Cardiovascular diseases represent the leading cause of death worldwide, with myocardial infarction and strokes being the most common complications. In both cases, the appearance of an enlarged artery wall as a consequence of a growing plaque is responsible for the disturbance of the blood flow. The formation of plaques is driven by a chronic inflammatory condition known as atherosclerosis, characterized by an initial step of endothelial cell (EC) dysfunction followed by the recruitment of circulating immune cells into the tunica intima of the vessel. Accumulation of lipids and cells lead to the formation of atheromatous plaques that will define the cardiovascular outcome of an individual.
The role of the immune system in the progression of atherosclerosis has been widely recognized. By far, macrophages constitute the most abundant cell type in lesions and are known to be the major source of the lipid-laden foam cell pool during the course of the disease. However, other immune cells types, including T cells, dendritic cells (DCs) or mast cells, among others, have been described to be present in human and mouse plaques. How these populations can modulate the atherogenic process is dependent on their specialized function.
DCs constitute a unique population with the ability to bridge innate and adaptive immune responses, mainly by their strong capacity to present antigens bound to a major histocompatibility complex (MHC) molecule. Given their ability to polarize T cells and secrete cytokines, their role in atherosclerosis has gained attention for the development of new therapeutic approaches that could impact lesion growth. Hence, knowing the effect of a specific subset is an initial key step to evaluate its potential for clinical purposes. For example, the basic leucine zipper ATF-like 3 transcription factor (Batf3) controls the development of conventional dendritic cells type 1 (cDCs1), characterized by the expression of the surface markers CD8 and CD103. Initially, they were described to promote both T-helper 1 (Th1) and regulatory T cell (Treg) responses, known to accelerate and to protect against atherosclerosis, respectively. The first part of this thesis aimed to elucidate the potential role of Batf3-dependent DCs in atherosclerosis and concluded that even though systemic immune responses were mildly altered they do not modify the course of the disease and may not represent an attractive candidate for clinical studies.
DCs also have the ability to impact lesion growth through the release of a broad range of cytokines, which can either directly impact atherosclerotic plaques by modulating resident cells, or by further polarizing T cell responses. Among others, interleukin (IL) 23, a member of the IL-12 family of cytokines, has received much attention during the past year due to its connection to autoimmunity.
IL-23 is known to induce pathogenicity of Th17 cells and is responsible for the development of several autoimmune diseases including multiple sclerosis, psoriasis or rheumatoid arthritis. Interestingly, these patients often present with an accelerated course of atherosclerosis and thus, are at higher risk of developing cardiovascular events. Several epidemiological studies have pointed toward a possible connection between IL-23 and its receptor IL-23R in atherosclerosis, although their exact contribution remains to be elucidated. The second part of this thesis showed that resident antigen-presenting cells (APCs) in the aorta produced IL-23 during the steady state but this secretion was greatly enhanced after incubation with oxidized low-density lipoprotein (oxLDL). Furthermore, disruption of the IL-23R signaling led to decreased relative necrotic plaque area in lesions of Ldlr-/-Il23r-/- mice fed a high-fat diet (HFD) for 6 and 12 weeks compared to Ldlr-/- controls. A proposed mechanism involves that increased IL-23 production in the context of atherosclerosis may promote the pathogenicity of IL-23-responding T cells, especially IL-23R+ γδ T cells in the aortic root. Response to IL-23 might increase the release of granulocyte-macrophage colony-stimulating factor (GM-CSF) and IL-17 and alter the pro- and anti-inflammatory balance of cytokines in the aortic root. Altogether, these data showed that the IL-23 / IL-23R axis play a role in plaque stability.
Lagrange Multiplier Methods for Constrained Optimization and Variational Problems in Banach Spaces

This thesis is concerned with a class of general-purpose algorithms for constrained minimization problems, variational inequalities, and quasi-variational inequalities in Banach spaces.
A substantial amount of background material from Banach space theory, convex analysis, variational analysis, and optimization theory is presented, including some results which are refinements of those existing in the literature. This basis is used to formulate an augmented Lagrangian algorithm with multiplier safeguarding for the solution of constrained optimization problems in Banach spaces. The method is analyzed in terms of local and global convergence, and many popular problem classes such as nonlinear programming, semidefinite programming, and function space optimization are shown to be included as special cases of the general setting.
The algorithmic framework is then extended to variational and quasi-variational inequalities, which include, by extension, Nash and generalized Nash equilibrium problems. For these problem classes, the convergence is analyzed in detail. The thesis then presents a rich collection of application examples for all problem classes, including implementation details and numerical results.
For the differentiation of a embryonic stem cells (ESCs) to neuronal cells (NCs) a complex and coordinated gene regulation program is needed. One important control element for neuronal differentiation is the repressor element 1 silencing transcription factor (REST) complex, which represses neuronal gene expression in non-neuronal cells. Crucial effector proteins of the REST complex are small phosphatases such as the CTDSPs (C-terminal domain small phosphatases) that regulate polymerase II activity by dephosphorylating the C-terminal domain of the polymerase, thereby repressing target genes. The stepwise inactivation of REST, including the CTDSPs, leads to the induction of a neuron-specific gene program, which ultimately induces the formation of neurons. The spatio-temporal control of REST and its effector components is therefore a crucial step for neurogenesis.
In zebrafish it was shown that the REST-associated CTDSP2 is negatively regulated by the micro RNA (miR) -26b. Interestingly, the miR-26b is encoded in an intron of the primary transcript of CTDSP2. This gives the fundament of an intrinsic regulatory negative feedback loop, which is essential for the proceeding of neurogenesis. This feedback loop is active during neurogenesis, but inactive in non-neuronal cells. The reason for this is that the maturation of the precursor miR (pre-miR) to the mature miR-26 is arrested in non neuronal cells, but not in neurons. As only mature miRs are actively repressing genes, the regulation of miR-26 processing is an essential step in neurogenesis.
In this study, the molecular basis of miR-26 processing regulation in the context of neurogenesis was addressed. The mature miR is processed from two larger precursors: First the primary transcript is cleaved by the enzyme DROSHA in the nucleus to form the pre-miR. The pre-miR is exported from the nucleus and processed further through the enzyme DICER to yield the mature miR. The mature miR can regulate gene expression in association with the RNA-induced silencing complex (RISC).
Multiple different scenarios in which miR processing was regulated were proposed and experimentally tested. Microinjection studies using Xenopus leavis oocytes showed that slowdown or blockage of the nucleo-cytoplasmic transport are not the reason for delayed pre-miR-26 processing. Moreover, in vitro and in vivo miR-processing assays showed that maturation is most likely regulated through a in trans acting factor, which blocks processing in non neuronal cells.
Through RNA affinity chromatographic assays using zebrafish and murine lysates I was able to isolate and identify proteins that interact specifically with pre-miR-26 and could by this influence its biogenesis. Potential candidates are FMRP/FXR1/2, ZNF346 and Eral1, whose functional characterisation in the context of miR-biogenesis could now be addressed.
The second part of my thesis was executed in close colaboration with the laboratory of Prof. Albrecht Müller. The principal question was addressed how miR-26 influences neuronal gene expression and which genes are primarily affected. This research question could be addressed by using a cell culture model system, which mimics ex vivo the differentiation of ESCs to NCs via neuronal progenitor.
For the functional analysis of miR-26 knock out cell lines were generated by the CRISPR/Cas9 technology. miR-26 deficient ESC keep their pluripotent state and are able to develop NPC, but show major impairment in differentiating to NCs. Through RNA deep sequencing the miR-26 induced transcriptome differences could be analysed.
On the level of mRNAs it could be shown, that the expression of neuronal gene is downregulated in miR-26 deficient NCs. Interestingly, the deletion of miR-26 leads to selectively decreased levels of miRs, which on one hand regulate the REST complex and on the other hand are under transcriptional control by REST themself. This data and the discovery that induction of miR-26 leads to enrichment of other REST regulating miRs indicates that miR-26 initiates neurogenesis through stepwise inactivation of the REST complex.
Bei der postoperativen Therapieplanung des Mammakarzinoms treten immer wieder Entscheidungsgrenzfälle auf, bei denen keine sicheren Argumente für oder gegen eine adjuvante Chemotherapie gefunden werden können. Bei 50 Hormonrezeptor-positiven, Her2/neu-negativen Mammakarzinomen ohne oder mit nur geringer nodaler Metastasierung (max. pT1a) wurde zusätzlich zu den konventionellen klinisch-pathologischen Risikofaktoren der OncotypeDX®-Multigentest veranlasst. In der Tumorkonferenz wurde bereits vor Eingang des Testergebnisses ein Votum für oder gegen eine Chemotherapie auf Basis konventioneller Parameter protokolliert; die definitive Therapieempfehlung erfolgte nach Vorliegen des Multigentest-Ergebnisses.
32 Mammakarzinome (64 %) zeigten einen niedrigen, 26 (32 %) einen mittleren und 3 (6 %) einen hohen Recurrence-Score (RS). In vielen Fällen konnte das OncotypeDX®-Ergebnis eine auf der Basis konventioneller Parameter getroffene Therapieentscheidung stützen. In fünf Fällen wurde eine zunächst favorisierte Entscheidung für eine adjuvante Therapie revidiert. In drei Fällen wurde eine zunächst nicht geplante Chemotherapie empfohlen. Allerdings führte in einigen Fällen auch eine niedrige oder intermediäre Risikokonstellation in der OncotypeDX®-Testung nicht dazu, von einer adjuvanten Chemotherapie abzuraten.
Insgesamt spricht das Ergebnis nicht dafür, einen Multigentest als Standardmethode einzusetzen. Vielmehr sollten zunächst die konventionellen, insbesondere die histopathologischen und immunhistochemischen Parameter mit großer Sorgfalt erhoben und analysiert werden. Im Zweifelsfall und nach Kosten-Nutzen-Abwägung kann ein Multigentest jedoch ein weiteres hilfreiches Argument für oder gegen eine bestimmte Therapieempfehlung liefern.
Das follikuläre Lymphom (FL) wird nach der aktuellen Klassifikation der WHO (World Health Organization Classification of Lymphoid Tumours) anhand der Zahl der Zentroblasten in drei Grade und der Grad 3 weiter in 3A und 3B eingeteilt. Bis heute ist die Rolle der FL3B aufgrund der morphologischen und genetischen Unterschiede zu den anderen FL umstritten, es wird eine eigene Entität und Pathogenese des FL3B diskutiert. Durch das Verbundprojekt „Molekulare Mechanismen in malignen Lymphomen“ (MMML) Daten zu FISH-, Genexpressionsanalysen und immunhistochemischen Färbungen bearbeitet werden.
Diesen Daten zufolge sind FL3B in ihrer Genexpression nicht von FL3A trennbar. Es konnte jedoch eine Abgrenzung der FL1/2 zu den FL3A/B durch die erhöhte Expression von 13 Genen in den FL3A/B gefunden werden, von denen Homolog, double strand break repair nuclease (MRE11A), Topoisomerase II alpha (TOP2A) und Thioredoxin (TXN) schon zuvor im Rahmen von FL und NHL diskutiert wurden.
This thesis discusses and proposes a solution for one problem arising from deformation quantization:
Having constructed the quantization of a classical system, one would like to understand its mathematical properties (of both the classical and quantum system). Especially if both systems are described by ∗-algebras over the field of complex numbers, this means to understand the properties of certain ∗-algebras:
What are their representations? What are the properties of these representations? How
can the states be described in these representations? How can the spectrum of the observables be
described?
In order to allow for a sufficiently general treatment of these questions, the concept of abstract O ∗-algebras is introduced. Roughly speaking, these are ∗ -algebras together with a cone of positive linear functionals on them (e.g. the continuous ones if one starts with a ∗-algebra that is endowed with a well-behaved topology). This language is then applied to two examples from deformation quantization, which will be studied in great detail.
Chemotherapeutika stellen nach wie vor eine der wichtigsten Behandlungs-optionen bei Krebs dar. Ihre akuten und chronischen Nebenwirkungen aber limitieren ihre Anwendung. Aktuelle klinische Studien deuten auf einen positiven Effekt von Kurzzeitfasten auf die Nebenwirkungen von Chemotherapeutika hin. Eine Erklärung hierfür könnte das unterschiedliche Ansprechen von normalen und malignen Zellen auf Chemotherapeutika in einer Mangelsituation sein, das als »differential stress resistance« (DSR) bezeichnet wird. Dieses Phänomen lässt nicht-maligne Zellen bei Restriktion von Glukose und Wachstumsfaktoren weniger sensitiv auf Chemotherapeutika reagieren als maligne Zellen. Das Ziel der vorliegenden Arbeit war zu untersuchen, ob ein Mangel an Glukose und Wachstumsfaktoren das Ansprechen nicht transformierten Zellen 5-FU abschwächen kann während das von kolorektalen Karzinomzellen (Colo741, LS174T, HCT116, HT29 und SW620) gleichbleibt.
Optimale Kulturbedingungen für Tumorzellen in vitro stellen 11 mmol/l Glukose und 10 % FCS dar, während 3 mmol/l Glukose und 1 % FCS Mangelbedingun¬gen repräsentieren. Glukosewerte von 3 mmol/l werden auch mit Kurzzeitfasten erreicht. Da der Großteil der soliden Tumoren mit Sauerstoff unterversorgt ist, wurden Untersuchungen auch bei tumorphysiologischen Sauerstoffbedingungen von u. a. 5 % durchgeführt. Der antiproliferative Effekt von 5-FU wurde als halbmaximale inhibitorische Konzentration (IC50) für eine Kultur¬dauer von 72 Stunden bestimmt.
Im Mangelmedium mit 3 mmol/l Glukose und 1 % FCS verstärkte sich der antiproliferative Effekt von 5-FU bei drei der fünf getesteten kolorektalen Karzinomzelllinien in Gegenwart von 5 % Sauerstoff im Vergleich zum Standard¬medium mit 11 mmol/l Glukose und 10 % FCS. Die Unterschiede in den IC50 Werten für 5-FU bei diesen drei Zelllinien (Colo741, HCT116, HT29) waren signifikant und bei den beiden anderen Zelllinien (LS174T, SW620) zeigten sich Tendenzen. Dagegen nahm bei Fibroblasten der antiproliferative Effekt von 5-FU im Mangelmedium ab, die Zellen waren somit besser vor dem Chemo¬therapeutikum geschützt.
Eine Restriktion von Glukose und Wachstumsfaktoren verändert den antiproliferativen Effekt von 5-FU bei kolorektalen Karzinomzellen nicht und verringert den der Fibroblasten. Damit zeigen die Zellen das Phänomen der »differential stress resistance« (DSR), dass bei 5 % und 21 % Sauerstoff beobachtet wurde, nicht aber bei und 1 % Sauerstoff. Bei 5 % Sauerstoff wurde bei 3/5 Zelllinien sogar ein besseres Ansprechen auf 5 FU nachgewiesen. Der Einfluss von Sauerstoff auf die »differential stress resistance« ist bisher wenig untersucht und basiert vermutlich auf HIF-1-abhängige intrazelluläre Signal¬wege. Die Bedeutung von Sauerstoff und seinem Transkriptionsfaktor HIF-1 für DSR ist bisher nicht verstanden und sollte deshalb weiter untersucht werden.
In this thesis, the synthesis and photophysics of a great variety of squaraine dyes are presented. This variety is based on four parent squaraines containing either indolenine or quinoline heterocycles. By a suitable choice of the donor and acceptor unit, the optical properties can already be adapted to the properties desired on the stage of the monomer.
To promote a further derivatisation of these dyes, diverse functional groups are attached to the monomers using transition metal-catalysed C-C coupling reactions. However, this has to be preceded by the synthesis of bromine-functionalised derivatives as a direct halogenation of squaraine dyes is not feasible. Therefore, the halogen function is already introduced in precursor molecules giving rise to a molecular building block system containing bromine-, boronic ester-, and alkyne-functionalised monomer units, which pave the way to a plethora of squaraine oligomers and polymers.
The indolenine homopolymer pSQB-1 as well as the corresponding small molecular weight oligomers dSQB-1 and tSQB were synthesized applying Ni-mediated Yamamoto and Pd-catalysed Suzuki coupling methodologies, respectively. The motivation for this project relied on the fundamental investigations by Völker et al. on pSQB-V. A progressive red-shift of the lowest energy absorption maximum from the dimer to the polymer was observed in CHCl3 compared to the monomer. With increasing number of monomer units, the exciton coupling decreases from the dimer to the polymer. In addition, the shape of the absorption band manifold shows a strong dependence on the solvent, which was also observed by Völker et al. J-type aggregate behavior is found in chlorinated solvents such as CHCl3 and DCM, whereas H-type aggregates are formed in acetone. Temperature-dependent absorption studies in PhCN reveals a reversible equilibrium of diverse polymer conformers, which manifests itself in a gradual change from H-aggregate behavior to a mixture with a more pronounced J-aggregate behavior upon raising the temperature. It isassumed that both characteristic aggregate bands correlate in borderline cases with two polymer structures which can be assigned to a zig-zag and a helical structure. As no experimental evidence for these structures could hitherto be provided by NMR, TD-DFT computations on oligomers (22-mers) can reproduce very closely the characteristic features of the spectra for the two conformational isomers.
The subsequent chapters are motivated by the goal to influence the optical properties through a control of the superstructure and thus of the intramolecular aggregate formation.
On the one hand, bulky groups are implemented in the 3-position of the indolenine scaffold to provoke steric repulsion and thus favoring J-aggregate behavior at the expense of helical arrangements. The resulting homopolymer pDiPhSQB bearing two phenyl groups per indolenine exhibits J-type aggregate behavior with red-shifted absorption maxima in all considered solvents which is explained to be caused by the formation of elongated zig-zag structures. Furthermore, single-crystal X-ray analysis of monomer DiPhSQB-2-Br2 reveals a torsion of the indolenine moieties as a consequence of steric congestion. The twist of the molecular geometry and the resulting loss of planarity leads to a serious deterioration of the fluorescence properties, however a significant bathochromic shift of ca. 1 200 cm-1 of the lowest absorption band was observed compared to parent SQB, which is even larger than the shift for dSQB-1 (ca. 1 000 cm-1).
On the other hand, a partial stiffening of the polymer backbone is attempted to create a bias for elongated polymer chains. In this respect, the synthetic approach is to replace every second biarylaxis with the rigid transoid benzodipyrrolenine unit. Despite a rather low average degree of polymerization < 10, exclusively red-shifted absorption maxima are observed in all solvents used.
In order to complete the picture of intramolecular aggregates through the selective design of H-aggregates, a squaraine-squaraine copolymer was synthesised containing the classic cisoid indolenine as well as the cisoid quinoline building block. Taking advantage of the highly structure directing self-assembly character of the quinoline moiety, the copolymer pSQBC indeed showes a broad, blue-shifted main absorption band in comparison with the monomer unit dSQBC. The shape of the absorption band manifold solely exhibited a minor solvent and temperature dependence indicating a persistent H-aggregate behaviour. Hence, as a proof of concept, it is shown that the optical properties of the polymers (H- and J-aggregate) and the corresponding superstructure can be inherently controlled by an adequate design of monomer precursors.
The last chapter of this work deals, in contrast to all other chapters, with intermolecular aggregates. It is shown that the two star-shaped hexasquarainyl benzenes hSQA-1 and hSQA-2 exhibit a strong propensity for self-organisation. Concentration- and temperature-dependent studies reveal a great driving force for self-assembly in acetone. While the larger hSQA-2 instantaneously forms stable aggregates, the aggregates of hSQA-1 shows a pronounced kinetic stability. Taking advantage of the kinetic persistency of these aggregates, the corresponding kinetic activation parameters for aggregation and deaggregation can be assessed. The absorption spectra of both hexasquarainyl benzenes in the aggregated state reveal some striking differences. While hSQA-1 features an intensive, very narrow and blue-shifted absorption band, two red-shifted bands are observed for hSQA-2, which are closely located at the monomer absorption. The very small bandwidth of hSQA-1 are interpreted to be caused by exchange narrowing and pointed towards highly ordered supramolecular aggregates. The concentration-dependent data of the two hexasquarainyl benzenes can be fitted to the dimer-model with excellent correlation coefficients, yielding binding constants in excess of 10^6 M-1, respectively. Such high binding constants are very surprising, considering the unfavourable bulky 3,3-dimethyl groups of the indolenine units which should rather prevent aggregation. Joint theoretical and NMR spectroscopic methods were applied to unravel the supramolecular aggregate structure of hSQA-1, which is shown to consist of two stacked hexasquarainyl benzenes resembling the picture of two stacked bowls.
Quantifizierung myokardialer Mikrostruktur und Perfusion mittels longitudinaler NMR Relaxation

Ziel der Arbeit war es die Quantifizierung funktioneller bzw. mikrostruktureller Parameter des Herzmuskels mit Hilfe T1-basierter Methoden zu verbessern. Diese Methoden basieren darauf, die gewünschte Information durch eine geeignete Präparation der Magnetisierung bzw. durch die Gabe von Kontrastmittel in den Zeitverlauf der longitudinalen Relaxation zu kodieren. Aus der Änderung der Relaxationszeit läßt sich dann die gewünschte Information bestimmen. Dafür sollte sowohl der Einfluß der Anatomie als auch derjenige der Meßmethodik auf die Bestimmung der longitudinalen Relaxationszeit und damit auf die Quantifizierung der Funktion bzw. Mikrostrukturparameter untersucht werden.
Speziell der Einfluß der Bildgebungssequenz führt dazu, daß nur eine scheinbare Relaxationszeit gemessen wird. Während dies keinen Einfluß auf die T1-basierte Bestimmung der untersuchten Mikrostrukturparameter hatte, ergab sich für die Perfusionsquantifizierung eine deutliche Abhängigkeit von den Parametern der verwendeten IRLL-Sequenz. Um diesen Einfluß gerecht zu werden, wurden an die Meßmethodik angepaßte Gleichungen zur Bestimmung der Perfusion gefunden mit denen die systematischen Abweichungen korrigiert werden können. Zusätzlich reduzieren die angepaßten Gleichungen die Anforderungen bezüglich der Inversionsqualität im schichtselektiven Experiment. Dies wurde in einem weiteren Projekt bei der Bestimmung der Nierenperfusion im Mausmodell ausgenutzt.
Neben der Untersuchung der Auswirkungen der Meßmethode wurde auch der Einfluß der anatomischen Besonderheiten des Blutkreislaufs am Herzen auf die Parameterquantifizierung mittels T1-basierter Methoden untersucht. Es konnte gezeigt werden, daß auf Grund der Anatomie des Herzens bei typischen Orientierungen der Bildgebungsschicht, auch bei der schichtselektiven Inversionspräparation der Magnetisierung des Herzmuskels ein Anteil des Blutpools invertiert wird. Daraus folgt, daß die vereinfachende Annahme, nach welcher bei schichtselektiver Präparation in Folge von Perfusion nur Blut mit Gleichgewichtsmagnetisierung den Herzmuskel erreicht, nicht erfüllt ist. Es konnte gezeigt werden, daß dies bei Perfusion zu einer deutlichen Unterschätzung der berechneten Perfusionswertes führt. Um mit diesem Problem umgehen zu können, wurde aufbauend auf einem vereinfachten Modell der zeitlichen Entwicklung der Blutmagnetisierung eine Korrektur für die Bestimmung der Perfusionswerte gefunden welche den Einfluß der anatomischen Besonderheiten berücksichtigt.
Das für die Perfusionskorrektur eingeführte Model prognostiziert ebenso, daß auch bei schichtselektiver Inversion die T1-basierte Bestimmung der untersuchten Mikrostrukturparameter von der Perfusion abhängig wird und eine systematische Überschätzung der quantifizierten Werte verursacht. Da die Perfusion im Kleintier deutlich höher ist als im Menschen, ist dieser Einfluß besonders in der präklinischen Forschung zu beachten. So können dort allein durch verminderte Perfusion deutliche Änderungen in den bestimmten Werten der Mikrostrukturparameter erzeugt werden, welche zu einer fehlerhaften Interpretation der Ergebnisse führen und somit ein falsches Bild für die Vorgänge im Herzmuskel suggerieren. Dabei bestätigt der Vergleich mit experimentellen Ergebnissen aus der Literatur die Vorhersagen für das Rattenmodell. Beim Menschen ist der prognostizierte Effekt deutlich kleiner. Der prognostizierte Fehler bspw. im RBV-Wert liegt in diesem Fall bei etwa 10% und wird üblicherweise in der aktuellen Forschung vernachlässigt. Inwieweit dies in er klinischen Forschung gerechtfertigt ist, muß in weiteren Untersuchungen geklärt werden.
Den untersuchten Methoden zur Bestimmung von funktionellen und mikrostrukturellen Parametern ist gemein, daß sie eine exakte Quantifizierung der longitudinalen Relaxationszeit T1 benötigen. Dabei ist im Kleintierbereich die klassische IRLL-Methode als zuverlässige Sequenz zur T1-Quantifizierung etabliert. In der klinischen Bildgebung werden auf Grund der unterschiedlichen Zeitskalen und anderer technischer Voraussetzungen andere Anforderungen an die Datenakquisition gestellt. Dabei hat in den letzten Jahren die MOLLI-Sequenz große Verbreitung gefunden. Sie ist eine Abwandlung der IRLL-Sequenz, bei der mit einer bSSFP-Bildgebungssequenz getriggert ganze Bilder während eines Herzschlages aufgenommen werden. Die MOLLI-Sequenz reagiert dabei empfindlich auf die Wartezeiten zwischen den einzelnen Transienten. Um mit diese Problematik in den Griff zu bekommen und gleichzeitig die Meßzeit verkürzen zu können wurde eine neue Methode zum Fitten der Daten entwickelt, welche die Abhängigkeit der scheinbaren Relaxationszeit von der Wartezeit zwischen den einzelnen Transienten, sowie der mittleren Herzrate fast vollständig eliminiert. Diese Methode liefert für das ganze klinisch Spektrum an erwarteten T1-Zeiten, vor und nach Kontrastmittelgabe, stabile Ergebnisse und erlaubte ein deutliche Verkürzung der Meßzeit, ohne die Anzahl der aufgenommenen Meßzeitpunkte zu reduzieren. Dies wurde in einer initialen klinischen Studie genutzt, um ECV-Werte in Patienten zu bestimmen.
Ein Nachteil der Verwendung der MOLLI-Sequenz ist, daß nur die scheinbare Relaxationszeit aus den Fit der Meßdaten bestimmt wird. Die standardmäßig genutzte Korrektur benutzt aber dem gefitteten Wert der Gleichgewichtsmagnetisierung um den wahren T1-Wert zu bestimmen. Somit ist es für die Bestimmung des T1-Wertes notwendig, die Qualität der Inversionspräparation zu kennen. Auf Basis der neuen Fitmethode wurde eine Anpassung der MOLLI-Sequenz demonstriert, welche die Bestimmung der Gleichgewichtsmagnetisierung unabhängig von der Qualität der Inversionspräparation erlaubt. Dafür verlängert sich die Meßdauer lediglich um einen Herzschlag um in geeigneter Weise ein zusätzliches Bild aufnehmen zu können.
Abschließend wurde in dieser Arbeit der Signal-Zeit-Verlauf der MOLLI-Sequenz eingehend theoretische untersucht um ein besseres Verständnis der getriggerten IRLL-Sequenzen zu entwickeln. In diesem Zusammenhang konnte eine einfache Interpretation der scheinbaren Relaxationszeit gefunden werden. Ebenso konnte erklärt werden, warum die für ungetriggerte IRLL-Sequenzen abgeleitete Korrekturgleichung auch im getriggerten Fall erstaunlich gute Ergebnisse liefert. Weiterhin konnten Fehlerquellen für die verbleibenden Abweichungen identifiziert werden, welche als Ausgangspunkt für die Ableitung verbesserter Korrekturgleichungen genutzt werden können.
Technische Schwierigkeiten während der Lipidapherese beeinflussen die Fraktionatorperformance. Aus diesem Grund wurde ein Großtiermodell zur Charakterisierung neuartiger Plasmafraktionatormembranen entwickelt. Vier Schafe wurden im Rahmen einer randomisierten, kontrollierten "Cross-over"-Studie einer Doppelfiltrationsplasmapherese mit drei Varianten der neuartigen FractioPES-Membran unterzogen. Diese Varianten unterschieden sich bezüglich ihrer HDL-Siebkoeffizienten (SK) (FPESa, 0.30, FPESb, 0.26, and FPESc, 0.22). Getestet wurden diese gegen eine Kontroll-Fraktionatormembran (EVAL). Siebkoeffizienten und Reduktionsraten wurden bestimmt für LDL, HDL, Fibrinogen, IgG und Albumin. Im Vergleich zu EVAL (0.42 0.04 zu 0.74 0.08) und FPESa (0.36 0.06 zu 0.64 0.04) waren die SK für HDL niedriger (p < 0,05) von FPESc (0.30 0.04 to 0.49 0.10). Die SK für Fibrinogen waren höher mit EVAL (p < 0,05; 0.02 0.01 zu 0.40 0.08) im Vergleich zu FPESb (0.05 0.02 zu 0.26 0.34) und FPESc (0.01 0.01 to 0.21 0.16). Das Tiermodell unterschied somit die minimalen Unterschiede der Fraktionatormembranen.
Coagulase-negative staphylococci, particularly Staphylococcus epidermidis, have been recognised as an important cause of health care-associated infections due to catheterisation, and livestock-associated infections. The colonisation of indwelling medical devices is achieved by the formation of biofilms, which are large cell-clusters surrounded by an extracellular matrix. This extracellular matrix consists mainly of PIA (polysaccharide intercellular adhesin), which is encoded by the icaADBC-operon. The importance of icaADBC in clinical strains provoking severe infections initiated numerous investigations of this operon and its regulation within the last two decades. The discovery of a long transcript being located next to icaADBC, downstream of the regulator gene icaR, led to the hypothesis of a possible involvement of this transcript in the regulation of biofilm formation (Eckart, 2006). Goal of this work was to characterise this transcript, named ncRNA IcaZ, in molecular detail and to uncover its functional role in S. epidermidis.
The ~400 nt long IcaZ is specific for ica-positive S. epidermidis and is transcribed in early- and mid-exponential growth phase as primary transcript. The promotor sequence and the first nucleotides of icaZ overlap with the 3' UTR of the preceding icaR gene, whereas the terminator sequence is shared by tRNAThr-4, being located convergently to icaZ. Deletion of icaZ resulted in a macroscopic biofilm-negative phenotype with highly diminished PIA-biofilm. Biofilm composition was analysed in vitro by classical crystal violet assays and in vivo by confocal laser scanning microscopy under flow conditions to display biofilm formation in real-time. The mutant showed clear defects in initial adherence and decreased cell-cell adherence, and was therefore not able to form a proper biofilm under flow in contrast to the wildtype. Restoration of PIA upon providing icaZ complementation from plasmids revealed inconsistent results in the various mutant backgrounds.
To uncover the functional role of IcaZ, transcriptomic and proteomic analysis was carried out, providing some hints on candidate targets, but the varying biofilm phenotypes of wildtype and icaZ mutants made it difficult to identify direct IcaZ mRNA targets. Pulse expression of icaZ was then used as direct fishing method and computational target predictions were executed with candidate mRNAs from aforesaid approaches. The combined data of these analyses suggested an involvement of icaR in IcaZ-mediated biofilm control. Therefore, RNA binding assays were established for IcaZ and icaR mRNA. A positive gel shift was maintained with icaR 3' UTR and with 5'/3' icaR mRNA fusion product, whereas no gel shift was obtained with icaA mRNA. From these assays, it was assumed that IcaZ regulates icaR mRNA expression in S. epidermidis. S. aureus instead lacks ncRNA IcaZ and its icaR mRNA was shown to undergo autoregulation under so far unknown circumstances by intra- or intermolecular binding of 5' UTR and 3' UTR (Ruiz de los Mozos et al., 2013). Here, the Shine-Dalgarno sequence is blocked through 5'/3' UTR base pairing and RNase III, an endoribonuclease, degrades icaR mRNA, leading to translational blockade. In this work, icaR mRNA autoregulation was therefore analysed experimentally in S. epidermidis and results showed that this specific autoregulation does not take place in this organism. An involvement of RNase III in the degradation process could not be verified here. GFP-reporter plasmids were generated to visualise the interaction, but have to be improved for further investigations.
In conclusion, IcaZ was found to interact with icaR mRNA, thereby conceivably interfering with translation initiation of repressor IcaR, and thus to promote PIA synthesis and biofilm formation. In addition, the environmental factor ethanol was found to induce icaZ expression, while only weak or no effects were obtained with NaCl and glucose. Ethanol, actually is an ingredient of disinfectants in hospital settings and known as efficient effector for biofilm induction. As biofilm formation on medical devices is a critical factor hampering treatment of S. epidermidis infections in clinical care, the results of this thesis do not only contribute to better understanding of the complex network of biofilm regulation in staphylococci, but may also have practical relevance in the future.
Historical maps are fascinating documents and a valuable source of information for scientists of various disciplines. Many of these maps are available as scanned bitmap images, but in order to make them searchable in useful ways, a structured representation of the contained information is desirable.
This book deals with the extraction of spatial information from historical maps. This cannot be expected to be solved fully automatically (since it involves difficult semantics), but is also too tedious to be done manually at scale.
The methodology used in this book combines the strengths of both computers and humans: it describes efficient algorithms to largely automate information extraction tasks and pairs these algorithms with smart user interactions to handle what is not understood by the algorithm. The effectiveness of this approach is shown for various kinds of spatial documents from the 16th to the early 20th century.
The success of semantic systems has been proven over the last years.
Nowadays, Linked Data is the driver for the rapid development of ever new intelligent systems.
Especially in enterprise environments semantic systems successfully support more and more business processes.
This is especially true for after sales service in the mechanical engineering domain.
Here, service technicians need effective access to relevant technical documentation in order to diagnose and solve problems and defects.
Therefore, the usage of semantic information retrieval systems has become the new system metaphor.
Unlike classical retrieval software Linked Enterprise Data graphs are exploited to grant targeted and problem-oriented access to relevant documents.
However, huge parts of legacy technical documents have not yet been integrated into Linked Enterprise Data graphs.
Additionally, a plethora of information models for the semantic representation of technical information exists.
The semantic maturity of these information models can hardly be measured.
This thesis motivates that there is an inherent need for a self-contained semantification approach for technical documents.
This work introduces a maturity model that allows to quickly assess existing documentation.
Additionally, the approach comprises an abstracting semantic representation for technical documents that is aligned to all major standard information models.
The semantic representation combines structural and rhetorical aspects to provide access to so called Core Documentation Entities.
A novel and holistic semantification process describes how technical documents in different legacy formats can be transformed to a semantic and linked representation.
The practical significance of the semantification approach depends on tools supporting its application.
This work presents an accompanying tool chain of semantification applications, especially the semantification framework CAPLAN that is a highly integrated development and runtime environment for semantification processes.
The complete semantification approach is evaluated in four real-life projects: in a spare part augmentation project, semantification projects for earth moving technology and harvesting technology, as well as an ontology population project for special purpose vehicles.
Three additional case studies underline the broad applicability of the presented ideas.
The thesis describes the development of new synthetic strategies towards planar nanometer-sized and electron-deficient polycyclic aromatic dicarboximides, which are rather unexplored compared with the large variety of electron-rich polycyclic aromatic hydrocarbons and nanographenes. Thus, new polycyclic aromatic systems containing a different number of dicarboximide groups were designed since this class of compounds has revealed its significance in the past due to a range of desirable molecular properties and its high thermal and photochemical stability. The synthetic concept towards these systems includes different C–C coupling techniques that were combined within coupling cascade reactions. Therefore, this thesis provides new insights into the reactivity of aromatic substrates and elucidates mechanistic aspects of C–C coupling cascade reactions to facilitate the precise design of new and desirable materials based on polycyclic aromatic dicarboximides. Furthermore, structure-property relationships as well as the optical and electrochemical properties were investigated by UV/Vis absorption and fluorescence spectroscopy and cyclic or square wave voltammetry. Insights into the molecular structures in the solid state were obtained by single-crystal X-ray analysis. In subsequent studies, highly electron-deficient perylene bisimides and their reduced species have been investigated in detail. Thus, core-functionalized perylene bisimides were synthesized and UV/Vis absorption spectroscopy, spectroelectrochemistry and cyclic or square wave voltammetry were used to determine their optical properties and the stability of the individual reduced species.
Durch das konkav gekrümmte mediale Tibiaplateau und das konvex gekrümmte laterale Tibiaplateau, welches zudem etwas nach dorsal versetzt ist, entsteht ein mechanisches Viergelenk. Dieses Viergelenk erzwingt im menschlichen Knie eine reibungsarme Roll-Gleitbewegung. Handelsübliche Knieendoprothesen haben dieser anatomischen Besonderheit bis dato keine Rechnung getragen.
In der seit 2003 eingeführten neuartigen Knieprothese „AEQUOS G1“ wird diese anatomische Besonderheit erstmalig berücksichtigt. Bei Belastung ist die Rollbewegung die dominierende Bewegungsform und minimiert so den Materialverschleiß des Polyethylen-Inlays. Durch eine dominierende Gleitbewegung bei Beugewinkeln > 60° ermöglich diese Prothese zugleich eine maximale Flexion.
Diese Arbeite beschäftigt sich mit dem klinischen Outcome und der Patientenzufriedenheit nach Implantation einer Aequos G1 Prothese in einer mittelfristigen Follow-up Studie. Die Frage, ob sich die ersten vielversprechenden Ergebnisse reproduzieren lassen, ist ebenfalls Gegenstand der vorliegend Arbeit.
Es wurde bei 54 Patienten eine Implantation einer Aequos G1 Prothese durchgeführt, diese wurden in einer Monocenterstudie nachuntersucht.
Die Patienten wurden nach mindestens 24 monatiger Prothesenstandzeit klinisch untersucht und mit Hilfe verschiedener Fragebögen Interviewt. Als Fragebögen wurden verwendet: der Kujala-Score (KS), der Turba-Score (TS), der Knee Society Score (KSS), die visuelle Analogskala Schmerz (VAS), der Oxford Knee Score (OKS) und der Short-Form-36 (SF-36).
Es wurden 46 Patienten nach durchschnittlich 40,89 Monaten nachuntersucht. Das Durchschnittsalter belief sich auf 70,30 Jahre, bei einem Frauenanteil von 73,90 % und folglich einem Männeranteil von 26,10 %. Der durchschnittliche BMI konnte mit 32,71 kg/m² bestimmt werden.
Das mittlere Bewegungsausmaß betrug in der vorliegenden Studie 108,48° ± 9,65°. Dieses Ergebnis zeigt sich nahezu identisch mit den Ergebnissen der kurzfristigen Vergleichsstudien von Frosch und Will. [15, 67]
Altersassoziierte und strahleninduzierte Veränderungen des genomweiten DNA-Methylierungs-Profils

Der Prozess des Alterns ist ein komplexer multifaktorieller Vorgang, der durch eine sukzessive Verschlechterung der physiologischen Funktionen charakterisiert ist. Ein hohes Alter ist der Hauptrisikofaktor für die meisten Krankheiten, einschließlich Krebs und Herz-Kreislauf-Erkrankungen. Das Verständnis der epigenetischen Mechanismen, die in den Prozess des Alterns involviert sind, könnte zur Entwicklung pharmakologischer Interventionen beitragen, die nicht nur die Lebenserwartung erhöhen, sondern auch den Beginn des altersassoziierten funktionellen Abbaus verzögern könnten. Durch die Langzeit-Kultivierung primärer humaner Fibroblasten wurde ein in vitro Modell für das Altern etabliert, das die Identifizierung altersassoziierter DNA-Methylierungs-Veränderungen ermöglichte. Die in vitro Alterung konnte mit einer globalen Hypomethylierung und einer erhöhten DNA-Methylierung der ribosomalen DNA assoziiert werden. Darüber hinaus konnten DNA-Methylierungs-Veränderungen in Genen und Signalwegen, die für das Altern relevant sind, und ein erhöhtes epigenetisches Alter nachgewiesen werden.
Das in vitro Modell für das Altern wurde verwendet, um neben den direkten Effekten ionisierender Strahlung auf die DNA-Methylierung auch deren Langzeit-Effekte zu untersuchen. Die Strahlentherapie ist ein entscheidendes Element der Krebstherapie, hat aber auch negative Auswirkungen und kann unter anderem das Risiko für die Entwicklung eines Zweittumors erhöhen. Bei externer Bestrahlung wird neben dem Tumor auch gesundes Gewebe ionisierender Strahlung ausgesetzt. Daher ist es wichtig zu untersuchen, wie Zellen mit intakten DNA-Reparatur-Mechanismen und funktionierenden Zellzyklus-Checkpoints durch diese beeinflusst werden. In der frühen Phase der DNA-Schadensantwort auf Bestrahlung wurden in normalen Zellen keine wesentlichen DNA-Methylierungs-Veränderungen beobachtet. Mehrere Populations-Verdoppelungen nach Strahlenexposition konnten dagegen eine globale Hypomethylierung, eine erhöhte DNA-Methylierung der ribosomalen DNA und ein erhöhtes epigenetisches Alter detektiert werden. Des Weiteren zeigten Gene und Signalwege, die mit Krebs in Verbindung gebracht wurden, Veränderungen in der DNA-Methylierung. Als Langzeit-Effekte ionisierender Strahlung traten somit die mit der in vitro Alterung assoziierten DNA-Methylierungs-Veränderungen verstärkt auf und ein epigenetisches Muster, das stark an das DNA-Methylierungs-Profil von Tumorzellen erinnert, entstand. Man geht davon aus, dass Veränderungen der DNA-Methylierung eine aktive Rolle in der Entwicklung eines Tumors spielen. Die durch ionisierende Strahlung induzierten DNA-Methylierungs-Veränderungen in normalen Zellen könnten demnach in die Krebsentstehung nach Strahlenexposition involviert sein und zu dem sekundären Krebsrisiko nach Strahlentherapie beitragen. Es ist bekannt, dass Patienten unterschiedlich auf therapeutische Bestrahlung reagieren. Die Ergebnisse dieser Arbeit weisen darauf hin, dass die individuelle Sensitivität gegenüber ionisierender Strahlung auch auf epigenetischer Ebene beobachtet werden kann.
In einem zweiten Projekt wurden Gesamtblutproben von Patienten mit Werner-Syndrom, einer segmental progeroiden Erkrankung, und gesunden Kontrollen analysiert, um mit dem vorzeitigen Altern in Verbindung stehende DNA-Methylierungs-Veränderungen zu identifizieren. Werner-Syndrom konnte nicht mit einer globalen Hypomethylierung, jedoch mit einer erhöhten DNA-Methylierung der ribosomalen DNA und einem erhöhten epigenetischen Alter assoziiert werden. Das vorzeitige Altern geht demzufolge mit spezifischen epigenetischen Veränderungen einher, die eine Beschleunigung der mit dem normalen Altern auftretenden DNA-Methylierungs-Veränderungen darstellen.
Im Rahmen dieser Arbeit konnte die Bedeutung epigenetischer Mechanismen im Prozess des Alterns hervorgehoben werden und gezeigt werden, dass sowohl exogene Faktoren, wie ionisierende Strahlung, als auch endogene Faktoren, wie das in Werner-Syndrom-Patienten mutiert vorliegende WRN-Gen, altersassoziierte DNA-Methylierungs-Veränderungen beeinflussen können.
Programmansätze und deren Einsatz in vorschulisch, schulisch und außerschulisch bildenden Kontexten erfreuen sich der zunehmenden Beliebtheit. Ein breites und nicht nachlassendes Interesse in Forschung und Praxis kommt insbesondere vorschulischen Trainingskonzepten zuteil, denen das Potenzial zugesprochen wird, später auftretenden Schwierigkeiten beim Erwerb der Schriftsprache wirksam vorzubeugen.
Das Würzburger Trainingsprogramm »Hören, lauschen, lernen« stellt einen konzeptionell auf schriftspracherwerbstheoretischen Annahmen fundierten und mit mehreren evaluierenden Studien erprobten Trainingsansatz dar. Dieser bezweckt, Kindern den Erwerb des Lesens und Schreibens zu erleichtern. Dem Anspruch, späteren Lese-Rechtschreibschwierigkeiten effektiv vorzubeugen, unterliegt die vorschulische Förderung bereichsspezifischer Kompetenzen des Schriftspracherwerbs, insbesondere der Kompetenz phonologische Bewusstheit. Die Förderung wird optimal ausgeschöpft, sofern Empfehlungen einer qualitativen Implementierung umgesetzt werden, die als Manualtreue, Durchführungsintensität, Programmdifferenzierung, Programmkomplexität, Implementierungsstrategien, Vermittlungsqualität und Teilnehmerreaktion spezifiziert sind.
Zunehmend diskutiert sind in der Trainingsforschung, neben der theoretischen Fundierung und dem zu erbringenden Nachweis an empirischer Evidenz von Programmansätzen, Kriterien der Praxistauglichkeit. Daher befasst sich die vorliegende Arbeit mit der Frage der Programmrobustheit gegenüber Trainereffekten. Es nahmen 300 Kinder an dem Würzburger Trainingsprogramm teil und wurden 64 Kindern gegenübergestellt, die dem regulären Kindergartenprogramm folgten. Angeleitet durch das erzieherische Personal fand das 5-monatig andauernde Training innerhalb des Vorschuljahres statt. Die kindliche Entwicklung in den bereichsspezifischen Kompetenzen der phonologischen Bewusstheit und der Graphem-Phonem-Korrespondenz wurde vor und nach der Trainingsmaßnahme sowie zum Schulübertritt und in den Kompetenzen des Rechtschreibens und Lesens zum Ende des ersten Schuljahres untersucht. Es ließen sich unmittelbar und langfristig Trainingseffekte des eingesetzten Programmes nachweisen; indessen blieb ein Transfererfolg aus.
Der Exploration von Trainereffekten unterlag eine Eruierung der Praxistauglichkeit des Trainingsprogrammes anhand der erfolgten Implementierung durch das anleitende erzieherische Personal. Aus der ursprünglich mit 300 Kindern aus 44 involvierten Kindergärten bestehenden Datenbasis wurden drei Subgruppen mit insgesamt 174 Kindern aus 17 Kindergärten identifiziert, bei denen deutliche Diskrepanzen zu unmittelbaren, langfristigen und transferierenden Effekten des Trainingsprogrammes auftraten. Exploriert wurden Unterschiede in der Durchführung, um Rückschlüsse auf qualitative Aspekte der Programmimplementierung zu ziehen. Die Befunde des Extremgruppenvergleichs deuteten an, dass weniger Aspekte der Manualtreue und Durchführungsintensität ausschlaggebend für die Programmwirksamkeit waren; vielmehr schien für die Wirksamkeit des Trainingsprogrammes die Implementierung in der Art und Weise, wie die Trainingsinhalte den Kindern durch das erzieherische Personal vermittelt waren, entscheidend zu sein. Befunde zur eruierten Teilnehmerreaktion, die auf differenzielle Fördereffekte verweisen, stellten die Trainingswirksamkeit insbesondere für Kinder heraus, bei denen prognostisch ein Risiko unterstellt war, später auftretende Schwierigkeiten mit der Schriftsprache zu entwickeln. Ferner zeichnete sich ab, dass neben der Qualität der Programmimplementierung scheinbar auch Unterschiede in der schulischen Instruktionsmethode des Lesens und Schreibens einen nivellierenden Einfluss auf den Transfererfolg des Programmes ausübten. Theoretische und praktische Implikationen für den Einsatz des Trainingsprogrammes wurden diskutiert.
Herzkreislauferkrankungen stellen die häufigsten Todesursachen in den Industrienationen dar. Die Entwicklung nichtinvasiver Bildgebungstechniken mit Hilfe der Magnetresonanz-Tomografie (MRT) ist daher von großer Bedeutung, um diese Erkrankungen frühzeitig zu erkennen und um die Entstehungsmechanismen zu erforschen. In den letzten Jahren erwiesen sich dabei genetisch modifzierte Mausmodelle als sehr wertvoll, da sich durch diese neue Bildgebungsmethoden entwickeln lassen und sich der Krankheitsverlauf im Zeitraffer beobachten lässt.
Ein große Herausforderung der murinen MRT-Bildgebung sind die die hohen Herzraten und die schnelle Atmung. Diese erfordern eine Synchronisation der Messung mit dem Herzschlag und der Atmung des Tieres mit Hilfe von Herz- und Atemsignalen. Konventionelle Bildgebungstechniken verwenden zur Synchronisation mit dem Herzschlag EKG Sonden, diese sind jedoch insbesondere bei hohen Feldstärken (>3 T) sehr störanfällig. In dieser Arbeit wurden daher neue Bildgebungsmethoden entwickelt, die keine externen Herz- und Atemsonden benötigen, sondern das MRT-Signal selbst zur Bewegungssynychronisation verwenden. Mit Hilfe dieser Technik gelang die Entwicklung neuer Methoden zur Flussbildgebung und der 3D-Bildgebung, mit denen sich das arterielle System der Maus qualitativ und quantitativ erfassen lässt, sowie einer neuen Methode zur Quantisierung der longitudinalen Relaxationszeit T1 im murinen Herzen. Die in dieser Arbeit entwickelten Methoden ermöglichen robustere Messungen des Herzkreislaufsystems. Im letzten Kapitel konnte darüber hinaus gezeigt werden dass sich die entwickelten Bildgebungstechniken in der Maus auch auf die humane Bildgebung übertragen lassen.
The rise of automated driving will fundamentally change our mobility in the near future. This thesis specifically considers the stage of so called highly automated driving (Level 3, SAE International, 2014). At this level, a system carries out vehicle guidance in specific application areas, e.g. on highway roads. The driver can temporarily suspend from monitoring the driving task and might use the time by engaging in so called non-driving related tasks (NDR-tasks). However, the driver is still in charge to resume vehicle control when prompted by the system. This new role of the driver has to be critically examined from a human factors perspective.
The main aim of this thesis was to systematically investigate the impact of different NDR-tasks on driver behavior and take-over performance. Wickens’ (2008) architecture of multiple resource theory was chosen as theoretical framework, with the building blocks of multiplicity (task interference due to resource overlap), mental workload (task demands), and aspects of executive control or self-regulation. Specific adaptations and extensions of the theory were discussed to account for the context of NDR-task interactions in highly automated driving.
Overall four driving simulator studies were carried out to investigate the role of these theoretical components. Study 1 showed that drivers focused NDR-task engagement on sections of highly automated compared to manual driving. In addition, drivers avoided task engagement prior to predictable take-over situations. These results indicate that self-regulatory behavior, as reported for manual driving, also takes place in the context of highly automated driving. Study 2 specifically addressed the impact of NDR-tasks’ stimulus and response modalities on take-over performance. Results showed that particularly visual-manual tasks with high motoric load (including the need to get rid of a handheld object) had detrimental effects. However, drivers seemed to be aware of task specific distraction in take-over situations and strictly canceled visual-manual tasks compared to a low impairing auditory-vocal task. Study 3 revealed that also the mental demand of NDR-tasks should be considered for drivers’ take-over performance. Finally, different human-machine-interfaces were developed and evaluated in Simulator Study 4. Concepts including an explicit pre-alert (“notification”) clearly supported drivers’ self-regulation and achieved high usability and acceptance ratings.
Overall, this thesis indicates that the architecture of multiple resource theory provides a useful framework for research in this field. Practical implications arise regarding the potential legal regulation of NDR-tasks as well as the design of elaborated human-machine-interfaces.
In der Studie dieser Dissertation wird untersucht, ob das biokompatible Kollagennetz Lyoplant® (B.Braun, Deutschland) ein geeignetes Biomaterial zur Harnblasenaugmentation ist. Es wurden 16 Wistar Ratten ein Lyoplant® -Netz in die Harnblasen implantiert. Nach sechs Wochen lang täglicher Visite wurden die Harnblasen explantiert und mikroskopisch, sowie immunhistologisch aufgearbeitet. Es zeigte sich eine Epithelialisierung und die Bildung von Bindegewebe, außerdem wenig Entzündungszellen, sodass Lyoplant® ein gut verträgliches Material zur Blasenaugmentation im Kleintiermodell ist.
In this thesis stability and robustness properties of systems of functional differential equations which dynamics depends on the maximum of a solution over a prehistory time interval is studied. Max-operator is analyzed and it is proved that due to its presence such kind of systems are particular case of state dependent delay differential equations with piecewise continuous delay function. They are nonlinear, infinite-dimensional and may reduce to one-dimensional along its solution. Stability analysis with respect to input is accomplished by trajectory estimate and via averaging method. Numerical method is proposed.
Fluorescence microscopy has become one of the most important techniques for the imaging of biological cells and tissue, since the technique allows for selective labeling with fluorescent molecules and is highly suitable for low-light applications down to the single molecule regime. The methodological requirements are well-defined for studying membrane receptors within a highly localized nanometer-thin membrane. For example, G-protein-coupled receptors (GPCRs) are an extensively studied class of membrane receptors that represent one of the most important pharmaceutical targets. Ligand binding and GPCR activation dynamics are suspected to take place at the millisecond scale and may even be far faster. Thus, techniques that are fast, selective, and live-cell compatible are required to monitor GPCR dynamics. Fluorescence resonance energy transfer (FRET) and total internal reflection fluorescence microscopy (TIRF-M) are methods of choice to monitor the dynamics of GPCRs selectively within the cell membrane.
Despite the remarkable success of these modalities, there are limitations. Most importantly, inhomogeneous illumination can induce imaging artifacts, rendering spectroscopic evaluation difficult. Background signal due to scattering processes or imperfect labeling can hamper the signal-to-noise, thus limiting image contrast and acquisition speed. Careful consideration of the internal physiology is required for FRET sensor design, so that ligand binding and cell compatibility are well-preserved despite the fluorescence labeling procedures. This limitation of labeling positions leads to very low signal changes in FRET-based GPCR analysis. In addition, microscopy of these systems becomes even more challenging in single molecule or low-light applications where the accuracy and temporal resolution may become dramatically low. Fluorescent labels should therefore be brighter, protected from photobleaching, and as small as possible to avoid interference with the binding kinetics. The development of new fluorescent molecules and labeling methods is an ongoing process. However, a complete characterization of new labels and sensors takes time. So far, the perfect dye system for GPCR studies has not been found, even though there is high demand.
Thus, this thesis explores and applies a different approach based on improved illumination schemes for TIRF-M as well as metal-coated coverslips to enhance fluorescence and FRET efficiency. First, it is demonstrated that a 360° illumination scheme reduces typical TIRF artifacts and produces a much more homogenously illuminated field of view. Second, membrane imaging and FRET spectroscopy are improved by metal coatings that are used to modulate the fluorescent properties of common fluorescent dyes. Computer simulation methods are used to understand the underlying photophysics and to design the coatings. Third, this thesis explores the operational regime and limitations of plasmonic approaches with high sectioning capabilities. The findings are summarized by three publications that are presented in the results section of this work. In addition, the theory of fluorescence and FRET is explained, with particular attention to its emission modulations in the vicinity of metal-dielectric layers. Details of the instrumentation, computer simulations, and cell culture are described in the method section. The work concludes with a discussion of the findings within the framework of recent technological developments as well as perspectives and suggestions for future approaches complete the presented work.
Platelets, small anucleated blood cells responsible for hemostasis, interact at sights of injury with several exposed extracellular matrix (ECM) proteins through specific receptors. Ligand binding leads to activation, adhesion and aggregation of platelets. Already megakaryocytes (MKs), the immediate precursor cells in bone marrow (BM), are in constant contact to these ECM proteins (ECMP). The interaction of ECMP with MKs is, in contrast to platelets, less well understood. It is therefore important to study how MKs interact with sinusoids via the underlying ECMP. This thesis addresses three major topics to elucidate these interactions and their role in platelet biogenesis.
First, we studied the topology of ECMP within BM and their impact on proplatelet formation (PPF) in vitro. By establishing a four-color immunofluorescence microscopy we localized collagens and other ECMP and determined their degree of contact towards vessels and megakaryocytes (MKs). In in vitro assays we could demonstrate that Col I mediates increased MK adhesion, but inhibits PPF by collagen receptor GPVI. By immunoblot analyses we identified that the signaling events underyling this inhibition are different from those in platelet activation at the Src family kinase level.
Second, we determined the degree of MK-ECM interaction in situ using confocal laser scanning microscopy of four-color IF-stained femora and spleen sections. In transgenic mouse models lacking either of the two major collagen receptors we could show that these mice have an impaired association of MKs to collagens in the BM, while the MK count in spleen increased threefold. This might contribute to the overall unaltered platelet counts in collagen receptor-deficient mice.
In a third approach, we studied how the equilibrium of ECMP within BM is altered after irradiation. Collagen type IV and laminin-α5 subunits were selectively degraded at the sinusoids, while the matrix degrading protease MMP9 was upregulated in MKs. Platelet numbers decreased and platelets became hyporesponsive towards agonists, especially those for GPVI activation.
Taken together, the results indicate that MK-ECM interaction differs substantially from the well-known platelet-ECM signaling. Future work should further elucidate how ECMP can be targeted to ameliorate the platelet production and function defects, especially in patients after BM irradiation.
Der Name des populären Chirurgen Professor Julius Hackethal (1921-1997) weckt bis heute Assoziationen mit den Schlagwörtern Medizinkritik, Sterbehilfe und alternative Krebsbehandlungen. In einem stetig größer werdenden Forschungsstand zur Geschichte der Alternativmedizin und deutschen Nachkriegsmedizin beschäftigt sich vorliegende Dissertation mit Julius Hackethals Medizinkritik im Allgemeinen und Krebs im Speziellen, seinem therapeutischen Gegenvorschlag EUBIOS, der Sterbehilfedebatte sowie der Darstellung seiner Medizinkritik und der entsprechenden Resonanz in deutschen Medien. Die vermeintlichen „Kardinalfehler“ der Schulmedizin bei Krebs werden dabei exemplarisch am Beispiel Prostatakarzinom erläutert.
Welche gesellschaftlichen und schulmedizinischen Rahmenbedingungen vorlagen und die Medizinkritik anfachten, was die Gründe für Julius Hackethals Abkehr von der Schulmedizin waren und inwiefern sich seine Medizinkritik von anderen Kritikern der damaligen schulmedizinischen Verhältnisse unterschied, waren wichtige Fragestellungen der Arbeit. Zudem wird unter Miteinbeziehung von Zeitzeugenberichten beantwortet, warum er mit seinem EUBIOS-Konzept und vermeintlichen Pauschalbehandlungen gerade bei Krebspatienten regen Zulauf fand. Zuletzt stand das Verhältnis von Julius Hackethal zu den Medien sowie das der Medien zu Julius Hackethal im Fokus.
Neben allen Publikationen Hackethals als Hauptquellen und Mikroebene wurde die Recherche um umfangreiche Quellen der Epoche, Forschungsliteratur zum Thema und audiovisuelle Medien als Makroebene erweitert. Hauptschlagwörter waren Medizinkritik und Krise der Krebstherapie, Alternativmedizin sowie das Thema Sterbehilfe. Zudem wurden alle im Zusammenhang mit Julius Hackethal erschienenen Artikel in vorselektionierten Medien, dem Deutschen Ärzteblatt, den Nachrichtenmagazinen Spiegel und Stern sowie den Illustrierten Quick und BUNTE, den Fragestellungen entsprechend, analysiert.
Vor einem sich wandelnden Gesundheitspanorama in der zweiten Jahrhunderthälfte mit enttäuschten Hoffnungen an rasche Behandlungserfolge chronischer (Krebs-)Erkrankungen und einem kritischen Hinterfragen von (Arzt-)Autoritäten wurden Forderungen nach einer posthippokratischen Medizin und Ethik laut. Schlagwörter wie Fünfminutenmedizin, Apparate- und Maschinenmedizin und anonyme Großkliniken machten die Runde. Als Gegenantwort kam es zu einer Renaissance alternativer Behandlungsrichtungen, die von verunsicherten, von der Schulmedizin enttäuschten Patienten aufgegriffen wurden.
Julius Hackethal war dabei nicht der einzige oder erste Kritiker der damaligen schulmedizinischen Praxis, allerdings war Kritik von einem bis dahin selbst praktizierenden Schulmediziner und Professor ein Novum. Mit bewusstem Verzicht auf „Medizinbabylonisch“ und Büchern sowie öffentlicher Kritik in teils vulgärer, aggressiver „Volkssprache“ wurden komplexe Sachverhalte der breiten Masse zugänglich gemacht. Bis heute ist sein Neologismus harmloser „Haustierkrebse“ ein Begriff und mit ihm verquickt. Durch derart provozierende Rhetorik, aber auch spektakuläres, medienwirksames Handeln polarisierte Julius Hackethal dabei zeitlebens. Seine Beihilfe zum Suizid Hermine Eckerts im Jahr 1984 ist hierfür Beispiel und wird in der Arbeit dargelegt. Zudem ließ er keine Möglichkeit aus, seine Thesen in Medien jedweder Couleur zu verbreiten, die großen medizinischen Themenfelder für sich zu reklamieren und gleichzeitig für eigene Kliniken und sein Behandlungsprogramm EUBIOS zu werben. Ein einzelner Querdenker habe es geschafft, die viel zu kompliziert denkende Schulmedizin zu entmystifizieren.
Die Position des Deutschen Ärzteblatts war zwangsläufig klar abgesteckt: Gegenüber Standeskritikern galt es eine klare Position aufrechtzuerhalten, um die bereits in der Kritik stehende Schulmedizin nicht noch weiter zu gefährden. Entsprechend einseitig und teils unseriös fielen die Artikel aus. Das Nachrichtenmagazin Spiegel begrüßte Hackethals anfängliche Medizinkritik, distanzierte sich dann aber ausdrücklich von ihm und seinen Krebsheilungsvisionen sowie seiner Forderung nach einer Legalisierung des ärztlich assistierten Suizids.
Im 21. Jahrhundert ist Medizinkritik weiterhin präsent, gleichzeitig sind alternative Behandlungsverfahren zu einem festen Bestandteil des Behandlungsrepertoires einst streng schulmedizinisch ausgerichteter Ärzte geworden. Julius Hackethal war dabei ein zeitgeschichtliches Phänomen auf einem kurz vor und vor allem nach ihm existenten Kontinuum deutscher Medizinkritik, dem weitere Persönlichkeiten mit neuen Heilsversprechungen oder Ideen zur Umstrukturierung der modernen Schulmedizin rasch nachfolgten und nachfolgen werden.
Die vorliegende Arbeit ist eine korpusbasierte und datenbankgestützte Fehleranalyse von schriftlichen Textproduktionen mazedonischsprachiger Deutschlerner. Sie ist zwischen zwei zentralen Zweigen der sprachwissenschaftlichen Fachliteratur situiert: einerseits die Fehlerlinguistik und andererseits die Studien zu Mehrsprachigkeit bzw. Tertiärspracherwerb (oder L3-Erwerb). Aus Sicht der Fehlerlinguistik setzt sie sich das Ziel, Fehler bzw. Auffälligkeiten zu identifizieren und sie nach linguistischen Kriterien zu typologisieren: orthografische, morphologische, morphosyntaktische, syntaktische, lexikalische und textspezifische. Aus Sicht der Forschung für Tertiärspracherwerb setzt sich die Arbeit das Ziel, Einflüsse aus der Muttersprache (L1) und weiteren Fremdsprachen, insbesondere die der zweiten Fremdsprache (L2 - Englisch, aus einem schulischen Erwerbskontext) zu ermitteln und möglichst deren wechselseitige Wirkung aufzuzeigen. Diese beiden Gesichtspunkte der Untersuchung lassen sich gut durch ein weiteres Ziel ergänzen, überprüfen und miteinander verknüpfen, nämlich das Ermitteln weiterer Fehlerursachen, die nicht nur auf interlinguale Einflüsse zurückzuführen sind, sondern auch auf intralinguale, wie z. B. Einflüsse aus der Sprache selbst oder Einflüsse aus dem Unterricht und den Unterrichtsmaterialien sowie Strukturen und Strategien des Spracherwerbsprozesses selbst. Überdies werden wichtige Faktoren – wie didaktische, lernerspezifische oder erwerbsspezifische – mitberücksichtigt, sodass sich diese Arbeit auch in Nischen der Sprachdidaktik positionieren lässt.
G protein-coupled receptors (GPCRs) form the biggest receptor family that is encoded in the human genome and represent the most druggable target structure for modern therapeutics respectively future drug development. Belonging to aminergic class A GPCRs muscarinic Acetylcholine receptors (mAChRs) are already now of clinical relevance and are also seen as promising future drug targets for treating neurodegenerative diseases like Alzheimer or Parkinson. The mAChR family consist of five subtypes showing high sequence identity for the endogenous ligand binding region and thus it is challenging until now to selectively activate a single receptor subtype. A well accepted method to study ligand binding, dynamic receptor activation and downstream signaling is the fluorescence resonance energy transfer (FRET) application. Here, there relative distance between two fluorophores in close proximity (<10 nm) can be monitored in a dynamic manner. The perquisite for that is the spectral overlap of the emission spectrum of the first fluorophore with the excitation spectrum of the second fluorophore. By inserting two fluorophores into the molecular receptor structure receptor FRET sensors can serve as a powerful tool to study dynamic receptor pharmacology.
Dualsteric Ligands consist of two different pharmacophoric entities and are regarded as a promising ligand design for future drug development. The orthosteric part interacts with high affinity with the endogenous ligand binding region whereas the allosteric part binds to a different receptor region mostly located in the extracellular vestibule. Both moieties are covalently linked. Dualsteric ligands exhibit a dynamic ligand binding. The dualsteric binding position is characterized by a simultaneous binding of the orthosteric and allosteric moiety to the receptor and thus by receptor activation. In the purely allosteric binding position no receptor activation can be monitored.
In the present work the first receptor FRET sensor for the muscarinic subtype 1 (M1) was generated and characterized. The M1-I3N-CFP sensor showed an unaltered physiological behavior as well as ligand and concentration dependent responses. The sensor was used to characterize different sets of dualsteric ligands concerning their pharmacological properties like receptor activation. It was shown that the hybrids consisting of the synthetic full agonist iperoxo and the positive allosteric modulator of BQCA type is very promising. Furthermore, it was shown for orthosteric as well as dualsteric ligands that the degree of receptor activation is highly dependent on the length of and the chemical properties of the linker moiety. For dualsteric ligands a bell-shaped activation characteristic was reported for the first time, suggesting that there is an optimal linker length for dualsteric ligands. The gained knowledge about hybrid design was then used to generate and characterize the first photo-switchable dualsteric ligand. The resulting hybrids were characterized with the M1-I3N-CFP sensor and were described as photo-inactivatable and dimmable. In addition to the ligand characterization the ligand application methodology was further developed and improved. Thus, a fragment-based screening approach for dualsteric ligands was reported in this study for the first time. With this approach it is possible to investigate dualsteric ligands in greater detail by applying either single ligand fragments alone or in a mixture of building blocks. These studies revealed the insights that the effect of dualsteric ligands on a GPCR can be rebuild by applying the single building blocks simultaneously. The fragment-based screening provides high potential for the molecular understanding of dualsteric ligands and for future screening approaches. Next, a further development of the standard procedure for measuring FRET by sensitized emission was performed. Under normal conditions single cell FRET is measured on glass coverslips. After coating the coverslips surface with a 20 nm thick gold layer an increased FRET efficiency up to 60 % could be reported. This finding was validated in different approaches und in different configurations. This FRET enhancement by plasmonic surfaces was until yet unreported in the literature for physiological systems and make FRET for future projects even more powerful.
Ionisierende Strahlung (IR) ist in der medizinischen Diagnostik und in der Tumortherapie von zentraler Bedeutung, kann aber Genominstabilität und Krebs auslösen. Strahleninduzierte Genominstabilität (RIGI) ist in den klonalen Nachkommen bestrahlter Zellen zu beobachten, die zugrundeliegenden Mechanismen sind jedoch noch unverstanden. Zur Erforschung von verzögerten Strahleneffekten wurden primäre embryonale Fibroblastenkulturen mit 2 Gray bestrahlt und für 20 Populationsverdopplungen klonal expandiert. Zellen, die keiner Strahlung ausgesetzt waren, dienten als Kontrolle für normale Alterungsprozesse. Die Klone wurden durch klassische Chromosomenbänderungstechniken analysiert und in Abhängigkeit der Stabilität ihres Genoms in Gruppen eingeteilt. Ein Klon wurde als stabil gewertet, wenn die analysierten Metaphasen keinerlei Auffälligkeiten zeigten, während instabile Klone ein Mosaik aus normalen und abnormalen Metaphasen waren. Die Zellen von zwei Spendern wurden untersucht, um interindividuelle Strahleneffekte zu beurteilen. Nach Bestrahlung hatten mehr als die Hälfte der Klone Metaphasen mit strukturellen Aberrationen und wurden dementsprechend als instabil eingestuft. Drei Klone zeigten zudem numerische Aberrationen, die ausschließlich das Y Chromosom betrafen. Fluoreszenz in situ Hybridisierungen verifizierten diese Beobachtung in weiteren Klonen und deuteten an, dass der Verlust des Y Chromosoms mit RIGI assoziiert ist.
Molekulare Karyotypisierungen mit SNP Arrays ergaben, dass IR in den Klonen Veränderungen der Kopienzahl auslöst. Ein Unterschied zwischen chromosomal stabilen und instabilen Klonen konnte jedoch nicht detektiert werden. Chromosomale Regionen, in denen sich bekanntermaßen fragile Stellen befinden, zeigten eine Anhäufung von CNVs. Ein RIGI Effekt konnte für die fragile Stelle 3B, in der sich das Gen FHIT befindet, identifiziert werden.
Exom Sequenzierungen von Klonen und der entsprechenden Massenkultur zeigten eine alterungsassoziierte Entstehung von Varianten. Der Effekt wurde durch die Einwirkung von Strahlung erhöht. Auf Ebene von einzelnen Nukleotiden konnten ebenfalls Anhäufungen von Schäden in bestimmten genomischen Bereichen detektiert werden, dieser Effekt ging ohne die typischen RIGI Endpunkte einher.
Die Ergebnisse der vorliegenden Arbeit zeigen, dass strahlenbedingte Veränderungen auf verschiedenen Ebenen (Chromosomen, Genkopienzahl und einzelnen Nukleotiden) beobachtet werden können, welche, unabhängig von RIGI, die Tumorentstehung begünstigen. Speziell Veränderungen im FRA3B Lokus und der Verlust des Y Chromosoms scheinen jedoch über die Destabilisierung des Genoms zur Krebsentstehung beizutragen.
Bei einem SHT handelt es sich um eine mechanische Schädigung des Hirngewebes, verursacht durch eine Gewalteinwirkung auf den Kopf. Diese initiale Schädigung des Hirngewebes weitet sich nachfolgend aus. Wirksame Therapien, um diese sekundären Pathomechanismen zu inhibieren, gibt es nicht. Sofern das SHT überlebt wird, ist es eine der häufigsten Ursachen für bleibende Behinderungen. Wichtige Pathomechanismen, welche zur Ausweitung der Hirnschädigung beitragen, sind das posttraumatische Hirnödem und Entzündungsreaktionen. Beides wird durch die Aktivierung des so-genannten Kallikrein-Kinin-Systems begünstigt. In der vorliegenden Arbeit wurde dieses System 1 Stunde nach experimentellem SHT durch die Applikation des C1-Inhibitors gehemmt und am nachfolgenden Tag die Auswirkungen bewertet. Die Ergebnisse zeigen, dass diese Behandlung nach der Hirnverletzung zu einer Reduktion des Hirnödems und der Entzündungsreaktion führt. Die Bildung von Thromben in den Hirngefäßen ist geringer als in Kontrolltieren, vermutlich da der C1-Inhibitor auch die intrinsische Gerinnungs-kaskade hemmt. Insgesamt führt die Behandlung zu kleineren Hirnläsionen als in entsprechenden Kontrolltieren. Hiermit stellt der C1-Inhibitor ein potenzieller Therapieansatz bei SHT dar. Jedoch bleibt es offen, inwiefern sich diese Ergebnisse auf das menschliche SHT übertragen lassen.
Hintergrund:
Die weltweit höchsten HIV-Prävalenzen finden sich im südlichen Afrika. In den letzten Jahren gelang in dieser Region die massive Ausweitung der Verfügbarkeit antiretroviraler Medikamente – ein großer Erfolg im Kampf gegen HIV und AIDS. Doch nur durch regelmäßiges Viruslastmonitoring können die Therapieerfolge nachhaltig gesichert werden. In vielen Ländern des südlichen Afrikas ist die Verfügbarkeit von Viruslasttests noch unzureichend. Gründe dafür sind die hohen Kosten und die Komplexität kommerzieller Testsysteme. Als Alternative für ressourcenknappe Regionen wurde eine qualitative PCR-Minipool-Strategie zum kombinierten Nachweis von virologischem Versagen und Medikamentenresistenzen vorgeschlagen. In dieser Arbeit sollte diese Methode an die Gegebenheiten des südlichen Afrikas adaptiert und im Rahmen einer Studie validiert werden.
Methoden:
Die qualitative PCR wurde zunächst für HIV-1 Subtyp C optimiert. Dafür wurden Primer designt und in eine Nested-PCR integriert. Das Detektionslimit der optimierten Methode wurde bestimmt, um den HIV-1 RNA-Eintrag in die RT-PCR so abstimmen zu können, dass trotz der qualitativen Natur der PCR eine Unterscheidung zwischen hoch- und niedrigpositiven Proben möglich war. Im Rahmen einer Studie an 50 südafrikanischen Patientenproben wurde die qualitative PCR-Minipool-Strategie mit der Referenzmethode verglichen. An einigen positiv getesteten Proben wurde eine genotypische Resistenztestung durchgeführt.
Ergebnisse:
Das Detektionslimit der Nested-PCR betrug 20 RNA-Kopien. Daraus wurde der optimale RNA-Eintrag in die RT-PCR abgeleitet. 50 Patientenproben wurden in 10 Minipools à 5 Proben getestet, anschließend erfolgte die Dekonvolution der 7 positiven Pools. Die qualitative PCR-Minipool-Strategie detektierte 10 der 11 Patientenproben mit einer Viruslast über 1.000 Kopien/ml. Die Übereinstimmung mit der Referenzmethode betrug 98 %, die diagnostische Sensitivität für die Detektion von Therapieversagern betrug 91 %, der negative prädiktive Wert 98 %. Die Spezifität und der positive prädiktive Wert betrugen jeweils 100 %. Durch die Minipool-Methode konnten 10 % der Tests eingespart werden. Vier positiv getestete Patientenproben wurden der genotypischen Resistenztestung zugeführt. Alle wurden erfolgreich sequenziert und resistenzassoziierte Mutationen wurden identifiziert.
Schlussfolgerung:
Die konsequente Virussuppression verbessert die Gesundheit der Patienten unter ART und ist eine Schlüsselmaßnahme auf dem Weg zur Beendigung der HIV-Pandemie. Viele Länder des südlichen Afrikas verfügen jedoch noch nicht über angepasste Technologien zum Monitoring der Viruslast. Die in dieser Arbeit optimierte Methode zeigte bei der qualitativen Viruslastbestimmung von 50 südafrikanischen Patientenproben eine hervorragende Übereinstimmung mit der Referenzmethode. Durch die Kombination mit einer genotypischen Resistenztestung bietet sie darüber hinaus wichtige Informationen über die Ätiologie des Therapieversagens. Dem behandelnden Arzt hilft das bei der Entscheidung über die Notwendigkeit einer Therapieumstellung. Durch ihr anpassungsfähiges und simples Testdesign ist die qualitative PCR-Minipool-Strategie für die Anwendung in ressourcenknappen Regionen des südlichen Afrikas geeignet.
In der vorliegenden Arbeit werden die Sensorikeigenschaften der lumineszierenden metallorganischen Gerüstverbindungen (metal-organic frameworks, MOFs) ■(3@∞)[Ba0.98Eu0.02(Im)2] (1), ■(3@∞)[Sr0.90Eu0.10(Im)2] (2), ■(3@∞)[Tb(Im)3] (3), ■(2@∞)[Tb2Cl6(4,4‘-bipy)3]•2(4,4‘-bipy) (4) und ■(2@∞)[Eu2Cl6(4,4‘-bipy)3]•2(4,4‘-bipy) (5), sowohl in Form der Bulksubstanzen als auch in Form von mixed-matrix membranes (MMMs) gegenüber den Analyten Wasser, Methanol, Ethanol und Isopropanol untersucht und mittels kinetischer Betrachtungen quantitativ beschrieben.
Hierfür wurde ein Versuchsaufbau konstruiert, der den Anforderungen einer quantitativen Erfassung der Sensorikeigenschaften genügt und einflussgebende externe und interne Parameter, wie die Konzentration des Analyten, Temperatur, Druck, Probenmenge und Probenoberfläche standardisiert, um eine möglichst genaue Erfassung der auftretenden Veränderung der Lumineszenzintensität zu ermöglichen. Zusätzlich wurde eine mathematische Betrachtung genutzt, um die Ergebnisse der Experimente zu interpretieren und in einen Gesamtkontext zu setzen.
Für jedes der fünf Modellsysteme wurde je eine Messreihe gegen ein breites Spektrum an relativen Feuchten (Wasser als Analyt) bei 25 °C durchgeführt. Der in allen Fällen auftretende Lumineszenzverlust ließ sich jeweils mittels einer Exponentialfunktion beschreiben und zur Bestimmung der konzentrationsabhängigen Reaktionskonstanten sowie der Halbwertszeiten nutzen. Die gewonnenen Informationen wurden verwendet, um die Reaktionskonstanten und die Schwellenwerte der Sensorik der jeweiligen Systeme zu bestimmten.
In weiteren Versuchsreihen wurde der Messaufbau genutzt und der Einfluss der Alkohole Methanol, Ethanol und Isopropanol auf die Lumineszenz der MOFs ■(3@∞)[Ba0.98Eu0.02(Im)2] (1), ■(2@∞)[Tb2Cl6(4,4‘-bipy)3]•2(4,4‘-bipy) (4) und ■(2@∞)[Eu2Cl6(4,4‘-bipy)3]•2(4,4‘-bipy) (5) bei verschie-denen Konzentrationen in der Gasphase und bei unterschiedlichen Temperaturen untersucht, quantitativ bestimmt und gemäß der vorangegangen Messreihen für Feuchte beschrieben. Zusätzlich wurde die Veränderung des Lumineszenzmusters der MOFs ■(2@∞)[Tb2Cl6(4,4‘-bipy)3]•2(4,4‘-bipy) (4) und ■(2@∞)[Eu2Cl6(4,4‘-bipy)3]•2(4,4‘-bipy) (5) untersucht.
Die Möglichkeiten der Prozessierung von ■(3@∞)[Ba0.98Eu0.02(Im)2] (1), und ■(2@∞)[Tb2Cl6(4,4‘-bipy)3]•2(4,4‘-bipy) (4) wurden im Hinblick auf eine praktische Anwendung im Bereich der Sensorik hin untersucht. Hierfür wurden die jeweiligen Bulksubstanzen via Dropcoating in die drei Polymere Polystyrol (PS), Polyvinychlorid (PVC) und Polymethylpenten (TPX) eingebettet. Die so dargestellten MMMs wurden auf ihre Sensorikeigenschaften hin untersucht und der Lumineszenzverlust wurde quantitativ beschrieben. Des Weiteren wurde die korrespondierende Bulksubstanz, in Bezug auf die (konzentrationsabhängigen) Reaktionskonstanten und Halbwertszeiten, mit den MMMs verglichen. Im Rahmen einer Kooperation mit der Arbeitsgruppe von Prof. Dr. Janiak der Heinrich-Heine-Universität Düsseldorf wurden die MOFs ■(3@∞)[Ba0.98Eu0.02(Im)2] (1) und ■(3@∞)[Sr0.90Eu0.10(Im)2] (2) in Polysulfon (PSF) eingebettet und ebenfalls auf ihre Fähigkeit zur Feuchtesensorik hin untersucht.
Im Rahmen der vorliegenden Arbeit war es erstmals möglich, die Sensoreigenschaften der genutzten MOFs und CPs (1 - 5) quantitativ zu erfassen und mathematisch zu beschreiben. Ebenso konnte das entwickelte Sensorikkonzept erfolgreich auf die Erfassung von verschiedenen Alkoholen in der Gasphase übertragen werden (für System 1, 4 und 5). Anschließend war es möglich, die genutzten Modellsysteme 1, 2 und 4 unter dem Erhalt der Lumineszenzeigenschaft in verschiedene Polymere einzubetten und deren Eigenschaften zur Feuchtesensorik ebenfalls quantitativ zu erfassen und mathematisch zu beschreiben.
Im letzten Abschnitt der Arbeit wurden die beiden porösen ZIFs (zeolitic imidazolate frameworks) Na-Rho-ZMOF (6) und Zn(nmIm)2 (7) mit einer Terbiumnitratlösung behandelt, um durch die Füllung der Kavitäten die Lumineszenzeigenschaft der Terbiumionen in das ZIF zu implementieren. Der Grad und die Qualität der Befüllung wurden anschließend analytisch auf Lumineszenzeigenschaften hin untersucht. Hierbei konnte die Befüllung von Na-Rho-ZMOF (6) erfolgreich nachgewiesen werden, während es bei Zn(nmIm)2 (7) nicht zu einer Befüllung des Porensystems kam.
Alzheimer’s disease is a complex network of several pathological hallmarks. These characteristics always occur concomitantly and cannot be taken as distinct features of the disease. While there are hypotheses trying to explain the origin and progression of the illness, none of them is able to pinpoint a definitive cause. This fact challenges researchers not to focus on one individual hallmark but, bearing in mind the big picture, target two or more indications at once. This work, therefore, addresses two of the major characteristics of AD: the cholinergic hypothesis and neurotoxic oxidative stress. The former was achieved by targeting the postsynaptic muscarinic M1 acetylcholine receptor to further investigate its pharmacology, and the latter with the synthesis of neuroprotective natural antioxidant hybrids.
The first aim was the design and synthesis of dualsteric agonists of the muscarinic M1 acetylcholine receptor. Activation of this receptor was previously shown to improve AD pathologies like the formation of Aβ and NFTs and protect against oxidative stress and caspase activation. Selectively targeting the M1 receptor is difficult as subtypes M1 – M5 of the muscarinic AChRs largely share the same orthosteric binding pocket. Orthosteric ligands are thus unsuitable for selective activation of one specific subtype. Secondary, allosteric binding sites are more diverse between subtypes. Allosteric ligands are, however, in most cases dependent on an orthosteric ligand to cause downstream signals. Dualsteric ligands thus utilize the characteristics of both orthosteric and allosteric ligands in form of a message-address concept. Bridged by an alkylene-linker, the allosteric part ensures selectivity, whereas the orthosteric moiety initiates receptor activation. Two sets of compounds were synthesised in this sense. In both cases, the orthosteric ligand carbachol is connected to an allosteric ligand via linkers of different chain length. The first set utilizes the selective allosteric M1 agonist TBPB, the second set employs the selective M1 positive allosteric modulator BQCA. Six compounds were obtained in twelve-step syntheses each. For each one, a reference compound lacking the carbachol moiety was synthesised. The dualsteric ligands 1a-c and 2a c were tested in the IP1 assay. The assay revealed that the TBPB-dualsterics 1 are not able to activate the receptor, whereas the respective TBPB-alkyl reference compounds 27 gave signals depending on the length of the alkylene-linker, suggesting allosteric partial agonism of alkyl compounds 27 and no dualsteric binding of the putatively dualsteric compounds 1. The dualsteric BQCA molecules 2, however, activated the receptor as expected. Efficacy of the C5 linked compound 2b was the highest, yet C3 and C8 compounds (2a and 2c) also showed partial agonism. In this case, the reference compounds 31 showed no receptor activation, implying the intended dualsteric binding mode of the BQCA-carbachol compounds 2. Further investigations will be conducted by the working group of Dr. Christian Tränkle at the Department of Pharmacology at the University of Bonn to confirm binding modes and determine affinities as well as selectivity of the synthesised dualsteric compounds.
The second project dealt with the design, synthesis and biological evaluation of neuroprotective esters of the flavonolignan silibinin. While silibinin is already a potent antioxidant, it has been observed that the 7-OH group has a pro-oxidative character, making this position attractive for functionalisation. In order to obtain more potent antioxidants, the pro-oxidative position was esterified with other antioxidant moieties like ferulic acid 35 and derivatives thereof. Seventeen esters of silibinin 32, including pure diastereomers of 7 O feruloylsilibinin (43a and 43b) and a cinnamic acid ester of 2,3-dehydrosilibinin 46, were synthesised by regioselective esterification using acyl chlorides under basic conditions. The physicochemical antioxidant properties were assessed in the FRAP assay. This assay revealed no improvement of the antioxidant properties except for 7-O-dihydrosinapinoylsilibinin 39b. These results, however, do not correlate with the neuroprotective properties determined in the HT-22 hippocampal neuronal cell model. The assay showed overadditive neuroprotective effects of the esters exceeding those of its components and equimolar mixtures with the most potent compounds being 7-O-cinnamoylsilibinin 37a, 7-O-feruloylsilibinin 38a and the acetonide-protected caffeic acid ester 40a. These potent Michael system bearing compounds may be considered as “PAINS”, but the assays used to assess antioxidant and neuroprotective activities were carefully chosen to avoid false positive readouts. The most potent compounds 37a and 38a, as well as the diastereomers 43a and 43b, were further studied in assays related to AD. In vitro ischemia, inhibition of microglial activation, PC12 cell differentiation and inhibition of Aβ42 and τ protein aggregation assays showed similar results in terms of overadditive effects of the synthesised esters. Moreover, the diastereomers 43a and 43b showed differences in their activities against oxytosis (glutamate-induced apoptosis), inhibition of Aβ42 and τ protein aggregation, and PC12 cell differentiation. The stereospecific effect or mode of action against Aβ42 and τ protein aggregation is more pronounced than that of silybin A (32a) and silybin B (32b) reported in literature and needs to be elucidated in future work. Stability measurements in cell culture medium revealed that the esters do not only get hydrolysed but are partially oxidised to their respective 2,3-dehydrosilibinin esters. Because dehydrosilibinin 45 itself is described as a more potent antioxidant than silibinin 32, 7 O cinnamoyl-2,3-dehydrosilibinin 46 was expected to be even more potent than its un-oxidised counterpart 37a in terms of neuroprotection. The oxytosis assay, however, showed that the neurotoxicity of 46 is much more pronounced, especially at higher concentrations, reducing its neuroprotective potential. Dehydrosilibinin esters are therefore inferior to the silibinin esters for application as neuroprotectants, because of the difficulty of their synthesis and their increased neurotoxicity. A synergistic effect of both species (silibinin and the oxidised form) might, however, be possible or even necessary for the pronounced neuroprotective effects of silibinin esters. As the dehydro-species show distinct neuroprotective properties at low concentrations, their continuous formation over time might make an essential contribution to the overall neuroprotection of the synthesised esters. Due to solubility issues for some of the ester compounds, 7-O-cinnamoylsilibinin 37a was converted into a highly soluble hemisuccinate. The vastly improved solubility of 7 O cinnamoyl-23-O-succinylsilibinin 48 was confirmed in shake-flask experiments. Contrary to expectation, stability examinations showed that the succinyl compound 48 is not cleaved to form 7-O-cinnamoylsilibinin 37a. Neuroprotection assays confirmed that 48 is not a prodrug of the corresponding ester. It was determined that the main site of hydrolysis is the 7-position, cleaving 37 to silibinin 32 and cinnamic acid thus reducing the compound’s neuroprotective effects. Nevertheless, the compound still showed neuroprotection at a concentration of 25 µM. The improved solubility might be more beneficial than the higher neuroprotection of the poorly soluble parent compound 37a in vivo. 7 O Cinnamoylsilibinin 37a was further investigated to reduce Aβ25 35 induced learning impairment in mice. While tendencies of improved short-term and long-term memory in the animals were observed, the effects are not yet statistically significant in both Y-maze and passive avoidance tests. A greater number of test subjects is necessary to ensure correctness of the preliminary results presented in this work. However, an effect of ester 37a is observable in vivo, showing blood-brain barrier penetration. The esters synthesised are a novel approach for the treatment of AD as they show strong neuroprotective effects and their hydrolysis products or metabolites are only non-toxic natural products.
Die vorliegende Arbeit ist in zwei Teile gegliedert und befasst sich im ersten Abschnitt mit der stöchiometrischen und katalytischen Aktivierung von Element-Element-Bindungen an NHC-stabilisierten Eisen(II)-Komplexen. Im Fokus der Untersuchungen steht hierbei sowohl die Isolierung und Charakterisierung neuartiger NHC-stabilisierter Eisen-Komplexe sowie deren Nutzung als Katalysatoren in der Hydrosilylierung von Carbonylverbindungen und der Hydrophosphanierung von Mehrfachbindungssystemen. Der zweite Teil dieser Arbeit ist der Reaktivität N-heterocyclischer Carbene gegenüber Hauptgruppenelement-Verbindungen wie beispielsweise Chlorsilanen, Stannanen, Phosphanen und Alanen gewidmet. Neben der Aufklärung mechanistischer Details der Reaktionen ist die übergangsmetallfreie Hydrodefluorierung von Fluoraromaten zentraler Bestandteil dieser Untersuchungen.
Die vorliegende Arbeit befasst sich mit der Synthese und Untersuchung V- und brettförmiger Flüssigkristalle zur Realisierung einer biaxialen nematischen Mesophase. Es wurde erfolgreich eine Serie neuer Mesogene mit hockeyschlägerförmiger und V-förmiger Struktur synthetisiert. Zusätzlich wurden Dimere aus einem dieser hockeyschlägerförmigen Verbindungen dargestellt. Als Kernbaustein wurde Benzo[1,2-b:4,3-b']dithiophen verwendet, dessen lokales Kerndipolmoment von 1.0 Debye sich nach theoretischen Vorgaben zusätzlich zum Bindungswinkel (108.9 °) positiv auf die Bildung einer Nb-Phase auswirken soll. Überraschenderweise bilden nur die hockeyschlägerförmigen Moleküle eine uniaxiale, optisch positive nematische Mesophase aus. Alle anderen V-förmigen Verbindungen und sogar die Dimere sind ausschließlich kristallin und keine Flüssigkristalle. Die Einkristallstrukturanalyse eines hockeyschlägerförmigen Mesogens sowie eines V-förmigen Moleküls zeigt bemerkenswerte Ähnlichkeiten auf. Ein Modell des Phasenübergangs wird präsentiert, welches die Abwesenheit der nematischen Mesophase in der Familie der V-förmigen, formstabilen Mesogene mit terminalen aliphatischen Ketten erklärt. Zudem befasst sich die Arbeit mit der Synthese und der Untersuchung brettförmiger Moleküle, welche dem optimalen Seitenverhältnis von 15 : 5 : 3 mit L > B > T zur Bildung biaxialer Mesophasen, relativ nahekommen. Ein Anthrachinon-Kernbaustein wurde dabei mit Armen bestehend aus einem Oligo(phenylenethinylen)-Grundgerüst entsprechender Länge verknüpft. Es konnten verschiedene dachförmige Mesogene dargestellt werden, bei denen die Art und Anzahl der Seitenketten sowie der terminalen Ketten variiert wurde. Thermische sowie mikroskopische Untersuchungen zeigen bei allen Verbindungen eine breite nematische Mesophase. Mittels spezieller Röntgenstreuung im magnetischen Feld kann die Bildung nematischer Domänen mit SmC-artigen biaxialen Aggregaten bestätigt werden.
Given points in the plane, connect them using minimum ink. Though the task seems simple, it turns out to be very time consuming. In fact, scientists believe that computers cannot efficiently solve it. So, do we have to resign? This book examines such NP-hard network-design problems, from connectivity problems in graphs to polygonal drawing problems on the plane. First, we observe why it is so hard to optimally solve these problems. Then, we go over to attack them anyway. We develop fast algorithms that find approximate solutions that are very close to the optimal ones. Hence, connecting points with slightly more ink is not hard.
I. Timing is a crucial feature in organisms that live within a variable and changing environment. Complex mechanisms to measure time are wide-spread and were shown to exist in many taxa. These mechanisms are expected to provide fitness benefits by enabling organisms to anticipate environmental changes and adapt accordingly. However, very few studies have addressed the adaptive value of proper timing. The objective of this PhD-project was to investigate mechanisms and fitness consequences of timing decisions concerning colony phenology and foraging activity in the honey bee (Apis mellifera), a social insect species with a high degree of social organization and one of the most important pollinators of wild plants and crops. In chapter II, a study is presented that aimed to identify the consequences of disrupted synchrony between colony phenology and the local environment by manipulating the timing of brood onset after hibernation. In a follow-up experiment, the importance of environmental factors for the timing of brood onset was investigated to assess the potential of climate change to disrupt synchronization of colony phenology (Chapter III). Chapter IV aimed to prove for the first time that honey bees can use interval time-place learning to improve foraging activity in a variable environment. Chapter V investigates the fitness benefits of information exchange between nest mates via waggle dance communication about a resource environment that is heterogeneous in space and time.
II. In the study presented in chapter II, the importance of the timing of brood onset after hibernation as critical point in honey bee colony phenology in temperate zones was investigated. Honey bee colonies were overwintered at two climatically different sites. By translocating colonies from each site to the other in late winter, timing of brood onset was manipulated and consequently colony phenology was desynchronized with the local environment. Delaying colony phenology in respect to the local environment decreased the capability of colonies to exploit the abundant spring bloom. Early brood onset, on the other hand, increased the loads of the brood parasite Varroa destructor later in the season with negative impact on colony worker population size. This indicates a timing related trade-off and illustrates the importance of investigating effects of climate change on complex multi-trophic systems. It can be concluded that timing of brood onset in honey bees is an important fitness relevant step for colony phenology that is highly sensitive to climatic conditions in late winter. Further, phenology shifts and mismatches driven by climate change can have severe fitness consequences.
III. In chapter III, I assess the importance of the environmental factors ambient temperature and photoperiod as well as elapsed time on the timing of brood onset. Twenty-four hibernating honey bee colonies were placed into environmental chambers and allocated to different combinations of two temperature regimes and three different light regimes. Brood onset was identified non-invasively by tracking comb temperature within the winter cluster. The experiment revealed that ambient temperature plays a major role in the timing of brood onset, but the response of honey bee colonies to temperature increases is modified by photoperiod. Further, the data indicate the involvement of an internal clock. I conclude that the timing of brood onset is complex but probably highly susceptible to climate change and especially spells of warm weather in winter.
IV. In chapter IV, it was examined if honey bees are capable of interval time-place learning and if this ability improves foraging efficiency in a dynamic resource environment. In a field experiment with artificial feeders, foragers were able to learn time intervals and use this ability to anticipate time periods during which feeders were active. Further, interval time-place learning enabled foragers to increase nectar uptake rates. It was concluded that interval time-place learning can help honey bee foragers to adapt to the complex and variable temporal patterns of floral resource environments.
V. The study presented in chapter V identified the importance of the honey bee waggle dance communication for the spatiotemporal coordination of honey bee foraging activity in resource environments that can vary from day to day. Consequences of disrupting the instructional component of honey bee dance communication were investigated in eight temperate zone landscapes with different levels of spatiotemporal complexity. While nectar uptake of colonies was not affected, waggle dance communication significantly benefitted pollen harvest irrespective of landscape complexity. I suggest that this is explained by the fact that honey bees prefer to forage pollen in semi-natural habitats, which provide diverse resource species but are sparse and presumably hard to find in intensively managed agricultural landscapes. I conclude that waggle dance communication helps to ensure a sufficient and diverse pollen diet which is crucial for honey bee colony health.
VI. In my PhD-project, I could show that honey bee colonies are able to adapt their activities to a seasonally and daily changing environment, which affects resource uptake, colony development, colony health and ultimately colony fitness. Ongoing global change, however, puts timing in honey bee colonies at risk. Climate change has the potential to cause mismatches with the local resource environment. Intensivation of agricultural management with decreased resource diversity and short resource peaks in spring followed by distinctive gaps increases the probability of mismatches. Even the highly efficient foraging system of honey bees might not ensure a sufficiently diverse and healthy diet in such an environment. The global introduction of the parasitic mite V. destructor and the increased exposure to pesticides in intensively managed landscapes further degrades honey bee colony health. This might lead to reduced cognitive capabilities in workers and impact the communication and social organization in colonies, thereby undermining the ability of honey bee colonies to adapt to their environment.
Die idiopathische dilatative Kardiomyopathie ist eine eher seltene Herzerkrankung (Inzidenz 8/100.000 Einwohner pro Jahr), jedoch eine der häufigsten Ursache für die Entstehung einer Herzinsuffizienz bei jüngeren Patienten und geht immer mit Veränderungen sowohl des humoralen als auch des zellulären Immunsystems einher.
Die der vorliegenden Arbeit zugrunde liegende ETiCS-Studie untersucht besonders die Rolle von 1-adrenergen Autoantikörpern bei der Ausbildung und Entwicklung einer Herzinsuffizienz sowie deren Einfluss auf den Verlauf der kardialen Pumpfunktion nach einem ersten akuten Myokardinfarkt (FAMI) oder einer ersten Episode einer akuten Myokarditis (AMitis).
Nach Studieneinschluss wurden diese beiden klar definierten Patientenkollektive über einen Zeitraum von 12 Monaten beobachtet. Im Fokus der vorliegenden Arbeit stand der Verlauf der mittels MRT erhobenen kardialen Funktionsdaten (LVEF, EDV, ESV, SV, HI und LV-Masse, Baseline und 12 Monats-Follow Up) und ihr Zusammenhang mit Biomarkern der Herzschädigung (CK/CK-MB) sowie der Ausbildung/dem Verlauf von 1-Autoantikörpern.
FAMI-Patienten wiesen innerhalb eines Jahres grundsätzlich eine Verbesserung der kardialen Pumpfunktion auf; Patienten mit erstem Hinterwandinfarkt zeigten im Vergleich zu Vorderwandinfarkt-Patienten bei generell besseren Ausgangswerten auch im Verlauf deutlich geringere Funktions- und Volumenänderungen. Patienten mit kleineren Myokardinfarkten (CK Werte <1000 U/l) zeigten, unabhängig von der Lokalisation, eine bessere Erholung der LV-Funktion, als Patienten mit größeren Herzinfarkten (CK Werte >1000 U/l). Bei den im Rahmen dieser Arbeit analysierten Infarktpatienten konnte allerdings kein wesentlicher Einfluss einer Ausbildung oder Gegenwart von herzspezifischen Autoantikörpern auf die Entwicklung der kardialen Pumpfunktion nachgewiesen werden.
AMitis-Patienten wiesen innerhalb eines Jahres ebenso grundsätzlich eine Verbesserung der kardialen Pumpfunktion auf; In dieser Patienten-Kohorte scheint die mittel- und langfristige Entwicklung der kardialen Funktionsparameter jedoch stark von der Ausbildung 1-adrenerger Autoantikörper beeinflusst zu sein: Antikörper-negative AMitis-Patienten zeigten zu jedem Beobachtungszeitpunkt deutlich bessere kardiale Funktionsparameter als Antikörper-positiv getestete AMitis-Patienten.
Dies bestätigt die initiale Hypothese der ETiCS-Studie und muss durch die abschließende Analyse aller ETiCS-Studienpatienten (erwartet 2019) noch bestätigt werden. Den Nachweis von 1-adrenergen Autoantikörpern in die Routine-Labordiagnostik einzuführen, erscheint aufgrund der Ergebnisse der vorliegenden Arbeit zumindest bei Myokarditispatienten sinnvoll. Bei einem Herzinfarkt sprechen die Ergebnisse der vorliegenden Arbeit jedoch nicht für einen Mehrwert eines routinemäßigen 1-Autoantikörper-Screenings.
Tiaprid wird bei Kindern und Jugendlichen im deutschsprachigen Raum als Mittel der ersten Wahl zur Behandlung von Ticstörungen off-label eingesetzt. Es gilt dabei die generelle Empfehlung, Therapeutischen Drug Monitoring (TDM) bei der Behandlung von Minderjährigen mit Neuro-/Psychopharmaka durchzuführen. Therapeutische Referenzbereiche für Tiaprid sind bisher jedoch nur für erwachsene Patienten mit Chorea Huntington definiert worden (1000 bis 2000 ng/ml) (Hiemke et al., 2011).
An ausgewählten Zentren im Rahmen des Kompetenznetzwerks Therapeutisches Drug Monitoring Kinder- und Jugendpsychiatrie (www.tdm-kjp.com) wurden von 2007 bis 2014 standardisiert TDM-Daten erfasst, um den Zusammenhang zwischen Dosis, Serumkonzentration, Wirksamkeit und UAW von Tiaprid zu untersuchen sowie Hinweise auf einen möglichen alters- und diagnosespezifischen therapeutischen Referenzbereich zu generieren.
Bei den 49 Patienten (mittleres Alter 12,5 Jahre; 84 % männlich) zeigte sich eine positive Korrelation (r= 0.76; p< .001) zwischen der Dosis (Mittelwert 354 mg) und der Serumkonzentration von Tiaprid (Mittelwert 1324 ng/ml) mit einer ausgeprägten interindividuellen Variabilität, jedoch keine Beziehung zwischen Serumkonzentration und Wirkeffekt (83,3 % profitierten) bzw. UAW in der Gesamtpopulation.
Die Auswertung der Verlaufsmessungen von Patienten mit mehreren Messungen der Tiaprid-Serumkonzentration ergab beim dritten Messzeitpunkt eine negative Korrelation zwischen Wirkeffekt und Serumkonzentration (r= -.68; p= .032). Bei Patienten mit Serumkonzentrationen unter 2000 ng/ml wurde ein günstigerer klinischer Effekt dokumentiert als bei solchen mit Konzentrationen oberhalb dieses Wertes. Die ROC-Analyse ergab eine Sensitivität von 86 %, ab einer Konzentration von 618 ng/ml zu respondieren (AUC= .524). Kein Patient litt an einer schweren UAW und nur wenige Patienten unter leichten oder mittelschweren UAW (n=13).
Diese Ergebnisse lassen vermuten, dass der untere therapeutische Referenzbereich für jugendliche Patienten mit einer Tic-Störung bei etwa 600 ng/ml liegt und die obere Grenze von 2000 ng/ml auch als Orientierungswert auf Kinder und Jugendliche gelten könnte. Bevor diesbezüglich gültige Empfehlungen für den klinischen Alltag formuliert werden, müssen Studien mit höheren Fallzahlen und mehr kontrollierten Studiendesigns abgewartet werden.
For cellular viability, transcription is a fundamental process. Hereby, the DNA plays the most elemental and highly versatile role. It has long been known that promoters contain conserved and often well-defined motifs, which dictate the site of transcription initiation by providing binding sites for regulatory proteins. However, research within the last decade revealed that it is promoters lacking conserved promoter motifs and transcribing constitutively expressed genes that constitute the majority of promoters in eukaryotes. While the process of transcription initiation is well studied, whether defined DNA sequence motifs are required for the transcription of constitutively expressed genes in eukaryotes remains unknown. In the highly divergent protozoan parasite Trypanosoma brucei, most of the proteincoding genes are organized in large polycistronic transcription units. The genes within one polycistronic transcription unit are generally unrelated and transcribed by a common transcription start site for which no RNA polymerase II promoter motifs have been identified so far. Thus, it is assumed that transcription initiation is not regulated but how transcription is initiated in T. brucei is not known. This study aimed to investigate the requirement of DNA sequence motifs and chromatin structures for transcription initiation in an organism lacking transcriptional regulation. To this end, I performed a systematic analysis to investigate the dependence of transcription initiation on the DNA sequence. I was able to identify GT-rich promoter elements required for directional transcription initiation and targeted deposition of the histone variant H2A.Z, a conserved component during transcription initiation. Furthermore, nucleosome positioning data in this work provide evidence that sites of transcription initiation are rather characterized by broad regions of open and more accessible chromatin than narrow nucleosome depleted regions as it is the case in other eukaryotes. These findings highlight the importance of chromatin during transcription initiation. Polycistronic RNA in T. brucei is separated by adding an independently transcribed miniexon during trans-splicing. The data in this work suggest that nucleosome occupancy plays an important role during RNA maturation by slowing down the progressing polymerase and thereby facilitating the choice of the proper splice site during trans-splicing. Overall, this work investigated the role of the DNA sequence during transcription initiation and nucleosome positioning in a highly divergent eukaryote. Furthermore, the findings shed light on the conservation of the requirement of DNA motifs during transcription initiation and the regulatory potential of chromatin during RNA maturation. The findings improve the understanding of gene expression regulation in T. brucei, a eukaryotic parasite lacking transcriptional Regulation.
Cadherin-13 (CDH13) is an atypical member of the cadherin superfamily, a group of membrane proteins mediating calcium-dependent cellular adhesion. Although CDH13 shows the classical extracellular cadherin structure, the typical transmembrane and cytoplasmic domains are absent. Instead, CDH13 is attached to the cell membrane via a glycosylphosphatidylinositol (GPI) anchor. These findings and many studies from different fields suggest that CDH13 also plays a role as a cellular receptor. Interestingly, many genome-wide association studies (GWAS) have found CDH13 as a risk gene for attention-deficit/hyperactivity disorder (ADHD) and other neurodevelopmental disorders. In previous work from our research group, strong expression of Cdh13 mRNA in interneurons of the hippocampal stratum oriens (SO) was detected. Therefore, double-immunofluorescence studies were used to evaluate the degree of co-expression of CDH13 with seven markers of GABAergic interneuron subtypes. For this purpose, murine brains were double stained against CDH13 and the respective marker and the degree of colocalization in the SO of the hippocampus was assessed. Based on the result of this immunofluorescence study, quantitative differences in interneuron subtypes of the SO between Cdh13 knockout (ko), heterozygote (het) and wildtype (wt) mice were investigated in this dissertation using stereological methods. In addition, genotype- dependent differences in the expression of genes involved in GABAergic and glutamatergic neurotransmission were analyzed by quantitative real-time PCR (qRT-PCR). Primers targeting different GABA receptor subunits, vesicular GABA and glutamate transporter, GABA synthesizing enzymes and their interaction partners were used for this purpose.
The results of the stereological quantification of the interneuron subtypes show no significant differences in cell number, cell density or volume of the SO between Cdh13 ko, het and wt mice. On the other hand, qRT-PCR results indicate significant differences in the expression of tropomyosin-related kinase B gene (TrkB), which encodes the receptor of brain-derived neurotrophic factor (BDNF), a regulator of GABAergic neurons. This finding supports a role for CDH13 in the regulation of BDNF signaling in the hippocampus.
The aim of this thesis was the development of a multifunctional coating system for AuNPs based on thioether polymers, providing both excellent colloidal stability and a variable possibility to introduce functionalities for biological applications.
First, two thioether-polymer systems were synthesised as a systematic investigation into colloidal stabilisation efficacy. Besides commonly used monovalent poly(ethylene glycol) (PEG-SR), its structural analogue linear poly(glycidol) (PG-SR) bearing multiple statistically distributed thioether moieties along the backbone was synthesised. Additionally, respective thiol analogues (PEG-SH and PG-SH) were produced and applied as reference.
Successive modification of varyingly large AuNPs with aforementioned thiol- and thioether-polymers was performed via ligand exchange reaction on citrate stabilised AuNPs. An increased stabilisation efficacy of both thioether-polymers against biological and physiological conditions, as well as against freeze-drying compared to thiol analogues was determined.
Based on the excellent colloidal stabilisation efficacy and multi-functionalisability of thioether-PG, a plethora of functional groups, such as charged groups, hydrophilic/hydrophobic chains, as well as bio-active moieties namely diazirine and biotin was introduced to the AuNP surface. Moreover, the generic and covalent binding of diazirine-modified PG-SR with biomolecules including peptides and proteins was thoroughly demonstrated.
Lastly, diverse applicability and bioactivity of aforementioned modified particles in various studies was displayed, once more verifying the introduction of functionalities. On the one hand the electrostatic interaction of charged AuNPs with hydrogels based on hyaluronic acid was applied to tune the release kinetics of particles from three-dimensional scaffolds. On the other hand the strong complexation of siRNA onto two positively charged AuNPs was proven. The amount of siRNA payload was tuneable by varying the surface charge, ionic strength of the surrounding medium and the N/P ratio. Moreover, the biological activity and selectivity of the biotin-streptavidin conjugation was verified with respectively functionalised particles in controlled agglomeration test and in laser-triggered cell elimination experiments. In the latter, streptavidin-functionalised AuNPs resulted in excellent depletion of biotinylated cells whereas unfunctionalised control particles failed, excluding unspecific binding of these particles to the cell surface.
Zielsetzung:
Die lymphogene Metastasierung bei Patienten mit Urothelkarzinom der Harnblase ist bekanntermaßen ein schlechter Prognosefaktor. Langzeitergebnisse dieser Patienten vor allem hinsichtlich Outcome, Prognosefaktoren und Einfluß der adjuvanten Chemotherapie sind in der Literatur rar.
Das Patientenkollektiv wurde hinsichtlich ihres Langzeitüberlebens und dessen Abhängigkeit von verschiedenen Variablen untersucht.
Material und Methoden:
Es wurden Daten aus zwei universitären Einrichtungen gesammelt. Alle Patienten mit lymphogen metastasiertem Urothelkarzinom der Harnblase, welche von 1991 bis 2008 radikal zystektomiert worden sind, wurden in die Studie eingeschlossen. Patienten mit neoadjuvanter Chemotherapie wurden ausgeschlossen.
Ergebnisse:
Es resultierten 170 Patienten. Die 5 und 10 – Jahres Überlebensraten betrugen 33.1% und 21.9 %. Die retrospektive statistische Analyse ergab, das Tumorausdehnung, Ausmaß der nodalen Beteiligung und adjuvante Chemotherapie unabhängige Prognosefaktoren hinsichtlich des Gesamtüberlebens darstellen.
Schlußfolgerung:
Trotz lymphogener Metastasierung aller Patienten dieses Kollektivs, hatten solche mit kleinerer Tumorausdehnung (pTa-pT2b) deutlich bessere Überlebensraten, als solche mit extravesikalem Tumor (pT3a-pT4b). Die adjuvante Chemotherapie war assoziiert mit verbesserten Überlebensraten, sodass diese Daten den Einsatz der adjuvanten Chemotherapie in dieser Gruppe von Patienten befürwortet.
The noninvasive magnetic resonance imaging technique allows for the investigation of functional processes in the living plant. For this purpose during this work, different NMR imaging methods were further developed and applied.
For the localisation of the intrusion of water into the germinating rape seed with the simultaneous depiction of the lipid-rich tissue via a 3D rendering, in Chap. 5 the technique of interleaved chemical selective acquisition of water and lipid was used in the germinating seed. The utilization of high-resolution MR images of germinated seeds enabled the localization of a predetermined water gap in the lipid-rich aleurone layer, which resides directly under the seed coat. The for a long time in biology prevalent discussion, whether such a gap exists or the seed soaks up the water from all sides, rather like a sponge, could hereby, at least for the rapeseed seed, be answered clearly. Furthermore, the segmentation and 3D visualization of the vascular tissue in the rapeseed seeds was enabled by the high-resolution datasets, a multiply branched structure preconstructed in the seed could be shown. The water is directed by the vascular tissue and thus awakens the seed gradually to life. This re-awakening could as well be tracked by means of invasive imaging via an oxygen sensor. In the re-awakened seeds, the lipid degradation starts, other than expected, not in the lipid-rich cotyledons but in the residual endosperm remaining from seed development and in the aleurone layer which previously protected the embryo. Within this layer, the degradation could be verified in the high-resolution MR datasets.
The method presented in Chap. 6 provides a further characteristic trait for phenotyping of seeds and lipid containing plants in general. The visualization of the compounds of fatty acids in plant seeds and fruits could be achieved by the distinct utilization of chemical shift-selective imaging techniques. Via the application of a CSI sequence the fatty acid compounds in an olive were localized in a 2D slice. In conjunction with an individually adjusted CHESS presaturation module Haa85 the high-resolution 3D visualization of saturated and unsaturated fatty acid compounds in different seeds was achieved. The ratio maps calculated from these datasets allow to draw conclusions from the developmental stage or the type of seed. Furthermore, it could be shown that the storage condition of two soybean seeds with different storage time durations lead to no degradation of the fatty acid content.
Additional structural information from inside of dry seeds are now accessible via MRI. In this work the imaging of cereal seeds could be significantly improved by the application of the UTE sequence. The hitherto existing depictions of the lipid distribution, acquired with the spin echo sequence, were always sufficient for examinations of the lipid content, yet defects in the starchy endosperm or differences in the starch concentration within the seed remained constantly unseen with this technique. In a direct comparison of the datasets acquired with the previous imaging technique (spin echo) and with UTE imaging, the advantage of data acquisition with UTE could be shown. By investigating the potential seed compounds (starch, proteins, sugar) in pure form, the constituent parts contributing to the signal could be identified as bound water (residual moisture) and starch. The application of a bi-exponential fit on the datasets of the barley seed enabled the separate mapping of magnetization and of relaxation time of two components contributing to the NMR signal. The direct comparison with histological stainings verified the previous results, thus this technique can be used for the selective imaging of starch in dry seeds.
Conclusions on the translocation characteristics in plants can be drawn by the technique proposed in Chap. 8. The associated translocation velocities can now, even in the range of several um/h, be determined in the living plant. Based on calculated concentrations of an MR contrast agent, which was taken up by the plant, these translocation velocities were estimated both in longitudinal direction, thus along the vascular bundle, and in horizontal direction, thus out of the bundle. The latter velocity is located below the contrast agent's velocity value of free diffusion. By adjusting a dynamic contrast-enhancing imaging technique (DCE-Imaging, Tof91) the acquisition duration of a T1-map was significantly reduced. By means of these maps, local concentrations of the contrast agent in plant stems and the siliques of the rapeseed plant could be determined.
Numerous questions in plant science can only be answered by non-invasive techniques such as MRI. For this reason, besides the experimental results achieved in this work, further NMR methods were tested and provided for the investigation of plants.
As an example, the study on the imaging of magnetic exchange processes are mentioned, which provided the groundwork for a possible transfer of CEST experiments (Chemical Exchange Saturation Transfer) to the plant. The results are presented in the bachelor thesis of A. Jäger Jae17, which was performed under my supervision, they find great interest under biologists.
The development of new technologies, which extend the possibilities for the investigation of living organisms, is of great importance. For this reason, I have contributed to the development of the currently unpublished method RACETE (Refocused Acquisition of Chemical Exchange Transferred Excitations [Jak17, Reu17, Gut18a]). By rephasing the transferred magnetization the utilization of properties which have not been available in chemical "`exchange"' experiments is enabled. With this method a positive contrast is generated, thus a reference experiment is not mandatory. Furthermore, the image phase, which in classical experiments contains no information about the exchanged protons, can be used for the distinct identification of multiple substances which have been excited simultaneously.
This recently at the Department of Experimental Physics V developed method can be used in particular for the identification of lipids and for the localization of sugars and amino acids, thus it can serve the enhancement and improvement of non-invasive analytical methods.
Assoziationsuntersuchungen zu schizophrenen und affektiven Psychosen im Bereich des EphA4 Gens

Die Schizophrenie ist eine schwerwiegende Erkrankung, deren Gesamtlebenzeitprävalenz ca. 1% beträgt. Da bei schizophrenen Erkrankungen die genetische Komponente eine erhebliche Rolle spielt und es außerdem in bisherigen Studien Hinweise für einen Zusammenhang von EphA4 mit diversen neuronalen Krankheitsformen gibt, ist dieser mögliche Zusammenhang Gegenstand der durchgeführten Untersuchungen. In der vorliegenden Arbeit sollte die Rolle des Eph receptor A4 bei der Ätiopathogenese von schizophrenen und affektiven Psychosen untersucht werden, da besonders zur Rolle der Eph- und Ephrin A-Familie bei schizophrenen Erkrankungen derzeit noch grundlegendes Wissen fehlt. Dabei wurde ein Patientenkollektiv von mehr als 1000 Probanden sowohl nach der ICD-10-Klassifikation als auch der Klassifikation von Leonhard in Unterformen eingeteilt und diese getrennt untersucht und mit einer gesunden Kontrollgruppe verglichen. Es wurden sowohl SNP-Analysen als auch Haplotypanalysen durchgeführt. Das Kandidatengen EphA4 liegt beim Menschen auf dem Chromosom 2 (Basenpaar 221.418.027 bis 221.574.202), besteht aus insgesamt 156.176 Basenpaaren und dient vor allem der Steuerung der Zellform und -bewegung durch Veränderungen am Aktinoskelett. Insgesamt wurden 9 SNPs auf Assoziation mit schizophrenen Psychosen und zykloiden Psychosen untersucht, um einen möglichen Einfluss von EphA4 bei der Ätiopathogenese oder im Krankheitsverlauf zu diagnostizieren. Zum einen wurden 4 Single SNP-Analysen durchgeführt, um einzelne SNPs auf Assoziation mit dem erkrankten Phänotyp zu untersuchen. Weiterhin wurden Haplotypanalysen für 9 SNPs durchgeführt, um die Vererbung von gemeinsamen Polymorphismen miteinander auf benachbarten Bereichen der DNA zu untersuchen. Hierbei stellte sich als Hauptbefund der durchgeführten Studie ein Haplotyp rs2052940T – rs3087584T als möglicher Risikofaktor für die Entstehung schizophrener Erkrankungsformen heraus, welcher wahrscheinlich über einen Zufallsbefund hinausgeht und nach der Leonhard-Klassifikation vor allem Patienten mit dem Phänotyp affektvolle Paraphrenie betrifft. Für die Single-SNP-Analysen ergaben sich einige nominell positive Befunde, die jedoch einer Korrektur auf multiples Testen nach Bonferroni nicht standhalten konnten, womit folglich nicht klar ist, ob es sich hierbei möglicherweise um Zufallsbefunde handelt.
Es ist nach Auswertung der vorliegenden Ergebnisse davon auszugehen, dass EphA4 zwar keinen gemeinsamen Risikofaktor für endogene Psychosen darstellt, jedoch einen Beitrag als spezifischer Risikofaktor für spezielle Unterformen schizophrener Psychosen leisten könnte. Dies konnte vor allem für die Unterform der affektvollen Paraphrenie nach Leonhard aufgezeigt werden. Um die Resultate dieser Studie zu verifizieren, wären weitere Untersuchungen wünschenswert, welche auf ein erweitertes Kollektiv mit einer höheren Anzahl von Fällen und Kontrollen zurückgreifen.
In den letzten zwei Jahrzehnten hat die Anzahl von Metastasenresektionen bei kolorektalem Karzinom signifikant zugenommen. Patienten, bei welchen die Lebermetastasen in kurativer Absicht reseziert werden, zeigen ein durchschnittliches 5-Jahres-Überleben von 30 %, Ergebnisse in der Literatur reichen sogar von 16 bis 74 %. In Kombination mit optimierter perioperativer und multimodaler Chemotherapie wurde somit ein deutlich verbessertes Gesamtüberleben für Patienten mit Metastasen bei kolorektalem Karzinom erreicht.
Viele verschiedene Faktoren beeinflussen das Gesamtüberleben der an kolorektalen Metastasen operierten Patienten. Auch neuartige Verfahren wie die zweizeitige Leberresektion werden in der Literatur noch kontrovers diskutiert. Neben der bereits etablierten chirurgischen Resektion von hepatischen kolorektalen Metastasen werden auch vermehrt Lungenmetastasen mit kurativem Behandlungsansatz reseziert.
Zweck dieser Arbeit war es, die Daten der Patienten, die in einem Zeitraum von 2003 bis 2013 am Universitätsklinikum Würzburg an kolorektalen Lebermetastasen operiert wurden, aufzuarbeiten. Die verschiedenen Einflussfaktoren auf das Gesamtüberleben dieser Patienten wurden erarbeitet und diskutiert, mit besonderem Augenmerk auf zweizeitige Leberresektionen und extrahepatische Metastasen (besonders Lungenmetastasen). Es wurde ein Kollektiv von 148 Patienten, welche mit kurativer Intention an Lebermetastasen bei kolorektalem Karzinom operiert wurden, untersucht.
Es konnte gezeigt werden, dass die Resektion von sowohl Lungen- als auch Lebermetastasen zu einem im Vergleich zum konservativen Vorgehen deutlich verbessertem Langzeitüberleben führt und daher – wenn möglich – immer in Betracht gezogen werden sollte. Insbesondere ein fortgeschrittenes Alter ist bei gutem Allgemeinzustand des Patienten kein Hindernis für eine Operation. Interessanterweise überlebten Patienten mit vermehrtem Einsatz von Blutprodukten deutlich schlechter.
Es konnte dargelegt werden, dass der neue Ansatz des zweizeitigen Resektionsverfahrens von Lebermetastasen für ein sorgfältig ausgewähltes Patientenkollektiv mit einem sehr guten Langzeitüberleben verbunden ist.
Darüber hinaus konnte nachgewiesen werden, dass die Entwicklung von zusätzlichen resektablen Lungenmetastasen bei zugrundeliegender Lebermetastasierung per se keinen negativen prognostischen Marker darstellt. Diese Beobachtungen sollen multidisziplinäre Tumorkonferenzen dazu ermutigen, alle Patienten mit resektablen synchronen und metachronen hepatischen und pulmonalen Metastasen und bei denen es Allgemeinzustand und Begleiterkrankungen erlauben, für einen kurativen chirurgischen Eingriff vorzuschlagen.
Ziel dieser Arbeit war es Veränderungen mitochondrialer Atmungskettenfunktionen von murinem Herz und muriner Niere unter einer inhalativen Anästhesie mit Sevofluran in Abhängigkeit von der inspiratorischen Sauerstoffkonzentration zu untersuchen.
In unserer in vivo Studie wurden männliche Black Six Mäuse 6 Stunden mit 1.0 MAC Sevofluran anästhesiert. Je nach Versuchsgruppe wurde mit einer inspiratorischen Sauerstoffkonzentration von 21%, 50% oder 100% ventiliert. Am Ende des Versuchsprotokolls wurden Herz und Nieren entnommen und direkt weiterverarbeitet. Es erfolgten photometrische Analysen und einer Blue Native Polyacrylamid Gel Elektrophorese zur Darstellung der mitochondrialen Komplexaktivitäten, der Proteincarbonylierung mitochondrialer Proteine und der Empfindlichkeit der isolierten Mitochondrien gegenüber der Ca2+ induzierten Schwellung.
Zusammenfassend zeigen unsere Ergebnisse, dass eine sechsstündige volatile Anästhesie zu einer deutlichen Änderung der mitochondrialen respiratorischen Komplexaktivitäten, sowohl in der Niere als auch im Herzen führt. Insbesondere wies Komplex I eine erhöhte enzymatische Aktivität auf. Die Veränderungen der mitochondrialen Respiration waren größtenteils unabhängig von der inspiratorischen Sauerstoffkonzentration und führten zu keiner mitochondrialen Schädigung. Sowohl die Superoxid Produktion als auch die Proteincarbonylierung blieben unverändert. Die mitochondriale Empfindlichkeit gegenüber der Ca2+ induzierten Schwellung zeigte keine Veränderung gegenüber den Kontrolltieren. Somit ergaben sich keine toxischen Auswirkungen hoher Sauerstoffkonzentrationen im Sinne eines erhöhten oxidativen Schadens des mitochondrialen Proteoms oder einer Membranschädigung.
In der vorliegenden Dissertation wurden die Folgen einer SPRED2-Defizienz in einem Knockout Mausmodell untersucht. Dabei wurde insbesondere die mögliche Verbindung zur Zwangsstörung, einer psychiatrischen Erkrankung beleuchtet. Das SPRED2-Protein kommt im menschlichen Körper in zahlreichen Geweben vor, besonders im Hirn wurde eine ubiquitäre Expression nachgewiesen und ein Zusammenhang mit der Neurogenese und neuronaler Differenzierung vermutet. Seine regulatorische Funktion besteht in einer inhibitorischen Wirkung auf den BDNF/TrkB-ERK-Signalweg, welcher u.a. für die Transkription neuronaler Gene verantwortlich ist. Die verwendeten SPRED2-defizienten Mäuse wurden durch Insertion eines Gene-Trap Vektors in das Spred2-Gen generiert. Die Insertion verhindert letztendlich die korrekte Translation des Proteins. Von der durch weitere Verpaarung entstehenden SPRED2-Knockout Mauslinie wurden ausschließlich männliche Tiere verwendet. Im Rahmen einer SPRED2-KO-Studie von der AG Schuh des Physiologischen Instituts der Universität Würzburg, die u.a. die Entgleisung der HHNA mit resultierendem erhöhten Stresshormonspiegel und eine Dysregulation des Mineralhaushaltshormons Aldosteron zeigte, wurden bei den Versuchstieren zwanghafte Verhaltensmuster beobachtet. Daraufhin wurden elektrophysiologische Messungen durchgeführt, die auf eine Anomalie in der synaptischen Übertragung zwischen Thalamus und Amygdala hindeuteten. Erhöhte Effizienz und Erregbarkeit der amygdaloiden Neuronen führten zu der morphologischen Untersuchung, die im Rahmen dieser Arbeit durchgeführt wurden. Da die Afferenzen des Thalamus vorwiegend in den lateralen Kern der Amygdala projizieren, wurde zunächst dieser betrachtet. Ziel der Untersuchung war es, Erkenntnisse darüber zu erlangen, ob der Knockout des SPRED2-Proteins in Mäusen zu einer veränderten Morphologie der Neuronen der lateralen Amygdala führt. Falls dies der Fall sein sollte, könnte damit zumindest ansatzweise das zwanghafte Verhalten der SPRED2-defizienten Mäusen erklärt werden. Die Hirne der Versuchstiere wurden nach der Golgi-Cox-Imprägnierung nach Glaser und Van der Loos und der Einbettung in Celloidin in 150 μm dicke Scheiben geschnitten und anschließend mithilfe eines Hellfeld-Mikroskops und des Neurolucida-Systems analysiert. Quantitativ erfasst und analysiert wurden pyramidale Klasse 1-Neuronen der lateralen Amygdala inklusive absoluter Anzahl und Dichte der Spines an ihren Dendriten. Die Untersuchung zeigte bei SPRED2-KO-Mäusen eine signifikante Erhöhung der mittleren Länge des apikalen Dendriten in Branch order 3 und eine tendenzielle Erhöhung der Gesamtzahl der Spines an den Dendriten in Branch order 1-3 gegenüber den Wildtyp-Mäusen. Daraus lässt sich folgern, dass ein Knockout des SPRED2-Proteins sich auf die Morphologie der Neuronen der lateralen Amygdala auswirkt. Die erhöhte mittlere Länge des apikalen Dendriten in Branch order 3 und die tendenziell erhöhte Spine-Anzahl korrelieren mit der gesteigerten synaptischen Übertragung und Erregbarkeit an amygdaloiden pyramidalen Neuronen. Auf molekularer Ebene kann die Hyperaktivität der lateralen Amygdala als Folge der fehlenden Inhibition des BDNF/TrkB-ERK-Signalwegs und der dadurch veränderten Expression zahlreicher synaptischer Proteine diskutiert werden. Die veränderte Morphologie der Neuronen in der lateralen Amygdala kann eine Ursache für das zwanghafte Verhalten der Mäuse sein, jedoch ist anzunehmen, dass Zwangsstörungen nicht bloß eine monokausale Ursache haben. Diese Arbeit identifiziert SPRED2 als neuen Regulator der Morphologie und Aktivität von Synapsen und die Amygdala als wichtige Hirnregion bei der Entstehung von Zwangsstörungen. SPRED2 ist somit ein vielversprechender Angriffspunkt für andere und spezifischere Untersuchungen der Hirnfunktion und eine potenzielle genetische Ursache für weitere neurologische Erkrankungen.
In der Revisionsendoprothetik des Hüftgelenks bietet ein modulares Schaftsystem die Möglichkeit der intraoperativen Anpassung an die individuellen anatomischen Gegebenheiten. Dies ist eine Untersuchung des klinisch-radiologischen Outcomes des MRP-Revisionsschafts mit diaphysärer Fixation in der Revisionschirurgie. Der MRP-Revisionsschaft zeigt ein zufriedenstellendes klinisches Outcome.
Improved treatment options for the degenerative joint disease osteoarthritis (OA) are of major interest, since OA is one of the main sources of disability, pain, and socioeconomic burden worldwide [202]. According to epidemiological data, already 27 million people suffer from OA in the US [23]. Moreover, the WHO expects OA to be the fourth most common cause of disability in 2020 [203], illustrating the need for effective and long-lasting therapy options of severe cartilage defects. Despite numerous clinically available products for the treatment of cartilage defects [62], the development of more cartilage-specific materials is still at the beginning.
Hyaluronic acid (HA) is a major component of the cartilaginous extracellular matrix (ECM) and inherently creates a cell-friendly niche by providing cell attachment and migration sites. Furthermore, it is known that the functional groups of HA are well suited for chemical modification. These characteristics render HA an attractive material for hydrogel-based tissue engineering approaches. Poly(glycidol) (PG) as chemical crosslinker basically features similar chemical characteristics as the widely used poly(ethylene glycol) (PEG), but provides additional side groups at each repeating unit that can be further chemically functionalized. With the introduction of PG as multifunctional crosslinker for HA gels, a higher cross-linking density and, accordingly, a greater potential for biomimetic functionalization may be achieved. However, despite the mentioned potential benefits, PG has not been used for cartilage regeneration approaches so far.
The initial aim of the study was to set up and optimize a HA-based hydrogel for the chondrogenic differentiation of mesenchymal stromal cells (MSCs), using different amounts and variations of cross-linkers. Therefore, the hydrogel composition was optimized by the utilization of different PEG diacrylate (PEGDA) concentrations to cross-link thiol-modified HA (Glycosil, HA-SH) via Michael addition. We aimed to generate volumestable scaffolds that simultaneously enable a maximum of ECM deposition. Histological and biochemical analysis showed 0.4% PEGDA as the most suitable concentration for these requirements (Section 5.1.2).
In order to evaluate the impact of a differently designed cross-linker on MSC chondrogenesis, HA-SH was cross-linked with PEGTA (0.6%) and compared to PEGDA (0.4%) in a next step. Following this, acrylated PG (PG-Acr) as multifunctional cross-linker alternative to acrylated PEG was evaluated. It provides around five times more functional groups when utilized in PG-Acr (0.6%) HA-SH hydrogels compared to PEGTA (0.6%) HA-SH hydrogels, thus enabling higher degrees of biomimetic functionalization. Determination of cartilage-specific ECM components showed no substantial differences between both cross-linkers while the deposition of cartilaginous matrix appeared more homogeneous in HA-SH PG-Acr gels. Taken together, we were able to successfully increase the possibilities for biomimetic functionalization in the developed HA-SH hydrogel system by the introduction of PG-Acr as cross-linker without negatively affecting MSC chondrogenesis (Section 5.1.3).
The next part of this thesis focused extensively on the biomimetic functionalization of PG-Acr (0.6%) cross-linked HA-SH hydrogels. Here, either biomimetic peptides or a chondrogenic growth factor were covalently bound into the hydrogels.
Interestingly, the incorporation of a N-cadherin mimetic (HAV), a collagen type II binding (KLER), or a cell adhesion-mediating peptide (RGD) yielded no improvement of MSC chondrogenesis. For instance, the covalent binding of 2.5mM HAV changed morphology of cell nuclei and reduced GAG production while the incorporation of 1.0mM RGD impaired collagen production. These findings may be attributed to the already supportive conditions of the employed HA-based hydrogels for chondrogenic differentiation. Most of the previous studies reporting positive peptide effects on chondrogenesis have been carried out in less supportive PEG hydrogels or in significantly stiffer MeHA-based hydrogels [99, 101, 160]. Thus, the incorporation of peptides may be more important under unfavorable conditions while inert gel systems may be useful for studying single peptide effects (Section 5.2.1).
The chondrogenic factor transforming growth factor beta 1 (TGF-b1) served as an example for growth factor binding to PG-Acr. The utilization of covalently bound TGF-b1 may thereby help overcome the need for repeated administration of TGF-b1 in in vivo applications, which may be an advantage for potential clinical application. Thus, the effect of covalently incorporated TGF-b1 was compared to the effect of the same amount of TGF-b1 without covalent binding (100nM TGF-b1) on MSC chondrogenesis. It was successfully demonstrated that covalent incorporation of TGF-b1 had a significant positive effect in a dose-dependent manner. Chondrogenesis of MSCs in hydrogels with covalently bound TGF-b1 showed enhanced levels of chondrogenesis compared to hydrogels into which TGF-b1 was merely mixed, as shown by stronger staining for GAGs, total collagen, aggrecan and collagen type II. Biochemical evaluation of GAG and collagen amounts, as well as Western blot analysis confirmed the histological results. Furthermore, the positive effect of covalently bound TGF-b1 was shown by increased expression of chondrogenic marker genes COL2A1, ACAN and SOX9. In summary, covalent growth factor incorporation utilizing PG-Acr as cross-linker demonstrated significant positive effects on chondrogenic differentiation of MSCs (Section 5.2.2).
In general, PG-Acr cross-linked HA hydrogels generated by Michael addition represent a versatile hydrogel platform due to their high degree of acrylate functionality. These hydrogels may further offer the opportunity to combine several biological modifications, such as the incorporation of biomimetic peptides together with growth factors, within one cell carrier.
A proof-of-principle experiment demonstrated the suitability of pure PG gels for studying single peptide effects. Here, the hydrogels were generated by the utilization of thiol-ene-click reaction. In this setting, without the supportive background of hyaluronic acid, MSCs showed enhanced chondrogenic differentiation in response to the incorporation of 1.0mM HAV. This was demonstrated by staining for GAGs, the cartilage-specific ECM molecules aggrecan and type II collagen, and by increased GAG and total collagen amounts shown by biochemical analysis. Thus, pure PG gels exhibit the potential to study the effects and interplay of peptides and growth factors in a highly modifiable, bioinert hydrogel environment.
The last section of the thesis was carried out as part of the EU project HydroZONES that aims to develop and generate zonal constructs. The importance of zonal organization has attracted increased attention in the last years [127, 128], however, it is still underrepresented in tissue engineering approaches so far. Thus, the feasibility of zonal distribution of cells in a scaffold combining two differently composed hydrogels was investigated. A HA-SH(FMZ) containing bottom layer was generated and a pure PG top layer was subsequently cast on top of it, utilizing both times thiol-ene-click reaction. Indeed, stable, hierarchical constructs were generated that allowed encapsulated MSCs to differentiate chondrogenically in both zones as shown by staining for GAGs and collagen type II, and by quantification of GAG amount. Thus, the feasibility of differently composed zonal hydrogels utilizing PG as a main component was successfully demonstrated (Section 5.4).
With the first-time utilization and evaluation of PG-Acr as versatile multifunctional cross-linker for the preparation of Michael addition-generated HA-SH hydrogels in the context of cartilage tissue engineering, a highly modifiable HA-based hydrogel system was introduced. It may be used in future studies as an easily applicable and versatile toolbox for the generation of biomimetically functionalized hydrogels for cell-based cartilage regeneration. The introduction of reinforcement structures to enhance mechanical resistance may thereby further increase the potential of this system for clinical applications.
Additionally, it was also demonstrated that thiol-ene clickable hydrogels can be used for the generation of cell-laden, pure PG gels or for the generation of more complex, coherent zonal constructs. Furthermore, thiol-ene clickable PG hydrogels have already been further modified and successfully been used in 3D bioprinting experiments [204]. 3D bioprinting, as part of the evolving biofabrication field [205], offers the possibilities to generate complex and hierarchical structures, and to exactly position defined layers, yet at the same time alters the requirements for the utilized hydrogels [159, 206–209]. Since a robust chondrogenesis of MSCs was demonstrated in the thiol-ene clickable hydrogel systems, they may serve as a basis for the development of hydrogels as so called bioinks which may be utilized in more sophisticated biofabrication processes.
Bei den Charcot-Marie-Tooth (CMT) Neuropathien handelt es sich um erbliche Erkrankungen des peripheren Nervensystems, die progredient zu motorischen und sensorischen Defiziten führen und für die bislang keine kausalen Therapieoptionen existieren. In verschiedenen Studien konnte gezeigt werden, dass Entzündungsreaktionen, insbesondere durch Lymphozyten und Makrophagen vermittelt, eine bedeutende Rolle bei der Pathogenese dieser Erkrankung spielen. Neben neuronaler und axonaler Schädigung, sowie Demyelinisierung ist in untersuchten Myelin Mutanten auch eine erhöhte Anzahl an denervierten neuromuskulärer Endplatten zu erkennen. Eine genetische Blockade der Makrophagen-Aktivierung konnte in den Studien eine Verbesserung sämtlicher neuropathologischer Merkmale bei gleichzeitig reduzierter Makrophagenanzahl zeigen. Ob und welche Rolle Makrophagen bei der Denervation neuromuskulärer Endplatten spielen, blieb bislang ungeklärt.
In dieser Studie konnte in allen untersuchten Myelin Mutanten im Vergleich zum Wildtyp eine Zunahme an neuromuskulären Synapsen beobachtet werden, die mit Makrophagen räumlich assoziiert waren. Daneben zeigten entsprechende Myelin Mutanten eine Zunahme denervierter und partiell denervierter Endplatten und zwar interessanterweise direkt proportional zur Anzahl an Synapsen in Assoziation mit Makrophagen. Das bedeutet, dass die Anzahl an Endplatten in Assoziation mit Makrophagen verhältnismäßig parallel zur Anzahl an denervierten Endplatten zunahm, während die Anzahl an Makrophagen im gesamten Muskel nahezu unverändert blieb. Dies deutet eine mögliche Rolle der räumlich mit Endplatten assoziierten Makrophagen an deren Denervation an. Dabei waren alle Synapsen in Assoziation mit Makrophagen innerviert und damit morphologisch intakt. Bei doppel-mutanten Mäusen mit genetischer Blockade der Makrophagen-Aktivierung waren die beschriebenen pathologischen Merkmale an der neuromuskulären Synapse deutlich reduziert bei gleichzeitig signifikanter Abnahme an Makrophagen in Assoziation mit Endplatten. Ähnliche pathologische Auffälligkeiten wie bei Myelin Mutanten fanden sich in geringerer Ausprägung auch im Wildtyp im Rahmen des Alterungsprozesses sowie auch bei Mäusen mit Defizienz des neurotrophen Faktors CNTF.
Zusammenfassend deuten die Ergebnisse darauf hin, dass sowohl in der Pathogenese der CMT Neuropathie wie auch im Rahmen altersbedingter Neurodegeneration ein Makrophagen-vermittelter Schaden an der neuromuskulären Endplatte entsteht. Wesentliche Mediatoren scheinen hierbei das von Fibroblasten und vermutlich auch perisynaptischen Fibroblasten exprimierte CSF-1 zu sein, sowie MCP-1, das durch Schwann Zellen und möglicherweise auch von terminalen Schwann Zellen freigesetzt wird. Auch eine Defizienz des neurotrophen Faktors CNTF bewirkt zumindest in geringem Ausmaß eine Zunahme der pathologischen Merkmale Denervation und Makrophagen-Endplatten-Assoziation im Vergleich zum Wildtyp. Diese Ergebnisse erweitern insbesondere das Wissen um Pathomechanismen an der neuromuskulären Endplatte und eröffnen neue Möglichkeiten der Behandlung für CMT und weitere neuromuskuläre Erkrankungen.
Ziel:
Unter dem Tyrosinkinaseinhibitor Dasatinib haben sich bei Patienten Nebenwirkungen gezeigt, die eine immunmodulierende Wirkung des Medikamentes vermuten lassen. Diese Arbeit hatte daher zum Ziel die Auswirkungen von Dasatinib auf dendritische Zellen zu untersuchen.
Material und Methoden:
Es wurden dendritische Zellen mittels Zugabe von IL4 und GM-CSF aus Monozyten gesunder Spender generiert. Anschließend wurden die Zellen nach Zugabe von Dasatinib mit Zymosan (25 µg/ml) oder LPS (100 ng/ml)zur Ausreifung gebracht und die Phosphorylierung der Signaltransduktionsproteine p38, ERK, Akt, c-jun sowie die Translokation der NFkB-Bestandteile RelA und RelB in den Zellkern mittels Westernblot gemessen. Zudem erfolgt eine Konzentrationsbestimmung der sekretierten Interleukine 10 und 12 im Kulturmedium mittels ELISA.
Ergebnisse:
Dasatinib inhibierte die die Phosphorylierung der Kinasen p38 und ERK nach Stimulation mit LPS und Zymosan. Zudem kam es zu einer reduzierten Sekretion von Interleukin 10 und einer vermehrten Sekretion von Interleukin 12 unter Dasatinib nach Stimulation mit LPS.
Fazit:
Dasatinib verstärkt die Sekretion von Interleukin 12 und vermindert die Sekretion von Interleukin 10 in ausgereiften dendritischen Zellen durch Hemmung der Kinasen p38 und ERK, vermutlich indirekt vermittelt über Src-Kinasen. Dies könnte eine Erklärung für immunmodulatorische Effekte wie das Auftreten von Autoimmunerkrankungen und eine LGL-Expansion bei mit Dasatinib behandelten Patienten bieten.
