’ UTR of the tlpB mRNA allows for a gradual, rather than ON/OFF, control of HP0102 expression, thereby affecting LPS biosynthesis in H. pylori. This fine-tuning of O-chain and Lewis x antigen expression modulates H. pylori antibiotics sensitivity and thus, might be advantageous for Helicobacter colonization and persistence.
Whole transcriptome analysis based on microarray and RNA sequencing was used to identify additional RepG target mRNAs and uncover the physiological role of this riboregulator in H. pylori. Altogether, repG deletion affected expression of more than 40 target gene candidates involved various cellular processes, including membrane transport and adhesion, LPS modification, amino acid metabolism, oxidative and nitrosative stress, and nucleic acid modification. The presence of homopolymeric G-repeats/G-rich sequences in almost all target mRNA candidates indicated that RepG hijacks a conserved motif to
recognize and regulate multiple target mRNAs in H. pylori.
Overall, this study demonstrates that H. pylori employs riboregulation in stress response and virulence control. In addition, this thesis has successfully established Helicobacter as a new model organism for investigating general concepts of gene expression control by Hfq-independent sRNAs and sRNAs in bacterial pathogens.
